The phytohormone auxin performs important functions in the initiation of plant tissues and organs, as well as in the control of root growth in conjunction with external stimuli such as gravity, water and nutrient availability. These functions are based primarily on the auxin-dependent regulation of cell division and elongation. Important for the latter is the control of the cell turgor by the vacuole. As storage for nutrients, metabolites and toxins, vacuoles are of vital importance. Vacuolar stored metabolites and ions are exchanged across the vacuolar membrane with the cytoplasm via active transport processes as well as passively through ion channels. In their function as second messenger, calcium ions are important regulators but also subject to vacuolar transport processes. Changes in the cytosolic calcium concentration not only act locally, but are also associated with signal transduction over longer distances. In this work, electrophysiological methods were combined with imaging techniques to gain insights into the interaction between cytosolic calcium signals, vacuolar transport processes and auxin physiology in the intact plant organism.
Calcium signals are involved in the regulation of vacuolar ion channels and transporters. In order to investigate this in the intact organism, intracellular microelectrode measurements were performed in the model system of bulging Arabidopsis thaliana root hairs. By means of the two-electrode voltage-clamp technique, it could be confirmed that the vacuolar membrane is the limiting electrical resistance during intravacuolar measurements and thus measured ion currents actually represent only the currents across the vacuolar membrane. The already known time-dependent decrease of vacuolar conductivity during intravacuolar experiments could be further correlated with an impalement-related, transient increase of the cytosolic calcium concentration. Intravacuolar voltage-clamp experiments in root hair cells of calcium reporter plants confirmed this relationship between vacuolar conductivity and the cytosolic calcium concentration.
However, the vacuole is not just a recipient of cytosolic calcium signals. Since the vacuole represents the largest intracellular calcium reservoir, it has long been argued that it is also involved in the generation of such signals. This could be confirmed in intact root hair cells. Changes in the vacuolar membrane potential affected the cytosolic calcium concentration in these cells. While depolarizing potentials led to an increase of the cytosolic calcium concentration, hyperpolarization of the vacuolar membrane caused the opposite. Thermodynamic considerations of passive and active calcium transport across the vacuolar membrane suggested that the results described herein reflect the behaviour of vacuolar H+/Ca2+ exchangers whose activity is determined by the proton motive force.
In addition, cytosolic calcium has been shown to be a key regulator of a rapid auxin-induced signaling pathway that regulates polar transport of the hormone.
In the same model system of bulging root hairs it could be shown that the external application of auxin results in a very fast, auxin concentration- and pH-dependent depolarization of the plasma membrane potential. Synchronous with the depolarization of the plasma membrane potential, transient calcium signals were recorded in the cytosol. These were caused by an auxin-activated influx of calcium ions through the ion channel CNGC14. Experiments on loss-of-function mutants as well as pharmacological experiments showed that the auxin-induced activation of the calcium channel requires auxin-perception by the F-box proteins of the TIR1/AFB family.
Investigations of auxin-dependent depolarization as well as the auxin-induced influx of protons into epidermal root cells of loss-of-function mutants showed that the secondary active uptake of auxin by the high-affinity transport protein AUX1 is responsible for the rapid depolarization
Not only the cytosolic calcium signals correlated with CNGC14 function, but also the AUX1-mediated depolarization of root hairs. An unchanged expression of AUX1 in the cngc14 loss-of-function mutant suggested that the activity of AUX1 must be post-translationally regulated. This hypothesis was supported by experiments in which treatment with the calcium channel blocker lanthanum led to inactivation of AUX1 in the wild type.
The cytosolic loading of individual epidermal root cells with auxin resulted in the spread of lateral and acropetal calcium waves. These correlated with a shift of the auxin gradient at the root apex and thus supported a hypothetical calcium-dependent regulation of polar auxin transport. A model for a rapid, auxin-induced and calcium-dependent signaling pathway is presented and its importance for gravitropic root growth is discussed. Since AUX1-mediated depolarization varied with external phosphate concentration, the importance of this rapid signaling pathway is also discussed for the adaptation of root hair growth to an inadequate availability of phosphate.
Der Hörsinn ist für uns Menschen von entscheidender Bedeutung, um mit der Umwelt kommunizieren zu können. Hörstörungen werden dabei in sensorineurale Erkrankungen der neuronalen Strukturen und nicht-sensorineurale Schallleitungsschwerhörigkeiten unterschieden.
In der vorliegenden Studie sollte es darum gehen zu untersuchen, inwiefern ein neues konditionelles Mausmodell, die Brn3.1 IRES Cre Maus, und ein bestehendes Mausmodell, die pmn-Maus, sich eignen, um die Erkrankung der auditorischen Neuropathie nachzubilden. Die Brn3.1 IRES Cre Maus wurde zur Evaluation der Expression von Cre-Rekombinase unter der Aktivität des Brn3.1 Promotors mit gefloxten Reporter-Mauslinien verkreuzt. Die pmn-Maus ist ein anerkanntes Modell einer Motoneuronerkrankung, hatte aber in vorherigen Untersuchungen erhöhte Hörschwellen gezeigt.
Alle verwendeten Mauslinien wurden mittels ABR und DPOAE frequenzspezifisch untersucht, um die Funktion der Haarzellen im Corti´schen Organ zu evaluieren. Bei der pmn-Linie wurden die audiologischen Untersuchungen wöchentlich zwischen P21 und P35 durchgeführt. Zusätzlich wurde das Corti´sche Organ zu diesen Zeitpunkten morphologisch untersucht.
Es zeigte sich, dass alle verwendeten Mauslinien unauffällige Hörschwellen verglichen zur Backgroundlinie C57BL/6 hatten sowie DPOAE-Antworten zeigten. Die Verkreuzung der Brn3.1 IRES Cre Linie mit den beiden Reporterlinien zeigte eine nachweisbare Cre-Rekombinase-Expression unter der Aktivität des Brn3.1 Promotors nur an den postnatalen Tagen P14 und P21. Diese Expression erfolgte mosaikartig in den äußeren Haarzellen. Mit Hilfe einer RT-PCR wurde eine Diskrepanz zwischen Genotyp und Expression des Reporterproteins im Gewebe festgestellt. Dies ließ vermuten, dass die Expression von Cre-Rekombinase durch gene silencing Prozesse unterdrückt wurde und Brn3.1 als Promotor nicht leistungsstark genug war, um eine Cre-Rekombinase-Expression steuern zu können.
Die pmn-Linie zeigte in ABR-Untersuchungen bereits zum Zeitpunkt P21 erhöhte Hörschwellen in allen untersuchten Frequenzen im Vergleich zum Wildtyp. DPOAE-Antworten waren in der pmn-Linie nur bedingt auslösbar. Es zeigte sich in der morphologischen Evaluation ein Verlust von äußeren Haarzellen über die gesamte Länge des Corti´schen Organes. Durch einen TUNEL-Assay konnte das Absterben dieser Zellen durch apoptotische Vorgänge nachgewiesen werden. Der pmn-Phänotyp entsteht durch eine Mutation im TBCE Gen. Dieses Gen kodiert für ein Protein, welches einen stabilisierenden Einfluss auf die Organisation der Mikrotubuli hat. Die TBCE-Verteilung im Corti´schen Organ zeigte, dass dieses hauptsächlich in den äußeren Haarzellen und inneren Stützzellen exprimiert wird und damit wahrscheinlich einen bedeutenden Einfluss auf die Erhaltung der Haarzellen hat. Eine Analyse des Hörnervs zeigte einen Verlust von Mikrotubuli.
Neben diesen in vivo Untersuchungen sollte außerdem eine gliazellfreie Kultur von dissoziierten auditorischen Neuronen etabliert werden. Hierfür wurden mehrere Faktoren einer Primärzellkultur in Bezug auf ihren Einfluss auf die Gesamtzellzahl, den prozentualen Neuronanteil und die Axonlänge der Neurone untersucht. Das Medium, die Beschichtung des Zellkulturgefäßes, die Gabe von Neurotrophinen/Zytokinen und der Einsatz eines Zytostatikums wurden separat untersucht. Es zeigte sich, dass das Medium, die Beschichtung und Neurotrophin-/Zytokingabe hauptsächlich einen Einfluss auf das axonale Längenwachstum von Neuronen haben. Den Prozentsatz der Neurone beeinflusste nur der Einsatz des Zytostatikums Cytosin-β-D-arabinofuranosid (AraC) signifikant. Die Ergebnisse wurden auch im Zusammenhang mit der reellen Neuronanzahl in Kultur gesehen.
Es ergab sich weiterhin eine Präferenz für DMEM- über NB-Medium sowie für zusätzliche Lamininbeschichtung, für den Einsatz des Zytokins LIF gegenüber den neurotrophen Faktoren BDNF und NT-3 und die Gabe des Zytostatikums AraC ab Tag 2 nach Ausplattierung in einer Konzentration von 5-10 µM. Ein kombinatorischer Einsatz dieser Präferenzen spiegelte in Summe die Ergebnisse der Versuchsreihen Neurotrophine/Zytokin und AraC wieder. Der Anteil der Neurone in der Kultur konnte im Durchschnitt auf 10-12 % gesteigert werden. Eine Verschiebung des Glia-/Neuronenanteils zugunsten letzterer ist vermutlich nur durch den Einsatz weiterer Faktoren oder anderer Methoden möglich.
Die untersuchten Mausmodelle zeigten auf Grund der Untersuchungsergebnisse nur teilweise Ähnlichkeiten mit der Erkrankung der auditorischen Neuropathie. Die Erkenntnisse zur pmn-Mauslinie über das frequenzspezifische Hörvermögen und die morphologische Degeneration im Corti´schen Organ im altersabhängigen Verlauf können aber hilfreich sein, um neben der motorischen auch die sensorische Degeneration dieser Pathologie besser zu verstehen. Zudem konnten auch umfassende Erkenntnisse für die Kultur von dissoziierten auditorischen Neuronen der Maus in Bezug auf verschiedene Variablen erhalten werden.
Functional and genetic dissection of mechanosensory organs of \(Drosophila\) \(melanogaster\)

In Drosophila larvae and adults, chordotonal organs (chos) are highly versatile mechanosensors
that are essential for proprioception, touch sensation and hearing. Chos share molecular,
anatomical and functional properties with the inner ear hair cells of mammals. These multiple
similarities make chos powerful models for the molecular study of mechanosensation.
In the present study, I have developed a preparation to directly record from the sensory neurons
of larval chos (from the lateral chos or lch5) and managed to correlate defined mechanical inputs
with the corresponding electrical outputs. The findings of this setup are described in several case
studies.
(1) The basal functional lch5 parameters, including the time course of response during continuous
mechanical stimulation and the recovery time between successive bouts of stimulation, was
characterized.
(2) The calcium-independent receptor of α-latrotoxin (dCIRL/Latrophilin), an Adhesion class G
protein-coupled receptor (aGPCR), is identified as a modulator of the mechanical signals
perceived by lch5 neurons. The results indicate that dCIRL/Latrophilin is required for the
perception of external and internal mechanical stimuli and shapes the sensitivity of neuronal
mechanosensation.
(3) By combining this setup with optogenetics, I have confirmed that dCIRL modulates lch5
neuronal activity at the level of their receptor current (sensory encoding) rather than their ability
to generate action potentials.
(4) dCIRL´s structural properties (e.g. ectodomain length) are essential for the mechanosensitive
properties of chordotonal neurons.
(5) The versatility of chos also provides an opportunity to study multimodalities at multiple levels.
In this context, I performed an experiment to directly record neuronal activities at different
temperatures. The results show that both spontaneous and mechanically evoked activity increase
in proportion to temperature, suggesting that dCIRL is not required for thermosensation in chos.
These findings, from the development of an assay of sound/vibration sensation, to neuronal
signal processing, to molecular aspects of mechanosensory transduction, have provided the first
insights into the mechanosensitivity of dCIRL.
In addition to the functional screening of peripheral sensory neurons, another
electrophysiological approach was applied in the central nervous system: dCIRL may impact the
excitability of the motor neurons in the ventral nerve cord (VNC). In the second part of my work,
whole-cell patch clamp recordings of motor neuron somata demonstrated that action potential
firing in the dCirl\(^K\)\(^O\) did not differ from control samples, indicating comparable membrane
excitability.
Accurate information transfer between neurons governs proper brain function. At chemical synapses, communication is mediated via neurotransmitter release from specialized presynaptic intercellular contact sites, so called active zones. Their molecular composition constitutes a precisely arranged framework that sets the stage for synaptic communication.
Active zones contain a variety of proteins that deliver the speed, accuracy and plasticity inherent to neurotransmission. Though, how the molecular arrangement of these proteins influences active zone output is still ambiguous. Elucidating the nanoscopic organization of AZs has been hindered by the diffraction-limited resolution of conventional light microscopy, which is insufficient to resolve the active zone architecture on the nanometer scale. Recently, super-resolution techniques entered the field of neuroscience, which yield the capacity to bridge the gap in resolution between light and electron microscopy without losing molecular specificity. Here, localization microscopy methods are of special interest, as they can potentially deliver quantitative information about molecular distributions, even giving absolute numbers of proteins present within cellular nanodomains.
This thesis puts forward an approach based on conventional immunohistochemistry to quantify endogenous protein organizations in situ by employing direct stochastic optical reconstruction microscopy (dSTORM). Focussing on Bruchpilot (Brp) as a major component of Drosophila active zones, the results show that the cytomatrix at the active zone is composed of units, which comprise on average ~137 Brp molecules, most of which are arranged in approximately 15 heptameric clusters. To test for a quantitative relationship between active zone ultrastructure and synaptic output, Drosophila mutants and electrophysiology were employed. The findings indicate that the precise spatial arrangement of Brp reflects properties of short-term plasticity and distinguishes distinct mechanistic causes of synaptic depression. Moreover, functional diversification could be connected to a heretofore unrecognized ultrastructural gradient along a Drosophila motor neuron.
Neurophysiologische Veränderungen der Hörbahn während und nach Vestibularisschwannom-Operationen

Aufgrund von Fallberichten bei Patienten mit operativer Resektion eines Vestibularisschwannoms über verzögerte Ertaubung bei endoperativ erhaltenem Resthörvermögen, war es das Ziel dieser Arbeit die intraoperative und frühe postoperative Phase mittels ABR-Monitoring näher zu charakterisieren und prädiktive Faktoren für die postoperative Entwicklung und die resultierende Hörfunktion zu untersuchen.
Dabei zeigte sich, dass der ABR-Befund sowohl zu Beginn der Operation als auch postoperativ mit dem jeweiligen Hörbefund korreliert. Fälle mit erhaltenem ABR und sekundärer Ertaubung müssen am ehesten auf das Vorhandensein weniger noch leitender Fasern ohne funktionellen Nutzen oder eine Veränderung der Elektrophysiologie zurückgeführt werden.
Die detaillierte Betrachtung des intraoperativen Verlaufes ergab eine Untergliederung in sechs Gruppen:
- Gruppe 1: ABR-Verbesserung
- Gruppe 2: ABR kontinuierlich erhalten
- Gruppe 3: ABR temporär abweichend, letztlich erhalten
- Gruppe 4: ABR verschlechtert im Vergleich zum Ausgangsniveau
- Gruppe 5: ABR-Verlust intraoperativ bei fluktuierendem Verlauf
- Gruppe 6: klarer intraoperativer ABR-Verlust,
welche eine sinnvolle Korrelation mit dem Hörergebnis und der postoperativen Entwicklung zeigten. Im Gegensatz zur bisher verbreiteten Auffassung, dass die endoperative ABR-Qualität einen bleibenden Zustand nach den intraoperativen Veränderungen darstellt, wurde in dieser Studie erstmals nachgewiesen, dass in den ersten Stunden und mit weiteren Fluktuationen in der ersten Woche erhebliche Veränderungen auftreten mit erheblichen Auswirkungen auf das Endergebnis. Darüber hinaus konnten verschiedene Typen dieser veränderlichen physiologischen Funktionszustände identifiziert und hier erstmals beschrieben werden:
- Gruppe mit stabilem und erhaltenem ABR
- Gruppe mit fluktuierendem und erhaltenem ABR
- Gruppe mit fluktuierendem und verlorenem ABR
- Gruppe mit stabilem ABR-Verlust.
In der Analyse und Korrelation der intra- mit den postoperativen Entwicklungen wurde die ABR-Qualität nach 60% Tumorresektion als prognostisch signifikantes Merkmal erkannt. Ferner konnte nachgewiesen werden, dass Patienten mit postoperativem Erholungspotential als typisches Merkmal Tumore der Klasse T3A oder kleiner nach der Hannover Klassifikation zeigen.
Diese Studie deutet darauf hin, dass sich der intraoperative Verlauf postoperativ fortsetzt. Während bei intraoperativ instabilem Verlauf (Gruppen 3-5) sich postoperativ eine fluktuierende Entwicklung anschließt oder die ABR-Qualität sich sogar dauerhaft im Vergleich zum endoperativen Status ändert, behalten Patienten mit intraoperativ stabilem Verlauf (Gruppen 1 und 2) oder mit verlorenem ABR (Gruppe 6) diesen Status auch in der frühen postoperativen Phase in der Regel bei.
Im Hinblick auf mögliche Empfehlungen zur mikrochirurgischen Strategie stellten sich die Phase der knöchernen Eröffnung des IAC sowie die direkte Präparation der Tumor-Nerven-Grenze als besonders gefahrenträchtig heraus. Unter Kenntnis der zwar gegebenen aber begrenzten postoperativen Erholungschancen verschlechterter ABRs muss das chirurgische Vorgehen also in dieser Phase auf schonendste Präparation und rasche Reaktion auf kritisches Monitoring-Feedback ausgerichtet sein.
Die Ergebnisse dieser Studie legen nahe bei Patienten mit auffälligen intraoperativen ABR-Fluktuationen (intraoperative Gruppen 3, 4 und 5) postoperativ ein ABR-Monitoring bis zum 5. Tag nach Duraschluss durchzuführen. Auf der Grundlage der hier vorgelegten Studie ist ein postoperatives ABR-Monitoring gleichermaßen für Patienten mit erhaltenem oder erloschenem ABR jeweils nach Fluktuationen zu empfehlen, da sich intraoperative Fluktuationen fortsetzen und postoperativ sowohl ein ABR-Verlust als auch eine ABR-Erholung erfolgen kann. Dies ist besonders indiziert, wenn in Zukunft die medikamentösen Interventionsmöglichkeiten näher evaluiert und weiterentwickelt sind, aber auch mit den aktuellen Standardmedikamenten HAES, Nimodipin und Kortison ist eine Unterstützung der Regeneration bei Feststellung einer postoperativen Verschlechterung zu empfehlen.
Channelrhodopsin 2 (ChR2) aus dem Augenfleck von C. rheinhardtii gehört zur Gruppe der mikrobiellen Rhodopsine (Typ1-Rhodopsine). ChR2 besteht aus einem extrazellulär gelegenen N-Terminus, 7 Transmembranhelices und einem zytosolisch gelegenen C-Terminus. Der lichtreaktive Bestandteil (Chromophor) all-trans-Retinal ist via Schiff´ Base kovalent an ein Lysinrest der siebten Transmembranhelix gebunden. Bei Applikation von Blaulicht isomerisiert all-trans- zu 13-cis-Retinal, was in einer Konformationsänderung und dem Öffnen des Kanals resultiert. Abhängig vom elektrochemischen Gradienten können ein- und zweiwertige Kationen in die Zelle ein- oder aus der Zelle herausströmen.
Eine retinalabhängige Stabilität konnte bereits für Bakteriorhodopsin (BR) bestätigt werden (Booth, Farooq et al. 1996, Turner, Chittiboyina et al. 2009, Curnow and Booth 2010), bezüglich ChR2 waren bisher nur wenige Daten verfügbar (Hegemann, Gartner et al. 1991, Lawson, Zacks et al. 1991). Die heterologe Expression von wildtypischem und modifiziertem ChR2 in Oozyten von X. laevis erlaubte einen detaillierteren Einblick in die retinalabhängige Stabilität und pH-abhängige Dunkelleitfähigkeit von Guanidinium.
Wildtypisches Chop2 zeigte bei Zugabe von Retinal zum Inkubationsmedium, direkt nach RNA-Injektion, Stromamplituden im µA-Bereich und deutliche Fluoreszenzintensitäten. Ausschließlich endogen vorhandenes Retinal hatte verminderten Fluoreszenzen und Stromamplituden zur Folge, was auf ein geringes Vorhandensein von Chop2-Proteinen in der Plasmamembran hindeutete. Da die Inkubation über Nacht in retinalsupplementierter Lösung nur eine minimale Erhöhung des resultierenden Stromes erbrachte, deuten die in dieser Arbeit erhaltenen Ergebnisse stark auf eine verminderte Stabilität des Proteins bei fehlender Bindung des Kofaktors Retinal.
Das Einfügen einer aromatischen Aminosäure (Y/F/W) an Position 159 führte zu einer, von der Retinalsupplementation unabhängigen, in beiden Ansätzen gleichwertigen Expressionsstärke. Diese äusserte sich in äquivalenten Fluoreszenzintensitäten. Die erhaltenen Stromamplituden wiesen eine starke Differenz auf: ohne Zugabe zusätzlichen Chromophors lag die Stromstärke bei nur wenigen Nanoampere, die bei Inkubation in einer retinalhaltigen Lösung über Nacht auf das Niveau von retinalsupplementierten Oozyten anstieg. Des Weiteren konnte die Zunahme der Stromamplitude innerhalb von 15 Minuten beobachtet werden, wenn die vermessenen Oozyten mit einer retinalhaltigen Lösung perfundiert wurden. Zusammengefasst weisen die Ergebnisse auf eine Stabilisierung des aromatisch substituierten Proteins hin. Bei der von Berndt et al. (2011) beschriebenen Mutante T159C konnten diese Eigenschaften nicht nachgewiesen werden.
Die Modifikation der Retinalbindestelle (K257) in Verbindung mit einer aromatischen Substitution an Position 159 resultierte in deutlichen Fluoreszenzintensitäten, unabhängig von der Retinalverfügbarkeit bei, in beiden Fällen, fehlenden lichtaktivierten Strömen. Diese und die gleichwertigen Bandenstärken des Proteinimmunoblots von aromatisch substituierten ChR2-Varianten unterstützen die Hypothese der retinalunabhängigen Stabilität zusätzlich.
Die Ergebnisse legen, im Falle von Chop2-WT, eine Degradation des Apoproteins nahe. Bei Einfügen einer aromatischen AS an Position 159 ist das Apoprotein davor geschützt (siehe Abb. 75). Infolge der strukturellen Similarität, dem Vorhandensein delokalisierter π-Elektronen und der räumlichen Größe der aromatischen AS ist eine strukturelle Veränderung des Apoproteins denkbar, die eine Degradation aufgrund von nunmehr unzugänglichen Ubiquitinierungsstellen verhindert.
Des Weiteren besteht die Möglichkeit, dass sich bei fehlender Bindung des Kofaktors Wassermoleküle in der Nähe der Bindetasche befinden, welche von umliegenden Aminosäuren (u.a. T159, D156) unter großem Energieaufwand koordiniert werden und die strukturelle Integrität bis hin zur Degradation beeinträchtigen können. Dies könnte durch eine Erhöhung der Hydrophobizität bei Einfügen einer aromatischen Aminosäure verhindert werden.
Bei Substitutionen durch eine aromatische AS (Y/W/F) an Position 159 zeigte sich ein weiteres, bisher nicht beschriebenes, Charakteristikum. Bei Perfusion der Oozyten mit einer guanidiniumhaltigen Lösung, konnten in Abhängigkeit des pH-Wertes ohne die Applikation von Licht Stöme im µA Bereich aufgezeichnet werden. Die Größe der Stromamplitude korreliert hierbei mit dem Anstieg des pH-Wertes und der Konzentration an Guanidiniumionen der perfundierten Lösung und kann durch das Hinzufügen von 1mM Lanthan reversibel geblockt werden. Des Weiteren konnten die vorgenommenen Messungen die Ergebnisse der retinalabhängigen Degradation verifizieren, da der Einstrom von Gua+ sowohl bei retinalsupplementierter Inkubation, als auch bei ausschließlich endogen vorhandenem Retinal zu beobachten war. Des Weiteren zeigte auch die Doppelmutante T159Y/K257R trotz ihres Unvermögens Retinal zu binden, die beschriebenen lichtunabhängigen Ströme.
Die Ergebnisse bei Substitution durch Phenylalanin (F) stellen eine Abweichung des Musters dar. Bei Inkubation von T159F-injizierten Zellen bei ausschließlich endogen vorhandenem Retinal konnte eine stark erhöhte Guanidiniumleitfähigkeit festgestellt werden, diese kam jedoch bei retinalsupplementierter Inkubation nicht zum Tragen. Dies könnte ein Hinweis auf eine sterische Hinderung durch das gebundene Chromophor sein, die bei den Substitutionen durch Tyrosin und Tryptophan, möglicherweise durch unterschiedliche chemische Eigenschaften der AS, nicht auftreten.
Die hervorgerufene pH-Abhängigkeit kann in zwei möglichen Ursachen begründet liegen:
• Vorhandensein einer (de)protonierbaren Gruppe wie Histidin, Arginin oder Lysin, die als pH-Sensor dienen könnte
• Deprotonierung der Schiff´ Base durch Guandininium
Das Vorhandensein eines pH-Sensors konnte durch die vorgenommenen Modifikationen von H114, R115, R120 und H249 nicht bestätigt werden.
Bei Substitution von K257 (in Verbindung mit T159Y) zu Arginin (R) konnte weiterhin ein pH-abhängiger Gua+-Dunkelstrom festgestellt werden. Die Modifikation zu Alanin (A) oder Glutamin (Q) hingegen resultierte im Ausbleiben der Ströme. Der Austausch einer basischen zu einer neutralen Gruppe ohne protonierbaren Rest deutet auf die Beteiligung der Schiff´ Base bzw. der Aminosäure an Position 257 am Mechanismus der Dunkelleitfähigkeit hin.
Serotonin (5-HT) has been implicated in the regulation of emotions as well as in its pathological states, such as anxiety disorders and depression. Mice with targeted deletion of genes encoding various mediators of central serotonergic neurotransmission therefore provides a powerful tool in understanding contributions of such mediators to homeostatic mechanisms as well as to the development of human emotional disorders. Within this thesis a battery of electrophysiological recordings were conducted in the dorsal raphe nucleus (DRN) and the hippocampus of two murine knockout lines with deficient serotonergic systems. Serotonin transporter knockout mice (5-Htt KO), which lack protein responsible for reuptake of 5-HT from the extracellular space and tryptophan hydroxylase 2 knockout (Tph2 KO) mice, which lack the gene encoding the neuronal 5-HT-synthesising enzyme. First, 5-HT1A receptor-mediated autoinhibition of serotonergic neuron firing in the DRN was assessed using the loose-seal cell-attached configuration. Stimulation of 5-HT1A receptors by a selective agonist, R-8-hydroxy-2-(di-n-propylamino)tetralin (R-8-OH-DPAT), showed a mild sensitisation and a marked desensitisation of these receptors in Tph2 KO and 5-Htt KO mice, respectively. While application of tryptophan, a precursor of 5-HT and a substrate of Tph2, did not cause autoinhibition in Tph2 KO mice due to the lack of endogenously produced 5-HT, data from 5-Htt KO mice as well as heterozygous mice of both KO mice lines demonstrated the presence of autoinhibitory mechanisms as normal as seen in wildtype (WT) controls. When the Tph2-dependent step in the 5-HT synthesis pathway was bypassed by application of 5-hydroxytryptophan (5-HTP), serotonergic neurons of both Tph2 KO and 5-Htt KO mice showed decrease in firing rates at lower concentrations of 5-HTP than in WT controls. Elevated responsiveness of serotonergic neurons from Tph2 KO mice correspond to mild sensitisation of 5-HT1A receptors, while responses from 5-Htt KO mice suggest that excess levels of extracellular 5-HT, created by the lack of 5-Htt, stimulates 5-HT1A receptors strong enough to overcome desensitisation of these receptors. Second, the whole-cell patch clamp recording data from serotonergic neurons in the DRN showed no differences in basic electrophysiological properties between Tph2 KO and WT mice, except lower membrane resistances of neurons from KO mice. Moreover, the whole-cell patch clamp recording from CA1 pyramidal neurons in the hippocampus of 5-Htt KO mice showed increased conductance both at a steady state and at action potential generation. Lastly, magnitude of long-term potentiation (LTP) induced by the Schaffer collateral/commissural pathway stimulation in the ventral hippocampus showed no differences among Tph2 KO, 5-Htt KO, and WT counterparts. Taken together, lack and excess of extracellular 5-HT caused sensitisation and desensitisation of autoinhibitory 5-HT1A receptors, respectively. However, this may not directly translate to the level of autoinhibitory regulation of serotonergic neuron firing when these receptors are stimulated by endogenously synthesised 5-HT. In general, KO mice studied here showed an astonishing level of resilience to genetic manipulations of the central serotonergic system, maintaining overall electrophysiological properties and normal LTP inducibility. This may further suggest existence of as-yet-unknown compensatory mechanisms buffering potential alterations induced by genetic manipulations.
In ihrer Evolution mussten Pflanzen Strategien entwickeln um sich sowohl gegen Pathogene aus der Luft als auch solche im Boden zu verteidigen. Diese Resistenzmechanismen der Pflanzen zu verstehen ist von höchster Wichtigkeit für die moderne Gesellschaft. Die Weltbevölkerung wächst schnell, was zu der Notwendigkeit führt, die landwirtschaftlichen Flächen möglichst optimal zu nutzen. Ohne die Weiterentwicklung der landwirtschaftlichen Methoden wird eine ausreichende Versorgung mit Grundnahrungsmitteln nicht möglich sein. Obwohl nicht viele Daten zu diesem Thema vorliegen, ist es sehr wahrscheinlich, dass ein hoher Prozentsatz der jährlichen Ernteverluste auf Pflanzenkrankheiten zurückzuführen ist (Orke et al. 1994, Pinstrup-Andersen; 2001). Der Ernteverlust ist nicht ausschließlich auf den Tod der infizierten Pflanze zurückzuführen, sondern vielmehr auf die sogenannten Resistenzkosten Walters und Heil; 2007). Um sich gegen das Pathogen zu schützen müssen Ressourcen genutzt werden, die sonst für die korrekte Entwicklung der Pflanze, sowie der Samen und Früchte verwendet würden. Die pflanzliche Cuticula, welche die Blattoberfläche bedeckt, ist die erste Verteidigungslinie gegen pathogene Microorganismen, die durch die Luft verbreitet werden. Um diese Barriere zu umgehen nutzen Bakterien und einige Pilze die Stomata als Eingang in den Apoplasten der Blätter. Dies kann durch die Pflanze allerdings verhindert werden, indem diese Poren geschlossen werden. Diese Schließzellantwort wurde zunächst als Teil der Immunantwort auf Bakterien angesehen (Melotto et al. 2006). Nichtsdestotrotz konnte beobachtet werden, dass die Stomata auch während der Infektion des Mehltaupilzes schließen, obwohl dieser nicht durch die Stomata in das Blatt eindringen. Daher haben wir Einzelzellstudien an intakten Gerstenpflanzen vorgenommen um zu klären, wie die Signale erkannt und weitergeleitet werden, die schließlich zum pathogen-induzierten Stomaschluss führen (Koers et al. 2011). Zusammengefasst kann gesagt werden, dass der Stomaschluss ein wichtiger Bestandteil der pflanzlichen Immunantwort ist. Innerhalb dieser Antwort der Stomata auf durch Wind übertragene Pathogene, spielt die Aktivierung der S-Typ Anionenkanäle eine entscheidende Rolle. Es konnte dabei gezeigt werden, dass die Immunantwort die Licht-induzierte Inhibierung dieser Anionenkanäle außer Kraft setzt. S-Typ Anionenkanäle sind aber nicht allein in der Pathogenabwehr von Bedeutung, sondern auch in der Reaktion der Pflanzen auf Trockenstress. Es ist jedoch nicht bekannt, in wie weit sich die beiden Signalwege überschneiden. Zusammen mit den neuen mutierten Gerstenlinien, werden die in dieser Arbeit beschriebenen Techniken zur Messung von Einzelzellen tiefere Einsichten in das Zusammenspiel zwischen Trockenstress und Pathogenabwehr in Pflanzen ermöglichen. Die daraus resultierenden Ergebnisse können zur Optimierung von Getreide für die moderne Landwirtschaft genutzt werden. Dies wird einer der wichtigsten Ansätze sein, um die Menschheit auch in Zukunft mit ausreichend Nahrung versorgen zu können.
Um eine mögliche elektrophysiologische, kardiale Ursache für den plötzlichen Tod von STIM2 Knock-Out Mäusen zu prüfen, wurde eine elektrophysiologische Charakterisierung mittels Ruhe- und Stress-EKG, telemetrischem Langzeit-EKG sowie Elektrophysiologischer Untersuchung durchgeführt. Hierbei konnte keine kardial-elektrophysiologische Grundlage für den plötzlichen Tod dieser Tiere gefunden werden.
Leaf-cutting ants have a highly developed thermal sense which the insects use to regulate the own body temperature and also to optimize brood and fungus development. Apart from the already described temperature guided behaviors inside the nest it is unknown to what extent the ants may use their thermal sense outside the nest. As part of the present thesis, the question was addressed whether leaf-cutting ants (Atta vollenweideri) are able to learn the position of a warm object as landmark for orientation during foraging. Using absolute conditioning, it was shown that ten training trials are sufficient to elicit the association be-tween food reward and the temperature stimulus. In the test situation (without reward) a significantly higher amount of ants preferred the heated site compared to the unheated con-trol. Importantly, thermal radiation alone was sufficient to establish the learned association and served as orientation cue during the test situation (chapter IV). Based on the experi-mental design used in the previous chapter, the localization of thermosensitive neurons, which detect the underlying thermal stimuli, is restricted to the head or the antennae of the ants. The antennal sensillum coeloconicum is a potential candidate to detect the thermal stimuli during the orientation behavior. In chapter V the sensillum coeloconicum of Atta vollenweideri was investigated concerning its gross morphology, fine-structure and the phy-siology of the associated thermosensitive neuron. The sensillum is predominantly located on the apical antennal segment (antennal tip) where around 12 sensilla are clustered, and it has a peg-in-pit morphology with a double walled, multiporous peg. The sensory peg is deeply embedded in a cuticular pit, connected to the environment only by a tiny aperture. The sen-sillum houses three receptor neurons of which one is thermosensitive whereas the sensory modality of the other two neurons remains to be shown. Upon stimulation with a drop in temperature, the thermosensitve neuron responds with a phasic-tonic increase in neuronal activity (cold-sensitive neuron) and shows rapid adaptation to prolonged stimulation. In ad-dition, it is shown that thermal radiation is an effective stimulus for the thermosensitive neuron. This is the first evidence that sensilla coeloconica play an important role during the thermal orientation behavior described in chapter IV. During the test situation of the classic-al conditioning paradigm, the ants showed rapid antennal movements, indicating that they scan their environment in order to detect the heated object. Rapid antennal movements will result in rapid discontinuities of thermal radiation that re-quire thermosensitive neurons with outstanding sensitivity and high temporal resolution. In Chapter VI the question was addressed whether the thermosensitive neuron of the sensilla coeloconica fulfils these preconditions. Extracellular recordings revealed that the neuron is extremely sensitive to temperature transients and that, due to the response dynamics, an estimated stimulus frequency of up to 5 Hz can be resolved by the neuron. Already a tem-perature increase of only 0.005 °C leads to a pronounced response of the thermosensitive neuron. Through sensory adaptation, the sensitivity to temperature transients is maintained over a wide range of ambient temperatures. The discovered extreme sensitivity, the high temporal resolution and the pronounced adaptation abilities are further evidence support-ing the idea that sensilla coeloconica receive information of the thermal environment, which the ants may use for orientation. In order to understand how the ants use their thermal environment for orientation, it is ne-cessary to know where and how thermal information is processed in their central nervous system. In Chapter VII the question is addressed where in the brain the thermal information, specifically received by the thermosensitive neuron of sensilla coeloconica, is represented. By selectively staining single sensilla coeloconica, the axons of the receptor neurons could be tracked into the antennal lobe of Atta vollenweideri workers. Each of the three axons termi-nated in a single functional unit (glomerulus) of the antennal lobe. Two of the innervated glomeruli were adjacent to each other and are located lateral, while the third one was clear-ly separate and located medial in the antennal lobe. Using two-photon Ca2+ imaging of an-tennal lobe projection neurons, the general representation of thermal information in the antennal lobe was studied. In 11 investigated antennal lobes up to six different glomeruli responded to temperature stimulation in a single specimen. Both, warm- and cold-sensitive glomeruli could be identified. All thermosensitive glomeruli were located in the medial half of the antennal lobe. Based on the correlative evidence of the general representation of thermal information and the results from the single sensilla stainings, it is assumed that thermal information received by sensilla coeloconica is processed in the medial of the three target glomeruli. This part of the thesis shows the important role of the antennal lobe in temperature processing and links one specific thermosensitive neuron to its target region (a single glomerulus). In chapter V it was shown that the sensilla coeloconica are clustered at the antennal tip and have an extraordinary peg-in-pit morphology. In the last chapter of this thesis (Chapter VIII) the question is addressed whether the morphology of the sensilla coeloconica predicts the receptive field of the thermosensitive neuron during the detection of thermal radiation. The sensory pegs of all sensilla coeloconica in the apical cluster have a similar orientation, which was not constraint by the shape of the antennal tip where the cluster is located. This finding indicates that the sensilla coeloconica function as a single unit. Finally the hypothesis was tested whether a single sensillum could be direction sensitive to thermal radiation based on its eye-catching morphology. By stimulating the thermosensitive neuron from various angles around the sensillum this indeed could be shown. This is the last and most significant evi-dence that the sensilla coeloconica may be adapted to detect spatially distributed heated objects in the environment during the thermal landmark orientation of ants.
