Franz von Kutschera über ungegenständliche Erfahrungen und ihre (religions-)philosophische Bedeutung

Zusammenfassung
Der Beitrag diskutiert im Anschluss an Franz von Kutschera die (religions-)philosophische und theologische Bedeutung ungegenständlicher Erfahrungen. Es handelt sich um Erfahrungen, die sich nicht oder nur bedingt als Erfahrungen eines Subjekts von einem Objekt beschreiben lassen. Besonders in den Blick genommen wird die Relevanz ungegenständlicher Erfahrungen sowohl für die Semantik religiöser Rede als auch für die Epistemologie religiöser Überzeugungen. In diesem Zusammenhang wird argumentiert, dass die kognitive Relevanz dieser Erfahrungen davon abhängt, ob sie innerhalb eines weltanschaulichen Paradigmas einen Beitrag zu einem kohärenten und umfassenden Verständnis der uns begegnenden Wirklichkeit leisten.
Abstract
Constructing evidence constitutes a practice to establish the speaker's authority at Prime Minister's Question Time (PMQT), a weekly half-hour session in the British House of Commons. Here the verb see constitutes a resource for both the questioning Leader of the Opposition (LO) and Members of Parliament (MP) as well as for the responding Prime Minister (PM) to claim first-hand perceptual experience. This paper takes an integrated approach, offering a combined analysis of the grammatical formatting, semantics and pragmatics of the verb see in the context of evidential moves at PMQT. It shows how the verb see is functional in referring to the perceptual basis of a claim made and how its grammatical formatting is reflective of the contingencies of the local interactional context. The analysis is grounded in 32 sessions of PMQT (ca. 16 hrs of video-recordings). The results can be summarised as follows: 1) The evidential function of the verb is achieved through its context-specific grammatical formatting and semantics. 2) The reference to the perceptual basis of a claim evoked by see may co-occur with epistemic qualification and evaluative expressions. 3) The formatting of the verb may be indexical of the political relationship between the questioner and the responding PM.
Enteric pathogens often cycle between virulent and saprophytic lifestyles. To endure these frequent changes in nutrient availability and composition bacteria possess an arsenal of regulatory and metabolic genes allowing rapid adaptation and high flexibility. While numerous proteins have been characterized with regard to metabolic control in pathogenic bacteria, small non-coding RNAs have emerged as additional regulators of metabolism. Recent advances in sequencing technology have vastly increased the number of candidate regulatory RNAs and several of them have been found to act at the interface of bacterial metabolism and virulence factor expression. Importantly, studying these riboregulators has not only provided insight into their metabolic control functions but also revealed new mechanisms of post-transcriptional gene control. This review will focus on the recent advances in this area of host-microbe interaction and discuss how regulatory small RNAs may help coordinate metabolism and virulence of enteric pathogens.
Das „Phänomen“ Shopping Center beschäftigt geographische Handelsforscher und Praktiker seit Jahren gleichermaßen. Dies liegt nicht zuletzt an vielfältigen Prozessen der raum-zeitlichen Ausbreitung, welche auf globaler Ebene zunehmende Internationalisierung, auf der regionalen Ebene rücken vermehrt mittelgroße und kleinere Städte in den Fokus der Projektentwickler und auf lokaler Ebene ist eine Verlagerung des Ansiedlungsgeschehens von Stadtrandlagen zu innerstädtischen Zentren zu beobachten.
Auf Akteursseite spielen Projektentwickler und Management eine wichtige Rolle, um Shopping Center zu erfolgreichen Investmentprodukten zu machen, während die räumliche Planung bei der Bewertung von Projektanträgen und bei der Einordnung in städtische Ansiedlungsstrategien und Leitbilder zu berücksichtigen hat, dass Shopping Center eine besondere Raumkonfiguration in den Stadtlandschaften darstellen - mit Auswirkungen auf verschiedene Aspekte der wirtschaftlichen Entwicklungen und des sozialen Lebens. Ihre Verträglichkeit mit bestehenden Einzelhandelsstrukturen, ihre Wirkungen auf die Stadtentwicklung sowie auf Kaufkraftströme mit geeigneten Methoden und unter Berücksichtigung aktueller Prozesse zu untersuchen, ist Anliegen der Forschung und Praxis.
Diesem Anliegen widmet sich der vorliegende Sammelband, wobei der Großteil der Beiträge auf den Vorträgen einer Fachsitzung im Rahmen des Deutschen Geographentages 2013 in Passau basiert.
Autophagy is a central process behind the cellular remodeling that occurs during differentiation of Leishmania, yet the cargo of the protozoan parasite's autophagosome is unknown. We have identified glycosomes, peroxisome-like organelles that uniquely compartmentalize glycolytic and other metabolic enzymes in Leishmania and other kinetoplastid parasitic protozoa, as autophagosome cargo. It has been proposed that the number of glycosomes and their content change during the Leishmania life cycle as a key adaptation to the different environments encountered. Quantification of RFP-SQL-labeled glycosomes showed that promastigotes of L. major possess ~20 glycosomes per cell, whereas amastigotes contain ~10. Glycosome numbers were significantly greater in promastigotes and amastigotes of autophagy-defective L. major Δatg5 mutants, implicating autophagy in glycosome homeostasis and providing a partial explanation for the previously observed growth and virulence defects of these mutants. Use of GFP-ATG8 to label autophagosomes showed glycosomes to be cargo in ~15% of them; glycosome-containing autophagosomes were trafficked to the lysosome for degradation. The number of autophagosomes increased 10-fold during differentiation, yet the percentage of glycosome-containing autophagosomes remained constant. This indicates that increased turnover of glycosomes was due to an overall increase in autophagy, rather than an upregulation of autophagosomes containing this cargo. Mitophagy of the single mitochondrion was not observed in L. major during normal growth or differentiation; however, mitochondrial remnants resulting from stress-induced fragmentation colocalized with autophagosomes and lysosomes, indicating that autophagy is used to recycle these damaged organelles. These data show that autophagy in Leishmania has a central role not only in maintaining cellular homeostasis and recycling damaged organelles but crucially in the adaptation to environmental change through the turnover of glycosomes.
The molecular basis of male infertility is poorly understood, the majority of cases remaining unsolved. The association of aberrant sperm DNA methylation patterns and compromised semen parameters suggests that disturbances in male germline epigenetic reprogramming contribute to this problem. So far there are only few data on the epigenetic heterogeneity of sperm within a given sample and how to select the best sperm for successful infertility treatment. Limiting dilution bisulfite sequencing of small pools of sperm from fertile donors did not reveal significant differences in the occurrence of abnormal methylation imprints between sperm with and without morphological abnormalities. Intracytoplasmic morphologically selected sperm injection was not associated with an improved epigenetic quality, compared to standard intracytoplasmatic sperm injection. Deep bisulfite sequencing (DBS) of 2 imprinted and 2 pluripotency genes in sperm from men attending a fertility center showed that in both samples with normozoospermia and oligoasthenoteratozoospermia (OAT) the vast majority of sperm alleles was normally (de)methylated and the percentage of epimutations (allele methylation errors) was generally low (<1%). However, DBS allowed one to identify and quantify these rare epimutations with high accuracy. Sperm samples not leading to a pregnancy, in particular in the OAT group, had significantly more epimutations in the paternally methylated GTL2 gene than samples leading to a live birth. All 13 normozoospermic and 13 OAT samples leading to a child had <1% GTL2 epimutations, whereas one (7%) of 14 normozoospermic and 7 (50%) of 14 OAT samples without pregnancy displayed 1–14% GTL2 epimutations.
Over the past three decades, China’s fast economic development has induced considerable changes in China’s university and research institution landscape, research financing and academic career incentives. This paper argues that these changes have affected the motivation and the ways in which Chinese scholars engage in international research cooperation. Most recently it has been observed that strong pressures on scholars and scientists – especially at leading academic institutions – to excel in international publications while simultaneously fulfilling their obligation to generate income for their institutions can lead to a dilemma with regard to international research cooperation: Those institutions and scholars most interesting for foreign scholars to cooperate with may be the ones with the least amount of both incentive and time to enter into serious cooperation. This article invites us to reflect on the implications of these changes in the incentive structure for cooperation in social science research on China.
In der Diskussion um eine gegenstandsadäquate Forschungsmethodik für die Pädagogik bzw. Erziehungswissenschaft stellt deren Bedeutung für eine theoriebasierte Verbesserung der Erziehungs- und Bildungspraxis ein wichtiges Thema dar. Da die Medienpädagogik als erziehungswissenschaftliche Teildisziplin zu verstehen ist, gelten für die medienpädagogische Forschung gleichermassen die Fragen nach ihrem Stellenwert für die Weiterentwicklung medienpädagogischer Praxis und für ihre theoretische Fundierung. Vor diesem Hintergrund werden in unserem Beitrag verschiedene erziehungswissenschaftliche Forschungsansätze mit medienpädagogischen Bezügen skizziert, die in besonderer Weise auf eine theoriebasierte Verbesserung der Erziehungs- und Bildungspraxis zielen: die Aktionsforschung, die praxis- und theorieorientierte Entwicklung und Evaluation von Konzepten für pädagogisches Handeln, der Design-Based Research-Ansatz sowie das Konzept der entwicklungsorientierten Bildungsforschung. In einer Zusammenschau werden zehn Merkmale entsprechender Forschungsansätze aufgezeigt und im Hinblick auf eine gestaltungsorientierte medienpädagogische Forschung kommentiert: Gestaltung als Bestandteil des Forschungsprozesses und der Wissensgenerierung, Praxisrelevanz und Praxistauglichkeit, Reflexion von Wertbezügen, Theoriefundierung und Theorieentwicklung, empirische Fundierung und Kontrolle, Forschung als zirkulärer und iterativer Prozess, Zusammenwirken von Praxis und Wissenschaft, Forschung als Beitrag zur Professionalisierung, Verknüpfung unterschiedlicher Forschungsmethoden, Diskussion von Gütekriterien und Qualitätsstandards.
The Staphylococcus aureus two component system (TCS) sae governs expression of numerous virulence factors, including Eap (extracellular adherence protein), which in turn among other functions also mediates invasion of host cells. The sae TCS is encoded by the saePQRS operon, with saeS coding for the sensor histidine kinase (SaeS) and saeR encoding the response regulator (SaeR). The saeRS system is preceded by two additional open reading frames (ORFs), saeP and saeQ, which are predicted to encode a lipoprotein (SaeP) and a membrane protein (SaeQ), respectively. Earlier, we have shown that SDS-containing subinhibitory concentrations of biocides (Perform®) and SDS alone activate sae transcription and increase cellular invasiveness in S. aureus strain Newman. The effect is associated with an amino acid exchange in the N-terminus of SaeS (L18P), specific to strain Newman.
In this work, the role of whether the two additional genes, saePQ coding for the accessory proteins SaeP and SaeQ, respectively, are involved in SDS-mediated saeRS was investigated. It could demonstrated that the lack of the SaeP protein resulted in an increased saeRS transcription without SDS stress in both SaeSL/P variants, while the SDS effect was less pronounced on sae and eap expression compared to the Newman wildtype, suggesting that the SaeP protein represses the sae system. Also, SDS-mediated inductions of sae and eap transcription along with enhanced invasion were found to be dependent on presence of the SaeSP variant in Newman wildtype. On the other hand, the study also shows that the saePQ region of the sae operon is required for fully functional two-component system saeRS under normal growth conditions, but it is not involved in SDS-mediated activation of the saeS signaling and sae-target class I gene, eap.
In the second approach, the study investigates whether SDS-induced sae expression and host cell invasion is common among S. aureus strains not carrying the (L18P) point mutation. To demonstrate this strain Newman, its isogenic saeS mutants, and various S. aureus isolates were analysed for sae, eap expression and cellular invasiveness. Among the strains tested, SDS exposure resulted only in an increase of sae transcription, Eap production and cellular invasiveness in strain Newman wild type and MRSA strain ST239-635/93R, the latter without an increase in Eap. Interestingly, the epidemic community-associated MRSA strain, USA300 LAC showed a biphasic response in sae transcription at different growth stages, which, however, was not accompanied by increased invasiveness. All other clinical isolates investigated displayed a decrease of the parameters tested. While in strain Newman the SDS effect was due to the saeSP allele, this was not the case in strain ST239-635/93R and the biphasic USA300 strains. Also, increased invasiveness of ST239-635/93R was found to be independent of Eap production. Furthermore, to investigate the global effect of SDS on sae target gene expression, strain Newman wild-type and Newman ∆sae were treated with SDS and analyzed for their transcription profiles of sae target genes using microarray assays. We could show that subinhibitory concentrations of SDS upregulate and downregulate gene expression of several signaling pathways involved in biosynthetic, metabolic pathways as well as virulence, host cell adherence, stress reponse and many hypothetical proteins.
In summary, the study sheds light on the role of the upstream region saePQ in SDS-mediated saeRS and eap expression during S. aureus SDS stress. Most importantly, the study also shows that subinhibitory SDS concentrations have pronounced strain-dependent effects on sae transcription and subsequent host cell invasion in S. aureus, with the latter likely to be mediated in some strains by other factors than the known invasin Eap and FnBP proteins. Moreover, there seems to exist more than the saeSP-mediated mechanism for SDS-induced sae transcription in clinical S. aureus isolates. These results help to further understand and clarify virulence and pathogenesis mechanisms and their regulation in S. aureus.
Atherosclerosis is considered a chronic inflammatory disease of the arterial vessel wall which is not only modulated by innate and adaptive immune responses but also by factors of the blood coagulation system.
In general hypercoagulability seems to increase the development and progression of experimental atherosclerosis in mice on an atherogenic background. In addition, the great majority of coagulation proteins including coagulation factor XII (FXII) have been detected in early and advanced human atherosclerotic lesions supporting the cross-link between the coagulation system and atherosclerosis. Moreover, FXII has been detected in close proximity to macrophages, foam cells and smooth muscle cells in these lesions and has been demonstrated to be functionally active in human plaques. Although these data indicate that factor XII may play a role in atherogenesis a direct contribution of FXII to atherogenesis has not been addressed experimentally to date. Furthermore, clinical studies examining the function of FXII in vascular disease have yielded conflicting results.
Hence, in order to investigate the function of coagulation factor XII in atherosclerosis apolipoprotein E and FXII-deficient (F12\(^{-/-}\) apoE\(^{-/-}\)) mice were employed. Compared to F12\(^{+/+}\)apoE\(^{-/-}\) controls, atherosclerotic lesion formation was reduced in F12\(^{-/-}\)apoE\(^{-/-}\) mice, associated with diminished systemic T-cell activation and Th1-cell polarization after 12 weeks of high fat diet. Moreover, a significant decrease in plasma levels of complement factor C5a was evidenced in F12\(^{-/-}\)apoE\(^{-/-}\) mice. Interestingly, C5a increased the production of interleukin-12 (IL-12) in dendritic cells (DCs) and enhanced their capacity to trigger antigen-specific interferon-gamma (IFNγ) production in OTII CD4\(^+\) T cells in vitro. Importantly, a reduction in frequencies of IL-12 expressing splenic DCs from atherosclerotic F12\(^{-/-}\)apoE\(^{-/-}\) versus F12\(^{+/+}\)apoE\(^{-/-}\) mice was observed in vivo, accompanied by a diminished splenic Il12 transcript expression and significantly reduced IL-12 serum levels.
Consequently, these data reveal FXII to play an important role in atherosclerotic lesion formation and to promote DC-induced and systemic IL 12 expression as well as pro-inflammatory T-cell responses likely at least in part via the activation of the complement system.
Was bedeutet eine Änderung der Kontrastmittelaufnahme in niedrig malignen Gliomen bei Kindern?

Bei niedriggradigen Gliomen WHO I° und II° ist das Kontrastmittelverhalten variabel und nicht mit der Prognose assoziiert. Andererseits wurde bei spontanen Regressionen von niedriggradigen Gliomen bei Patienten sowohl mit als auch ohne eine Neurofibromatose Typ I eine Abnahme der Kontrastmittelaufnahme berichtet. Vielleicht deswegen verleitet eine Zunahme des Enhancements oder ein neu aufgetretenes Enhancement nicht selten zur Diagnose einer Tumorprogression. Es stellt sich also die Frage, ob eine Kontrastmittelaufnahme bei LGGs mit einer Größenzunahme somit einem Tumorprogress assoziiert ist.
Es lässt sich eine überdurchschnittliche Assoziation einer Änderung der Kontrastmittelaufnahme mit dem Wachstumsverhalten bei LGGs nachweisen. Allerdings ist das Maß für eine Progression oder Regression weiterhin die Größenänderung des Tumors. Auch eine neue Kontrastmittelaufnahme innerhalb eines Tumors hat keine Bedeutung für das aktuelle Staging. Sie findet sich zwar häufiger bei Wachstum aber auch bei regredienten Tumoren.
Der Myokardinfarkt (MI) gehört nach wie vor zu den führenden Todesursachen weltweit. Eine Minimierung der Infarktgröße, die durch die Dauer der Ischämie bestimmt wird, ist wesentlich für das Überleben und die Lebensqualität des Myokardinfarkt-Patienten. Die Reperfusion stellt aktuell eine zentrale klinische Intervention dar, um den myokardialen Schaden einzugrenzen. Dennoch führt die Reperfusion per se zu zusätzlichem Schaden am Herzen. Somit ist die Erforschung neuer Strategien zur Minimierung des myokardialen Reperfusionsschadens international von Interesse. Die Pathophysiologie des myokardialen Reperfusionsschadens ist vielschichtig und einige Komponenten sind auch heute in ihrer Wirkweise noch nicht vollständig mechanistisch verstanden. Die vorliegende Arbeit untersucht die Rolle von CD4+ T-Zellen und insbesondere deren Subpopulation der regulatorischen T-Zellen im myokardialen Reperfusionsschaden und stellt neue, auf T-Zellen abzielende, Therapien in Ergänzung zur myokardialen Reperfusion vor.
Zunächst wurde eine Infiltration von T-Zellen in das Myokard nach Ischämie-Reperfusion (I/ R) untersucht. Nach der Ischämie-Reperfusion wurden infiltrierende CD4+ T-Zellen als quantitativ führend und aktiviert identifiziert und erwiesen sich in der Infarktgrößenbestimmung als relevante Mediatoren des Reperfusionsschadens. CD25+Foxp3+ regulatorische T-Zellen (Treg) stellen eine Subpopulation von CD4+ T-Zellen mit immunsuppressiven Eigenschaften dar, die schnell und niederschwellig aktiviert werden können und kommen somit als zum Reperfusionsschaden beitragend in Frage. Mit Hilfe des DEREG (DEpletion of REGulatory T cells) -Mausmodells wurde gezeigt, dass regulatorische T-Zellen zum myokardialen Reperfusionsschaden beitragen; Treg-depletierte DEREG-Mäuse waren vor dem Reperfusionsschaden geschützt und zeigten kleinere Infarktgrößen als die Kontrolltiere. Zudem wurde mittels Transferexperimenten gezeigt, dass für den Treg-vermittelten Reperfusionsschaden die Anwesenheit von CD25- konventionellen T-Zellen (Tconv) erforderlich ist. Regulatorische T-Zellen stellen also einen in der vorliegenden Arbeit identifizierten potentiellen Angriffspunkt zur Reduktion des myokardialen Reperfusionsschadens dar.
Anhand von T-Zell-Rezeptor transgenen OT-II Mäusen und MHC (Major Histocompatibility Complex) Klasse II Knockout (KO) Tieren wurde gezeigt, dass Autoantigenerkennung im myokardialen Reperfusionsschaden eine Rolle spielt. Zur vollen T-Zell-Aktivierung notwendig ist neben dem MHC Klasse II-Signalweg und Kostimulatoren auch das Moleküle CD154 (CD40L). Die Gabe eines inhibitorischen anti-CD154-Antikörpers reduzierte die Infarktgröße in Wildtyp-Tieren sigifikant. Der myokardiale Reperfusionsschaden kann neben Zellen der adaptiven Immunität auch durch Neutrophile Granulozyten, Plättchen oder Inflammation des Endothels verstärkt werden. Knockout Mäuse mit einer Defizienz an CD4+ T-Zellen verfügten über eine verbesserte Mikroperfusion. Mechanistisch war nach 24h Reperfusion die absolute Zellzahl an Neutrophilen Granulozyten im CD4 KO im Vergleich zu Wildtyp-Mäusen unverändert; in Endothelzellen war die Regulation bestimmter Gene (VEGFα, TIMP-1 und Eng) nach I/ R im CD4 KO jedoch verändert.
Zusammengefasst zeigt die vorliegende Arbeit eine zentrale Rolle der Antigen-Erkennung durch den T-Zell-Rezeptor zur Aktivierung von CD4+ T-Zellen im myokardialen Reperfusionsschaden. In Anwesenheit von CD4+Foxp3+ T-Zellen ist der Reperfusionsschaden erhöht. Somit können CD4+Foxp3+ T-Zellen potentiell als Ziel für neuartige Therapien des Myokardinfarkts genutzt werden.
Aktive Zonen (AZs) sind hoch spezialisierte, subzelluläre Kompartimente von Neuronen, die der synaptischen Übertragung dienen. Sie enthalten Gerüstproteine wie RIM (Rab3 interacting molecule) sowie elektronendichte Projektionen bestehend aus Bruchpilot bei Drosophila melanogaster oder Bassoon im Säuger, welche Schlüsselkomponenten des Vesikelverkehrs darstellen. Bei der Fliege sind Anzahl und Verteilung von Bruchpilot-Molekülen in AZs relevant für die funktionelle Differenzierung. Ihre Anordnung wird im Abstand von weniger als einem Mikrometer innerhalb einer präsynaptischen Endigung reguliert.
Im Rahmen der vorliegenden Arbeit wurden elektrophysiologische Ableitungen und konfokale sowie höchstauflösende, immunhistochemische Bildgebung mit dem dSTORM (direct Stochastic Optical Reconstruction Microscopy) Verfahren an larvalen, neuromuskulären Synapsen von Drosophila durchgeführt. Dabei wurde das genetische Potenzial des Modellorganismus genutzt, um relevante Proteinfunktionen und -interaktionen zu analysieren.
RIM als zentrale Komponente Aktiver Zonen ist relevant für synaptische Plastizität. Eine als CORD7 (cone-rod dystrophy type 7) bezeichnete Punktmutation (Arginin zu Histidin) innerhalb der 310 Helix der C2A-Domäne von RIM wurde mit erhöhten kognitiven Fähigkeiten einer Patientengruppe in Verbindung gebracht. Weil die Drosophila C2A-Domäne eine hohe Homologie zur Säugerdomäne aufweist, konnte der Einfluss dieser Mutation auf Struktur und Funktion von Synapsen untersucht werden. Es zeigte sich, dass der Aminosäureaustausch der CORD7-Position und des benachbarten Arginin-Restes die synaptische Organisation und Transmission beeinflussen.
In einer Reihe weiterer Experimente wurde das Zusammenspiel von Bruchpilot und Synaptotagmin, dem Calciumsensor der evozierten Transmitterfreisetzung, analysiert. Während AZs ohne Bruchpilot auch ohne Synaptotagmin funktionieren, führt dessen Reduktion zu einer Umverteilung von Bruchpilot-Molekülen innerhalb von AZs und zu dramatischen Änderungen in ihrer Anzahl. Abschließend wurde so ein Beitrag zum Verständnis der molekularen Organisation synaptischer Informationsverarbeitung und Plastizität geleistet, wobei zu klären bleibt, wie die zuverlässige Speicherung von Informationen an AZs erreicht werden kann.
The haloacid dehalogenase (HAD) family of phosphatases is an ancient, ubiquitous group of enzymes, and their emerging role in human health and disease make them attractive targets for detailed analyses.
This thesis comprises the biochemical and structural characterization of chronophin, an HAD-type
phosphatase, which has been shown to act on Ser3-phosphorylated cofiln-1, a key regulator of actin dynamics, and on the Ser/Thr-phosphorylated steroid receptor co-activator 3 (SRC-3). Besides being a specific phosphoprotein phosphatase, chronophin also acts on the small molecule pyridoxal 5'-phosphate (PLP, vitamin B6), implying that chronophin serves as a regulator of a variety important physiological pathways. The analysis of chronophin was performed on different levels, ranging from intrinsic regulatory mechanisms, such as the allosteric regulation via dimerization or the characterization of specificity determinants, to modes of extrinsic modulation, including the association with putative interacting proteins or the generation of chronophin-specific inhibitors.
The association of the previously identified putative chronophin interactors calcium- and integrinbinding protein 1 (CIB1) and calmodulin was investigated using recombinantly expressed and purified proteins. These studies revealed that the interaction of chronophin with CIB1 or calmodulin is mutually exclusive and regulated by calcium. Neither CIB1 nor calmodulin had an effect on the in vitro chronophin phosphatase activity towards PLP or phospho-cofilin-1, but might regulate other functions of this important phosphatase.
The role of chronophin dimerization was studied by generating a constitutively monomeric variant,
which showed reduced PLP hydrolyzing activity. X-ray crystallographic studies revealed that dimerization is essential for the positioning of the substrate specificity loop in chronophin, unraveling a previously unknown mechanism of allosteric regulation through a homophilic interaction. This mechanism potentially applies to other enzymes of the C2a subfamily of HAD-type phosphatases, as all structurally characterized members show a conserved mode of dimerization.
The general determinants of substrate specificity in the C2a subfamily of HAD phosphatases were
investigated by performing domain swapping experiments with chronophin and its paralog AUM and
subsequent biochemical analyses of the hybrid proteins. The X-ray crystallographic structure
determination of the chronophin catalytic domain equipped with the AUM capping domain revealed the first partial structure of AUM. This structural information was then used in subsequent studies that analyzed the divergent substrate specificities of AUM and chronophin in an evolutionary context.
Finally, a set of four chronophin inhibitors were generated based on the structure of PLP and
characterized biochemically, showing moderate inhibitory effects with IC50-values in the micromolar range. These compounds nevertheless constitute valuable tools for future in vitro experiments, such as studies concerning the structure-function relationship of chronophin as a PLP phosphatase. In addition, the crystal structure of one inhibitor bound to chronophin could be solved. These results provide the basis for the further development of competitive chronophin inhibitors with increased specificity and potency.
The number of fungal infections is rising in Germany and worldwide. These infections are mainly caused by the opportunistic fungal pathogen C. albicans, which especially harms immunocompromised people. With increasing numbers of fungal infections, more frequent and longer lasting treatments are necessary and lead to an increase of drug resistances, for example against the clinically applied therapeutic fluconazole. Drug resistance in C. albicans can be mediated by the Multidrug resistance pump 1 (Mdr1), a membrane transporter belonging to the major facilitator family. However, Mdr1-mediated fluconazole drug resistance is caused by the pump’s regulator, the transcription factor Mrr1 (Multidrug resistance regulator 1). It was shown that Mrr1 is hyperactive without stimulation or further activation in resistant strains which is due to so called gain of function mutations in the MRR1 gene.
To understand the mechanism that lays behind this constitutive activity of Mrr1, the transcription factor should be structurally and functionally (in vitro) characterized which could provide a basis for successful drug development to target Mdr1-mediated drug resistance caused by Mrr1. Therefore, the entire 1108 amino acid protein was successfully expressed in Escherichia coli. However, further purification was compromised as the protein tended to form aggregates, unsuitable for crystallization trials or further characterization experiments. Expression trials in the eukaryote Pichia pastoris neither yielded full length nor truncated Mrr1 protein. In order to overcome the aggregation problem, a shortened variant, missing the N-terminal 249 amino acids named Mrr1 ‘250’, was successfully expressed in E. coli and could be purified without aggregation. Similar to the wild type Mrr1 ‘250’, selected gain of function variants were successfully cloned, expressed and purified with varying yields and with varying purity. The Mrr1 `250’ construct contains most of the described regulatory domains of Mrr1. It was used for crystallization and an initial comparative analysis between the wild type protein and the variants. The proposed dimeric form of the transcription factor, necessary for DNA binding, could be verified for both, the wild type and the mutant proteins. Secondary structure analysis by circular dichroism measurements revealed no significant differences in the overall fold of the wild type and variant proteins. In vitro, the gain of function variants seem to be less stable compared to the wild type protein, as they were more prone to degradation. Whether this observation holds true for the full length protein’s stability in vitro and in vivo remains to be determined. The crystallization experiments, performed with the Mrr1 ‘250’ constructs, led to few small needle shaped or cubic crystals, which did not diffract very well and were hardly reproducible. Therefore no structural information of the transcription factor could be gained so far.
Infections with M. tuberculosis, the causative agent of tuberculosis, are the leading cause of mortality among bacterial diseases. Especially long treatment times, an increasing number of resistant strains and the prevalence of for decades persisting bacteria create the necessity for new drugs against this disease. The cholesterol import and metabolism pathways were discovered as promising new targets and interestingly they seem to play an important role for the chronic stage of the tuberculosis infection and for persisting bacteria.
In this thesis, the 3-ketoacyl-CoA thiolase FadA5 from M. tuberculosis was characterized and the potential for specifically targeting this enzyme was investigated. FadA5 catalyzes the last step of the β-oxidation reaction in the side-chain degradation pathway of cholesterol. We solved the three dimensional structure of this enzyme by X-ray crystallography and obtained two different apo structures and three structures in complex with acetyl-CoA, CoA and a hydrolyzed steroid-CoA, which is the natural product of FadA5. Analysis of the FadA5 apo structures revealed a typical thiolase fold as it is common for biosynthetic and degradative enzymes of this class for one of the structures. The second apo structure showed deviations from the typical thiolase fold. All obtained structures show the enzyme as a dimer, which is consistent with the observed dimer formation in solution. Thus the dimer is likely to be the catalytically active form of the enzyme. Besides the characteristic structural fold, the catalytic triad, comprising two cysteines and one histidine, as well as the typical coenzyme A binding site of enzymes belonging to the thiolase class could be identified. The two obtained apo structures differed significantly from each other. One apo structure is in agreement with the characteristic thiolase fold and the well-known dimer interface could be identified in our structure. The same characteristics were observed in all complex structures. In contrast, the second apo structure followed the thiolase fold only partially. One subdomain, spanning 30 amino acids, was in a different orientation. This reorientation was caused by the formation of two disulfide bonds, including the active site cysteines, which rendered the enzyme inactive. The disulfide bonds together with the resulting domain swap still permitted dimer formation, yet with a significantly shifted dimer interface. The comparison of the apo structures together with the preliminary activity analysis performed by our collaborator suggest, that FadA5 can be inactivated by oxidation and reactivated by reduction. If this redox switch is of biological importance requires further evaluation, however, this would be the first reported example of a bacterial thiolase employing redox regulation.
Our obtained complex structures represent different stages of the thiolase reaction cycle. In some complex structures, FadA5 was found to be acetylated at the catalytic cysteine and it was in complex with acetyl-CoA or CoA. These structures, together with the FadA5 structure in complex with a hydrolyzed steroid-CoA, revealed important insights into enzyme dynamics upon ligand binding and release. The steroid-bound structure is as yet a unique example of a thiolase enzyme interacting with a complex ligand. The characterized enzyme was used as platform for modeling studies and for comparison with human thiolases. These studies permitted initial conclusions regarding the specific targetability of FadA5 as a drug target against M. tuberculosis infection, taking the closely related human enzymes into account. Additional analyses led to the proposal of a specific lead compound based on the steroid and ligand interactions within the active site of FadA5.
HIV verursacht eine progressive Zerstörung des Immunsystems und führt zusätzlich durch Veränderungen im ZNS zu neurokognitiven Störungen (HIV-associated neurocognitive disorders, HAND). Die HIV-Infektion geht mit einer Dysfunktion von dopaminergen Signalwegen einher, die sich unter anderem in einer erhöhten Dopamin-Verfügbarkeit im Liquor von Therapie-naiven HIV-Patienten äußert. Der Grund für die Dysregulation der dopaminergen Signalwege in HIV-Patienten ist nicht geklärt. Aufgrund dessen war das Hauptziel dieser Arbeit die Identifizierung des pathogenetischen Mechanismus, der zu einer erhöhten Dopamin-Konzentration im Liquor von HIV-Patienten führt. Die primäre Hypothese war, dass die erhöhte Dopamin-Verfügbarkeit nicht durch das Virus selbst, sondern vielmehr durch die genetische Konstitution der HIV-Patienten hervorgerufen wird. Deshalb wurden Polymorphismen untersucht, die die dopaminerge Neurotransmission beeinflussen. Es wurde vermutet, dass a) verschiedene Genotypen dieser Polymorphismen in nicht-infizierten und HIV-infizierten Personen mit anderen Häufigkeiten auftreten, b) verschiedene Genotypen mit veränderten Dopamin-Verfügbarkeiten assoziiert sind, c) unterschiedliche Genotypen Auswirkungen auf Marker der Progression der HIV-Infektion haben und d) verschiedene Genotypen die Immunaktivierung beeinflussen. Dazu wurden in 190 HIV-infizierten und nicht-infizierten Teilnehmern unterschiedlicher Ethnien die Polymorphismen BDNF Val66Met, COMT Val108/158Met, DAT 3‘-UTR VNTR, DRD2 TaqIα, DRD3 Ser9Gly und DRD4 VNRT mit PCR, ggf. Restriktionsverdau und Agarose-Gelelektrophorese analysiert und die Expression des Dopamin-Transporters mit real time PCR bestimmt. Darüber hinaus wurden zur weiteren klinischen Charakterisierung die Immunmarker MCP-1, sCD14, suPAR und RANTES mit ELISA analysiert, da eine Erhöhung dieser Parameter mit einer beschleunigten HIV-Progression assoziiert ist. Die Bestimmung der T-Zell-Aktivierung (CD3/CD8/CD38/HLA-DR) wurde mit einer durchflusszytometrischen Analyse durchgeführt. In dieser Arbeit haben wir gezeigt, dass HIV-Patienten hochsignifikant häufiger homozygot für das 10-repeat Allel des Dopamin-Transporter-Polymorphismus sind als nicht-infizierte Personen (57,1 % bzw. 26,8 %, p = 0,001, OR = 3,93, 95 % CI 1,72 – 8,96, direkte logistische Regression). HIV-Patienten und nicht-infizierte Personen mit diesem Genotyp weisen eine signifikant höhere Dopamin-Verfügbarkeit im Liquor auf als Personen mit dem 9/10-Genotyp (p = 0,03) und eine signifikant geringere Expression des Dopamin-Transporters auf PBMCs (p = 0,05). Der DAT 10/10-Genotyp ist im Gegensatz zu anderen Genotypen in HIV-Patienten jedoch weder mit unterschiedlichen CD4+-Zellzahlen und Viruslasten noch mit einer veränderten Häufigkeit von HAND verbunden.
Zusätzlich weisen deutsche und südafrikanische nicht-infizierte und HIV-infizierte Personen mit dem DAT 10/10-Genotyp eine signifikant höhere MCP-1-Konzentration im Plasma auf als Personen mit anderen DAT-Genotypen (p = 0,0076). Keiner der Immunmarker ist mit der Dopamin-Verfügbarkeit assoziiert. Dennoch ist die Immunaktivierung in südafrikanischen HIV-Patienten im Vergleich zu nicht-infizierten Südafrikanern signifikant erhöht: HIV-Patienten zeigen im Vergleich zu nicht-infizierten Personen eine stärkere T-Zell-Aktivierung (p = 0,0001), eine erhöhte Plasma-Konzentration von MCP-1 (p = 0,0014), eine gesteigerte sCD14-Konzentration (p = 0,0004) und eine vermehrte suPAR-Konzentration im Plasma (p = 0,006). In der vorliegenden Arbeit konnte kein Nachweis erbracht werden, dass die erhöhte Immunaktivierung in den südafrikanischen HIV-Patienten durch die Koinfektion mit Echinoccocus oder durch genetische Polymorphismen bei Chemokinen hervorgerufen wird. Eine chronisch erhöhte Immunaktivierung stellt eine treibende Kraft für die Virusreplikation dar und kann letztendlich zu einer Erschöpfung des Immunsystems führen.
Der 10/10-Genotyp des DAT VNTR könnte einen Risiko-Faktor für die HIV-Infektion darstellen, da dieser eine erhöhte Dopamin-Verfügbarkeit nach sich zieht. Dopamin aktiviert HIV in chronisch infizierten T-Lymphoblasten und führt zudem zu einer erhöhten Expression und Sezernierung von TNF-α, das wiederum die Expression von HIV induziert. Diese Ergebnisse untermauern den Zusammenhang von Dopamin und HIV. Es ist jedoch nicht völlig geklärt, ob die erhöhte Dopamin-Konzentration ausschließlich durch den Genotyp hervorgerufen oder auch durch die HIV-Infektion begünstigt wird.
Proteine bestehen aus einer spezifischen Sequenz verschiedener Aminosäuren, die ihre charakteristische Funktion bestimmt. Die große Variabilität an Aminosäuresequenzen ermöglichte die Evolution einer nahezu unbegrenzten Anzahl an Proteinen. Meistens nehmen diese Schlüsselpositionen ein, von robusten Baustoffen bis hin zu molekularen Maschinen. Daher kann eine Fehlfunktion gravierende Auswirkungen auf das Leben haben, z.B. Krankheiten wie Alzheimer oder Epilepsi. Um die Funktionen und Fehlfunktionen zu verstehen, ist eine umfassende Kenntnis der Proteinfaltung, der Protein-Protein Assoziation, sowie den Dynamiken innerhalb von Proteinen erforderlich. Diese Vorgänge wurden in dieser Arbeit an drei isolierten Proteindomänen durch die Anwendung der Fluoreszenzlöschmechanismen der H-Dimerbildung und des photoinduzierten Elektronentransfers untersucht.
Der entfaltete Zustand der Bindungsdomäne BBL, das Teil des 2-oxo-acid Dehydrogenasekomplexes ist, wurde unter physiologischen Bedingungen mit Zirkulardichroismus (CD) und einer Kombination aus photoinduziertem Elektronentransfer und Fluoreszenzkorrelationsspektroskopie analysiert. Beide Methoden zeigten übereinstimmend anhand von 20 in BBL einzeln eingefügten konservativen Punktmutationen, dass Seitenketteninteraktionen keine Auswirkungen auf die Sekundärstruktur des denaturierten Zustandes, den Ausgangspunkt der Faltung, haben. Mit Hilfe der Dekonvolation der CD-Spektren wurde zudem gezeigt, dass die Reststruktur im denaturierten Zustand der helikalen Proteindomäne von β-Strängen und β-Kehren dominiert wird, die eine entscheidende Funktion bei der Faltung in den nativen Zustand haben könnten.
Die N-terminale Domäne (NTD), der für die Materialforschung hochinteressanten Spinnen-seidenfaser, ist für die Polymerisation des Spinnenseidenfadens auf den pH-Wechsel von pH 7 auf pH 6 hin verantwortlich. Dieser für die Proteinfunktion wichtige Prozess wurde durch die Einbringung eines extrinsischen Fluoreszenzschalters, basierend auf der H-Dimerbildung, mit der Stopped-Flow-Technik untersucht. Es wurde gezeigt, dass die NTDs

mit einer Rate von 3 x 10^8 M-1 s-1 assoziieren und somit nahezu das Geschwindigkeitslimit der Protein-Protein Assoziation erreicht wird. Zwei geladenen Seitenketten, der D39 und D40, kommt eine entscheidende Funktion in dem Prozess zu, da eine Mutation dieser die Assoziation verhindert. Des Weiteren wurde gezeigt, dass sich die NTD auf eine Erhöhung der Ionenstärke entgegengesetzt zu anderen Proteinen verhält: die Dissoziation wird beschleunigt, die Assoziation nicht beeinflusst. Gleiches Verhalten wurde auf den einzelnen Austausch der übrigen protonierbaren Aminosäureseitenketten hin beobachtet, ausgenommen die Mutation der E119, welche die Dissoziation verlangsamt. Daher scheint der makromolekulare Dipol, der auf Grund der Ladungsverteilung in der NTD entsteht, die Assoziation maßgeblich zu beeinflussen.
Glutamatrezeptoren sind an der schnellen synaptischen Signalweiterleitung im Nervensys-tem von Vertebraten beteiligt. Die Konformationen der Ligandenbindungsdomäne (LBD) haben dabei entscheidende Auswirkungen auf die Funktion des Gesamtrezeptors. Diese wurden mit einer Kombination aus photoinduziertem Elektronentransfer und Fluoreszenzkorrelationsspektroskopie untersucht. Mit dieser Methode wurde ein dynamisches Bild der gebundenen sowie ungebundenen Form der AMPA-spezifischen Glutamatrezeptor 2-LBD gezeigt. Es wurde zudem gezeigt, dass sich die Dynamiken in Abhängigkeit der Bindung von den Agonisten Glutamat und AMPA, dem partiellen Agonisten Kainate oder Cyclothiazid (CTZ), welches eine Dimerisierung der LBDs bewirkt, unterschiedlich verändern. Dies könnte eine Auswirkung auf die Funktion der Rezeptoren haben.
Die Anwendung der Fluoreszenzlöschmechanismen der H-Dimerbildung und des photoinduzierten Elektronentransfers in dieser Arbeit hat gezeigt, dass diese die Möglichkeit bieten, unterschiedlichste Fragestellungen zu beantworten und so Einblicke in dynamische Funktionsweisen von Proteinen eröffnen. Kombiniert mit etablierten Fluoreszenzmethoden ist es so möglich quantitativ Kinetiken auf unterschiedlichen Zeitskalen zu untersuchen.
Die TGF-β-Proteinfamilie umfasst eine Vielzahl von zumeist homodimeren sezernierten Liganden in höheren Tieren, die viele Vorgänge und Entwicklungen im Embryo wie im adulten Lebewesen über absolute oder graduelle Einflussnahme steuern. Die Signalweiterleitung ins Zytoplasma und den Nukleus erfolgt über promisk paarig rekrutierte Typ-I- und Typ-II-Rezeptoren, ehe vorwiegend rezeptorabhängig verschiedene SMAD-Proteine von Typ-I-Kinasen der Rezeptoren aktiviert werden, in den Kern translozieren und die Transkription induzieren. Zu jedem Zeitpunkt dieser Signalweiterleitung kann mittels verschiedener endogener Inhibitoren regulatorisch eingegriffen werden. Dem bisher einzig bekannten membranständigen Pseudorezeptor Bambi (BMP and Activine membrane bound inhibitor) wurde in vorangegangenen Arbeiten inhibitorisches Potential gegenüber dem BMP- und Activin-vermittelten Signalweg über Bindung an distinkte ligandenadressierte Rezeptoren zugeschrieben, wobei die genaue Wirkweise bislang noch vollkommen unklar war.
In der vorliegenden Arbeit wurde initial ein Homologiemodell der extrazellulären Domäne von hBambi anhand der gelösten Kristallstruktur der extrazellulären Domäne von BR1A im gebundenen Zustand (PDB-ID: 1REW) erstellt. Anhand dieses Modells wurde eine Arbeitshypothese entwickelt und es gelang in der Folge, biologisch aktives rekombinantes Protein zum einen aus transfizierten Insektenzellen sowie aus der Renaturierung aus bakteriellen Einschlusskörpern in hinreichender Menge herzustellen und chromatographisch aufzureinigen. Nach einer vergleichenden Qualitätskontrolle beider Exprimate wurden mittels CD-Spektroskopie und analytischer Gelfiltration der Anteil der Sekundärstrukturelemente sowie der Oligomerisierungsgrad erfolgreich bestimmt. In SPR-Bindestudien wurde der Beweis erbracht, dass hBambi-ECD Affinität zu annähernd allen getesteten Liganden der BMP-/GDF-Gruppe, die den SMAD-1/-5-/-8-Signalweg aktivieren, zeigt. Bekannte Typ-I- und Typ-II-Bindungsmutanten von BMP-2 wurden ebenfalls von hBambi-ECD quasi wildtypisch gebunden. Verschiedene Rezeptorektodomänen sowie ActivinA wurden, wie bisher in der Literatur fälschlich angenommen wurde, hingegen nicht gebunden. Die propagierte Homooligomerisierung von Bambi wird überdies nicht über die extrazelluläre Domäne vermittelt. Eingesetzt in Stimulationsversuche mit BMP-responsiven Zellen wurde eine konzentrationsabhängige inhibierende Wirkung von freier hBambi-ECD auf die BMP-2-vermittelte Signalweiterleitung mit unterschiedlichen Nachweismethoden ermittelt, welche die Ergebnisse aus den SPR-Versuchen erfolgreich bestätigten.
In einem weiteren Teil dieser Arbeit wurden verschiedene chimäre Konstrukte aus für Bambi- und BR1A-Domänen kodierenden Sequenzen kloniert, in HEK Ad293-Zellen zusammen mit BMP- und Activin-responsiven Reportergenkonstrukten transient transfiziert und Stimulationsversuche mit BMP-2 und ActivinA durchgeführt. Wildtypisches Bambi zeigte hierbei ein ambivalentes Verhalten in Bezug auf die Regulation des BMP-2-Signals: geringe Mengen wirken agonistisch, höhere Mengen antagonistisch auf die Ausbildung des Reporters. Im Fall von ActivinA zeigte sich hingegen kein antagonistischer Einfluss von Bambi. In den Experimenten mit chimären Varianten erfolgte durch die erhaltenen Daten die Eingrenzung der Bindestelle von hBambi-ECD an BMP-2 auf den Bereich der Typ-I-Bindestelle. Ein direkter Einfluss der intrazellulären Domäne auf den BMP-2-Signalweg wurde ausgeschlossen. Weiterhin konnte gerade in Versuchen mit einem Antikörper gegen BR1A-ECD eine weitere Eigenheit der Bindung von Bambi an den Liganden offenbart werden: so bildet das Konstrukt aus hBambi-ECD und der intrazellulären BR1A-Domäne mit zugehöriger GS-Box und Typ-I-Kinase einen korrekt in den signalaktiven heterohexameren Komplex rekrutierten funktionellen Typ-I-Rezeptor.
Mit den in dieser Arbeit erzielten Ergebnissen, nämlich der gelungenen Erstellung eines Herstellungsprotokolls der ECD, deren erfolgreich identifizierten Bindepartnern sowie der Charakterisierung der Bindung an BMP-2 ist der Grundstein für die Strukturaufklärung von hBambi-ECD gelegt, welche weitere Klarheit in die Funktionalität dieses Modulators der BMP-/GDF-vermittelten Signalweiterleitung bringen wird. Ebenso sind erste das Verständnis der ICD aufklärende Ergebnisse erzielt worden, die das Fundament für weitere Experimente und darauf folgende Kenntnisgewinne darstellen werden.
Due to the rotation of the earth in the solar system all inhabitants of our planet are exposed to regular environmental changes since more than 3.5 billion years. In order to anticipate these predictable changes in the environment, evolutionarily conserved biological rhythms have evolved in most organisms – ranging from ancient cyanobacteria up to human beings – and also at different levels of organization – from single cells up to behavior. These rhythms are endogenously generated by so called circadian clocks in our body and entrained to the 24 h cycle by external timing cues. In multi-cellular organisms the majority of the cells in the body is equipped with such an oscillator. In mammals, the circadian system is structured in a hierarchical fashion: A central pacemaker resides in the bilateral suprachiasmatic nucleus (SCN) of the hypothalamus, while subsidiary peripheral clocks exist in nearly every tissue and organ.
In contrast to the aforementioned recurrent environmental changes most organisms are also exposed to unpredictable changes in the environment. In order to adapt to these sudden alterations the acute activation of the stress response system, involving the hypothalamic-pituitary-adrenal (HPA) axis and the sympathetic nervous system, displays a fundamental survival mechanism. However, if activation of the stress system becomes chronic, devastating somatic and affective disorders might be the consequence.
At first glance, the circadian and the stress system seem to represent two separate bodily control systems that are involved in adaptation to predictable and unpredictable stimuli, respectively. However, both systems are fundamental for survival, and thus, communicate with each other at various levels. Early studies already demonstrated that stressor exposure at different times of the diurnal cycle generates different stress effects, whereupon the type of stressor plays a pivotal role. Moreover, alterations in the SCN and peripheral circadian clocks could be shown following stressor exposure.
In cooperation with various co-workers, I investigated whether the stress responsiveness is modulated by the endogenous clock in a diurnal fashion and whether repeated psychosocial stress impacts the circadian clock depending on the time of day of stressor exposure. Therefore, male C57BL/6 mice were repeatedly exposed to a psychosocial stressor, either at the beginning of the inactive/light phase (SDL mice) or active/dark phase (SDD mice).
Subsequently, different behavioral, physiological/endocrine and immunological/ inflammatory consequences were assessed. It could be shown that the effects of repeated psychosocial stressor exposure strongly depend on the time of day of stressor exposure. The present results demonstrate that repeated daily stressor exposure has a more negative outcome when applied during the active/dark phase compared to the inactive/light phase. Stressor exposure during the active phase resulted in a loss of general activity, decreased interest in an unfamiliar conspecific, a shift towards a more pro-inflammatory body milieu, and rhythm disturbances in plasma hormones, all representing well-accepted hallmarks of depression. In contrast, C57BL/6 mice exposed to the stressor in their inactive phase exhibited minor physiological alterations that might prevent the formation of the maladaptive consequences mentioned above, thus representing beneficial adaptations.
The second focus of this thesis was put on the investigation of the effects of repeated psychosocial stressor exposure at different times of the light-dark cycle on various levels of the circadian system. An increased expression of the PERIOD2 (PER2) protein, which represents an essential core clock component, could be found in the SCN of mice repeatedly exposed to the stressor during their active phase. In consistence with the alterations in the central circadian pacemaker, the daily rhythm of different hormones and the activity rhythm were considerably affected by SDD. Mice exposed to the psychosocial stressor in their active phase showed a shifted, or absent, rhythm of the hormones corticosterone and leptin. Moreover, their activity was found to be phase-delayed, which seems to be attributable to the Period (Per) gene since Per1/Per2 double-mutants still exhibited their normal activity rhythm following 19 days of stressor exposure during the active phase. In contrast, a phase-advance in the peripheral adrenal gland clock could be seen in C57BL/6 mice subjected to the stressor during their inactive phase. This phase-shift might be required for maintaining the normal rhythmicity in hormonal release and activity.
It has previously been suggested that activation of the HPA axis upon stressor exposure at different times of the light-dark cycle is depending on whether the stressor is of physical or psychological nature. Data from the HPA axis analysis now refine previous findings, indicating that psychosocial stressors also modulate HPA axis responses based on the time of day of stressor presentation. The present results demonstrate that HPA axis activity was reduced following repeated stressor exposure during the active phase. It is reasonable to speculate that this reduced basal activity of the stress system represents a failure in HPA axis adjustment, which could contribute to the negative consequences of repeated psychosocial stressor exposure during the dark phase.
Taken together, it can be concluded that the endogenous clock in mice modulates the stress responsiveness in a circadian fashion and that repeated psychosocial stressor exposure affects the biological clock depending on the time of day of stressor presentation. Thereby, stressor exposure during the active phase results in a more negative outcome as compared to stressor experience during the inactive phase. It is assumed that the interaction between the circadian clock and the stress system is a complex issue that might ensure that the endogenous clock does not get out of synchrony in any order.
Vergleich zweier Verfahren zur Rekonstruktion des vorderen Kreuzbandes:
Ligamentum patellea vs. Semitendinosussehnentransplantat.
10-jahres Ergebnisse einer prospektiven, randomisierten klinischen Studie.
Verfahren:
Diese Studie vergleicht die Ergebnisse einer 10-Jahres Nachuntersuchung nach arthroskopisch unterstütztem Ersatz des vorderen Kreuzbandes mittels zweier verschiedener autologer Transplantate. Zwischen November 1998 und November 1999 wurden in der Orthopädischen Universitätsklinik König-Ludwig-Haus in Würzburg 62 Studienteilnehmer (45 Männer, 17 Frauen) mit Ruptur des vorderen Kreuzbandes operiert. Bei 31 Patienten erfolgte die ACL-Ersatzplastik mittels Lig. Patellae Transplantat (BTB-Transplantat), bei der anderen Hälfte mittels Semitendinosussehnentransplantat. Die Gruppenzusammensetzung zeigte vor dem Verletzungszeitpunkt keinen signifikanten Unterschied bezüglich Alter, Geschlecht und Aktivitätsniveau. Alle Patienten erhielten die gleiche postoperative Behandlung mit Don-Joy Schiene (Ext/Flex 0/0/90), Teilbelastung bis 20 kg während der ersten 6 Wochen und externe Rehabilitationsmaßnahmen. Nach über 10 Jahren konnten 47 (24 LP/23 ST) Patienten vollständig klinisch untersucht und bei 9 (5 LP/4 ST) weiteren zumindest standardisierte Fragebögen ausgewertet werden. 2 Studienteilnehmer (beide ST) schieden wegen einer Transplantatruptur, ein weiterer Patient (ebenfalls ST) aufgrund einer postoperativen Kniegelenksinfektion vorzeitig aus der Studie aus.
Ergebnisse:
Die klinische Untersuchung ergab bezüglich der verwendeten Parameter IKDC- und Lysholm-Score, Tegener-Aktivitätsskala, KT-1000-Arthrometer-Messung und radiologischer Kontrolle keine signifikanten Unterschiede zwischen den beiden Gruppen. Auch in den verwendeten Fragebögen mit Evaluation der subjektiven Kniefunktion, möglicher Schmerzen im Kniegelenk, Entnahmestellenmorbidität und der Zufriedenheit mit dem Operationsergebnis konnten keine signifikanten Gruppenunterschiede festgestellt werden.
In der vorliegenden Studie fanden sich somit auch über 10 Jahre nach vorderem Kreuzbandersatz keine relevanten Unterschiede oder Vorteile für eines der beiden untersuchten autologen Sehnentransplantate.
Die autoimmune Thyreoiditis nach Hashimoto stellt aktuell eine der häufigsten Autoimmunerkrankungen eines Organs und die häufigste Ursache der Hypothyreose dar. Die Hashimotothyreoiditis (HT) weist eine hohe Prävalenz und Inzidenz auf. Es existieren Hinweise, dass die Inzidenz der HT aus noch nicht geklärten Gründen gestiegen sein könnte.
Die Kohlenhydrate Fruktose, Laktose und Sorbitol werden in der Lebensmittelproduktion umfassend eingesetzt. Insbesondere die industrielle Verwendung sowie der weltweite Konsum von Fruktose und Laktose unterlagen in den letzten Jahrzehnten einer rasanten Steigerung, obwohl ein hoher Prozentsatz der Bevölkerung zur Malassimilation jener Kohlenhydrate prädisponiert ist.
In einer internistischen Praxis (Praxis Frau Dr. med. I. Heckl, Bad Homburg) zeigte sich, dass HT-Patienten trotz verifizierter Euthyreose vermehrt über gastrointestinale Symptome berichteten. Unter anderem wurden eine bakterielle Fehlbesiedelung des Dünndarmes und eine Zöliakie ausgeschlossen. In der weiteren Abklärung durch die Praxis Dr. I. Heckl wurde eine deutliche Häufung der Malassimilation der Kohlenhydrate Fruktose, Laktose oder Sorbitol unter euthyreoten HT-Patienten ersichtlich. In Abhängigkeit von einer konsequenten Nahrungsumstellung normalisierten sich regelmäßig das Befinden der Patienten sowie die sonographischen, die serologischen und die laborchemischen Marker der HT, sodass man einen ursächlichen Zusammenhang empirisch vermuten konnte.
Im Rahmen einer prospektiven Studie sollte dieser neu beobachtete Zusammenhang zwischen der HT und der Kohlenhydratmalassimilation in der Klinik und Poliklinik für Nuklearmedizin der Universität Würzburg untersucht werden.
In einem unizentrischen Fall-Kontroll-Studiendesign wurden 45 euthyreote HT-Patienten und 38 schilddrüsengesunde Kontrollpersonen auf das Vorliegen einer Kohlenhydratmalassimilation mittels des Wasserstoffatemtests (H2-Atemtest) untersucht. Alle Probanden erhielten einen Fruktose-H2-Atemtest sowie einen Laktose-H2-Atemtest inklusive einer kapillären Blutglukosemessung. Im Falle eines positiven Ergebnisses des Fruktose-H2-Atemtests wurde ein Glukose-H2-Atemtest zum Ausschluss einer bakteriellen Fehlbesiedelung des Dünndarmes durchgeführt. Lieferte der Fruktose-H2-Atemtest ein negatives Ergebnis, so folgte ein H2-Atemtest mit Sorbitol. Das Auftreten gastrointestinaler Symptome während der Testdurchführung wurde dokumentiert. Symptomfragebögen und semiquantitative Ernährungsfragebögen im retrospektiven Design dienten der Erfassung alltäglicher Symptome und Ernährungsgewohnheiten. Blutproben dienten der Messung von Schilddrüsenhormonen, Schilddrüsenautoantikörpern, Gewebstransglutaminase-Antikörpern und Antiparietalzell-Autoantikörpern.
Unter den euthyreoten HT-Patienten konnte ein signifikant häufigeres Auftreten der Fruktose- sowie der Laktosemalassimilation im Vergleich zu den schilddrüsengesunden Kontrollpersonen demonstriert werden. Die Fruktosemalassimilation wurde bei den HT-Patienten mit 48,9% signifikant häufiger als in der Kontrollgruppe nachgewiesen (p=0,035). Im Kontrollgruppenkollektiv hatte eine Fruktosemalassimilation lediglich bei 26,3% der Probanden bestanden. Eine Laktosemalassimilation wurde bei den HT-Patienten mit 42,2% signifikant häufiger als im Kontrollkollektiv diagnostiziert, welches in 21,1% der Fälle eine Laktosemalassimilation aufwies (p=0,04). Insgesamt lag eine Fruktose- und / oder Laktosemalassimilation bei 73,3% der HT-Patienten und bei 42,1% der Kontrollgruppenprobanden vor. Somit vertrugen nur 26,7% der Fallgruppe, jedoch 57,9% der Kontrollgruppe beide Kohlenhydrate (p=0,004). Hinsichtlich der Prävalenz der Sorbitolmalassimilation oder eines positiven Glukose-H2-Atemtestes kam kein signifikanter Unterschied zur Darstellung. Die Auswertung der Ernährungsfragebögen zeigte für beide Kollektive eine vergleichbare durchschnittliche Konsummenge der jeweiligen Kohlenhydrate auf.
Gastrointestinale Symptome waren während des Laktose-H2-Atemtests sowie während des Fruktose-H2-Atemtests jeweils in der Fallgruppe signifikant häufiger anzutreffen als in der Kontrollgruppe. Auch im Hinblick auf das Alltagsleben beschrieben die euthyreoten HT-Patienten signifikant häufiger unter den folgenden Symptomen zu leiden: Weicher Stuhlgang, Oberbauchschmerzen, Meteorismus, laute Darmgeräusche, „Kugelbauch“, Sodbrennen, Schleimauflagerungen des Stuhlgangs, Obstipation, Müdigkeit, postprandiale Kraftlosigkeit, Depressionen, Heißhunger auf Süßes, Migräne, Konzentrationsmangel und eine vermehrte Infektanfälligkeit.
Zur Kausalität des hier erstmals beschriebenen Zusammenhangs existieren mehrere Hypothesen. Die Einteilung der HT-Patienten gemäß ihrer Schilddrüsenautoantikörper-Titer in Subkollektive ergab keinen Hinweis auf einen Einfluss der Aktivität des Autoimmungeschehens auf die Häufigkeit der Kohlenhydratmalassimilation. Es steht zur Diskussion, ob die HT zur Entstehung einer Kohlenhydratmalassimilation führen, oder ob eine vorbestehende Kohlenhydratmalassimilation, im Sinne eines neu identifizierten Risikofaktors, zur Genese einer HT prädisponieren könnte.
In der vorliegenden Studie konnte erstmalig eine signifikante Häufung der Kohlenhydratmalassimilation bei euthyreoten HT-Patienten aufgezeigt werden. Vor dem Hintergrund der weitreichenden lebensmittelindustriellen Verwendung und des hohen Konsums der Kohlenhydrate Fruktose, Laktose und Sorbitol sowie der hohen Prävalenz und Inzidenz der HT, ergibt sich eine hohe Relevanz des hier nachgewiesenen Zusammenhangs. In der differenzialdiagnostischen Abklärung gastrointestinaler Beschwerden bei euthyreoten HT-Patienten nimmt die hier beschriebene Assoziation zwischen der HT und der Kohlenhydratmalassimilation einen besonderen Stellenwert ein. Die Kohlenhydratmalassimilation verkörpert einen neuen, sowohl klinisch, als auch potentiell pathogenetisch relevanten Aspekt der Hashimotothyreoiditis.
Seit den 60-er Jahren beherbergt das Schloss Ditterswind im Landkreis Haßberge erwachsene Menschen mit mehrfachen und schweren Behinderungen und einem hohen Hilfebedarf. Neben den Wohneinheiten für ca. 70 Personen (8 Wohngruppen) stehen auf dem Gelände auch beschäftigungsbezogene Angebote zur Verfügung (Arbeitsvorbereitung, Tagesförderstätte, Seniorentagesstätte). Vor dem Hintergrund der Forderungen der UN-Konvention (2011, Art. 19) und des Paradigmenwechsels in der Behindertenhilfe (vgl. Theunissen 2006) soll die Komplexeinrichtung im Rahmen eines Regionalisierungsprozesses der Rummelsberger Dienste für Menschen mit Behinderung gGmbH bis 2015 in kleinere, gemeindenahe Wohneinheiten u.a. in Zeil am Main und Ebelsbach überführt werden. Die Vorbereitungsphase dieses Prozesses wurde durch die Universität Würzburg wissenschaftlich begleitet.
Der vorliegende Bericht fasst die Untersuchungen und Ergebnisse einer von den Rummelsberger Diensten für Menschen mit Behinderung RDB gGmbH in Auftrag gegebenen und von der "Aktion Mensch" finanzierten Forschung zusammen, mit dem Auftrag einer wissenschaftliche Begleitung des Projekts: „Dezentralisierung und Konversion“ der Komplexeinrichtung Schloss Ditterswind.
Since more than two centuries naturalists are fascinated by the profound changes in biodiversity observed along climatic gradients. Although the theories explaining changes in the diversity and the shape of organisms along climatic gradients belong to the foundations of modern ecology, our picture on the spatial patterns and drivers of biodiversity is far from being complete. Ambiguities in theory and data are common and past work has been strongly concentrated on plants and vertebrates. In the last two decades, interest in the fundamental processes structuring diversity along climatic gradients gained new impetus as they are expected to improve our understanding about how ecosystems will respond to global environmental changes. Global temperatures are rising faster than ever before; natural habitats are transformed into agricultural land and existing land use systems get more and more intensified to meet the demands of growing human populations. The fundamental shifts in the abiotic and biotic environment are proclaimed to affect ecosystems all over the world; however, precise predictions about how ecosystems respond to global changes are still lacking. We investigated diversity, traits and ecosystem services of wild bees along climate and land use gradients on Mount Kilimanjaro (Tanzania, East Africa). Wild bees play a major role in ecosystems, as they contribute to the reproduction and performance of wild and crop plants. Their responsiveness to environmental changes is therefore of high ecological and economic importance.
Temperature and energy resources have often been suggested to be the main determinants of global and local species richness, but the mechanisms behind remain poorly understood. In the study described in chapter II we analyzed species richness patterns of wild bees along climate and land use gradients on Mount Kilimanjaro and disentangled the factors explaining most of the changes in bee richness. We found that floral resources had a weak but significant effect on pollinator abundance, which in turn was positively related to species richness. However, temperature was the strongest predictor of species richness, affecting species richness both directly and indirectly by positively influencing bee abundances. We observed higher levels of bee-flower-interactions at higher temperatures, independently of flower and bee abundances. This suggests that temperature restricts species richness by constraining the exploitation of resources by ectotherms. Current land use did not negatively affect species richness. We conclude that the richness of bees is explained by both temperature and resource availability, whereas temperature plays the dominant role as it limits the access of ectotherms to floral resources and may accelerate ecological and evolutionary processes that drive the maintenance and origination of diversity.
Not only species numbers, but also morphological traits like body size are expected to be shaped by both physiological and energetic constraints along elevational gradients. Paradoxically, Bergmann´s rule predicts increases of body sizes in cooler climates resulting from physiological constraints, while species-energy theory suggests declines in the mean body size of species caused by increased extinction probabilities for large-bodied species in low-energy habitats. In chapter III we confronted this ambiguity with field data by studying community-wide body size variation of wild bees on Mt. Kilimanjaro. We found that along a 3680 m elevational gradient bee individuals became on average larger within species, while large species were increasingly absent from high-elevational communities. This demonstrates, on the one hand, how well-established, but apparently contrasting ecological theories can be merged through the parallel consideration of different levels of biological organization. On the other hand it signals that the extinction risk in the course of environmental change is not equally distributed among species within a community.
Land use intensification is known to threaten biodiversity, but the consequences for ecosystem services are still a matter of debate. In chapter IV, we experimentally tested the single and combined contributions of pest predators and pollinators to coffee production along a land use intensification gradient on Mount Kilimanjaro. We found that pest predation increased fruit set by on average 9%, while pollination increased fruit weight of coffee by on average 7.4%. Land use had no significant effect on both ecosystem services. However, we found that in coffee plantations with most intensified land use, pollination services were virtually exclusively provided by the honey bee (Apis mellifera). The reliance on a single pollinator species is risky, as possible declines of that species may directly lower pollination services, resulting in yield losses. In contrast, pollination services in structurally complex homegardens were found to be provided by a diverse pollinator community, increasing the stability of pollination services in a long term.
We showed that on Mount Kilimanjaro pollinator communities changed along elevational gradients in terms of species richness (chapter II) and trait composition (chapter III). Temperature and the temperature-mediated accessibility of resources were identified as important predictors of these patterns, which contributes to our fundamental understanding about the factors that shape ectothermic insect communities along climatic gradients. The strong temperature-dependence of pollinators suggests that temperature shifts in the course of global change are likely to affect pollinator communities. Pollinators might either profit from rising temperatures, or shift to higher elevations, which could result in related biotic attrition in the lowland with consequences for the provision of ecosystem services in cropping systems. Up to now, land use intensification had no significant impact on the diversity of pollinator communities and their ecosystem services. Pollinators might profit from the strong landscape heterogeneity in the region and from the amount of flower resources in the understory of cropping systems. However,progressing homogenization of the landscape and the pronounced application of pesticides could result in reduced diversity and dominance of single species, as we already found in sun coffee plantations. Such shifts in community compositions could threaten the stability of ecosystem services within cropping and natural systems in a long term.
1. Pollination of sexually reproducing plants requires pollen transfer agents, which can be biotic, abiotic or a combination of biotic and abiotic agents. The dominance of one of pollination system in wild plant communities depends on climatic factors and/or degrees of anthropogenic influences, which have effects on pollinator diversity and pollination function. Anthropogenic activities and climate change are also considered as main causes of ongoing invasion of invasive species into wild and managed habitats which can bring up competition for pollinators with possible negative consequences for the reproduction of co-occurring native plant species.
2. The study aimed to determine pollination systems and pollination limitation of invasive and native plant communities in natural savannah between 870 – 1130 m and semi-natural (managed) grassland between 1300 – 1750 m above sea level; effects of flower density and pollinator abundance on seed production of cross-pollinated and self-pollinated plants; and relationships of bee abundance and the proportion of cross- pollinated plants at the southern slope of Mount Kilimanjaro, Tanzania.
3. Pollinator-exclusion, open pollination and supplemental hand-pollination treatments were applied to 27 plant species in savannah and grassland habitats. Flowers were counted in each clusters based upon their species. Pollinators were sampled by using pan traps. Information-theory-based multi-model averaging and generalized linear mixed effects models were used to identify and analyze the effects of flower density, pollinator abundance, pollination treatments and habitat types on seed production. Regression models were used to determine relationships of altitude with bee abundance, and with proportion of cross-pollinated plants.
4. My results show that mean seed numbers of native plants were significantly lower in pollinator-exclusion treatments than in open-pollination treatments, indicating their reliance on pollinators for reproductive success. In contrast, seed numbers of invasive plants were similar in pollinator-exclusion and open-pollination treatments, demonstrating an ability of reproduction without pollinators. Despite of higher levels of self-pollination in invasive plants, supplemental hand-pollination treatments revealed pollen limitation in grassland and marginally in savannah habitats. There were no significant difference in seed numbers between supplemental hand pollination and open pollination treatments of native plant communities in savannah and grassland, which indicates no pollination limitation in the studied ecological system for native communities. Besides, grassland plants produced comparatively more seeds than savannah plants, however seeds in grasslands were lighter than those of the savannah which may be due to nutrient limitation in grassland.
5. I found 12 cross-pollinated and 15 self-pollinated plants along altitudinal gradient after comparing seeds from pollinator-excluded and open-pollinated experiments. I also found that proportions of cross-pollinated plants and bee abundance simultaneously decreased with increasing altitude. All cross-pollinated plants were native and grew in savannah habitats, with an exception of one species.
6. Neither effects of focal flower density nor a significant interaction between focal flower densities and bee abundance for self-pollinated plants were observed. However, there were effects of focal flower densities and interactions of flower density with bee abundance for cross-pollinated plants. Non-focal flower density has no significant effects on seed production of cross-pollinated and self-pollinated plants.
7. The results show that native plants depend more on cross-pollination than invasive plants, despite of most native plants in managed habitat (grassland) rely on self-pollination for reproduction. The tendency of having more cross-pollinated plants in natural savannah which are in low altitude coincides with other finding that the cross-pollinated plants and bee abundance simultaneously decrease with increasing altitude. Therefore, our findings support the hypotheses that self-fertilization of flowering plants increases with increasing altitude, and pollinator limitation is most pronounced in managed or disturbed habitats. Despite of reduction of pollinators in grassland, only invasive plants experience pollen limitation, which may be due to poor integration with available pollinator networks.
8. I also found bee abundance and flower density are not the main pollination factors required by self-pollinated plants during reproduction. However, focal flower density, which influences pollinator diversity, is more applicable to cross-pollinated plants. Climate change and anthropogenic activities in natural habitats are factors that influence pollinator abundance and functioning, which lead to a shift of mating systems in plant communities so as to assure their reproduction.
Search for the \(X_b\) and other hidden-beauty states in the \(π^+π^−ϒ\)(1S) channel at ATLAS

This Letter presents a search for a hidden-beauty counterpart of the X(3872) in the mass ranges of 10.05–10.31 GeV and 10.40–11.00 GeV, in the channel X\(_b\)→π\(^+\)π\(^−\)ϒ(1S)(→μ\(^+\)μ\(^−\)), using 16.2 fb\(^{−1}\) of s=8 TeV \(pp\) collision data collected by the ATLAS detector at the LHC. No evidence for new narrow states is found, and upper limits are set on the product of the X\(_b\) cross section and branching fraction, relative to those of the ϒ(2S), at the 95% confidence level using the CLSCLS approach. These limits range from 0.8% to 4.0%, depending on mass. For masses above 10.1 GeV, the expected upper limits from this analysis are the most restrictive to date. Searches for production of the ϒ(1\(^3\)D\(_J\)), ϒ(10860), and ϒ(11020) states also reveal no significant signals.
GAS2L3 was identified recently as a target gene of the DREAM complex (Reichert et al., 2010; Wolter et al., 2012). It was shown that GAS2L3 is expressed in a cell cycle specific manner and that depletion of the protein leads to defects in cytokinesis and genomic instability (Wolter et al., 2012).
Major aim of this thesis was, to further characterize the biochemical properties and physiological function of GAS2L3.
By in vitro co-sedimentation and bundling assays, GAS2L3 was identified as a cytoskeleton associated protein which bundles, binds and crosslinks F-actin and MTs. GST pulldown assays and co-immunoprecipitation experiments revealed that GAS2L3 interacts in vitro and in vivo with the chromosomal passenger complex (CPC), a very important regulator of mitosis and cytokinesis, and that the interaction is mediated by the GAR domain of GAS2L3 and the C-terminal part of Borealin and the N-terminal part of Survivin. Kinase assays showed that GAS2L3 is not a substrate of the CPC but is strongly phosphorylated by CDK1 in vitro. Depletion of GAS2L3 by shRNA influenced protein stability and activity of the CPC. However pharmacological studies showed that the decreased CPC activity is not responsible for the observed cytokinesis defects upon GAS2L3 depletion. Immunofluorescence experiments revealed that GAS2L3 is localized to the constriction zone by the CPC in a GAR dependent manner and that the GAR domain is important for proper protein function.
New interacting proteins of GAS2L3 were identified by stable isotope labelling by amino acids in cell culture (SILAC) in combination with tandem affinity purification and subsequent mass spectrometrical analysis. Co-immunoprecipitation experiments further confirmed the obtained mass spectrometrical data.
To address the physiological function of GAS2L3 in vivo, a conditional and a non-conditional knockout mouse strain was established. The non-conditional mouse strain showed a highly increased mortality rate before weaning age probably due to heart failure. The physiological function of GAS2L3 in vivo as well as the exact reason for the observed heart phenotype is not known at the moment.
Volumenregulatorische Transportwege von anorganischen und organischen Osmolyten in Säugetierzellen

Die Aufrechterhaltung des Zellvolumens unter variablen osmotischen Bedingungen stellt für nahezu alle tierischen Zellen eine essenzielle Aufgabe dar. Um regulatorische Volumenanpassungen vorzunehmen besitzen sie daher effektive Mechanismen, mit deren Hilfe der zelluläre Gehalt an organischen und anorganischen Osmolyten erhöht (= regulatorische Volumenzunahme; RVI) oder gesenkt (= regulatorische Volumenabnahme; RVD) werden kann. Trotz langjähriger Forschung auf diesem Gebiet konnten die hieran beteiligten Transportwege für Osmolyte bisher nur unvollständig aufgeklärt werden.
Insbesondere bei T-Lymphozyten sind wichtige Zellfunktionen wie die Proliferation, Migration und die T-Zell-Aktivierung eng mit volumenregulatorischen Mechanismen verbunden. Bei all diesen Prozessen sind u. a. unterschiedliche Kaliumkanäle beteiligt, die insbesondere für die pharmakologische Manipulation von Immunsystemprozessen von wissenschaftlichem Interesse sind. Bisherige Modelle der hypotonen Volumenregulation von T-Lymphozyten berücksichtigen lediglich den spannungsabhängigen KV1.3 sowie den Ca2+-aktivierten IKCa1-Kanal, die zur Klasse der 6TM/P-K+-Kanäle gehören.
Im ersten Teil der vorliegenden Arbeit wurde eine potentielle Rolle von kürzlich entdeckten Zwei-Poren Domänen Kaliumkanälen (K2P) am RVD von murinen und humanen primären CD4+-T-Lymphozyten untersucht. In einem kombinierten genetischen und pharmakologischen Ansatz mittels knockout-Tiermodellen und dem Einsatz kanalspezifischer Inhibitoren konnte mithilfe zellvolumetrischer Analysen gezeigt werden, dass die K2P-Vertreter TASK1, TASK2, TASK3 und TRESK maßgeblich am schwellungsaktivierten Efflux von K+ beteiligt sind. Beurteilt an den Ergebnissen dieser Untersuchung sind der spannungsabhängige TASK2- und der Ca2+-aktivierte TRESK-Kanal für die hypotone Volumenregulation in T-Zellen deutlich bedeutender als TASK1 und TASK3. Der Beitrag der Kanäle TASK2 und TRESK am RVD-Prozess war über dies vergleichbar mit dessen des bisher bekannten KV1.3-Kanals. In dieser Arbeit wurde damit erstmals eine Beteiligung der K2P-Kanäle am RVD muriner und humaner CD4+-Lymphozyten identifiziert. Aufgrund der engen Verbindung zwischen T-Zell-Funktion und der Volumenregulation können Zwei-Poren Domänen K+-Kanäle damit in den engeren Kreis potentieller immunmodulierende Angriffspunkte aufgefasst werden.
Im zweiten und umfangreicheren Teil dieser Arbeit wurden darüber hinaus die schwellungsaktivierten Transportwege für organische Osmolyte (small organic osmolytes; SOOs) untersucht. SOOs stellen chemisch inerte Verbindungen dar, zu denen vor allem Polyole (Sorbitol, myo-Inositol), Methylamine (Betain, α-Glycerophosphocholin) sowie Aminosäuren (α- bzw. β-Alanin und Prolin) und deren Derivate (Taurin) zählen. Da SOOs weder die zelluläre Struktur noch die Funktion von Makromolekülen beeinträchtigen, sind sie wichtige Instrumente der Volumenregulation, die sich in hohen Konzentrationen im Zytosol nahezu aller Zellen wiederfinden. Werden tierische Zellen mit hypotonen Bedingungen konfrontiert, dann ist bei nahezu allen Zellen die Freisetzung organischer Osmolyte zu beobachten, wodurch die zelluläre Osmolarität unabhängig von Elektrolyten angepasst werden kann. Trotz der wichtigen Funktion der SOOs in der Osmoregulation tierischer Zellen konnte die molekulare Identität beteiligter Effluxwege (Kanäle bzw. Transporter) bisher nicht aufgeklärt werden.
Ungeachtet der molekularen Identität der SOO-Effluxwege war es aus zahlreichen biotechnologischen Anwendungen zu Beginn dieser Arbeit bekannt, dass die schwellungsaktivierten Transportwege für organische Osmolyte eine größenselektive Permeabilität für eine Reihe monomerer Zucker und verwandter Verbindungen aufweisen. Um diese Größenselektivität näher zu charakterisieren, wurde im ersten Schritt die schwellungsaktivierte Membranpermeabilität für eine Reihe strukturell homogener Polyethylenglykole unterschiedlicher Polymerlänge (PEG200–1500; hydrodynamische Radien zwischen ~0,5-1,5 nm) unter iso- und hypotonen Bedingungen in Jurkat-Lymphozyten untersucht. Unter milden hypotonen Bedingungen (200 mOsm) war die Plasmamembran der untersuchten Lymphozyten für PEG300-1500 undurchlässig, was aus der Fähigkeit der Zellen zur hypotonen Volumenregulation geschlossen werden konnte. Darüber hinaus wurde RVD in stark hypotonen Lösungen (100 mOsm) mit PEG600-1500 beobachtet, während PEG300-400 unter vergleichbaren osmotischen Bedingungen die Volumenregulation der Zellen inhibierten. Dieses Ergebnis deutet darauf hin, dass starkes hypotones Zellschwellen der Lymphozyten zur Permeabilisierung der Plasmamembran für PEG300-400, nicht jedoch für PEG600-1500, führt. Anhand der hydrodynamischen Radien Rh der verwendeten PEGs konnte ein cutoff-Radius von ~0,74 nm für schwellungsaktivierte Transportwege organischer Osmolyte bestimmt werden. Da diese schwellungsaktivierten Transportwege vielfältig für Zellbeladungstechniken verwendet werden, könnte dieses Ergebnis für zahlreiche biotechnologische und biomedizinische Anwendungen von Interesse sein.
Im zweiten Schritt wurde der Versuch unternommen, potentielle Transportwege für organische Osmolyte im RVD-Prozess molekular zu identifizieren. Da es grundlegend ungeklärt war, wie viele unterschiedliche Transporter bzw. Kanäle am Efflux der zahlreichen organischen Osmolyte beteiligt sind, erfolgte zunächst die vergleichende Analyse des schwellungsaktivierten Membrantransports strukturell verschiedener SOOs einschließlich der Aminosulfonsäure Taurin und des Polyols myo-Inositol. Hierbei wurde erstmals gezeigt, dass die schwellungsaktivierten Transportwege für Taurin und myo-Inositol deutlich unterschiedliche Aktivitätsprofile aufweisen. Während der Taurintransport bereits unter milden hypotonen Bedingungen, d.h. nach einer geringen Absenkung der Osmolalität von 300 auf ~230 mOsm, aktiviert wurde, erfolgte die Aktivierung der Membranpermeabilität für myo-Inositol bei einer viel niedrigeren Osmolalität von ~150 mOsm. Darüber hinaus wiesen die beiden Transportwege unter vergleichbarem hypotonen Stress von 100 mOsm deutlich unterschiedliche Aktivitätsdauern auf (Transport von Taurin ~95 min und myo-Inositol ~40 min). Somit deuteten diese Ergebnisse erstmals auf substrat-spezifische Transportwege für SOOs hin, die voneinander stark abweichende osmotische Aktivierungsprofile besitzen.
Als aussichtsreiche Kandidaten für diese Transportwege wurden zwei Mitglieder der Gruppe der Solute Carrier (SLC) untersucht, die klare Übereinstimmungen mit den gesuchten Transportern für SOOs aufweisen. Daher wurde im Weiteren eine RVD-Beteiligung dieser Transportergruppe mit einer Kombination aus molekularbiologischer und konventioneller bzw. hochaufgelöster mikroskopischen Techniken überprüft. Die semiqantitativen RT-PCR-Ergebnisse dieser Arbeit zeigen dabei, dass die Gentranskription der potentiellen SOO-Transporter SLC5A3 und SLC6A6 in den untersuchten Zelllinien Jurkat, HEK wie auch HepG2-Zellen durch hypotone Bedingungen deutlich verstärkt wird. Hierbei nimmt der zelluläre mRNA-Gehalt der Gene SLC5A3 zwischen 20-60% und SLC6A6 um 30-100% innerhalb von 10-20 min zu, was auf eine potentielle RVD-Beteiligung von SLC-Transportern hindeutet. Ausgehend von diesem Ergebnis wurde daraufhin die zelluläre Lokalisation des SLC5A3-Transporters unter isotonen und hypotonen Bedingungen mikroskopisch untersucht. Wie anhand der konfokalen lasermikroskopischen Untersuchung zu erkennen ist, findet unter hypotoner Stimulation eine zelluläre Umverteilung des mit EGFP fluoreszenzmarkierten Proteins SLC5A3 statt. Innerhalb von 10 min wird der Transporter dabei von intrazellulären Regionen in Richtung Plasmamembran verlagert. Darüber hinaus konnte mit Hilfe der hochauflösenden Mikroskopie-Technik dSTORM gezeigt werden, dass der Transporter SLC5A3 unter hypotoner Stimulation verstärkt mit der Plasmamembran assoziiert vorliegt. Diese verstärkte Membranassoziation des SLC5A3-Proteins deutet damit auf einen schwellungsinduzierten exozytotischen Einbau des Transporters hin.
Die Ergebnisse dieser Arbeit zeigen damit erstmals, dass SLC-Transporter wie SLC5A3, SLC6A6 und vermutlich andere Vertreter der SLC-Superfamilie potentiell am Mechanismus der hypotonen Volumenregulation beteiligt sind. Da SLC-Transporter als wichtige Transportsysteme für Therapeutika angesehen werden und die Mechanismen der Volumenregulation bereits in zahlreichen biotechnologischen Anwendungen implementiert sind, könnte der hier aufgedeckte Zusammenhang einen Erkenntnisgewinn für zahlreiche biomedizinische Forschungsgebiete darstellen.
Emotionale Esser neigen dazu, in emotional belastenden Situationen überwiegend süße und fettreiche Nahrung häufig in Abwesenheit von Hunger zu essen, um negative Gefühle zu bewältigen. In unangenehmen emotionalen Zuständen setzen sie sich kaum mit den Emotionen auseinander und essen stattdessen. Es fällt ihnen oft schwer, ihren Emotionen, aber auch ihren Hunger- und Sättigungsgefühlen Aufmerksamkeit zu schenken und diese zu erkennen. Emotionales Essverhalten kann Betroffene davon abhalten, einen konstruktiven Umgang mit den emotionalen Belastungen zu erlernen, und kann zu Übergewicht, den mit Übergewicht einhergehenden ernährungsbedingten Erkrankungen oder gar zu Essstörungen führen. Um diese langfristigen Folgen gar nicht erst entstehen zu lassen, ist es von zentraler Bedeutung, problematischen Formen des emotionalen Essverhaltens vorzubeugen oder sie zu verändern. Die vorliegende Dissertation umfasst 3 empirische Studien, in denen anhand von standardisierten und selbst entwickelten Fragebögen sowie mithilfe der Experience-Sampling-Methode die Auslöser und die Modifikation emotionalen Essverhaltens untersucht wurden. Die Experience-Sampling-Methode basiert auf alltagsnahen, zeitlich präzisen, multiplen Messungen im Feld. In der 1. Studie wurde bei gesunden Personen die Wirkung der negativen Gefühle als Auslöser des emotionalen Essverhaltens im Alltag beobachtet. Die Befunde deuten darauf hin, dass keine bestimmten negativen Emotionen zu existieren scheinen, die einen stärkeren Einfluss auf das emotionale Essverhalten haben als andere, und dass alle negativen Gefühle nahezu gleichermaßen das Potenzial bergen, emotionales Essverhalten hervorzurufen. Diese Erkenntnis ist in die Konzeption eines Trainingsprogramms zur Veränderung des emotionalen Essverhaltens für emotionale Esser eingeflossen, das achtsamkeitsbasierte Konzepte mit verhaltenstherapeutischen Behandlungsmethoden kombiniert. Die 2. und die 3. Studie überprüften mit einem randomisierten, kontrollierten Design die Machbarkeit und die Wirksamkeit des Gruppentrainings im ambulanten und klinischen Setting. Das Training ließ sich ambulant und in einer Klinik sehr gut umsetzen und wurde von den Teilnehmern weitgehend positiv bewertet. Im Anschluss an das Training aßen emotionale Esser weniger emotional, indem sie durch die achtsame Selbstbeobachtung lernten, die Essauslöser besser zu erkennen und sie von den körperlichen Hungerempfindungen zu unterscheiden. Vielmehr fingen sie bis zu einem gewissen Grad an, achtsamer zu essen und das Essen zu genießen. Die Emotionsregulation verbesserte sich ebenfalls in vielen Aspekten. Die Trainingsteilnehmer entwickelten die Kompetenzen oder tendierten dazu, ihre Gefühle aufmerksamer wahrzunehmen, klarer zu erkennen und zu benennen, sie stärker positiv zu beeinflussen und sie leichter zu akzeptieren, wenn sie im Augenblick nicht verändert werden konnten. Somit konnten die Machbarkeit und die Wirksamkeit des entwickelten Trainingsprogramms sowohl im ambulanten Setting als auch in den bedeutendsten Aspekten im klinischen Kontext als Teil eines breiter angelegten Therapiekonzepts nachgewiesen werden. Die drei vorgelegten Arbeiten liefern einen Beitrag zum Verständnis und zur Veränderung des emotionalen Essverhaltens. Weitergehende Untersuchungen sollten die Merkmale der äußeren Situation als Risikofaktoren für das emotionale Essverhalten analysieren sowie die zeitliche Stabilität der Trainingseffekte testen, die außerhalb einer Katamnese von 3 Monaten liegt. Die Wirksamkeit des Trainingsprogramms könnte ferner gegenüber anderen Therapieverfahren und multizentrisch in unterschiedlichen Kliniken unter zahlreichen Rahmenbedingungen geprüft werden.
Die Lokalisationsmikroskopie ist eine neue, vielversprechende Methode der hochauflösenden Fluoreszenzmikroskopie. Sie ermöglicht detaillierte Einblicke in die Organisation und den strukturellen Aufbau von Zellen. Da die Vorbereitung der Proben und das Aufnehmen der Bilder im Vergleich zu herkömmlichen Methoden höhere Anforderungen stellt, mussten ihr Potential und ihre Zuverlässigkeit erst noch überzeugend gezeigt werden. Bis vor kurzem wurde das Auflösungsvermögen vor allem an Mikrotubuli gezeigt, deren filamentöse Struktur allerdings schon in konfokalen Bildern zu erkennen ist. Deswegen wurde in dieser Dissertation der Kernporenkomplex (NPC), dessen Struktur in der konventionellen Fluoreszenzmikroskopie nicht auflösbar ist, als Modellstruktur für die hochauflösende Fluoreszenzmikroskopie eingeführt.
Dazu wurden Kernporenkomplexe aus Kernhüllen von Xenopus laevis Oocyten mit dSTORM (direct stochastic optical reconstruction microscopy), einer Methode der Lokalisationsmikroskopie, hochaufgelöst. Damit konnte nun erstmals die Achtfachsymmetrie dieses Proteinkomplexes lichtmikroskopisch dargestellt werden. Desweiteren konnte der Zentralkanal mit einem Durchmesser von ca. 40 nm aufgelöst werden. Die Daten eigneten sich außerdem für eine automatisierte Bildanalyse nach dem sogenannten "particle averaging" - einer aus der Elektronenmikroskopie bekannten Methode, um eine Durchschnittsstruktur zu ermitteln.
Darüber hinaus wurden Zweifach-Färbungen von NPCs benutzt, um verschiedene Ansätze für Zweifarben-Aufnahmen mit dSTORM zu testen. Neben dem mittlerweile standardmäßig benutzten, sequentiellen Ansatz mit zwei spektral getrennten Farbstoffen, wurde auch ein simultaner Ansatz mit zwei spektral überlappenden Farbstoffen erfolgreich angewandt. Auch für 3D-Messungen mit den Ansätzen Biplane und Astigmatismus eignete sich die Markierung der Kernhülle. Hier wurden jedoch A6-Zellen benutzt und die Krümmung des Zellkerns über die gefärbten Kernporen dargestellt.
dSTORM-Messungen können nicht nur an fixierten, sondern auch in lebenden Zellen durchgeführt werden. Hierzu eignen sich vor allem sehr immobile Proteine, wie H2B oder Lamin C. Anhand von SNAP-Tag- und Halo-Tag-Konstrukten konnte gezeigt werden, dass sich kommerziell erhältliche, organische Farbstoffe auch in endogener zellulärer Umgebung schalten lassen, wodurch Lebendzell-Aufnahmen mit dSTORM möglich sind.
Ein weiterer Teil dieser Arbeit befasst sich mit korrelativen Aufnahmen aus dSTORM und Rasterelektronenmikroskopie (SEM). Hierzu wurden Xenopus laevis Kernhüllen zuerst mit dSTORM hochaufgelöst und danach für die EM präpariert. Anschließend wurden zugehörige Bereiche am Rasterelektronenmikroskop aufgenommen. Mit den erhaltenen korrelativen Bildern konnte gezeigt werden, dass sich dSTORM und SEM bei geeigneten Proben durchaus kombinieren lassen. Proteine können somit spezifisch markiert und im Rahmen ihrer strukturellen Umgebung mit nahezu molekularer Auflösung dargestellt werden.
Da hochwertige Aufnahmen eine ausgereifte Probenpräparation voraussetzen, darf deren Etablierung nicht zu kurz kommen. Unter dieser Prämisse wurde ein optimiertes Markierungsprotokoll mit dem Namen ClickOx entwickelt. Mit ClickOx bleibt bei der kupferkatalysierten Azid-Alkin-Cycloaddition die Feinstruktur von Aktinfilamenten, sowie die Fluoreszenz fluoreszierender Proteine, deutlich sichtbar erhalten. Während bei den klassischen Click-Protokollen auf Grund der Entstehung von reaktiven Sauerstoff-Spezies (ROS) feine zelluläre Strukturen, wie Aktinfilamente, angegriffen oder zerstört werden, schützt das neue Protokoll mit enzymatischem Sauerstoffentzug Proteine und somit Strukturen vor Reaktionen mit ROS. Das unterstreicht, wie wichtig es ist auch sogenannte "etablierte" Protokolle weiterzuentwickeln, denn bestimmte Nebeneffekte in Präparationen werden unter Umständen erstmals in der Hochauflösung sichtbar.
Ein weiterer Aspekt war die Untersuchung des Einflusses von D1 auf die Chromatinorganisation. Mit verschiedenen mikroskopischen Methoden konnten Hinweise auf eine mögliche DNA-Cross-Linking-Fähigkeit dieses Proteins gesammelt werden. Hier wurde die Einzelmolekülinformation der dSTORM-Filme genutzt, um unterschiedliche Grade von DNA- bzw. Chromatin-Akkumulation zu vergleichen. Die Ergebnisse deuten darauf hin, dass wildtypisches D1 DNA vernetzen kann. Dies erfolgt über die sogenannten AT-Haken-Motive. Sobald diese alle durch Mutation funktionsunfähig gemacht werden - wie bei der verwendeten R10xG-Mutante - lässt sich keine Akkumulation der DNA mehr beobachten. Neben der Chromatinaggregation durch D1-Expression konnte in FRAP-Experimenten gezeigt werden, dass nur die "echten" AT-Haken eine hohe Affinität zum Chromatin aufweisen, die sogenannten "potentiellen" hingegen nicht.
Mammalian haloacid dehalogenase (HAD)-type phosphatases are an emerging family of enzymes with important functions in physiology and disease. HAD phosphatases can target diverse metabolites, lipids, DNA, and serine/threonine or tyrosine phosphorylated proteins with often high specificity (Seifried et al., 2013). These enzymes thus markedly enlarge the repertoire and substrate spectrum of mammalian phosphatases. However, the basis of HAD phosphatase substrate specificity is still elusive and a number of mammalian HAD phosphatases remain uncharacterized to date. This study characterizes the biochemical and structural properties of AUM (aspartate-based, ubiquitous, Mg2+-dependent phosphatase), a previously unexplored mammalian HAD phosphatase.
In vitro phosphatase assays of purified, recombinant AUM showed phosphatase activity towards para-nitrophenyl phosphate and adenine and guanine nucleotide di- and triphosphates. Inhibitor studies indicated that similar to other HAD superfamily members, the AUM-catalyzed dephosphorylation reaction proceeds via a pentacovalent phosphoaspartate intermediate. In line with an aspartate-based catalytic mechanism, AUM was insensitive to inhibitors of serine/threonine phosphatases. The characterization of the purified recombinant murine enzyme also revealed that AUM exists in equilibrium between dimers and tetramers.
AUM was identified as the closest, yet functionally distinct relative of chronophin, a pyridoxal 5’-phosphate and serine/threonine-directed phosphatase. Phylogenetic analyses showed that AUM and chronophin evolved via duplication of an ancestral gene at the origin of the vertebrates. In contrast to chronophin, AUM acts as a tyrosine-specific HAD-type phosphatase in vitro and in cells. To elucidate how AUM and chronophin achieve these distinct substrate preferences, comparative evolutionary analyses, biochemical approaches and structural analyses were combined. Swapping experiments of less homologous regions between AUM and chronophin were performed. The mutational analysis revealed residues important for AUM catalysis and specificity. A single differently conserved residue in the cap domain of AUM or chronophin is crucial for phosphatase specificity (AUML204, chronophinH182). The X-ray crystal structure of the AUM cap fused to the catalytic core of chronophin (CAC, PDB: 4BKM) was solved to 2.65 Å resolution. It presents the first crystal structure of the murine AUM capping domain. The detailed view of the catalytic clefts of AUM and chronophin reveals the structural basis of the divergent substrate specificities. These presented findings provide insights into the design principles of capped HAD phosphatases and show that their substrate specificity can be encoded by a small number of predictable residues. In addition, the catalytic properties of AUM were investigated, identifying a mechanism of reversible oxidation regulating the activity of AUM in vitro. AUM phosphatase activity is inhibited by oxidation and can be recovered by reduction. The underlying molecular mechanism was revealed by mutational analyses. The cysteines C35, C104 and C243, located in the AUM core domain, are responsible for the inhibition of AUM by oxidation. C293 mediates the redox-dependent tetramerization of AUM in vitro. Based on the chronophin and CAC structure, a direct impact of the oxidation of C35 on the nucleophile D34 is proposed. In addition, a redox-dependent disulfide bridge (C104, C243), connecting the core and cap domain of AUM may be important for an open/close-mechanism. This hypothesis is supported by CD spectroscopy experiments that demonstrate a structural change in AUM upon reduction. These data present the first evidence for the regulation of AUM catalysis by reversible oxidation. This finding is so far unique in the field of HAD phosphatases.
In this context, the first cell-based AUM activity assay was developed. For this, the artificial substrate pNPP was combined with the reducing agent DTT to create a specific AUM activity readout. This fractionation-based assay is the first tool to differentiate between cell lines or tissues with different AUM concentrations or activities.
Taken together, the presented biochemical characterization reveals the specificity determinants and catalytic properties of AUM. General insights into structural determinants of mammalian HAD phosphatase substrate recognition are provided and reversible oxidation as possible regulatory mechanism for AUM is proposed. These findings constitute a framework for further functional analyses to elucidate the biomedical importance of AUM.
In der vorliegenden Studie wurden 220 Patientinnen und Patienten prospektiv 1 Jahr lang nach der Anlage einer PEG-Sonde verfolgt. Die Studie sollte der Frage nachgehen, welche Patienten bzw. welche Indikationsgruppen in welchem Umfang von einer PEG-Anlage kurz- oder langfristig profitieren.
Außerdem wurde die Zufriedenheit der Patienten, ihrer Angehörigen, Betreuer und Hausärzte sowie auf den Einfluss einer PEG-Anlage und der Sondenernährung auf die Lebensqualität der Betroffenen untersucht. Die Befragung wurde über strukturierte Telefoninterviews nach 7 Tagen, 4 Wochen und dann im Abstand von jeweils 3 Monaten bis abschließend ein Jahr nach Sondenanlage durchgeführt.
Die Gesamtheit der Patienten setzte sich zu 41% aus Frauen und 59% aus Männern zusammen. Das Durchschnittsalter lag bei 72 Jahren, die Altersspanne bewegte sich zwischen 39 und 97 Jahren.
Es zeigte sich eine außergewöhnlich hohe Zufriedenheit aller Befragten mit der PEG-Sonde und deren Auswirkung auf die Lebensqualität. 95,7% der befragten Patienten beurteilten ihre Zufriedenheit mit der PEG als sehr gut oder gut. Der Grad der Zufriedenheit war auch bei den Angehörigen, Pflegekräften und Ärzten sehr hoch: 94%, 95,2% und 92% bewerteten die PEG mit sehr gut/ gut. Die wenigen negativen Beurteilungen basierten im Wesentlichen auf Komplikationen, die in Verbindung mit der Sondenernährung auftraten, und auf dem Hinterfragen der Notwendigkeit der PEG-Anlagen. Unterschiede zwischen den einzelnen Indikationsgruppen gaben die Befragten mit einer Ausnahme nicht an. Während Angehörige und Pflegekräfte ihre hohe Zustimmung zur PEG auch bei den Demenzkranken zum Ausdruck brachten (100%, bzw. 95% sehr gute/gute Bewertung), lassen die Hausärzte in dieser Indikation gewisse Vorbehalte erkennen. 9,1% vergaben die Noten mangelhaft/ungenügend und weitere 13,6% die Bewertung befriedigend/ausreichend. Sie begründen ihre Kritik mit ethischen und medizinischen Überlegungen.
Zusammenfassend bestätigt sich die PEG in dieser prospektiven Studie als sichere und effektive Maßnahme zur enteralen Ernährung. Die Indikationsstellung darf nicht nur den Erhalt oder die Steigerung des Körpergewichtes berücksichtigen, sondern sollte auch die Aspekte wie Komorbiditäten, Prognose und nicht zuletzt Allgemeinzustand und Lebensqualität der Patienten beachten. Nach einer PEG-Anlage ist die allgemeine Zufriedenheit bei Patienten Angehörigen, Pflegekräften und Ärzten fast ausnahmslos sehr hoch.
Die Verwendung des Motivs der Schönheit in Vg einerseits und in LXX/ Hs 151 andererseits zeigt eine auffällige Differenz. Während Judit sich in Jdt 10,2-4 LXX / Hs 151 nur schön macht und ursprünglich bereits als Gott gegebene Voraussetzung schön war (Jdt 8,7), lässt Idt 10,4 Vg sie durch das Handeln Gottes erst schön werden.
Schönheit ist damit in der Vg nicht mehr länger fester Bestandteil der Figurencharakterisierung Iudiths und auf einer Linie mit innerer Rechtschaffenheit und göttlichem Segen zu sehen, wie es noch für LXX und Hs 151 galt. In der Vg verliert die Schönheit vielmehr an Bedeutung, denn Iudith erhält sie einzig und allein, um die Begierde des feindlichen Feldherrn zu wecken, damit dieser dadurch zu Fall kommt. Während Holofernis seiner negativ bewerteten Begierde unterliegen wird, ist es Iudith, die sich durch das Gegenteil, durch ihre „Keuschheit" (castitas), moralisch abhebt und darum zunächst nicht als schön, sondern als „anständig" (elegans) charakterisiert wird (Idt 8,7). In Idt 10,4 wird ihr dann die Tugend (virtus) statt der „Begierde" (libido) zugeordnet. Aufgrund ihrer keuschen Lebensweise wird Iudith von Gott zur rettenden Tat bestimmt, der sie zur Verwirklichung derselben schön macht. Die Keuschheit ist es, die den Leserinnen und Lesern, wie in der praefatio beschrieben, als nachahmungswürdiges Vorbild dienen soll.
Iudith verkörpert damit all das, was in den hieronymianischen Schriften vielfach als rechtes Leben vor Gott beschrieben ist und wird dafür von Gott belohnt. Die Abänderungen in Idt 8 7 und Idt 10 ,4 im Vergleich zu den anderen Textfassungen zeigen das hieronymianische Profil der Iudith-Erzählung und zielen auf diese Botschaft hin.
In einer ständig älter werdenden Bevölkerung, in der auch die Senioren einen unverändert hohen Anspruch an ihre Lebensqualität aufrechterhalten, kommt dem Ersatz verschlissener Gelenke und hier besonders der Hüftendoprothetik eine ständig wachsende Bedeutung zu. Ausgehend von den Erfolgen der minimal-invasiven Chirurgie in anderen chirurgischen Disziplinen entwickelte sich dieser Zweig seit Beginn des Jahrtausendes auch in der Hüftchirurgie: 2005 stellte Rachbauer eine minimal-invasive Variante des seit 1887 bekannten anterioren Zugangs zur Hüfttotalendoprothesenimplantation vor. Zum kurzfristigen postoperativen Intervall existieren bereits zahlreiche Studien. Hier zeigt der anteriore Zugang entscheidende Vorteile im Vergleich zu anderen Zugängen. Hervorzuheben sind ein geringeres postoperatives Schmerzausmaß, eine kürzere stationäre Liegedauer und eine schnellere Rehabiliation. Ein gravierender Nachteil des anterioren Zugangs besteht darin, dass er aufgrund des schmalen Operationskanals eine hohe Qualifikation des Operateurs erfordert und mit einer anfänglich erhöhten Komplikationsrate vergesellschaftet sein kann.
Ziel der vorliegenden Arbeit war es, die Resultate des minimal-invasiven anterioren Zugangs in einem mittelfristigen postoperativen Intervall mit den Resultaten des lateralen Zugangs, den man als eine weithin etablierte konventionelle Methode betrachten kann, zu vergleichen. Untersucht wurden 85 über einen minimal-invasiven anterioren Zugang implantierte primäre Hüfttotalendoprothesen nach durchschnittlich 3,7 Jahren post operationem, denen 86 Fälle mit einem lateralen Zugang nach durchschnittlich 5,5 Jahren entgegengestellt wurden. Beide Gruppen unterschieden sich nicht signifikant in Alter und Geschlechterverteilung. Der signifikante Unterschied der Nachuntersuchungsintervalle ist ohne Relevanz, da sich sich beide Gruppen in einer komplikationsarmen postoperativen Phase befinden, die in etwa vom 2. bis zum 10. Postoperativen Jahr geht.
Die Gruppen wurden in Funktion (HHS), Aktivität (UCLA activity score), Schmerzen (abgeleitet aus dem HHS), Komplikationsraten und Gesundheitsempfinden (SF-36) miteinander verglichen.
Die vorliegende Arbeit kam zu den folgenden Ergebnissen über den mittelfristigen Zeitraum: Die Unterschiede in der Funktion waren marginal, die HHS-Durchschnittswerte in beiden Gruppen waren exzellent und nicht signifikant unterschiedlich. Ebenso zeigten beide Gruppen gleichwertige Ergebnisse im subjektiven Gesundheitsempfinden (SF-36). Ein weiteres wichtiges Erfolgskriterium war die Frage nach der durch die Operation erreichten Aktivität. Hier fand sich eine Diskrepanz zwischen UCLA und TWB. Der UCLA ist zur Aktivitätsmessung weit verbreitet. Im UCLA zeigten sich die Patienten der lateralen Gruppe aktiver. Der TWB ergab gleiche Aktivität in beiden Gruppen. Nach dem eingehenden Vergleich beider Fragebögen, kamen wir in dieser Arbeit jedoch zu der Auffassung, dass der UCLA zwar als ein schnell erhebbarer Test für den klinischen Alltag praktisch ist, der wesentlich aufwendigere TWB aber zuverlässigere Ergebnisse liefert.
Die Komplikationsraten waren in beiden Gruppen gering. Die Wahrscheinlichkeit, eine der erfassten Komplikationen zu erleiden, betrug 10,7% für den minimal-invasiven und 10,5% für den klassischen Zugang. Um aber nicht nur die Quantität, sondern auch die Qualität der aufgetretenen Komplikationen erfassen zu können, wurde in dieser Arbeit der Komplikationsindex eingeführt, der die registrierten Komplikationen in Gruppen verschiedener Schweregrade unterteilt. Eine schwere Komplikation floss mit 10 Punkten, eine mittlere mit 5 und eine leichte Komplikation mit einem Punkt in den Komplikationsindex ein. Der anteriore Zugang zeigte so mit insgesamt 26 Punkten einen deutlich geringeren Komplikationsindex als der laterale mit insgesamt 60 Punkten. Dem entsprach eine Reoperationsrate von 1,2% unter den anterioren Patienten im Vergleich zu 3,3% bei den lateralen. Die Schaft- und die Pfannenüberlebensrate lag in beiden Gruppen bei 100%.
Zusammenfassend zeigt die vorliegende Arbeit, dass der anteriore Zugang viele Vorteile gegenüber dem lateralen Zugang aufweist. Die entscheidenden Unterschiede finden sich im kurzfristigen postoperativen Zeitraum. Zu einem mittelfristigen Zeitpunkt erzielen beide Zugänge gleichwertige Ergebnisse.
Welche langfristigen Resultate das noch relativ junge Verfahren des minimal-invasiven anterioren Zugangs hervorbringen wird, muss durch künftige prospektiv randomisierte Studien großer Fallzahlen belegt werden.
SPRED proteins are inhibitors of the Ras/ERK/MAPK signaling pathway, an evolutionary highly conserved and very widespread signaling cascade regulating cell proliferation, differentiation, and growth. To elucidate physiological consequences of SPRED2 deficiency, SPRED2 KO mice were generated by a gene trap approach. An initial phenotypical characterization of KO mice aged up to five months identified SPRED2 as a regulator of chondrocyte differentiation and bone growth. Here, the loss of SPRED2 leads to an augmented FGFR-dependent ERK activity, which in turn causes hypochondroplasia-like dwarfism. However, long term observations of older KO mice revealed a generally bad state of health and manifold further symptoms, including excessive grooming associated with severe self-inflicted wounds, an abnormally high water uptake, clear morphological signs of kidney deterioration, and a reduced survival due to sudden death. Based on these observations, the aim of this study was to discover an elicitor of this complex and versatile phenotype.
The observed kidney degeneration in our SPRED2 KO mice was ascribed to hydronephrosis characterized by severe kidney atrophy and apoptosis of renal tubular cells. Kidney damage prompted us to analyze drinking behavior and routine serum parameters. Despite polydipsia, which was characterized by a nearly doubled daily water uptake, the significantly elevated Na+ and Cl- levels and the resulting serum hyperosmolality could not be compensated in SPRED2 KOs. Since salt and water balance is primarily under hormonal control of aldosterone and AVP, we analyzed both hormone levels. While serum AVP was similar in WTs and KOs, even after experimental water deprivation and an extreme loss of body fluid, serum aldosterone was doubled in SPRED2 KO mice. Systematic investigation of contributing upstream hormone axes demonstrated that hyperaldosteronism developed independently of an overactivated Renin-Angiotensin system as indicated by halved serum Ang II levels in KO mice. However, aldosterone synthase expression in the adrenal gland was substantially augmented. Serum corticosterone, which is like aldosterone released from the adrenal cortex, was more than doubled in SPRED2 KOs, too. Similar to corticosterone, the production of aldosterone is at least in part under control of pituitary ACTH, which is further regulated by upstream hypothalamic CRH release. In fact, stress hormone secretion from this complete hypothalamic-pituitary-adrenal axis was upregulated because serum ACTH, the mid acting pituitary hormone, and hypothalamic CRH, the upstream hormonal inductor of HPA axis activity, were also elevated by 30% in SPRED2 KO mice. This was accompanied by an upregulated ERK activity in paraventricular nucleus-containing hypothalamic brain regions and by augmented hypothalamic CRH mRNA levels in our SPRED2 KO mice. In vitro studies using the hypothalamic cell line mHypoE-44 further demonstrated that both SPRED1 and SPRED2 were able to downregulate CRH promoter activity, CRH secretion, and Ets factor-dependent CRH transcription. This was in line with the presence of various Ets factor binding sites in the CRH promoter region, especially for Ets1.
Thus, this study shows for the first time that SPRED2-dependent inhibition of Ras/ERK/MAPK signaling by suppression of ERK activity leads to a downregulation of Ets1 factor-dependent transcription, which further results in inhibition of CRH promoter activity, CRH transcription, and CRH release from the hypothalamus. The consecutive hyperactivity of the complete HPA axis in our SPRED2 KO mice reflects an elevated endogenous stress response becoming manifest by excessive grooming behavior and self-inflicted skin lesions on the one hand; on the other hand, in combination with elevated aldosterone synthase expression, this upregulated HPA hormone release explains hyperaldosteronism and the associated salt and water imbalances. Both hyperaldosteronism and polydipsia very likely contribute further to the observed kidney damage.
Taken together, this study initially demonstrates that SPRED2 is essential for the appropriate regulation of HPA axis activity and of body homeostasis.
To further enlighten and compare consequences of SPRED2 deficiency in mice and particularly in humans, two follow-up studies investigating SPRED2 function especially in heart and brain, and a genetic screen to identify human SPRED2 loss-of-function mutations are already in progress.
Galectin-1 (hGal-1) is overexpressed by numerous cancer types and previously conducted studies confirmed that the β-galactoside-binding protein mediates various molecular interactions associated with tumor growth, spread and survival. Upon interaction with carbohydrate-based binding epitopes of glycan structures on human cell surfaces galectin-1 induces proliferative, angiogenetic and migratory signals and modulates negative T cell regulation which essentially helps the tumor to evade the immune response. These findings attributed galectin-1 a pivotal role in tumor physiology and strongly suggest the protein as target for diagnostic and therapeutic applications.
Within the scope of this work a strategy was elaborated for designing tailor-made galectin-1 ligands by functionalizing selected hydroxyl groups of the natural binding partner N-acetyllactosamine (LacNAc) that are not involved in the sophisticated interplay between the disaccharide and the protein. Synthetic modifications intended to introduce chemical groups i) to address a potential binding site adjacent to the carbohydrate recognition domain (CRD) with extended hGal-1-ligand interactions, ii) to implement a tracer isotope for diagnostic detection and iii) to install a linker unit for immobilization on microarrays.
Resulting structures were investigated regarding their targeting ability towards galectin-1 by cocrystallization experiments, SPR and ITC studies. Potent binders were further probed for their diagnostic potential to trace elevated galectin-1 levels in microarray experiments and for an application in positron emission tomography (PET).
Abstract
Tobacco addiction is considered as a chronic relapsing disorder, characterized by compul-sive drug seeking and intake. Learning processes are stressed to account for the situational-specific expression of core features of the disorder, e.g., craving for drug, tolerance and ex-cessive consumption. According to incentive theories, smoke conditioned stimuli are hy-pothesized to be appetitive in nature, promoting craving, approach and consummatory be-havior. Commonly, smoking cues are treated as simple excitatory conditioned stimuli formed by a close and reliable overlap with the drug effect. However, the smoking ritual comprises a multitude of stimuli which may give rise to different forms of learning and con-ditioned responses partially opposing each other. Previous research suggests the predictive content and the temporal proximity of smoking stimuli to the drug effect as important de-terminants of cue reactivity. In contrast to stimuli related to the preparatory stage of smok-ing and the start of consumption (BEGIN stimuli), stimuli from the terminal stage of smok-ing (END stimuli) apparently lack high cue reactivity. Several lines of evidence suggest the poor cue properties of terminal stimuli to be related to their signaling of poor smoke availa-bility. Indeed, cue reactivity is commonly decreased when smoking appears to be unavaila-ble. Moreover, the learning literature suggests that stimuli predictive for the non-availability of reward may acquire the capacity to modulate or oppose the responses of ex-citatory conditioned stimuli. Therefore, the aim of the present thesis was to enhance our knowledge of stimulus control in human drug addiction and incentive motivation by running a series of conditioning studies with smoke intake and monetary reward as reinforcer. Sub-jective report and physiological measures of motivational valence and consummatory re-sponse tendencies were used as dependent variables.
The first experiment of this thesis used a differential conditioning paradigm to reveal evi-dence for the conditioning of preparatory and consummatory responses to a CS+ for smok-ing. Neutral pictograms served as CSs and single puffs on a cigarette as US. In line with the predictions of incentive theories, the excitatory CS+ for smoking acquired the ability to evoke an appetitive conditioned response, as indicated by enhanced activity of the M. zy-gomaticus major. Moreover, anticipation of puffing on the cigarette increased the activity of the M. orbicularis oris (lip muscle), indicating the activation of consummatory response tendencies. Finally, the CS+ evoked stronger skin conductance responses, indicative of in-creased autonomic arousal and orienting in preparation for action. In contrast, the rating data were apparently unaffected by the experimental contingency. In sum, the physiological data provide support for the notion that excitatory smoke conditioning gives rise to appeti-tive and consummatory conditioned responses, which may at least partially contribute to the maintenance of tobacco addiction.
The second experiment of this thesis adapted the conditioning protocol of the first study to probe the functional significance of terminal stimuli in the control of addictive behavior. This study manipulated the predictive relationship of BEGIN and END stimuli to smoke rein-forcement to provide further support for the differential reactivity to both stimuli and the retarded (i.e., delayed) conditioning of END stimuli. Overall, the results of the first study of this thesis were conceptually replicated as the association of a BEGIN stimulus with smoke intake resulted in the acquisition of appetitive and consummatory physiological responses. Importantly, the results revealed evidence for a retarded excitatory conditioning of END stimuli. Thus, pairing of an END stimulus with smoke intake failed to produce a conditioned discrimination in terms of motivational valence and autonomic arousal, as indicated by the activity of the M. corrugator supercilii and the skin conductance data. These results provide further support for the notion that END stimuli may be weak cues for smoking. Moreover, in light of the results of the first study of this thesis, the retarded excitatory conditioning of terminal stimuli may be suggestive of an inhibitory response component, which may be re-lated to their signaling of poor smoke availability. In sum, these results add to a growing body of data, which suggest that the expression of cue reactivity may be modulated by the temporal proximity and the availability of the drug effect.
The aim of the third study of this thesis was to provide “proof of concept” for an inhibi-tory conditioning notion of terminal stimuli. In this analog study BEGIN and END stimuli were emulated as discriminative SD and S for monetary reward. During an acquisition phase conditioned inhibition was established to the S predictive of the non-availability of re-ward. Subsequently a retardation test was used to substantiate conditioned inhibition. In this test, excitatory conditioning of the previous S was compared to the excitatory condi-tioning of a novel control stimulus. Importantly, the results revealed evidence for reward conditioned inhibition as indicated by the retarded acquisition of subjective (pleasure and reward expectancy) and physiological (skin conductance and activity of the M. orbicularis oculi) responses. In sum, these results provide support for the notion that stimuli predictive for the non-availability of reward may acquire the capacity to oppose the responses of ex-citatory conditioned stimuli. Thus, future research may benefit from the consideration of inhibitory conditioning processes in drug addiction, which may be of theoretical, methodo-logical and clinical importance.
In sum, the present thesis revealed evidence for 1) an appetitive nature of excitatory condi-tioned smoking cues, 2) the dependency of this learning process on the temporal position of the conditioned stimuli in the intake ritual and 3) the acquisition of conditioned inhibition by a stimulus predictive for the non-availability of reward, as evident in retarded excitatory conditioning. Overall, these studies made a novel contribution to the field of human drug addiction and incentive motivation and provided valuable suggestions for further research.
Structural and biochemical characterization of gephyrin and various gephyrin-ligand complexes

Efficient synaptic neurotransmission requires the exact apposition of presynaptic terminals and matching neurotransmitter receptor clusters on the postsynaptic side. The receptors are embedded in the postsynaptic density, which also contains scaffolding and regulatory proteins that ensure high local receptor concentrations. At inhibitory synapses the cytosolic scaffolding protein gephyrin assumes an essential organizing role within the postsynaptic density by the formation of self-oligomers which provide a high density of binding sites for certain -amino butyric acid type A (GABAA) and the large majority of glycine receptors (GlyR). Gephyrin contains two oligomerization domains: In isolation, the 20 kDa N-terminal G domain (GephG) and the 46 kDa E domain (GephE) trimerize and dimerize, respectively. In the full-length protein the domains are interconnected by a central ~150 amino acid linker, and only GephG trimerization is utilized, whereas GephE dimerization is prevented, thus suggesting the need for a trigger to release GephE autoinhibition, which would pave the way for the formation of higher oligomers and for efficient receptor clustering. The structural basis for this GephE autoinhibition has remained elusive so far, but the linker was reported to be sufficient for autoinhibition. This work dealt with the biochemical and structural characterization of apo-gephyrin and gephyrin in complexes with ligands which are known to promote the formation of synaptic gephyrin clusters (collybistin and neuroligin 2) and reorganize them (dynein light chain 1).
For full-length gephyrin no structural information has been available so far. Atomic force microscopy (AFM) and small-angle X-ray scattering (SAXS) analyses described in this thesis disclosed that the gephyrin trimer forms a highly flexible assembly, which, due to the long linker, can switch between compact and extended conformational states in solution, with a preference for compact states. This partial compaction and potentially GephE autoinhibition are achieved by interactions of parts of the linker with the G and E domains, as suggested by circular dichroism spectroscopy. However, the linker on its own cannot account for GephE blockage, as size exclusion chromatography experiments coupled with multi angle light scattering detection (SEC-MALS) and SAXS analyses revealed that a gephyrin variant only encompassing the linker and GephE (GephLE) forms dimers and not monomers as suggested by an earlier study. The oligomeric state of GephLE and the observation that several gephyrin variants, in which linker segments of varying length were deleted, predominantly formed trimers, suggested the presence of a linker independent mechanism of GephE dimerization blockade. Taken together, the data indicated that linker-dependent and linker-independent mechanisms mediate gephyrin autoinhibition.
In the second project gephyrin’s interaction with DYNLL1 (Dynein LC8 Light Chain 1) was characterized. DYNLL1 is a 25 kDa dimer incorporated into the dynein motor and provides two binding sites, each of which can accommodate an octapeptide derived from gephyrin’s linker region (referred to as GephDB). Originally, DYNLL1 was regarded as a cargo adaptor, linking gephyrin-GlyR complexes to the dynein motor, thus driving their retrograde transport and leading to a decrease of synaptic gephyrin-GlyR complexes.
Building on these studies, this thesis assessed the cargo hypothesis as well as the so far unclear stoichiometry of the gephyrin-DYNLL1 complex. The cargo scenario would require ternary complex formation between gephyrin, DYNLL1 and the dynein intermediate chain (DIC) of the dynein motor. However, such a complex could not be detected by analytical size exclusion chromatography (aSEC) experiments – presumably because gephyrin and DIC competed for a common binding site in DYNLL1. This finding was consistent with a single DYNLL1 dimer capturing two linker segments of a single gephyrin trimer as suggested by a 26 kDa mass increase of the gephyrin species in the presence of DYNLL1 in SEC-MALS experiments. aSEC experiments at even higher concentrations (~20 µM gephyrin and ~80 µM DYNLL1) indicated that the affinity of GephDB was significantly impaired in the context of full-length gephyrin but also in a variant that bears only GephG and the first 39 residues of the linker (GephGL220). Presumably due to avidity effects two linkers stably associated with a single DYNLL1 dimer, whereas the third DYNLL1 binding motif remained predominantly unoccupied unless high concentrations of GephGL220 (50 µM) and DYNLL1 (200 µM) were used. These findings indicate that an interplay between GephG and the N-terminal linker segment mediates the attenuation of GephDB affinity towards DYNLL1 and that preventing DYNLL1 from the induction of higher gephyrin oligomers is either advantageous for DYNLL1-mediated reorganization of gephyrin-GlyR clusters or that DYNLL1 exerts possibly two (concentration-dependent) actions on gephyrin.
The gephyrin-collybistin-neuroligin 2 complex was the subject of the third project. Previously, collybistin and gephyrin were observed to mutually trigger their translocation to the postsynaptic membrane, where the disordered cytoplasmic tail of the postsynaptic cell adhesion molecule NL2 (NL2cyt) causes the anchoring of collybistin 2 (CB2) by binding to its SH3 domain, thereby releasing SH3 domain mediated autoinhibiton of CB2 binding to the membrane phospholipid phosphatidylinositol-3-phosphate. Critical for this event is the binding of gephyrin to both CB2 and NL2, presumably via GephE.
Following up on these previous studies biochemical data presented in this thesis confirm the formation of the ternary complex. Unexpectedly, analyses by means of native polyacrylamide gel electrophoresis pointed to: (1) The existence of a complex containing NL2cyt and CB2 lacking the SH3 domain and consequently an additional NL2 binding site in CB2. (2) Attenuated gephyrin-collybistin complex formation in the presence of the SH3 domain. (3) A requirement for high NL2cyt concentrations (> 30 µM) during the formation of the ternary complex. This might allow for the regulation by other factors such as additional binding partners or posttranslational modifications. Although of preliminary character, these results provide a starting point for future studies, which will hopefully elucidate the interplay between gephyrin, collybistin, NL2 and certain GABAA receptors.
Die zunehmende Versalzung des Bodens führt weltweit zu starken Ernteeinbußen. Ob- wohl die Wurzeln der Pflanzen als erstes mit dem Salzstress in Berührung kommen, ist noch nicht viel über Signaltransduktionswege in Wurzeln zur Anpassung der Pflanze an Salzstress bekannt. Die bZIP-Transkriptionsfaktoren der Gruppe S1, bZIP1 und bZIP53, werden gewebespezifisch in der Wurzel nach Salzstress aktiviert. In dieser Arbeit werden diese bZIPs in ein Netzwerk eingeordnet, von der Aktivierung der Tran- skriptionsfaktoren bis zur Funktion in der Regulation des Stoffwechsels in der salzgest- ressten Pflanze.
Die Aktivierung von bZIP1 kann über verschiedene sowohl ionische als auch osmotische Stimuli erfolgen und ist abhängig von Calcium, der HEXOKINASE 1 und SnRK1- Kinasen (Snf1 RELATED PROTEIN KINASE 1). Die dunkelinduzierte Expression von bZIP1 wird HXK1-abhängig durch Glucose inhibiert, bei Energiemangelbedingungen ist die Aktivierung von bZIP1 SnRK1-abhängig. Beide Enzyme spielen auch in der salzinduzierten Expression von bZIP1 eine Rolle. Über Transkriptom- und Me- tabolomanalysen kann gezeigt werden, dass bZIP1 und bZIP53 an der Umprogram- mierung des Kohlenhydrat- und Aminosäuremetabolismus teilhaben. Besonders Gene der Glukoneogenese (PYRUVAT ORTHOPHOSPHAT DIKINASE und FRUCTOSE- 1,6-BISPHOS- PHATASE) bzw. des Aminosäurekatabolismus (BRANCHED- CHAIN AMINO ACID TRANSAMINASE 2, METHYLCROTONYL- COA-CARBOXYLASE A und HOMOGENTISATE 1,2-DIOXYGENASE ) werden von den Transkriptionsfaktoren reguliert. Das spricht für eine Umprogrammierung des Metabolismus und der Mobilisierung von Energie aus Aminosäuren zur Anpassung an die Stressbedingungen. Die Transkriptionsfaktoren der Gruppe S1 bilden vorzugsweise Heterodimere mit der Gruppe C. Mit Mutantenanalysen, die zum einen die Transkriptionsfaktoren des C/S1-Netzwerks und zum anderen Komponenten der Abscisinsäure (ABA) abhängigen Signaltransduktion beinhalten, konnte ein Signaltransduktionsnetzwerk aufgestellt werden, das die Antwort auf abiotischen Stress mittels des Signalwegs über ABA, SnRK2 und AREB (ABA RESPONSIVE ELEMENTS-BINDING PROTEIN) mit der SnRK1-vermittelten Antwort auf Energiemangelbedingungen in der Pflanze verknüpft. Die gefundenen stress- bzw. energieresponsiven Gene konnten nach den Mutantenana- lysen auf Grund ihrer unterschiedlichen Regulation in vier Klassen eingeteilt werden, wovon nur eine, die Klasse 4, von dem C/S1 Netzwerk reguliert wird. Die Klassen 1- 3 sind unabhängig von den bZIP-Transkriptionsfaktoren der Gruppe C. Die Klasse 1 bilden typische ABA-responsive Gene, die von den Gruppe A-bZIPs reguliert werden. Faktoren der Gruppe A sind auch an der Expression der Gene der Klasse 2 beteiligt, diese werden aber auch durch bZIP1 und bZIP53 induziert. Dieser Klasse konnten Gene zugeordnet werden, die im Abbau verzweigtkettiger Aminosäuren eine Rolle spielen. Am Aminosäureabbau sind außerdem die Gene der Klasse 2 beteiligt. Für diese Gene konnte eine Expressionsregulation durch bZIP1 und bZIP53 gezeigt werden. Für die Bestimmung möglicher Heterodimerisierungspartner bedarf es noch weiterer Analysen. Dieses Model, das den abitoschen Stress abhängigen ABA-Signalweg mit dem ener- gieabhängigen SnRK1-Signaltransduktionsweg verknüpft, zeigt die präzise Regulation von mindestens 4 Gen-Klassen, deren Expression durch die Kombination verschiedener bZIP-Transkriptionsfaktoren aktiviert wird.
Invasive Zygomykosen verzeichnen in den letzten Jahren eine steigende Inzidenz, insbesondere im Risikokollektiv immunsupprimierter Patienten. Aufgrund des häufig letalen Verlaufs dieser Infektionen ist eine rasche, korrekte Diagnosestellung essentiell, um rechtzeitig eine adäquate Therapie einzuleiten. Jedoch sieht sich die konventionelle, mikrobiologische Diagnostik mit vielen Problemen konfrontiert, so dass molekularbiologische Nachweisverfahren zunehmend in den Fokus der Aufmerksamkeit rücken. Eine zuverlässige, mit relativ geringem Zeit- und Kostenaufwand praktizierbare Methode stellt in diesem Zusammenhang die Real-time-PCR dar, deren Aussagekraft durch anschließende Speziesidentifizierung mittels Sequenzierung noch verstärkt werden kann.
Aus diesem Grund wurden im Rahmen dieser Arbeit 3 PCR-Assays entwickelt und deren Sensitivität, Spezifität und klinische Anwendbarkeit evaluiert. Alle 3 Systeme nutzten Multi-copy-Gene des ribosomalen Operons der Zygomyzeten als Target und erwiesen sich als zuverlässige Werkzeuge zur Amplifikation fungaler DNA. Sie wurde sowohl an Pilzkulturen, als auch an klinischen Proben und einem Quasi-Tiermodell mit Erfolg ausgetestet und werden möglicherweise in Zukunft der klinischen Routinediagnostik zur Verfügung stehen.
Bedingt durch die Seltenheit invasiver Zygomykosen besteht in diesem Bereich noch ein großer Forschungsbedarf, auch, um die noch nicht optimale Therapie dieser Erkrankungen zu verbessern. Es bleibt daher zu hoffen, dass sich in absehbarer Zeit mehr Forschungsgruppen mit diesen Erregern beschäftigen, damit den schwer kranken Patienten eine echte Heilungschance geboten werden kann.
Organisms use different resources in different habitat types during their life cycle. Thereby, they connect habitats and provide ecosystem services or disservices in several habitat types. In agricultural landscapes, the spillover of organisms, i.e. movement of an organism and its function from one habitat to another, especially from semi-natural to managed habitats, is one of the most important processes that influence population dynamics and community composition. Importantly, spillover connects habitats not only spatially, but also on different temporal scales, because availability of resources changes over time in agricultural landscapes, e.g. by mass-flowering events of crops, harvesting or crop rotation. Most often, semi-natural habitats are seen as beneficial source of organisms, but also managed habitats can provide valuable resources, and thereby initiate spillover to other habitats. Mass-flowering crops, like oil-seed rape, are such valuable feeding resources for pollinators, and pollinators might spillover from oil-seed rape to other habitats which provide alternative foraging resources. The focus of this dissertation was to evaluate the influence of oil-seed rape on pollinators in agricultural landscapes by studying effects (1) on different temporal scales (from effects during the flowering period of oil-seed rape, Chapter II & IV, to intermediate effects on a second mass-flowering crop, Chapter III, to spillover effects to the flowering period in the next year, Chapter IV), (2) semi-natural (Chapter II) and crop (Chapter III, IV) habitats, and (3) on various pollinator groups which differ in their life cycle (Chapter II, III, IV).
In this dissertation effects from oil-seed rape on all temporal scales – in the short term during mass-flowering and in the long term on a late-flowering crop and even in the next year on oil-seed rape fields ─ were found. These effects might be important for crop and wild plant pollination, and pollinator conservation. Importantly, the effects on different temporal scales depend on the considered habitat (managed or different semi-natural habitats) and on the investigated pollinator group. The more pollinators match the flowering period of oil-seed rape in their activity period and the more dependent they are on flowering resources in their life cycle, the more pronounced are their responses. Effects were found for wild bees, but not for hoverflies and honey bees. Moreover, the availability of semi-natural habitats in the landscape is important and may modulate effects from oil-seed rape. The longevity of effects of oil-seed rape shows the importance of including several temporal scales into ecosystem-service studies, not only for pollinators, but also for other ecosystem-service providing species groups.
