Der Gram-positive Erreger Staphylococcus aureus ist ein Bestandteil der normalen Haut und Schleimhautflora des Menschen, kann aber auch ein weites Spektrum von Krankheitsbildern hervorrufen. Ein besonderes Charakteristikum dieses Pathogens besteht in der Expression von Oberflächenstrukturen, welche eine hohe Affinität für Proteine der extrazellulären Matrix (ECM) von eukaryontischen Organismen aufweisen und die kollektiv als MSCRAMM (microbial surface components recognizing adhesive matrix molecules) bezeichnet werden. Das auf der Bakterienoberfläche gebundene Fn kann in der Folge als eine Art molekulare Brücke zwischen FnBP exprimierenden S. aureus und dem Fn-Rezeptor auf der Wirtszellseite, dem Integrin 51, dienen. Neben der Anheftung an das Wirtsgewebe kann die indirekte Assoziation mit Integrin 51 die Aufnahme der Bakterien durch die eukaryontische Zelle auslösen. Wie die bakterielle Adhäsion an Integrin 51 und die Aggregation der Integrine durch die mit Fn-beschichteten Bakterien in ein Signal zur Aufnahme der Pathogene durch die Zelle umgesetzt wird, ist nicht vollständig geklärt und sollte im Rahmen dieser Arbeit untersucht werden. Zu diesem Zweck wurde ein neues und effektives Protokoll zur fluoreszenzmikroskopischen Differenzierung von extra- und intrazellulären Bakterien entwickelt. Diese Methode besitzt den Vorteil, von Bakterien-spezifischen Antikörpern unabhängig zu sein. Dadurch bietet sich die Möglichkeit, Bakterien, gegen die es noch keine spezifischen Antiseren gibt, dennoch auf ihre zelluläre Lokalisation und Invasivität mittels mikrobiologischer Methoden untersuchen zu können. Im Hinblick auf die nähere Untersuchung der Signaltransduktion bei der Invasion von S. aureus war die kritische Rolle von Tyrosinkinasen für die Integrin-vermittelte Invasion ein erster wichtiger Hinweis. Diese Befunde führten zu weiteren spezifischeren Untersuchungen, wobei eine wichtige Rolle für Kinasen der Src Familie gezeigt werden konnte. Ein weiterer Hinweis auf die Bedeutung der Src-Kinasen für die Internalisierung von S. aureus war ein dramatischer Rückgang der Aufnahmerate in Src/Yes/Fyn-defizienten Maus-Fibroblasten, verglichen mit Src-rekonstituierten Zellen. Auf biochemischer Ebene konnte eine deutliche Aktivierung der Src-Kinase nach einer Infektion mit S. aureus, nicht aber nach Infektion mit dem nicht-pathogenen S. carnosus festgestellt werden. Integrin-reiche fokale Kontakte (FK) sind angereichert mit Proteinen wie Talin, Vinculin, Paxillin, Tensin, -Actinin oder Zyxin sowie Signalenzymen wie der Fokalen Adhäsions Kinase (FAK) oder Kinasen der Src Familie. Die Protein Tyrosin Kinase (PTK) FAK ist nach Integrinstimulierung eines der Schlüsselenzyme in FK. Dies war der Anlass nach der Bedeutung von FAK für die Integrin-vermittelte Internalisierung von S. aureus zu fragen. Ebenfalls ein wichtiger Hinweis waren die starken Rekrutierungen von Markerproteinen von fokalen Komplexen zum Ort von zellgebunden S. aureus nicht aber von S. carnosus. Daraufhin wurde mittels dominant-negativer FAK-Mutanten und FAKdefizienter Mausfibroblasten der Einfluss von FAK für die Internalisierung von S. aureus untersucht. Bei beiden Versuchsansätzen konnte ein starker Rückgang der Aufnahme beobachtet werden. Zusammengefasst bestätigten diese Ergebnisse die essentielle Rolle von FAK für die Integrin vermittelte Aufnahme der pathogenen S. aureus. Bei der Reorganisation des Aktin-Zytoskeletts spielen eine Reihe von Proteinen eine wichtige Rolle, darunter auch Cortactin. Cortactin ist ein bekanntes Substrat der Src-Kinasen und es lag nahe, nach einer funktionellen Verbindung von Src, FAK und Cortactin zu suchen. Dominant-negative Cortactin-Mutanten, die keine Assoziation mit dem Arp2/3 Komplex oder mit Dynamin aufweisen, oder welche die von Src-vermittelte Phosphorylierung am C-Terminus verhindern, blockierten die Aufnahme von S. aureus. Mikroskopisch konnte eine starke Rekrutierung von Cortactin zu zellgebundenen S. aureus beobachtet werden, jedoch wurde die Rekrutierung nicht von FAK beeinflusst. Die Phosphorylierung von Cortactin aufgrund S. aureus-Infektion war allerdings FAK- und Src-abhängig. Diese Ergebnisse legen nahe, dass ine bisher unbeschriebene FAK/Src Cortactin Signalachse für die Regulation der Integrin-Internalisierung verantwortlich ist. Die detaillierten Untersuchungen der rezeptorvermittelten Aufnahme und der dabei induzierten Signaltransduktion in Wirtszellen gaben neue Erkenntnisse über die Pathogenitätsstrategien von S. aureus. Darüber hinaus ermöglichen diese Arbeiten neue Einblicke in die molekularen Vorgänge, welche die Internalisierung von Integrinen steuern.
Activated platelets and coagulation jointly contribute to physiological hemostasis. However, pathological conditions can also trigger unwanted platelet activation and initiation of coagulation resulting in thrombosis and precipitation of ischemic damage of vital organs such as the heart or brain. The specific contribution of procoagulant platelets, positioned at the interface of the processes of platelet activation and coagulation, in ischemic stroke had remained uninvestigated. The first section of the thesis addresses this aspect through experiments conducted in novel megakaryocyte- and platelet-specific TMEM16F conditional KO mice (cKO). cKO platelets phenocopied defects in platelets from Scott Syndrome patients and had severely impaired procoagulant characteristics. This led to decelerated platelet-driven thrombin generation and delayed fibrin formation. cKO mice displayed prolonged bleeding times and impaired arterial thrombosis. However, infarct volumes in cKO mice were comparable to wildtype (WT) mice in an experimental model of ischemic stroke. Therefore, while TMEM16F-regulated platelet procoagulant activity is critical for hemostasis and thrombosis, it is dispensable for cerebral thrombo-inflammation in mice.
The second section describes the generation and initial characterization of a novel knockin mouse strain that expresses human coagulation factor XII (FXII) instead of endogenous murine FXII. These knockin mice had normal occlusion times in an experimental model of arterial thrombosis demonstrating that human FXII is functional in mice. Therefore, these mice constitute a valuable tool for testing novel pharmacological agents against human FXII – an attractive potential target for antithrombotic therapy.
Glycoprotein (GP)VI and C-type lectin-like receptor 2 (CLEC-2)-mediated (hem)immunoreceptor tyrosine-based activation motif (ITAM) signaling represent a major pathway for platelet activation. The last section of the thesis provides experimental evidence for redundant functions between the two members of the Grb2 family of adapter proteins - Grb2 and Gads that lie downstream of GPVI and CLEC-2 stimulation. In vitro and in vivo studies in mice deficient in both Grb2 and Gads (DKO) revealed that DKO platelets had defects in (hem)ITAM-stimulation-specific activation, aggregation and signal transduction that were more severe than the defects observed in single Grb2 KO or Gads KO mice. Furthermore, the specific role of these adapters downstream of (hem)ITAM signaling was essential for maintenance of hemostasis but dispensable for the known CLEC-2 dependent regulation of blood-lymphatic vessel separation.
In mammals, the RAF family of serine/threonine kinases consists of three members, A-, B- and C-RAF. Activation of RAF kinases involves a complex series of phosphorylations. Although the most prominent phosphorylation sites of B- and C-RAF are well characterized, little is known about regulatory phosphorylation of A-RAF. Using mass spectrometry, we identified here a number of novel in vivo phosphorylation sites in A-RAF. The physiological role and the function of these sites were investigated subsequently by amino acid exchange at the relevant positions. In particular, we found that S432 participates in MEK binding and is indispensable for A-RAF signaling. On the other hand, phosphorylation within the activation segment does not contribute to epidermal growth factor-mediated activation. Regarding regulation of A-RAF activity by 14-3-3 proteins, we show that A-RAF activity is regulated differentially by its C-terminal and internal 14-3-3 binding domain. Furthermore, by use of SPR technique, we found that 14-3-3 proteins associate with RAF in an isoform-specific manner. Of importance, we identified a novel regulatory domain in A-RAF (referred to as IH-segment) positioned between amino acids 248 and 267, which contains seven putative phosphorylation sites. Three of these sites, serines 257, 262 and 264, regulate A-RAF activation in a stimulatory manner. The spatial model of the A-RAF fragment including residues between S246 and E277 revealed a “switch of charge” at the molecular surface of the IH-region upon phosphorylation, suggesting a mechanism in which the high accumulation of negative charges may lead to an electrostatic destabilization of protein/membrane interaction resulting in depletion of A-RAF from the plasma membrane. Activation of B- and C-RAF is regulated by phosphorylation at conserved residues within the negative-charge regulatory region (N-region). Identification of phosphopeptides covering the sequence of the N-region led to the conclusion that, similar to B- and C-RAF, kinase activity of A-RAF is regulated by phosphorylation of the N-region. Abrogation of A-RAF activity by S299A substitution and elevated activity of the A-RAF-Y301D-Y302D mutant confirmed this conclusion. In addition, we studied the role of the non-conserved residues within the N-region in the activation process of RAF kinases. The non-conserved amino acids in positions –3 and +1 relative to the highly conserved S299 in A-RAF and S338 in C-RAF have so far not been considered as regulatory residues. Here, we demonstrate that Y296R substitution in A-RAF led to a constitutively active kinase. In contrast, G300S substitution (mimicking B- and C-RAF) acts in an inhibitory manner. These data were confirmed by analogous mutations in C-RAF. Based on the three-dimensional structure of the catalytic domain of B-RAF, a tight interaction between the N-region residue S339 and the catalytic domain residue R398 was identified in C-RAF and proposed to inhibit the kinase activity of RAF proteins. Furthermore, Y296 in A-RAF favors a spatial orientation of the N-region segment, which enables a tighter contact to the catalytic domain, whereas a glutamine residue at this position in C-RAF abrogates this interaction. Considering this observation, we suggest that Y296, which is unique for A-RAF, is a major determinant of the low activating potency of this RAF isoform. Finally, the residues R359 in A-RAF and R398 in C-RAF, which interact with the N-region, are also involved in binding of phosphatidic acid. Substitution of this conserved arginine by alanine resulted in accumulation of hyper-phosphorylated form of RAF, suggesting that this residue play a crucial role in phosphorylation-mediated feedback regulation of A- and C-RAF. Collectively, we provide here for the first time a detailed analysis of in vivo A-RAF phosphorylation status and demonstrate that regulation of A-RAF by phosphorylation exhibits unique features compared with B- and C-RAF.
Ein Weg, der von Rezeptor-Tyrosin-Kinasen benutzt wird um Signale auf "downstream" gelegene Effektormoleküle zu übertragen, erfolgt über Adaptorproteine, die Bindungsstellen für verschiedene Proteine zur Verfügung stellen. Das daughter of sevenless (dos) Gen wurde in einem Screen nach Downstream-Komponenten der Sevenless (Sev) Rezeptor-Tyrosin-Kinase gefunden. Dos besitzt eine N-terminale PH-Domäne und mehrere Tyrosinreste in Konsensussequenzen für SH2-Domänen Bindungsstellen von verschiedenen Proteinen. Die strukturellen Merkmale von Dos und Experimente, die zeigten, daß Tyrosine im Dos Protein nach der Aktivierung von Sev phosphoryliert werden, legen den Schluß nahe, daß Dos zur Familie der Multi-Adaptor-Proteine gehört. Zu dieser Familie werden die Insulin-Rezeptor-Substrat (IRS) Proteine, Gab1 und Gab2 gerechnet. In dieser Arbeit wurde ein monoklonaler Maus anti-Dos Antikörper etabliert. Das Epitop dieses Antikörpers liegt im Bereich der C-terminalen 416 Aminosäuren des Dos Proteins. Mittels Westernblot Analysen wurde für Dos ein Molekulargewicht von 115 kD ermittelt. Antikörperfärbungen von wildtypischen Augenimaginalscheiben dritter Larven zeigten, daß das Dos Protein in Zellen in und posterior der morphogenetischen Furche exprimiert wird und in diesen Zellen apikal lokalisiert ist. Zur Charakterisierung des homozygot letalen dosR31 Allels, wurde der genomische Bereich sequenziert und die erhaltenen Daten mit der cDNA Sequenz verglichen. Die so etablierte Aminosäuresequenz für das DosR31 Protein hat sechs Aminosäuresubstitutionen, die möglicherweise die Tertiärstruktur beeinflussen. Zusätzlich wurde ein Stopcodon in Position 463 der Aminosäuresequenz gefunden. Bei dosR31 handelt es sich um ein "loss of function" Allel, das nicht in der Lage ist, die normale Dos Funktion zu erfüllen. Um die funktionelle Rolle der potentiellen SH2-Domänen Bindungsstellen für die Dos Funktion in der Rezeptor-Tyrosin-Kinasen vermittelten Signaltransduktion zu untersuchen, wurden mutierte dos Transgene in Fliegen exprimiert. Die potentiellen Bindungsstellen für die SH2-Domänen des SH2/SH3 Adaptorproteins Shc, der PhospholipaseC-g (PLCg), der regulatorische Untereinheit der Phosphatidylinositol-3-Kinase (PI3Kinase) und der Corkscrew (Csw) Tyrosin Phosphatase wurden durch den Austausch des für die Bindung wichtigen Tyrosins gegen ein Phenylalanin mutiert. Die ektopische Expression der mutierten Konstrukte ohne Bindungsstellen für die Shc, PLCg und PI3Kinasen SH2-Domänen konnte in Abwesenheit von endogenem Dos die fehlende Dos Funktion während der Entwicklung vollständig ersetzen. Im Gegensatz dazu ist das Tyrosin 801 als nachgewiesene Bindungsstelle für Csw SH2-Domänen essentiell für die Funktion von Dos. Ektopische Expression von Transgene durch Hitzeschock kann zu phänotypischen Effekten führen, die nicht auf das Transgen zurückzuführen sind. Um dieses Problem zu umgehen wurde das endogene dos Enhancer/Promotor Element kloniert, damit die Funktion von mutierten Transgenen auch im endogenen Expressionsmuster untersucht werden konnte. Das klonierte genE-dos Minigen war in der Lage, den Verlust von endogenem Dos in dosR31 und dosP115 Tieren vollständig zu ersetzen und zeigte eine völlig wildtypische Expression in Augenimaginalscheiben. Zur Untersuchung, welche Rolle die mutierten SH2-Domänen Bindungsstellen bei der Dos Funktion in der Augenentwicklung spielen, wurde ein neues in vivo Testsystem basierend auf der Flp/FRT Flipase Rekombinase Technik etabliert. Dieses klonale Testsystem erlaubt die Expression mutierter Transgene unter der Kontrolle der dos Enhancer/Promotor Sequenzen in Klonen von Zellen, denen die endogene Dos Funktion fehlt. Die klonale Analyse der mutierten Konstrukte konnte zeigen, daß das Tyrosin 801, als Bindungsstelle für eine Csw SH2-Domäne, eine essentielle Rolle für die Dos Funktion spielt. Die Tyrosinreste in den potentiellen SH2-Domänen Bindungsstellen für Shc, PLCg und PI3Kinase spielen hingegen keine essentielle Rolle für die Dos Funktion bei der Augenentwicklung. Das etablierte klonale Testsystem kann allgemein zur Untersuchung der in vivo Funktion von potentiellen Protein-Protein Interaktionsregionen im Dos Protein bei der Augenentwicklung eingesetzt werden unabhängig von deren Erfordernis für andere Entwicklungsprozesse.
Hintergrund: Der weitverbreitete und karzinogene Pilzmetabolit Ochratoxin A (OTA) beeinflußt die Funktion und das Wachstumsverhalten renaler Zellen. In höheren Konzentrationen reduziert OTA auch die Integrität der Zellen. Untersucht wurde die mögliche Beteiligung von Änderungen der zellulären Calciumhomöostase an den Wirkungen von OTA in nanomolaren Konzentrationen. Methoden: Immortalisierte menschliche Nierenepithelzellen (IHKE) wurden verwendet, um die Effekte von OTA auf die zytosolische Calciumhomöostase ([Ca2+]i), Zellwachstum und und -integrität zu untersuchen. 1 nmol/l OTA potenzierte Ca2+-abhängig die EGF- und Ang II-induzierte Zellproliferation. Ca2+-unahängige Zelluntergänge und Reduktion der Zellzahl konnte nur nach 24-stündiger Inkubation mit einer Schwellenkonzentration von >10 nmol/l beobachtet werden. Innerhalb von Sekunden wurden durch OTA reversible und konzentrationsabhänige [Ca2+]i-Oszillationen mit einer Schwellenkonzentration von 0.1 nmol/l hervorgerufen. Die Oszillationen wurden durch Reduktion des extrazellulären Ca2+, den Ca2+-Kanalblocker SK&F 96365 und durch Hemmung der der Phospholipase C verhindert. Der durch OTA hervorgerufene Ca2+-Einstrom war auch nach Entleerung von Ca2+-Speichern durch Schwellenkonzentration das ebenfalls die Oszollationen hemmte, noch vorhanden. Zusätzlich steigerte OTA den Füllungszustand von Thapsigargin-empfindlichen Ca2+-Speichern und stimulierte die Aktivität der Thapsigargin-empfindlichen Ca2+-ATPase. Eine 10-minütige Inkubation mit OTA erhöhte den zellulären cAMP-Gehalt dosisabhängig. Der Proteinkinase A Inhibitor H-89 unterdrückte die OTA-induzierten Ca2+- Oszillationen. 1 nM OTA potenzierte die Effekte von Angiotensin II und EGF auf [Ca2+]i. Schlußfolgerungen: (i) OTA beeinträchtigt in niedrig-nanomolaren Konzentrationen, die im Rahmen der natürlichen Exposition auftreten können, reversibel die Ca2+-Homöostase in menschlichen proximalen Tubuluszellen. (ii) OTA verursacht dosis-abhängige [Ca2+]i-Oszillationen die auf OTA-induzierten Ca2+-Einstrom und Thapsigargin-sensitive Ca2+-Speicher angewiesen sind. (iii) Ferner interagieren niedrig-nanomoler Konzentrationen von OTA mit hormonellen Ca2+-Signalen, was z.B. zu einem veränderten zellulären Proliferationsverhalten führt. (iv) Die Verminderung der Zellintegrität durch höhere OTA-Konzentrationen hängt nicht von Veränderungen der Ca2+-Homöostase ab. (v) Die durch OTA hervogerufene renale Dysfunktion scheint, zumindest teilweise, auf Wechselwirkungen mit zellulären Signaltransduktionsmechanismen zu beruhen und nicht auf Zellzerstörung.
Mechanismen der TZR-abhängigen und -unabhängigen Aktivierung primärer T-Zellen der Ratte durch CD 28

Zur Aktivierung ruhender T-Zellen sind zwei Signale erforderlich: Eines wird antigen-abhängig über den T-Zellrezeptor (TZR) gegeben, ein zweites erfolgt über kostimulatorische Rezeptoren. Unter den bekannten kostimulatorischen Molekülen ist CD28 das potenteste. Die mitogene Aktivität einiger monoklonaler Antikörper (mAb) spezifisch für CD28 der Ratte, die alle ruhenden primären Ratten-T-Zellen ohne TZR-Ligation zu Proliferation und IL-2-Produktion anregen können, erlaubte, die Signalwege nach dieser direkten CD28-Stimulation und nach Kostimulation zu analysieren und zu vergleichen. Die erzielten Ergebnisse deuten auf unterschiedliche Signalwege nach Kostimulation mit TZR-Beteiligung und direkter CD28-Stimulation hin und zeigen, daß direkte CD28-Stimulation nicht die Signaltransduktion durch den TZR imitiert. Daher unterstützt CD28 als kostimulatorisches Molekül nicht nur TZR-vermittelte Signaltransduktion, sondern fungiert auch als eigenständiges Signalmolekül, das spezifische mitogene Signale generiert. Diese Eigenschaft von CD28 könnte für die Art der induzierten Immunreaktion von Bedeutung sein, da das Verhältnis der Stärke von TZR- und CD28-Signal die funktionelle Differenzierung von T-Zellen in Th1- oder Th2-Zellen bestimmt.
Summary: I previously demonstrated that conditional overexpression of the neuronal nitric oxide synthase (nNOS) inhibited L-type Ca2+-channels and decreased myocardial contractility1 (Burkard N. et al. (2007). Circ Res 100, 32-44). However, nNOS has multiple targets within the cardiac myocyte and it is possible that interesting biological functions of this protein remain to be elucidated. In this study, I showed that nNOS overexpression has a cardioprotective effect after ischemia-reperfusion injury by inhibiting mitochondrial function and reducing the generation of reactive oxygen species (ROS). The effect of conditional nNOS overexpression in cardiac myocytes in ischemiareperfusion injury was assessed. Ischemia-reperfusion injury in WT mice resulted in nNOS accumulation in the mitochondria. Similary, transgenic nNOS overexpression caused nNOS abundance in mitochondria. Electron microscopy of mouse myocardium from nNOS overexpressing mice showed that after induction of its expression, nNOS is additionally localised in mitochondria. nNOS translocation into mitochondria was dependent on HSP90. Ischemia-reperfusion experiments in isolated hearts showed a cardioprotective effect of nNOS overexpression (30min post-ischemia, LVDP 27.0±2.5mmHg in non-induced animals vs. 45.2±1.9mmHg in nNOS overexpressing mice, n=12, p<0.05). Consistently with this finding, in vivo the infarct size within the area at risk was significantly decreased in nNOS overexpressing mice compared to non-induced animals (36.6±8.4 relative % vs. 61.1±2.9 relative %, n=12, p<0.05). nNOS overexpression also caused a significant increase in mitochondrial nitrite levels accompanied by a decrease of cytochrome c oxidase activity (72.0±8.9units/ml in nNOS overexpressing mice vs. 113.2±17.1units/ml in non-induced mice, n=12, p<0.01) resulting in an inhibition of mitochondrial function. Accordingly, O2-consumption (MVO2) in isolated heart muscle stripes was decreased in nNOS overexpressing mice, already under resting conditions (0.016±0.0015 vs. 0.024±0.006ml[O2] x mm-3 x min-1, n=13, p<0.05). Additionally, this study showed that the ROS concentration was significantlydecreased in hearts of nNOS overexpressing mice compared to non-induced animals (6.14±0.685 vs. 14.53±1.7μM, n=8, p<0.01). Application of different inhibitors, Western Blot analysis and activity assays showed that the lower ROS concentration in nNOS overexpressing mice was caused by inhibition of the xanthine oxidoreductase (XOR) activity by the increased abundance of nNOS expression. In summary, this study demonstrated that the conditional transgenic overexpression of nNOS resulted in myocardial protection after ischemia-reperfusion injury. Besides reduction of myocardial Ca2+-overload after reperfusion this might be caused by inhibition of mitochondrial function through nNOS, which reduced myocardial oxygen consumption already under baseline conditions (Burkard N. conditionally accepted by
Chlamydia are Gram-negative obligate intracellular bacteria responsible for a wide spectrum of relevant diseases. Due to their biphasic developmental cycle Chlamydia depend on an intact host cell for replication and establishment of an acute infection. Chlamydia have therefore evolved sophisticated strategies to inhibit programmed cell death (PCD) induced by a variety of stimuli and to subvert the host immune system. This work aimed at elucidating whether an infection with C. trachomatis can influence the cellular response to double-stranded RNA (dsRNA). The synthesis of dsRNA is a prominent feature of viral replication inside infected cells that can induce both PCD and the activation of a cellular innate immune response. In order to mimic chlamydial and viral co-infections, Chlamydia-infected cells were transfected with polyinosinic:polycytidylic acid (polyI:C), a synthetic dsRNA. In the first part of this work it was investigated whether C. trachomatis-infected host cells could resist apoptosis induced by polyI:C. A significant reduction in apoptosis, determined by PARP cleavage and DNA fragmentation, could be observed in infected cells. It could be shown that processing of the initiator caspase-8 was inhibited in infected host cells. This process was dependent on early bacterial protein synthesis and was specific for dsRNA because apoptosis induced by TNFalpha was not blocked at the level of caspase-8. Interestingly, the activation of cellular factors involved in apoptosis induction by dsRNA, most importantly PKR and RNase L, was not abrogated in infected cells. Instead, RNA interference experiments revealed the crucial role of cFlip, a cellular caspase-8 inhibitor, for chlamydial inhibition of dsRNA-induced apoptosis. First data acquired by co-immunoprecipitation experiments pointed to an infection-induced concentration of cFlip in the dsRNA-induced death complex of caspase-8 and FADD. In the second part of this work, the chlamydial influence on the first line of defense against viral infections, involving expression of interferons and interleukins, was examined. Activation of the interferon regulatory factor 3 (IRF-3) and the NF-kappaB transcription factor family member p65, both central regulators of the innate immune response to dsRNA, was altered in Chlamydia-infected epithelial cells. polyI:C-induced degradation of IkappaB-alpha, the inhibitor of NF-kappaB, was accelerated in infected cells which was accompanied by a change in nuclear translocation of the transcription factor. Translocation of IRF-3, in contrast, was significantly blocked upon infection. Together the data presented here demonstrate that infection with C. trachomatis can drastically alter the cellular response to dsRNA and imply an impact of chlamydial infections on the outcome of viral super-infections.
Analyse der Expression und möglicher signalinduzierender Eigenschaften des CD1d-Moleküls der Ratte

Wie MHC Klasse I und II-Moleküle präsentieren CD1d-Moleküle dem TCR Antigene, allerdings Lipide und Glykolipide und nicht Proteinfragmente. Die Entdeckung der massiven TH1- und TH2-Zytokinproduktion von Typ I-NKT-Zellen nach CD1d-vermittelter Erkennung von α-Galactosylceramid, einem aus dem Meeresschwamm gewonnenen Glykosphingolipid, weckte großes Interesse an ihrem immunregulatorischen Potential und ihrem möglichen Nutzen für neue Immun- und Tumortherapien. Um die Funktion und die Bedeutung von CD1d besser zu verstehen, wurden in dieser Arbeit die Expressionslevel der lymphatischen Gewebe der Ratte und der Maus untersucht. Hierfür wurden die neu generierten monoklonalen Antikörper 232 und 58/4 verwendet, die die CD1d-Moleküle von Ratte und Maus binden und so den direkten Vergleich beider Spezies ermöglichen. Sowohl die isolierten Zellen des Thymus und der Milz als auch des Lymphknotens waren in der LEW- und F344-Ratte sowie in der BALB/c-Maus schwach bis stark CD1d positiv. In der LEW-Ratte und in der F344-Ratte wiesen jeweils ca. 18% der Milzzellen eine vergleichsweise erhöhte CD1d-Expression auf. Dabei handelte es sich in erster Linie um Marginalzonen-B-Zellen. Bestimmte Subpopulationen der Dendritischen Zellen und vermutlich Makrophagen stellten die restlichen CD1d stark positiven Populationen dar. Nur ca. 2% der isolierten Zellen der Lymphknoten der LEW-Ratte waren stark CD1d positiv, wohingegen der LEW-Thymus gemäß dem noch geringeren Anteil an APC kaum Zellen mit erhöhter CD1d-Expression enthielt. In der BALB/c-Maus war der Anteil CD1d stark positiver Milzzellen mit 4% deutlich geringer als in der LEW- oder F344-Milz. Abgesehen von MZ-B-Zellen konnten in der Maus kaum Populationen mit starker CD1d-Expression in den verschiedenen Färbungen festgestellt werden. Demnach stellt CD1d sowohl in der Ratte als auch in der Maus einen guten Marker für MZ-B-Zellen dar. Demgegenüber zeigten vereinzelt kleine Populationen der Milz, des Lymphknotens und des Thymus beider Spezies eine verminderte oder gar keine CD1d-Expression. Zur Analyse möglicher signalinduzierender Eigenschaften der verschiedenen Anti- CD1d-Antikörper wurden ihre Effekte auf rCD1d+ Transduktanten und primäre Zellen untersucht. 58/4 konnte im Gegensatz zu 232 spezifisch über Bindung an Ratten- CD1d Zelltod und Aggregatbildung in Tumor-B-Zellen des Menschen und der Maus, aber nicht in Tumor-T-Zellen, induzieren. Der zytoplasmatische Schwanz der CD1d-Moleküle scheint an der Aggregatbildung beteiligt zu sein. Die Bindung von 58/4 oder 232 führte in überlebenden rCD1d+ Raji-Zellen zu einer ähnlich starken Internalisierung der CD1d-Moleküle. Während nach 5-stündiger Inkubation mit 232 und erneuter CD1d-Färbung wieder die vorherige CD1d- Expression festgestellt wurde, konnte nach Inkubation mit 58/4 eine bleibende Herunterregulierung beobachtet werden. Folglich bewirkte 58/4 ein anderes bzw. stärkeres Signal in den Zellen als 232. Diese Beobachtungen stützen die Signaltransduktion als mögliche weitere Funktion der CD1d-Moleküle neben der Antigenpräsentation und definieren die monoklonalen Antikörper 232 und 58/4 als nützliche Werkzeuge für weitere Studien zur Analyse der molekularen Mechanismen der CD1d-vermittelten Signaltransduktion. Das Verständnis solcher Mechanismen bildet wiederum die Grundlage für die Entwicklung neuer Therapien z. B. zur Eliminierung CD1d exprimierender Tumore.
Gegenstand dieser Arbeit ist die Untersuchung von Aspekten der Funktion von CD22, einem B-Zell spezifischen Transmembran-Rezeptor der Siglec-Familie (Sialinsäure-bindende Immunglobulin-ähnliche Lectine). Mit der äußersten der 7 extrazellulären Ig-ähnlichen Domänen kann CD22 spezifisch mit a2,6-Sialinsäure interagieren. In der cytoplasmatischen Domäne von CD22 befinden sich 6 konservierte Tyrosine, 3 davon in ITIMs (Immunrezeptor tyrosinhaltige inhibitorischen Motiven). Nach Kreuzvernetzung des B-Zell Rezeptors wird CD22 tyrosinphosphoryliert. Die cytosolische Tyrosin-Phosphatase SHP-1 bindet in der Folge an die phosphorylierten ITIMs, wird aktiviert, und inhibiert das BCR Ca2+-Signal. Gleichzeitig binden jedoch auch positive Modulatoren des BCR-Signals (Lyn, Syk, PLCg PI3K, und Grb-2) an CD22, deren Rolle im Zusammenhang mit CD22 bislang ungeklärt ist. 1. In einem Hauptteil der Arbeit sollten zwei Knockin Mausmodelle generiert werden. Das eine Mausmodell (CD22-ITIM-KO) sollte zerstörte ITIM-Motive enthalten. Bei dem anderen (CD22-Tailless) sollte die gesamte cytoplasmatische Domäne von CD22 fehlen. Beide Modelle sollten der Untersuchung der Rolle der an CD22 bindenden positiven Modulatoren des BCR-Signals, und des Zusammenhangs zwischen Signaltransduktion und Ligandenbindung in vivo dienen. Die Klonierung der Targeting-Vektoren für CD22-ITIM-KO (pCD22-ITIM-KO) und CD22-Tailless (pCD22-Tailless) wurde abgeschlossen. Mit Hilfe ebenfalls klonierter Kontrollvektoren wurden PCRs zur Identifizierung homolog rekombinanter ES-Zell Klone etabliert. Für beide Targeting-Konstrukte wurden nach Transfektion von C57BL/6 ES-Zellen homolog rekombinante Klone erhalten, und mittels Southern Blot und Sequenzierung der eingeführten Mutationen vollständig charakterisiert. Nach Cre/lox-vermittelter Deletion der Selektionskassette des Targeting-Konstrukts folgte Injektion voll charakterisierter CD22-ITIM-KO Klone in BALB/c-Blastozysten. Es wurden 5 chimäre Tiere erhalten, von denen keines die Mutationen durch die Keimbahn weitergab. Transfektionen der C57BL/6 Targeting-Konstrukte in anderen, nicht-isogenen ES-Zell Linien ergaben keine homologen Rekombinanten. Das Auffinden der genomischen Sequenz von CD22 in einer Internet-Datenbank ermöglichte die Verlängerung von pCD22-ITIM-KO um ca. 4 kb mit 129/ola-DNA. Eine Transfektion dieses neuen Konstruktes in eine 129/ola ES-Zelllinie ergab keine homologen Rekombinanten. Jedoch öffnet die nun bekannte genomische CD22-Sequenz den Weg zu einfacher Neukonstruktion von pCD22-ITIM-KO mit 129/ola-DNA, oder zu einer Veränderung und Verbesserung der vorhandenen C57BL/6-Vektoren. 2. Zur Untersuchung der Auswirkung der zerstörten ITIMs auf Tyrosinphosphorylierung und SHP-1 Assoziation von CD22 in vitro in einer Zelllinie wurde ein CD22-ITIM-KO-Expressionsvektor konstruiert, und Sialyltransferase/CD22-ITIM-KO Doppeltransfektanten der Plasmozytom-Zelllinie J558L gewonnen. CD22-ITIM-KO wurde nach BCR-Stimulation nicht tyrosinphosphoryliert, SHP-1 konnte entsprechend nicht mit CD22-ITIM-KO assoziieren. Die Ergebnisse zeigen die Funktionalität des CD22-ITIM-KO Konstrukts hinsichtlich ITIM-Phosphorylierung und SHP-1 Bindung. Weiterhin zeigten die Ergebnisse, daß die ITIM-Tyrosine wichtig für die Phosphorylierung der nicht-ITIM-Tyrosine sind. 3. Interaktion von CD22 mit a2,6-Sialinsäure auf der selben Zelloberfläche (in Cis) spielt eine wichtige Rolle bei der Zell-Zell-Interaktion und bei der intrazellulären Signaltransduktion. In dieser Arbeit wurden erstmals mittels Durchflußcytometrie B-Zellen mit CD22, dessen Liganden-Bindungsstelle nicht durch endogene a2,6-Sialinsäure besetzt ist (demaskiertes CD22), identifiziert. Ca. 10,5% aller B220+ Milzzellen von Wildtyp-Mäusen, aber nur ca. 4,5% der B220+ Milzzellen aus CD22-/- Mäusen waren in der Lage, exogene a2,6-Sialinsäure zu binden. Dieser Effekt ist zum Großteil auf CD22 zurückzuführen. Genauere FACS-Analyse zeigte, daß Zellen mit demaskiertem CD22 in der Fraktion der Transitionalen B-Zellen Typ 2 (T2-Zellen) angereichert sind, und Zeichen von Aktivierung (B7.2, CD25, CD69) zeigen. In Übereinstimmung damit führte in vitro Aktivierung von B-Zellen mit LPS oder IL4 zu CD22-abhängiger Demaskierung. 4. FACS-Färbungen zeigten, daß das Marginalzonen (MZ) B-Zell Kompartiment in CD22-/- Mäusen um ca. 70-80% gegenüber wt-Mäusen verkleinert ist. In Bestätigung früherer Arbeiten war die Immunantwort gegen i.p.-injizierte Thymusunabhängige Antigene Typ 2 (TI2-Antigene) in CD22-/- Mäusen 2-fach reduziert. Die Antwort war jedoch signifikant stärker reduziert (3-4-fach), wenn die gleiche Antigen-Menge i.v.-injiziert wurde, eine Situation, in der bevorzugt die MZ B-Zellen der Milz in Kontakt mit im Blutstrom transportierten Antigenen kommen. Es ist wahrscheinlich, daß die bekannte Defizienz in TI2-Immunantworten in CD22-/- Mäusen auf die verringerte MZ B-Zell Anzahl zurückzuführen ist.
RKIP reguliert Proteinkinasen der Signaltransduktionskaskaden von G Protein-gekoppelten Rezeptoren, der Raf/MEK/ERK-MAPK, des Transkriptionsfaktors NFκB und von GSK3β. Unklar war bisher, wie die spezifische Interaktion von RKIP mit seinen mannigfaltigen Interaktionspartnern ermöglicht und reguliert wird. Raf1 und GRK2 sind die einzigen bekannten direkten Interaktionspartner von RKIP und wurden deshalb gewählt, um die zugrundeliegenden molekularen Mechanismen dieser Interaktion genauer zu untersuchen. In dieser Arbeit wurde gezeigt, dass RKIP nach PKC-vermittelter Phosphorylierung von Serin153 dimerisiert und dass diese Dimerisierung für die RKIP/Raf1-Dissoziation und die RKIP/GRK2-Interaktion essentiell ist. Co-Immunpräzipitationsexperimente mit einer phosphorylierungsdefizienten Mutante zeigten, dass für diese Dimerisierung die Phosphorylierung von beiden RKIP-Molekülen notwendig ist. Als Dimerinteraktionsfläche wurden die Aminosäuren 127-146 von RKIP identifiziert, da das Peptid RKIP127-146 die Dimerisierung von RKIP spezifisch und effizient hemmte. Um die Bedeutung dieser phosphorylierungsinduzierten Dimerisierung von RKIP für seine Interaktion mit Raf1 und GRK2 zu untersuchen, wurden eine phosphomimetische Mutante (RKIPSK153/7EE) und eine Mutante von RKIP generiert, welche bereits unphosphoryliert dimerisiert (RKIP∆143-6). Folgende Ergebnisse legen nahe, dass die Dimerisierung von RKIP für die spezifische Interaktion mit Raf1 bzw. GRK2 entscheidend ist: (i) Die Dimerisierung von phosphoryliertem RKIP ging mit der Dissoziation von RKIP und Raf1 und der Assoziation von RKIP und GRK2 einher; (ii) die Mutanten RKIPSK153/7EE und RKIP∆143-6, die bereits in unstimulierten Zellen eine starke Dimerisierung zeigten, hatten eine höhere Affinität zu GRK2 als zu Raf1; (iii) die Hemmung der RKIP-Dimerisierung interferierte nur mit der RKIP/GKR2- aber nicht mit der RKIP/Raf1-Interaktion; (iv) in vitro und in Mausherzen konnte ein RKIP- und GRK2-immunreaktiver Komplex nachgewiesen werden; (v) Untersuchungen zur RKIP-vermittelten Hemmung der Kinaseaktivität von GRK2 und Raf implizierten, dass dimerisiertes RKIP nur die Aktivität von GRK2, nicht aber von Raf hemmt. Diese Arbeit zeigt, dass die phosphorylierungsinduzierte Dimerisierung von RKIP die spezifische Interaktion von RKIP mit Raf1 und GRK2 koordiniert. Die Aufklärung dieses Mechanismus erweitert unser Verständnis der spezifischen Interaktion von Kinasen mit ihren Regulatorproteinen.
The phytohormone auxin performs important functions in the initiation of plant tissues and organs, as well as in the control of root growth in conjunction with external stimuli such as gravity, water and nutrient availability. These functions are based primarily on the auxin-dependent regulation of cell division and elongation. Important for the latter is the control of the cell turgor by the vacuole. As storage for nutrients, metabolites and toxins, vacuoles are of vital importance. Vacuolar stored metabolites and ions are exchanged across the vacuolar membrane with the cytoplasm via active transport processes as well as passively through ion channels. In their function as second messenger, calcium ions are important regulators but also subject to vacuolar transport processes. Changes in the cytosolic calcium concentration not only act locally, but are also associated with signal transduction over longer distances. In this work, electrophysiological methods were combined with imaging techniques to gain insights into the interaction between cytosolic calcium signals, vacuolar transport processes and auxin physiology in the intact plant organism.
Calcium signals are involved in the regulation of vacuolar ion channels and transporters. In order to investigate this in the intact organism, intracellular microelectrode measurements were performed in the model system of bulging Arabidopsis thaliana root hairs. By means of the two-electrode voltage-clamp technique, it could be confirmed that the vacuolar membrane is the limiting electrical resistance during intravacuolar measurements and thus measured ion currents actually represent only the currents across the vacuolar membrane. The already known time-dependent decrease of vacuolar conductivity during intravacuolar experiments could be further correlated with an impalement-related, transient increase of the cytosolic calcium concentration. Intravacuolar voltage-clamp experiments in root hair cells of calcium reporter plants confirmed this relationship between vacuolar conductivity and the cytosolic calcium concentration.
However, the vacuole is not just a recipient of cytosolic calcium signals. Since the vacuole represents the largest intracellular calcium reservoir, it has long been argued that it is also involved in the generation of such signals. This could be confirmed in intact root hair cells. Changes in the vacuolar membrane potential affected the cytosolic calcium concentration in these cells. While depolarizing potentials led to an increase of the cytosolic calcium concentration, hyperpolarization of the vacuolar membrane caused the opposite. Thermodynamic considerations of passive and active calcium transport across the vacuolar membrane suggested that the results described herein reflect the behaviour of vacuolar H+/Ca2+ exchangers whose activity is determined by the proton motive force.
In addition, cytosolic calcium has been shown to be a key regulator of a rapid auxin-induced signaling pathway that regulates polar transport of the hormone.
In the same model system of bulging root hairs it could be shown that the external application of auxin results in a very fast, auxin concentration- and pH-dependent depolarization of the plasma membrane potential. Synchronous with the depolarization of the plasma membrane potential, transient calcium signals were recorded in the cytosol. These were caused by an auxin-activated influx of calcium ions through the ion channel CNGC14. Experiments on loss-of-function mutants as well as pharmacological experiments showed that the auxin-induced activation of the calcium channel requires auxin-perception by the F-box proteins of the TIR1/AFB family.
Investigations of auxin-dependent depolarization as well as the auxin-induced influx of protons into epidermal root cells of loss-of-function mutants showed that the secondary active uptake of auxin by the high-affinity transport protein AUX1 is responsible for the rapid depolarization
Not only the cytosolic calcium signals correlated with CNGC14 function, but also the AUX1-mediated depolarization of root hairs. An unchanged expression of AUX1 in the cngc14 loss-of-function mutant suggested that the activity of AUX1 must be post-translationally regulated. This hypothesis was supported by experiments in which treatment with the calcium channel blocker lanthanum led to inactivation of AUX1 in the wild type.
The cytosolic loading of individual epidermal root cells with auxin resulted in the spread of lateral and acropetal calcium waves. These correlated with a shift of the auxin gradient at the root apex and thus supported a hypothetical calcium-dependent regulation of polar auxin transport. A model for a rapid, auxin-induced and calcium-dependent signaling pathway is presented and its importance for gravitropic root growth is discussed. Since AUX1-mediated depolarization varied with external phosphate concentration, the importance of this rapid signaling pathway is also discussed for the adaptation of root hair growth to an inadequate availability of phosphate.
The MEK5/ ERK5 kinase module is a relatively new discovered mitogen-activated protein kinase (MAPK) signalling pathway with a poorly defined physiological function. Since ERK5 and its upstream activator MEK5 are abundant in skeletal muscle a function of the cascade during muscle differentiation was examined. ERK5 becomes activated upon induction of differentiation in mouse myoblasts. The selective activation of the pathway results in promoter activation of differentiation-specific genes, such as the cdk-inhibitor p21 gene, the myosin light chain (MLC1A) gene, or an E-box containing promoter element, where myogenic basic-helix-loop-helix proteins such as MyoD or myogenin bind. Moreover, myogenic differentiation is completely blocked, when ERK5 expression is inhibited by antisense RNA. The effect can be detected also on the expression level of myogenic determination and differentiation markers such as p21, MyoD and myogenin. Another new finding is that stable expression of ERK5 in C2C12 leads to differentiation like phenotype and to increased p21 expression levels under growth conditions. These results provide first evidence that the MEK5/ERK5 MAP kinase cascade is critical for early steps of muscle cell differentiation.
Um Änderungen in seiner Umwelt wahrnehmen zu können, benötigt S. aureus unterschiedliche Signaltransduktionssysteme. In dieser Arbeit wurde erstmals die Eukaryoten-ähnliche Serin/Threonin-Proteinkinase (STPK) PknB umfassend charakterisiert. Die posttranslationale Proteinmodifikation mittels Phosphorylierung spielt sowohl in Eukaryoten als auch in Prokaryoten eine wichtige Rolle. Man glaubte lange, dass die Phosphorylierung von Serin-, Threonin- und Tyrosinresten ein nur auf Eukaryoten beschränkter Regulationsmechanismus ist. Dagegen wurde die Phosphorylierung an Histidin- und Aspartatresten durch die Zweikomponenten-Systeme allein den Prokaryoten zugeordnet. Die Genomanalysen der letzten Jahre identifizierten jedoch STPKs und Serin/Threonin-Proteinphosphatasen (STPP) in nahezu allen prokaryotischen Genomen. Auch S. aureus codiert für eine STPK, die eine hohe Homologie zu den beschriebenen STPKs aufweist. In dieser Arbeit wurden mittels Microarray-Analyse einer ΔpknB-Mutante im Stamm 8325 erste Hinweise zur Funktion von PknB als Regulator der Zellwandsynthese sowie zentraler Stoffwechselwege gewonnen. Es wurden mittels Phosphopreoteom-Analysen in vivo-Substrate identifiziert und weiterhin die Kinase biochemisch charakterisiert.
Multiple sclerosis is an autoimmune disease of the central nervous system characterized by inflammatory, demyelinating lesions and neuronal death. Formerly regarded as a variant of MS, neuromyelitis optica (NMO)/Devic’s disease is now recognized as a distinct neurological disorder exhibiting characteristic inflammatory and demyelinated foci in the optic nerves and the spinal cord sparing the brain. With the introduction of the double-transgenic “Devic mouse” model featuring spontaneous, adjuvant-free incidence of autoimmune neuroinflammation due to the interaction of transgenic MOG-specific T and B cells, a promising tool was found for the analysis of factors triggering or preventing autoimmunity. The co-inhibitory molecule B7-H1 has been proposed to contribute to the maintenance of peripheral tolerance and to confine autoimmune inflammatory damage via the PD-1/B7-H1 pathway. Compared to Devic B7-H1+/+ mice, Devic B7-H1-/- mice developed clinical symptoms with a remarkably higher incidence rate and faster kinetics emphasized by deteriorated disease courses and a nearly quadrupled mortality rate. Remarkably enlarged immune-cell accumulation in the CNS of Devic B7-H1-/- mice, in particular of activated MOG-specific CD4+ T cells, correlated with the more severe clinical features. Our studies showed that the CNS not only was the major site of myelin-specific CD4+ T-cell activation but also that B7-H1 expression within the target organ significantly influenced T-cell activation and differentiation levels. Analysis at disease maximum revealed augmented accumulation of MOG-specific CD4+ T cells in the peripheral lymphoid organs of Devic B7-H1-/- mice partly due to increased T-cell proliferation rates. Transgenic MOG-specific B cells of Devic B7-H1-/- mice activated MOG-specific CD4+ T cells more efficiently than B cells of Devic B7-H1+/+ mice. This observation indicated a relevant immune-modulating role of B7-H1 on APCs (antigen-presenting cells) in this mouse model. We also assumed altered thymic selection processes to be involved in increased peripheral CD4+ T-cell numbers of Devic B7-H1-/- mice as we found more thymocytes expressing the transgenic MOG-specific T-cell receptor (TCR). Moreover, preliminary in vitro experiments hinted on an enhanced survival of TCRMOG-transgenic CD4+ T cells of Devic B7-H1-/- mice; a mechanism that might as well have led to higher peripheral T-cell accumulation. Elevated levels of MOG-specific CD4+ T cells in the periphery of Devic B7-H1-/- mice could have entailed the higher quantities in the CNS. However, mechanisms such as CNS-specific proliferation and/or apoptosis/survival could also have contributed. This should be addressed in future investigations. Judging from in vitro migration assays and adoptive transfer experiments on RAG-1-/- recipient mice, migratory behavior of MOG-specific CD4+ T cells of Devic B7-H1+/+ and Devic B7-H1-/- mice seemed not to differ. However, enhanced expression of the transmigration-relevant integrin LFA-1 on CD4+ T cells in young symptom-free Devic B7-H1-/- mice might hint on temporally differently pronounced transmigration capacities during the disease course. Moreover, we attributed the earlier conversion of CD4+ T cells into Th1 effector cells in Devic B7-H1-/- mice during the initiation phase to the lack of co-inhibitory signaling via PD-1/B7-H1 possibly leading to an accelerated disease onset. Full blown autoimmune inflammatory processes could have masked these slight effects of B7-H1 in the clinical phase. Accordingly, at peak of the disease, Th1 and Th17 effector functions of peripheral CD4+ T cells were comparable in both mouse groups. Moreover, judging from titers of MOG-specific IgG1 and IgM antibodies, alterations in humoral immunity were not detected. Therefore, clinical differences could not be explained by altered T-cell or B-cell effector functions at disease maximum. B7-H1 rather seemed to take inhibitory effect in the periphery during the initiation phase only and consistently within the target organ by parenchymal expression. Our observations indicate that B7-H1 plays a relevant role in the regulation of T-cell responses in this mouse model for spontaneous CNS autoimmunity. By exerting immune-modulating effects in the preclinical as well as the clinical phase of the disease, B7-H1 contributed to the confinement of the immunopathological tissue damage in Devic B7-H1+/+ mice mirrored by later disease onsets and lower disease scores. As a model for spontaneous autoimmunity featuring a close to 100 % incidence rate, the Devic B7-H1-/- mouse may prove instrumental in clarifying disease-triggering and -limiting factors and in validating novel therapeutic approaches in the field of autoimmune neuroinflammation, in particular the human Devic’s disease.
Durch die im Labor der AG Topp standardisierte Verfahren konnten im Rahmen dieser Arbeit stabile T Zell Linien aus primär humanen CMV spezifischen T Zellen und CMV spezifischen T Zellen mit chimärischen Rezeptoren hergestellt werden. Die Anzahl der chimärischen Rezeptoren auf den unterschiedlichen T Zellinien ist nicht signifikant unterschiedlich und beträgt ca. 17000 Rezeptoren pro Zelle. Bei der Überprüfung des Lyseverhaltens CMV spezifischer T Zellen gegenüber CMV spezifischer T Zellen transduziert mit einem chimärischen Rezeptor zeigt sich das gleiche Lyseverhalten. Um die zeitliche Dynamik dieses Verhaltens besser darzustellen, wurde ein FACS basierter Lysierungsversuch selbstständig entwickelt und etabliert. Somit konnte herausgestellt werden, dass das Lyseverhalten der T Zelllinien sowohl der CMV spezifischen T Zellen als auch das der Erst- und Zweitgeneration- T Zelllinien CAR CD19z und CAR CD19/28z vergleichbar war. Alle genannten Zelllinien lysieren ihre Targetzellen sowohl in der Quantität als auch in der zeitlichen Dynamik vergleichbar. Dies ist die einzige Gemeinsamkeit der verglichenen T Zell Aktivierungssysteme. In allen anderen funktionellen Versuchen ist die Aktivierung der T Zellen über einen chimärischen Rezeptor dem der physiologischen Aktivierung über den T Zell Rezeptor unterlegen. Der Proliferationsversuch mit CFSE zeigt eindeutig, dass nur die T Zellen, die über den TCR aktiviert werden, das Potential haben, sich zu teilen. Die T Zellen, die über den CAR sowohl der ersten als auch der zweiten Generation aktiviert werden, teilen sich nicht. Der Defekt der CAR T Zellen zeigt sich auch in der Hochregulation von CD25, einem Aktivierungsmarker für T Zellen. Die T Zellen, die durch die chimärischen Rezeptoren sowohl der ersten als auch der zweiten Generation aktiviert werden, regulieren CD25 nicht so stark nach oben wie die T Zellen, die physiologisch durch den TCR aktiviert werden. Diese Minderaktivierung spiegelt sich auch in der Zytokinproduktion wieder. Auch hier zeigt sich die unvollständige Aktivierung der CAR T Zellen. In der intrazellulären Zytokinmessung sieht man, dass bei CAR T Zellen der ersten und zweiten Generation zum einen die Einzelzellen weniger IFNy produzieren, zum anderen die Gesamtzahl der Zellen, die Zytokine produzieren, weniger ist als im Vergleich zur TCR Aktivierung. Dieses Phänomen der defizienten Aktivierung und der funktionellen Defekte von CAR T Zellen ist bekannt. Dies konnte noch einmal unter standardisierten und kontrollierten Bedingungen bestätigt werden. Durch die relativ neue Methode der durchflusszytometrischen Bead Technologie konnte dies nun zum ersten Mal realisiert werden. Es zeigte sich, dass ein signifikanter Unterschied zwischen den verschiedenen Aktivierungsmodi der T Zellen besteht. Eine physiologische Aktivierung von T Zellen führt zu einer höheren Maximum-Phosphorylierung als eine Aktivierung über den Erstgeneration CAR. Der signifikante Unterschied zeigt sich bei den MAP Kinasen ERK, JNK und p38. Somit ist dies ein Hinweis, dass die Signaltransduktion auf breiter Ebene aufgrund des alleinigen Vorhandenseins der Zeta Kette bei den CAR CD19z T Zellen, defizient ist und nicht ausreicht für eine volle Aktivierung, die eine volle Funktionalität der T Zelle nach sich ziehen würde. Diese unzureichende Aktivierung soll durch die kostimulatorische Komponente CD28 beim Zweitgenerationrezeptor CAR CD19/28z aufgehoben werden. In der Proliferation, Zytokineproduktion und den Aktivierungsmarkern zeigt sich keine Verbesserung im Vergleich zum Erstgenerationrezeptor bei CMV spezifischen T Zellen. Dieses Ergebnis wird bestätigt und korreliert damit, dass es keinen Unterschied in der maximalen Phosphorylierung von ERK, JNK und p38 zwischen Erstgeneration CAR CD19z und Zweitgeneration CAR CD19/28z gibt. Somit ist dieses System geeignet, um aus dem Phosphorylierungsstatus von MAP Kinasen von CAR T Zellen auf die Funktionalität dieser T Zellen zu schließen.
Echinococcus multilocularis besitzt als Lebergewebe - infiltrierender Parasit und Erreger der Alveolären Echinokokkose evolutionär konservierte Faktoren des TGF-β / BMP Signalsystems, die im Konzept der hormonellen Wirt - Parasit Kreuzkommunikation nicht nur hinsichtlich des Wirts - Einflusses auf die Entwicklung des Parasiten sondern auch umgekehrt in Bezug auf Immunantwort und Physiologie des Wirts eine mögliche regulierende Funktion besitzen. Die vorliegende Arbeit befasst sich primär mit den Auswirkungen von TGF-β / BMP Signaling auf den Fuchsbandwurm E. multilocularis. Dazu wurde zunächst durch Immunlokalisation das Vorliegen aller Echinokokken TGF-β / BMP Rezeptoren während einer natürlich aufgetretenen Alveolären Echinokokkose überprüft, sowie die funktionelle Interaktion von EmRSK3 und EmRSK4 mit humanem TGF-β1 in vitro nachgewiesen. Mit Hilfe von humanen Zytokinen (BMP2 und TGF β1) und Rezeptor - spezifischen Inhibitoren (SB431542, Dorsomorphin und LY364947) wurde die Rolle von TGF β / BMP Signaling in Bezug auf Vitalität, Wachstum, Differenzierung und Regeneration von E. multilocularis in verschiedenen Stadien untersucht. Zusätzlich wurde der Einfluss von humanem BMP2 und von SB431542 auf die Genexpression in E. multilocularis analysiert. Die Möglichkeit einer intra - Spezies Kommunikation durch Echinokokken Faktoren des TGF-β / BMP Signaling wurde ebenfalls adressiert. Dabei konnte gezeigt werden, dass Echinokokken TGF-β / BMP Rezeptoren in vitro durch sekretierte Faktoren aus Metazestoden aktiviert werden können. Auf Basis von Genomsequenzierungsdaten wurden zudem weitere Komponenten des TGF-β / BMP Signaling in E. multilocularis, wie die bisher unbekannten TGF β / BMP Homologe EmBMP2 und EmAct, die Noggin-like Homologe EmNlg1 und 2, ein I-Smad (EmSmadF) sowie ein zweiter BR-Smad (EmSmadE) identifiziert, strukturell charakterisiert und die jeweilige Genexpression in verschiedenen Stadien überprüft. EmSmadE wurde zudem in weiterführenden Studien funktionell charakterisiert. Fragestellungen bezüglich der Spezifität, Funktionalität und Möglichkeit der konstitutiven Aktivierung von TGF-β / BMP Signaling in E. multilocularis wurden auf Basis von EmSmadA, EmRSK3 und EmRSK3b in weiteren molekularbiologischen Studien untersucht und dienen dem tieferen Verständnis von TGF-β / BMP Signaling. In der vorliegenden Arbeit konnten damit alle grundlegenden Faktoren von TGF-β / BMP Signaling in E. multilocularis identifiziert werden und es wurden erstmals die eindeutige Sensierung von Wirts TGF-β / BMP Zytokinen durch Metazestoden sowie die Möglichkeit einer intra - Spezies Kommunikation über Echinokokken TGF-β / BMP Faktoren belegt. Zusätzlich deuten die neu identifizierten potenziell sekretierten TGF-β / BMP Faktoren auf eine mögliche Beeinflussung von Physiologie und Immunantwort des Wirts durch Echinokokken TGF-β / BMP Zytokine hin.
Transforming-Growth-Factor-beta1 (TGF-b1) is a multifunctional cytokine that regulates cell growth and differentiation in many types of cells. TGF-b1 is especially known to exert a variety of regulatory functions in the immune system, such as T cell differentiation and T cell function. Signal transduction of TGF-b1 is mediated by phosphorylation of receptorassociated Smad proteins (R-Smads). R-Smads are phosphorylated by the activated type I receptor, which is itself phosphorylated by the high affinity type II receptor upon ligand binding. The phosphorylated R-Smads then associate with Co-Smads. Heterooligomers of R- and Co-Smads translocate into the nucleus where they regulate transcription of target genes in concert with other transcription factors such as CBP/p300 or AP-1. Recent findings suggest that the pleiotropic effects of TGF-b1 are conferred by crosstalks to other signal transduction pathways such as the MAP-kinases or the STAT-pathway. Here we describe the effect of long-term exposure to TGF-b1 on the effector function of differentially stimulated primary murine splenocytes and purified primary murine CD8+ cytotoxic T cells. Long-term exposure to TGF-b1 results in non-responsiveness to TGF-b1- induced Smad2 phosphorylation. This is seen either by no phosphorylation or sustained phosphorylation of Smad2. Furthermore, we observed a strong correlation between sustained Smad2 phosphorylation and resistance to TGF-b1 mediated growth inhibition. In contrast, splenocyte cultures strongly growth inhibited by TGF-b1 showed no Smad2 phosphorylation. Lytic activity of these cultures, however, was found to be suppressed regardless of proliferation properties and Smad2 phosphorylation pattern. We also describe that a functional MEK-1 pathway is a prerequisite for rendering murine splenocytes unresponsive to TGF-b1 mediated growth inhibition, and that inhibition of the MEK-1 cascade alters the Smad2 phosphorylation pattern. In addition, we show that resistance to TGF-b1 mediated growth inhibition correlates with the activation of the JNK pathway. However, the resistant phenotype was found unable to be reverted upon administration of exogeneous IFNg and/or aCD28 antibody. In human or mouse T cell lines, however, the described correlation between the type of stimulation and TGF-b growth resistance or growth sensitivity is not present. Thus, this correlation is specific for primary T cells. We also cloned a chimeric dominantnegative TGF-b receptor which is coupled to a suicide gene, in order to render T cells resistant to TGF-b mediated effects.These findings shed light on how TGF-b1 mediates its immunosuppressive role, and may help to gain knowledge of averting these TGF-b1 effects in the course of tumor therapy.
Members of the RAF protein kinase family are key regulators of diverse cellular processes. The need for isoform-specific regulation is reflected by the fact that all RAFs not only display a different degree of activity but also perform isoform-specific functions at diverse cellular compartments. Protein-protein-interactions and phosphorylation events are essential for the signal propagation along the Ras-RAF-MEK-ERK cascade. More than 40 interaction partners of RAF kinases have been described so far. Two of the most important regulators of RAF activity, namely Ras and 14-3-3 proteins, are subject of this work. So far, coupling of RAF with its upstream modulator protein Ras has only been investigated using truncated versions of RAF and regardless of the lipidation status of Ras. We quantitatively analyzed the binding properties of full-length B- and C-RAF to farnesylated H-Ras in presence and absence of membrane lipids. While the isolated Ras-binding domain of RAF exhibit a high binding affinity to both, farnesylated and nonfarnesylated H-Ras, the full-length RAF kinases demonstrate crucial differences in their affinity to Ras. In contrast to C-RAF that requires carboxyterminal farnesylated H-Ras for interaction at the plasma membrane, B-RAF also binds to nonfarnesylated H-Ras in the cytosol. For identification of the potential farnesyl binding site we used several fragments of the regulatory domain of C-RAF and found that the binding of farnesylated H-Ras is considerably increased in the presence of the cysteine-rich domain of RAF. In B-RAF a sequence of 98 amino acids at the extreme N terminus enables binding of Ras independent of its farnesylation status. The deletion of this region altered Ras binding as well as kinase properties of B-RAF to resemble C-RAF. Immunofluorescence studies in mammalian cells revealed essential differences between B- and C-RAF regarding the colocalization with Ras. In conclusion, our data suggest that that B-RAF, in contrast to C-RAF, is also accessible for nonfarnesylated Ras in the cytosolic environment due to its prolonged N terminus. Therefore, the activation of B-RAF may take place both at the plasma membrane and in the cytosolic environment. Furthermore, the interaction of RAF isoforms with Ras at different subcellular sites may also be governed by the complex formation with 14-3-3 proteins. 14-3-3 adapter proteins play a crucial role in the activation of RAF kinases, but so far no information about the selectivity of the seven mammalian isoforms concerning RAF association and activation is available. We analyzed the composition of in vivo RAF/14-3-3 complexes isolated from mammalian cells with mass spectrometry and found that B-RAF associates with a greater variety of 14-3-3 proteins than C- and A-RAF. In vitro binding assays with purified proteins supported this observation since B-RAF showed highest affinity to all seven 14-3-3 isoforms, whereas C-RAF exhibited reduced affinity to some and A-RAF did not bind to the 14-3-3 isoforms epsilon, sigma, and tau. To further examine this isoform specificity we addressed the question of whether both homo- and heterodimeric forms of 14-3-3 proteins participate in RAF signaling. By deleting one of the two 14-3-3 isoforms in Saccharomyces cerevisiae we were able to show that homodimeric 14-3-3 proteins are sufficient for functional activation of B- and C-RAF. In this context, the diverging effect of the internal, inhibiting and the activating C-terminal 14-3-3 binding domain in RAF could be demonstrated. Furthermore, we unveil that prohibitin stimulates C-RAF activity by interfering with 14-3-3 at the internal binding site. This region of C-RAF is also target of phosphorylation as part of a negative feedback loop. Using tandem MS we were able to identify so far unknown phosphorylation sites at serines 296 and 301. Phosphorylation of these sites in vivo, mediated by activated ERK, leads to inhibition of C-RAF kinase activity. The relationship of prohibitin interference with 14-3-3 binding and phosphorylation of adjacent sites has to be further elucidated. Taken together, our results provide important new information on the isoform-specific regulation of RAF kinases by differential interaction with Ras and 14-3-3 proteins and shed more light on the complex mechanism of RAF kinase activation.
Die klassische Signaltransduktionskaskade, auch MAP Kinase Kaskade genannt, ist wesentlich an der Regulation zellulärer Vorgänge wie Proliferation, Differenzierung und Apoptose beteiligt. Proteinkinasen der Raf-Familie wirken dort als signalübertragende Elemente, welche Membranrezeptoren nachgeschaltet sind. Diese Proteine fungieren als Proto-Onkogene, eine Veränderung dieser Proteine kann sie in Onkogene überführen und sind wesentlich an der Krebsentstehung beteiligt. Während die Rolle von c-Raf als MEK-Aktivator innnerhalb des klassischen Signaltransduktionsweges gut charakterisiert ist, so ist nur wenig über die beiden anderen Isoformen A-Raf und B-Raf bekannt. Im Rahmen dieser Arbeit wurden zwei PC12 cDNA-Bibliotheken unter Verwendung des Two-Hybrid Systems mit A-Raf und mit c-Raf zur Isolierung neuer Raf-Interaktionspartner untersucht. Für c-Raf wurden die Wechselwirkungen mit den bekannten Interaktionspartnern bestätigt, es wurden jedoch keine neuen Bindungspartner identifiziert. Im A-Raf Two Hybrid Screen konnte zum einen Prolyl 4-Hydroxylase als Schlüsselenzym der Kollagensynthese isoliert werden. Es wurde keine Wechselwirkung zwischen Prolyl 4-Hydroxylase und c-Raf oder B-Raf beobachtet. Die Prolyl 4-Hydroxylase Bindungsstelle konnte innerhalb der N-terminalen variablen Region von A-Raf lokalisiert werden. Zum anderen wurde die Pyruvatkinase M2 als A-Raf spezifischer Bindungspartner identifiziert. Für diese Wechselwirkung war die c-terminale Region von A-Raf ausreichend. Durch Mutation zweier Aminosäuren im c-terminalen Teil von A-Raf konnte diese Wechselwirkung verhindert werden, wobei die Interaktion zu MEK und Ras dadurch nicht beeinträchtigt wurde. Ein kooperativer Effekt auf die Zelltransformation wurde durch Co-Transfektion von NIH Zellen mit onkogenem A-Raf und Pyruvatkinase M2 gezeigt. Diese führte zu doppelt so vielen Foci wie die Transfektion mit A-Raf alleine. Die Mutation der mutmaßlichen ATP-Bindungsstelle der Pyruvatkinase M2, welche die A-Raf Pyruvatkinase M2 Kooperation blockieren sollte, verhinderte diesen synergistischen Effekt. Diese Ergebnisse weisen auf eine Regulation der Pyruvatkinase M2 durch A-Raf hin und lassen den Schluss zu, dass die funktionelle Interaktion mit der Pyruvatkinase M2 durch onkogenes A-Raf für die Zelltransformation notwendig ist. Als zwei neue Raf-Interaktionspartner wurden Prolyl 4-Hydroxylase und Pyruvatkinase M2 identifiziert, welche Raf-Isoform spezifische Bindung zeigen. Auf diese Weise konnte eine direkte Verbindung zwischen der transformierenden MAP Kinase Kaskade und dem Energiestoffwechsel hergestellt werden.
Parasitic helminths share a large degree of common genetic heritage with their various hosts. This includes cell-cell-communication mechanisms mediated by small peptide cytokines and lipophilic/steroid hormones. These cytokines are candidate molecules for host-parasite cross-communication in helminth diseases. In this work the function of two evolutionary conserved signaling pathways in the model cestode Echinococcus multilocularis has been studied. First, signaling mechanisms mediated through fibroblast growth factors (FGF) and their cognate receptors (FGFR) which influence a multitude of biological functions, like homeostasis and differentiation, were studied. I herein investigated the role of EmFR which is the only FGFR homolog in E. multilocularis. Functional analyses using the Xenopus oocyte expression system clearly indicate that EmFR can sense both acidic and basic FGF of human origin, resulting in an activation of the EmFR tyrosine kinase domain. In vitro experiments demonstrate that mammalian FGF significantly stimulates proliferation and development of E. multilocularis metacestode vesicles and primary cells. Furthermore, DNA synthesis and the parasite’s Erk-like MAPK cascade module was stimulated in the presence of exogenously added mammalian FGF. By using the FGFR inhibitor BIBF1120 the activity of EmFR in the Xenopus oocyte system was effectively blocked. Addition of BIBF1120 to in vitro cultivated Echinococcus larval material led to detrimental effects concerning the generation of metacestode vesicles from parasite stem cells, the proliferation and survival of metacestode vesicles, and the dedifferentiation of protoscoleces towards the metacestode. In conclusion, these data demonstrate the presence of a functional EmFR-mediated signaling pathway in E. multilocularis that is able to interact with host-derived cytokines and that plays an important role in larval parasite development. Secondly, the role of nuclear hormone receptor (NHR) signaling was addressed. Lipophilic and steroid hormone signaling contributes to the regulation of metazoan development. By means of in silico analyses I demonstrate that E. multilocularis expresses a set of 17 NHRs that broadly overlaps with that of the related flatworms Schistosoma mansoni and S. japonicum, but also contains several NHR encoding genes that are unique to this parasite. One of these, EmNHR1, is homolog to the DAF-12/HR-96 subfamily of NHRs which regulate cholesterol homeostasis in metazoans. Modified yeast-two hybrid analyses revealed that host serum contains a ligand which induces homodimerization of the EmNHR1 ligand-binding domain. Also, a HNF4-like homolog, EmHNF4, was characterized. Human HNF4 plays an important role in liver development. RT-PCR experiments showed that both isoforms of the EmHNF4 encoding gene are expressed stage-dependently suggesting distinct functions of the two isoforms in the parasite. Moreover, specific regulatory mechanisms on the convergence of NHR signaling and TGF-β/BMP signaling pathways in E. multilocularis have been identified. On the one hand, EmNHR1 directly interacted with the EmSmadC and on the other hand EmHNF4b interacted with EmSmadD, EmSmadE which are all downstream signaling components of the TGF-β/BMP signaling pathway. This suggests cross-communication in order to regulate target gene expression. With these results, further studies on the role of NHR signaling in the cestode will be facilitated. Also, the first serum-free in vitro cultivation system for E. multilocularis was established using PanserinTM401 as medium. Serum-free co-cultivation with RH-feeder cells and an axenic cultivation method have been established. With the help of this serum-free cultivation system investigations on the role of specific peptide hormones, like FGFs, or lipophilic/steroid hormones, like cholesterol, for the development of helminths will be much easier.
Die NFκB-Signalwege, Apoptose und Nekroptose sind essentielle Prozesse in der Immunantwort. Außerdem sind diese Signalwege Teil der Regulation von Zelldifferenzierung, -proliferation, -tod und Entzündungsreaktionen. Dabei wird zuerst der Rezeptor (TNFR1 oder TRAILR 1/2) aktiviert, die rekrutierten DD-Adapterproteine TRADD, FADD und RIPK1 leiten dann die entsprechende Signalkaskade weiter und bestimmen durch ihre Zusammenwirkung, ob der NFκB-Signalweg, Apoptose oder Nekroptose induziert wird.
TNFR1 und TRAILR 1/2 benötigen die DD-Adapterproteine TRADD, FADD und RIPK1 für die Zelltodinduktion, deren konkrete Bedeutung in Bezug auf Rezeptor-Spezifität, Zusammenwirken und Relevanz allerdings noch unklar ist. Um das Zusammenspiel dieser Proteine besser zu verstehen, wurden in dieser Arbeit Nekroptose-kompetente RIPK3-exprimierende HeLa-Zellen verwendet, bei denen die DD-Adapterproteine FADD, TRADD und RIPK1 einzeln oder in Kombination von zweien ausgeknockt wurden. Es stellte sich heraus, dass RIPK1 essentiell für die TNFR1- und TRAILR 1/2-vermittelte Nekroptose-Induktion ist, doch RIPK1 alleine, d.h. ohne FADD- oder TRADD-Mitbeteiligung, nur bei der TNFR1-Nekroptose-Induktion ausreicht. Wiederum inhibiert TRADD die TNFR1- und TRAILR 1/2-induzierte Nekroptose. RIPK1 und TRADD sind aber unverzichtbar für die NFκB-Aktivierung durch TNFR1 oder TRAILR 1/2 und spielen eine wichtige Rolle bei TNFR1-induzierter Apoptose. Andererseits ist FADD alleine ausreichend für die TRAILR 1/2-bezogene Caspase-8 Aktivierung. Zudem ist FADD notwendig für die TRAIL-induzierte NFκB-Signalaktivierung. In Abwesenheit von FADD und TRADD vermittelt RIPK1 die TNF-induzierte Caspase-8 Aktivierung. FADD wird für die TRAIL-induzierte Nekroptose benötigt, aber gegenläufig wirkt die TNF-induzierte Nektroptose in einer Caspase-8 abhängigen und unabhängigen Weise. Zudem sensitiviert TWEAK die TNF- und TRAIL-induzierte Nekroptose.
Zusammenfassend wurde in dieser Arbeit die Auswirkung von TNFR1 und TRAILR 1/2 auf die Aktivierung der unterschiedlichen Signalkaskaden untersucht. Des Weiteren wurde gezeigt, in welcher Weise sich das Zusammenspiel von TRADD, FADD und RIPK1 auf die Induktion von NFκB, Apoptose und Nekroptose auswirkt.
Investigation on Distinct Roles of Smad Proteins in Mediating Bone Morphogenetic Proteins Signals

Bone morphogenetic proteins (BMPs) belong to the transforming growth factor-β (TGF-β) superfamily and play important roles in numerous biological events in the development of almost all multi-cellular organisms. Dysregulated BMP signaling is the underlying causes of numerous heritable and non-heritable human diseases including cancer. The vast range of biological responses induced by BMPs converges on three closely related Smad proteins that convey intracellular signals from BMP receptors to the nucleus. The specificity of BMP signaling has been intensively investigated at the level of ligand-receptor interactions, but how the different Smad proteins contribute to differential signals elicited by BMPs remains unclear. In this work, we investigated the BMP/Smad signaling in different aspects. In search for an appropriate fluorescence reporter in zebrafish, we compared different photo-switchable proteins and found EosFP the best candidate this model system for its fast maturation and fluorescence intensity. We modified and created appropriate vectors enabling Tol2-transposon based trangenesis in zebrafish, with which transgenic zebrafish lines were generated. We combined fluorescence protein tagging with high resolution microscopy and investigate the dynamics of Smad proteins in model system zebrafish. We observed that Smad5 undergoes nucleo-translocation as BMP signal transmitter during zebrafish gastrulation. We explored the Smad involvement during myogenic-to-osteogenic conversion of C2C12 cell line induced by BMP4. We created transient loss-of-function of Smads by siRNA-mediated knockdowns and analyzed the effects on these coupled yet distinct procedures by quantitative real-time PCR and terminal marker staining. We found that different Smad-complex stoichiometry might be responsible for distinct cellular signals elicited by BMPs.
In der vorliegenden Arbeit wurde das Thema "Die Rolle des Signalmoleküls Ca2+ in der Signaltransduktion der SC-1-induzierten Apoptose" anhand von immunhisto-chemischen, proteinbiochemischen und fluoreszenzmikroskopischen Methoden erarbeitet. Die SC-1-induzierte Apoptose stellt einen neuen intrinsischen, „death domain“-unabhängigen Signalweg in der Vermittlung des programmierten Zelltodes dar, der über die Bindung dieses Antikörpers an CD55SC-1 ausgelöst wird. Dem Signalmolekül Ca2+ galt besonderes Interesse bei der Charakterisierung der durch SC-1 ausgelösten Signaltransduktion. Nach Aktivierung von CD55SC-1 durch SC-1 steigt das intrazelluläre Ca2+ um Faktor 2,7 an, stabilisiert sich danach auf dem 1,5fachen Ausgangsniveau. Dies hat keinerlei Einfluss auf die Ausführung des Apoptoseprogramms, ist aber maßgeblich in das Expressionsverhalten involviert. Der Ca2+ - Einstrom ist somit nicht direkt in die Ausführung der Apoptose beteiligt, durch die Hochregulation der Expression des Apoptoserezeptors verstärkt es jedoch die Wahrscheinlichkeit der Tumorzelle, den programmierten Zelltod zu vollziehen, da durch die verstärkte Expression dieses Apoptose-induzierenden Rezeptors auch die Wahrscheinlichkeit steigt, dass mehrere SC-1-Antikörper binden können. Da die Quervernetzung mehrerer Rezeptoren mittels des SC-1-Antikörpers in der Apoptoseinduktion von Nöten ist, stellt diese durch Ca2+ getriggerte Überexpression von CD55SC-1 einen essentiellen Bestandteil in der Exekution dieses Selbstmordprogrammes dar. Dieses Ergebnis gibt einen interessanten Einblick in den Signalweg der durch SC-1 induzierten Apoptose. Weiterführende Experimente sind notwendig, um genauere Erkenntnisse dieser einzigartigen Form einer gewebespezifischen Apoptose zu erlangen.
Echinococcus multilocularis is the causative agent of alveolar echinococcosis (AE), a life-threatening disease with limited options of chemotherapeutic treatment. Anti-AE chemotherapy is currently based on a single class of drugs, the benzimidazoles. Although acting parasitocidic in vitro, benzimidazoles are merely parasitostatic during in vivo treatment of AE and cause severe site effects. In the case of operable lesions, the resection of parasite tissue needs to be supported by a prolonged chemotherapy. Thus, the current treatment options for AE are inadequate and require alternatives. In the present work, the flatworm signaling pathways were analyzed to establish potential targets for novel therapeutic approaches. I focused on factors that are involved in development and proliferation of E. multilocularis using molecular, biochemical and cell biological methods. Among the analysed factors were three MAP kinases of the parasite, EmMPK1, an Erk-1/2 orthologue, EmMPK2, a p38 orthologue and EmMPK3, an Erk7/8 orthologue. Further, I identified and characterized EmMKK2, a MEK1/2 orthologue of the parasite, which, together with the known kinases EmRaf and EmMPK1, forms an Erk1/2-like MAPK module. Moreover, I was able to demonstrate several influences of host growth factors such as EGF (epidermal growth factor) and insulin on worm signaling mechanisms and larval growth, including the phosphorylation of Elp, an ezrin-radixin-moesin like protein, EmMPK1, EmMPK3 and increased mitotic activity of Echinococcus cells. In addition, several substances were examined for their efficacy against the parasite including (i) general tyrosine kinase inhibitors (PP2, leflunamide), (ii) compounds designed to inhibit the activity of receptor tyrosine kinases, (iii) anti-neoplastic agents (miltefosine, perifosine), (iv) serine/threonine kinase inhibitors that have been designed to block the Erk1/2 MAPK cascade and (v) inhibitors of p38 MAPKs. In these studies, EmMPK2 proved to be a promising drug target for the following reasons. Amino acid sequence analysis disclosed several differences to human p38 MAPKs, which is likely to be the reason for the observed enhanced basal activity of recombinant EmMPK2 towards myelin basic protein in comparison to human recombinant p38 MAPK-α. In addition, the prominent auto-phosphorylation activity of the recombinant EmMPK2 protein together with the absence of an interaction with the Echinococcus MKKs suggest a different mechanism of regulation compared to the human enzyme. EmMPK2 activity could be effectively inhibited in vitro and in cultivated metacestode vesicles by treatment with SB202190 and ML3403, two ATP-competitive pyridinyl imidazole inhibitors of p38 MAPKs, in a concentration-dependent manner. Moreover, both compounds, in particular ML3403, caused parasite vesicle inactivation at concentrations which did not affect cultured mammalian cells. Likewise, during the cultivation of Echinococcus primary cells, the presence of ML3403 prevented the generation of new vesicles. Targeting members of the EGF signaling pathway, particulary of the Erk1/2-like MAPK cascade, with Raf and MEK inhibitors prevented the phosphorylation of EmMPK1 in metacestodes cultivated in vitro. However, although parasite growth was prevented under these conditions, the structural integrity of the metacestode vesicles maintained during long-term cultivation in the presence of the MAPK cascade inhibitors. Similar results were obtained when studying the effects of other drugs mentioned above. Taken together, several targets could be identified that reacted with high sensitivity to the presence of inhibitory substances, but did not cause the parasite’s death with one exception, the pyridinyl imidazoles. Based on the presented data, I suggest pyridinyl imidazoles as a novel class of anti-Echinococcus drugs and imply EmMPK2 as survival signal mediating factor, the inhibition of which could be used for the treatment of AE.
Der inhibitorische Einfluss von CD22 auf das B-Zellrezeptorsignal nach Stimulation der B-Zelle

CD22 ist ein B-zellspezifisches Transmembranprotein der Immunglobulin (Ig)-Superfamilie. Es übernimmt zwei unterschiedliche Aufgaben. Zum einen hat CD22 eine inhibitorische Wirkung auf das BZR-Signal. Nach BZR-Ligation lagert sich die Tyrosin-Phosphatase SHP-1 an die cytoplasmatische Domäne von CD22 an, wodurch SHP-1 aktiviert wird. Durch die dephosphorylierende Aktivität der Phosphatase moduliert sie das BZR-Signal. Zum anderen ist CD22 ein Adhäsionsmolekül, das in die Gruppe der Siglecs (Sialic acid-binding Ig-like lectin) gehört. Die N-terminale Ig-Domäne von CD22 weist die Bindungseigenschaften eines Lectins auf und bindet spezifisch 2,6-gekoppelte Sialinsäure. Das Ziel der vorliegenden Doktorarbeit war es, die inhibitorische Wirkung von CD22 auf das BZR-Signal molekular zu definieren. Daher wurde das Substrat der an CD22 rekrutierten und aktivierten Phosphatase SHP-1 in vergleichenden Analysen von CD22-defizienten und Kontroll-B-Zellen nach BZR-Stimulation gesucht. Wir konnten zeigen, dass in CD22-defizienten B-Zellen nach BZR-Stimulation das Adaptermolekül SLP-65, auch BLNK oder BASH genannt, früher und stärker Tyrosin-phosphoryliert vorliegt, als in Kontroll-B-Zellen. Transfektionsexperimente mit der CD22-defizienten Plasmazytom-Zelllinie J558Lm3 wurden begonnen, um den molekularen Zusammenhang zwischen CD22, SHP-1 und SLP-65/BLNK zu bestätigen. Das in J558Lm3 Zellen ektopisch exprimierte CD22 wurde Tyrosin-phosphoryliert, und SHP-1 konnte mit CD22 co-präzipitiert werden. Jedoch war in den CD22-Transfektanten keine Reduktion der Tyrosin-Phosphorylierung von SLP-65/BLNK nach BZR-Stimulation im Vergleich zu untransfizierten Zellen nachweisbar. Da in unabhängigen Experimenten die Liganden-Bindung von CD22 als Voraussetzung für die inhibitorische Wirkung von CD22 deutlich wurde, etablierten wir 2,6 Sialinsäure- und CD22-positive J558Lm3 Doppeltransfektanten. Auf diesen konnte die cis-Maskierung von CD22 nachgewiesen werden. In Stimulationsexperimenten der Doppeltransfektanten wurde die reduzierte Tyrosin-Phosphorylierung von SLP-65/BLNK in Abhängigkeit von CD22 in der Mehrzahl der Klone bestätigt. Allerdings mussten die Resultate in Frage gestellt werden, als in den meisten Klonen einer Kontrolltransfektion ebenfalls eine Reduktion der Tyrosin-Phosphorylierung von SLP-65/BLNK festgestellt wurde. Um den Einfluss von CD22 und SLP-65 auf das BZR-Signal zu klären, wurden CD22- und SLP-65-defiziente Mäuse gekreuzt. Durch die zusätzliche Deletion von CD22 in SLP-65-/- Mäusen konnte das Ca2+-Signal nach BZR-Stimulation wieder hergestellt werden. Jedoch zeigte die weitere Analyse der doppel-defizienten Mäuse, dass in der Regel der Phänotyp der SLP-65-defizienten Mäuse dominiert. Diese Ergebnisse verdeutlichten, dass das Adaptermolekül SLP-65/BLNK zwar ein Substrat des CD22/SHP-1 Signalweges ist, aber keine essentielle Rolle in der inhibitorischen Wirkung von CD22 übernimmt. Weiterhin wurde der Einfluss der Ligandenbindung von CD22 auf dessen intrazelluläre, inhibitorische Wirkung auf das BZR-Signal untersucht. Ein synthetisches Sialoside stand zur Verfügung, das hochspezifisch die Interaktion von CD22 mit dessen Liganden stört. Wurden die Zellen einer humanen B-Zelllinie in Gegenwart des Sialosids über den BZR stimuliert, konnte ein erhöhtes Ca2+-Signal gemessen werden. Dieses Resultat erinnerte an die stärkere Ca2+-Mobilisierung in CD22-defizienten B-Zellen. Entsprechend war die Tyrosin-Phosphorylierung von CD22 nach Vorbehandlung mit dem Sialosid in den humanen B-Zellen verringert, und weniger SHP-1 konnte mit CD22 co-präzipitiert werden. Mit diesen Ergebnissen wurde deutlich, dass die Adhäsionsdomäne von CD22 einen direkten, positiven Einfluss auf die intrazelluläre, inhibitorische Domäne von CD22 hat. Als Nebenprojekt wurde die Rolle von CD22 in knock-out Mäusen des transkriptionellen Co-Aktivators BOB.1/OBF.1 untersucht. Ein Entwicklungsblock im transitionalen B-Zellstadium im Knochenmark der BOB.1/OBF.1-defizienten Mäuse verursacht eine deutliche Reduktion reifer B-Zellen in der Milz. Die Analyse des Knochenmarks der BOB.1/OBF.1-defizienten Mäuse zeigte, dass ausschließlich die Expression von CD22 auf den B-Vorläufer Zellen erhöht war. Nach zusätzlicher Deletion von CD22 in BOB.1/OBF.1-/- Mäusen war nach BZR-Stimulation ein deutliches Ca2+-Signal in den doppel-defizienten B-Zellen messbar. Dieses könnte die normalisierte Anzahl transitionaler B-Zellen im Knochenmark und die gestiegene Anzahl reifer B-Zellen in der Milz der doppel-defizienten Mäuse bewirken. Allerdings waren die doppel-defizienten Mäuse, entsprechend den BOB.1/OBF.1-/- Mäusen, nicht in der Lage, eine humorale Immunantwort einzuleiten oder Keimzentren zu bilden. Die Proliferation von CD22-defizienten B-Zellen nach LPS-Stimulation verlief unabhängig von der An- oder Abwesenheit von BOB.1/OBF.1. Mit den Untersuchungen konnten wir zeigen, dass die Differenzierungsschwierigkeiten der BOB.1/OBF.1-defizienten B-Zellen vom BZR-Signal abhängen. Allerdings muss das Ausbleiben der Keimzentrenbildung auf einen anderen Mechanismus zurückgeführt werden.
Whereas G-protein coupled receptors (GPCRs) have been long believed to signal through cyclic AMP exclusively at cell surface, our group has previously shown that GPCRs not only signal at the cell surface but can also continue doing so once internalized together with their ligands, leading to persistent cAMP production. This phenomenon, which we originally described for the thyroid stimulating hormone receptor (TSHR) in thyroid cells, has been observed also for other GPCRs. However, the intracellular compartment(s) responsible for such persistent signaling and its consequences on downstream effectors were insufficiently characterized. The aim of this study was to follow by live-cell imaging the trafficking of internalized TSHRs and other involved signaling proteins as well as to understand the consequences of signaling by internalized TSHRs on the downstream activation of protein kinase A (PKA). cAMP and PKA
activity was measured in real-time in living thyroid cells using FRET-based sensors Epac1-camp and AKAR2 respectively. The results suggest that TSH co-internalizes with its receptor and that the internalized TSH/TSHR complexes traffic retrogradely to the trans-Golgi network (TGN). This study also provides evidence that these internalized TSH/TSHR complexes meet an intracellular pool of Gs proteins in sorting endosomes and in TGN and activate it there, as visualized in real-time using a conformational biosensor nanobody, Nb37. Acute Brefeldin A-induced Golgi collapse hinders the retrograde trafficking of TSH/TSHR complexes, leading to reduced cAMP production and PKA signaling. BFA pretreatment was also able to attenuate CREB phosphorylation suggesting that an intact Golgi/TGN organisation is essential
for an efficient cAMP/PKA signaling by internalized TSH/TSHR complexes. Taken together this data provides evidence that internalized TSH/TSHR complexes meet and activate Gs proteins in sorting endosomes and at the TGN, leading to a local activation of PKA and consequently increased CREB activation. These findings suggest unexpected functions for receptor internalization, with major pathophysiological and pharmacological implications.
Mechanismen der Toxoplasma-gondii-vermittelten Inhibierung der Apoptose in humanen Wirtszelllinien

Als obligat intrazellulärer Parasit und Erreger von lebenslang persistierenden Infektionen in Mensch und Tier dürfte Toxoplasma gondii von der Integrität seiner Wirtszelle in besonderem Maße abhängig sein. Ziel der Arbeit war es, den Einfluss des Parasiten auf die Wirtszellapoptose zu untersuchen. T. gondii inhibiert die in vitro induzierte Apoptose in humanen HL-60- und U937-Zellen. Dabei muss der Parasit aktiv in die Zelle eindringen, jedoch nicht in dieser replizieren können. Er interferiert dabei mit mindestens zwei Komponenten der Apoptose-Signalkaskade: Erstens vermindert T. gondii die Herunterregulation der Mcl-1-Expression nach Apoptoseinduktion. Das führt dazu, dass trotz Apoptoseinduktion die Translokation von Cytochrom c aus den Mitochondrien in das Zytoplasma inhibiert wird und daraufhin die Caspasen 9 und 3 sowie deren Substrate weniger stark aktiviert werden. Zweitens wird die Expression der Poly-(ADP-Ribose)Polymerase (PARP) durch T. gondii inhibiert. Beide Mechanismen könnten an der Inhibierung der Wirtszellapoptose durch T. gondii beteiligt sein und dem Parasiten damit sein intrazelluläres Überleben sichern.
The Transforming Growth Factor (TGF) superfamily of cytokines and their serine/threonine kinase receptors play an important role in the regulation of cell division, differentiation, adhesion, migration, organization, and death. Smad proteins are the major intracellular signal transducers for the TGF receptor superfamily that mediate the signal from the membrane into the nucleus. Bone Morphogenetic Protein-4 (BMP-4) is a representative of the TGF superfamily, which regulates the formation of teeth, limbs and bone, and also plays a role in fracture repair. Binding of BMP-4 to its receptor stimulates phosphorylation of Smad1, which subsequently recruits Smad4. A hetero-oligomeric complex consisting of Smad1 and Smad4 then translocates into the nucleus and regulates transcription of target genes by interacting with transcription factors. Although the individual steps of the signaling cascade from the receptor to the nucleus have been identified, the exact kinetics and the rate limiting step(s) have remained elusive. Standard biochemical techniques are not suitable for resolving these issues, as they do not offer sufficiently high sensitivity and temporal resolution. In this study, advanced optical techniques were used for direct visualization of Smad signaling in live mammalian cells. Novel fluorescent biosensors were developed by fusing cyan and yellow fluorescent proteins to the signaling molecules Smad1 and Smad4. By measuring Fluorescence Resonance Energy Transfer (FRET) between the two fluorescent proteins, the kinetics of BMP/Smad signaling was unraveled. A rate-limiting delay of 2 - 5 minutes occurred between BMP receptor stimulation and Smad1 activation. A similar delay was observed in the complex formation between Smad1 and Smad4. Further experimentation indicated that the delay is dependent on the Mad homology 1 (MH1) domain of Smad1. These results give new insights into the dynamics of the BMP receptor – Smad1/4 signaling process and provide a new tool for studying Smads and for testing inhibitory drugs.
Klassischerweise ist der Aldosteron-gebundene MR an der Regulation des Blutdruckes und des Wasser-Elektrolyt-Haushaltes beteiligt. Neuere klinische Studien zeigen allerdings, dass Aldosteron auch an pathophysiologischen Remodelingprozessen im kardiovaskulären und renalen System mitwirkt. Die zugrundeliegenden Mechanismen sind noch weitgehend unbekannt. Der EGFR ist ein Wachstumsfaktor und heterologer Signaltransduktor für G-Protein-gekoppelte Rezeptoren von beispielsweise Angiotensin II, Phenylephrin und Endothelin-1. In der Literatur gibt es Hinweise für eine Interaktion zwischen den Signaltransduktionswegen von Aldosteron/MR und EGFR. So können Mineralocorticoide nach zerebraler Ischämie zu einem vermehrten vaskulären Remodeling und einem Anstieg der EGFR-mRNA-Konzentration führen und außerdem eine EGF-induzierte Vasokonstriktion verstärken. Daher wäre eine mögliche Erklärung für die pathophysiologische Wirkung von Aldosteron eine Induktion der EGFR-Expression mit vermehrter Wirksamkeit von vasoaktiven Peptiden. Um diese Hypothese zu überprüfen untersuchten wir in verschiedenen Modellsystemen, ob Aldosteron die EGFR-Proteinexpression erhöht. Dies war sowohl im heterologen CHO-Expressionsystem also auch in MR-exprimierenden Zelllinien und Primärkulturen der Fall. Auch in adrenalektomierten Ratten mit osmotischen Minipumpen bestätigte sich die Aldosteron-induzierte EGFR-Expression in der Aorta, im linken Herzen und der Niere. Über den eng verwandten Glucocorticoidrezeptor ließ sich keine EGFR-Expressionssteigerung auslösen, so dass es sich um einen MR-spezifischen Effekt handelt. Zur Charakterisierung des zugrundeliegenden molekularen Mechanismus, der besonders für therapeutische Interventionen von Interesse ist, wurde die Promotoraktivität des EGFR untersucht. Es zeigte sich bei Aldosteroninkubation eine gesteigerte EGFR-Promotoraktivität im Reporter-Gen-Assay. Die beteiligten Promotoranteile konnten mit Deletionskonstrukten auf zwei DNA-Fragmente eingegrenzt werden. Von Seiten des MR ist die A/B-Domäne für die Interaktion bedeutend, denn ein trunkierter MR mit den Domänen C, D, E und F genügt nicht, um den EGFR-Promoter vollständig zu aktivieren. Um Hinweise für die physiologische und pathophysiologische Bedeutung der Interaktion zwischen MR und EGFR zu erhalten, untersuchten wir sowohl den Einfluß auf die Bildung von Extrazellulärmatrix in glatten Gefäßmuskelzellen als auch auf die Natriumresorption im Sammelrohr der Niere. Als Anhaltspunkt für die vermehrte Bildung von extrazellulärer Matrix wie sie bei Remodelingprozessen vorkommt, quantifizierten wir die Fibronektinsekretion in glatten Muskelzellen der humanen Aorta (HAoSMC). Nach Aldosteroninkubation und besonders bei Koinkubation mit EGF zeigte sich eine vermehrte Fibronektinsekretion ins Medium, die sich durch Hemmer der EGFR-Kaskade normalisieren ließ. Dies unterstützt die Hypothese, dass die Aldosteron-EGFR-Interaktion an der Entstehung von Remodelingprozessen im kardiovaskulären und renalen System beteiligt ist. Neben einem Einfluss auf die Entstehung pathophysiologischer Prozesse im kardiovaskulären und renalen System kommt es über eine Aldosteron-induzierte EGFR-Expression im Sammelrohr der Niere auch zu physiologischen Effekten, nämlich einer Hemmung der Natriumresorption. Diese wirkt der klassischerweise durch Aldosteron vermittelten vermehrten Natriumresoprtion über den epithelialen Natriumkanal (ENaC) entgegen und könnte daher als negative Feedbackschleife Dauer und Ausmaß der Aldosteron-induzierten Natriumresorption limitieren. Zusätzlich zu den klassischen genomischen Wirkungen zeigen Steroide nicht-genotrope Effekte. Beim Aldosteron führen diese MR- und EGFR-vermittelt zu einer Aktivierung der ERK1/2- und JNK-1/2-Kinasen. Die nicht-genotrope Aldosteron-induzierte ERK-Aktivierung ist ferner durch c-Src-Inhibitoren hemmbar und führt zu einer Stimulation der Kerntranslokation des MR. Nicht-genotrope Effekte können folglich unter Beteiligung der EGFR-Signalkaskade die genomischen modulieren. Aldosteron führt ebenfalls zu einem Anstieg der zytosolischen Calciumkonzentration, allerdings ist dieser Effekt unabhängig vom MR. Hieraus folgt, dass die nicht-genotropen Effekte teilweise MR-vermittelt und teilweise MR-unabhängig sind. Insgesamt konnte also auf verschiedenen Ebenen eine Interaktion zwischen Aldosteron/MR und der EGFR-Signalkaskade gezeigt werden, mit Hinweisen für eine Bedeutung bei sowohl physiologischen als auch pathophysiologische Vorgängen.
Der Transkriptionsfaktor Stat6 vermittelt zentrale Wirkungen von IL-4 und IL-13, die in der Pathologie atopischer Erkrankungen eine Rolle spielen. Seine Spezifität für diese beiden allergieassoziierten Cytokine ist eine wesentliche Motivation ihn näher zu untersuchen. In dieser Arbeit sollte mehr über die Funktion von Stat6 herausgefunden werden. Außerdem wurden Möglichkeiten untersucht dieses Verhalten zu beinflussen. Einen Schwerpunkt der Arbeit bildete die Regulation des Eotaxin-1-Promotors. Eotaxin-1 ist einer der stärksten Rekrutierungsfaktoren für Eosinophile, die eine zentrale Rolle bei der Immunpathologie allergischer Erkrankungen spielen. Mit Hilfe der Daten konnte eine neue Hypothese zur Regulation des Eotaxin-1-Promotors entwickelt werden. Zum Vergleich wurde mit der Untersuchung des Promotors eines weiteren Chemokins, des MCP-4, begonnen. In Zusammenarbeit mit Dr. Sascha Stolzenberger wurde ein Weg untersucht den Stat6-Signalweg zu hemmen. Dabei wurden mit Hilfe des Antennapedia-Peptides Stat6-Bindepeptide in die Zelle transportiert, um dort über eine kompetitive Hemmung die Signaltransduktion zu unterbinden. Ergebnis dieser Arbeiten ist ein hochspezifischer, aber nur transient wirkender Stat6 Inhibitor. Die Stat6/DNA-Wechselwirkung wurde mit der Magnetobead-Technik untersucht. Dabei werden Promotorfragmente an Magnetkügelchen gekoppelt und unter Ausnutzung der Magnetisierung an die DNA bindende Proteine isoliert und über SDS-PAGE/Immunoblotanalyse untersucht. Mit dem Verfahren konnte die Stat6-Bindung an acht verschiedene Promotoren nachgewiesen werden. In Zusammenarbeit mit der Arbeitsgruppe Pallardy aus Paris wurde die Wechselwirkung von Stat6 mit dem Glucocorticoid-Rezeptor untersucht. Glucocorticoide kontrollieren Entzündungen und Interaktionen des aktivierten Rezeptors mit anderen Proteinen aus der Stat-Familie sind seit längerem bekannt. Wie in dieser Arbeit gezeigt wurde, interagiert Stat6 mit dem Glucocorticoidrezeptor unabhängig von einer Bindung an DNA. Zusätzlich wurde der Mucin-2-Promotor auf Stat6-Regulierung untersucht. Mucine sind wichtige Bestandteile des Schleimes. Verstärkte Schleim-Sekretion ist ein klinisches Symptom asthmatischer Erkrankungen und trägt zur Zerstörung der Lunge bei. Ein potentiell Stat6 reguliertes Fragment aus dem Mucinpromoter wurde mit Hilfe von PCR-Techniken isoliert und in Reportergenvektoren kloniert.
In a variety of established tumour cell lines, but also in primary mammary epithelial cells metalloprotease-dependent transactivation of the EGFR, and EGFR characteristic downstream signalling events were observed in response to stimulation with physiological concentrations of GPCR agonists such as the mitogens LPA and S1P as well as therapeutically relevant concentrations of cannabinoids. Moreover, this study reveals ADAM17 and HB-EGF as the main effectors of this mechanism in most of the cancer cell lines investigated. However, depending on the cellular context and GPCR agonist, various different members of the ADAM family are selectively recruited for specific ectodomain shedding of proAR and/or proHB-EGF and subsequent EGFR activation. Furthermore, biological responses induced by LPA or S1P such as migration in breast cancer and HNSCC cells, depend on ADAM17 and proHB-EGF/proAR function, respectively, suggesting that highly abundant GPCR ligands may play a role in tumour development and progression. Moreover, EGFR signal transactivation could be identified as the mechanistic link between cannabinoid receptors and the activation of mitogen activated protein kinases (MAPK) ERK1/2 as well as pro-survival Akt/PKB signalling. Depending on the cellular context, cannabinoid-induced signal cross-communication was mediated by shedding of proAmphiregulin and/or proHB-EGF by ADAM17. Most importantly, our data show that concentrations of THC comparable to those detected in the serum of patients after THC administration accelerate proliferation of cancer cells instead of apoptosis and thereby may contribute to cancer progression in patients.
Bone Morphogenetic Proteins (BMPs) are secreted multifunctional signaling proteins that play an important role during development, maintenance and regeneration of tissues and organs in almost all vertebrates and invertebrates. BMPs transmit their signals by binding to two types of serine-/threonine-kinase receptors. BMPs bind first to their high affinity receptor, thereby recruiting their low affinity receptor into the complex. This receptor assembly starts a Smad (Small mothers against decapentaplegic) protein signaling cascade which regulates the transcription of responsive genes. Up to date, only seven type I and five type II receptors are known for more than 30 ligands. Therefore, many BMP ligands can recruit more than one receptor subtype. Vice versa, receptors can bind to several ligands, indicating a highly promiscuous ligand-receptor interaction. This raises the following questions: (i) How are BMPs able to induce ligand-specific signals, despite forming complexes with identical receptor composition and (ii) how are they able to recognize and bind various binding partners in a highly specific manner. From the ligand’s point of view, heterodimeric BMPs are valuable tools for studying the interplay between different sets of receptors, thereby providing new insights into how the various BMP signals can be generated. This study describes the expression and purification of the heterodimers BMP-2/6 and -2/7 from E.coli cells. BIAcore interaction studies and various in vitro cell activity assays revealed that the generated heterodimers are biologically active. Furthermore, BMP-2/6 and -2/7 exhibit a higher biological activity in most of the cell assays compared to their homodimeric counterparts. In addition, the BMP type I receptor BMPR-IA is involved in heterodimeric BMP signaling. However, the usage of other type I receptor subtypes (e.g. ActR-I) building a heteromeric ligand-receptor type I complex as indicated in previous works could not be determined conclusively. Furthermore, BMP heterodimers seem to require only one type I receptor for signaling. From the receptors’ point of view, the BMP type I receptor BMPR-IA is a prime example for its promiscuous binding to different BMP ligands. The extracellular binding interface of BMPR-IA is mainly unfolded in its unbound form, requiring a large induced fit to adopt the conformation when bound to its ligand BMP-2. In order to unravel whether the binding promiscuity of BMPR-IA is linked to structural plasticity of its binding interface, the interaction of BMPR-IA bound to an antibody Fab fragment was investigated. The Fab fragment was selected because of its ability to recognize the BMP-2 binding epitope on BMPR-IA, thus neutralizing the BMP-2 mediated receptor activation. This study describes the crystal structure of the complex of the extracellular domain of BMPR-IA bound to the antibody Fab fragment AbyD1556. The crystal structure revealed that the contact surface of BMPR-IA overlaps extensively with the contact surface of BMPR-IA for BMP-2 interaction. Although the contact epitopes of BMPR-IA to both binding partners coincide, the three-dimensional structures of BMPR-IA in both complexes differ significantly. In contrast to the structural differences, alanine-scanning mutagenesis of BMPR-IA showed that the functional determinants for binding to both the antibody and BMP-2 are almost identical. Comparing the structures of BMPR-IA bound to BMP-2 or to the Fab AbyD1556 with the structure of unbound BMPR-IA revealed that binding of BMPR-IA to its interaction partners follows a selection fit mechanism, possibly indicating that the ligand promiscuity of BMPR-IA is inherently encoded by structural adaptability.
MAPK Kaskaden spielen eine Schlüsselrolle bei der Übertragung und der Umwandlung von extrazellulären Reizen in intrazelluläre Signale und regulieren dadurch unterschiedliche Prozesse wie Differenzierung, Zellproliferation oder Stress Antworten. 'Mixed Lineage Kinasen' (MLKs) bilden eine Familie von Serin/Threonin Proteinkinasen mit mehreren Protein/Protein Interaktionsdomänen (SH3, Cdc42 Rac interactive binding sequence/CRIB, Leuzin Zipper), die am engsten mit der Familie der MAPKK Kinasen verwandt ist. In der voliegenden Arbeit zeigen wir, dass MLK3 Cdc42/Rac induzierte JNK/SAPK Aktivität vermittelt, welches zu einer anschließenden Phosphorylierung und verstärkter transkriptioneller Aktivität des zur AP-1 Familie gehörenden immediate early Gens c-Jun führt. Zusätzlich phosphoryliert MLK3 die dualspezifischen Kinasen MEK1/2 direkt in vitro und in vivo an den für die Aktivierung wichtigen Serinen 217/221. Wohingegen dies nur zu einer ERK Aktivierung in Überexpressionssystemen führte, demonstrieren die Analysen überzeugend, dass endogenes MEK1/2, welches von MLK3 phosphoryliert worden ist, nicht in der Lage ist, diese Aktivität auf das physiologische Substrat ERK1/2 in vitro wie auch in vivo zu übertragen. Wir postulieren daher, dass MLK3 vermittelte MEK1/2 Phosphorylierung die dualspezifischen Kinasen von ERK1/2 Aktivierung entkoppelt. Zusätzlich zeigen wir, dass Überexpression von Wild Typ MLK3 zur morphologischen Transformation von NIH 3T3 Fibroblasten und Wachstum in Soft Agar führt. In Übereinstimmung mit den anderen Analysen ist MEK1/2 stark phosphoryliert jedoch von ERK1/2 Aktivierung in MLK3 transformierten Zellen entkoppelt. Weiterhin kooperiert MLK3 mit aktivierter MEK1 in Foci Bildung, was zeigt, dass MLK3 andere Signalwege als ERK1/2 in der Zelltransformation benutzt. Vielmehr ist in MLK3 transformierten Fibroblasten Wachstumsfaktor-induzierte ERK1/2 Aktivierung und Expression des immediate early Gens junB partiell blockiert. Unsere Analysen zeigen zum ersten Mal für eine MAPKK Kinase, dass sie MAPK Signalwege unterschiedlich reguliert. Während MLK3 auf der einen Seite JNK/SAPK aktiviert, entkoppelt es MEK1/2 induzierte Phosphorylierung von ERK1/2 Aktivierung und blockiert sogar Wachstumsfaktor-induzierte ERK1/2 Ativierung partiell.
BMPs influence a variety of cellular processes. They have been shown to regulate proliferation, differentiation, migration and apoptosis and thus play central roles during developmental processes and tissue homeostasis. Ligand mediated signal transduction is transmitted via BMP type I and BMP type II receptors, both members of the serine/threonine kinase superfamily. The BMP receptor mediated signal transduction is not explored in detail. Therefore our aim was to address different aspects of BMP mediated signal transduction with main focus on BRII and its regulation. Due to the existence of two alternative splice variants, a long and a short form, the function of the two variants and the impact of the C-terminal extension are of general interest. Moreover, mutations in the BMPR2 gene were identified to be responsible for PPH, a autosomal dominant lung disease. In this thesis, BRII phosphorylation and signalling mediated by different receptor oligomers were investigated and multiple BRII associated proteins were identified. We could show that the oligomerization pattern of BMP receptors exhibits a higher degree of flexibility compared to other receptors of that superfamily. In the present work the BMP2 mediated signal transduction should be examined, depending on the receptor oligomerization pattern. Using kinase-deficient mutants, it could be demonstrated, that signalling via preformed BMP receptor complexes is mediated by the well characterized Smad1/5/8 pathway, whereas signalling initiated by BMP2 induced recruitment of the receptors activates the p38 pathway and leads to Alkaline Phosphatase production. To further study signalling events triggered directly from the BRII a proteomics-based screen for BRII associated proteins was performed. 53 associated proteins were found, the majority being signal transducing molecules, but in addition metabolic proteins, transcriptional regulators and others were identified. These proteins enable to gain a deeper insight in BMP mediated signalling. One of the interactors, the receptor tyrosine kinase c-kit, was characterized in more detail. It could be demonstrated, that BRII and c-kit form a complex in vitro and in vivo, and the interaction is enhanced upon BMP2 stimulation. 2D phosphopeptid mapping showed that BRII is phosphorylated at S757 upon activation of c-kit by SCF. Moreover, c-kit and its ligand SCF are modulating BMP2 pathways, by enhancing Smad1/5 phosphorylation, Smad-transcriptional activity, Alkaline Phosphatase production and expression of Cbfa1. All these pathways hint towards modulation of the osteoblast development via c-kit. Thus, we were able to develop a novel paradigm for the BMP2 meditated signalling. One of the initial triggers for BRII is the auto-phosphorylation of BRII. Here we analyze ligand-independent as well as ligand-dependent phosphorylation of BRII. Some phosphorylation sites in BRII were identified. The general phosphorylation occurs mostly on serines. S815, S818 and Y825 are identified targets of phosphorylation whose function is still unclear. However phosphorylation of S336 is demonstrated to be essential for BRII activation. The elucidation of BMP receptor phosphorylation and oligomerization as well as the impact of a number of BRII associated proteins (such as c-kit), demonstrated in this thesis that BMP signalling has to be regulated precisely on multiple levels. This can be useful for the development of selective signalling inhibitors for basic research and therapeutic approaches of PPH and other diseases.
Platelet activation and aggregation at sites of vascular injury is critical to prevent excessive blood loss, but may also lead to life-threatening ischemic diseases, such as myocardial infarction and stroke. Extracellular agonists induce platelet activation by stimulation of platelet membrane receptors. Signal transduction results in reorganization of the cytoskeleton, shape change, platelet adhesion and aggregation, cumulating in thrombus formation. Several Rho GTPases, including Rac1, Cdc42 and RhoA, are essential mediators of subsequent intracellular transduction of ITAM- and GPCR-signaling. Therefore, inhibition or knockout can result in severely defective platelet signaling.
Mice with platelet specific Rac1-deficiency are protected from arterial thrombosis. This benefit highlights further investigation of Rac1-specific functions and its potential as a new pharmacological target for prevention of cardiovascular diseases. Two newly developed synthetic compounds, NSC23766 and EHT1864, were proposed to provide highly specific inhibition of Rac1 activity, but both drugs have never been tested in Rac1-deficient cell systems to rule out potential Rac1-independent effects.
This study revealed significant off-target effects of NSC23766 and EHT1864 that occurred in a dose-dependent fashion in both wild-type and Rac1-deficient platelets. Both inhibitors individually affected resting platelets after treatment, either by altering membrane protein expression (NSC23766) or by a marked decrease of platelet viability (EHT1864). Platelet apoptosis could be confirmed by enhanced levels of phosphatidylserine exposure and decreased mitochondrial membrane potential. Phosphorylation studies of the major effector proteins of Rac1 revealed that NSC23766 and EHT1864 abolish PAK1/PAK2 activation independently of Rac1 in wild-type and knockout platelets, which may contribute to the observed off-target effects.
Additionally, this study demonstrated the involvement of Rac1 in G protein-coupled receptor-mediated platelet activation and GPIb-induced signaling. Furthermore, the data revealed that Rac1 is dispensable in the process of integrin IIb 3-mediated clot retraction.
This study unveiled that new pharmacological approaches in antithrombotic therapy with Rac1 as molecular target have to be designed carefully in order to obtain high specificity and minimize potential off-target effects.
In der vorliegenden Dissertation gelang es, die strukturellen und molekularen Determinanten der PTH-Rezeptoren für die Ankopplung an intrazelluläre Signalwege näher zu charakterisieren. Die Regulation des Kalzium-, Phosphat- und Knochenstoffwechsels wird zum erheblichen Teil über den PTH1-Rezeptor (P1R) vermittelt. Parathormon (PTH) aktiviert am P1R mindestens zwei Signalwege: den durch zyklisches Adenosinmonophosphat (cAMP) vermittelten Weg und den Phospholipase C-Signalweg (PLC). Der nahe verwandte PTH2-Rezeptor (P2R) kann außer durch PTH auch über das tuberoinfundibiläre Peptid (TIP39) aktiviert werden. Jedoch besitzt dieser Rezeptor keine Ankopplung an den PLC-Signalweg. Zur Aufklärung der strukturellen und molekularen Determinaten der intrazellulären Signalwegankopplung wurden verschiedene Versuchsansätze ausgewählt, die eine Untergliederung dieser Arbeit in drei Teilprojekte ermöglicht: 1) Die PTH-Rezeptoren sind wichtige Vertreter der Klasse II der G-Protein gekoppelten Rezeptoren. Ein Vergleich der Aminosäuresequenzen der siebten Transmembrandomäne dieser Klasse zeigt ein hoch konserviertes, cytosolnahes „YCFXN“-Motiv in diesem Bereich. Im ersten Teil dieser Arbeit konnte durch Punktmutationen der einzelnen Aminosäuren dieses Motivs gezeigt werden, dass dieser Abschnitt eine entscheidende Determinierungsregion dieser Rezeptorfamilie sowohl für die Ankopplung an den cAMP-Weg, als auch an den PLC-Signalweg darstellt. Die Untersuchungen legen die Vermutung nahe, dass dieser Bereich für die Stabilisierung der Konformation dieser Rezeptoren von großer Bedeutung ist. 2) In einem zweiten Abschnitt dieser Arbeit wurde durch stufenweise Angleichung des P2R an den P1R eine Reihe von funktionell exprimierten P2R/P1R Hybridrezeptoren hergestellt, die eine Übereinstimmung des intrazellulären Bereichs des P1R von bis zu 95 % erreichen. Der Nachweis des PLC-Signalwegs durch die Bestimmung der akkumulierten Gesamtinositolphosphate und des intrazellulären Kalziums zeigte eindrucksvoll, dass trotz einer weitgehenden intrazellulären Übereinstimmung der Aminosäuresequenz des P2R mit dem P1R die Eigenschaft des P1R an den PLC-Signalweg zu koppeln nicht auf den P2R übertragen werden kann. Dies legt nahe, dass auch extrazelluläre Bereiche und Transmembranabschnitte die Ankopplung an intrazelluläre Signalwege steuern. Im Weiteren konnte für den cAMP-Signalweg durch diese Hybridrezeptoren gezeigt werden, dass im Kontext des P2R eingefügte Teilabschnitte des P1R (C-Terminus, zweite und dritte intrazelluläre Schleife ) zusammenwirken und eine effizientere Ankopplung an den cAMP-Weg ermöglichen. Weiterführende Untersuchungen der Membrantranslokation von beta Arrestin2 mit einer anschließenden Internalisierung des Rezeptors zeigten, dass sowohl der P2R als auch die hiervon abgeleiteten Hybridrezeptoren selektiv durch Stimulation mit TIP39, nicht jedoch nach einer Stimulation mit PTH, eine Translokation bewirken. Dieses Ereignis ist von den untersuchten Signalwegen (cAMP-, PLC- und Mitogen-aktivierte Proteinkinase (MAPK)-Signalweg) unabhängig. Erstmals wurde hier gezeigt, dass der P2R eine Phosphorilierung von MAPK bewirkt, wobei hierfür einer beta-Arrestin2 Translokation nicht notwendig ist. Diese Ergebnisse lassen den Schluss zu, dass der PTH-Rezeptor in unterschiedlichen Rezeptorkonformationen existiert, so dass einzelne Rezeptorabschnitte unabhängig voneinander verschiedene Signale aktivieren können. 3) Der letzte Teil dieser Arbeit beschäftigte sich mit der Identifikation von intrazellulären Interaktionspartnern des humanen P1R. Neue Protein–Interaktionen mit dem humanen P1R wurden mit einem „Yeast-two-Hybrid“ System identifiziert. Mit Hilfe dieser Methode konnte, als ein potentiell bedeutender intrazellulärer Interaktionspartner des humanen P1R, das PDZ-Protein PDZK1 identifiziert werden. Es gelang mit Hilfe von Koimmunpräzipitations-Experimenten und von GST-pull-down-Assays die Interaktion von PDZK1 mit dem P1R zu verifizieren. PDZK1 bindet an eine Liganden-Bindungsdomäne innerhalb des C-terminalen Abschnitts des P1R, vermutlich an die letzten vier Aminosäuren. Durch einen Hefe-Interaktionstest konnte von den vier in diesem Protein vorkommenden PDZ-Domänen die PDZ1-Domäne als einzige selektiv mit dem P1R interagierende Domäne identifiziert werden. In der Niere interagiert PDZK1 über die PDZ3-Domäne mit dem Na/Pi-Transporter IIa. Eine attraktive Hypothese ist daher die Funktion von PDZK1 als einem Bindeglied zwischem dem Rezeptor und dem Transporter und die damit einhergehende PTH- vermittelte Regulation des Phosphattransports in der Niere. Diese Hypothese bedarf aber nochweiterer funktioneller Analysen (z.B. durch Untersuchungen an PDZK1 Knock-Out Mäusen).
Bone Morphogenetic Proteins (BMPs) bilden zusammen mit den Activinen, Growth and Differentiation Factors (GDFs) und Transforming Growth Factor β (TGF-β) die Transforming Growth Factor β-Superfamilie von sekretierten Signalproteinen. Sie spielen eine wichtige Rolle in der Entwicklung, Erhaltung und Regeneration von Geweben und Organen. Die Signalvermittlung dieser Proteine erfolgt durch die Bindung von zwei verschiedenen Typen von Serin-/Threonin-Kinaserezeptoren, die als Typ-I- und Typ-II-Rezeptoren bezeichnet werden. Im ersten Schritt erfolgt die Bindung an den hochaffinen Rezeptor (im Fall von BMP-2 der Typ-I-Rezeptor), im nächsten Schritt wird der niederaffine Rezeptor in den Komplex rekrutiert. Bis heute sind lediglich sieben Typ-I- und fünf Typ-II-Rezeptoren bekannt, was auf eine Promiskuität in der Liganden-Rezeptor-Interaktion schließen lässt. Die Architektur beider Rezeptorsubtypen ist dabei relativ ähnlich. Beide bestehen aus einer ligandenbindenden extrazellulären Domäne, einer Transmembrandomäne sowie einer intrazellulären Kinasedomäne. Eine nacheinander ablaufende Transphosphorylierung der intrazellulären Domänen führt zu einer Phosphorylierung von SMAD-Proteinen, die dann als nachgeschaltete Vermittler fungieren und die Transkription regulierter Gene auslösen. Im Hauptteil dieser Arbeit wurden die initialen Schritte der Rezeptorkomplexformierung sowie die Mobilität der Rezeptoren mit Hilfe von fluoreszenzmikroskopischen Methoden untersucht. Dabei konnte festgestellt werden, dass für die Bildung eines Signalkomplexes eine bestimmte Schwellenkonzentration des Liganden nötig ist und dass der Mechanismus nach einem Alles-oder-Nichts-Prinzip wie ein Schalter funktioniert. Außerdem konnten Unterschiede in der Nutzung der gleichen Rezeptoren durch verschiedene Liganden festgestellt werden. Die anderen Teile der Arbeit befassen sich mit der Funktionalität der verschiedenen Rezeptordomänen in der Signalübermittlung, der Analyse von hoch- und niederaffinen Ligandenbindestellen auf ganzen Zellen sowie dem Einfluss des SMAD- und des MAPK-Signalwegs auf die Induktion der Alkalischen Phosphatase. Dabei konnte gezeigt werden, dass die Art der SMAD-Phosphorylierung allein vom Typ der Kinasedomäne abhängig ist, dass auf einer Zelle verschiedene Rezeptorpopulationen existieren, welche von unterschiedlichen Ligandenkonzentrationen angesprochen werden, und dass die Induktion der Alkalischen Phosphatase stark vom zeitlichen Verlauf der SMAD- und MAPK-Aktivierung abhängig ist.
Zur Durchführung peripherer Nervenblockaden werden im klinischen Alltag nichtselektive Lokalanästhetika verwendet, die neben sensorischen auch motorische Nervenfasern blockieren. Diese Arbeit untersucht und beschreibt Grundlagen für die Verwendung selektiv wirksamer Co-Analgetika. Ziel dieser Arbeit war in diesem Kontext die Analyse der intrazellulären Signalwege, welche nach Applikation von rtPA am peripheren Nerven zur Öffnung der perineuralen Barriere und so zu einer opiat- vermittelten Analgesie führen. Gemäß unserer Hypothese bindet rtPA an den LRP-1- Rezeptor und löst eine intrazelluläre Signalkaskade aus: Erk wird phosphoryliert und inhibiert über bislang unklare Mechanismen die Claudin-1-Transkription. Claudin-1 wird weniger in die Zellmembran eingebaut und/oder verlässt durch Endozytose/ Internalisierung die Zellmembran, was zur Öffnung der perineuralen Barriere führt und den Durchtritt selektiv wirksamer Analgetika erlaubt. In der späteren Phase steht die Analyse der Wiederherstellung der Barrierefunktion der Zellmembran im Vordergrund. Die ist von zentraler Bedeutung um eine Schädigung des Nervens durch das Umgebungsmilieu zu verhindern. Vermutlich wird die Wiederherstellung der Barrierefunktion über den Wnt-Signalweg gesteuert. Die Akkumulation von b-Catenin und Cdx2 führt zu einem erneuten Anstieg der Claudin-1-Transkription. Der Claudin-1- Gehalt steigt in Western Blot-Untersuchungen jedoch bereits zu einem früheren Zeitpunkt in der Zellmembran wieder an. Dies legt nahe, dass weitere von der Transkription unabhängige Mechanismen zur Wiederherstellung der Barrierefunktion beitragen. Eine mögliche Alternative zu rtPA stellt katalytisch inaktives rtPAi dar, welches in Untersuchungen ähnliche Ergebnisse wie rtPA zeigte. Dabei könnte die Verwendung von rtPAi anstatt rtPA pathophysiologisch denkbare Komplikationen wie beispielsweise Blutungen verhindern.
In Versuchen anderer Mitglieder der Arbeitsgruppe wurde die Öffnung der perineuralen Barriere mittels immunhistochemischer und funktioneller Untersuchungen bestätigt. Auch konnten keine akute Neurotoxizität oder Blutungsgefahr beobachtet werden. Somit stellt rtPA in Kombination mit Opioiden eine mögliche Alternative zur Verbesserung der postoperativen Analgesie dar, die jedoch weiterer Untersuchungen hinsichtlich von Nutzen, Risiken und Nebenwirkungen bedarf.
Plants have evolved an elaborate system to cope with a variety of biotic and abiotic stresses. Typically, under stress conditions an appropriate defense response is invoked which is accompanied by changes in the metabolic status of the plant. Photosynthesis is downregulated and sucrose is imported into the tissue, which provides a faster and more constant flux of energy and carbon skeletons to perform the defense response. Interestingly, these processes are co-ordinately regulated and the signal transduction chains underlying these cellular programs appear to share at least some common elements. Both the induction of sink metabolism and defense response is dependent on signal transduction pathways involving protein phosphorylation. Furthermore, regulation of extracellular invertase (INV) and phenylalanine ammonia lyase (PAL) which are markers for sink metabolism and defense response is preceded by the transient activation of MAP kinases. In depth analysis of MAP kinase activation by partial purification led to the discovery that, depending on the stimulus, different subsets of MAP kinases are activated. This differential MAPK activation is likely to possess a signal encoding function. In addition, the partial purification of MAP kinases was found to be suitable to address specific cellular functions to individual MAP kinase isoenzymes. By this way, LpWIPK was identified as the major MAP kinase activity induced after stimulation of tomato cells with different elicitors. LpWIPK is thus considered as a key regulator of defense response together with sink induction in tomato. A study using nonmetabolisable sucrose analogs revealed that the regulation of photosynthesis is not directly coupled to this signal transduction pathway since it is independent of MAP kinase activation. Nonetheless, downregulation is induced by the same stimuli that induce the defense response and sink metabolism and it will therefore be interesting to uncover the branch points of this signalling network in the future. MAP kinases are not only central components regulating the response to biotic stresses. In addition to e.g. pathogens, MAP kinases are as well involved in signal transduction events invoked by abiotic stresses like cold and drought. In a recent study, we could show that a MAP kinase is activated by heat stress, under conditions a plant will encounter in nature. This previously unknown MAP kinase is able to specifically recognise the heat stress transcription factor HsfA3 as a substrate, which supports a role of this MAP kinase in the regulation of the heat stress response. Moreover, the observation that HsfA3 is phosphorylated by the heat activated MAP kinase in vitro provides a promising basis to identify HsfA3 as the first physiological substrate of a plant MAP kinase. Intracellular protons have been implicated in the signal transduction of defense related signals. In a study using Chenopodium rubrum cells, we could show that cytosolic changes in pH values do not precede the regulation of the marker genes INV and PAL. Depending on the stimulus applied, cytosolic acidification or alkalinisation can be observed, which excludes a role for protons as signals in this pathway. Together with the concomitant changes of the pH value of the extracellular space, these variations can thus be considered as terminal part of the defense response itself rather than as a second messenger. WRKY transcription factors have only recently been identified as indirect targets of a central plant MAP kinase cascade. In addition, the identification of cognate binding sites in the promoters of INV and PAL supports a role for these proteins in the co-ordinate regulation of defense response and sink induction. A novel elicitor responsive WRKY transcription factor, LpWRKY1, was cloned from tomato and characterised with respect to its posttranslational modification. This immediate early transcription factor is transiently induced upon pathogen attack and the induction is dependent on phosphorylation. Furthermore, it was shown for the first time with respect to WRKY transcription factors, that LpWRKY1 is phosphorylated in vivo. Analysis of the role of this phosphorylation by in gel assays using recombinant WRKY protein as the substrate revealed two protein kinases that are transiently activated during the defense response to phosphorylate LpWRKY1. This data demonstrates that WRKY proteins require phosphorylation to modulate their DNA binding or transactivating activity.
Activation of mitogen-activated protein (MAP) kinases is a common reaction of plant cells in defense-related signal transduction pathways. Since the downstream events after the activation of MAP kinases are largely unknown in plants, the role of MAP kinases in the co-ordinate regulation of defense reactions and primary carbon metabolism by stress related stimuli has been analyzed in tomato. Thus, the relationship between mitogen activated protein kinases (LpMPK2 and LpMPK3) and extracellular invertases Lin6, as the key enzyme of an apoplasmic phloem unloading pathway, has been analyzed. The results showed that the mRNAs of LpMPK3 and Lin6 are sequentially induced by the same set of stress related stimuli (E-Fol, PGA,wounding, and KCl). The induction of the Lin6 promotor, as revealed by an increase in β-glucuronidase activity after 2 hours, was dependent both on the expression and activation of LpMPK3. These data suggest that the induction of extracellular invertase Lin6 by stress related stimuli requires LpMPK3. Glucose, metabolic molecule, was shown to result in the simultaneous induction of AtMPK4 and AtMPK6 activities that could be separated by anion-exchange chromatography, and characterized by differential cross-reaction with MAP kinase antibodies. Taken together, these data suggest that the activation of MAP inases play central roles in the regulation of sugar signaling. Stomatal movement is controlled by environmental signals including light intensity,humidity and atmospheric CO2 level. In Arabidopsis, a complete MAP kinase signaling cascade regulates stomatal development and patterning. However, the movement of stomata mediated by CO2 induced signaling pathways is not fully studied in higher plants. Here, we show that elevated levels of CO2 induce rapid and transient activation of SIPK and NtMPK4. The activation of both MAP kinases may regulate the anion channel activation for stomatal movement by the elevated level CO2. Up to now, the non-antioxidant function of tocopherol is not clear in higher plant,whereas the ability of tocopherol to modulate the stress tolerance mediated by function of antioxidant has been described in numerous studies. Thus, the function of α-tocopherol in stimuli-induced signal transduction pathways mediated by MAP kinase has been analyzed in tobacco. It has been shown that the activation of MAP kinase was induced by treatment of fungal elicitor and α-tocopherol phosphate but not α-tocopherol. Interestingly, α-tocopherol showed the transient inhibitory effect on the activation of stimuli-induced MAP Kinases in BY2 cells and tobacco plants, whereas ascorbate did not inhibit the activation of MAP kinases. The inhibitory activity test indicated that current application may indirectly affect the activity of MAP kinases. These results suggest that α-tocopherol can negatively regulate stimuliinduced signal transduction pathways via inactivation of MAP kinases. The purine-analogues have been tested and reported to be specific inhibitors of protein kinases mediated by structural-based selectivity in mammalian. Here, we tested C2, N6, N9-trisubstituted purines to determine basic relationship between their chemical structure and inhibitory activity using a particular plant MAP kinase. The modification of substitution in position C2 and N9 caused the increased inhibitory activity of 6-(benzylamino) purine analogue. In addition, 6-(isopentenylamino) purine analogues suggested that addition of a methyl group to position N9 caused at least 2-fold increased inhibitory activity compared with the addition of isopropyl group.Taken together, our study suggests that the selectivity and potency of inhibitors can be improved by structure modification. In addition, we have characterized the physiological function of Arabidopsis thaliana PLAT domain protein 1 (AtPDP1) in modulating the interaction of defense pathways mediated by biotic and abiotic factors. Interestingly, overexpression of AtPDP1 resulted in increasing susceptibility of virulent pathogens and necrotrophic fungus, and developing necrosis induced by unknown biotic factors. However, these overexperssion lines showed the significantly delayed senescence and higher level of phosystem II quantum yield compared with control plants against high salt stress. Our results strongly indicate that AtPDP1 positively regulate with salt tolerance, and enhances the sensitivity to biotic stresses. We propose that the AtPDP1 might be regulated with the complex pathways of interplay among various signaling during stress adaptation.
Die vorliegende Arbeit hatte zum Ziel, neuroendokrine Zellen in den Atemwegen bei Mäusen zu untersuchen, welche Kontakt zu sensorischen Nervenfasern ausbilden. In vorangegangenen Versuchen konnte bereits die Menge des ausgeschütteten CGRPs nach Stimulation mit Bitterstoffen bestimmt werden. Die Methode zur Messung der Freisetzung von CGRP aus verschiedenen Organen wurde von Prof. Reeh und seiner Arbeitsgruppe etabliert. Ziel der vorliegenden Arbeit war es, zu untersuchen, woher das ausgeschüttete CGRP kommt und ob die Stimulation von Bürstenzellen mit Bitterstoffen zur Ausschüttung von CGRP aus den neuroendokrinen Zellen führt. Anhand der elektronenmikroskopischen Auswertung und der dreidimensionalen Rekonstruktion konnte gezeigt werden, dass es Kontakt zwischen den neuroendokrinen Zellen im Epithel der Trachea und sensorischen Nervenfasern gibt. Die immunhistochemischen Versuche zeigten, dass es nach Stimulation mit Denatonium höchstwahrscheinlich zur Ausschüttung von CGRP durch die intraepithelialen Fasern gekommen ist. Diese Annahme spiegelt sich in der veränderten Morphologie sowie der geringeren Quantität der intraepithelialen Fasern nach Stimulation mit Denatonium deutlich wider. Dass es weder bei der Anzahl der neuroendokrinen Zellen, noch bei der Erscheinung und Anzahl der extraepithelialen Fasern nach Denatoniumstimulation zu einer Veränderung gekommen ist, unterstützt diese Annahme ebenfalls. Im Hinblick auf die durchgeführten Versuche mit den TRPM5-gendefizienten Mäusen zeigte sich, dass die Stimulation mit Denatonium keine Auswirkungen auf die Anzahl der neuroendokrinen Zellen hatte. Dieses Ergebnis unterstützt die Erkenntnisse der vorangegangenen Untersuchungen, welche gezeigt haben, dass das CGRP nicht von den neuroendokrinen Zellen ausgeschüttet wurde. Des Weiteren lässt das Ergebnis darauf schließen, dass die Ausschüttung von CGRP nicht abhängig von der Anwesenheit von Bürstenzellen ist. Insgesamt zeigen die Untersuchungen, dass es nach Stimulation mit Bittersubstanzen zu einer CGRP-Ausschüttung durch die intraepithelialen Fasern gekommen ist. Interessant wäre es weiterhin zu klären, welche Effekte diese Ausschüttung bewirkt und welche Bedeutung der Freisetzung von Substanz P in diesem Zusammenhang zukommt.
Flagellar motility and chemotaxis are essential virulence traits required for the ability of Helicobacter pylori to colonize the gastric mucosa. The flagellar regulatory network and the complex chemotaxis system of H. pylori are fundamentally different from other bacteria, despite many similarities. In H. pylori expression of the flagella is controlled by a complex regulatory cascade involving the two-component system FlgR-HP244, the sigma factors 54 and 28 and the anti-sigma 28 factor FlgM. Thus far, the input signal for histidine kinase HP244, which activates the transcriptional regulator FlgR, which triggers sigma factor 54-dependent transcription of the flagellar class 2 genes, is not known. Based on a yeast two-hybrid screen a highly significant protein-protein interaction between the H. pylori protein HP137 and both the histidine kinase HP244 and the flagellar hook protein HP908 (FlgE´) has been reported recently (Rain et al., 2001). So far, no function could be assigned to HP137. Interestingly, the interaction between HP137 and histidine kinase HP244 was observed in the characteristic block N sequence motif of the C-terminal ATP-binding kinase domain. In this work a potential role of HP137 in a feedback regulatory mechanism controlling the activity of histidine kinase HP244 in the flagellar regulation of H. pylori was investigated. Although the substitution of the gene encoding HP137 by a kanamycin cassette resulted in non-motile bacteria, the failure to restore motility by the reintroduction of hp137 in cis into the mutant strain, and the observation that HP137 has no significant effect on the activity of histidine kinase HP244 in vitro indicated that HP137 is not directly involved in flagellar regulation. Therefore, it was demonstrated that HP137 does not participate in the regulation of flagellar gene expression, neither in H. pylori nor in the closely related bacterium C. jejuni. Chemotactic signal transduction in H. pylori differs from the enterobacterial paradigm in several respects. In addition to a CheY response regulator protein (CheY1) H. pylori contains a CheY-like receiver domain (CheY2) which is C-terminally fused to the histidine kinase CheA. Furthermore, the genome of H. pylori encodes three CheV proteins consisting of an N-terminal CheW-like domain and a C-terminal receiver domain, while there are no orthologues of the chemotaxis genes cheB, cheR, and cheZ. To obtain insight into the mechanism controlling the chemotactic response of H. pylori the phosphotransfer reactions between the purified two-component signalling modules were investigated in vitro. Using in vitro phosphorylation assays it was shown that both H. pylori histidine kinases CheAY2 and CheA´ lacking the CheY-like domain (CheY2) act as ATP-dependent autokinases. Similar to other CheA proteins CheA´ shows a kinetic of phosphorylation represented by an exponential time course, while the kinetics of phosphorylation of CheAY2 is characterized by a short exponential time course followed by the hydrolysis of CheAY2~P. Therefore, it was demonstrated that the presence of the CheY2-like receiver domain influences the stability of the phosphorylated P1 domain of the CheA part of the bifunctional protein. Furthermore, it was proven that both CheY1 and CheY2 are phosphorylated by CheAY2 and CheA´~P and that the three CheV proteins mediate the dephosphorylation of CheA´~P, although with a clearly reduced efficiency as compared to CheY1 and CheY2. Moreover, CheA´ is capable of donating its phospho group to the CheY1 protein from C. jejuni and to CheY protein from E. coli. Retrophosphorylation experiments indicated that CheY1~P is able to transfer the phosphate group back to the HK CheAY2 and the receiver domain present in the bifunctional CheAY2 protein acts as a phosphate sink fine tuning the activity of the freely diffusible CheY1 protein, which is thought to interact with the flagellar motor. Hence, in this work evidence of a complex phosphorelay in the chemotaxis system was obtained which has similarities to other systems with multiple CheY proteins. The role of the CheV proteins remain unclear at the moment, but they might be engaged in a further fine regulation of the phosphate flow in this complex chemotaxis system and the independent function of the two domains CheA´ and CheY2 is not sufficient for normal chemotactic signalling in vivo.
Chronische Drucküberlastung des adulten Herzens führt zu Umbauvorgängen in der Extrazellulärmatrix und zur Hypertrophie der Herzmuskelzellen. Dabei kommt es zur Aktivierung von Integrinrezeptoren und der dadurch induzierten Entstehung des sogenannten fokalen Adhäsionskomplexes im Zytoskelett der Herzmuskelzelle. Dies konnte kürzlich im Modell der rechtsventrikulären Drucküberlastung an der Katze gezeigt werden. (Laser M et al., J. Biol. Chem, 2000). Um zu testen, ob die Integrinaktivierunng und der Umbau des Zytoskeletts im Hypertrophieprozess des linken Ventrikels eine Rolle spielen, wurden ausgewachsene Mäuse (30g Körpergewicht) nach Konstriktion der transversalen Aorta analysiert und mit scheinoperierten Mäusen verglichen. Morphologisch zeigte sich bei den drucküberlasteten Tieren eine signifikante Herzhypertrophie bezogen auf Körpergewicht und Tibialänge innerhalb eines Monats. Keine Unterschiede fanden sich bei der hämodynamischen Charakterisierung für die in vivo gemessene Herzfrequenz, während der systolische Druck und der enddiastolische Druck signifikant erhöht waren. Zur Western Blot-Analyse wurden jeweils eine Detergens-lösliche sowie eine unlösliche Fraktion (Zytoskelett) der Herzen präpariert. Dabei zeigten sich im Zytoskelett verschiedene tyrosinphosphorylierte Proteine bei 160, 125, 75 und 60 kD, beginnend drei Tage nach Drucküberlastung. Zwei dieser Banden konnten wir als Zytoskelett-assoziierte Tyrosinkinasen, „focal adhesion kinase“(Fak) und c-Src, identifizieren, welche beide zum sogenannten Integrin-abhängigen fokalen Adhäsionskomplex gehören. Weitere Analysen zeigten Phosphorylierungen von Fak bei Tyr-397 und Tyr-925, c-Src bei Tyr-418. Dies lässt darauf schließen, dass beide Kinasen aktiviert sind. Aus diesen Daten wird deutlich, dass eine Aktivierung von Integrinrezeptoren und die Formation des fokalen Adhäsionskomplexes auch im Hypertrophieprozess des linken Ventrikels eine Rolle spielen. Um den Einfluss der Integrin-vermittelten Signalwege auf die Entwicklung der Hypertrophie und deren Signaltransduktion weiter zu testen, wurde eine konditionale „knock-out“-Mauslinie (Cre/loxP-System mit MLC-2v-Promotor) etabliert. Bei diesen Mäusen ist das beta.1-Integrin-Gen selektiv im Myokard ausgeschaltet. Die Ergebnisse aus Phänotypisierung und Charakterisierung dieser Mauslinie im Hypertrophieprozess stehen noch aus.
The Immune system exerts its response against invading pathogens via a cumulative, sequential cooperation of immune cells coordinated by their secreted products. Immune cells, such as macrophages and dendritic cells (DCs), express toll-like receptors (TLRs) to sense the presence of pathogens through pathogen-associated molecular patterns (PAMPs). The interaction of PAMPs with TLRs elicits a cytosolic signaling cascade that enhances the expression of genes to stimulate inflammation. Interleukin 1 receptor-associated kinase 2 (IRAK2) is a component of the TLR signaling pathway. IRAK2 transduces the TLR signal via a direct interaction with TNF receptor-associated factor 6 (TRAF6) and subsequent enhancement of its ubiquitination.
During my PhD thesis, I determined that a 55-amino acid long stretch at the C-terminal end of IRAK2 is important for TLR signaling. Overexpression of C-terminal truncated IRAK2 (IRAK2Δ55) in the murine macrophage cell line RAW 264.7 led to impaired CD40 expression after TLR4 stimulation by Lipopolysaccharide (LPS). I observed attenuated competency of IRAK2Δ55 in restoring a full TLR signaling response i.e. IL-6 secretion, NO production and CD40 expression in IRAK2-deficient RAW cells generated via CRISPR-Cas9 approach. Additionally, diminished TLR4 induced activation of nuclear factor κB (NF-κB) and extracellular signal related kinase (ERK) was observed with IRAK2Δ55 reconstituted RAW cells as compared to cell reconstituted with wildtype IRAK2.
IRAK2Δ55 reconstituted RAW cells also exhibited reduced TLR4-induced cell death and phosphorylation of receptor interacting protein kinase 3 (RIP3). Co-immunoprecipitation experiments in HEK 293T cells showed that IRAK2Δ55 was still able to bind to TRAF6 alike IRAK2 but failed to induce ubiquitination of TRAF6. In conclusion, the results suggest that the IRAK2 TRAF6 interaction is not sufficient to sustain full TLR signaling. An C-terminus-dependent unknown molecular mechanism is also involved.
Through my PhD work, I also analyzed a B cell lineage-specific HECTD1 knock-out mice. HECTD1 is an E3 ubiquitin ligase for various substrate proteins, such as heat shock protein 90 (HSP90), adenomatous polyposis coli and phosphatidylinositol phosphate kinase type 1 γ. Hsp90 regulates a variety of signaling molecules in NF-κB activation pathways which are essential for an optimal B cell response.
HECTD1-deficient pro-B cells developed normally into mature B cells. However, TLR4 stimulated HECTD1-deficient B cells displayed reduced immunoglobulin (Ig) production in in vitro cultures. In addition, mice with HECTD1-deficient B cells showed a diminished Ig response after nitrophenylacetyl-keyhole limpet hemocyanin immunization. Thus, HECTD1 is necessary for efficient Ig secretion.
In physiological conditions platelets have a major role in maintaining haemostasis. Platelets prevent bleeding from wounds by distinguishing normal endothelial cells in vasculature from areas with lesions to which they adhere. Interaction of platelet agonists and their receptors is controlled by intracellular signaling molecules that regulate the activation state of platelets. Very important intracellular signaling molecules are cyclic nucleotides (cGMP and cAMP), both involved in inhibition of platelet activation. Formation of cGMP and cAMP in platelets is stimulated by endothelial-derived NO and prostacyclin (PGI2), which then mediate inhibition of platelets by activating protein kinase G (PKG) and protein kinase A (PKA). Recently, it has been suggested that reactive oxygen species (ROS) represent new modulators of cell signaling within different cell types. The work summarized here describes the involvement of platelet ROS production in platelet activation, the relation of NO/cGMP/PKG I pathway to ROS and to mitogen-activated protein kinases (MAP kinase) signaling, and the involvement of cyclic nucleotides in megakaryocyte and platelet development. Platelets activated with different agonists produce intracellular but not extracellular ROS by activation of NAD(P)H oxidase. In addition, ROS produced in platelets significantly affects αIIbβ3 integrin activation but not alpha/dense granule secretion and platelet shape change. Thrombin induced integrin αIIbβ3 activation is significantly decreased after pretreatment of platelets with NAD(P)H oxidase inhibitors and superoxide scavengers. These inhibitors also reduce platelet aggregation and thrombus formation on collagen under high shear and achieve their effects independently of the NO/cGMP pathway. ADP secreted from platelet dense granules with subsequent activation of P2Y12 receptors as well as thromboxane A2 release are found to be important upstream mediators of p38 MAP kinase activation by thrombin. However, p38 MAP kinase activation does not significantly contribute to calcium mobilization, P-selectin expression, αIIbβ3 integrin activation and aggregation of human platelets in response to thrombin. Finally, PKG activation does not stimulate, but rather inhibit, p38 and ERK MAP kinases in human platelets. Further study revealed that cyclic nucleotides not only inhibit platelet activation, but are also involved, albeit differentially, in megakaryocyte and platelet development. cAMP is engaged in haematopoietic stem cell differentiation to megakaryocytes, and cGMP has no impact on this process. While PKA is already present in stem cells, expression of proteins involved in cGMP signaling (soluble guanylyl cyclase, sGC; PKG) increases with maturation of megakaryocytes. In the final step of megakaryocyte maturation that includes release of platelets, cGMP and cAMP have mild but opposing effects: cGMP increases platelet production while cAMP decreases it indicating a finely regulated process that could depend on stimulus coming from adjacent endothelial cells of sinusoids in bone marrow. The results of this thesis contribute to a better understanding of platelet regulation and of the possible molecular mechanisms involved in megakaryocyte maturation in bone marrow vascular microenvironment.
Das zentrale Paradigma der Systembiologie zielt auf ein möglichst umfassendes Ver-ständnis der komplexen Zusammenhänge biologischer Systeme. Die in dieser Arbeit angewandten Methoden folgen diesem Grundsatz.
Am Beispiel von drei auf Basis von Datenbanken und aktueller Literatur rekonstruier-ten Netzwerkmodellen konnte in der hier vorliegenden Arbeit die Gültigkeit analyti-scher und prädiktiver Algorithmen nachgewiesen werden, die in Form der Analy-sesoftware Jimena angewandt wurden. Die daraus resultierenden Ergebnisse sowohl für die Berechnung von stabilen Systemzuständen, der dynamischen Simulation, als auch der Identifikation zentraler Kontrollknoten konnten experimentell validiert wer-den. Die Ergebnisse wurden in einem iterativen Prozess verwendet werden um das entsprechende Netzwerkmodell zu optimieren.
Beim Vergleich des Verhaltens des semiquantitativ ausgewerteten regulatorischen Netzwerks zur Kontrolle der Differenzierung humaner mesenchymaler Stammzellen in Chondrozyten (Knorpelbildung), Osteoblasten (Knochenbildung) und Adipozyten (Fett-zellbildung) konnten 12 wichtige Faktoren (darunter: RUNX2, OSX/SP7, SOX9, TP53) mit Hilfe der Berechnung der Bedeutung (Kontrollzentralität der Netzwerkknoten identifi-ziert werden). Der Abgleich des simulierten Verhaltens dieses Netzwerkes ergab eine Übereinstimmung mit experimentellen Daten von 47,2%, bei einem widersprüchlichen Verhalten von ca. 25%, dass unter anderem durch die temporäre Natur experimentel-ler Messungen im Vergleich zu den terminalen Bedingungen des Berechnung der stabilen Systemzustände erklärt werden kann.
Bei der Analyse des Netzwerkmodells der menschlichen Immunantwort auf eine Infek-tion durch A. fumigatus konnten vier Hauptregulatoren identifiziert werden (A. fumi-gatus, Blutplättchen, hier Platelets genannt, und TNF), die im Zusammenspiel mit wei-teren Faktoren mit hohen Zentralitätswerten (CCL5, IL1, IL6, Dectin-1, TLR2 und TLR4) fähig sind das gesamte Netzwerkverhalten zu beeinflussen. Es konnte gezeigt werden, dass sich das Aktivitätsverhalten von IL6 in Reaktion auf A. fumigatus und die regulato-rische Wirkung von Blutplättchen mit den entsprechenden experimentellen Resultaten deckt.
Die Simulation, sowie die Berechnung der stabilen Systemzustände der Immunantwort von A. thaliana auf eine Infektion durch Pseudomonas syringae konnte zeigen, dass die in silico Ergebnisse mit den experimentellen Ergebnissen übereinstimmen. Zusätzlich konnten mit Hilfe der Analyse der Zentralitätswerte des Netzwerkmodells fünf Master-regulatoren identifiziert werden: TGA Transkriptionsfaktor, Jasmonsäure, Ent-Kaurenoate-Oxidase, Ent-kaurene-Synthase und Aspartat-Semialdehyd-Dehydrogenase.
Während die ersteren beiden bereits lange als wichtige Regulatoren für die Gib-berellin-Synthese bekannt sind, ist die immunregulatorische Funktion von Aspartat-Semialdehyd-Dehydrogenase bisher weitgehend unbekannt.
Die vorliegende Arbeit behandelt TRAIL-induzierte Apoptose und Nekroptose in verschiedenen Zelllinien. Im Speziellen wurden die verschiedenen Funktionen des TNF receptor-associated factor 2 (TRAF2) untersucht. Hierzu wurde ein transienter Knockdown etabliert und dessen Wirkung auf die Suszeptibilität der Zellen gegenüber dem Zytokin TRAIL untersucht. Es konnte gezeigt werden, dass ein Knockdown von TRAF2 nicht nur zur Sensitivierung für Apoptose führt, sondern auch in Nekroptose-kompetenten Zellen zu einer Verstärkung der durch Caspaseinhibition mittels zVAD-fmk nach TRAIL-Stimulation induzierten Nekroptose führt. Mittels des Zytokins Fc-TWEAK wurde Fn14-vermittelt TRAF2 aus dem Zytosol in ein Triton X100-unlösliches Kompartiment rekrutiert und dadurch physiologisch depletiert. Dies führte zwar kaum zu gesteigerter TRAIL-abhängiger Apoptose, sensitivierte jedoch analog zum TRAF2-Knockdown RIP3-exprimierende Zellen für Nekroptose. Durch Vergleich RIP3-negativer (HeLa-Leervektor) mit RIP3-exprimierenden Zellen (HeLa RIP3, HT29, HaCaT) konnte die Essentialität von RIP3 für die Nekroptose herausgestellt werden und Einsatz des RIP1-Kinase-Inhibitors Necrostatin-1 sowie des MLKL-Inhibitors Necrosulfonamide belegte die Beteiligung der Nekroptosomkomponenten RIP1 und MLKL. Antagonismus putativen autokrinen TNFs bewies, dass es sich bei dem durch Fc-TWEAK verstärkten Zelltod um einen direkten TRAIL-Effekt handelte und Inhibition kanonischen NFkBs durch IKK2-Inhibitor TPCA-1, dass die TRAF2-Knockdown-vermittelte Sensitivierung gegenüber TRAIL nicht auf verändertes NFkB-Signalling zurückzuführen ist. Einsatz des SMAC-Mimetikums BV6 rekapitulierte zudem stark das im TRAF2-Knockdown Gesehene und unterstrich die Bedeutung der cIAPs. Immunpräzipitation von Caspase 8 unter nekroptotischen Bedingungen zeigte bei TRAF2-Knockdown eine Depletion von TRAF2 und cIAP1/2 sowie RIP1 und RIP3 aus dem Komplex mit Caspase 8. Insgesamt wird deutlich, dass TRAF2 einerseits antiapoptotisch wirkt als K48-Ubiquitinligase, die die Halbwertszeit aktiver Caspase 8-Komplexe determiniert und andererseits eine antinekroptotische Funktion hat, da es durch Rekrutierung von cIAP1/2 an RIP1 die TRAIL-induzierte Nekroptose verhindert, wenn die Caspasen inhibiert sind.
Das Hauptthema der hier vorliegenden Arbeit befaßt sich mit dem B-Zell spezifischen Oberflächenprotein CD22, einem Mitglied der Siglec (Sialinsäure bindende Igähnliche Lektine) Proteinfamilie. Dieses Transmembranprotein besitzt sieben extrazelluläre Immunoglobulin-ähnliche Domänen und kann über die äußerste V-set Domäne seine Liganden: α2,6 verknüpfte Sialinsäuren binden. CD22 hat eine Transmembrandomäne und eine cytoplasmatische Domäne mit sechs potentiellen Tyrosin Phosphorylierungsstellen, von denen drei eine ITIM-Sequenz (engl. immunoreceptor tyrosine-based inhibitory motif) aufweisen. CD22 defiziente Mäuse zeigten eindeutig, daß das Siglec CD22 ein negativer Regulator des BCR-Signals ist. Durch BCR-Kreuzvernetzung wird CD22 tyrosinphosphoryliert, die inhibitorische Tyrosinphosphatase SHP-1 gebunden, aktiviert, und ist nun in der Lage das BCR Ca2+ Signal zu inhibieren. Um die Rolle der CD22Ligandenbindungsdomäne, in vivo zu untersuchen, sollte in dieser Arbeit eine CD22 knock -in Maus erzeugt werden (CD22R130E Maus), in der die Ligandenbindungsdomäne von CD22 durch eine Punktmutation funktionell ausgeschaltet ist. In der hieraus resultierenden Mauslinie sollte dann die BZellentwicklung, Signaltransduktion und der Immunstatus analysiert werden. Der Vergleich des Phänotyps der CD22R130E Maus und der CD22 defizienten Maus sollte dann zeigen, wie die Adhäsions- und Signalleitungseigenschaften von CD22 zusammenhängen. Der „Targeting“ Vektor für die „Gene Targeting“ Experimente wurde von der Arbeitsgruppe Dr. Anton van der Merwe (von Christina Piperi) angefertigt. Ursprünglich wurde ein „Targeting“ Vektor aus genomischer C57BL/6-DNA verwendet, um den genetischen Hintergrund der CD22-defizienten Maus beizubehalten. Dieser Vektor wurde von mir für ES-Zell Transfektionen in der C57Bl/6 ES-Zellline verwendet. Aus den Gene Targeting Experimenten mit der C57Bl/6-III ES-Zelllinie konnten zwei ES-Zellklone isoliert werden, die eine korrekte homologe Integration des Targetvektors trugen. Aus einem Blastozysteninjektions- Experiment mit einem Cre-deletierten C57BL/6-III Subklon wurden sechs hochchimäre Mäuse erhalten, mit denen allerdings keine Keimbahntransmission erzielt werden konnte. Nach Problemen mit Keimbahntransmission von Klonen aus der C57BL/6-III ESZelllinie, wurden noch die BALB/c und die E14Tg2a ES-Zelllinie für neue Gene Targeting Experimente verwendet. Die Experimente mit der BALB/c ES-Zelllinie ergaben keine ES-Zellklone mit korrekter homologer Integration, dies beruhte wahrscheinlich auf dem nicht isogenen Hintergrund. Alle folgenden Experimente mit der E14Tg2a ES-Zelllinie (genetischer Hintergrund: 129/ola) wurden mit dem verlängerten R130E-Targetvektor (Targetvektor 2), der mit 129/ola DNA um 2,3 Kb in 5’-Richtung verlängert wurde, um den isogenetischen Anteil des Targetvektors zu erhöhen, durchgeführt. Aus diesen Experimenten resultierten wiederum zwei ESZellklone, deren korrekte homologen Rekombination durch Southern Blot bestätigt werden konnten. Bei den darauffolgenden Blastozysten-Injektionsexperimenten mit diesen zwei E14Tg2a Klonen konnten fünf chimäre Tiere gewonnen werden. Ein 80 %ig chimäres Männchen erzeugte eine hohe Anzahl von Nachkommen mit Keimbahntransmission. Bei der Analyse dieser Tiere trat das Resultat zutage, daß alle diese Tiere mit Keimbahntransmission einen wildtypischen Genotyp besaßen. Ein weiteres Mitglied der Siglecproteinfamilie, das murine SiglecG (ein Ortholog zu humanem Siglec10), wurde in dieser Arbeit untersucht. In Zusammenarbeit mit dem Labor von Dr. Paul Crocker sollte eine SiglecG knock out Maus hergestellt werden. Die Strategie für die Gene Targeting Experimente für einen SiglecG knock out basierten auf der Verwendung der BalbI ES-Zelllinie (aus BALB/c Mäusen), da hiermit sehr gute Erfahrungen vorlagen, was die Stabilität ihrer Pluripotenz und des Keimbahntransmissionspotenzials angeht. Daher wurde im Labor von Paul Crocker (von Sheena Kerr) ein Kontroll- und ein Targetvektor kloniert, mit dem große Teile der ersten und zweiten Ig-Domäne von SiglecG ausgeschaltet werden sollte. Mit diesem Vektor führte ich mehrere ES-Zell Transfektionsexperimente durch. Innerhalb der Zeitspanne meiner Doktorarbeit konnten keine ES-Zellklone mit einem korrekten homologen Integrationsereignis gewonnen werden. Mittels der ursprünglichen Strategie konnte die mir nachfolgende Doktorandin jedoch ES-Zell Klone isolieren, nach Blastozysteninjektion Keimbahntransmission erzielen und somit eine SiglecGdefiziente Maus generieren. Eine andere Zusammenarbeit kam mit Dr. Burkhard Kneitz (Physiologisches Chemie I, Biozentrum, Universität Würzburg) zustande. Seine Intention war es, die Rolle des TGF-β Signalmediators SMAD2 auf B-Zellebene näher zu untersuchen. Von Erwin Böttinger bekamen wir eine Mauslinie, in der das Smad2-Gen gefloxt ist, die mit der CD19-Cre Maus gekreuzt wurde. So wurde eine B-Zell spezifische SMAD2 knock out Maus (bSmad2-/-) erzeugt. Meine Aufgabe bestand darin, die B-Zellkompartmente und die Immunantworten der B-Zell spezifischen Smad2-defizienten Maus zu analysieren. Faßt man alle gewonnenen Daten aus den hier generierten B-Zell spezifischen Smad2 knock out Tieren zusammen, so kann man zu dem klaren Ergebnis kommen, daß der TGF-β Signalmediator Smad2 eine entscheidende Rolle bei der Weiterleitung von TGF-β Signalen in das Zellinnere von B-Zellen spielt. Hierbei zeigten sich klare Veränderungen, im Vergleich zu Kontrolltieren, eine Erhöhung der Zellzahl in den Peyerschen Plaques (PP), und der B1-Zellen im Peritoneum. Die IgA-Immunantwort war in bSmad-/- Tieren stark erniedrigt. Der für TGF-β beschriebene Effekt der Proliferationshemmung von aktivierten B-Zellen war bei aktivierten B-Zellen der bSmad2-/- Mäuse hingegen nicht beeinträchtigt.
