The past decades have witnessed the development of new pharmaceutical compounds that modulate receptor function by targeting allosteric sites. Allosteric sites are, by definition, domains topographically distinct from the orthosteric binding pocket where the natural ligand binds. Exploring the possibilities of linking orthosteric and allosteric pharmacophores in one compound to yield ‘bitopic’ compounds is a strategy derived from the “message-address” concept by Schwyzer , first applied to GPCRs by Portoghese et al. This concept explicitly underlines the orthosteric/allosteric combination, in opposite to the more general umbrella term bivalent. The broad possibilities of bitopic ligands in the pharmaceutical field are under continuous study. Bitopic compounds are promising pharmaceutical tools for taking advantage of the allosteric binding to achieve subtype selectivity while preserving high affinity at the receptor. The development of bitopic ligands, based on the idea of combining high affinity (via orthosteric sites) with high selectivity (via allosteric sites), have led to the development of highly selective bivalent ligands for GPCRs , such as for the opioid receptors , muscarinic acetylcholine receptors (mAChRs), serotonin receptors, cannabinoid receptors, and gonadotropin-releasing hormone receptors. This concept has even been extended to other receptors, for examples nicotinic receptors and other proteins, such as acetylcholinesterases and the tyrosine kinase receptors TrkA and TrkC. The reasons to pursue a bitopic ligand approach are various. An improved affinity for the target GPCR and/or an improved selectivity either at the level of receptor subtype, or at the level of signaling pathway. Another advantage of bitopic ligands over purely allosteric ligands is that the former rely on the appropriate presence of endogenous agonist tone to mediate their effects, whereas a bitopic ligand would engage the orthosteric site irrespective of the presence or absence of endogenous tone. By way of introduction to the hybrid approach, a review of the concept of hybrids compounds targeting the cholinergic system is presented in section A of this thesis. Recent updates in hybrid molecule design as a strategy for selectively addressing multiple target proteins involved in Alzheimer's disease (AD) is here reported . This represents the potential and the growing interest in hybrid compound as pharmacological tools to achieve receptor subtype selectivity and/or, to study the overall functional activity of the receptor. Until now, muscarinic acetylcholine receptors (mAChRs) have proved to be a particularly fruitful receptor model for the development and characterization of bitopic ligands. In this thesis, several examples of new muscarinic bitopic approach are reported in the results section. A study of bipharmacophoric ligands composed of the muscarinic positive allosteric modulators (BQCAderived compounds) linked with chain of various lengths to different orthosteric building blocks is reported in the result part 1. Synthesis and examination of the potential pharmacological characteristic of Oxotremorine-BQCAd compounds and Xanomeline-BQCAd hybrid derivatives are described in results parts 2 and 4, respectively. Moreover, the bitopic concept has even been extended to other proteins, such as acetylcholinesterase. In the result part 5 an overview of the new Tacrine-Xanomeline hybrids aiming to improve the inhibitory potency of the acetylcholinesterase and simultaneously to increase the cholinergic tone, via the xanomelinic portion acting on the M1 receptor is given. A new trivalent approach is presented for the first time to deepen the study of the M1 muscarinic receptor in the result part 6. Moreover, the synthesis of a new series of iperoxo-derived alkane, bis(ammonio)alkane-type and rigidified chain ligands is given in the result part 7 together with some prospects for further research.
Die muscarinischen Rezeptoren sind ein wichtiger Bestandteil des parasympathischen Nervensystems. Sie gehören zur großen Gruppe der G-Protein-gekoppelten Rezeptoren, die nach ihrer Verwandtschaft in drei große Klassen eingeteilt werden können. Die muscarinischen Rezeptoren gehören zur Klasse A, den rhodopsinähnlichen Rezeptoren. Durch die im Jahr 2000 vorgenommene Aufklärung der hochauflösenden Röntgenkristallstruktur des Rinderrhodpsins und die hohe Aminosäuresequenzähnlichkeit der G-Protein-gekoppelten Rezeptoren hat man eine sehr gute Modellvorstellung über den Aufbau der G-Protein-gekoppelten Rezeptoren. Die Rezeptoren bestehen aus sieben transmembranalen Helices, die von drei intrazellulären und drei extrazellulären Loops stabilisiert werden. Bis heute konnten fünf Rezeptorsubtypen gentechnisch klassifiziert werden, die sich durch ihre Gewebeverteilung und Funktion unterscheiden. Allen Subtypen ist eine hohe Sequenzhomologie im Bereich der orthosteren Bindungsstelle gemeinsam, so dass die Entwicklung von subtyp-spezifischen orthosteren Liganden sehr schwierig ist. Außer der orthosteren Bindungsstelle konnte noch eine weitere Bindungsstelle am muscarinischen Rezeptor identifiziert werden. Diese befindet sich weiter außerhalb im Rezeptor in einem Bereich, der über die fünf Rezeptorsubtypen nicht sehr stark konserviert ist, so dass die Entwicklung von subtyp-spezifischen Liganden möglich ist. An dieser zweiten Bindungsstelle binden allostere Modulatoren. Hierbei handelt es sich um Substanzen, die ohne den orthosteren Liganden keinen Effekt am Rezeptor auslösen, dafür aber die Gleichgewichtsbindung des orthosteren Liganden beeinflussen können. Der Einfluss auf die Gleichgewichtsbindung geschieht wechselseitig und kann positiv, neutral oder negativ kooperativ sein. Zusätzlich üben allostere Modulatoren einen Effekt auf die Dissoziation des orthosteren Liganden aus. Die meisten bisher gefunden allosteren Modulatoren erniedrigen die Dissoziationsgeschwindikeit des orthosteren Liganden vom Rezeptor. Die Summe dieser Eigenschaften machen die allosteren Modulatoren sehr interessant für die Arzneimitteltherapie. Das Ziel dieser Arbeit war die Synthese strukturell neuer allosterer Modulatoren des muscarinischen Rezeptors unter Anwendung des postulierten Pharmakophormodells. Als Ausgangspunkt sollten geländerhelicale Moleküle dienen, die strukturell abgewandelt dieses Pharmakophormodell sehr gut erfüllen. Die geländerhelicalen Moleküle ähneln in ihrem dreidimensionalen Aufbau dem Geländer einer Wendeltreppe. Sie sind durch die Brücken zwischen den aromatischen Bereichen sehr rigide Moleküle, so dass es nur wenige genau definierte Konformationen gibt. Grundsätzlich können drei Atropisomere unterschieden werden, wobei zwei zueinander enantiomer sind. Geplant war die Synthese eine Reihe von tertiären Aminen oder quartären Ammoniumsalzen. Die Synthese der Ausgangsverbindung konnte nach der Vorschrift von Kiupel erfolgen, war aber nur mit geringer Ausbeute möglich. Deshalb wurde dieser Syntheseweg nicht weiterverfolgt. Als Alternative bot sich an, auf die Brücken zwischen den aromatischen Ringen zu verzichten. Die so entstandenen Verbindungen sind weniger rigide und können sich deshalb gegebenenfalls besser an den Rezeptor anpassen. Grundsätzlich können je nach Substitutionsmuster zwei Synthesewege verfolgt werden. Beide Varianten erfüllen das postulierte Pharmakophormodell. Der Aufbau des Grundgerüstes erfolgt mittels einer nickelkatalysierten Grignard-Kupplung. Danach erfolgen eine Wohl-Ziegler-Seitenkettenbromierung und eine Verlängerung der Seitenkette im Sinne einer Alkylierung mittels Malonsäurediethylester und einer Hilfsbase. Anschließend erfolgen die Decarboxylierung und die Umsetzung zum Amid, das zum Amin reduziert werden kann. Betrachtet man die Lage der Pharmakophorelemente so variiert der Abstand der positiv geladenen Stickstoffe je nach Konformation zwischen 5 Å und 15 Å, so dass ein weiter Bereich abgedeckt werden kann. Der Abstand der aromatischen Bereiche bleibt relativ stabil. Die pharmakologische Testung der Verbindungen auf ihre allostere Potenz und Affinität zum muscarinischen Rezeptor erfolgte in der Arbeitsgruppe von Prof. Mohr in Bonn. Hierzu werden Membranhomogenate vom Herzventrikelgewebe des Hausschweines verwendet. Diese enthalten mit großer Prävalenz muscarinische M2-Rezeptoren. Es wurden Gleichgewichtsbindungs- und Dissoziationsexperimente durchgeführt. Bis jetzt sind noch nicht alle Verbindungen getestet worden. Die bisher getesteten Verbindungen weisen alle eine Affinität zum mit [3H]-N-Methylscopolamin besetzten muscarinischen M2-Rezeptor im mikro-molaren Bereich auf. Sie liegen damit im oberen Bereich der bisher synthetisierten allosteren Modulatoren. Das postulierte Pharmakophormodell konnte also mit Hilfe der synthetisierten Substanzen bestätigt werden.
Alzheimer’s disease is a complex network of several pathological hallmarks. These characteristics always occur concomitantly and cannot be taken as distinct features of the disease. While there are hypotheses trying to explain the origin and progression of the illness, none of them is able to pinpoint a definitive cause. This fact challenges researchers not to focus on one individual hallmark but, bearing in mind the big picture, target two or more indications at once. This work, therefore, addresses two of the major characteristics of AD: the cholinergic hypothesis and neurotoxic oxidative stress. The former was achieved by targeting the postsynaptic muscarinic M1 acetylcholine receptor to further investigate its pharmacology, and the latter with the synthesis of neuroprotective natural antioxidant hybrids.
The first aim was the design and synthesis of dualsteric agonists of the muscarinic M1 acetylcholine receptor. Activation of this receptor was previously shown to improve AD pathologies like the formation of Aβ and NFTs and protect against oxidative stress and caspase activation. Selectively targeting the M1 receptor is difficult as subtypes M1 – M5 of the muscarinic AChRs largely share the same orthosteric binding pocket. Orthosteric ligands are thus unsuitable for selective activation of one specific subtype. Secondary, allosteric binding sites are more diverse between subtypes. Allosteric ligands are, however, in most cases dependent on an orthosteric ligand to cause downstream signals. Dualsteric ligands thus utilize the characteristics of both orthosteric and allosteric ligands in form of a message-address concept. Bridged by an alkylene-linker, the allosteric part ensures selectivity, whereas the orthosteric moiety initiates receptor activation. Two sets of compounds were synthesised in this sense. In both cases, the orthosteric ligand carbachol is connected to an allosteric ligand via linkers of different chain length. The first set utilizes the selective allosteric M1 agonist TBPB, the second set employs the selective M1 positive allosteric modulator BQCA. Six compounds were obtained in twelve-step syntheses each. For each one, a reference compound lacking the carbachol moiety was synthesised. The dualsteric ligands 1a-c and 2a c were tested in the IP1 assay. The assay revealed that the TBPB-dualsterics 1 are not able to activate the receptor, whereas the respective TBPB-alkyl reference compounds 27 gave signals depending on the length of the alkylene-linker, suggesting allosteric partial agonism of alkyl compounds 27 and no dualsteric binding of the putatively dualsteric compounds 1. The dualsteric BQCA molecules 2, however, activated the receptor as expected. Efficacy of the C5 linked compound 2b was the highest, yet C3 and C8 compounds (2a and 2c) also showed partial agonism. In this case, the reference compounds 31 showed no receptor activation, implying the intended dualsteric binding mode of the BQCA-carbachol compounds 2. Further investigations will be conducted by the working group of Dr. Christian Tränkle at the Department of Pharmacology at the University of Bonn to confirm binding modes and determine affinities as well as selectivity of the synthesised dualsteric compounds.
The second project dealt with the design, synthesis and biological evaluation of neuroprotective esters of the flavonolignan silibinin. While silibinin is already a potent antioxidant, it has been observed that the 7-OH group has a pro-oxidative character, making this position attractive for functionalisation. In order to obtain more potent antioxidants, the pro-oxidative position was esterified with other antioxidant moieties like ferulic acid 35 and derivatives thereof. Seventeen esters of silibinin 32, including pure diastereomers of 7 O feruloylsilibinin (43a and 43b) and a cinnamic acid ester of 2,3-dehydrosilibinin 46, were synthesised by regioselective esterification using acyl chlorides under basic conditions. The physicochemical antioxidant properties were assessed in the FRAP assay. This assay revealed no improvement of the antioxidant properties except for 7-O-dihydrosinapinoylsilibinin 39b. These results, however, do not correlate with the neuroprotective properties determined in the HT-22 hippocampal neuronal cell model. The assay showed overadditive neuroprotective effects of the esters exceeding those of its components and equimolar mixtures with the most potent compounds being 7-O-cinnamoylsilibinin 37a, 7-O-feruloylsilibinin 38a and the acetonide-protected caffeic acid ester 40a. These potent Michael system bearing compounds may be considered as “PAINS”, but the assays used to assess antioxidant and neuroprotective activities were carefully chosen to avoid false positive readouts. The most potent compounds 37a and 38a, as well as the diastereomers 43a and 43b, were further studied in assays related to AD. In vitro ischemia, inhibition of microglial activation, PC12 cell differentiation and inhibition of Aβ42 and τ protein aggregation assays showed similar results in terms of overadditive effects of the synthesised esters. Moreover, the diastereomers 43a and 43b showed differences in their activities against oxytosis (glutamate-induced apoptosis), inhibition of Aβ42 and τ protein aggregation, and PC12 cell differentiation. The stereospecific effect or mode of action against Aβ42 and τ protein aggregation is more pronounced than that of silybin A (32a) and silybin B (32b) reported in literature and needs to be elucidated in future work. Stability measurements in cell culture medium revealed that the esters do not only get hydrolysed but are partially oxidised to their respective 2,3-dehydrosilibinin esters. Because dehydrosilibinin 45 itself is described as a more potent antioxidant than silibinin 32, 7 O cinnamoyl-2,3-dehydrosilibinin 46 was expected to be even more potent than its un-oxidised counterpart 37a in terms of neuroprotection. The oxytosis assay, however, showed that the neurotoxicity of 46 is much more pronounced, especially at higher concentrations, reducing its neuroprotective potential. Dehydrosilibinin esters are therefore inferior to the silibinin esters for application as neuroprotectants, because of the difficulty of their synthesis and their increased neurotoxicity. A synergistic effect of both species (silibinin and the oxidised form) might, however, be possible or even necessary for the pronounced neuroprotective effects of silibinin esters. As the dehydro-species show distinct neuroprotective properties at low concentrations, their continuous formation over time might make an essential contribution to the overall neuroprotection of the synthesised esters. Due to solubility issues for some of the ester compounds, 7-O-cinnamoylsilibinin 37a was converted into a highly soluble hemisuccinate. The vastly improved solubility of 7 O cinnamoyl-23-O-succinylsilibinin 48 was confirmed in shake-flask experiments. Contrary to expectation, stability examinations showed that the succinyl compound 48 is not cleaved to form 7-O-cinnamoylsilibinin 37a. Neuroprotection assays confirmed that 48 is not a prodrug of the corresponding ester. It was determined that the main site of hydrolysis is the 7-position, cleaving 37 to silibinin 32 and cinnamic acid thus reducing the compound’s neuroprotective effects. Nevertheless, the compound still showed neuroprotection at a concentration of 25 µM. The improved solubility might be more beneficial than the higher neuroprotection of the poorly soluble parent compound 37a in vivo. 7 O Cinnamoylsilibinin 37a was further investigated to reduce Aβ25 35 induced learning impairment in mice. While tendencies of improved short-term and long-term memory in the animals were observed, the effects are not yet statistically significant in both Y-maze and passive avoidance tests. A greater number of test subjects is necessary to ensure correctness of the preliminary results presented in this work. However, an effect of ester 37a is observable in vivo, showing blood-brain barrier penetration. The esters synthesised are a novel approach for the treatment of AD as they show strong neuroprotective effects and their hydrolysis products or metabolites are only non-toxic natural products.
Die vorliegende Arbeit befasst sich mit der Synthese von Liganden muscarinerger Rezeptoren, die zur Superfamilie der G-Protein-gekoppelten Rezeptoren gehören. Die fünf Muscarinrezeptor-Subtypen weisen im Bereich der orthosteren Bindungsstelle, an die u. a. der endogene Ligand Acetylcholin bindet, einen hohen Konservierungsgrad der Aminosäuresequenz auf. Alle Muscarinrezeptoren weisen neben der orthosteren Bindungsstelle auch eine oder mehrere allostere Bindungsstellen auf. Da diese sich im weniger konservierten extrazellulären Bereich des Rezeptors befinden, ist es möglich subtypspezifische Liganden zu entwickeln. Allostere Modulatoren binden an diese topographisch andere Stelle des Rezeptors und sind in der Lage, gezielt die Bindung eines orthosteren Agonisten oder Antagonisten zu modulieren. Dies bedeutet, dass sie die Assoziation und die Dissoziation orthosterer Agonisten und Antagonisten beeinflussen können. Die Gleichgewichtsbindung des Orthosters kann erhöht (d. h. positive Kooperativität), erniedrigt (d. h. negative Kooperativität) bzw. nicht beeinflusst (d. h. neutrale Kooperativität) werden. Das Ziel der Arbeit bestand zunächst darin, die Synthese allosterer Modulatoren des M2-Rezeptors vom Bis(ammonium)alkan-Typ, wie W84 und Naphmethonium, zu optimieren. Da die Synthesen dieser bisquartären Verbindungen zeitaufwändig sind, wurden die Synthesen durch Einsatz einer Synthesemikrowelle optimiert, um neue Bis(ammonium)alkan-Verbindungen effizienter synthetisieren zu können. Der Vergleich beider Synthesemethoden (konventionell bzw. Mikrowellen-unterstützt) zeigt sehr deutlich, dass die Reaktionszeiten durch Einsatz einer Synthesemikrowelle drastisch verkürzt werden konnten, insbesondere bei Verbindungen, die sehr lange Reaktionszeiten beanspruchen. Zusätzlich konnten die Ausbeuten aller synthetisierten Verbindungen durch Mikrowellen-unterstützte Synthese z. T. deutlich gesteigert werden. Im Verlauf der Arbeit wurden unsymmetrisch und symmetrisch nitrosubstituierte Bisnaphthalimide hergestellt. Durch Mikrowellen-unterstützte Hydrierung unter Palladium/Kohle-Katalyse wurden anschließend die analogen aminosubstituierten Derivate erhalten. Ein weiteres Ziel der Arbeit bestand darin, ein Naphmethonium-Derivat herzustellen, das über einen Spacer mit einem geeigneten Fluoreszenzfarbstoff gekoppelt werden kann. Ein Farbstoff-markierter allosterer Modulator könnte als ein wichtiges pharmakologisches Werkzeug zur direkten Charakterisierung allosterer Interaktionen und zur Verfolgung des „Rezeptor-Traffickings“ mittels Fluoreszenzkorrelations-spektroskopie genutzt werden. Als Spacer diente eine Alkylkette mit endständiger primärer Aminofunktion, die mit Alexa-Fluor 532 gekoppelt werden sollte. Zur Einführung des Spacers wurden verschiedene Strategien verfolgt. Schließlich konnte Verbindung 3h über eine Chlorzwischenstufe hergestellt werden, über deren freie primäre Aminogruppe der Fluoreszenzfarbstoff in weiteren Arbeiten gekoppelt werden kann. Ein weiteres Ziel der Arbeit war die Syntheseoptimierung des hochpotenten unselektiven Agonisten Iperoxo, das eine für akademische Zwecke wichtige Substanz in der pharmakologischen Testung und ein wichtiges Zwischenprodukt in der Synthese bivalenter Agonist/Alloster-Hybridverbindungen ist. Die Ausbeuten aller Stufen konnten erhöht und die Gesamtausbeute signifikant von 13% auf 22% gesteigert werden. Außerdem wurden neue Agonisten hergestellt, die sich im Heterozyklus unterscheiden. Ein weiteres Ziel der Arbeit war die Synthese bivalenter Hybridverbindungen nach dem Nachrichten-Adressen-Modell von Schwyzer. Zum einen sollten Agonist/Alloster-Hybridverbindungen synthetisiert werden, wobei die in der Literatur beschriebene Hybridverbindung Hybrid 1 (W84 und Iperoxo) als Leitstruktur diente. So wurde im ersten Schritt eine auf der allosteren Seite verkürzte Substanz ohne Phthalimidopropyl-Rest hergestellt (Hexamethonium und Iperoxo). Danach wurden verschiedene Funktionalitäten an der lateralen quartären Ammoniumfunktion eingeführt. Daneben wurden erstmals Antagonist/Alloster-Hybridverbindungen hergestellt, die aus einem M2-selektiven allosteren Modulator (W84, Naphmethonium bzw. Hexamethonium) und einem subtypunspezifischen Antagonisten (Atropin bzw. Scopolamin) bestehen. Im ersten Schritt wurde der allostere Molekülteil nach der optimierten Mikrowellen-unterstützten Synthese hergestellt. Die erhaltenen Zwischenstufen 2 wurden im Anschluss mit Atropin und Scopolamin in Acetonitril umgesetzt und die Antagonist/Alloster-Hybridverbindungen 34 und 35 erhalten. Die pharmakologische Testung der synthetisierten Verbindungen erfolgte durch Radioligandbindungsstudien an Herzventrikelgewebe des Hausschweins. Der allostere Effekt der Testsubstanzen wurde indirekt über die Verzögerung der Dissoziation des radioaktiv markierten orthosteren Antagonisten [3H]N-Methylscopolamin.
Da die Kontrolle der Kontraktion am Myometrium in der klinischen Praxis eine bedeutende Rolle spielt, sind Kenntnisse über die Physiologie zur Blockierung der Wehentätigkeit und Vermeidung von Frühgeburtlichkeit unverzichtbar. Der Einfuß des sympathischen Nervensystems mit Alpha und Beta-Rezeptoren ist gut untersucht, weshalb wir in der vorliegenden Arbeit eine Untersuchung zur Verteilung der muscarinischen Rezeptoren M1-M5 im schwangeren und nicht schwangeren Myometrium durchführten. Diese G-Protein gesteuerten parasympathischen Rezeptoren sind beispielsweise an Kontraktionsvorgängen in Blasen-, Magen-Darm- und Bronchialmuskulatur beteiligt und könnten so auch im Myometrium eine Rolle spielen. Mittels RT-PCR wurden 21 Myometriumbiopsien analysiert, wovon 11 Myometriumproben von Patientinnen in der 40. Schwangerschaftswoche, sowie jeweils 3 Proben von Patientinnen in der 32. Schwangerschaftswoche und von Patientinnen in der 25. Schwangerschaftswoche stammten, desweiteren eine Probe einer in der 10. Schwangerschaftswoche durch Hysterektomie abgebrochenen Schwangerschaft. Die drei letzten Proben stammten von nicht schwangeren Patientinnen nach vaginaler Hysterektomie. Dabei konnte gezeigt werden, dass sowohl zu verschiedenen Zeitpunkten in der Schwangerschaft, als auch am nicht schwangeren Myometrium jeweils m-RNA für M2, M3 und M5 muscarinische Rezeptoren existiert. Die durch die hier angewendeten Verfahren zur Gewebepräparation, Gewinnung von RNA und Anwendung der RT-PCR vorliegenden Ergebnisse sollten auf Proteinebene bestätigt werden, die Durchführung funktioneller Studien wäre wünschenswert.
GPCRs, particularly muscarinic receptors (mAChRs), are significant therapeutic targets in many physiological conditions. The significance of dualsteric hybrids selectively targeting mAChR subtypes is their great advantage in avoiding undesired side effects. This is attained by exploitation of the high affinity of ligand-binding to the orthosteric site and the structural diversity of the allosteric site to target an individual mAChR subtype, as well as offering signal bias to avoiding undesired transduction pathways. Furthermore, dualsteric targeting of mAChR subtypes helps in the elucidation of the physiological role of each individual mAChR subtype.
The first project was the attempt of synthesis of the M2-preferring ligand AFDX-384. AFDX-384 is known to preferentially bind to the M2 receptor subtype as an orthosteric antagonist, with partial interaction with residues in the allosteric site. This project aimed to re-trace the synthesis route of AFDX-384, to open the door to its upscaling and the future synthesis of AFDX-type dualsteric ligands. The multi-step synthesis of AFDX-384 is achieved through the synthesis of its 2 precursors, the chloro acyl derivative VIII and the piperidinyl derivative IV. Upscaled synthesis of the piperidinyl derivative IV was attained. Synthesis of the chloro acyl compound VIII was attempted. Several trials to synthesize the benzopyridodiazepine nucleus as well as its chloro-acylation resulted in the production of the novel crystal structures V and VI. X-ray crystallography was also done for crystallized molecules of the closed-ring benzopyridodiazepine VII that was previously synthesized. Chloro-acylation reactions of compound VII using phosgene seem to be attainable when done using reflux overnight. However, the use of methanol to aid in elution during silica gel column chromatography converted the expected product to the carbamate analogue IX. Hence, further attempts in purification should refrain from the use of methanol. The use of triphosgene instead of phosgene demonstrates a cleaner route for further upscaled synthesis.
The second project was the synthesis of dualsteric ligands involving variable orthosteric and allosteric moieties. Four different types of hybrids have been created over multiple steps. Dualsteric ligands have been synthesized using either a phthalimido- or 1,8-naphthalimidopropylamino moiety as the allosteric-binding group, coupled to either N-desmethyl pirenzepine or N-desmethyl clozapine using variable chain lengths. Furthermore, the synthesis of the dualsteric ligands involving N-desmethyl clozapine linked to either the super-agonist iperoxo or acetylcholine, and being connected using variable alkane chain lengths. Several reaction conditions have been investigated throughout the analysis of the optimal condition to conduct the critical final step of synthesis of these dualsteric hybrids, which involves the linking of the two segments of the hybrid together. The optimal method, which produced the least side products and highest yield, was to connect the two intermediates of the compound in absence of base, catalyst or microwaves while stirring at 35 °C for several days using acetonitrile as solvent (silica gel TLC monitoring, 0.2 M aqueous KNO3/MeOH 2:3). The ideal purification methods for the final compounds were found to be either crystallization from the reaction medium or using C18 reverse phase silica gel flash chromatography (using H2O/MeOH solvent system). All the hybrids will be subjected to pharmacological testing using the appropriate FRET assays.
Die vorliegende Arbeit befasst sich mit der Synthese und Charakterisierung allosterer Modulatoren muscarinischer Rezeptoren. Allostere Modulatoren binden an einer topographisch anderen Stelle am Rezeptor als klassische orthostere Liganden und sind so in der Lage, die Dissoziation und die Assoziation orthosterer Agonisten und Antagonisten zu beeinflussen. Die fünf Subtypen des Muscarinrezeptors M1-M5 unterscheiden sich vor allem in der Aminosäuresequenz der in den äußeren Bereichen des Rezeptorproteins vorhandenen Loops, während sie im Bereich des Rezeptorkanals, wo die orthostere Bindungsstelle lokalisiert ist, eine hohe Sequenzhomologie aufweisen. Die gemeinsame Bindungsstelle allosterer Modulatoren des M2-Rezeptors befindet sich im weniger konservierten extrazellulären Bereich. Somit sind allostere Modulatoren in der Lage, spezifisch an einen der Rezeptorsubtypen zu binden. Als Leitstruktur zum Entwurf der im Rahmen dieser Arbeit synthetisierten Verbindungen diente die Bis(ammonium)alkanverbindung W84. Über Weg A wurden Phthalsäure- bzw. Naphthalsäureanhydridderivate in einer Kondensationsreaktion mit dem entsprechenden N,N-Dimethylpropan-1,3-diaminderivat zum jeweiligen Phthalimidopropylaminderivat umgesetzt. Durch die Reaktion von zwei Äquivalenten des Amins mit einem Äquivalent 1,6-Dibromhexan wurden dann die symmetrischen W84-Derivate erhalten. Um die unsymmetrischen W84-Derivate zu erhalten, musste zunächst das jeweilige Phthalimidopropylamin einseitig durch 1,6-Dibromhexan alkyliert werden. Im letzten Schritt wurden äquimolare Mengen der alkylierten Verbindung und eines Phthalimidopropylamins umgesetzt. Da sich im Laufe der Arbeit die Methylierung an Position 2 der Propylketten als kritische Position zur Beeinflussung der Gleichgewichtsbindung herausstellte, wurden Verbindungen hergestellt, die an den Propylketten Alkylgruppen verschiedener Länge tragen. Aus diesem Grund wurde Syntheseweg B entwickelt. Zunächst wurden in mehreren Stufen, ausgehend von Malonsäurediethylester, einfach und zweifach mit Alkylgruppen substituierte 1,3-Dibrompropanderivate hergestellt. Diese wurden dann mit Kaliumphthalimid zu den jeweiligen 3-Brompropylphthalimidderivaten umgesetzt. Zwei Äquivalente dieser 3-Brompropylphthalimide reagierten mit einem Äquivalent N,N,N’,N’-Tetramethyl-1,6-hexandiamin zu den entsprechenden symmetrischen W84-Derivaten. Ein weiteres Ziel der Arbeit bestand darin, stark fluoreszierende W84-Derivate herzustellen. Die fluoreszierenden Eigenschaften N-substituierter Naphthalimide könnten zur direkten Charakterisierung allosterer Interaktionen oder zur Verfolgung des „Rezeptor-Traffickings“ mittels Fluoreszenzkorrelationsspektroskopie genutzt werden. Deshalb wurden in Position 3 und 4 des Naphthalimidringes des potentesten allosteren Modulators Aminogruppen eingeführt. Hexamethonio-Derivate beeinflussen in nennenswertem Maße bisher nur die Bindung von Antagonisten am M2-Rezeptor. Da die allostere und die orthostere Bindungsstelle räumlich nahe zusammenliegen, wurde der Versuch unternommen, einen orthosteren Agonisten und einen allosteren Modulator in einem Molekül miteinander zu verknüpfen. Es wurden zwölf Hybridmoleküle aus einem Teil des hochaffinen allosteren Modulators 3a und Derivaten des Muscarinagonisten Oxotremorin-M, verbunden durch aliphatische Spacer verschiedener Länge, hergestellt. In pharmakologischen Testungen soll aufgeklärt werden, ob es möglich ist, mit einem Agonist/Alloster-Hybridmolekül gleichzeitig die orthostere und die allostere Bindungsstelle zu besetzen. Die pharmakologische Testung der synthetisierten Verbindungen erfolgte durch Radioligandbindungsstudien. Der allostere Effekt der Testsubstanzen wurde indirekt über die Verzögerung der Dissoziation des radioaktiv markierten orthosteren Antagonisten [3H]N-Methylscopolamin bestimmt. Alle bisquartären Testverbindungen weisen deutlich höhere Affinitätswerte als die Leitstruktur W84 auf. Die 1,8-Naphthalimid-substituierten Verbindungen mit gleichzeitiger zweifacher Methylierung erwiesen sich als hochaffin und zugleich positiv kooperativ. Die wirksamste Verbindung dieser Serie ist Verbindung 3a (Naphmethonium), deren Affinität zum NMS-besetzten Rezeptor im einstelligen nanomolaren Bereich liegt (pEC50 = 8.36). Somit stellt Naphmethonium den potentesten in der Literatur bekannten allosteren Modulator des M2 Rezeptors dar. Mittels QSAR-Analysen wurden die ermittelten Affinitäten zum freien und zum NMS-besetzten Rezeptor in Zusammenhang mit verschiedenen physikochemischen Parametern gebracht. Die Affinität zum NMS-besetzten Rezeptor der Verbindungen der Serie 2 lässt sich mit hoher Güte durch das Volumen eines lateralen N-Methylimids in Kombination mit der benachbarten Dimethylierung der Propylkette beschreiben. Somit wird deutlich, dass zur Erzielung von positiver Kooperativität die Kombination aus einem hochaffinen aromatischen Imid in direkter Nachbarschaft zu einer 2,2-Alkylpropylkette essentiell ist.
Synthesis of Dualsteric Ligands for Muscarinic Acetylcholine Receptors and Cholinesterase Inhibitors

The study is dealing with the synthesis and pharmacological investigation of newly designed dualsteric ligands of muscarinic acetylcholine receptors belonging to the superfamily of G protein-coupled receptors. Such bipharmacophoric ligands combine the advantages of the orthosteric binding site (high-affinity) and of the topographically distinct allosteric binding site (subtype-selectivity) resulting in compounds with reduced side effects. This opens the way to a new therapeutic approach in the treatment of e.g. chronic pain, drug withdrawal, Parkinson`s and Alzheimer`s disease. Furthermore, the newly synthesized dualsteric compounds were pharmacologically investigated in order to get a better understanding of the activation and signaling processes in muscarinic acetylcholine receptors, especially with regard to partial agonism.
The development of the “dynamic ligand binding” concept offers new perspectives for ligand binding and signaling at G protein-coupled receptors. GPCRs are no longer considered as simple on/off switches. Dualsteric ligands can bind in a dualsteric pose, reflecting an active receptor state as well as in a purely allosteric binding pose, characterized by an inactive receptor state resulting in partial agonism. The degree of partial agonism depends on the ratio of active versus inactive receptor populations. On this basis, orthosteric/orthosteric hybrid ligands consisting of the antagonist atropine and scopolamine, respectively, as well as of the agonist iperoxo and isoxazole, respectively, linked via different alkyl chain length were synthesized in order to investigate partial agonism (Figure 1).

Furthermore, different sets of quaternary and tertiary homodimers consisting either of two iperoxo or two acetylcholine units were synthesized in order to study their extent on partial agonism (Figure 2). The two agonists were connected by varying alkyl chain length. Binding studies on CHO-hM2 cells of the quaternary compounds revealed that dimerization of the agonist results in a loss of potency. The iperoxo-dimers reached higher maximum effects on the Gi- as well as on the Gs pathway in comparison to the acetylcholine-dimers. Besides the choice of the orthosteric building block (potency of the agonist), the alkyl chain length is also crucial for the degree of partial agonism.

Quinolone-based hybrids connected to the superagonist iperoxo and to the endogenous ligand acetylcholine, respectively, linked through an alkyl chain of different length were synthesized in order to develop further partial agonists (Figure 3). FRET studies confirmed M1 subtype-selectivity as well as linker dependent receptor response. The greatest positive FRET signal was observed with quinolone-C6-iper resulting from a positive cooperativity between the two separated moieties, alloster and orthoster. However, the corresponding hybrids with a longer linker led to an inverse FRET signal indicating a different binding mode, e.g. purely allosteric, in contrast to the shorter linked hybrids. Furthermore, the flexible alkyl spacer was replaced by a rigidified linker resulting in the hybrid quinolone-rigid-iperoxo (Figure 3). FRET studies on the M1 receptor showed reduced FRET kinetics, resulting from interactions between the bulky linker and the aromatic lid, located between the orthosteric and allosteric binding site. A bitopic binding mode of the rigidified hybrid is presumed. For further clarity, mutational studies are necessary.

Another aim of this work was the design and synthesis of new hybrid compounds, acting as agonists at the M1 and M2 receptor and as inhibitors for AChE and BChE in the context of M. Alzheimer. Several sets of hybrid compounds consisting of different pharmacophoric units (catalytic active site: phthalimide, naphthalimide, tacrine; peripheric anionic site: iperoxo, isoxazole) linked through a polymethylene chain of varying length were synthesized. Tac-C10-iper (Figure 4), consisting of tacrine and the superagonist iperoxo linked by a C10 polymethylene spacer, was found to have excellent anticholinesterase activity for both AChE (pIC50 = 9.81) and BChE (pIC50 = 8.75). Docking experiments provided a structural model to rationalize the inhibitory power towards AChE. Additionally, the tacrine related hybrids showed affinity to the M1 and M2 receptor. Such compounds, addressing more than one molecular target are favorable for multifactorial diseases such as Alzheimer.

In summary, the choice of the pharmacophoric units, their connecting point as well as the nature, length, and flexibility of the linker play an important role for the activity of designed bivalent ligands. A shorter linker length cannot bridge both binding sites simultaneously in contrast to longer linker chains. On the other hand, too long linker chains can result in unwanted steric interactions. Further investigations with respect to structural variations of hybrid compounds, with or without quaternary ammonium groups, are necessary in the light of drug development.
Die Subtypselektivität von Liganden für einzelne Rezeptoren, deren Aktivierung und die anschließende Signalweiterleitung sind bis heute weitestgehend ungeklärt. Die hier synthetisierten Liganden-Gruppen sollen helfen, die verschiedenen Prozesse am muskarinischen Rezeptor und seinen Subtypen zu verstehen. Die Einzelprojekte werden im Folgenden vorgestellt. 1) Um mittels FRET-Mikroskopie den Einfluss von allosteren Modulatoren, die sich von W84 bzw. Naphmethonium ableiten, in Bezug auf die Konformationsänderung aktivierter Rezeptoren untersuchen zu können, wurden die bekannten allosteren Bausteine sowie eine Reihe neuer Derivate synthetisiert. Alle untersuchten Substanzen zeigten einen hemmenden Effekt auf die mit dem Agonisten Iper-oxo vorstimulierten Rezeptoren. Das heißt, die Verbindungen ließen sich als negative allostere Modulatoren charakterisieren. 2) Da Iperoxo aufgrund seiner großen agonistischen Aktivität ein interessantes Werkzeug für die Grundlagenforschung darstellt, war es von großer Bedeutung, eine schnelle und reproduzierbare Synthese zu gewährleisten. Ausgehend von Propargylalkohol wurde in einer Mannich-Reaktion 4-Dimethylamino-but-2-en-1-ol gebildet, was mit dem zuvor hergestellten 3-Nitro-Δ2-isoxazolin zur Iperoxo-Base umgesetzt wurde. Neben einer deutlichen Ausbeutesteigerung ist nun die Reproduzierbarkeit im Gegensatz zu der von Dallanoce et al. publizierten Synthese gewährleistet. 3) Um den Einfluss der Kettenlänge der Hybride Iper-6-Phth und Iper-6-Naph auf die agonistische Aktivität und Subtypselektivität der Substanzen untersuchen zu können, wurde versucht, Hybride verschiedener Kettenlängen herzustellen. Dabei konnte Iper-4-Phth erhalten werden. 4) Weiterhin sollte der Einfluss der Alkylkette am Stickstoff-Atom auf die Wirksamkeit in Bezug auf Affinität und Zellantwort des Iperoxo-Moleküls ohne allosteren Modulator analysiert werden. Hierzu wurden die N-alkylierten Iperoxo-Derivate mit den Kettenlängen C2 bis C10 synthetisiert.. Mithilfe von Radioligand-Bindungsstudien und der dynamischen Massenumverteilung sollte die konformative Änderung des Rezeptors durch Aktivierung und die Signalweiterleitung untersucht werden. Durch Verlängerung der N-Alkylkette zeigte sich ein Wirksamkeitsverlust, d. h. die Dosis-Wirkungs-Kurven der prozentualen Zellantwort wurden im Vergleich zu denen des Iperoxos nach rechts verschoben. In Untersuchungen an der Rezeptor-Mutante CHO-hM2-Y1043.33A zeigte sich zudem, dass für den maximalen Effekt eine deutlich höhere Konzentration der Iperoxo-Derivate benötigt wird, wobei dieser Wirksamkeitsverlust im Vergleich zum Rezeptor-Wildtyp für Iperoxo selbst am stärksten ausgeprägt ist. Allerdings weisen Iperoxo und seine N-alkylierten Derivate an dieser Mutante eine höhere intrinsische Aktivität auf als die Kontrollverbindungen Oxotremorin M, C1-IP-C1 und Acetylcholin. Weiterhin konnte gezeigt werden, dass Iperoxo ebenso wie Oxotremorin, Acetylcholin, aber auch die kurzkettigen Iperoxo-Derivate neben dem Gi-Signalweg auch den Gs-Weg aktivieren können, wohingegen die langkettigen Derivate eine Gi-Signalwegs-Selektivität aufweisen. 5) In Analogie zu den N-alkylierten-Iperoxo-Derivaten sollten Untersuchungen mit den Antagonisten N-Alkyl-Atropin und -Scopolamin durchgeführt werden. In beiden Fällen zeigte das N-Methyl-Derivat eine höhere Affinität zum Rezeptor als der jeweilige Antagonist selbst, was auf die durch Alkylierung generierte positive Ladung zurückzuführen ist. Durch Verlängerung der Alkylketten ist jeweils eine Abnahme der Affinität zu beobachten. Die Affinität findet ebenfalls in beiden Fällen mit dem Butyl-Derivat ihr Minimum. Der anfängliche Affinitätsverlust lässt sich durch die zunehmende sterische Hinderung des Moleküls erklären, der spätere Anstieg bei einer Alkylkette länger als C4 deutet auf eine Wechselwirkung dieser langen Alkylkette mit einer weiteren (allosteren) Bindungsstelle hin. 6) Ausgehend von Iperoxo sollten dualstere Liganden entwickelt werden, die durch geeignete allostere Modulatoren selektiv nur einen Rezeptor-Subtyp adressieren und diesen durch Iperoxo aktivieren sollten. Als allostere Bausteine sollten die M4-selektiven Thienopyridine und die M1-selektiven Chinolone verwendet werden. 7) Zur Fluoreszenzmarkierung sollte Iperoxo-Base zudem mit dem Farbstoff Py-1 umgesetzt werden.
The study deals with the area of the allosteric modulation of the muscarinic M2 receptors. The allosteric modulators have an influence on binding of orthosteric ligands (agonists and antagonists) to the classical orthosteric binding site of the muscarinic M2-receptors. The modulators are able to enhance (positive cooperativity) or decrease (negative cooperativity)the affinity of ligands to the orthosteric binding site. The allosteric binding site is located at the entrance of the receptor binding pocket. It is less conserved than the orthosteric binding site which is located in a narrow cavity created by the seven transmembrane domains. Consequently, development of subtype selective allosteric ligands is easier than subtypeselective muscarinic agonists or antagonists. Furthermore, subtype selectivity can be achieved by differently cooperative interactions between the allosteric and orthosteric ligand at different receptor subtypes. For example, the allosteric modulators that are positively cooperative with ACh at M1 receptors and neutrally cooperative at the other receptor subtypes could be beneficial for treatment of the Alzheimer’s disease. Bisquaternary analogues of the Strychnos alkaloid caracurine V are among the most potent allosteric modulators of muscarinic M2-receptors. The very rigid ring skeleton comprises the pharmacophoric elements of two positively charged nitrogens at an approximate distance of 10 surrounded by two aromatic ring systems in a distinct spatial arrangement. Owing to the close structural relationship of caracurine V salts to the strong muscle relaxants toxiferine and alcuronium, they are likely to exhibit neuromuscular blocking activity, which would limit their usefulness as research tools and make the therapeutical use impossible. Reduction of the caracurine V ring skeletons to structural features responsible for good allosteric potency could possibly lead to compounds with negligible neuromuscular blocking activity and very high affinity to the allosteric binding site at M2 receptor. Thus, the aim of this study was to synthesize and pharmacologically evaluate analogues of a novel heterocyclic ring system, which comprises the pharmacophoric elements mentioned previously. The key step of the synthesis of the desired 6,7,14,15-tetrahydro[1,5]diazocino[1,2-a:6,5-a]-diindole ring system (6) involved the intermolecular double N-alkylation of the bromoethylindole (5), which was prepared from the known indolyl methylacetate (3) by reduction of the ester group to alcohol and subsequent substitution by bromine. 3 could be prepared in three steps involving N,N-dibenzylation of tryptamine followed by introduction of the dimethyl malonate moiety at C-2 of indole ring and a subsequent demethoxycarbonylation. The total synthesis of 6,7,14,15-tetrahydro[1,5]diazocino[1,2-a:6,5-a]diindole ring system (6) is shown in Scheme 24. In order to examine the influence of the length of the side-chain on muscarinic activity,exchange of the ethylamine moieties of 14 by the methylamino groups was planned. This should be accomplished by dimerization of the unsubstituted 2-bromoethylindole (32), and subsequent Mannich aminomethylation of the resulting unsubstituted pentacyclic ring. The total synthesis of the 6,7,14,15-tetrahydro-15aH-azocino[1,2-a:6,5-b]diindole ring system(35) is shown in Scheme 25. 32 was prepared from indole-2-carboxylic acid in six steps involving reduction of the acid to the corresponding alcohol 26, benzoylation of 26 followed by nucleophilic substitution with KCN, hydrolysis of the cyanide 28 to indolyl acetic acid 29,reduction of 29 to the corresponding alcohol 30, and finally bromination of 30 to give the bromide 32. Since dimerization attempts of 32 provided only 2-vinylindole (33), the tosylate 34 was used as starting material for the intermolecular alkylation to give exclusively an isomeric pentacyclic ring system, 7,14,15-tetrahydro-15aH-azocino[1,2-a:6,5-b]diindole (35). The formation of the novel, asymmetric ring skeleton can be explained by the ambident nucleophilic character of the indolyl anion that can be alkylated either at nitrogen or at C-3 of indole ring. 35 was subjected to a Mannich reaction to give 2,13-dimethylaminoalkylated product 37 as well as small amounts of the 13-monosubstituted compound (36). The geometry of novel ring systems 6 was elucidated by means of NMR spectroscopy and semiempirical calculations. The diazocinodiindole ring skeleton of 6 exists in chloroform solution at room temperature in a twisted-boat conformation, as indicated by 600 MHz ROESY experiment, vicinal coupling constants within the eight-membered ring, and AM1 calculations. In order to obtain potent allosteric ligands, the new heterocycles 6 and 37 were quarternized with methyliodide to the corresponding ammonium salts 14 and 38, respectively. Additionally, the N,N -diallylsalts of 37 (compound 39) was prepared. The allosteric effect of 14, 38, and 39 on the dissociation of the orthosteric radioligand [3H]Nmethylscopolamine([3H]NMS) and their effects on [3H]NMS equilibrium binding were studied in homogenates of porcine heart ventricles. The concentration of an allosteric agent for a half-maximum effect on orthosteric ligand dissociation (EC50,diss) corresponds to a 50 % occupancy of the liganded receptors by the respective allosteric test compounds. Due to the presence of two benzyl groups on each nitrogen in the side chains of 14, its binding affinity can be best compared with that of N,N -dibenzylcaracurinium V dibromide (EC50,diss = 69 nM). Compound 14 exhibited the comparable affinity to N,N -dibenzylcaracurinium V dibromide with EC50,diss = 54 nM. This result suggested that replacement of the bulky benzyl groups of 14 by smaller substitutents will probably increase the allosteric potency, since dimethyl- and diallylcaracurinium salts showed a 5-fold increase of binding affinity relative to the dibenzyl analogue. Even though the new azocinodiindole ring system of 38 and 39, is not included in the caracurine V ring skeleton, it comprises the essentially pharmacophoric elements of allosteric potency. Due to the different spatial arrangements of the aromatic rings, as well as to different internitrogen distances in both ring systems, compound 38 and 39 exhibited 4-fold lower M2 binding affinity (EC50,diss = 35 and 48 nM, respectively) than the corresponding caracurine V analogues. This study deals with the synthesis of the first representative (Compound 6) of a novel pentacyclic ring system derived from caracurine V. The high allosteric potency of its dimethyl analogue reveals the [1,5]diazocino[1,2-a:6,5-a]-diindole ring system as a new promising lead structure for allosteric modulators of muscarinic M2 receptors. Future research will be focused on structural modifications of the new ring system in order to increase the affinity to the muscarinic receptors. Furthermore, the binding affinities of the new synthesized compounds to the muscle type of nicotinic ACh-receptor should reveal structural features responsible for the muscarinic/nicotinic selectivity.
G protein-coupled receptors (GPCRs) form the biggest receptor family that is encoded in the human genome and represent the most druggable target structure for modern therapeutics respectively future drug development. Belonging to aminergic class A GPCRs muscarinic Acetylcholine receptors (mAChRs) are already now of clinical relevance and are also seen as promising future drug targets for treating neurodegenerative diseases like Alzheimer or Parkinson. The mAChR family consist of five subtypes showing high sequence identity for the endogenous ligand binding region and thus it is challenging until now to selectively activate a single receptor subtype. A well accepted method to study ligand binding, dynamic receptor activation and downstream signaling is the fluorescence resonance energy transfer (FRET) application. Here, there relative distance between two fluorophores in close proximity (<10 nm) can be monitored in a dynamic manner. The perquisite for that is the spectral overlap of the emission spectrum of the first fluorophore with the excitation spectrum of the second fluorophore. By inserting two fluorophores into the molecular receptor structure receptor FRET sensors can serve as a powerful tool to study dynamic receptor pharmacology.
Dualsteric Ligands consist of two different pharmacophoric entities and are regarded as a promising ligand design for future drug development. The orthosteric part interacts with high affinity with the endogenous ligand binding region whereas the allosteric part binds to a different receptor region mostly located in the extracellular vestibule. Both moieties are covalently linked. Dualsteric ligands exhibit a dynamic ligand binding. The dualsteric binding position is characterized by a simultaneous binding of the orthosteric and allosteric moiety to the receptor and thus by receptor activation. In the purely allosteric binding position no receptor activation can be monitored.
In the present work the first receptor FRET sensor for the muscarinic subtype 1 (M1) was generated and characterized. The M1-I3N-CFP sensor showed an unaltered physiological behavior as well as ligand and concentration dependent responses. The sensor was used to characterize different sets of dualsteric ligands concerning their pharmacological properties like receptor activation. It was shown that the hybrids consisting of the synthetic full agonist iperoxo and the positive allosteric modulator of BQCA type is very promising. Furthermore, it was shown for orthosteric as well as dualsteric ligands that the degree of receptor activation is highly dependent on the length of and the chemical properties of the linker moiety. For dualsteric ligands a bell-shaped activation characteristic was reported for the first time, suggesting that there is an optimal linker length for dualsteric ligands. The gained knowledge about hybrid design was then used to generate and characterize the first photo-switchable dualsteric ligand. The resulting hybrids were characterized with the M1-I3N-CFP sensor and were described as photo-inactivatable and dimmable. In addition to the ligand characterization the ligand application methodology was further developed and improved. Thus, a fragment-based screening approach for dualsteric ligands was reported in this study for the first time. With this approach it is possible to investigate dualsteric ligands in greater detail by applying either single ligand fragments alone or in a mixture of building blocks. These studies revealed the insights that the effect of dualsteric ligands on a GPCR can be rebuild by applying the single building blocks simultaneously. The fragment-based screening provides high potential for the molecular understanding of dualsteric ligands and for future screening approaches. Next, a further development of the standard procedure for measuring FRET by sensitized emission was performed. Under normal conditions single cell FRET is measured on glass coverslips. After coating the coverslips surface with a 20 nm thick gold layer an increased FRET efficiency up to 60 % could be reported. This finding was validated in different approaches und in different configurations. This FRET enhancement by plasmonic surfaces was until yet unreported in the literature for physiological systems and make FRET for future projects even more powerful.
Im Rahmen der vorliegenden Arbeit wurden neuartige siliciumhaltige Muscarinrezeptor-Antagonisten, Dopaminrezeptor-Antagonisten, Ca2+-Kanal-Blocker sowie alpha1-Rezeptor-Antagonisten synthetisiert, welche Sila-Analoga (C/Si-Austausch) bekannter organischer Pharmaka darstellen. Die C/Si-Analoga wurden pharmakologisch charakterisiert und damit Beiträge zur Thematik der C/Si-Bioisosterie geleistet.
As part of the parasympathetic nervous system, muscarinic receptors are involved in the regulation of numerous functions in the human body. However, targeting a specific subtype of muscarinic receptors is challenging due to the high degree of similarity within the binding site of the endogenous neurotransmitter acetylcholine. Therefore, this study focused on the investigation of dualsteric ligands. Such hybrid ligands target the orthosteric acetylcholine binding site and, simultaneously, a distinct allosteric binding site. Since allosteric binding regions show significant structural differences throughout muscarinic receptor subtypes, it was aimed to produce selective ligands by means of combination of two pharmacophores in one molecule. Herein, the thienopyridine derivatives LY2033298 and LY2119620 were chosen as allosteric moieties. Based on literature studies, the investigated allosteric modulators were analyzed in terms of adequate attachment points for the combination with an orthosteric agonist. As orthosteric units, muscarinic superagonist iperoxo, xanomeline, and TMA were applied in this work. Since the distance between orthosteric and allosteric moieties plays a crucial role for dualsteric ligand binding, the linker chain length was also varied. Pharmacological investigations of the synthesized hybrid ligands were perfomed via FRET- and BRET-assay measurements.
Einwärtsgleichrichtende Kaliumkanäle (Kir), aktuell in die 7 Unterfamilien Kir1-Kir7 eingeteilt, sind an der Regulation einer Vielzahl von Körperfunktionen, beispielsweise Herzfrequenz, Erregbarkeit von Nervenzellen, Tonus von Gefäßmuskelzellen, Hormonsekretion oder Aktivierung von Immunzellen, beteiligt. Für die Kontrolle dieser Funktionen ist es von entscheidender Bedeutung, dass die Leitfähigkeit dieser Kanäle beeinflusst werden kann. Die Kir3-Unterfamilie (früher GIRK für G-protein-activated-K+-channels) wird beispielsweise obligat durch die direkte Bindung der beta/gamma-Untereinheit des trimeren Gi/0-Proteins aktiviert (Karschin, 1999). Es gibt Hinweise in der Literatur, dass auch die stark einwärts gleichrichtenden Kanäle der Kir2-Familie durch G-Proteine der Gq-Familie reguliert sein können. Dabei widersprechen sich insbesondere zwei Untersuchungen zur Spezifität der Interaktion (Jones, 1996; Chuang et al., 1997). Ebenso ist der intrazelluläre Signalweg bislang nicht hinreichend geklärt. Um dies genauer zu untersuchen, wurden in dieser Arbeit die Kir-Kanäle Kir2.1-Kir2.4 jeweils mit 5 verschiedenen Gq-gekoppelten Rezeptoren in Xenopus-Oozyten koexprimiert und mit der Technik der „Zwei-Elektroden-Spannungsklemme“ der Strom über die Kir-Kanäle vor und nach Rezeptoraktivierung mit dem jeweils physiologischen Rezeptoragonisten gemessen. Es zeigte sich, dass ausschließlich Kir2.3 nach Aktivierung des M1-Acetylcholinrezeptors inhibiert wird. Eine Sequenzanalyse zeigte in der Extrazellulärregion von Kir2.3 eine zu den anderen Kir2-Kanälen abweichende Aminosäuresequenz, welche durch Mutation aber als potentielle Bindestelle zur Vermittlung des inhibitorischen Effektes ausgeschlossen werden konnte. Nachdem bereits gezeigt werden konnte, dass die Koexpression von Kir2.3 und M1-Acetylcholinrezeptor in bestimmten Gehirnregionen der Kontrolle neuronaler Erregbarkeit dient (Shen et al., 2007), ist es wahrscheinlich, dass derselbe Mechanismus auch in ventrikulären Kardiomyozyten existiert und dort als Schutzmechanismus vor vagaler Überstimulation fungiert.
Zahlreiche experimentelle Befunde lassen vermuten, dass G-Protein gekoppelte Rezeptoren (GPCR) nach ihrer Aktivierung einer ligandenselektiven Änderung der Rezeptorkonformation unterliegen. Ziel der vorliegenden Arbeit war es dieses Phänomen am Subtyp 2 der muskarinischen Acetylcholinrezeptoren (M2 AChR) zu untersuchen. Muskarinische Acetylcholinrezeptoren (mAChR) können in fünf Subtypen (M1-M5) unterschieden werden. Durch die Beteiligung der mAChR an zahlreichen physiologischen Prozessen stellen sie wichtige Zielstrukturen pharmakologischer Therapien dar. Da die orthosterische Ligandenbindestelle (= Bindestelle des endogenen Liganden) in allen fünf Subtypen hoch konserviert ist, wird ihr pharmakologischer Nutzen derzeit allerdings durch die unselektive Rezeptormodulation und dem damit verbundenen Auftreten unerwünschter Arzneimittelwirkungen stark limitiert. Ein Ansatz zur Erzielung subtypselektiver Effekte besteht in der Verwendung allosterischer Modulatoren. Da die allosterische Bindestelle der mAChR eine geringere Sequenzhomologie aufweist, können so gezielt einzelne Subtypen der mAChR reguliert werden. Der M2 AChR stellt hinsichtlich allosterischer Modulation ein gut charakterisiertes Modellsystem dar. Für ihn wurde bereits eine Vielzahl allosterischer Liganden entwickelt. Auch bitopische Liganden, die sowohl einen allosterischen als auch einen orthosterischen Anteil enthalten, wurden für den M2 AChR bereits beschrieben. Im ersten Teil der vorliegenden Arbeit wurden verschiedene FRET-Sensoren des M2 AChR generiert und charakterisiert. Als FRET-Paar wurden das cyan fluoreszierende Protein (CFP) und der niedermolekulare fluorescein-basierte Fluorophor FlAsH (fluorescein arsenical hairpin binder) gewählt. CFP wurde in den Sensoren am Ende des C-Terminus angefügt. Die zur Markierung mit FlAsH nötige Tetracysteinsequenz wurde in verschiedenen Bereichen der dritten intrazellulären Rezeptorschleife (IL) eingebracht. Die auf diese Weise erstellten Re-zeptorsensoren trugen das Tetracysteinmotiv in der N terminalen (M2i3-N) bzw. in der C terminalen Region (M2i3-C) von IL 3. Die Charakterisierung der Rezeptorsensoren bezüglich Ligandenbindung, Gi-Protein Aktivierung und β-Arrestin2 Translokation ergab keine signifikanten Unterschiede zwischen M2i3-N, M2i3 C und M2CFP oder Wildtyp M2 AChR. Zunächst wurden sowohl unterschiedliche orthosterische, als auch allosterische Liganden hinsichtlich ihrer mittleren effektiven Konzentration und ihrer maximalen Wirkstärke an den Rezeptorsensoren untersucht. Mit Hilfe von FRET-Messungen konnte ein superago-nistisches Verhalten des orthosterischen Testliganden Iperoxo gezeigt werden. Die Eigenschaften der allosterischen Substanzen wurden durch Messung der Rezeptordeakti-vierungskinetik und des maximalen Hemmeffekts auf einen vorstimulierten Rezeptor charakterisiert. Alle allosterischen Liganden deaktivierten den vorstimulierten M2 AChR mit einer schnelleren Kinetik als Atropin. Die EC50-Werte der unterschiedlichen Substanzen waren unabhängig von der Markierungsposition im verwendeten Rezeptorsensor vergleich-bar. Ausnahmen bildeten die allosterischen Liganden JK 289, JK 338, ½ W84 und EHW 477, die liganden- und sensorabhängig unterschiedliche mittlere effektive Konzentrationen aufwie-sen. Bei der Untersuchung der Konformationsänderung des M2 AChR konnte kein liganden-selektiver Unterschied zwischen den FRET-Signalen für 19 getestete orthosterische Liganden beobachtet werden. Dies deutet darauf hin, dass alle orthosterischen Testliganden eine dem Acetylcholin (ACh) vergleichbare Änderung der M2 AChR Konformation induzier-ten. Um zu untersuchen, ob für die orthosterischen Testliganden eine Korrelation zwischen ihrer maximalen Wirkstärke hinsichtlich Rezeptoraktivierung in FRET-Experimenten und der Aktivierung nachgeschalteter Signalwege besteht, wurde die orthosterisch-vermittelte Translokation von β-Arrestin2 mit Hilfe der Konfokalmikroskopie bestimmt. Bis auf 5-Methyl-furmethiodid translozierten alle orthosterischen Liganden β-Arrestin2 in einem Ausmaß, das mit der maximalen Rezeptoraktivierung vergleichbar war. Dagegen rief 5 Methylfurmethiodid verglichen mit dem endogenen Liganden ACh zwar eine ca. halbmaximale Rezeptorakti-vierung, aber nur eine äußerst geringe β-Arrestin2 Translokation hervor. Im zweiten Teil der Arbeit wurde der Einfluss verschiedener Allostere auf eine ligandenselektive Konformationsänderung des M2 AChR untersucht. Die allosterischen Liganden JK 337 und Seminaph beeinflussten den M2i3-C Sensor signifikant stärker, als das M2i3-N Konstrukt. Dagegen zeigte EHW 477 eine stärkere Beeinflussung der Rezeptorkon-formation des M2i3-N-, als des M2i3-C Sensors. Dies erlaubt die Vermutung, dass JK 337 und Seminaph eine stärkere Bewegung unterhalb von Transmembrandomäne (TM) 6, als unterhalb von TM 5 hervorriefen. Die Ergebnisse für EHW 477 legen nahe, dass TM 5 eine größere Bewegung eingeht, als TM 6. FRET-basierte Untersuchungen der Einflüsse der allosterischen Testliganden auf nachgeschaltete Signalwege ergaben, dass sowohl die Akti-vierung des Gi Proteins, als auch die β-Arrestin2 Translokation selektiv durch einzelne allosterische Liganden beeinflusst werden. Auch ein Zusammenhang zwischen Rezeptor-aktivierung und der Regulation nachgeschalteter Signalwege war erkennbar. Allerdings waren auf Grund der Versuchsbedingungen keine quantitativen Aussagen möglich. Im Folgenden wurden die bitopischen Liganden Hybrid 1 und 2 (H 1, H 2) hinsichtlich ihres Effekts auf die Konformationsänderung des M2 AChR untersucht. Während eine Stimulation mit H 1 vergleichbare FRET-Signale an beiden Sensoren ergab, konnte mit H 2 weder am M2i3-N-, noch an M2i3-C Sensor eine FRET-Änderung detektiert werden. Um den mangeln-den Effekt der Hybridsubstanzen in FRET-mikroskopischen Untersuchungen aufzuklären, wurden verschiedene Ansätze gewählt: Mit kettenverlängerten Derivaten der Hybridsubstanzen konnte in FRET-Messungen eine Änderung des FRET-Signals detektiert werden. Die Entfernung des allosterischen Bausteins führte in FRET-Experimenten zu einer verglichen mit dem endogenen Liganden ACh etwa halbmaximalen Aktivierung beider Sensoren. Dagegen resultierte die Mutation der alloste-rischen Bindestelle in nachfolgenden FRET-Untersuchungen mit H 1 und H 2 in keiner Signaländerung des FRET-Ratio. Diese Beobachtungen führten zu der Annahme, dass die Linkerkette, die orthosterischen und allosterischen Baustein der Hybride miteinander verbindet, zu kurz war um eine gleichzeitige Bindung an die allosterische und orthosterische Bindestelle zu ermöglichen. Ein anderer Erklärungsansatz besteht darin, dass nach Bindung des Orthosters der Kanal zwischen orthosterischer und allosterischer Bindestelle durch die Konformationsänderung des Rezeptors verschlossen wird, weshalb keine dauerhafte, dualsterische Bindung der Hybridsubstanzen an den M2 AChR möglich ist. Im Rahmen der vorliegenden Arbeit ist es gelungen mittels FRET-Experimenten die Existenz einer ligandenselektiven Rezeptorkonformation des M2 AChR mit allosterischen Liganden nachzuweisen. Darüber hinaus konnte auch ein Bezug zum Auftreten einer funktionellen Selektivität mit allosterischen Liganden hergestellt werden. Die Untersuchung von 19 orthosterischen Liganden hinsichtlich ihres Einflusses auf die Rezeptorkonformation des M2 AChR ergab keinen Hinweis auf eine ligandenselektive Konformationsänderung. Die Betrachtung der orthosterisch-vermittelten Translokation von β-Arrestin2 zeigte, dass zwischen der Effizienz der orthosterischen Testliganden, den M2 AChR zu aktivieren und dem Ausmaß, in dem sie eine β Arrestin2 Translokation induzierten eine direkte Korrelation besteht. Lediglich 5-Methylfurmethiodid rief eine ungleich geringere β-Arrestin2 Translokation hervor, verglichen mit dem Ausmaß an Rezeptoraktivierung. Diese Beobachtung deutet auf die Existenz eines signaling-bias für diesen Liganden hin. Die Untersuchung der dualsterischen Liganden H 1 und 2 bezüglich ihrer Fähigkeit zur Rezeptoraktivierung ergab, dass erst durch eine Verlängerung der Linkerkette, durch die orthosterischer und alloste-rischer Baustein miteinander verbunden sind eine Konformationsänderung des M2 AChR hin zu einer aktiven Konformation erreicht werden kann. Es kann somit angenommen werden, dass in den ursprünglichen Hybridsubstanzen H 1 und H 2 eine zu kurze Linkerkette, durch die keine dualsterische Bindung der Hybride an die allosterische und orthosterische Bindestelle möglich ist, ursächlich für die mangelnde Rezeptoraktivierung des M2 AChR war.
G protein-coupled receptor research looks out for new technologies to elucidate the complex
processes of receptor activation, function and downstream signaling with spatiotemporal
resolution, preferably in living cells and organisms. A thriving approach consists in making use
of the unsurpassed properties of light, including its high precision in space and time, noninvasiveness
and high degree of orthogonality regarding biological processes. This is realized
by the incorporation of molecular photoswitches, which are able to effectively respond to light,
such as azobenzene, into the structure of a ligand of a given receptor. The muscarinic
acetylcholine receptors belong to class A GPCRs and have received special attention in this
regard due to their role as a prototypic pharmacological system and their therapeutic potential.
They mediate the excitatory and inhibitory effects of the neurotransmitter acetylcholine and
thus regulate diverse important biological processes, especially many neurological functions in
our brain.
In this work, the application of photopharmacological tool compounds to muscarinic receptors
is presented, consisting of pharmacophores extended with azobenzene as light-responsive
motif. Making use of the dualsteric concept, such photochromic ligands can be designed to bind
concomitantly to the orthosteric and allosteric binding site of the receptor, which is
demonstrated for BQCAAI (M1) and PAI (M2) and may lead to subtype- and functionalselective
photoswitchable ligands, suitable for further ex vivo and in vivo studies.
Moreover, photoswitchable ligands based on the synthetic agonist iperoxo were investigated
extensively with regard to their photochemical behavior and pharmacological profile, outlining
the advantages and challenges of using red-shifted molecular photoswitches, such as tetraortho-
fluoro azobenzene. For the first time on a GPCR it was examined, which impact the
different substitution pattern has on both the binding and the activity on the M1 receptor. Results
show that substituted azobenzenes in photopharmacological compounds (F4-photoiperoxo and
F4-iper-azo-iper) not just represent analogs with other photophysical properties but can exhibit
a considerably different biological profile that has to be investigated carefully.
The achievements gained in this study can give important new insights into the binding mode
and time course of activation processes, enabling precise spatial and temporal resolution of the
complex signaling pathway of muscarinic receptors. Due to their role as exemplary model
system, these findings may be useful for the investigation into other therapeutically relevant
GPCRs.
