Climate change and associated extreme weather events are a threat not only for agricultural
yields but the plant kingdom in general. Therefore, there is a great necessity to better
understand the plants' intrinsic mechanisms to combat heat stress. The plant heat stress
response already has been investigated in many studies, including the role of HSFA1
transcription factors as the central regulators. Other aspects such as the initial perception of
heat and the role of heat-induced changes in plant metabolism are rather unknown.
In this thesis, the natural variation of 250 different accessions of Arabidopsis thaliana was
investigated regarding the temperature-dependent accumulation of raffinose and
triacylglycerols. A connection between these phenotypes and respective genotypes was
established using genome-wide association studies. As a result, the candidate gene
TREHALOSE-6-PHOSPHATE SYNTHASE 1 (TPS1), was identified. Enzymatic TPS1 is responsible
for the synthesis of trehalose 6-phosphate (T6P), which serves as an indicator and regulator
of sucrose homeostasis.
Subsequent analyses using tps1 tilling mutants demonstrated a link between T6P metabolism
and an increased accumulation of various soluble carbohydrates and starch, including
raffinose both under control conditions and during heat exposure. Furthermore, the mutant
lines displayed enhanced thermotolerance and survival rates following long-term heat stress.
Transcriptome analyses, however, did not show any difference in the regulation of canonical
heat stress-associated genes. Instead, genes related to photosynthesis were overrepresented
among the differentially upregulated genes in tps1 tilling lines during heat exposure. In this
work, a direct connection of T6P signaling, sucrose homeostasis, and thermotolerance is
shown for the first time.
In a second project, two Arabidopsis thaliana accessions (Oberursel-0, accession ID: 7276;
Nieps-0, accession ID: 7268) showing distinct capacities to acquire short-term
thermotolerance were compared to identify the putative causative regulators or mechanisms
that lead to the different levels of thermotolerance.
An examination of the transcriptomes of 7268 and 7276 showed that several hundreds of
genes were already differentially regulated within 10 minutes of exposure to 32 °C or 34 °C.
Among these, several genes associated with sulfur metabolism were more highly induced in
the more thermotolerant accession 7268. However, experimental as well as genetic
manipulation of sulfur availability and metabolism did not result in altered thermotolerance.
In addition to sulfur-related genes, most of the canonical heat stress-associated genes were
more highly expressed in 7268 than in 7276. While we could not identify a causative regulator
or mechanism of differential thermotolerances, the data strongly suggests that 7268 either
has a higher overall sensitivity, i.e., the heat stress response is initiated at lower temperatures,
or stronger overall heat stress response when exposed to a certain elevated temperature.
The interaction between circadian clocks and metabolism is of increasing interest, since clock dysfunction often correlates with metabolic pathologies. Many research articles have been published analysing the impact of factors such as circadian clock, light, feeding time and diet-type on energy homeostasis in various tissues/organs of organisms with most of the findings done in mammals. Little is known about the impact of circadian clock and the above-mentioned factors on circulating lipids, especially the transport form of lipids - diacylglycerol (DG) and membrane lipids such as phosphatidylethanolamine (PE) and phosphatidylcholine (PC) in the Drosophila hemolymph. The fruit fly Drosophila is a prime model organism in circadian, behaviour and metabolism research.
To study the role of circadian clock and behaviour in metabolism, we performed an extensive comparative hemolymph lipid (diacylglycerol: DG, phosphatidylethanolamine: PE, phosphatidylcholine: PC) analysis using ultra performance liquid chromatography coupled to time-of-flight mass spectrometry (UPLC-MS) between wild-type flies (WTCS) and clock disrupted mutants (per01). In addition, clock controlled food intake– feeding behaviour was investigated. Time-dependent variation of transport (DG) and membrane lipids (PE and PC) were not rhythmic in WTCS under constant darkness and in per01 under LD, suggesting an impact of light and clock genes on daily lipid oscillations. Day-time and night-time restriction of food led to comparable lipid profiles, suggesting that lipid oscillations are not exclusively entrained by feeding but rather are endogenously regulated. Ultradian oscillations in lipid levels in WTCS under LD were masked by digested fatty acids since lipid levels peaked more robustly at the beginning and end of light phase when flies were fed a lipid- and protein-free diet. These results suggest that metabolite (DG, PE and PC) oscillation is influenced by complex interactions between nutrient-type, photic conditions, circadian clock and feeding time.
In conclusion, the results of this thesis suggest that circadian clocks determine transport and membrane lipid oscillation in Drosophila hemolymph in complex interactions between nutrient-type, photic conditions and feeding behaviour.
In Brassicaceae werden bei einer Gewebszerstörung unreaktive Glukosinolate durch das Enzym Myrosinase hydrolysiert. Es entstehen reaktive Substanzen wie Isothiocyanate (ITCs). Da diese Reaktion sehr schnell erfolgt wird sie auch als Senföl-Glukosid-Bombe bezeichnet. In Arabidopsis thaliana erfolgt nach Verwundung und Pathogeninfektion eine massive Akkumulation des ITCs Sulforaphan (SF), welches eine reaktive elektophile Spezies (RES) darstellt. Zu der Gruppe der RES zählen auch einige Oxylipine mit einer α,β-ungesättigten Carbonylgruppen wie 12-oxo-Phytodiensäure (OPDA) oder Phytoprostan A1 (PPA1). Die Fähigkeit der kovalenten Modifikation von Peptiden und Proteinen gilt als essentiell sowohl für die toxischen als auch die Gen-induzierenden Eigenschaften der RES. Neben ihrer Reaktivität spielt auch die Lipophilie eine Rolle für die Fähigkeit über Membranen zu diffundieren und unspezifisch an Proteine zu binden.
Die in der vorliegenden Arbeit durchgeführten Transkriptomanalysen an Arabidopsis-Keimlingen mit sub-toxischen Konzentrationen von SF, Benzylisothiocyanat (BITC) und dem Oxylipin Prostaglandin A1 (PGA1) zeigten, dass strukturell sehr verschiedene RES einen gemeinsamen Satz von 55 Genen induzieren. Unter diesen befanden sich verschiedene Hitzeschock-, Stressassoziierte- und Detoxifizierungsgene. Diese Ergebnisse deuten darauf hin, dass die Aktivierung über eine Muster-spezifische Erkennung der RES erfolgt. Als einen möglichen Mechanismus der RES-vermittelten Geninduktion wird die Regulation durch die Veränderung des zellulären Redox-Potentials als Folge kovalenter Modifikation von GSH durch RES diskutiert. Die Untersuchung der GSH-Gehalte sowie des Redox-Potential nach Behandlung mit sub-toxischen RES-Konzentrationen in Arabidopsis-Keimlingen zeigte jedoch unter den getesteten Bedingungen keine Veränderung.
Neben dem Erkennungs- und Signaltransduktionsmechanismus ist auch die biologische Bedeutung von RES für die Vermittlung einer Stresstoleranz noch weitgehend unklar. Durch die Untersuchung der Genexpression in Arabidopsis-Pflanzen nach Verwundung konnte gezeigt werden, dass eine wundinduzierte Akkumulation von SF zur Induktion einiger Gene der Hitzeschockreaktion (HSR) im Wildtyp, jedoch nicht in der myrosinase-defiziten tgg1tgg2-Mutante führte. Auch in der Transkriptomanalyse war nach RES-Gabe ebenfalls eine starke Induktion hitze-responsiver Gene, deren Regulation über den Masterregulator dem Hitzeschock-TF A1 vermittelt wird, zu beobachten. Besonders die Induktion der HSPs, welche als Chaperone fungieren und damit Thiolgruppen von Proteinen vor Modifikation schützen können, haben vermutlich bei chemischer Intoxikation protektive Eigenschaften für die Zellen. Tatsächlich zeigte sich unter den gewählten Bedingungen die hsfa1a,b,d,e-Mutante empfindlicher gegenüber ITCs als der Wildtyp. Die Fähigkeit, eine HSR ausbilden zu können, scheint in Arabidopsis bei chemischer Intoxikation eine bedeutende Rolle zu spielen. Eine Vorbehandlung mit RES wie SF, BITC oder dem HSP90-Inhibitor Radicicol in Arabidopsis-Keimlingen konnte eine Schutzwirkung vor chemischer Intoxikation vermitteln. Dies erfolgte jedoch nicht nach Behandlung mit moderater Hitze (zwei Stunden, 37 °C). Somit scheint die HSR alleine nicht ausreichend für den Aufbau eines effektiven Schutzes vor BITC-Intoxikation zu sein.
Als metabolische Antwort von Arabidopsis-Keimlingen auf Intoxikation mit RES konnte eine konzentrationsabhängige Senkung der maximalen Quantenausbeute am Photosystem II (PSII), sowie gleichzeitig eine Akkumulation an TAG-Spezies beobachtet werden. Diese metabolische Reaktion ist in der Literatur bereits als Schutz gegen Hitzestress beschrieben. Die Bedeutung der TAG-Akkumulation nach chemischem ITC-Stress ist noch unklar.
Effects of timing and herbivory on a grass-endophyte association and its trophic interactions

I.) Plant associated microorganisms can affect the plant`s interaction with herbivores and higher trophic levels. For instance, endophytic fungi infecting aerial plant parts of grass species produce bioactive alkaloids that can negatively affect species from higher trophic levels, indicating a defensive mutualism between the grass and the endophyte. However, beneficial insects can also be negatively affected by the endophyte, which might question the mutualistic effect of endophytic fungi. On the other hand, grass-endophytes are affected by environmental conditions and species interactions. Grazing can increase endophyte frequencies in natural habitats. Furthermore, endophyte mediated effects on herbivores are most pronounced during warm summers following rainy springs. In this study, we investigated whether endophyte derived alkaloids cascade up a food chain (chapter II) and whether their concentrations depend on plant age and season (chapter III). Further we analysed, whether altered herbivore phenology affects the endophytic fungus (chapter IV) and whether endophyte derived alkaloid production is induced by different herbivore species (chapter V).
II.) In our first experimental study we analysed whether grass-endophyte derived alkaloids decreased the performance of two ladybird species feeding on aphids exclusively reared on endophyte infected grass (6 weeks young grass). Further, we screened species from three trophic levels (grass, herbivores and aphid predators) for their alkaloid content using two year old infected grass as diet for herbivores. We established an UPLC-MS method to detect and quantify the amount of the endophyte derived alkaloids peramine and lolitrem B extracted from the organic plant and insect material. Performance parameters of ladybirds revealed little differences between ladybirds fed on aphids reared on endophyte infected and non-infected grass, which probably resulted from low alkaloid concentrations in the young (6-weeks old) endophyte infected grass used in this part of the study. Alkaloid quantification of the two year old endophyte infected grass, herbivores and aphid predators revealed similar concentrations between grass and aphids, while aphid predators contained approximately half of that amount which still exceeded the bioactive threshold. We conclude that alkaloids produced by grass-endophytes cascade up the food chain and are responsible for fitness disadvantages of higher trophic levels.
III.) In the second study we investigated the impact of plant age and seasonal timing on grass-endophyte growth and alkaloid production. Plants were sown in April of 2013 and sampled monthly over 30 consecutive months. Endophyte growth was quantified with real-time PCR (qPCR) and alkaloid concentrations with UPLC-MS. We showed that alkaloid concentrations and fungal growth followed a seasonal rhythmicity and that alkaloid concentrations increased with plant age. Alkaloid concentrations peak during summer, when also herbivore abundances are high. Consequently, we conclude that plant age and season contribute to the toxicity of endophytes on grass herbivores
IV.) In the third study we simulated earlier spring arrival of aphids by enhancing aphid abundance on endophyte infected and endophyte-free grass in spring and analysed responses across three trophic levels. Enhanced aphid abundance in spring caused higher aphid abundances during the study period. Predators stayed unaffected by increased herbivore abundances; however they did level aphid numbers within two weeks after arrival on the plants, independent of aphid abundance. Grass-endophyte showed a time delayed growth, two weeks after aphid abundance peak and after predators already controlled aphid infestations on the plants. We conclude that phenology shifts of herbivorous insects can affect multi-trophic interactions leading to desynchronizations between phenologies of interacting species and mismatches in food-webs.
V.) In the fourth study we analysed whether herbivores induce endophyte growth and alkaloid production and whether different types of herbivores induce specific alkaloid production. We applied three different herbivore treatments on endophyte infected grass over 18 weeks. Locust herbivory increased the insect deterring alkaloid peramine and clipping of plants (simulation of grazing livestock) increased the vertebrate toxic alkaloid lolitrem B. Aphid herbivory did not affect endophyte derived alkaloid concentrations. Endophyte responses to herbivory were species specific which indicates a primarily plant protecting role of alkaloid synthesis in endophyte infected plants and a close chemical crosstalk between interacting species.
VI.) In summary, we showed that endophyte derived alkaloids affect higher trophic levels and that alkaloid concentrations in the plant depend on prevalent herbivore species, plant age and seasonal timing. Our results indicate a close chemical crosstalk between the host plant and the endophytic fungus which is susceptible to environmental changes altering the endophyte`s alkaloid production in plants. We gained insights into the grass-endophyte symbiosis in ecological contexts and conclude that several factors determine the herbivore toxic potential of endophytic fungi and thereby their plant mutualistic or parasitic character. Future studies should investigate the mechanisms behind the herbivore induced alkaloid concentration increase, shown in this thesis, especially whether plant signals mediate the endophyte response. Furthermore it would be interesting to study the induction of indirect endophyte mediated defence and how it affects multi-trophic level interactions.
Oxylipine sind Signalmoleküle, die durch enzymatische Oxidation oder durch Autoxidation von mehrfach ungesättigten Fettsäuren entstehen. Sie akkumulieren während einer Vielzahl von biotischen und abiotischen Stressen und spielen eine bedeutende Rolle bei der Abwehr verschiedener Stressoren. In vielen physiologischen Entwicklungsprozessen sind Oxylipine ebenfalls wichtig.
Eine bisher wenig erforschte Untergruppe dieser Oxylipine bilden reaktive elektrophile Spezies, die sog. RES-Oxylipine. Hierzu gehören unter anderem der Jasmonsäure-Vorläufer 12 Oxophytodiensäure (OPDA), aber auch (E)-2-Hexenal oder Phytoprostan A1 (PPA1). Diese Substanzen sind aufgrund einer α,β ungesättigten Carbonylgruppe elektrophil und damit chemisch reaktiv. Diese Reaktivität wird als Grund für ihre biologische Aktivität angesehen: RES-Oxylipine sind Induktoren einer Reihe von Genen. Allerdings ist bisher wenig über den Signalweg sowie die Funktionen der RES-Oxylipine in Arabidopsis thaliana bekannt.
Fast die Hälfte (40 %) aller durch OPDA-induzierten Gene in A. thaliana sind abhängig von TGA-Transkriptionsfaktoren, jedoch werden OPDA-responsive Hitzeschockgene (z.B. Hitzeschockproteine) unabhängig von TGA-Transkriptionsfaktoren induziert. Außerdem gibt es Hinweise auf eine Akkumulation des RES-Oxylipins OPDA, aber auch des non-RES-Oxylipins Jasmonsäure (JA) durch eine Behandlung mit 38° C in A. thaliana. Eine exogene Applikation von JA bewirkt jedoch, im Gegensatz zu OPDA, keine Genexpression von Hitzeschockgenen in Arabidopsis.
Ziel dieser Arbeit war es, die Funktion der RES-Oxylipine OPDA und Prostaglandin A1 (PGA1, ein Analogon zu PPA1) während der Hitzeschockantwort in Arabidopsis thaliana, sowie die TGA-unabhängige Signaltransduktion der Hitzeschockgene, aufzuklären.
Durch einen Vergleich zweier bereits veröffentlichter Transkriptomdaten in silico konnte die Überschneidung des Hitze-induzierten- (1 h, 37 °C) und des OPDA-induzierten-Transkriptoms (4 h, 75 µM) genau analysiert werden. Es werden 30 Gene sowohl von OPDA als auch durch 37 °C mehr als dreifach hochreguliert. Dieses Ergebnis konnte durch realtime qPCR vier repräsentativer Gene (HSP101, HSP26.5, DREB2A, HSFA2) bestätigt werden. Allerdings zeigten sich deutliche Unterschiede in der Stärke und Kinetik der Induktion: Hitze (37 °C) hat einen sehr viel stärkeren Einfluss auf die Hochregulation der Genexpression als die getesteten RES-Oxylipine OPDA und PGA1 (unter 10 % der Induktion durch 37 °C, Ausnahme DREB2A). Zudem resultiert eine Hitzebehandlung in einer schnellen und transienten Genexpression, das Maximum ist nach 1 bis 2 h erreicht während die Addition von RES-Oxylipinen eine langsamere Induktion der Genexpression bewirkt (Maximum nach 4 bis 6 h).
Eine Genexpressionsanalyse mit verschiedenen Signaltransduktionsmutanten half bei der Aufklärung möglicher Signaltransduktionskomponenten der RES-Oxylipine. So konnte gezeigt werden, dass der putative OPDA-Rezeptor Cyclophilin 20-3 sowie sein Interaktionspartner, das Protein Serin-Acetyltransferase 1, keine Bedeutung in der Regulation von Hitzeschockgenen durch RES-Oxylipine haben. Die Hitze-Masterregulatoren HSFA1 a,b,d (und e) jedoch sind für die Induktion der Hitzeschockgene HSP101, HSP26.5 und HSFA2 durch RES-Oxylipine essentiell und für DREB2A zumindest teilweise notwendig. Dennoch spielt der durch Hitze induzierbare Transkriptionsfaktor HSFA2 in der Signaltransduktion von RES-Oxylipinen (bezüglich der Hitzeschockgeninduktion) keine Rolle.
Durch ein Screening strukturell verschiedener RES hinsichtlich ihrer Induktion von HSP101 konnte geklärt werden, dass nicht die Anwesenheit einer α,β ungesättigten Carbonylgruppe, sondern vielmehr die Eigenschaft der Elektrophilie für die Induktion des HSP101 verantwortlich ist. Auch das RES Sulforaphan vermittelt, wie die RES-Oxylipine OPDA und PGA1, die Induktion der Hitzeschockgene über die HSFA1-Transkritionsfaktoren.
Ein weiterer Schwerpunkt dieser Arbeit lag in der Quantifizierung der endogenen Oxylipine in zehn Tage alten Arabidopsis-Keimligen nach einer Hitzebehandlung unter Kurztag-Lichtbedingungen. Weder während eines kurzzeitigen Hitzestresses (bis zu 8 h) noch während einer längerfristigen Hitzebehandlung (bis zu 7 Tage) steigt der Gehalt des RES Oxylipins OPDA signifikant an. Das non-RES-Oxylipin Jasmonsäure hingegen akkumuliert transient (Maximum 2 h nach Beginn eines Hitzestresses) und signifikant, allerdings ist dieser Anstieg in seiner Stärke (13fach) nicht vergleichbar mit einer Akkumulation beispielsweise nach Verwundung (hier ist ein 1000facher Anstieg möglich).
In weiteren Experimenten wurde eine mögliche Korrelation der endogenen Oxylipin-Akkumulation (verursacht durch Verwundung oder osmotischen Stress) mit der Genexpression von Hitzeschockgenen untersucht. …
