Die NO/cGMP-vermittelte Signalkaskade ist im vaskulären System entscheidend an der Regulation des Blutdrucks beteiligt. Innerhalb der Kaskade nimmt die NO-sensitive Guanylyl-Cyclase (NO-GC) eine Schlüsselfunktion als wichtigster Rezeptor für das Signalmolekül Stickstoffmonoxids (NO) ein. NO wird endogen von verschiedenen Isoformen der NO Synthase produziert. Die Bindung von NO an die NO GC führt zur Produktion des sekundären Botenstoffs cyclisches Guanosinmonophosphat (cGMP). Dieser Botenstoff aktiviert verschiedene Effektor-Moleküle und bewirkt letztlich eine Relaxation der glatten Muskulatur. Ein weiterer sekundärer Botenstoff, das Signalmolekül cyclisches Adenosinmonophosphat (cAMP), ist ebenfalls an der Regulation des Tonus der glatten Muskulatur und dadurch an der Blutdruckregulation beteiligt. Unterschiedliche Phosphodiesterasen (PDE) bauen die sekundären Botenstoffe ab und beenden dadurch die Signalkaskaden. Die PDE3 spielt hierbei eine besondere Rolle, da sie eine gemischte Substratspezifität besitzt. Um den Einfluss der NO-GC auf das kardiovaskuläre System zu untersuchen, wurden NO-GC Knockout(KO)-Mäuse mit globaler (GCKO) oder Glattmuskel-spezifischer (SMC-GCKO) Deletion der NO-GC generiert.
Um das Zusammenspiel von cAMP und cGMP näher zu beleuchten, wurde im ersten Teil dieser Arbeit die PDE3 genauer untersucht. Im Gefäßsystem wird lediglich die PDE3A und nicht die PDE3B exprimiert. Die Aorten von GCKO- und SMC-GCKO-Tieren reagieren sensitiver auf PDE3A-Blockade als die Kontroll-Tiere. Auch die akute Blockade der NO-GC führt zu diesem Sensitivitätseffekt. Die PDE3A ist in Folge der NO-GC-Deletion sowohl in ihrer Expression, als auch ihrer Aktivität um die Hälfte reduziert. Dies dient vermutlich kompensatorisch dazu, das cAMP-Signal weitgehend zu erhalten und so eine cAMP-induzierte Relaxation der Gefäße zu gewährleisten. Ohne Rückkopplung zwischen den beiden Signalwegen käme es vermutlich zu weiteren negativen Konsequenzen für das Herz-Kreislaufsystem. Diese Daten weisen auf eine direkte Regulation der PDE3 in glatten Muskelzellen durch die NO/cGMP-Signalkaskade und einen PDE3-vermittelten cAMP/cGMP-Crosstalk hin. Der genaue Mechanismus dieser Expressionsregulation ist noch unklar. Denkbar wäre eine cGMP-vermittelte Transkriptionsregulation oder eine Modulation der Translation der PDE3A.
Der Verlust der NO-GC führt in GCKO- und SMC-GCKO-Mäusen zu einem erhöhten systolischen Blutdruck von ~30 mmHg. Bei der Entwicklung der arteriellen Hypertonie könnte eine erhöhte Aortensteifigkeit beteiligt sein, die im zweiten Teil dieser Arbeit näher untersucht wurde. In GCKO-Mäusen ist die aortale Steifigkeit und daraus resultierend die Pulswellengeschwindigkeit (PWV) deutlich erhöht. Die Steigerung der PWV wird in den GCKO-Tieren zusätzlich durch den verminderten Aorten-Durchmesser bedingt. Außerdem weisen die Aorten dieser Tiere eine veränderte Wandstruktur auf, die zu einer Verminderung der aortalen Windkesselfunktion führt. Diese Veränderungen könnten die Blutdruckerhöhung in GCKO-Mäusen erklären. In SMC-GCKO-Tieren tritt keine dieser Gefäß-Modifikationen auf. Eine Aortensteifigkeit als mögliche Ursache für den erhöhten systolischen Blutdruck in den SMC-GCKO-Tieren kann somit ausgeschlossen werden. Zur Aufklärung müssen weitere Versuche zum Aufbau der Gefäßwände und zur Bestimmung des peripheren Widerstands gemacht werden. Auch der Einfluss anderer Zelltypen, wie z.B. Perizyten oder Fibroblasten, auf die Blutdruckregulation sollte untersucht werden.
Ziel der Arbeit ist es, verschiedene Knochenersatzmaterialien der Tympanoplastik Typ 3
(autogenes Gewebe, Titan, Ionomerzement) bezüglich ihres Langzeitverhaltens im Mittelohr zu vergleichen. Es werden zwischen dem 21.12.1995 und dem 30.04.2011 in der Hals-Nasen-Ohrenklinik des Städtischen Klinikums Solingen operierte Patienten nachuntersucht. Insgesamt handelt es sich um 957 mit einer Tympanoplastik Typ III versorgte Patienten, die in diesem Zeitraum insgesamt 1093mal operiert worden sind. 676mal ist die Kette mit einer Titanprothese rekonstruiert worden, davon 301mal mit einer PORP und 375mal mit einer TORP (davon 21 bei intakter Stapessuprastruktur). Zu Beginn des Beobachtungszeitraums sind 56 Ionomerzement-prothesen eingesetzt worden, so dass 40 Ionomerzement-PORP und 16 Ionomerzement-TORP mit berücksichtigt worden sind. In 19 Fällen sind „sonstige“ Methoden (z.B. Knorpelüberhöhung des Steigbügels) zur Gehörknöchelchenkettenrekonstruktion gewählt worden.
Die Untersuchung zeigt, dass zur Kettenrekonstruktion eine Transposition autogener Ossikel angestrebt werden sollte. Stehen diese nicht zur Verfügung, empfiehlt sich die Verwendung von Titan-Prothesen. Aufgrund ihres hervorragenden In-Situ-Verhaltens sowie der nachgewiesen guten audiologischen Resultate sind sie derzeit das Mittel der Wahl.
Multiple Sklerose (MS) ist die häufigste neurologische Erkrankung, die bei jungen Erwachsenen zu dauerhaften körperlichen Einschränkungen führt. Ein Kennzeichen der MS sind zeitlich und örtlich disseminierte entzündliche Läsionen im zentralen Nervensystem (ZNS). Die Läsionsart, die am häufigsten auftritt, ist u. a. durch Antikörperablagerungen charakterisiert. Die häufigste Verlaufsform der MS tritt in Schüben auf. Im Laufe der Erkrankung bilden sich die Symptome in der Mehrzahl der Patienten unvollständig zurück und es entwickelt sich ein chronischer Verlauf. Trotz intensiver Forschung ist die Ätiologie der MS bisher unbekannt Bis heute gibt es keine Biomarker, um den Therapieerfolg oder das Therapieversagen der MS-Basistherapeutika (Glatirameracetat und β-Interferon) zu bestimmen. Aktuelle Studien, bei denen B-Zellen depletiert wurden, zeigten eine signifikante Reduktion MS-typischer Läsionen und der Schubrate bei der schubförmigen MS. Man vermutet, dass autoreaktive B-Zellen vielfältige Aufgaben in der Pathogenese der MS übernehmen: sie produzieren Autoantikörper, präsentieren autoreaktiven T-Zellen Autoantigene und sezernieren Mediatoren, die zur Aktivierung anderer Immunzellen führen. Es ist noch unklar, welche B-Zell-Untergruppe bei der MS besondere Relevanz hat. Vor kurzem wurden B1-Zellen beim Menschen beschrieben. Eine Studie zeigte, dass die Anzahl der B1-Zellen in unbehandelten MS-Patienten signifikant erniedrigt war. Des Weiteren wurden im ZNS von chronisch erkrankten MS-Patienten B-Zell-Aggregate nachgewiesen. Diese B-Zell-Aggregate ähneln sekundären lymphatischen Organen und könnten zur Progredienz der Erkrankung beitragen. Eine ex vivo-Studie zeigte, dass die B-Zell-Aggregat-Bildung durch das Adhäsionsmolekül CEACAM1-(carcinoembryogenic antigen-related cell adhesion molecule 1) vermittelt wird. Überdies ist die Koexpression von CEACAM1 und TIM-3 (T-cell immunoglobulin- and mucin-domain containing-3) für immunerschöpfte und tolerante T-Zellen charakteristisch. Schließlich konnte unsere Arbeitsgruppe zeigen, dass ZNS-reaktive B-Zellen nur im Blut von Patienten mit einem klinisch isolierten Syndrom und MS-Patienten nachweisbar waren.
In meiner Studie habe ich den Einfluss von MS-Basistherapeutika und einer MS-Eskalationstherapie auf die B-Zell-Untergruppen untersucht. Dabei habe ich die naive B-Zell-, B-Gedächtniszell-, B1-Zell- und Plasmablasten-Zahl von gesunden Probanden sowie unbehandelten und behandelten MS-Patienten miteinander verglichen. Die B-Zell-Untergruppen wurden durchflusszytometrisch untersucht. Die B1-Zell-Zahl war bei behandelten und unbehandelten MS-Patienten signifikant erniedrigt. In einer weiteren Studie konnte ich zeigen, dass die Anwesenheit von ZNS-reaktiven B-Zellen im Blut von glatirameracetat-behandelten MS-Patienten mit dem Therapieerfolg assoziiert war. Die ZNS-reaktiven B-Zellen wurden durch einen ZNS-Lysat-ELISPOT detektiert. Schließlich habe ich in einer dritten Studie die Expression von CEACAM1 und TIM-3 auf B-Zellen bei natalizumab-behandelten MS-Patienten durchflusszytometrisch untersucht. Im Vergleich zu gesunden Probanden zeigte sich, dass im Blut der MS-Patienten die CEACAM1+- und die CEACAM1\(^+\)TIM-3\(^+\)-B-Zell-Zahl signifikant erhöht war. Im Gegensatz dazu waren CEACAM1\(^+\)TIM-3\(^+\)-T-Helferzellen signifikant erniedrigt in behandelten MS-Patienten.
Meine Arbeit belegt, dass die B1-Zell-Population unabhängig von der MS-Therapie in MS-Patienten erniedrigt ist. Ungeklärt bleibt, ob diese Erniedrigung eine Folge oder eine Ursache der Erkrankung ist. B1-Zellen sind die Quelle von natürlichen Antikörpern in Mensch und Tier. Sie haben protektive Eigenschaften und sind bei der B-Zell-Toleranzinduktion beteiligt. Die protektiven Funktionen der natürlichen Antikörper könnten durch die Erniedrigung der B1-Zell-Zahl ausbleiben. Zusätzlich waren B-Zellen mit einem immunerschöpften Phänotyp im Blut von MS-Patienten erhöht. Trotz Stimulation konnte kein Phänotyp bei T-Helferzellen induziert werden, der für tolerante und immunerschöpfte T-Zellen beschrieben worden ist. In zukünftigen Studien sollte man die B1-Zell-Zahl und die CEACAM1\(^+\)TIM-3\(^+\)-B- und -T-Zell-Zahl bei Patienten mit einem klinisch isolierten Syndrom im Liquor und im Blut untersuchen. Damit könnte man feststellen, ob B1-Zellen aus der Peripherie bei MS-Patienten in das ZNS migrieren. Die Anwesenheit ZNS-reaktiver B-Zellen im Blut von behandelten MS-Patienten zeigte sich in meiner Arbeit als ein Marker, um den Therapieerfolg zu dokumentieren. Eine weiterführende Querschnittstudie (COPSELECT) wird ZNS-reaktive B-Zellen mittels ZNS-Lysat-ELISPOT als zukünftige Therapie-Biomarker ausführlicher untersuchen. MS-Biomarker wären für den einzelnen Betroffenen von großer Bedeutung und hätten ebenfalls gesundheitsökonomisch eine hohe Relevanz.
Costly signaling with mobile devices: An evolutionary psychological perspective on smartphones

In the last decade, mobile device ownership has largely increased. In particular, smartphone ownership is constantly rising (A. Smith, 2015; Statista, 2016a), and there is a real hype for luxury brand smartphones (Griffin, 2015). These observations raise the question of which functions smartphones serve in addition to their original purposes of making and receiving calls, searching for information, and organizing. Beyond these obvious functions, studies suggest that smartphones express fashion, lifestyle, and one’s economic status (e.g., Bødker et al., 2009; Statista, 2016b; Vanden Abeele, Antheunis, & Schouten, 2014). Specifically, individuals seem to purchase and use conspicuous luxury brand smartphones to display and enhance status (D. Kim et al., 2014; Müller-Lietzkow et al., 2014; Suki, 2013). But how does owning a conspicuous, high-status smartphone contribute to status, and which benefits may these status boosts provide to their owners? From an evolutionary perspective, status carries a lot of advantages, particularly for males; high status grants them priority access to resources and correlates with their mating success (van Vugt & Tybur, 2016). In this sense, research suggests that men conspicuously display their cell phones to attract mates and to distinguish themselves from rivals (Lycett & Dunbar, 2000). In a similar vein, evolutionarily informed studies on conspicuous consumption indicate that the purchase and display of conspicuous luxuries (including mobile phones and smartphones) relate to a man’s interest in uncommitted sexual relationships and enhance his desirability as a short-term mate (Hennighausen & Schwab, 2014; Saad, 2013; Sundie et al., 2011). Drawing on these findings, this doctoral dissertation investigated how a man is perceived given that he is an owner of a high-status (vs. nonconspicuous, low-status) smartphone as a romantic partner and male rival. This was done in three experiments. In addition, it was examined how male conspicuous consumption of smartphones interacted with further traits that signal a man’s mate quality, namely facial attractiveness (Studies 1 and 2) and social dominance (Study 3). Study 1 revealed that men and women perceived a male owner of a conspicuous smartphone as a less desirable long-term mate and as more inclined toward short-term mating. Study 2 replicated these results and showed that men and women assigned traits that are associated with short-term mating (e.g., low loyalty, interest in flirts, availability of tangible resources) to a male owner of a conspicuous smartphone and perceived him as a stronger male rival and mate poacher, and less as a friend. The results of Study 2 further suggested that specifically more attractive men might benefit from owning a conspicuous smartphone in a short-term mating context and might be hence considered as stronger male rivals. Study 3 partially replicated the findings of Studies 1 and 2 pertaining to the effects of owning a conspicuous smartphone. Study 3 did not show different effects of conspicuous consumption of smartphones on perceptions of a man dependent on the level of his social dominance.
To conclude, the findings of this doctoral dissertation suggest that owning a conspicuous, high-status smartphone might not only serve proximate functions (e.g., making and receiving calls, organization) but also ultimate functions, which relate to mating and reproduction. The results indicate that owning a conspicuous smartphone might yield benefits for men in a short-term rather than in a long-term mating context. Furthermore, more attractive men appear to benefit more from owning a conspicuous smartphone than less attractive men. These findings provide further insights into the motivations that underlie men’s purchases and displays of conspicuous, high-status smartphones from luxury brands that reach beyond the proximate causes frequently described in media and consumer psychological research. By applying an evolutionary perspective, this doctoral dissertation demonstrates the power and utility of this research paradigm for media psychological research and shows how combining a proximate and ultimate perspective adds to a more profound understanding of smartphone phenomena.
Virtualization allows the creation of virtual instances of physical devices, such as network and processing units. In a virtualized system, governed by a hypervisor, resources are shared among virtual machines (VMs). Virtualization has been receiving increasing interest as away to reduce costs through server consolidation and to enhance the flexibility of physical infrastructures. Although virtualization provides many benefits, it introduces new security challenges; that is, the introduction of a hypervisor introduces threats since hypervisors expose new attack surfaces.
Intrusion detection is a common cyber security mechanism whose task is to detect malicious activities in host and/or network environments. This enables timely reaction in order to stop an on-going attack, or to mitigate the impact of a security breach. The wide adoption of virtualization has resulted in the increasingly common practice of deploying conventional intrusion detection systems (IDSs), for example, hardware IDS appliances or common software-based IDSs, in designated VMs as virtual network functions (VNFs). In addition, the research and industrial communities have developed IDSs specifically designed to operate in virtualized environments (i.e., hypervisorbased IDSs), with components both inside the hypervisor and in a designated VM. The latter are becoming increasingly common with the growing proliferation of virtualized data centers and the adoption of the cloud computing paradigm, for which virtualization is as a key enabling technology.
To minimize the risk of security breaches, methods and techniques for evaluating IDSs in an accurate manner are essential. For instance, one may compare different IDSs in terms of their attack detection accuracy in order to identify and deploy the IDS that operates optimally in a given environment, thereby reducing the risks of a security breach. However, methods and techniques for realistic and accurate evaluation of the attack detection accuracy of IDSs in virtualized environments (i.e., IDSs deployed as VNFs or hypervisor-based IDSs) are lacking. That is, workloads that exercise the sensors of an evaluated IDS and contain attacks targeting hypervisors are needed. Attacks targeting hypervisors are of high severity since they may result in, for example, altering the hypervisors’s memory and thus enabling the execution of malicious code with hypervisor privileges. In addition, there are no metrics and measurement methodologies
for accurately quantifying the attack detection accuracy of IDSs in virtualized environments with elastic resource provisioning (i.e., on-demand allocation or deallocation of virtualized hardware resources to VMs). Modern hypervisors allow for hotplugging virtual CPUs and memory on the designated VM where the intrusion detection engine of hypervisor-based IDSs, as well as of IDSs deployed as VNFs, typically operates. Resource hotplugging may have a significant impact on the attack detection accuracy of an evaluated IDS, which is not taken into account by existing metrics for quantifying IDS attack detection accuracy. This may lead to inaccurate measurements, which, in turn, may result in the deployment of misconfigured or ill-performing IDSs, increasing
the risk of security breaches.
This thesis presents contributions that span the standard components of any system
evaluation scenario: workloads, metrics, and measurement methodologies. The scientific contributions of this thesis are:
A comprehensive systematization of the common practices and the state-of-theart on IDS evaluation. This includes: (i) a definition of an IDS evaluation design space allowing to put existing practical and theoretical work into a common context in a systematic manner; (ii) an overview of common practices in IDS evaluation reviewing evaluation approaches and methods related to each part of the design space; (iii) and a set of case studies demonstrating how different IDS evaluation approaches are applied in practice. Given the significant amount of existing practical and theoretical work related to IDS evaluation, the presented systematization is beneficial for improving the general understanding of the topic by providing an overview of the current state of the field. In addition, it is beneficial for identifying and contrasting advantages and disadvantages of different IDS evaluation methods and practices, while also helping to identify specific requirements and best practices for evaluating current and future IDSs.
An in-depth analysis of common vulnerabilities of modern hypervisors as well as a set of attack models capturing the activities of attackers triggering these vulnerabilities. The analysis includes 35 representative vulnerabilities of hypercall handlers (i.e., hypercall vulnerabilities). Hypercalls are software traps from a kernel of a VM to the hypervisor. The hypercall interface of hypervisors, among device drivers and VM exit events, is one of the attack surfaces that hypervisors expose. Triggering a hypercall vulnerability may lead to a crash of the hypervisor or to altering the hypervisor’s memory. We analyze the origins
of the considered hypercall vulnerabilities, demonstrate and analyze possible attacks that trigger them (i.e., hypercall attacks), develop hypercall attack models(i.e., systematized activities of attackers targeting the hypercall interface), and discuss future research directions focusing on approaches for securing hypercall interfaces.
A novel approach for evaluating IDSs enabling the generation of workloads that contain attacks targeting hypervisors, that is, hypercall attacks. We propose an approach for evaluating IDSs using attack injection (i.e., controlled execution of attacks during regular operation of the environment where an IDS under test is deployed). The injection of attacks is performed based on attack models that capture realistic attack scenarios. We use the hypercall attack models developed as part of this thesis for injecting hypercall attacks.
A novel metric and measurement methodology for quantifying the attack detection accuracy of IDSs in virtualized environments that feature elastic resource provisioning. We demonstrate how the elasticity of resource allocations in such environments may impact the IDS attack detection accuracy and show that using existing metrics in such environments may lead to practically challenging and inaccurate measurements. We also demonstrate the practical use of the metric we propose through a set of case studies, where we evaluate common conventional IDSs deployed as VNFs.
In summary, this thesis presents the first systematization of the state-of-the-art on IDS evaluation, considering workloads, metrics and measurement methodologies as integral parts of every IDS evaluation approach. In addition, we are the first to examine the hypercall attack surface of hypervisors in detail and to propose an approach using attack injection for evaluating IDSs in virtualized environments. Finally, this thesis presents the first metric and measurement methodology for quantifying the attack detection accuracy of IDSs in virtualized environments that feature elastic resource provisioning.
From a technical perspective, as part of the proposed approach for evaluating IDSsthis thesis presents hInjector, a tool for injecting hypercall attacks. We designed hInjector to enable the rigorous, representative, and practically feasible evaluation of IDSs using attack injection. We demonstrate the application and practical usefulness of hInjector, as well as of the proposed approach, by evaluating a representative hypervisor-based IDS designed to detect hypercall attacks. While we focus on evaluating the capabilities of IDSs to detect hypercall attacks, the proposed IDS evaluation approach can be generalized and applied in a broader context. For example, it may be directly used to also evaluate security mechanisms of hypervisors, such as hypercall access control (AC) mechanisms. It may also be applied to evaluate the capabilities
of IDSs to detect attacks involving operations that are functionally similar to hypercalls,
for example, the input/output control (ioctl) calls that the Kernel-based Virtual Machine (KVM) hypervisor supports. For IDSs in virtualized environments featuring elastic resource provisioning, our approach for injecting hypercall attacks can be applied in combination with the attack detection accuracy metric and measurement methodology we propose. Our approach for injecting hypercall attacks, and our metric and measurement methodology, can also be applied independently beyond the scenarios considered in this thesis. The wide spectrum of security mechanisms in virtualized environments whose evaluation can directly benefit from the contributions of this thesis (e.g., hypervisor-based IDSs, IDSs deployed as VNFs, and AC mechanisms) reflects the practical implication of the thesis.
Die Messung der Herzfrequenzvariabilität (HRV) stellt ein sensitives Verfahren dar, um die Aktivität des autonomen Nervensystems zu erfassen. Die HRV beschreibt die sich ständig wechselnden zeitlichen Unterschiede aufeinanderfolgender Herzschläge und unterliegt vor allem der Steuerung des sympathischen und des parasympathischen Nervensystems. Die Ermittlung der HRV erfolgt über Kurz- und Langzeit-EKG-Aufzeichnungen.
Störungen in der autonomen Kontrolle wurden mit vielen Krankheiten, v.a. mit einem erhöhten Risiko für kardiovaskuläre Erkrankungen, in Verbindung gebracht. Die Imbalance des autonomen Nervensystems, welche durch eine Hyperaktivität des sympathischen Nervensystems und einer verminderten Aktivität des parasympathischen Nervensystems charakterisiert ist, könnte einer der entscheidenden Faktoren sein, welcher zu einer erhöhten Morbidität und Mortalität von kardiovaskulären Erkrankungen führt. Ob diese Störungen in der autonomen Kontrolle zudem auch eine wichtige Rolle für das gehäufte Auftreten kardiovaskulärer Erkrankungen bei kleinwüchsigen Menschen einnehmen, ist Gegenstand heutiger Forschung.
Für ein besseres Verständnis der Verbindung zwischen Körpergröße und kardiovaskulärem Risiko wurden in dieser Arbeit HRV-Analysen aus 24h-Langzeit-EKG-Aufzeichungen ausgewertet.
Die Herzfrequenzvariabilität von 30 kleinwüchsigen Kindern (MWAlter = 6,3 ± 3,6 Jahre), darunter 17 ehemalige hypothrophe Neugeborene (sog. „Small for gestational age“, SGA), 11 Kinder mit einer konstitutionellen Entwicklungsverzögerung (KEV) und 2 Kinder mit einem nachgewiesenen idiopathischen isolierten Wachstumshormonmangel (Growth hormone deficiency, GHD) wurden hierfür mit einer Kontrollgruppe von 121 normwüchsigen und herzgesunden Kindern verglichen.
Zusätzlich wurde die HRV von 30 Kindern unter langjähriger Wachstumshormontherapie (MWAlter = 10,8 ± 3,7 Jahre), darunter 20 Kinder mit einer GHD und 10 SGA-Kinder, mit dieser Kontrollgruppe verglichen, um den Einfluss einer Substitutionstherapie auf das autonome Nervensystem zu ergründen.
Es zeigte sich, dass kleinwüchsige Kindern ab einem Alter von 9 Jahren eine signifikant herabgesetzte Herzfrequenzvariabilität haben. Die SDNN (Standard deviation of normal RR-intervals) als Maß der Gesamtvariabilität und die vagal modulierten HRV-Parameter RMSSD (Root mean squared of successive difference) und pNN50 (Percent NN differences over 50 ms) waren signifikant erniedrigt. Zudem zeigte sich bei diesen Kindern eine signifikant erhöhte Herzfrequenz bei Tag und Nacht.
Kleinwüchsige präpubertäre Kinder unter 9 Jahren zeigten dagegen keine Veränderungen der HRV und der Herzfrequenz im Vergleich zur Kontrollgruppe. Eine Therapie mit Wachstumshormonen bei Kindern mit einem idiopathischen isolierten Wachstumshormonmangel und SGA-Kindern ohne Aufholwachstum scheint dagegen keinen Einfluss auf die Herzfrequenzvariabilität zu haben. Diese blieb trotz Substitutionstherapie auch weiterhin erniedrigt.
Durch die Aufzeichnung von 24h-Langzeit-EKGs und anschließender HRV-Analyse während eines Clonidin-Wachstumshormonstimulationstests bei kleinwüchsigen Kindern sollte zudem die Rolle des zentralen α2-Adrenorezeptors in der Pathogense des Kleinwuchses untersucht werden.
Erstaunlicherweise kam es nur bei den Kindern mit einer konstitutionellen Entwicklungsverzögerung (KEV) zu einem erwarteten signifikanten Anstieg der vagal modulierten HRV-Parameter sowie zu einem Absinken des sympathisch modulierten HRV-Parameters LFn. Kinder mit einer GHD zeigten weder einen adäquaten Anstieg der Wachstumshormon-Ausschüttung noch eine Reaktion des sympathischen und des parasympathischen Nervensystems auf die Clonidingabe. Bei den SGA-Kindern konnte nur ein Anstieg der vagal modulierten Parameter, nicht jedoch ein Abfall der sympathisch modulierten Parameter, gemessen werden.
Es muss daher davon ausgegangen werden, dass der zentrale α2-Adrenorezeptor eine wichtige Rolle bei der Entstehung des Kleinwuchses, zumindest bei Kindern mit einem idiopathischen Wachstumshormonmangel und evtl. auch bei SGA-Kindern, einnimmt.
The present work addressed the influence of spins on fundamental processes in organic
semiconductors. In most cases, the role of spins in the conversion of sun light
into electricity was of particular interest. However, also the reversed process, an electric
current creating luminescence, was investigated by means of spin sensitive measurements.
In this work, many material systems were probed with a variety of innovative
detection techniques based on electron paramagnetic resonance spectroscopy.
More precisely, the observable could be customized which resulted in the experimental
techniques photoluminescence detected magnetic resonance (PLDMR), electrically
detected magnetic resonance (EDMR), and electroluminescence detected magnetic
resonance (ELDMR). Besides the commonly used continuous wave EPR spectroscopy,
this selection of measurement methods yielded an access to almost all intermediate
steps occurring in organic semiconductors during the conversion of light into electricity
and vice versa. Special attention was paid to the fact that all results were applicable
to realistic working conditions of the investigated devices, i.e. room temperature application and realistic illumination conditions.
Motoneuron diseases form a heterogeneous group of pathologies characterized by the progressive degeneration of motoneurons. More and more genetic factors associated with motoneuron diseases encode proteins that have a function in RNA metabolism, suggesting that disturbed RNA metabolism could be a common underlying problem in several, perhaps all, forms of motoneuron diseases. Recent results suggest that SMN interacts with hnRNP R and TDP-43 in neuronal processes, which are not part of the classical SMN complex. This point to an additional function of SMN, which could contribute to the high vulnerability of spinal motoneurons in spinal muscular atrophy (SMA) and amyotrophic lateral sclerosis (ALS). The current study elucidates functional links between SMN, the causative factor of SMA (spinal muscular atrophy), hnRNP R, and TDP-43, a genetic factor in ALS (amyotrophic lateral sclerosis). In order to characterize the functional interaction of SMN with hnRNP R and TDP-43, we produced recombinant proteins and investigated their interaction by co-immunoprecipitation. These proteins bind directly to each other, indicating that no other co-factors are needed for this interaction. SMN potentiates the ability of hnRNP R and TDP-43 to bind to ß-actin mRNA. Depletion of SMN alters the subcellular distribution of hnRNP R in motoneurons both in SMN-knockdown motoneurons and SMA mutant mouse (delta7 SMA). These data point to functions of SMN beyond snRNP assembly which could be crucial for recruitment and transport of RNA particles into axons and axon terminals, a mechanism which may contribute to SMA pathogenesis and ALS.
ALS and FTLD (frontotemporal lobar degeneration) are linked by several lines of evidence with respect to clinical and pathological characteristics. Both sporadic and familial forms are a feature of the ALS-FTLD spectrum, with numerous genes having been associated with these pathological conditions. Both diseases are characterized by the pathological cellular aggregation of proteins. Interestingly, some of these proteins such as TDP-43 and FUS have also common relations not only with ALS-FTLD but also with SMA. Intronic hexanucleotide expansions in C9ORF72 are common in ALS and FTLD but it is unknown whether loss of function, toxicity by the expanded RNA or dipeptides from non ATG-initiated translation is responsible for the pathophysiology. This study tries to characterize the cellular function of C9ORF72 protein. To address this, lentiviral based knockdown and overexpression of C9ORF72 was used in isolated mouse motoneurons. The results clearly show that survival of these motoneurons was not affected by altered C9ORF72 levels, whereas adverse effects on axon growth and growth cone size became apparent after C9ORF72 suppression. Determining the protein interactome revealed several proteins in complexes with C9ORF72. Interestingly, C9ORF72 is present in a complex with cofilin and other actin binding proteins that modulate actin dynamics. These interactions were confirmed both by co-precipitation analyses and in particular by functional studies showing altered actin dynamics in motoneurons with reduced levels of C9ORF72. Importantly, the phosphorylation of cofilin is enhanced in C9ORF72 depleted motoneurons and patient derived lymphoblastoid cells with reduced C9ORF72 levels. These findings indicate that C9ORF72 regulates axonal actin dynamics and the loss of this function could contribute to disease pathomechanisms in ALS and FTLD.
Der zur Familie der pentameren ligandengesteuerten Ionenkanäle zugehörige Glycinrezeptor (GlyR) ist ein wichtiger Vermittler synaptischer Inhibition im Zentralnervensystem von Säugetieren. GlyR-Mutationen führen zur neurologischen Bewegungsstörung Hyperekplexie. Aufgrund fehlender struktureller Daten ist die intrazelluläre Loop-Struktur zwischen den Transmembransegmenten 3 und 4 (TM3-4 Loop) eine weitgehend unerforschte Domäne des GlyR. Innerhalb dieser Domäne wurden Rezeptortrunkierungen sowie Punktmutationen identifiziert. Rezeptortrunkierung geht mit Funktionslosigkeit einher, welche jedoch durch Koexpression des fehlenden Sequenzabschnitts zum Teil wiederhergestellt werden kann. Innerhalb dieser Arbeit wurde die Interaktion zwischen trunkierten, funktionslosen GlyR und sukzessiv verkürzten Komplementationskonstrukten untersucht. Dabei wurden als Minimaldomänen für die Interaktion das C-terminalen basische Motive des TM3-4 Loops, die TM4 sowie der extrazelluläre C-Terminus identifiziert. Die Rückkreuzung transgener Mäuse, die das Komplementationskonstrukt iD-TM4 unter Kontrolle des GlyR-Promotors exprimierten, mit der oscillator-Maus spdot, die einen trunkierten GlyR exprimiert und 3 Wochen nach der Geburt verstirbt, hatte aufgrund fehlender Proteinexpression keinen Effekt auf die Letalität der Mutation. Des Weiteren wurde die Bedeutsamkeit der Integrität beider basischer Motive 316RFRRKRR322 und 385KKIDKISR392 im TM3-4 Loop in Kombination mit der Loop-Länge für die Funktionalität und das Desensitisierungsverhalten des humanen GlyRα1 anhand von chimären Rezeptoren identifiziert. Eine bisher unbekannte Patientenmutation P366L innerhalb des TM3-4 Loops wurde mit molekularbiologischen, biochemischen und elektrophysiologischen Methoden charakterisiert. Es wurde gezeigt, dass die mutierten Rezeptorkomplexe in vitro deutlich reduzierte Glycin-induzierte Maximalströme sowie eine beschleunigte Schließkinetik aufweisen. P366L hat im Gegensatz zu bereits charakterisierten Hyperekplexiemutationen innerhalb des TM3-4 Loops keinen Einfluss auf die Biogenese des Rezeptors. P366 ist Teil einer möglichen Poly-Prolin-Helix, die eine Erkennungssequenz für SH3-Domänen darstellt. Ein potenzieller Interaktionspartner des TM3-4 Loops des GlyRα1 ist Collybistin, welches eine wichtige Rolle bei der synaptischen Rezeptorintegration spielt und die Verbindung zum Zytoskelett vermittelt. An der inhibitorischen Synapse verursacht P366L durch die Reduzierung postsynaptischer Chloridströme, das beschleunigte Desensitisierungsverhalten des GlyRα1 sowie ein verändertes Interaktionsmotiv Störungen der glycinergen Transmission, die zur Ausprägung phänotypischer Symptome der Hyperekplexie führen.
Die Verwendung von Dried Blood Spots zur serologischen Diagnostik auf Antikörper gegen opportunistische Erreger im Rahmen einer HIV Infektion, wurde unter simulierten afrikanischen Extrembedingungen validiert. Ziel war es zu zeigen, dass Dried Blood Spots unter idealen Lagerungsbedingungen für 4 Wochen stabil bleiben und anschliessend eine valide Antikörperdiagnostik möglich ist.
Diese Methode kann als Möglichkeit für einen einfachen und kostengünstigen Probentransport in Tansania dienen.
Oxylipine sind Signalmoleküle, die durch enzymatische Oxidation oder durch Autoxidation von mehrfach ungesättigten Fettsäuren entstehen. Sie akkumulieren während einer Vielzahl von biotischen und abiotischen Stressen und spielen eine bedeutende Rolle bei der Abwehr verschiedener Stressoren. In vielen physiologischen Entwicklungsprozessen sind Oxylipine ebenfalls wichtig.
Eine bisher wenig erforschte Untergruppe dieser Oxylipine bilden reaktive elektrophile Spezies, die sog. RES-Oxylipine. Hierzu gehören unter anderem der Jasmonsäure-Vorläufer 12 Oxophytodiensäure (OPDA), aber auch (E)-2-Hexenal oder Phytoprostan A1 (PPA1). Diese Substanzen sind aufgrund einer α,β ungesättigten Carbonylgruppe elektrophil und damit chemisch reaktiv. Diese Reaktivität wird als Grund für ihre biologische Aktivität angesehen: RES-Oxylipine sind Induktoren einer Reihe von Genen. Allerdings ist bisher wenig über den Signalweg sowie die Funktionen der RES-Oxylipine in Arabidopsis thaliana bekannt.
Fast die Hälfte (40 %) aller durch OPDA-induzierten Gene in A. thaliana sind abhängig von TGA-Transkriptionsfaktoren, jedoch werden OPDA-responsive Hitzeschockgene (z.B. Hitzeschockproteine) unabhängig von TGA-Transkriptionsfaktoren induziert. Außerdem gibt es Hinweise auf eine Akkumulation des RES-Oxylipins OPDA, aber auch des non-RES-Oxylipins Jasmonsäure (JA) durch eine Behandlung mit 38° C in A. thaliana. Eine exogene Applikation von JA bewirkt jedoch, im Gegensatz zu OPDA, keine Genexpression von Hitzeschockgenen in Arabidopsis.
Ziel dieser Arbeit war es, die Funktion der RES-Oxylipine OPDA und Prostaglandin A1 (PGA1, ein Analogon zu PPA1) während der Hitzeschockantwort in Arabidopsis thaliana, sowie die TGA-unabhängige Signaltransduktion der Hitzeschockgene, aufzuklären.
Durch einen Vergleich zweier bereits veröffentlichter Transkriptomdaten in silico konnte die Überschneidung des Hitze-induzierten- (1 h, 37 °C) und des OPDA-induzierten-Transkriptoms (4 h, 75 µM) genau analysiert werden. Es werden 30 Gene sowohl von OPDA als auch durch 37 °C mehr als dreifach hochreguliert. Dieses Ergebnis konnte durch realtime qPCR vier repräsentativer Gene (HSP101, HSP26.5, DREB2A, HSFA2) bestätigt werden. Allerdings zeigten sich deutliche Unterschiede in der Stärke und Kinetik der Induktion: Hitze (37 °C) hat einen sehr viel stärkeren Einfluss auf die Hochregulation der Genexpression als die getesteten RES-Oxylipine OPDA und PGA1 (unter 10 % der Induktion durch 37 °C, Ausnahme DREB2A). Zudem resultiert eine Hitzebehandlung in einer schnellen und transienten Genexpression, das Maximum ist nach 1 bis 2 h erreicht während die Addition von RES-Oxylipinen eine langsamere Induktion der Genexpression bewirkt (Maximum nach 4 bis 6 h).
Eine Genexpressionsanalyse mit verschiedenen Signaltransduktionsmutanten half bei der Aufklärung möglicher Signaltransduktionskomponenten der RES-Oxylipine. So konnte gezeigt werden, dass der putative OPDA-Rezeptor Cyclophilin 20-3 sowie sein Interaktionspartner, das Protein Serin-Acetyltransferase 1, keine Bedeutung in der Regulation von Hitzeschockgenen durch RES-Oxylipine haben. Die Hitze-Masterregulatoren HSFA1 a,b,d (und e) jedoch sind für die Induktion der Hitzeschockgene HSP101, HSP26.5 und HSFA2 durch RES-Oxylipine essentiell und für DREB2A zumindest teilweise notwendig. Dennoch spielt der durch Hitze induzierbare Transkriptionsfaktor HSFA2 in der Signaltransduktion von RES-Oxylipinen (bezüglich der Hitzeschockgeninduktion) keine Rolle.
Durch ein Screening strukturell verschiedener RES hinsichtlich ihrer Induktion von HSP101 konnte geklärt werden, dass nicht die Anwesenheit einer α,β ungesättigten Carbonylgruppe, sondern vielmehr die Eigenschaft der Elektrophilie für die Induktion des HSP101 verantwortlich ist. Auch das RES Sulforaphan vermittelt, wie die RES-Oxylipine OPDA und PGA1, die Induktion der Hitzeschockgene über die HSFA1-Transkritionsfaktoren.
Ein weiterer Schwerpunkt dieser Arbeit lag in der Quantifizierung der endogenen Oxylipine in zehn Tage alten Arabidopsis-Keimligen nach einer Hitzebehandlung unter Kurztag-Lichtbedingungen. Weder während eines kurzzeitigen Hitzestresses (bis zu 8 h) noch während einer längerfristigen Hitzebehandlung (bis zu 7 Tage) steigt der Gehalt des RES Oxylipins OPDA signifikant an. Das non-RES-Oxylipin Jasmonsäure hingegen akkumuliert transient (Maximum 2 h nach Beginn eines Hitzestresses) und signifikant, allerdings ist dieser Anstieg in seiner Stärke (13fach) nicht vergleichbar mit einer Akkumulation beispielsweise nach Verwundung (hier ist ein 1000facher Anstieg möglich).
In weiteren Experimenten wurde eine mögliche Korrelation der endogenen Oxylipin-Akkumulation (verursacht durch Verwundung oder osmotischen Stress) mit der Genexpression von Hitzeschockgenen untersucht. …
Neuropathic pain, caused by neuronal damage, is a severely impairing mostly chronic condition. Its underlying molecular mechanisms have not yet been thoroughly understood in their variety. In this doctoral thesis, I investigated the role of microRNAs (miRNAs) in a murine model of peripheral neuropathic pain. MiRNAs are small, non-coding RNAs known to play a crucial role in post-transcriptional gene regulation, mainly in cell proliferation and differentiation. Initially, expression patterns in affected dorsal root ganglia (DRG) at different time points after setting a peripheral nerve lesion were studied. DRG showed an increasingly differential expression pattern over the course of one week. Interestingly, a similar effect, albeit to a smaller extent, was observed in corresponding contralateral ganglia. Five miRNA (miR-124, miR-137, miR-183, miR-27b, and miR-505) were further analysed. qPCR, in situ hybridization, and bioinformatical analysis point towards a role for miR-137 and -183 in neuropathic pain as both were downregulated. Furthermore, miR-137 is shown to be specific for non-peptidergic non-myelinated nociceptors (C fibres) in DRG. As the ganglia consist of highly heterocellular tissue, I also developed a neuron-specific approach. Primarily damaged neurons were separated from intact adjacent neurons using fluorescence-activated cell-sorting and their gene expression pattern was analysed using a microarray. Thereby, not only were information obtained about mRNA expression in both groups but, by bioinformatical tools, also inferences on miRNA involvement. The general expression pattern was consistent with previous findings. Still, several genes were found differentially expressed that had not been described in this context before. Among these are corticoliberin or cation-regulating proteins like Otopetrin1. Bioinformatical data conformed, in part, to results from whole DRG, e.g. they implied a down-regulation of miR-124, -137, and -183. However, these results were not significant.
In summary, I found that a) miRNA expression in DRG is influenced by nerve lesions typical of neuropathic pain and that b) these changes develop simultaneously to over-expression of galanin, a marker for neuronal damage. Furthermore, several miRNAs (miR-183, -137) exhibit distinct expression patterns in whole-DRG as well as in neuron-specific approaches. Therefore, further investigation of their possible role in initiation and maintenance of neuropathic pain seems promising.
Finally, the differential expression of genes like Corticoliberin or Otopetrin 1, previously not described in neuropathic pain, has already resulted in follow-up projects.
Attention-Deficit/Hyperactivity Disorder (ADHD) is characterized by symptoms of inattentiveness and hyperactivity/impulsivity. Besides, increasing evidence points to ADHD patients showing emotional dysfunctions and concomitant problems in social life. However, systematic research on emotional dysfunctions in ADHD is still rare, and to date most studies lack conceptual differentiation between emotion processing and emotion regulation. The aim of this thesis was to systematically investigate emotion processing and emotion regulation in adult ADHD in a virtual reality paradigm implementing social interaction. Emotional reactions were assessed on experiential, physiological, and behavioral levels.
Experiment 1 was conducted to develop a virtual penalty kicking paradigm implying social feedback and to test it in a healthy sample. This paradigm should then be applied in ADHD patients later on. Pleasant and unpleasant trials in this paradigm consisted of hits respectively misses and subsequent feedback from a virtual coach. In neutral trials, participants were teleported to different spots of the virtual stadium. Results indicated increased positive affectivity (higher valence and arousal ratings, higher zygomaticus activations, and higher expression rates of positive emotional behavior) in response to pleasant compared to neutral trials. Reactions to unpleasant trials were contradictory, indicating increased levels of both positive and negative affectivity, compared to neutral trials. Unpleasant vs. neutral trials revealed lower valence ratings, higher arousal ratings, higher zygomaticus activations, slightly lower corrugator activations, and higher expression rates of both positive and negative emotional behavior. The intensity of emotional reactions correlated with experienced presence in the virtual reality.
To better understand the impact of hits or misses per se vs. hits or misses with coach feedback healthy participants’ emotional reactions, only 50% of all shots were followed by coach feedback in experiment 2. Neutral trials consisted of shots over the free soccer field which were followed by coach feedback in 50 % of all trials. Shots and feedback evoked more extreme valence and arousal ratings, higher zygomaticus activations, lower corrugator activations, and higher skin conductance responses than shots alone across emotional conditions. Again, results speak for the induction of positive emotions in pleasant trials whereas the induction of negative emotions in unpleasant trials seems ambiguous. Technical improvements of the virtual reality were reflected in higher presence ratings than in experiment 1.
Experiment 3 investigated emotional reactions of adult ADHD patients and healthy controls after emotion processing and response-focused emotion regulation. Participants successively
went through an ostensible online ball-tossing game (cyber ball) inducing negative emotions, and an adapted version of the virtual penalty kicking game. Throughout cyber ball, participants were included or ostracized by two other players in different experimental blocks. Participants were instructed to explicitly show, not regulate, or hide their emotions in different experimental blocks. Results provided some evidence for deficient processing of positive emotions in ADHD. Patients reported slightly lower positive affect than controls during cyber ball, gave lower valence ratings than controls in response to pleasant penalty kicking trials, and showed lower zygomaticus activations than controls especially during penalty kicking. Patients in comparison with controls showed slightly increased processing of unpleasant events during cyber ball (higher ratings of negative affect, especially in response to ostracism), but not during penalty kicking. Patients showed lower baseline skin conductance levels than controls, and impaired skin conductance modulations. Compared to controls, patients showed slight over-expression of positive as well as negative emotional behavior. Emotion regulation analyses revealed no major difficulties of ADHD vs. controls in altering their emotional reactions through deliberate response modulation. Moreover, patients reported to habitually apply adaptive emotion regulation strategies even more frequently than controls. The analyses of genetic high-risk vs. low-risk groups for ADHD across the whole sample revealed similar results as analyses for patients vs. controls for zygomaticus modulations during emotion processing, and for modulations of emotional reactions due to emotion regulation.
To sum up, the virtual penalty kicking paradigm proved to be successful for the induction of positive, but not negative emotions. The importance of presence in virtual reality for the intensity of induced emotions could be replicated. ADHD patients showed impaired processing of primarily positive emotions. Aberrations in negative emotional responding were less clear and need further investigation. Results point to adult ADHD in comparison to healthy controls suffering from baseline deficits in autonomic arousal and deficits in arousal modulation. Deficits of ADHD in the deliberate application of response-focused emotion regulation could not be found.
Human Vγ9Vδ2 T cells are the main γδ T cell subset in the circulation, accounting for up to 5% of the total peripheral blood lymphocyte population. They have been suggested to be important in response to tumors and infections. Their immune mechanisms encompass cell killing via cytotoxicity and secretion of pro-inflammatory cytokines such as IFNγ and tumor necrosis factor (TNF). The main stimulators of Vγ9Vδ2 T cells are isopentenyl pyrophosphate (IPP) and (E)-4-hydroxy-3-methyl-but-2-enyl pyrophosphate (HMBPP), denominated phosphoantigens (PAg).
A major advance in the understanding of PAg detection and Vγ9Vδ2 T cell activation has been the identification of the butyrophlin 3A (BTN3A) proteins as key mediators in these processes. In humans, three isoforms constitute the BTN3A family: BTN3A1, BTN3A2, and BTN3A3; and their genes are localized on the short arm of chromosome 6. The role of BTN3A1 has been highlighted by BTN3A-specific monoclonal antibody 20.1 (mAb 20.1), which has an agonist effect and causes proliferation, expansion, and activation of primary human Vγ9Vδ2 T cells. On the other hand, BTN3A-specific monoclonal antibody 103.2 (mAb 103.2) is antagonistic, inhibiting the Vγ9Vδ2 T cell response. The actual mechanism underlying both PAg- and mAb 20.1-mediated activation is not completely elucidated, but the importance of BTN3A1 is clear.
The main objective of this dissertation was to characterize the role of BTN3A1 in the PAg-dependent and PAg-independent Vγ9Vδ2 T cell activation and to evaluate its contribution in the response to influeza A virus infected cells. This research work demonstrated, by using Vγ9Vδ2 TCR MOP-transduced murine cells (reporter cells), that human chromosome 6 (Chr6) is mandatory for PAg-induced stimulation, but not for stimulation with mAb 20.1. The reporter cells responded to mAb 20.1 in cultures with BTN3A1-transduced Chinese hamster ovary cells (CHO BTN3A1) as antigen presenting cells. Nevertheless, for PAg-dependent activation the presence of Chr6 in CHO BTN3A1 was mandatory.
Although reporter cells expressing clonotypically different Vγ9Vδ2 TCRs showed similar PAg response, they clearly differed in the mAb 20.1 response. The reporter cell line transduced with Vγ9Vδ2 TCR D1C55 demonstrated essentially no response to mAb 20.1 compared to Vγ9Vδ2 TCR MOP cells. These findings were further supported by experiments performed with human PBMCs-derived Vγ9Vδ2 T cell clones. The results indicate heterogeneity in the PAg- and 20.1-dependent responses, in terms of CD25 and CD69 expression, among three different Vγ9Vδ2 T cells clones.
Co-cultures of reporter cells with Raji RT1BI and PAg plus mAb 20.1 or single chain antibody 20.1 (sc 20.1) revealed no additive or synergistic activating effects. In contrast, mAb 20.1 or sc 20.1 inhibited the PAg-mediated activation of the reporter cells.
The comparison of the relative contribution of the isoforms BTN3A2 and BTN3A3, in the activation of Vγ9Vδ2 T cells, was undertaken by overexpression of these isoforms in CHO cells. The results showed that BTN3A2 contributes to both PAg- and mAb-induced Vγ9Vδ2 T cell activation. On the contrary, BTN3A3 does not support PAg-mediated γδ T cell response.
Additionally, mutations in the proposed PAg- and mAb 20.1-binding sites of the extracellular BTN3A1 domain were generated by means of site-directed mutagenesis. These mutations revoked the mAb 20.1-induced Vγ9Vδ2 T cell activation, but not that induced by PAg.
Finally, co-cultures of Vγ9Vδ2 TCR MOP-transduced murine reporter cells with influenza A/PR/8/34-infected cells, or infection of PBMCs with this virus strain indicated that BTN3A1 might be dispensable for the Vγ9Vδ2 T cell response against influenza A.
The data of this research work points out that: i) in addition to BTN3A1, other Chr6-encoded genes are necessary for Vγ9Vδ2 T cell activation with PAg; ii) clonotypical (CDR3) differences influence the PAg- and mAb 20.1-mediated Vγ9Vδ2 T cell activation; iii) the PAg- and mAb 20.1-induced responses are not synergistic and interfere with each other; iv) BTN3A2 and BTN3A3 isoforms differ in the ability to support PAg- or mAb 20.1-dependent Vγ9Vδ2 T cell activation; v) the importance of the intracellular B30.2 domain of BTN3A1, in the Vγ9Vδ2 T cell activation, might be higher than that of the extracellular domain; and vi) in spite of the importance of BTN3A1 in the activation of Vγ9Vδ2 T cells, it is possible that many molecules with redundant functions are involved in the elimination of influenza virus infection by these cells.
In summary, it is possible to hypothesize a model in which BTN3A1 detects prenyl pyrophosphates in the cytoplasm via its B30.2 domain and in association with another protein(s). The binding of PAg to this domain induces a multimerization of BTN3A1 or a conformational change of its extracellular domain (mimicked by mAb 20.1). These modifications might be recognized by the Vγ9Vδ2 TCR or by an associated T cell protein. In the case that the TCR directly recognizes BTN3A1, the intensity of the response will depend on the Vγ9Vδ2 TCR clonotype. Future research will allow to gain a better understanding of BTN3A1, its interaction with other proteins, its actual role in the activation of Vγ9Vδ2 T cells, and its importance in specific models of cancer or infection. This knowledge will be necessary to transform these cells into effective tools in the clinic.
Ziel dieser Arbeit ist es, weitere Einblicke in die Aktivierung von FGF19 und
FXR durch diverse nukleäre Agonisten und deren spezifischer Rezeptoren zu
gewinnen. Hierbei soll im humanen Zellmodell versucht und mittels DNA-Analyse
untersucht werden, welche messbaren molekularbiologischen
Auswirkungen eine Behandlung mit unterschiedlichen Substanzen in
variierenden Konzentrationen bewirkt. Genauer soll betrachtet werden, ob sich
Vitamin A und Vitamin D als Induktoren von FGF19 in menschlichen
Darmzelllinien eignen, da dies bereits im Mausmodel demonstriert werden
konnte.
Dieser initialen Vermutung folgend, sollen auch die möglichen
Wechselwirkungen und Synergismen untersucht werden – welche
Mechanismen liegen diese zu Grunde und über welche molekularen
Signalwege werden dies vermuteten Effekte vermittelt.
Hierdurch soll ein besseres Verständnis für die Rezeptor und Agonistenabhängigen
Abläufe ermöglicht werden, um mögliche Rückschlüsse auf weitere
Funktionen bereits bekannter Vertreter zu erlauben.
Aufgrund der bereits oben beschriebenen Tiermodelle und der daraus
gewonnenen Einsichten würde sich durch ein noch besseres Verständnis des
FGF15/19 und des Farnesoid X Rezeptors in menschlichen Zellen, auf eine
zukünftige Anwendung in analytischen und/oder therapeutischen Bereichen
hoffen lassen.
Diese Arbeit soll sich deshalb den Fragen widmen, ob eine FGF19 Induktion in

Molecular Beam Epitaxy and Characterization of Bi-Based V\(_2\)VI\(_3\) Topological Insulators

The present thesis is addressed to the growth and characterization of Bi-based V2VI3 topological insulators (TIs). The TIs were grown by molecular beam epitaxy (MBE) on differently passivated Si(111) substrates, as well as InP(111) substrates. This allows the study of the influence of the substrate on the structural and electrical properties of the TIs.
The Bi2Se3 layers show a change of mosaicity-tilt and -twist for growth on the differently prepared Si(111) substrates, as well as a significant increase of crystalline quality for growth on the lateral nearly lattice matched InP(111). The rocking curve FWHMs observed for thick layers grown on InP are comparable to these of common zincblende layers, which are close to the resolution limit of standard high resolution X-ray diffraction (HRXRD) setups. The unexpected high structural crystalline quality achieved in this material system is remarkable due to the presence of weak van der Waals bonds between every block of five atomic layers, i.e. a quintuple layer (QL), in growth direction.
In addition to the mosaicity also twin domains, present in films of the V2VI3 material system, are studied. The twin defects are observed in Bi2Se3 layers grown on Si(111) and lattice matched InP(111) suggesting that the two dimensional surface lattice of the substrates can not determine the stacking order ABCABC... or ACBACB... in locally separated growth seeds. Therefore the growth on misoriented and rough InP(111) is analyzed.
The rough InP(111) with its facets within a hollow exceeding the height of a QL is able to provide its stacking information to the five atomic layers within a QL. By varying the roughness of the InP substrate surface, due to thermal annealing, the influence on the twinning within the layer is confirmed resulting in a complete suppression of twin domains on rough InP(111).
Focusing on the electrical properties of the Bi2Se3 films, the increased structural quality for films grown on lattice matched flat InP(111)B results in a marginal reduction of carrier density by about 10% compared to the layers grown on H-passivated Si(111), whereas the suppression of twin domains for growth on rough InP(111)B resulted in a reduction of carrier density by an order of magnitude. This implies, that the twin domains are a main crystal defect responsible for the high carrier density in the presented Bi2Se3 thin films.
Besides the binary Bi2Se3 also alloys with Sb and Te are fabricated to examine the influence of the compound specific point defects on the carrier density. Therefore growth series of the ternary materials Bi2Te(3-y)Se(y), Bi(2-x)Sb(x)Se3, and Bi(2-x)Sb(x)Te3, as well as the quaternary Bi(2-x)Sb(x)Te(3-y)Se(y) are studied.
To further reduce the carrier density of twin free Bi2Se3 layers grown on InP(111)B:Fe a series of Bi(2-x)Sb(x)Se3 alloys were grown under comparable growth conditions. This results in a reduction of the carrier density with a minimum in the composition range of about x=0.9-1.0.
The Bi(2-x)Sb(x)Te3 alloys exhibit a pn-transition, due to the dominating n-type and p-type point defects in its binary compounds, which is determined to reduce the bulk carrier density enabling the study the TI surface states. This pn-transition plays a significant role in realizing predicted applications and exotic effects, such as the quantum anomalous Hall effect.
The magnetic doping of topological insulators with transition metals is studied by incorporating Cr and V in the alloy Bi(2-x)Sb(x)Te3 by codeposition. The preferential incorporation of Cr on group-V sites is confirmed by EDX and XRD, whereas the incorporation of Cr reduces the crystalline quality of the layer. Magnetotransport measurements of the Cr-doped TIs display an anomalous Hall effect confirming the realization of a magnetic TI thin film. The quantum anomalous Hall effect is observed in V-doped Bi(2-x)Sb(x)Te3, where the V-doping results in higher Curie temperatures, as well as higher coercive fields compared to the Cr-doping of the TIs.
Moreover the present thesis contributes to the understanding of the role of the substrate concerning the crystalline quality of van der Waals bonded layers, such as the V2VI3 TIs, MoS2 and WoTe2. Furthermore, the fabrication of the thin film TIs Bi(2-x)Sb(x)Te(3-y)Se(y) in high crystalline quality serves as basis to explore the physics of topological insulators.
A novel growth method has been developed, allowing for the growth of strained HgTe shells on CdTe nanowires (NWs). The growth of CdTe-HgTe core-shell NWs required high attention in controlling basic parameters like substrate temperature and the intensity of supplied material fluxes. The difficulties in finding optimized growth conditions have been successfully overcome in this work.
We found the lateral redistribution of liquid growth seeds with a ZnTe growth start to be crucial to trigger vertical CdTe NW growth. Single crystalline zinc blende CdTe NWs grew, oriented along [111]B. The substrate temperature was the most critical parameter to achieve straight and long wires. In order to adjust it, the growth was monitored by reflection high-energy electron diffraction, which was used for fine tuning of the temperature over time in each growth run individually. For optimized growth conditions, a periodic diffraction pattern allowed for the detailed analysis of atomic arrangement on the surfaces and in the bulk. The ability to do so reflected the high crystal quality and ensemble uniformity of our CdTe NWs. The NW sides were formed by twelve stable, low-index crystalline facets. We observed two types stepped and polar sides, separated by in total six flat and non-polar facets.
The high crystalline quality of the cores allowed to grow epitaxial HgTe shells around. We reported on two different heterostructure geometries. In the first one, the CdTe NWs exhibit a closed HgTe shell, while for the second one, the CdTe NWs are overgrown mainly on one side. Scanning electron microscopy and scanning transmission electron microscopy confirmed, that many of the core-shell NWs are single crystalline zinc blende and have a high uniformity. The symmetry of the zinc blende unit cell was reduced by residual lattice strain. We used high-resolution X-ray diffraction to reveal the strain level caused by the small lattice mismatch in the heterostructures. Shear strain has been induced by the stepped hetero-interface, thereby stretching the lattice of the HgTe shell by 0.06 % along a direction oriented with an angle of 35 ° to the interface.
The different heterostructures obtained, were the base for further investigation of quasi-one-dimensional crystallites of HgTe. We therefore developed methods to reliably manipulate, align, localize and contact individual NWs, in order to characterize the charge transport in our samples. Bare CdTe cores were insulating, while the HgTe shells were conducting. At low temperature we found the mean free path of charge carriers to be smaller, but the phase coherence length to be larger than the sample size of several hundred nanometers. We observed universal conductance fluctuations and therefore drew the conclusion, that the trajectories of charge carriers are defined by elastic backscattering at randomly distributed scattering sites. When contacted with superconducting leads, we saw induced superconductivity, multiple Andreev reflections and the associated excess current. Thus, we achieved HgTe/superconductor interfaces with high interfacial transparency.
In addition, we reported on the appearance of peaks in differential resistance at Delta/e for HgTe-NW/superconductor and 2*Delta/e for superconductor/HgTe-NW/superconductor junctions, which is possibly related to unconventional pairing at the HgTe/superconductor interface. We noticed that the great advantage of our self-organized growth is the possibility to employ the metallic droplet, formerly seeding the NW growth, as a superconducting contact. The insulating wire cores with a metallic droplet at the tip have been overgrown with HgTe in a fully in-situ process. A very high interface quality was achieved in this case.
Wilms tumor protein 1 (WT1) is a suitable target to develop an immunotherapeutic approach against high risk acute myeloid leukemia (AML), particularly their relapse after allogeneic hematopoietic stem cell transplantation (HSCT). As an intracellular protein traversing between nucleus and cytoplasm, recombinant expression of WT1 is difficult. Therefore, an induction of WT1-specific T-cell responses is mostly based on peptide vaccination as well as dendritic cell (DC) electroporation with mRNA encoding full-length protein to mount WT1-derived peptide variations presented to T cells. Alternatively, the WT1 peptide presentation could be broadened by forcing receptor-mediated endocytosis of DCs.
In this study, antibody fusion proteins consisting of an antibody specific to the human DEC205 endocytic receptor and various fragments of WT1 (anti-hDEC205-WT1) were generated for a potential DC-targeted recombinant WT1 vaccine. Anti-hDEC205-WT1 antibody fusion proteins containing full-length or major parts of WT1 were not efficiently expressed and secreted due to their poor solubility and secretory capacity. However, small fragment-containing variants: anti-hDEC205-WT110-35, anti-hDEC205-WT191-138, anti-hDEC205-WT1223-273, and anti-hDEC205-WT1324-371 were obtained in good yields.
Since three of these fusion proteins contain the most of the known immunogenic epitopes in their sequences, the anti-hDEC205-WT191-138, anti-hDEC205-WT1223-273, and anti-hDEC205-WT1324-371 were tested for their T-cell stimulatory capacities. Mature monocyte-derived DCs loaded with anti-hDEC205-WT191-138 could induce ex vivo T-cell responses in 12 of 16 blood samples collected from either healthy or HSC transplanted individuals compared to included controls (P < 0.01). Furthermore, these T cells could kill WT1-overexpressing THP-1 leukemia cells in vitro after expansion.
In conclusion, alongside proving the difficulty in expression and purification of intracellular WT1 as a vaccine protein, our results from this work introduce an alternative therapeutic vaccine approach to improve an anti-leukemia immune response in the context of allogeneic HSCT and potentially beyond.
Hintergrund
In Südafrika sind ca. 30 % aller schwangeren Frauen mit HIV infiziert. Physiologische Veränderungen in der Schwangerschaft alterieren die Metabolisierung der antiretroviralen Medikamente, was subtherapeutische oder toxische Serumspiegel zur Folge haben kann. Konsequenzen ergeben sich in Form einer Risikoerhöhung einer Mutter-Kind-Übertragung und Resistenzentwicklung einerseits sowie toxischer Wirkungen andererseits. Über die Pharmakokinetik antiretroviraler Substanzen bei Schwangeren ist bislang wenig bekannt. In der vorliegenden Untersuchung wurden deshalb im Setting der routinemäßigen Patientenversorgung Nevirapin (NVP)- und Efavirenz (EFV)-Serumkonzentrationen bei antiretroviral behandelten Schwangeren gemessen.
Methoden
Die Untersuchungen erfolgten im Rahmen des DFG-geförderten Internationalen Graduiertenkollegs IRTG1522, einem Partnerprojekt der Universitäten Stellenbosch und Kapstadt/Südafrika und Würzburg, prospektiv im Tygerberg Academic Hospital in Kapstadt. Bei 108 HIV-infizierten Schwangeren, die mindestens seit 4 Wochen eine NVP- (n=52) oder EFV-Medikation (n=56) erhielten, wurden Serumproben entnommen. Die Messung der NVP- und EFV-Serumkonzentrationen erfolgte im TDM-Labor der Infektiologie am Universitätsklinikum Würzburg mittels einer etablierten Gaschromatographie bzw. Hochflüssigkeitschromatographie-Methode.
Ergebnisse
Nur 28 Proben (54%) der NVP-Konzentrationen lagen im therapeutischen Bereich (3.000 - 8.000 ng/ml) 36 Proben (64%) der EFV-Konzentrationen lagen im therapeutischen Bereich (1.000 - 4.000 ng/ml). Es zeigte sich ein signifikanter negativer Zusammenhang zwischen EFV-Konzentrationen und einem BMI >25.
In den univariaten Regressionen der einzelnen Variablen gab es einen signifikanten negativen Zusammenhang zwischen NVP-Konzentrationen und der Anzahl an Schwangerschaftswochen sowie einen signifikant positiven Zusammenhang zwischen NVP-Konzentrationen bei einer Begleitmedikation mit Valproat.
Es konnte ein signifikanter Zusammenhang zwischen NVP-Konzentrationen <3.000 ng/ml und einem virologischen Versagen nachgewiesen werden. Bei EFV konnte dieser signifikante Zusammenhang zu Konzentrationen <1.000 ng/ml gefunden werden.
Mittels Therapeutischem Drug Monitoring konnten 6 NVP- und 4 EFV-Serumspiegel detektiert werden, die eine unzureichende Medikamenteneinnahme der Patientinnen widerspiegelten. Bei einer Patientin fand eine vertikale Transmission statt.
Mit dieser Untersuchung konnte gezeigt werden, dass eine große Anzahl an schwangeren Frauen Serumspiegel außerhalb des therapeutischen Bereiches aufwiesen. Die pharmakokinetischen Veränderungen im Laufe der Schwangerschaft waren am ehesten Ursache für subtherapeutische Serumspiegel. Weiterhin zeigten sich subtherapeutische Konzentrationen als Auslöser von virologischem Versagen.
Schlussfolgerungen
Die Ergebnisse dieser Untersuchung legen nahe, dass ein Therapeutisches Drug Monitoring integraler Bestandteil der antiretroviralen Therapie von schwangeren Frauen im dritten Trimenon in Südafrika sein sollte.
Die vorliegende Arbeit stellt einen Beitrag zur Chemie höherkoordinierter Silicium(II) und Silicium(IV)-Verbindungen dar. Ein wesentlicher Teilaspekt der durchgeführten Untersuchungen betraf das Studium der Reaktivität der beiden donorstabilisierten Silylene 1 und 2.
Im Einzelnen wurden die folgenden Teilprojekte bearbeitet: Die neutrale, hexakoordinierte Silicium(IV)-Verbindung 10 und die ionische, pentakoordinierte Silicium(IV)-Verbindung 11 wurden Umsetzung von 5 (dem Chloro-Analogon von 10) mit Me3SiBr bzw. Me3SiI in Transsilylierungsreaktionen dargestellt. Die mit 10 verwandten Verbindungen 5–9 wurden bereits früher synthetisiert und im Rahmen dieser Arbeit zusammen mit 10 erstmalig bezüglich ihrer Moleküldynamik in Lösung untersucht. Die Verbindungen 5–10 zeigten in Lösung bei Raumtemperatur unterschiedlich stark ausgeprägte Dynamikphänomene, die mittels VT-NMR-Experimenten untersucht wurden.
Die neutralen, hexakoordinierten Silicium(IV)-Verbindungen 12 und 16 wurden durch sequentielle Umsetzung der entsprechenden sekundären Amine Ph2NH bzw. iPr2NH mit n-Butyllithium und Kohlenstoffdisulfid sowie anschließende Umsetzung mit Tetrachlorsilan dargestellt und als die Acetonitrilsolvate 12·MeCN bzw. 16·MeCN isoliert. Es handelt sich hierbei um die ersten hexakoordinierten Silicium(IV)-Komplexe mit einem SiS4Cl2-Gerüst.
Die neutrale, hexakoordinierte Silicium(IV)-Verbindung 17 mit einem SiN4Cl2-Gerüst wurde durch Umsetzung des Silylens 2 mit Chlor dargestellt. Im Gegensatz zu dieser oxidativen Addition schlug die Synthese von 17 durch Umsetzung von Tetrachlorsilan mit zwei Moläquivalenten des entsprechenden Lithiumguanidinats [iPrNC(NiPr2)NiPr]Li fehl: Es entstand lediglich der entsprechende pentakoordinierte Mono(guanidinato)silicium(IV)-Komplex mit drei Chloroliganden. Die Umsetzung von 1,2-Diphenylethin mit dem Silylen 1 lieferte den neutralen, hexakoordinierten Silicium(IV)-Komplex 19.
Der neutrale, pentakoordinierte Silicium(IV)-Komplex 20 wurde in einer Redoxreaktion durch Umsetzung des Silylens 2 mit Dimangandecacarbonyl dargestellt. Dabei wurde das Silicium(II)- zu einem Silicium(IV)-Fragment oxidiert und das Dimanganfragment unter Verlust von zwei Carbonylliganden reduziert. Die neutralen, tetrakoordinierten Silicium(II)-Übergangsmetallkomplexe 22, 23 und 24 (isoliert als 24·THF) konnten durch Umsetzung des Silylens 2 mit den entsprechenden Übergangsmetalldibromiden bzw. Nickel(II)-bromid–1,2-Dimethoxyethan dargestellt werden. Im Fall von Nickel gelang die Umsetzung mit dem freien NiBr2 nicht. Die Verbindungen 22 und 23 stellen paramagnetische Komplexe mit jeweils tetraedrisch koordinierte Übergangsmetallatomen dar. Das Nickelatom in Verbindung 24·THF ist dagegen quadratisch-planar koordiniert und damit diamagnetisch, wie es für d8-Metalle auch zu erwarten ist. Den drei Verbindungen 22, 23 und 24·THF gemeinsam ist der besondere Bindungsmodus einer der beiden Guanidinatoliganden, der das Siliciumatom und das Übergangsmetallatom miteinander verbrückt, was zur Ausbildung einer spirocyclischen Struktur führt. Der neutrale, pentakoordinierte Zink–Silylen-Komplex 25 wurde in einer Lewis-Säure/Base-Reaktion durch Umsetzung des Silylens 2 mit Zink(II)-bromid dargestellt und als das Solvat 25·0.5Et2O isoliert. Obwohl sich das Reaktionsprodukt wie auch bei den Verbindungen 22–24 als ein Lewis-Säure/Base-Addukt verstehen lässt, ist der Koordinationsmodus von Verbindung 25 anders: Beide Guanidinatoliganden sind bidentat an das Siliciumatom gebunden.
Die neutralen Bis(silylen)palladium(0)- bzw. Bis(silylen)platin(0)-Komplexe 28 und 29 repräsentieren die ersten homoleptischen, dikoordinierten Bis(silylen)-Komplexe dieser Metalle mit N-heterocyclischen Silylenliganden und im Fall des Platin(0)-Komplexes 29 den ersten homoleptischen, dikoordinierten Platin(0)–Silylen-Komplex überhaupt. Verbindung 28 wurde durch Umsetzung von drei Moläquivalenten des Silylens 2 mit dem Palladium(II)-Komplex [PdCl2(SMe2)2] dargestellt. Dabei reduziert ein Moläquivalent des Silylens den Palladium(II)-Komplex und wird selbst zu Verbindung 17 oxidiert und die beiden verbliebenen Moläquivalente des Silylens substituieren die Dimethylsulfidliganden am Palladiumatom. Dieselbe Synthesestrategie ließ sich jedoch nicht auf die Darstellung von Verbindung 29 übertragen. Offenbar reicht das Reduktionspotenzial des Silylens 2 hier nicht aus. Zur Darstellung von Verbindung 29 wurde zunächst der Platin(II)-Komplex [PtCl2(PiPr3)2] mit Natrium/Naphthalin reduziert und anschließend wurden die beiden Triisopropylphosphanliganden durch Silylenliganden substituiert.
Ziel dieser Arbeit war die Reduktion des Modenvolumens in Mikrokavitäten. Ein klei-nes Modenvolumen ist für die Stärke der Licht-Materie-Wechselwirkung wesentlich, weil dadurch z.B. die Schwelle für kohärente Lichtemission gesenkt werden kann [1]. Der Purcell-Faktor, ein Maß für die Rate der spontanen Emission, wird durch ein mi-nimales Modenvolumen maximiert [2, 3]. Im Regime der starken Kopplung steigt mit Abnahme des Modenvolumens die Rabi-Aufspaltung und damit die maximale Tempe-ratur, bei der das entsprechende Bauteil funktioniert [4, 5]. Spektrale Eigenschaften treten deutlicher hervor und machen die Funktion der Struktur stabiler gegenüber stö-renden Einflüssen.
Der erste Ansatz, das Modenvolumen einer Mikrokavität zu reduzieren, zielte darauf, die Eindringtiefe der optischen Mode in die beiden Bragg-Spiegel einer Mikrokavität zu minimieren. Diese hängt im Wesentlichen vom Kontrast der Brechungsindizes der alternierenden Schichten eines Bragg-Spiegels ab. Ein maximaler Kontrast kann durch alternierende Schichten aus Halbleiter und Luft erreicht werden. Theoretisch kann auf diese Weise das Modenvolumen in vertikaler Richtung um mehr als einen Faktor 6 im Vergleich zu einer konventionellen Galliumarsenid/Aluminiumgalliumarsenid Mikro-kavität reduziert werden. Zur Herstellung dieser Strukturen wurden die aluminiumhal-tigen Schichten einer Galliumarsenid/Aluminiumgalliumarsenid Mikrokavität voll-ständig entfernt und so der Brechungsindexkontrast maximiert. Die Schichtdicken sind dabei entsprechend anzupassen, um weiterhin die Bragg-Bedingung zu erfüllen. Die Herstellung einer freitragenden Galliumarsenid/Luft-Mikrokavität konnte so erfolg-reich demonstriert werden. Die Photolumineszenz der Bauteile weist diskrete Reso-nanzen auf, deren Ursache in der begrenzten lateralen Größe der Strukturen liegt. In leistungsabhängigen Messungen kann durch ausgeprägtes Schwellenverhalten und auf-lösungsbegrenzte spektrale Linienbreiten Laseremission nachgewiesen werden. Wegen der Abhängigkeit der photonischen Resonanz vom genauen Brechungsindex in den freitragenden Schichten eignen sich die vorgestellten Strukturen auch zur Bestimmung von Brechungsindizes. Alternativ kann die photonische Resonanz durch Einbringen verschiedener Gase in die freitragenden Schichten abgestimmt werden. Beides konnte mit Erfolg nachgewiesen werden. Der Nachteil dieses Ansatzes liegt vor allem darin, dass ein elektrischer Betrieb der so gefertigten Strukturen nicht möglich ist. Hier bie-tet der zweite Ansatz eine bestmögliche Lösung.
Das alternative Konzept für den oberen Bragg-Spiegel einer konventionellen Galli-umarsenid/Aluminiumgalliumarsenid Mikrokavität ist das der Tamm-Plasmonen. Der photonische Einschluss wird hier durch einen unteren Bragg-Spiegel und einem dün-nen oberen Metallspiegel erreicht. An der Grenzfläche vom Halbleiter zum Metall bil-den sich die optischen Tamm-Plasmonen aus. Dabei kann der Metallspiegel gleichzei-tig auch als elektrischer Kontakt genutzt werden. Die Kopplung von Quantenfilm-Exzitonen an optische Tamm-Plasmonen wird in dieser Arbeit erfolgreich demons-triert. Im Regime der starken Kopplung wird mittels Stark-Effekt eine vollständige elektro-optische Verstimmung, d.h. vom Bereich positiver bis hin zur negativen Ver-stimmung, des Quantenfilm-Exzitons gegenüber der Tamm-Plasmonen Mode nachge-wiesen. Die Messungen bestätigen entsprechend des reduzierten Modenvolumens (Faktor 2) eine erhöhte Rabi-Aufspaltung. Dabei sind die spektrale Verschiebung und die Oszillatorstärke des Quantenfilm-Exzitons konsistent mit der Theorie und mit Li-teraturwerten. Der wesentliche Nachteil des Ansatzes liegt in der maximalen Güte, die durch den großen Extinktionskoeffizienten des Metallspiegels limitiert ist.
Die postovulatorische Alterung sowie die ovarielle Alterung konnten bei der Anwendung assistierter Reproduktionstechniken (ARTs) als entscheidende Faktoren identifiziert werden, die den Reproduktionserfolg nachhaltig beeinträchtigen. Die postovulatorische Alterung tritt ein, sobald die reife Eizelle nicht mehr innerhalb ihres physiologischen Zeitfensters befruchtet wird. Die ovarielle Alterung beschreibt hingegen die Abnahme des Follikel-Vorrats mit zunehmendem Alter des weiblichen Individuums bzw. des Ovars. Sowohl die postovulatorische Alterung als auch die ovarielle Alterung führen u.a. zu einer reduzierten Oozytenqualität und einer geringeren Blastozystenrate. Die Zielsetzung dieser Arbeit bestand darin, den Einfluss der postovulatorischen Alterung und der ovariellen Alterung im Holstein-Rind (Bos taurus) auf die DNA-Methylierung entwicklungsrelevanter Gene in Eizellen und Embryonen zu untersuchen. Aus Schlachthof-Ovarien wurden Antralfollikeln unterschiedlicher Größe (<2 mm, 3-5 mm und >6 mm) isoliert. Eizellen aus Follikeln der Größe 3-5 mm wurden für 24h (physiologisch) und 48h (gealtert) in vitro gereift (IVM). Die gereiften Oozyten wurden anschließend in vitro fertilisiert und Embryonen im 4-6 Zellstadium generiert. Sowohl in den unreifen Eizellen aus Antralfollikeln unterschiedlicher Größe als auch in den gereiften Oozyten und den Embryonen wurde die Promotormethylierung der Gene bH19, bSNRPN, bZAR1, bDNMT3A, bOCT4, bDNMT3Lo und bDNMT3Ls analysiert. Zur Untersuchung der ovariellen Alterung wurden mittelgroßen Antralfollikel aus Ovarien lebender Rinder (in vivo) unterschiedlichen Alters (9-12 Monate, 3-7 Jahre und 8-11 Jahre) gewonnen. In den daraus isolierten unreifen Eizellen wurde die DNA-Methylierung der Promotorregionen der Gene bTERF2, bREC8, bBCL-XL, bPISD, bBUB1, bDNMT3Lo, bH19 und bSNRPN bestimmt. Als Methode zur Analyse der Promotormethylierung wurde die Limiting Dilution Bisulfit-Sequenzierung angewendet.
In unreifen Eizellen aus Antralfollikeln unterschiedlicher Größe (<2 mm, 3-5 mm und >6 mm) konnte ein erhöhtes Auftreten abnormal methylierter Allele in den geprägten Genen bH19 und bSNRPN von Eizellen kleiner Follikel (<2 mm) identifiziert werden. Dieses Ergebnis könnte eine mögliche Ursache einer bereits bekannten und mehrfach beschriebenen geringeren Entwicklungskompetenz von Eizellen kleiner Follikel (<2 mm) auf epigenetischer Ebene darstellen.
Die verlängerte Reifungsdauer der IVM-Eizellen hatte eine signifikante Hypermethylierung in der Promotorregion des Gens DNMT3Lo von 48h-gereiften Eizellen zur Folge. Beim Übergang von 48h-gereiften Eizellen zum Embryo konnte eine signifikante Hypomethylierung von CpG7 des stammzellspezifischen Transkripts DNMT3Ls beobachtet werden. Diese CpG-Stelle wies ebenfalls einen signifikanten Anstieg von CpGs mit nicht-eindeutigem Methylierungszustand in unreifen Eizellen mit steigender Follikelgröße auf. Da sich die CpG-Position innerhalb eines Sequenz-Motivs einer Bindungsstelle des Transkriptionsfaktors CREB befindet, könnten die Methylierungsdaten auf eine Interaktion zwischen dem Transkriptionsfaktor CREB und der DNA-Methylierung während der Entwicklung und Reifung der Eizelle sowie der Transition von der Eizelle zum Embryo hindeuten.
Die DNA-Methylierungsprofile der untersuchten Gene in unreifen Eizellen aus Kühen unterschiedlichen Alters (9-12 Monate, 3-7 Jahre und 8-11 Jahre) wiesen keine signifikanten Unterschiede zwischen den Altersgruppen auf. Die ovarielle Alterung bei Rindern zwischen 9 Monaten und 11 Jahren zeigte damit keinen Effekt auf die DNA-Methylierung der untersuchten Promotorregionen der Gene bTERF2, bREC8, bBCL-XL, bPISD, bBUB1, bDNMT3Lo, bH19 und bSNRPN.
Nach einer simulierten postovulatorischen Alterung durch eine in vitro Reifung für 48h konnte eine Veränderung der DNA-Methylierung der Oozyten-spezifischen (DNMT3Lo) und Stammzell-spezifischen (DNMT3Ls) Promotoren des katalytisch inaktiven Cofaktors von DNMT3A, DNMT3L, beobachtet werden. Die veränderte DNA-Methylierung von DNMT3Ls tritt dabei erst im frühen Embryo in Erscheinung und interagiert vermutlich mit dem Transkriptionsfaktor CREB. Die Veränderungen von DNMT3Lo in Eizellen und DNMT3Ls in den daraus generierten Embryonen lässt vermuten, dass es sich hierbei um eine dynamische Anpassung des Embryos auf äußere Umweltbedingungen der Eizelle über die Methylierung der DNA handelt.
Traditional species identification based on morphological characters is laborious
and requires expert knowledge. It is further complicated in the case of
species assemblages or degraded and processed material. DNA-barcoding,
species identification based on genetic data, has become a suitable alternative,
yet species assemblages are still difficult to study. In the past decade
meta-barcoding has widely been adopted for the study of species communities,
due to technological advances in modern sequencing platforms and
because manual separation of individual specimen is not required. Here,
meta-barcoding is put into context and applied to the study of bee-collected
pollen as well as bacterial communities. These studies provide the basis
for a critical evaluation of the powers and limitations of meta-barcoding. Advantages
identified include species identification without the need for expert
knowledge as well as the high throughput of samples and sequences. In
microbiology, meta-barcoding can facilitate directed cultivation of taxa of interest
identified with meta-barcoding data. Disadvantages include insufficient
species resolution due to short read lengths and incomplete reference
databases, as well as limitations in abundance estimation of taxa and functional
profiling. Despite these, meta-barcoding is a powerful method for the
analysis of species communities and holds high potential especially for automated
biomonitoring.
Although the physiological roles of BChE are not yet determined to date, the importance of this enzyme is continuously increasing as it was found to be associated with several disorders like diabetes mellitus type 2, cardiovascular diseases, obesity and especially with Alzheimer’s disease (AD). In consequence, for investigations of BChE’s pathological role in these diseases and to find new medication strategies, the development of selective and potent inhibitors is necessary.
For this purpose, the current work progresses in five chapters on the exploration of the chemical, physical and biochemical properties of tetrahydroquinazoline based carbamates which were previously reported to be selective BChE inhibitors with potency in the low nanomolar range.
1) A Novel Way to Radiolabel Human Butyrylcholinesterase for PET through Irreversible Transfer of the Radiolabeled Moiety:
PET-radiotracers represent an innovative tool to determine the distribution and the expression of a biological target in vivo. BChE lacks to a large degree of such tracers with a few exceptions. In this work, methods were developed to incorporate the radioisotopes 11C and 18F into the carbamate moiety of an tetrahydroquinazoline based inhibitor. In contrast to reversibly acting PET-probes, the described radiotracers were proven by kinetic studies to transfer the radioisotope covalently onto the active site of BChE, thus labeling the enzyme directly and permanently.
2) Discovery of Highly Selective and Nanomolar Carbamate-Based Butyrylcholinesterase Inhibitors by Rational Investigation into Their Inhibition Mode:
To investigate the role of the tetrahydroquinazoline carrier scaffold on BChE inhibition, carbamate based inhibitors were synthesized. These compounds were successively used to perform kinetic investigations to determine their inhibition mode. Based on these data, a plausible binding model was postulated explaining the influence of the tetrahydroquinazoline carrier scaffold for binding at BChE’s active site just before carbamate transfer takes place. Additionally, these compounds feature neuroprotective properties and prevent oxidative stress induced cell death in their carbamate form as well as after the release of the tetrahydroquinazoline carrier scaffold.
3) Dual Addressing of Butyrylcholinesterase by Targeting the Catalytic Active Site (CAS) and the Peripheral Anionic Site (PAS):
Compounds which are dual-targeting the CAS and the PAS of BChE are the most potent and selective BChE inhibitors to date with inhibition values in the picomolar range. In this work, a strategy is described how to turn tetrahydroquinazoline based carbamates into dual binding BChE inhibitors. These inhibitors feature a carbamate moiety which is covalently transferred onto the CAS of BChE, and in addition provide a second pharmacophore connected via a linker to the carbamate moiety which is proposed to target the PAS. Preliminary results reveal a high tolerance of BChE towards different linker lengths without decrease in affinity.
4) Investigation into Selective Debenzylation and Ring Cleavage of Quinazoline based Heterocycles:
The tetrahydroquinazoline system is well investigated in terms of its synthesis and its selective oxidation. To explore the reactivity of this system, a tetracyclic tetrahydroquinazoline was exposed to common reduction agents. These experiments revealed a high sensitivity of the tetrahydroquinazoline core towards several reduction conditions
5) Experimental and Theoretical Investigation into the Stability of Cyclic Aminals:
Tetrahydroquinazolines are known to degrade in acidic media through hydrolysis of their aminal system; but literature is lacking of a systematic investigation into this behavior. Therefore, different tetrahydroquinazolines were synthesized and exposed to phosphate buffered systems with defined pH-values. A clear increase of the hydrolysis rate of the aminal system was determined in dependency of an increasing acidic media. Computational studies predicted and experimental studies proved that hydrolysis takes place in an acidic environment while the condensation of this system is preferred in neutral or basic aqueous media.
Das Glioblastoma multiforme (GBM) ist der primäre maligne Hirntumor mit der höchsten Prävalenz bei Erwachsenen. Dieser astrozytäre Tumor ist durch ein besonders schnelles Wachstum und ein äußerst invasives Verhalten charakterisiert. Deshalb beträgt das mediane Überleben nach Diagnose trotz interdisziplinärer Therapie nur ungefähr 14,6 Monate.
Metastasis Associated in Colon Cancer-1 (MACC1) ist ein neuer prognostischer Marker für Metastasierung im kolorektalen Karzinom. Es ist ein transkriptioneller Regulator von Met, dem Rezeptor des Hepatocyte Growth Factor (HGF). Überexpression von MACC1 führt zur Induktion von Migration und Proliferation. Es wurde gezeigt, dass MACC1 auch in anderen Tumorentitäten wie dem Magenkarzinom, Bronchialkarzinom und hepatozellulärem Karzinom verstärkt exprimiert ist. Jedoch gab es bisher noch keine Daten über die Rolle von MACC1 in astrozytären Tumoren. Obwohl GBM nur selten metastasieren, ist ihr aggressives und invasives Verhalten mit dem von metastasierenden Tumoren vergleichbar. Deshalb war das Ziel dieser Arbeit zu zeigen, dass MACC1 auch eine wichtige Rolle in der Pathogenese von Glioblastomen spielen könnte.
Die MACC1-Expression von Glioblastomen wurde zunächst in silico mit frei zugänglichen Microarray-Platformen analysiert. Von Gewebeproben humaner niedergradiger Astozytome (LGA) und GBM wurde die MACC1- und Met-Expression mittels PCR bestimmt. Die Analyse der Expression auf Proteinebene wurde durch Immunhistochemie (IHC) von Patientengewebe durchgeführt. Funktionelle Analysen folgten in Form eines Sphäroidmigrationsassays von primären GBM Zellkulturen. Weiterhin wurde MACC1 in zwei GBM-Zelllinien stabil überexprimiert und deren Migration und Proliferation in Echtzeit gemessen. Komplettiert wurden die funktionellen Versuche durch einen Koloniebildungsassay.
Die Expression von MACC1 stieg mit zunehmendem WHO-Grad auf mRNA- und Proteinebene an. Die Analyse von MACC1 durch IHC erlaubte eine Differenzierung nicht nur zwischen ruhenden LGA und LGA welche später ein Rezidiv bildeten, bzw. Progress zeigten, sondern auch zwischen primären und sekundären GBM. Eine hohe MACC1-Expression war mit einer ungünstigen klinischen Prognose der Patienten assoziiert. Die endogene Expression von MACC1 korrelierte mit der Migrationsaktivität primärer GBM-Zellkulturen. Die Überexpression von MACC1 in GBM-Zelllinien induzierte Proliferation, Migration und Koloniebildung und korrelierte somit mit Schlüsseleigenschaften maligner Zellen.
Zusammenfassend zeigen die Ergebnisse dieser Arbeit zum ersten Mal eine essentielle Rolle von MACC1 für die Pathogenese von Glioblastomen. Deshalb könnte MACC1 ein potentielles neues therapeutisches Ziel für die Behandlung von Glioblastomen sein und eventuell sogar als neuer prognostischer Marker dienen.
Die Interaktion mit Gehirnendothelzellen stellt ein zentraler Schritt in der Infektionspathogenese von Neisseria meningitidis dar. In dieser Promotionsarbeit konnte gezeigt werden, dass die Infektion von menschlichen Gehirnendothelzellen mit N. meningitidis zu einer transienten Aktivierung der sauren Sphingomyelinase (ASM) gefolgt von einer vermehrten Ceramidproduktion führt. Als Antwort auf die Infektion mit N. meningitidis kommt es zu einer vermehrten Präsentation der ASM und von Ceramiden an der äusseren Seite der Plasmamembran und zu einer Ausbildung von großen Ceramid-reichen Membran-Domänen, welche mit cortical plaque assoziierten Proteinen kolokalisieren. Bei dieser N. meningitids vermittelten Aktivierung der ASM spielt das bakterielle Aussenmembranprotein Opc sowie die Aktivierung der Phosphatidylcholin-spezifische Phospholipase C über die Interaktion von Opc mit Heparansulfat-Proteoglykane eine entscheidende Rolle. Die pharmakologische oder genetische Inhibition der ASM Funktion führt zu einer geringeren Invasivität der Meningokokken ohne dabei die Adhärenz zu beeinflussen. Im Einklang mit diesen Ergebnissen steht die Beobachtung, dass die geringere Invasivität von ausgewählten Isolaten des ST-11/ST-8 Komplex in menschlichen Gehirnendothelzellen direkt mit ihrer eingeschränkter Fähigkeit korreliert, die ASM zu aktivieren bzw. eine Ceramidproduktion zu induzieren. Schlussfolgernd ist die ASM Aktivierung und eine nachfolgende Ceramidproduktion essenziell für die Internalisierung von Opc-exprimierende Meningokokken in Gehirnendothelzellen und bietet einen Erklärungsansatz für die unterschiedliche Invasivität von verschiedenen N. meningitidis Stämmen.
To simplify a judgment, people often base it on easily accessible information. One cue that is usually readily available is processing fluency – a metacognitive feeling of ease of cognitive processing. Consequently, processing fluency is used as a cue for many different types of judgment, such as judgment of truth, confidence, and novelty. The present work describes results of three studies investigating various aspects of processing fluency effects on judgment.
Processing fluency has been sometimes equated with speed of a cognitive process. Therefore, response times have been used for evaluation of processing fluency. However, response times in experimental tasks often do not encompass only the time needed for a given process, but also the time needed for a decision based on the resulting information. The study described in Chapter II uses a novel experimental method that enables separation of reading and decision times. The results show that people make a decision about liking of pseudowords faster when the pseudowords are hard-to-pronounce (i.e., disfluent) than when they are moderate in pronounceability. This suggests that response times cannot be used as a proxy for processing fluency when they include the time needed to make a decision.
One of the studies of judgmental effects of processing fluency showed that food additives with easier pronounceable names are judged to be less harmful than those with hard-to-pronounce names. While people encounter food additives that are safe more often, this environmental association may be in the opposite direction for some categories of objects. For example, people are more likely to see names of especially dangerous criminals in the news. Chapter III describes a study which initially tested whether the fluency-safety association may be in the opposite direction for some categories of objects as a consequence of this selective exposure to especially dangerous exemplars. The results did not show support for this hypothesis. Furthermore, subsequent studies suggest that the previously found association between fluency and safety is replicable with the original stimuli used in the previous research, but not with newly constructed stimuli.
Chapter IV describes a study which applied a finding from the processing fluency literature to a positive psychology exercise in order to increase its effectiveness. Namely, the experiment manipulated the number of good things that participants listed daily for two weeks as part of the exercise. While listing more things was considered harder, the number of things listed each day had no effect on effectiveness of the exercise.
Das Ziel der Arbeit war zu untersuchen, ob der Stoffwechsel kolorektaler Karzi-nomzellen geeignete Targetstrukturen für mögliche therapeutische Ansätze aufweist. In Krebszellen induziert sowohl der Warburg-Effekt bei Normoxie als auch die anaerobe Glykolyse bei Hypoxie eine massive Bildung von Laktat. Wird die Krebszelle dauerhaft daran gehindert, die für die Glykolyse notwendi-gen Reduktionsäquivalente NADH+H+ mit Hilfe der Laktatdehydrogenase zu reoxidieren und/oder Laktat über die Transporter MCT1 und MCT4 nach außen zu schleusen, dann löst diese Kombination aus Mangelsituation und intrazellulärer Ansäuerung den apoptotischen Zelltod aus. Für die Situation in vivo ist entscheidend, dass auch Zellen von Normalgeweben zwar Laktat in Hypoxie bilden, dies jedoch keine vorherrschende physiologische Situation darstellt.
Die Hemmstoffe Natriumoxamat (NaOx) für die Laktatdehydrogenase und α-Cyano-4-Hydroxycinnamat (αCHC) für MCT1 und MCT4 wurden an den sechs humanen kolorektalen Karzinomzelllinien Colo741, HCT116, HT29, LS174T, SW620 und WiDr untersucht. Zusätzlich wurde der Glukoseverbrauch und die Laktatbildung bestimmt und die Funktion der Atmungskette überprüft. Die IC50-Werte für 5-FU, NaOx und αCHC wurden bestimmt und danach NaOx in einer Konzentration von 40x10-3 mol/L, αCHC in einer Konzentration von 2x10-3 mol/L und 5-FU in einer Konzentration von 5x10-6 mol/L eingesetzt. Die Zellen wurden bei tumorphysiologischen Sauerstoffkonzentrationen von 5 % und 1 % Sauerstoff für bis zu 120 Stunden inkubiert.
Die Funktion der Atmungskette in den Mitochondrien der kolorektalen Karzi-nomzellen wurde u. a. durch Bestimmung wichtiger Kenngrößen wie dem P:O Quotienten und des respiratorischen Kontrollindex (RKI) nachgewiesen. Fünf der sechs Karzinomzelllinien wiesen im Vergleich zur Kontrollzelllinie J774 einen verringerten P:O-Quotienten und respiratorischen Kontrollindex (RKI) auf, was darauf hindeutet, dass die Funktion der Mitochondrien dieser Zellen im Vergleich zu Kontrollzellen zwar verringert war, aber nicht vollständig aufgehoben. Dieses Ergebnis stützt die allgemein akzeptierte Auffassung, dass die meisten Tumore über funktionelle Mitochondrien verfügen.
Durch die Analyse des Glukosestoffwechsels wurden die sechs kolorektalen Zelllinien, die einen unterschiedlich stark ausgeprägten glykolytischen Phänotyp aufwiesen, nach der Stärke der Laktatbildung bei 5 % Sauerstoff in drei Kategorien eingeordnet. Zudem wurde für jede der sechs Zelllinien die Expression von LDH-A, LDH-B sowie MCT-1 und MCT-4 auf Proteinebene nachgewiesen.
Wesentliches Ziel der Untersuchungen war die Überprüfung des antiprolife-rativen Potentials der beiden Inhibitoren NaOx und αCHC einzeln oder in Kombination mit 5-FU bei den tumorspezifischen Sauerstoffkonzentrationen von 5 % und 1 %. Die Kombination aus NaOx und αCHC induzierte bei 1 % Sauerstoff nach 9 Tagen in Kultur zytotoxische Effekte und war damit so wirksam wie 5x10-6 mol/L 5-FU. Die Zugabe von 5-FU zur Kombination aus NaOx und αCHC führte zu keiner Steigerung des zelltoxischen Effektes. Die beiden Inhibitoren NaOx und αCHC waren für SW620 Zellen weniger wirksam als für Zellen der anderen fünf Zelllinien. Das mehr „oxidative“ Profil von SW620 Zellen (bester P:O-Quotient, geringste Laktatbildung bei 5 % und 1 % Sauerstoff; zudem die höchsten IC50-Werte für NaOx und αCHC) könnte erklären, warum die beiden Stoffwechselinhibitoren, die einen glykolytischen Phänotyp (starke Bildung von Laktat) erfordern, für SW620 Zellen von geringerer Wirksamkeit waren.
Für die Hemmstoffe NaOx und αCHC wurden zytostatische bzw. zytotoxische Effekte in kolorektalen Karzinomzellen gezeigt. Dies deutet darauf hin, dass Krebszellen auf einen ungehinderten glykolytischen Stoffwechsel angewiesen sind. Für beide Hemmstoffe wurde ebenfalls gezeigt, dass sie auch bei tumorre-levanten Sauerstoffkonzentrationen von 5 % und 1 % wirksam sind.
Das Ziel der vorliegenden Arbeit ist es, den Stellenwert von Trost im Umgang mit Patienten und Angehörigen aufzuzeigen und mittels einer empirischen Untersuchung zur Sterbebegleitung festzustellen, wie dies in der Realität im Klinikalltag umgesetzt wird. Hierfür wurde die Sterbebegleitung auf zwei unterschiedlichen Stationen innerhalb eines Krankenhauses qualitativ ausgewertet.
Der theoretische Teil der vorliegenden Arbeit zeigt anhand wissenschaftlicher Daten, welche unterschiedlichen Bedürfnisse schwerstkranke und sterbende Patienten und ihre Angehörigen an den Arzt im Hinblick auf Trost haben und wie diesen angemessen begegnet werden kann.
Mittels teilstrukturiertem Leitfadeninterview wurden Ärzte und Pflegekräfte als Experten dazu befragt, wie die Begleitung sterbender Patienten und ihrer Angehörigen aussieht und wie sie den Betroffenen Trost spenden. Die Aspekte Zeit, Raum, Personal und Ausbildung und ihr Einfluss auf die Begleitung wurden thematisiert. Zuletzt wurden die Experten nach ihrer Vorstellung von einem würdevollen Sterben im Krankenhaus und Ansätzen zur Verbesserung des Umgangs mit sterbenden Patienten und ihren Angehörigen gefragt.
Nach dem Prinzip des Theoretical Sampling der Grounded Theory nach Glaser und Strauss wurde die Sterbebegleitung auf einer Normal- und einer Palliativstation gegenübergestellt. Insgesamt wurden vier Ärzte und acht Pflegekräfte interviewt. Das Sampling pro Gruppe wurde beendet, nachdem die theoretische Sättigung erreicht war.
Die Auswertung der Interviews erfolgte nach dem Prinzip von Meuser und Nagel. Es wurde untersucht, wie Trost in der Begleitung sterbender Patienten und ihrer Angehörigen gestaltet wird. Unterschiede zwischen den beiden Stationen wurden herausgearbeitet und analysiert, worauf diese zurückzuführen sind. Lösungsansätze für eine Verbesserung der Situation im Krankenhaus wurden konzipiert.
Das Ergebnis der Untersuchung zeigt, dass sich alle befragten Ärzte und Pflegekräfte der existentiellen Ausnahmesituation von Sterbenden und Angehörigen bewusst sind und ein hohes Maß an Bereitschaft vorhanden ist, eine adäquate Begleitung zu gewährleisten.
Die Möglichkeiten der Sterbebegleitung auf der Palliativstation werden insgesamt als gut bewertet. Im Mittelpunkt steht die individuelle Begleitung des sterbenden Patienten und seiner Angehörigen. Bemängelt werden ein teilweise zu hoher Patientendurchlauf und eine zu geringe pflegerische Besetzung im Nachtdienst.
Im Gegensatz dazu wird die Arbeit der Begleiter auf der Normalstation durch den niedrigeren Personalschlüssel und die gegebenen Räumlichkeiten limitiert. Problematisch ist vor allem die mangelnde Ausbildung im Umgang mit Sterbenden und Angehörigen.
Um die Situation in Krankenhäusern, insbesondere auf den Normalstationen zu verbessern, sollte ein gesellschaftliches Umdenken stattfinden. Voraussetzung hierfür ist das Bewusstsein und die Akzeptanz, dass Sterben unabdingbar zum Leben gehört und somit auf jeder Station eines Krankenhauses stattfindet. Auf politischen Ebenen können entsprechende Maßnahmen in die Wege geleitet und die notwendigen Mittel bereitgestellt werden, damit nicht nur auf Palliativ- sondern auch auf Normalstationen geschultes Personal und geeignete Räumlichkeiten zur Verfügung stehen, um allen sterbenden Patienten und ihren Angehörigen eine bestmögliche Begleitung zuteilwerden zu lassen.
Die kongenitale Zwerchfellhernie ist eine seltene aber dramatische Fehlbildung, die bis heute mit einer hohen prä- und postnatalen Mortalität vergesellschaftet ist. Der Defektverschluss ist ein entscheidender Schritt in der Therapie.Bei größeren Zwerchfellhernien muss ein Patch zum Verschluss verwendet werden. Hierbei ist die Verwendung von Gore-Tex©, ein Mesh aus PTFE, ein weit verbreiteter Standard. Obwohl biokompatible Materialien Vorteile gegenüber synthetischen Materialien wie PTFE aufweisen, konnte eine echte biokompatible Alternative zum Patchverschluss noch nicht gefunden werden.
Auf Grund guter Ergebnisse im Klein- und Großtiermodell für den Einsatz von Lyoplant an Bauchwanddefekten sollte geprüft werden, ob Lyoplant® auch für den Einsatz am Zwerchfell geeignet ist. Da es sich um die erstmalige Anwendung von Lyoplant® am Zwerchfell handelte, wurde ein Kleintiermodell zur Erprobung gewählt.
Verglichen wurde die Verwendung von Lyoplant® mit dem momentanen Standardmaterial Gore-Tex©. Als Versuchstiere dienten 15 junge, gesunde Wistar Furth-Ratten, denen ein 1,0 x 0,5 cm großer Defekt im linken Zwerchfell gesetzt wurde, welcher anschließend mit dem Fremdmaterial (Gore-Tex©: n = 5; Lyoplant®: n = 6; Direktnaht-Kontrollgruppe: n = 2) verschlossen wurde. Nach einem Versuchszeitraum von 12 Wochen, in denen die Tiere regelmäßig untersucht wurden, wurde der Patch mitsamt umgebendem Gewebe zur histologischen Analyse entnommen. Alle Tiere zeigten eine physiologische Gewichtsentwicklung. Bei keinem Tier kam es zur Hernienbildung. Starke Adhäsionen zur Leber konnten bei allen Versuchstieren inklusive der Kontrollgruppe festgestellt werden, zur Pleura zeigten lediglich 50% der Lyoplant®-Versuchsgruppe Verwachsungen. Die Adhäsionstendenz von Lyoplant® ist damit höher als die von Gore-Tex© oder bei Primärverschluss. In der mikroskopischen Auswertung konnte gezeigt werden, dass sowohl Lyoplant®, als auch Gore-Tex© gut in das Nachbargewebe integriert werden. Auch der Umbauprozess in körpereiniges Kollagen für das biokompatible Material konnte bestätigt werden. Lyoplant® wird mit einer maximalen Umbaurate von 1,1%Flächananteil/Tag umgewandelt. Innerhalb des Lyoplant®-Patchs konnte zudem eine kontinuierliche Neovaskularisation, vergleichbar mit der der Übergangsbereiche festgestellt werden. Die immunologischen Prozesse konnten nur teilweise beurteilt werden, sodass hier kein abschließendes Urteil erlaubt ist, gaben jedoch Hinweise auf einen Ablauf des Immunantwort über den TH2-Pathway.
Diese Ergebnisse zeigen, dass der Einsatz von Lyoplant® zur Defektdeckung am Zwerchfell möglich ist. Um die Frage nach einer möglichen zukünftigen klinischen Anwendung beantworten zu können, sind weitere tierexperimentelle Studien im Klein- und Großtiermodell nötig.
Neue Erkenntnisse über elektronische Eigenschaften von Festkörpern legen den Grundstein für innovative Anwendungen der Zukunft. Von zentraler Bedeutung sind insbesondere die Eigenschaften der Elektronenspins. Um diese besser zu verstehen, befasst sich die vorliegende Arbeit mit der experimentellen Analyse der elektronischen Struktur von topologischen Isolatoren (Sb$_2$Te$_3$ , Bi$_2$Se$_x$Te$_{3−x}$, Bi$_{1.5}$Sb$_{0.5}$Te$_{1.8}$Se$_{1.2} und Bi$_{1.4}$Sb$_{1.1}$Te$_{2.2}$S$_{0.3}$) und Kristallen mit starker Spin-Bahn-Wechselwirkung (BiTeI) mittels Photoelektronenspektroskopie. Zu Beginn werden die zum Verständnis dieser Arbeit benötigten Grundlagen erklärt sowie die unterschiedlichen zum Einsatz kommenden Techniken eingeführt. Der Hauptteil der Arbeit teilt sich in drei Forschungsschwerpunkte. Der erste Teil befasst sich mit den elektronischen Eigenschaften der Valenzbandstruktur von Sb2Te3 und den auftretenden Oberflächenzuständen. Durch gezielte Variation der Energie der anregenden Strahlung wird der Charakter der Wellenfunktion des topologischen Oberflächenzustands und dessen Wechselwirkung mit Valenzzuständen erforscht. Dabei spielt die Topologie der Volumenbandstruktur eine grundlegende Rolle. Der zusätzliche Vergleich zu Photoemissionsrechnungen ermöglicht detaillierte Einblicke in die Wechselwirkung zwischen Oberflächen- und Volumenzuständen und gibt Aufschluss darüber, wie diese vermittelt werden.
Im zweiten Abschnitt wird durch die Analyse des gemessenen Photoelektronenspins das Zusammenspiel der Spintextur des Grundzustands und Endzuständen in Bi2Te3 untersucht. Dabei treten, im Gegensatz zu Grundzustandsrechnungen, Radialkomponenten des Polarisationsvektors in nichtsymmetrischer Messgeometrie auf. Sowohl deren Energieabhängigkeit als auch deren Auftreten in Photoemissionsrechnungen (1-Schritt-Modell) deutet darauf hin, dass diese ihren Ursprung in Übergangsmatrixelementen des Photoemissionsprozesses haben. Dieses Ergebnis wird mit Spinpolarisationsmessungen am Oberflächenzustand des nicht-topologischen Schichtsystems BiTeI verglichen.
Im dritten Teil werden Auswirkungen unterschiedlicher Manipulationen der untersuchten Materialien auf deren elektronische Eigenschaften beschrieben. Die Adsorption von Bruchteilen einer monoatomaren Lage des Alkalimetalls Caesium auf die Oberfläche des topologischen Isolators Sb2Te3 wird systematisch untersucht. Dadurch kann dessen intrinsische p-Dotierung teilweise abgebaut werden, wobei die Valenzbandstruktur trotz der Reaktivität des Adsorbats intakt bleibt. Des Weiteren werden Auswirkungen von Änderungen der Kristallstöchiometrie durch Volumendotierung vergleichend diskutiert.
Ausblickend befasst sich das Kapitel mit dem Verhalten geringer Mengen ferromagnetischer
Materialen (Fe, Ni) auf den Oberflächen der topologischen Isolatoren. Für die verschiedenen Adsorbate werden Trends aufgezeigt, die von Temperatur und Zusammensetzung des Substratkristalls abhängen.
This dissertation focuses on the drivers of international capital flows to emerging markets, as well as the determinants of crises in emerging markets. Particular emphasis is devoted to the role of U.S. monetary policy. The dissertation consists of three independent chapters.
Chapter 1 is a survey of the voluminous empirical literature on the drivers of capital flows to emerging markets. The contribution of the survey is to provide a comprehensive assessment of what we can say with relative confidence about the empirical drivers of EM capital flows. The evidence is structured based on the recognition that the drivers of capital flows vary over time and across different types of capital flows. The drivers are classified using the traditional framework for external and domestic factors (often referred to as “push versus pull” drivers), which is augmented by a distinction between cyclical and structural factors. Push factors are found to matter most for portfolio flows, somewhat less for banking flows, and least for foreign direct investment (FDI). Pull factors matter for all three components, but most for banking flows. A historical perspective suggests that the recent literature may have overemphasized the importance of cyclical factors at the expense of longer-term structural trends.
Chapter 2 undertakes an empirical analysis of the drivers of portfolio flows to emerging markets, focusing on the role of Fed policy. A time series model is estimated to analyze two different concepts of high frequency portfolio flows, including monthly data on flows into investment funds and a novel dataset on monthly portfolio flows obtained from individual national sources. The evidence presented in this chapter suggests a more nuanced interpretation of the role of U.S. monetary policy. In the existing literature, it is traditionally argued that Fed policy tightening is unambiguously negative for capital flows to emerging markets. By contrast, the findings presented in this dissertation suggest that it is the surprise element of monetary policy that affects EM portfolio inflows. A shift in market expectations towards easier future U.S. monetary policy leads to greater foreign portfolio inflows and vice versa. Given current market expectations of sustained increases in the federal funds rate in coming years, EM portfolio flows could be boosted by a slower pace of Fed tightening than currently expected or could be reduced by a faster pace of Fed tightening.
Chapter 3 examines the role of U.S. monetary policy in determining the incidence of emerging market crises. A negative binomial count model and a panel logit model are estimated to analyze the determinants of currency crises, banking crises, and sovereign defaults in a group of 27 emerging economies. The estimation results suggest that the probability of crises is substantially higher (1) when the federal funds rate is above its natural level, (2) during Fed policy tightening cycles, and (3) when market participants are surprised by signals that the Fed will tighten policy faster than previously expected. These findings contrast with the existing literature, which generally views domestic factors as the dominant determinants of emerging market crises. The findings also point to a heightened risk of emerging market crises in the coming years if the Fed continues to tighten monetary policy.
Polymorphonuclear neutrophils (PMNs) are phagocytic cells of the innate immune system that efficiently kill bacteria. However, they also have regulatory effects on other immune cells and contribute to immunosuppression in cancer, which worsens the outcome. In particular, this has been demonstrated for a subset of granulocytic cells called myeloid- derived suppressor cells (MDSCs), but its distinction from PMNs is controversial. Most authors have explored the suppressive effects of MDSCs on T cells, but recent data suggest that NK cells are also affected. NK cells are crucial for the combat of tumor cells, in particular leukemic cells. There is hardly data available on the interaction between NK cells and suppressive granulocytic cells. Therefore, the aim of this thesis was to explore the effects of MDSCs and PMNs on the NK cell function against the leukemia cell line K562.
In co-culture experiments, I demonstrate that granulocytic MDSCs and PMNs had similar effects on NK cell function and homeostasis. On the one hand, they positively influenced the survival and maturation of NK cells. On the other, they inhibited the activation, cytotoxicity and cytokine production of NK cells, both IFNγ and TNFα, in response to K562 target cells. Furthermore, I show a down-regulation of the activating receptor NKp30 on NK cells in the presence of MDSCs or PMNs, which may form part of the underlying suppressive mechanisms.
However, there is also evidence for the involvement of other molecules. Further investigations are needed to confirm a relevant suppression of NK cells by granulocytic cells in cancer patients, and to identify therapeutic targets. The recognition that regular PMNs have similar effects on NK cells as MDSCs could simplify future experiments, since MDSCs are heterogeneous and laborious to isolate and identify.
NKcells and granulocytes are among the first immune cells to reconstitute after hematopoietic stem cell transplantation, and NK cells may be particularly exposed to suppressive effects of granulocytes this scenario. Modulating these suppressive effects of granulocytes on NK cells therapeutically may yield a better NK cell function and an improved cancer prognosis.

Staphylococcus aureus is a prevalent commensal bacterium which represents one of the leading causes in health care-associated bacterial infections worldwide and can cause a variety of different diseases ranging from simple abscesses to severe and life threatening infections including pneumonia, osteomyelitis and sepsis.
In recent times multi-resistant strains have emerged, causing severe problems in nosocomial as well as community-acquired (CA) infection settings, especially in the United States (USA). Therefore S. aureus has been termed as a superbug by the WHO, underlining the severe health risk originating from it. Today, infections in the USA are dominated by S. aureus genotypes which are classified as USA300 and USA400, respectively. Strains of genotype USA300 are responsible for about 70% of the CA infections.
The molecular mechanisms which render S. aureus such an effective pathogen are still not understood in its entirety. For decades S. aureus was thought to be a strictly extracellular pathogen relying on pore-forming toxins like α-hemolysin to damage human cells and tissue. Only recently it has been shown that S. aureus can enter non-professional phagocytes, using adhesins like the fibronectin-binding proteins which mediate an endocytotic uptake into the host cells. The bacteria are consequently localized to endosomes, where the degradation of enclosed bacterial cells through phagosome maturation would eventually occur.
S. aureus can avoid degradation, and translocate to the cellular cytoplasm, where it can replicate. The ability to cause this so-called phagosomal escape has mainly been attributed to a family of amphiphilic peptides called phenol soluble modulins (PSMs), but as studies have shown, they are not sufficient.
In this work I used a transposon mutant library in combination with automated fluorescence microscopy to screen for genes involved in the phagosomal escape process and intracellular survival of S. aureus. I thereby identified a number of genes, including a non-ribosomal peptide synthetase (NRPS). The NRPS, encoded by the genes ausA and ausB, produces two types of small peptides, phevalin and tyrvalin. Mutations in the ausAB genes lead to a drastic decrease in phagosomal escape rates in epithelial cells, which were readily restored by genetic complementation in trans as well as by supplementation of synthetic phevalin. In leukocytes, phevalin interferes with calcium fluxes and activation of neutrophils and promotes cytotoxicity of intracellular bacteria in both, macrophages and neutrophils. Further ausAB is involved in survival and virulence of the bacterium during mouse lung pneumoniae.
The here presented data demonstrates the contribution of the bacterial cyclic dipeptide phevalin to S. aureus virulence and suggests, that phevalin directly acts on a host cell target to promote cytotoxicity of intracellular bacteria.
High-Resolution X-ray Imaging based on a Liquid-Metal-Jet-Source with and without X-ray Optics

With increasing miniaturization in industry and medical technology, non-destructive testing techniques are an area of everincreasing importance. In this framework, X-ray microscopy offers an efficient tool for the analysis, understanding and quality assurance of microscopic species, in particular as it allows reconstructing three-dimensional data sets of the whole sample’s volumevia computed tomography (CT).
The following thesis describes the conceptualization, design, construction and characterization of a compact laboratory-based X-ray microscope in the hard X-ray regime around 9 keV, corresponding to a wavelength of 0.134 nm. Hereby, the main focus is on the optimization of resolution and contrast at relatively short exposure times. For this, a novel liquid-metal-jet anode source is the basis. Such only recently commercially available X-ray source reaches a higher brightness than other conventional laboratory sources, i.e. the number of emitted photons (X-ray quanta) per area and solid angle is exceptionally high. This is important in order to reach low exposure times. The reason for such high brightness is the usage of the rapidly renewing anode out of liquid metal which enables an effective dissipation of heat, normally limiting the creation of high intensities on a small area.
In order to cover a broad range of different samples, the microscope can be operated in two
modes. In the “micro-CT mode”, small pixels are realized with a crystal-scintillator and an
optical microscope via shadow projection geometry. Therefore, the resolution is limited by the emitted wavelength of the scintillator, as well as the blurring of the screen. However, samples in the millimeter range can be scanned routinely with low exposure times. Additionally, this mode is optimized with respect to in-line phase contrast, where edges of an object are enhanced and thus better visible.
In the second “nano-CT mode”, a higher resolution can be reached via X-ray lenses. However,
their production process is due to the physical properties of the hard X-ray range - namely high absorption and low diffraction - extremely difficult, leading typically to low performances. In combination with a low brightness, this leads to long exposure times and high requirements in terms of stability, which is one of the key problems of laboratory-based X-ray microscopy. With the here-developed setup and the high brightness of its source, structures down to 150 nm are resolved at moderate exposure times (several minutes per image) and nano-CTs can be obtained.
In order to select the appropriate behavior, it is important to choose the right behavior at the right time out of many options. It still remains unclear nowadays how exactly this is managed. To address this question, I expose flies (Drosophila melanogaster) to uncontrollable stress to study their behavior under restrictive circumstances by using the so-called shock box. Exposing animals to uncontrollable stress may have an impact on subsequent behavior and can last for some time. The animal learns that whatever it does, it cannot change the situation and therefore can develop something called learned helplessness. The term was first conceptualized by two American psychologists Maier and Seligman (1967), who discovered this phenomenon while doing experiments with dogs. They found out that dogs which are exposed to inescapable stress, later fail in a learning task (‘shuttle box’).
In this work the walking patterns of three different types of experimental flies, walking in a small dark chamber, were evaluated. Using the triadic design (Seligman and Maier, 1967), flies were either exposed to electric shock randomly (yoked), could turn it off by being active (master) or did not receive punishment at all (control). Master flies were shocked whenever they sat for more than 0.9 seconds. At the same time yoked flies received a shock as well independent of what they were doing, to ensure the same amount of shocks received and to create random punishment pattern for the yoked group. With this so-called no-idleness paradigm flies were conditioned either 10 minutes, which resulted in a short (3 minutes) after-effect, or 20 minutes that turned out to be more stable (10 minutes).
In a second part, the behavior during the 20 minute conditioning and a 10 minutes post-test was described in detail. Female flies of the yoked group developed lower activity levels, longer pauses and walked more slowly than master and control flies during conditioning. In the time after the shocks while still in the box, the yoked flies also reduced the frequency and duration of walking bouts as well as their walking speed. Additionally, they took more time to resume walking after the onset of an electric shock than master flies (escape latency) and turned out to make less pauses lasting between 1-1.5 seconds which supports the finding concerning the escape latency.
Male flies, tested under the same conditions, showed a slightly weaker after-effect regarding the difference between master and yoked during conditioning and post-test when compared to female flies.
When comparing the 20 minutes conditioning with subsequent 10 minutes test in the heat and the shock box in parallel, one finds the same effect: Flies which do not have control over the shocks, lower their activity, make less but longer pauses and walk more slowly than their respective master flies. Despite the similar effect of heat and shock on the flies, some differences between the devices occurred, which can partly be explained by different humidity conditions as well as by different surfaces within the chambers.
When the control over the shocks is given back to the yoked flies, it takes them about seven minutes to realize it. One could also show that dopamine levels in the brain were reduced in comparison to flies which did not receive shocks. Yoked flies also were impaired in a place learning task (place learning) and their reaction to light (exit from the box towards the light) directly after conditioning.
After characterizing the walking behavior in the chambers, the study deals with the question whether the effects observed in the chambers transfer to different environments.
In free walk they only differed from flies which did not receive electric shocks and no effect of uncontrollability was transferred to courtship behavior. Handling as the cause could be excluded. Since handling could be exclude to be the cause of losing the effect, I assumed that the behavior shown in the boxes are context depend.
Not only were the after-effects of inescapable shock subject of the current research also the impact of the rearing situation on the response to electric shock was investigated in the present study. Flies which grew up in a single-reared situation turned out to be less affected by inescapable stress in both sexes.
In the next part, the first steps to unravel the neuronal underpinning were taken. A mutant – fumin – which is defective in the dopamine re-uptake transporter showed less reaction to inescapable foot shocks, while a mutant for the gene which encodes an adenylate cyclase (rutabaga2080) resulted in a good score during conditioning, but showed no stable after-effect. Downregulating the expression of the adenylate cyclase gene (rutabaga) in different parts of the mushroom bodies showed, that rutabaga is necessary in the α’β’-lobes for expressing the differences between master and yoked flies in the no-idleness paradigm. The study further confirmed previous findings, that rutabaga is needed in operant but not in classical conditioning.
As a result, the study could show that not the stimulus itself causes the state of uncontrollability but the fact that the fly learned that it was not in control of the stimulus. This state turned out to be context and time dependent.
Die Sequenzierungstechnologien entwickeln sich stetig weiter, dies ermöglicht eine zuvor nicht erreichte Ausbeute an experimentellen Daten und auch an Neuentwicklungen von zuvor nicht realisierbaren Experimenten. Zugleich werden spezifische Datenbanken, Algorithmen und Softwareprogramme entwickelt, um die neu entstandenen Daten zu analysieren. Während der Untersuchung bioinformatischer Methoden für die Identifizierung und Klassifizierung somatischer Mutationen in hämatologischen Erkrankungen, zeigte sich eine hohe Vielfalt an alternativen Softwaretools die für die jeweiligen Analyseschritte genutzt werden können. Derzeit existiert noch kein Standard zur effizienten Analyse von Mutationen aus Next-Generation-Sequencing (NGS)-Daten. Die unterschiedlichen Methoden und Pipelines generieren Kandidaten, die zum größten Anteil in allen Ansätzen identifiziert werden können, jedoch werden Software spezifische Kandidaten nicht einheitlich detektiert.
Um eine einheitliche und effiziente Analyse von NGS-Daten durchzuführen war im Rahmen dieser Arbeit die Entwicklung einer benutzerfreundlichen und einheitlichen Pipeline vorgesehen. Hierfür wurden zunächst die essentiellen Analysen wie die Identifizierung der Basen, die Alignierung und die Identifizierung der Mutationen untersucht. Des Weiteren wurden unter Berücksichtigung von Effizienz und Performance diverse verfügbare Softwaretools getestet, ausgewertet und sowohl mögliche Verbesserungen als auch Erleichterungen der bisherigen Analysen vorgestellt und diskutiert. Durch Mitwirken in Konsortien wie der klinischen Forschergruppe 216 (KFO 216) und International Cancer Genome Consortium (ICGC) oder auch bei Haus-internen Projekten wurden Datensätze zu den Entitäten Multiples Myelom (MM), Burkitt Lymphom (BL) und Follikuläres Lymphom (FL) erstellt und analysiert. Die Selektion geeigneter Softwaretools und die Generierung der Pipeline basieren auf komparativen Analysen dieser Daten, sowie auf geteilte Ergebnisse und Erfahrungen in der Literatur und auch in Foren. Durch die gezielte Entwicklung von Skripten konnten biologische und klinische Fragestellungen bearbeitet werden. Hierzu zählten eine einheitliche Annotation der Gennamen, sowie die Erstellung von Genmutations-Heatmaps mit nicht Variant-Calling-File (VCF)-Syntax konformen Dateien. Des Weiteren konnten nicht abgedeckte Regionen des Genoms in den NGS-Daten identifiziert und analysiert werden. Neue Projekte zur detaillierten Untersuchung der Verteilung von wiederkehrender Mutationen und Funktionsassays zu einzelnen Mutationskandidaten konnten basierend auf den Ergebnissen initiiert werden.
Durch eigens erstellte Python-Skripte konnte somit die Funktionalität der Pipeline erweitert werden und zu wichtigen Erkenntnissen bei der biologischen Interpretation der Sequenzierungsdaten führen, wie beispielsweise zu der Detektion von drei neuen molekularen Subgruppen im MM. Die Erweiterungen, der in dieser Arbeit entwickelten Pipeline verbesserte somit die Effizienz der Analyse und die Vergleichbarkeit unserer Daten. Des Weiteren konnte durch die Erstellung eines eigenen Skripts die Analyse von unbeachteten Regionen in den NGS-Daten erfolgen.
Ernährungsprobleme haben eine hohe Prävalenz auf Palliativstationen und ihre Bedeutung für die Lebensqualität ist nicht zu unterschätzen. Die Behandlung wird im Rahmen der frühen palliativmedizinischen Betreuung wichtiger. In der vorliegenden Arbeit wurden verschiedene Screeningtools auf ihre Leistungsfähigkeit im Setting der Palliativstation überprüft, sowie der Bedarf und die Auswirkungen ernährungstherapeutischer Interventionen evaluiert. Im Zeitraum Mai 2011 bis Juli 2012 wurden 125 stationäre Patienten in die Studie eingeschlossen (46,9 % der in diesem Zeitraum stationär behandelten Patienten). Ernährungsstatus, Ernährungsprobleme und subjektive Bewertung wurden bei Aufnahme und Entlassung erfragt. Das Mangelernährungsrisiko wurde bei Aufnahme mit drei etablierten Screeningbögen vergleichend erfasst (NRS 2002, AKE und SGA) und abschließend bewertet.
64 % der Patienten gaben bei der stationären Aufnahme Ernährungs- und Verdauungsprobleme an. Ein Risiko für eine Mangelernährung lag bei ca. 74 % der Patienten vor, im Wesentlichen übereinstimmend nach AKE, SGA und NRS 2002 (73 %, 75 %, 74 %). Gemäß Arzteinschätzung nach klinischer Erstuntersuchung zeigten 72 % der Patienten einen Bedarf an ernährungstherapeutischen Interventionen. Nach dem AKE waren 58 % der Patienten manifest mangelernährt, nach DGEM-Kriterien 60 %.
Von 94 Patienten liegen Erst- und Zweitbefragung vor Entlassung vor (75,2 %). Im Vergleich zur Aufnahme zeigte sich eine signifikante Minderung der Appetitlosigkeit und ein Zugewinn an Genuss bei den nach AKE primär mangelernährten und bei den Patienten mit einer oralen Nahrungsaufnahme von weniger als 50 % im Vergleich zur Nahrungsaufnahme vor Erkrankungsbeginn. Im prä-post-Vergleich verbesserte sich die Lebensqualität signifikant.
Zusammenfassend ist die Inzidenz von NNK bei Patienten mit einer chronischen
Nebennierenrindeninsuffizienz auch bei geschulten Patienten hoch. Das Auftreten der NNK
ist zudem mit einer erheblichen Morbidität und Mortalität verknüpft. In der heutigen Zeit
sterben weiterhin Patienten an den Folgen ihrer chronischen NNRI trotz einer adäquaten
Behandlung. Legt man die oben genannten Zahlen zugrunde, werden in den nächsten zehn
Jahren zwischen 5526 und 10647 Patienten an einer behandelbaren NNK versterben. Dies
macht die Notwendigkeit, die genauen Umstände und Ursachen einer NNK zu verstehen,
noch wichtiger. Die Analysen der Risikofaktoren einer NNK haben jedoch nur ein begrenztes
Potential, Patienten mit einem besonders hohen Risiko für das Auftreten von NKK zu
identifizieren. Patientenaufklärungen im Hinblick auf Dosisanpassungen der Glukokortikoide
in Stresssituationen werden die intravenösen Gaben von Glukokortikoiden nicht unnötig
machen, um eine drohende NNK zu verhindern. Anstrengungen um eine einheitliche
Patientenaufklärung mit dem Training der Selbstbehandlung mit parenteralem Hydrocortison
werden essentiell sein, um die NNK als Todesursache zu verhindern. Die aktuelle Studie
zeigt, dass die bisherigen Anstrengungen, eine einheitliche und breite Informationsgrundlage
zu vermitteln, nicht ausreichend sind. Denn auch bei medizinischem Fachpersonal besteht
anscheinend nach wie vor die Notwendigkeit einer intensiveren Schulung und Aufklärung. In
mehreren telefonischen Kontakten berichteten Patienten, dass sich ärztliche Kollegen nur
zurückhaltend auf eine ausreichend hohe intravenöse Cortisongabe einlassen konnten, in
einem Fall sogar, obwohl der betroffene Patient einen Notfallausweis bei sich trug. Auch in
der Laienpresse wurde von einem Fall berichtet, in dem laut Aussage der Patientin eine
Cortisonanpassung nicht ausreichend erfolgte.[40] Da die meisten tätigen Ärzte nur sehr
selten mit dem Krankheitsbild einer NNK konfrontiert sind, reagieren diese rezidivierend
nicht adäquat.[8] Auch zeigen sich die Symptome einer NNK häufig sehr unspezifisch,
weshalb es vielen Ärzten schwer fallen mag, diese zu erkennen. Da eine frühzeitige
Intervention für ein gutes Out Come jedoch unverzichtbar ist, sollten weitere Anstrengungen
gemacht werden, auch ärztliche Kollegen über diese Erkrankung und deren Behandlung
aufzuklären. Weiterhin zeigt sich auch die Schulung von Familie und Freunden als sinnvoll,
um das Aktionspotential im Falle einer akuten Verschlechterung der
Nebennierenrindenerkrankung zu verstärken.
423 Fragebögen waren für die Ersterhebung verfügbar, insgesamt schlossen 364 Patienten
(84%) das gesamte follow up über 2 Jahre ab. 767,5 Patientenjahre konnten über die Studie
erfasst werden. Innerhalb der Erhebungszeit wurde von 64 NNK berichtet. Dies entspricht
einer Häufigkeit von 8,3 Krisen/100 Patientenjahre. Als Hauptauslösefaktoren konnten
gastrointestinale Infektionen, Fieber und emotionaler Stress erkannt werden. Die Häufigkeit
entsprach 20%. Jedoch auch andere Stressoren konnten identifiziert werden. Hier zeigten sich
zum Beispiel Operationen, starke Schmerzen, Hitze, anstrengende körperliche Betätigung und
Schwangerschaft als ursächlich. In 7% der Fälle konnte bei einer plötzlichen
Verschlechterung des Zustandes keine auslösende Ursache gefunden werden. Es fand sich
jedoch, dass Patienten, die in ihrer Anamnese bereits eine oder mehrere NNK erlitten hatten,
ein höheres Risiko für eine erneute Entgleisung auswiesen. (Odds ratio 2,85, 95%
Konfidenzinterval 1.5–5.5, p 0,01) Andere Risikofaktoren konnten in der aktuellen Studie
nicht identifiziert werden. Während des Erhebungszeitraumes von zwei Jahren verstarben
insgesamt zehn Patienten. Vier dieser Todesfälle konnte einer NNK als Todesursache
zugeordnet werden. Dies entspricht einer Mortalitätshäufigkeit von 0,5/100 Patientenjahre.
Somit war in unserer Studie die NNK-assoziierte Mortalität 6% der NNK.
Unsere Studienteilnehmer wurden zu Beginn des Follow ups detailliert über die
Notwendigkeit von Dosisanpassungen und der Inanspruchnahme von professionellen Helfern
aufgeklärt. Dennoch zeigte sich im Vergleich zu anderen Studien mit 8,3 NNK/100
Patientenjahre keine reduzierte NNK-Häufigkeit. Jedoch konnte durchaus eine Reduktion der
NNK-Häufigkeit im Vergleich zu den Daten im Ersterhebungsbogen gezeigt werden. Neben
der Notwendigkeit, die Patientenaufklärung zu verbessern, zeigte die vorliegende Studie
jedoch auch, dass weitere Anstrengungen gemacht werden müssen, um das Vorgehen vor und
bei einer NNK weiterhin zu optimieren. So sind die genauen Umstände, die zu einer NNK
führen, bis heute noch nicht detailliert geklärt. Zwar konnten einige Risikofaktoren und
auslösende Situationen identifiziert werden, jedoch nicht die Frage, warum einige Patienten
eine NNK bekommen und Andere nicht. In diesem Zusammenhang besteht auch weiterhin die
Frage, warum einige Patienten gut auf eine Erhöhung der oralen Cortisondosis reagieren und
Andere trotz der Erhöhung in eine NNK kommen. Hieraus ergibt sich die Frage, ob es
Patienten gibt, die sensitiver auf das Cortison reagieren, und wenn dies der Fall ist, warum.
Hierfür könnte sprechen, dass es Patienten gibt, die rezidivierend in eine NNK kommen,
während Andere nie ein Krise erleiden. Auch wird es weiterhin Anstrengungen und neue
Strategien brauchen, eine schnellstmögliche Intervention im Falle einer Verschlechterung des
Allgemeinzustandes zu gewährleisten. Hier sind bessere, flächendeckende Aufklärungen für
medizinisches Fachpersonal und/oder die Verbesserung der mitgeführten Notfallkarten
notwendig. Eine Strategie, die Interventionszeit zu verkürzen, wäre der Ausbau der
Selbstinjektionen durch den betroffenen Patienten oder dessen Angehörige.
In der Dissertation wird vornehmlich die Perspektive des Römischen Staates eingenommen, aus der heraus er zu Maßnahmen gegen die Institution der Kirche, deren Amtsträger und Anhänger aus dem Laienstand schritt. Es wird zu zeigen versucht, welche (religions-)politischen Überlegungen und Hintergründe die antichristliche Agenda der einzelnen Verfolgerkaiser hervorriefen und v.a. inwiefern die einmal gemachten Erfahrungen das Vorgehen der späteren Verfolger beeinflussten, so dass eine sich steigernde, auf einem "Vorwissen" um die Beschaffenheit des "Gegners" aufbauende Logik der Verfolgungsmaßnahmen erkennbar wird.
Der Schwerpunkt der Arbeit liegt auf den Christenverfolgungen von Kaiser Decius bis Maximinus Daia, wobei ein umfangreicheres Kapitel über die Gepflogenheiten der römischen Religion und des Kaiserkultes vorausgeschickt wird.
Computer systems have replaced human work-force in many parts of everyday life, but there still exists a large number of tasks that cannot be automated, yet. This also includes tasks, which we consider to be rather simple like the categorization of image content or subjective ratings. Traditionally, these tasks have been completed by designated employees or outsourced to specialized companies. However, recently the crowdsourcing paradigm is more and more applied to complete such human-labor intensive tasks. Crowdsourcing aims at leveraging the huge number of Internet users all around the globe, which form a potentially highly available, low-cost, and easy accessible work-force.
To enable the distribution of work on a global scale, new web-based services emerged, so called crowdsourcing platforms, that act as mediator between employers posting tasks and workers completing tasks. However, the crowdsourcing approach, especially the large anonymous worker crowd, results in two types of challenges. On the one hand, there are technical challenges like the dimensioning of crowdsourcing platform infrastructure or the interconnection of crowdsourcing platforms and machine clouds to build hybrid services. On the other hand, there are conceptual challenges like identifying reliable workers or migrating traditional off-line work to the crowdsourcing environment. To tackle these challenges, this monograph analyzes and models current crowdsourcing systems to optimize crowdsourcing workflows and the underlying infrastructure. First, a categorization of crowdsourcing tasks and platforms is developed to derive generalizable properties. Based on this categorization and an exemplary analysis of a commercial crowdsourcing platform, models for different aspects of crowdsourcing platforms and crowdsourcing mechanisms are developed. A special focus is put on quality assurance mechanisms for crowdsourcing tasks, where the models are used to assess the suitability and costs of existing approaches for different types of tasks. Further, a novel quality assurance mechanism solely based on user-interactions is proposed and its feasibility is shown. The findings from the analysis of existing platforms, the derived models, and the developed quality assurance mechanisms are finally used to derive best practices for two crowdsourcing use-cases, crowdsourcing-based network measurements and crowdsourcing-based subjective user studies. These two exemplary use-cases cover aspects typical for a large range of crowdsourcing tasks and illustrated the potential benefits, but also resulting challenges when using crowdsourcing.
With the ongoing digitalization and globalization of the labor markets, the crowdsourcing paradigm is expected to gain even more importance in the next years. This is already evident in the currently new emerging fields of crowdsourcing, like enterprise crowdsourcing or mobile crowdsourcing. The models developed in the monograph enable platform providers to optimize their current systems and employers to optimize their workflows to increase their commercial success. Moreover, the results help to improve the general understanding of crowdsourcing systems, a key for identifying necessary adaptions and future improvements.
The key hypothesis of this work represented the question, if mimicking the zonal composition and structural porosity of musculoskeletal tissues influences invading cells positively and leads to advantageous results for tissue engineering. Conventional approaches in tissue engineering are limited in producing monolithic “scaffolds” that provide locally variating biological key signals and pore architectures, imitating the alignment of collagenous fibres in bone and cartilage tissues, respectively. In order to fill this gap in available tissue engineering strategies, a new fabrication technique was evolved for the production of scaffolds to validate the hypothesis.
Therefore, a new solidification based platform procedure was developed. This process comprises the directional solidification of multiple flowable precursors that are “cryostructured” to prepare a controlled anisotropic pore structure. Porous scaffolds are attained through ice crystal removal by lyophilisation. Optionally, electrostatic spinning of polymers may be applied to provide an external mesh on top or around the scaffolds. A consolidation step generates monolithic matrices from multi zonal structures. To serve as matrix for tissue engineering approaches or direct implantation as medical device, the scaffold is sterilized.
An Adjustable Cryostructuring Device (ACD) was successively developed; individual parts were conceptualized by computer aided design (CAD) and assembled. During optimisation, a significant performance improvement of the ACDs accessible external temperature gradient was achieved, from (1.3 ± 0.1) K/mm to (9.0 ± 0.1) K/mm. Additionally, four different configurations of the device were made available that enabled the directional solidification of collagenous precursors in a highly controlled manner with various sample sizes and shapes.
By using alginate as a model substance the process was systematically evaluated. Cryostructuring diagraphs were analysed yielding solidification parameters, which were associated to pore sizes and alignments that were determined by image processing. Thereby, a precise control over pore size and alignment through electrical regulation of the ACD could be demonstrated.
To obtain tissue mimetic scaffolds for the musculoskeletal system, collagens and calcium phosphates had to be prepared to serve as raw materials. Extraction and purification protocols were established to generate collagen I and collagen II, while the calcium phosphates brushite and hydroxyapatite were produced by precipitation reactions.
Besides the successive augmentation of the ACD also an optimization of the processing steps was crucial. Firstly, the concentrations and the individual behaviour of respective precursor components had to be screened. Together with the insights gained by videographic examination of solidifying collagen solutions, essential knowledge was gained that facilitated the production of more complex scaffolds. Phenomena of ice crystal growth during cryostructuring were discussed. By evolutionary steps, a cryostructuring of multi-layered precursors with consecutive anisotropic pores could be achieved and successfully transferred from alginate to collagenous precursors. Finally, very smooth interfaces that were hardly detectable by scanning electron microscopy (SEM) could be attained. For the used collagenous systems, a dependency relation between adjustable processing parameters and different resulting solidification morphologies was created.
Dehydrothermal-, diisocyanate-, and carbodiimide- based cross linking methods were evaluated, whereby the “zero length” cross linking by carbodiimide was found to be most suitable. Afterwards, a formulation for the cross linking solution was elaborated, which generated favourable outcomes by application inside a reduced pressure apparatus. As a consequence, a pore collapse during wet chemical cross linking could be avoided.
Complex monolithic scaffolds featuring continuous pores were fabricated that mimicked structure and respective composition of different areas of native tissues by the presence of biochemical key stimulants. At first, three types of bone scaffolds were produced from collagen I and hydroxyapatite with appropriate sizes to fit critical sized defects in rat femurs. They either featured an isotropic or anisotropic porosity and partly also contained glycosaminoglycans (GAGs). Furthermore, meniscus scaffolds were prepared by processing two precursors with biomimetic contents of collagen I, collagen II and GAGs. Here, the pore structures were created under boundary conditions, which allowed an ice crystal growth that was nearly orthogonal to the external temperature gradient. Thereby, the preferential alignment of collagen fibres in the natural meniscus tissue could be mimicked. Those scaffolds owned appropriate sizes for cell culture in well plates or even an authentic meniscus shape and size. Finally, osteochondral scaffolds, sized to either fit well plates or perfusion reactors for cell culture, were fabricated to mimic the composition of subchondral bone and different cartilage zones. Collagen I and the resorbable calcium phosphate brushite were used for the subchondral zone, whereas the cartilage zones were composed out of collagen I, collagen II and tissue mimetic contents of GAGs. The pore structure corresponded to the one that is dominating the volume of natural osteochondral tissue.
Energy dispersive X-ray spectroscopy (EDX) and SEM were used to analyse the composition and pore structure of the individual scaffold zones, respectively. The cross section pore diameters were determined to (65 ± 25) µm, (88 ± 35) µm and(93 ± 42) µm for the anisotropic, the isotropic and GAG containing isotropic bone scaffolds. Furthermore, the meniscus scaffolds showed pore diameters of (93 ± 21) µm in the inner meniscus zone and (248 ± 63) µm inside the outer meniscus zone. Pore sizes of (82 ± 25) µm, (83 ± 29) µm and (85 ± 39) µm were present inside the subchondral, the lower chondral and the upper chondral zone of osteochondral scaffolds. Depending on the fabrication parameters, the respective scaffold zones were also found to feature a specific micro- and nanostructure at their inner surfaces.
Degradation studies were carried out under physiological conditions and resulted in a mean mass loss of (0.52 ± 0.13) %, (1.56 ± 0.10) % and (0.80 ± 0.10) % per day for bone, meniscus and osteochondral scaffolds, respectively. Rheological measurements were used to determine the viscosity changes upon cooling of different precursors. Micro computer tomography (µ-CT) investigations were applied to characterize the 3D microstructure of osteochondral scaffolds. To obtain an osteochondral scaffold with four zones of tissue mimetic microstructure alignment, a poly (D, L-lactide-co-glycolide) mesh was deposited on the upper chondral zone by electrostatic spinning. In case of the bone scaffolds, the retention / release capacity of bone morphogenetic protein 2 (BMP-2) was evaluated by an enzyme linked immunosorbent assay (ELISA). Due to the high presence of attractive BMP binding sites, only less than 0.1 % of the initially loaded cytokine was released. The suitability of combining the cryostructuring process with 3D powder printed calcium phosphate substrates was evaluated with osteochondral scaffolds, but did not appear to yield more preferable results than the non-combined approach.
A new custom build confined compression setup was elaborated together with a suitable evaluation procedure for the mechanical characterisation under physiological conditions. For bone and cartilage scaffolds, apparent elastic moduli of (37.6 ± 6.9) kPa and (3.14 ± 0.85) kPa were measured. A similar behaviour of the scaffolds to natural cartilage and bone tissue was demonstrated in terms of elastic energy storage. Under physiological frequencies, less than 1.0 % and 0.8 % of the exerted energy was lost for bone and cartilage scaffolds, respectively. With average relaxation times of (0.613 ± 0.040) sec and (0.815 ± 0.077) sec, measured for the cartilage and bone scaffolds, they respond four orders of magnitude faster than the native tissues. Additionally, all kinds of produced scaffolds were able to withstand cyclic compression at un-physiological frequencies as high as 20 Hz without a loss in structural integrity.
With the presented new method, scaffolds could be fabricated whose extent in mimicking of native tissues exceeded the one of scaffolds producible by state of the art methods. This allowed a testing of the key hypothesis: The biological evaluation of an anisotropic pore structure in vivo revealed a higher functionality of immigrated cells and led finally to advantageous healing outcomes. Moreover, the mimicking of local compositions in combination with a consecutive anisotropic porosity that approaches native tissue structures could be demonstrated to induce zone specific matrix remodelling in stem cells in vitro. Additionally, clues for a zone specific chondrogenic stem cell differentiation were attained without the supplementation of growth factors.
Thereby, the hypothesis that an increased approximation of the hierarchically compositional and structurally anisotropic properties of musculoskeletal tissues would lead to an improved cellular response and a better healing quality, could be confirmed. With a special focus on cell free in situ tissue engineering approaches, the insights gained within this thesis may be directly transferred to clinical regenerative therapies.
Chemoselective poly(oxazolines) (POx) and poly[(oligo ethylene glycol) acrylates] were synthesized. An initiator was produced for the preparation of poly(oxazoline)s capable of participating in click chemistry reactions which allows the functionalization of the polymer at the α terminus which was confirmed by 1H NMR spectroscopy. The initiator was used for the polymerization of hydrophilic 2 methyl 2 oxazoline (MeOx), whereby chemoselective, alkyne functionalized polymers could be prepared for Cu-catalyzed azide–alkyne cycloaddition. The desired molecular weight could be achieved through the living, ring opening cationic polymerization and was confirmed by 1H NMR, SEC and MALDI ToF measurements. Polymers were terminated with piperidine if no further functionalization was needed, or with an ester derivate for enabling amine attachment in a subsequent step. In addition, polymers were functionalized by termination with NaN3 in order to provide the counterpart to the azide–alkyne reaction. IR spectroscopy was suitable for the azide detection. The coupling of polymers showed the reactivity and could be confirmed by SEC, 1H NMR and IR spectroscopy.
The composition of cysteine functionalized POx was completed by thiol–ene chemistry. Since the commercially available iso 2 propyl 2 oxazoline is not available for the cationic polymerization, 2 butenyl and 2 decenyl 2 oxazoline (ButenOx and DecenOx) were first prepared. The synthesis of both copolymers, based on MeOx could be confirmed by 1H NMR as well as with SEC, whereby narrow distributions with dispersities of 1.06 could be achieved. The cysteine functionalization of the copolymers was enabled by the creation of a thiazolidine component which could be synthesized by acetal and formyl protection of cysteine and subsequent functionalization with a thiol. The component enabled the reaction with a polymer by thiol–ene reaction which was started by the addition of dimethoxyphenyl-acetophenone and was catalyzed by irradiation with UV light. Both copolymers, with a shorter (polymers with BuenOx) and longer (polymers with DecenOx) hydrophobic sidechain could be functionalized. 1H NMR spectroscopic analysis showed a quantitative reaction with the thiazolidine derivate. After deprotection by acidic workup the desired, cysteine functionalized polymer could be isolated. Quantification of cysteine functions was ensured by a modified TNBSA assay, whereby the thiols were first oxidized in order to confirm an independent measurement of amine functions. Both, the TNBSA assay as well as the NMR measurement showed the desired number of cysteine residues.
The cytotoxicity of functionalized polymers with different compositions was tested by a luminescent cell viability assay (LCVA). Both, the amount of cysteine functions (5–10%) in the copolymers as well as the length of the hydrophobic side chain were varied. All polymers did not show cytotoxicity up to concentrations of 10 mg∙mL-1. The cell activity and cell numbers only decreased below 50% and 20% respectively, when copolymers with 5% cysteine and longer sidechains were measured, which was attributed to a contamination of the sample itself. The cooperation partner performed Native Chemical Ligation (NCL) with model peptides and purified the products by HPLC. A sterically non demanding peptide was synthesized, consisting of an aromatic amino acid and four glycine units. The aromatic unit was used for the quantification of the polymer–peptide conjugate in the 1H NMR spectroscopy. A polymer having five cysteine side chains has been fully implemented by NCL to a conjugate of one polymer with five peptides. A sterically more demanding peptide was additionally used and MALDI ToF measurements confirmed the successful conjugation.
Furthermore the cysteine functionalized polymer was used for nanogel synthesis. The thiol of the cysteine function was oxidized in an inverse mini-emulsion by H2O2, resulting in nanogels (~500 nm) which could be confirmed by SEM, AFM, DLS and NTA measurements.
Besides POx, oligo (ethylene glycol)acrylates (OEGA) were polymerized; by copolymerization with the reactive pentafluorophenyl acrylate (PFPA) reactive and amphiphilic polymers were obtained. The synthesis of PFPA could be confirmed spectroscopically by 1H , 19F NMR, and by FT IR. Copolymers were synthesized by RAFT polymerization with narrow dispersities. Functionalization with an amine functionalized thiazolidine led to a hydrophilic cysteine functionalized polymer after acidic deprotection. Apart from this polymer, a thioester functionalization was successfully performed by reaction of the active polymer with a cyclic amine functionalized thioester which does not release a toxic by product (such as the resulting thiol) during NCL and thus features a very high potential to replace former thioester.
The requirements for the impurity profiling of substances for pharmaceutical use have become greater over time. They can be accomplished by the use of modern instrumental analysis techniques, which have been evolved in the last decades. New types of columns with HILIC, mixed-mode and chiral stationary phases are suitable for the separation of all kinds of substances mixtures, that were previously hardly possible with the use of common reversed phase columns. Modern, almost universal detectors like CAD, ELSD and CNLSD can be applied for a sensitive detection of substances without a chromophore. However, in addition to some small individual disadvantages to these methods, the costs are high and applications are still kind of rare. Thus, the introduction of these devices at a broader level has not yet taken place. While this presumably will change over time, there is a need for methods that enable the impurity profiling of challenging substances with widespread analytics devices.
Methionine is a substance with hydrophobic and hydrophilic impurities. With the help of a mixed-mode stationary phase, which is a combination of a reversed phase and a strong cationic exchanger, the separation of all putative impurities was found possible with good sensitivity and selectivity. The method requires apart from the column only standard isocratic HPLC equipment and was successfully validated.
The evaluation of the enantiomeric purity of amino acids is challenging. Two approaches were made. The first method utilizes CE by means of in-capillary derivation with OPA and the subsequent separation with a cyclodextrin. With the use of OPA/NAC and γ-cyclodextrin, a simple and cost-effective method for the indirect enantioseparation of 16 amino acids was developed. With the second approach, racemic amino acids can be analyzed with HPLC and in-needle derivatization. For this, different columns and chiral thiols were evaluated and the chromatographic parameters were optimized. A method with OPA/NIBLC, a pentafluorophenyl column made the enantioseparation of 17 amino acids feasible. A LOQ of the minor enantiomer down to 0.04 % can be achieved with UV spectrophotometric detection. A similar method was developed for impurity profiling of L-amino acids. This can be used alternatively for the amino acid analysis performed by the European Pharmacopoeia.
A simple, robust, precise and accurate method for the evaluation of impurities in glyceryl trinitrate solution was developed and validated. The four impurities of glyceryl trinitrate are separated by means of an acetonitrile-water gradient and the assay for this substance is also possible.
Die Produktionsplanung und -steuerung (PPS) ist für nahezu jedes fertigende Unternehmen – sowohl im Hinblick auf Lagerbestands- und Kostenoptimierung, als auch für eine termintreue Lieferbereitschaft sowie die dadurch bedingte Kundenzufriedenheit – von zentraler Bedeutung und leistet somit einen erheblichen Beitrag für den Erhalt bzw. den Ausbau der Wettbewerbsfähigkeit. Dabei stellen die Interdependenzen der verschiedenen Teilbereiche innerhalb der PPS sowie zwischen den vor- und nachgelagerten Planungsaufgaben eine – im Zuge der zunehmend angestrebten Integration der gesamten Wertschöpfungskette – immer größer werdende Herausforderung dar.
Diese Arbeit widmet sich mit der Planungsaufgabe der Ermittlung kostenminimaler Losgrößen bei simultaner Festlegung der optimalen Produktionsreihenfolge (Economic Lot Scheduling Problem (ELSP) oder Lossequenzproblem) einem zentralen Teilbereich der PPS. Diese Problemstellung ist insbesondere für den Fall einer Serien- und Sortenfertigung von Relevanz, bei dem mehrere, artverwandte Erzeugnisse im Wechsel auf einer Fertigungsanlage mit beschränkter Kapazität bearbeitet werden. Da die Bestimmung der Fertigungslosgrößen und der Produktionsreihenfolge bei der Ermittlung einer überschneidungsfreien Maschinenbelegung unmittelbar miteinander korrelieren, sollte deren Planung zur bestmöglichen Ausnutzung der Kapazitäten und Minimierung der Kosten nicht sukzessiv, sondern weitestgehend simultan erfolgen. Durch diesen Zusammenhang entsteht eine im Allgemeinen nicht triviale und lediglich mittels spezieller Heuristiken adäquat lösbare Planungsaufgabe. Letztere soll in dieser Arbeit um die Möglichkeit des Lossplittings im Sinne einer überlappenden Fertigung (Teil- oder Transportlosbildung) erweitert werden. Dieses logistische Konzept innerhalb der Produktion geht im Allgemeinen sowohl mit einer Verkürzung der Durchlaufzeiten, als auch mit einer Verringerung der Lagerbestände einher.
Vor diesem Hintergrund findet eingangs zunächst eine allgemeine Einordung bzw. Abgrenzung der Aufgaben und des Umfelds der simultanen Losgrößen- und Reihenfolgeplanung im Rahmen der PPS statt. Anschließend werden die prinzipiell unterschiedlichen Ansätze zur Lösung des ELSP, mit ihren jeweils individuellen Annahmen und Eigenschaften dargestellt. Hierbei wird insbesondere auf die chronologische Entwicklung des Basisperiodenansatzes (BPA) in der Literatur eingegangen, da dieser im weiteren Verlauf der Arbeit eine zentrale Rolle einnimmt. Abschließend werden die Zusammenhänge zwischen den strukturell verschiedenen Lösungsansätzen zum ELSP nochmals zusammenfassend erörtert sowie eine Auswertung zu deren relativer Verbreitung in der wissenschaftlichen Literatur präsentiert.
Nach der Erörterung zweier alternativer Lagerhaltungsmodelle zur Berücksichtigung von Lossplitting im Sinne einer überlappenden Fertigung bildet deren Integration in ausgewählte Lösungsansätze zum ELSP den Hauptteil der Arbeit. Hierfür wird zur Identifizierung und Eingrenzung potentiellen Forschungsbedarfs zunächst ein dedizierter Literaturüberblick gegeben, der eine Kategorisierung der bis dato im engeren Sinne relevanten Veröffentlichungen beinhaltet. Die daraus abgeleiteten Forschungsziele bzw. -fragen werden anschließend in fünf Punkten konkretisiert und beinhalten im Kern die Entwicklung von Modellen zur Berücksichtigung des Lossplittings im ELSP. Dabei wird sowohl das Common Cycle Modell (CCM), als auch der Ansatz variierender Losgrößen (TVL) einbezogen, jedoch steht vor allem eine Heuristik nach dem BPA im Fokus der Ausführungen. Des Weiteren werden bestehende Ansätze zur Integration der Teillosbildung im CCM aus einer neuen Perspektive betrachtet und bezüglich eines eventuellen Optimierungspotentials des Lösungswegs analysiert. Zu den neu entwickelten bzw. erweiterten Modellen werden für die Lösungsfindung Algorithmen formuliert und implementiert, die für beide Alternativen der Teillosbildung eine für alle Produkte einheitliche oder sortenindividuelle Transporthäufigkeit erlauben.
Die Evaluation der entwickelten Modelle erfolgt sowohl anhand von ausgewählten Referenzdatensätzen aus der Literatur als auch auf Basis von insgesamt 4000 zufallsgenerierten Parameterkonstellationen. Dabei liegt der Schwerpunkt der Auswertungen auf einer Ergebnisanalyse hinsichtlich der Höhe des Kosteneinsparungspotentials, das durch die Teillosbildung im BPA zum einen gegenüber der „geschlossenen Fertigung“ und zum anderen im Vergleich zu bestehenden Ansätzen mit Lossplitting im CCM erzielbar ist. Die diesbezüglich gewonnenen Erkenntnisse sowie weitere, aus den Resultaten ableitbare Zusammenhänge werden umfassend diskutiert und interpretiert, so dass letztendlich eine Grundlage zur Ableitung von Handlungsempfehlungen gelegt wird. Die Arbeit schließt mit einem Resümee und der kritischen Würdigung der Forschungsziele bzw. -fragen sowie einem Ausblick auf weiteren Forschungsbedarf.
Motilitätsstörungen des Magen-Darm-Traktes können bei kritisch kranken Patienten auf der Intensivstation zu einem lebensbedrohlichen Krankheitsbild führen. Dabei spielen eine Vielzahl von Pathomechanismen eine Rolle, wobei das Interesse dieser Arbeit den Wirkungen des Tag-Nacht-Hormons Melatonin gilt. Da aus anderen Untersuchungen eine protektive Funktion des Melatonins postuliert werden kann, ist sein Einfluss auf die Peristaltik am Meerschweinchendünndarm untersucht worden. Dabei wurde durch kontinuierliche Perfusion eines Dünndarmsegments im Organbad eine gerichtete Peristaltik induziert. Der Schwellendruck, bei dem eine Kontraktionswelle ausgelöst wurde, als Messparameter herangezogen. Durch Zugabe von Melatonin (in den Konzentrationen: 10 pM, 1nM, 0,1µM und 10 µM) in das Organbad konnte kein Einfluss auf dem Schwellendruck nachgewiesen werden. Auch der Melatoninrezeptorantagonist Luzindol führte zu keiner Änderung des Schwellendruckes. Ein signifikanter Anstieg des Schwellendruckes und damit ein inhibitorischer Einfluss auf die Dünndarmperistaltik konnte lediglich durch den partiellen Agonisten 2Phenylmelatonin nachgewiesen werden. Wesentliche Ergebnisse dieser Arbeit zeigen den Einfluss von Melatonin unter Hypoxiebedingungen des Dünndarmes, bei dem Luzindol den inhibitorischen Effekt auf die Darmperistaltik verstärkt. Die Melatoningabe führt zu keiner protektiven Wirkung auf die Darmperistaltik unter Hypoxiebedingungen. Damit ist zu vermuten, dass der protektive Effekt des Melatonins auf die Darmperistaltik nicht durch seine Eigenschaften als Radikalfänger, sondern über Melatoninrezeptoren vermittelt wird. In den Versuchen mit dem Opioid Fentanyl ist eine signifikante Hemmung der Dünndarmperistalik ebenso unter Blockade des Melatoninrezeptorantagonisten Luzindol festzustellen. Bei Versuchen mit Propofol wurde durch Zugabe von Melatonin oder Melatoninrezeptoragonisten eine Verstärkung der Hemmung der Dünndarmmotilität durch Propofol nachgewiesen. In unseren Versuchen bestätigten wir, dass Midazolam eine hemmende Wirkung auf die Dünndarmperistalik hat. Eine vorherige Zugabe von Melatonin hatte dabei keinen Einfluss auf die hemmende Wirkung von Midazolam, wohingegen Luzindol die Hemmwirkung von Midazolam verstärkte. Somit hat das endogene Melatonin möglicherweise einen protektiven Einfluss, der jedoch durch exogene Zugabe von Melatonin nicht verstärkt wird und nicht nachgeahmt werden kann. Insgesamt zeigen die Untersuchungen, dass Melatonin per se keinen gesicherten Einfluss auf die Peristaltik hat, möglicherweise aber in Wechselwirkung mit Anästhetika.
Viele Forschergruppen konzentrieren sich derzeit auf die Entwicklung von neuartigen Technologien, welche den Weg für die kommerzielle Nutzung einer Quantenkommunikation bereiten sollen. Erste Erfolge konnten dabei insbesondere auf dem Gebiet der Quantenschlüsselverteilung erzielt werden. In diesem Bereich nutzt man die Eigenschaft einzelner, ununterscheidbarer Photonen nicht kopiert werden zu können, um eine abhörsichere Übertragung sensibler Daten zu realisieren. Als Lichtquellen dafür eignen sich Halbleiter-Quantenpunkte. Diese Quantenpunkte lassen sich außerdem leicht in komplexe Halbleiter-Mikrostrukturen integrieren und sind somit besonders interessant für die Entwicklung solch fortschrittlicher Technologien, welche für eine abhörischere Kommunikation notwendig sind. Basierend auf diesem Hintergrund wurden in der vorliegenden Arbeit Halbleiter-Quantenpunkte spektroskopisch hinsichtlich ihres Potentials als Quanten-Lichtquelle für die Quantenkommunikation untersucht. Dabei wurden die Quantenpunkte aus InAs/GaAs und InP/GaInP unter anderem in einem speziellen Verfahren deterministisch positioniert und letztendlich in eine photonische Mikrostruktur integriert, welche aus einer Goldscheibe und einem dielektrischen Spiegel besteht. Als Grundcharakterisierungsmittel kam hauptsächlich die Mikrophotolumineszenzspektroskopie zur Bestimmung der Emissionseigenschaften zum Einsatz. Weiterführend wurden Photonen-Korrelationsmessungen zweiter Ordnung durchgeführt, um den Nachweis einer Quanten-Lichtquelle zu erbringen.
Einfluss eines RTA-Prozesses auf die Emissionseigenschaften von InAs/GaAs-Quantenpunkten
Zur Untersuchung des Einflusses eines Rapid-Thermal-Annealing-Prozesses auf die elektronischen Eigenschaften und die Oszillatorstärke selbstorganisierter InAs/GaAs-Quantenpunkte wurden Mikrophotolumineszenzmessungen an verschiedenen Proben im externen Magnetfeld von bis zu 5 T durchgeführt. Die Quantenpunkte wurden dabei in einem besonderen Verfahren gewachsen, bei dem die nominelle Quantenpunkthöhe durch eine bestimmte Bedeckungsschichtdicke vorgegeben wurde. Insgesamt wurden drei Proben mit Schichtdicken von 2 nm, 3 nm und 4 nm hergestellt, die jeweils nachträglich bei Temperaturen von 750° C bis 850° C für fünf Minuten ausgeheilt wurden. Anhand polarisationsaufgelöster Spektroskopie konnten aus den aufgenommenen Quantenpunktspektren die Zeemanaufspaltung und die diamagnetische Verschiebung extrahiert und damit der effektive Landé g-Faktor sowie der diamagnetische Koeffizient bestimmt werden. Die Auswertung der Zeemanaufspaltung zeigte, dass sowohl höhere Ausheiltemperaturen als auch dickere Bedeckungsschichten zu einer drastischen Abnahme der absoluten g-Faktoren sorgen. Dies lässt darauf schließen, dass eine dickere Bedeckungsschicht zu einer stärkeren Interdiffusion der Atome und einer steigenden Ausdehnung der Quantenpunkte für ex-situ Ausheilprozesse führt. Im Gegensatz dazu steigen die diamagnetischen Koeffizienten der Quantenpunkte mit zunehmender Ausheiltemperatur, was auf eine Ausdehnung der Exzitonwellenfunktion hindeutet. Außerdem wurden mittels zeitaufgelöster Mikrophotolumineszenzspektroskopie die Lebensdauern am Quantenpunktensemble bestimmt und eine Abnahme dieser mit steigender Temperatur festgestellt. Sowohl über die Untersuchungen des diamagnetischen Koeffizienten als auch über die Analyse der Lebensdauer konnte schließlich die Oszillatorstärke der Quantenpunkte ermittelt werden. Beide Messverfahren lieferten innerhalb der Fehlergrenzen ähnliche Ergebnisse. Die höchste Oszillatorstärke \(f_{\chi}=34,7\pm 5,2\) konnte für eine Schichtdicke von d = 3 nm und einer Ausheiltemperatur von 850° C über den diamagnetischen Koeffizienten berechnet werden. Im Falle der Bestimmung über die Lebensdauer ergab sich ein maximaler Wert von \(f_{\tau}=25,7\pm 5,7\). Dies entspricht einer deutlichen Steigerung der Oszillatorstärke im Vergleich zu den Referenzproben um einem Faktor größer als zwei. Des Weiteren konnte eine Ausdehnung der Schwerpunktswellenfunktion der Exzitonen um etwa 70% festgestellt werden. Insgesamt betrachtet, lässt sich durch ex-situ Rapid-Thermal-Annealing-Prozesse die Oszillatorstärke nachträglich deutlich erhöhen, wodurch InAs/GaAs-Quantenpunkte noch interessanter für Untersuchungen im Regime der starken Kopplung werden.
Temperatur- und Leistungsabhängigkeit der Emissionseigenschaften positionierter InAs/GaAs Quantenpunkte
Um einen Einblick in den Ablauf des Zerfallsprozesses eines Exzitons in positionierten Quantenpunkten zu bekommen, wurden temperatur- und leistungsabhängige Messungen durchgeführt. Diese Quantenpunkte wurden in einem speziellen Verfahren deterministisch an vorher definierten Stellen gewachsen. Anhand der Temperaturserien konnten dann Rückschlüsse auf die auftretenden Verlustkanäle in einem Quantenpunkt und dessen Emissionseigenschaften gezogen werden. Dabei wurden zwei dominante Prozesse als Ursache für den Intensitätsabfall bei höheren Temperaturen identifiziert. Die Anhebung der Elektronen im Grundzustand in die umgebende Barriere oder in delokalisierte Zustände in der Benetzungsschicht sorgt für die anfängliche Abnahme der Intensität bei niedrigeren Temperaturen. Der starke Abfall bei höheren Temperaturen ist dagegen dem Aufbruch der exzitonischen Bindung und der thermischen Aktivierung der Ladungsträger in das umgebende Substratmaterial geschuldet. Hierbei lassen sich exemplarisch für zwei verschiedene Quantenpunkte die Aktivierungsenergien \(E_{2A}=(102,2\pm 0,4)\) meV und \(E_{2B}=(163,2\pm 1,3)\) meV bestimmen, welche in etwa den Lokalisierungsenergien der Exzitonen in dem jeweiligen Quantenpunkt von 100 meV bzw. 144 meV entsprechen. Weiterhin deckte die Auswertung des Intensitätsprofils der Exzitonemission die Streuung der Exzitonen an akustischen und optischen Phononen als Hauptursache für die Zunahme der Linienbreite auf. Für hohe Temperaturen dominierte die Wechselwirkung mit longitudinalen optischen Phononen den Verlauf und es konnten für das InAs/GaAs Materialsystem typische Phononenenergien von \(E_{LOA}=(30,9\pm 4,8)\) meV und \(E_{LOB}=(32,2\pm 0,8)\) meV bestimmt werden. In abschließenden Messungen der Leistungsabhängigkeit der Linienbreite wurde festgestellt, dass spektrale Diffusion die inhärente Grenze für die Linienbreite bei niedrigen Temperaturen setzt.
Optische Spektroskopie an positionierten InP/GaInP-Quantenpunkten
Weiterhin wurden positionierte InP/GaInP-Quantenpunkte hinsichtlich der Nutzung als Quanten-Lichtquelle optisch spektroskopiert. Zunächst wurden die Emissionseigenschaften der Quantenpunkte in grundlegenden Experimenten analysiert. Leistungs- und polarisationsabhängige Messungen ließen dabei die Vermutung sowohl auf exzitonische als auch biexzitonische Zerfallsprozesse zu. Weiterhin brachten die Untersuchungen der Polarisation einen ungewöhnlich hohen Polarisationsgrad der Quantenpunktemission hervor. Aufgrund von lokalen Ordnungsphänomenen in der umgebenden GaInP-Matrix wurden im Mittel über 66 Quantenpunkte der Grad der Polarisation von Exziton und Biexziton zu \(p_{Mittel}=(93^{+7}_{-9})\)% bestimmt. Des Weiteren wiesen die Quantenpunkte eine sehr hohe Feinstrukturaufspaltung von \(\Delta_{FSS}^{Mittel}=(300\pm 130)\) µeV auf, welche sich nur durch eine stark anisotrope Quantenpunktform erklären lässt. Durch Auto- und Kreuzkorrelationsmessungen zweiter Ordnung wurden dann sowohl der nicht-klassische Einzelphotonencharakter von Exziton und Biexziton als auch erstmalig für diese Strukturen der kaskadierte Zerfall der Biexziton-Exziton-Kaskade demonstriert. Hierbei wurden \(g^{(2)}(0)\)-Werte von bis 0,08 erreicht. Diese Ergebnisse zeigen das Potential von positionierten InP/GaInP-Quantenpunkten als Grundbausteine für Quanten-Lichtquellen, insbesondere in Bezug auf den Einsatz in der Quantenkommunikation.
Realisierung einer Einzelphotonenquelle auf Basis einer Tamm-Plasmonen-Struktur
Nachdem die vorangegangen Untersuchungen die Eignung der positionierten InP/GaInP-Quantenpunkte als Emitter einzelner Photonen demonstrierten, befasst sich dieser Teil nun mit der Integration dieser Quantenpunkte in eine Tamm-Plasmonen-Struktur zur Realisierung einer effizienten Einzelphotonenquelle. Diese Strukturen bestehen aus einem dielektrischen Spiegel aus 30,5 AlGaAs/AlAs-Schichtpaaren und einer einigen Zehn Nanometer dicken Goldschicht, zwischen denen die Quantenpunkte eingebettet sind. Anhand von Messungen an einer planaren Tamm-Plasmonen-Struktur wurde das Bauteil charakterisiert und neben der Exziton- und Biexzitonemission der Zerfall eines Trions beobachtet, was durch Polarisations- und Korrelationsmessungen nachgewiesen wurde. Um eine Verstärkung der Einzelphotonenemission durch die Kopplung der Teilchen an eine lokalisierte Tamm-Plasmonen-Mode demonstrieren zu können, wurde ein Bereich der Probe mit mehreren Goldscheiben von Durchmessern von 3-6 µm abgerastert und die Lichtintensität aufgenommen. Unterhalb der untersuchten Goldscheiben konnte eine signifikante Erhöhung des Lumineszenzsignals festgestellt werden. Eine quantitative Analyse eines einzelnen Quantenpunktes mittels einer Temperaturserie lieferte dabei eine maximale Emissionsrate von \(\eta_{EPQ}^{Max}=(6,95\pm 0,76)\) MHz und damit eine Effizienz von \((6,95\pm 0,76)\)% solch einer Einzelphotonenquelle unter gepulster Anregung bei 82 MHz. Dies entspricht einer deutlichen Verbesserung der Effizienz im Vergleich zu Quantenpunkten im Volumenmaterial und sogar zu denen in einer planaren DBR-Resonatorstruktur. Positionierte InP/GaInP-Quantenpunkte in einer Tamm-Plasmonen-Struktur bilden somit eine vielversprechende Basis für die Realisierung hocheffizienter Einzelphotonenquellen.
Comparative transcriptomics and post-transcriptional regulation in \(Campylobacter\) \(jejuni\)

The transcriptome is defined as the set of all RNA molecules transcribed in a cell. These include protein-coding messenger RNAs (mRNAs) as well as non-coding RNAs, such as ribosomal RNAs (rRNAs), transfer RNAs (tRNAs), and small non-coding RNAs (sRNAs). sRNAs are known to play an important role in regulating gene expression and virulence in pathogens. In this thesis, the transcriptome of the food-borne pathogen Campylobacter jejuni was characterized at single nucleotide resolution by use of next-generation sequencing approaches. The first genome of a C. jejuni strain was published in the year 2000. However, its transcriptome remained uncharacterized at large.
C. jejuni can survive in a variety of ecological niches and hosts. However, how strain-specific transcriptional changes contribute to such adaptation is not known. In this study, the global transcriptome maps of four closely related C. jejuni strains were defined using a differential RNA-seq (dRNA-seq) approach. This analysis also included a novel automated method to annotate the transcriptional start sites (TSS) at a genome-wide scale. Next, the transcriptomes of four strains were simultaneously mapped and compared by the use of a common coordinate system derived from whole-genome alignment, termed as SuperGenome. This approach helped to refine the promoter maps by comparison of TSS within strains. Most of the TSS were found to be conserved among all four strains, but some single-nucleotide-polymorphisms (SNPs) around promoter regions led to strain-specific transcriptional output. Most of these SNPs altered transcription only slightly, but some others led to a complete abrogation of transcription leading to differential molecular phenotypes. These in turn might help the strains to adapt to their specific host or microniche. The transcriptome also unveiled a plethora of sRNAs, some of which were conserved among the four strains while others were strain specific. Furthermore, a Cas9-dependent minimal type-II CRISPR-Cas system with only three Cas genes and multiple promoters to drive the transcription of the CRISPR locus was also characterized in C. jejuni using the dRNA-seq dataset.
Apart from sRNAs, the role of global RNA binding proteins (RBPs) is also unclear in C. jejuni. Aided by the global transcriptome data, the role of RBPs in post-transcriptional regulation of C. jejuni was studied at a global scale. Two of the most widely studied RNA binding proteins in bacteria are Hfq and CsrA. The RNA interactome of the translational regulator CsrA was defined using another global deep-sequencing technique that combines co-immunoprecipitation (coIP) with RNA sequencing (RIP-seq). Using this interactome dataset, the direct targets of this widespread global post-transcriptional regulator were defined, revealing a significant enrichment for mRNAs encoding genes involved in flagella biosynthesis. Unlike Gammaproteobacteria, where sRNAs such as CsrB/C, antagonize CsrA activity, no sRNAs were enriched in the CsrA-coIP in C. jejuni, indicating absence of any sRNA antagonists and novel modes of CsrA activity regulation. Instead, the CsrA regulatory pathway revealed flaA mRNA, encoding the major flagellin, as a dual-function mRNA. flaA mRNA was the main target of CsrA but it also served to antagonize CsrA activity along with the protein antagonist FliW previously identified in the Gram-positive bacterium Bacillus subtilis. Furthermore, this regulatory mRNA was also shown in this thesis to localize to the poles of elongating C. jejuni cells in a translation-dependent manner. It was also shown that this localization is dependent on the CsrA-FliW regulon, which controls the translation of flaA mRNA. The role and mechanism of flaA mRNA localization or mRNA localization in general is not yet clear in bacteria when compared to their eukaryotic counterparts.
Overall, this study provides first insights into riboregulation of the bacterial pathogen C. jejuni. The work presented in this thesis unveils several novel modes of riboregulation in C. jejuni, which could be applicable more generally. Moreover, this study also lays out several unsolved intriguing questions, which may pave the way for interesting studies to come.
Functional and genetic dissection of mechanosensory organs of \(Drosophila\) \(melanogaster\)

In Drosophila larvae and adults, chordotonal organs (chos) are highly versatile mechanosensors
that are essential for proprioception, touch sensation and hearing. Chos share molecular,
anatomical and functional properties with the inner ear hair cells of mammals. These multiple
similarities make chos powerful models for the molecular study of mechanosensation.
In the present study, I have developed a preparation to directly record from the sensory neurons
of larval chos (from the lateral chos or lch5) and managed to correlate defined mechanical inputs
with the corresponding electrical outputs. The findings of this setup are described in several case
studies.
(1) The basal functional lch5 parameters, including the time course of response during continuous
mechanical stimulation and the recovery time between successive bouts of stimulation, was
characterized.
(2) The calcium-independent receptor of α-latrotoxin (dCIRL/Latrophilin), an Adhesion class G
protein-coupled receptor (aGPCR), is identified as a modulator of the mechanical signals
perceived by lch5 neurons. The results indicate that dCIRL/Latrophilin is required for the
perception of external and internal mechanical stimuli and shapes the sensitivity of neuronal
mechanosensation.
(3) By combining this setup with optogenetics, I have confirmed that dCIRL modulates lch5
neuronal activity at the level of their receptor current (sensory encoding) rather than their ability
to generate action potentials.
(4) dCIRL´s structural properties (e.g. ectodomain length) are essential for the mechanosensitive
properties of chordotonal neurons.
(5) The versatility of chos also provides an opportunity to study multimodalities at multiple levels.
In this context, I performed an experiment to directly record neuronal activities at different
temperatures. The results show that both spontaneous and mechanically evoked activity increase
in proportion to temperature, suggesting that dCIRL is not required for thermosensation in chos.
These findings, from the development of an assay of sound/vibration sensation, to neuronal
signal processing, to molecular aspects of mechanosensory transduction, have provided the first
insights into the mechanosensitivity of dCIRL.
In addition to the functional screening of peripheral sensory neurons, another
electrophysiological approach was applied in the central nervous system: dCIRL may impact the
excitability of the motor neurons in the ventral nerve cord (VNC). In the second part of my work,
whole-cell patch clamp recordings of motor neuron somata demonstrated that action potential
firing in the dCirl\(^K\)\(^O\) did not differ from control samples, indicating comparable membrane
excitability.
