In this thesis we have used Drosophila melanogaster as a model organism to investigate proteins and their putative interacting partners that are directly or indirectly involved in the release of neurotransmitters at the synapse. We have used molecular techniques to investigate conserved synaptic proteins, synapsin and synapse associated protein of 47 kD (SAP47), and a putative interaction partner of SAP47, tubulin binding chaperone E-like (TBCEL). SAP47 and synapsins are highly conserved synaptic vesicle associated proteins in Drosophila melanogaster. To further investigate the role and function of Sap47 and Syn genes, we had earlier generated the null mutants by P-element mutagenesis (Funk et al., 2004; Godenschwege et al., 2004). Western blots and ELISA of brain homogenates from Sap47156 null mutants showed the presence of up-regulated phospho-synapsin in comparison to wild-type (CS) and the presence of up-regulated phospho-synapsin was partially abolished when a pan-neuronal rescue of SAP47 was performed by the Gal4- UAS technique. Thus, the results suggest a qualitative and quantitative modulation of synapsin by SAP47. At the transcript level, we did not observe any difference in content of Syn transcript in Sap47156 and wild-type CS flies. The question of a direct molecular interaction between SAP47 and synapsin was investigated by co-immunoprecipitation (Co-IP) experiments and we did not find any stable interactions under the several IP conditions we tested. The possibility of Sap47 as a modifier of Syn at the genetic level was investigated by generating and testing homozygous double null mutants of Sap47 and Syn. The Syn97, Sap47156 double mutants are viable but have a reduced life span and decreased locomotion when compared to CS. In 2D-PAGE analysis of synapsins we identified trains of spots corresponding to synapsins, suggesting that synapsin has several isoforms and each one of them is posttranslationally modified. In an analysis by Blue native-SDS-PAGE (BN-SDS-2D- PAGE) and Western blot we observed synapsin and SAP47 signals to be present at 700-900 kDa and 200-250 kDa, respectively, suggesting that they are part of large but different complexes. We also report the possibility of Drosophila synapsin forming homo- and heteromultimers, which has also been reported for synapsins of vertebrates. In parallel to the above experiments, phosphorylation of synapsins in Drosophila was studied by IP techniques followed by 1D-SDS gel electrophoresis and mass spectrometry (in collaboration with S. Heo and G. Lubec). We identified and verified 5 unique phosphorylation sites in Drosophila synapsin from our MS analysis. Apart from phosphorylation modifications we identified several other PTMs which have not been verified. The significance of these phosphorylations and other identified PTMs needs to be investigated further and their implications for synapsin function and Drosophila behavior has to be elucidated by further experiments. In a collaborative project with S. Kneitz and N. Nuwal, we investigated the effects of Sap156 and Syn97 mutations by performing a whole Drosophila transcriptome microarray analysis of the individual null mutants and the double mutants (V2 and V3). We obtained several candidates which were significantly altered in the mutants. These genes need to be investigated further to elucidate their interactions with Sap47 and Syn. In another project, we investigated the role and function of Drosophila tubulin- binding chaperone E-like (Tbcel, CG12214). The TBCEL protein has high homology to vertebrate TBCE-like (or E-like) which has high sequence similarity to tubulin-binding chaperone E (TBCE) (hence the name TBCE-Like). We generated an anti-TBCEL polyclonal antiserum (in collaboration with G. Krohne). According to flybase, the Tbcel gene has only one exon and codes for two different transcripts by alternative transcription start sites. The longer transcript RB is present only in males whereas the shorter transcript RA is present only in females. In order to study the gene function we performed P- element jump-out mutagenesis to generate deletion mutants. We used the NP4786 (NP) stock which has a P(GawB) insertion in the 5’ UTR of the Tbcel gene. NP4786 flies are homozygous lethal due to a second-site lethality as the flies are viable over a deficiency (Df) chromosome (a deletion of genomic region spanning the Tbcel gene and other upstream and downstream genes). We performed the P-element mutagenesis twice. In the first trial we obtained only revertants and the second experiment is still in progress. In the second attempt, jump-out was performed over the deficiency chromosome to prevent homologous chromosome mediated double stranded DNA repair. During the second mutagenesis an insertion stock G18151 became available. These flies had a P-element insertion in the open reading frame (ORF) of the Tbcel gene but was homozygous viable. Western blots of fresh tissue homogenates of NP/Df and G18151 flies probed with anti-TBCEL antiserum showed no TBCEL signal, suggesting that these flies are Tbcel null mutants. We used these flies for further immunohistochemical analyses and found that TBCEL is specifically expressed in the cytoplasm of cyst cells of the testes and is associated with the tubulin of spermatid tails in wild-type CS, whereas in NP/Df and G18151 flies the TBCEL staining in the cyst cells was absent and there was a disruption of actin investment cones. We also found enrichment of TBCEL staining around the actin investment cone. These results are also supported by the observation that the enhancer trap expression of the NP4786 line is localised to the cyst cells, similar to TBCEL expression. Also, male fertility of NP/Df and G18151 flies was tested and they were found to be sterile with few escapers. Thus, these results suggest that TBCEL is involved in Drosophila spermatogenesis with a possible role in the spermatid elongation and individualisation process.
Zur Gattung Bordetella zählen derzeit neun verschiedene Arten Gram-negativer Bakterien. Der strikt humanpathogene Keim B. pertussis ist der Erreger des Keuchhustens und stellt das wohl wichtigste Mitglied der Gattung dar. B. holmesii gehört zu den sogenannten neuen Bordetella-Arten und wurde 1995 erstmals von Weyant et al. beschrieben. In den letzten Jahren gewann B. holmesii als humanpathogener Keim, der Keuchhusten-ähnliche Erkrankungen verursacht, zunehmend an Bedeutung. Mit Ausnahme des BvgAS-Systems (Gerlach et al., 2004) konnten in B. holmesii bislang keine für die „klassischen“ Bordetella-Arten bekannten Virulenzfakoren nachgewiesen werden. Im Rahmen dieser Arbeit wurde ein zum Filamentösen Hämagglutinin homologer Faktor in B. holmesii identifiziert. Der fhaB-Locus aus B. holmesii unterscheidet sich deutlich von dem der anderen Bordetella-Arten, da die fhaB-Sequenz in B. holmesii (fhaBBH) stromaufwärts von einem putativen Membranprotein und stromabwärts von einem IS1001-ähnlichen IS-Element flankiert wird. Sowohl auf DNA- als auch auf Proteinebene weist das fhaBBH-Gen die größte Ähnlichkeit zu dem fhaB-Homolog aus B. avium auf, was die Vermutung bestärkt, dass B. holmesii phylogenetisch im Umfeld von B. avium anzusiedeln ist. Durch in silico-Analysen der FhaBBH-Aminosäuresequenz konnte eine N-terminale Signalpeptiddomäne sowie eine TPS-Domäne identifiziert werden. Dies lässt vermuten, dass FHABH an die bakterielle Oberfläche transportiert und möglicherweise sekretiert wird. Anhand von in silico-Analysen wurde zudem ein KGD-Motiv, nicht jedoch eine Heparinsulfat- und eine Kohlenhydratbindedomäne im FhaBBH identifiziert. Das KGD-Motiv könnte das RGD-Motiv im FHA der „klassischen“ Bordetella-Arten bei der Erkennung von Rezeptoren ersetzen. In Zellkultur-Experimenten konnte die Adäsionsfähigkeit von B. holmesii G7702 an A549-Zellen nachgewiesen werden. Eine fhaB-Deletionsmutante des entsprechenden Stammes zeigte dagegen eine signifikant niedrigere Adhäsionsrate an die menschlichen Lungenepithelzellen. Somit konnte gezeigt werden, dass FHA in B. holmesii eine Rolle als Adhäsionsfaktor spielt. Die im Vergleich zum Wildtyp stark verringerte Adhäsionsrate der bvgA-Mutante von B. holmesii G7702 lässt zudem auf die Beteiligung weiterer Adhäsionsfaktoren an der Wirtszelladhäsion in B. holmesii und deren Regulation durch das BvgAS-Systems schließen. Die Regulationsstudien haben gezeigt, dass die fhaB-Transkription in B. holmesii über zwei konstitutiv aktive Promotoren und einen bvg-abhängigen Promotor erfolgt. In vitro ist phosphoryliertes BvgABH in der Lage, spezifisch an die fhaB-upstream-Region aus B. holmesii zu binden. Innerhalb der fhaBBH-upstream-Region wurden drei putative BvgA-Bindestellen BS2-4 identifiziert, die Ähnlichkeiten zu inverted repeat-Anordnungen der BvgA-Konsensussequenz 5’- T/A T T C C/T T A -3’ aufweisen. Basierend auf den Ergebnissen der in vitro-Binde- und in vivo-Expressionsstudien zur Charakterisierung der einzelnen putativen BvgABH-Bindestellen wird ein Modell für die BvgABH-Bindung innerhalb des fhaB-Promotors in B. holmesii vorgeschlagen. Demnach wird zunächst die primäre BvgABH-Bindestelle BS2 mit der höchsten Affinität von BvgABH-P gebunden, wobei für BvgABH-P eine Dimerisierung angenommen wird. Anschließend bindet ein zweites BvgABH-P-Dimer an die als sekundäre Bindestelle bezeichnete BS3-Region, die sich stromabwärts von BS2 befindet. Eine möglicherweise darauf folgende unspezifische Anlagerung von zwei weiteren BvgABH-P-Dimeren an den 37 bp großen DNA-Abschnitt zwischen BS2 und BS3 ist, ebenso wie die Besetzung der niedrigaffinen Bindestelle BS4 durch ein weiteres BvgABH-P-Dimer, hauptsächlich auf kooperative Protein-Protein-Wechselwirkungen zurückzuführen. Das an BS4 gebundene BvgABH-P-Dimer befindet sich am weitesten stromabwärts und könnte zusammen mit der RNA-Polymerase die Initiation der Transkription gewährleisten. Ergebnisse aus den in vitro-Bindestudien sowie die unterschiedliche Zusammensetzung und Anordnung der putativen BvgA-Bindestellen der fhaB-Promotoren aus B. holmesii und B. pertussis deuten auf eine unterschiedliche Regulation dieser beiden Promotoren hin. So scheint der fhaB-Promotor aus B. holmesii, im Vergleich zum fhaB-Promotor aus B. pertussis, erst bei höheren Konzentrationen an phosphoryliertem BvgA gebunden und somit aktiviert zu werden. In B. holmesii wird vermutlich die konstitutive FHA-Synthese nach BvgAS-Stimulation durch die bvg-abhängige fhaBBH-Expression unterstützt, um eine erfolgreiche Infektion des Wirtes zu gewährleisten.
Best disease, also termed vitelliform macular dystrophy type 2, VMD2, (OMIM #153700), is an autosomal dominant, early onset macular dystrophy associated with a remarkable accumulation of lipofuscin-like material within and beneath the retinal pigment epithelium (RPE). The VMD2 gene mutated in Best disease encodes a 585 amino acid putative transmembrane protein named bestrophin, and is preferentially expressed in the RPE. The protein has a complex membrane topology with 4-6 putative transmembrane domains (TMDs) and is presumably involved in Ca2+-dependent transport of chloride ions across the membrane. The vast majority of known disease-associated alterations are missense mutations nonrandomly distributed across the highly conserved N-terminal half of the protein with clusters near the predicted TMDs. The mechanism connecting Best disease pathology with the identified mutations or the Cl- channel function is not yet clear. To further elucidate the biological function of the bestrophin protein and to identify the molecular mechanisms underlying the disease, a search for interacting partners of bestrophin was performed using the GAL4-based yeast two hybrid system (Y2H). Screening of a bovine RPE cDNA library with various truncated bestrophin baits resulted in the identification of 53 putative interacting partners of bestrophin. However, verification of the interaction has excluded all candidate clones. Our comprehensive Y2H analyses suggest that bestrophin may not be suitable for traditional yeast two hybrid screens likely due to the fact that the protein is integral to the membrane and even fragments thereof may not be transported to the nucleus which is, however a prerequisite for protein interaction in the yeast system. Bestrophin belongs to a large family of integral membrane proteins with more than 100 members identified to date originating from evolutionarily diverse organisms such as mammals, insects and worms. The most distinctive feature of the bestrophin family, besides the invariant RFP (arginine-phenylalanine-proline) domain, is an evolutionarily highly conserved N-terminal region. To clarify the phylogenetic relationship among bestrophin homologues and to identify structural and functional motifs conserved across family members, a bioinformatics/phylogenetic study of the conserved N-terminal region was conducted. Phylogenetic analysis of the bestrophin homologues reveals existence of four evolutionary conserved family members in mammals, with high homology to the human VMD2, VMD2-L1 to L3 proteins. The significant level of protein sequence similarity between divergent species suggests that each of the bestrophin family members has a unique, Chapter One: Summary 2 evolutionarily conserved function and that the divergence of bestrophin into several family members occurred before the divergence of individual mammalian species.
The bacteriophage Lambda is a virus which infects bacteria carrying LamB protein in their outer membrane. GpJ, a protein of the tail of the phage, is involved in the binding to LamB. The study of the interaction between GpJ expressed as fusion protein and LamB was performed in order to investigate the interaction between the bacteriophage Lambda and LamB. The fusion proteins are called MBP-gpJ and His-gpJ. MBP-gpJ is a chimeric protein representing Maltose Binding Protein connected to the Cterminal part of the GpJ protein (residue 684 until 1132), graciously given by Pr. Charbit (Paris, France). MBP-gpJ, expressed in E.coli and purified, bound to the exoplasmic side of LamB and LamB variants in planar lipid bilayer experiments and allowed a complete and reversible blockage of LamB channels. In order to obtain data about the binding of the GpJ fragment alone to LamB, an other fusion protein without MBP was created, called His-gpJ. His-gpJ is the C-terminal part of GpJ (684-1132) in fusion with a 6×Histidine-tag, produced as insoluble form in E.coli. After renaturation, a soluble protein can be obtained. Without MBP, the GpJ fragment still bound to LamB in planar lipid bilayer experiments, but did not block significantly its channels, as previously observed after addition of MBP-gpJ. The interaction between His-gpJ and LamB or LamB mutants was also demonstrated on SDSPAGE and immunodetection by the presence of high molecular mass bands. Furthermore, the use of variants of lamB allowed to demonstrate that the C-terminal fragment of GpJ does not bind to the same area on the surface of LamB than GpJ involved in the tail of the Lambda phage.
Funktionelle Charakterisierung neuer Proteine der inneren Kernmembran von Drosophila melanogaster: Drosophila Lamin B Rezeptor (dLBR), ein integrales Membranprotein der inneren Kernmembran; Bocksbeutel alpha und Bocksbeutel beta, LEM-Domänen Proteine sowie deren potentiellen Interaktionspartner Drosophila Barrier-to-Autointegration Factor (dBAF).
Listeria monocytogenes, ein Gram-positives, fakultativ intrazelluläres Bakterium, kann bei immunsupprimierten Menschen schwere Infektionen des Zentralnervensystems auslösen. In Folge seines ubiquitären Vorkommens, sowie seiner hohen Resistenz gegenüber Lebensmittel-Konservierungsmethoden besteht ein großes Interesse an seiner raschen Identifizierung und Differenzierung gegenüber den apathogenen Spezies seiner Gattung. Sein P60 als essentielles Housekeeping-Gen bietet sich auf Grund der zwischen den einzelnen Spezies konservierten und variablen Bereiche für die Etablierung gattungs- und speziesspezifischer Nachweissysteme an. Mit Hilfe des EPITOP-SPOT-Mappings wurde eine Immunogenitätskarte des P60 von L. innocua bzw. L. monocytogenes erstellt, P60-spezifische CD4-T-Zellinien charakterisiert und das Epitop eines dieser T-Zellklone exakt bestimmt. Transferexperimente mit diesen T-Zelllinien und Boosterinfektionen mit L. monocytogenes bzw. L. innocua zeigten, dass L. innocua alleine zwar nicht in der Lage ist, einen Immunschutz gegen L. monocytogenes zu etablieren, diesen jedoch in vitro und in vivo erhalten kann, indem es durch kreuzreaktive Epitope Gedächtnis-T-Zellen stimuliert. Die in vitro-Transkription von Phly, PplcA und PactA erfolgt strikt PrfA-abhängig, während Piap, PprfA1/2 und - unerwarteter Weise - auch Pmpl PrfA-unabhängig transkribiert werden. Sie erfolgt - ausgenommen bei PprfA - nur bei ausreichender Verfügbarkeit von ATP nicht jedoch GTP. Um eine effiziente Transkription zu gewährleisten, müssen die ersten drei initiierenden Nukleotide in höherer Konzentration vorliegen. Die verschiedenen, über eine Heparinsäule aufgetrennten RNAP-Fraktionen von L. monocytogenes zeigten je nachdem, ob die Kulturen einer Hitzeschockbehandlung bei 48°C (RNAP48) ausgesetzt, in MEM geshiftet (RNAPMEM) oder aber direkt aus dem BHI-Medium (RNAPBHI) geerntet wurden, mit den oben genannten Promotoren unterschiedliche Aktivitätsprofile. Demnach benötigen actA und hly für ihre optimale Transkription womöglich einen alternativen Sigmafaktor (als Sigma-43).
The identification of NRAGE

The inhibitor of apoptosis proteins (IAPs) have been shown to interact with a growing number of intracellular proteins and signalling pathways in order to fulfil their anti-apoptotic role. In order to investigate in detail how the avian homologue ITA interfered with both TNF induced apoptosis and the NGF mediated differentiation in PC12 cells, a two hybrid screen was performed with a PC12 library using ITA as a bait. The screen resulted in the identification of several overlapping fragments of a previously unknown gene. The complete cDNA for this gene was isolated, the analysis of which revealed a high homology with a large family of tumour antigens known as MAGE (melanoma associated antigens). This newly identified member of the MAGE family, which was later named NRAGE, exhibited some unique characteristics that suggested for the first time a role in normal cellular physiology for this protein family. MAGE proteins are usually restricted in their expression to malignant or tumour cells, however NRAGE was also expressed in terminally differentiated adult tissue. NRAGE also interacted with the human XIAP in direct two-hybrid tests. The interactions observed in yeast cells were confirmed in mammalian cell culture, employing both coimmunoprecipitation and mammalian two-hybrid methods. Moreover, the results of the coimmunoprecipitation experiments indicated that this interaction requires the RING domain. The widely studied 32D cell system was chosen to investigate the effect of NRAGE on apoptosis. NRAGE was stably transduced in 32D cells, and found to augment cell death induced by the withdrawal of Interleukin-3. One reason for this reduced cell viability in NRAGE expressing cells could be the binding of endogenous XIAP, which occurred inducibly after growth factor withdrawal. Interestingly, NRAGE was able to overcome the protection afforded to 32D cells by the exogenous expression of human Bcl-2. Thus NRAGE was identified during this research doctorate as a novel pro-apoptotic, IAP-interacting protein, able to accelerate apoptosis in a pathway independent of Bcl-2 cell protection.
B-Zell-Lymphome vom mukosa-assoziierten lymphatischen Gewebe (MALT)-Typ sind die größte Gruppe der extranodalen Lymphome. Sie enstehen vor dem Hintergrund einer chronischen Entzündung, wie etwa einer Helicobacter pylori-assoziierten Gastritis im Magen. Die Mechanismen der Lymphomgenese sind weitgehend unverstanden. Der Befund, dass die malignen B-Zellen der MALT-Typ Lymphome autoreaktiv sind und durch Antigen oder T-Zell-vermittelte Signale stimuliert werden, weist auf ein mögliches Ver-sagen der T-Zell-Kontrolle hin. Zur Prüfung dieser Hypothese wurden in der vorliegenden Arbeit sowohl tumor-infiltrierende T-Zellen als auch maligne B-Zellen aus MALT-Typ Lymphomen untersucht. Mit Hilfe von Expressionsanalysen der Vb -Kette des T-Zell-Rezeptors (TCR) wurden antigen-induzierte klonale Expansionen in den tumor-infiltrierenden T-Zellen nachgewiesen. Weiterhin wurden ähnliche antigen-erkennende Regionen (CDR3) des TCR in tumor-infiltrierenden T-Zellen aus zwei verschiedenen MALT-Typ Lymphomen gefunden. Diese Ergebnisse charakterisieren die T-Zellen als funktionell und auch als potenziell tumor-reaktiv. Um mögliche Defekte bei malignen B-Zellen zu untersuchen, wurde eine in vitro T/B-Zell-Kokultur entwickelt. Damit war es möglich, die Interaktion des apoptose-induzierenden Oberflächenmoleküls FasL auf aktivierten T-Zellen mit dem entsprechenden Todesrezeptor Fas auf malignen B-Zellen zu untersuchen. Drei von sieben MALT-Typ Lymphomen und vier von fünf DLBL erwiesen sich als resistent gegen FasL-vermittelte Apoptose. Meine Untersuchungen deuten darauf hin, dass hierfür eine mutationsbedingte funktionelle Inaktivierung des Fas-Rezeptors verantwortlich ist. In Fas-Transkripten aller untersuchten malignen B-Zellen wurden insgesamt 14 verschiedene Punktmutationen gefunden, die zu Aminosäureaustauschen bei der Translation führen. Zehn dieser Mutationen waren mit der Apoptose-Resistenz maligner B-Zellen assoziiert. Durch ergänzende Untersuchungen konnten alternative Mechanismen der Apoptose-Resistenz wie etwa reduzierte Fas-Expression, Produktion von löslichem Fas (sFas) oder Störungen in der Fas-Signalkaskade weitgehend ausgeschlossen werden. Aus diesen Ergebnissen lässt sich folgender Schluss ziehen: Resistenz gegen FasL/Fas-vermittelte Apoptose ist ein Mechanismus der frühen MALT-Typ Lymphomgenese und möglicherweise auf bestimmte Fas-Mutationen zurückzuführen. Durch Apoptose-Resistenz entkommen die malignen B-Zellen der MALT-Typ Lymphome der vorhandenen T-Zell-Kontrolle. Gleichzeitig nehmen sie die parakrin angebotene T-Zell-Hilfe solange in Anspruch, bis sie völlige Autonomie erreicht haben. Durch das abnorm verlängerte Überleben der B-Zellen steigt dann die Wahrscheinlichkeit, weitere Aberrationen wie etwa die in 50 Prozent aller Fälle auftretende Chromosomentranslokation t(11;18)(q21;q21) zu erwerben.
