The role of cuticular waxes in the prepenetration processes of Blumeria graminis f.sp. hordei

The obligate biotrophic fungus Blumeria graminis f.sp. hordei is the causative agent of barley powdery mildew, a destructive foliar disease. The fungus infests barley (Hordeum vulgare), an important crop plant, which causes remarkable yield losses. Leaf cuticular wax of barley consists mainly of primary alcohols (80%), alkyl esters (10%) and minor constituents such as fatty acids (2%), alkanes (2%) and aldehydes (1%). The asexual airborne conidia have an initial contact to the leaf surface, in an environment dominated by cuticular waxes, which trigger germination and differentiation. The conidia undergo a sequential morphogenesis during that phase, the so-called prepenetration processes. The conidium initially forms a short primary germ tube, followed by a secondary elongated germ tube, which swells and finally forms a septate appressorium. The fungal appressorium infests the epidermal cell of the host plant and establishes an initial haustorium, the feeding structure of the fungus. In order to assess the effects of single host plant wax constituents on the prepenetration processes a novel in vitro assay based on Formvar® resin was established. This system permits the setting up of homogeneous surfaces as substrata, at which the adsorbed amounts and the surface hydrophobicity are highly reproducible, independently of the tested substance classes and chain lengths of the molecules. In this system, very-long-chain aldehydes promoted germination and differentiation of B. graminis f.sp. hordei conidia. The appressorium formation rates were decreasing in a concentration and chain-length dependent manner compared to n-hexacosanal (C26), which was the most effective aldehyde (C22<<C24<C26>C28>>C30). The tested alkanes with even and odd numbers (C24-C33), fatty acids (C20-C28), alkyl esters (C40-C44) and primary alcohols (C20-C30) did not induce germination and appressorium formation. The primary alcohol n-hexacosanol (C26) was an exception, as it was capable of significantly stimulating conidial germination and appressorial germ tube formation. To elucidate the impact of very-long-chain aldehydes on an intact plant surface in vivo, B. graminis f.sp. hordei conidia were inoculated on glossy11 mutant leaves of the non-host plant maize (Zea mays), which are - unlike the wildtype - completely devoid of very-long-chain aldehydes. On glossy11 leaves 60% of B. graminis f.sp. hordei conidia remained ungerminated and 10% developed a mature appressorium, which is three times less than on wildtype plants. Spraying of synthetic n-hexacosanal or wildtype leaf wax on glossy11 leaves fully restored the fungal prepenetration processes. In contrast, spraying of non-inducing n-alkanes, primary alcohols or very-long-chain fatty acids on wildtype leaves of maize mimicked the aldehyde deficient phenotype of glossy11. During the prepenetration processes an appressorium is formed, which is a newly formed specialized cell. Germination and subsequent morphogenesis are linked to the cell cycle in certain phytopathogenic fungi. It was investigated to what extent the prepenetration processes of B. graminis f.sp. hordei are synchronized with cell cycle progression. Hence, a distinct staining procedure of nuclei for fixed samples of B. graminis f.sp. hordei conidia based on DAPI (4,6-diamidino-2-phenylindole) was developed. In combination with a pharmacological approach it was possible to trace mitosis in dependency of conidial germination and differentiation in vivo and in vitro. The uninucleate conidium germinated and after formation of the appressorial germ tube, a single mitosis occurred in the primordial conidium six hours after inoculation. The inhibition of S-phase with hydroxyurea or M-phase with benomyl prevented appressorium formation, but not the development of the appressorial germ tube. These results indicate that mitosis and a successful cytokinesis are necessary prerequisites for the appressorium formation but not for conidial morphogenesis. In order to identify genes that are expressed in response to certain host plant wax constituents, which may be critical for the prepenetration phase, cDNA clone libraries were constructed by suppression subtractive hybridization (SSH) after inoculation. The Formvar® resin based in vitro system provided a stable platform to enrich cDNA sequences that were expressed in B.graminis f.sp. hordei conidia incubated on n-hexacosanal coated surfaces for 22 minutes. Among various candidates, a cDNA sequence was identified, which was upregulated on barley leaves and on surfaces coated with n-hexacosanal or extracted barley leaf wax. The hexacosanal responsive transcript was cloned by 3’ and 5’ RACE. The cDNA sequence showed no homologies to genes of known function in fungal development and fungal pathogenicity in plants.
Pflanzen der Gattung Nepenthes decken einen erheblichen Anteil ihres Nährstoffbedarfs durch den Fang und die Verdauung tierischer Beute, insbesondere von Insekten. Als Fangorgane dienen kannenförmig umgewandelte Blattspreiten. Die Kanneninnenseiten sind in einer breiten Zone dicht mit epikutikulären Wachskristallen besetzt. Die Oberflächen dieser so genannten Gleitzone sind extrem rutschig für die meisten Insekten und spielen eine zentrale Rolle beim Fang und der Zurückhaltung der Beute in der Kanne. Frühere Untersuchungen beschrieben die Kristalle dabei als extrem fragil, wodurch diese unter der mechanischen Belastung eines Insekts leicht abrechen und somit der Kontakt zur Pflanzenoberfläche verloren geht. Um diese Hypothese zu überprüfen und den Mechanismus der Rutschigkeit verstehen zu können, hatte die vorliegende Arbeit zum Ziel, sowohl die strukturellen als auch die physikalisch-chemischen Eigenschaften der Wachskristalle in den Kannen von drei Nepenthes-Arten vergleichend zu charakterisieren. Diese Eigenschaften können jedoch nur dann bewertet werden, wenn die chemische Zusammensetzung der Wachskristalle verlässlich bestimmt werden kann. Um die gaschromatographische Trennung und massenspektrometrische Analyse der Komponenten zu erleichtern, werden hydroxyl-haltige Verbindungen häufig durch eine Derivatisierung mit N,O-Bis(trimethylsilyl)trifluoracetamid (BSTFA) in die entsprechenden Trimethylsilyl-Ether bzw. -Ester überführt. Dabei können jedoch auch unerwünschten Nebenreaktionen carbonyl-haltiger Verbindungen auftreten, die eine quantitative Analyse der ursprünglichen Komponenten erschweren. Im ersten Teil dieser Arbeit ergab die Überprüfung der Derivatisierungsreaktion, dass aliphatische Aldehyde mit BSTFA zu cis-trans isomeren Alkenyl-Trimethylsilyl-Ethern und Alkenyl-Acetamid-Addukten reagierten. Weiterhin bildeten sich aus Aldehyden und primären Alkoholen unter den gegebenen Bedingungen, cis-trans isomere Alkenyl-Alkyl-Ether. Es konnte gezeigt werden, dass eine verlässliche und quantitative Bestimmung der ursprünglich vorhandenen Aldehyd- und Alkoholmengen nur unter einer Quantifizierung der in den resultierenden Nebenprodukten gebundenen Mengen möglich war. Im zweiten Teil dieser Arbeit zeigten rasterelektronenmikroskopische Untersuchungen an den Gleitzonenoberflächen von drei Nepenthes-Arten, dass die epikutikulären Wachskristalle ein Netzwerk aus glattrandigen Plättchen bilden und senkrecht aus den Oberflächen herausstehen. Es wurden Methoden etabliert, die eine mechanische Präparation der Wachs-kristalle von den Gleitzonenoberflächen erlaubten. Dabei zeigten die Kristalle eine hohe strukturelle Integrität. Die Beprobungsstrategien erwiesen sich als selektiv für die epikutiku-lären Wachse und somit für die Schnittstelle der Pflanze-Insekten-Wechselwirkung. Die anschließenden chemischen Analysen zeigten deutliche Gradienten zwischen den Zusammen-setzungen der epikutikulären und intrakutikulären Wachskompartimente. Die epikutikulären Kristalle bestanden aus Mischungen aliphatischer Komponenten und waren von sehr lang-kettigen Aldehyden dominiert. Triacontanal war in allen Fällen die Hauptkomponente und weitgehend für die Kristallbildung verantwortlich. Diese Ergebnisse quantifizierten erstmalig direkt die Zusammensetzung epikutikulärer Wachskristalle und bestätigten die für deren Bildung ursprüngliche Hypothese einer spontanen Phasentrennung eines hochkonzentrierten Bestandteils. Die schlechte Löslichkeit der Kristalle von verschiedenen Nepenthes-Arten in Chloroform wies zudem darauf hin, dass sie polymere Formen der Aldehyde beinhalteten. Diese Vermutung konnte im dritten Teil dieser Arbeit durch ATR-FTIR-spektroskopische Untersuchungen bestätigt werden. Die Kombination dieser Analysetechnik mit einer der mechanischen Beprobungsstrategien zeigte, dass weder isolierte Kristalle, noch Kristalle auf nativen Oberflächen, monomere Aldehyde beinhalteten. Diese konnten jedoch durch Tempe-raturerhöhung oder Lösen in Chloroform unter erhöhter Temperatur freigesetzt werden. Auf Grund charakteristischer Absorptionseigenschaften, der molekularen Anordnung sowie dem Phasenverhalten der beteiligten Komponenten konnte geschlossen werden, dass die Aldehyde in nativen Kristallen in Form von Polyacetalen vorliegen. Dies lässt vermuten, dass die epikutikulären Wachskristalle dadurch nicht nur thermisch und chemisch, sondern auch mechanisch verstärkt werden. Werden alle Daten zusammengefasst, können die strukturellen sowie physikalisch-chemischen Eigenschaften der epikutikulären Wachskristalle auf den Gleitzonenoberflächen verschiedener Nepenthes-Arten im Kontext ihrer ökologischen Funktion neu beurteilt werden. Auf diesen Ergebnissen basierend kann die Hypothese aufgestellt werden, dass die Kristalle im Kräftebereich, den ein Haftorgan eines Insektes auf sie ausübt, mechanisch stabil sind und somit andere Mechanismen die Rutschigkeit verursachen.
In food and pharmaceutical analysis, the classical indices peroxide value (PV), acid value (AV) and p-anisidine value (ANV) still play an important role as quality and authenticity control parameters of fats and oils. These indices are sum parameters for certain deterioration products (PV for hydroperoxides, AV for free fatty acids, ANV for aldehydes) and are obtained using volumetric or UV/VIS spectroscopic analytical approaches. 1H NMR spectroscopy provides a fast and simple alternative to these classical approaches. In the present work, novel 1H NMR methods to determine hydroperoxides, free fatty acids and aldehydes in fats and oils were developed.
Hydroperoxides:
The influence of solvent, water, free fatty acids and sample weight on the hydroperoxide group proton (OOH) signal was investigated. On the basis of the obtained results, the sample preparation procedure of the new 1H NMR method was established. A rough assignment of the hydroperoxide group signals in edible fats and oils to methyl oleate, methyl linoleate and methyl linolenate was conducted. Furthermore, to gain information on how many different hydroperoxide species originate from trioleate autoxidation, a kinetic study on trioleate monohydroperoxides was performed. The evaluation of the data strongly indicates that all of the conceivable 18 trioleate monohydroperoxides were formed during trioleate autoxidation. The analytical performance of the NMR method was compared to that of the classical PV approach by means of the so-called “relative sensitivity” according to Mandel. It was shown that both methods exhibit a similar analytical performance. A total of 444 edible oil samples were analysed using both methods. For some oil varieties considerable discrepancies were found between the results. In the case of black seed oil and olive oil two substances were identified that influence the classical PV determination and thus cause positive (black seed oil) and negative (olive oil) deviations from the theoretical PV expected from the NMR values.
Free fatty acids:
In order to find the optimal solvent mixture to measure the carboxyl group protons (COOH) of free fatty acids in fats and oils, the effect of solvent on the COOH signal was investigated for different mixtures of CDCl3 and DMSO-d6. The comparison of the NMR method with the classical AV method by means of the relative sensitivity revealed that both methods exhibit a similar analytical performance. 420 edible oil samples were analysed by both approaches. Except for pumpkin seed oil, where slight deviations were observed, there was a good compliance between the results obtained from the two methods. Furthermore, the applicability of the 1H NMR assay to further lipids with relevance in pharmacy was tested. For hard fat, castor oil, waxes and oleyl oleate modifications of the original sample preparation procedure of the NMR method were necessary to achieve comparable results for both methods.
Aldehydes:
The new 1H NMR method enables the determination of the molar amounts of n-alkanals, (E)-2-alkenals and (E,E)-2,4-alkadienals. It was illustrated that the ANV can be modelled as a linear combination of the NMR integrals of these aldehyde species. A functional relationship was derived on the basis In conclusion, the new 1H NMR methods provide an excellent alternative to of calibration experiments. The suitability of the model was shown by comparing the NMR-determined ANVs with the measured classical ANVs of 79 commercially available edible oils of different oil types.
In conclusion, the new 1H NMR methods provide an excellent alternative to the determination of the classical indices PV, AV and ANV. They have several advantages over the classical methods including the consumption of small solvent amounts, the ability to automatize measurement and to acquire several different parameters out of the same NMR spectrum. Especially concerning their selectivity, the 1H NMR methods are highly superior to the classical methods.
