Die Chlorophylle stellen in der Natur die wichtigsten Pigmente dar, weil sie verantwortlich für die Photosynthese sind und hierbei vielfältige Funktionen wahrnehmen, die sich aus ihrer Selbstassemblierung sowie den vorteilhaften optischen und Redox-Eigenschaften ergeben. Die in dieser Arbeit untersuchten semisynthetischen Zinkchlorine stellen Modellverbindungen des natürlichen Bacteriochlorophylls c (BChl c) der Lichtsammelsysteme (light-harvesting: LH) in Chlorosomen von Bakterien, jedoch ohne Proteingerüst, dar. Die entscheidenden Vorteile dieser Zinkchlorine (ZnChl) gegenüber den natürlichen BChls bestehen im einfachen semisynthetischen Zugang ausgehend von Chlorophyll a (Chl a), ihrer gesteigerten chemischen Stabilität sowie der Möglichkeit ihre Selbstassemblierung durch gezielte chemische Modifizierung der Seitenketten in der Peripherie zu steuern. Während bereits mehrfach über die vielversprechenden Redox- und excitonischen Eigenschaften von Aggregaten von ZnChl und natürlichem BChl c und den damit verbundene Voraussetzungen für Excitontransport über große Distanzen berichtet wurde, sind die Ladungstransporteigenschaften von Aggregaten der biomimetischen ZnChl bis heute unerforscht. Die vorliegende Arbeit beschäftigt sich mit der Aufklärung der Struktur von Aggregaten einer Vielzahl von semisynthetischen Zinkchlorophyllderivaten im Feststoff, in Lösung und auf Oberflächen durch die Kombination verschiedenster spektroskopischer, kristallographischer und mikroskopischer Techniken an die sich Untersuchungen zum Ladungstransport in den Aggregaten anschließen. Schema 1 zeigt die verschiedenen, in dieser Arbeit synthetisierten ZnChls, die entweder mit einer Hydroxy- oder Methoxygruppe in der 31-Position funktionalisiert sind sowie Substituenten unterschiedlicher Art, Länge und Verzweigung an der Benzylestergruppe in 172-Position tragen.Die Packung dieser Farbstoffe hängt entscheidend von ihrer chemischen Struktur ab. Während die ZnChls 1a, 2a, 3 mit 31-Hydroxygruppe und Alkylseitenketten (Dodecyl bzw. Oligoethylenglykol) gut lösliche stabförmige Aggregate bilden, lagern sich die analogen Verbindungen mit 31-Methoxygruppe (1b, 2b) zu Stapeln in Lösung und auf Oberflächen zusammen. Diese supramolekularen Polymere wurden im Detail in Kapitel 3 mit Hilfe von UV/Vis- und CD-Spektroskopie (circular dichroism: CD) sowie dynamische Lichtstreuung (dynamic light scattering: DLS) untersucht. Darüber hinaus lieferten temperaturabhängige UV/Vis- in Kombination mit DLS-Messungen wertvolle Informationen über die Aggregationsprozess dieser beiden Sorten von Aggregaten. Während sich die ZnChl 1a mit 31 Hydroxygruppe entsprechend dem isodesmischen Modell zu röhrenförmigen Aggregaten zusammenlagern, bilden sich die stapelförmigen Aggregate von 1b nach einem kooperativen Keimbildungs-Wachstums-Mechanismus (nucleation-elongation mechanism). Detaillierte elektronenmikroskopische Studien lieferten erstmals überzeugende Beweise für röhrenförmige Nanostrukturen der Aggregate des wasserlöslichen 31-Hydroxy Zinkchlorin 3. Die gemessenen Durchmesser der Röhren von ~ 5-6 nm dieser Aggregate liegen in hervorragender Übereinstimmung mit den Elektronenmikroskopie-Daten von BChl c Stabaggregaten in Chlorosomen (Chloroflexus aurantiacus, Durchmesser ~ 5-6 nm) und entsprechen damit dem von Holzwarth und Schaffner postulierten röhrenförmigen Modell... Im Einklang mit ihren hoch geordneten, robusten Strukturen, die sich eindimensional in einer Größenordnung von Mikrometeren erstrecken, sowie ihrer Fähigkeit zum effizienten Ladungs-trägertransport stellen diese selbstassemblierten Nanoröhren von ZnChls vielversprechende Ausgangsmaterialien für die Fertigung supramolekularer elektronischer Bauteile dar. Wissenschaftliche Bemühungen einige dieser Moleküle und ihre entsprechenden supramolekularen Polymere für die Fertigung von (opto-)elektronischen Bauteilen wie organischen Feldeffekttransistoren zu benutzten, stellen lohnende Aufgaben für die Zukunft dar...
Synthesis and Characterization of an Oligo(Phenylene Ethynylene)-Based Perylene Bisimide Foldamer

The present work is part of the currently only rudimentary understanding of the structure-property relationships in the self-assembly of pi-conjugated organic molecules. Such structures may reveal favorable photophysical and semiconducting properties due to the weak non-covalent pi-pi interactions between the monomer units. The specific mutual orientation of the dyes is known to evoke individual functional properties for the condensed matter, however, the related electronic processes are still not well-understood and further enhancements of functional properties are seldom triggered by rational design. The pi-pi self-assembly structures of perylene bisimide (PBI) dyes are promising, versatile materials for organic electronic devices and have been elected for this thesis as an archetype aggregate system to investigate the dye-dye interactions in more detail. In cooperation with experts in the field of spectroscopy and theory the development of reliable routines towards a better understanding of the origins of the functional properties may be feasible, and, on a longer time-line, such knowledge may enable optimization of functional organic materials. Having designed such structures entailed the challenge of developing feasible synthesis strategies, and to actually generate the targeted molecules by synthesis. Several synthesis approaches were conducted until finally a perylene bisimide foldamer was obtained based on a Sonogashira co-polymerization reaction. After purification and enrichment of the larger-sized species by means of semi-preparative gel permeation chromatography (GPC) the average size of an octamer (8500 Da) species was determined by analytical GPC. The low polydispersity index (PD) of 1.1 is indicative of a sharp size distribution of the oligomers. This average size was confirmed by performing diffusion ordered NMR spectroscopy (DOSY). Furthermore, MALDI-TOF mass analysis substantiated the structural integrity of the co-polymerization product. Solvent-dependent UV/vis spectroscopic investigations demonstrated that intramolecular PBI aggregates are reversibly formed, indicating that this oligomer is able to fold and unfold in the intended manner upon changing external conditions. In the unfolded states, the PBI moieties are closely arranged due to the short OPE bridges (< 2.4 nm), which is expressed by an exciton coupling interaction of the dyes and therefore the characteristic monomer absorption pattern of the PBI chromophore cannot be obtained in the unfolded states. More interestingly, the folded state revealed a pronounced aggregate spectrum of the PBIs, however, striking differences in the shape of the absorption spectrum compared to our previously investigated PBI self-assembly were obtained.
In dieser Arbeit wurde die Kupfer(I)-katalysierte Cycloaddition von Alkinen und Aziden ("Click-Chemie") als effiziente, vielseitige und milde Reaktion für die Funktionalisierung von Nanodiamantpartikeln etabliert. Es wurden verschiedene Alkin- oder Azid-funktionalisierte Nanodiamantsysteme hergestellt und deren Reaktivität in Click-Reaktionen anhand vielfältiger Beispiele demonstriert. Hierzu wurden neben einfachen Testverbindungen auch komplexere Substanzen (z. B. ein Mannose-Derivat, Polytriazole, Fluoreszenzfarbstoffe, ein Thiazolium-Organokatalysator) durch Triazolsynthese immobilisiert. Zusätzlich wurde eine Methode entwickelt, die die Herstellung funktionalisierter und vollständig dispergierter Nanodiamant-Primärteilchen ermöglicht, wobei gezeigt wurde, dass dieser Weg auch für die Funktionalisierung durch Click-Chemie geeignet ist. Die Analyse der Nanodiamantpartikel erfolgte u. a. durch FT-IR, TGA, Elementaranalyse, Partikelgrößen- und Zetapotentialbestimmung, NMR, HR-TEM, UV / Vis sowie Fluoreszenzspektroskopie und -mikroskopie.
n this work the synthesis and analysis of chromophore functionalized spherical gold nanoparticles is presented. The optical, electrochemical and spectroelectrochemical properties of these hybrid materials are furthermore studied. The work therefore is divided into two parts. The first part deals with triarylamine and PCTM-radical functionalized gold nanoparticles. The focus thereby was on the synthesis and on the investigations of chromophore-chromophore interactions and gold core-chromophore interactions. The chromopores, especially triarylamines, were attached to the gold core via different bridging units and were studied with optical and electrochemical methods. The purity and dimensions of the nanoparticles was determined by 1H-NMR spectroscopy, diffusion ordered NMR spectroscopy (DOSY), TGA, XPS and STEM. Furthermore a cyclic voltammetry technique was used to determine the composition of the particles via the Randles-Sevcik equation. An analysis of these parameters led to a model of a sea urchin-shaped nanoparticle. Optical measurements of the particles revealed an anisotropic absorption behavior of the triarylamine units due to gold core-chromophore interaction. However this behavior depends strongly on the relative orientation of the transition dipole moment of the chromophore to the gold surface and the distance of the chromophore to the surface. Hence, the anisotropic behavior was exclusively detected in the spectra of the Au-Tara1 particles. The short and rigid pi-conjugated bridging unit thereby facilitates this gold core-chromophore interaction. It was shown from electrochemical investigations that the triarylamine units can be chemically reversibly oxidized to the triarylamine monoradical cation. Furthermore, the measurements revealed a strong interligand triarylamine-triarylamine interaction which was only seen for the Au-Tara1 particles. The long pi-conjugated bridging units of the Au-Tara2 and Au-Tara3 particles as well as the aliphatic bridging unit of Au-Tara4 prevent any detectable interligand interactions. One may conclude that both the gold core-chromophore and the interligand triarylamine-triarylamine interaction depend on the length and the rigidity of the bridging unit. The electron transfer behavior of the triarylamine units adsorbed onto the gold core was additionally studied via spectroelectrochemical (SEC) measurements which are able to reveal weaker interactions. The investigations of Au-Tara1 and Au-Tara2 revealed a significant strong coupling between neighboring triarylamine units which is due to through-space intervalence interactions. This behavior was not detected for Au-Tara3 or for Au-Tara4. The SEC analysis also revealed that these observed interligand interactions depend on the length and the rigidity of the bridging unit. Thus, the systematic variation of the bridging unit gave a basic insight in the optical and electrochemical properties of triarylamines, located in the vicinity of a gold nanoparticle. The second part of this work aimed at the synthesis of new molecules, denoted as SERS-markers, for immuno SERS applications. For this purpose, the SERS-markers were designed to have a Raman-active unit and a thiol group for chemisorptions to Au/Ag nanoshells. In cooperation with the group of Schlücker (University of Osnabrück) the SERS-markers were absorbed onto Au/Ag nanoshells, denoted as SERS-labels, and characterized. The SERS spectra of the SERS-labels exhibited intense and characteristic SERS-signals for each marker. For immuno SERS investigations SEMA3 was functionalized with a hydrophilic end unit. This marker was adsorbed onto an Au/Ag nanoshell and encapsulated with silica. An anti-p63 antibody was bound to the silica surface in order to generate a SERS-labeled antibody for the detection of the tumor suppressor p63 in benign prostate. Immuno-SERS imaging of prostate tissue incubated with SERS-labeled anti-p63 antibodies demonstrated the selective detection of p63 in the basal epithelium. The results show the potential of the method for the detection of several biomolecules in a multiplexing SERS experiment.
Einleitung: Die α-Hydroxycarbonsäuren 2 sind nicht nur wichtige chirale Bausteine für die organische Synthese,[20–25] sondern ebenfalls für den Menschen von großer Bedeutung aufgrund ihrer vielfältigen Anwendungen in der Lebensmittelbranche, Kosmetikindustrie und Medizin.[13–15] Aus diesen Gründen ist ein effizienter Zugang auch zu enantiomenreinen Säuren 2 notwendig. Viele natürlich vorkommende Vertreter sind aus dem chiral pool isolierbar, doch gerade für wissenschaftliche Zwecke sind häufig spezielle Strukturen von Interesse, die nur synthetisch darstellbar sind. Deshalb wurden, basierend auf unterschiedlichen Ansätzen, zahlreiche stereoselektive Routen zu α-Hydroxycarbonsäuren 2 entwickelt.[26–79] Eine schnelle und zugleich breit anwendbare Methode, 2 durch Hydroxylierung unfunktionalisierter Carbonsäuren 20 zu synthetisieren, sucht man jedoch vergebens. Das Ziel des ersten Teils dieser Arbeit bestand in der Entwicklung einer stereoselektiven Eintopfsynthese zur α-Hydroxylierung α-unfunktionalisierter Carbonsäurederivate 20 ... Fazit: Mit der MHACU von chiralen O-acylierten Oxazolin-N-oxiden (S)-37 wurde eine schnelle und einfache Eintopf-Synthese für α-Acyloxyoxazoline (S,S)-39, α-Hydroxymethylester (S)-30 und -säuren (S)-2 ausgehend von unfunktionalisierten Orthoestern 29 erarbeitet, die exzellente Diastereomeren- und Enantiomerenüberschüsse erlaubt (94–98% de/ee). Einleitung zum zweiten Teil der Arbeit: L-Prolin [(S)-222] gilt als einer der bedeutendsten Organokatalysatoren überhaupt und besticht durch seine strukturelle Einfachheit und seine erstklassigen katalytischen Eigenschaften.[230–232] Zudem stehen bei keinem zweiten Katalysator die materielle Verfügbarkeit und die chemische Anwendbarkeit in einem so idealen Verhältnis. Für einen Grundlagenforscher ist es daher eine besonders interessante Aufgabe, strukturelle Merkmale von L-Prolin [(S)-222] mit Erkenntnissen aus mechanistischen Untersuchungen zu verschmelzen, um so neue, noch leistungsstärkere Katalysatoren zu entwickeln. Fazit: Die vorgelegten Konzepte zur Verbesserung der Reaktivität von Pyrrolidin-basierten Katalysatoren erschienen zwar aussichtsreich, jedoch lieferten die bisherigen Umsetzungen noch keine brauchbaren Ergebnisse. Die in diesem Rahmen synthetisierten Aminosäuren und Isoxazolidine besaßen nur sehr geringe katalytische Aktivitäten. Weitere Arbeiten, zum Beispiel Veränderungen an den Katalysatorstrukturen, sind nötig, um eine erfolgreiche Umsetzung dieser Konzepte noch zu realisieren.
Quantum chemical calculations of circular dichroism (CD) spectra in combination with experimental CD studies are one of the most efficient analytical tools for the elucidation of the three-dimensional structure of a chiral molecule. In the present work 18 chiral compounds of most different molecular structures and origins were investigated using various theoretical methods (the semiempirical CIS methods, the time-dependent DFT and DFT/MRCI approaches). The advantages and limitations of the applied methods were discussed in the context of the studied compounds. Furthermore, the last part of this work deals with the CD investigations of a chiral compound in the crystalline state. A well-known natural product with a specific conformation/CD spectrum behavior was used as a model compound to examine a novel solid-state CD method and to investigate the possibility of its improvement to provide a higher reliability for the assignment of the absolute configuration.
Für Diamantpartikel basiert diese Funktionalisierungschemie auf den bereits vorhanden Oberflächengruppen, die durch die Synthese und Reinigung entstanden sind. Die stabilste Anbindung wäre eine C-C-Verknüpfung der Diamantatome auf der Oberfläche mit den organischen Reagenzien. Um diese Verknüpfung zu erreichen, wurde die Oberfläche mittels Diels-Alder-Reaktion funktionalisiert. Grundvoraussetzung hierfür ist eine Oberfläche, die über eine ausreichende Menge an C=C-Doppelbindungen verfügt. Dies wurde durch eine thermische Behandlung erreicht. Die gewonnenen Ergebnisse zeigen, dass ab einer Behandlungstemperatur von 750 °C keine funktionellen Gruppen durch die eingesetzten Analysemethoden mehr nachgewiesen werden können. Die Elementaranalyse der einzelnen Proben zeigt einen deutlichen Anstieg des Kohlenstoffgehaltes. Nach erfolgreicher thermischer Behandlung des Diamanten wurde durch die Verwendung von Cyclopentadien und Anthracen als Dien gezeigt, dass eine Funktionalisierung über eine Diels-Alder-Reaktion generell möglich ist. Daher wurde als alternatives Dien ein o-Chinodimethan eingesetzt. Hierzu wurde eine systematische Untersuchung der Umsetzung der unterschiedlichen thermisch behandelten Proben mit α,α’-Dibrom-o-xylol durchgeführt. Hierdurch wurde gezeigt, dass der Ausgangsdiamant bei mindestens 750 °C behandelt werden muss, um eine messbare Oberflächenbelegung zu erhalten. Im Anschluss wurde die Funktionalisierbarkeit der eingeführten Arylgruppen überprüft. Durch die Einführung unterschiedlicher funktioneller Gruppen ist es dann möglich, Moleküle für spezielle Anwendungen anzubinden. Durch die Anwendung der klassischen Aromatenchemie war es möglich, die aromatischen Oberflächen zu nitrieren und sulfonieren. Die Sulfonsäuregruppe konnte partiell zum Thiol reduziert werden. Durch Umsetzung dieser Thiolgruppen war es möglich, einen mannosemodifizierten sowie einen farbstoffmodifizierten Diamanten herzustellen. Da die nachträgliche Einführung von Carboxylgruppen mit hohem synthetischem Aufwand verbunden ist, wurde alternativ ein carboxyltragender Vorläufer mit dem thermisch behandelten Diamant umgesetzt. Hierbei wurde eine Oberflächenbelegung von ca. 0.8 mmol g-1 erreicht (Ausgangsdiamant thermisch behandelt bei 750 °C). Im Anschluss wurde die Säuregruppe mit einem einfach geschützten Diamin weiterfunktionalisiert. Hierbei wurden unterschiedliche Methoden zur Amidbildung getestet. Als effektivste Methode stellte sich die Synthese über ein Säurechlorid heraus. Infolgedessen war eine weitgehende Umsetzung (ca. 94 - 88 %) der Säuregruppe möglich. Eine weitere wichtige funktionelle Gruppe für die Anbindung größerer Einheiten stellen die Amine dar. Diese lassen sich einfach darstellen, z. B. durch Reduktion von Cyanogruppen. Daher wurde 2,3-Bis-(brommethyl)-5,6-dicyanopyrazin als Edukt für die Funktionalisierung eingesetzt. Nach anschließender Reduktion der Cyanogruppen wurden primäre Amine erhalten. Die so erzeugte Aminogruppe wurde mit Biotin weiter funktionalisiert. Die bisher eingesetzten o-Chinodimethane sind ausschließlich durch 1,4-Eliminierung zugänglich. Eine sinnvolle Alternative zu diesem Vorläufer ist die Verwendung eines Cyclobutenderivates. Der Vorteil liegt in einer einfacheren Reinigung des Produktdiamanten. Als Testverbindung wurde 2,4-Bis-(methylsulfonyl)-5,6-dihydro-cyclobuta-[d]-pyrimidin verwendet. Nach erfolgreicher Umsetzung mit dem Diamanten wurden die Thioethergruppen in Sulfone überführt und in 2-Position wurde das Sulfon gegen eine Aminogruppe ausgetauscht. Neben den durchgeführten Funktionalisierungsreaktionen wurde eine nasschemische Nachweismethode zur Quantifizierung von primären Aminen entwickelt. Ein Vorteil des Verfahrens ist, dass der Nachweis auf einfache Weise durchführbar ist und nur 1 – 2 mg Diamantprobe nötig sind. Die gewonnen Ergebnisse zeigen, dass durch den Nachweis verlässliche Angaben über die genaue Aminmenge erhalten werden.
Nanodiamant weist aufgrund seiner herausragenden mechanischen, optischen und biokompatiblen Eigenschaften ein enormes Potential auf. Für eine erfolgreiche Anwendung muss dieser jedoch zunächst desagglomerisiert und an seiner Oberfläche funktionalisiert werden. In dieser Arbeit werden zwei unterschiedlichen Methoden zur Deagglomerierung und Oberflächenfunktionalisierung vorgestellt. Im ersten Teil wird die thermochemische Methode beschrieben, um den ungewöhnlich stark agglomerierten Detonationsnanodiamant zu deagglomerieren. Dabei wird die Strategie verfolgt, den Nanodiamant erst durch eine thermische Behandlung im Vakuum die vorhandenen Oberflächengruppe zu entfernen, π-Bindungen zu etablieren und dann dessen Oberfläche durch kovalente Bindungen zu modifizieren. Im zweiten Teil wird eine mechanochemische Methode für die Deagglomerierung und Oberflächenfunktionalisierung vorgestellt. Dabei wird ein völlig neuer Ansatz (BASD-Verfahren) entwickelt. Die Ultraschallbehandlung in Kombination mit zusätzlichen Mikrokeramikpartikeln eignet sich hervorragend, um Nanodiamant-agglomerate aufzubrechen. Zwei unterschiedliche Reaktionen mit zwei unterschiedlichen Nanodiamantsorten werden getestet: Eine einfache Kondensationsreaktion durch Silanisierung und eine Radikalreaktion über das Diazoniumsalz. In weiteren Experimenten kann gezeigt werden, dass sich die zuerst vorgestellte thermochemische Methode auch eignet, um aus fluoreszierenden NV-Nanodiamant kolloidalen NV-Nanodiamant mit funktionellen Gruppen herzustellen. Der letzte Teil der vorliegenden Arbeit beschäftigt sich mit der Herstellung von Nanodiamanten aus Diamantfilmen mit Hilfe des Top-down Ansatzes, da der gezielte und kontrollierte Aufbau der Gitterstruktur des Diamanten sowie die kontrollierte Einführung bisher nur durch CVD oder Ionenimplantationstechnik möglich ist.
In the first part of the work three polycarbazoles poly[N-((4-dimesitylboryl)-3,5-dimethylphenyl)-carbazole]-2,7-diyl P1, poly[N-((4-dimesitylboryl)-3,5-dimethylphenyl)-carbazole]-3,6-diyl P2 and poly[N-(4-(diphenylmethylene)-phenyl)- carbazole]-2,7-diyl P3 were synthesized by Yamamoto coupling reaction and their spectroscopic and electrochemical properties were investigated. Absorption and fluorescence characteristics of P1 and P3 were found to be similar to other 2,7-linked polycarbazoles, whereas P2 shows a CT absorption band arising from a shift of electron density from the nitrogen of the carbazole donor to the triarylborane acceptor. This causes a negative solvatochromic absorption and a positive solvatochromic fluorescence behaviour and is responsible for the significantly enlarged fluorescence quantum efficiency in solution and solid state compared to other 3,6-linked polycarbazoles. Thus the spectroscopic properties are governed by the connection pattern: the 2,7-linked polycarbazoles are not affected by the acceptor substituent due to the rigid poly-para-phenylene-like backbone structure, whereas the 3,6-linked polycarbazole P2 is dominated by the properties of the monomer unit due to its more flexible (less conjugated) structure. The oxidative processes of P1-P3 have been investigated in detail by cyclic voltammetry, which are similar to known 2,7- and 3,6-polycarbazoles. The reversible reduction found for P1 and P2, respectively, is attributed to the reduction of the triarylborane moiety. No reduction process referring to the carbazole moiety was observed. Due to its better solubility compared to P1 and P3 only P2 was used as active layer in an OLED device (ITO/P2/Al). The electroluminescence spectrum revealed CIE coordinates of (0.17, 0.21). In the second part of the work the low band gap polyradical poly{[((2,3,4,5,6-pentachlorophenyl)-bis(2,3,5,6-tetrachlorophenyl)methyl radical)-4,4’-diyl]-alt-4,4’-bis(vinylphenyl)-4-(2-ethylhexyloxy)phenylamin} P4 was synthesized by Horner-Emmons reaction. It shows an IV-CT band in the NIR, which arises from an ET from the triarylamine donor to the PCTM radical acceptor. This transition is confined to one monomer unit as deduced from comparison with the monomer spectra. HOMO and LUMO of P4 determined by cyclic voltammetry are at -5.5 and -4.5 eV, respectively. The smaller electrochemical band gap (1.0 eV) compared to the optical band gap (1.2 eV) is probably caused by ion pairing effects in the electrochemical experiments and indicates a low exciton binding energy. Femtosecond-pump-probe transient absorption spectroscopy revealed the spectral features of the oxidized triarylamine donor and the reduced PCTM acceptor similar to the spectra obtained separately for positive and negative potentials by spectroelectrochemistry. Thus the ET event causing the IV-CT absorption band could unambiguously be identified. The decay of the IV-CT state was found to be biexponential. The fast solvent dependent decay component is ascribed to the direct decay from the IV-CT state to the ground state, whereas the slow solvent independent decay component is tentatively attributed to an equilibrium formation of the IV-CT state and a completely charge separated state formed by charge migration along the polymer backbone. Well balanced ambipolar charge transport with hole and electron mobilities of ca. 3 × 10-5 cm2 V-1 s-1 was found in OFET devices (BG/TC structure) comprising an additional insulating organic PPcB layer. Polymer/polymer BHJ solar cell devices with the structure glass/ITO/PEDOT:PSS/(P3HT/P4)/Ca/Al yielded a power conversion efficiency of 3.1 × 10-3 %, VOC = 0.38 V, JSC = 2.8 × 10-2 mA cm-2 and FF = 0.29 for the 1:4 (P3HT/P4) blend ratio. The improper solid state morphology of P4 that causes the unsatisfying performance of OFET and solar cell devices renders P4 less suitable for these applications, whereas the hypothesis of charge migration in the excited state is worth to be investigated in more detail.
Seit der Strukturaufklärung der grünen Blattpigmente Chlorophyll a und Chlorophyll b sowie des roten Blutfarbstoffes Häm durch Richard Willstätter und Hans Fischer zu Beginn des 20. Jahrhunderts stehen tetrapyrrolische Naturstoffe weltweit im Fokus unzähliger biologischer, medizinischer, physikalischer und chemischer Forschungsarbeiten. Heute spielen insbesondere Porphyrine – die prominentesten Vertreter der synthetischen Tetrapyrrol-Makrocyclen – eine bedeutende Rolle in der modernen angewandten Chemie, etwa als metallorganische Katalysatoren, als Photosensibilisatoren in der photodynamischen Krebstherapie oder auf dem Gebiet der Materialwissenschaften. Neben monomeren Porphyrinen sind dabei v.a. Multiporphyrine mit maßgeschneiderten photophysikalischen Eigenschaften und definierter dreidimensionaler Struktur höchst attraktive Syntheseziele. Im Gegensatz zum immensen Forschungsinteresse an achiralen Porphyrin-Systemen wurde der Darstellung und stereochemischen Charakterisierung chiraler Porphyrinoide bislang vergleichsweise wenig Beachtung geschenkt. Insbesondere optisch aktive Vertreter mit stereo-genen Porphyrin-Aryl-Achsen und intrinsisch axial-chirale Oligoporphyrine wurden bislang kaum untersucht. Aufgrund eines Mangels an geeignet funktionalisierten tetrapyrrolischen Vorläufern sind hierbei Strukturmotive mit β-Verknüpfung besonders unterrepräsentiert. Die generell spärliche Beschreibung axial-chiraler Porphyrin-Systeme und ihrer chiroptischen Eigenschaften liegt hauptsächlich in der oft extrem schweren Zugänglichkeit entsprechender Verbindungen – insbesondere in optisch reiner Form – begründet. Aufgrund der derzeit rapide ansteigenden Bedeutung chiraler Porphyrinoide sind die Synthese und stereochemische Analyse sowie eine Erweiterung des bis dato mehr als begrenzten methodischen Repertoires zur stereoselektiven Darstellung von chiralen Porphyrin-Derivaten von größtem Interesse. Ziel der vorliegenden Arbeit war daher die Etablierung effizienter und vielseitig ein-setzbarer Verfahren zum Aufbau komplexer axial-chiraler Mono- und Multiporphyrine mit maßgeschneiderten chemischen, physikalischen und chiroptischen Eigenschaften sowie unter-schiedlicher räumlicher Anordnung der Chromophore. Desweiteren sollten erstmals verschiedene Konzepte zur stereoselektiven Synthese axial-chiraler Porphyrin-Systeme entwickelt und vergleichend erprobt werden. Bei allen bearbeiteten Fragestellungen standen ein tieferes Verständnis stereochemischer Aspekte sowie die eingehende Untersuchung der chiroptischen Eigenschaften (z.B. unter Anwendung moderner HPLC-Kopplungstechniken) der neuartigen synthetisierten Verbindungen im Vordergrund.
In conclusion, the present thesis demonstrates that the highly desired J-type aggregation of functional perylene bisimide chromophores can be achieved by proper design of monomeric building blocks that direct self-assemble by mutual effects of hydrogen bonding and pi-pi interaction, and on the other hand, are prevented to assemble in columnar stacks owing to their twisted pi-conjugated core and sterically demanding substituents. Furthermore, the self-assembly studies gave new insights into the dynamic aggregation process of low-dimensional extended assemblies with strongly excitonically coupled chromophores. The relationship between commonly known cyanine dye aggregates like that of THIATS and that of the present PBI 1a was investigated by absorption and fluorescence spectroscopy at low temperatures down to 5 K. The formerly unprecedented functional properties of PBI aggregates that are expressed in J-type excitonic coupling hold promise for application in optoelectronic and photovoltaic devices.
This thesis is divided into three parts with the main goal allocating novel antimicrobial compounds that could be used as future antibiotics. The first part aimed to evaluate the potential of plant suspension cultures for the production of antimicrobial proteins. The extracellular, intracellular and cell wall bound fractions of seven heterotrophic and photomixotrophic plant cell suspension cultures treated with nine different elicitors were tested for the elicitor dependent production of antimicrobial proteins. Bioactivities were tested against a selected panel of human isolates including Gram-positive and Gram-negative bacteria as well as fungi using the disc diffusion assay. The intracellular fractions of elicited cell cultures were more active than extracellular fractions while the cell wall bound fractions showed lowest activities. Among the 21 fractions tested, the intracellular fraction of Lavendula angustifolia elicited with DC3000 was most active against Candida maltosa. The second most active fraction was the intracellular fraction of Arabidopsis thaliana elicited with salicylic acid which was moreover active against all test strains. The antimicrobial activity of elicited Arabidopsis thaliana cell cultures was tested by bioautography to locate the antimicrobial proteins in the crude extract. The intracellular fraction of photomixotrophic Arabidopsis thaliana cells elicited with salicylic acid was selected for further gel filtration chromatography on S-200 column leading to the purification of one 19 kDa antimicrobially active protein, designated, AtAMP. Our findings suggest that elicited plant cell cultures may present a new promising alternative source of antimicrobial proteins. The second part comprises the isolation of actinomycetes associated with marine sponges and testing the bioactivities of new species for further investigations. Actinobacterial communities of eleven taxonomically different sponges that had been collected from offshore Ras Mohamed (Egypt) and from Rovinj (Croatia) were investigated by a culture-based approach using different standard media for isolation of actinomycetes and media enriched with aqueous sponge extract to target rare and new actinomycete species. Phylogenetic characterization of 52 representative isolates out of 90 based on almost complete sequences of genes encoding 16S rRNA supported their assignment to 18 different actinomycete genera. Altogether 14 putatively new species were identified based on sequence similarity values below 98.2% to other strains in the NCBI database. The use of M1 agar amended with aqueous sponge extract yielded a putative new genus related to Rubrobacter which highlighting the need for innovative cultivation protocols. Biological activity testing showed that five isolates were active against Gram-positives only, one isolate was active against Candida albicans only and one isolate showed activity against both groups of pathogens. Moreover, the antiparasistic activity was documented for four isolates. These results showed a high diversity of actinomycetes associated with marine sponges as well as highlighted their potential to produce anti-infective agents. The third part of the thesis focused on the isolation and structure elucidation of new bioactive compounds. Streptomyces strain RV15 recovered from sponge Dysidea tupha, was selected for further chemical analysis by virtue of the fact that it exhibited the greatest antimicrobial potential against Staphylococcus aureus as well as Candida albicans among the all tested strains. Moreover, members of the genus Streptomyces are well known as prolific producers of interesting pharmacologically active metabolites. Chemical analysis of the methanolic crude extract using different chromatographic tools yielded four new compounds. The structures of the new compounds were spectroscopically elucidated to be four new cyclic peptides, namely, cyclodysidins A-D. Their bioactivity was tested against different proteases, bacteria and Candida as well as tumor cell lines. The compounds did not show any significant activities at this point.
Der bekannteste Vertreter der Bispidine ist das Lupinenalkaloid (–)-Spartein, das vor allem in enantioselektiven Deprotonierungen aber auch Übergangsmetall-katalysierten asymmetrischen Reaktionen als Ligand der Wahl eingesetzt wird. Daneben gibt es nur wenige weitere synthetische Vertreter, da keine flexiblen Darstellungsmethoden zu enantiomerenreinen Bispidinen mit variablen Substituenten in 2-endo-Position existieren. Ein zielgerichtetes Design solcher Verbindungen war daher bisher nur eingeschränkt möglich. So sollte in dieser Arbeit eine neue Substanzklasse als chirale Liganden in der asymmetrischen Synthese etabliert werden: 2-endo-substituierte 9-Oxabispidine. Das Brücken-Sauerstoffatom sollte die Synthese stark vereinfachen. Insgesamt wurden drei strategisch unterschiedliche Methoden zur enantioselektiven Synthese von 2-endo-substituierten 9-Oxabispidinen entwickelt. Zunächst ist die sehr kurze Route zu 2-endo-Phenyl-substituierten Derivaten mit diversen Resten R' an den Stickstoff-Funktionen zu nennen. Ausgehend von käuflichem (R,R)-Phenylglycidol wurde (S,S)-3-Benzylamino-3-phenyl-1,2-propandiol dargestellt, das in einer Dreistufen-Eintopf-Reaktion mit (S)-Epichlorhydrin kondensiert und zum all-cis-konfigurierten 2,6-Dimesyloxymethyl-3-phenylmorpholin mesyliert wurde. Die finale Cylisierung erfolgte mit primären Aminen zu verschiedenen 2-endo-Phenyl-9-oxabispidinen in insgesamt drei bis fünf Stufen. Die Darstellung des tricyclischen 9-Oxa-Derivats eines bekannten (+)-Spartein-Ersatzstoffs gelang nach einem verwandten Syntheseprotokoll. Für eine effiziente Variation des 2-endo-Substituenten auf einer späten Synthesestufe wurde zunächst enantiomerenreines 3N-Boc-7N-Methyl-9-oxabispidin-2-on als Schlüsselintermediat ausgewählt, das aus (R)-Epichlorhydrin und racemischem Glycidsäuremethylester dargestellt wurde. Die weitere Überführung in die 9-Oxabispidine wurde durch Grignard-Addition, Abspaltung der N-Boc-Gruppe vom resultierenden, ringoffenen N-Boc-Aminoketon, Cyclisierung zum Imin und abschließende, exo-selektive Reduktion oder Hydrierung erreicht. So wurden bi- und tricyclische 9-Oxabispidine in nur drei Stufen und 51% Ausbeute synthetisiert. Ein größeres Potenzial besitzt jedoch der primär von David Hein parallel zu den eigenen Arbeiten entwickelte Zugang über ein cis-konfiguriertes 6-(N-Boc-Aminomethyl)morpholin-2-carbonitril als zentrale Zwischenstufe, das auch im 10-g-Maßstab problemlos erhältlich war. Die allgemeine Anwendbarkeit und Flexibilität dieser Methode wurde anhand der Darstellung einer Reihe an 9-Oxabispidinen demonstriert. Die dargestellten chiralen 9-Oxabispidine wurden erstmalig in den folgenden fünf unterschiedlichen Gebieten der asymmetrischen Synthese getestet: Organolithium- und Organomagnesium-vermittelte Umsetzungen sowie Pd(II)-, Cu(II) und Zink(II)-katalysierte Reaktionen. Für enantioselektive Deprotonierungen schwach C-H-acider Verbindungen mit sBuLi erwiesen sich die 9-Oxabispidine als ungeeignet, da sie selbst in Brückenkopfposition lithiiert wurden. Die Stabilität der resultierenden -Lithioether war unerwartet hoch; sie ließen sich beispielsweise bei -78 °C in guten Ausbeuten mit Elektrophilen abfangen. Umlagerungen traten erst beim Aufwärmen ein, wenn kein Elektrophil als Reaktionspartner zur Verfügung stand. Als definierte Produkte wurden dabei Ring-kontrahierte N,O-Acetale erhalten. Zusammen mit den weniger basischen Grignard-Reagenzien wurden die 9-Oxabispidine erfolgreich zur Desymmetrisierung von meso-Anhydriden verwendet. Bei Pd(II)-katalysierten oxidativen kinetischen Racematspaltungen sekundärer Alkohole konnten mit einem 9-Oxabispidin-Pd(II)-Katalysator Selektivitätsfaktoren s vergleichbar zu denen mit (–)-Spartein erzielt werden. Die Ursache für die geringere Reaktivität der 9-Oxabispidin-Komplexe liegt gemäß quantenchemischen Berechnungen in der Elektronegativität des Brücken-Sauerstoffatoms, was die Elektronendichte am Palladiumatom verringert. In Kooperation mit David Hein wurde ein von einem tricyclischen 9-Oxabispidin abgeleiteter Cu-Katalysator entwickelt, der bei der Addition von Nitromethan an zahreiche aromatische, heteroaromatische und aliphatische Aldehyde exzellente Enantioselektivitäten im Bereich von 91–97% lieferte. Mit bicyclischen, 2-endo-substituierten 9-Oxabispidinen als chiralen Liganden wurden hingegen lediglich 33-57% ee erreicht bemerkenswerterweise entstanden hierbei bevorzugt die enantiokomplementären β-Nitroalkohole. In Zusammenarbeit mit Janna Börner aus der Arbeitsgruppe von S. Herres-Pawlis wurde der erste chirale, neutrale Diamin-Zink(II)-Katalysator für die Ringöffnungs-Polymerisation von D,L-Lactid entwickelt. Die Reaktion benötigte kein weiteres anionisches Additiv und konnte ohne Lösungsmittel mit nicht-aufgereinigtem, käuflichem Lactid durchgeführt werden.
Kaum ein Bereich der menschlichen Tätigkeit hat sich jemals so stürmisch entwickelt, wie die Mikro- und Nanoelektronik in den letzten Jahrzehnten. Der rasche Fortschritt dieser Gebiete war möglich, weil die Vorteile in der Anwendung der Mikroelektronik den gewaltigen Entwicklungs- und Forschungsaufwand rechtfertigten. Eine besondere Rolle spielt dabei die Herstellung von Halbleiterbauelementen durch Kristallzüchtungsmethoden. In dieser Arbeit wurden Prozesse untersucht, die sich auf der Kristalloberfläche abspielen und somit das Wachstum von hochgeordneten Kristallstrukturen bestimmen. In den vergangenen Jahren wurden mehrere Methoden zur Untersuchung dieser Prozesse entwickelt, deren Präzision sich von Jahr zu Jahr unablässig steigerte. In der Reihe der theoretischen Ansätze stehen quantenchemische Methoden im Vordergrund. Eine von diesen Methoden, die Dichtefunktionaltheorie, ist aufgrund ihrer Anschaulichkeit und des relativ niedrigen Rechenaufwands das Hauptwerkzeug der vorliegenden Arbeit. Im ersten Teil dieser Arbeit wurden die Wanderungsmöglichkeiten eines Adsorbatatoms (Cd oder Te) auf der (001) Oberfläche von CdTe (Substrat) auf DFT-Niveau im Rahmen der GGA-Näherung untersucht. Dies erforderte es, die Gesamtenergie des Systems Adsorbat-Kristall an verschiedenen Adsorptionsstellen zu berechnen. Dabei wurde nur ein Teil des Kristalls - das Adsorbat selbst und die nächste Umgebung der Adsorptionsstelle (Quantencluster) - auf DFT-Niveau berechnet. Der Einfluss des übrigen Kristalls auf den Cluster wurde mit einem Gitter aus Punktladungen angenähert, wobei die Te- und Cd-Atome die Ladungen −2 bzw. +2 trugen. Bei dem Einsatz dieses Modells ergab sich allerdings das Problem, dass es eigentlich nur auf Ionenkristalle anwendbar ist, die in guter Näherung volle Ionizität besitzen. CdTe stellt aber laut vielen experimentellen und theoretischen Untersuchungen eine Abstufung zwischen ionischen und kovalenten Kristallen dar, was eine gründliche Analyse der Abhängigkeit unserer Ergebnisse von der Clustergröße und der Entfernung der Adsorptionsstelle von den Clusterrändern erforderte. Als Ergebnis wurde ein Modell entworfen, das dazu in der Lage ist, die Struktur der (2X1) Te-terminierten CdTe Oberfläche mit ausreichender Genauigkeit wiederzugeben. Durch geeignete Wahl des Quantenclusters (ausreichende Größe in den Richtungen parallel zur Oberfläche und Platzierung der weniger polarisierbaren Cd-Kationen an den Außenflächen) gelang es, den Einfluss der Clusteroberfläche auf die untersuchten Eigenschaften auf ein akzeptables Maß zu verringern. Die durchgeführten Berechnungen der Cd-Potentialenergiefläche zeigen zwei Potentialtöpfe, mit den Tiefen 2.1 eV und 1.7 eV. Die Existenz dieser beiden Minima ist eng mit der Dimerisierung von Te-Atomen an der adsorbatfreien Te-Oberfläche verbunden. Das erste, der Struktur =Te-Cdad-Te= entsprechende Minimum entsteht durch den Bruch einer Te-Te Dimerbindung beim Cd-Angriff an diese Stelle. Der zweite Potentialtopf kommt dadurch zustande, dass das Cd-Adsorbatatom mit zwei entlang der [110]-Richtung angeordneten Te2-Dimeren reagiert. Die Potentialenergiefläche des Te-Adsorbats unterscheidet sich zwar wesentlich von der des Cd-Atoms, es gibt aber auch Ähnlichkeiten. Das gilt vor allem für das der Struktur =Te-Tead-Te= entsprechende Minimum, das ungefähr 2.8 eV tief ist. Wie im Fall der Cd-Adsorption entsteht diese Struktur infolge der Wechselwirkung eines adsorbierten Te-Atoms mit einem Te2-Dimer auf der Oberfläche. Die Ergebnisse unserer Berechnungen bestätigen experimentelle Hinweise, gemäß denen Te- und Cd-Atome aus dem Teilchenfluss, dem die (2X1)Te Oberfläche während der MBE ausgesetzt ist, leicht adsorbiert werden. Außerdem wurden die relativ genauen Werte der Potentialbarrieren bekommen, die für ein besseres Verständnis des Wachstumsprozesses zum Beispiel mit Hilfe von Monte-Carlo-Simulationen notwendig sind. Im Vordergrund des zweiten Teils der vorliegenden Arbeit stand die Strukturbestimmung von ZnO-Nanoclustern, die durch spezielle Kristallisationsprozesse erzeugt werden und wegen ihrer eigenartigen optischen und elektronischen Eigenschaften von großem Interesse sind. Zwei grundsätzlich unterschiedliche Atomanordnungen wurden betrachtet, wobei festgestellt werden sollte, welche dieser Strukturen in Abhängigkeit von der Clustergröße und der Umgebung stabiler ist. Angenommen wurde dabei, dass diese Tendenz bei der weiteren Vergrößerung der Atomanzahl von Hundert bis mehreren Tausenden erhalten bleibt. Die Clustermodelle erster Art besaßen die für ZnO-Verbindungen typische Wurtzitstruktur, die anderen, sogenannten Käfigcluster, bestanden aus Zn3O3- und (oder) Zn2O2-Ringen, die so verknüpft sind, dass sie kugel- oder zylinderförmige Strukturen bilden. Charakteristisch für letztere Cluster ist eine Homogenität der Atomumgebung, da alle Zn- und O-Atome dreifach koordiniert sind, während sie in Wurtzitstrukturen im Wesentlichen vierfach koordiniert sind. Durch Knüpfung zusätzlicher Zn-O Bindungen konnte die Anzahl der in Frage kommenden Strukturen nennenswert vergrößert werden. Dabei entstehen vierfach koordinierten Atome und, laut den Berechnungen, deutlich stabilere Cluster. Die Rechnungen wurden sowohl im Vakuum als auch im Rahmen des COSMO Verfahrens (im „Wasser“) durchgeführt. Sie ergaben, dass die Wurtzitstrukturen bei der Zunahme der Atomanzahl stabiler werden als ihre Käfig-Analoge. Dieses Ergebnis ist allerdings eher von theoretischem Interesse, da die experimentell in einer Lösung gezüchteten ZnO-Nanocluster an ihrer Oberfläche mit Molekülen aus der Lösung bedeckt sind. Ein weiterer Schritt war daher, den Einfluss der Umgebung auf die Bildungsenergie durch die Absättigung der Oberfläche mit H+- und OH−-Ionen zu simulieren. Als Bezugspunkt für die Berechnung der Bildungsenergie der verschiedenen Cluster wurde der Molekülkomplex Zn(OH)2(H2O)2 verwendet. Mit anderen Worten wurde angenommen, dass ein freies Zn2+-Ion in der Lösung von zwei OH−-Gruppen und zwei H2O-Molekülen umgeben ist. Die Ergebnisse zeigen, dass die Absättigung einen starken Einfluss auf die Randbereiche der wurtzitartigen Cluster ausübt. Bei fast allen Clustermodellen sind diese stark verformt, während bei den Käfigstrukturen nur deutlich geringere Verzerrungen beobachtet werden. Ebenso stark ist der Einfluss auf die Bildungsenergie: Verglichen mit ihren unabgesättigten Analogen werden alle abgesättigte Strukturen erheblich stabiler, was auf die Tatsache zurückzuführen ist, dass durch die OH− -Gruppen und H+-Kationen die freien Valenzen an der Clusteroberfläche abgesättigt werden. Ansonsten lassen sich bei den abgesättigten Strukturen dieselben Tendenzen erkennen, wie bei nicht abgesättigten. So werden Wurtzitstrukturen mit zunehmender Clustergröße energetisch günstiger als Käfigstrukturen mit der gleichen Anzahl an Atomen. Da es die im Rahmen dieser Arbeit festgestellten Regelmäßigkeiten ermöglichen, die stabilsten ZnO-Atomanordnungen auf die hier Betrachteten einzuschränken, ergibt sich, dass die stabilste Struktur für Nanocluster wurtzitartig ist. Dies stimmt auch mit allen verfügbaren experimentellen Daten überein.
Perylene bisimide (PBI) dyes are a widely used class of industrial pigments, and currently have gained significant importance for organic-based electronic and optical devices. Structural modification at the PBI core results in changes of the optical and electronic properties, which enable tailored functions. Moreover, the aggregation behavior of PBIs is alterable and controllable to achieve new materials, among which organogels are of particular interest because of their potential for applications as supramolecular soft materials. In this work, new PBI-based organic gelators were designed, synthesized, and characterized, and the aggregation behaviors under different conditions were intensively studied by various spectroscopic and microscopic methods. In chapter 2, a brief overview is given on the structural and functional features of organogel systems. The definition, formation and reversibility of organogels are introduced. Some examples on dye based organogel are selected, among which PBI-based organogelators reported so far are especially emphasized. Some basic knowledges of supramolecular chirality are also overviewed such as characterization, amplification, and symmetry breaking of the chiral aggregates. According to our former experiences, PBIs tend to form aggregates because the planer aromatic cores interact with one another by pi-pi interaction. In chapter 3, a new PBI molecule is introduced which possesses amide groups between the conjugated core and periphery alkyl chains. It is found that well oriented aggregates are formed by hydrogen bonding and the pi-pi interaction of the cores. These interactions enable the aggregates to grow in one-dimension forming very long fibers, and these fibers further intercross to 3D network structures, e.g., organogels. In comparison to the very few PBI-based gelators reported before, one advantage of this gelator is that, it is more versatile and can gelate a wide range of organic solvents. Moreover, the well-organized fibers that are composed of extended π-stacks provide efficient pathways for n-type charge carriers. Interestingly, AFM studies reveal that the PBI molecules form well-defined helical fibers in toluene. Both left-handed (M) and right-handed (P) helicities can be observed without any preference for one handedness because the building block is intrinsically achiral. In chapter 4, we tried to influence the M/P enantiomeric ratio by applying external forces. For example, we utilized chiral solvents to generate chiral aggregates with a preferential handedness. AFM analysis of the helices showed that a enantiomeric ratio of about 60: 40 can be achieved by aggregation in chiral solvents R- or S-limonene. Moreover, the long aggregated fibres can align at macroscopic level in vortex flows upon rotary stirring In chapter 5, bulky tetra-phenoxy groups are introduced in the bay area of the PBI gelator. The conjugated core of the new molecule is now distorted because of the steric hindrance. UV/Vis studies reveal a J-type aggregation in apolar solvents like MCH due to intermolecular pi-pi-stacking and hydrogen-bonding interactions. Microscopic studies reveal formation of columnar aggregates in apolar solvent MCH, thus this molecule lacks the ability to form gels in this solvent, but form highly fluorescent lyotropic mesophases at higher concentration. On the other hand, in polar solvents like acetone and dioxane, participation of the solvent molecules in hydrogen bonding significantly reduced the aggregation propensity but enforced the gel formation. The outstanding fluorescence properties of the dye in both J-aggregated viscous lyotropic mesophases and bulk gel phases suggest very promising applications in photonics, photovoltaics, security printing, or as fluorescent sensors. In chapter 6, we did some studies on combining PBI molecules with inorganic gold nanorods. Gold nanorods were synthesized photochemically. By virtue of the thioacetate functionalized PBIs, the rods were connected end to end to form gold nanochains, which were characterized by absorption spectra and TEM measurement. Such chromophore-nanorod hybrids might be applied to guide electromagnetic radiation based on optical antenna technology.
In the first part of this work we presented the synthesis and photophysical properties of a series of transition metal donor-acceptor Ir(III)complexes of the type [(C^N)2Ir(N^N)][PF6]. The Ir(III) was connected with hole conducting donor-moieties like carbazole (CZ) and triarylamine (TAA) linked via a methylene and ethylene bridge to the cyclometalating C^N ligands phenylpyrazole (ppz) and phenylpyridine (ppy). Bidentate N^N and P^P ligands like 2,2’-bipyridyl (bpy), 3,4,7,8-tetramethyl-1,10-phenanthroline (tmp) and cis-1,2-bis(diphenylphosphino)ethylene (bdppe) were used as acceptor units. In order to analyse the influence of the electron density of the bpy ligand, TAA-complexes with acceptor- and donor-substituted bpy acceptor units were synthesised. Therefore, 4,4’-dinitro-2,2’-bipyridyl, 4,4’-dichloro-2,2’-bipyridyl, 4,4’-dimethoxy-2,2’-bipyridyl and 4,4’-dimethylamino-2,2’bipyridyl were used as neutral N^N ligands. In order to compare the photophysical properties, all reference compounds without hole conducting component were syntesised. All the carbazole compounds, except the bdppe complexes, exhibit emission and transient absorption properties similar to their reference compounds that make them interesting for OLED (organic light emitting device) applications. LEC (light emitting electrochemical cell) studies show a red shifted luminescence. The triarylamine compounds do not luminesce at RT but they exhibit an intense, blue-shifted and long-lived luminescence at 77 K in a rigid matrix. The transient absorption spectra differ strongly from that of their reference compounds. The spectra display characteristic features of the spectra of the isolated radical anions and cations supported by spectroelectrochemical measurements. Thus, it can be assumed that the transient states are charge separated (CS) states in which the positive charge is localised at the TAA donor units and the negative charge at the N^N acceptor units. The decays of the transient states are biexponentially what indicates the presence of two transient states, the 1CS and the 3CS state. To understand this behaviour the differently substituted bipyridyl-complexes were synthesised and analysed. Temperature dependent transient absorption measurements showed that all rate constants are indepentend of the temperature, except for the complex with OMe subsituents at the bpy ligand. The equilibrium constant K = k1 / k2 is nearly one for all complexes. For the OMe-compound it decreases with increasing temperature. Plotting the rate constants vs. the free energy differences (determined by cyclovoltammetry measurements) shows that all constants are decreasing with increasing donor strength of the bpy ligand. DFT calculations on the OMe-compound are already in work. In the second part of this work, neutral Ir(III) and Pt(II) complexes of the type [(O^O)Ir(N^N)2] and [(O^O)Pt(N^N)] were introduced. There, TTA was connected directly or via a CH2 bridge to acectylacetonate (acac = O^O) in order to probe the influence of the different kinds of connection on the photophysics of the complexes. As the bidentate N^N ligand 2,2’-bipyridyl (bpy) was chosen. All the corresponding reference compounds without triarylamine were obtained in order to compare with the TAA substituted analoga. Furthermore, the homoleptic fac Ir(N^N)3 complex with triarylamine connected via a methylene and ethylene bridge to phenylpyrazole as introduced in the first part of this work was synthesised. The synthesis of the Ir(III) compound with the TAA substituted acac ligand connected via the CH2 group was not successful. All the neutral triarylamine-substituted -diketonato Pt(II) and Ir(III) complexes do not luminesce at RT, except the Pt(II)-complex with CH2 bridge. This compound shows transient state characteristics that are in good agreement with the luminescence lifetimes at RT and that are similar to the reference compound, what suggests to a 3Pt(N^N)(O^O) state. The complexes without the CH2 bridging unit show no transient signals what may be caused by charge-transfer quenching due to the direct linkage between donor and acceptor unit. The homoleptic fac Ir(N^N)3 complex exhibits no emission at RT and no transient signals. At 77 K it shows a highly structured emission with 14 s lifetime. Compared to the literature-known reference compound this emission is caused by the population of a 3Ir(ppz)3 state. Our findings are important for designing complexes with stronger acceptor units (i.e. naphthaleneimide) for long CS states lifetimes to be used as photosynthesisers in solar cells and other optoelectronic devices. Besides, LEC and OLED studies on the carbazole complexes are still of interest to analyse the degree of triplet-triplet-annihiliation in these devices.
Im Rahmen dieser Arbeit wurde ein synthetischer Membrantransporter für acetylierte Aminosäurecarboxylate entworfen und hergestellt. Als Bindungsstelle für die Carboxylate wurde das Guanidiniocarbonylpyrrol-Motiv von Schmuck verwendet. In den Seitenarm des Pyrrols wurde ein L-Valinamid-Rest eingebracht, um die Möglichkeit zu zusätzlichen Wasserstoffbrückenbindungen zu bieten und gegebenenfalls Substrat- und Enantioselektivität zu erreichen. Zur Herstellung der Löslichkeit in unpolaren Medien wie dem Inneren der Zellmembran musste eine lipophile Gruppe eingebracht werden. Als löslichkeitsvermittelnder Rest wurde Tris-(Dodecyloxy)phenylmethylen ausgewählt, das drei lange unpolare Alkylreste trägt. Zusammengenommen ergab sich so ein Rezeptor für Oxo-Anionen und speziell für Aminosäurecarboxylate mit erhöhter Löslichkeit in organischen Medien. Somit war die Fähigkeit zu Membrantransport gegeben. In Kraftfeldrechnungen erhielt man die vermutliche Struktur des Rezeptor-Substrat-Komplexes, der eine Kombination aus einer Salzbrücke, Wasserstoffbrückenbindungen und einer Stapelwechselwirkung von Guanidinum-Kation, Benzylgruppe und ggf. aromatischem Rest des Aminosäuresubstrates aufweist. Nach erfolgreicher Synthese wurde in Extraktionsexperimenten die Fähigkeit des Rezeptors erprobt, Aminosäurecarboxylate aus einer wässrigen in eine organische Phase aus zu überführen. Man erhielt das beste Extraktionsvermögen für Ac-Trp-OH, gefolgt von Ac Phe OH und Ac Tyr OH. Es wurde eine neue Formel aufgestellt, mit der aus den pKS-Werten der Substrate und den Extraktionsdaten mit und ohne Rezeptor die Bindungskonstanten der Rezeptor-Substrat-Komplexe berechnet werden konnten. Die Größe der Bindungkonstanten entsprach der Reihenfolge Trp > Tyr > Phe ~ Val mit den höchsten Bindungskonstanten für das Tryptophanderivat mit 1.5*10E4 1/M. Zur Bestätigung der Bindungskonstanten wurden ITC-Messungen durchgeführt. Es wurden Messungen des Rezeptors in Chloroform mit den tert-Butylammoniumsalzen der acetylierten Aminosäuren Phenylalanin, Tyrosin und Valin durchgeführt. Für die Werte von Enthalpie und Entropie konnten bei dieser Auswertung konsistente Werte ermittelt werden. Die höchsten Werte der Enthalpie erhielt man für das Tyrosinderivat mit 3.7*10E3 cal/mol, gefolgt vom Phenylalaninderivat mit 2.8*10E3 cal/mol und Valinderivat mit 1.3*10E3 cal/mol. Diese Abstufung entspricht dem Einfluss des aromatischen Restes, der durch die Stapelwechselwirkung mit dem Guanidinium-Kation die Bindungswärme erhöht und durch den damit verbundenen engeren Komplex den Wert für die Entropie senkt. Für die Evaluierung des Transportvermögens wurden U-Rohr-Versuche verschiedener Art durchgeführt. Es wurde ein Gradient von pH 6 in der Ausgangsphase auf pH 8 in der Zielphase eingesetzt, wodurch der Rezeptor an der Grenzfläche zur Zielphase deprotoniert wurde, was zu gerichtetetem Transport führte. Es ergaben sich recht starke Unterschiede für die Fluxwerte der einzelnen Substraten, die der Reihenfolge Val > Phe > Ala > Trp > Tyr folgten. Dabei wurde das Valinderivat um den Faktor 17 schneller als das Tyrosinderivat befördert, mit dem recht hohen Flux von 1.11*10E-6 mol/m2*s, was nahe an den höchsten literaturbekannten Wert für acetylierte Aminosäuren heranreicht. Durch Verwendung gleicher Substratkonzentrationen in Start- und Zielphase konnte aktiver Transport nachgewiesen werden, d.h. Transport gegen das Konzentrationsgefälle. Die Triebkraft des Transportes war der Gradient von pH 6 auf pH 8 zwischen Ausgangs- und Zielphase, der durch den Symport von Substrat und einem Proton ausgeglichen wurde. Bei einem kompetitiven Versuch mit einer Mischung der verschiedenen Substrate in der Ausgangsphase wurden veränderte Fluxwerte und Selektivitäten festgestellt. Die neue Reihenfolge der Transportgeschwindigkeit war nun Trp > Phe > Val > Tyr > Ala, wobei die Fluxwerte fast durchgehend niedriger waren als im Einzelversuch. Die Veränderung der Werte erschließt sich bei Vergleich mit den thermodynamischen Daten aus den Extraktionsexperimenten. Bei direkter Konkurrenz um den Rezeptor wurden diejenigen Substrate mit den höchsten Bindungskonstanten bevorzugt, unabhängig von ihrer Transportgeschwindigkeit. Die schwächer bindenden Substrate wurden aus dem Komplex verdrängt und wiesen deswegen niedrigere Transportwerte auf. Der kompetitive Versuch ist somit eine stärkere Abbildung der Bindungsstärke und entspricht eher der Situation in einer realen Zelle.
The main focus of this thesis was the synthesis and analysis of multifunctional oligopeptides. The study of their non-covalent interactions with various counterparts revealed interesting new results, leading to both methodological and application related progress. The first project of this thesis concentrated on the in-depth analysis of the peptide receptor CBS-Lys-Lys-Phe-NH2 to acquire a better understanding of its binding mode upon complexation with a substrate. In this context it was possible to develop—in cooperation with the group of Prof. Sebastian Schlücker—a direct and label free spectroscopic detection of immobilized compounds which are often found in combinatorial libraries. This new screening method utilizes the advantages of the surface enhanced Raman spectroscopy and allowed for the first time a surface mapping of a single polystyrene bead for the identification of peptides in femtomolar concentrations. Hence, this method allows a very fast and sensitive detection of resin bound compounds. The development of this promising new approach set the starting point for future experiments to enable on-bead library screenings and to investigate the complex formation of immobilized compounds. After the comprehensive analysis of the basic structural features of small peptide receptors in the first part of this thesis, the second big block focused on its in vitro evaluation using biological relevant targets. Therefore, several different modifications of the initial peptide structures were synthesized. These modifications provided a molecular toolkit for the tailor made synthesis of structures individually designed for the respective target. The first tests addressed the interaction with Alzheimer’s related amyloid fibrils. During these experiments, the successful SPPS syntheses of tri- and tetravalent systems were achieved. The comparison of the multivalent form with the corresponding monovalent version was then under special investigations. These concentrated mainly on the interaction with various bacteria strains, as well as with different parasites. To localize the compounds within the organisms, the synthesis of fluorescence labelled versions was achieved. In addition, several compounds were tested by the Institute for Molecular Infection Biology of the University of Würzburg for their antibacterial activity. This thorough evaluation of the biological activity generated precious information about the influence of small structural changes in the peptide receptors. Especially the distinct influence of the multivalency effect and the acquired synthetic skills led to the development of an advanced non-covalent recognition event, as described in the final project of this thesis. The last part of this thesis discussed the development of a novel inhibitor for the serine protease beta-tryptase based on a tailor-made surface recognition event. It was possible to study and analyze the complex interaction with the unique structure of tryptase, that features a tetrameric frame and four catalytic cleavage sites buried deep inside of the hollow structure. However, the point of attack were not the four binding pockets, as mostly described in the literature, but rather the acidic areas around the cleavage sites and at the two circular openings. These should attract peptides with basic residues, which then can block the accessibility to the active sites. A combinatorial library of 216 tetravalent peptide compounds was synthesized to find the best structural composition for the non-covalent inhibition of beta-tryptase. For the screening of the library a new on-bead assay was applied. With this method a simultaneous readout of the total inhibition of all library members was possible, thus allowing a fast and direct investigation of the still resin bound inhibitors. Several additional experiments in solution unveiled the kinetics of the inhibition process. In conclusion, both mono- and multivalent inhibitors interact in a non-destructive and reversible way with the tryptase.
This thesis deals with the synthesis of improved organic semiconductors, the detailed investigation of the molecular properties and the solid state arrangements revealed by single crystal X-ray diffraction and finally the development of structureperformance dependencies by measuring of the charge carrier mobilities of the derivatives in thin film transistors. The two main-goals of this thesis were achieved. Well soluble acene derivatives for spin-coated TFTs were obtained, showing charge carrier mobilities in the range of polymer p-type materials. Novel core-fluorinated perylene bisimide dyes were synthesized particularly and the use of electron deficient substituents lead to PBIs with outstanding air-stable mobilities in thin film transistors prepared by vacuum deposition techniques. The relationship between performance, air stability and solid state packing was elucidated in detail by single crystal X-ray diffraction analysis.
Ziel der Dissertation „Entwicklung von Cysteinproteaseinhibitoren – ein klassischer und ein kombinatorischer Ansatz zur Inhibitoroptimierung“ war die Optimierung von neuen Inhibitoren von Falcipain-2 und Rhodesain als neue potentielle Wirkstoffe gegen Malaria bzw. die Schlafkrankheit über zwei verschiedene Methoden. Es handelt sich hierbei um einen klassischen und einen kombinatorischen Ansatz. Der klassische Ansatz basiert auf einer Struktur, deren Aktivität per Zufall entdeckt wurde. In Screenings von synthetisierten Strukturanaloga, gestützt durch virtuelle Docking-Experimente am aktiven Zentrum der Cysteinproteasen, wurden Struktur-Aktivitäts-Beziehungen erarbeitet. Bei der kombinatorischen Methode wurde zunächst ein peptidischer Inhibitor entworfen, der durch Festphasensynthese an einem geeigneten Harz synthetisiert wurde. Durch den kombinatorischen Einsatz von Aminosäuren konnte auf diese Weise unter enormer Zeitersparnis eine Bibliothek von 150 Inhibitoren synthetisiert werden. In einem Screening dieser harzgebundenen Inhibitoren wurden anschließend die potentesten Inhibitoren identifiziert. Die Aktivität der gefundenen Inhibitoren aus beiden Ansätzen an den protozoischen Erregern wurde durch in-vitro-Experimente an Plasmodien und Trypanosomen untersucht. Beim klassischen Ansatz wurde eine neue Substanzklasse entwickelt, die sehr gute Hemmeigenschaften an beiden Cysteinproteasen mit IC50-Werten im niedrigen mikromolaren Bereich zeigten. Außerdem besaßen sie eine hohe in-vitro-Aktivität gegenüber den Erregern im gleichen Konzentrationsbereich. Einige der Inhibitoren zeigten keine Zytotoxizität an Makrophagen. Aus dem klassischen Ansatz konnten also hochaktive Substanzen mit geringer Zytotoxizität entwickelt werden, deren Einsatz als Wirkstoffe gegen Malaria oder der Schlafkrankheit denkbar ist. Für den kombinatorischen Ansatz wurde zur Inhibitoroptimierung eine Screeningmethode für Falcipain-2 und Rhodesain direkt an einem geeigneten Harz zur Festphasensynthese entwickelt. Neu bei dieser Screeningmethode war es, dass erstmals ein quantitatives Screening einer Inhibitorbibliothek möglich sein sollte, und nicht nur die besten Inhibitoren identifiziert werden können. Aus den Ergebnissen der Festphasenscreenings an beiden Proteasen wurden 14 besonders interessante Inhibitoren der Bibliothek ausgewählt und synthetisiert. Diese Verbindungsklasse zeigte ebenfalls sehr gute Ergebnisse an den isolierten Enzymen, in den mikrobiologischen Tests an den Erregern jedoch fielen alle Ergebnisse vergleichsweise schlechter aus. Die schlechte Löslichkeit, die Bioverfügbarkeit und der Metabolismus durch den Erreger der peptidischen Inhibitoren schienen von großer Bedeutung zu sein. Der bearbeitete kombinatorische Ansatz lieferte eine neuartige Screeningmethode, die auch auf andere Targets anwendbar ist.
Ziel der Dissertation „Oktapeptide als neue Organokatalysatoren zur Hydrolyse von Phosphaten und Estern“ war es neue Oktapeptide zu finden, die die Fähigkeit besitzen Phosphate und Ester zu Hydrolysieren. In der Natur ist bei den Katalysereaktionen die Sekundärstruktur von entscheidender Bedeutung. Aus diesem Grunde wurde im Rahmen dieser Arbeit zunächst ein Modellsystem entwickelt, mit dem gezeigt werden konnte, dass es möglich ist mit einfachen Bausteinen wie Diaminobutan und dem in der Gruppe von Schmuck entwickelten Guanidiniocarbonylpyrrol ein Molekül zu entwickeln, welches eine stabile intramolekulare Schleife in polaren Lösungsmitteln ausbildet. Aufgrund der geringen Größe des Moleküls, konnte kein β-Faltblatt gebildet werden. Dennoch konnte mit Hilfe der NMR-Spektroskopie gezeigt werden, dass in dem polaren Lösungsmittel Methanol eine stabile Schleifenbildung mit einer intramolekularen Komplexierung stattfindet. Nach erfolgreicher Synthese des oben beschriebenen Testsystems wurde als nächster Schritt eine kombinatorische Organokatalysatorbibliothek mit 625 Mitgliedern aufgebaut. Die Struktur der Peptide kann man in drei Teile untergliedern. Der erste Teil ist die feste Phase, das Amino-TentaGel, an das nacheinander die einzelnen Aminosäuren gekuppelt wurden. An Stelle der Butylgruppe als Schleifenelement wurde Aib-D-Pro als β-Turn Element eingesetzt. Den dritten Teil bilden die bei der Synthese der Oktapeptide eingesetzten Aminosäuren AA1, AA3, AA6, AA8, die ein β-Faltblatt ausbilden sollten. Die kombinatorische Synthese der Bibliothek erfolgte nach der „Split and Mix“ Methode. Zum Unterscheiden der einzelnen Mitglieder untereinander, wurde die feste Phase zusammen mit einem Radiofrequenzchip in IRORI MikroKans gegeben. Durch die unterschiedlichen Aminosäuren ist die Bibliothek für die Katalyse in polaren Lösungsmitteln wie Wasser konzipiert worden. Als exemplarische Katalysereaktionen wurden dabei zwei Hydrolysereaktionen (Phosphatspaltung und Esterspaltung) ausgesucht. Zunächst wurde für beide Reaktionen ein Screening mit unterschiedlichen Bedingungen durchgeführt. Dabei hat sich gezeigt, dass bei der Phosphatspaltung nur dann die Reaktion katalysiert wurde, wenn das künstliche Argininanalogon, welches in unserer Arbeitsgruppe synthetisiert wurde, vorhanden war. Der beste Katalysator hat die Reaktion 175-mal schneller katalysiert als die unkatalysierte Reaktion. Bei dem Screening der Esterspaltung hat sich herausgestellt, dass die Aminosäure Histidin essentiell für die katalytische Aktivität ist. Der beste Katalysator bei der Esterspaltung hat die Hydrolyse des Esters 345-mal schneller katalysiert als die unkatalysierte Reaktion. Bei beiden Reaktionen hat sich gezeigt, dass die Sequenz der Katalysatoren sehr wichtig für die Katalyse ist. So verringert z.B. bei der Esterhydrolyse der Austausch zweier Aminosäuren eine Verringerung der Aktivität von dem Faktor 294 auf den Beschleunigungsfaktor 35. Auch konnten beide Katalysereaktionen in wässrigem gepufferten Lösung durchgeführt werden. Damit ist es möglich gewesen neue Oktapeptide für die Katalyse von Phosphat- und Esterspaltung zu finden und diese erfolgreich im Screening als auch in Lösung zu untersuchen.
Ziel der Dissertation „Neue zwitterionische Halbschalen als Bausteine für supramolekulare Kapseln“ war die Verknüpfung zweier Guanidiniocarbonylpyrrolcarboxylat-Bindungsmotive von Schmuck über starre, sowohl aromatische als auch nichtaromatische Linker. Die so erhaltenen zwitterionische Halbschalen sollten in Lösung zu supramolekulare Kapseln aggregieren, welche einen Hohlraum ausweisen, in den Gastmoleküle eingelagert werden können. Dieses Bindungsmotiv ist selbstkomplementär und daher in der Lage Homodimere auszubilden. Durch die Kombination aus Wasserstoffbrücken und Ionenbindungen sind diese selbst in polaren Lösemitteln wie DMSO oder Wasser stabil, im Gegensatz zu Systemen, welche z.B. nur über Wasserstoffbrücken verfügen und in polaren Medien wieder dissoziieren. Zur Synthese wurden zwei Bindungsmotive mittels Tetrahydroxybenzol verbrückt. Die eindeutige Charakterisierung erfolgte über NMR-Spektroskopie, Massen-Spektrometrie und Röntgenstrukturanalyse. Anschließend wurde die Verbindung in die zwitterionische Form überführt und auf Kapselbildung hin untersucht (NMR, DOSY, Masse, Molecular Modelling). Die theoretischen Berechnungen wiesen darauf hin, dass die synthetisierten Halbschalen in der Lage sein sollten, Kapseln zu bilden. Trotz der erfolgreichen Synthese dieses neuartigen zwitterionischen Makrozyklus steht der experimentelle Nachweise auf Grund der schlechten Löslichkeit der Zwitterionen in allen verwendeten Lösemitteln noch aus. Auch wurde Glucoluril als nichtaromatisches Linkermolekül erfolgreich verwendet. Als erstes wurde das 4,4’-Diphenylglucoluril erfolgreich in der Kupplung eingesetzt. Es war möglich, die so erhaltenen cis/trans-Makrozyklen säulenchromatographisch zu isolieren und mittels Röntgenstrukturanalyse zu charakterisieren. Nach Überführung in die Zwitterionen wurden diese wiederum auf die Kapselbildung hin untersucht (NMR, DOSY, Masse, Molecular Modelling). Berechnungen zufolge sollte die Kapselbildung möglich sein, jedoch steht auch hier trotz erfolgreicher Synthese der experimentelle Nachweis auf Grund der Unlöslichkeit noch aus. Zur Verbesserung der Löslichkeit wurden zwei neue Glucolurilderivate entwickelt, welche am Phenylring mit Octyl- bzw. Triethylenglykolketten substituiert waren. Dadurch sollte die Löslichkeit der Zwitterionen in organischen bzw. wässrigen Lösungen erhöht werden. Jedoch zeigte die Einführung dieser Ketten keine wesentliche Verbesserung der Löslichkeit und somit konnte auch bei diesen neuen zwitterionischen Halbschalen keine Kapselbildung nachgewiesen werden. Im Rahmen dieser Dissertation wurden sieben neue zwitterionische makrozyklische Halbschalen synthetisiert und die daraus gewonnenen Erkenntnisse können als Ausgangspunkt verwendet werden, die Löslichkeit weiter zu verbessern.
The present thesis demonstrates the potential of dipolar aggregation of merocyanine dyes as novel directional and specific supramolecular binding motif for the creation of more elaborate supramolecular architectures beyond simple dimers. Furthermore, the self-assembly studies into bis(merocyanine) nanorods gave new insights into the kinetics of morphogenesis in supramolecular aggregates.
The subject of this thesis is the synthesis and characterization of PBI-based fluorescent metallosupramolecular polymers and cyclic arrays. Terpyridine receptor functionalized PBIs of predesigned geometry have been used as building blocks to construct desired macromolecular structures through metal-ion-directed self-assembly. These metallosupramolecular architectures have been investigated by NMR, UV/Vis and fluorescence spectroscopy, mass spectrometry, and atomic force microscopy.
Neue künstliche Guanidiniocarbonylpyrrol-Rezeptoren zur Komplexierung von Oxo-Anionen in Wasser

Ziel der Dissertation „Neue künstliche Guanidiniocarbonylpyrrol-Rezeptoren zur Komplexierung von Oxo-Anionen in Wasser“ war die Weiterentwicklung dieser Rezeptoren nach Schmuck für die Komplexierung insbesondere von Carboxylaten, um sie hinsichtlich Bindungsaffinität und Substratspezifität zu optimieren. Dazu wurde zunächst die Synthese zweier wichtiger Grundbausteine in einzelnen Schritten vollständig überarbeitet, wobei veränderte Reaktionsbedingungen und Aufarbeitungsschritte zu gesteigerten Ausbeuten führten. Dadurch ist es nun möglich, diese Bausteine effizienter zu synthetisieren und im Multigramm-Maßstab für die Darstellung von Rezeptoren zur Oxo-Anionen-Erkennung einzusetzen. Weiterhin wurde die Verbesserung der Komplexierungseigenschaften gegenüber Carboxylaten auf zwei Arten untersucht: zum einen durch das Anbringen eines zusätzlichen Seitenarms an der Guanidinio-Einheit zur Bildung von Guanidiniocarbonylpyrrol-Tweezer-Rezeptoren, zum anderen durch das Einführen einer zweiten positiven Ladung neben der Carboxylat-Bindungsstelle (CBS) zur Darstellung biskationischer Guanidiniocarbonylpyrrol-Rezeptoren. Zur Darstellung von Tweezer-Rezeptoren wurde ein zusätzlicher Seitenarm an der N’-Position der Guanidinio-Einheit angebracht. Die beiden Arme sollten ein Substrat pinzettenartig von zwei Seiten, mit der CBS als Kopfgruppe, komplexieren können. Durch zusätzliche Wechselwirkungen des neuen Seitenarms sollte neben einer stärkeren Komplexierung vor allem eine höhere Substratspezifität erzielt werden. Die experimentell ermittelten Bindungskonstanten lagen allerdings im Bereich der N’-unsubstituierten Rezeptoren. Somit stellen die Tweezer-Modifikationen daher keine Verbesserung der Guanidiniocarbonylpyrrol-Rezeptoren dar. In einem weiteren Projekt zur Rezeptor-Optimierung wurden, durch Einführung einer zweiten positiven Ladung in Form einer terminalen Ammonium-Gruppe, biskationische Guanidiniocarbonylpyrrol-Rezeptoren erfolgreich synthetisiert. Die Komplexierungseigenschaften dieser biskationischen Rezeptoren wurden in Bindungsstudien vornehmlich mit Aminosäurecarboxylaten mittels UV- und Fluoreszenz-Spektroskopie, Massenspektrometrie, NMR-Spektroskopie, ITC und Molecular Modeling Berechnungen untersucht. Anhand der Substratspezifität der biskationischen Rezeptoren wurde deutlich, dass die Spacerlänge, an der die zusätzliche positive Ladung angebracht ist, eine entscheidende Rolle bei der Komplexierung spielte. Galten eigentlich starre, präorganisierte, kurze Linker als vorteilhaft hinsichtlich der Entropie, so ist hier zu erkennen, dass längere, flexiblere Linker zu einer besseren Komplexierung führen können, wenn geeignete zusätzliche nichtkovalente Wechselwirkungen möglich sind. Die biskationischen Rezeptoren stellen damit eine Optimierung des Carboxylat-Bindungsmotivs der Guanidiniocarbonylpyrrol-Rezeptoren nach Schmuck in der Anionen-Erkennung dar.
This work encompasses three parts. The first part provides a concise review of the most prominent metaheuristic concepts currently available and gives essential preliminaries together with definition of the combinatorial optimization problems. It substantiates the choice of the investigation direction and basis idea of the developed methods. In the second part the new nonlinear global optimization routines based on the TS strategy are described. The new approaches are the Gradient Tabu Search (GTS), the Gradient Only Tabu Search (GOTS), and the Tabu Search with Powell’s Algorithm (TSPA). In the last part of the work the GOTS is applied for such chemical optimization problems. The chapter provides a systematic approach how the variables are chosen and the adjustable parameters are set. As test cases the global minimum energy conformation of some amino acids, of two angiotensin converting enzyme (ACE) inhibitors, of 2-acetoxy-N,N,N-trimethylethanaminium, and of a HIV-1 protease inhibitor is determined.
This PhD thesis introduced several concepts for the construction of new supramolecular assem-blies in polar solvents. Although the building blocks differ in their binding mode and association strength they follow the same principle: one main driving force for the self-assembly in polar solutions in combination with one texturing force. The main self-assembly process is based on the mutual interaction of hydrogen-bond enforced ion pairs which deliver the association energy needed for stable, supramolecular structures even in polar solvents. The texturing force itself is represented by the linkers between the zwitterionic building blocks or parts of them. The different length and functionalization of the linkers have a tremendous influence on the mode of self-assembly leading to cyclic dimers, vesicles, layers or solid spheres. Hence, this principle is suitable for the construction of programmable monomers. Since the derivatisation of the main binding motive is rather simple it offers a great number of new and undoubtedly fascinating structures with potential applications in material and biomimetic science.

Im Rahmen dieser Disseration wurden im Einzelnen folgende Ergebnisse erzielt: A. Isolierung und Strukturaufklärung von Naphthylisochinolin-Alkaloiden aus verschiedenen Ancistrocladus-Spezies: • Die bisher noch nicht phytochemisch untersuchten Rindenextrakte der vietnamesischen Unterart Ancistrocladus tectorius ssp. cochinchinensis wurden im Hinblick auf ihre Sekundärmetabolite analysiert. Dabei identifizierte man vier bereits bekannte und drei neuartige Naphthylisochinoline-Alkaloide. Die Strukturen dieser drei Metabolite wurden nach Isolierung unter Verwendung diverser 2D-NMR-Techniken aufgeklärt. Die entdeckten Substanzen – Ancistrocladinium A (30) und seine beiden O-Demethylderivate 31 und 32 – waren die drei ersten Vertreter des neuartigen N,8'-Naphthyldihydroisochinolin-Kupplungstyps. Diese Naturstoffe verfügen über vielversprechende pharmakologische Wirkungen – vor allem gegen den Erreger der Leishmaniose. • Die botanisch noch nicht vollständig charakterisierte Lianenart ''A. ikela'', die aus der Demokratischen Republik Kongo stammt, wurde im Laufe der Arbeit morphologisch und phytochemisch untersucht und beschrieben. Neben den beiden N,C-verknüpften Naphthylisochinolinen Ancistrocladinium A (30) und Ancistrocladinium B [(M/P)-39] wurde bei der phytochemischen Analyse ein neuartiges C,C-gekuppeltes Alkaloid – 8-O-Methylancistrogriffin C (40) – isoliert. Des weiteren wurde ein Gradient entwickelt, der die vollständige Trennung der beiden Atrop-Diastereomere von 39 und dadurch HPLC-NMR- und HPLC-CD-Analysen der einzelen Epimere ermöglichte, so dass die Rotationsbarriere der bei Raumtemperatur langsam drehenden Biarylachse bestimmt werden konnte. • Aus Blättern der bereits gut untersuchten indischen Ancistrocladus-Art A. heyneanus wurde mit 6-O-Methyl-8,4'-O-didemethylancistrocladin (42) ein weiteres neues Naphthylisochinolin-Alkaloid isoliert. • Eine phytochemische Untersuchung der Familie der Ancistrocladaceae auf das Vorkommen von N,C-verküpften Naphtylisochinolinen ergab, dass diese strukturell außergewöhnlichen Alkaloide in diesen Lianen weit verbreitet sind. B: Die Rolle des Phloems bei der Pathogen-vermittelten Ausbreitung von Signalen: • Im Rahmen des Teilprojektes B8 des SFBs 567 wurden Untersuchungen zur Rolle des Phloems bei der Weiterleitung von Langstreckensignalen nach Infektion von Arabidopsis-thaliana-Pflanzen mit virulenten oder avirulenten Stämmen von Pseudomonas syringae pv. tomato durchgeführt. Zunächst wurde dazu eine im nL-Maßstab anwendbare Analysenmethode für die Hauptmetabolite von A. thaliana – die Glucosinolate – entwickelt. Mit Hilfe dieser empfindlichen Methode wurden in Pflanzenextrakten von A. thaliana viele bekannte und einige neue Glucosinolate (8-Methylsulfonyl-n-octyl-, 2-Hydroxy-4-methylsulfinyl-n-butyl-, 2-Hydroxy-4-methylsulfonyl-n-butyl- und 4-Hydroxy- benzoyloxymethylglucosinolat) identifiziert. Des weiteren wurden MS/MS-Analysen der Glucosinolate durchgeführt, bei denen neben mehreren typischen Fragmenten für die Thiozucker-Einheit auch einige charakteristische Fragmente für die unterschiedlichen Seitenketten (z.B. Methylsulfinyl-n-alkyl- oder Methylthio-n-alkyl-Struktur) detektiert wurden. Leider ergaben vor allem die aromatischen und heteroaromatischen Seitenketten-Typen kein typisches Fragmentierungs-muster. • Bei der Analyse der Phloemexsudate konnte in Phloemsäften von unbehandelten Pflanzen neben Methoxyglucobrassicin (73) ein für Pflanzen neuartiges Phosphat 87 (1-Glycero-1-myo-inositolphosphat) identifiziert werden. In den Phloemsäften der unterschiedlich behandelten Pflanzen (infiltriert mit MgCl2, einem virulentem oder einem avirulentem Pseudomonas-Stamm) kamen sämtliche Hauptmetabolite der Blätter vor. Lediglich ein leichter, nicht signifikanter Konzentrationsanstieg von Methoxyglucobrassicin (73) wurde im Phloemsaft von mit avirulenten Pathogenen infizierten Pflanzen festgestellt. Dieser Anstieg muss aber kritisch betrachtet werden, da er auch ein Artefakt des starken mechanischen Reizes des Infiltrationsprozesses sein könnte. Andere kleine Konzentrationsänderungen könnten außerdem durch das starke ''Grundrauschen'' der Infiltration überlagert werden. C: Strukturaufklärung polyketidischer Sekundärmetabolite aus Mikroorganismen: • Zwei niedermolekulare Naturstoffe aus dem extremophilen Streptomyceten-Stamm KC 1030, die in der Arbeitsgruppe von Prof. H.-P. Fiedler (Universität Tübingen) isoliert worden waren, wurden strukturell aufgeklärt. Bei dem einen handelt es sich um das bereits bekannte Frigocyclinon (89), bei dem anderen um ein neues Angucyclinon 88 mit Fridamycin-E-Grundkörper. Darüber hinaus wurden aus einem weiteren Streptomyces-Stamm (AK 671) zwei neue (97, 98) und drei (96, 99, 100) bekannte biosynthetisch interessante Sekundärmetabolite isoliert.
Wasserstoffbrückengesteuerte Ausrichtung von Merocyaninfarbstoffen für photorefraktive Materialien

Merocyaninchromophore spielen eine herausragende Rolle bei der Entwicklung von photorefraktiven Materialien für Anwendungen in der Holographie. Der photorefraktive Effekt beruht auf einer Orientierung der dipolaren Merocyanine in einem elektrischen Feld. Diese können umso effektiver ausgerichtet werden, je größer ihr Dipolmoment ist. Folglich sollten Merocyanine mit sehr großen Dipolmomenten den gewünschten Effekt hervorbringen. Es hat sich jedoch gezeigt, dass solche Merocyanine Dimere mit antiparalleler zentrosymmetrischer Struktur bilden. In dieser Anordnung addieren sich die Dipolmomente destruktiv, so dass die dipolare Eigenschaft des Materials verloren geht. In dieser Arbeit ist es gelungen, Merocyanine über sechsfache Wasserstoffbrückenbindungen zu supramolekularen Strukturen mit großen resultierenden Dipolmomenten zu assoziieren. Diese Komplexe werden in schwach polaren Lösungsmitteln sogar bei sehr niedrigen Farbstoffkonzentrationen gebildet.
Artificial light-harvesting (LH) systems have been obtained by self-assembly of naphthalene diimide-functionalized zinc chlorin dyads and triad in nonpolar, aprotic solvents. UV-vis, CD, and steady-state emission spectroscopy as well as atomic force microscopy showed that rod-like structures are formed by excitonic interactions of zinc chlorin units, while the appended naphthalene diimide dyes do not aggregate at the periphery of the cylinders. In all cases, photoexcitation of the enveloping naphthalene diimides at 540 and 620 nm, respectively, was followed by highly efficient energy-transfer processes to the inner zinc chlorin backbone, as revealed by time-resolved fluorescence spectroscopy on the picosecond time-scale. As a consequence, the LH efficiencies of zinc chlorin rod aggregates were increased by up to 63%. The effective utilization of solar energy recommends these biomimetic systems for an application in electronic materials on the nanoscale.
Die Natur entwickelte im Laufe der Evolution eine unvorstellbare Vielfalt an unterschiedlichsten Lebewesen. Viele diese Organismen besitzen die Fähigkeit, biologisch aktive Sekundärstoffe zu produzieren, die ihnen im täglichen Überlebenskampf einen Vorteil gegenüber ihren Konkurrenten bieten. Die Effizienz, mit der solche Naturstoffe in lebenden Organismen biosynthetisch dargestellt werden, wurde von der organisch-chemischen Synthese bislang nicht annähernd erreicht. Die Untersuchung von Biosynthese-Routen verspricht daher nicht nur die Entdeckung wissenschaftlich interessanter Phänomene, sondern bietet zudem die Chance, von der Natur zu lernen. Die Ergebnisse der vorliegenden Arbeit zeigen dabei deutlich, dass sowohl die genaue Analyse des Aufbaus bereits lange bekannter Metabolite, wie z.B. des Furanonaphthochinons I (FNQ I) oder des Chrysophanols, als auch die Untersuchung der Biosynthese strukturell neuartiger Sekundärstoffe, wie etwa des Sorbicillactons A, von großem Interesse sein können. Die gewonnenen Informationen können dann zur optimierten biotechnologischen oder synthetischen Produktion viel versprechender bioaktiver Substanzen genutzt werden. Auch die Gewinnung neuer Substanzen aus der Natur, z.B. als Leitstrukturen für die Pharma-Forschung, ist ein lohnendes Ziel. Eine stete Verbesserung der Methoden zur Charakterisierung von Naturstoffen, z.B. unter Anwendung von Online-Analyse-Verfahren, hilft dabei, die gezielte Entdeckung noch unbekannter Metabolite schneller und einfacher zu gestalten. Für die Aufklärung der Konstitution von Substanzen nützlich ist hier vor allem die Kopplung von HPLC mit NMR und MS, wie beispielsweise im Rahmen der Identifizierung von Secohyperforin und neuer mariner Macrolactame gezeigt. Die Kombination von HPLC mit CD bietet zudem die Chance zur effizienten Aufklärung der absoluten Stereostruktur chiraler Verbindungen direkt am Peak im Chromatogramm. Ziel der vorliegenden Arbeit war die Isolierung, die biosynthetische und strukturelle Charakterisierung sowie die Produktion bioaktiver Sekundärstoffe unter Anwendung und Weiterentwicklung unterschiedlichster Konzepte und Methoden der Naturstoffchemie. Die vielfältigen Ergebnisse sind das Resultat von interdisziplinären Kooperationen innerhalb des SPP 1152 (DFG Schwerpunktprojekts 'Evolution metabolischer Diversität') und des vom BMBF geförderten Exzellenzzentrums BIOTECmarin ('Nachhaltige Nutzung mariner Schwämme').
Neuartige Wirkstoffe gegen Infektionskrankheiten : N,C-gekuppelte Naphthylisochinolin-Alkaloide

Infektionskrankheiten sind nach wie vor weltweit die Todesursache Nummer eins. Aufgrund der zunehmenden Resistenzbildung der Erreger gegen gängige Medikamente verlieren diese immer mehr an Wirksamkeit und bereits besiegt geglaubte Krankheiten, wie Tuberkulose und Malaria, erleben derzeit ein comeback. Aus diesem Grund ist die Suche nach neuartigen Wirkstoffen nach wie vor ein wichtiges Ziel. Eine aussichtsreiche Quelle neuer Leitstrukturen gegen Infektionskrankheiten sind Pflanzen, die ein breites Spektrum an strukturell facettenreichen Sekundärmetaboliten bieten. Eine solche viel versprechende neue Wirkstoffklasse phytochemischen Ursprungs sind die Naphthylisochinolin-Alkaloide, die ausgeprägte In-vitro-Aktivitäten gegen protozoische Erreger wie Plasmodien, Leishmanien und Trypanosomen aufweisen. Kürzlich wurde eine neuartige Unterklasse dieser Alkaloide entdeckt. Es handelte sich dabei um die ersten N,C-verknüpften Naphthylisochinoline, wie z.B. Ancisheynin sowie Ancistrocladinium A und B . Diese Alkaloide weisen als strukturelle Besonderheit eine bis dato beispiellose Hetero-'Biarylachse' auf, genauer die erste rotationsgehinderte Iminium-Stickstoff-Arylachse. Des Weiteren zeichnen sie sich durch eine hohe antileishmaniale Aktivitäten aus, bei vergleichsweise geringen Cytotoxizitäten gegen menschliche Zellen. Das Ziel der vorliegenden Dissertation war daher die erstmalige totalsynthetische Erschließung dieser neuartigen Strukturunterklasse der Naphthylisochinoline. Ebenfalls sollte die ausgezeichnete antiinfektive Aktivität der N,C-verknüpften Alkaloide in Studien zur Struktur-Aktivitäts-Beziehung (SAR) sowie in Untersuchungen zum Wirkmechanismus in enger Zusammenarbeit mit unseren Partnern innerhalb des Sonderforschungsbereiches 630 sowie mit externen Kooperationspartnern evaluiert werden.
The covalent linkage of the aryloxy-substituents through macrocyclisation was applied for the synthesis of perylene bisimide atropo-enantiomers. The synthesis of macrocyclic perylene bisimides was achieved by using a tetra(3-hydroxyphenoxy)-functionalized perylene bisimide with achiral 2,6-diisopropylphenyl as imide substituent through Williamson´s etherfication which could be realized for four different oligoethylene glycol bridging units. Two regioisomeric macrocycles, namely the diagonally bridged (1,7- and 6,12- linkage) and the laterally bridged (1,12- and 6,7-linkage) isomers, were obtained for each bridging unit. The structural assignment of the isolated regioisomeric macrocycles was unambiguously accomplished by X-ray analysis of two macrocycles and by 1H NMR spectroscopy for all isomers. The conformational influence of the aryloxy-substituents on the functional properties of this class of chromophores could be derived by comparison of the optical and electrochemical properties of all isolated macrocylces with those of an open-chained reference compound. It was shown that the aryloxy-substituents prefer a lateral conformation in solution. Furthermore, solvent dependent fluorescence studies indicated that a photoinduced electron transfer process is of importance for the fluorescence quenching of electron-rich aryloxy-substituted perylene bisimides. The resolution of the atropo-diastereomers of diagonally bridged macrocyclic perylene bisimides with chiral 2-(R)-octylamine as imide substituent and diethylene glycol bridging units could be accomplished by semi-preparative HPLC on a chiral column. The chiroptical properties of the isolated epimerically pure macrocycles were determined by CD spectroscopy. Based on the experimental CD spectra, the stereochemical assignment of the isolated epimers was accomplished by application of the excition chirality method and confirmed by quantum chemical calculation of the CD spectra. The synthetical concept was extended successfully to 1,7-diaryloxy-substituted perylene bisimides. The structure of the diagonally bridged macrocycle was unambiguously confirmed by X-ray analysis and NMR spectroscopy. The atropo-enantiomers of this macrocycle could be resolved by semi-preparative HPLC on a chiral column and the assignment of the absolute configuration was achieved by comparison of the CD spectra of the resolved enantiomers with those of epimerically pure bis(macrocycles) reported before. By comparison of the X-ray structures obtained for the racemic mixture as well as one enantiomer important information could be extracted for the formation of p-dimers of perylene bisimides. The dependence of the interconversion barrier on the bulkiness of the bay-substituents was investigated for four halogen-substituted perylene bisimides. The dynamic properties were investigated by temperature-dependent NMR spectroscopy and kinectic measurements using CD spectroscopy. By applying the concept of the “apparent overlap” a convincing linear relationship between the size of the substituents and the free enthalpy of activation could be derived. Furthermore, the resolution of the atropo-diastereomers or enantiomers of the tetrachloro and tetrabromo-substituted derivates was accomplished, whereupon especially the 1,6,7,12-tetrabromosubstituted perylene bisimide provided at room temperature stable enantiomers. Additionally, the derived structure-property relationship allows the design of conformationally stable perylene bisimide enantiomers by proper choice of the bay substituents. In order to utilize the reversibility of self-assembly for the quantitative formation of macrocyclic perylene bisimides, a tetrazinc porphyrin-functionalized perylene bisimide was synthesized. The self-assembly of the zinc porphyrin perylene bisimide bichromophoric building block and diazabicyclo-[2.2.2]-undecane into the desired 1:2 sandwich complex was investigated by UV/Vis and 1H NMR spectroscopy and the macrocyclic structure was unequivocally proven by diffusion-ordered NMR spectroscopy (DOSY NMR). Furthermore, the controlled deposition of these well-defined macrocycles on highly ordered pyrolitic graphite (HOPG) was demonstrated by atomic force microscopy (AFM) investigations. The alignment of a linear amino functionalised p-conjugated polymers upon addition of the bichromphoric tetrazinc porphyrin-perylene bisimide was investigated by UV/Vis spectroscopy and AFM measurement. The surface analysis by AFM investigations revealed that the bichromophoric system composed of perylene bisimide and zinc porphyrin is able to cross-link the linear p-conjugated polymer over a wide range of the graphite surface which provided a defined arrangement of three different functional p-systems.
Although known about and investigated since the late 1970’s, the picture of the basic principles governing inhibitor strengths and the structure-activity relationships of the cysteine protease inhibition mechanism is still very incomplete. Computational approaches can be a very useful tool for investigating such questions, as they allow the inspection of single, specific effects in isolation from all others, in a manner very difficult to achieve experimentally. The ab initio treatments of such large systems like proteins are still not feasible. However, there is a vast number of computational approaches capable of dealing with protein structures with reasonable accuracy. This work presents a summary of theoretical investigations into cysteine protease cathepsin B using a range of methods. We have concentrated on the investigation of cysteine protease inhibition by epoxide- and aziridine-based inhibitors in order to obtain better insight into these important topics. Various model systems are simulated by means of pure quantum mechanical methods and by hybrid (QM/MM) methods. Both approaches provide a static picture. Dynamical effects are then accounted for by additional molecular dynamics (MD) simulations, using both classical and QM/MM MD approaches. The quantum mechanical approach was used to study very small model systems consisting only of the electrophilic warhead of the inhibitor (both substitituted and not) and molecular moieties simulating a very simplified protein active site (methylthiolate instead of Cys29 and methylimidazolium instead of His199 residue) and solvent surroundings (two waters or two ammonium ions, in combination with a continuum solvent model). Although simple, such a system provides a good description of the most important interactions involved in the inhibition reaction. It also allows investigation of the influence of the properties of the electrophilic warhead on the reaction rate. Beside the properties of the electrophilic warhead, the protein and solvent environment is also an important factor in the irreversible deactivation of the enzyme active site by the inhibitor. The non-covalent interactions of the inhibitor with the oxyanion hole and other subsites of the enzyme, as well as its interaction with the solvent molecules, need to be explicitly taken into account in the calculations, because of their possible impact on the reaction profile. As molecular modeling methods allow the treatment of such large systems, but lack the possibility of describing covalent interactions, our method of choice was the combined quantum mechanics/molecular modeling approach. By splitting the system into a smaller part that undergoes the bond cleavage/formation process and must be treated quantum mechanically, and a larger part, comprised of the rest of the protein, which could be treated using force fields, we managed to simulate the system at the desired precision. Our investigations concentrated on the role of His199 in the inhibition mechanism as well as on the structure-reactivity relationships between cysteine protease and various inhibitors, yielding new insight into the kinetics, regio- and stereospecificity of the inhibition. In particular, our calculations provide the following insights: i.) an explanation for the regioselectivity of the reaction, and original insight into which interactions affect the stereoselectivity; ii.) a clear model which explains the known structure-activity relationships and connects these effects with the pH-dependency of the inhibition; iii.) our computations question the generally accepted two-step model by showing that substituent effects accelerate the irreversible step to such an extent that the achievement of an equilibrium in the first step is doubtful; iv.) by way of theoretical characterizations of aziridine models, the reasons for similarities and differences in the mode of action of epoxide- and aziridine-based inhibitors are elucidated; and finally, v.) combining our results with experimental knowledge will allow rational design of new inhibitors. To account for dynamical effects as well, molecular dynamics (MD) computations were also performed. In these calculations the potential energy was computed at the force field level. The results not only supported and clarified the QM/MM results, but comparison with previous X-ray structures helped correct existing errors in the available geometrical models and resolved inconsistencies in the weighting of various factors governing the inhibition. In the work the first QM/MM MD calculations on the active site of the cysteine proteases are presented. In contrast to the MD simulations, these calculations used potential energies computed at the QM/MM-level. With the help of these computations we sought to address strongly disputed questions about the reasons for the existence of the active site ion pair and its role in the high activity of the enzyme.
In the first part of this work a new approach to measure transient absorption spectra of fluorescent compounds by means of laser flash photolysis technique was presented. Generally, the recorded transient absorption signal consists of transient absorption, fluorescence and ground state bleaching. Thus, for fluorescent chromophores a fluorescence correction is indispensable in order to obtain undisturbed absorption decay curves as well as accurate transient absorption spectra. Due to time response characteristics of the PMT detector the fluorescence contribution cannot be corrected by recording the fluorescence separately. Measuring two transient absorption signals with probe light differing in intensity, compounds with quantum yields up to ~ 35 % can be investigated. This is a major improvement because transient absorption spectroscopy is a powerful method to gain insight into the kinetics and the energy of excited states and information in the time domain of fluorescence are no longer lost. In the second part the synthesis and the photophysical characterisation of redox cascades were reported. These cascades consist of an acridine acceptor and up to three triarylamine donor subunits. The redox potentials of the triarylamines were tuned by adequate substituents in the para-position of the phenyl ring to ensure a directed redox gradient. Upon photoexcitation a locally excited state or a CT state is populated which then injects a hole onto the adjacent donor and consequently results in a CS state. Fluorescence and transient absorption measurements revealed that HT depends strongly on donor strength and solvent polarity. Formation of a CS state was only observed in case of strong terminal donors or polar solvents. A low lying localised triplet state acts as an energy trap and quenches all CS states even in case of the cascade with the strongest terminal donor in very polar solvents. Furthermore, population of a CS state catalyses the formation of this triplet states which results in a shorter lifetime of the CS state compared to the lifetime of the CT state of the corresponding reference compound. Compared to redox cascades already reported in literature, the electronic coupling between the redox centres was decreased by sterical as well as electronic effects. To prolong the lifetime of the CS state saturated spacers on the one hand and a perpendicular orientation of the acceptor and the adjacent donor on the other hand were selected. The twisting of the subunits forming the CT state results in a higher degree of charge separation but its contribution to increase the lifetimes of the CS states is of minor importance. The longer lifetime of the CS states can be ascribed to the saturated spacers. Experimental data in combination with calculated values indicate that charge recombination takes place in the Marcus normal region by a superexchange mechanisms. Although charge recombination of the known cascades is located in the Marcus inverted region, these CS states decay faster than the CS states of the compounds investigated in this work.
The present work deals with the synthesis and the investigation of the photophysical properties of covalently constructed calix[4]arene–perylene bisimide dye arrays containing various PBI units. The obtained conjugates are characterized with respect towards their application in a new, zigzag-type architecture of artificial light-harvesting systems. For this purpose, orange (core-unsubstituted), red (6,7,11,12-tert-butylphenoxy-functionalized) and green (1,7-pyrrolidino-substituted) perylene bisimide building blocks have been attached to the calix[4]arene scaffold. First, the monochromophoric reference systems have been studied, and second, the photophysical properties of a comprehensive series of newly synthesized, multichromophoric calix[4]arene–perylene bisimide conjugates showing efficient energy transfer processes between the individual dye subunits have been investigated. Furthermore, a series of bichromophoric compounds containing identical chromophoric units has been obtained. Towards this goal, a variety of spectroscopic techniques such as UV/vis absorption, steady state and time-resolved fluorescence emission, and femtosecond transient absorption spectroscopy as well as a spectrotemporal analysis of the obtained data has been applied. This work presents a new concept for an artificial light-harvesting system positioning the dye units by means of calix[4]arene spacers along a zigzag chain. The investigations start with the syntheses and optical properties of the monochromophoric building blocks and result in an elaborate study on the energy and electron transfer processes occurring after photoexcitation in a comprehensive series of multichromophoric calix[4]arene–perylene bisimide conjugates. Finally, the photophysical properties of a series of compounds containing each two identical PBI units are discussed.
Diese Arbeit beschäftigt sich mit der Selbstorganisation von Zinkchlorin-Farbstoffen, welche sich strukturell von Chlorophyllen ableiten. Im Gegensatz zu allen anderen bakteriellen und pflanzlichen Lichtsammelpigmenten ist es den Bakteriochlorophyllen c, d und e der Lichtsammelsysteme grüner phototropher Bakterien möglich, allein durch nichtkovalente Wechselwirkungen zwischen den Farbstoff-Molekülen, ohne die Beteiligung von Proteinen, röhrenförmige Antennensysteme auszubilden, welche die am dichtest gepackten und effizientesten Lichtsammelsysteme in der Natur darstellen. Um einen Betrag zur Aufklärung dieser biologisch wichtigen Aggregate zu leisten, wurden im ersten Teil dieser Arbeit Zinkchlorine als Modellverbindungen für BChl c hergestellt. Mit den neu synthetisierten Zinkchlorinen ist es gelungen, Modellsysteme der natürlichen BChl-Selbstorganisate herzustellen, welche sich im Gegensatz zu den bisher in der Literatur beschriebenen Zinkchlorin-Aggregaten durch eine gute und dauerhafte Löslichkeit auszeichnen. Diese Eigenschaft erlaubte es sowohl spektroskopische als auch mikroskopische Untersuchungen zur Aufklärung der Aggregatstruktur durchzuführen. Durch Rasterkraftmikroskopie an den Zinkchlorin Aggregaten konnte erstmals ein mikroskopischer Beweis der stabförmigen Struktur von Aggregaten dieser Substanzklasse erhalten werden. Der zweite Teil dieser Arbeit beschäftigt sich mit Zinkchlorinen, denen aufgrund einer methylierten 31-Hydroxy-Gruppe die Fähigkeit zur Röhrenbildung fehlt, die aber durch Koordinationsbindungen und p-p-Wechselwirkungen weiterhin Stapel bilden können. Temperaturabhängige UV/Vis- und CD-spektroskopische Studien offenbarten die reversible Bildung von löslichen, chiralen Zinkchlorin-Stapelaggregaten. Rasterkraft- und rastertunnelmikroskopische Untersuchungen zeigen die Bildung von zwei Typen p-gestapelter Aggregate auf hoch geordnetem Graphit.
The effective binding of anions like carboxylates and phosphates in aqueous solutions is of particular interest for various reasons. The natural archetypes of effective anion receptors are enzymes that contain often arginine as relevant amino acid in the binding pocket. For this reason, one class of artificial anion receptors that emerged more than two decades ago mimics the anion binding with the guanidinium group present in the amino acid side chain. In 1999, Schmuck and coworkers developed a new class of guanidinium-based oxo anion receptor that binds carboxylates even in aqueous media. The binding modes of the 2-(guanidiniocarbonyl)-1H-pyrroles are based on individually weak non-covalent interaction between artificial host and substrate like ion pairing and multiple hydrogen bonds. The zwitterionic derivative with substitution of a carboxylate group in position 5 of the pyrrole ring system shows a strong self-assembly to discrete dimers (dimer 1) with an estimated association constant of 170 M-1 even in water. In order to further improve the structure motif for an effective oxo anion binding it is therefore of great interest to quantify the different intermolecular interactions between two monomeric units of 1. Against this background several theoretical ab initio studies were conducted in order to elucidate the influences of intrinsic properties as well as solvent effects on the stability of self-assembled dimers. In chapter 4.1 the molecular interactions in dimer 1 were investigated by comparison to various “knock-out” analogues. In these analogues single hydrogen bonds were switched off by substitution of hydrogen donor atoms with either methylene groups or ether bridges. The calculations were done for vacuum and solvation, as represented by a conductor-like polarizable continuum. It could be shown that the application of a simple continuum solvent model fails to predict the absolute energies of the knock-out analogues in strongly polar solvents. However, the calculated trends can explain the relative stabilities. In chapter 4.2 the structural similarity of arginine with structure 1 was used in order to examine the dependence of self-assembly from the flexibility of the molecular structure. In chapter 4.2.1 new global minimum structures of the canonical and zwitterionic arginine in gas phase were found by means of exhaustive force field based conformational searches in conjunction with ab initio structure optimizations of the lowest energy conformers. Most of the newly identified minimum conformers of both the zwitterionic and canonical tautomer revealed geometrical arrangements with hitherto unreported stacked orientations of the terminal groups. Finally a novel global minimum structure was detected that is more than 8 kJ mol-1 lower in energy than the previously published conformers. The same strategy for finding minimum energy conformers of the arginine monomer has also been employed for the arginine dimer structures. While previous theoretical studies favoured directed hydrogen bonds the new global minimum structure MMFF1 is about 60 kJ mol-1 more stable and exhibits a stacked orientation of the guanidinium and carboxylate groups. The importance of rigidity on the dimer stability was proven by calculations of an artificially stiffened arginine dimer system. The high binding affinity dimer 1 results by about 50% from the rigidity of the monomers which prevents any intramolecular stabilization. In chapter 4.3 novel structure motifs with varying ring systems have been examined on a DFT level of theory in order to make proposals for an improved carboxylate binding motif. The direct dependency of the dimerization energy on an increasing dipole moment was demonstrated by various anellated ring structures. The influence of the delocalization in the monomer on the dimerization energy was examined by variation of the electronic structure of electronically decoupled biphenylenes. With the aid of various substituted 7-guanidinioindole-2-carboxylate derivatives we could show that the carbonyl function is mainly responsible for the advantageous preorganisation, whereas the effect on the acidity seems to be only of minor importance. In the last chapter cooperativity effects in supramolecular assemblies have been investigated. This was achieved by NMR shift calculations of adenosine-carboxylic acid complexes as model systems and comparison to experimental low-temperature NMR studies. We could demonstrate that only by applying vibrational averaged NMR shifts the experimental proton shifts obtained at very low temperatures in the hydrogen bond exchange regime could be reproduced.
Oxygen-centered radicals are important intermediates in photobiological, mechanistic and synthetic studies. The majority of precursors of reactive oxyl radicals are labile and thus delicate to handle. Therefore N-(alkoxy)-pyridinethiones and N-(Alkoxy)-thiazolethiones have attracted attention as "mild'' photochemical source of alkoxyl radicals, in the last few years. A disadvantage of the pyridine compounds, is their sensibility to daylight. Despite of their similarities, both molecules behave surprisingly different, if photolyzed in the absence of trapping reagents. The pyridinethione compounds undergo highly efficient radical chain reactions under such conditions while the corresponding thiazolethiones react surprisingly sluggish and give rise to several unwanted side products. The properties of both compounds should be understood and optimized in the frame of this work. Additionally new compounds should be suggested that can also be applied in the photochemical alkoxyl radical generation. Some background information about the generation and application of alkoxyl radicals is provided in chapter 2. Electronic excitations and UV/vis spectroscopy together with a description of quantum chemical approaches that are able to calculate such phenomena are outlined in chapter 3. Chapter 4 deals with the description of the vertical excitation spectra. During the validation CASSCF, CASPT2, TD-DFT and RI-CC2 were tested with respect to their ability to describe the vertical excitations in both compounds. The CASPT2 approach gives accurate descriptions of the electronic excitation spectra of all compounds. The time-dependent DFT results are very sensitive on the choice of the functional and a validation of the results should be always done. On the basis of these computations the spectroscopic visible absorption bands of both compounds were assigned to a pi-->pi* transition in the thiohydroxamic acid functionality. In chapter 5 the mechanism of the thermally and the photochemically induced N,O homolysis in both compounds is unveiled. The near UV-induced N,O homolysis will start from the S2 state. The expected relaxation from the S2- to the S1-state and the dissociation process is expected to be very fast in the case of the thiazolethione compound. The potential surfaces of the pyridine compound in contrast point to a slower N,O bond dissociation. Due to the resulting faster dissociation process the excess energy which results from the photochemical activation is quenched only to small amounts. The maximal possible excess energy of the fragments is lower and a quenching is much more likely in the case of the pyridinethione compounds. This explaines the different reactivities of both compounds. For the also already successfully applied precursor system N-(alkoxy)-pyridineones the computed dissociation paths show courses that clearly predict a slow bond dissociation process. Chapter 6 deals with the tuning of the initial excitation wave length of the known pyridinethiones und thiazolethiones. In the first part the effects of substituents on the thiazolethione heterocycle was examined. The UV/vis spectra of 4 and 5 substituted thiazolethiones can be interpreted like the spectrum of the parent compound. The second part of chapter 6 deals with the identification of a substitution pattern on the pyridine heterocycle which induces a blue shift of the photo active band. The computations showed that electron rich and electron poor substituents result the same effects on the electronic excitation spectra. These substituent effects are additive, but the steric orientation of the substituents has to be taken into account. Chapter 7 describes a computer aided design of new alkoxyl radical precursors. Combining the advantages of both compounds the radical formation should be initiated by an irradiation with light at about 350 nm, and the amount of side products during the radical formation process should be small. To achieve this 18 test candidates were obtained by a systematic variation of the parent compound of the thiazolethione precursor. To identify the promising new precursor systems a screening of the lower electronic excitations of all resulting 18 systems was performed with TD-DFT. For promising systems the N,O or P,O dissociation paths, respectively, were analyzed according to the developed model. N-(methoxy)-azaphospholethione and N-(methoxy)-pyrrolethione seem to be the most promising candidates. The computations predict a strong absorption at about 350 nm respectively 320 nm. Due to the amounts of maximal excess energy and the shapes of the potential surfaces of the N,O bond dissociation paths their reactivity should resemble more the behavior of the pyridinethiones.
Aus dem Bromfluorcarben-Addukt des Indens wurden durch HPLC an Chiralcel OD die reinen Enantiomere erhalten. Damit stand erstmals eine enantiomerenreine Vorstufe für die Freisetzung eines Sechsringallens im Sinne der Doering-Moore-Skattebol Reaktion zur Verfügung. Die Behandlung dieser Vorstufen, die in 2,5-Dimethyl, 2-tert-Butyl-5-methyl- or 2,5-Di-tert-butylfuran gelöst waren, mit Methyllithium lieferte die [4+2]-Cycloaddukte des Isonaphthalins an die Furane. Durch HPLC an Chiralcel OD wurde gezeigt, dass die Produkt-Enantiomerenpaare mit ca. 40% ee anfielen und dass dieser Wert praktisch unabhängig von der Art des Furans und seiner Konzentration, sowie von der Raktionstemperatur war. Die absoluten Konfigurationen der Vorstufe sowie der [4+2]-Cycloaddukte wurden durch Vergleich der gemessenen und berechneten CD-Spektren ermittelt, was in Zusammenarbeit mit der Arbeitsgruppe von Prof. Dr. B. Engels erfolgte. Erzeugt durch Methyllithium aus dem Dibromcarben-Addukt des Indens wurde das Isonaphthalin erstmals durch Inden abgefangen. Die Konstitution diese [2+2]-Cycloaddukts wurde durch Röntgenstrukturanalyse bestimmt. Erstmals wurde das Bromfluorcarben-Addukt des 1-Phenylcyclopentens hergestellt und erfolgreich daraufhin geprüft, ob es als Vorstufe für 1-Phenylcyclohexa-1,2-dien geeignet ist. Mit Hilfe von HPLC an Chiralcel OJ-H wurden die reinen Enantiomere erhalten. Die Umsetzung eines solchen reinen Enantiomers mit Methyllithium in Gegenwart von 2,5-Dimethylfuran erbrachte das [4+2]-Cycloaddukt als reines Enantiomer. Die Umsetzung einer reinen Vorstufe mit Methyllithium in Gegenwart von Inden erbrachte das [2+2]-Cycloaddukt mit einem Enantiomerenverhältnis von 95:5. Überraschend wurden auch relativ hohe Enantioselektivitäten beim Abfang dieser Vorstufe mit Styrol beobachtet. Bei der Umsetzung des reinen Enantiomers mit Methyllithium und Styrol ergab sich ein Gemisch der zwei endo- und exo-Enantiomere des [2+2]-Cycloaddukts von 50:3:40:7, was ee-Werten von 89 bzw. 79% entspricht. Geht man von einem zweistuifigen Mechanismus der Reaktion von 1-Phenylcyclohexa-1,2-dien mit Styrol aus, dann spielt die achirale Konformation des intermediären Diradikals, wenn überhaupt, keine große Rolle. Die Reduktion des bekannten 1,6-Dibrom-2-phenylcyclohex-1-ens mit Lithiumaluminiumhydrid erbrachte mit 78% Ausbeute die Monobromide 1-Brom-2-phenylcyclohexen and 1-Brom-6-phenylcyclohexen, die bei der Behandlung mit Kalium-tert-butoxid ebenfalls das Cycloallen 1-Phenylcyclohexa-1,2-dien liefern. Dies zeigte die Ausführung dieser Reaktion in Gegenwart von Furan und 2,5-Dimethylfuran.
Ausgehend von Benzvalen wurden zwei an der Doppelbindung difunktionalisierte Bicyclo[2.1.1]hex-2-ene synthetisiert, die in Anlehnung an Literaturmethoden jeweils zu einem Benzolderivat trimerisiert werden sollten. Durch dreifachen Ringschluss in den Anellanden unter Ausbildung von Bicyclobutansystemen sollte daraus dann ein Octahydro-trimetheno-trinden hervorgehen, von dem aufgrund der Spannungsenergie in den anellierten Systemen eine signifikante Bindungslängenalternanz im Benzolring erwartet wurde. Zur Untersuchung der Natur der zentralen Bindung in [1.1.1]Propellanen wurde aus Benzvalen und Adamantanon in Anlehnung an die Literatur ein neues [1.1.1]Propellan dargestellt. Weiterhin wurden verschiedene aromatische Azide in einer 1,3-dipolaren Cycloaddition an Hexamethyl-Dewar-Benzol (HMDB) addiert. Unter Thermolysebedingungen lagerten die resultierenden Produkte in Abhängigkeit vom aromatischen Rest zu unterschiedlichen Verbindungen um.
This thesis deals with the isolation and structural elucidation of bioactive naphthylisoquinoline alkaloids and related analogs. The mode of action of the antiplasmodial activity exhibited by the naphthylisoquinoline alkaloids was explored and compared to that of the antimalarial drug chloroquine. Furthermore, the phase 1 and 2 metabolism of dioncophyllines A and C and dioncopeltine A were investigated. In detail the following results have been obtained: • From the leaves of the recently discovered East African liana A. tanzaniensis six naphthylisoquinoline alkaloids were isolated. • The leaves of a botanical yet undescribed Ancistrocladus species, collected by Prof. Dr. V. Mudogo in the Democratic Republic of Congo in the habitat Yeteto near the town Ikela, were analyzed for naphthylisoquinoline alkaloids for the first time. The isolation work led to the first identification of an N,C-coupled naphthyldihydroisoquinoline alkaloid; ancistrocladinium B. Phytochemical investigation of the roots of the Congolese Ancistrocladus species (habitat Yeteto), , afforded five new derivatives of known naphthylisoquinoline alkaloids, namely 5'-O-demethylhamatine, 5'-O-demethylhamatinine, 6-O-demethylancistroealaine A, 6,5'-O,O-didemethylancistroealaine A, and 5-epi-6-O-methylancistrobertsonine A, along with six known naphthylisoquinoline alkaloids. • The antiplasmodial activity guided purification of 60Co irradiated samples containing commercially available naphthylisoquinoline related substances, afforded the isolation of the irradiation products 3,4-dihydro-1-isoquinolinone, 3,4-dihydro-1-isoquinolineamine, and 1,2,3,4-tetrahydro-1,2-diazirino-isoquinoline. The compounds were found to be more active than the starting material, although only exhibiting weak antiplasmodial activity against P. falciparum. • The effect on the absorption spectrum of FPIX due to complex formation with the naphthylisoquinoline alkaloids dioncophyllines A and C, dioncopeltine A korupensamine A, and ancistrocladine was examined by a titration study. Job's plot analyses by UV-spectroscopy determined the stoichiometry for the complex formation of FPIX and naphthylisoquinoline alkaloids to be 2:1. Furthermore, the dissociation constants for the complexation with FPIX were determined for each of the naphthylisoquinoline alkaloids investigated. Dioncophylline C and dioncopeltine A were found to possess dissociation constants, which are comparable to the one reported for the antimalarial drug chloroquine. The ability of ESI to transfer noncovalent solution-phase assemblies intact into the gas phase, was conducted on solution mixtures of naphthylisoquinoline alkaloid and FPIX, as well as on mixtures of chloroquine and FPIX. The mass spectrometry analyses revealed several peaks, which corresponded to the complex formation of FPIX to the respective ligands investigated. The most interesting results obtained were the detection of peaks corresponding to the complex formation between a chelated dimer of FPIX and dioncophylline Cand of peaks corresponding to a double protonated tetramer of FPIX – consisting of two chelated -oxo dimers of FPIX – in complex formation with two molecules of chloroquine. • Two phase 1 metabolism products of dioncophylline A were identified. Coelution in combination with HPLC-MS/MS, NMR, and CD investigations assigned the major metabolic product as 5'-O-demethyldioncophylline A. The minor metabolic product was only present in small amounts, which disabled an unambiguous structural characterization of the compound. However, as deduced from the mass spectrometry analyses and exclusion of a possible metabolic oxidation product by coelution with authentic reference material, the metabolite should possess a 4-hydroxylated isoquinoline portion and is assumed to be represented by structure. Dioncophylline C and dioncopeltine A were found to be stable to phase 1 metabolism reactions caused by rat liver microsomes.
Die absoluten Konfigurationen von mehr als 20 neuartigen Naturstoffen und Syntheseprodukten mit unterschiedlichen Chiralitätselementen (stereogene Zentren, chirale Achsen und chirale Ebenen) wurden durch Vergleich ihrer experimentellen CD-Spektren mit den quantenchemisch berechneten der jeweils möglichen Stereoisomere aufgeklärt. Zur Simulation des molekularen CD kamen dabei semiempirische Verfahren (CNDO/S und OM2) und die zeitabhängige Dichtefunktionaltheorie (TDDFT) zum Einsatz.
The functionalities of DNA and RNA are mainly determined by the various interactions between the pairing nucleobases. To understand the complex interplay of the various interactions model systems are needed in which the interstrand pairing is less restricted by the backbone. Such systems are peptide nucleo acids (PNA) in which the sugar phosphate backbone of DNA or RNA is replaced by a peptide backbone. Diederichsen et al. were able to synthesize a large number of systems with an alpha-alanyl backbone to which canonical and non-canonical nucleobases were attached (alpha-alanyl-PNA). These systems formed aggregates with various binding motifs which do not appear in DNA or RNA. Especially the unusual binding motifs would allow a deep insight into the complex interplay of the interactions between nucleobases but the small solubility of alpha-alanyl PNA oligomers hampers the experimental determination of the geometrical arrangement by X-Ray or NMR. Only the overall stability of the various aggregates could be determined by measurements of melting temperatures via UV spectroscopy. Since a detailed knowledge about the geometrical structure and bonding motifs are necessary to obtain insight into the interplay of the various interactions it is the goal of the present work to achieve such information with the help of theoretical approaches. Additionally we are interested in the effects which govern the trends in the stabilities of the systems. This task should be simpler than an investigation of the absolute stabilities since many contributions (e.g. entropic and dynamic effects) can be expected to be similar for similar systems. Consequently, such effects are less important for our goal. For the investigation of all experimentally tested alpha-alanyl-PNA oligomers it was essential to parameterize the noncanonical nucleobases since they were not implemented in the standard version of the Amber4.1 force field. This was achieved by adding the missing parameters to the Amber Force Field. The charges of each nucleobase were determined by the R.E.D program package. The investigation started with the construction of all possible pairing modes for alpha-alanyl-PNA dimer. It could be observed that certain pairing modes were not realizable due to the geometrical arrangement of the dimer and the restriction of the backbone. For other pairing modes a construction was possible, but due to the geometrical restrictions of the backbone the strain in the system is so high that they fall apart during a first geometry optimization. Stable systems were then simulated by various molecular dynamics (MD)-runs. Information about their geometrical arrangements for T=0 K were obtained from geometry optimizations which were started from various points of the MD-run. The resulting geometries were found to be virtually identical. Information about the interactions within a dimer at T=0 K were obtained from a two step procedure in which the effects connected with the nucleobases and the influence of the backbone are determined separately. It was performed for the optimized geometries. In a first step the backbone was removed and the resulting dangling bonds were saturated by methyl groups. The total interaction energy between the nucleobases can now be estimated by the difference between the energy of the complete system and the sum of the energies of the single nucleobases computed at the geometries they take in the whole system. According to the carried out investigation and the resulting correlation of the melting temperature with the calculated stabilization energies the presented method seems to represent a reliable tool for the description of the PNA systems. Despite this success additional experimental verifications of our method are necessary to ensure its applicability. Such verifications could be based on geometrical information obtained via X-Ray or NMR investigations. More detailed data about entropic an enthalpic contribution to the stability of the various complexes would also be very helpful to verify and improve our approach. Such information could be either obtained from a careful analysis of shape of the melting temperature curve or from microcalorimetric investigations. If such tests confirm our predictions the approach could be extended and applied to neighboring fields as for examples beta-alanyl-PNA, DNA or RNA systems with unusual nucleobases. Such information is also necessary to extend our approach in a way that dynamic and/or entropic effects are also taken into account.
Die vorliegende Arbeit beschäftigt sich mit der Entwicklung von künstlichen Rezeptoren für biologisch relevante Oligopeptide und besteht aus drei Teilen. Im ersten Teil wurde auf der Basis von computergestützten de novo Berechnungen ein künstlicher Rezeptor für den D-Alanin-D-Alanin-C-Terminus entwickelt. Diese Peptidsequenz befindet sich in bakteriellen Zellwänden und nimmt eine Schlüsselfunktion in der Wirkungsweise des Antibiotikums Vancomycin ein. Zur Entwicklung dieses Rezeptors wurde ein Guanidiniocarbonylpyrrol als Bindungsmotiv für Carboxylate mit einer Cyclotribenzylen-Einheit verknüpft. Letztere ist entsprechend der theoretischen Berechnungen in der Lage, die Methylreste des Alanins größenselektiv durch hydrophobe Wechselwirkungen zu koordinieren. Dieser Rezeptor wurde in umfangreichen Bindungsstudien bezüglich seiner Affinität in Wasser und seiner Substratselektivität untersucht. Zur Erhöhung der Löslichkeit und zur Bestimmung der Komplexstruktur mit NMR-Techniken in Wasser wurde ein weiteres Derivat des Rezeptors synthetisiert, welches in peripherer Position mit Triethylenglykolseitenketten substituiert ist. Auf diese Weise gelang es, einen hoch affinen (log K = 4,7) und hoch selektiven künstlichen Rezeptor für den D-Ala-D-Ala-Terminus darzustellen und umfassend zu charakterisieren. So konnte gezeigt werden, dass ein de novo Design derartiger Rezeptoren prinzipiell möglich ist. In einem weiteren Teilprojekt wurde ein künstlicher Rezeptor für die interne RGD-Peptidsequenz entwickelt. Diese nimmt eine zentrale Funktion in Zell-Zell- und Zell-Matrix-Erkennungsprozessen ein. Dieses Teilprojekt wurde in Zusammenarbeit mit dem Arbeitskreis Schrader (Universität Marburg) durchgeführt. Dazu wurde ein Bindungsmotiv für Alkylguanidine (in der Seitenkette von Arg, R) über einen geeigneten Spacer mit einem Bindungsmotiv für Carboxylate (in der Seitenkette von Asp, D) verknüpft. Nach der Synthese und Charakterisierung einer Reihe von vier Rezeptoren konnte die grundsätzliche Anwendbarkeit dieses Ansatzes bestätigt werden. Dabei konnte gezeigt werden, dass der verwendete Spacer für die Effektivität der Koordinierung von besonderer Bedeutung ist. Auf der Grundlage dieser Ergebnisse wurde in einem dritten Teilprojekt ein kombinatorisches Festphasenprotokoll zur Optimierung derartiger Spacer entwickelt. Dabei wurde das Carboxylat-Bindungsmotiv (ein Guanidiniocarbonylpyrrol) auf einem polymeren Träger immobilisiert. Zu diesem Zweck wurden umfangreiche Studien zur Synthese von Pyrrol-Tricarboxylaten und zur Verwendung verschiedener Schutzgruppen unternommen. Die Eigenschaften von drei Schutzgruppen unterschiedlicher Sensitivität (basisch, stark sauer und photolytisch spaltbar) auf dem Acylguanidin wurden in Lösung und an der festen Phase untersucht. Zu diesem Zweck wurde ein umfangreiches HPLC-Protokoll zur Charakterisierung der Reaktion entwickelt. So gelang die Entwicklung und Etablierung eines universell einsetzbaren Protokolls zur Optimierung derartiger Rezeptoren, womit zahlreiche Anwendungsmöglichkeiten in der kombinatorischen Chemie aber auch in weiteren Teilbereichen wie der Katalyse oder der Chromatographie ermöglicht werden.
Polyketide stellen aufgrund ihrer großen strukturellen Vielfalt nach wie vor Leit- und Wirkstoffe für die Pharma- und Pflanzenschutzforschung in den Industrieländern dar und bilden außerdem eine der wichtigsten Klassen von Naturstoffen (Sekundärmetaboliten) überhaupt. Besonders die Biosynthese aromatischer Polyketide und die hierbei involvierten Enzyme, die Polyketidsynthasen (PKS), wurden von Biosyntheseforschern als hervorragendes Modellsystem zur Untersuchung von Struktur-Funktions-Beziehungen von Multienzymkomplexen erkannt. Für annelierte aromatische Polyketide existiert seit dem Jahr 2001 eine biosynthetische Klassifizierung auf Metabolitebene, das sogenannte Modus-F/S-System, mit dessen Hilfe man zwischen pro- und eukaryotischen Produzenten unterscheiden kann. Die Erforschung der detaillierten Biosynthese von aromatischen Polyketiden ist somit in mehrfacher Hinsicht ein lohnendes Ziel. In der vorliegenden Dissertation sollten die Biosynthese und die Faltungsmodi ausgewählter aromatischer Polyketide einschließlich der Charakterisierung potentieller Vorstufen in verschiedensten biologischen Systemen untersucht werden. Die dabei gewonnenen Resultate sind das Ergebnis interdisziplinärer Zusammenarbeit.
As a traditional industrial pigment, perylene bisimide (PBI) dyes have found wide-spread applications. In addition, PBI dyes have been considered as versatile and promising functional materials for organic-based electronic and optic devices, such as transistors and solar cells. For these novel demands, the control of self-organization of this type of dye and the investigation of the relationship between the supramolecular structure and the relevant optical and electronic properties is of great importance. The objective of this thesis focuses on gaining a better understanding of structural and functional properties of pi-stacks based on self-assembling PBIs. Studies include the synthesis and characterization of new functional PBI dyes, their aggregation in solution, in liquid crystalline state and on surfaces, and their fluorescence and charge transport properties. An overview of the formation, thermodynamics and structures of pi-stacks of functional pi- conjugated molecules in solution and in liquid crystalline phases is given in Chapter 2. Chapters 3 and 4 deal with the pi-pi aggregates of new, highly fluorescent PBIs without core-substituents. In Chapter 3, the self-assembly of a PBI with tridodecylphenyl substituents at imide N atoms both in solution and condensed phase has been studied in great detail. In condensed state, the dye exhibits a hexagonal columnar liquid crystalline (LC) phase as confirmed by DSC, OPM and X-ray diffraction analysis. The columnar stacking of this dye has been further confirmed by atomic force microscopy (AFM) where single columns could be well resolved The charge transport properties this dye have been investigated by pulse radiolysis-time resolved microwave conductivity (PR-TRMC) measurements. To shed more light on the nature of the pi-pi interaction of the unsubstituted PBIs, solvent depend aggregation properties have been investigated in Chapter 4. The studies are further extended from core-unsubstituted PBIs to core-substituted ones (Chapter 5 and 6). In Chapter 5, a series of highly soluble and fluorescent core-twisted PBIs that bear the same trialkylphenyl groups at the imide positions but different bay-substituents and were synthesized. These compounds are characterized by distortions of the perylene planes with dihedral angles in the range of 15-37° according to crystallographic data and molecular modeling studies. In contrast to the extended oligomeric aggregates formed for planar unsubstituted PBIs, this family of dyes formed discrete pi-pi-stacked dimers in apolar methylcyclohexane as concentration-dependent UV/Vis measurements and VPO analysis revealed. The Gibbs free energy of dimerization can be correlated with the twist angles of the dyes linearly. In condensed state, several of these PBIs form luminescent rectangular or hexagonal columnar liquid crystalline phases with low isotropization temperatures. The core-twisting effect on semiconducting properties has been examined in Chapter 6. In this chapter, a comparative study of the electrochemical and the charge transport properties of a series of non-substituted and chlorine-functionalized PBIs was performed. While Chapters 3-6 focus on one-component dye systems, Chapter 7 explored the possibility of a supramolecular engineering of co-aggregates formed by hydrogen-bonded 2:1 and 1:1 complex of oligo(p-phenylene vinylene)s (OPVs) and PBIs. Covalently linked donor-acceptor dye arrays have been prepared for comparison. Concentration and temperature-dependent UV/Vis spectroscopy revealed all hydrogen-bonded and covalent systems form well-ordered J-type aggregates in methylcyclohexane. With these hydrogen-bonded OPV-PBI complexes, fibers containing p-type and n-type molecules can be prepared on the nano-scale (1-20 nm). For the 2:1 OPV-PBI hydrogenbonded arrays hierarchically assembled chiral superstructures consisting of left-handed helical pi-pi co-aggregates (CD spectroscopy) of the two dyes that further assemble into right-handed nanometer-scale supercoils in the solid state (AFM study) have been observed. All of these well-defined OPV-PBI assemblies presented here exhibit photoinduced electron transfer on sub-ps timescale, while the electron recombination differs for different systems.Thus, it was suggested that such assemblies of p- and n-type semiconductors might serve as valuable nanoscopic functional units for organic electronics.
Eine molekulare Fliegenfalle zur Erkennung von biologisch relevanten (poly)-anionischen Substraten

Im Rahmen dieser Arbeit konnte ein multivalenter Rezeptor auf Basis von Guanidiniocarbonylpyrrolen zur Komplexierung von biologisch relevanten (poly)- anionischen Substraten wie Citrat, Malat und Tatrat dargestellt werden. Der Rezeptor bindet Citrat selbst in Gegenwart eines 1000fachen Überschusses an NaCl und einem 10fachen Überschuss an Bis-tris Puffer mit einer hohen Bindungskonstante von KAss = 86000 M-1 in Wasser. Wenn man Rezeptoren auf Metall- und Boronsäurebasis nicht berücksichtigt, handelt es sich nach meinem Wissen um den besten Citrat-Rezeptor, der in der Literatur bisher publiziert ist. Außerdem zeigt der Rezeptor mit einem Faktor von mindestens 8 eine hohe Selektivität für Citrat gegenüber anderen biologisch relevanten Dicarboxylaten wie Malat und Tatrat. Mithilfe von Bindungsstudien und Molecular Modeling Rechnungen konnte hergeleitet werden, welchen Einfluss die verschiedenen nicht-kovalenten Wechselwirkungen auf die Bindungskonstante haben. Dabei konnte gezeigt werden, dass Flexibilität, Hydroxy-Funktionen, pi-Stacking und Symmetrie bei den Substraten Einfluss auf die Bindungskonstanten zeigen, wobei vor allem die unpolaren Wechselwirkungen und die zusätzliche Hydroxy-Funktionen einen hohen Anteil an der Bindung zu haben scheinen. Neben der selektiven Erkennung von Citrat durch den Rezeptor konnte zusätzlich ein Sensorsystem mit Carboxyfluorescein auf Basis eines indicator displacement assays entwickelt werden, mit dem die Anwesenheit von Citrat im Gegensatz zu anderen Carboxylaten mit dem bloßen Auge (naked eye) erkannt werden kann. Neben den Polycarboxylaten zeigt der Rezeptor außerdem noch hohe Bindungskonstanten für polyanionische Zucker. So konnten z.B. für Glucophosphate mit UV-Spektroskopie Bindungskonstanten von KAss = ca. 20000 M-1 in dem sehr polaren Lösemittelgemisch 10 % DMSO/Wasser (pH = 4, Acetat-Puffer) gefunden werden.
Electroactive Conjugated Polymers as Charge-Transport Materials for Optoelectronic Thin-Film Devices

In this work the electrochemical and spectroelectrochemical properties of a series of pi-conjugated organic polymers were studied. The polymers were deposited on platinum electrodes or ITO-coated glass substrates by potentiodynamic electro-polymerisation of the corresponding monomeric precursor molecules. The electro-chemical and photophysical properties of the triarylborane monomers were studied in detail in order to estimate possible influences on the behaviour of the corresponding polymer. The first part of this work aimed at the synthesis and investigation of conjugated donor–acceptor polymers which combine the prerequisites of an OLED within one material: the transport of positive and negative charges and the formation of emissive excited states. With the carbazole-substituted oxadiazoles 1–3 it was shown that on the one hand the carbazole functionality is suitable for enabling the electrochemical polymerisation of the monomers and on the other hand it facilitates reversible p-doping of the resultant polymers. Although n-doping of poly-1–poly-3 is possible due to the electron-deficient oxadiazole rings, it causes the continuous degradation of these electron-acceptor units. Interestingly, this process does not influence the capability of p-doping of the polymers. With respect to its electrochemical and spectroelectrochemical properties the behaviour of the borane polymer poly-4 is absolutely identical with that of the oxadiazole polymers. Moreover, the optical excitation of poly-4 in the solid state leads to the emission of blue-green light which suggests that this polymer might also possess electroluminescent properties. AFM-measurements of poly-4 films on ITO-coated glass substrates revealed, that the film thickness can be controlled to a certain extent by the number of polymerisation redox cycles. It was shown from the electrochemical and photophysical properties of the triarylboranes 4–6 that the pi–pi-interaction between boron and nitrogen atoms is comparably weak in these molecules. This leads to an unexpected ground-state polarisation with a partially positive boron atom and a partially negative nitrogen atom. Moreover, it was found that TAB 4 possesses a lower symmetry than D3 in solution and that excitation energy can be transferred amongst the three subchromophores of 4. By titration experiments it was also demonstrated that TAB 4 can reversibly bind fluoride ions and that the binding event significantly influences the optical absorption characteristics of the chromophore. It can be assumed, that the above mentioned properties, which have a profound influence on the photophysical behaviour of these triarylborane chromophores, also determine the behaviour of the corresponding polymer in a solid state environment. The aim of the second part of this work was the investigation of purely n-conducting materials based on electron-deficient borane and viologen polymers. The corresponding precursor molecules should be polymerised on platinum electrodes by reductive electropolymerisation. However, a reductive polymerisation was not possible for the borane monomer 19 which is thought to be due to a strong localisation of the unpaired electron on the central boron atom of the radical anion. An electropolymerisation of the cyano-substituted bispyridinio-compound 17 failed because of the poor quality of CN– as a leaving group. Thus, a synthesis of the analogous isomer 18 was developed, in which the cyano-substituents were exchanged by the better leaving group Cl–. The viologen polymer poly-18, which can be regarded as an electron-deficient iso-electronic analogue of poly(para-phenylene), was successfully deposited on a platinum electrode by reductive electropolymerisation of 18. Poly-18 can be reversibly n-doped at comparably low potentials; however, at higher potentials the polymer is overcharged and destroyed irreversibly. As the synthetic strategy for 18 allows the variation of both spacer unit and leaving group in the last two steps of the reaction sequence, a series of analogous compounds can be easily synthesised using this route.
