In the first part of his work, the causes for the sudden degradation of useable capacity of lithium-ion cells have been studied by means of complementary methods such as computed tomography, Post-Mortem studies and electrochemical analyses. The results obtained point unanimously to heterogeneous aging as a key-factor for the sudden degradation of cell capacity, which in turn is triggered by differences in local compression.
At high states of health, the capacity fade rate is moderate but some areas of the graphite electrode degrade faster than others. Still, the localized changes are hardly noticeable on cell level due to averaging effects. Lithium plating occurs first in unevenly compressed areas, creating patterns visible to the human eye. As lithium plating leads to rapid consumption of active lithium, a sudden drop in capacity is observed on cell level. Lithium plating appears to spread out from the initial areas over the whole graphite electrode, quickly consuming the remaining useful lithium and active graphite. It can be hypothesized that a self-amplifying circle of reciprocal acceleration of local lithium loss and material loss causes rapid local degradation.
Battery cell designers can improve cycle life by homogeneous pressure distribution in the cell and using negative active materials that are resilient to elevated discharge potentials such as improved carbons or lithium titanate. Also, a sufficiently oversized negative electrode and suitable electrolyte additives can help to avoid lithium plating. When packs are designed, care must be taken not to exert local pressure on parts of cells and to avoid both very high and low states of charge.
In the second part of this dissertation the resilience of cylindrical and pouchbag cells to shocks and different vibrations was investigated. Stresses inflicted by vibration and shock tests according to the widely recognized UN38.3 transport test were compared to a long-time test that exposed cells to a 186 days long ordeal of sine sweep vibrations with a profile based on real-world applications. All cells passed visual and electric inspection performed by TU München after the vibration tests. Only cylindrical cells subjected to long-term vibrations in axial direction showed an increase in impedance and a loss of capacity that could be recuperated in part.
The detailed analyses presented in this thesis gave more details on the damages inflicted by vibrations and shocks and revealed drastic damages in some cases. In cylindrical cells, only movement in axial direction caused damage. Long term vibrations were found to be especially detrimental.
No damage whatsoever could be detected for pouch cells, regardless of the test protocol and the direction of movement. The extreme resilience of pouchbag cells shows that the electrode stack of lithium-ion cells is resistant to vibrations, and that damages are caused by design imperfections that can be improved at low cost.
The findings of this work, and the general state of research show that it is most crucial to control the lithiation and thus potential of the graphite electrode.
In the last part of this work, a new, direct method for charge estimation based on changing transmission is presented. A correlation between transmission of short ultrasonic pulses and state of charge is found. This new technology allows direct measurement of the state of charge. The method is demonstrated for batteries with different positive active materials, showing its versatility. As the observed changes can be traced to the lithiation of graphite, it can be determined without a reference electrode. Already at this early stage of development, the found correlations allow estimation of state of charge. The present hysteresis in the signal height of the slow wave, which is unneglectable especially during discharging at higher currents, will be subject to further investigation.
The observed effects can be explained by effects on different length scales. Biot’s theory explains the second wave’s slowness based on the active material particles size in the range of 0.01 mm and electrolyte-filled pores. Lithiation of graphite changes the porosity of the electrode and thereby the velocity and wavelength of the impulse. When the wavelength approaches the length scale of the layers, 0.1 mm, scattering effects dampen the transmitted signal. Finally, the wavelength of the pulse should be shorter than the transducers diameter to obtain a homogeneous wave front.
To conclude, the new method allows the control of each individual cell in a pack independent from the electrical connections of the cells.
As the method shows great promise, further studies regarding factors such as long-term behavior, temperature and current rates should be conducted. In this thesis hysteresis was observed and a deeper understanding of the reasons behind it may allow further improvements of measurement precision.
Die vorliegende Arbeit beschreibt die Entwicklung von SiO2-basierten nano- und mikroskalig strukturierten Schichten, welche eine Temperaturstabilität > 500 °C sowie eine Strukturtreue > 90 % aufzeigen. Diese wurden unter Verwendung von hybridpolymeren, partikelmodifizierten Sol-Gel-Vorstufen via UV-basierter Nanoimprintlithographie (UV-NIL) hergestellt. Der Einfluss chemischer und verfahrenstechnischer Syntheseparameter auf die anorganische Netzwerkbildung der flüssigen Vorstufe (hybridpolymerer Binder), die Polymerisation und Verdichtung der hybridpolymeren Sol-Gel-Schichten während der thermischen Härtung sowie deren Strukturierung via UV-NIL wurden im Detail untersucht und die erhaltenen Zusammenhänge mit den resultierenden Materialeigenschaften korreliert. Dabei dienten die Kenntnisse hinsichtlich des Schrumpf- und Verdichtungsverhaltens von planaren Schichten als Basis für die daraus hergestellten, strukturierten Schichten.
In der vorliegenden Arbeit wird der Einfluss von Metallkomplexverbindungen auf der Basis von monotopen und ditopen Terpyridin-Liganden auf Zellen behandelt. Es können mehrere Möglichkeiten aufgezeigt werden, wie MEPE als kontrollierte Freisetzungssysteme für Zel-lanwendungen eingesetzt werden können. Es werden 2D-Beschichtungen, 3D-Knochenzemente und Terpyridin funktionalisierte Alginate hergestellt. Es ist möglich, definier-te, homogene Fe-MEPE Schichten auf Borosilikatglas mithilfe der Layer by Layer Technik und mittels Tauschbeschichtung abzuscheiden. Um die Oberfläche und somit die Freisetzung von Metallionen zu erhöhen, werden zusätzlich poröse SiO2-Schichten hergestellt, welche mit Fe-MEPE infiltriert werden. Um die Anwendbarkeit von Metallkomplexverbindungen auf der Basis von monotopen und ditopen Terpyridin-Liganden als Knochenersatzmaterial zu testen werden Hydroxylapatit Knochenzemente synthetisiert. Ziel ist eine retardierende Freisetzung der Metallionen ohne Burst Effekt und ohne den Verlust der Druckstabilitäten der HA Zemen-te. Die Funktionalisierung von Alginat mit 1-Amino-5-(2,2ʹ:6ʹ,2ʹʹ-terpyrid-4ʹ-yl-oxy)pentan resultiert in Hydrogelen, welche ein anderes Gelierverhalten als das unfunktionalisierte Alginat zeigen. Zudem ist es möglich mit Fe(II)- /Ca(II)-Salzmischungen Hydrogele auszubilden. Die funktionalisierten Alginate sind zudem bioaktiv.
Zum grundlegenden Verständnis der MEPE Zell Wechselwirkung werden zunächst Zytotoxo-zitätsuntersuchungen mittels WST-1 Test von L929 und C2C12-Zellen mit wässrigen M(II)MEPE Lösungen (Metallionen M= Fe(II), Co(II), Ni(II), Zn(II)) in einem Konzentrationsbe-reich von 1,56x10-11 bis 1,6x10-5 mol L-1 durchgeführt. Fe-MEPE zeigt im betrachteten Kon-zentrationsbereich keine zytotoxischen Eigenschaften auf die eingesetzte Fibroblastenzelllinie. Bei Konzentrationen über 1x10-6 mol L-1 Fe-MEPE sinkt die Mitochondrienaktivität der C2C12-Zellen auf 40%. Dagegen wirken Co- und Zn-MEPE ab einer Konzentration von 1x10-7 mol L-1 stark zytotoxisch auf L929 und C2C12-Zellen.
Um selektiv die Differenzierung von C2C12, MG63, humanen mesenchymalen Stammzellen (hMSCs) und humanen Endothelzellen anzuregen, werden die Zellen auf den hergestellten 2D Beschichtungen ausgesät. Es kann gezeigt werden, dass Fe-MEPE die Proliferation zu-gunsten der Stoffwechselaktivität von C2C12, MG63-Zellen und hMSCs hemmt. Bei weiterer Betrachtung der spezifischen myogenen Differenzierungsmarker der C2C12-Zellen bzw. der spezifischen Gene der osteogenen Differenzierung (Osteocalcin und ALP) mithilfe qRT-PCR können erhebliche Stimulierungen auf der mRNA Basis detektiert werden. Auch auf
enzymatischer Ebene zeigen Fe-MEPE modifizierte Oberflächen einen stimulierenden Effekt auf die Aktivität der alkalischen Phosphatase der MG63 Zelllinie und humaner mesenchyma-ler Stammzellen.
Somit kann eine Stimulierung der myogenen Differenzierung von C2C12-Zellen, sowie oste-ogenen Differenzierung von MG63-Zellen und hMSCs mittels Fe-MEPE beschichteten Ober-flächen innerhalb von drei Tagen nachgewiesen werden. Die Ergebnisse zeigen, dass Fe-MEPE funktionalisierte Oberflächen als innovative Scaffolds für die Behandlung von Kno-chendefekten eingesetzt werden können.
Background: Culturing of cells is typically performed on standard tissue culture plates generating growth conditions, which in general do not reflect the native three-dimensional cellular environment. Recent investigations provide insights in parameters, which strongly affect the general cellular behavior triggering essential processes such as cell differentiation. The physical properties of the used material, such as stiffness, roughness, or topology, as well as the chemical composition of the cell-surface interface are shown to play a key role in the initiation of particular cellular responses. Methods: We extended our previous research, which identified thin films of metallo-supramolecular coordination polyelectrolytes (MEPEs) as substrate to trigger the differentiation of muscular precursor cells. Results: Here, we show that the same MEPEs similarly stimulate the osteogenic differentiation of pre-osteoblasts. Remarkably, MEPE modified surfaces also trigger the differentiation of primary bone derived mesenchymal stem cells (BMSCs) towards the osteogenic lineage. Conclusion: This result leads to the conclusion that these surfaces individually support the specification of cell differentiation toward lineages that correspond to the natural commitment of the particular cell types. We, therefore, propose that Fe-MEPEs may be used as scaffold for the treatment of defects at least in muscular or bone tissue.
Aim of this thesis was the development of functionalizable hydrogel coatings for melt electrowritten PCL scaffolds and of bioprintable hydrogels for biofabrication.
Hydrogel coatings of melt electrowritten scaffolds enabled to control the surface hydrophilicity, thereby allowing cell-material interaction studies of biofunctionalized scaffolds in minimal protein adhesive environments. For this purpose, a hydrophilic star- shaped crosslinkable polymer was used and the coating conditions were optimized. Moreover, newly developed photosensitive scaffolds facilitated a time and pH independent biofunctionalization.
Bioprintable hydrogels for biofabrication were based on the allyl-functionalization of gelatin (GelAGE) and modified hyaluronic acid-products, to enable hydrogel crosslinking by means of the thiol-ene click chemistry. Optimization of GelAGE hydrogel properties was achieved through an in-depth analysis of the synthesis parameters, varying Ene:SH ratios, different crosslinking molecules and photoinitiators. Homogeneity of thiol-ene crosslinked networks was compared to free radical polymerized hydrogels and the applicability of GelAGE as bioink for extrusion-based bioprinting was investigated. Purely hyaluronic acid-based bioinks were hypothesized to maintain mechanical- and rheological properties, cell viabilities and the processability, upon further decreasing the overall hydrogel polymer and thiol content.
Hydrogel coatings: Highly structured PCL scaffolds were fabricated with MEW and subjected to coatings with six-armed star-shaped crosslinkable polymers (sP(EO-stat-PO)). Crosslinking results from the aqueous induced hydrolysis of reactive isocyanate groups (NCO) of sP(EO-stat-PO) and increased the surface hydrophilicity and provided a platform for biofunctionalizations in minimal protein adhesive environments. Not only the coating procedure was optimized with respect to sP(EO-stat-PO) concentrations and coating durations, instead scaffold pre-treatments were developed, which were fundamental to enhance the final hydrophilicity to completely avoid unspecific protein adsorption on sP(EO-stat-PO) coated scaffolds. The sP(EO-stat-PO) layer thickness of around 100 nm generally allows in vitro studies not only in dependence on the scaffold biofunctionalization but also on the scaffold architecture. The hydrogel coating extent was assessed via an indirect quantification of the NCO-hydrolysis products. Knowledge of NCO-hydrolysis kinetics enabled to achieve a balance of sufficiently coated scaffolds while maintaining the presence of NCO-groups that were exploited for subsequent biofunctionalizations. However, this time and pH dependent biofunctionalization was restricted to small biomolecules. In order to overcome this limitation and to couple high molecular weight biomolecules another reaction route was developed. This route was based on the photolysis of diazirine moieties and enabled a time and pH independent scaffold biofunctionalization with streptavidin and collagen type I. The fibril formation ability of collagen was used to obtain different collagen conformations on the scaffolds and a preliminary in vitro study demonstrated the applicability to investigate cell-material interactions.
The herein developed scaffolds could be applied to gain deeper insights into the fundamentals of cellular sensing. Especially the complexity by which cells sense e.g. collagen remain to be further elucidated. Therefore, different hierarchies of collagen-like conformations could be coupled to the scaffolds, e.g. gelatin or collagen-derived peptide sequences, and the activation of DDR receptors in dependence on the complexity of the coupled substances could be determined. Due to the strong streptavidin-biotin bond, streptavidin functionalized scaffolds could be applied as a versatile platform to allow immobilization of any biotinylated molecules.
Gelatin-based bioinks: First the GelAGE products were synthesized with respect to molecular weight distributions and amino acid composition integrity. A detailed study was conducted with varying molar ratios of reactants and synthesis durations and implied that gelatin degradation was most dominant for high alkaline synthesis conditions with long reaction times. Gelatin possesses multiple functionalizable groups and the predominant functionalization of amine groups was confirmed via different model substances and analyses. Polymer network homogeneity was proven for the GelAGE system compared to free radical polymerized hydrogels with GelMA. A detailed analysis of hydrogel compositions with varying functional group ratios and UV- or Vis-light photoinitiators was executed. The UV-initiator concentration is restricted due to cytotoxicity and potential cellular DNA damages upon UV-irradiation, whereas the more cytocompatible Vis- initiator system enabled mechanical stiffness tuning over a wide range by controlling the photoinitiator concentration at constant Ene:SH ratios and polymer weight percentages. Versatility of the GelAGE bioink for different AM techniques was proved by exploiting the thermo-gelling behavior of differently degraded GelAGE products for stereolithography and extrusion-based printing. Moreover, the viability of cell-laden GelAGE constructs was demonstrated for extrusion-based bioprinting. By applying different multifunctional thiol-macromolecular crosslinkers the mechanical and rheological properties improved concurrently to the processability. Importantly, lower thiol-crosslinker concentrations were required to yield superior mechanical strengths and physico-chemical properties of the hydrogels as compared to the small bis-thiol-crosslinker. Extrusion-based bioprinting with distinct encapsulated cells underlined the need for individual optimization of cell-laden hydrogel formulations.
Not only the viability of encapsulated cells in extrusion-based bioprinted constructs should be assessed, instead other parameters such as cell morphology or production of collagen or glycosaminoglycans should be considered as these represent some of the crucial prerequisites for cartilage Tissue Engineering applications. Moreover, these studies should be expanded to the stereolithographic approach and ultimately the versatility and cytocompatibility of formulations with macromolecular crosslinkers would be of interest. Macromolecular crosslinkers allowed reducing polymer weight percentages and amounts of thiol groups and are thus expected to contribute to increased cytocompatibility, especially in combination with the more cytocompatible Vis-initiator system, which remains to be elucidated.
Hyaluronic acid-based bioinks: Different molecular weight hyaluronic acid (HA) products were synthesized to bear ene- (HAPA) or thiol-functionalities (LHASH) to enable pure HA thiol-ene crosslinked hydrogels. Depending on the molecular weight of modified HA products, polymer weight percentages and Ene:SH ratios, a wide range of mechanical stiffness was covered. However, the manageability of high molecular weight HA (HHAPA) product solutions (HHAPA + LHASH) was restricted to 5.0 wt.-% as a consequence of the high viscosity. Based on the same HA thiol component (LHASH), hybrid hydrogels of HA with GelAGE were compared to pure HA hydrogels. Although the overall polymer weight percentage of HHAPA + LHASH hydrogels was significantly lowered compared to hybrid hydrogels (GelAGE + LHASH), similar mechanical and physico-chemical properties of pure HA hydrogels were determined with maintained Ene:SH ratios. Low viscous low molecular weight HA precursor solutions (LHAPA + LHASH) prevented the applicability for extrusion-based bioprinting, whereas the non-thermoresponsive HHAPA + LHASH system could be bioprinted with only one-fourth of the polymer content of hybrid formulations. The high viscous behavior of HHAPA + LHASH solutions, lower polymer weight percentages, decreased printing pressures and consequently declined shear stress during printing, were hypothesized to contribute to high cell viabilities in extrusion-based bioprinted constructs compared to the hybrid bioink.
The low molecular weight HA precursor formulation (LHAPA + LHASH) was not applicable for extrusion-based printing, but this system has potential for other AM techniques such as stereolithography. Similar to the GelAGE system a more detailed study on the functions of encapsulated cells would be useful to further develop this system. Moreover, the initiation with the Vis-initiator should be conducted.
Die Arbeit beschäftigte sich mit Untersuchungen zu photovernetzbaren und –strukturierbaren (Hybrid-)Polymeren, um Grundlagen für die Herstellung von Trägergerüststrukturen (Scaffolds) auf Basis photovernetzbarer (Hybrid-)Polymere zu legen und damit in der Zukunft patientenindividuelle medizinische Werkstücke, die beliebig durch Zwei-Photonen-Absorptionsprozesse in drei Dimensionen strukturierbar sind, für die Regenerative Medizin zu ermöglichen.
Dafür wurden zunächst die zum Teil in der Literatur unbekannten unterschiedlichen Monomere Acr-1, MAcr-2, Acr-3, MAcr-4 und DiMAcr-5 synthetisiert. Dabei handelt es sich um einfache und gut vergleichbare organische (Meth-)Acrylat-Monomere, die mono- bzw. difunktional in ihren photochemisch reaktiven Gruppen sind. Die synthetisierten organischen Monomere Acr-3, MAcr-4 und DiMAcr-5 wurden in verschiedenen Verhältnissen mit dem anorganisch-organischen Methacrylat-basierten Hybridpolymers ORMOCER® I kombiniert.
Die (Co-)Polymerisation der unterschiedlichen Formulierungen wurde in situ mittels UV-DSC-Untersuchung verfolgt. Dabei wurden bei diesen Untersuchungen zum Teil deutliche Unterschiede im Reaktionsverlauf der einzelnen Materialformulierungen festgestellt. So konnten zum Beispiel bei Monomermischungen ein schnellerer Polymerisationsverlauf sowie eine höhere maximale Polymerisationsrate als bei den jeweiligen Einzelkomponenten beobachtet werden (Synergieeffekt). Diese Beobachtungen wurden anhand der Monomerstruktur (unterschiedliche Diffusionsfähigkeiten im vergelten, aber noch nicht erstarrten System durch Mono- bzw. Difunktionalität) und der Art der funktionellen Gruppe (Acrylat- bzw. Methacrylatgruppe) erklärt.
Weiterhin wurden der Einfluss des verwendeten Photoinitiators und dessen eingesetzte Konzentration auf die photochemisch-induzierte Copolymerisation eines ausgewählten Systems beleuchtet. Dazu wurden verschiedene Einflussfaktoren der Initiation betrachtet. Neben der eingesetzten Initiatorkonzentration spielen auch die Absorptionseigenschaften, die umgebende Matrix und die Initiatoreffizienz eine große Rolle für den Reaktionsverlauf der photochemischen Vernetzung. Weiterhin wurden die Photoinitiatoren in unterschiedlichen Konzentrationen eingesetzt, um die dadurch induzierte Veränderung des Reaktionsverlaufs zu betrachten. Aus den Einflüssen auf die Reaktionsverläufe konnte geschlossen werden, dass diese sowie auch die maximale Polymerisationsrate RP,max und damit die Reaktionskinetik nicht in jedem Fall linear mit der Initiatorkonzentration zunehmen muss.
Erste generelle 2PP-Strukturierungen wurden zudem an ausgewählten Material-formulierungen durchgeführt. Dabei zeigte sich, dass alle Formulierungen bei bestimmten Parameterkombinationen aus Laserleistung und Schreibgeschwindigkeit mittels 2PP strukturiert werden konnten. Außerdem wurden bei den verschiedenen Formulierungen bei gleicher Parameterkombination unterschiedliche Strukturbreiten und damit erstmalig unterschiedliche Strukturvolumina beobachtet. Diese unterschiedlichen Volumina konnten erstmalig mit den unterschiedlichen Reaktionsverläufen der Materialformulierungen korreliert werden. Dabei zeigte sich, dass das chemische Wechselwirkungsvolumen von der Funktionalität der eingesetzten Materialkomponenten abhängig ist, da davon der Grad an Quervernetzung abhängt, der bestimmt, ob ausreichend vernetzte Voxel und Strukturen entstehen, die durch einen Entwicklungsschritt nicht mehr entfernt werden.
Im zweiten Teil der Arbeit wurde ein biokompatibles und photostrukturierbares Hybridpolymer (RENACER® MB-I) entwickelt, welches mittels 2PP strukturiert werden konnte, was anhand kleiner wie auch großer Scaffolds mit dem Material demonstriert wurde. Dazu wurde das kommerziell erhältliche Alkoxysilan-Molekül
O-(Methacryloxyethyl)-N-(triethoxysilylpropyl)urethan als Precursor verwendet. Durch eine bewusst unvollständige Hydrolyse- und Kondensationsreaktion konnte aus dem Precursorsilan ein Hybridpolymerharz hergestellt werden, welches anorganisch vorverknüpft war. Weiterhin wies es sowohl als Volumenpolymer, als auch in Form von Scaffold-Strukturen eine sehr gute Biokompatibilität auf. Um zu untersuchen, ob die im Hybridpolymer enthaltenen prinzipiell degradierbaren Gruppen unter physiologischen Bedingungen tatsächlich degradieren und Teile aus dem Polymerverband herausgelöst werden können, wurde ein selbstentwickeltes Verfahren für stationäre Degradations-untersuchungen in phosphat-gepufferter Saline (PBS, pH = 7,4) verwendet. Die durch die photochemische Polymerisation neu entstandenen Ketten besaßen ihrer Natur gemäß keine hydrolysierbaren Einheiten, weshalb das Hybridpolymer nicht vollständig degradieren kann. Es konnte jedoch ein prinzipieller Zugang zu Gerüstträgerstrukturen auf Basis photovernetzbarer Polymere für die Regenerative Medizin geschaffen werden.
Im Rahmen der vorliegenden Arbeit wurde ein neuer Beschichtungstyp für die Elektrodenmaterialien von Lithium-Ionen-Akkumulatoren entwickelt und charakterisiert. Dieser besteht aus einem speziellen anorganisch-organischen Hybridpolymer, das sich bezüglich seiner Zusammensetzung und Funktion gegenüber bestehenden Beschichtungsmaterialien abhebt. Das anorganisch-organische Netzwerk des Hybridpolymers konnte mittels Feststoff-NMR-Messungen vollständig aufgeklärt werden. Dabei zeigte sich ein stabiles anorganisches Gerüst aus hoch vernetzten Polysiloxan-Einheiten. Zusätzliche organische Modifizierungen liegen als lange bewegliche Ketten mit funktionellen Polyethylenoxid-Einheiten vor oder sind in Form von Polyethern und Diolen vernetzt. Mit dieser speziellen Netzwerkstruktur ist es möglich, Materialeigenschaften zu erzeugen, die über solche von rein anorganischen und rein organischen Beschichtungen hinausgehen. Zu den mit verschiedenen Methoden nachgewiesenen Eigenschaften zählen eine hohe ionische Leitfähigkeit von 10\(^{-4}\) S/cm, eine hohe Elastizität mit E = 63 kPa, eine hohe elektrochemische Stabilität bis 5,0 V vs. Li/Li\(^+\) und eine hohe thermische Stabilität.
Eine weitere Besonderheit des neuen Beschichtungsmaterials ist die mehrstufige Vernetzung der anfänglichen Prekursoren zu einem Hybridpolymer-Sol und dem abschließenden Hybridpolymer-Gel. Die im Beschichtungssol vorliegende Teilvernetzung der Vorstufen konnte detailliert mittels Flüssig-NMR-Messungen untersucht und beschrieben werden. Aus den Messungen ließ sich folgern, dass die organisch und anorganisch vernetzbaren Gruppen im Sol teilweise vernetzt vorliegen. Die sterisch erreichbaren Si-OR-Gruppen der so entstandenen Oligomere sind vorwiegend nicht hydrolysiert, wodurch deren anorganische Anbindung an die OH-Gruppen der Partikeloberflächen kinetisch bevorzugt ist. Damit lassen sich besonders homogene und vollständig bedeckende Beschichtungen der Elektrodenmaterialien erzeugen. Dies konnte mit verschiedenen physikalischen und chemischen Methoden nachgewiesen werden: simulationsgestützte Rückstreuanalysen mittels REM, hochaufgelöste TEM-Aufnahmen sowie Elementanalysen durch EDX und XPS.
Nach der Optimierung des nasschemischen Beschichtungsprozesses über Rotationsverdampfen ergaben sich für die verschiedenen Elektrodenmaterialien Li\(_4\)Ti\(_5\)O\(_{12}\), Li(Ni,Co,Mn)O\(_2\) und Li(Mn,Ni)\(_2\)O\(_4\) jeweils etwa 20 nm dicke Beschichtungen mit Hybridpolymer. Die Frage nach deren Lösungsmittelbeständigkeit konnte durch die Analyse von behandelten Proben mit TG, REM, XPS und ICP-OES aufgeklärt werden. Dabei zeigte sich sowohl für die Behandlung mit NMP, dem klassischen Lösungsmittel bei der Elektrodenfertigung mit PVDF-Binder, als auch für die Behandlung mit dessen umweltschonenderem Ersatzstoff Aceton eine gute Beständigkeit der Beschichtung. Die Beschichtung löste sich in den Lösungsmitteln an, blieb allerdings als geschlossene nanoskalige Beschichtung erhalten. Lediglich gegenüber dem Lösungsmittel H\(_2\)O, das in Kombination mit dem neuen Binder CMC eingesetzt wird, wurde eine mangelnde Schichtstabilität deutlich. Das dafür verantwortliche Quellverhalten der Beschichtung konnte mittels Dünnschicht-Modellsystem und daran durchgeführten REM-, IR- und EPA-Untersuchungen aufgeklärt werden. Die Optimierung des Hybridpolymer-Materials bezüglich einer besseren H\(_2\)O-Beständigkeit übersteigt den Rahmen dieser Arbeit und liefert die Grundlage für weitere künftige Forschungsarbeiten.
Aufgrund der vollständigen Bedeckung der neuen Beschichtung, ihrer besonderen Eigenschaften und ihrer Beständigkeit bei der klassischen Elektrodenfertigung ist es möglich, die Elektrodenmaterialien grundlegend hinsichtlich ihrer wichtigsten Eigenschaften zu verbessern. Hierfür wurden sowohl über die NMP- als auch über die Aceton-Route Elektroden gefertigt und zu Halbzellen und Vollzellen verarbeitet. Die REM-Analyse der Elektroden zeigte, dass die Partikelbeschichtungen keinen negativen Einfluss auf die Homogenität und Morphologie der Elektroden ausüben. Damit war es möglich, jeweils einen direkten Vergleich von beschichteten und unbeschichteten Materialien hinsichtlich ihrer elektrochemischen Performance anzustellen. Für die Kathodenmaterialien Li(Ni,Co,Mn)O\(_2\) und Li(Mn,Ni)\(_2\)O\(_4\) ergaben die Zyklenfestigkeits- und Impedanzmessungen klare Verbesserungen durch die Beschichtung. Verbunden mit einer Verbesserung der Energiedichte erhöhte sich bei beiden Materialien die Zyklenfestigkeit um mehr als 60 %. Bei Li(Mn,Ni)\(_2\)O\(_4\) zeigt sich die Verbesserung in einer erhöhten Zellspannung durch das vergleichsweise hohe Redoxpotential des Materials von etwa 4,7 V vs. Li/Li\(^+\), während sich bei Li(Ni,Co,Mn)O\(_2\) die Hochvoltfähigkeit des Materials verbessert, was mit einer vergrößerten Speicherkapazität verbunden ist. Dabei ist herauszustellen, dass für keines der Materialien ein negativer Einfluss der dünnen Beschichtung auf die Leistungsdichte festgestellt werden konnte.
Der erwartete Mechanismus für die verbesserte Elektrodenfunktion durch das Hybridpolymer ist die Bildung einer physikalischen Schutzschicht in Form einer Li\(^+\)-leitfähigen Membran. Diese umgibt das Elektrodenmaterial vollständig, ermöglicht die Ladungsträgerinterkalation und schützt die Elektrode gleichzeitig vor irreversiblen Reaktionen mit dem Elektrolyten. Damit verbunden ist eine verminderte Mn-Auslösung und eine verminderte Entwicklung von isolierenden Deckschichten aus Reaktionsprodukten wie LiF, Li\(_2\)O, Li\(_2\)CO\(_3\), was sich positiv auf die Alterung der Batteriezellen auswirkt. Die Funktion der Beschichtung wurde primär auf den Kathodenmaterialien demonstriert. Doch auch auf der Anodenseite wurde ihre Anwendungstauglichkeit aufgezeigt, was das große Potential der Beschichtung für eine breite Anwendung in Lithium-Ionen-Batterien verdeutlicht.
The main focus of this work was to get a deeper understanding of the relationship between the structure of sol-gel films, their densification and their macroscopic cracking. First of all titania was chosen as model system. Therefore a synthesis route starting from the preparation of long-term stable amorphous redissoluble precursor powders based on acetylacetone as chelate ligand was utilized. The solubility and stability of the powders in various solvents can be determined by chemical synthesis and technological parameters. When dissolved in a solvent mixture of ethanol and 1,5-pentanediol, thin films can be easily prepared by dip-coating technique. Thereby the quality of the titania films enormously depends on the calcinations temperature and the solvent mixture is used. In order to investigate the influence of different solvents and solvent mixtures on the microstructure and densification of the precursors, the coating solutions were stripped off (sol powder) and analyzed as function of annealing temperature. It was pointed out that a high densification rate caused by the addition of 1,5-pentanediol, results in dense microstructure with trapped residual carbon. These impurities can retard the phase transformation of anatase to rutile. The analysis of so-called “film powders” scraped off multiple dip-coated substrates provides valuable information on the effect of air moisture and unidirectional densification during drying and aging on the structure of thin films. The high surface-to-volume ratio and access to air moisture determine the chemical composition of the as-prepared film, which controls shrinkage, crystallization and defect structure of the coatings. Further it was shown, that drying as a thin film results in the formation of closed pores and much denser microstructure than the respective sol powder. Without the addition of 1,5-pentanediol all –OEt moieties undergo hydrolysis reactions, which causes the formation of a rigid network. The presence of 1,5-pentanediol retards this hydrolysis reactions and provides some network plasticity. Generally the microstructure of thin films is comparatively close to the microstructure of the film powders. The addition of 1,5-pentandiol prevents hydrolysis and condensation reactions as like in the film powders. However even at 700 °C, thin films never transform to rutile, which was attributed to the tensile stresses in thin films. In thin films and in film powders as well a comparable amount of closed pores are formed during annealing. Further it was shown that most of the thin sol-gel films investigated form a dense crust on their tops during annealing. This explains why crack free films exhibit only closed pores. However, when cracks appear during thin film shrinkage in the coating, this crust is burst, which generates open porosity. The defect density in the coatings was determined by an automated analysis of surface images. The crack formation and quantity can be directly referred to tensile stresses in the coatings, which arise from hydrolysis and condensation during thin film drying and aging. Therefore when 1,5-pentanediol is added to the sol, thin film cracking was avoided, because hydrolysis and condensation reactions are retarded, which preserves a higher network flexibility. Furthermore the crack formation was significantly influenced by the atmospheric humidity that was used during the coating process, which was explained by different drying and condensation rates. Under certain chemical starting conditions water soluble precursor powders can be also obtained. In general the observations made with the water based coating solutions are mostly in agreement with the former results based on ethanol based coating solutions. For example the high surface-to-volume ratio of film powders compared to sol powders also significantly enhances film drying and densification. The addition of 1,5-pentanediol also clearly contributes to their densification behavior and phase evolution. As seen before in the case of ethanol based coatings, 1,5-pentanediol enhances the stability towards hydrolysis and condensation reactions and preserves some network plasticity. Therefore coatings prepared without the addition of 1,5-pentanediol already form cracks during film drying and aging because of tensile stresses. Thus, the addition of 1,5-pentanediol results in a reduction/prevention of crack formation. Nevertheless some differences were observed, i.e. the critical single coating film thickness of ethanol based coatings is nearly twice that of water based coatings. This was explained by the different surface tensions of the basis solvents, which during thin film drying causes significantly higher capillary forces and tensile stresses in water based coatings. When acetylacetone is replaced by triethanolamine as chelating ligand for titanium also re-dissolvable precursor powders can be synthesized. The film powders combine a high hydrolytic stability of the precursor with sufficient intermediate network flexibility. The different type of organics changes the drying and densification behavior: i.e. in contrast to film powders obtained from acetylacetone based precursor powders the structure of triethanolamine based film powders is unaffected by the thin film drying process. This high hydrolytic stability and plasticity of this precursor allows the preparation of defect free coatings up to single film thickness of 300 nm. However triethanolamine based thin films present at intermediary annealing temperatures a distinctively different microstructure compared to acetylacetone based films. The general validity of the conclusions was proved on the basis of zirconia coatings that were also prepared by the use of re-dissolvable precursor powders. In principle all conclusions concerning the interconnection of precursor chemistry, film formation, densification and structure were transferable to the respective zirconia coatings. Differences mainly arise only from differential material properties i.e. bulk density. Finally, it has been pointed out that the findings obtained on the densification behavior of thinsol-gel films are also a valuable tool for improved explanations of other important scientific questions concerning sol-gel films, i.e. scratch resistance of sol-gel coatings, fiber -bridging and – degradation of sol-gel coated fibers.
Here, a postpolymerization modification method for an α-terminal functionalized poly-(N-methyl-glycine), also known as polysarcosine, is introduced. 4-(Methylthio)phenyl piperidine-4-carboxylate as an initiator for the ring-opening polymerization of N-methyl-glycine-N-carboxyanhydride followed by oxidation of the thioester group to yield an α-terminal reactive 4-(methylsulfonyl)phenyl piperidine-4-carboxylate polymer is utilized. This represents an activated carboxylic acid terminus, allowing straightforward modification with nucleophiles under mild reaction conditions and provides the possibility to introduce a wide variety of nucleophiles as exemplified using small molecules, fluorescent dyes, and model proteins. The new initiator yielded polymers with well-defined molar mass, low dispersity, and high end-group fidelity, as observed by gel permeation chromatography, nuclear magnetic resonance spectroscopy, and matrix-assisted laser desorption/ionization time-of-flight mass spectroscopy. The introduced method can be of great interest for bioconjugation, but requires optimization, especially for protein conjugation.
Hydrogels that can mimic mechanical properties and functions of biological tissue have attracted great interest in tissue engineering and biofabrication. In these fields, new materials and approaches to prepare hydrogels without using toxic starting materials or materials that decompose into toxic compounds remain to be sought after. Here, we report the crosslinking of commercial, unfunctionalized hydrophilic poly(2-ethyl-2-oxazoline) using peroxide copolymers in their melt. The influence of temperature, peroxide copolymer concentration, and duration of the crosslinking process has been investigated. The method allows to create hydrogels from unfunctionalized polymers in their melt and to control the mechanical properties of the resulting materials. The design of hydrogels with a suitable mechanical performance is of crucial importance in many existing and potential applications of soft materials, including medical applications.
Die experimentelle Verbesserung der makroskopischen Eigenschaften (z. B. thermische oder mechanische Eigenschaften) von Keramiken ist aufgrund der zahlreichen erforderlichen Experimente zeitaufwändig und kostenintensiv. Simulationen hingegen können die Korrelation von Mikrostruktur und makroskopischen Eigenschaften nutzen, um die Eigenschaften von beliebigen Gefügekompositionen zu berechnen. In bisherigen Simulationen wurden meist stark vereinfachte Modelle herangezogen, welche die Mikrostruktur einer Keramik nur sehr grob widerspiegeln und deshalb keine zuverlässigen Ergebnisse liefern.
In der vorliegenden Arbeit wird die Mikrostruktur-Eigenschafts-Korrelation der drei wichtigsten Nicht-Oxid-Keramiken untersucht. Dies sind Aluminiumnitrid (AlN), Siliciumnitrid (Si3N4) und Siliciumcarbid (SiC). Diese drei Keramiktypen vertreten die häufigsten Mikrostrukturtypen, welche bei Nicht-Oxid-Keramiken auftreten können. Zu jedem Keramiktyp liegen zwei verschiedene Proben vor.
Alle drei untersuchten Keramiktypen sind zweiphasig. Die Hauptphase von AlN und Si3N4 besteht aus keramischen Körnern, die Nebenphase erstarrt während der Sinterung aus den zugesetzten Sinteradditiven. Die Restporosität von AlN und Si3N4 wird als vernachlässigbar angesehen und in den Simulationen nicht berücksichtigt. Bei den SiC-Proben handelt es sich um Keramiken mit bimodaler Korngröÿenverteilung. Durch Infiltration mit flüssigem Silicium wurden die Hohlräume zwischen den Körnern aufgefüllt, um porenfreie SiSiC-Proben zu erhalten.
Anhand von Simulationen werden zunächst reale Mikrostrukturen in Anlehnung an vorliegende Vergleichsproben nachgebildet. Diese Modelle werden durch Abgleich mit rasterelektronenmikroskopischen 2D-Aufnahmen der Proben verifiziert. An den Modellen werden mit der Methode der Finite-Element-Simulation makroskopische Eigenschaften (Wärmeleitfähigkeit, Elastizitätsmodul und Poisson-Zahl) der Keramiken simuliert und mit experimentellen Messungen an den vorliegenden Proben abgeglichen.
Der Vergleich der Mikrostruktur von den computergenerierten Gefügen und den vorliegenden Proben zeigt in der Mustererkennung durch das menschliche Auge und quantitativ in den Gefügeparametern eine gute Übereinstimmung. Für die makroskopischen Eigenschaften wird auf der Basis einer ausführlichen Literaturrecherche zu den Materialparametern der beteiligten Phasen eine gute Übereinstimmung zwischen den experimentell gemessenen und den simulierten Eigenschaften erreicht. Evtl. auftretende Abweichungen zwischen Experiment und Simulation können damit erklärt werden, dass die Proben Verunreinigungen enthalten, da aus der Literatur bekannt ist, dass Verunreinigungen eine Verschlechterung der Wärmeleitfähigkeit bewirken.
Nachdem die Gültigkeit der Modelle verifiziert ist, wird der Einfluss von charakteristischen
Mikrostrukturparametern und Phaseneigenschaften auf die Wärmeleitfähigkeit, den Elastizitätsmodul und die Poisson-Zahl der Keramiken untersucht. Hierzu werden die Mikrostrukturparameter von AlN und Si3N4 gezielt um die Parameter der vorliegenden Vergleichsproben variiert. Bei beiden Keramiktypen werden die Volumenanteile der beteiligten Phasen sowie die mittlere Sehnenlänge der keramischen Körner verändert. Bei den AlN-Keramiken wird zusätzlich der Dihedralwinkel variiert, welcher Auskunft über den Benetzungsgrad der Flüssigphase gibt; bei den Si3N4-Keramiken ist das Achsenverhältnis der langgezogenen Si3N4-Körner von Interesse und wird deshalb ebenfalls variiert. Es zeigt sich, dass die Aufteilung der Teilvolumina zwischen den zwei Phasen den größten Einfluss auf die Eigenschaften der Keramik hat, während die übrigen Mikrostrukturparameter nur eine untergeordnete Rolle spielen.
Um die Qualität der Simulationen zu überprüfen, wird die Simulationsreihe an AlN mit unterschiedlicher Aufteilung der Volumina zwischen den beiden Phasen in Relation zu etablierten Modellen aus der Literatur (Mischungsregel und Modell nach Ondracek) gesetzt. Alle Simulationsergebnisse für die Wärmeleitfähigkeit und den Elastizitätsmodul liegen innerhalb der jeweils oberen und unteren Grenze beider Modelle. Es konnte also eine Verbesserung gegenüber den etablierten Modellen erzielt werden.
An allen drei Keramiktypen wird der Einfluss der Materialeigenschaften der Haupt- und Nebenphase auf die makroskopischen Eigenschaften der Keramik untersucht. Hierfür werden die Wärmeleitfähigkeit, der Elastizitätsmodul und die Poisson-Zahl der Phasen getrennt voneinander über einen größeren Bereich variiert. Es stellt sich heraus, dass es vom Keramiktyp und dem Volumenanteil der Nebenphase abhängt, wie stark der Einfluss einer Komponenteneigenschaft auf die Eigenschaft der Keramik ist.
Mit den im Rahmen dieser Arbeit durchgeführten Simulationen wird der Einfluss von Mikrostrukturparametern und Phaseneigenschaften berechnet. Auf der Grundlage dieser Simulationen können die Architektur des Gefüges simuliert und die Eigenschaften von Keramiken für individuelle Anwendungen berechnet werden. Dies ist die Basis für die Produktion von maßgeschneiderten Keramiken. Zudem können mit den validierten Mikrostrukturmodellen die Eigenschaften von unbekannten Mischphasen ermittelt werden, was experimentell oft nicht möglich ist.
Die vorliegende Arbeit beschäftigt sich mit Kompositschichten aus Zeolithen und Hybridpolymeren,
die mittels des Sol-Gel-Prozesses aus Alkyltrialkoxysilanen hergestellt werden. Am Beispiel
von dealuminiertem Zeolith Y und Solen aus Bis(triethoxysilyl)ethan wurde untersucht, wie
sich die Zugänglichkeit der Zeolithporen in Kompositschichten erhalten lässt. Zur Analyse der
Porenzugänglichkeit kamen Gasadsorptionsmessungen zum Einsatz. Zur weiteren Charakterisierung
wurden elektronenmikroskopische Aufnahmen und ausführliche spektroskopische Untersuchungen
der erhaltenen Hybridpolymer-Sole durchgeführt. Die Ermittlung der mechanischen Eigenschaften
erfolgte über die Messung der Wischfestigkeit.
Die im Rahmen diverser Experimente erhaltenen Kompositschichten wiesen eine hohe Zeolithporenerreichbarkeit
auf, sofern der Zeolithanteil mindestens 70 Volumenprozent betrug, und das
jeweilige Sol einen hohen Hydrolyse- und Kondensationsgrad aufwies. Im Zusammenhang mit
den genannten Studien wurden Hybridpolymere verglichen, die bei unterschiedlichen pH-Bedingungen
mit verschiedenen Mengen an Wasser zur Hydrolysereaktion hergestellt wurden, oder bei
denen neben Bis(triethoxysilyl)ethan Methacryloxypropyltrimethoxysilan als zweites Monomer
eingesetzt wurde.
Letztendlich konnten mit einfachen Mitteln Kompositschichten hergestellt werden, die auf flexible
Oberflächen aufgebracht werden konnten und beim Biegen nicht vom Substrat abplatzten.
Ferner waren sie wischfest und zeigten bei passender Zusammensetzung eine nahezu vollständige
Zeolithporenerreichbarkeit (Zeolithanteil: ≥ 70 Vol.-%; Monomer: Bis(triethoxysilyl)ethan;
Hydrolyse- und Polykondensationsreaktion: pH-Wert ≤ 2, Überschuss an Wasser). Ihr Anwendungspotential
als Adsorbensschicht für die Aufnahme organischer Schadstoffe wurde beispielhaft
anhand der reversiblen Adsorption von Formaldehyd demonstriert.
In dieser Arbeit konnten ethanolische Sole aus TEOT und der metabolisierbaren α-Hydroxycarbonsäure Milchsäure (LA) in spinnfähige viskose Spinnmassen überführt werden und erstmalig über die Methode des Druckspinnens zu Mikrofasern prozessiert werden.
Die hybriden Fasern sind intrinsisch stabil. Über FTIR- und 13C-MAS-NMR-Untersuchungen konnte gezeigt werden, dass in der Faser der Koordinationsmodus von LA an Ti sowohl im mono- als auch im bidentaten Modus (Nomenklatur bezogen auf die Säureeinheit) vorliegt.
Die nähere Untersuchung des Degradationsverhaltens einer LA-Faser zeigte hauptsächlich die Freisetzung von Lactat und Ethanol innerhalb weniger Stunden. Danach kann kaum noch ein Massenverlust der Fasern nachgewiesen werden. Vermutlich ist die Degradationsgeschwindigkeit abhängig von der Sättigungskonzentration der wasserlöslichen Titanoxid-Spezies Ti(OH)4 und Ti(O)(OH)2. Die Löslichkeit dieser Verbindungen beträgt ca. 1 µmol/L. Die Freisetzung von Titanverbindungen an das Degradationsmedium konnte über ICP-Messungen und indirekt auch über NMR-Messungen der Degradationsprodukte in Lösung nachgewiesen werden. Nach ca. einer Woche in Lösung bildet sich der wasserlösliche metallorganische Komplex TiBALDH. Dieser Komplex zeigt keinen negativen Einfluss auf die Umwelt, so dass Zellkulturmedien, die in Kontakt mit den Fasermaterialien getreten sind, in Zukunft nach dem Autoklavieren gefahrlos entsorgt werden können.
Zudem sollte keines der detektierten Abbauprodukte in den abgegebenen Mengen toxisch auf den humanen Organismus bei in vivo-Anwendungen wirken. Lactat und Ethanol können im menschlichen Organismus verstoffwechselt werden. TIBALDH ist dem im menschlichen Serum nachweisbaren Titan(IV)citrat-Komplex strukturell sehr ähnlich. Aufgrund der Tatsache, dass die Bildung von TiBALDH ca. 1 Woche dauert, ist die vorherige Bildung des Titan(IV)citrat-Komplexes im humanen Organismus wahrscheinlich.
Weiterhin konnte das hybride Fasermaterial durch den Zusatz von basischen Stoffen neutralisiert werden und nach Vorkonditionierung der Fasern als nicht zytotoxisch eingestuft werden. Als Gegenionen wurde Ammonium, das biogene Amin Phenethylamin, die Aminosäure Phenylalanin und das Biopolymer CHI getestet. Für zukünftige Weiterentwicklungen können auch basische Wirkstoffe als Gegenionen herangezogen werden. Somit könnte das hybride Zellträgermaterial zusätzlich eine Drug-Delivery-Funktion erhalten.
Die LA-Fasern verhalten sich nach dem Verspinnen sehr flexibel. Bei einer Lagerung bei RT jedoch verspröden diese sehr schnell innerhalb von 3 d. Diese Materialeigenschaft wurde im zweiten Teil der Arbeit näher untersucht und optimiert.
Tempern des Fasermaterials bei 170 °C bewirkte eine Umlagerung der LA-Liganden zu AA-Liganden, aber keine Verbesserung der mechanischen Eigenschaften. Versuche einer getemperten LA-Faser mit CHI als Gegenion zeigte durchwegs positive Eigenschaften in den Zytotoxizitätstests und auf deren Oberfläche konnten Zellen der Zelllinien L929, 16HBE, HTB94 und MG63 erfolgreich kultiviert werden.
Durch die Verwendung anderer metabolisierbarer α Hydroxycarbonsäuren konnten Rückschlüsse auf die chemische Zusammensetzung der Fasern gezogen werden. Die Fasern scheinen aus wenig untereinander vernetzen Titan-oxo-carboxo-Clustern der Summenformel [Ti6O6(OR)6(Carboxylat)6] (mit R = H2+, H, Et oder „Ti6O6(OR)5(Carboxylat)6“) zu bestehen. Durch Variation der verwendeten Säuren konnten die Wechselwirkungen der Cluster untereinander verstärkt werden, so dass beispielsweise eine Faser mit MA bedeutend flexiblere Eigenschaften – auch bei einer Lagerung für 3d bei RT aufweist. Des Weiteren konnte durch Lagerung dieser Faser bei 4 °C der Versprödungsprozess für mind. 1 Monat gestoppt werden. Eine Lagerung von Medizinprodukten bei 4 °C stellt in Ländern mit ausreichender Infrastruktur kein Problem dar.
Aufbauend auf diesen Tatsachen und TGA-MS-Messungen konnte die These aufgestellt werden, dass sich zwischen den wenig untereinander vernetzten Titan-oxo-carboxo-Cluster direkt nach dem Verspinnen noch Wassermoleküle befinden. Diese Reste an Wasser verleihen – vermutlich aufgrund der Ausbildung von Wasserstoffbrückenbindungen – der Faser flexible Eigenschaften. Bei einer Lagerung bei RT entweichen diese Wasserreste und die Faser versprödet; bei einer Lagerung bei 4°C wird das Verdampfen des restlichen Wassers bedeutend verlangsamt.
Die Faser mit den flexibelsten Eigenschaften konnte letztendlich durch die Verwendung des zweizähnigen Carboxylat-Liganden MalA erhalten werden.
Zusammenfassend konnte in dieser Arbeit ein neuartiges faserförmiges Material auf Basis von Titan-oxo-carboxo-Clustern produziert werden, welches großes Potential besitzt als Zellträgermaterial Anwendung zu finden. Aufbauend auf den hier gewonnenen Ergebnissen können die mechanischen Eigenschaften weiter optimiert und die Anforderungen des gewünschten Zielgewebes feinjustiert werden. Zudem besteht die Möglichkeit dem Material Drug-Delivery-Eigenschaften zu verleihen. Somit könnte das Scaffold aus Mikrofasern neben den bereits integrierten chemischen und physikalischen Stimuli (die Oberflächenfunktionalitäten und die Oberflächentopographie der Fasern) auch durch freigesetzte Wirkstoffe Zellen zur gewünschten Differenzierung anregen.
The development of novel fibrous biomaterials and further processing of medical devices is still challenging. For instance, titanium(IV) oxide is a well-established biocompatible material, and the synthesis of TiO\(_x\) particles and coatings via the sol-gel process has frequently been published. However, synthesis protocols of sol-gel-derived TiO\(_x\) fibers are hardly known. In this publication, the authors present a synthesis and fabrication of purely sol-gel-derived TiO\(_x\) fiber fleeces starting from the liquid sol-gel precursor titanium ethylate (TEOT). Here, the α-hydroxy-carboxylic acid lactic acid (LA) was used as a chelating ligand to reduce the reactivity towards hydrolysis of TEOT enabling a spinnable sol. The resulting fibers were processed into a non-woven fleece, characterized with FTIR, \(^{13}\)C-MAS-NMR, XRD, and screened with regard to their stability in physiological solution. They revealed an unexpected dependency between the LA content and the dissolution behavior. Finally, in vitro cell culture experiments proved their potential suitability as an open-mesh structured scaffold material, even for challenging applications such as therapeutic medicinal products (ATMPs).
Im Rahmen der vorliegenden Arbeit wurde untersucht, wie sich organisch polymerisierbare Titan(IV)- und Zirkonium(IV)komplexe, die bei der Synthese anorganisch-organischer Hybrid-polymere verwendet werden, in Precursor-Solen für anorganische Beschichtungen verhalten. Dabei sollte ein Konzept zur Herstellung photochemisch strukturierter, anorganischer Dünnschichten erarbeitet werden. Als Beispiel für das anorganische Dünnschichtmaterial wurde Bleizirkonattitanat (PZT) und für die polymerisierbaren Liganden Methacrylsäure gewählt. Der Schwerpunkt der Arbeit lag dabei besonders auf den Untersuchungen der photochemischen Polymerisation mittels UV-lithographischer und Mehrphotonenpolymerisation sowie der in situ Untersuchung während der Pyrolyse des polymerisierten Materials und der Reaktion zur anorganischen Dünnschicht.
Der Prozess zur Herstellung photostrukturierter, anorganischer Dünnschichten kann in zwei Schritte eingeteilt werden. Nach der Synthese des Precursor-Sols erfolgten zunächst die licht-induzierte organische Polymerisation und die Entfernung der nicht polymerisierten Bereiche mit Hilfe eines Entwicklerbads. Im Anschluss daran wurden im zweiten Schritt die organischen Bestandteile durch eine thermische Behandlung entfernt und die Reaktion zum anorganischen Oxidmaterial induziert. Die homogensten Schichten wurden dabei mit einem n Butanol-basierten, methacrylat¬funktionalisierten PZT-Sol mit 25 Gew. % Feststoffgehalt auf Stahlsubstraten bei Pyrolyse¬bedingungen ab 500 bis 700 °C mit einer Heizrate von 5 K/min erreicht. An diesen Schichten konnte ferroelektrisches Verhalten nachgewiesen werden. Allerdings reichen die Eigenschaften noch nicht an in der Literatur beschrieben ferro- und piezoelektrische Dünnschichten heran, die ebenfalls aus PZT-Solen hergestellt wurden. Dies liegt zum einen an den zu geringen Schichtdicken und der damit verbundenen erhöhten Durchschlagsgefahr dieser Sol-Gel-basierten PZT-Schichten. Zum anderen reduzieren Grenzflächenreaktionen des chromhaltigen Substrats und der PZT-Solschicht zu Pb2(CrO4)O sehr drastisch die ferroelektrischen Eigenschaften, indem sich nicht-ferroelelektrische Oxidschichten mit niedrigen Permittivitäten bilden und die Beweglichkeit der Domänenwände durch Diffusion von Cr3+-Akzeptorionen in die PZT-Schicht verringert wird.
Zunächst wurde die PZT-Solsynthese untersucht. Dabei konnte gezeigt werden, dass die Nebenproduktbildung von verschiedenen Estern vermieden werden kann, wenn die leicht flüchtigen Bestandteile nach der Ligandenaustauschreaktion von Alkoxid gegen Methacrylsäure bei reduziertem Druck und damit verbundenen niedrigeren Reaktionstemperaturen, erfolgte. Die Bindung der Methacrylatliganden ist in jedem Fall bidentat. Bei den n Butanol-basierten PZT-Solen konnte trotz höherer Siedetemperatur und niedrigerem Dampfdruck ein größerer Anteil an Alkohol entfernt werden, als bei n-Propanol-basierten PZT-Solen. Dies wurde auf den etwas höheren +I Effekt des n Butanols und in der Folge der bevorzugten Ligandenaustauschreaktion von Methacryl¬säure gegen den n Butanol im Vergleich zu n Propanol zurückgeführt. Die Größe der PZT-Cluster war langzeitstabil, reproduzierbar herstellbar und betrug ca. 2,0 – 2,5 nm. Aus der Partikelgröße und dem spektros¬kopischen Nachweis von Metalloxobindungen konnte ein Strukturvorschlag basierend auf den zugrundeliegenden Titan- und Zirkoniumkomplexen erstellt werden. Dabei bilden MO6 Komplexe das Grundgerüst stäbchenförmiger, heterometallischer Cluster. Die polymerisierbaren Methacrylatliganden befinden sich auf der Oberfläche und sind gut zugänglich für die Polymerisationsreaktion. Die mittlere Anzahl an Methacrylatliganden pro PZT-Cluster wurde zu 5 - 25 je nach Gewichtung abgeschätzt.
Bei der UV-lithographischen Strukturierung ergaben sich zwei unerwartete Effekte. Zum einen trat nach der Schichtherstellung eine reversible Trübung des PZT-Sols auf, welches auf konzentrations¬bedingte Phasenseparation zurückgeführt werden konnte. Zum anderen setzte sich unter N2 Atmosphäre die lichtinduzierte radikalische Kettenreaktion in den methacrylat-funktionalisierten PZT-Solschichten auch in die unbelichteten Bereiche fort. Dieser Effekt wird sonst in dieser Deutlichkeit nur von kationischen Polymerisationsreaktionen beobachtet und auf lange Radikal¬lebensdauern zurückgeführt und wurde mittels Photo-Differentialkalorimetrie weitergehend untersucht. Allerdings konnte aufgrund der Probengeometrie der Einfluss der Atmosphäre mit der Photo-DSC-Methode nicht eindeutig geklärt werden. Die Qualität der pyrolysierten PZT-Schichten konnte durch Variation der Feststoffgehalte und der Pyrolysebedingungen optimiert werden. So wurden durch die Verdünnung der PZT-Sole auf 25 Gew. % Feststoffgehalt bei ansonsten analogen Pyrolyse¬bedingungen dichtere und homogenere Schichten erhalten, wohingegen PZT-Solschichten mit einem Feststoffgehalt von 31 Gew.-% eine xerogelartige Struktur durch agglomerierte Partikel zeigten. Die untersuchten Substrate wiesen einen deutlichen Einfluss auf die nach der Pyrolyse erhaltenen PZT-Schichten auf.
Unmetallisierte Silizium-Wafer eigneten sich zwar zur Untersuchung der UV-lithographischen Herstellung strukturierter Solschichten, bei der Pyrolyse bildeten sich jedoch unerwünschte Bleisilicat¬schmelzen, die der PZT-Bildung entgegen wirkten. Weiterhin wurden platinierten Silizium-Wafer mit Titanhaftvermittler¬schicht und SiO2-Sperrschicht verwendet. Diese Substrate zeigten eine durch die hohen Pyrolyse¬temperaturen induzierten Diffusion der Haftvermittlerschicht in die Platinmetallisierung und in der Folge eine Delamination aufgrund hoher Druckspannung in der PZT-Schicht, welche durch die Entfernung der organischen Bestandteile aus der Solschicht resultierte. In der Folge entstanden mechanisch instabile Proben. Die Delamination konnte durch Zwischensinterschritte bei 360 °C bzw. durch die Pyrolyse auf Heizplatten reduziert, jedoch nicht vollständig vermieden werden. Aus diesem Grund konnten keine ferroelektrischen Eigenschaften der PZT-Schichten auf diesen Substraten ermittelt werden. Lediglich auf Stahlsubstraten konnten elektrisch dichte Schichten hergestellt werden.
Die Polymerisationsreaktion eines ausgesuchten PZT-Sols, welches bei den Strukturierungs- und Pyrolyseuntersuchungen die besten Ergebnisse zeigte, wurde näher untersucht, um ein Verständnis zu den ablaufenden Reaktionen zu erhalten. Die Reaktion zeigt den für radikalische Kettenpolymerisationen typischen Trommsdorf- bzw. Gel-Effekt. Durch die Verdünnung der PZT-Cluster konnten trotz hoher Anzahl an C=C-Bindungen pro Monomer, d. h. stark quervernetzender Spezies, sehr hohe Umsetzungsgrade erreicht werden. Die initiator¬gehalts-abhängige Steigerung der Reaktionsenthalpie während der Belichtung konnte mit steigenden Umsetzungs¬graden korreliert werden. Dieses Verhalten wird in der Literatur für eine Vielzahl an methacrylatbasierten Monomeren beschrieben. Der empirische Zusammenhang zwischen der Reaktionsrate und der Initiatorkonzentration konnte für das methacrylat¬funktionalisierte PZT-Sol zu bestimmt werden. Die Gesamtaktivierungenergie konnte ebenfalls in Abhängigkeit vom Reaktionsumsatz ermittelt werden. Die Polymerisation eines konzentrierten PZT-Sols mit difunktionellen Monomeren zeigte höhere Umsetzungsgrade als die entsprechenden Reinsubstanzen und ein späteres Eintreten des Gelpunkts. Dies wird zurückgeführt auf die niedrigere Viskosität sowie die geringere Molekülgröße des Reaktivverdünners und die in der Folge bessere Infiltration des durch das PZT-Sol aufgespannten Netzwerks mit zusätzlichen kleinen und beweglichen Monomeren.
Da bei der thermischen Behandlung der polymerisierten PZT-Schichten die experimentellen Bedingungen, wie z. B. Heizrate oder Anfangstemperatur der Pyrolyse, einen signifikanten Einfluss besitzen, wurden energiedispersive in-situ-Röntgenbeugungsexperimente mit Hilfe von Synchrotron¬strahlung durch¬geführt. Hierbei konnte vor allem der Einfluss der organischen Polymerisation und der Anfangstemperatur der Pyrolyse auf die Phasenentwicklung des Precursor-Sols untersucht werden. Dabei zeigte sich besonders deutlich, dass die PZT-Bildungstemperatur durch die Belichtung der Probe und damit der Bildung des organischen Netzwerks steigt. Dies wird auf das starke organische Netzwerk und den erhöhten Energiebedarf zur Reorganisation zurückgeführt. Durch eine erhöhte Anfangstemperatur der Pyrolyse von 500 °C kann die in der Literatur beschriebene Verringerung der unerwünschten Pyrochlor-Phase beobachtet werden. Zudem ist die PZT-Bildungstemperatur hierbei niedriger als bei Pyrolysen ab Raumtemperatur. Dies wird mit einer durch den geringeren Anteil an Pyrochlor-Phase erhöhten Triebkraft zur PZT-Bildung erklärt. Die konsekutive Phasenumwandlung von Pyrochlor in die erwünschte PZT-Phase ist bei Pyrolysen ab Raumtemperatur später abgeschlossen als bei Pyrolysen ab 500 °C. Dies kann mit dem geringeren Anteil an Pyrochlor-Phase erklärt werden, da sich ab 500 °C bereits die PZT-Phase zu bilden beginnt. Der Vergleich der Halbwertsbreiten des intensitätsstarken Reflexes bei 31 ° zeigte, dass die Belichtung der Solschichten zu größeren Kristalliten führt, was auf eine niedrigere Nukleationsrate während der Kristallisation zurückzuführen ist.
Bei der Auswertung und Diskussion der Ergebnisse der im Rahmen dieser Arbeit untersuchten Thematik ergaben sich einige Ansatzpunkte für weitere Untersuchungen, die jedoch nicht im zeitlichen und thematischen Rahmen dieser Arbeit betrachtet werden konnten. So wäre es interessant weitere Substrate auf ihre Eignung bei der, für die Herstellung strukturierten PZT-Schichten notwendigen, hohen thermischen Belastung zu testen, da die hier untersuchten platinierten Silizium-Wafer bzw. die Stahlsubstrate deutliche Nachteile zeigten. Des Weiteren können neben den hier behandelten polymerisierbaren Carbonsäureliganden weitere mehrzähnige Liganden und dessen Einfluss auf die Precursormaterialien für die Herstellung strukturierter anorganischer Schichten untersucht werden. β-Diketone, Aminoalkohole und auch Phosphonate bilden ebenfalls stabile Metall-Ligand-Bindungen und könnten so die Integration polymerisierbarer Gruppen in die PZT-Cluster analog zu der im Rahmen dieser Arbeit verwendeten Carbonsäure ermöglichen.
Das hier erarbeitete Konzept könnte neben den untersuchten ferroelektrischen Schichtmaterialien ebenfalls auf andere dielektrische oder magnetische Materialklassen, wie z. B. Bariumtitanat oder Magnetit übertragen werden. Die Herstellung photostrukturierbarer anorganischer Schichten durch den Einbau polymerisierbarer Liganden eignet sich für zahlreiche Materialklassen, die über flüssige Sol-Gel-Vorstufen synthetisiert werden können.
Darüber hinaus können weitere Pyrolysebedingungen mit den hier vorgestellten Analysemethoden untersucht werden. So könnten z. B. verschiedene Atmosphären oder unterschiedliche Heizraten innerhalb eines Pyrolysezyklus variiert werden, um ihren Einfluss auf die Herstellung rissfreier und phasenreiner Schichten zu untersuchen. Dazu müsste der hier vorgestellte experimentelle Aufbau dahingehend erweitert werden, dass eine Gaszufuhr während der Pyrolyse möglich ist.
The demand for LIB with enhanced energy densities leads to increased utilization of the space within the confinements of the battery housing or to the use of electrode material with increased intrinsic specific energy densities. Both requirements result in more stress on the battery electrodes and separator during cycling or aging. However, the effect of mechanical strain on the cell’s electrochemistry and thus the performance of batteries is rather unexplored compared to the impact of current or temperature, for example. The objective of this thesis was to give a better understanding of the electrochemical and mechanical interplay in current- and next-generation lithium based battery cells. Therefore, the thesis was structured into the investigations on SoA and next-generation LIBs. For SoA LIBs, the investigations of the interplay started at laboratory scale. Here, the expansion of various electrodes and also the impact of mechanical pressure and its distribution on the performance of the cells were
studied. The investigations at laboratory scale was followed by an examination of the electrochemical and mechanical interactions on large format commercial LIBs which are used in BEVs. Accordingly, the effect of bracing and its effect on the performance was studied in an aging and post-mortem study. To gain a deeper understanding of the mechanical changes in LIBs, an ultrasonic study was performed for pouch cells. Here, the mechanical changes were further investigated in dependence of SoC and SoH. The effects of the mechanical stress on the performance for next-generation batteries were studied at laboratory scale. In the beginning, the expansion of next-generation anode materials such as silicon and lithium was compared with today’s anode materials. Furthermore, the effect of mechanical pressure and electrolyte on the irreversible dilation and performance was investigated for lithium metal cells. Overall, it was shown that pressure has a significant effect on the performance of today’s and also future LIBs. The interplay of the electrochemical and mechanical effects inside a LIB has a considerable impact on the lifetime, capacity fading and impedance increase of the batteries.
In the framework of this thesis, new UV-patternable organic-inorganic hybrid polymers with higher refractive indices than reported in the literature for photonic applications were developed and studied with respect to their chemical structure, their optical properties, and their ability of being patterned by 1PP and 2PP. Particularly with 2PP, one could create 3D structures using the novel hybrid materials. The materials were prepared from hydrolysis and polycondensation reactions of · organo-alkoxysilanes and titanium alkoxide precursors, modified with and without CL and organo-alkoxysilanes precursors, and · organo-alkoxysilanes, titanium alkoxide and organophosphorus precursors. The major scope of this work was to increase the refractive index of ORMCER® materials based on only organo-alkoxysilanes. Thus, the parameters which influence the refractive index were investigated thoroughly. In particular, the synthesis parameters such as the introduction of titanium alkoxide and its concentration, the organo-alkoxysilanes, the catalyst concentration, the solvent used, but, also the processing parameters such as, the UV exposure dose, initiator concentration, and developer were investigated.
This work deals with the sintering of multi-material composites. It aims at the establishment of an alternative to the existing complex models for sintering. The development of the associated experimental procedure is also included in this work. The developed material model must be able to predict (i) the sintering kinetics and (ii) the viscous moduli of a material. An experimental approach with free sintering and hot-forging measurements is favoured in this work. The prediction of the sintering kinetics is addressed with the construction of a map of sintering kinetics data: the Master Sintering Diagram (MSD). The MSD is based on a generalized equation for solid-state diffusion, thus is suitable for any thermal activated diffusion. The MSD allows the prediction of sintering kinetics for a large range of temperatures and external loads. A novel approach to the determination of the viscous moduli is developed in this work: the cyclic unloading method. It is a hot-forging measurement (sintering under uniaxial compression) where the applied load is released for short periods. The measurements are carried out with continuous heating, so that the viscous moduli are determined over large ranges of temperatures and densities. The advantage of this method is the measurement of the viscous moduli in anisotropic microstructures. The material model is validated in two steps. Firstly, the predictions of sintering kinetics with the MSD are compared with experimental results: changes of thermal profile and changes of load are predicted with a maximum deviation of 10%. Secondly, the experimentally determined viscous moduli are used for the prediction of a bi-layer curvature using models for warpage from literature. The prediction is qualitatively good for a maximum deviation of 27%. The study of a sintering glass-ceramic tape on a rigid substrate is presented. It shows that this co-sintering problem can be qualitatively investigated with requirement of the material model. The formation of anisotropy intrinsic to the hot-forging experiments is also reported in this work. It appears to be a important point to address in the future for a better understanding of the cosintering.
Das Mischkristallsystem PbZrxTi1-xO3 (PZT) gehört durch seine ausgeprägten piezo- und ferroelektrischen Eigenschaften zu den meist verwendeten Funktionskeramiken. In Form von Dünnschichten auf flexiblen Metallsubstraten können sie für unterschiedlichste Anwendungen als Tastschalter, Vibrationsdämpfer, Mikroaktuator oder Ultraschallwandler eingesetzt werden. Ziel der vorliegenden Arbeit ist es, den Gefügeaufbau und den Phasenbestand von PZT-Schichten, die in einem mehrstufigen Sol-Gel-Prozeß auf Blechen der säure- und temperaturbeständigen Chrom-Nickel-Legierung Hastelloy abgeschieden worden sind, zu analysieren und mit ihren ferroelektrischen und die-lektrischen Eigenschaften zu korrelieren. Es wird nachgewiesen, daß das Gefüge gezielt mittels verschiedener Temperaturbehandlungen und unter-schiedlicher Neodymdotierung variiert werden kann. Durch Nd-Dotierung wird das Maximum der Keimbil-dungsrate zu niedrigeren Temperaturen hin verschoben und die Wachstumsgeschwindigkeit gegenüber undotierten Schichten verringert. Die Kristallisation in dotierten und undotierten Schichten ist heterogen und erfolgt bevorzugt an den Grenz- und Oberflächen sowie an den Porenrändern im Innern der Schichten. Die Zusammensetzung der PZT-Sol-Gel-Beschichtungen liegt im Bereich der morphotropen Phasengrenze (x=0,53) zwischen tetragonaler und rhomboedrischer Phase. Erstmals wurde die während der Temperatur-behandlung auftretende Gradientenbildung im Zr/Ti-Verhältnis systematisch mit dem Gefüge und dem Phasenbestand auf Nanometerskala in Verbindung gesetzt. Hierbei konnte aufgezeigt werden, dass langreichweitige Zr:Ti-Fluktuationen vorrangig während der Kri-stallisation der Pyrochlorphase entstehen. Bei der nachfolgenden Perowskitkristallisation wachsen die Kri-stalle über die entmischten Bereiche hinweg, so daß Schwankungen im Zr:Ti-Verhältnis innerhalb der Kristallite erhalten bleiben. Es wird dargelegt, daß die Fluktuationen im Zr:Ti-Verhältnis infolge der starken Spannungen innerhalb der Schicht nur geringe Auswirkungen auf den Verzerrungsgrad der Kristallite und die Zugehörigkeit zur rhomboedrischen oder tetragonalen Perowskitphase haben. Beim Übergang in den ferroelektrischen Zustand ist das Ausmaß der Gitterverzerrung unabhängig von der Kristallitgröße und in Nd-dotierten Schichten generell geringer als in undotierten Schichten. Es wird die Ausbildung einer Zwischenschicht zwischen Metallsubstrat und PZT-Schicht untersucht, die die resultierenden elektrischen Eigenschaften maßgeblich beeinflußt. Sie wird als Oxidschicht identifiziert, be-stehend aus kristallinem NiO und NiCr2O4, verschiedenen Chromoxiden und Pb2(CrO4)O, die als nicht-ferroelektrisches Dielektrikum die resultierende Dielektrizitätskonstante des Substrat-Schichtverbundes stark herabsetzt. Durch Aufbringen einer unterstöchiometrischen La0,75Sr0,2MnO3 (ULSM) - Elektrodierung vor der PZT-Beschichtung gelingt es, die Kontaktierung zu verbessern, die (001)-Orientierung in undotierten Schichten zu steigern und sehr schmale P-E-Hysteresekurven zu erhalten.
Control of living cells is vital for the survival of organisms. Each cell inside an organism is exposed to diverse external mechano-chemical cues, all coordinated in a spatio-temporal pattern triggering individual cell functions. This complex interplay between external chemical cues and mechanical 3D environments is translated into intracellular signaling loops. Here, we describe how external mechano-chemical cues control cell functions, especially cell migration, and influence intracellular information transport. In particular, this work focuses on the quantitative analysis of (1) intracellular vesicle transport to understand intracellular state changes in response to external cues, (2) cellular sensing of external chemotactic cues, and (3) the cells' ability to migrate in 3D structured environments, artificially fabricated to mimic the 3D environment of tissue in the human body.
The introduction of novel bioactive materials to manipulate living cell behavior is a crucial topic for biomedical research and tissue engineering. Biomaterials or surface patterns that boost specific cell functions can enable innovative new products in cell culture and diagnostics. This study aims at investigating the interaction of living cells with microstructured, nanostructured and nanoporous material surfaces in order to identify distinct systematics in cell-material interplay. For this purpose, three different studies were carried out and yielded individual effects on different cell functions.
Cell migration processes are controlled by sensitive interaction with external cues such as topographic structures of the cell's environment. The first part of this study presents systematically controlled assays to investigate the effects of spatial density and local geometry of micron scale topographic cues on amoeboid migration of Dictyostelium discoideum cells in quasi-3D pillar fields with systematic variation of inter-pillar distance and pillar lattice geometry. We can extract motility parameters in order to elucidate the details of amoeboid migration mechanisms and consolidate them in a two-state contact-controlled motility model, distinguishing directed and random phases. Specifically, we find that directed pillar-to-pillar runs are found preferably in high pillar density regions, and cells in directed motion states sense pillars as attractive topographic stimuli. In contrast, cell motion in random probing states is inhibited by high pillar density, where pillars act as obstacles for cell motion. In a gradient spatial density, these mechanisms lead to topographic guidance of cells, with a general trend towards a regime of inter-pillar spacing close to the cell diameter. In locally anisotropic pillar environments, cell migration is often found to be damped due to competing attraction by different pillars in close proximity and due to lack of other potential stimuli in the vicinity of the cell. Further, we demonstrate topographic cell guidance reflecting the lattice geometry of the quasi-3D environment by distinct preferences in migration direction.
We further investigate amoeboid single-cell migration on intrinsically nano-structured, biodegradable silica fibers in comparison to chemically equivalent plain glass surfaces. Cell migration trajectories are classified into directed runs and quasi-random migration by a local mean squared displacement (LMSD) analysis. We find that directed movement on silica fibers is enhanced in a significant manner by the fibers' nanoscale surface-patterns. Further, cell adhesion on the silica fibers is a microtubule-mediated process. Cells lacking microtubules detach from the fibers, but adhere well to glass surfaces. Knock-out mutants of myosin II migrating on the fibers are as active as cells with active myosin II, while the migration of the knock-out mutants is hindered on plain glass.
We investigate the influence of the intrinsically nano-patterned surface of nanoporous glass membranes on the behavior of mammalian cells. Three different cell lines and primary human mesenchymal stem cells (hMSCs) proliferate readily on nanoporous glass membranes with mean pore sizes between 10 nm and 124 nm. In both proliferation and mRNA expression experiments, L929 fibroblasts show a distinct trend towards mean pore sizes > 80 nm. For primary hMSCs, excellent proliferation is observed on all nanoporous surfaces. hMSC on samples with 17 nm pore size display increased expression of COL10, COL2A1 and SOX9, especially during the first two weeks of culture. In upside down culture, SK MEL-28 cells on nanoporous glass resist the gravitational force and proliferate well in contrast to cells on flat references. The effect of paclitaxel treatment of MDA MB 321 breast cancer cells is already visible after 48 h on nanoporous membranes and strongly pronounced in comparison to reference samples.
The studies presented in this work showed novel and distinct effects of micro- and nanoscale topographies on the behavior of various types of living cells. These examples display how versatile the potential for applications of bioactive materials could become in the next years and decades. And yet this variety of different alterations of cell functions due to topographic cues also shows the crucial part of this field of research: Carving out distinct, robust correlations of external cues and cell behavior is of utmost importance to derive definitive design implications that can lead to scientifically, clinically and commercially successful products.
Die vorliegende Arbeit befasste sich mit der bisher relativ unbekannten Polymerklasse der Polypeptoide, die hinsichtlich ihrer Verwendung als Biomaterial näher untersucht werden sollte. Hierbei war die Untersuchung des Polymerisationssystems ein wesentlicher Schwerpunkt. Dies beinhaltete zum einen die Synthesen verschiedener Monomere sowie deren Polymerisationskinetiken und zum anderen Studien über die Stabilität des aktiven Kettenendes. Um mehr über die Polypeptoide zu erfahren, wurden die erhaltenen Homopolymere nach der Strukturanalyse hinsichtlich ihrer physikochemischen Eigen-schaften untersucht. Im Anschluss erfolgte die Synthese von (amphiphilen) Blockco-polypeptoiden, die sich in wässrigen Lösungen zu definierten Morphologien zusammen-lagern. Die resultierenden Morphologien, sowohl mizellare als auch vesikuläre Strukturen, wurden mit verschiedenen Methoden, wie z. B. der Pyren-Fluoreszenz-Spektroskpie und der dynamischen Lichtstreuung, untersucht. Erste Erkenntnisse über die Biokompatibilität der Polypeptoide sollte die Bestimmung der Zellviabilität in verschiedenen Polymerlösungen liefern.
Die verschiedenen Studien über die Polypeptoide zeigten, dass diese Polymerklasse über eine besonders lebende Polymerisation synthetisiert werden kann. Dabei resultieren Produkte, die sich durch eine Poisson-Verteilung und eine hohe Endgruppengenauigkeit auszeichnen. Zusätzlich bestehen Polypeptoide aus einem abbaubaren Rückgrat und, im Vergleich zu den Polypeptiden, besitzen sie eine erhöhte proteolytische Stabilität. Amphiphile Blockcopolypeptoide sind zudem in der Lage, sich in Lösung zu verschiedenen Morphologien anzuordnen. Durch die Variierung der Seitenkette und des f kann sowohl die Selbstorganisation als auch das Mikroumfeld der Aggregate abgestimmt werden. Darüber hinaus können die amphiphile Blockcopolymere, die sich zu Mizellen anordnen, hydrophobe Substanzen solubilisieren.
Polypeptoide liefern all die nötige chemische Vielseitigkeit und potentielle Biokompatibilität, um bestehende sowie neuartige Probleme in biomedizinischen Anwendungen zu bewältigen. Zukünftige in vivo und in vitro Test werden das Potential, aber auch die Grenzen dieser neuen Polymerklasse als Biomaterial zeigen.
Polypeptoids are an old but recently rediscovered polymer class with interesting synthetic, physico-chemical and biological characteristics. Here, we introduce new aromatic monomers, N-benzyl glycine N-carboxyanhydride and N-phenethyl glycine N-carboxyanhydride and their block copolymers with the hydrophilic polysarcosine. We compare their self-assembly in water and aqueous buffer with the self-assembly of amphiphilic block copolypeptoids with aliphatic side chains. The aggregates in water were investigated by dynamic light scattering and electron microscopy. We found a variety of morphologies, which were influenced by the polymer structure as well as by the preparation method. Overall, we found polymersomes, worm-like micelles and oligo-lamellar morphologies as well as some less defined aggregates of interconnected worms and vesicles. Such, this contribution may serve as a starting point for a more detailed investigation of the self-assembly behavior of the rich class of polypeptoids and for a better understanding between the differences in the aggregation behavior of non-uniform polypeptoids and uniform peptoids.
In der vorliegenden Dissertation - Kathoden für Metall-Luft Batterien - steht die Komponente Gasdiffusionselektrode (GDE) – oftmals auch als Luft-Kathode bezeichnet – einer wässrigen Metall-Luft Batterie im Fokus.
Ziel dieser Arbeit ist die Synthese und Charakterisierung verschiedener Katalysatorsysteme für die Sauerstoffreduktion und -evolution. Dabei soll auf die Verwendung von Edelmetallen verzichtet und der Einsatz von verfügbaren und günstigen Materialien bzw. Herstellungsprozessen favorisiert werden. Auf Basis von bekannten Materialklassen sollen repräsentative Katalysatoren synthetisiert und ihre katalytischen Aktivitäten für die Sauerstoffreduktion und -evolution bestimmt werden. Im Detail wird eine mögliche Korrelation der strukturellen Eigenschaften der Katalysatoren auf die katalytische Aktivität untersucht. Auf Basis dieser Erkenntnisse sollen die Katalysatoren modifiziert werden, um die katalytischen Eigenschaften weiter zu optimieren. Um einen geschlossenen Entwicklungszyklus in dieser Arbeit realisieren zu können, wird parallel ein kostengünstiger und skalierbarer Herstellungsprozess von GDEs entwickelt.
Ein weiteres Ziel dieser Arbeit ist es, Konzepte für sekundäre Zink-Luft Energiespeicher zu erarbeiten und deren Umsetzung zu untersuchen. Dabei kommen die zuvor entwickelten Katalysatoren zum Einsatz.
Die vorliegende Arbeit gliedert sich, nach der Darlegung der relevanten Grundlagen mit Stand der Wissenschaft und Technik, in vier Teilkapitel, in denen die einzelnen Ziele adressiert sind. Dies sind die Erforschung reiner Katalysatoren und hybrider Katalysatoren sowie die Etablierung eines Herstellungsprozesses für GDEs und die Implementierung dieser in sekundäre Zink-Luft Energiespeicher. Die experimentellen Grundlagen befinden sich im darauffolgenden Kapitel.
Here, we report a continuous flow synthesis of nano LDH, comprising a continuous precipitation process using static mixers and followed by an immediate cleaning process via a semi-continuous centrifuge to obtain the final product in one-go. Via this synthesis setup, it is possible to independently vary the concentrations of the reactants during precipitation and at the same time ensure constant reaction conditions and an immediate "quenching" of the precipitate due to "on the flow"-washing. We found that this paves the way to adjust the synthesis parameters in a way that the final morphology of the nano-LDH particles can be controlled to be either round or platelet-like.
This thesis investigates different ligand designs for Ru(II) complexes and the activity of the complexes as photosensitizer (PS) in photocatalytic hydrogen evolution. The catalytic system typically contains a catalyst, a sacrificial electron donor (SED) and a PS, which needs to exhibit strong absorption and luminescence, as well as reversible redox behavior. Electron-withdrawing pyridine substituents on the terpyridine metal ion receptor result in an increase of excited-state lifetime and quantum yield (Φ = 74*10-5; τ = 3.8 ns) and lead to complex III-C1 exhibiting activity as PS. While the turn-over frequency (TOFmax) and turn-over number (TON) are relatively low (TOFmax = 57 mmolH2 molPS-1 min-1; TON(44 h) = 134 mmolH2 molPS-1), the catalytic system is long-lived, losing only 20% of its activity over the course of 12 days. Interestingly, the heteroleptic design in III-C1 proves to be beneficial for the performance as PS, despite III-C1 having comparable photophysical and electrochemical properties as the homoleptic complex IV-C2 (TOFmax = 35 mmolH2 molPS-1 min-1; TON(24 h) = 14 mmolH2 molPS-1). Reductive quenching of the excited PS by the SED is identified as rate-limiting step in both cases.
Hence, the ligands are designed to be more electron-accepting either via N-methylation of the peripheral pyridine substituents or introduction of a pyrimidine ring in the metal ion receptor, leading to increased excited-state lifetimes (τ = 9–40 ns) and luminescence quantum yields (Φ = 40–400*10-5). However, the more electron-accepting character of the ligands also results in anodically shifted reduction potentials, leading to a lack of driving force for the electron transfer from the reduced PS to the catalyst. Hence, this electron transfer step is found to be a limiting factor to the overall performance of the PS. While higher TOFmax in hydrogen evolution experiments are observed for pyrimidine-containing PS (TOFmax = 300–715 mmolH2 molPS-1 min-1), the longevity for these systems is reduced with half-life times of 2–6 h.
Expansion of the pyrimidine-containing ligands to dinuclear complexes yields a stronger absorptivity (ε = 100–135*103 L mol-1 cm-1), increased luminescence (τ = 90–125 ns, Φ = 210–350*10-5) and can also result in higher TOFmax given sufficient driving force for electron transfer to the catalyst (TOFmax = 1500 mmolH2 molPS-1 min-1). When comparing complexes with similar driving forces, stronger luminescence is reflected in a higher TOFmax. Besides thermodynamic considerations, kinetic effects and electron transfer efficiency are assumed to impact the observed activity in hydrogen evolution. In summary, this work shows that targeted ligand design can make the previously disregarded group of Ru(II) complexes with tridentate ligands attractive candidates for use as PS in photocatalytic hydrogen evolution.

Der erste Teil der Arbeit untersucht den Einsatzes von 4,6-Di-(pyrid-2´-yl)-1,3,5-triazin als Baustein für Metallo-supramolekulare Polyelektrolyte. Die dafür nötigen ditopen Liganden werden mittels Stille Kreuzkupplungen dargestellt. Die Absorptions- und Fluoreszenzeigenschaften können durch den Einbau von Oligothiophenen eingestellt werden.
Im zweiten Teil der Arbeit werden die elektrorheologischen Eigenschaften von Metallo-supramolekularen Polyelektrolyten untersucht. Zu diesem Zweck werden die Koordinationspolymere in das Schichtsilikat Montmorillonit interkaliert. Die Interkalation wird mittels verschiedener analytischer Methoden wie Pulverdiffraktometrie, Thermoanalyse oder Infrarotspektroskopie untersucht. Die entstehenden Nanokomposite zeigen einen elektrorheologischen Effekt bei einer geringen Stromdichte.
Oxidative precipitation is a facile synthesis method to obtain ferromagnetic iron oxide nanoparticles from ferrous salts—with unexplored potential. The concentration of base and oxidant alone strongly affects the particle's structure and thus their magnetic properties despite the same material, magnetite (Fe\(_{3}\)O\(_{4}\)), is obtained when precipitated with potassium hydroxide (KOH) from ferrous sulfate (FeSO\(_{4}\)) and treated with potassium nitrate (KNO\(_{3}\)) at appropriate temperature. Depending on the potassium hydroxide and potassium nitrate concentrations, it is possible to obtain a series of different types of either single crystals or mesocrystals. The time‐dependent mesocrystal evolution can be revealed via electron microscopy and provides insights into the process of oriented attachment, yielding faceted particles, showing a facet‐dependent reactivity. It is found that it is the nitrate and hydroxide concentration that influences the ligand exchange process and thus the crystallization pathways. The presence of sulfate ions contributes to the mesocrystal evolution as well, as sulfate apparently hinders further crystal fusion, as revealed via infrared spectroscopy. Finally, it is found that nitrite, as one possible and ecologically highly relevant reduction product occurring in nature in context with iron, only evolves if the reaction is quantitative.
Starting off with solubility experiments of possible precursors, the present study reveals the whole development of a sol gel processing route for transparent p type semiconductive thin films with delafossite structure right to the fabrication of functional p-n junctions. The versatile sol formulation could successfully be modified for several oxide compositions, enabling the synthesis of CuAlO2, CuCrO2, CuMnO2, CuFeO2 and more. Although several differences in the sintering behaviour of powders and thin films could be observed, the powder experiments significantly contributed to the clearification of the intricate phase development during thermal annealing and also to optimization of the annealing sequence for thin film processing. Two different ternary systems turned out to be the most promising candidates for p-TCO application: Copper aluminum oxide for its high optical transmittance and copper chromium oxide for its low synthesis temperature, which allowed thin film deposition on low-cost borosilicate substrates. In order to combine the advantages of these two systems, the quaternary oxide composition CuAl1-xCrxO2 was investigated. With a higher optical transmittance than CuCrO2, a lower synthesis temperature than CuAlO2 and a lower resistivity than both parent systems, the optimum composition of the quaternary oxide is reached for x = 0.50. Compared to physical vapour deposition techniques, the undoped thin films presented here still need to make up some deficites in their optoelectronic performance. Although the best sol-gel samples are able to compete with RF sputtered samples or sampes deposited by PLD in transmittance, their resistivity is almost two orders of magnitude higher. The most probable reasons for this are the characteristic imperfections of sol-gel thin films like porosity and small crystallite size, which create barriers like grain boundaries and bottlenecks like barely connected particles. By additional effort such shortcomings can be repelled to a certain extend, but nevertheless the density of undoped sol-gel material always stays behind its pendants processed by physical vapour deposition.[246] Furthermore, such additional endeavour is likely to annihilate the advantage of sol-gel technique in processing costs. Extrinsic doping is a common method to decrease the resistivity of delafossite materials. Partially replacing the trivalent cations by divalent ones creates additional holes and thus generates additional charge carriers for p-type semiconductivity. This can improve the conductivity of delafossites by up to three orders of magnitude. Due to the compositorial flexibility of sol-gel processing, dopants could be introduced easily in this study by soluble precursors. However, improving the conductivity of CuAlO2 and CuAl0.5Cr0.5O2 via this method failed. Actually, this seems to be due to the fact that instead of being incorporated into the delafossite phase the dopant ions form intransparent phase impurities like spinels, which interfere with optical transmittance of the thin films. On the contrary, doping had a positive effect on the conductivity and the optical transmittance of copper chromium oxide, with magnesium being the most effective dopant. The resistivity could be decreased by more than three orders of magnitude, but in order to achieve this, much higher Mg concentrations than by other thin film deposition methods were necessary. This indicates a low doping efficiency in sol gel processed thin films, but also the ability of sol gel processing to incorporate more magnesium into the oxide than any other processing method. The extensive substitution of the chromium ions also increases the optical transmittance and allows sol gel processed thin films to draw level with thin films deposited by sputtering methods or PLD. Finally, the applicability of the delafossite thin films was proven by the asymmetric current voltage characteristics of heterojunctions between ITO and the delafossites. Shunting problems of the metallic contacts, on the other hand, reveal structural deficites of the delafossites, which should be the subject of further investigations.
Hydrogels are key components in bioink formulations to ensure printability and stability in biofabrication. In this study, a well-known Diels-Alder two-step post-polymerization modification approach is introduced into thermogelling diblock copolymers, comprising poly(2-methyl-2-oxazoline) and thermoresponsive poly(2-n-propyl-2-oxazine). The diblock copolymers are partially hydrolyzed and subsequently modified by acid/amine coupling with furan and maleimide moieties. While the thermogelling and shear-thinning properties allow excellent printability, trigger-less cell-friendly Diels-Alder click-chemistry yields long-term shape-fidelity. The introduced platform enables easy incorporation of cell-binding moieties (RGD-peptide) for cellular interaction. The hydrogel is functionalized with RGD-peptides using thiol-maleimide chemistry and cell proliferation as well as morphology of fibroblasts seeded on top of the hydrogels confirm the cell adhesion facilitated by the peptides. Finally, bioink formulations are tested for biocompatibility by incorporating fibroblasts homogenously inside the polymer solution pre-printing. After the printing and crosslinking process good cytocompatibility is confirmed. The established bioink system combines a two-step approach by physical precursor gelation followed by an additional chemical stabilization, offering a broad versatility for further biomechanical adaptation or bioresponsive peptide modification.
Thermoresponsive polymers are frequently involved in the development of materials for various applications. Here, polymers containing poly(2- benzhydryl-2-oxazine) (pBhOzi) repeating units are described for the first time. The homopolymer pBhOzi and an ABA type amphiphile comprising two flanking hydrophilic A blocks of poly(2-methyl-2-oxazoline) (pMeOx) and the hydrophobic aromatic pBhOzi central B block (pMeOx-b-pBhOzi-b-pMeOx) are synthesized and the latter is shown to exhibit inverse thermogelling properties at concentrations of 20 wt.% in water. This behavior stands in contrast to a homologue ABA amphiphile consisting of a central poly(2-benzhydryl-2-oxazoline) block (pMeOx-b-pBhOx-b-pMeOx). No inverse thermogelling is observed with this polymer even at 25 wt.%. For 25 wt.% pMeOx-b-pBhOzi-b-pMeOx, a surprisingly high storage modulus of ≈22 kPa and high values for the yield and flow points of 480 Pa and 1.3 kPa are obtained. Exceeding the yield point, pronounced shear thinning is observed. Interestingly, only little difference between self-assemblies of pMeOx-b-pBhOzi-b-pMeOx and pMeOx-b-pBhOx-b-pMeOx is observed by dynamic light scattering while transmission electron microscopy images suggest that the micelles of pMeOx-b-pBhOzi-b-pMeOx interact through their hydrophilic coronas, which is probably decisive for the gel formation. Overall, this study introduces new building blocks for poly(2-oxazoline) and poly(2-oxazine)-based self-assemblies, but additional studies will be needed to unravel the exact mechanism.
In the past decade, poly(2-oxazoline)s (POx) and very recently poly(2-oxazine)s (POzi) based amphiphiles have shown great potential for medical applications. Therefore, the major aim of this thesis was to further explore the pharmaceutical and biomedical applications of POx/POzi based ABA triblock and AB diblock copolymers, respectively with the special emphasis on structure property relationship (SPR). ABA triblock copolymers (with shorter side chain length in the hydrophobic block) have shown high solubilizing capacity for hydrophobic drugs. The issue of poor aqueous solubility was initially addressed by developing a (micellar) formulation library of 21 highly diverse, hydrophobic drugs with POx/POzi based ABA triblock copolymers. Theoretically, the extent of compatibility between polymers and drug was determined by calculating solubility parameters (SPs). The SPs were thoroughly investigated to check their applicability in present systems. The selected formulations were further characterized by various physico-chemical techniques. For the biomedical applications, a novel thermoresposive diblock copolymer was synthesized which has shown promising properties to be used as hydrogel bioink or can potentially be used as fugitive support material. The most important aspect i.e. SPR, was studied with respect to hydrophilic block in either tri- or di-block copolymers. In triblock copolymer, the hydrophilic block played an important role for ultra high drug loading, while in case of diblock, it has improved the printability of the hydrogels. Apart from the basic research, the therapeutic applications of two formulations i.e. mitotane (commercially available as tablet dosage form for adrenocortical carcinoma) and BT-44 (lead compound for nerve regeneration) were studied in more detail.
Adrenocortical carcinoma (ACC) is a malignant tumor originating from the adrenal gland cortex with a heterogeneous but overall dismal prognosis in advanced stages. For more than 50 years, mitotane has remained a cornerstone for the treatment of ACC as adjuvant and palliative therapy. It has a very poor aqueous solubility of 0.1 mg/l and high partition coefficient in octanol/water (log P) value of 6. The commercially available dosage form is 500 mg tablets (Lysodren®). Even at doses up to 6 g/day (12 tablets in divided doses) for several months, > 50% patients do not achieve therapeutic plasma concentration > 14 mg/l due to poor water solubility, large volume of distribution and inter/intra-individual variability in bioavailability. This article aims to give a concise update of the clinical challenges associated with the administration of high-dose mitotane oral therapy which encompass the issues of poor bioavailability, difficult-to-predict pharmacokinetics and associated adverse events. Moreover, we present recent efforts to improve mitotane formulations. Their success has been limited, and we therefore propose an injectable mitotane formulation instead of oral administration, which could bypass many of the main issues associated with high-dose oral mitotane therapy. A parenteral administration of mitotane could not only help to alleviate the adverse effects but also circumvent the variable oral absorption, give better control over therapeutic plasma mitotane concentration and potentially shorten the time to achieve therapeutic drug plasma concentrations considerably.
Mitotane as tablet form is currently the standard treatment for adrenocortical carcinoma. It has been used for 5 decades but suffers from highly variable responses in patients, subsequent adverse effects and overall lower response rate. This can be fundamentally linked to the exceedingly poor water solubility of mitotane itself. In terms of enhancing water solubility, a few research groups have attempted to develop better formulations of mitotane to overcome the issues associated with tablet dosage form. However, the success rate was limited, and these formulations did not make it into the clinics. In this article, we have comprehensively reviewed the properties of these formulations and discuss the reasons for their limited utility. Furthermore, we discuss a recently developed mitotane nanoformulation that led us to propose a novel approach to mitotane therapy, where intravenous delivery supplements the standard oral administration. With this article, we combine the current state of knowledge as a single piece of information about the various problems associated with the use of mitotane tablets, and herein we postulate the development of a new injectable mitotane formulation, which can potentially circumvent the major problems associated to mitotane's poor water solubility.
As one kind of “smart” material, thermogelling polymers find applications in biofabrication, drug delivery and regenerative medicine. In this work, we report a thermosensitive poly(2-oxazoline)/poly(2-oxazine) based diblock copolymer comprising thermosensitive/moderately hydrophobic poly(2-N-propyl-2-oxazine) (pPrOzi) and thermosensitive/moderately hydrophilic poly(2-ethyl-2-oxazoline) (pEtOx). Hydrogels were only formed when block length exceeded certain length (≈100 repeat units). The tube inversion and rheological tests showed that the material has then a reversible sol-gel transition above 25 wt.% concentration. Rheological tests further revealed a gel strength around 3 kPa, high shear thinning property and rapid shear recovery after stress, which are highly desirable properties for extrusion based three-dimensional (3D) (bio) printing. Attributed to the rheology profile, well resolved printability and high stackability (with added laponite) was also possible. (Cryo) scanning electron microscopy exhibited a highly porous, interconnected, 3D network. The sol-state at lower temperatures (in ice bath) facilitated the homogeneous distribution of (fluorescently labelled) human adipose derived stem cells (hADSCs) in the hydrogel matrix. Post-printing live/dead assays revealed that the hADSCs encapsulated within the hydrogel remained viable (≈97%). This thermoreversible and (bio) printable hydrogel demonstrated promising properties for use in tissue engineering applications.
Ziel dieser Arbeit war es, ein Schichtsystem auf Basis des Sol-Gel-Prozesses zu entwickeln, um Lighttrapping in Si-Dünnschichtsolarzellen zu erzeugen. Die Grundlage dieses Schichtsystems bilden SiO2-Partikel, die über den Stöber-Prozess hergestellt werden. Es zeigte sich, dass sich die Rauheit und der Haze der Schichten über die Partikelgröße und Schichtdicke einstellen lassen.
Um die mechanische Stabilität der reinen Stöber-Schichten zu verbessern, kamen verschiedene Binder zum Einsatz. Beste Ergebnisse zeigten Binder basierend auf löslichen Vorstufenpulvern, da diese dem Stöber-Sol beigemischt werden konnten und so Binder und Partikel gleichzeitig aufgebracht werden konnten.
Auf diese Weise entstehen mechanisch stabile, lichtstreuende Schichten. Zum Einsatz kam zunächst ein TiO2-Binder. Durch eine anschließende Glättung der Stöber-TiO2-Streuschichten mit SiO2 entsteht eine defektfreie, aber dennoch raue Oberfläche. Zusätzlich wird ein beträchtlicher Teil des Lichts in große Winkel gestreut.
Es konnte gezeigt werden, dass sich auf den SiO2-geglätteten Stöber-TiO2-Streuschichten ZnO:Al deponieren lässt, wobei die elektrischen Eigenschaften von der Dicke der Glättung abhängen. Auch die elektrischen Eigenschaften der Si-Dünnschichtsolarzellen hängen von der Glättung bzw. der Dicke der Glättung ab. Dies gilt insbesondere für die von der Materialqualität abhängigen Parameter Füllfaktor FF und offen Klemmenspannung VOC. Insgesamt fallen die Parameter jedoch noch gegenüber Referenzzellen auf geätztem Frontkontakt zurück. Vor allem aber wurde die hohe Zellreflexion aufgrund der Glas-TiO2-Grenzfläche als primäres Problem identifiziert. Dennoch konnte bei einer Glättungsdicke von 200 nm sehr gutes Lighttrapping beobachtet werden. Verantwortlich hierfür ist sehr wahrscheinlich die Großwinkelstreuung der Stöber-TiO2-Streuschichten.
Um die Zellreflexion zu verringern, wurde der Brechungsindex des Binders und der Glättungsschichten an den Stack aus Substrat, Streuschicht und ZnO:Al-Schicht angepasst. Idee war es, durch Einbringen eines Al2O3-Vorstufenpulvers eine niedrigbrechende Komponente bereitzustellen, um durch Mischung von Al2O3- und TiO2-Vorstufenpulver freie Hand über den Brechungsindex des Binders und der Glättung zu erhalten.
Da sich das Volumenverhältnis von SiO2-Partikeln zu Binder bei verschiedenen Al2O3-TiO2-Verhältnissen nur schwer bestimmen lässt, wurde lediglich ein reiner Al2O3-Binder in den Streuschichten eingesetzt. Die Einstellung des Brechungsindex beschränkte sich allein auf die Glättungsschichten. Um Stöber-Al2O3-Streuschichten mit hoher Rauigkeit und geringen Defekten zu erzielen, muss das Binder-zu-Partikel-Verhältnis angepasst werden. Beste Ergebnisse ergaben sich bei einem Al2O3-Gehalt von 2% im Sol. Aufgrund der hohen Rauigkeit besitzen die Streuschichten einen hohen Haze und wegen des geringen Brechungsunterschied zwischen Glas und Binder eine hohe Transmission. Die Glättung der Streuschichten im Al2O3-TiO2-System ist nur mit Hilfe einer zusätzlichen SiO2-Glättungsschicht und einer reduzierten Dicke auf 50 nm möglich. Auf den reinen defektreichen Streuschichten tendieren die Al2O3-TiO2-Schichten selbst zu Rissbildung.
Zur Untersuchung der ZnO:Al-Deposition wurde eine Glättungsdicke von 200 nm gewählt. Die erwies sich als zu gering. Die aufgebrachten ZnO:Al-Schichten wiesen größere Poren und kleinere Oberflächendefekte auf. Die Anpassung des Brechungsindex der Glättungsschichten an die ZnO:Al-Schicht erwies sich nicht als vorteilhaft. Die reine Al2O3-Glättung zeigt auch nach der ZnO:Al-Deposition die höchste Transmission. Die Winkelverteilung des Streulichts der Stöber-Al2O3-Streuschichten ist gegenüber den Stöber-TiO2-Streuschichten zu kleineren Winkeln verschoben. Dennoch wird ein größerer Anteil des Lichts in große Winkel gestreut, als es bei der geätzten ZnO:Al-Referenz der Fall ist.
Trotz der Defekte in den ZnO:Al-Schichten konnten auf den Stöber-Al2O3-Streuschichten funktionierende Tandemzellen hergestellt werden. Der Füllfaktor und die offene Klemmenspannung fallen nur geringfügig hinter die der Referenzzelle zurück. In der Kurzschlussstromdichte machen sich die verringerte Zellreflexion und das sehr gute Lighttrapping bemerkbar, so dass das Niveau der Referenz erreicht werden konnte. Zu beachten ist allerdings, dass gerade im langwelligen Lighttrapping-Spektralbereich die gleiche EQE erreicht wurde, trotz immer noch leicht erhöhter Zellreflexion.
Die letzte Versuchsreihe konnte zeigen, dass die entwickelten Schichten sich sehr gut zur Erzeugung von Lighttrapping in Si-Dünnschichtsolarzellen eignen.
In order to mimic the extracellular matrix for tissue engineering, recent research approaches often involve 3D printing or electrospinning of fibres to scaffolds as cell carrier material. Within this thesis, a micron fibre printing process, called melt electrospinning writing (MEW), combining both additive manufacturing and electrospinning, has been investigated and improved. Thus, a unique device was developed for accurate process control and manufacturing of high quality constructs. Thereby, different studies could be conducted in order to understand the electrohydrodynamic printing behaviour of different medically relevant thermoplastics as well as to characterise the influence of MEW on the resulting scaffold performance.
For reproducible scaffold printing, a commonly occurring processing instability was investigated and defined as pulsing, or in extreme cases as long beading. Here, processing analysis could be performed with the aim to overcome those instabilities and prevent the resulting manufacturing issues. Two different biocompatible polymers were utilised for this study: poly(ε-caprolactone) (PCL) as the only material available for MEW until then and poly(2-ethyl-2-oxazoline) for the first time. A hypothesis including the dependency of pulsing regarding involved mass flows regulated by the feeding pressure and the electrical field strength could be presented. Further, a guide via fibre diameter quantification was established to assess and accomplish high quality printing of scaffolds for subsequent research tasks.
By following a combined approach including small sized spinnerets, small flow rates and high field strengths, PCL fibres with submicron-sized fibre diameters (fØ = 817 ± 165 nm) were deposited to defined scaffolds. The resulting material characteristics could be investigated regarding molecular orientation and morphological aspects. Thereby, an alignment and isotropic crystallinity was observed that can be attributed to the distinct acceleration of the solidifying jet in the electrical field and by the collector uptake. Resulting submicron fibres formed accurate but mechanically sensitive structures requiring further preparation for a suitable use in cell biology. To overcome this handling issue, a coating procedure, by using hydrophilic and cross-linkable star-shaped molecules for preparing fibre adhesive but cell repellent collector surfaces, was used.
Printing PCL fibre patterns below the critical translation speed (CTS) revealed the opportunity to manufacture sinusoidal shaped fibres analogously to those observed using purely viscous fluids falling on a moving belt. No significant influence of the high voltage field during MEW processing could be observed on the buckling phenomenon. A study on the sinusoidal geometry revealed increasing peak-to-peak values and decreasing wavelengths as a function of decreasing collector speeds sc between CTS > sc ≥ 2/3 CTS independent of feeding pressures. Resulting scaffolds printed at 100 %, 90 %, 80 % and 70 % of CTS exhibited significantly different tensile properties, foremost regarding Young’s moduli (E = 42 ± 7 MPa to 173 ± 22 MPa at 1 – 3 % strain). As known from literature, a changed morphology and mechanical environment can impact cell performance substantially leading to a new opportunity of tailoring TE scaffolds.
Further, poly(L-lactide-co-ε-caprolactone-co-acryloyl carbonate) as well as poly(ε-caprolactone-co-acryloyl carbonate) (PCLAC) copolymers could be used for MEW printing. Those exhibit the opportunity for UV-initiated radical cross-linking in a post-processing step leading to significantly increased mechanical characteristics. Here, single fibres of the polymer composed of 90 mol.% CL and 10 mol.% AC showed a considerable maximum tensile strength of σmax = 53 ± 16 MPa. Furthermore, sinusoidal meanders made of PCLAC yielded a specific tensile stress-strain characteristic mimicking the qualitative behaviour of tendons or ligaments. Cell viability by L929 murine fibroblasts and live/dead staining with human mesenchymal stem cells revealed a promising biomaterial behaviour pointing out MEW printed PCLAC scaffolds as promising choice for medical repair of load-bearing soft tissue.
Indeed, one apparent drawback, the small throughput similar to other AM methods, may still prevent MEW’s industrial application yet. However, ongoing research focusses on enlargement of manufacturing speed with the clear perspective of relevant improvement. Thereby, the utilisation of large spinneret sizes may enable printing of high volume rates, while downsizing the resulting fibre diameter via electrical field and mechanical stretching by the collector uptake. Using this approach, limitations of FDM by small nozzle sizes could be overcome. Thinking visionary, such printing devices could be placed in hospitals for patient-specific printing-on-demand therapies one day. Taking the evolved high deposition precision combined with the unique small fibre diameter sizes into account, technical processing of high performance membranes, filters or functional surface finishes also stands to reason.
Die Terahertz-Technologie erschließt permanent neue Forschungsbereiche und wird auch vermehrt im kommerziellen Bereich eingesetzt. Diese Entwicklung wird in eigenen Analysen dargestellt. Als Kernaspekte dieses Buchs werden technologische Verbesserungen für gepulste Terahertz-Systeme und der spektroskopische Einsatz für die Polymeranalytik vorgestellt. Einen besonderen Stellenwert nehmen dabei Untersuchungen an Polymermischungen im Schmelzezustand während der Prozessierung ein. Bemerkenswert ist hierbei das temperaturabhängige Verhalten von verschiedensten Polyamiden. Die Änderung der optischen und spektroskopischen Eigenschaften und die Einflussfaktoren werden tiefergehend betrachtet. Dabei werden verschiedene klassische und neuere Experimentalmethoden, Modellierungen und quantenchemische Simulationsmethoden eingesetzt, um insbesondere die intermolekularen Wechselwirkungen aufzuklären.
Overcoming Obstacles in the Aqueous Processing of Nickel-rich Layered Oxide Cathode Materials

The implementation of a water-based cathode manufacturing process is attractive, given the prospect of improved sustainability of future lithium-ion batteries. However, the sensitivity of many cathode materials to water poses a huge challenge.
Within the scope of this work, a correlation between the water sensitivity of cathode materials from the class of layered oxides and their elemental composition was identified. In particular for the cathode material LiNi0.8Co0.15Al0.05O2 (NCA), the processes taking place in aqueous medium were clarified in detail. Based on this knowledge, the surface of NCA particles could be specifically modified, which led to a reduced water sensitivity. As a result, the electrochemical performance of cells with water-based NCA cathodes was significantly improved and a remarkable long-term cycling performance was achieved.
The present work contributes to a deeper understanding of the water sensitivity of cathode materials and at the same time presents a promising approach to overcome this obstacle. Consequently, this work advances the successful widespread realization of water-based cathode manufacturing.
Motivated by the great potential offered by the combination of additive manufacturing technology and hydrogels, especially in the field of tissue engineering and regenerative medicine, a series of novel hybrid hydrogel inks were developed based on the recently described thermogelling poly(2-oxazoline)s-block-poly(2-oxazine)s diblock copolymers, which may help to expand the platform of available hydrogel inks for this transformative 3D printing technology (Fig. 5.1).
In the present thesis, the first reported thermogelling polymer solely consisting of POx and POzi, i.e., the diblock copolymer PMeOx-b-PnPrOzi comprising a hydrophilic block (PMeOx) and a thermoresponsive block (PnPrOzi), was selected and used as a proof-of-concept for the preparation of three novel hybrid hydrogels. Therefore, three batches of the diblock copolymers with a DP of 100 were synthesized for the study of three different hybrid hydrogels with a special focus on their suitability as (bio)inks for extrusion-based 3D printing. The PMeOx-b-PnPrOzi diblock copolymer solution shows a temperature induced reversible gelation behavior above a critical polymer concentration of 20 wt%, as described for the Pluronic F127 solution but with a unique gelation mechanism, working through the formation of a bicontinuous sponge-like structure from the physically crosslinked vesicles. Specially, its intrinsic shear thinning behavior and excellent recovery property with a certain yield point make it a promising ink candidate for extrusion-based printing technology.
Increasing the polymer concentration is the most traditional approach to improve the printability of an ink material, and serve as the major strategy available to improve the printability of PMeOx-b-PnPrOzi systems prior to this work. From the analysis of rheological properties related to printability, it came a conclusion that increasing the copolymer concentration does improve the hydrogel strength and thus the printability. However, such improvement is very limited and usually leads to other problems such as more viscous systems and stringent requirements on the printers, which are not ideal for the printing process and applications especially in the cell-embedded biofabrication field.
POx-b-POzi/clay Hybrid Hydrogel
An alternative method proposed to improve the printability of this thermoresponsive hydrogel ink is through nanoclay (Laponite XLG) addition, i.e., the first hybrid hydrogel system of PMeOx-b-PnPrOzi/clay (also named shortly as POx-b-POzi/clay) in this thesis. To optimize the viscoelastic properties of the ink material, Laponite XLG acted as a reinforcement additive and a physically crosslinker was blended with the copolymers. Compared with the pristine copolymer solution of PMeOx-b-PnPrOzi, the hybrid PMeOx-b-PnPrOzi/clay solution well retained the temperature induced gelation performance of the copolymers.
The obtained hybrid hydrogels exhibited a rapid in situ reversible thermogelation at a physiological relevant Tgel of around 15 ℃ and a rapid recovery of viscoelastic properties within a few seconds. More importantly, with the addition of only a small amount of 1.2 wt% clay, it exhibited obviously enhanced shear thinning character (n = 0.02), yield stress (240 Pa) and mechanical strength (storage modulus over 5 kPa). With this novel hybrid hydrogel, real three-dimensional constructs with multiple layers and various geometries are generation with greatly enhanced shape fidelity and resolution. In this context, the thermogelling properties of the hybrid hydrogels over a copolymer concentration range of 10-20 wt% and a clay concentration of 0-4 wt% were systematically investigated, and from which a printable window was obtained from the laboratory as a reference.
In fact, the printing performance of an ink is not only determined by the intrinsic physicochemical properties of the material, but is also influenced by the external printing environments as well as the printer parameter settings. All the printing experiments in this study were conducted under a relatively optimized conditions obtained from preliminary experiments. In future work, the relationship between material rheology properties, printer parameters and printing performance could be systematically explored. Such a fundamental study will help to develop models that allows the prediction and comparison of printing results from different researches based on the parameters available through rheology, which is very beneficial for further development of more advanced ink systems.
Although the printability has been significantly improved by the addition of nanoclay Laponite XLG, the hybrid hydrogels and their printed constructs still suffer from some major limitations. For example, these materials are still thermoresponsive, which will cause the printed constructs to collapse when the environment temperature changes below their Tgel. In addition, the formed hydrogel constructs are mechanical too weak for load-bearing applications, and the allowed incubation time is very limited during media exchange/addition as it will lead to dissolution of the hydrogels due to dilution effects. Therefore, it is essential to establish a second (chemical or physical) crosslinking mechanism that allows further solidification of the gels after printing. It should be kept in mind that the second crosslinking step will eliminate the thermoresponsive behavior of the gels and thus the possibility of cell recovery. In this case, besides through the traditional approach of copolymer modification to realize further crosslinking, like one of the well-known post-polymerization modification approach Diels-Alder reaction,[430] designing of interpenetrating networks (IPN) hydrogels serves as one of the major strategy for advanced (bio)ink preparation.[311] Therefore, the second hybrid hydrogel system of PMeOx-b-PnPrOzi/PDMAA/clay (also named shortly as POx-b-POzi/PDMAA/clay) was developed in this thesis, which is a 3D printable and highly stretchable ternary organic-inorganic IPN hydrogel.
POx-b-POzi/PDMAA/clay Hybrid Hydrogel
The nanocomposite IPN hydrogel combines a thermoresponsive hydrogel with clay described above and in situ polymerized poly(N, N-dimethylacrylamide). Before in situ polymerization, the thermoresponsive hydrogel precursors exhibited thermogelling behavior (Tgel ~ 25 ℃, G' ~ 6 kPa) and shear thinning properties, making the system well-suited for extrusion-based 3D printing. After chemical curing of the 3D-printed constructs by free radical polymerization, the resulting IPN hydrogels show excellent mechanical strength with a high stretchability to a tensile strain at break exceeding 550%. The hybrid hydrogel can sustain a high stretching deformation and recover quickly due to the energy dissipation from the non-covalent interactions. With this hybrid hydrogel, integrating with the advanced 3D-printing technique, various 3D constructs can be printed and cured successfully with high shape fidelity and geometric accuracy.
In this context, we also investigated the possibility of acrylic acid (AA) and 2-hydroxyethylmethacrylate (HEMA) as alternative hydrogel precursors. However, the addition of these two monomers affected the thermogelation of POx-b-POzi in an unfavorable manner, as these monomers competed more effectively with water molecules, preventing the hydration of nPrOzi block at lower temperatures and therefore, the liquefaction of the gels. Furthermore, the influence of the printing process and direction on the mechanical properties of the hydrogel was investigated and compared with the corresponding bulk materials obtained from a mold. No significant effects from the additive manufacturing process were observed due to a homogeneously adhesion and merging between sequentially deposited layers. In the future, further studies on the specific performance differences among hydrogels fabricated at different printing directions/speeds would be of great interest to the community, as this allows for a more accurately control and better predict of the printed structures.
This newly developed hybrid IPN hydrogel is expected to expand the material toolbox available for hydrogel-based 3D printing, and may be interesting for a wide range of applications including tissue engineering, drug delivery, soft robotics, and additive manufacturing in general. However, in this case, the low toxicity from the monomer DMAA and other small molecules residuals in the polymerized hydrogels made this hybrid hydrogel not ideal for bioprinting in the field of biofabrication. For this problem, cyto-/biocompatible monomers such as polyethylene glycol diacrylate (PEGDA) can be used as an alternative, while the overall properties of the hydrogels including mechanical properties should be re-evaluated accordingly. Moreover, the swelling behavior of the hydrogels should also be taken into account, as it may most likely affect the mechanical strength and geometry size of the printed scaffold, but is often be overlooked after printing. For example, regarding the specific hybrid hydrogel POx-b-POzi/PDMAA/clay in this work, an equilibrium swelling ratio of 1100% was determined. The printed hydrogel cuboid experienced a volume increasing over 6-fold after equilibrium swelling in water, and became mechanical fragile due to the formation of a swollen hydrogel network absorbing large amount of water.
POx-b-POzi/Alg/clay Hybrid Hydrogel
In the final part of this dissertation, to enable the cell-loaded bioprinting and long-term cell culture, the third hybrid hydrogel system POx-b-POzi/Alg/clay was introduced by replacing the monomer DMAA to the natural polysaccharides alginate. Initially, detailed rheological characterization and mechanical tests were performed to evaluate their printability and mechanically properties. Subsequently, some simple patterns were printed with the optimized hydrogel precursor solutions for the preliminary filament fusion and collapse test before proceeding to more complex printings. The fibers showed a sufficient stability which allows the creation of large structures with a height of a few centimeters and a suspended filament up to centimeter. Accordingly, various 3D constructs including suspended filaments were printed successfully with high stackability and shape fidelity. The structure after extrusion was physical crosslinked easily by soaking in CaCl2 solution and, thereafter exhibited a good mechanical flexibility and long-term stability. Interestingly, the mechanical strength and geometry size of the generated scaffolds were well maintained over a culture period of weeks in water, which is of great importance for clinical applications. In addition, the post-printing ionic crosslinking of alginate could also be realized by other di/trivalent cations such as Fe3+ and Tb3+.
Subsequently, the cell-laden printing with this hybrid hydrogel and post-printing crosslinking by Ca2+ ions highlighting its feasibility for 3D bioprinting. WST-1 assay of fibroblast suggested no-dose dependent cytocompatibility of the hydrogel precursor solution. The cell distribution was uniform throughout the printed construct, and proliferated with high cell viability during the 21 days culture. The presented hybrid approach, utilizing the beneficial properties of the POx-b-POzi base material, could be interesting for a wide range of bioprinting applications and potentially enabling also other biological bioinks such as collagen, hyaluronic acid, decellularized extracellular matrix or cellulose based bioinks. Although the results look promising and the developed hydrogel is an important bioink candidate, the long-term in vitro cell studies with different cell lines and clinical model establishment are still under investigation, which remains a long road but is of great importance before realizing real clinical application.
Last but not least, the improvement to the printability of thermogelling POx/POzi-based copolymers by the clay Laponite XLG was also demonstrated in another thermogelling copolymer PEtOx-b-PnPrOzi. This suggests that the addition of clay may be a general strategy to improve the printability of such polymers. Despite these advances in this work which significantly extended the (bio)material platform of additive manufacturing technology, the competition is still fierce and more work should be done in the further to reveal the potential and limitations of this kind of new and promising candidate (bio)ink materials. It is also highly expected for further creative works based on the thermogelling POx/POzi polymers, such as crosslinking in Ca2+ solution containing monomer acrylamide to prepare printable and mechanically tough hydrogels, research on POx-based support bath material, and print of clinically more relevant sophisticated structures such as 3D microvascular networks omnidirectionally.
As a promising biofabrication technology, extrusion-based bioprinting has gained significant attention in the last decade and major advances have been made in the development of bioinks. However, suitable synthetic and stimuli-responsive bioinks are underrepresented in this context. In this work, we described a hybrid system of nanoclay Laponite XLG and thermoresponsive block copolymer poly(2-methyl-2-oxazoline)-b-poly(2-n-propyl-2-oxazine) (PMeOx-b-PnPrOzi) as a novel biomaterial ink and discussed its critical properties relevant for extrusion-based bioprinting, including viscoelastic properties and printability. The hybrid hydrogel retains the thermogelling properties but is strengthened by the added clay (over 5 kPa of storage modulus and 240 Pa of yield stress). Importantly, the shear-thinning character is further enhanced, which, in combination with very rapid viscosity recovery (~ 1 s) and structure recovery (~ 10 s), is highly beneficial for extrusion-based 3D printing. Accordingly, various 3D patterns could be printed with markedly enhanced resolution and shape fidelity compared to the biomaterial ink without added clay.
Melt electrowriting, a high-resolution additive manufacturing technique, is used in this study to process a magnetic polymer-based blend for the first time. Carbonyl iron (CI) particles homogenously distribute into poly(vinylidene fluoride) (PVDF) melts to result in well-defined, highly porous structures or scaffolds comprised of fibers ranging from 30 to 50 µm in diameter. This study observes that CI particle incorporation is possible up to 30 wt% without nozzle clogging, albeit that the highest concentration results in heterogeneous fiber morphologies. In contrast, the direct writing of homogeneous PVDF fibers with up to 15 wt% CI is possible. The fibers can be readily displaced using magnets at concentrations of 1 wt% and above. Combined with good viability of L929 CC1 cells using Live/Dead imaging on scaffolds for all CI concentrations indicates that these formulations have potential for the usage in stimuli-responsive applications such as 4D printing.
Die Wandmaterialien innerhalb des Plasmagefäßes zukünftiger Fusionsreaktoren sind teilweise extremen thermischen und mechanischen Belastungen ausgesetzt. Der thermisch höchstbelastete Bereich der Wand des Torusgefäßes ist der Divertor. Hier werden die anfallende Fusionsasche (Helium) und erodierte Wandpartikel aus dem Plasma entfernt, wodurch aufgrund erhöhter Teilchen-Wand-Interaktion Wärmeflüsse von bis zu 15 MW/m² erreicht werden. Wolfram gilt momentan als ideales Wandmaterial mit direktem Plasmakontakt (Plasma-Facing-Material, PFM) für diese Beanspruchungen. Unterhalb des PFM muss die Wärme möglichst effizient in das Kühlmedium übertragen werden. Im zukünftigen Experimentalreaktor ITER wird dafür eine Kupferlegierung (CuCrZr) verwendet, welche eine hohe Wärmeleitfähigkeit besitzt und für Temperaturen von bis zu 350°C unter fusionsrelevanten Bedingungen einsetzbar ist. In dieser Konfiguration kann ITER mit einer Kühlmitteltemperatur von 150°C betrieben werden. Zur kommerziellen Energiegewinnung ist dies unzureichend, da die thermische Effizienz durch eine deutliche Anhebung der Kühlmitteltemperatur verbessert werden muss. Wird der konventionelle Ansatz einer Wasserkühlung zu Grunde gelegt, ist das Ziel die Kühlmitteltemperatur auf mindestens 300°C anzuheben. In der Folge ist CuCrZr als Wärmesenkenmaterial nicht mehr einsetzbar, da verstärkte Alterung und Festigkeitsverlust im Material auftritt. Zusätzlich vergrößern sich die thermisch induzierten Spannungen in der Komponente mit höheren Temperaturen, durch unterschiedlich große thermische Ausdehnungskoeffizienten der beteiligten Materialien. Für höhere Temperaturen stellt faserverstärktes Kupfer eine mögliche Alternative dar. Die Kombination der hohen Wärmeleitfähigkeit der Kupfermatrix mit der hohen Steifigkeit und Festigkeit von Siliziumcarbidfasern soll die nötigen thermischen und mechanischen Eigenschaften des Wärmesenkenmaterials auch für Temperaturen über 350°C gewährleisten. Im Rahmen dieser Arbeit wurden zwei unterschiedlich hergestellte SiC-Verstärkungsfasertypen hinsichtlich ihrer Eignung für die Herstellung eines Kupfer-Matrix-Komposits (CuMMC) untersucht. Die Zielstellung für das CuMMC beinhaltet eine Festigkeit von 300 MPa bei 300°C sowie eine möglichst hohe Wärmeleitfähigkeit von über 200 W m-1 K-1. Beide Parameter werden stark von der Faserfestigkeit und der Anbindung zwischen Faser und Matrix beeinflusst. Die Wärmeleitfähigkeit durch das CuMMC wird von der Kupfermatrix dominiert, wodurch geringere Faservolumenanteile von Vorteil sind. Höhere Faserfestigkeit erfordert geringere Faseranteile zum Erreichen mechanischer Vorgaben, womit die erzielbare Wärmeleitfähigkeit des CuMMCs steigt. Die Faserfestigkeit wird durch Einzel-Faser- Zugversuche validiert. Darüber hinaus ist die Anbindung zwischen Faser und Matrix essentiell, um die optimale Verstärkungswirkung durch die Fasern im CuMMC zu erzielen. Zur Faser-Matrix-Anbindung werden für jeden Fasertyp unterschiedliche Zwischenschichtsysteme verwendet, die anschließend durch Einzelfaser-Push-Out-Versuche validiert werden. Sind die Voraussetzungen von Faserfestigkeit und Anbindung für einen Fasertyp erfüllt, wird dieser für die Herstellung eines unidirektional verstärkten CuMMCs verwendet, welches bezüglich seiner mechanischen und thermischen Eigenschaften charakterisiert wird. Die mechanische Charakterisierung des CuMMCs erfolgt durch Zugversuche und dehnungsgeregelte, zyklische Versuche, wobei der Fokus neben der Festigkeit auf der Plastifizierung, Verfestigung und Schädigung innerhalb des CuMMCs liegt. Die thermische Charakterisierung erfolgt anhand der Wärmeleitfähigkeitsbestimmung sowohl parallel, als auch transversal zur Faserrichtung. Die mechanischen und thermischen Eigenschaften werden in Abhängigkeit von Faservolumenanteil und Temperatur untersucht. Um den Einfluss von längeren Betriebsphasen unter hoher thermischer Belastung analysieren zu können, wird das CuMMC bei 550°C für 400 h ausgelagert und anschließend wiederum mittels Vergleich seiner mechanischen und thermischen Eigenschaften auf mögliche Schädigungen untersucht. Zur Begutachtung von Schliff- und Bruchflächen zur Schadensanalyse stehen als bildgebende Untersuchungsmethoden neben Lichtmikroskopen ebenso Rasterelektronenmikroskope (REM) zur Verfügung.
In der vorliegenden Arbeit wurden neue Materialien für die additive Fertigung für opti-sche Anwendungen entwickelt. Hierbei wurde zunächst ein ORMOCER® Harz für den LCD/DLP 3D-Druck synthetisiert und charakterisiert. Das Material zeigte eine gute Druckbarkeit, gute optische Eigenschaften und eine hohe Stabilität gegenüber Belas-tungen mit UV-Licht, Temperatur und Luftfeuchtigkeit. Die prozessbedingte Stufenbil-dung für gekrümmte Oberflächen beim LDC/DLP Druck erforderte eine Weiterentwick-lung zu einem Harz, das auch mittels Inkjet-Verfahren gedruckt werden kann. Hierfür mussten die Viskosität des ORMOCER®s und die Einflüsse darauf untersucht werden. Zu diesem Zweck wurde die Synthese entsprechend verändert und die Produkte cha-rakterisiert. Variationen des Wasseranteils, des Katalysators, der Reaktionszeit, der Re-aktionsführung und der Edukte wurden durchgeführt. Harze mit resultierender niedriger Viskosität dürfen zusammenfassend nur zweifach anorganisch vernetzende Edukte mit niedrigem Reibungskoeffizienten beinhalten. Ein H2O-Verhältnis von 0,5 zu den vorlie-genden Si-O-Gruppen resultiert in akzeptablen Viskositäten und einer ausreichenden Stabilität. Als zuverlässiger Katalysator stellte sich HCl heraus. Die Reaktionszeit muss so gewählt werden, dass die Sol-Gel-Synthese abgeschlossen ist. Kürzere Zeiten füh-ren zwar zu kleineren Viskositäten, jedoch auch zu eventuell schlechter Langzeitstabili-tät. Veränderungen in der Reaktionsführung, durch Zutropfen der Edukte, resultierten jedoch vorwiegend zur Erhöhung der Viskositäten. Mit diesen Erkenntnissen wurde an-schließend ein Harz synthetisiert, das erfolgreich ohne weitere Verdünnungsschritte am Inkjet-Drucker prozessiert werden konnte. Dieses Harz ist zusätzlich auch am LCD/DLP Drucker einsetzbar.
Als ergänzender Schritt konnte im Anschluss noch gezeigt werden, dass Partikel in Harze zusätzliche Eigenschaften, wie Fluoreszenz, einbringen können.
Thermoplastische Kunststoffe (sog. Thermoplaste) lassen sich in einem be-stimmten Temperaturbereich beliebig oft schmelzen und in einer gewünschten Form erstarren. Grundvoraussetzung für eine bestimmte Anwendung eines thermoplastischen Bauteils sind die Gebrauchseigenschaften des Materials, die im Wesentlichen vom Ablauf der Erstarrung abhängen.
Die Moleküle einiger Thermoplaste können bei der Erstarrung geordnete kristalline Bereiche bilden. Dies sind die sog. teilkristallinen Kunststoffe, deren Erstarrungsprozess Kristallisation genannt wird. Die dabei entstehenden Kristallstrukturen werden zusammen mit deren Charakteristiken allgemein als Morphologie der teilkristallinen Kunststoffe bezeichnet. Die Morphologie hat einen signifikanten Einfluss auf die mechanischen, thermischen und optischen Eigenschaften des Materials. Dementsprechend stellen Kenntnisse über die Kristallisation eine wertvolle Hilfe bei der Vorhersage der Gebrauchseigenschaften eines teilkristallinen Kunststoffs dar.
Um die Kristallisation zu starten, muss zunächst eine Energiebarriere überwunden werden, die an erster Stelle vom molekularen Aufbau des Kunststoffs abhängt. Somit weisen beispielsweise Kunststoffe mit linearen, regelmäßigen Molekülen und kleinen Seitengruppen eine niedrigere Energiebarriere und aus diesem Grund eine starke Neigung zur Kristallisation auf. Einige Zusatzstoffe wie z. B. unterschiedliche Additive, Farbstoffe oder Füllstoffe können die Energiebarriere und infolgedessen die Kristallisation eines teilkristallinen Kunststoffs wesentlich beeinflussen.
Das Ziel dieser Dissertation war es, ein bestehendes Kristallisationsmodell zu erweitern und es an gefüllte oder additivmodifizierte teilkristalline Kunststoffe anzupassen. Das erweiterte Modell soll die Ermittlung eines Kristallisationspa-rameters, des sog. Keimbildungsexponenten, eines gefüllten oder additivmodifizierten teilkristallinen Kunststoffs bei der nicht-isothermen Kristallisation ermöglichen. Der Keimbildungsexponent ist mit der erwähnten Energiebarriere eng verbunden und bestimmt somit den Ablauf des Kristallisationsprozesses bzw. die daraus folgende Morphologie. Ein wesentlicher Schwerpunkt der Arbeit lag darin, die vorgeschlagene Modellerweiterung bei verschiedenen Abkühlgeschwindigkeiten zu überprüfen. Im Anschluss sollten die Beziehungen zwischen den berechneten Keimbildungsexponenten und experimentell ermittelten me-chanischen Eigenschaften (E-Modul, Streckspannung und Schlagzähigkeit) überprüft werden. Für die Untersuchungen wurden drei verschiedene Polymersysteme verwendet: PP / Talkum, HDPE / Talkum sowie PA6 / Bentonit. Hierbei weist der Füllstoff eine stark positive, schwach positive bzw. inhibierende Wirkung auf die Kristallisation der entsprechenden Polymermatrix auf.
Hinsichtlich reiner Polymere wurde eine gute Übereinstimmung zwischen den ermittelten und Literaturwerten des Keimbildungsexponenten festgestellt. Die Zugabe von positiv wirkendem Talkum in PP bzw. HDPE führt zu einer Abnah-me des Keimbildungsexponenten, was zu dickeren Kristallen des jeweiligen Kunststoffs führte. Im Gegensatz dazu bewirkte die Bentonitzugabe einen zu-nehmenden Keimbildungsexponenten, was anschließend dünnere PA6-Kristalle zur Folge hat. Die durchgeführten Untersuchungen zeigen außerdem, dass die Füllstoffpartikelgröße einen ausgeprägten Einfluss auf den ermittelten Keimbildungsexponenten hat.
Weiterhin wurde festgestellt, dass der ermittelte Keimbildungsexponent durch die (DSC)-Abkühlgeschwindigkeit beeinflusst wird. Es wurde ferner gezeigt, dass sich dieser Einfluss ab einer bestimmten Abkühlgeschwindigkeit (20 K/min im Falle des PP und HDPE bzw. 15 K/min im Falle des PA6) nicht mehr ändert, was zu einem konstanten Keimbildungsexponenten führt. Um den Einfluss der Abkühlgeschwindigkeit auf die modellierte Größe zu berücksichtigen, sind weitere Untersuchungen nötig.
Die Ergebnisse der Arbeit zeigen weiterhin, dass der berechnete Keimbildungsexponent mit den experimentell ermittelten Werten für E-Modul, Streckspannung und Charpy-Schlagzähigkeit bei talkumgefülltem PP gut korreliert. Solche Korrelationen wurden jedoch bei den HDPE- und PA6-Proben nicht gefunden. Der Grund hierfür könnte eine ausgeprägte Orientierung der HDPE-Makromoleküle bzw. ein starker mikromechanischer Effekt des exfolierten Bentonits sein. Diese Effekte konnten im Rahmen der Arbeiten bestätigt werden.
Die in dieser Arbeit erzielten Ergebnisse zeigen, dass die vorgeschlagene Mo-dellerweiterung auch bei gefüllten oder additivmodifizierten Kunststoffen zufriedenstellende Resultate liefert. Die entsprechende Berechnung erfordert dabei lediglich eine DSC-Messung, was im Vergleich zum Stand der Technik in einen niedrigeren Messaufwand resultiert. Die vorliegende Arbeit liefert daher einen signifikanten Beitrag zur Erstellung des Zusammenhangs zwischen der Kristallisation, der Morphologie und dem mechanischen Verhalten von teilkristallinen Polymeren.
Im Rahmen der vorliegenden Dissertation wurden ORMOCER®-basierte Materialsysteme für dentale Versorgungen entwickelt, die additiv mittels Digital Light Processing (DLP) verarbeitbar sind und ein hochwertiges, auf die vorgesehene Zielanwendung abgestimmtes Eigenschaftsprofil besitzen. Zunächst wurden grundlegende Untersuchungen zum DLP-Druck des Harzsystems und einfachen Kompositen durchgeführt, um auftretende Herausforderungen zu identifizieren und die weitere Vorgehensweise festzulegen. Ausgehend davon konzentrierte sich die Arbeit neben der Vermeidung der klebrigen Sauerstoffinhibierungsschicht auf der Bauteiloberfläche einerseits darauf, die Maßhaltigkeit bei DLP-gedruckten Bauteilen mit überhängenden Strukturen zu steigern. Insbesondere wurde das Augenmerk hier auf die Verwendung von organischen Lichtabsorbern zur Realisierung von hochtransluzenten Harz-basierten Bauteilen gelegt. Andererseits lag ein weiterer Schwerpunkt der Arbeit auf der Entwicklung von DLP-druckbaren Kompositen mit hoher Transluzenz. Die dafür nötige Brechzahlanpassung von Harzsystem und Füllstoff wurde zum einen durch die Synthese neuer, höherbrechender Harzsysteme und zum anderen durch die Verwendung hochbrechender ZrO2-Nanopartikel realisiert. Die resultierenden hochtransluzenten Komposite wurden umfassend mechanisch charakterisiert sowie erfolgreich DLP-gedruckt.
Motivated by the great potential which is offered by the combination of additive manufacturing and tissue engineering, a novel polymeric bioink platform based on poly(2 oxazoline)s was developed which might help to further advance the young and upcoming field of biofabrication. In the present thesis, the synthesis as well as the characteristics of several diblock copolymers consisting of POx and POzi have been investigated with a special focus on their suitability as bioinks.
In general, the copolymerization of 2-oxazolines and 2-oxazines bearing different alkyl side chains was demonstrated to yield polymers in good agreement with the degree of polymerization aimed for and moderate to low dispersities.
For every diblock copolymer synthesized during the present study, a more or less pronounced dependency of the dynamic viscosity on temperature could be demonstrated. Diblock copolymers comprising a hydrophilic PMeOx block and a thermoresponsive PnPrOzi block showed temperature induced gelation above a degree of polymerization of 50 and a polymer concentration of 20 wt%. Such a behavior has never been described before for copolymers solely consisting of poly(cyclic imino ether)s.
Physically cross linked hydrogels based on POx b POzi copolymers exhibit reverse thermal gelation properties like described for solutions of PNiPAAm and Pluronic F127. However, by applying SANS, DLS, and SLS it could be demonstrated that the underlying gel formation mechanism is different for POx b POzi based hydrogels. It appears that polymersomes with low polydispersity are formed already at very low polymer concentrations of 6 mg/L. Increasing the polymer concentration resulted in the formation of a bicontinuous sponge like structure which might be formed due to the merger of several vesicles. For longer polymer chains a phase transition into a gyroid structure was postulated and corresponds well with the observed rheological data.
Stable hydrogels with an unusually high mechanical strength (G’ ~ 4 kPa) have been formed above TGel which could be adjusted over a range of 20 °C by changing the degree of polymerization if maintaining the symmetric polymer architecture. Variations of the chain ends revealed only a minor influence on TGel whereas the influence of the solvent should not be neglected as shown by a comparison of cell culture medium and MilliQ water.
Rotationally as well as oscillatory rheological measurements revealed a high suitability for printing as POx b POzi based hydrogels exhibit strong shear thinning behavior in combination with outstanding recovery properties after high shear stress.
Cell viability assays (WST-1) of PMeOx b PnPrOzi copolymers against NIH 3T3 fibroblasts and HaCat cells indicated that the polymers were well tolerated by the cells as no dose-dependent cytotoxicity could be observed after 24 h at non-gelling concentrations up to 100 g/L.
In summary, copolymers consisting of POx and POzi significantly increased the accessible range of properties of POx based materials. In particular thermogelation of aqueous solutions of diblock copolymers comprising PMeOx and PnPrOzi was never described before for any copolymer consisting solely of POx or POzi. In combination with other characteristics, e.g. very good cytocompatibility at high polymer concentrations and comparably high mechanical strength, the formed hydrogels could be successfully used for 3D bioprinting. Although the results appear promising and the developed hydrogel is a serious bioink candidate, competition is tough and it remains an open question which system or systems will be used in the future.
Aluminium-Kupfer-Legierungen des Typs 2xxx erhalten ihre Festigkeit während der Auslagerung an Raumtemperatur oder erhöhter Temperatur – auch Alterung genannt – durch die Bildung kupferhaltiger Ausscheidungen. Größe, räumliche Verteilung und Kristallstruktur dieser Ausscheidungen sind maßgeblich für die mechanischen Eigenschaften dieser Legierungen. Die Zugabe geringer Mengen (100 ppm) an Elementen wie In oder Sn kann das Ausscheidungsverhalten und damit auch die mechanischen Eigenschaften des Systems stark verändern. Die vorliegende Arbeit beschäftigt sich mit dem Einfluss dieser Spurenelemente auf die Ausscheidungsbildung in Al-Cu(-Mg)-Legierungen und der damit verbundenen Veränderung der Festigkeit. Hauptaugenmerk liegt dabei auf der in-situ Charakterisierung der vorherrschenden Ausscheidungen und deren Kinetik, welche durch die Interaktion der Spurenelemente mit den eingeschreckten Leerstellen signifikant beeinflusst wird. Ziel ist es, ein fundamentales Verständnis der zugrundeliegenden Mechanismen zu erlangen. Durch den Einsatz vieler komplementärer Methoden wie Dynamischer Differenzkalorimetrie (DSC), in-situ Röntgenabsorptionsspektroskopie (XAFS) und in-situ Röntgenkleinwinkelstreuung (SAXS) kann ein umfassendes Bild der vorherrschenden Prozesse gewonnen werden. Als unterstützende Messmethoden dienen Transmissionselektronenmikroskopie (TEM) und Positronenlebensdauerspektroskopie (PALS) bzw. Dopplerspektroskopie (DBS). Um die Eigenschaften der Spurenelemente in Bezug auf Löslichkeit und Wechselwirkung mit Leerstellen in der Aluminiummatrix zu charakterisieren, werden zunächst binäre Aluminium- Spurenelement-Legierungen untersucht. Die ausgewählten Elemente (Bi, In, Pb, Sb, Sn) weisen alle eine, nach ab-intio Rechnungen, hohe Bindungsenergie zu Leerstellen auf. Man stellt nur bei In und Sn eine signifikante Bindung zu Leerstellen fest, da aufgrund der geringen Löslichkeiten von Bi, Pb und Sb keine für den Nachweis ausreichende Anzahl an Leerstellen gebunden werden. Die gelösten In- und Sn-Atome bilden mit den eingeschreckten Leerstellen sogenannte Komplexe, welche thermisch bis ca. 150 � C stabil sind. Die so konservierten Leerstellen können dann in einem komplexen Prozess durch Erwärmen der Legierung freigesetzt werden und tragen dadurch zur Diffusion bei. Grundsätzlich ist der Transport von Legierungsatomen in Aluminiumlegierungen weitestgehend durch die verfügbaren thermischen sowie eingeschreckten Leerstellen gesteuert. Durch die Bindung der Leerstellen an die Spurenelementatome in den ternären Al-Cu-X-Legierungen stehen nur noch wenige Leerstellen für den Cu-Transport zur Verfügung und man beobachtet eine Unterdrückung des Entmischungsprozesses bei Raumtemperatur. Lagert man die Legierungen bei erhöhter Temperatur (z.B. 150 � C) aus, werden Leerstellen aus den Spurenelement- Leerstellen-Komplexen freigesetzt, was zu einer Bildung der metastabilen q0-Phase bei deutlich erniedrigter Temperatur führt. Die kleinen und homogen verteilten Ausscheidungen steigern die Festigkeit der Legierung deutlich. Durch die Zugabe von Mg zum Al-Cu-Legierungssystem beschleunigt und verstärkt man die Entmischung der Legierungsatome bei Raumtemperatur. Ursache ist die rasche Bildung von thermisch stabilen Cu-Mg-Clustern, die für eine effektive Blockierung der Versetzungsbewegung im Kristallgitter – und damit für eine Erhöhung der Festigkeit – sorgen. Die Zugabe von In oder Sn zu einer Al-Cu-Mg-Legierung hat überraschenderweise keinen Einfluss auf die Ausscheidungsbildung bei Raumtemperatur. Gründe hierfür könnten zum einen in der durch Mg herabgesetzten Löslichkeit der In- bzw. Sn-Atome liegen. Zum anderen könnte die zahlreiche Bildung von Cu-Mg-Leerstellen-Komplexen in Konkurrenz zum Bindungseffekt der Spurenelementatome stehen. Auch bei erhöhter Temperatur lässt sich kein nennenswerter Einfluss von In oder Sn, beispielsweise durch bevorzugte Bildung der härtenden S-Phase, beobachten.
A series of polypeptoid homopolymers bearing short (C1–C5) side chains of degrees of polymerization of 10–100 are studied with respect to thermal stability, glass transition and melting points. Thermogravimetric analysis of polypeptoids suggests stability to >200 °C. The study of the glass transition temperatures by differential scanning calorimetry revealed two dependencies. On the one hand an extension of the side chain by constant degree of polymerization decrease the glass transition temperatures (Tg) and on the other hand a raise of the degree of polymerization by constant side chain length leads to an increase of the Tg to a constant value. Melting points were observed for polypeptoids with a side chain comprising not less than three methyl carbon atoms. X-ray diffraction of polysarcosine and poly(N-ethylglycine) corroborates the observed lack of melting points and thus, their amorphous nature. Diffractograms of the other investigated polypeptoids imply that crystalline domains exist in the polymer powder.
Polymer micelles are an attractive means to solubilize water insoluble compounds such as drugs. Drug loading, formulations stability and control over drug release are crucial factors for drug‐loaded polymer micelles. The interactions between the polymeric host and the guest molecules are considered critical to control these factors but typically barely understood. Here, we compare two isomeric polymer micelles, one of which enables ultra‐high curcumin loading exceeding 50 wt.%, while the other allows a drug loading of only 25 wt.%. In the low capacity micelles, steady‐state fluorescence revealed a very unusual feature of curcumin fluorescence, a high energy emission at 510 nm. Time‐resolved fluorescence upconversion showed that the fluorescence life time of the corresponding species is too short in the high‐capacity micelles, preventing an observable emission in steady‐state. Therefore, contrary to common perception, stronger interactions between host and guest can be detrimental to the drug loading in polymer micelles.
Structure-property relationships in poly(2-oxazoline)/poly(2-oxazine) based drug formulations

According to estimates, more than 40% of all new chemical entities developed in pharmaceutical industry are practically insoluble in water. Naturally, the demand for excipients which increase the water solubility and thus, the bioavailability of such hydrophobic drugs is enormous. Poly(2-oxazoline)s (POx) are currently intensively discussed as highly versatile class of biomaterials. Although selected POx based micellar drug formulations exhibit extraordinarily high drug loadings > 50 wt.% enabling high anti-tumor efficacies in vivo, the formulation of other hydrophobic compounds has failed. This casts doubt on the general understanding in which a hydrophobic active pharmaceutical ingredient is dissolved rather unspecifically in the hydrophobic core of the micelles following the fundamental concept of “like dissolves like”. Therefore, a closer look at the interactions between all components within a formulation becomes increasingly important. To do so, a large vehicle platform was synthesized, loaded with various hydrophobic drugs of different structure, and the formulations subsequently characterized with conventional and less conventional techniques. The obtained in-depth insights helped to develop a more thorough understanding about the interaction of polymer and incorporated API finally revealing morphologies deviating from a classical core/shell structure. During these studies, the scarcely investigated polymer class of poly(2-oxazine)s (POzi) was found as promising drug-delivery vehicle for hydrophobic drugs. Apart from this fundamental research, the anti-tumor efficacy of the two APIs curcumin and atorvastatin has been studied in more detail. To increase the scope of POx and POzi based formulations designed for intravenous administration, a curcumin loaded hydrogel was developed as injectable drug-depot.
