Phytoprostane F1

Isoprostane F2 sind Autoxidationsprodukte der Arachidonsäure, die über radikal-katalysierte Oxidation entstehen. Bei einigen Erkrankungen im Tier konnte gezeigt werden, daß die Konzentration von Isoprostanen F2 mit dem verstärkten Vorkommen von freien Radikalen korreliert, weshalb Isoprostane heute als Marker des oxidativen Streß genutzt werden. Darüber hinaus weisen einige Isoprostane eine biologische Aktivität auf, weshalb sie heute als Signalstoffe des oxidativen Streß im Tier diskutiert werden. Pflanzen hingegen können keine Isoprostane F2 synthetisieren, da ihnen der Precursor Arachidonsäure fehlt. In der vorliegenden Arbeit wird gezeigt, daß analog zu den Isoprostanen F2 Phytoprostane F1 in Pflanzen aus Linolensäure gebildet werden. Hierfür wurden HPLC- und Gaschromatographie-Massenspektroskopie-Methoden entwickelt, die eine Quantifizierung von Phytoprostanen F1 in Pflanzen ermöglichten. In frischen Pflanzenorganen wurden Phytoprostane F1 sowohl in freier als auch in veresterter Form detektiert. Darüber hinaus stieg die Konzentration sowohl freier als auch veresterter Phytoprostane F1 in Pfefferminzblättern nach Verwundung und in pflanzlichen Zellkulturen nach Zusatz von Agentien, von denen bekannt ist, daß sie pflanzliche Zellen oxidativ schädigen, an. In getrockneten Pflanzenmaterialien wurden extrem hohe Konzentrationen an Phytoprostanen F1 quantifiziert. Daher steht die Vermutung nahe, daß Phytoprostane F1 ähnlich wie die Isoprostane F2 im Tier als sensitiver Marker der oxidativen Zellverletzung in der Pflanze eingesetzt werden können. Darüber hinaus konnte gezeigt werden, daß der Zusatz von Phytoprostanen F1 zu Eschscholzia californica-, Crotalaria cobalticola- and Thalictrum tuberosum-Zellsuspensionskulturen zu einer Phytoalexinakkumulation führte.
Mit den Experimenten dieser Arbeit konnte erstmals gezeigt werden, dass ein Phytohormon wie Abscisinsäure mit dem "Solvent-drag" des Wasserflusses apoplastisch durch den Wurzelzellwandbereich in die Xylemgefäße transportiert werden kann. Es konnte ein Bypass-Fluss für ABA durch den gesamten Zellwandapoplasten, auch durch lipophile Barrieren wie Exo- und Endodermis nachgewiesen werden. Dies ist durch die speziellen Moleküleigenschaften von Abscisinsäure möglich: (i) der geringe Durchmesser des Moleküls (8 - 11 nm) und (ii) die hohe Lipophilie von ABA bei schwach sauren pH-Werte. Mit einer Penetration apoplastischer Barrieren ist demnach zu rechnen. Weiterhin wurde gezeigt, dass die Ausbildung solcher lipophilen Zellwandnetze einen signifikanten Einfluss auf den apoplastischen ABA-Transport besitzt. Die Ausbildung einer Exodermis in Mais, wie sie unter natürlichen Bedingungen zu beobachten ist, konnte den ABA-Fluss in das Xylem um die Faktoren 2 bis 4 reduzieren. Da gleichzeitig eine Verminderung der hydraulischen Wurzelleitfähigkeit um denselben Betrag auftrat, blieb das Wurzel-Spross-ABA-Signal, die Phytohormonkonzentration, im Xylem gleich. Die zu den Stomata geleitete Information über den Wasserzustand der Wurzel änderte sich also nicht. Im natürlichen System ist sogar eine Verstärkung des Signals zu erwarten, da eine Exodermis nicht als Aufnahme-Barriere für gewebeproduzierte ABA wirkt. Gleichzeitig verringert sie den Verlust von apoplastischer ABA an die Rhizosphäre. Außerdem wird der Wasserverlust aus dem Gewebe durch eine Exodermis signifikant reduziert wird. Somit sind solche Wurzeln gut an die Bedingungen eines eintrocknenden Bodens angepasst. Apoplastische Barrieren sind demnach, neben membran-lokalisierten Tranportern, wichtige Parameter für die Beurteilung von Wurzeltransporteigenschaften für Wasser und darin gelöste Substanzen. Der Beitrag der apoplastischen Komponente zum Gesamt-ABA-Transport ist abhängig von der untersuchten Pflanzenart, der aktuellen Transpirations- oder Wasserflussrate und von Umwelteinflüssen wie erhöhter ABA-Konzentration im Wurzelgewebe (z.B. durch Trockenstress), pH-Wert der Rhizosphäre und den Ernährungsbedingungen der Pflanze. Erhöhter radialer Wasserfluss, erhöhte ABA-Wurzelgewebegehalte und niedriger pH-Wert der Rhizosphäre verstärken den apoplastischen Bypass-Fluss unter physiologischen Bedingungen. Geringe Wassertransportraten, niedrige ABA-Konzentrationen im Gewebe, alkalische pH-Werte der Rhizosphäre und Ammoniumernährung verstärken dagegen den symplastischen Beitrag zum ABA-Transport. In der vorliegenden Arbeit konnten die sich widersprechenden Theorien bezüglich des ABA-Effektes auf die hydraulische Leitfähigkeit von Wurzeln erklärt werden. ABA erhöht über einen Zeitraum von 2 Stunden die Zellleitfähigkeit (Lp) mit einem Maximum 1 Stunde nach ABA-Inkubation. Dies wirkt sich in einem verstärktem Lpr von intakten Wurzelsystemen aus, das einem ähnlichen Zeitmuster folgt. Pflanzen sind demnach in der Lage, mittels ABA den zellulären Wassertransportweg reversibel zu optimieren, um so unter mildem Trockenstress, wie er in einem gerade eintrocknenden Boden auftritt, die Pflanze mit ausreichend Wasser zu versorgen. Tritt ein länger andauernder Wassermangel ein, versperrt die Pflanze diesen Weg wieder. Dieser transiente Effekt erklärt auch die aus der Literatur bekannten stimulierenden und inhibierenden ABA-Wirkungen. Durch den verstärkten Wasserfluss zu Beginn der Stresssituation erzeugt ABA auf diese Weise ein sich selbst verstärkendes, wurzelbürtiges Hormonsignal in den Spross. Das Blatt erreicht in effektiver Weise eine ABA-Menge, die ausreichend ist, um die Stomata zu schließen. Es folgt eine Reduktion der Transpiration. Eine weiter andauernde Erhöhung des symplastischen Wassertransportweges wäre ohne physiologische Bedeutung. Regulierende Membranstrukturen für diesen Vorgang könnten ABA-sensitive Wasserkanäle (Aquaporine) der Plasmamembran sein. Es wurde gezeigt, dass der Rezeptor für diesen Vorgang innerhalb von corticalen Maiswurzelzellen lokalisiert und hochspezifisch für (+)-cis-trans-ABA ist. Die Signaltransduktion für diesen Kurzzeiteffekt erfolgt nicht mittels verstärkter Aquaporintranskription, könnte aber über ABA-induzierte Aktivierung (Phosphorylierung), oder Einbau von Aquaporinen in die Zellmembran ablaufen. Der Abscisinsäure-Transport ist ein komplexer Vorgang. Er wird beeinflusst durch Umwelteinflüsse, Wurzelanatomie, ist gekoppelt mit dem Wasserfluss und durch sich selbst variierbar. Herkömmliche Vorstellungen einer simplen Hormondiffusion können diesen regulierbaren Vorgang nicht mehr beschreiben. Pflanzen besitzen ein ABA-Transportsystem, das schnell, effektiv und an sich verändernde Umweltbedingungen adaptierbar ist.
Aufgrund seiner potentiell gesundheitsfoerdernden Wirkung wurde das Falvonol Quercetin in den letzten Jahren intensiv untersucht. Daten zur Bioverfuegbarkeit nach oraler Applikation sind jedoch selten und widerspruechlich. Fruehere Untersuchungen deuteten darauf hin, dass die Disposition von Quercetin von der Zuckerkomponente des Glykosids oder der Pflanzenmatrix abhaengen koennte. Um den Einfluss der Zuckerkomponente oder der Matrix auf die Resorption von Quercetin festzustellen, wurden zwei isolierte Quercetinglykoside sowie zwei Pflanzenextrakte in einer vierarmigen, randomisierten cross-over Studie an 12 gesunden Probanden getestet. Jeder Proband erhielt eine Zwiebelzubereitung oder Quercetin-4'-O-glucosid, jeweils entsprechend 100 mg Quercetinaglykon, sowie Quercetin-3-O-rutinosid oder Buchweizenkrauttee entsprechend 200 mg Quercetinaglykon. Die Proben wurden mittels HPLC und Coulometrischer Arraydetektion analysiert. Im Plasma wurden ausschliesslich Quercetinglucuronide detektiert. Freies Quercetin und die Glykoside waren nicht nachweisbar. Die Bioverfuegbarkeit und Pharmakokinetik nach Applikation von Zwiebeln und Quercetin-4'-glucosid zeigte keine signifikanten Unterschiede. Maximale Plasmakonzentrationen von 2.3±1.5 µg·mL-1 and 2.1±1.6 µg·mL-1 (MW±SD) wurden nach 0.7±0.2 h und 0.7±0.3 h erreicht. Nach Einnahme von Buchweizenkraut und Rutin wurden maximale Plasmakonzentrationen (trotz der doppelten Dosis) von nur 0.6±0.7 µg·mL-1 und 0.3±0.3 µg·mL-1 nach 4.3±1.8 h bzw. 7.0±2.9 h erreicht. Die terminale Halbwertszeit lag bei ca. 11 h fuer alle vier Pruefpraeparate. Die Disposition von Quercetin ist daher primaer von der Zuckerkomponente abhaengig. Zu einem geringern Anteil beeinflusst die Pflanzenmatrix im Falle von Buchweizenkrauttee sowohl Geschwindigkeit als auch Ausmass der Resorption. Der Resorptionsort scheint fuer Quercetin-4‘-O-glucoside und Quercetin-3-O-rutinoside unterschiedlich zu sein. Die bedeutung spezifischer carrier fuer die Resorption von Quercetinglykosiden sowie von intestinalen ß-Glucosidasen muss in weiteren Untersuchungen geklaert werden.
Um einen Beitrag zum besseren Verständnis der Rolle der Bienenwachse in der Kommunikation der Honigbienen leisten zu können, wurden Wabenwachse unterschiedlichen Alters und Kutikulawachse unterschiedlicher Kasten,Geschlechter und Berufsgruppen mit Hilfe von Gaschromatographie, Massenspektroskopie und FTIR-Spektroskopie untersucht. Die chemischen Analysen zeigten mittels Diskriminantenfunktionsanalysen hochsignifikante Unterschiede in den aliphatischen Kohlenwasserstoffen zwischen Wabenwachsen unterschiedlichen Alters und Kutikulawachsen unterschiedlicher Kasten und Geschlechter. Erstmals konnte für ein komplexes Substanzgemisch (Bienenwachs) eine lineare Abhängigkeit zwischen dem Schmelzverhalten und der chemischen Zusammensetzung der Wachse nachgewiesen werden.Mit Hilfe von Verhaltensversuchen wurde der Frage nachgegangen, ob die chemischen Unterschiede für die Bienen überhaupt relevant sind. Mit Hilfe der differentielle Konditionierung des Rüsselreflexes wurde getestet, inwieweit Bienen die verschiedenen Wachse unterscheiden können. Eine Diskriminierung der Wachse aufgrund der aliphatischen Kohlenwasserstoffe war den Honigbienen nicht möglich. Dies ergab einen neuen und interessanten Einblick in die Kommunikation der Honigbienen

The author presents the first detailed review of the pharmacological therapy of parkinsonism from ancient times until the near present (1980). It is not clear whether parkinsonism as it is now defined – a progressive neurodegenerative disorder of the basal ganglia characterized by sharply reduced striatal dopamine levels, particularly in the striatum – has always affected a significant minority of aged persons, but suggestive evidence to this effect in the older literature is reviewed. The major discussion commences, however, with the administration of various plant alkaloids to parkinsonian patients in the second half of the 19th century. Antiparkinsonian therapy since this time may be divided into a number of phases: 1. The employment of alkaloids derived from solanaceous plants: initially hyoscyamine, then hyoscine/scopolamine and atropine. The discovery and characterization of these alkaloids, and the gradual recognition that other pharmacologically useful solanaceous alkaloids (such as duboisine) were identical with one or other of these three compounds, is discussed. 2. With the outbreak of encephalitis lethargica following the First World War, parkinsonian patient numbers increased dramatically, leading to a multiplicity of new directions, including the use of another solanaceous plant, stramonium, of extremely high atropine doses, and of harmala alkaloids. 3. The so-called “Bulgarian treatment” was popularized in western Europe in the mid-1930s. It was also a belladonna alkaloid-based therapy, but associated with greater efficacy and fewer side effects. This approach, whether as actual plant extracts or as defined combinations of belladonna alkaloids, remained internationally dominant until the end of the 1940s. 4. Synthetic antiparkinsonian agents were examined following the Second World War, with the aim of overcoming the deficiencies of belladonna alkaloid therapy. These agents fell into two major classes: synthetic anticholinergic (= antimuscarinic) agents, such as benzhexol, and antihistaminergic drugs, including diphenhydramine. These agents were regarded as more effective than plant-based remedies, but certainly not as cures for the disease. 5. A complete change in direction was heralded by the discovery in 1960 of the striatal dopamine deficit in parkinsonism. This led to the introduction of L-DOPA therapy for parkinsonism, the first approach directed against an identified physiological abnormality in the disorder. 6. Subsequent developments have thus far concentrated on refinement or supplementation of the L-DOPA effect. Recent attempts to develop neuroprotective or -restorative approaches are also briefly discussed. The thesis also discusses the mechanisms by which the various types of antiparkinsonian agent achieved their effects, and also the problems confronting workers at various periods in the design and assessment of novel agents. The impact of attitudes regarding the etiology and nature of parkinsonism, particularly with regard to symptomatology, is also considered. Finally, the history of antiparkinsonian therapy is discussed in context of the general development of both clinical neurology and fundamental anatomical, physiological and biochemical research. In particular, the deepening understanding of the neurochemical basis of central nervous system function is emphasized, for which reason the history of dopamine research is discussed in some detail. This history of antiparkinsonian therapy also illustrates the fact that the nature of experimental clinical pharmacology has markedly changed throughout this period: No longer the preserve of individual physicians, it is now based firmly on fundamental laboratory research, the clinical relevance of which is not always immediately apparent, and which is only later examined in (large scale) clinical trials. It is concluded that antiparkinsonian therapy was never irrational or without basis, but has always been necessarily rooted in current knowledge regarding neural and muscular function. The achievements of L-DOPA therapy, the first successful pharmacological treatment for a neurodegenerative disorder, derived from the fruitful union of the skills and contributions of different types by laboratory scientists, pharmacologists and clinicians.
Die Methodik und Technik der Gaswechselmessung von Pflanzen wurde für den Modellorganismus Arabidopsis thaliana optimiert und für Untersuchungen zur Beteiligung des Aquaporins PIP1b an Wassertransportvorgängen während der Stomaöffnung verwendet. Die Messungen der Transpirationsraten von PIP1b-Antisense-Pflanzen ergaben keine Hinweise auf Veränderungen des zeitlichen Verlaufs der Stomaöffnung. Die Wasserpermeabilitäten von Schließzell-Plasmamembranen scheinen somit nicht von der Expression des Aquaporins PIP1b beeinflußt zu sein. - Gaswechselmessungen an Nicotiana tabacum NtAQP1-Antisense-Pflanzen zeigten eine Verringerung der Transpirationsraten im Licht und eine geringere Grundtranspiration im Dunkeln. Dies deutet auf eine Beteiligung von NtAQP1 am Wassertransport hin. - Ausgewählte Arabidopsis thaliana-Mutanten wurden hinsichtlich ihrer stomatären Antwort auf Rot- und Rot-/Blaulicht-Bestrahlung analysiert. Hierfür wurde ein Doppelbestrahlungs-Protokoll entwickelt. Vergleiche mit den Wildtypen ergaben signifikante Unterschiede bei der Phytochrom-Mutante phyA-103, der Abscisinsäure-Mutante aba3-2 und der Auxin-resistenten Mutante axr1-3. Ferner zeigte die Mutante npq1-2 nicht die beschriebene Abweichung der stomatären Antwort auf Blaulicht. - Die Expressionsmuster eines PIP1b-GFP-Reportergens in transgenen Arabidopsis thaliana-Pflanzen wurden analysiert. Hohe Promotor-Aktivitäten konnten in meristematischen Bereichen von Wurzel und Sproß, in Elementen der Leitbündel, in jungen Kotyledonen und in Staubblättern beobachtet werden. Es zeigte sich eine enge Korrelation zwischen PIP1b-Promotoraktivität und Streckungswachstum. - Eine Sequenzanalyse des NtAQP1-Promotors ergab Übereinstimmungen mit spezifischen Bindungsmotiven von MYB-ähnlichen Transkriptionsfaktoren. Mit Promotor-Reportergenen konnte die Beteiligung eines dieser Sequenzmotive an der GA- und ABA-induzierten Aktivierung des NtAQP1-Promotors gezeigt werden. Zur Analyse der Phytohormon-Wirkungen auf deletierte Promotorbereiche wurde ein duales Vektorsystem entwickelt und bei der transienten Transformation von BY2-Protoplasten eingesetzt. - Die Expression eines GFP::NtAQP1-Fusionsgens in BY2-Zellen zeigte die subzelluläre Lokalisation des Aquaporins in der Zytoplasmamembran. Ferner wurde Fusionsprotein in Vesikel-ähnlichen Strukturen beobachtet.
