Um eine Signaltransduktion mittels agnostischer Antikörper an Rezeptoren der TNFRSF zu bewirken, ist eine vorherige Immobilisation über des Fc Anteil des Antikörpers Grundvorraussetzung. In dieser Arbeit sollte die Möglichkeit der Verankerung über eine andere Bindungsdomäne untersucht werden. Es konnte gezeigt werden, dass eine Immobilisation mittels scFv:CD70 zu einer starken Signalaktivierung führt.
The immune system is responsible for the preservation of homeostasis whenever a given organism is exposed to distinct kinds of perturbations. Given the complexity of certain organisms like mammals, and the diverse types of challenges that they encounter (e.g. infection or disease), the immune system evolved to harbor a great variety of distinct immune cell populations with specialized functions. For instance, the family of T cells is sub-divided into conventional (Tconv) and unconventional T cells (UTCs). Tconv form part of the adaptive arm of the immune system and are comprised of αβ CD4+ or CD8+ cells that differentiate from naïve to effector and memory populations upon activation and are essential during infection and cancer. Furthermore, UTCs, which include γδ T cells, NKT and MAIT, are involved in innate and adaptive immune responses, due to their dual mode of activation, through cytokines (innate-like) or TCR (adaptive), and function. Despite our understanding of the basic functions of T cells in several contexts, a great number of open questions related to their basic biology remain. For instance, the mechanism behind the differentiation of naïve CD4+ and CD8+ T cells into effector and memory populations is not fully understood. Moreover, the exact function and relevance of distinct UTC subpopulations in a physiological context have not been fully clarified. Here, we investigated the factors mediating naïve CD8+ T cell differentiation into effector and memory cells. By using flow cytometry, mass spectrometry, enzymatic assays, and transgenic mouse models, we found that the membrane bound enzyme sphingomyelin-phosphodiesterase acid-like 3b (Smpdl3b) is crucial for the maintenance of memory CD8+ T cells. Our data show that the absence of Smpdl3b leads to diminished CD8+ T cell memory, and a loss of stem-like memory populations due to an aggravated contraction. Our scRNA-seq data suggest that Smpdl3b could be involved in clathrinmediated endocytosis through modulation of Huntingtin interacting protein 1 (Hip1) levels, likely regulating TCR-independent signaling events. Furthermore, in this study we explored the role of UTCs in lymph node-specific immune responses. By using transgenic mouse models for photolabeling, lymph node transplantation models, infection models and flow cytometry, we demonstrate that S1P regulates the migration of tissue-derived UTC from tissues to draining lymph nodes, resulting in heterogeneous immune responses mounted by lymph nodes draining different tissues. Moreover, our unbiased scRNAseq and single lineage-deficient mouse models analysis revealed that all UTC lineages (γδ T cells, NKT and MAIT) are organized in functional units, based on transcriptional homogeneity, shared microanatomical location and migratory behavior, and numerical and functional redundancy. Taken together, our studies describe additional cell intrinsic (Smpdl3b) and extrinsic (S1Pmediated migration) functions of sphingolipid metabolism modulating T cell biology. We propose the S1P/S1PR1/5 signaling axis as the potential survival pathway for Smpdl3b+ memory CD8+ T cells and UTCs, mainly in lymph nodes. Possibly, Smpdl3b regulates S1P/S1PR signaling by balancing ligandreceptor endocytosis, while UTCs migrate to lymph nodes during homeostasis to be exposed to specific levels of S1P that assure their maintenance. Our results are clinically relevant, since several drugs modulating the S1P/S1PR signaling axis or the levels of Smpdl3b are currently used to treat human diseases, such as multiple sclerosis and B cell-mediated diseases. We hope that our discoveries will inspire future studies focusing on sphingolipid metabolism in immune cell biology.
Wilms tumor protein 1 (WT1) is a suitable target to develop an immunotherapeutic approach against high risk acute myeloid leukemia (AML), particularly their relapse after allogeneic hematopoietic stem cell transplantation (HSCT). As an intracellular protein traversing between nucleus and cytoplasm, recombinant expression of WT1 is difficult. Therefore, an induction of WT1-specific T-cell responses is mostly based on peptide vaccination as well as dendritic cell (DC) electroporation with mRNA encoding full-length protein to mount WT1-derived peptide variations presented to T cells. Alternatively, the WT1 peptide presentation could be broadened by forcing receptor-mediated endocytosis of DCs.
In this study, antibody fusion proteins consisting of an antibody specific to the human DEC205 endocytic receptor and various fragments of WT1 (anti-hDEC205-WT1) were generated for a potential DC-targeted recombinant WT1 vaccine. Anti-hDEC205-WT1 antibody fusion proteins containing full-length or major parts of WT1 were not efficiently expressed and secreted due to their poor solubility and secretory capacity. However, small fragment-containing variants: anti-hDEC205-WT110-35, anti-hDEC205-WT191-138, anti-hDEC205-WT1223-273, and anti-hDEC205-WT1324-371 were obtained in good yields.
Since three of these fusion proteins contain the most of the known immunogenic epitopes in their sequences, the anti-hDEC205-WT191-138, anti-hDEC205-WT1223-273, and anti-hDEC205-WT1324-371 were tested for their T-cell stimulatory capacities. Mature monocyte-derived DCs loaded with anti-hDEC205-WT191-138 could induce ex vivo T-cell responses in 12 of 16 blood samples collected from either healthy or HSC transplanted individuals compared to included controls (P < 0.01). Furthermore, these T cells could kill WT1-overexpressing THP-1 leukemia cells in vitro after expansion.
In conclusion, alongside proving the difficulty in expression and purification of intracellular WT1 as a vaccine protein, our results from this work introduce an alternative therapeutic vaccine approach to improve an anti-leukemia immune response in the context of allogeneic HSCT and potentially beyond.
IL-6 spielt eine wichtige Rolle bei der Immunantwort, Entzündung und Hämatopoese.
Das Glykoprotein 130 (gp130) wird ubiquitär exprimiert und bildet als Dimer die signaltransduzierende Rezeptoreinheit für das IL-6 Signal.
Die biologische Wirkung des IL-6 ist abhängig von der Dauer und Stärke des induzierten Signals. Die gp130 Rezeptorexpression stellt einen bedeutenden Faktor zur Beeinflussung des IL-6 Signals dar.
Die im Rahmen dieser Arbeit untersuchte gp130LLAA Maus weist eine Punktmutation im gp130 Rezeptor auf, bei der das Dileucin-Motiv (L874, L785) im zytoplasmatischen Bereich von gp130 zu Dialanin verändert wurde.
Für die Endozytose ist das intrazelluläre Dileucin-Motiv erforderlich, da das Adapterprotein AP-2 an dieses Motiv bindet und dadurch den Transport mittels Clathrin-umhüllter Vesikel begünstigt. Die beschriebene Punktmutation hat zur Folge, dass die veränderte Form von gp130 resistent gegenüber der Liganden- und crosstalk-vermittelten Endozytose ist.
Da IL-6 generell eine wichtige Rolle bei der Differenzierung hämatopoetischer Zellen spielt, so auch bei T- und B-Zellen, wurde der Einfluss der gp130LLAA Mutation auf die Homöostase dieser lymphoiden Zellen untersucht. Für die Versuche wurden sowohl B- und T-Zellen und jeweilige Subpopulationen aus der Milz von WT Mäusen und gp130LLAA Mäusen untersucht.
MDSCs are suppressive immune cells with a high relevance in various pathologies including cancer, autoimmunity, and chronic infections. Surface marker expression of MDSCs resembles monocytes and neutrophils which have immunostimulatory functions instead of suppressing T cells. Therefore, finding specific surface markers for MDSCs is important for MDSC research and therapeutic MDSC manipulation. In this study, we analyzed if the integrin VLA-1 has the potential as a novel MDSC marker. VLA-1 was expressed by M-MDSCs but not by G-MDSCs as well as by Teff cells. VLA-1 deficiency did not impact iNOS expression, the distribution of M-MDSC and G-MDSC subsets, and the suppressive capacity of MDSCs towards naïve and Teff cells in vitro. In mice, VLA-1 had no effect on the homing capability of MDSCs to the spleen, which is a major reservoir for MDSCs. Since the splenic red pulp contains collagen IV and VLA-1 binds collagen IV with a high affinity, we found MDSCs and Teff cells in this area as expected. We showed that T cell suppression in the spleen, indicated by reduced T cell recovery and proliferation as well as increased apoptosis and cell death, partially depended on VLA-1 expression by the MDSCs. In a mouse model of multiple sclerosis, MDSC injection prior to disease onset led to a decrease of the disease score, and this effect was significantly reduced when MDSCs were VLA-1 deficient. The expression of Sema7A by Teff cells, a ligand for VLA-1 which is implicated in negative T cell regulation, resulted in a slightly stronger Teff cell suppression by MDSCs compared to Sema7A deficient T cells. Live cell imaging and intravital 2-photon microscopy showed that the interaction time of MDSCs and Teff cells was shorter when MDSCs lacked VLA 1 expression, however VLA-1 expression had no impact on MDSC mobility. Therefore, the VLA-1-dependent interaction of MDSC and Teff cells on collagen IV in the splenic red pulp is implicated MDSC-mediated Teff cell suppression.
Untersuchungen zur Autophagieinduktion in Leishmania major-infizierten Knochenmarksmakrophagen

Die von der WHO zu den 17 wichtigsten NTDs gezählte Leishmaniose wird durch intrazelluläre Parasiten der Gattung Leishmania hervorgerufen. Der Lebenszyklus der Parasiten besteht aus zwei Phasen. Die länglichen und beweglichen Promastigoten kennzeichnen die Phase in der Sandmücke – der Vektor der Leishmaniose. Hingegen ist die Phase im Säugerwirt durch runde unbewegliche Amastigoten charakterisiert. Aufgrund des Mangels an potenten antileishmanialen Therapien wurde in der vorliegenden Arbeit die Interaktion zwischen L. m. Parasiten und der Hauptwirtszelle, der Makrophage, v. a. in Hinblick auf autophage Prozesse in den infizierten Makrophagen näher untersucht, um demgemäß neue Erkenntnisse zu gewinnen, welche bei der Herstellung zukünftiger anti-leishmanialer Medikamente helfen könnten.
Bei der Autophagie handelt es sich um einen katabolen Prozess, wodurch Zellen bei Nahrungsmangel oder zellulärem Stress ihre Homöostase erhalten können. Durch diesen Prozess können überflüssige oder beschädigte Organellen recycelt werden, um die Funktionen der Zelle aufrechtzuerhalten. Daneben übernimmt Autophagie auch eine essenzielle Rolle bei der Abwehr von ins Zytosol eindringenden Pathogenen.
Mittels des neu etablierten totalen Autophagiescore konnte festgestellt werden, dass Autophagie in L. m.-infizierten BMDM induziert wird. Die intrazellulären Amastigoten werden durch Autophagie in den BMDM verdaut. Die erhöhte autophage Aktivität konnte zudem durch Western-Blot-Analysen der autophagierelevanten Proteine ATG5, LC3B und UB bestätigt werden. Die molekulargenetischen Untersuchungen von L. m.-infizier-ten BMDM mithilfe von Affymetrix Microarrays führten zu einem Netzwerk aus autophagierelevanten und infektionsspezifischen Genen, welches als LISA bezeichnet worden ist. Hier hat sich ebenfalls eine starke Verknüpfung von autophagierelevanten Genen und den Genen der Glykolyse, einem zweiten katabolen Prozess, gezeigt. Zudem konnten zwei weitere autophagierelevante und infektionsspezifische Gene außerhalb von LISA identifiziert werden, nämlich Bnip3 und Ctse, welche im Anschluss genauer untersucht worden sind. Bei beiden Genen konnte auf Proteinebene gezeigt werden, dass sie in L. m.-infizierten BMDM signifikant erhöht sind. Durch siRNA-Analysen konnte überdies beobachtet werden, dass beide für die erfolgreiche Elimination der Amastigoten essenziell sind.
Somit konnte mit den Proteinen BNIP3 und CTSE zwei potenzielle neue Ansatzpunkte für mögliche zukünftige antileishmaniale Therapien gefunden werden. Auch die in LISA enthaltenen Gene stellen prinzipiell vielversprechende Ziele für künftige Medikamente gegen Leishmaniose dar. Durch all diese Untersuchungen kommt man dem Ziel einer neuen, gezielten und nebenwirkungsärmeren Behandlung der Leishmaniose einen Schritt näher.
In this work we expanded upon a study from our group where a ligand-based TNF-α mutein was developed to engage specifically TNFR2 and not TNFR1 activating Tregs and expanding them, which in an allo-HCT context conferred protection from GvHD. Fusing TNF trimers to the heavy chain of an Fc-dead and mouse irrelevant antibody, a new generation of this agonist was developed called NewSTAR2. It is believed that other members of the TNFSF can also target Tregs, therefore additional agonists against DR3 and GITR were developed under the same principles as for NewSTAR2. Phenotyping analysis of the expression of these three receptors were done to confirm their specificity for Tregs before in vitro and in vivo testings with mice or murine splenic cells. A potent expansion of Tregs was seen with NewSTAR2 and the other agonists as well as upregulation of activation markers on Tregs. Thorough analyses with NewSTAR2-treated mice showed how Tregs in several immune and non-immune organs were expanded and upregulated immunomodulatory receptors. A miniature suppressive assay and other cocultures with responder cells confirmed their enhanced suppression over unstimulated Tregs through contact dependent and independent mechanisms. Despite other myeloid cells also being increased after treatment, no undesired effects were observed under steady-state and prophylactic administration of a single dose of NewSTAR2 improved survival frequencies and lessened development of clinical symptoms. Prophylactic treatment with the other TNFRSF agonists showed similar protection yet Fc(DANA)-muTL1A was superior in in terms of less death events and lower clinical score. It was found that not all the three TNFSF members have redundant functions as development of skin lesions was observed with GITRL-based agonist Fc(DANA)-muGITRL, although its expansion of Tregs in steady-state was remarkable with no apparent adverse effects. Neither agonist had an impact on donor cell engraftment or allorective T cell response, however NewSTAR2-treatmend proved to reduce inflammation in small intestine and liver. This work is proof of concept of the effectivity of selectively engaging TNFSF to activate Tregs and expand them systemically allowing them to control strong and complex immune interactions like those governing GvHD.
Cardiovascular disease and the acute consequence of myocardial infarc- tion remain one of the most important causes of morbidity and mortality in all western societies. While much progress has been made in mitigating the acute, life-threatening ischemia caused by infarction, heart failure of the damaged my- ocardium remains prevalent. There is mounting evidence for the role of T cells in the healing process after myocardial infarction, but relevant autoantigens, which might trigger and regulate adaptive immune involvement have not been discov- ered in patients.
In this work, we discovered an autoantigenic epitope in the adrenergic receptor beta 1, which is highly expressed in the heart. This autoantigenic epitope causes a pro-inflammatory immune reaction in T cells isolated from pa- tients after myocardial infarction (MI) but not in control patients. This immune reaction was only observed in a subset of MI patients, which carry at least one allele of the HLA-DRB1*13 family. Interestingly, HLA-DRB1*13 was more com- monly expressed in patients in the MI group than in the control group.
Taken together, our data suggests antigen-specific priming of T cells in MI patients, which leads to a pro-inflammatory phenotype. The primed T cells react to a cardiac derived autoantigen ex vivo and are likely to exhibit a similar phenotype in vivo. This immune phenotype was only observed in a certain sub- set of patients sharing a common HLA-allele, which was more commonly ex- pressed in MI patients, suggesting a possible role as a risk factor for cardiovas- cular disease.
While our results are observational and do not have enough power to show strong clinical associations, our discoveries provide an essential tool to further our understanding of involvement of the immune system in cardiovascu- lar disease. We describe the first cardiac autoantigen in the clinical context of MI and provide an important basis for further translational and clinical research in cardiac autoimmunity.
NFATc3 in der akuten GvHD

Bei Leukämien, Lymphomen und dem Multiplen Myelom stellt die allogene hämatopoetische Stammzelltransplantation (allo-HCT) oft die letzte kurative Therapieoption dar. Spender-T-Zellen (v.a. CD8+-T-Zellen), die im Transplantat enthalten sind, erkennen nach Chemo-/Strahlentherapie verbliebene Reste des entarteten Empfängergewebes, eradizieren dieses und verhindern somit ein Tumorrezidiv (Graft-versus-Leukämie Reaktion/GvL). Häufig attackieren Spender-T-Zellen (v.a. CD4+-Th1-Zellen) aber auch nicht-malignes Gewebe (z.B. Haut, Leber und Darm), was bis zum Tod des Patienten führen kann (Graft-versus-Host Disease/GvHD). Calcineurin-Inhibitoren wie Cyclosporin A (CsA) und Tacrolimus, die oft schon prophylaktisch verabreicht werden, verhindern über eine unselektive Inhibition aller Mitglieder der NFAT-Transkriptionsfaktorfamilie (Nuclear factor of activated T-cells) die Aktivierung der Spender-T-Zellen. Es folgt eine klinische Besserung der GvHD-Symptomatik, während jedoch der GvL-Effekt ebenfalls supprimiert wird. Bisherige Untersuchungen unserer Arbeitsgruppe am Mausmodell hatten gezeigt, dass die selektive Inhibition eines NFAT-Familienmitgliedes (NFATc1 oder NFATc2) in den Donor-T-Zellen zu einer signifikanten Besserung der aGvHD bei jedoch erhaltener GvL führt. Es wurde nun der Einfluss des dritten, in Lymphozyten exprimierten NFAT-Mitglieds NFATc3 im Kontext der aGvHD untersucht.
Zur Basisanalyse der neu kreierten Nfatc3fl/fl.Cd4cre- und Nfatc1fl/fl.Nfatc3fl/fl.Cd4cre-Mauslinien erfolgten durchflusszytometrische und Western-Blot-Analysen. Anschließend wurden In-vivo-Untersuchungen unter Verwendung eines etablierten major-mismatch-aGvHD-Modells (H-2b→H-2d) durchgeführt.
Es konnte gezeigt werden, dass durch eine NFATc3- (+/- NFATc1-) Defizienz direkt ex vivo die CD4+/CD8+-Ratio durch Abnahme der CD4+- hin zu den CD8+-T-Zellen verschoben wird. Auch zeigte sich in den entsprechenden Genotypen eine Abnahme der naiven- und dafür vice versa eine Zunahme der Effektor-T-Zellen. In den wiederholt durchgeführten aGvHD-Versuchen zeigte sich in vivo als Korrelat der (ebenfalls erneut nachgewiesenen) Abnahme des CD4+/CD8+-Quotienten in den Zielorganen eine geringere Expansion der NFAT-defizienten als der wildtypischen T-Zellen. Leider spiegelte sich dies nicht in dem clinical score zur Quantifizierung der aGvHD-Symptomatik wider. Auch das Körpergewicht der Versuchsgruppe nahm rapide ab. Ursächlich hierfür ist – als Korrelat zur direkt ex vivo nachgewiesenen Aktivierungsneigung – ein vermehrter Th1-Shift der NFATc3 (+/-NFATc1-) defizienten T-Zellen.
Eine Inhibierung von NFATc3 – im Gegensatz zu NFATc1 und NFATc2 – ist demzufolge kein sinnvoller Ansatzpunkt für eine mögliche, zielgerichtetere aGvHD-Therapie. Der positive Effekt der reduzierten Proliferationsneigung der NFATc3-defizienten Lymphozyten wird durch deren vermehrte Aktivierungsneigung mit erhöhter Sekretion von pro-inflammatorischen Zytokinen zunichte gemacht.
Veränderung der Tumorimmunumgebung muriner Mamma-Karzinome durch Inhibierung der Kollagensynthese

Das Mamma-Karzinom gehört zu den sogenannten desmoplastischen Tumorarten. Hierbei handelt es sich um Tumoren mit erhöhter Ansammlung von Bindegewebszellen und einer Akkumulation von Extrazellulärer Matrix (EZM). Diese verdichtete EZM wirkt sowohl auf mechanischer als auch auf Signalweg-vermittelter Ebene als eine Barriere, welche die therapeutische Wirksamkeit erheblich vermindert.
Einer der Hauptbestandteile der EZM ist Kollagen. Durch Anwendung von Präparaten, welche die Kollagensynthese und -reifung inhibieren, kann die rigide Struktur aufgelockert werden. Daraus ergibt sich eine verbesserte Versorgung mit Nährstoffen und eine verbesserte Infiltrationsmöglichkeit für Immunzellen. Dies ist für die Effizienz der Immuntherapie, welche sich in den letzten Jahren als vielversprechende Alternative zu den Grundsäulen der Krebstherapie entwickelt hat, unabdinglich.
In der vorliegenden Arbeit wurden murine Mamma-Karzinome der 4T1-Linie nach Behandlung mit EZM-destabilisierenden Kollageninhibitoren auf ihre Immunumgebung hin untersucht.
Verwendet wurden drei Wirkstoffe, welche an unterschiedlichen Punkten in die Kollagensynthese und -reifung eingreifen: βAPN als LOX(L)-Inhibitor, 1,4-DPCA als P4HA-Inhibitor und Minoxidil als LH-Inhibitor. Die Behandlung führte zu einem deutlichen Anstieg aller untersuchten Immunzellen und deutet somit auf eine verbesserte Infiltrationsmöglichkeit hin. Zudem wurde die Expression maligner Signalwege, wie die der Angiogenese, Hypoxie, Metastasierungsneigung, Invasivität und Immunsuppression, verringert und tumorsuppressive Immunantworten verstärkt.
Die Kollageninhibition hatte zusätzlich ein verringertes Tumorwachstum und eine Reduktion der Blutgefäßdichte zufolge.
Als Fazit gilt es festzuhalten, dass die Verwendung von Kollageninhibitoren in der Immuntherapie eine vielversprechende Option zur Verbesserung der Effizienz dieser Therapeutika darstellt. Diese Erkenntnis gilt es im Rahmen künftiger wissenschaftlicher Untersuchungen weiterzuentwickeln.
