In der vorliegenden Arbeit wurden die Reaktionen elektrophiler und nucleophiler Schwefeldonoren mit cyclischen Alkenen und Alkinen untersucht, wobei ungewöhnliche und neuartige Schwefelchemie aufgedeckt wurde. Als elektrophile Schwefeldonoren wurden ein Sulten und ein Thiophenendoperoxid eingesetzt. Als Schwefelakzeptoren standen die cyclische Alkene, Enolether und Alkine zur Verfügung. Das Sulten überträgt unter Lewissäure-Katalyse das Schwefelatom auf verschiedene gespannte cyclische Olefine, wobei diastereoselektiv die entsprechenden Episulfide sowie ein Oxetan und/oder ein Aldehyd gebildet werden. Es kamen dabei verschiedene Lewissäuren wie z. B. Metallhalogenide (BF3•Et2O, ZnCl2 und SnCl4), Metallkomplexe [Mn(salen*)PF6] und Porphyrine zum Einsatz. Als beste Lewissäure erweist sich das Zinnporphyrin Sn(tpp)(ClO4)2, mit der die Olefine bei Raumtemperatur in 30 Prozent bis > 95 Prozent Ausbeute episulfidiert werden. Beim Schwefeltransfer vom Sulten auf 1-Methoxycycloocten entsteht das Produkt einer Insertion des Enolethers in die O-S-Bindung des Sultens in 69 Prozent Ausbeute. Wird das Sulten mit Cyclooctin und einem Äquivalent Trifluoressigsäure oder einer anderen starken Säure umgesetzt, wird ein Thiireniumion erhalten, das in stark saurer Lösung bis zu 24 h persistent ist und unter neutralen Bedingungen zu einem Dien umlagert. Die Bildung des Thiireniumions ist unter Einwirkung von Base reversibel. Mit Dithiacyclononin wird ein analoges Thiireniumion postuliert, das jedoch nicht direkt beobachtet werden kann. Persistentes Endprodukt dieser Reaktion ist ein Thioacetal. Das Thiophenendoperoxid wurde durch Tieftemperatur-Photooxygenierung des entsprechenden Thiophens in situ generiert. Bei der Thermolyse in Gegenwart von Cyclooctin bildet sich diastereoselektiv in 70 Prozent Ausbeute ein Episulfid. Bei der Reaktion des nucleophilen Schwefeldonors Thiotosylat mit Ninhydrin oder Indantrion in Gegenwart von trans-Cycloocten entsteht ein Cycloaddukt in bis zu 63 Prozent Ausbeute, während das Episulfid des trans-Cyclooctens nur in maximal 18 Prozent Ausbeute erhalten wird. Mit dem Schwefelnucleophil Diethylphosphorothioat und Indantrion wird neben dem Cycloaddukt das Diethylphosphat generiert. Wird Alloxanhydrat als Substrat verwendet, entsteht ein analoges Cycloaddukt in 33 Prozent Ausbeute. Indantrion geht mit 1 Methoxycycloocten eine Carbonyl-En-Reaktion ein, bei der ausschließlich ein Regioisomer in 51 Prozent Ausbeute entsteht.
Mittels Laserblitz-Photolyse wurden die Triplettlebenszeiten sowie die Löschraten der Triplettzustände verschiedener Acetophenonderivate durch dG, 8-oxodG, DNA, molekularen Sauerstoff und die Ketone selbst bestimmt. Für AP-OAc, AP und BP wurden Triplettlebensdauern von 7-9 µs gemessen, während die Triplettzustände von AP-OH und AP-OtBu aufgrund alpha Spaltung deutlich kurzlebiger waren (ca. 1 µs); die alpha Spaltung konnte EPR-spektroskopisch durch Spinabfangexperimente mit DMPO und TEMPO belegt werden. Im Fall von AP-OMe wurde weder dessen Triplettzustand noch die Bildung von Radikalen detektiert, was auf einer schnell ablaufenden Norrish-Typ-II-Spaltung beruht. Aufgrund dieses photochemischen Verhaltens wurden die Ketone (mit Ausnahme von AP-OMe) in zwei Gruppen klassifiziert, nämlich die „Gruppe A“-Ketone (keine Radikalbildung) und die „Gruppe B“-Ketone (Radikalbildner). Während die „Gruppe A“-Ketone gegenüber niedrigen Konzentrationen von DNA (62.5 µM) inaktiv waren, verursachten die bei der Bestrahlung der „Gruppe B“-Ketone generierten Peroxylradikale, neben wenigen direkt induzierten Strangbrüchen, hauptsächlich die Guaninoxidationsprodukte 8-oxoGua und guanidinfreisetzende Produkte (GRP). Erst wenn die DNA-Konzentration zehnfach erhöht wird (625 µM), tritt bei der Photolyse der „Gruppe A“-Ketone auch DNA-Oxidation durch einen Elektronentransfer von der Guaninbase auf das angeregte Keton ein. Ein analoger Konzentrationseffekt wurde auch in der dG-Oxidation beobachtet, bei niedrigen Substratkonzentrationen sind nur die radikalbildenden „Gruppe B“-Ketone aktiv. Die Tatsache, dass in der dG-Oxidation durch die „Gruppe A“-Ketone kein 8-oxodG detektiert wurde, wurde auf dessen effiziente Oxidation durch dG•+-Radikalkationen zurückgeführt. Die „Gruppe B“-Ketone sind in Abwesenheit von O2 gegenüber dG und DNA oxidativ inaktiv, da die in der alpha Spaltung generierten kohlenstoffzentrierten Radikale keine Peroxylradikale bilden können. Die „Gruppe A“-Ketone sind gegenüber DNA in Abwesenheit wie auch in Anwesenheit von Sauerstoff genauso reaktiv, da der Elektronentransfer von DNA zum Keton unabhängig von Sauerstoff ist. Um mechanistische Einblicke in die oxidative DNA-Schädigung zu erlangen, wurden photochemische Modellstudien mit dem Nukleosid dG sowie 8-oxodG durchgeführt, wobei zusätzlich Spiroiminodihydantoin gebildet wird. Bis vor kurzem wurde die Struktur dieses Oxidationsproduktes als 4-HO-8-oxodG angenommen, dass zuerst in der dG Oxidation mit Singulettsauerstoff (1O2) beobachtet wurde. Weder Spiroiminodihydantoin noch 4 HO-8-oxodG sind als authentische Verbindungen bekannt, so dass eine zweifelsfreie Strukturaufklärung die Bestimmung der Konnektivität der markierten Positionen erforderte. Diese Zuordnung erfolgte mittels eines SELINQUATE-NMR Spektrums, mit dem schlüssig die 4 HO-8-oxodG-Struktur ausgeschlossen wurde. Wie alle „Gruppe B“-Ketone sind auch alle „Gruppe A“-Ketone in Abwesenheit von O2 mit Ausnahme von AP-OAc gegenüber dG inert. Dies ist ein Beleg dafür, dass der Elektronentransferschritt von dG zum Keton in Abwesenheit von Sauerstoff (im Gegensatz zur DNA-Oxidation) reversibel ist und daher keine Oxidation möglich ist, wenn die Ketylradikale nicht durch O2 abgefangen werden. Das aus AP-OAc gebildete Ketylradikal besitzt als einziges einen effektiven unimolekularen Deaktivierungsweg, nämlich die Acetation-abspaltung, so dass die Reversibilität nicht mehr möglich ist.
In der vorliegenden Dissertation werden optisch aktive Hydroperoxide I als enantioselektive Oxidationsmittel in der Weitz-Scheffer-Epoxidierung von Enonen II eingesetzt. Dabei sollten zunächst die besten Reaktionsbedingungen für eine effektive asymmetrische Induktion gefunden werden, um anhand dieser den Mechanismus des enantioselektiven Sauerstofftransfers aufzuklären. In einer weiteren Studie werden Chinconin- und Chinconidin-abgeleitete optisch aktive Phasentransferkatalysatoren (PTK) IV zur asymmetrischen Epoxidierung von Enonen II mit racemischen Hydroperoxiden I genutzt, wobei vordergründig die kinetische Racematspaltung der verwendeten Hydroperoxide I untersucht werden sollte. Darauf aufbauend wurde eine höchst effektive Methode zur enantioselektiven Epoxidierung von Isoflavonen V mit kommerziell erhältlichen, achiralen Hydroperoxiden entwickelt. 1. Die Optimierung der Reaktionsbedingungen an Chalkon IIa zeigt, dass die höchste Enantioseitendifferenzierung mit (S)-(-)-1-Phenylethylhydroperoxid (Ia) und KOH in Schema A: Asymmetrische Weitz-Scheffer-Epoxidierung mit optisch aktiven Hydroperoxiden I und den Basen KOH oder DBU als Katalysatoren Acetonitril bei –40 °C möglich ist. Dabei bildet sich das (alphaS,betaR)-Epoxid IIIa in 51 Prozent ee. Im Gegensatz dazu wird in Toluol bei 20 °C mit der Base DBU das entgegengesetzt konfigurierte (alphaR,betaS)-Epoxid IIIa in einem Enantiomerenüberschuss von 40 Prozent gebildet. Die Art der Base beeinflusst demnach grundlegend den stereochemischen Verlauf der Reaktion. Um diesen Effekt mechanistisch zu ergründen wird der elektronische Charakter der Arylreste im Enon II systematisch variiert, was allerdings nur zu einer geringen Veränderung der Enantioselektivität führt. Einen größeren Einfluss auf das Ausmaß der Enantioseitendifferenzierung in dieser asymmetrischen Weitz-Scheffer-Epoxidierung hat, sowohl bei der Reaktionsführung mit DBU in Toluol als auch mit KOH in CH3CN, der sterische Anspruch des beta-Substituenten im Enon II. Aufgrund der maßgeblichen Signifikanz der Größe des beta-Substituenten wird eine Templatstruktur T+ (Abbildung A) vorgeschlagen, in der eine sterische Wechselwirkung zwischen dem beta-Substituenten des Enons II und dem Hydroperoxyanion I- den Abbildung A: Bevorzugte Anordnungen in der Templatstruktur für die KON-vermittelte und die DBU-vermittelte Epoxidierung stereochemischen Verlauf der Epoxidierung bestimmt. Das Aggregat aus Substrat, Hydroperoxid und Gegenion wird in Form eines Templats T+ durch das K+-Ion oder das protonierte Amin DBU-H+ zusammengehalten. Dadurch wird den entgegengesetzten Enantioselektivitäten Rechnung getragen, die für diese beiden Basen beobachtet werden. Aus Abbildung A wird ersichtlich, dass die unterschiedliche Größe der K+- oder DBU-H+-Kationen und des beta-Substituenten im Templat wichtig für eine effektive Diskriminierung der beiden möglichen Angriffe T+-(Si) und T+-(Re) ist. Für das relativ kleine Kaliumion dominiert die Wechselwirkung zwischen dem beta-Substituenten und dem Hydroperoxid I. Diese wird im T+-(Si)-Angriff minimiert, indem das Wasserstoffatom am stereogenen Zentrum des Hydroperoxids I auf der Seite des Enons II steht. In der Epoxidierung mit der sterisch anspruchsvolleren Base DBU tritt die Wechselwirkung zwischen DBU-H+ und dem Hydroperoxid im Templat in den Vordergrund, was den Angriff auf der Re-Seite bedingt. Demnach werden mit KOH die besten Enantioselektivitäten für große beta-Substituenten beobachtet, wohingegen für die Amin-vermittelte Epoxidierung eine große Base, wie DBU, vorteilhaft ist. Sowohl für KOH als auch für DBU als Basenkatalysatoren wird die Validität der Templatstruktur durch weitere Variation der Reaktionsbedingungen geprüft. Wenn K+ durch den Kronenether 18-Krone-6 komplexiert wird oder anstelle von DBU-H+ eine nicht-koordinierende Schwesinger Base verwendet wird, das Templat also nicht durch Koordination gebildet werden kann, werden deutlich niedrigere Enantioselektivitäten in der Epoxidierung beobachtet. Die Notwendigkeit der S-cis-Konformation des Enons II für die Bildung des Templats, wird durch Untersuchungen mit konformationell fixierten Enonen untermauert. So wird die Enantioselektivität bei der Epoxidierung eines S-cis-fixierten Enons (IIb) auf bis zu 90 Prozent ee erhöht, während sie bei einer S-trans-Fixierung des Enons deutlich auf < 5 Prozent ee abfiel. Fazit: Mit den optisch aktiven Hydroperoxiden I wird in der Weitz-Scheffer-Epoxidierung durch die Wahl geeigneter Basen, KOH oder DBU, sowohl das (alphaS,betaR)-Epoxid III (bis zu 90 Prozent ee) als auch das (alphaR,betaS)-Epoxid (bis zu 72 Prozent ee) erhalten. Welches Enantiomer überwiegt kann dabei allein durch die Wahl der Base gesteuert werden. Die Enantioseitendifferenzierung wird durch sterische Wechselwirkungen in einem Templat aus Enon II, Hydroperoxid I und den Kationen K+ oder DBU-H+ bestimmt. Die kinetische Racematspaltung chiraler Hydroperoxide I durch Weitz-Scheffer-Epoxidierung mit optisch aktiven Chinconin-basierten Phasentransferkatalysatoren (PTK) IV wird untersucht, bei der als willkommenes „Nebenprodukt" optisch aktive Isoflavonepoxide VI (Schema B) mit bis zu 92 Prozent ee entstehen. Die Racematspaltung ist Schema B: Kinetische Racematspaltung des chiralen Hydroperoxids Ia mittels Weitz-Scheffer-Epoxidierung und dem optisch aktiven PTK IV jedoch nicht effektiv, es werden ee-Werte von maximal 33 Prozent erzielt. Auf dieser Basis wird eine Methode zur asymmetrischen Epoxidierung der Isoflavonen (V) (Schema C) mit dem Schema C: Enantioselektivitäten für die Epoxidierung der Enone IIb,c und des Isoflavons Vb in Anwesenheit des PTK IV kommerziell verfügbaren Cumylhydroperoxid entwickelt, die für das Isoflavon Vb bis zu 98 Prozent ee zu Gunsten des (1aR,7aS)-Epoxids ergibt. Die hohe Enantioselektivität wird mit dem Templat A (Schema D) erklärt, in dem eine H-Brücke von der Hydroxy-Funktion des PTK IV Schema D: Wasserstoffbrückengebundene Templatstrukturen A und B zum endocyclischen Ethersauerstoffatom des Isoflavons V ausgeht. Die Relevanz einer solchen H-Brücke ist durch Methylierung der Hydroxy-Funktion des PTK IV demonstriert. Zudem ist die Wichtigkeit dieses Ethersauerstoffatoms durch die Tatsache untermauert, dass das konformationell ähnliche Enon IIc (Schema C) nahezu unselektiv epoxidiert wird (18 Prozent ee). Eine analoge H-Brücke nunmehr zum Carbonylsauerstoffatom des S-cis-fixierten Enons IIb wird als Erklärung für dessen hoch enantioselektive Epoxidierung (95 Prozent ee) postuliert (Templat B, Schema D). Fazit: Die asymmetrische Weitz-Scheffer-Epoxidierung mit dem optisch aktiven Phasentransferkatalysator IV wird zur Herstellung fast enantiomerenreiner Epoxide (bis zu 98 Prozent ee) genutzt. Für die Enantioseitendifferenzierung zeigt sich die Ausbildung einer H-Brücke zwischen PTK IV und Substrat II oder V als essentiell. In der kinetischen Racematspaltung chiraler Hydroperoxide I ist diese Epoxidierung nicht effektiv.
Die kausalen Ursachen, die zur Auslösung der neurodegenerativen Erkrankung Morbus Parkinson führen, sind noch immer unklar. Man nimmt heute an, daß das Absterben dopaminerger Neurone im Mittelhirn von Parkinsonpatienten multifaktoriell ausgelöst wird. Genetische Prädisposition sowie endogene und exogene Umweltgifte wie etwa Substanzen, die strukturelle Ähnlichkeit mit dem bekanntesten dopaminergen Neurotoxin MPTP (1-Methyl-4-phenyl-1,2,3,5-tetrahydropyridin) besitzen, werden als Hauptursachen für die Entstehung des Parkinsonsyndroms diskutiert. Diese Arbeit beschäftigt sich mit einer neuen Klasse von neurotoxisch wirksamen Tetrahydro-b-carbolinen, die sich von Trichloracetaldehyd (Chloral) ableiten. Die wohl prominenteste Verbindung in dieser Reihe ist TaClo (1-Trichlormethyl-1,2,3,4-tetrahydro-b-carbolin), das im menschlichen Körper nach Aufnahme des Schlafmittels Chloralhydrat durch Pictet-Spengler-Kondensation mit dem endogen vorhandenen Tryptamin gebildet wird. Zusätzlich scheint die Bildung von TaClo aus dem Industrielösungsmittel TRI (Trichlorethylen), das im Organismus zu Chloral metabolisiert wird, möglich. Die über Chloral eingeführte große CCl3-Gruppe erhöht die Lipophilie von TaClo, die Passage der Blut-Hirn-Schranke ist erleichtert. In der Tat haben zahlreiche Untersuchungen in vitro und in vivo gezeigt, daß TaClo toxische Prozesse in dopaminergen und serotonergen Systemen zu induzieren vermag. Ein wesentliches Ziel dieser Arbeit bestand darin, die in-vivo-Entstehung und Metabolisierung von TaClo im Menschen sowie den Einfluß dieses Neurotoxins auf die DNA herauszuarbeiten und näher zu untersuchen. Im einzelnen wurden folgende Ergebnisse erzielt: TaClo schädigt die DNA, wie Versuche an zellfreier DNA und in-vitro-Experimente an PC12-Zellen belegen. Die endogene Bildung von TaClo in Chloralhydrat-behandelten Patienten und die in-vitro-Entstehung von TaClo aus Trichlorethylen wurde mittels HPLC-ESI-MS-MS-Analytik eindeutig bewiesen. Außerdem wurden erste Hinweise auf eine Anreicherung des Neurotoxins im menschlichen Körper erhalten. Stereostrukturelle Aspekte der Bildung und Verstoffwechslung von TaClo wurden aufgeklärt und TaClo-Metabolite in in-vitro- und in-vivo-Proben identifiziert. Das Tetrahydro-b-carbolin entsteht in racemischer Form, wird also nicht enzymatisch sondern spontan durch Pictet-Spengler-Reaktion gebildet. Zusätzlich wurden Hinweise auf eine enzymatische Metabolisierung von TaClo gefunden. Außerdem gelang es, eine etablierte Methode des oxidativen Abbaus zur Aufklärung der absoluten Konfiguration von Tetrahydropyridin-Heterocyclen [z.B. Eleagnin] auf Substanzen mit benzylischer Hydroxy- oder Metylether-Gruppe zu erweitern. Geringe Mengen (1-2 mg) an Substanzen, die in benzylischer Position eine chirale Sauerstoffunktion besitzen, wurden durch Ruthenium-katalysierte Oxidation zu GC-gängigen Säuren abgebaut, deren absolute Konfiguration nach Trennung an chiraler Phase durch Vergleich mit enantiomerenreinem Referenzmaterial bestimmt wurde. Diese Zuordnung erlaubte den Rückschluß auf die absolute Konfiguration einer Reihe Ausgangsverbindungen.
Tropische Pflanzen gelten als wertvolle Quellen biologisch aktiver Sekundärmetaboliten. Im Rahmen dieser Dissertation wurden mehrere tropische Arten unter verschiedenen naturstoffchemischen Gesichtspunkten bearbeitet. Einen Schwerpunkt bildeten die tropischen Lianenfamilien der Ancistrocladaceen und Dioncophyllaceen. Diese produzieren die Naphthylisochinolin-Alkaloide, Biarylnaturstoffe mit faszinierenden strukturellen und pharmakologischen Eigenschaften. In dieser Arbeit wurde die Biogenese der Naphthylisochinolinalkaloide am Beispiel von Dioncophyllin A aus Triphyophyllum peltatum mittels Verfütterung von [13C2]-Acetat untersucht. 13C-NMR-Experimente offenbarten einen Einbau von diskreten [13C2]-Einheiten im ganzen Kohlenstoffgerüst. Damit sind die Naphthylisochinolin-Alkaloide die ersten bekannten Tetrahydroisochinolin-Alkaloide mit einem polyketidischen Ursprung. Als weiteres Forschungsobjekt diente das strukturell und biogenetisch hochinteressante Pyridon-Alkaloid Antidesmon aus Antidesma membranaceum. Untersuchungen zur Biogenese führten zur Entdeckung eines neuen Biogeneseweges. Antidesmon wird ebenfalls aus einer Polyketid-Vorstufe gebildet. Überraschenderweise dient Glycin als Vorstufe für einen C2N-Baustein. Im analytischen Bereich standen der Einsatz und die methodische Verbesserung der online-"Triade" HPLC-MS/MS, HPLC-NMR und HPLC-CD im Vordergrund. Diese neuen Techniken dienten im Anschluß der phytochemischen Untersuchung verschiedener Pflanzenextrakte. Nach der erstmaligen online-Analyse von Ancistrocladus griffithii wurden drei neue Alkaloide, Ancistrogriffin A, B und C, detektiert und strukturell aufgeklärt. Weiterhin wurde das erste dimere Naphthylisochinolin-Alkaloid aus einer asiatischen Ancistrocladus-Spezies, Ancistrogriffithin A, detektiert und vor seiner Isolierung strukturell mit online-Methoden aufgeklärt.
