Das Cytokin Interleukin-4 (IL-4) ist ein essentieller Faktor bei der Entstehung von Sofort-Typ Allergien. Die Bindung von IL-4 an seinen Rezeptor und die anschließende Phosphorylierung des IL-4 aktivierten Transkriptionsfaktors Stat6 ist ein Schlüsselereignis bei der allergischen Immunantwort. In der vorliegenden Arbeit werden Ergebnisse zur Hemmung der Stat6 vermittelten Signaltransduktion des IL-4 Rezeptors vorgestellt. Dazu wurde ein Vektorsystem etabliert, bei dem ein von dem Drosophila-Transkriptionsfaktor Antennapedia abgeleitetes 16 AS langes Peptid benutzt wird. Dieses Antennapediapeptid kann Plasmamembranen lebender Zellen energie- und rezeptorunabhängig durchqueren und dabei andere hydrophile Moleküle mittransportieren. Stat6 bindet über eine SH2 Domäne an phosphorylierte Reste von IL4Ra und bildet, nachdem es selbst phosphoryliert ist, mit anderen Stat6-Molekülen aktive Dimere. Ein aus der Stat6-Bindestelle des IL-4Ra abgeleitetes phosphoryliertes Peptid (Stat6BP) wurde mit Hilfe des Antennapediapeptids in verschiedene humane und murine Zellinien transportiert. Für Stat6BP konnte mit Hilfe von spezifischer Immunpräzipitation und Western-Blot gezeigt werden, dass es IL-4 induzierte Phosphorylierung und Aktivierung von Stat6 transient hemmen kann. Durch zusätzliche Applikation des Tyrosinphosphataseinhibitors Natriumpervanadat gelang es, die hemmende Wirkung von Stat6BP zu verlängern. Unter gleichen Bedingungen konnte auch gezeigt werden, dass Stat6BP spezifisch die Aktivierung von Stat6 hemmt, da die durch IL-4 oder IL-3 induzierte Phosphorylierung des eng verwandten Stat5 völlig unbeeinträchtigt bleibt. Ferner wurde durch das Peptid die Expression eines Stat6 kontrollierten Reportergens gehemmt. Im Rahmen dieser Arbeit wurde außerdem die Rolle der Src-Typ Kinasen p56lck und p59fyn in der IL-4 Signaltransduktion in unterschiedlichen T-Zellinien untersucht. Es zeigte sich, dass die Aktivierung der beide Kinasen stark von der getesteten Zellinie abhängt. In einigen T-Zellinien aktiviert IL-4 eher p56lck, in anderen eher p59fyn.
Bone Morphogenetic Proteins (BMPs) are key regulators for a lot of diverse cellular processes. During embryonic development these proteins act as morphogens and play a crucial role particularly in organogenesis. BMPs have a direct impact on distinct cellular fates by means of concentration-gradients in the developing embryos. Using the diverse signaling input information within the embryo due to the gradient, the cells transduce the varying extracellular information into distinct gene expression profiles and cell fate decisions. Furthermore, BMP proteins bear important functions in adult organisms like tissue homeostasis or regeneration. In contrast to TGF-ß signaling, currently only little is known about how cells decode and quantify incoming BMP signals. There is poor knowledge about the quantitative relationships between signal input, transducing molecules, their states and location, and finally their ability to incorporate graded systemic inputs and produce qualitative responses. A key requirement for efficient pathway modulation is the complete comprehension of this signaling network on a quantitative level as the BMP signaling pathway, just like many other signaling pathways, is a major target for medicative interference. I therefore at first studied the subcellular distribution of Smad1, which is the main signal transducing protein of the BMP signaling pathway, in a quantitative manner and in response to various types and levels of stimuli in murine c2c12 cells. Results indicate that the subcellular localization of Smad1 is not dependent on the initial BMP input. Surprisingly, only the phospho-Smad1 level is proportionally associated to ligand concentration. Furthermore, the activated transducer proteins were entirely located in the nucleus. Besides the subcellular localization of Smad1, I have analyzed the gene expression profile induced by BMP signaling. Therefore, I examined two endogenous immediate early BMP targets as well as the expression of the stably transgenic Gaussia Luciferase. Interestingly, the results of these independent experimental setups and read-outs suggest oscillating target gene expression. The amplitudes of the oscillations showed a precise concentration-dependence for continuous and transient stimulation. Additionally, even short-time stimulation of 15’ activates oscillating gene-expression pulses that are detectable for at least 30h post-stimulation. Only treatment with a BMP type I receptor kinase inhibitor leads to the complete abolishment of the target gene expression. This indicated that target gene expression oscillations depend directly on BMP type I receptor kinase activity.
BMPs influence a variety of cellular processes. They have been shown to regulate proliferation, differentiation, migration and apoptosis and thus play central roles during developmental processes and tissue homeostasis. Ligand mediated signal transduction is transmitted via BMP type I and BMP type II receptors, both members of the serine/threonine kinase superfamily. The BMP receptor mediated signal transduction is not explored in detail. Therefore our aim was to address different aspects of BMP mediated signal transduction with main focus on BRII and its regulation. Due to the existence of two alternative splice variants, a long and a short form, the function of the two variants and the impact of the C-terminal extension are of general interest. Moreover, mutations in the BMPR2 gene were identified to be responsible for PPH, a autosomal dominant lung disease. In this thesis, BRII phosphorylation and signalling mediated by different receptor oligomers were investigated and multiple BRII associated proteins were identified. We could show that the oligomerization pattern of BMP receptors exhibits a higher degree of flexibility compared to other receptors of that superfamily. In the present work the BMP2 mediated signal transduction should be examined, depending on the receptor oligomerization pattern. Using kinase-deficient mutants, it could be demonstrated, that signalling via preformed BMP receptor complexes is mediated by the well characterized Smad1/5/8 pathway, whereas signalling initiated by BMP2 induced recruitment of the receptors activates the p38 pathway and leads to Alkaline Phosphatase production. To further study signalling events triggered directly from the BRII a proteomics-based screen for BRII associated proteins was performed. 53 associated proteins were found, the majority being signal transducing molecules, but in addition metabolic proteins, transcriptional regulators and others were identified. These proteins enable to gain a deeper insight in BMP mediated signalling. One of the interactors, the receptor tyrosine kinase c-kit, was characterized in more detail. It could be demonstrated, that BRII and c-kit form a complex in vitro and in vivo, and the interaction is enhanced upon BMP2 stimulation. 2D phosphopeptid mapping showed that BRII is phosphorylated at S757 upon activation of c-kit by SCF. Moreover, c-kit and its ligand SCF are modulating BMP2 pathways, by enhancing Smad1/5 phosphorylation, Smad-transcriptional activity, Alkaline Phosphatase production and expression of Cbfa1. All these pathways hint towards modulation of the osteoblast development via c-kit. Thus, we were able to develop a novel paradigm for the BMP2 meditated signalling. One of the initial triggers for BRII is the auto-phosphorylation of BRII. Here we analyze ligand-independent as well as ligand-dependent phosphorylation of BRII. Some phosphorylation sites in BRII were identified. The general phosphorylation occurs mostly on serines. S815, S818 and Y825 are identified targets of phosphorylation whose function is still unclear. However phosphorylation of S336 is demonstrated to be essential for BRII activation. The elucidation of BMP receptor phosphorylation and oligomerization as well as the impact of a number of BRII associated proteins (such as c-kit), demonstrated in this thesis that BMP signalling has to be regulated precisely on multiple levels. This can be useful for the development of selective signalling inhibitors for basic research and therapeutic approaches of PPH and other diseases.
Transforming-Growth-Factor-beta1 (TGF-b1) is a multifunctional cytokine that regulates cell growth and differentiation in many types of cells. TGF-b1 is especially known to exert a variety of regulatory functions in the immune system, such as T cell differentiation and T cell function. Signal transduction of TGF-b1 is mediated by phosphorylation of receptorassociated Smad proteins (R-Smads). R-Smads are phosphorylated by the activated type I receptor, which is itself phosphorylated by the high affinity type II receptor upon ligand binding. The phosphorylated R-Smads then associate with Co-Smads. Heterooligomers of R- and Co-Smads translocate into the nucleus where they regulate transcription of target genes in concert with other transcription factors such as CBP/p300 or AP-1. Recent findings suggest that the pleiotropic effects of TGF-b1 are conferred by crosstalks to other signal transduction pathways such as the MAP-kinases or the STAT-pathway. Here we describe the effect of long-term exposure to TGF-b1 on the effector function of differentially stimulated primary murine splenocytes and purified primary murine CD8+ cytotoxic T cells. Long-term exposure to TGF-b1 results in non-responsiveness to TGF-b1- induced Smad2 phosphorylation. This is seen either by no phosphorylation or sustained phosphorylation of Smad2. Furthermore, we observed a strong correlation between sustained Smad2 phosphorylation and resistance to TGF-b1 mediated growth inhibition. In contrast, splenocyte cultures strongly growth inhibited by TGF-b1 showed no Smad2 phosphorylation. Lytic activity of these cultures, however, was found to be suppressed regardless of proliferation properties and Smad2 phosphorylation pattern. We also describe that a functional MEK-1 pathway is a prerequisite for rendering murine splenocytes unresponsive to TGF-b1 mediated growth inhibition, and that inhibition of the MEK-1 cascade alters the Smad2 phosphorylation pattern. In addition, we show that resistance to TGF-b1 mediated growth inhibition correlates with the activation of the JNK pathway. However, the resistant phenotype was found unable to be reverted upon administration of exogeneous IFNg and/or aCD28 antibody. In human or mouse T cell lines, however, the described correlation between the type of stimulation and TGF-b growth resistance or growth sensitivity is not present. Thus, this correlation is specific for primary T cells. We also cloned a chimeric dominantnegative TGF-b receptor which is coupled to a suicide gene, in order to render T cells resistant to TGF-b mediated effects.These findings shed light on how TGF-b1 mediates its immunosuppressive role, and may help to gain knowledge of averting these TGF-b1 effects in the course of tumor therapy.
Das pleiotrope Zytokin TNF (tumor necrosis factor) kann an den TNF-Rezeptor 1 (TNFR1) und den TNF-Rezeptor 2 (TNFR2) binden und mit deren Hilfe seine biologischen Funktionen über verschiedene Signalwege, wie z.B. NFB- und MAPK-Aktivierung bzw. Apop¬toseinduktion, vermitteln. In früheren Arbeiten konnte gezeigt werden, dass die Aktivierung des TNFR2 zur proteasomalen Degradation des Adaterproteins TRAF2 führt und dadurch die TNFR1-induzierte Apoptose verstärkt wird. TWEAK (tumor necrosis like weak inducer of apoptosis), das ebenfalls der TNF-Ligandenfamilie angehört und die Interaktion mit dessen Rezeptor Fn14 (fibroblast growth factor-inducible 14), der wie der TNFR2 zur Untergruppe der TRAF-bindenden Rezeptoren der TNF-Rezeptorfamilie gehört, zeigten in verschiedenen Arbeiten auch eine TRAF2-degradierende Wirkung. In der vorliegenden Arbeit konnte nun gezeigt werden, dass dies auch im Falle des TWEAK/Fn14-Systems mit einem verstärkenden Effekt auf die TNFR1-vermittelte Apoptose einhergeht. Darüber hinaus konnte gezeigt werden, dass TWEAK zusätzlich auch die TNFR1-induzierte Nekrose verstärkt, die den Zelltod durch andere Mechanismen als bei der Apoptose induziert. Von anderen Arbeiten unserer Gruppe war bekannt, dass lösliches TWEAK (sTWEAK) und membranständiges TWEAK (mTWEAK) bezüglich der TRAF2-Depletion wirkungs¬gleich sind. Da der apoptotische Fn14-TNFR1-„crosstalk“ auf der Depletion von TRAF2-Komplexen beruht wurden auch keine signifikanten Unterschiede zwischen sTWEAK und mTWEAK in Bezug auf die Verstärkung der TNFR1-induzierten Apoptose beobachtet. Interessanter¬weise zeigte sich in der vorliegenden Arbeit jedoch, dass sTWEAK den klassischen NFB-Signalweg gar nicht bzw. nur schwach aktiviert, wohingegen mTWEAK diesen stark induziert. Bei der Aktivierung des alternativen NFB-Signalweges hingegen ließen sich keine Unterschiede zwischen sTWEAK und mTWEAK erkennen. Die Aktivierung eines Signalweges wird also durch die Oligomerisierung des Liganden nicht moduliert, demgegenüber aber erwies sich die Aktivierung eines anderen Signalweges als stark abhängig von der Liganden-Oligomerisierung. Vor dem Hintergrund, dass das Adapterprotein TRAF1 (TNF-receptor-associated factor 1) Heterotrimere mit TRAF2 bildet, wurde weiterhin untersucht, ob dieses Molekül einen Einfluss auf die Aktivität der TWEAK-induzierten Signalwege hat. Tatsächlich zeigte sich in TRAF1-exprimie¬renden Zellen eine Verstärkung der TWEAK-induzierten Aktivierung des klassischen NFB-Signalweges Zukünftige Studien müssen nun aufklären, inwieweit die hier gefundenen Mecha-nismen das Zusammenspiel von TNF und TWEAK in vivo bestimmen.
Bakterien müssen ständig in der Lage sein auf Veränderungen in ihrer Umwelt reagieren zu können. Zur Wahrnehmung dieser Veränderungen haben sich unterschiedliche Signaltransduktionssysteme entwickelt. Ein weit verbreiteter und gut charakterisierter Mechanismus zur Signaltransduktion sind die so genannten Zwei-Komponentensysteme. Im Genom von H. pylori konnten nur wenige Bestandteile von Zwei-Komponentensystemen identifiziert werden. Dazu zählen neben dem Chemotaxis-System lediglich drei Histidin-Kinasen, ArsS, CrdS und FlgS, und fünf Response-Regulatoren HP1021, HP1043, ArsR, CrdR und FlgR, die vermutlich Transkriptions-regulatorische Funktionen haben. Zwei der Response-Regulatoren, HP1043 und ArsR als essentiell für das Überleben von H. pylori, während HP1021 einen deutlichen Einfluss auf das Zellwachstum hat, da ein Wachstums-Defekt zu erkennen ist, wenn das entsprechende Gen hp1021 deletiert wird. Eine Deletion von arsS, dem Gen der zugehörigen Histidin-Kinase von ArsR, hat unter Standard-Wachstumsbedingungen keine Auswirkung auf das Zellwachstum von H. pylori. Diese Beobachtung spricht für die Hypothese, dass der Response-Regulator ArsR die Transkription zweier unterschiedlicher Sets von Zielgenen kontrolliert. Demzufolge reguliert der Response-Regulator ArsR nach Säure-induzierter Phosphorylierung durch ArsS die Transkription von Genen, die zur Säureresistenz beitragen, während ArsR im nicht-phosphorylierten Zustand die Transkription von weiteren Zielgenen kontrolliert, von denen mindestens eines für das Zellwachstum essentiell sein sollte. Das durch ArsR~P kontrollierte Regulon konnte bereits weitgehend charakterisiert werden allerdings sind die Zielgene des unphosphorylierten Regulators bislang unbekannt. In der vorliegenden Arbeit konnte die zuvor beschriebene Hypothese bestätigt werden, da gezeigt wurde, dass ein Derivat von ArsR, mit einer Mutation der Phosphorylierungsstelle D52 zu N52, das wildtypische Protein bezüglich des Zellwachstums unter Standardbedingungen funktionell ersetzen kann. Für die Response-Regulatoren HP1021 und HP1043 konnte bislang keine zugehörige Histidin-Kinase identifiziert werden und interessanterweise findet man in der Receiver-Domäne dieser Response-Regulatoren atypische Abweichungen von der Konsensus-Sequenz. Um die Bedeutung dieser atypischen Primärsequenzen für die Funktion dieser Response-Regulatoren zu untersuchen wurden mutierte H. pylori-Stämme konstruiert, die ausschließlich Derivate von HP1021 bzw. HP1043 exprimieren, die in ihrer Receiver-Sequenz der Konsensus-Sequenz entsprachen. Da diese Mutanten sich bezüglich ihres Zellwachstums nicht vom Wildtyp unterscheiden, konnte nachgewiesen werden, dass die atypischen Receiver-Sequenzen der beiden Response-Regulatoren nicht entscheidend für die Funktionen der Response-Regulatoren sind. Weiterhin konnten Indizien dafür gesammelt werden, dass HP1021 und HP1043 hinsichtlich ihrer Aktivierung vermutlich vom üblichen Zwei-Komponentenparadigma abweichen. Derivate von HP1021 und HP1043 mit Mutationen ihrer putativen atypischen Phosphorylierungsstelle sind in der Lage ihre wildtypischen Pendants hinsichtlich der bekannten Phänotypen funktionell zu ersetzen. Somit ist eine Phosphorylierung der Receiver-Domäne dieser Response-Regulatoren keine Voraussetzung für ein normales Zellwachstum von H. pylori. Diese Hypothese wird gestützt durch die Beobachtung, dass ein Ortholog von HP1043 aus C. jejuni CJ0355, das natürlicherweise an der potentiellen Phosphorylierungsstelle einen nicht phosphorylierbaren Aminosäurerest trägt, HP1043 in seiner Funktion ersetzen kann. Es konnte gezeigt werden, dass in vitro keine Phosphorylierung durch radioaktiv markiertes Acetylphosphat stattfindet und dass ein H. pylori-Stamm mit einer Deletion der Gene pta und ackA, welche Proteine kodieren, die bei der Synthese von zellulärem Acetylphosphat benötigt werden, einen normalen Wachstums-Phänotyp zeigt. Zusätzlich konnten in einer massenspektrometrischen Analyse des Proteins HP1021, welches nach Zweidimensionaler Gelelektrophorese von Gesamtzellproteinlysaten aus H. pylori isoliert wurde, keine Hinweise auf eine Serinphosphorylierung entdeckt werden. Es ist daher fraglich ob in vivo eine funktionell relevante Phosphorylierung stattfindet. Die Mechanismen zur Modulation der Regulator-Aktivität von HP1043 und HP1021 bleiben unklar. In der vorliegenden Arbeit konnte demonstriert werden, dass eine strikte Transkriptionskontrolle nicht für die Zellwachstums-assoziierten Funktionen von HP1021 von Bedeutung ist. Dagegen wurden Hinweise darauf erzielt, dass die Expression von HP1043 auf einem posttranskriptionellen und/oder auf einem posttranslationellen Level reguliert wird. Es waren bislang keine Zielgene von HP1021 bekannt. Durch vergleichende Zweidimensionale Gelelektrophorese der H. pylori Stämme 26695 und 26695 1021Δ konnten einige potentielle Zielgene des Response-Regulators HP1021 identifiziert werden.
Bone Morphogenetic Proteins (BMPs) are secreted multifunctional signaling proteins that play an important role during development, maintenance and regeneration of tissues and organs in almost all vertebrates and invertebrates. BMPs transmit their signals by binding to two types of serine-/threonine-kinase receptors. BMPs bind first to their high affinity receptor, thereby recruiting their low affinity receptor into the complex. This receptor assembly starts a Smad (Small mothers against decapentaplegic) protein signaling cascade which regulates the transcription of responsive genes. Up to date, only seven type I and five type II receptors are known for more than 30 ligands. Therefore, many BMP ligands can recruit more than one receptor subtype. Vice versa, receptors can bind to several ligands, indicating a highly promiscuous ligand-receptor interaction. This raises the following questions: (i) How are BMPs able to induce ligand-specific signals, despite forming complexes with identical receptor composition and (ii) how are they able to recognize and bind various binding partners in a highly specific manner. From the ligand’s point of view, heterodimeric BMPs are valuable tools for studying the interplay between different sets of receptors, thereby providing new insights into how the various BMP signals can be generated. This study describes the expression and purification of the heterodimers BMP-2/6 and -2/7 from E.coli cells. BIAcore interaction studies and various in vitro cell activity assays revealed that the generated heterodimers are biologically active. Furthermore, BMP-2/6 and -2/7 exhibit a higher biological activity in most of the cell assays compared to their homodimeric counterparts. In addition, the BMP type I receptor BMPR-IA is involved in heterodimeric BMP signaling. However, the usage of other type I receptor subtypes (e.g. ActR-I) building a heteromeric ligand-receptor type I complex as indicated in previous works could not be determined conclusively. Furthermore, BMP heterodimers seem to require only one type I receptor for signaling. From the receptors’ point of view, the BMP type I receptor BMPR-IA is a prime example for its promiscuous binding to different BMP ligands. The extracellular binding interface of BMPR-IA is mainly unfolded in its unbound form, requiring a large induced fit to adopt the conformation when bound to its ligand BMP-2. In order to unravel whether the binding promiscuity of BMPR-IA is linked to structural plasticity of its binding interface, the interaction of BMPR-IA bound to an antibody Fab fragment was investigated. The Fab fragment was selected because of its ability to recognize the BMP-2 binding epitope on BMPR-IA, thus neutralizing the BMP-2 mediated receptor activation. This study describes the crystal structure of the complex of the extracellular domain of BMPR-IA bound to the antibody Fab fragment AbyD1556. The crystal structure revealed that the contact surface of BMPR-IA overlaps extensively with the contact surface of BMPR-IA for BMP-2 interaction. Although the contact epitopes of BMPR-IA to both binding partners coincide, the three-dimensional structures of BMPR-IA in both complexes differ significantly. In contrast to the structural differences, alanine-scanning mutagenesis of BMPR-IA showed that the functional determinants for binding to both the antibody and BMP-2 are almost identical. Comparing the structures of BMPR-IA bound to BMP-2 or to the Fab AbyD1556 with the structure of unbound BMPR-IA revealed that binding of BMPR-IA to its interaction partners follows a selection fit mechanism, possibly indicating that the ligand promiscuity of BMPR-IA is inherently encoded by structural adaptability.
A subtly regulated and controlled course of cellular processes is essential for the healthy functioning not only of single cells, but also of organs being constituted thereof. In return, this entails the proper functioning of the whole organism. This implies a complex intra- and inter-cellular communication and signal processing that require equally multi-faceted methods to describe and investigate the underlying processes. Within the scope of this thesis, mathematical modeling of cellular signaling finds its application in the analysis of cellular processes and signaling cascades in different organisms. ...
Zars and co-workers were able to localize an engram of aversive olfactory memory to the mushroom bodies of Drosophila (Zars et al., 2000). In this thesis, I followed up on this finding in two ways. Inspired by Zars et al. (2000), I first focused on the whether it would also be possible to localize memory extinction.While memory extinction is well established behaviorally, little is known about the underlying circuitry and molecular mechanisms. In extension to the findings by Zars et al (2000), I show that aversive olfactory memories remain localized to a subset of mushroom body Kenyon cells for up to 3 hours. Extinction localizes to the same set of Kenyon cells. This common localization suggests a model in which unreinforced presentations of a previously learned odorant intracellularly antagonizes the signaling cascades underlying memory formation. The second part also targets memory localization, but addresses appetitive memory. I show that memories for the same olfactory cue can be established through either sugar or electric shock reinforcement. Importantly, these memories localize to the same set of neurons within the mushroom body. Thus, the question becomes apparent how the same signal can be associated with different events. It is shown that two different monoamines are specificaly necessary for formation of either of these memories, dopamine in case of electric shock and octopamine in case of sugar memory, respectively. Taking the representation of the olfactory cue within the mushroom bodies into account, the data suggest that the two memory traces are located in the same Kenyon cells, but in separate subcellular domains, one modulated by dopamine, the other by octopamine. Taken together, this study takes two further steps in the search for the engram. (1) The result that in Drosophila olfactory learning several memories are organized within the same set of Kenyon cells is in contrast to the pessimism expressed by Lashley that is might not be possible to localize an engram. (2) Beyond localization, a possibible mechanism how several engrams about the same stimulus can be localized within the same neurons might be suggested by the models of subcellular organisation, as postulated in case of appetitive and aversive memory on the one hand and acquisition and extinction of aversive memory on the other hand.
Die molekularen Mechanismen der Wirt-Parasit-Interaktion bei der durch den Zestoden Echinococcus multilocularis ausgelösten Erkrankung der alveolären Echinokokkose sind bislang ungeklärt. Zudem liegen keine Daten über Entwicklungs- und Differenzierungsmechanismen dieses Parasiten vor, die für die Entwicklung neuer Antiparasitika genutzt werden könnten. Ein bei der Evolution der Metazoen bereits frühzeitig entstandener Signaltransduktionsmechanismus zur Steuerung von Entwicklungsvorgängen ist das TGFβ/BMP-System, das aus strukturell verwandten Zytokinen der TGFβ (transforming growth factor β) bzw. BMP (bone morphogenetic protein)-Familie, oberflächenständigen Rezeptoren der TGFβ-Rezeptorfamilie (Typ I und Typ II) und intrazellulären Signaltransduktoren der Smad-Familie besteht. Außer an Entwicklungsvorgängen tierischer Organismen könnte diesem System eine wichtige Rolle bei der Wirt-Helminth-Kommunikation während Infektionsprozessen zukommen, wie in vorherigen Studien am Nematoden Brugia malayi und am Trematoden Schistosoma mansoni gezeigt werden konnte. Erste, wichtige Schritte zur Charakterisierung von TGFβ und BMP-Signalsystemen in Zestoden wurden in der vorliegenden Arbeit getan. Aufbauend auf einem vorherigen Bericht zu einem Transmembranrezeptor (EmRSK1) und einem Smad-Homologen (EmSmadA) aus Echinococcus multilocularis wurde die Liste der TGFβ/BMP Signaltransduktionsfaktoren in E. multilocularis in dieser Arbeit deutlich erweitert und erstmals umfangreiche funktionelle Studien durchgeführt. Die hier charakterisierten Faktoren umfassen zwei weitere Serin/Threonin-Kinasen der TGFβ/BMP-Rezeptorfamilie (EmRSK2, EmRSK3) sowie intrazelluläre Transduktoren der R-Smad-Subfamilie (EmSmadB, EmSmadC) und ein Homologes zur MAP-kinase-kinase-kinase TAK1 (TGFβ activated kinase 1), genannt EmTAK1. Zudem konnte erstmals für einen parasitären Helminthen ein Zytokin der BMP-Subfamilie, EmBMP, auf molekularer Ebene charakterisiert werden. Strukturelle und funktionelle Untersuchungen legen nahe, dass E. multilocularis sowohl ein TGFβ wie auch ein BMP-Signalsystem exprimiert. Ersteres wird sehr wahrscheinlich durch die Kinase EmRSK2 und den Smad-Faktor EmSmadC gebildet, letzteres durch EmRSK1 und EmSmadB. EmSmadA nimmt eine Sonderstellung ein, da es sowohl durch TGFβ- wie auch durch BMP-Rezeptoren aktiviert werden kann. Die genaue Rolle von EmRSK1 und EmTAK1 wäre durch weitere Untersuchungen zu klären. Signifikante funktionelle Homologien zwischen den TGFβ/BMP-Signalsystemen des Parasiten und Säugern konnten nachgewiesen werden, die sich u.a. darin äußern, dass die Echinococcus Smad-Proteine durch entsprechende Rezeptoren des Menschen aktiviert werden können. Darüber hinaus konnten jedoch auch einige deutliche Unterschiede zwischen den Systemen aus Parasit und Wirt nachgewiesen werden, die sich als Angriffspunkte zur Entwicklung von Chemotherapeutika eignen könnten. So fehlt den Smad-Faktoren EmSmadA und EmSmadC eine MH1-Domäne, die sonst unter allen R-Smads hoch konserviert ist. Zudem sind einige bislang noch nie beschriebene, strukturelle Besonderheiten der Echinococcus TGFβ/BMP-Rezeptoren zu verzeichnen. Auch die Regulation dieser Faktoren und die Kreuz-Interaktion mit weiteren intrazellulären Signalwegen (z.B. der MAP Kinase Kaskade) scheint in E. multilocularis anders zu verlaufen als bislang für Vertebraten, Insekten oder Nematoden beschrieben. Schließlich konnte, als sehr wichtiger Befund, auch nachgewiesen werden dass mindestens ein Rezeptor des Parasiten, EmRSK1, mit einem Zytokin des Wirts (BMP2) in vitro funktionell interagiert. Da BMP2 in Zellkultursystemen, die das Wachstum des Parasiten am befallenen Wirtsorgan nachstellen, einen deutlichen Effekt auf E. multilocularis ausübt, könnte die hier beschriebene EmRSK1/BMP2 – Interaktion von entscheidender Bedeutung für die Wirt-Parasit-Interaktion bei der alveolären Echinokokkose sein.
