Palladium‐catalyzed [5+2] annulation of 1‐boraphenalenes with ortho‐dihaloarenes afforded negatively curved π‐extended pleiadienes. Two benzo[1,2‐i:4,5‐i’]dipleiadienes (BDPs) featuring a seven‐six‐seven‐membered ring arrangement were synthesized and investigated. Their crystal structure revealed a unique packing arrangement and theoretical calculations were employed to shed light onto the dynamic behavior of the BDP moiety and its aromaticity. Further, a naphthalene‐fused pleiadiene was stitched together by oxidative cyclodehydrogenation to yield an additional five‐membered ring. This formal azulene moiety led to distinct changes in optical and redox properties and increased perturbation of the aromatic system.

The γ-oxoketenes, which are formed from oxadiazinone Ja and strained cyclopentene der1vat1ves, are shown to undergo a pericyclic ring enlargement to give the title compounds 2a, 2b, and 5. In the case of 5, two configurations, one having a cis and the other a trans Iactone functionality, are in equilibrium.

Aus dem Bromfluorcarben-Addukt des Indens wurden durch HPLC an Chiralcel OD die reinen Enantiomere erhalten. Damit stand erstmals eine enantiomerenreine Vorstufe für die Freisetzung eines Sechsringallens im Sinne der Doering-Moore-Skattebol Reaktion zur Verfügung. Die Behandlung dieser Vorstufen, die in 2,5-Dimethyl, 2-tert-Butyl-5-methyl- or 2,5-Di-tert-butylfuran gelöst waren, mit Methyllithium lieferte die [4+2]-Cycloaddukte des Isonaphthalins an die Furane. Durch HPLC an Chiralcel OD wurde gezeigt, dass die Produkt-Enantiomerenpaare mit ca. 40% ee anfielen und dass dieser Wert praktisch unabhängig von der Art des Furans und seiner Konzentration, sowie von der Raktionstemperatur war. Die absoluten Konfigurationen der Vorstufe sowie der [4+2]-Cycloaddukte wurden durch Vergleich der gemessenen und berechneten CD-Spektren ermittelt, was in Zusammenarbeit mit der Arbeitsgruppe von Prof. Dr. B. Engels erfolgte. Erzeugt durch Methyllithium aus dem Dibromcarben-Addukt des Indens wurde das Isonaphthalin erstmals durch Inden abgefangen. Die Konstitution diese [2+2]-Cycloaddukts wurde durch Röntgenstrukturanalyse bestimmt. Erstmals wurde das Bromfluorcarben-Addukt des 1-Phenylcyclopentens hergestellt und erfolgreich daraufhin geprüft, ob es als Vorstufe für 1-Phenylcyclohexa-1,2-dien geeignet ist. Mit Hilfe von HPLC an Chiralcel OJ-H wurden die reinen Enantiomere erhalten. Die Umsetzung eines solchen reinen Enantiomers mit Methyllithium in Gegenwart von 2,5-Dimethylfuran erbrachte das [4+2]-Cycloaddukt als reines Enantiomer. Die Umsetzung einer reinen Vorstufe mit Methyllithium in Gegenwart von Inden erbrachte das [2+2]-Cycloaddukt mit einem Enantiomerenverhältnis von 95:5. Überraschend wurden auch relativ hohe Enantioselektivitäten beim Abfang dieser Vorstufe mit Styrol beobachtet. Bei der Umsetzung des reinen Enantiomers mit Methyllithium und Styrol ergab sich ein Gemisch der zwei endo- und exo-Enantiomere des [2+2]-Cycloaddukts von 50:3:40:7, was ee-Werten von 89 bzw. 79% entspricht. Geht man von einem zweistuifigen Mechanismus der Reaktion von 1-Phenylcyclohexa-1,2-dien mit Styrol aus, dann spielt die achirale Konformation des intermediären Diradikals, wenn überhaupt, keine große Rolle. Die Reduktion des bekannten 1,6-Dibrom-2-phenylcyclohex-1-ens mit Lithiumaluminiumhydrid erbrachte mit 78% Ausbeute die Monobromide 1-Brom-2-phenylcyclohexen and 1-Brom-6-phenylcyclohexen, die bei der Behandlung mit Kalium-tert-butoxid ebenfalls das Cycloallen 1-Phenylcyclohexa-1,2-dien liefern. Dies zeigte die Ausführung dieser Reaktion in Gegenwart von Furan und 2,5-Dimethylfuran.

Diese Arbeit behandelt die Entwicklung von Methoden zur Synthese halogenierter O-Heterocyclen ausgehend von substituierten Bishomoallylalkoholen. Jene acyclischen Substrate können durch Reaktion mit elektrophilen Bromierungsreagenzien Cyclisierungsreaktionen zu O-Heterocyclen eingehen. Diese Strategie ist an die Chemie Vanadium-abhängiger Halogenidperoxidasen (VHPOs) angelehnt, die in der Lage sind, mit H2O2 als Primäroxidans, Halogenide oxidativ zu aktivieren. Dazu wurden sowohl Reaktionen Vanadium-abhängiger Bromidperoxidasen (VBPOs) als auch ihrer funktionaler Modelle untersucht. Als funktionale Modelle in wasserfreien Medien wurden neue Vanadium-Schiffbasekomplexe synthetisiert, charakterisiert (51V-NMR, IR, UV/Vis) und hinsichtlich ihrer Eignung als Oxidationskatalysatoren untersucht. Mittels mechanistischer Untersuchungen konnte gezeigt werden, dass Vanadium-Schiffbasekomplexe mit tert-Butylhydroperoxid (TBHP) zu Peroxykomplexen (ESI-MS, 51V-NMR) reagieren. Diese dienen in wasserfreien Medien als Katalysatoren um Bromid (aus PyHBr) in situ in Br2 umzuwandeln. Das erzeugte elementare Brom kann für Bromcyclisierungsreaktionen substituierter Bishomoallylalkohole zum Aufbau halogenierter cyclischer Ether genutzt werden. Die Ergebnisse aus den Untersuchungen der funktionalen Modelle in wasserfreien Medien wurden mit Enzym-katalysierten Reaktionen verglichen. Bei enzymatischen Reaktionen dominiert vermutlich zunächst eine Bildung von HOBr. Letzteres dient als elektrophiles Bromintermediat, um Bishomoallylalkohole bevorzugt in Halohydrine umzuwandeln. Durchgeführte methodischen Arbeiten zur oxidativen und radikalischen Halogenierung fanden schließlich Anwendung als Schlüsselschritte in Totalsynthesen von Aplysiapyranoid A, einem hexasubstituierten, dreifachhalogenierten Tetrahydropyran-abgeleiteten Naturstoff aus Aplysia kurodai, sowie dessen 5-Epimer.

In this communication, we demonstrate a novel approach to prepare a discrete dimer of chiral phthalocyanine (Pc) by exploiting the flexible molecular geometry of helicenes, which enables structural interlocking and strong aggregation tendency of Pcs. Synthesized [7]helicene-Pc hybrid molecular structure, zinc-[7]helicenocyanine (Zn-7HPc), exclusively forms a stable dimeric pair consisting of two homochiral molecules. The dimerization constants were estimated to be as high as 8.96×10\(^6\) M\(^{−1}\) and 3.42×107 M\(^{−1}\) in THF and DMSO, respectively, indicating remarkable stability of dimer. In addition, Zn\(^{-7}\)HPc exhibited chiral self-sorting behavior, which resulted in preferential formation of a homochiral dimer also in the racemic sample. Two phthalocyanine subunits in the dimeric form strongly communicate with each other as revealed by a large comproportionation constant and observation of an IV-CT band for the thermodynamically stable mixed-valence state.
Although solid-state nuclear magnetic resonance (NMR) is a versatile analytical tool to study polymorphs and phase transitions of pharmaceutical molecules and products, this work summarizes examples of spontaneous and unexpected (and unwanted) structural rearrangements and phase transitions (amorphous-to-crystalline and crystalline-to-crystalline) under magic angle spinning (MAS) conditions, some of them clearly being due to the pressure experienced by the samples. It is widely known that such changes can often be detected by X-ray powder diffraction (XRPD); here, the capability of solid-state NMR experiments with a special focus on \(^{1}\)H-\(^{13}\)C frequency-switched Lee–Goldburg heteronuclear correlation (FSLG HETCOR)/MAS NMR experiments to detect even subtle changes on a molecular level not observable by conventional 1D NMR experiments or XRPD is presented. Furthermore, it is shown that a polymorphic impurity combined with MAS can induce a crystalline-to-crystalline phase transition. This showcases that solid-state NMR is not always noninvasive and such changes upon MAS should be considered in particular when compounds are studied over longer time spans.
This work is concerned with the syntheses and photophysical properties of para-xylylene bridged macrocycles nPBI with ring sizes from two to nine PBI units, as well as the complexation of polycyclic aromatic guest compounds.
With a reduced but substantial fluorescence quantum yield of 21% (in CHCl3) the free host 2PBI(4-tBu)4 can be used as a dual fluorescence probe. Upon encapsulation of rather electron-poor guests the fluorescence quenching interactions between the chromophores are prevented, leading to a significant fluorescence enhancement to > 90% (“turn-on”). On the other hand, the addition of electron-rich guest molecules induces an electron transfer from the guest to the electron-poor PBI chromophores and thus quenches the fluorescence entirely (“turn-off”). The photophysical properties of the host-guest complexes were studied by transient absorption spectroscopy. These measurements revealed that the charge transfer between guest and 2PBI(4-tBu)4 occurs in the “normal region” of the Marcus-parabola with the fastest charge separation rate for perylene. In contrast, the charge recombination back to the PBI ground state lies far in the “inverted region” of the Marcus-parabola.
Beside complexation of planar aromatic hydrocarbons into the cavity of the cyclophanes an encapsulation of fullerene into the cyclic trimer 3PBI(4-tBu)4 was observed. 3PBI(4-tBu)4 provides a tube-like structure in which the PBI subunits represent the walls of those tubes. The cavity has the optimal size for hosting fullerenes, with C70 fitting better than C60 and a binding constant that is higher by a factor of 10. TA spectroscopy in toluene that was performed on the C60@3PBI(4-tBu)4 complex revealed two energy transfer processes. The first one comes from the excited PBI to the fullerene, which subsequently populates the triplet state. From the fullerene triplet state a second energy transfer occurs back to the PBI to generate the PBI triplet state.
In all cycles that were studied by TA spectroscopy, symmetry-breaking charge separation (SB-CS) was observed in dichloromethane. This process is fastest within the PBI cyclophane 2PBI(4-tBu)4 and slows down for larger cycles, suggesting that the charge separation takes place through space and not through bonds. The charges then recombine to the PBI triplet state via a radical pair intersystem crossing (RP-ISC) mechanism, which could be used to generate singlet oxygen in yields of ~20%.
By changing the solvent to toluene an intramolecular folding of the even-numbered larger cycles was observed that quenches the fluorescence and increases the 0-1 transition band in the absorption spectra. Force field calculations of 4PBI(4-tBu)4 suggested a folding into pairs of dimers, which explains the remarkable odd-even effect with respect to the number of connected PBI chromophores and the resulting alternation in the absorption and fluorescence properties. Thus, the even-numbered macrocycles can fold in a way that all chromophores are in a paired arrangement, while the odd-numbered cycles have open conformations (3PBI(4-tBu)4, 5PBI(4-tBu)4, 7PBI(4-tBu)4) or at least additional unpaired PBI unit (9PBI(4-tBu)4).
With these experiments we could for the first time give insights in the interactions between cyclic PBI hosts and aromatic guest molecules. Associated with the encapsulation of guest molecules a variety of possible applications can be envisioned, like fluorescence sensing, chiral recognition and photodynamic therapy by singlet oxygen generation. Particularly, these macrocycles provide photophysical relaxation pathways of PBIs, like charge separation and recombination and triplet state formation that are hardly feasible in monomeric PBI dyes. Furthermore, diverse compound specific features were found, like the odd-even effect in the folding process or the transition of superficial nanostructures of the tetrameric cycle influenced by the AFM tip. The comprehensive properties of these macrocycles provide the basis for further oncoming studies and can serve as an inspiration for the synthesis of new macrocyclic compounds.
Water‐soluble cationic perylene diimide dyes as stable photocatalysts for H\(_2\)O\(_2\) evolution

Photocatalytic generation of hydrogen peroxide, H\(_2\)O\(_2\), has gained increasing attention in recent years, with applications ranging from solar energy conversion to biophysical research. While semiconducting solid‐state materials are normally regarded as the workhorse for photogeneration of H\(_2\)O\(_2\), an intriguing alternative for on‐demand H\(_2\)O\(_2\) is the use of photocatalytic organic dyes. Herein we report the use of water‐soluble dyes based on perylene diimide molecules which behave as true molecular catalysts for the light‐induced conversion of dissolved oxygen to hydrogen peroxide. In particular, we address how to obtain visible‐light photocatalysts which are stable with respect to aggregation and photochemical degradation. We report on the factors affecting efficiency and stability, including variable electron donors, oxygen partial pressure, pH, and molecular catalyst structure. The result is a perylene diimide derivative with unprecedented peroxide evolution performance using a broad range of organic donor molecules and operating in a wide pH range.
In terms of the need of environmentally benign renewable and storable energy sources, splitting of water into hydrogen and oxygen by using sunlight is a promising approach. Hereby, water oxidation catalysts (WOCs) are required to perform the water oxidation comprising the transfer of four electrons to provide the reducing equivalents for producing hydrogen. The class of Ru(bda) (bda = 2,2'-bipyridine-6,6'-dicarboxylate) catalysts has proven to be efficient for this reaction.
In this thesis, ligand exchange processes in Ru(bda) complexes have been analyzed and the formation of multinuclear macrocyclic WOCs was studied. Based on the knowledge acquired by these studies, new multinuclear cyclic Ru(bda) complexes have been synthesized and their catalytic efficiencies in homogeneous water oxidation have been investigated. Going one step further for setting up functional devices, molecular WOCs have been immobilized on conducting or semiconducting supporting materials. Direct anchoring on carbon nanotubes generated a promising materials for further applications.
Wasserstoffbrückengesteuerte Ausrichtung von Merocyaninfarbstoffen für photorefraktive Materialien

Merocyaninchromophore spielen eine herausragende Rolle bei der Entwicklung von photorefraktiven Materialien für Anwendungen in der Holographie. Der photorefraktive Effekt beruht auf einer Orientierung der dipolaren Merocyanine in einem elektrischen Feld. Diese können umso effektiver ausgerichtet werden, je größer ihr Dipolmoment ist. Folglich sollten Merocyanine mit sehr großen Dipolmomenten den gewünschten Effekt hervorbringen. Es hat sich jedoch gezeigt, dass solche Merocyanine Dimere mit antiparalleler zentrosymmetrischer Struktur bilden. In dieser Anordnung addieren sich die Dipolmomente destruktiv, so dass die dipolare Eigenschaft des Materials verloren geht. In dieser Arbeit ist es gelungen, Merocyanine über sechsfache Wasserstoffbrückenbindungen zu supramolekularen Strukturen mit großen resultierenden Dipolmomenten zu assoziieren. Diese Komplexe werden in schwach polaren Lösungsmitteln sogar bei sehr niedrigen Farbstoffkonzentrationen gebildet.
Vom Monomer zum Polymer: Iterative Synthese und optische Spektroskopie von Squarain-Oligomeren

Mittels einer Schutzgruppenstrategie wurden Squarain-basierte monodisperse Oligomere synthetisiert. Die lösungsmittelabhängigen Konformationen (Random Coil vs. Helix) wie auch der Faltungsprozess der Homooligomere wurden mittels optischer Spektroskopie, verschiedener NMR-Experimenten, Kleinwinkelneutronenstreuungsexperimenten sowie quantenchemischen Berechnungen näher beleuchtet. Die optisch-spektroskopischen Beobachtungen wurden mithilfe der Exzitonenkopplungstheorie und einer Orientierungs- und Winkelabhängigkeit der Übergangsdipolmomente der Oligomere erklärt. Der hohe Windungsabstand der helikalen Konformation führt zu einer Interkalation von Lösungsmittel, wodurch eine Art Klathrat gebildet wird. Zusätzlich wurden mittels eines Frenkel-Exzitonenmodells die Absorptions- und Fluoreszenzspektren modelliert. Es konnten die Exzitonendelokalisationslängen abgeschätzt und die Auswirkung der energetischen und strukturellen Unordnungen auf die Absorptions- und Fluoreszenzspektren bestimmt werden. Die Absorptionsspektren werden vorwiegend durch strukturelle Unordnungen verbreitert, die Fluoreszenzspektren dagegen von energetischen Übergangsenergieabweichungen.
Weiterhin wurden auch alternierende Squarain-Cooligomere synthetisiert und mittels optischer Spektroskopie untersucht. Es wurde, abhängig von dem gewählten Lösungsmittel, eine Verschiebung der Hauptbande beobachtet, was durch einen Random Coil vs. helikale-/schlaufenartige Konformation erklärt wird. Gestützt wurde dies mittels quantenchemischen Berechnungen der jeweiligen Konformationen.
Abschließend wurden alternierende Squarain-Copolymere synthetisiert, in verschiedenen Größen aufgetrennt und mittels optischer Spektroskopie untersucht. Mittels EEI2D-Experimenten wurde die Exzitonendynamik in Abhängigkeit von der Kettenlänge eingehender untersucht. Hierbei wird eine steigende, aber relativ abnehmende Kohärenzlänge bestimmt, die Auswirkungen auf die Exzitonendynamik hat. Der Exzitonentransport weist erst wellenförmiges und dann subdiffuses Verhalten auf.
Kohlenhydrat-Protein-Wechselwirkungen sind häufig entscheidend beteiligt an verschiedenen einer Infektion oder malignen Erkrankung zugrunde liegenden molekularen Erkennungs-prozessen, die zu Adhäsion, Zell-Zell-Interaktion sowie Immunreaktion und -toleranz führen. Trotz der hohen Relevanz für Diagnostik und Therapie dieser Erkrankungen sind die betreffenden Strukturen und Mechanismen bisher nur ungenügend untersucht und verstanden. Ziel dieser stark interdisziplinär angelegten Arbeit war es daher, Methoden der Fachbereiche Chemie und Pharmazie, Biologie und Medizin, aber auch Physik zu kombinieren, um Kohlenhydraterkennungsprozesse im Detail zu untersuchen und auf dieser Basis strukturell neuartige diagnostische und therapeutische Anwendungen zu entwerfen.
Die hochkomplexe Zusammensetzung einer Zelloberfläche wurde zunächst auf ihren Glycan-anteil reduziert und stark vereinfacht auf der Oberfläche sogenannter Glycochips imitiert. Die verwendeten Systeme auf Basis einer Gold- bzw. Glasoberfläche ergänzen sich optimal in ihrer Eignung für komplementäre analytische Methoden wie Massenspektrometrie sowie quantifizierbare Fluoreszenzspektroskopie.
Der Übergang auf die lebende Zelloberfläche gelang mit Hilfe des Metabolic Glyco-engineering, das die kovalente Präsentation definierter Motive durch eine Cycloaddition zwischen zwei bioorthogonalen Reaktionspartnern (z.B. Azid und Alkin) ermöglicht.
Auf diese Weise wurden in Zusammenarbeit mit der Arbeitsgruppe Sauer (Universität Würzburg) zunächst die Dichte und Verteilung verschiedener Oberflächenglycane auf humanen Zellen mittels hochauflösender Fluoreszenzmikroskopie (dSTORM) bestimmt. Diese Parameter zeigten im Modell des Glycochips einen entscheidenden Einfluss auf Bindungsereignisse und multivalente Erkennung und zählen auch auf natürlichen Zelloberflächen – in engem Zusammenhang mit der lateralen und temporalen Dynamik der Motive – zu den wichtigen Faktoren molekularer Erkennungsprozesse.
Die gezielte Modifikation zellulärer Oberflächenglycane eignet sich aber auch selbst als Methode zur Beeinflussung molekularer Wechselwirkungsprozesse. Dies wurde anhand des humanpathogenen Bakteriums S. aureus gezeigt, dessen Adhäsion auf Epithelzellen der Blasenwand durch Metabolic Glycoengineering partiell unterdrückt werden konnte.
In einem ergänzenden Projekt wurden zwei potentielle Metabolite eines konventionellen Antibiotikums – des Nitroxolins – mit bakteriostatischer sowie antiadhäsiver Wirksamkeit dargestellt. Diese dienten als Referenzsubstanzen zur Verifizierung der postulierten Struktur der Derivate, werden aber auch selbst auf ihr Wirkprofil hin untersucht. Gleichzeitig stehen sie zusammen mit der Grundverbindung zudem als Referenz für die Wirkstärke potentieller neu entwickelter Antiadhäsiva zur Verfügung.
It is demonstrated that the di‐\(\pi\)‐methane (DPM) rearrangement of carbonyl‐substituted dibenzobarrelene (9,10‐dihydro‐9,10‐ethenoanthracene) derivatives is induced by visible‐light‐induced triplet photosensitization with Ir(ppy)\(_{3}\), Ir(dFppy)\(_{3}\) or 1‐butyl‐7,8‐dimethoxy‐3‐methylalloxazine as catalysts, whereas derivatives that lack carbonyl substituents are photoinert under these conditions. Notably, the products are formed almost quantitatively.

In der vorligenden Arbeit wurden Vanadium-abhängige Bromidperoxidase- (VBPO-) Modelle zur stereoselektiven Synthese funktionalisierter O-Heterocyclen entwickelt, die durch Vanadium-katalysierte Oxygenierung von Bishomoallylalkoholen mechanistisch untersucht wurden. Weiterhin wurden Bromcyclisierungen von Bishomoallylalkoholen auf enzymatischem (VBPO), oxidativem und radikalischem Weg für Referenzprodukte einer neuen Variante der Bromcyclisierung durch Vanadium-katalysierte Bromidoxidation durchgeführt. Die Selektivitätsmuster aus den Synthesen ß-hydroxylierter und ß-bromierter Tetrahydrofurane wurden anschließend innerhalb einfacher Naturstoffsynthesen genutzt. Anhand eigener Vorarbeiten wurden neue Vanadium(V)-Komplexe aus Triethoxyvanadat mit tridentaten Schiffbaseliganden, basierend auf Salicylaldehyd und Aminoalkoholen mit strukturell unterschiedlichen Seitenketten in quantitativen Ausbeuten synthetisiert und charakterisiert (51V-NMR, UV und IR). In Test-Cyclisierungen unterschiedlicher Bishomoallylalkohole eignete sich VO(salin)(OEt) mit hohen Umsätzen und guten Regio- wie Stereoselektivitäten am besten. Die relative Geschwindigkeitskonstante (krel = 120±20) der Vanadium-katalysierten Oxidation des Testsystems konnte über Konkurrenzkinetik (Alkenol versus Alken) ermittelt werden und weist deutlich auf eine Bindung des Alkenols an Vanadium während der Oxidation hin. Um die Regio- und Stereoselektivitäten Vanadium-Schiffbase-katalysierter Oxidationen von Bishomoallylalkoholen verstehen zu können, wurden stereochemische Studien anhand des Testsystems durchgeführt. Dessen Oxidation wird demnach im selektivitätsbestimmenden Schritt dem Metallzentrum abgewandt in like-Position bevorzugt gebildet und führt zu dem cis-konfigurierten Tetrahydrofuran als Hauptprodukt. Im Folgenden wurden Vanadium-katalysierte Oxidationen unterschiedlich substituierter bishomoallylischer Alkohole durchgeführt, sämtliche Oxidationen führten regioselektiv zu Tetrahydrofuranen als Hauptprodukte, die Oxygenierung Dimethyl-substituierter Pentenole lieferte durch Substitution an C-1 selektiv cis-konfigurierte Tetrahydrofurane, 2-Substitution führte ebenso wie 3-Substitution zu trans-konfigurierte Oxolanen. Alkohole nicht aktivierter Olefine wurden in der Reihenfolge C-1 ? C-3 mit höherer Selektivität zu trans-konfigurierten Tetrahydrofuranen gebildet. Die Regio- und Stereoselektivitäten der radikalischen Bromcyclisierungen folgen den schon in früheren Arbeiten unserer Arbeitsgruppe aufgestellten Richtlinien (5-exo-trig; 2,5-trans, 2,4-cis und 2,3-trans). Die ionischen Bromcyclisierungen 5,5-dimethylierter Bishomoallyl-alkohole mittels NBS verliefen komplementär zu den Radikal-Cyclisierungen regioselektiv und in Abhängigkeit der Phenylsubstituenten an C-1 - C-3 stereoselektiv zu den 2,5-trans-, 3,5-cis- und 4,5-trans-konfigurierten Tetrahydropyranen. Aus der Bromcyclisierung prochiraler Pentenole in Gegenwart eines Acetonpulvers aus Ascophyllum nodosum (A.n.A.P.) konnte das b-bromierte Tetrahydrofuran racemisch in 87proz. Ausbeute erhalten werden. Ebensowenig führte der Einsatz chiraler Liganden in der Vanadium-katalysierten Oxygenierung prochiraler Penten-1-ole bei guten Ausbeuten (>80%) zu Enantiomeren-angereicherten Tatrahydrofuranen. Innerhalb einfacher Naturstoffsynthesen wurden cis-Pityol, Linalooloxid sowie (-)-epi-Bisabololoxid selektiv unter Standardbedingungen der Vanadium-Schiffbase-katalysierten Oxidationen mit VO(salin)(OEt) und TBHP dargestellt. Die Stereoselektivitäten steigen proportional zu dem Größenunterschied der Substituenten an Position 1 der Bishomoallylalkohole. Abschließend wurde durch Vanadium-katalysierte Bromidoxiadtion mit TBHP eine neue dreistufige Totalsynthese der vier "natürlichen" Muscarin-Isomere ausgearbeitet. Die Gesamtausbeuten dieser Synthesen liegen zwischen 3.0 und 19.9%.
Carbon-13 shieldings and one-bond \(^{13}\)C-H coupling constants of bicydo[2.1.1]hexane, bicydo[2.l.l]hex- 2-ene, tricydo[3.1.1.0\(^{2.4}\)]heptane and benzvalene are presented and compared. to the data of related. compounds. H a bicydo[3.1.0]hexane system is part of a rigid skeleton, the cydopropane ring exerts spedfk: 'Y substituent eflects of two ldnds. In the case of the bicyclobexane boat form an upfield shift of the C-3 signal is observed and in tbe esse of the chair form a downfield shift of 15-20 ppm. Compared to the corresponding cydopentanes the double bond in strained cydopentenes causes downfield shifts of the C-4 absorption. 1bis eftect increases witb increasing strain, reaching 8 45.9 ppm maximum in benzvalene. Hence it is tbe only known bicydo[l.l.O]butane baving 8 reversed order of carbon shieldings. The downfield shifts are e:xplained by means of simple orbital interaction schemes.
Gegenstand der vorliegenden Arbeit ist die Untersuchung der Typ-, Regio- und Stereoselektivität photochemischer Reaktionen in organisierten Medien. Es wird anhand ausgewählter Beispiele gezeigt, daß bei der Festkörperbestrahlung von kronenetherverknüpften Substraten sowie von photoaktiven Verbindungen, die mit Polyaminosäuren, Cyclodextrinen oder Zeolithen assoziiert sind, häufig andere Produkte gebildet und höhere Selektivitäten erreicht werden als in Lösung.
In dieser Arbeit wurde ein geeigneter Zugang zu TBTQ-Derivaten erarbeitet, der eine paarweise Funktionalisierung der aromatischen Positionen in direkter Nachbarschaft zum Triquinacen-Kern zulässt. Hierfür wurde sowohl die doppelte Cyclodehydratisierungsroute als auch die dreifache Cyclisierungsvariante zur Darstellung von Tribenzotriquinacenen dahingehend modifiziert, dass die Einführung der „ortho“-Substituenten bereits zu Beginn der Synthese mit den Edukten erfolgte, da eine nachträgliche Funktionalisierung der gewünschten Positionen am TBTQ aufgrund sterischer Faktoren nicht möglich ist. Es wurden unter anderem die ersten literaturbekannten „ortho“-substituierten TBTQ-Derivate erhalten und vollständig charakterisiert.
Zur Erweiterung des aromatischen Systems wurden außerdem Benzodiphenanthrenotriquinacen-Vorstufen synthetisiert, die im Falle der doppelten Cyclodehydratisierungsroute Acenapthylen-Derivate lieferten. Benzodiphenanthrenotriquinacen konnte mit Hilfe der dreifachen Cyclisierungsvariante synthetisiert werden.
Die Zielmoleküle sollen als Vorstufen für überbrückte aromatisch anellierte Triquinacene dienen.
Ziel dieser Arbeit war die Synthese geeigneter Modellsysteme für die Untersuchung gekrümmter Graphenfragmente. Dafür bot sich die Synthese von substituierten Pyracylenen an. Diese sollten in eine Aromatensphäre eingebunden sein, um so den Einfluss der Graphenumgebung zu simulieren. Ein geeignetes einfaches Untersuchungsobjekt stellt das Bisbenzo[d,j]pyracylen/ Dicyclopenta[fg,op]tetracen (10) dar. Auch die Einbettung in größere „Nanographene“ ist ein lohnendes Syntheseziel zur Untersuchung von Struktur-Eigenschaftsbeziehungen. Gegebenenfalls kann nach erfolgreicher Darstellung auch das Auftreten der Stone-Wales-Umlagerung untersucht werden. Das heißt, die Systeme sollen mit Hilfe der FVP auf ihr Vermögen zur Kohlenstoffgerüstumlagerung untersucht werden. Substituierte Tetrahydrofurane sind Bausteine vieler pharmakologisch interessanter Natur- und Wirkstoffe. Deshalb ist die Entwicklung effizienter Methoden zur Darstellung dieser Verbindungsklasse von großer Bedeutung. Bei den Übergansmetall-katalysierten Verfahren erwiesen sich sowohl Vanadium(V)-, als auch Cobalt(II)-Komplexe als besonders vielversprechende Oxidationskatalysatoren. Bis dato ist allerdings kein universelles Verfahren bekannt, mit welchem nur durch Wahl der Reaktionsbedingungen ein Substrat unter vollständiger Regio- und Stereokontrolle selektiv in einen definierten cyclischen Ether umgewandelt werden kann. Ziel dieser Arbeit war daher die Ausarbeitung von Vanadium(V)- und Cobalt(II)-Komplex-katalysierten Oxidationen zur selektiven Synthese substituierter Tetrahydrofurane. Die Reaktionen wurden ausgehend von Bishomoallylalkoholen entwickelt, optimiert und mechanistisch untersucht. Für die Vanadium(V)-katalysierten Oxidationen wurden Reaktionen evaluiert, die nach dem Peroxo- (H2O2 als Primäroxidans) und solche, die nach dem Peroxy-Mechanismus (TBHP als Primäroxidans) verlaufen. Bei den Cobalt(II)-katalysierten Oxidationen wurde molekularer Sauerstoff als Primäroxidans eingesetzt. Im Zentrum des Interesses stand weiterhin die breit angelegte Synthese neuartiger Komplexe, welche auf ihre Eignung als Katalysatoren hin überprüft wurden. Die Metallionen wurden durch strukturell unterschiedliche Liganden stabilisiert, die auf Grund ihrer sterischen und elektronischen Natur Reaktivitäts- und Selektivitätseinflüsse bei den oxidativen Cyclisierungen ausüben sollten. Hierbei kamen Picolin- und Hydroxamsäure-, Diketonat-, sowie macrocyclische Diamin-Schiffbase-Liganden zum Einsatz. In Test-Oxidationen unterschiedlicher Bishomoallylalkohole zur Optimierung der Reaktionsbedingungen hinsichtlich Solvens, Primäroxidans und Reaktionstemperatur erwiesen sich für Vanadium-katalysierte Oxidationen TBHP in Dichlormethan und H2O2 in tert-Butanol bei jeweils 25°C am geeignetsten. Für Cobalt(II)-katalysierte Oxidationen konnten molekularer Sauerstoff als Primäroxidans, Isopropanol als Solvens und eine Reaktionstemperatur von 60°C als Reaktionsparameter der Wahl ermittelt werden. Im Folgenden wurden Vanadium(V)-katalysierte Oxidationen unterschiedlich substituierter bishomoallylischer Alkohole unter optimierten Reaktionsbedingungen durchgeführt. Sämtliche Oxidationen führten regioselektiv zu Tetrahydrofuranen als Hauptprodukte. Die gewonnenen Erkenntnisse zur Vanadium(V)-katalysierten Darstellung substituierter Tetrahydrofurane wurden im Rahmen der Synthese eines Naturstoff-Derivats angewandt. Ziel war es dabei in einer vierstufigen Totalsynthese Derivate der natürlich vorkommenden Carboline Cyclocapitellin und Isocyclocapitellin zu synthetisieren. Die auf diese Weise erhaltenen enantiomerenreinen Indolalkaloide werden zur Zeit pharmakologischen Testungen unterzogen. In einem weiteren Projekt wurden aerobe Cobalt(II)-katalysierte Oxidationen unterschiedlich substituierter Bishomoallylalkohole durchgeführt. Wie bei den Vanadium-katalysierten Reaktionen konnten auch hier regioselektiv Tetrahydrofurane als Hauptprodukte erhalten werden. Abschließend wurden durch Oxidation prochiraler Alkenole in Gegenwart chiraler Cobalt(II)-Diketonat-Komplexe enantiomerenangereicherte Tetrahydrofurane dargestellt. Diese Oxidationen lieferten nur geringe Enantioselektivitäten, welche durch Verwendung von Propionaldehyddiethylacetal als Solvens gesteigert werden konnten. Zusammenfassend kann festgestellt werden, dass sich für Vanadium(V)- und Cobalt(II)-katalysierte Oxidationen, im Hinblick auf die Substituenten in unmittelbarer Nähe der Alkenol-Doppelbindung, ein komplementäres Reaktivitäts- und Selektivitätsbild ergibt. So werden bei Cobalt(II)-katalysierten Oxidationen Alkenole ohne terminale Methylgruppen in höheren Ausbeuten und Stereoselektivitäten zu Tetrahydrofuranen oxidiert, bei Vanadium(V)-Katalysen hingegen 5,5-Dimethyl-substituierte Alkenole.
Die kausalen Ursachen, die zur Auslösung der neurodegenerativen Erkrankung Morbus Parkinson führen, sind noch immer unklar. Man nimmt heute an, daß das Absterben dopaminerger Neurone im Mittelhirn von Parkinsonpatienten multifaktoriell ausgelöst wird. Genetische Prädisposition sowie endogene und exogene Umweltgifte wie etwa Substanzen, die strukturelle Ähnlichkeit mit dem bekanntesten dopaminergen Neurotoxin MPTP (1-Methyl-4-phenyl-1,2,3,5-tetrahydropyridin) besitzen, werden als Hauptursachen für die Entstehung des Parkinsonsyndroms diskutiert. Diese Arbeit beschäftigt sich mit einer neuen Klasse von neurotoxisch wirksamen Tetrahydro-b-carbolinen, die sich von Trichloracetaldehyd (Chloral) ableiten. Die wohl prominenteste Verbindung in dieser Reihe ist TaClo (1-Trichlormethyl-1,2,3,4-tetrahydro-b-carbolin), das im menschlichen Körper nach Aufnahme des Schlafmittels Chloralhydrat durch Pictet-Spengler-Kondensation mit dem endogen vorhandenen Tryptamin gebildet wird. Zusätzlich scheint die Bildung von TaClo aus dem Industrielösungsmittel TRI (Trichlorethylen), das im Organismus zu Chloral metabolisiert wird, möglich. Die über Chloral eingeführte große CCl3-Gruppe erhöht die Lipophilie von TaClo, die Passage der Blut-Hirn-Schranke ist erleichtert. In der Tat haben zahlreiche Untersuchungen in vitro und in vivo gezeigt, daß TaClo toxische Prozesse in dopaminergen und serotonergen Systemen zu induzieren vermag. Ein wesentliches Ziel dieser Arbeit bestand darin, die in-vivo-Entstehung und Metabolisierung von TaClo im Menschen sowie den Einfluß dieses Neurotoxins auf die DNA herauszuarbeiten und näher zu untersuchen. Im einzelnen wurden folgende Ergebnisse erzielt: TaClo schädigt die DNA, wie Versuche an zellfreier DNA und in-vitro-Experimente an PC12-Zellen belegen. Die endogene Bildung von TaClo in Chloralhydrat-behandelten Patienten und die in-vitro-Entstehung von TaClo aus Trichlorethylen wurde mittels HPLC-ESI-MS-MS-Analytik eindeutig bewiesen. Außerdem wurden erste Hinweise auf eine Anreicherung des Neurotoxins im menschlichen Körper erhalten. Stereostrukturelle Aspekte der Bildung und Verstoffwechslung von TaClo wurden aufgeklärt und TaClo-Metabolite in in-vitro- und in-vivo-Proben identifiziert. Das Tetrahydro-b-carbolin entsteht in racemischer Form, wird also nicht enzymatisch sondern spontan durch Pictet-Spengler-Reaktion gebildet. Zusätzlich wurden Hinweise auf eine enzymatische Metabolisierung von TaClo gefunden. Außerdem gelang es, eine etablierte Methode des oxidativen Abbaus zur Aufklärung der absoluten Konfiguration von Tetrahydropyridin-Heterocyclen [z.B. Eleagnin] auf Substanzen mit benzylischer Hydroxy- oder Metylether-Gruppe zu erweitern. Geringe Mengen (1-2 mg) an Substanzen, die in benzylischer Position eine chirale Sauerstoffunktion besitzen, wurden durch Ruthenium-katalysierte Oxidation zu GC-gängigen Säuren abgebaut, deren absolute Konfiguration nach Trennung an chiraler Phase durch Vergleich mit enantiomerenreinem Referenzmaterial bestimmt wurde. Diese Zuordnung erlaubte den Rückschluß auf die absolute Konfiguration einer Reihe Ausgangsverbindungen.
Die Infektion mit dem Masernvirus (MV) stellt weltweit immer noch ein großes Problem dar. Trotz des vorhandenen Lebendimpfstoffs, der eine Erkrankung sicher zu verhindern vermag, haben nicht nur die Entwicklungsländer, in denen ein flächendeckender Impfschutz schwieriger zu erreichen ist, mit der Erkrankung und ihren Komplikationen zu kämpfen. Hat sich die Erkrankung klinisch manifestiert gibt es keine kausalen Therapiemöglichkeiten und es kann nur noch symptomatisch behandelt werden. Dies ist v.a. auch in Hinblick auf die schweren Komplikationen der Maserninfektion von Bedeutung. Bei Erstkontakt mit dem Masernvirus ist die Suszeptibilität nicht geimpfter Menschen sehr hoch. Das bedeutet, dass es in 95-98 % der Fälle nach einer Infektion mit dem Masernvirus auch zum klinischen Bild der Masern kommt, unabhängig von Alter und Geschlecht. Das Ziel dieser Arbeit war es daher, potentielle Hemmstoffe der Maserninfektion auf ihre Wirkung zu testen und zu verstehen, wo im Infektions- und Replikationszyklus des MV sie eingreifen. Es wurden eine Reihe Substanzen mit potentiell-inhibitorischen Eigenschaften in Infektions-Hemmtests und im Zytotoxizitätstest untersucht, von denen im Anschluss die drei besten Inhibitoren (JK80, QD6-8 und Droseron) weiter untersucht wurden. JK80 und QD6-8 waren beide mit IC50-Werten um 30 µM und SI-Werten von über 2 nur mäßig spezifisch antiviral wirksam. Während JK80 vermutlich den Eintritt des MV in die Zellen verhindert, hemmt QD6-8 die intrazelluläre Virusreplikation und wäre im Hinblick auf die Entwicklung neuartiger, spezifischer Medikamente gegen die Maserninfektion von grossem Interesse. Eine Zielmolekülanalyse der Substanz und die Testung anderer Derivate könnten Aufschluss darüber geben, wie Substanzen aussehen müssten, die eine spezifische Hemmung der intrazellulären Replikation bewirken können. Der Naturstoff Droseron könnte mit einer spezifischen Hemmung (IC50 ca. 10 µM; SIWert 6 im Fluoreszenzreader, bzw. IC50 ca. 2 µM; SI-Wert 30 in der Titration) eine mögliche Leitsubstanz für einen neuen MV-Inhibitor darstellen. Allerdings waren alle bisher getesteten Droseron-Derivate entweder weniger inhibitorisch wirksam oder deutlich zytotoxischer als Droseron selbst. Die Ergebnisse der Infektionshemmversuche mit Zugabe von Droseron vor, während oder nach der Infektion mit MV sprechen dafür, dass Droseron den Eintritt des Virus in die Zelle stört.
Oligophenyleneethynylenes (OPEs) are prominent building blocks with exciting optical and supramolecular properties. However, their generally small spectroscopic changes upon aggregation make the analysis of their self-assembly challenging, especially in the absence of additional hydrogen bonds. Herein, by investigating a series of OPEs of increasing size, we have unravelled the role of the conjugation length on the self-assembly properties of OPEs.
Polymer micelles are an attractive means to solubilize water insoluble compounds such as drugs. Drug loading, formulations stability and control over drug release are crucial factors for drug‐loaded polymer micelles. The interactions between the polymeric host and the guest molecules are considered critical to control these factors but typically barely understood. Here, we compare two isomeric polymer micelles, one of which enables ultra‐high curcumin loading exceeding 50 wt.%, while the other allows a drug loading of only 25 wt.%. In the low capacity micelles, steady‐state fluorescence revealed a very unusual feature of curcumin fluorescence, a high energy emission at 510 nm. Time‐resolved fluorescence upconversion showed that the fluorescence life time of the corresponding species is too short in the high‐capacity micelles, preventing an observable emission in steady‐state. Therefore, contrary to common perception, stronger interactions between host and guest can be detrimental to the drug loading in polymer micelles.
In der vorliegenden Dissertation wurde die Selektivität der Oxidation von chiralen oxazolidinonsubstituierten Encarbamten I mit Singulettsauerstoff, Dimethyldioxiran (DMD) und meta-Chloroperbenzoesäure (mCPBA) untersucht. Durch die Variation der Encarbamatstruktur wurde die Typselektivität (En-Reaktion versus [2+2]-Cycloaddition) kontrolliert. Bei der [2+2]-Cycloaddition der Encarbamate I mit Singulettsauerstoff (1O2) und der Epoxidierung mit DMD und mCPBA wurden hohe Diastereoselektivitäten beobachtet. Die folgenden Ergebnisse wurden erzielt: 1. Die Typselektivität (En-Reaktion versus [2+2]-Cycloaddition) der Reaktion von 1O2 mit den Encarbamaten Ia-c ist in Tabelle I aufgeführt. Daraus ist ersichtlich, dass die E-Isomere vorzugsweise die En-Reaktion eingehen, während die Z-Isomere die [2+2]-Cycloaddition bevorzugen. Diese diastereomerenabhängige Typselektivität wird durch den orbitaldirigierenden Effekt des vinylischen Stickstoffatoms verständlich. Der Angriff des Singulettsauerstoffs wird durch die vorteilhafte bindende Orbitalwechselwirkung zwischen dem LUMO des Singulettsauerstoffs und dem HOMO des Encarbamates Ib auf die Seite der Stickstofffunktionalität gesteuert. Da im Encarbamat Z-I kein cis-ständiges allylisches Wasserstoffatom für den angreifenden 1O2 zur Verfügung steht, findet vorzugsweise die [2+2]-Cycloaddition mit hoher Diastereoselektivität (> 95:5) statt. Bei den Encarbamaten E-I sind die Stickstofffunktionalität und die allylische Methylgruppe auf der Seite der Doppelbindung angeordnet, die von 1O2 angegriffen wird, so dass der sich annähernde Singulettsauerstoff ein allylisches Wasserstoffatom von der Methylgruppe abstrahiert und eine hohe Typselektivität für die En-Reaktion beobachtet wird. 2. In der [2+2]-Cycloaddition der Encarbamate Id-e mit Singulettsauerstoff, dem kleinstmöglichen Enophil, wird eine vollständige Diastereoseitendifferenzierung (d.r. >95:5) durch die Oxazolidinonkonfiguration bestimmt. Der R1-Substituent des Oxazolidinonrings besetzt aufgrund konformationeller Effekte einen Halbraum der Doppelbindung (Abbildung A), so dass der bevorzugte -faciale Angriff alleine durch die sterische Abschirmung des R1-Substituenten kontrolliert wird, solange R1 ≠ H. Die Gegenwart eines weiteren stereogenen Zentrums im allylischen Substituenten der Encarbamate beeinflusst die Richtung des Singulettsauerstoffangriffs nicht: Der Angriff des Singulettsauerstoffs findet von derselben -Seite statt, unabhängig davon, ob (4R,3’S)-Ie oder (4R,3’R)-Ie als Substrat eingesetzt wird. Offensichtlich rufen (R)-konfigurierte R1-Substituenten die (S)-Konfiguration der zwei neu gebildeten stereogenen Zentren im resultierenden Dioxetan hervor. 3. Mit den chiralen Oxazolidinonauxiliaren kann nicht nur die Diastereoselektivität der [2+2]-Cycloaddition von Singulettsauerstoff gesteuert werden, sondern auch die der En-Reaktion. Die Konfiguration der Doppelbindung der Encarbamate spielt in der En-Reaktion eine entscheidende Rolle. Im Fall der Isomere Z-Ia, b hängt die Diastereoselektivität stark von der Größe des R1-Substituenten am Oxazolidinon ab. Dies ist verständlich, denn der Angriff des Singulettsauerstoffs muß auf der Seite der allylischen Methylgruppe erfolgen, um die En-Produkte zu erhalten (Abbildung B). Für kleinere R1-Substituenten, z.B. Methyl (Tabelle 1), wird diese Seite der Doppelbindung im Fall der Substrate Z-1 weniger effektiv abgeschirmt als für die räumlich anspruchsvollere Phenylgruppe. Für letztere wird das unlike En-Produkt daher in hoher Diastereoselektivität erhalten Im Fall der Substrate E-I, ist der Oxazolidinonring auf derselben Seite der Doppelbindung wie die allylische Methylgruppe. Daher treten intensivere sterische Wechselwirkungen des R1-Substituenten mit Singulettsauerstoff auf und sorgen für die höhere Diastereoselektivität als bei den Diastereomeren Z-I. 4. Bei der Epoxidierung der Encarbamate Ic, f wird ebenfalls eine hohe Diastereo-selektivität beobachtet, diese ist jedoch weniger ausgeprägt als bei der Photooxygenierung derselben Substrate. Zudem wird in diesem Fall nicht nur die Richtung, sondern auch das Ausmaß der Diastereo-selektivität durch den R1-Substituenten im Oxazolidinonring beeinflusst. Deshalb wird für den Sauerstoff-Transfer der Mechanismus in Abbildung A vorgeschlagen. Wenn X = H, sind die sterischen Wechselwirkungen für den (1’Re,2’Re)- und den (1’Si,2’Si)-Angriff vergleichbar und daher ist die Diastereoselektivität niedrig (60:40). Für X = CH3 ist die Doppelbindung vollständig abgeschirmt, wodurch der (1’Re,2’Re)-Angriff erschwert wird und die Diastereoselektivität steigt deutlich (93:7). Weiterhin ist das Ausmaß der diastereo-facialen Differenzierung nicht nur von der Größe von R1 abhängig, sondern auch von der Konfiguration der Doppelbindung. Die Epoxidierung des Z-Isomers ist wegen der besser abgeschirmten Doppelbindung der Z-Encarbamate immer selektiver als die des E-Isomers.
The chirality of the interlocked bay-arylated perylene motif is investigated upon its material prospect and the enhancement of its chiroptical response to the NIR spectral region. A considerable molecular library of inherently chiral perylene bisimides (PBIs) was utilized as acceptors in organic solar cells to provide decent device performances and insights into the structure-property relationship of PBI materials within a polymer blend. For the first time in the family of core-twisted PBIs, the effects of enantiopurity on the device performance was thoroughly investigated. The extraordinary structural sensitivity of CD spectroscopy served as crucial analytical tool to bridge the highly challenging gap between molecular properties and device analytics by proving the excitonic chirality of a helical PBI dimer. The chirality of this perylene motif could be further enhanced on a molecular level by both the expansion and the enhanced twisting of the π-scaffold to achieve a desirable strong chiroptical NIR response introducing a new family of twisted QBI-based nanoribbons. These achievements could be substantially further developed by expanding this molecular concept to a supramolecular level. The geometrically demanding supramolecular arrangement necessary for the efficient excitonic coupling was carefully encoded into the molecular design. Accordingly, the QBIs could form the first J-type aggregate constituting a fourfold-stranded superhelix of a rylene bisimide with strong excitonic chirality. Therefore, this thesis has highlighted the mutual corroboration of experimental and theoretical data from the molecular to the supramolecular level. It has demonstrated that for rylene bisimide dyes, the excitonic contribution to the overall chiroptical response can be designed and rationalized. This can help to pave the way for new organic functional materials to be used for
chiral sensing or chiral organic light-emitting devices.
Exciton coupling between two or more chromophores in a specific environment is a key mechanism associated with color tuning and modulation of absorption energies. This concept is well exemplified by natural photosynthetic proteins, and can also be achieved in synthetic nucleic acid nanostructures. Here we report the coupling of barbituric acid merocyanine (BAM) nucleoside analogues and show that exciton coupling can be tuned by the double helix conformation. BAM is a nucleobase mimic that was incorporated in the phosphodiester backbone of RNA, DNA and GNA oligonucleotides. Duplexes with different backbone constitutions and geometries afforded different mutual dye arrangements, leading to distinct optical signatures due to competing modes of chromophore organization via electrostatic, dipolar, - stacking and hydrogen-bonding interactions. The realized supramolecular motifs include hydrogenbonded BAM–adenine base pairs and antiparallel as well as rotationally stacked BAM dimer aggregates with distinct absorption, CD and fluorescence properties.

In T cells, as in all other cells of the body, sphingolipids form important structural components of membranes. Due to metabolic modifications, sphingolipids additionally play an active part in the signaling of cell surface receptors of T cells like the T cell receptor or the co-stimulatory molecule CD28. Moreover, the sphingolipid composition of their membranes crucially affects the integrity and function of subcellular compartments such as the lysosome. Previously, studying sphingolipid metabolism has been severely hampered by the limited number of analytical methods/model systems available. Besides well-established high resolution mass spectrometry new tools are now available like novel minimally modified sphingolipid subspecies for click chemistry as well as recently generated mouse mutants with deficiencies/overexpression of sphingolipid-modifying enzymes. Making use of these tools we and others discovered that the sphingolipid sphingomyelin is metabolized to ceramide to different degrees in distinct T cell subpopulations of mice and humans. This knowledge has already been translated into novel immunomodulatory approaches in mice and will in the future hopefully also be applicable to humans. In this paper we are, thus, summarizing the most recent findings on the impact of sphingolipid metabolism on T cell activation, differentiation, and effector functions. Moreover, we are discussing the therapeutic concepts arising from these insights and drugs or drug candidates which are already in clinical use or could be developed for clinical use in patients with diseases as distant as major depression and chronic viral infection.
The precise interplay between the mRNA codon and the tRNA anticodon is crucial for ensuring efficient and accurate translation by the ribosome. The insertion of RNA nucleobase derivatives in the mRNA allowed us to modulate the stability of the codon-anticodon interaction in the decoding site of bacterial and eukaryotic ribosomes, allowing an in-depth analysis of codon recognition. We found the hydrogen bond between the N1 of purines and the N3 of pyrimidines to be sufficient for decoding of the first two codon nucleotides, whereas adequate stacking between the RNA bases is critical at the wobble position. Inosine, found in eukaryotic mRNAs, is an important example of destabilization of the codon-anticodon interaction. Whereas single inosines are efficiently translated, multiple inosines, e.g., in the serotonin receptor 5-HT2C mRNA, inhibit translation. Thus, our results indicate that despite the robustness of the decoding process, its tolerance toward the weakening of codon-anticodon interactions is limited.
In this thesis, the usage of onion-like carbon (OLC) for energy storage applications was researched regarding sustainability, performance and processability. This work targets to increase the scientific understanding regarding the role of OLC in electrodes and to facilitate a large-scale production, which is the foundation for commercial application. Research was devoted to increase the knowledge in the particular field, to yield synergistic approaches and a shared value regarding sustainability and performance.
Tropische Infektionskrankheiten sind noch immer die Haupttodesurache in vielen Ländern der Dritten Welt. Unter ihnen ist Malaria neben der Immunschwächekrankheit AIDS und Tuberkulose am weitesten verbreitet. Laut WHO erkrankten allein im Jahr 2016 rund 216 Millionen Menschen an Malaria und weltweit verstarben 445.000 Menschen an den Folgen dieser Infektion. Solange die Wirksamkeit des Impfstoffs RTS,S/AS01 gegen Malaria noch Schwachstellen aufweist und andere Impfstoff-Kanditaten sich noch in präklinischen Testphasen befinden, ist vor allem die Entwicklung neuer Wirkstoffe, auch im Hinblick auf die rasante Ausbreitung von Resistenzen gegen herkömmliche Medikamente, weiterhin eine dringende Aufgabe.
Eine vielversprechende Wirkstoffklasse mit interessanten strukturellen Eigenschaften und einer ungewöhnlichen Biosynthese aus Acetat-Einheiten sind die Naphthylisochinolin-Alkaloide aus den beiden paläotropischen Pflanzenfamilien der Dioncophyllaceae und Ancistrocladaceae. Die Naphthylisochinolin-Alkaloide sind hervozuheben aufgrund ihrer exzellenten Aktivität gegen Plasmodium falciparum, den Erreger der Malaria tropica, sowie wegen ihrer Wirksamkeit gegen Erreger weiterer Krankheiten wie beispielsweise AIDS, Leishmaniose und Afrikanische Trypanosomiasis. Auch im Kampf gegen maligne Erkrankungen wie Leukämie und pankreatischen Krebs werden sie aufgrund ihrer cytotoxischen Eigenschaften als vielversprechende Leitstrukturen betrachtet.
Die strukturell beeindruckendsten Naphthylisochinolin-Alkaloide sind die dimeren Mbandakamine, die von unserer Arbeitsgruppe vor einiger Zeit aus einer kongolesischen Ancistrocladus-Liane isoliert wurden. Sie besitzen sieben stereogene Elemente und sind die ersten natürlich vorkommenden Dimere mit einer höchst unsymmetrischen 6',1''-gekuppelten zentralen Biarylachse. Diese impliziert eine außergewöhnlich hohe sterische Hinderung an der zentralen Achse, wie sie noch in keinem anderen dimeren Naphthylisochinolin-Alkaloid gefunden wurde. Verbunden mit ihren bemerkenswerten und vielseitigen pharmakologischen Wirkeigenschaften sind sie ausgesprochen interessante Moleküle für eine synthetische Erschließung.
Ziel dieser Arbeit war die erstmalige Totalsynthese von Mbandakamin A und B sowie die Synthese ihrer monomeren Hälften 5-epi-Korupensamin E und 8-O-Methylkorupensamin A. Zudem sollten weitere Naphthylisochinolin-Dimere, die bei der Synthese der Mbandakamine anfallen, isoliert und charakterisiert werden. Alle neuen mono- und dimeren Naphthylisochinoline sollten abschließend am Schweizerischen Tropen- und Public-Health-Institut auf ihre biologische Aktivität getestet werden.
Zusätzlich gelang im Rahmen eines Kooperationsprojekts erstmals die stereochemische Charakterisierung des strukturell ganz neuartigen, inhärent chiralen Wolframbiscorrols durch online HPLC-ECD-Analyse in Kombination mit quantenchemischen Rechnungen.
Die paläotropischen Pflanzenfamilien der Ancistrocladaceae und Dioncophyllaceae sind die bisher einzig bekannten Produzenten von Naphthylisochinolin-Alkaloiden. Diese spezielle Klasse acetogeniner Sekundärmetabolite weist durch die verschiedenen Kupplungspositionen der beiden namensgebenden Molekülbausteine eine breite strukturelle Diversität auf und zeichnet sich durch vielfältige pharmakologische Wirksamkeiten, z.B. antiplasmodiale, antileishmaniale oder antitrypanosomale Aktivitäten, aus. Zur Synthese dieser Naturstoffe wurde im Arbeitskreis Bringmann eigens eine Methodik entwickelt, das Lacton -Konzept. Diese Methode erlaubt durch eine Vorfixierung der beiden Molekülhälften durch eine Esterbrücke, anschließender intramolekularer Kupplungsreaktion und der stereoselektiven Öffnung des erhaltenen Lactons den atropselektiven Aufbau der Naphthylisochinoline. Als Ziele dieser Arbeit ergaben sich somit die Synthese pharmakologisch und strukturell interessanter Naphthylisochinolin-Alkaloide mittels des Lacton-Konzepts sowie die Isolierung und Strukturaufklärung weiterer Sekundärmetabolite aus Triphyophyllum peltatum (Dioncophyllaceae), welche anschließend auf ihre Bioaktivität hin untersucht werden sollten, um potenziell neue Leitstrukturen für neue Wirkstoffe zu finden.
Our research group focusses on the isolation, structural elucidation, and synthesis of bioactive natural products, among others, the naphthylisoquinoline alkaloids from tropical lianas. This intriguing class of compounds comprises representatives with activities against, e.g. P. falciparum, the cause of Malaria tropica, against the neglected disease leishmaniasis, and, as discovered more recently, against different types of cancer cells. Based on the high potency of theses extraordinary secondary metabolites, this thesis was devoted to the total synthesis of bioactive natural products and closely related analogs.
Chemical processes mostly happen in fluid environments where reaction partners encounter via diffusion. The bimolecular encounters take place at a nanosecond time scale. The chemical environment (e.g., solvent molecules, (counter)ions) has a decisive influence on the reactivity as it determines the contact time between two molecules and affects the energetics. For understanding reactivity at an atomic level and at the appropriate dynamic time scale, it is crucial to combine matching experimental and theoretical data. Here, we have utilized all-atom molecular-dynamics simulations for accessing the key time scale (nanoseconds) using a QM/MM-Hamiltonian. Ion pairs consisting of a radical ion and its counterion are ideal systems to assess the theoretical predictions because they reflect dynamics at an appropriate time scale when studied by temperature-dependent EPR spectroscopy. We have investigated a diketone radical anion with its tetra-ethylammonium counterion. We have established a funnel-like transition path connecting two (equivalent) complexation sites. The agreement between the molecular-dynamics simulation and the experimental data presents a new paradigm for ion–ion interactions. This study exemplarily demonstrates the impact of the molecular environment on the topological states of reaction intermediates and how these states can be consistently elucidated through the combination of theory and experiment. We anticipate that our findings will contribute to the prediction of bimolecular transformations in the condensed phase with relevance to chemical synthesis, polymers, and biological activity.
Dipolar merocyanines are very attractive supramolecular building blocks, as they combine interesting functional properties with strong, directional intermolecular interactions. The pyridine dioxocyano-pyridine (PYOP) chromophore (Chapter 2.2), used in this thesis, stands out because of its exceptionally high ground state dipole moment (g ~ 17 D), in combination with the option to retain good solubility also in unpolar solvents, by decoration with solubilizing groups.
The reliable binding motif of anti-parallel -stacking due to dipole-dipole interactions has allowed the design of molecular building blocks that form assemblies of predictable geometry. The intense unstructured charge transfer UV/Vis absorption band (eg ~ 10.7 D) is a result of the dominant contribution of the zwitterionic resonance structure which brings the PYOP chromophore just beyond the cyanine limit in solvents of low polarity (c2 = 0.60, 1,4 dioxane). The high sensitivity of the S0 – S1 UV/Vis absorption band to the environment manifests itself in a pronounced negative solvatochromism and strong H-type exciton coupling within -stacked PYOP assemblies. In accordance with the classical molecular exciton theory, an increasing hypsochromic shift of the dominant absorption band of these H aggregates can be observed as the stack size increases up to about six chromophores, where it levels out at about max ~ 440 nm (CHCl3). This allows a uniquely simple estimation of the number of interacting chromophores within the self-assembled structure from a single UV/Vis absorption spectrum of an aggregate.
The defined and well investigated PYOP dimer formation was employed in this thesis to probe the applicability and limitations of concentration-, temperature-, and solvent-dependent self-assembly studies (Chapter 3). Straightforward theoretical models to evaluate datasets of concentration-, temperature-, and solvent-dependent UV/Vis absorption by nonlinear regression analysis were derived for the case of dimer formation (Chapter 2.1). Although the dimer model is well known and widely applied in literature, this detailed derivation is helpful to understand assumptions and potential problems of the different approaches for the determination of thermodynamic parameters. This helps to decide on the most appropriate method to analyse a system of interest. In this regard it should be noted that covering a large portion of the self-assembly process with the experimental data is a prerequisite for the accuracy of the analysis. Additionally, many of the insights can also be transferred to other self-assembly systems like supramolecular polymerization or host-guest interactions.
The concentration-dependent analysis is the most straightforward method to investigate self-assembly equilibria. No additional assumptions, besides mass balance and mass action law, are required. Since it includes the least number of parameters (only K, if M/D are known), it is the most, or even only, reliable method, to elucidate the self-assembly mechanism of an unknown system by model comparison. To cover a large concentration range, however, the compound must be soluble enough and generally sample amounts at least in the low mg scale must be available.
The temperature-dependent analysis has the advantage that all thermodynamic parameters G0, H0 and S0 can be obtained from a single sample in one automated measurement. However, the accessible temperature-range is experimentally often quite limited and dependent on the solvent. For systems which do not show the transition from monomer to aggregate in a narrow temperature range, as given for, e.g., cooperative aggregation or processes with a high entropy contribution, often not the entire self-assembly process can be monitored. Furthermore, the assumptions of temperature-independent extinction coefficients of the individual species as well as temperature-independent H0 and S0 must be met. Monte Carlo simulations of data sets demonstrated that even minor changes in experimental data can significantly impact the optimized values for H0 and S0. This is due to the redundancy of these two parameters within the model framework and even small thermochromic effects can significantly influence the results. The G0 value, calculated from H0 and S0, is, however, still rather reliable.
Solvent-dependent studies can often cover the entire self-assembly process from monomeric (agg = 0) to the fully aggregated state (agg = 1). However, for dyes with strong solvatochromic effects, such as the dipolar merocyanines investigated in this thesis, the results are affected. Also, the assumption of a linear relation of the binding energy G0 and the fraction of denaturating solvent f, which is based on linear free energy relationships between G0 and the solvent polarity, can lead to errors. Especially when specific solvent effects are involved.
For the evaluation of experimental data by nonlinear regression, general data analysis software can be used, where user-defined fit models and known parameters can be implemented as desired. Alternatively, multiple specialized programs for analysing self-assembly data are available online. While the latter programs are usually more user-friendly, they have the disadvantage of being a “black box” where only pre-implemented models can be used without the option for the user to adapt models or parameters for a specific system.
In Chapter 3 comprehensive UV/Vis absorption datasets are presented for the dimerization of merocyanine derivative 1 in 1,4-dioxane, which allowed for the first time a direct comparison of the results derived from concentration-, temperature-, and solvent-dependent self-assembly studies.
The results for the binding constant K and corresponding G0 from the concentration- and temperature-dependent analysis were in very good agreement, also in comparison to the results from ITC. For the temperature-dependent analysis, though, multiple datasets of samples with different concentration had to be evaluated simultaneously to cover a meaningful part of the self-assembly process. Furthermore, a significant dependence of the optimized parameters H0 and S0 on the wavelength chosen for the analysis was observed. This can be rationalized by the small thermochromic shifts of both the monomer and the dimer UV/Vis absorption band. The results from the solvent-dependent evaluation showed the largest deviation, as expected for the highly solvatochromic merocyanine dye.
However, even here by evaluation at 491 and 549 nm the deviation for G0 was only 2.5 kJ mol1 (9%) with respect to the results from the concentration-dependent analysis (G0 = 29.1 kJ mol1). Thus, despite the strong solvatochromism of the dipolar chromophore, it can still be considered a reliable method for estimating the binding strength. Furthermore, multiple repetitions of the concentration-, temperature-, and solvent-dependent studies provided insight into the reproducibility of the results and possible sources of experimental errors. In all cases, the deviations of the results were small (G0 < 0.4 kJ mol1) and within the same range as the fit error from the nonlinear regression analysis.
The insights from these studies were an important basis for the in-depth investigation of a more complex supramolecular system in Chapter 4, as a single method is often not enough to capture the full picture of a more complicated self-assembly process. To elucidate the anti-cooperative self-assembly of the chiral merocyanine 2, a combination of multiple techniques had to be applied.
Solvent-dependent UV/Vis absorption studies in CH2Cl2/MCH mixtures showed the step-wise assembly of the merocyanine monomer (max(M) = 549 nm, CH2Cl2) to first a dimer (max(D) = 498 nm, CH2Cl2/MCH 15:85) by dipole-dipole interactions, and then a -stacked higher aggregate (max(H) = 477 nm, MCH), with pronounced H-type coupling.
The thermodynamic evaluation of this data, however, suffered from the severe solvatochromism, especially of the monomeric species (max(M, CH2Cl2) = 549 nm, max(M, MCH) = 596 nm). Therefore, concentration-dependent studies were performed at three different temperatures (298, 323, 353 K) to elucidate the self-assembly mechanism and determine reliable thermodynamic parameters. The studies at elevated temperatures were hereby necessary, to obtain experimental data over a larger agg--range. Due to the pronounced difference in the thermodynamic driving force for dimerization and higher aggregate formation (KD/K5 = 6500) a concentration range exists in MCH where almost exclusively the dimer species of 2 is present, before further self-assembly by dispersion interactions occurs. Therefore, the data could be evaluated independently for the two self-assembly steps. The self-assembly of dimers into the higher aggregate could not be described by the isodesmic model but was fitted satisfactorily to a pentamer model. This rather small size of about ten -stacked PYOP chromophores was, furthermore, consistently indicated by AFM, VPO and DOSY NMR measurements. Based on 1D and 2D NMR data as well as the strong bisignate CD signal of the higher aggregate in combination with TD-DFT calculations, a P-helical stack is proposed as its structure. The small size can be rationalized by the anti-cooperative self-assembly mechanism and the sterical demand of the solubilizing trialkoxyphenyl and the chiral tetralin substituents. Additionally, the aliphatic shell formed by the solubilizing chains around the polar chromophore stack, can account for the exceptionally high solubility of 2 in MCH (> 15 mg mL1). These combined studies of the self-assembly process enabled the identification of suitable conditions for the investigation of fluorescence properties of the individual aggregate species. Aggregation-induced emission enhancement was observed for the almost non-emissive monomer (Fl(M) = 0.23%), which can be rationalized by the increasing rigidification within the dimer (Fl(D) = 2.3%) and the higher aggregate (Fl(H) = 4.5%). The helical chirality of the PYOP decamer stack, furthermore, gave rise to a strong CPL signal with a large glum value of 0.011.
The important conclusion of this thesis is that the temperature- and solvent-dependent analyses are valid alternatives to the classical concentration-dependent analysis to determine thermodynamic parameters of self-assembly equilibria. Although, for a specific supramolecular system, one approach might be favourable over the others for a variety of reasons. The experimental limitations often demand a combination of techniques to fully elucidate a self-assembly process and to gain insights in the aggregate structure. The anti-cooperative merocyanine self-assembly, which was described here for the first time for the PYOP merocyanine 2, is no exception. Besides the interest in the merocyanine assemblies from a structural and functional point of view, the insights gained from the presented studies can also be transferred to other self-assembly systems and be a guide to find the most appropriate analysis technique.
