Die Amygdala ist ein Kernkomplex, der dicht von serotonergen Afferenzen innerviert wird. Sowohl bei Tieren als auch beim Menschen spielen Interaktionen zwischen dem serotonergen System und der Amygdala bei der Verarbeitung von Reizen, die mit Angst oder Stress assoziiert sind, eine zentrale Rolle. Genetische Variationen im serotonergen System und/oder dauerhafter Stress können dazu führen, dass diese Verarbeitungsprozesse fehlerhaft ablaufen, wodurch Verhaltensanormalitäten bzw. die Entstehung psychiatrischer Erkrankungen begünstigt werden. Die Zielneurone der serotonergen Transmission in der Amygdala, die molekularen Mechanismen möglicher Interaktionen und strukturelle Konsequenzen der Störungen dieser Interaktionen sind jedoch bis zum heutigen Zeitpunkt noch nicht vollständig bekannt. Daher bestand ein Ziel der vorliegenden Arbeit darin, den Einfluss eines Ungleichgewichts im serotonergen System (5-Htt KO) sowie von wiederholtem, sozialem Stress auf die neuronale Morphologie der Amygdala zu analysieren und Zielneurone serotonerger Afferenzen zu identifizieren und zu charakterisieren, um die neuronalen Netzwerke der Emotionsverarbeitung besser verstehen zu können. Um vom 5-Htt–Genotyp abhängige und stressbedingte neuromorphologische Veränderungen zu untersuchen, wurden dreidimensionale Rekonstruktionen von Neuronen der laterobasalen Amygdala von männlichen, adulten Wildtyp (WT)- und 5-Htt KO-Mäusen angefertigt und bezüglich verschiedener morphologischer Parameter ausgewertet. An den Pyramidenzellen wurden nur geringfügige Veränderungen der dendritischen Komplexität, jedoch, im Vergleich zu WT-Mäusen, eine wesentliche Erhöhung der Dornendichte an spezifischen dendritischen Kompartimenten bei gestressten WT-Mäusen, sowie nicht gestressten und gestressten 5-Htt KO-Mäusen nachgewiesen. Im Vergleich zu nicht gestressten WT–Mäusen war die dendritische Dornendichte aller anderen Gruppen gleichermaßen erhöht. Die Sternzelle, zeigten bezüglich der untersuchten Parameter keine morphologischen Veränderungen auf. Eine besondere Subpopulation der Interneurone stellen die NeuropeptidY (NPY)–Neurone der laterobasalen Amygdala dar, da sie in diesen Nuclei anxiolytisch wirken. Es gibt nur wenige Anhaltspunkte darüber, durch welche Systeme NPY–Neurone moduliert werden. Da sowohl NPY–Neurone in der laterobasalen Amygdala als auch das serotonerge System an angstregulierenden Prozessen beteiligt sind, sollte im zweiten Teil der vorliegenden Arbeit untersucht werden, ob es sich bei diesen Neuronen um Zielstrukturen des serotonergen Systems handelt. Mittels licht- und elektronenmikroskopischer Analysen wurden synaptische Kontakte zwischen serotonergen Afferenzen und NPY-immunreaktiven Neuronen in der laterobasalen Amygdala von Ratten verifiziert. Da der funktionelle Einfluss der serotonergen Innervation auf diese Zielneurone von deren Serotoninrezeptor (5-HTR)-Ausstattung abhängt, wurden Koexpressionsanalysen von NPY mRNA mit den mRNAs verschiedener 5-HTR durchgeführt. Die Analysen ergaben, dass NPY mRNA–reaktive Neurone in der laterobasalen Amygdala 5-HT1A und 5-HT2C, jedoch nicht 5-HT3 mRNA koexprimieren. Die in der vorliegenden Arbeit erzielten Resultate liefern neue Erkenntnisse über den Einfluss des serotonergen Systems auf die laterobasale Amygdala von Mäusen und Ratten. Bei den Veränderungen der dendritischen Dornendichte nach sozialen Stresserfahrungen könnte es sich um neuroadaptive bzw. kompensatorische Mechanismen der Pyramidenzellen handeln, die WT-Mäusen eine Anpassung an sich ändernde, negative Umweltbedingungen ermöglicht. Die erhöhte Dornendichte könnte dabei die Ausbildung eines „emotionalen Gedächtnisses“ repräsentieren, das eine flexible Verhaltensantwort auf ein erneutes Auftauchen von Gefahr erlaubt. Eine solche Modulation der Erregbarkeit der laterobasalen Amygdala könnte beispielsweise über eine situationsentsprechende Hemmung des Outputs der Pyramidenzellen durch differentiell aktive inhibitorische Netzwerke erfolgen. Eine differentielle Aktivierung kann z. B. über unterschiedliche Rezeptorausstattungen, wie es in der Subpopulation der NPY–Neurone in der vorliegenden Arbeit nachgewiesen wurde, erfolgen. Das erhöhte angstähnliche Verhalten der 5-Htt KO-Mäuse nach wiederholtem Stress könnte mit der Unfähigkeit zusammenhängen, in entsprechenden Situationen durch Neubildung von Dornen zu reagieren, da die Dornendichte bei diesen Tieren schon unter stressarmen Umweltbedingungen ihr Maximum erreicht hat. Sowohl Fehlfunktionen der neuronalen Plastizität als auch mögliche Fehlfunktionen der differentiellen Inhibierung der Pyramidenzellen durch Interneurone, die durch genetische Variationen und/oder Stress bedingt sein können, könnten eine „offene Tür“ repräsentieren, die zu manifesten Auffälligkeiten im Verhalten bei Tieren führt bzw. auch zur Entstehung bestimmter psychiatrischer Erkrankungen beim Menschen beiträgt.
In der Diagnostik und Therapie der KHK sind das frühzeitige Erkennen und die Beurteilung funktioneller Folgen atherosklerotischer Veränderungen von großer Bedeutung. Die First-Pass MR-Bildgebung ermöglicht Aussagen über die myokardiale Perfusion und damit die hämodynamische Relevanz einer Koronarstenose. In der vorliegenden Arbeit wurden quantitative Werte für die myokardiale Durchblutung gesunder Probanden unter Adenosin-induziertem Stress und in Ruhe unter Einsatz der Präbolustechnik bestimmt. Eine exakte Darstellung der arteriellen Inputfunktion wurde durch einen Kontrastmittelbolus in niedriger Dosierung erreicht, die Verwendung höherer Kontrastmitteldosen führte dagegen zu einem verbesserten Signal-zu-Rausch-Verhältnis im Myokard. Die Absolutwerte der myokardialen Perfusion unter Stressbedingungen und in Ruhe wie auch die myokardiale Perfusionsreserve zeigten vergleichbare Mittelwerte, wiesen aber eine geringere Streubreite im Vergleich zu früheren MR Studien auf und waren vergleichbar mit in PET-Studien erzielten Ergebnissen. Weiterhin wurden unter Verwendung dieser Methode Werte für das myokardiale Verteilungsvolumen des Kontrastmittels als wichtiger Parameter in der Differenzierung von gesundem und infarziertem Herzmuskelgewebe ermittelt und die Laufzeit der Boluspassage nach Injektion in Ruhe und unter Stress bestimmt, die zur Unterscheidung von antegrad perfundiertem und von über Kollateralen versorgtem Myokard dienen kann. Mit Hilfe der MRT war es auch möglich, Unterschiede zwischen subendo- und subepimyokardialer Perfusion zu quantifizieren. Die erzielten Ergebnisse entsprechen bisher publizierten Werten, die mit anderen Modalitäten gewonnen wurden. Der Vergleich der absoluten Perfusion bei verminderter zeitlicher Auflösung mit den bei hoher zeitlicher Auflösung gemessenen Werten ergab nur geringfügige Abweichungen der Ergebnisse voneinander. Dadurch eröffnet sich die Möglichkeit, durch die Zeitersparnis mehrere Schichten abwechselnd bei verschiedenen Herzschlägen zu messen und damit eine erweiterte Abdeckung des linksventrikulären Myokards zu erreichen. Durch die quantitative Auswertung der First-Pass MR-Perfusionsmessung stellt die beschriebene Methode eine vielversprechende Option im Bereich der nichtinvasiven Diagnostik verschiedener myokardialer Erkrankungen dar.
Climate and land-use changes cause increasing stress to pollinators but the molecular pathways underlying stress responses are poorly understood. Here, we analyzed the transcriptomic response of Bombus lucorum workers to temperature and livestock grazing. Bumblebees sampled along an elevational gradient, and from differently managed grassland sites (livestock grazing vs unmanaged) in the German Alps did not differ in the expression of genes known for thermal stress responses. Instead, metabolic energy production pathways were upregulated in bumblebees sampled in mid- or high elevations or during cool temperatures. Extensive grazing pressure led to an upregulation of genetic pathways involved in immunoregulation and DNA-repair. We conclude that widespread bumblebees are tolerant toward temperature fluctuations in temperate mountain environments. Moderate temperature increases may even release bumblebees from metabolic stress. However, transcriptome responses to even moderate management regimes highlight the completely underestimated complexity of human influence on natural pollinators.
Studienobjekte: Um angemessene Reaktionen auf die gebotene Stresssituation zu bekommen wurden 29 Probanden, davon 14 Männer und 15 Frauen verglichen. Dabei wurde auf einen breiten Querschnitt der beruflichen Tätigkeit geachtet. Versuchsaufbau: Die Probanden wurden anhand von Rechenaufgaben an einem Computer in Stress gesetzt, die dadurch auftretenden Reaktionen der Atmung wurden mittels Spirometrie aufgezeichnet. Gleichzeitig wurden durchgehend der Blutdruck und die Herzfrequenz bestimmt, zudem in regelmäßigen Abständen die Blutgase sowie Urin-Katecholamine abgenommen. Versuchsort: Universität Würzburg, Abteilung für Betriebsmedizin Versuchspersonen: 15 Frauen im Alter von 17-31 sowie 15 Männer von 19-33 Jahren nahmen an den Versuchen teil. Ergebnisse: Bezugnehmend auf die Urin-Katecholamine zeigte sich ein Anstieg während der Stressphase, sowohl bei Frauen, als auch bei Männern. RQ, VO2, VCO2, Vt sowie VE ergaben allesamt einen Anstieg während der Stressphase bei beiden Geschlechtern. Die Atemfrequenz fiel stressinduziert ab. Der Blutdruck stieg erwartungsgemäß, sowohl systolisch als auch diastolisch an. Der periphere Widerstand verzeichnete seinen höchsten Wert direkt im Anschluss an das Testende. Zusammenfassung: Während Stressbelastung kam es wohl als Ausdruck einer vermehrten Glucoseutilisation zu einem RQ Anstieg, gleichzeitig im Rahmen der Kompensationsmechanismen zu einem Abfall der Atemfrequenz bei gleichzeitigem Anstieg des Atemzugvolumens. Trotz Anstiegs der Blutdruckwerte bei gleichzeitig erhöhten Katecholaminen kam es zu einem Abfall des peripheren Widerstandes, was als Zentralisationsmechanismus zu deuten ist. Im Anschluss an die Stressbelastung erreichte dieser seinen höchsten Wert, somit ist von einer vermehrten Gefährdung eines gefäßstenosierenden Prozesses akut nach Beendigung einer psychischen Belastungssituation auszugehen.
In der vorgestellten Arbeit wurden das Wachstum und das physiologische Verhalten von Zea mays auf Müllheizkraftwerk (MHKW) -Schlacke im Vergleich zu Gartenerde als Kulturmedium untersucht. Dabei stand das Wurzelsystem der Maispflanzen im Mittelpunkt des Interesses. Da feste Bodensubstrate verwendet wurden, mußten diese zu Beginn der Experimente chemisch, physikalisch und bodenbiologisch charakterisiert werden. Die Analyse der Schlacke zeigte, daß Schlacke ein multifaktorielles Streßsystem darstellt: Sie enthält einen hohen Gehalt an leicht löslichen Salzen, v.a. NaCl (bis zu 220 mM in der Bodenlösung). MHKW-Schlacke ist dagegen arm an Stickstoff und pflanzenverfügbarem Phosphat. Der pH-Wert der Bodenlösung von Schlacke ist stark alkalisch (pH 8.4 - 9.0). Darüber hinaus besitzt Schlacke einen hohen Gehalt an potentiell toxischen Schwermetallen und weist im Vergleich zum Kontrollsubstrat Gartenerde eine verdichtete Bodenstruktur mit erhöhtem mechanischen Widerstand auf. Im Vergleich zu der Kontroll-Anzucht auf Gartenerde reagierten die auf Schlacke kultivierten Mais-Pflanzen mit vermindertem Wachstum: Sproß und Wurzel erreichten nur die Hälfte der Länge der Kontrollpflanzen. Ein Vergleich der Biomassen von Sproß und Wurzel zeigte, daß das Sproßwachstum der Schlacke-Pflanzen stärker eingeschränkt ist als das Wurzelwachstum, woraus ein vergrößertes Wurzel / Sproß-Verhältnis resultiert. Das Wachstum von jungen Mais-Pflanzen auf Schlacke ist jedoch nicht in dem Maß eingeschränkt, wie es aufgrund der hohen Salzbelastung zu erwarten wäre. In einem Vergleichsexperiment mit Mais-Pflanzen, die in einer Nährlösung mit Zusatz von 100 mM NaCl kultiviert wurden, war das Wachstum erheblich schlechter und in den Blättern akkumulierte weitaus mehr Natrium als in Schlacke-Pflanzen. Hier wird der positive Einfluß des hohen Calciumgehaltes der Schlacke deutlich. Die Beeinträchtigung des Wachstums von Mais bei Kultur auf Schlacke wird hauptsächlich auf Phosphatmangel zurückgeführt, da durch Düngung eine beträchtliche Wachstumsverbesserung erzielt werden kann. Zudem wurden keine toxischen Konzentrationen an Schwermetallen im Blattgewebe von auf Schlacke kultivierten Pflanzen gefunden. Der Photosynthese-Apparat der Schlacke-kultivierten Pflanzen war sehr leistungsfähig: Es bestand keine Beeinträchtigung in der Energieverfügbarkeit (Quantenausbeute des Photosystems II) und die Lichtsättigung der photo-synthetischen Elektronentransportrate lag sogar höher als bei den Kontrollpflanzen. Die Bestimmung des „adenylate energy charge“ bestätigte diesen Sachverhalt. Das Wurzelsystem von Zea mays auf Schlacke wies strukturelle Veränderungen auf. Neben der verkürzten Wurzellänge und dem vergrößerten Wurzeldurchmesser der Schlacke-Pflanzen ergaben mikroskopische Untersuchungen, daß die Wurzeln durch Kultur auf Schlacke mit einer mechanischen Verstärkung reagieren: Stärker ausgeprägte tangentiale Zellwandverdickungen der Endodermis im tertiären Zustand und Zellwandmodifikationen in den radialen Zellwänden der Rhizodermis (Phi-Verdickungen). Für monokotyle Arten, insbesondere für Mais, gibt es bisher keine Beschreibung von Phi-Verdickungen in der Literatur. Gaschromatographische und massenspektrometrische Untersuchungen belegen, daß sich die Zellwände von auf Erde und Schlacke kultivierten Maiswurzeln im Hinblick auf den Gesamtgehalt an Lignin (endodermale Zellwandisolate) und in der Ligninzusammensetzung (hypodermale Zellwandisolate) unterscheiden: In Schlacke-kultivierten Maiswurzeln wurde ein höherer Anteil an dem Lignin-Monomer p Hydroxyphenyl gefunden, was zu einem höher verdichteten Lignin führt (Streßlignin). Die endodermalen Zellwände von auf Schlacke-kulivierten Pflanzen hatten dagegen einen höheren Gesamtlignin-Gehalt als die entsprechenden Kontrollen, was ebenfalls eine mechanische Verstärkung der Wurzel bewirkt. In Bezug auf Suberin konnten keine Unterschiede zwischen den verschiedenen Anzuchten gefunden werden, weder in den hypodermalen noch in den endodermalen Zellwandisolaten. Die verschiedenen Streßfaktoren führen demnach nicht zu einer verstärkten Imprägnierung der Zellwände mit lipophilem Material. Die Zellwände von Mais spielen eine wichtige Rolle bei der Immobilisierung von Schwermetallen. Die Zellwandisolate von auf Erde und Schlacke kultivierten Mais-Pflanzen wiesen je nach Schwermetall-Element 43 - 100 % des Gesamtgehaltes auf. Die absoluten Gehalte in den Zellwandisolaten von auf Schlacke angezogenen Pflanzen waren dabei höher als die entsprechenden Werte der Kontrolle. Eine Anreicherung in den Zellwänden wurde hauptsächlich für die Schwermetalle Zink, Blei, Nickel und Chrom beobachtet. Als unspezifische Streßantwort reagierten Maispflanzen auf die Kultur in Schlacke mit einer erhöhten Peroxidaseaktivität in der interzellulären Waschflüssigkeit. Die Peroxidaseaktivität des Symplastens der Wurzel unterscheidet sich zwischen den beiden Anzuchten dagegen nicht. Die Konzentration des Phytohormons Abscisinsäure (ABA) war in Blättern von auf Schlacke kultivierten Pflanzen von Zea mays und Vicia faba im Vergleich zu den Kontrollpflanzen erhöht. Dieser Anstieg ist eine Folge der erhöhten Salzbelastung der Schlacke, da die ABA-Gehalte entsprechender Blätter von auf gewaschener Schlacke kultivierten Pflanzen annähernd den Kontrollwerten entsprachen. Bei der Verteilung von ABA zwischen der Wurzel und der Bodenlösung der umliegenden Rhizosphäre konnte das als Anionenfalle bekannte Prinzip bestätigt werden. Nach diesem Modell reichert sich ABA im alkalischten Kompartiment an (hier: Schlacke-Bodenlösung). In den Wurzeln konnte nur in der Maiskultur auf Schlacke ein erhöhter Gehalt gefunden werden, nicht dagegen in der Vicia faba-Kultur. Dieser Unterschied liegt daran, daß Mais im Gegensatz zu Vicia faba eine exodermale Spezies ist und die Exodermis für ABA eine Barriere darstellt, was den ABA-Efflux in die Rhizosphäre verhindert. Im Wurzelgewebe von auf Schlacke kultivierten Maispflanzen wurde ein im Vergleich zur Kontrolle 15-facher Gehalt an wasserlöslichen, nicht proteingebundenen Sulfhydrylgruppen nachgewiesen. Diese auf Schwermetallstreß zurückzuführende Reaktion impliziert, daß die in der Schlacke-Bodenlösung vorhandenen Schwermetalle nicht ausreichend im Apoplasten zurückgehalten werden und bis in den Symplasten vordringen können.
Background
The onset of mental illness such as depression and anxiety disorders in pregnancy and postpartum period is common. The coronavirus induced disease 2019 (COVID-19) pandemic and the resulting public policy responses represent an exceptional situation worldwide and there are hints for adverse psychosocial impact, hence, the study of psychological effects of the pandemic in women during hospitalization for delivery and in the postpartum period is highly relevant.
Methods
Patients who gave birth during the first wave of the COVID-19 pandemic in Germany (March to June 2020) at the Department of Obstetrics and Gynecology, University of Würzburg, Germany, were recruited at hospital admission for delivery. Biosamples were collected for analysis of SARS-CoV-2 infection and various stress hormones and interleukin-6 (IL-6). In addition to sociodemographic and medical obstetric data, survey questionnaires in relation to concerns about and fear of COVID-19, depression, stress, anxiety, loneliness, maternal self-efficacy and the mother–child bonding were administered at T1 (delivery stay) and T2 (3–6 months postpartum).
Results
In total, all 94 recruited patients had a moderate concern of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) at T1 with a significant rise at T2. This concern correlated with low to low-medium general psychosocial stress levels and stress symptoms, and the women showed a significant increase of active coping from T1 to T2. Anxiety levels were low and the Edinburgh Postnatal Depression Scale showed a medium score of 5 with a significant (T1), but only week correlation with the concerns about SARS-CoV-2. In contrast to the overall good maternal bonding without correlation to SARS-CoV-2 concern, the maternal self-efficiency correlated negatively with the obstetric impairment caused by the COVID-19 pandemic.
Conclusion
Obstetric patients` concerns regarding SARS-CoV-2 and the accompanying pandemic increased during the course of the pandemic correlating positively with stress and depression. Of note is the increase in active coping over time and the overall good mother–child-bonding. Maternal self-efficacy was affected in part by the restrictions of the pandemic.
Designing of implant surfaces using a suitable ligand for cell adhesion to stimulate specific biological responses of stem cells will boost the application of regenerative implants. For example, materials that facilitate rapid and guided migration of stem cells would promote tissue regeneration. When seeded on fibronectin (FN) that was homogeneously immmobilized to NCO-sP(EO-stat-PO), which otherwise prevents protein binding and cell adhesion, human mesenchymal stem cells (MSC) revealed a faster migration, increased spreading and a more rapid organization of different cellular components for cell adhesion on fibronectin than on a glass surface. To further explore, how a structural organization of FN controls the behavior of MSC, adhesive lines of FN with varying width between 10 mu m and 80 mu m and spacings between 5 mu m and 20 mu m that did not allow cell adhesion were generated. In dependance on both line width and gaps, cells formed adjacent cell contacts, were individually organized in lines, or bridged the lines. With decreasing sizes of FN lines, speed and directionality of cell migration increased, which correlated with organization of the actin cytoskeleton, size and shape of the nuclei as well as of focal adhesions. Together, defined FN lines and gaps enabled a fine tuning of the structural organization of cellular components and migration. Microstructured adhesive substrates can mimic the extracellular matrix in vivo and stimulate cellular mechanisms which play a role in tissue regeneration.
Introduction: National and international guidelines recommend early integration of evidence-based multimodal interventions and programs, especially with a focus on relaxation techniques and other Mind–Body-based methods to maintain the quality of life of oncology patients, improve treatment tolerability, and promote healthy lifestyle behaviors. Consequently, we aim to understand what drives patients and how they navigate integrative medicine to best advise them. This study aimed to detect possible topics of particular interest to patients and identify the patient groups that could benefit most from further programs. Furthermore, we aimed to investigate if patients are open-minded toward integrative oncology concepts and learn about their motivational level to maintain or change behavior.
Methods: Between August 2019 and October 2020 we surveyed patients undergoing oncological therapy in a university oncological outpatient center using a custom-developed questionnaire based on established Mind–Body Medicine concepts.
Results: We included 294 patients with various cancers. More than half reported problems sleeping through (61%) and 42% felt stressed frequently, invariably rating this as detrimental to their health. Moreover, a slight majority (52%) felt physically limited due to their disease and only 30% performed defined exercise programs. Women were significantly more likely to feel stressed and reported with alarming frequency that they often feel “everything was up to them.” The 40–65-year-olds reported significantly less restful sleep, more stress and were more dissatisfied with their situation. However, this group already used natural remedies most frequently and was most often motivated to use relaxation techniques in the next 6 months. The lower the perceived individual energy level (EL), the less frequently patients did sport, the more frequently they felt their disease impaired their activity, mostly feeling stressed and tense. We also found significant associations between negative emotions/thoughts and the variables “sleep,” “use of relaxation techniques,” “personal stress perception,” and “successful lifestyle modification.”
Conclusion: Mind–Body programs that focus on patient’s individual resources, with tools to explore impairing patterns of self-perception and cognitive biases, can be a valuable resource for oncology patients and should therefore be part of an integrative medical treatment concept.
The RNA-binding protein RC3H1 (also known as ROQUIN) promotes TNF\(\alpha\) mRNA decay via a 3'UTR constitutive decay element (CDE). Here we applied PAR-CLIP to human RC3H1 to identify ~3,800 mRNA targets with >16,000 binding sites. A large number of sites are distinct from the consensus CDE and revealed a structure-sequence motif with U-rich sequences embedded in hairpins. RC3H1 binds preferentially short-lived and DNA damage-induced mRNAs, indicating a role of this RNA-binding protein in the post-transcriptional regulation of the DNA damage response. Intriguingly, RC3H1 affects expression of the NF-\(\kappa\)B pathway regulators such as I\(\kappa\)B\(\alpha\) and A20. RC3H1 uses ROQ and Zn-finger domains to contact a binding site in the A20 3'UTR, demonstrating a not yet recognized mode of RC3H1 binding. Knockdown of RC3H1 resulted in increased A20 protein expression, thereby interfering with I\(\kappa\)B kinase and NF-\(\kappa\)B activities, demonstrating that RC3H1 can modulate the activity of the IKK/NF-\(\kappa\)B pathway.
The hoatzin (Opisthocomus hoazin) is an enigmatic bird that lives in the riparian lowlands of northern South America. Among its peculiar attributes are 1) microbial foregut fermentation, unique in birds, to convert plant cellulose in the foliage which it consumes into simple sugars, 2) an ongoing debate about the puzzling taxonomic position, although a relationship to the Cuculiformes appears likely, 3) adaptive wing claws in the young which are used for climbing, and 4) co-operative breeding behaviour. Despite the information available on digestive mode and taxonomy little has been published on its breeding biology and behaviour and until now almost all knowledge was based on a study in the savannah of Venezuela. This is the first detailed study of the hoatzin’s nesting ecology in a rainforest habitat. From 1995-1998 and in 2000 I monitored a hoatzin population which consisted of approximately 700 individuals in an Amazonian rainforest in Ecuador situated in the Cuyabeno Wildlife Reserve (between 0°02’ N, 76°0’ W, 0°03’ S, and 76°14’ W). The area is composed of various black water lagoons and small rivers, flooded forests and terra firme forest. Primarily, I examined group composition and breeding pattern and success related to traits such as clutch and egg size, offspring sex ratio and the number of parents involved in a common breeding attempt. Apart from standardised observations and monitoring I took blood samples from chicks, which were later used for molecular sexing and for DNA fingerprints. Food plants were collected and determined and a rough habitat mapping was conducted. Since the impacts of boat tourism in the area became apparent I investigated the interactions of adult and young hoatzins with tourists and measured the plasma concentration of the hormone corticosterone in chicks as an indicator of stress. Each chapter has its own introduction to the specific topic and can be read independently. The main findings of this study are: The reproduction of the hoatzin was timed strictly following the bimodal rainy pattern in the area. There was only one breeding attempt per year. Only 18% of breeding attempts ended successfully with at least one fledgling. Incubation started with the first egg laid and led to hatching asynchrony. In most cases only the A-chick survived and there is evidence for a brood reduction strategy. I observed egg size variation patterns both within the clutches and between the clutches. Approximately 80% of breeding attempts were carried out with auxiliaries. Units with alloparentals had a higher breeding success than single pairs. The results indicate a trade-off between helping and group size. DNA band-sharing comparisons revealed the existence of joint-nests, where several females laid their eggs in one single nest. The clutches of these joint-nests suffered severe egg loss during all stages of incubation. Breeding success did not differ between single- and joint-nests. The primary offspring sex ratio was biased towards daughters. There was no differential mortality between the sexes until fledging. Individual breeding units employed an adaptive production of offspring of each sex according to their current group size. Rainforest tourism negatively influenced the survival and growth of young, not yet fledged hoatzins. In addition tourist-exposed young showed a stronger hormonal stress response than their conspecifics from undisturbed sites. In contrast, breeding adults appear to have habituated to tourist boats and exposure to observers.
Schutzfaktoren bei Kindern vor dem Übergang in die Sekundarstufe – Ergebnisse aus der STRESSStudie

Der Übergang in die Sekundarstufe stellt für Kinder ein normativ kritisches Lebensereignis dar, das unterschiedlich bewertet und bewältigt wird. Der vorliegende Beitrag befasst sich mit der Frage, welche Schutzfaktoren dazu beitragen, dass Kinder dem Übergang mit Gefühlen der Vorfreude begegnen und sie dieses Ereignis problemorientiert bewältigen. Untersucht werden 596 ViertklässlerInnen aus Bayern und Hessen vor dem Übergang in die Sekundarstufe. In Anlehnung an die kognitiv-transaktionale Theorie von Lazarus und Folkman (1984) werden die Zusammenhänge von primärer Bewertung als Herausforderung und problemorientierter Stressbewältigung unter Berücksichtigung von personalen (akademisches Selbstkonzept, Selbstwirksamkeit) und sozialen Schutzfaktoren (Familienzusammenhalt, allgemeines förderndes Elternverhalten) analysiert. Mithilfe von Strukturgleichungsmodellen lassen sich Mediationseffekte der Schutzfaktoren nachweisen. Insbesondere das akademische Selbstkonzept und ein förderliches Familienumfeld erweisen sich als bedeutsame protektive Faktoren in der Phase vor dem Übergang.
Chronic stress, even stress of a moderate intensity related to daily life, is widely acknowledged to be a predisposing or precipitating factor in neuropsychiatric diseases. There is a clear relationship between disturbances induced by stressful stimuli, especially long-lasting stimuli, and cognitive deficits in rodent models of affective disorders. Regular physical activity has a positive effect on the central nervous system (CNS) functions, contributes to an improvement in mood and of cognitive abilities (including memory and learning), and is correlated with an increase in the expression of the neurotrophic factors and markers of synaptic plasticity as well as a reduction in the inflammatory factors. Studies published so far show that the energy challenge caused by physical exercise can affect the CNS by improving cellular bioenergetics, stimulating the processes responsible for the removal of damaged organelles and molecules, and attenuating inflammation processes. Regular physical activity brings another important benefit: increased stress robustness. The evidence from animal studies is that a sedentary lifestyle is associated with stress vulnerability, whereas a physically active lifestyle is associated with stress resilience. Here, we have performed a comprehensive PubMed Search Strategy for accomplishing an exhaustive literature review. In this review, we discuss the findings from experimental studies on the molecular and neurobiological mechanisms underlying the impact of exercise on brain resilience. A thorough understanding of the mechanisms underlying the neuroprotective potential of preconditioning exercise and of the role of exercise in stress resilience, among other things, may open further options for prevention and therapy in the treatment of CNS diseases.
Recent human and animal studies suggest that epigenetic mechanisms mediate the impact of environment on development of mental disorders. Therefore, we hypothesized that polymorphisms in epigenetic-regulatory genes impact stress-induced emotional changes. A multi-step, multi-sample gene-environment interaction analysis was conducted to test whether 31 single nucleotide polymorphisms (SNPs) in epigenetic-regulatory genes, i.e. three DNA methyltransferase genes DNMT1, DNMT3A, DNMT3B, and methylenetetrahydrofolate reductase (MTHFR), moderate emotional responses to stressful and pleasant stimuli in daily life as measured by Experience Sampling Methodology (ESM). In the first step, main and interactive effects were tested in a sample of 112 healthy individuals. Significant associations in this discovery sample were then investigated in a population-based sample of 434 individuals for replication. SNPs showing significant effects in both the discovery and replication samples were subsequently tested in three other samples of: (i) 85 unaffected siblings of patients with psychosis, (ii) 110 patients with psychotic disorders, and iii) 126 patients with a history of major depressive disorder. Multilevel linear regression analyses showed no significant association between SNPs and negative affect or positive affect. No SNPs moderated the effect of pleasant stimuli on positive affect. Three SNPs of DNMT3A (rs11683424, rs1465764, rs1465825) and 1 SNP of MTHFR (rs1801131) moderated the effect of stressful events on negative affect. Only rs11683424 of DNMT3A showed consistent directions of effect in the majority of the 5 samples. These data provide the first evidence that emotional responses to daily life stressors may be moderated by genetic variation in the genes involved in the epigenetic machinery.
Das Ziel der vorliegenden Arbeit war die Untersuchung der Reaktionen von Migränepatientinnen mit episodischer (EM) und häufiger Migräne (HM) auf verschiedene Aspekte des Triggerfaktors „Negativer Affekt“ wie Stress und negative Emotionen. Die Ergebnisse der beiden Gruppen wurden mit denen gesunder Kontrollpersonen verglichen (KG). Zur Ermittlung des Aufmerksamkeitsverhaltens gegenüber emotionalen Reizen wurden zwei Emotionale Stroop Tests (EST) durchgeführt. Erwartet wurde ein Aufmerksamkeitsbias der Patientinnen hinsichtlich negativer emotionaler Reize. Im EST 1 wurden allgemeine affektive Wörter der Valenzen positiv, neutral und negativ verwendet. Die Probandinnen sollten auf die Wortfarbe mit Tastendruck reagieren und den Wortinhalt ignorieren. Im EST 2 wurden emotionale Gesichtsausdrücke (ärgerlich, freundlich, neutral) als Reize verwendet. Dabei sollte die Rahmenfarbe der Bilder per Tastendruck bestimmt werden und der Inhalt ignoriert werden. Zur Auswertung wurden Emotionale Stroop Interferenzen (ESI) zum Vergleich Reaktionszeitdifferenzen negativ-neutral und negativ-positiv berechnet. Der erwartete Aufmerksamkeitsbias der HM für negative emotionale Reize wurde dabei nicht gefunden. Dafür zeigten im EST 2 die KG einen Aufmerksamkeitsbias für ärgerliche Gesichter. Ein signifikanter Gruppenunterschied in EST 2 mit sehr niedrigen, im Vergleich negativ-positiv sogar negativen ESI der HM ließ auf ein Vermeidungsverhalten dieser Gruppe ärgerlichen Gesichtern gegenüber schließen. Dieses wurde als Vermeidung negativer sozialer Reize interpretiert und zum gelernten, möglicherweise dysfunktionalen Vermeidungsverhalten von Migränepatienten potentiellen Triggersituationen gegenüber in Bezug gesetzt. Weiterhin wurden die Probandinnen mit dem „Paradigma der Öffentlichen Rede“ psychosozialem Stress ausgesetzt, indem sie vor einer Videokamera unter Beobachtung eine Rede halten sowie eine Kopfrechenaufgabe lösen sollten. Vorher und nachher wurden insgesamt vier Speichelproben zur Bestimmung des Stresshormons Kortisol genommen. Zudem wurden die Druckschmerzschwellen vor und nach dem Experimentalteil gemessen. Die erwartete Kortisolreaktion als Antwort auf die psychosoziale Stressaufgabe blieb aus. Ursache dafür kann die Stichprobenzusammensetzung mit 98% Frauen sein, deren Kortisolreaktion auf Stress durch hormonelle Schwankungen im Experiment nur unzuverlässig stimulierbar ist. Bei der Berechnung der Gesamtkortisolausschüttung über die Zeit zeigte sich im Gegensatz zu dem erwarteten erhöhten Kortisolspiegel der Migränepatientinnen ein linearer Abfall des Spiegels von KG, über EM zu HM, mit den niedrigsten Werten der HM. Diese Ergebnisse könnten auf Veränderungen der Hypophysen-Nebennieren (HHN)-Achse im Sinne eines Hypokortisolismus bei Migränepatientinnen widerspiegeln, der weiterer Klärung bedarf, z.B. durch die Bestimmung eines Kortisoltagesprofils bei Patientinnen. Eine veränderte Funktion der HHN-Achse könnte außerdem zu einer inadäquaten Reaktion auf Stresssituationen beitragen. Die bei Patientinnen ausbleibende Veränderung der Druckschmerzschwelle in Reaktion auf Stress lässt ebenfalls auf eine ungenügende Stressreaktion der Patientinnen schließen. Am Ende der Untersuchung, nach einer Entspannungsphase von 50 Minuten, wurde den Probandinnen Blut abgenommen, in dem die mRNA- und Proteinkonzentrationen ausgewählter pro- und antiinflammatorischer Zytokine bestimmt wurden. Die Analyse der Zytokinkonzentrationen mit Luminex ergab für die Proteindaten aufgrund zu geringer verwertbarer Daten kein interpretierbares Bild. Die mittels Real Time Quantitativer PCR erhaltenen mRNA-Konzentrationen spiegelten die Schmerzfreiheit der Patienten wieder, mit im Vergleich zu KG verringerten proinflammatorischen Zytokinen (TNF-alpha, IL-1beta, IL-2, IL-6) und dem ebenfalls verringerten antiinflammatorischen Zytokin IL-10, sowie dem deutlich erhöhten antiinflammatorischen IL-4. Die im Vergleich zur KG überregulierten Zytokine im schmerzfreien Intervall weisen auf veränderte Regulierungsmechanismen des Immunsystems für die Schmerzmediatoren Zytokine hin. Weitere Schmerzmediatoren könnten ebenfalls verändert sein, was weiterer Klärung in nachfolgenden Studien bedarf. Alles in allem konnten verschiedene Veränderungen in den psychologischen und endokrinen Reaktionen der Migränepatientinnen auf Bestandteile des Triggers „Negativer Affekt“ sowie in der Schmerzregulierung gefunden werden, wobei die Veränderungen bei Patientinnen mit Häufiger Migräne stärker auftraten. Dies weist auf eine mögliche Rolle der einzelnen untersuchten Komponenten bei der Migränechronifizierung hin, was in weiteren Studien vertiefend untersucht werden sollte.
Background
Chronic stress is detrimental to health, and children and young people have had to cope with significantly more stress since the start of the COVID-19 pandemic. In particular, stress at school and in relation to learning is a major problem in this age group. Studies in Germany have indicated that the pandemic has led to a reduced quality of life (QoL) and an increased risk for psychiatric disorders in children and adolescents. Schools are an ideal setting for interventions against stress, which is one of the strongest predictors for the development of psychosocial problems. The present study seeks to address stress by means of a short prevention training programme in schools, including emotion regulation, mindfulness, and self-compassion. In addition to information material for self-study, students should have the opportunity to actively deal with the topic of stress and develop coping strategies within a short space of time. In contrast to very long stress reduction programmes that often last several weeks, the programme is delivered in just 90 min.
Methods
The effectiveness of the short and economical prevention programme LessStress will be examined in a cluster-randomised controlled trial (RCT) encompassing 1894 students. At several measurement time points, students from two groups (intervention and control) will be asked about their subjectively perceived stress levels, among other aspects. Due to the clustered nature of the data, mainly multilevel analyses will be performed.
Discussion
In Germany, there are no nationwide universal prevention programmes for students against stress in schools, and this gap has become more evident since the outbreak of the pandemic. Universal stress prevention in schools may be a starting point to promote resilience. By dealing with stress in a healthy way, mental health can be strengthened and maintained. Moreover, to reach at-risk students at an early stage, we advocate for a stronger networking between child psychiatry and schools.
Tinnitus is a phantom sound perception in the ears or head and can arise from many different medical disorders. Currently, there is no standard treatment for tinnitus that reliably reduces tinnitus. Individual patients reported that acupressure at various points around the ear can help to reduce tinnitus, which was investigated here. With this longitudinal observational study, we report a systematic evaluation of auricular acupressure on 39 tinnitus sufferers, combined with a self-help smartphone app. The participants were asked to report on tinnitus, stress, mood, neck, and jaw muscle tensions twice a day using an ecological momentary assessment study design for six weeks. On average, 123.6 questionnaires per person were provided and used for statistical analysis. The treatment responses of the participants were heterogeneous. On average, we observed significant negative trends for tinnitus loudness (Cohen's d effect size: −0.861), tinnitus distress (d = −0.478), stress (d = −0.675), and tensions in the neck muscles (d = −0.356). Comparison with a matched control group revealed significant improvements for tinnitus loudness (p = 0.027) and self-reported stress level (p = 0.003). The positive results of the observational study motivate further research including a randomized clinical trial and long-term assessment of the clinical improvement.
The presented work shows the analysis of the correlation between the spatial and temporal expression pattern of NtAQP1 and its function in water relation in planta. In situ immunological studies indicated NtAQP1-protein accumulation in the root exodermis and endodermis, in the cortex, close to vascular bundles, in the xylem parenchyma and in cells of the stomatal cavities. The aquaporin was also found to be abundant in longitudinal cell-rows in the petioles. Expression studies with generated transgenic plants (Ntaqp1-promoter::gus or luc) confirmed the Ntaqp1 accumulation in the root, stem and petioles but also revealed further localization in pollen grains, adventitious roots and leaf glandular hairs. Ntaqp1-expression was induced during growth processes, like stem bending after gravistimulation or photostimulation, seed germination and hypocotyl elongation as well as during the comparatively fast circadian leaf movement. The expression was further stimulated by phytohormones, especially gibberellic acid (GA) and osmotic stress. Further analysis displayed a diurnal and even circadian expression of Ntaqp1 in roots and petioles. The functional analysis of the aquaporin was accomplished by reverse genetics and biophysical studies. The antisense technique was used to reduce NtAQP1-expression in tobacco plants. The antisense (AS) plants exhibited a severe reduction of Ntaqp1-mRNA, less reduction of the highly homologous NtPIP1a RNA and no effect on expression of other aquaporin family genes (PIP2, TIP). The function of NtAQP1 at the cellular level was investigated by a newly developed experimental setup to record the osmotically induced increase in protoplast volume. The reduction of NtAQP1 by the antisense expression decreased the overall cellular waterpermeability Pos for more than 50 %. Function of NtAQP1 at the whole plant level was e.g. measured by the “high-pressure flow meter method”. Those measurements revealed that the root hydraulic conductivity per unit root surface area (KRA) of roots from the AS-lines was reduced by more than 50 %. KRA displayed a strong diurnal and circadian variation with a maximum in the middle of the light period, similar to the expression pattern of Ntaqp1 in roots. Gas exchange-, stem (Ystem) and leaf (Yleaf) water potential measurement gave dissimilar values in AS and control plants under well-watered conditions. Under a water-limiting environment the Y of AS-plants remained at more negative water values, even though a further decrease in transpiration of AS-plants was detected. Quantitative analysis displayed a much stronger wilting reaction in the AS than in the control plants. Quantitative studies of the leaf movement in AS compared to control plants exhibited a dramatic reduction in velocity and also in the extent of the process. The following conclusions can be drawn. NtAQP1 was expressed at sites of anticipated high water fluxes from and to the apoplast or symplast. Additionally, the specific distribution pattern and temporal expression of NtAQP1 in petioles and the bending stem strongly indicate a role in transcellular movement of water. The reduction of NtAQP1 by the antisense expression decreased the overall cellular Pos. Conclusively, NtAQP1-function increases membrane water permeability of tobacco root protoplasts. The decrease of the specific root hydraulic conductivity (KRA) was in the same order of magnitude as the mean cellular water permeability reduction, indicating that aquaporin expression is essential in maintaining a natural root hydraulic conductance. Reduction of KRA in AS plants might be the first definitive proof that the pathway of water uptake from the root surface to the xylem involves passage across membranes. The absence of NtAQP1 resulted in a water stress signal, causing a certain stomatal closure. NtAQP1 seems to contribute to water stress avoidance in tobacco. NtAQP1 plays an essential role in fast plant movements and transcellular water shift.
Based on recent findings that show that depletion of factor XII (FXII) leads to better posttraumatic neurological recovery, we studied the effect of FXII-deficiency on post-traumatic cognitive and behavioral outcomes in female and male mice. In agreement with our previous findings, neurological deficits on day 7 after weight-drop traumatic brain injury (TBI) were significantly reduced in FXII\(^{−/−}\) mice compared to wild type (WT) mice. Also, glycoprotein Ib (GPIb)-positive platelet aggregates were more frequent in brain microvasculature of WT than FXII\(^{−/−}\) mice 3 months after TBI. Six weeks after TBI, memory for novel object was significantly reduced in both female and male WT but not in FXII\(^{−/−}\) mice compared to sham-operated mice. In the setting of automated home-cage monitoring of socially housed mice in IntelliCages, female WT mice but not FXII\(^{−/−}\) mice showed decreased exploration and reacted negatively to reward extinction one month after TBI. Since neuroendocrine stress after TBI might contribute to trauma-induced cognitive dysfunction and negative emotional contrast reactions, we measured peripheral corticosterone levels and the ration of heart, lung, and spleen weight to bodyweight. Three months after TBI, plasma corticosterone levels were significantly suppressed in both female and male WT but not in FXII\(^{−/−}\) mice, while the relative heart weight increased in males but not in females of both phenotypes when compared to sham-operated mice. Our results indicate that FXII deficiency is associated with efficient post-traumatic behavioral and neuroendocrine recovery.
Stress impacts negatively on plant growth and crop productivity, causing extensive losses to agricultural production worldwide. Throughout their life, plants are often confronted with multiple types of stress that affect overall cellular energy status and activate energy-saving responses. The resulting low energy syndrome (LES) includes transcriptional, translational, and metabolic reprogramming and is essential for stress adaptation. The conserved kinases sucrose-non-fermenting-1-related protein kinase-1 (SnRK1) and target of rapamycin (TOR) play central roles in the regulation of LES in response to stress conditions, affecting cellular processes and leading to growth arrest and metabolic reprogramming. We review the current understanding of how TOR and SnRK1 are involved in regulating the response of plants to low energy conditions. The central role in the regulation of cellular processes, the reprogramming of metabolism, and the phenotypic consequences of these two kinases will be discussed in light of current knowledge and potential future developments.
Multiple lines of evidence implicate brain serotonin (5-hydroxytryptamine; 5-HT) system dysfunction in the pathophysiology of stressor-related and anxiety disorders. Here we investigate the influence of constitutively deficient 5-HT synthesis on stressor-related anxiety-like behaviors using Tryptophan hydroxylase 2 (Tph2) mutant mice. Functional assessment of c-Fos after associated foot shock, electrophysiological recordings of GABAergic synaptic transmission, differential expression of the Slc6a4 gene in serotonergic neurons were combined with locomotor and anxiety-like measurements in different contextual settings. Our findings indicate that constitutive Tph2 inactivation and consequential lack of 5-HT synthesis in Tph2 null mutant mice (Tph2\(^{-/-}\)) results in increased freezing to associated foot shock and a differential c-Fos activity pattern in the basolateral complex of the amygdala. This is accompanied by altered GABAergic transmission as observed by recordings of inhibitory postsynaptic currents on principal neurons in the basolateral nucleus, which may explain increased fear associated with hyperlocomotion and escape-like responses in aversive inescapable contexts. In contrast, lifelong 5-HT deficiency as observed in Tph2 heterozygous mice (Tph\(^{+/-}\)) is able to be compensated through reduced GABAergic transmission in the basolateral nucleus of the amygdala based on Slc6a4 mRNA upregulation in subdivisions of dorsal raphe neurons. This results in increased activity of the basolateral nucleus of the amygdala due to associated foot shock. In conclusion, our results reflect characteristic syndromal dimensions of panic disorder and agoraphobia. Thus, constitutive lack of 5-HT synthesis influence the risk for anxiety- and stressor-related disorders including panic disorder and comorbid agoraphobia through the absence of GABAergic-dependent compensatory mechanisms in the basolateral nucleus of the amygdala.
Mobile health interventions (i.e., “apps”) are used to address mental health and are an increasingly popular method available to both individuals and organizations to manage workplace stress. However, at present, there is a lack of research on the effectiveness of mobile health interventions in counteracting or improving stress-related health problems, particularly in naturalistic, non-clinical settings. This project aimed at validating a mobile health intervention (which is theoretically grounded in the Job Demands-Resources Model) in preventing and managing stress at work. Within the mobile health intervention, employees make an evidence-based, personalized, psycho-educational journey to build further resources, and thus, reduce stress. A large-scale longitudinal randomized control trial, conducted with six European companies over 6 weeks using four measurement points, examined indicators of mental health via measures of stress, wellbeing, resilience, and sleep. The data were analyzed by means of hierarchical multilevel models for repeated measures, including both self-report measures and user behavior metrics from the app. The results (n = 532) suggest that using the mobile health intervention (vs. waitlist control group) significantly improved stress and wellbeing over time. Higher engagement in the intervention increased the beneficial effects. Additionally, use of the sleep tracking function led to an improvement in sleeping troubles. The intervention had no effects on measures of physical health or social community at work. Theoretical and practical implications of these findings are discussed, focusing on benefits and challenges of using technological solutions for organizations to support individuals’ mental health in the workplace.
