In der vorliegenden Arbeit werden die positiven Effekte des Phosphodiesterase-5-Inhibitors Sildenafil auf die Gefäßfunktion in einem experimentellen Diabetes-Modell (Streptozotocin-induziert) beschrieben. Diabetes und seine kardiovaskulären Folgeerkrankungen, welche bis zu 80 % der Todesfälle bei diabetischen Patienten verursachen, gehen mit einer Endotheldysfunktion einher, bei der eine verminderte NO-Bioverfügbarkeit sowie eine verstärkte Sauerstoffradikalbildung eine Schlüsselrolle spielen. Endotheliales NO aktiviert die lösliche Guanylatzyklase in glatten Gefäßmuskelzellen, welche über erhöhte zyklische Guanosinmonophosphat-Konzentrationen eine Gefäßrelaxation bewirkt. Sildenafil reduziert als potenter und selektiver PDE 5-Inhibitor den Abbau von cGMP. Bei diabetischen und nicht-diabetischen Ratten führte Sildenafil in-vitro zu einer direkten dosisabhängigen Gefäßrelaxation an vorkontrahierten Aortenringen und zu einer Verbesserung der endothelabhängigen und –unabhängigen Gefäßrelaxation. Auch nach akuter und chronischer Behandlung zeigte sich eine verbesserte endothelabhängige und -unabhängige Gefäßrelaxation. Ferner zeigte sich nach chronischer Behandlung mit Sildenafil eine signifikante Reduktion des bei diabetischen Tieren erhöhten oxidativen Stresses. Die signifikant höhere Expression der NAD(P)H-Oxidase Untereinheit gp91phox und der Rac-Untereinheit Rac-1 in den Aortenringen diabetischer Ratten - welche für erhöhten oxidativen Stress bei Diabetes spricht -, sowie die zum anderen bei diabetischen Ratten verstärkte Superoxid(O2-)-Produktion konnten durch eine chronische Behandlung mit Sildenafil signifikant reduziert werden. Desweiteren konnte Sildenafil das im diabetischen Tiermodell verminderte eNOS-Dimer/Monomer-Verhältnis zurück auf ein Niveau ähnlich dem von Kontrolltieren erhöhen. Eine chronische Behandlung mit Sildenafil könnte demnach die vaskuläre Hämostase im Hinblick auf die Sauerstoffradikalbildung und die Gefäßfunktion verbessern. Jedoch bleibt der klinische Einsatz aufgrund der kurzen Wirkdauer der Substanz limitiert; aktuell wird Sildenafil in der Therapie der erektilen Dysfunktion beim Diabetiker und der pulmonalen arteriellen Hypertonie verwendet.
Mit einer Prävalenz von 15% der Gesamtbevölkerung und sogar über 30% bei den Frauen über dem 50. Lebensjahr zählt die Osteoporose zu den Erkrankungen welcher bei steigender Lebenserwartung zunehmend sozioökonomisches Gewicht zukommt. Die Folgekosten bei Komplikationen wie Frakturen nach Stürzen übersteigen die einer Therapie um einiges. Ein Motor der Entstehung von Osteoporose ist oxidativer Stress. Dieser regt über Enzymkaskaden die Bildung und Aktivierung von Osteoklasten an, was einen erhöhten Knochenabbau zur Folge hat. Zu den körpereigenen Verteidigungstrategien gegen Oxidation gehören Selenoproteine wie Glutathionperoxidasen. In einer vorangegangenen Arbeit von Prof.Jakob und C. Becker wurde bereits ein Zusammenhang der GPx-Aktivität mit der Ausbildung von Osteoporose nachgewiesen. Osteoporotiker hatten eine signifikant verminderte Aktivität dieses Enzyms. Ein essentielles Spurenelement für die Herstellung der Gluthationperoxidasen ist Selen. Deutschland gilt diesbezüglich als Mangelversorgungsgebiet. Es ist bekannt, dass ein Mangel an Selen Krankheiten begünstigen kann. Als Beispiele seien hier die Keshan-Krankheit oder Kashin-Beck- Krankheit genannt. Auch in der Intensivmedizin wurde ein Nutzen der Selengabe bei Sepsispatienten nachgewiesen. Die Selenspiegel aller eingeschlossenen 163 Probanden wurden untersucht. Es zeigte sich durchgehend eine leichte Erniedrigung. Signifikante Unterschiede ergaben sich nicht. Ein mutmaßlicher Mangel an Selen war als Ursache für eine verminderte Gluthationperoxidaseaktivität nicht nachzuweisen. Da der Hauptproduktionsort der GPx die Niere ist, wurde als Arbeitsthese die Vermutung aufgestellt, dass eine Niereninsuffizienz über eine verminderte Produktion dieser antioxidativ wirkenden Enzyme als Ursache einer Osteoporose in Frage kommt. Berechnet wurde die Niereninsuffizienz in Form der GFR nach der Cockroft-Gault- Formel. Es ergab sich eine signifikante Reduzierung der GFR zwischen Kontrolle und Osteoporosegruppe (p<0,05) nicht aber zwischen Kontrolle und Osteopeniegruppe und Osteopenie- und Osteoporosegruppe. Bezüglich des Alters und GFR konnte eine signifikante Korrelation nachgewiesen werde. Zwischen der GPx-Aktivität und der glomerulären Filtrationsrate bestand eine signifikante Korrelation ( Korrelation nach Pearson 0.149 mit einer Signifikanz von 0.018; Spearman-Rho 0.162 mit Signifikanz 0.010 beides p< 0,05). Es bestätigt sich also die in der Literatur bekannte positive Assoziation der gestörten Nierenfunktion mit der Manifestation der Osteoporose. Mit der GPx wird in dieser ersten Arbeit ein Sekretionsprodukt der Niere identifiziert, welches über eine Verminderung der Sekretion in der gestörten Nierenfunktion die antioxidative Kapazität des Organismus einschränkt und damit die Entstehung einer Osteoporose begünstigen könnte.
Cysteines play important roles in the biochemistry of many proteins. The high reactivity, redox properties, and ability of the free thiol group to coordinate metal ions designate cysteines as the amino acids of choice to form key catalytic components of many enzymes. Also, cysteines readily react with reactive oxygen and nitrogen species to form reversible oxidative thiol modifications. Over the last few years, an increasing number of proteins have been identified that use redox-mediated thiol modifications to modulate their function, activity, or localization. These redox-regulated proteins are central players in numerous important cellular processes. First aim of this study was to discover nitric oxide (NO) sensitive proteins in E. coli, whose redox-mediated functional changes might explain the physiological alterations observed in E. coli cells suffering from NO-stress. To identify E. coli proteins that undergo reversible thiol modifications upon NO-treatment in vivo, I applied a differential thiol trapping technique combined with two-dimensional gel analysis. 10 proteins were found to contain thiol groups sensitive to NO-treatment. Subsequent genetic studies revealed that the oxidative modifications of AceF & IlvC are, in part, responsible for the observed NO-induced growth inhibition. Noteworthy, the majority of identified protein targets turned out to be specifically sensitive towards reactive nitrogen species. This oxidant specificity was tested on one NO-sensitive protein, the small subunit of glutamate synthase. In vivo and in vitro activity studies demonstrated that glutamate synthase rapidly inactivates upon nitric oxide treatment but is resistant towards other oxidative stressors. These results imply that reactive oxygen and nitrogen species affect distinct physiological processes in bacteria. The second aim of my study was to identify redox-sensitive proteins in S. cerevisiae and to use their redox state as in vivo read-out to assess the role of oxidative stress during the eukaryotic aging process. I first determined the precise in vivo thiol status of almost 300 yeast proteins located in the cytosol and sub-cellular compartments of yeast cells using a highly quantitative mass spectrometry based thiol trapping technique, called OxICAT. The identified proteins can be clustered in four groups: 1) proteins, whose cysteine residues are oxidation resistant; 2) proteins with structurally or functionally important cysteine modifications 3) proteins with highly oxidation-sensitive active site cysteines, which are partially oxidized in exponentially growing yeast cells due to their exquisite sensitivity towards low amounts of ROS; 4) proteins that are reduced in exponentially growing cells but harbor redox-sensitive cysteine(s) that affect the catalytic function of the protein during oxidative stress. These oxidative stress sensitive proteins were identified by exposure of yeast cells to sublethal concentrations of H2O2 or superoxide. It was shown that the major targets of peroxide- and superoxide-mediated stress in the cell are proteins involved in translation, glycolysis, TCA cycle and amino acid biosynthesis. These targets indicate that cells rapidly redirect the metabolic flux and energy towards the pentose phosphate pathway in an attempt to ensure the production of the reducing equivalent NADPH to counterattack oxidative stress. These results reveal that the quantitative assessment of a protein’s oxidation state is a valuable tool to identify catalytically active and redox-sensitive cysteine residues. The OxICAT technology was then used to precisely determine extent and onset of oxidative stress in chronologically aging S. cerevisiae cells by utilizing the redox status of proteins as physiological read-out. I found that chronological aging yeast cells undergo a global collapse of the cellular redox homeostasis, which precedes cell death. The onset of this collapse appears to correlate with the yeast life span, as caloric restriction increases the life span and delays the redox collapse. These results suggest that maintenance of the redox balance might contribute to the life expanding benefits of regulating the caloric intake of yeast. Clustering analysis of all oxidatively modified proteins in chronological aging yeast revealed a subset of proteins whose oxidative thiol modifications significantly precede the general redox collapse. Oxidation of these early target proteins, which most likely results in a loss of their activity, might contribute to or even cause the observed loss of redox homeostasis (i.e., thioredoxin reductase) in chronologically aging yeast. These studies in aging yeast expand our understanding how changes in redox homeostasis affect the life span of yeast cells and confirm the importance of oxidative thiol modifications as key posttranslational modifications in pro- and eukaryotic organisms.
Several epidemiological studies found that hypertensive patients have an increased risk to develop kidney cancer. Hyperaldosteronism frequently results in arterial hypertension and contributes to the development and progression of kidney injury, with reactive oxygen species (ROS) playing an important role. ROS are thought to be associated with many pathological conditions such as cancer and other disorders, like cardiovascular complications , which often go along with hypertension. The aim of the present work was to investigate whether the effects of elevated aldosterone concentrations might be involved in the increased cancer incidence of hypertensive individuals. First, the potential capacity of aldosterone to induce oxidative stress and DNA damage was investigated in vitro and in vivo. In LLC-PK1 porcine kidney cells and MDCK canine kidney cells the significant formation of ROS, and especially of superoxide (O2˙ˉ) was assessed. With two genotoxicity tests, the comet assay and the micronucleus frequency test, the DNA damaging potential of aldosterone was quantified. In both genotoxicity tests a dose-dependent increase in aldosterone-induced structural DNA damage was observed. Oxidative stress and DNA damage were prevented by antioxidants, suggesting ROS as a major cause of DNA damage. Furthermore, the oxidatively modified DNA lesion 8-oxo-7,8-dihydro-2´-deoxyguanosine (8-oxodG), was found to be significantly elevated. In kidneys of rats with desoxycorticosterone acetate (DOCA)/salt-induced hypertension, which is a model of severe mineralocorticoid-dependent hypertension, elevated levels of ROS and superoxide were found, compared to kidneys of sham rats. Also DNA strand breaks, measured with the comet assay and double strand breaks, visualized with antibodies against the double strand break-marker gamma-H2AX were significantly elevated in kidneys of DOCA/salt-treated rats. In addition, significantly increased amounts of 8-oxodG were detected. Proliferation of kidney cells was found to be increased, which theoretically enables the DNA damage to manifest itself as mutations, since the cells divide. Second, the effects of aldosterone on the activation of transcription factors and signaling pathways were investigated. A significant activation of the potentially protective transcription factor Nrf2 was observed in LLC-PK1 cells. This activation was triggered by an increase of ROS or reactive nitrogen species (RNS). In response to oxidative stress, glutathione synthesis and detoxifying enzymes, such as the subunits of the glutathione-cysteine-ligase or heme oxygenase 1 were rapidly induced after 4 h. Nevertheless, after 24 h a decrease of glutathione levels was observed. Since ROS levels were still high after 24 h, but Nrf2 activation decreased, this adaptive survival response seems to be transient and quickly saturated and overwhelmed by ROS/RNS. Furthermore, Nrf2 activation was not sufficient to protect cells against oxidative DNA damage, because the amounts of double strand breaks and 8-oxodG lesions steadily rose up to 48 h of aldosterone treatment. The second transcription factor that was time- and dose-dependently activated by aldosterone in LLC-PK1 and MDCK cells was NF-kappaB. Furthermore, a significant cytosolic and nuclear activation of ERK was detected. Aldosterone induced the phosphorylation of the transcription factors CREB, STAT1 and STAT3 through ERK. Third, the underlying mechanisms of oxidant production, DNA damage and activation of transcription factors and signaling pathways were studied. Aldosterone exclusively acted via the MR, which was proven by the MR antagonists eplerenone, spironolactone and BR-4628, whereas the glucocorticoid receptor (GR) antagonist mifepristone did not show any effect. Furthermore, aldosterone needed cytosolic calcium to exert its negative effects. Calcium from intracellular stores and the influx of calcium across the plasma membrane was involved in aldosterone signaling. The calcium signal activated on the one hand, the prooxidant enzyme complex NAD(P)H oxidase through PKC, which subsequently caused the generation of O2˙ˉ. On the other hand, nitric oxide synthase (NOS) was activated, which in turn produced NO. NO and O2˙ˉ can react to the highly reactive species ONOO- that can damage the DNA more severely than the less reactive O2˙ˉ. In the short term, the activation of transcription factors and signaling pathways could be a protective response against aldosterone-induced oxidative stress and DNA damage. However, a long-term NF-B and ERK/CREB/STAT activation by persistently high aldosterone levels could unfold the prosurvival activity of NF-kappaB and ERK/CREB/STAT in aldosterone-exposed cells. DNA damage caused by increased ROS might become persistent and could be inherited to daughter cells, probably initiating carcinogenesis. If these events also occur in patients with hyperaldosteronism, these results suggest that aldosterone could be involved in the increased cancer incidence of hypertensive individuals.
Rauchen stellt in den Industrienationen das bedeutendste vermeidbare Gesundheitsrisiko dar. Die Rolle des suchtauslösenden Alkaloids Nikotin in der Tabak assoziierten Kanzerogenese wird kontrovers diskutiert. Ziel dieser Arbeit war die Untersuchung genotoxischer Effekte von Nikotin in Zellen des oberen und unteren Aerodigestivtrakt sowie deren intrazellulärer Mechanismen. Dazu wurden Zellen aus humaner Nasenschleimhaut und humaner Bronchialschleimhaut enzymatisch isoliert sowie bronchiales Zelllinienepithel kultiviert und mit Nikotin unterschiedlicher Dosierungen für eine Stunde inkubiert. Zur Untersuchung beteiligter Signalkaskaden wurden Koinkubationen von Nikotin und dem nicht-kompetitiven nikotinergen Acetylcholinrezeptorblocker Mecamylamin und dem Antioxidans N-Acetylcystein durchgeführt. Die Erfassung Nikotin induzierter DNASchäden erfolgte mit Hilfe des Comet Assays. Zur Untersuchungen von Zellzyklusalterationen sowie Apoptoseinhibition durch Nikotin kam die Durchflusszytometrie zum Einsatz. Die Ergebnisse der Einzelzellgelelektrophorese zeigten eine dosisabhängige DNASchädigung im einstündigen Inkubationsversuch durch Nikotin. Diese Schäden waren gewebeabhängig ab einer Konzentration von 100μM in Zelllinienepithel (n=5) und 1mM in Nasenschleimhautzellen (n=8) signifikant. In humanem Bronchialzellepithel konnte bei dem Stichprobenumfang von n=4 keine signifikante DNA-Schädigung durch die getesteten Nikotindosierungen nachgewiesen werden. Durch eine Koinkubation mit dem Antioxidans N-Acetylcystein sowie dem nicht kompetitiven nACh Rezeptorblocker Mecamylamin konnte eine im Comet Assay nachweisbare Nikotin induzierte DNA-Schädigung verhindert werden. Durchflusszytometrische Untersuchungen zur Klärung einer möglichen Modulation der Apoptose durch Nikotin an bronchialem Zelllinienepithel zeigten keine signifikante Induktion oder Inhibition. Eine Beeinflussung des Zellzyklus durch Nikotin konnte in der Durchflusszytometrie nicht erfasst werden. Zusammenfassend induziert Nikotin DNA-Schäden in Epithelien des Atemtraktes. An diesem Effekt sind oxidative sowie nAch-Rezeptor abhängige Stoffwechselschritte beteiligt. Vor dem Hintergrund einer potentiellen Beteiligung von Nikotin an der Tumorinitiation und -progression muss eine Nikotinersatztherapie besonders kritisch abgewogen werden.
