The present thesis demonstrates the importance of the solid state packing of dipolar merocyanine dyes with regard to charge transport and exciton coupling.
Due to the charge transport theory for disordered materials, it is expected that high ground state dipole moments in amorphous thin films lead to low mobility values due to a broadening of the density of states. However, due to their inherent dipolarity, merocyanine dyes usually align in antiparallel dimers in an ordered fashion. The examination of twenty different molecules with ground state dipole moments up to 15.0 D shows that by a high dipolarity and well-defined sterics, the molecules pack in a highly regular two-dimensional brickwork-type structure, which is beneficial for hole transport. Utilization of these molecules for organic thin-film transistors (OTFTs) leads to hole mobility values up to 0.21 cm²/Vs. By fabrication of single crystal field-effect transistors (SCFETs) for the derivative showing the highest mobility values in OTFTs, even hole mobilities up to 2.34 cm²/Vs are achieved. Hence, merocyanine based transistors show hole mobility values comparable to those of conventional p-type organic semiconductors and therefore high ground state dipole moments are not necessarily disadvantageous regarding high mobility applications.
By examination of a different series of ten merocyanine dyes with the same chromophore backbone but different donor substituents, it is demonstrated that the size of the donor has a significant influence on the optical properties of thin films. For small and rigid donor substituents, a hypsochromic shift of the absorption compared to the monomer absorption in solution is observed due to the card stack like packing of the molecules in the solid state. By utilization of sterical demanding or flexible donor substituents, a zig-zag type packing is observed, leading to a bathochromical shift of the absorption. These packing motifs and spectral shifts with an offset of 0.93 eV of the H- and J-bands comply with the archetype examples of H- and J-aggregates from Kasha’s exciton theory.
In der vorliegenden Doktorarbeit konnte gezeigt werden, dass eine starke Exzitonenkopplung nicht nur zwischen gleichen Chromophoren, sondern auch zwischen Chromophoren mit unterschiedlichen Energien der angeregten Zustände möglich ist. Diese beeinflusst maßgeblich die Absorptionsspektren der Heterostapel bestehend aus Merocyanin- bzw. Perylenbisimidfarbstoffen und deutet außerdem auf einen kohärenten Energientransfer zwischen den Chromophoren hin. Weiterhin wurden Bis(merocyanin)-C60-Konjugate synthetisiert, die in unpolaren Lösungsmitteln selbst assemblieren und auf diese Weise wohldefinierte supramolekulare p/n-Heterogrenzflächen gebildet werden. An diesen wurde mithilfe von femtosekundenaufgelöster transienter Absorptionsspektroskopie der photoinduzierte Elektronentransfer untersucht, was ein wichtiger Schritt bei der Erzeugung von Ladungsträgern in organischen Solarzellen darstellt.
Im Rahmen der vorliegenden Arbeit wurde eine Serie von dodekatopischen [60]Fullerenhexakisaddukten, die mit zwölf Carbonsäuregruppen dekoriert sind, auf ihre Eigenschaften hin untersucht, ausgedehnte, kristalline Polymernetzwerke mit einer eventuellen Porosität darzustellen. Hierbei wurden die Fähigkeiten der synthetisierten Dodekasäuren ausgenutzt über Wasserstoffbrückenbindungen und Metallkoordinationen supramolekulare Kontakte auszubilden und ausgedehnte Netzwerke zu knüpfen.
In Kapitel 2 werden zunächst die grundlegenden physikalischen und chemischen Eigenschaften des sphärischen [60]Fullerenmoleküls, als Ausgangsverbindung für die Darstellung der supramolekularen Bausteine, vorgestellt. Insbesondere wird die chemische Funktionalisierbarkeit von C60 in verschiedenen Reaktionstypen unter Einbeziehung der selektiven, multiplen Funktionalisierbarkeit und der Fähigkeit Th-symmetrische Hexakisaddukte auszubilden, beschrieben. Danach folgt in dem Unterkapitel 2.5 ein kurzer Literaturüberblick über das intermolekulare Vernetzen von C60 und dessen Derivaten zu größeren Molekülverbänden und polymeren Strukturen mit besonderem Augenmerk auf metallorganische Hybridarchitekturen, die aus funktionalisierten Fullerenen und Metallionen oder Metallclustern aufgebaut sind.
Die Synthese der vier dodekatopischen, Th-symmetrischen [60]Fullerenhexakis-addukte C2-H, C3-H, C4-H und C5-H mit unterschiedlich langen Alkylketten in den Seitenarmen wird in Kapitel 4.1 beschrieben. Der Strukturtyp ist in Abbildung 114 gezeigt. Auszugsweise wird hier auch die Identifizierung der Moleküle und Kontrolle ihrer Reinheit mittels spektroskopischer Methoden vorgestellt.
In Kapitel 4.2 wird die Darstellung von Wasserstoffbrückenbindungsnetzwerken aus
den synthetisierten Dodekasäuren beschrieben und deren erhaltenen Einkristallstrukturdaten
diskutiert. Das Unterkapitel 4.2.2 beschäftigt sich zusätzlich mit der
Kristallstruktur eines VAN-DER-WAALS-Netzwerkes des Dodekasäuremethylester C2-
Me, welcher in situ bei Kristallisationsversuchen von C2-H erhalten wurde.
Ein Vergleich der supramolekularen Netzwerke untereinander zeigt, dass das
Packungsverhalten der Fullerenderivate, trotz Interaktion der Carbonsäuren mit
benachbarten Fullerenbausteinen und Lösungsmittelmolekülen, maßgeblich von den
großen, sphärischen Fullerengrundkörpern bestimmt wird. Die erhaltenen Netzwerke
weisen dabei alle kubisch-dichteste ABC-Packungsmuster auf, wie es auch bei reinem
C60
[244] im Festkörper oder bei den Fulleriden[214] beobachtet wird. Die unterschiedlich
langen Seitenarme bestimmen dabei lediglich die Dimensionen der Packungen, eine
mögliche Verzerrung, sowie die Ausprägungen der entstehenden Tetraeder- und
Oktaederlücken. Im Fall von C4-H richten die Wasserstoffbrückenbindungen der
Carbonsäuregruppen die Seitenarme aus und bilden somit ein geordnetes, poröses
Netzwerk aus. In den supramolekularen Netzwerken wird überwiegend die
Raumgruppe R 3 ̅
beobachtet, außer für C3-H, bei der die kritische Länge der
Seitenketten, mit der Raumgruppe P1 ̅
, eine geringere Symmetrie erzwingt. Alle
dargestellten supramolekularen Netzwerke sind in Abbildung 115 zusammengefasst.
Obwohl die Anzahl der Säuregruppen in den Bausteinen jeweils gleich ist, wird in jedem Wasserstoffbrückennetzwerk ein eigener Typus an verknüpfenden H-Brückenbindungsclustern beobachtet. Bei C2-H erfolgt die Knüpfungsbindung durch die Bereitstellung und Auffüllung von hydrophilen und hydrophoben Taschen, wobei die Distanz zwischen den Säuregruppen durch die Interkalation von Lösungsmittelmolekülen überbrückt wird. In C3-H führt die dreidimensionale Vernetzung über „S“-förmige Säuredimere. Und bei C4-H handelt es sich um zwei interpenetrierende Teilgitter, bei der zwei helikale H-Brückennetzwerke ineinander verschachtelt sind. Gemäß der „goldenen Regel“ des Kristalldesigns[212] (siehe Kapitel 4.2) maximieren die Netzwerke die Anzahl der klassischen Säuredimere mit dem steigenden Grad der geometrischen Flexibilität der Seitenarme. Bei C2-H sind die Arme noch zu kurz, so dass die Verknüpfung über H-Brückencluster verläuft. C3-H bildet mit acht Armen Säuredimere aus und C4-H verwendet alle Seitenarme für die
Ausbildung von Säuredimeren. Der Vergleich des raumausfüllenden VAN-DER-WAALS-Netzwerkes von C2-Me mit dem H-Brückennetzwerk von C2-H legt zudem nahe, dass die Ausprägung von Hohlräumen ein Effekt der gerichteten Wasserstoffbrückenbindungen sein muss.
Aktivierungsversuche der Porenstruktur des H-Brückennetzwerks von C4-H und die Bestimmung der inneren Oberfläche durch Gasadsorption runden das Kapitel ab. Die innere Oberfläche konnte auf 40 m2g–1 für die BET-Adsorptionsisotherme mit Stickstoff bestimmt werden. Durch den Vergleich der Pulverdiffraktogramme vor und nach der Aktivierung konnte eine Phasenumwandlung festgestellt werden, die ein Kollabieren der Poren nahelegt.
Die Implementierung von Metallen und Metallclustern in die Netzwerkstrukturen der Dodekasäuren wird im Kapitel 4.3 beschrieben. Hier konnte durch den Einbau von Zinkoxid-cluster in die Netzwerke von C2-H und C3-H die Hypothese eines „inversen MOFs“ aufgestellt werden. Da sich die Zinkoxid-Cluster formal in die vorhandenen H-Brückencluster der Fullerennetzwerke implementieren ließen, ohne dass sich das Packungsverhalten der Fullerengrundkörper wesentlich veränderte, kann geschlussfolgert werden, dass die strukturdirigierende Wirkung nicht wie in der klassischen MOF-Chemie üblich vom Metall, sondern vom organischen Bestandteil ausgeht. Das heißt Metall und Ligand tauschen hier ihre Funktionalität in Bezug auf ihre strukturdirigierende Wirkung. Die Zink-Fullerennetzwerke sind in Abbildung 116 dargestellt.
Das Prinzip des „inversen MOFs“ ist jedoch nicht auf die Metallfullerennetzwerke CdC2 und CdC4 übertragbar. Die Struktur wird hier durch hohe Bereitschaft von Cadmium mit den Carbonsäuregruppen Komplexe zu bilden dominiert. Cadmium bildet „zick-zack“-förmige, lineare Metallstränge aus, an denen die Seitenarme der Fullerenbausteine über Koordination mit den Carbonsäuregruppen aufgespannt werden. In Abbildung 117 sind die beiden erhaltenen, porösen Cadmium-Netzwerke dargestellt.
Im Netzwerk von CuC2, das in Abbildung 118 gezeigt ist, kann die strukturdirigierende Wirkung weder dem Metall, noch der Dodekasäure zugesprochen werden. Es kommt zur Ausbildung von zweidimensionalen metallorganischen Polymeren, indem je vier Fullerenbausteine über ein Kupferdimer koordiniert werden. Die Koordination von zwei weiteren Kupferionen, die jeweils endständig das Dimer zu einem Tetramer erweitern, führen zu einer vollständigen Inklusion der Metallionen in das Carbonsäurenetzwerk. Die freien Koordinationsstellen an den Kupferionen sind mit Wassermolekülen abgesättigt. Daraus resultiert die Ausbildung von Wasserstoffbrückenbindungen zwischen Wasser und den Carboxylgruppen der Seitenarme und somit die Ausbildung eines dreidimensionalen Netzwerkes mit einer sehr effektiven Raumausfüllung.

Das Metallfullerennetzwerk CaC2, das am Ende von Kapitel 4.3 behandelt wird, stellt einen Grenzfall zwischen H-Brücken- und Metallfullerennetzwerk dar. Die Struktur ist in Abbildung 119 gezeigt. Sie weist bezüglich der Clusterbildungen in den Oktaeder- und Tetraederlücken viele Parallelen zu den zinkhaltigen Netzwerken ZnC2 und ZnC3 auf. Die Clusterbildung von Kalzium erfolgt jedoch nur in jeder zweiten Oktaederlücke und die entstehenden Tetraederlücken werden, wie in dem H-Brückennetzwerk von C2-H von drei Carbonsäuren aus der oberen Schicht gefüllt. Die jeweils andere Oktaederlücke bleibt hingegen frei und schließt einen Hohlraum ein. Zudem ist CaC2 ein Hybridnetzwerk, da jeweils zwei Schichten zu einer metallorganischen Doppelschicht verknüpft sind und die Doppelschichten untereinander über Wasserstoffbrückenbindungen miteinander verbunden sind. Dabei entsteht die Koordination der Säuregruppen in hydrophilen Taschen, analog zum H-Brückennetzwerk von C2-H.
Die erhaltenen Metallfullerennetzwerke wurden jeweils durch Pulverdiffraktometrie-untersuchungen in verschiedenen Aktivierungsversuchen untersucht. Die Netzwerke ZnC2 und CaC2 haben keine sinnvoll auswertbaren BET-Adsorptionsisothermen gezeigt. Von ZnC3 konnte eine geringe innere Oberfläche von 25 m2g–1, bei CdC2 30 m2g–1 und bei CdC4 29 m2g–1 bestimmt werden. Größere innere Oberflächen mit stabileren Porositäten können vermutlich dann erhalten werden, wenn eine Möglichkeit gefunden wird Fullerenhexakisaddukte mit rigideren Seitenarmen zu synthetisieren.
Trotz des starken, multivalenten Einflusses der zwölf Säuregruppen und ihrer Ausbildung von Wasserstoffbrückenbindungs- und Metallcluster, konnte beobachtet werden, dass die strukturdirigierende Wirkung in den Netzwerken von C2-H, C3-H, C4-H, ZnC2, ZnC3 und CaC2 durch die Ausbildung eines jeweils kubisch dichtesten ABC-Packungsmusters vom nanoskaligen, sphärischen Fullerengerüst ausgeht.
Es konnten in der vorliegenden Arbeit neue, vielseitige molekulare Bausteine für den Aufbau von dreidimensional vernetzten, kristallinen Strukturen entwickelt werden. Mit Hilfe dieser Bausteine konnten, in ihrer Komplexizität und ihrem Vernetzungsgrad einzigartige, Wasserstoffbrückennetzwerke im Einkristall untersucht werden. Durch den Einbau der oktaedrischen Bausteine in Metallfullerennetzwerke gelang hier zum ersten Mal die Implementierung von [6:0]Hexakisaddukten bei denen die isotrope, sphärische Funktionalisierung effizient für eine echte, dreidimensionale Vernetzung der Fullerengrundkörper genutzt wurde. Die wenigen bekannten fullerenhaltigen MOFs beinhalteten bisher Hexakisaddukte lediglich als lineare Linker oder waren, wie bei {[Cd(36)2](NO3)2}∞, lediglich zweidimensional verknüpft. Die
neuen, außergewöhnlichen Strukturen der Metallfullerennetzwerke wurden beschrieben und diskutiert. Die Verwendung der Dodekasäuren als dodekatopische Linkermoleküle führte zusätzlich zu einer Ausweitung der Topizitätspalette in der MOF-Synthese, bei der bisher in der Literatur lediglich Linkermoleküle mit einer maximal oktatopischen[200] Qualität zum Einsatz kamen. Zusätzlich konnte der Begriff des „inversen MOFs“ eingeführt werden, bei dem der strukturdirigierende Einfluss vom organischen Baustein ausgeht und dadurch organischer Linker und anorganisches Koordinationszentrum ihre Funktion in der klassischen MOF-Synthese tauschen.
Merocyanine Dyes as Organic Semiconductors for Vacuum-processed Solar Cell and Transistor Devices

The present thesis comprises the synthesis of new functional merocyanine dyes, the study of their electro-optical properties as well as solid state packing and their application as p-type semiconductor materials in transistor and solar cell devices. The absorption properties of the obtained compounds could be modified by variation of the donor unit, the introduction of electron-withdrawing substituents in the acceptor unit or elongation of the polymethine chain. For a particular dye, the absorption band could be shifted by more than 160 nm by increasing the solvent polarity due to a conformational switch between a merocyanine-like and a cyanine-like structure. Single crystal analyses revealed that the studied dyes tend to pack either in an antiparallel fashion forming dimers with no overall dipole moment or in a staircase-like pattern where the dipole moments point to the same direction and are only balanced by another staircase oriented in the opposite direction (stair dimer). With respect to application as semiconductor materials, the latter packing arrangement resulted most favorable for charge carrier mobility. We concluded that this packing motif is preserved in the solar cell devices, where the selenium-containing dye afforded the highest performance of this series for an optimized planar-mixed heterojunction solar cell (6.2 %).
The thesis discusses aspects of the photocatalytic water oxidation reaction. The first chapter deals with a supramolecular macrocycle which contains three ruthenium metal centers. This novel catalyst shows promising catalytic activity and provides insides into the mechanism of the water oxidation reaction. After this part, the focus lies on the light interacting components of the photocatalytic water oxidation. In this regard, the azabenz-annulated perylene derivatives appeared to be a promising dye class. The combination of these chromophores and metal complexes result in metal organic compounds, which have photosensitizer potential.
Die Selbstorganisation von amphiphilen Molekülen zählt zu den am intensivsten bearbeiteten Forschungsgebieten der Supramolekularen Chemie. Die faszinierenden supramolekularen Architekturen der Natur zeigen eindrucksvoll, wie neuartige Funktionen durch das Zusammenspiel wohl-definierter Molekülensembles in einer wässrigen Umgebung entstehen. Es ist bekannt, dass der hydrophobe Effekt dabei eine entscheidende Rolle in der Selbstorganisation spielt und somit die Funktion eines Systems wesentlich bestimmt. Obwohl die Komplexität der bekannten Beispiele aus der Natur unerreicht ist, wurden in den letzten Jahren unzählige künstliche supramolekulare Architekturen basierend auf amphiphilen Molekülen erschaffen, mit vielfältigen Anwendungsmöglichkeiten an der Schnittstelle von Chemie, Biologie und Physik.
Darunter eröffnen insbesondere amphiphile π-konjugierte Systeme einen einfachen Zugang zu vielfältigen Strukturen, da im wässrigen Medium starke π-π-Wechselwirkungen als strukturbildendes Element ausgenutzt werden können. Vor allem wegen vielversprechender Anwendungsmöglichkeiten spiegelt sich die Selbstorganisation solcher Systeme in einem hohen Forschungsinteresse wider. Dennoch ist das Wirken des hydrophoben Effekts in der Selbstassemblierung amphiphiler π-konjugierter Moleküle weitgehend unverstanden.
Die vorliegende Arbeit befasste sich daher mit der Frage, welche physikochemischen Grundprinzipien die Bildung von supramolekularen Polymeren basierend auf amphiphilen π-konjugierten Molekülen in Wasser steuern und wie der hydrophobe Effekt die Funktionalität solcher Strukturen beeinflusst. Bolaamphiphile Perylenbisimide (PBIs) und Naphthalinbisimide (NBIs) erwiesen sich dabei für das Moleküldesign als besonders geeignet, weil sie vergleichsweise einfach dargestellt werden können, ihre hohe Symmetrie weniger komplexe Assoziationsprozesse begründet und die Untersuchung ihrer Selbstassemblierung im wässrigen Medium weiterhin einen Vergleich erlaubt, wie sich der hydrophobe Effekt bezüglich unterschiedlich großen π-Systemen auswirkt.
Es konnte gezeigt werden, dass OEG-basierte Rylenbisimide unter bestimmten strukturgeometrischen Voraussetzungen eine entropiegetriebene Aggregation aufweisen, wenn die Freisetzung von Wassermolekülen aus einer wohl-definierten Hydrathülle ausreicht, um den Enthalpiegewinn aus den im wässrigen Medium verstärkten Dispersionswechselwirkungen zwischen den π-Flächen zu übertreffen. Im vorliegenden Fall wurde dies durch das Pinsel-Strukturmotiv der symmetrisch angebrachten Imidsubstituenten erreicht, für die sich damit einhergehend eine gänzlich andere Temperaturabhängigkeit beobachten ließ und somit eine gänzlich andere Funktionalität, als man sie aus organischen Lösungsmitteln kennt. Wasser als Lösungsmittel führt also nicht nur zu einer signifikanten Bindungsverstärkung, sondern öffnet Zugang zu supramolekularen Systemen mit neuartigen Funktionen. Die entropiegetriebene Freisetzung von Wassermolekülen konnte daher im Rahmen dieser Arbeit ausgenutzt werden, um gleichzeitig die intrinsische Ordnung im π-Stapel von PBIs zu erhöhen, was anhand eines temperatursensorischen Hydrogels anschaulich demonstriert wurde.
Darüber hinaus stellte sich heraus, dass Wasser ein geeignetes Lösungsmittel zur Darstellung supramolekularer Kompositmaterialien ist. Wie anhand sich instantan bildender Co-Aggregate gezeigt wurde, ist die entropiegetriebene Assemblierung der entscheidende Faktor zur Darstellung von komplexeren supramolekularen Strukturen, die überdies einen Schritt hin zu den hochkomplexen multimolekularen Anordnungen der Natur darstellen.
Tetrapyrrole sind nicht nur in natürlichen Systemen von herausragender Bedeutung, sondern haben sich im letzten Jahrhundert zu einem zentralen Forschungsgegenstand verschiedener Naturwissenschaften entwickelt. Chirale Vertreter sind, trotz ihrer Bedeutung in der Natur, hierbei nur am Rande untersucht worden.
Im Rahmen der vorliegenden Arbeit wurde, ausgehend von p-Xylylen-verbrückten Porphyrinen, sogenannten Henkelporphyrinen, eine Vielzahl an chiralen Strukturen synthetisiert und stereochemisch untersucht.
Die monomeren Vertreter waren abhängig vom Substitutionsmuster der Henkeleinheit achiral oder planar-chiral. Über eine kurze und effiziente Syntheseroute wurden die Grundkörper im Multigramm-Maßstab aufgebaut und anschließend sowohl strukturell modifiziert als auch funktionalisiert. Die chiralen Vertreter wurden mittels HPLC an chiraler Phase in die Enantiomere getrennt und mittels online-ECD-Spektroskopie wurden die ECD-Spektren im stopped-flow-Modus gemessen. Der Vergleich mit quantenchemisch berechneten ECD-Kurven erlaubte die sichere Zuordnung der Absolutkonfigurationen. Des Weiteren wurden die Monomere röntgenkristallographisch, UV-Vis-spektroskopisch und elektrochemisch eingehend untersucht.
Ausgehend von den monomeren Vertretern wurden mittels direkter oxidativer Kupplung oder über Übergangsmetall-katalysierte Verfahren di- und trimere Systeme aufgebaut. Diese lagen als axial-, helikal- oder planar-chirale Strukturen vor. HPLC-ECD-Kopplung in Verbindung mit quantenchemischen Rechnungen erlaubte auch hier die Aufklärung der absoluten Stereostrukturen.
Neben klassischen Porphyrin-Vertretern wurden im Rahmen dieser Arbeit erstmals Henkelchlorine durch OsO4-vermittelte Dihydroxylierung der Henkelporphyrine synthetisiert und vollständig charakterisiert. Außerdem gelang in Kooperation mit Prof. Brückner (UConn) die Aufklärung der Absolutkonfigurationen helikal-chiraler Indachlorine.
Die Arbeit liefert insgesamt eine breite Plattform an chiralen Porphyrinoiden, die für weitere Untersuchungen zu Reaktivität und Struktur von größtem Interesse sind, sowie erste detaillierte Einblicke in die faszinierenden Eigenschaften dieser Substanzklasse.
The self-assembly of molecules based on π-π-interactions and hydrogen bonding is of significant importance in nature. These processes enable the formation of complex supramolecular structures with diverse functions. For the transfer of the concepts from nature to artificial supramolecular structures, a basic understanding of those processes is needed. For this purpose, π-conjugated aromatic molecules with an easy synthetic access are suitable as their functionalities can be changed effortless. Perylene bisimide (PBIs) dyes are attractive candidates since they fulfill these requirements owing to their tendency to self-assemble in solution due to their large aromatic π-surfaces. Furthermore, the changes of the optical properties (for instance absorption, emission or circular dichroism) of PBI dyes, caused by their self-assembly, are easy to study experimentally. Structural variations of PBI dyes including additional non-covalent interactions, such as hydro-gen bonding, enable to direct their self-assembly process. Thus, the formation of interesting su-pramolecular structures of PBI dyes could be realized, although, often of undefined size. The aim of this thesis was to develop strategies to restrict the aggregate size of PBI dyes. Therefore, de-fined structural features of PBI molecules were combined and a variation of external influences such as solvent and concentration included. Furthermore, DNA was utilized as a template for the limitation of the aggregate size of PBI dyes.
Chapters 1 and 2 provide general information and describe examples from literature which are necessary to understand the following experimental work. The first chapter is based on the inter-actions of various molecules with DNA. Therefore, DNA is considered as a supramolecular biom-acromolecule containing specific structural and functional features to interact with small mole-cules. Afterwards, the main interaction modes of small molecules with DNA such as electrostatic interaction, intercalation and groove binding with corresponding examples are discussed. Among all techniques applied to study the interaction of ligands with DNA, UV/Vis absorption, fluores-cence and circular dichroism spectroscopy were described in detail. At the end of this chapter, examples of already pre-associated systems showing interactions with DNA are presented.
The second chapter is focused on the determination and mathematic evaluation of the self-assembly processes. The simplest models such as monomer-dimer and isodesmic model are de-scribed and supplemented by examples. Furthermore, the simplest modification of the isodesmic model, the K2-K model, is presented. Additionally, experimental problems, which may arise dur-ing the investigations of the self-assembly processes, are addressed. For the description of the entire self-assembly process, a sufficiently large concentration range and an appropriate measure-ment method that is sensitive in this concentration range is necessary. Furthermore, the full transi-tion from the monomeric to the aggregated species has to be spectroscopically ascertainable. This enables an accurate mathematic evaluation of the self-assembly process and provides meaningful binding constants. The self-assembly pathway can be controlled by the variation of solvent, con-centration or temperature. However, this pathway can also be directed by a rational design of the molecular structure of the considered system. For example, a specific interplay of π-π-interactions and hydrogen bonding may promote isodesmic as well as cooperative growth into large struc-tures.
The main focus of this thesis is to develop strategies to control the aggregate size of PBI dyes (Chapter 3). For this purpose, a PBI scaffold was designed which contains hydrogen bonding amide functions at the imide positions derived from the amino acid L-alanine and solubilizing side groups in the periphery (Figure 81). The variations of the residues R/R’ range from didodecylox-yphenyl, didodecylphenyl, dioligo(ethylene glycol)phenyl to branched and linear alkyl chains.
The most extensive study of the aggregation behavior was performed for the PBI dye 5. Concen-tration-dependent 1H NMR and UV/Vis absorption measurements clearly revealed the formation of dimers in chloroform. Further investigations by means of 2D NMR, VPO and ITC confirmed the exclusive presence of dimer aggregates of PBI 5 in the investigated concentration range. Mo-lecular modelling studies, supported by NMR and FT-IR experiments, provided structural reasons for the absence of further growth into larger aggregates. The specific combination of π-π interac-tions and hydrogen bonds between the NH groups of the amide groups and the carbonyl oxygen atoms of the PBI core are decisive for the formation of the discrete dimer stack (see Figure 82). The investigations of the aggregation behavior of PBIs 6-9 were less extensive but consistent with the results obtained for PBI 5. However, the determined binding constants vary over a considera-ble range of 1.1 x 102 M-1 (PBI 8) to 1.4 x 104 M-1 (PBI 5). These differences could be attributed to structural variations of the dyes. The electron-rich phenyl substituent promoted the aggregation tendency of PBIs 5-7 compared with 8 and 9 that carry only alkyl side chains. Thus, the π-π in-teractions of bay-unsubstituted PBI cores in combination with hydrogen bonding of the amide functions control the formation of discrete dimers of these PBI dyes.
The variation of conditions, such as solvent, change the aggregation behavior of PBI dyes. In the solvents toluene and/or methylcyclohexane, anti-cooperative growth into larger aggregates of PBI 5 was observed (Chapter 4). The important feature of this self-assembly process is the absence of isosbestic points over the whole concentration range in the UV/Vis absorption measurements. The preference for the dimeric species of PBI 5 remained in both solvents as well as in mixtures of them, but upon increasing the concentration these dimers self-assemble into larger aggregates.
An important feature of the self-assembly process is the preferred formation of even-numbered aggregates compared to the odd-numbered ones (see Figure 83). Although, the conventional K2-K model provides plausible binding constants, it is not capable to describe the aggregation behavior adequately, since it considers a continuous size distribution. The gradual aggregation process over dimers, tetramers, hexamers, etc. was therefore analyzed with a newly developed K2-K model for anti-cooperative supramolecular polymerization. By the global analysis of the UV/Vis absorption spectra a very good agreement between the experimental and simulated spectra, which were based on the new K2-K model, was obtained. Furthermore, the calculated UV/Vis absorption spectra of a dimer and an aggregate highlighted the most important structural differences. The absorption spectrum of the dimer still has a pronounced vibronic structure which gets lost in the spectrum of the aggregate.
In another part of this work, a series of water soluble PBI dyes were described which contain similar PBI scaffolds as PBIs 5-8 (Chapter 5). These PBI dyes self-assemble into similar dimer aggregates in water due to their positively charged side chains causing electrostatic repulsion be-tween the molecules (see Figure 84). Here, however, the self-assembly behavior has not been studied thoroughly in water due to the similarities of already reported PBI dyes.
Instead, the focus here is on the characterization of the interactions of these dyes with DNA/RNA. The comprehensive studies using thermal denaturation experiments showed the high stability of these PBI/polynucleotide complexes. The spermine-functionalized PBI dyes having six positive charges showed strong interactions with DNA/RNA which was expressed in a signif-icant increase of the melting temperatures of DNA/RNA (ΔTm values between 7 and > 35 ° C). The dioxa analogues containing only two positive charges had lower enhancement of the melting temperature of DNA/RNA (ΔTm values between 3 and 30 ° C). A similar trend has been observed in the fluorimetric titrations. The spermine-functionalized PBI dyes showed high binding con-stants (log Ks = 9.2 - 9.8), independently of the used polynucleotides. In contrast, the dioxa ana-logues displayed smaller binding constants (log Ks = 6.5 - 7.9) without any correlation between binding affinity and binding strength of the PBI dyes and the applied polynucleotides. The CD-spectroscopic measurements revealed significant differences in the binding properties of the dyes with DNA/RNA. They were dependent on the steric hindrance of the amino acid residues at the imide position and their configuration on one side and the grooves properties of ds-DNA/RNA on the other side. The spectroscopic results confirmed the formation of excitonically coupled PBI dimers in the minor groove of ds-DNA and the major groove of ds-RNA. Depending on the se-quence, the grooves of the polynucleotides provide different amount of space for embedding molecules. The guanine amino groups protrude into the minor groove of the polynucleotide poly(dG-dC)2 increasing the steric hindrance, which is not the case for poly(dA-dT)2. Molecular modeling studies showed that the PBI dimers penetrate deeper into the groove of poly(dA-dT)2 due to the absence of the steric hindrance, in comparison to the groove of poly(dG-dC)2 (see Figure 85).
In this work the successful synthesis, the linear and nonlinear spectroscopic properties as well as the electrochemical behaviour of some linear and star-shaped squaraine superchromophores that are based on indolenine derivatives were presented. The attempt to synthesise similar chromophores which contained only benzothiazole squaraines failed unfortunately. However, one trimer that contained mixed benzothiazole indolenine squaraines could be synthesised and investigated as well.
The linear spectroscopic properties, like red-shift and broadening of the absorption, of all superchromophores could be explained by exciton coupling theory. The heterochromophores (SQA)2(SQB)-N, (SQA)(SQB)2-N and (SQA)(SQB)-NH displayed additional to the typical squaraine fluorescence from the lowest excited state some properties that could be assigned to localised states. While the chromophores with N-core showed very small emission quantum yields, the chromophores with the other cores and the linear oligomers display an enhancement compared to the monomers.
Transient absorption spectroscopy experiments of the star-shaped superchromophores showed, that their formally degenerated S1 states are split due to a deviation of the ideal C3 symmetry. This is also the reason for the observation of an absorption band for the highest exciton state, which is derived from the S1-state of the monomers, as its transition-dipole moment would be zero in the symmetrical case.
The linear oligomers and the star-shaped superchromophores with a benzene or triarylamine core showed at least additive, sometimes even weak cooperative, behaviour in the two-photon absorption experiments. Additional to higher two-photon absorption cross sections the chromophores showed a pronounced broadening of the nonlinear absorption, due to symmetry breaking and a higher density of states.
Unfortunately it was not possible to solve the problem of the equilibrium of the cisoid and the transoid structure of donor substituted azulene squaraines, due to either instability of the squaraines or steric hindrance.
Galectin-1 (hGal-1) is overexpressed by numerous cancer types and previously conducted studies confirmed that the β-galactoside-binding protein mediates various molecular interactions associated with tumor growth, spread and survival. Upon interaction with carbohydrate-based binding epitopes of glycan structures on human cell surfaces galectin-1 induces proliferative, angiogenetic and migratory signals and modulates negative T cell regulation which essentially helps the tumor to evade the immune response. These findings attributed galectin-1 a pivotal role in tumor physiology and strongly suggest the protein as target for diagnostic and therapeutic applications.
Within the scope of this work a strategy was elaborated for designing tailor-made galectin-1 ligands by functionalizing selected hydroxyl groups of the natural binding partner N-acetyllactosamine (LacNAc) that are not involved in the sophisticated interplay between the disaccharide and the protein. Synthetic modifications intended to introduce chemical groups i) to address a potential binding site adjacent to the carbohydrate recognition domain (CRD) with extended hGal-1-ligand interactions, ii) to implement a tracer isotope for diagnostic detection and iii) to install a linker unit for immobilization on microarrays.
Resulting structures were investigated regarding their targeting ability towards galectin-1 by cocrystallization experiments, SPR and ITC studies. Potent binders were further probed for their diagnostic potential to trace elevated galectin-1 levels in microarray experiments and for an application in positron emission tomography (PET).
The presented work in the field of supramolecular chemistry describes the synthesis and detailed investigation of (bi)pyridine-based oligo(phenylene ethynylene) (OPE) amphiphiles, decorated with terminal glycol chains. The metal-ligating property of these molecules could be exploited to coordinate to Pd(II) and Pt(II) metal ions, respectively, resulting in the creation of novel metallosupramolecular π-amphiphiles of square-planar geometry.
The focus of the presented studies is on the self-assembly behaviour of the OPE ligands and their corresponding metal complexes in polar and aqueous environment. In this way, the underlying aggregation mechanism (isodesmic or cooperative) is revealed and the influence of various factors on the self-assembly process in supramolecular systems is elucidated. In this regard, the effect of the molecular design of the ligand, the coordination to a metal centre as well as the surrounding medium, the pH value and temperature is investigated.
Die vorliegende Arbeit behandelt die Synthese sowie die Eigenschaften einer Serie von organofunktionellen α-, β-, γ- und ζ-Silanen, mit einem Fokus auf Alkoxy(aminoalkyl)silanen. Der Großteil dieser Modellstrukturen wurde anschließend hinsichtlich ihrer Hydrolysekinetik in Abhängigkeit der Art der funktionellen Gruppe X (NMe3+, N(H)COOMe, N(Me)COOMe, NH2, N(H)Me, NMe2, Pip, Me), des Abstandes des Substituenten X zu dem Silicium-Atom
(α-, β-, γ- und ζ-Position), der Alkoxy-Abgangsgruppe am Silicium-Atom (MeO, iPrO, tBuO) und des pD-Wertes der Reaktionslösung systematisch untersucht.
Eine große Herausforderung dieser Studie war die Synthese von β-Amino-funktionalisierten Alkoxysilanen, deren Chemie aufgrund ihrer Labilität bisher kaum erforscht ist. Die einzigen literaturbekannten Vertreter stellten bislang das Trialkoxysilan (EtO)3Si(CH2)2NH2 (1) und sein Dialkoxy-Derivat (EtO)2SiMe(CH2)2NH2 (2) dar, welche durch Reaktion des entsprechenden 2-(Chlorethyl)silans mit Ammoniak unter hohem Druck im Autoklaven zugänglich sind. Unter Verwendung dieser Synthesemethode konnte sowohl die Synthese der Silane 1 und 2 reproduziert, als auch das Trimethoxy-Analogon (MeO)3Si(CH2)2NH2 (3) erstmals dargestellt werden.
Darüber hinaus wurde eine Serie von organofunktionellen Monoalkoxysilanen des Typs RORSiMe(CH2)2X und ROSiMe2C(H)MeCH2X (4b–18b) im präparativen Maßstab analyserein dargestellt.
Des Weiteren wurden die entsprechenden α-Silane 8a, 11a, 14a und 15a, die γ-Silane 6c, 8c, 11c, 13c–15c und 18c sowie die ζ-Silane 19 und 20 erstmals dargestellt.
Weiterhin wurden die bereits literaturbekannten α-Silane 16a–18a und γ-Silane 7c, 16c und 17c für die Verwendung in den Hydrolyseexperimenten synthetisiert.
Die Charakterisierung aller im Rahmen dieser Arbeit synthetisierten Verbindungen erfolgte mittels NMR-Spektroskopie (1H-, 13C-, 15N- und 29Si-NMR) und Elementaranalysen (C, H, N) bzw. HRMS-Experimente.
Die hydrolytische Spaltung der Si–OC-Bindung in Alkoxy(aminoalkyl)silanen stellt einen technisch sehr wichtigen Schlüsselschritt in der Synthese von Amino-funktionalisierten Polysiloxanen dar. Um den Mechanismus dieser Si–OC-Bindungsspaltung besser zu verstehen, wurden die Alkoxysilane 4b, 4c, 5b, 6b, 6c, 7b, 7c, 8a–8c, 9b, 11a–11c, 12b, 14a–14c,
15a–15c, 16a–16c, 17a–17c, 18a–18c, 19 und 20 hinsichtlich ihrer Hydrolysekinetik in CD3CN/D2O unter sauren und basischen Bedingungen mittels 1H-NMR-Spektroskopie untersucht.
Die Ergebnisse dieser Struktur–Reaktivitäts-Studie zeigten, dass die beobachteten unterschiedlichen Hydrolysegeschwindigkeiten das Resultat mehrerer Faktoren sind, wie beispielsweise elektronische und sterische Effekte, der große Einflusses des pD-Wertes und auch intramolekulare N–H∙∙∙O-Wasserstoffbrückenbindungen zwischen der protonierten Amino-Gruppe und der Alkoxy-Abgangsgruppe. Da der Einfluss dieser Effekte auf die Reaktivität der untersuchten α-, β-, γ- und ζ-Silane sehr unterschiedlich ist, kann kein klarer Zusammenhang zwischen der Hydrolysereaktivität und der Positionierung der stickstoff-haltigen funktionellen Gruppe (α-, β-, γ- und ζ-Position) erkannt werden. Die jeweils beobachtete Reaktivität entspricht vielmehr einer Summe aller zuvor genannten Teileffekte.
Die Erkenntnisse, die im Rahmen dieser Arbeit erhalten wurden, ermöglichen ein verbessertes grundlegendes Verständnis der Reaktivität von funktionalisierten α-, β-, γ- und ζ-Silanen, und sind für die Silicon-Industrie von großem Interesse, da sie eine gezieltere Anwendung der α-, β- und γ-Aminosilane in der Synthese von technisch wichtigen Amino-funktionalisierten Polysiloxanen erlauben.
Das Ziel dieser Arbeit war die Herstellung von Diamantmaterialien, deren Oberflächen mit Alkinen, Aziden oder Aldehyden modifiziert waren. Diese funktionellen Gruppen sollten die einfache Anbindung verschiedener katalytisch aktiver Systeme mit Hilfe der 1,3-dipolaren Cycloaddition nach Huisgen bzw. Iminbildung ermgöglich.
Da in einer vorangegangenen Arbeit Hinweise darauf gefunden wurde, dass die hochgradig funktionalisierte Oberfläche von Detonationsnanodiamant dazu in der Lage ist, die Aktivität von immobilisierten Katalysatoren zu behindern. Darum wurde in dieser Arbeit verglichen, ob die Verwendung von starren Linkern auf Tolanbasis einen Vorteil gegenüber ihren flexiblen Gegenstücken liefert. Dazu wurde für jede der oben genannten Funktionalisierungsarten je ein Diamantmaterial mit flexibler sowie mindestens eines mit unbiegsamer Verbindungseinheit hergestellt und getestet. Dadurch konnte das Konzept der starren Linker für Enzyme bestätigt werden und es wurde eine signifikant höhere Aktivität erhalten, als wenn flexible Anbindungsbrücken verwendet wurden. Bei Organokatalysatoren und metallorganischen Systemen konnten jedoch keine erfolgreichen Katalysen durchgeführt werden.
Seit Anfang des 20. Jahrhunderts ist die Krankheit Krebs die zweithäufigste Todesursache.
Die immer besser werdende Diagnostik zur Früherkennung von Krebserkrankungen bringt
zudem mit sich, dass eine steigende Anzahl an Krankheitsfällen bekannt wird; gleichzeitig hat
eine sichere Erkennung im Frühstadium einen hohen Stellenwert, da nur so frühzeitig
therapeutische Maßnahmen ergriffen werden können. Obwohl zur kurativen oder zur
palliativen Behandlung schon lange viele ausgereifte Therapiemethoden und -konzepte
erfolgreich angewendet werden, sind bis heute viele Krebserkrankungen noch immer
unheilbar und es gibt bislang in vielen Fällen noch keine potenten Arzneistoffe für eine
wirkungsvolle Therapie. Die Suche nach neuen Wirkstoffen ist daher ein Schwerpunkt
aktueller Forschung. Auch unsere Arbeitsgruppe beschäftigt sich mit dieser Thematik und hat
in Zusammenarbeit mit dem Universitätsklinikum Würzburg ein neuartiges Pro-Prodrug-
Konzept für antitumorale Wirkstoffe entwickelt.
Daher war es ein Ziel der vorliegenden Arbeit, aufbauend auf medizinischen und
biochemischen Vorarbeiten, eine Leitstruktur für antitumorale Wirkstoffe zu erarbeiten, die auf
dem hier vorgestellten Pro-Prodrug-Konzept basiert, und eine entsprechende Syntheseroute zur
Gewinnung erster Fluoreszenz-markierter Pro-Prodrug-Modellverbindungen zu etablieren.
Eine andere interessante Substanzklasse, die nicht nur aufgrund ihrer Wirkeigenschaften
als Photosensibilisatoren in der photodynamischen Therapie bereits Gegenstand intensiver
Forschung ist, sondern auch in weiteren Bereichen der angewandten Chemie, wie
beispielsweise in der metallorganischen Katalyse oder in den Materialwissenschaften
Anwendung findet, sind die Porphyrine. Neben den achiralen Vertretern finden die chiralen
Porphyrinoide Anwendung in der chiralen Erkennung oder in der asymmetrischen
Synthese.
Auf dem Gebiet der Porphyrine war das Ziel der vorliegenden Arbeit die gezielte Synthese
N,C- und C,C-verknüpfter dimerer und trimerer Porphyrine, einhergehend mit intensiven
Untersuchungen der Reaktivität der verwendeten Bausteine, des Reaktionsmechanismus und
der hohen Regioselektivität. Da einige der synthetisierten Substanzen Axial- oder
Helikalchiralität aufweisen, wurden diese durch HPLC-CD eingehend auf ihre chiroptischen
Eigenschaften untersucht.
In this thesis the syntheses and detailed investigations on two foldable PBI systems were presented. The reversible, solvent-dependet folding/unfolding-behavior was used to study the ground and excited states properties of folda-dimer and folda-trimer by means of different spectroscopic methods as well as theoretical studies. The switching between charge transfer or excimer formation pathways of photoexcited molecules influenced by the spatial arrangement of chromophores within defined dye systems illustrates the impact of conformational preferences on functional properties.
Herstellung dotierter Nanodiamantpartikel und ihre Funktionalisierung mit schwefelhaltigen Gruppen

In dieser Dissertation werden Methoden zur Darstellung von Stickstoff- und Bor-dotierten Nanodiamantpartikeln durch Vermahlung von makroskopischem HPHT- und CVD-Diamant mit unterschiedlichen Mühlen und Mahlverfahren beschrieben. Untersucht wird dabei der Zusammenhang von erzielten Teilchengrößen nach dem Mahlen und den Kristallitgrößen der Ausgangsdiamanten sowie der angewandten Mahlmethode. Durch Anwendung verschiedener oxidativer Methoden wird während des Mahlens entstehender sp2-Kohlenstoff entfernt. Ebenfalls wird deren Einfluss auf das Fluoreszenzverhalten NV-haltiger Nanodiamantpartikel analysiert.
Des Weiteren werden Syntheserouten zur kontrolliert ablaufender Oberflächenflächenfunktionalisierung von Nanodiamant mit schwefelhaltigen Gruppen wie Thiolen, Trithiocarbonaten und Disulfiden gezeigt, welche eine selektive Anbindung der Diamantpartikel an Goldstrukturen ermöglicht. Diese Verfahren werden an Detonationsdiamant sowie fluoreszierendem HPHT- und CVD-Diamantpartikeln angewandt. Gegebenfalls kann nach erfolgreicher Anbindung fluoreszierender Nanodiamantpartikel an Goldnanostrukturen unter Ausnutzung von Plasmonenresonanz die einzigartigen Spineigenschaften der NV-Zentren mit Hinblick auf einen möglichen Einsatz Stickstoff-dotierter Diamantnanopartikel in der Quanteninformationsverarbeitung untersucht werden.
The main objective of this thesis was the design and synthesis of perylene bisimide dyes with sufficient water-solubility for the construction of self-assembled architectures in aqueous solutions. Beside these tasks another goal of this project was the control over the self-assembly process in terms of aggregate size and helicity, respectively. Within this thesis an appropriate synthesis for spermine-functionalized perylene bisimide dyes was developed and conducted successfully. The characterization of these building blocks and their course of self-assembly were investigated by NMR, UV/Vis and fluorescence spectroscopy as well as by atomic force and transmission electron microscopy. For the better understanding of the experimental results theoretical calculations were performed.
The successful synthesis of a family of donor-iridium complex-acceptor triads (T1–T6, pMV1 and mMV1) and their electrochemical and photophysical properties were presented in this work. Triarylamines (TAA) were used as donors and naphthalene diimide (NDI) as acceptor. A bis-cyclometalated phenylpyrazole iridium dipyrrin complex acts as a photosensitiser. In addition, a molecular structure of T1 was obtained by single crystal X-ray diffraction.
Transient absorption spectroscopy experiments of these triads resembled that upon excitation a photoinduced electron transfer efficiently generates long-lived, charge-separated (CS) states. Thereby, the electron-transfer mechanism depends on the excitation energy.
The presence of singlet and triplet CS states was clarified by magnetic-field dependent transient-absorption spectroscopy in the nanosecond time regime. It was demonstrated that the magnetic field effect of charge-recombination kinetics showed for the first time a transition from the coherent to the incoherent spin-flip regime.
The lifetime of the CS states could be drastically prolonged by varying the spacer between the iridium complex and the NDI unit by using a biphenyl instead of a phenylene unit in T4.
A mixed-valence (MV) state of two TAA donors linked to an iridium metal centre were generated upon photoexcitation of triad pMV1 and mMV1. The mixed-valence character in these triads was proven by the analysis of an intervalence charge-transfer (IV-CT) band in the (near-infrared) NIR spectral region by femtosecond pump-probe experiments. These findings were supported by TD-DFT calculations.
The synthesis of dyads (D1–D4) was performed. Thereby the dipyrrin ligand was substituted with electron withdrawing groups. The electrochemical and photophysical characterisation revealed that in one case (D4) it was possible to generate a CS state upon photoexcitation.
Die Produktion von Abwehr-, Signal- und Botenstoffen sichert vielen Pflanzen und Mikroorganismen das Überleben in einer sich ständig wandelnden Umwelt mit zahlreichen Konkurrenten und Feinden. Diese Sekundärmetabolite können oft medikamentös gegen Pathogene eingesetzt werden, die den Menschen befallen und Krankheiten verursachen. Die Herausforderung besteht dabei in der selektiven und sensitiven Detektion, der schonenden Isolierung und der richtigen und kompletten Strukturaufklärung dieser Moleküle, sowie der eventuellen synthetischen Modifikation, um eine bessere Verträglichkeit oder Wirkung für den menschlichen Körper zu erreichen. Leistungsfähige chromatographische Instrumente zur Trennung wie HPLC und CZE, emfindliche Detektoren wie UV- und Massenspektrometer, sowie aussagekräftige Messverfahren zur Charakterisierung struktureller Merkmale wie NMR- und CD-Spektroskopie und quantenchemische Rechnungen sind dabei von essentieller Bedeutung.
Mit diesen – und weiteren – Methoden gelang in der vorliegenden Arbeit die Detektion, Isolierung und Strukturaufklärung neuer Naphthylisochinolin-Alkaloide aus zwei tropischen Ancistrocladus-Lianen, die Charakterisierung von bekannten und neuen Polyketiden aus einem Pilz der Gattung Streptomyces, sowie die Analyse von Glucosinolaten im Phloemsaft der Modellpflanze Arabidopsis thaliana.
Analog zu den auf hochgeordneten Farbstoffarchitekturen in den biologischen Photosyntheseapparaten basierenden Energiekonversionssystemen sollte die exakte Einstellung zwischenmolekularer Wechselwirkungen auch in künstlichen Halbleitern eine entscheidende Rolle für die Weiterentwicklung organischer Elektronikmaterialien spielen. Für eine derartige, präzise Steuerung der nanoskaligen Anordnung in organischen Materialien erscheinen Merocyaninfarbstoffe wegen ihrer hochgerichteten, dipolaren Aggregation äußerst aussichtsreich. In diesem Zusammenhang war das Ziel der vorliegenden Arbeit die Ausnutzung funktionaler, stark selbstorganisierender Merocyanine, um eine gezielte Beeinflussung der Morphologie in der aktiven Schicht von BHJ-Solarzellen zu erreichen. Hierzu sollte zunächst eine umfangreiche Serie komplexer Merocyanine synthetisiert und vollständig charakterisiert werden. Im Folgenden wurde angestrebt, die optischen und elektrochemischen Eigenschaften der molekular gelösten Farbstoffe zu bestimmen und für ausgewählte, geeignete Strukturen das Selbstorganisationsverhalten im Detail zu studieren. Zuletzt sollte durch eine sorgfältige Optimierung der Prozessierungsbedingungen ein Transfer der in Lösung gefundenen, supramolekularen Strukturen in den Blend lösungsprozessierter BHJ-Solarzellen erreicht werden. Die organischen Elektronikbauteile wurden dabei im Arbeitskreis von Prof. Dr. Klaus Meerholz (Universität Köln) gefertigt und charakterisiert.
Zusammenfassend zeichnet die vorliegende Arbeit ein umfassendes Bild von der Synthese funktionaler Merocyanine, dem Studium ihrer molekularen und Selbstorganisationseigenschaften sowie ihrer Anwendung als p-Halbleitermaterialien in organischen Solarzellen. Der komplexe Molekülaufbau der dargestellten Farbstoffe führte dabei zur Ausbildung verschiedener Farbstofforganisate, deren Struktur sowohl in Lösung als auch teilweise im Festkörper aufgeklärt werden konnte. Die erfolgreiche Implementierung von H-aggregierten Spezies der Verbindung 67b in die aktive Schicht organischer BHJ-Solarzellen resultierte in der Bildung effizienter Perkolationspfade für Exzitonen und freie Ladungsträger, wodurch diese Bauteile merklich höhere Stromdichten generieren konnten und gegenüber Zellen ohne H-Spezies über 20 % gesteigerte Effizienz aufwiesen. Diese Befunde verifizieren die postulierte Hypothese, dass eine gezielte Einstellung der zwischenmolekularen Wechselwirkungen bei organischen Halbleitern zu einer Optimierung der Funktionalität organischer Elektronikmaterialien beitragen kann.
Kohlenhydrat-Protein-Wechselwirkungen sind häufig entscheidend beteiligt an verschiedenen einer Infektion oder malignen Erkrankung zugrunde liegenden molekularen Erkennungs-prozessen, die zu Adhäsion, Zell-Zell-Interaktion sowie Immunreaktion und -toleranz führen. Trotz der hohen Relevanz für Diagnostik und Therapie dieser Erkrankungen sind die betreffenden Strukturen und Mechanismen bisher nur ungenügend untersucht und verstanden. Ziel dieser stark interdisziplinär angelegten Arbeit war es daher, Methoden der Fachbereiche Chemie und Pharmazie, Biologie und Medizin, aber auch Physik zu kombinieren, um Kohlenhydraterkennungsprozesse im Detail zu untersuchen und auf dieser Basis strukturell neuartige diagnostische und therapeutische Anwendungen zu entwerfen.
Die hochkomplexe Zusammensetzung einer Zelloberfläche wurde zunächst auf ihren Glycan-anteil reduziert und stark vereinfacht auf der Oberfläche sogenannter Glycochips imitiert. Die verwendeten Systeme auf Basis einer Gold- bzw. Glasoberfläche ergänzen sich optimal in ihrer Eignung für komplementäre analytische Methoden wie Massenspektrometrie sowie quantifizierbare Fluoreszenzspektroskopie.
Der Übergang auf die lebende Zelloberfläche gelang mit Hilfe des Metabolic Glyco-engineering, das die kovalente Präsentation definierter Motive durch eine Cycloaddition zwischen zwei bioorthogonalen Reaktionspartnern (z.B. Azid und Alkin) ermöglicht.
Auf diese Weise wurden in Zusammenarbeit mit der Arbeitsgruppe Sauer (Universität Würzburg) zunächst die Dichte und Verteilung verschiedener Oberflächenglycane auf humanen Zellen mittels hochauflösender Fluoreszenzmikroskopie (dSTORM) bestimmt. Diese Parameter zeigten im Modell des Glycochips einen entscheidenden Einfluss auf Bindungsereignisse und multivalente Erkennung und zählen auch auf natürlichen Zelloberflächen – in engem Zusammenhang mit der lateralen und temporalen Dynamik der Motive – zu den wichtigen Faktoren molekularer Erkennungsprozesse.
Die gezielte Modifikation zellulärer Oberflächenglycane eignet sich aber auch selbst als Methode zur Beeinflussung molekularer Wechselwirkungsprozesse. Dies wurde anhand des humanpathogenen Bakteriums S. aureus gezeigt, dessen Adhäsion auf Epithelzellen der Blasenwand durch Metabolic Glycoengineering partiell unterdrückt werden konnte.
In einem ergänzenden Projekt wurden zwei potentielle Metabolite eines konventionellen Antibiotikums – des Nitroxolins – mit bakteriostatischer sowie antiadhäsiver Wirksamkeit dargestellt. Diese dienten als Referenzsubstanzen zur Verifizierung der postulierten Struktur der Derivate, werden aber auch selbst auf ihr Wirkprofil hin untersucht. Gleichzeitig stehen sie zusammen mit der Grundverbindung zudem als Referenz für die Wirkstärke potentieller neu entwickelter Antiadhäsiva zur Verfügung.
Der erste Teil dieser Arbeit beschäftigte sich mit dem Ziel, amphiphile Oligoethylenglykol-funktionalisierte Perylenmonoimiddiester und Dicarbonsäure- bzw. Amino-funktionalisierte Perylenbisimide zu synthetisieren. Weiterhin wurden die optischen Eigenschaften in Lösung und das Aggregationsverhalten der Ester-, Dicarbonsäure- und der Amino-funktionalisierten Perylenfarbstoffe untersucht. (...)
Der zweite Teil dieser Arbeit widmete sich dem Ziel, die amphiphilen Oligoethylenglykol-funktionalisierten Zinkchlorine, 31-Hydroxy-Zinkchlorin (ZnChl-OH) und 31-Methoxy-Zinkchlorin (ZnChl-OCH3), herzustellen und desweiteren deren Aggregationsverhalten in wässriger Lösung vergleichend zu studieren, um den Einfluss der Lösungsmittel und des Substituenten in 31-Position auf die thermodynamischen und kinetischen Eigenschaften und auf die Aggregatstruktur zu bestimmen. The focus of this work was the investigation of energy transfer between charge transfer states. For this purpose the multidimensional chromophores HAB-S, HAB-A, B1 and B2 were synthesised, each consisting of three electron donor and three electron acceptor redox centres linked symmetrically or asymmetrically by the hexaarylbenzene framework. Triarylamines represent in all these compounds the electron donors, whereas the electron poor centres were triarylboranes in B1 and B2 and PCTM centres in HAB-S and HAB-A, respectively. The hexaarylbenzenes were obtained by cobalt catalysed cyclotrimerisation of the respective tolan precursors. In addition, Star was synthesised, which consists of a central PCTM linked to three triarylamin centres by tolan bridging units in a star-like configuration. The hexaarylbenzene S1a/b substituted with six squaraine chromophores could not be realised. It is assumed that the cyclotrimerisation catalyst Co2(CO)8 does not tolerate the essential hydroxyl groups in the tolan precursor S2a. The alternative reaction pathway to execute the cyclotrimerisation reaction first and introduce the hydroxyl groups thereafter failed as well, because the required hexaarylbenzene substituted by six semisquaric acid moieties could not be synthesised. However, energy transfer interactions could be investigated in the tolan precursor S2a with two squaraine units to obtain information about the electronic coupling provided by the tolan bridge. For all multidimensional compounds model molecules were synthesised with only a single donor-acceptor pair (B3, Star-Model and HAB-Model). This allows a separate consideration of energy and charge transfer processes. It has to be stressed that in all before mentioned multidimensional compounds the “through bond” energy transfer interaction between neighbouring IV-CT states is identical to a transfer of a single electron between two redox centres of the same kind (e.g. TAA -> TAA+). The latter can be analysed by electron transfer theory. This situation is observed when the two IV-CT states transferring energy share one redox centre.
All compounds containing PCTM centres were characterised by paramagnetic resonance spectroscopy. Thereby, a weak interaction between the three PCTM units in HAB-S and HAB-A was observed. In addition, when oxidising Star-Model, a strongly interacting singlet or triplet state was obtained. In contrast, signals corresponding to a weakly interacting biradical were obtained for HAB-Model+. This indicates a strong electronic coupling between the redox centres provided by the tolan bridge and a weak coupling when linked by the hexaarylbenzene. This trend is supported by UV/Vis/NIR absorption measurements. The analysis of the observed IV-CT absorption bands by electron transfer theory reveals a weak electronic coupling of V = 340 cm-1 in HAB-Model and a distinctly stronger coupling of V = 1190-2900 cm-1 in Star-Model. In the oxidised HAB-S+, Star+ and Star-Model+ a charge transfer reversed from that of the neutral species, that is, from the PCTM radical to the electron poorer cationic TAA centre, was observed by spectroelectrochemistry. The temporal evolution of the excited states was monitored by ultrafast transient absorption measurements. Within the first picosecond stabilisation of the charge transfer state was observed, induced by solvent rotation. Anisotropic transient absorption measurements revealed that within the lifetime of the excited state (tau = 1-4 ps) energy transfer does not occur in the HABs whereas in the star-like system ultrafast and possibly coherent energy redistribution is observed. Taken this information together the identity between energy transfer and electron transfer in the specific systems were made apparent. It has to be remarked that neither energy transfer nor charge transfer theory can account for the very fast energy transfer in Star.
The electrochemical and photophysical properties of B1 and B2 were investigated by cyclic voltammetry, absorption and fluorescence measurements and were compared to B3 with only one neighbouring donor-acceptor pair. For the asymmetric B2 CV measurements show three oxidations as well as three reduction peaks whose peak separation is greatly influenced by the conducting salt due to ion-pairing and shielding effects. Consequently, peak separations cannot be interpreted in terms of electronic couplings in the generated mixed valence species. Transient absorption, fluorescence solvatochromism and absorption spectra show that charge transfer states from the amine to the boron centres are generated after optical excitation. The electronic donor-acceptor interaction is weak though as the charge transfer has to occur predominantly through space. The electronic coupling could not be quantified as the CT absorption band is superimposed by pi-pi* transitions localised at the amine and borane centres. However, this trend is in good agreement to the weak coupling measured for HAB-Model. Both transient absorption and fluorescence upconversion measurements indicate an ultrafast stabilisation of the charge transfer state in B1- B3 similar to the corresponding observations in HAB-S and Star. Moreover, the excitation energy of the localised excited charge transfer states can be redistributed between the aryl substituents of these multidimensional chromophores within fluorescence lifetime (ca. 60 ns). This was proved by steady state fluorescence anisotropy measurements, which further indicate a symmetry breaking in the superficially symmetric HAB. Anisotropic fluorescence upconversion measurements confirm this finding and reveal a time constant of tau = 2-3 ps for the energy transfer in B1 and B2. It has to be stressed that, although the geometric structures of B1 and HAB-S are both based on the same framework and furthermore the neighbouring CT states show in both cases similar Coulomb couplings and negligible “through bond” couplings, very fast energy transfer is observed in B1 whereas in HAB-S the energy is not redistributed within the excited state lifetime. To explain this, it has to be kept in mind that the energy transfer and the relaxation of the CT state are competing processes. The latter is influenced moreover by the solvent viscosity. Hence, it is assumed that this discrepancy in energy transfer behaviour is caused by monitoring the excited state in solvents of varying viscosity. Adding fluoride ions causes the boron centres to lose their acceptor ability due to complexation. Consequently, the charge transfer character in the donor-acceptor chromophores vanishes which could be observed in both the absorption and fluorescence spectra. However, the fluoride sensor ability of the boron centre is influenced strongly by the moisture content of the solvent possibly due to hydrogen bonding of water to the fluoride anions.
UV/Vis/NIR absorption measurements of S2a show a red-shift by 1800 cm-1 of the characteristic squarain band compared to the model compound S20. From exciton theory a Coulomb coupling of V = 410 cm-1 is calculated which cannot account for this strong spectral shift. Consequently, “through-bond” interactions have to contribute to the strong communication between the two squaraine chromophores in S2a. This is in accordance with the strong charge transfer coupling calculated for the tolan spacer in Star-Model.
Im Rahmen der vorliegenden Arbeit wurden promesogene Arme sowie eine Bibliothek von Sternmesogenen mit Anthracen als Donor- und Anthrachinon als Akzeptorbaustein synthetisiert und untersucht. Ein Schwerpunkt der Arbeit lag auf der Synthese, dem Upscaling, der selektiven Schützung und weiteren Umsetzung der 2,6-substituierten Anthracen- und Anthrachinon-Chromophore zu den Armbausteinen. Besondere Herausforderungen ergaben sich nicht nur in der Entwicklung einer effizienten Synthesestrategie zur Gewinnung der Chromophore, sondern auch in der Wahl geeigneter Schutzgruppen. Die sternförmigen Verbindungen, die im Rahmen der vorliegenden Arbeit präpariert wurden, enthalten 1,3,5-Trihydroxybenzen (Phloroglucin) als Kerneinheit und sind Multiarmmesogene mit der kleinstmöglichen Zahl an Armen. Durch geeignete Schutzgruppenstrategien gelang neben den C3-symmetrischen Verbindungen die gezielte Darstellung der C2-symmetrischen und unsymmetrischen Verbindungen. Die Gesamtausbeuten der semiperfluorierten Verbindungen fallen deutlich geringer aus als die der ausschließlich mit Alkylketten dekorierten Derivate, da ihre Isolierung sehr anspruchsvoll ist. Alle Verbindungen bilden ausnahmslos kolumnare Phasen. Semiperfluorierte Ketten wurden eingeführt, um eine Trennung des Donors Anthracen und des Akzeptors Anthrachinon zu erreichen. Die Kolumnendurchmesser sind bei allen kolumnaren Mesophasen wesentlich kleiner als die Durchmesser der sternförmigen Konformere der Mesogene. Angelehnt an die früher untersuchten Oligobenzoatsterne werden Modelle mit gefalteten, E-förmigen Konformeren aufgestellt. So ist es möglich, die erforderliche Anzahl an Molekülen pro Elementarzelle in einer dichten, nanosegregierten Packung anzuordnen. Mit Absorptions- und Emissionsmessungen konnte dieses Modell bestätigt werden. In allen Donor- und Akzeptor-substituierten Verbindungen wird die Fluoreszenz durch Energietransferprozesse nach Förster und Dexter fast vollständig gelöscht. Restfluoreszenz wird in dem Bereich beobachtet, der nur noch den Transfer nach Dexter zulässt und ist für die Derivate am höchsten, die in den E-förmigen Konformeren Donor und Akzeptor am besten trennen können. Die Ergebnisse dieser Arbeit zeigen, dass Anthracen- und Anthrachinonderivate eine Vielzahl komplexer zwei- und dreidimensional hochgeordneter kolumnarer Strukturen ausbilden und damit hochinteressant sind als flüssigkristalline organische Halbleitermaterialien.
Ziel dieser Arbeit war die Synthese geeigneter Modellsysteme für die Untersuchung gekrümmter Graphenfragmente. Dafür bot sich die Synthese von substituierten Pyracylenen an. Diese sollten in eine Aromatensphäre eingebunden sein, um so den Einfluss der Graphenumgebung zu simulieren. Ein geeignetes einfaches Untersuchungsobjekt stellt das Bisbenzo[d,j]pyracylen/ Dicyclopenta[fg,op]tetracen (10) dar. Auch die Einbettung in größere „Nanographene“ ist ein lohnendes Syntheseziel zur Untersuchung von Struktur-Eigenschaftsbeziehungen. Gegebenenfalls kann nach erfolgreicher Darstellung auch das Auftreten der Stone-Wales-Umlagerung untersucht werden. Das heißt, die Systeme sollen mit Hilfe der FVP auf ihr Vermögen zur Kohlenstoffgerüstumlagerung untersucht werden. Die folgende Abbildung zeigt die Zielmoleküle und mögliche Stone-Wales-Umlagerung. ...
Für die Aufklärung der absoluten Stereostruktur von chiralen Molekülen, die ein Chromophorsystem besitzen, hat sich die Kombination der experimentellen und theoretischen Spektroskopie des elektronischen Circulardichroismus (ECD) als Methode bewährt. In der vorliegenden Arbeit wurden die chiroptischen Eigenschaften von Bisbibenzyl-Makrocyclen, Mono- und Bis(cycloketo)porphyrinen, der Mohnblütenpigmente Nudicaulin I und II sowie von Bordipyrrol-Dimeren mit quantenchemischen Methoden untersucht. Zu diesem Zweck wurden verschiedene dichtefunktionaltheoretische (DFT) Ansätze und post-HF-Methoden, wie z. B. der Coupled-Cluster-Ansatz RI-CC2, bezüglich ihrer Eignung, die Grund- und angeregten Zustände (UV/vis- und ECD-Eigenschaften) der einzelnen Verbindungen korrekt wiederzugeben, evaluiert. Da bei quantenchemischen UV- und ECD-Rechnungen an ausgedehnten π-Systemen aufgrund energetisch nah beieinander liegender Anregungen die Wahrscheinlichkeit für ghost states und charge-transfer-Übergänge sowie Multireferenz-Problematiken steigt, wurden diese Aspekte genauer betrachtet. Die ersten zwei Phänomene lassen sich bereits auf TD-DFT-Niveau durch genaue Analyse der theoretischen spektroskopischen Daten ermitteln und unter Umständen durch entsprechend korrigierte Funktionale sogar sehr gut beschreiben. Im Gegensatz dazu können Doppelanregungsanteile überhaupt erst durch Rechnungen mit geeigneten Methoden, wie z. B. das hier verwendete SORCI-Verfahren, erfasst werden. Zusätzlich wurde das zur Auswertung von UV und ECD-Daten entwickelte Programm SpecDis um Funktionalitäten erweitert, welche die Berechnung des Übereinstimmungsgrades zweier UV- bzw. ECD-Kurven ermöglichen, und dadurch ein zusätzliches quantitatives Kriterium für die Verlässlichkeit des Spektrenvergleichs und folglich für die Zuordnung der absoluten Konfiguration bieten.
In dieser Arbeit wurde die Erstfunktionalisierung des Diamanten durch die Diels-Alder-Reaktion realisiert und anschließend ein Thiol als weiterführende Bindungsstelle etabliert. Die Thiolfunktion konnte in einer radikalischen Reaktion mit einem terminalen Alken umgesetzt werden. Als Reaktionspartner wurde ein Allylglucosid synthetisiert und mit Hilfe eines Radikalstarters an den Diamanten gebunden. Für nicht-radikalische Verknüpfungsreaktionen eines vorfunktionalisierten Diamanten mit dem Zielmolekül konnte die Diazoniumsalz-Methode genutzt werden. Die Wahl der 4 Aminobenzoesäure als Edukt der Reaktion erlaubte in einer einstufigen Reaktion die Einführung einer Carbonsäuregruppe auf der Oberfläche. Durch Umsetzung mit einem Amin (Mannosecluster) konnte eine stabile Amidbindung erzeugt werden. Die erhaltenen Saccharid-Diamant-Konjugate wurden anschließend auf deren Inhibitionsvermögen gegenüber Escherichia coli getestet. Um eine Erweiterung der Methoden zur stabilen Erstfunktionalisierung der Diamantoberfläche zu erreichen, wurde untersucht ob die aus der Fulleren-Chemie bekannte Bingel-Hirsch-Reaktion auf Nanodiamant übertragbar ist. Für die Untersuchungen am Diamanten wurden unterschiedliche Malonsäurederivate eingesetzt, welche signifikante Banden im Infrarotspektrum besitzen, um eine eindeutige Funktionalisierung nachweisen zu können.
Im Rahmen dieser Arbeit wurde gezeigt, dass die elektronischen Eigenschaften und das Packungsverhalten von Naphthalindiimid (NDI)- und Diketopyrrolopyrrol (DPP)- Derivaten durch Einführen geeigneter Substituenten sowie durch Erweiterung des konjugierten Pi-Systems zur Optimierung der Eigenschaften als organische Halbleitermaterialien eingestellt werden können. Während DPP-Halbleiter zwar in Polymeren, nicht jedoch als niedermolekulare Halbleiter, für die organische Elektronik von Bedeutung sind, stellen vor allem die hier vorgestellten cyanierten DPP-Derivate eine synthetisch leicht zugängliche Klasse an niedermolekularen p-Halbleitern mit exzellenten Lochtransporteigenschaften dar. Die Expansion des NDI- und DPP-Kerns eröffnet zudem den synthetischen Zugang zu neuen Verbindungsklassen mit veränderten elektronischen Eigenschaften. Gerade das für die Carbazolocarbazoldiimide postulierte Konzept einer elektronenreichen p-Transportachse konnte durch Wahl geeigneter Imidsubstituenten zur Entwicklung zweidimensionaler p-Halbleiter mit sehr guten Mobilitäten führen. Schließlich stellen 2,6-kernhalogenierte NDI-Derivate mit fluorierten Imidgruppen aufgrund der herausragenden Elektronenmobilitäten und der sehr hohen Luftstabilität außergewöhnliche Kandidaten für den Einsatz als n-Halbleiter in organischen Dünnschichttransistoren dar.
Diverse synthetische Zugänge zu einer neuen Verbindungsklasse, nämlich den chiralen 9-
Oxabispidinen wurden realisiert. Als gemeinsame Ausgangsverbindung diente das Aminodiol. Die Route über ein Bispidinlactam stellte sich aufgrund der geringen Ausbeute (~5%) über 8 Stufen als wenig effektiv heraus. Ein Nitril eignete sich wesentlich besser als Zwischenstufe und lieferte die Zielverbindungen in 9-27% Gesamtausbeute über 8-12 Stufen.
Auch ein Syntheseweg zur Darstellung von Bispidinen konnte ausgehend vom Naturstoff (–)-Cytisin, der aus den Samen des Gemeinen Goldregens isoliert wurde, etabliert werden. Hierzu wurde in Analogie zu O’Briens Arbeiten zunächst die freie Aminofunktion als Carbamat geschützt und der aromatische Ring zum Lactam reduziert, um dann in 5-Position derivatisieren zu können. Verschiedene Substituenten (Me, iPr, F, cPr) konnten sowohl in axialer als auch in äquatorialer Position eingeführt werden. Ein weiterer Reduktionschritt
am Ende der Sequenz lieferte die trizyklischen Diamine in 15-38% Ausbeute über nur 4-6
Stufen ausgehend von (–)-Cytisin. Eine Cyclopropylgruppe wurde durch Einführung einer
Methoxymethylgruppe, Eliminierung und eine Cyclopropanierung mit Diiodmethan/ Diethylzink generiert.
Alle synthetisierten Verbindungen wurden als chirale Liganden in verschiedenen enantioselektiven Reaktionen evaluiert. Im Fokus standen hierbei zunächst die 9-Oxabispidine, die sich–im Gegensatz zu den bekannten Bispidinen–leider nicht als chirale Liganden in enantioselektiven Deprotonierungen einsetzen lassen, da sie selbst deprotoniert werden. In enantioselektiven Henry-Reaktionen stellte sich das trizyklische 9-Oxabispidin als hervorragender Ligand heraus, der als Kupferkomplex mit einer Vielzahl an Aldehyden (aliphatisch, aromatisch) und Nitromethan umgesetzt wurde. Sowohl die Ausbeuten (44-95%) als auch die Enantiomerenüberschüsse (91-98%) waren exzellent und können mit den Ergebnissen der besten Literatur-bekannten Katalysatoren mithalten. Auch in einer diastereoselektiven Umsetzung mit Nitroethan konnte ein dr von 80:20 mit sehr guten 94% ee für das Hauptdiastereomer erzielt werden. Die bizyklischen 9-Oxabispidine eignen sich weniger für Henry-Reaktionen, da die Verbindungen mit sterisch weniger anspruchsvollen Reste nur mittelmäßige Resultate (33-46% ee) ergeben. Mit zunehmender Sterik ist auch eine steigende Tendenz zu einem höheren Enantiomerenüberschuss [z.B. R = Ph, 56/57% ee] zu erkennen. Interessant ist die Stereoselektivität der bizyklischen Oxabispidine: Während das trizyklische Bispidin zu einer S-Konfiguration im Produkt führt–wie es von einem (+)-Spartein-Ersatzstoff zu erwarten ist–liefern R = Et, iPr. cHex und Ph das R-Produkt - genau wie (–)-Spartein. Der Grund hierfür ist wahrscheinlich die exponierte Stellung der äußeren Methylgruppe im annelierten Piperidinring. Ein ähnlicher Effekt trat mit dem 9-Oxabispidin R = tBuPh, das ebenfalls einen weit ausladenden aromatischen Ring besitzt, ein. Auch hier wurden unter Induktionsumkehr im Vergleich zu R = Et, iPr, cHex, Ph das S-konfigurierte Produkt in 38-39 % ee erhalten.
Im Gegensatz zu den 9-Oxabispidinen können die Bispidine in enantioselektiven Deprotonierungsreaktionen eingesetzt werden. Die bei der Umsetzung erzielten Ergebnisse mit den Monomethyl-Verbindungen (92% ee, äquatoriale Me-Gruppe und 79% ee, axiale Me-Gruppe) zeigen, dass eine axiale Methylgruppe den Chiralitätstransfer eher negativ beeinflusst, wohingegen eine äquatoriale Methylgruppe keinen Einfluss ausübt auf den ee (gleicher ee mit 92% wie das trizyklische Bispidin). Bei größeren äquatorialen Substituenten wie iPr (51%) sinkt ebenfalls der Enantiomerenüberschuss. Zwei gleichartige Substituenten (≠ H) in 5-Position stören die Reaktion so sehr, dass der ee-Wert stark abfällt (34% ee bei zwei 5-Methylgruppen). Bei Einführung von kleinen Substituenten in äquatorialer und axialer Position, wie F (65% ee) oder -CH2-CH2- (40% ee) liegen die erzielten ee-Werte auch viel niedriger.
The objective of this thesis focuses on the development of strategies for precise control of perylene bisimide (PBI) self-assembly and the in-depth elucidation of structural and optical features of discrete PBI aggregates by means of NMR and UV/Vis spectroscopy. The strategy for discrete dimer formation of PBIs is based on delicate steric control that distinguishes the two facets of the central perylene surface. The strategy applied in this thesis for accessing discrete PBI quadruple and further oligomeric stacks relies on backbone-directed PBI self-assembly. For this purpose, two tweezer-like PBI dyads bearing the respective rigid backbones, diphenylacetylene (DPA) and diphenylbutydiyne (DPB), were synthesized. The distinct aggregation behavior of these structurally similar PBI dyads can be ascribed to the intramolecular distance between the two PBI chromophores imparted by the DPA and DPB spacers.
In this work, a series of redox cascades was synthesised and investigated in view of their photophysical and electrochemical properties. The cascades are based on a perchlorinated triphenylmethyl radical acceptor and two triarylamine donors. Absorption spectra showed the presence of charge-transfer bands in the NIR range of the spectra, which pointed to the population of a charge-transfer state between a triarylamine donor and the radical acceptor. A weak to moderate emission in the NIR range of the spectra was observed for all compounds in cyclohexane. Spectroelectrochemical measurements were used to investigate the characteristic spectral features of the oxidised and reduced species of all compounds. Transient absorption spectra in the ns- and fs-time regime revealed an additional hole transfer in the cascades between the triarylamine donors, resulting in a charge-separated state. Charge-separation and -recombination processes were found to be located in the ps-time regime.
Ausgangspunkt war die aus der Fulleren-Chemie bekannte Prato-Reaktion, bei welcher das Ylid in situ aus einer Aminosäure und einem Aldehyd generiert wird und anschließend mit den C=C-Bindungen des Fullerens reagiert. Diese Funktionalisierungsmethode wurde nun auf Detonationsnanodiamant übertragen.
Um zusätzliche π-Bindungen auf der Oberfläche der Diamantteilchen zu schaffen, wurden diese i.Vak. bei 750 °C ausgeheizt (ND750). Für die Immobilisierung wurde die Aminosäure Sarcosin gewählt. Dodecanal und 2,4,6-Tris(hexadecyloxy)-benzaldehyd dienten jeweils als Reaktionspartner.
Da bereits in früheren Studien gezeigt wurde, dass bei dieser Reaktion der Aldehyd selbst unspezifisch an den Diamanten binden kann und so möglicherweise Teile der Oberfläche für die spezifische Funktionalisierung blockiert, wurden für die weitere Betrachtung Azomethinylidvorstufen synthetisiert, die selbst nicht in der Lage sind, mit der Diamantoberfläche zu reagieren. Diesen Zweck erfüllten N-heterocyclische Iminiumbromide, die durch Umsetzung des jeweiligen Heteroaromaten mit Bromessigsäureethylester bzw. Bromacetonitril erhalten wurden.
Alle Ylidvorstufen wurden in Gegenwart von NEt3 in situ zu den gewünschten Dipolen umgesetzt und auf Nanodiamant immobilisiert.
Neben ND750 wurden auch oxidierter und unbehandelter Diamant (NDox bzw. NDunb) sowie Diamant, der bei 900 °C i.Vak. ausgeheizt wurde (ND900), als Substrat eingesetzt, um den Einfluss der Oberflächenterminierung und des Graphitisierungsgrades auf das Reaktionsverhalten zu studieren. Durch Raman- und IR-Spektroskopie wurde gezeigt, dass NDox sehr viele Carbonylgruppen und wenig C=C-Doppelbindungen auf seiner Oberfläche trägt. Durch das Ausheizen i.Vak wurden hingegen zusätzliche π-Bindungen erzeugt, die bei ND900 bereits ausgedehntere Bereiche mit sp2-Kohlenstoff bilden.
Der Erfolg der Immobilisierung wurde IR-spektroskopisch nachgewiesen. Die Oberflächenbeladung aller hergestellten Diamantaddukte wurde thermogravimetrisch bestimmt.
NDox immobilisierte unabhängig vom Reaktionspartner stets die wenigsten Moleküle auf seiner Oberfläche. Deren Terminierung wird von Carbonylgruppen dominiert, die grundsätzlich schlechtere Dipolarophile darstellen als C=C-Doppelbindungen.
Die übrigen Diamantmaterialien NDunb, ND750 und ND900 ließen keine eindeutige Tendenz bezüglich ihrer Reaktionsfreudigkeit erkennen. Die Oberfläche des unbehandelten Diamanten NDunb besitzt sowohl Carbonylfunktionen als auch einzelne Bereiche graphitischen Kohlenstoffs. Diese konkurrieren vermutlich um die angebotenen Dipole, sodass die resultierenden Oberlächenbeladungen ihrer Konjugate in einem mittleren Wertebereich liegen. Durch das Ausheizen i.Vak. werden viele Carbonylgruppen unter Ausbildung weiterer C=C-Doppelbindungen von der Oberfläche entfernt. Bei 750 °C sind diese räumlich sehr beschränkt, stark gekrümmt und daher sehr reaktiv. Trotzdem erreichte ND750 selten eine Oberflächenbelegung, welche jene von NDunb übertrifft. Die π-Bindungen auf seiner Oberfläche sind in Fünf- und Sechsringe eingebaut, um die gekrümmte Struktur zu realisieren. Wahrscheinlich besteht für die Cycloaddition an Nanodiamant eine dem Fulleren C60 ähnliche Regioselektivität bezüglich der angegriffen Doppelbindung. Somit stehen nicht alle frisch erzeugten C=C-Bindungen für die Reaktion zur Verfügung.
Bei 900 °C ist die Graphitisierung der Diamantoberfläche weiter fortgeschritten. Es entstehen nicht nur neue C=C-Bindungen, sondern bereits gebildete Kohlenstoffkappen beginnen zu koaleszieren, wobei ausgedehntere sp2-Bereiche mit geringerer Krümmung und somit verminderter Reaktivität entstehen. So nimmt die Oberflächenbeladung der meisten ND900-Konjugate nicht weiter zu.
Wie aus den Ergebnissen dieser Arbeit hervorgeht, ist die Funktionalisierung von Nanodiamantpartikeln nicht trivial. Sowohl die Oberflächenbeschaffenheit des Diamantmaterials als auch die Struktur des eingesetzten Azomethinylids beeinflussen das Immobilisierungsverhalten.
Die vorliegende Arbeit zeigt aber, dass die 1,3-dipolare Cycloaddition von Azomethinyliden eine nützliche Methode zur Funktionalisierung von Nanodiamantpartikeln ist. Sie ermöglicht des Weiteren die simultane Einführung mehrerer unterschiedlicher funktioneller Gruppen. Dies macht die untersuchte Reaktion zu einem wertvollen Werkzeug für die Herstellung funktionalisierter Nanodiamantmaterialien, z. B. für biomedizinische Anwendungen.
In dieser Arbeit wurde ein geeigneter Zugang zu TBTQ-Derivaten erarbeitet, der eine paarweise Funktionalisierung der aromatischen Positionen in direkter Nachbarschaft zum Triquinacen-Kern zulässt. Hierfür wurde sowohl die doppelte Cyclodehydratisierungsroute als auch die dreifache Cyclisierungsvariante zur Darstellung von Tribenzotriquinacenen dahingehend modifiziert, dass die Einführung der „ortho“-Substituenten bereits zu Beginn der Synthese mit den Edukten erfolgte, da eine nachträgliche Funktionalisierung der gewünschten Positionen am TBTQ aufgrund sterischer Faktoren nicht möglich ist. Es wurden unter anderem die ersten literaturbekannten „ortho“-substituierten TBTQ-Derivate erhalten und vollständig charakterisiert.
Zur Erweiterung des aromatischen Systems wurden außerdem Benzodiphenanthrenotriquinacen-Vorstufen synthetisiert, die im Falle der doppelten Cyclodehydratisierungsroute Acenapthylen-Derivate lieferten. Benzodiphenanthrenotriquinacen konnte mit Hilfe der dreifachen Cyclisierungsvariante synthetisiert werden.
Die Zielmoleküle sollen als Vorstufen für überbrückte aromatisch anellierte Triquinacene dienen.
In summary, it can be stated that the herein studied set of acceptor-substituted squaraine dyes can be seen as potent candidates for OTFTs. Furthermore, their transistor performance can be easily tuned to obtain hole mobilities up to 0.45 cm2/Vs from solution and 1.3 cm2/Vs from sublimation by choosing adequate deposition techniques. In the end, a probable structural model derived from studies of the thin-film morphology by methods such as optical spectroscopy, AFM and X-ray even facilitated the clarification of the observed charge transport behavior.
Infektionskrankheiten gehören weltweit immer noch zu den häufigsten Todesursachen, und auch wenn die Gefährdung in den Industriestaaten erheblich reduziert werden konnte, nimmt die Bedeutung von übertragbaren Krankheiten wieder zu. Verursacht wird dies zum einen durch die Fähigkeit der Keime gegen die eingesetzten Arzneistoffe verschiedenartige Resistenzmechanismen zu entwickeln, zum anderen auch dadurch, dass neuartige Infektionskrankheiten entstehen. Aus diesem Grund bleibt die Entwicklung neuer Medikamente ein ständiger Wettlauf mit der Anpassungsfähigkeit der Infektionserreger, und gerade dies spielt eine große Rolle für vernachlässigte und armutsassoziierte Krankheiten wie z.B. Tuberkulose, Malaria und HIV/AIDS, die in den Entwicklungsländern große Krankheitslasten und so auch hohen volkswirtschaftlichen Schaden verursachen. Protozoische Parasiten wie die Erreger der Malaria und der Leishmaniose sind besonders trickreich, denn sie wechseln zwischen Vektor (z.B. Mücke) und Wirt (z.B. Mensch) und durchleben so verschiedene Stadien eines komplexen Entwicklungszyklus, von denen sich jedes einzelne Stadium wie ein 'anderer' Organismus verhält. Hierdurch ist die therapeutische Behandlung erschwert, und für die dauerhafte Eradikation der Parasiten und für die Hemmung ihrer Transmission, um letztlich eine Resistenzentwicklung der Medikamente zu verhindern, müssen Wirkstoffe möglichst gegen alle Stadien ähnlich gut wirken. Die Konzeptionierung solcher Verbindungen, ihr strukturelles Design und schließlich ihre Synthesen waren Ziel der hier vorliegenden Arbeit, um neue aktive Vertreter gegen protozoische und bakterielle Erreger und Toxine bereitzustellen. Die Konzeptionierung und Synthese von Hybridmolekülen aus bewährten Arzneistoffen wurde als innovativer Ansatz zur Behandlung der Malaria verfolgt. Eine strukturell neue Wirkstoffklasse mit sehr guten spezifischen Aktivitäten und interessanten Struktur-Aktivitäts-Beziehungen gegen Promastigoten und gegen Amastigoten von L. major wurde entdeckt. Auf der Suche nach neuen Verbindungen, die binäre Toxine von Bacillus anthracis Anthrax-Toxin), Clostridium perfringens (Iota-Toxin) und Clostridium botulinum C2-Toxin) hemmen können, wurden neben 4-Aminochinolin-Verbindungen neue Aminochinolinium-Salze konzipiert, synthetisiert und in Target-basierten Assays durch Titrationsexperimente und Stromfluktuationsanalysen bzw. in In-vitro-Experimenten auf ihre Wirksamkeit getestet.
The SARS virus is the etiological agent of the severe acute respiratory syndrome, a deadly disease that caused more than 700 causalities in 2003. One of its viral proteins, the SARS coronavirus main protease, is considered as a potential drug target and represents an important model system for other coronaviruses. Despite extensive knowledge about this enzyme, it still lacks an effective anti-viral drug. Furthermore, it possesses some unusual features related to its active-site region. This work gives atomistic insights into the SARS coronavirus main protease and tries to reveal mechanistic aspects that control catalysis and inhibition. Thereby, it applies state-of-the-art computational methods to develop models for this enzyme that are capable to reproduce and interpreting the experimental observations. The theoretical investigations are elaborated over four main fields that assess the accuracy of the used methods, and employ them to understand the function of the active-site region, the inhibition mechanism, and the ligand binding. The testing of different quantum chemical methods reveals that their performance depends partly on the employed model. This can be a gas phase description, a continuum solvent model, or a hybrid QM/MM approach. The latter represents the preferred method for the atomistic modeling of biochemical reactions. A benchmarking uncovers some serious problems for semi-empirical methods when applied in proton transfer reactions. To understand substrate cleavage and inhibition of SARS coronavirus main protease, proton transfer reactions between the Cys/His catalytic dyad are calculated. Results show that the switching between neutral and zwitterionic state plays a central role for both mechanisms. It is demonstrated that this electrostatic trigger is remarkably influenced by substrate binding. Whereas the occupation of the active-site by the substrate leads to a fostered zwitterion formation, the inhibitor binding does not mimic this effect for the employed example. The underlying reason is related to the coverage of the active-site by the ligand, which gives new implications for rational improvements of inhibitors. More detailed insights into reversible and irreversible inhibition are derived from in silico screenings for the class of Michael acceptors that follow a conjugated addition reaction. From the comparison of several substitution patterns it becomes obvious that different inhibitor warheads follow different mechanisms. Nevertheless, the initial formation of a zwitterionic catalytic dyad is found as a common precondition for all inhibition reactions. Finally, non-covalent inhibitor binding is investigated for the case of SARS coranavirus main protease in complex with the inhibitor TS174. A novel workflow is developed that includes an interplay between theory and experiment in terms of molecular dynamic simulation, tabu search, and X-ray structure refinement. The results show that inhibitor binding is possible for multiple poses and stereoisomers of TS174.
Die paläotropischen Pflanzenfamilien der Ancistrocladaceae und Dioncophyllaceae sind die bisher einzig bekannten Produzenten von Naphthylisochinolin-Alkaloiden. Diese spezielle Klasse acetogeniner Sekundärmetabolite weist durch die verschiedenen Kupplungspositionen der beiden namensgebenden Molekülbausteine eine breite strukturelle Diversität auf und zeichnet sich durch vielfältige pharmakologische Wirksamkeiten, z.B. antiplasmodiale, antileishmaniale oder antitrypanosomale Aktivitäten, aus. Zur Synthese dieser Naturstoffe wurde im Arbeitskreis Bringmann eigens eine Methodik entwickelt, das Lacton -Konzept. Diese Methode erlaubt durch eine Vorfixierung der beiden Molekülhälften durch eine Esterbrücke, anschließender intramolekularer Kupplungsreaktion und der stereoselektiven Öffnung des erhaltenen Lactons den atropselektiven Aufbau der Naphthylisochinoline. Als Ziele dieser Arbeit ergaben sich somit die Synthese pharmakologisch und strukturell interessanter Naphthylisochinolin-Alkaloide mittels des Lacton-Konzepts sowie die Isolierung und Strukturaufklärung weiterer Sekundärmetabolite aus Triphyophyllum peltatum (Dioncophyllaceae), welche anschließend auf ihre Bioaktivität hin untersucht werden sollten, um potenziell neue Leitstrukturen für neue Wirkstoffe zu finden.
In this study, a double-donor concept is used to improve the performance of thermally evaporated merocyanine(s)/C60 bulk heterojunction (BHJ) solar cells. It is shown that the co-evaporation of two merocyanine dyes with absorption bands at ~ 500 nm (SW dye) and ~ 650 nm (LW dye), respectively, together with C60 fullerene results in an improvement of open-circuit voltage (VOC), short-circuit current (JSC) as well as total power conversion efficiency (PCE) compared to the best single-donor cell. The enhancement of JSC is attributed to a higher photon harvesting efficiency of the mixed-donor devices due to a better spectral coverage.
Zusammenfassend lässt sich festhalten, dass die in dieser Abreit vorgestellten Squaraine herausragend gute NIR-Absorptions- und NIR-Emissionseigenschaften aufweisen, die sie für zahlreiche Anwendungen interessant machen. Darüber hinaus konnte gezeigt werden, dass ihre besondere cis-Konfiguration und ihr daraus resultierendes Dipolmoment zu vorteilhaften Anordnungen in dünnen Filmen und in Blends mit PCBM führen. Diese Strukturen zeigen für dipolare Moleküle beeindruckende Exzitonen- und Ladungstransporteigenschaften, die vielversprechende Anwendungen in der organischen Elektronik wie in hier untersuchten lösungsprozessierten BHJ-Solarzellen oder auch in OFETs erwarten lassen.
Die Malaria und andere Infektionskrankheiten sind immer noch die Haupttodesursache in Entwicklungsländern. Durch das jahrzehntelange Versäumnis, neue Wirkstoffe zu entwickeln, und durch die rasante Ausbreitung von Resistenzen gegen herkömmliche Medikamente sind in vielen Regionen der Erde besorgniserregende Zahlen über Neuinfektionen und Todesfälle zu beobachten. Die Suche nach neuen Wirkstoffen ist daher dringend erforderlich und die Hauptaufgabe des Sonderforschungsbereichs 630 an der Universität Würzburg. An diesem interdisziplinären Projekt beteiligt sich unsere Forschungsgruppe vor allem mit der Naturstoffklasse der Naphthylisochinolin-Alkaloide. Neben ihren interessanten strukturellen Eigenschaften haben mehrere Vertreter dieser Sekundärmetabolite vielversprechende Aktivitäten gegen Plasmodien, Leishmanien und Trypanosomen. Dioncophyllin C (24), das bisher wirksamste Naphthylisochinolin gegen P. falciparum, zeigt nicht nur eine exzellente Aktivität in vitro, sondern auch in vivo. In Kooperation mit der Forschergruppe von K. Baumann (Braunschweig) führte man QSAR-Studien durch, um die für die biologische Wirkung entscheidenden Strukturmerkmale zu identifizieren und neue vereinfachte Analoga der Leistruktur 24 vorzuschlagen. Ziel der vorliegenden Arbeit war aufbauend auf Vorarbeiten in unserer Gruppe die Darstellung von strukturell vereinfachten Derivaten des Naturstoffs 24. Die Ergebnisse der biologischen Untersuchungen sollten ausgewertet und somit neue Struktur-Wirkungs-Beziehungen aufgestellt werden. Weiterhin sollten auch Chinolinium-Salze, die man als Analoga der N,C-verknüpften Naphthylisochinoline ansehen kann, synthetisiert werden und innerhalb des SFB 630 und bei unseren Partnern am Schweizerischen Tropen- und Public-Health-Institut auf ihre biologische Aktivität untersucht werden. Man erhoffte sich neben möglichen antiinfektiven Eigenschaften auch Rückschlüsse auf Struktur-Wirkungs-Beziehungen. Zusätzlich sollte die synthetische und analytische chemische Expertise unseren Kooperationspartnern in zwei Projekten außerhalb des SFB 630 zur Verfügung gestellt werden. Dabei handelte es sich einerseits um die Strukturaufklärung von Biosyntheseintermediaten mit Hilfe der HPLC-NMR-Kopplung und andererseits um die Darstellung langkettiger Aldehyde für die biologische Untersuchung des Prä-Penetrationsprozesses eines getreideschädigenden Pilzes.
Tropische Infektionskrankheiten wie Malaria, Leishmaniose oder auch die Afrikanische Trypanosomiase sind aufgrund von zunehmenden Resistenzen der Erreger, globaler Erwärmung, aber auch von Versäumnissen in der Vergangenheit bei der kontinuierlichen Weiterentwicklung bestehender sowie der Erforschung neuer Medikamente auch im 21. Jahrhundert noch eine große Bedrohung für Millionen von Menschen. Die Suche nach neuartigen Wirkstoffen und deren Weiterentwicklung zu potenziellen Medikamenten ist daher zwingend erforderlich. Insbesondere Produkte des Sekundärstoffwechsels wie etwa die Alkaloide bilden wichtige Grundlagen als Leitstrukturen für pharmazeutische Wirkstoffe. Eine solche Klasse phytochemischen Ursprungs sind die Naphthylisochinolin-Alkaloide mit interessanten strukturellen Eigenschaften sowie pharmakologischen Wirksamkeiten. Einige Vertreter zeigen ausgeprägte In-vitro-Aktivitäten gegen protozoische Erreger wie Plasmodien, Leishmanien und Trypanosomen. Besonders die neuartige Unterklasse ionischer N,C-verknüpfter Naphthylisochinolin-Alkaloide, wie z.B. Ancistrocladinium A und Ancistrocladinium B, zeichnen sich durch gute antileishmaniale Wirkungen aus. In Vorarbeiten zeigten erste Studien zu Struktur-Aktivitäts-Beziehungen (SAR-Studien) mit vereinfachten N,C-gekuppelten Arylisochinolinen, dass sich durch gezielte Strukturvariation die Aktivität gegen einen Erreger verbessern lässt. Zusätzlich wurde mit ersten Untersuchungen zum Wirkmechanismus dieser interessanten Verbindungen begonnen. Darüber hinaus ermöglicht die kontinuierliche Verbesserung der analytischen Methoden inzwischen die schnelle und gezielte Suche nach neuen Verbindungen aus der Natur. Durch die Anwendung von Online-Analyse-Verfahren, wie z.B. die Kopplung von HPLC mit NMR und MS, gelingt die Aufklärung der Konstitution von Substanzen direkt aus Extrakten. Ziel der vorliegenden Arbeit war die Verbesserung der biologischen Aktivitäten der N,C-verknüpften Arylisochinoline durch strukturelle Derivatisierung sowie Beiträge zur Aufklärung des Wirkmechanismus mittels markierter Verbindungen. Zusätzlich sollten Naturstoffe unter Verwendung moderner HPLC-Kopplungstechniken untersucht und strukturell aufgeklärt werden.
Nach wie vor ist die Zahl der Malaria-Neuerkrankungen mit ca. 500 Millionen Menschen weltweit sehr besorgniserregend. Durch zunehmende Resistenzen der Erreger gegen zahlreiche Arzneimittel wird die Situation zusätzlich verschärft. Daher ist die Suche und Entwicklung neuartiger Medikamente wichtiger denn je. Die Natur ist immer noch das größte Reservoir an neuen Wirkstoffen, welche als Leitstrukturen für Arzneistoffe fungieren. In den letzten Jahren wurde eine große Zahl neuartiger, biologisch aktiver Naturstoffe identifiziert und hinsichtlich ihres Potenzials für eine pharmazeutische Weiterentwicklung analysiert. Die Naphthylisochinolin-Alkaloide gehören zu solch einer vielversprechenden Wirkstoffklasse, da sie sich v.a. durch ihre hervorragenden pharmakologischen Eigenschaften auszeichnen. Kürzlich wurden die ersten N,C-gekuppelten Naphthyldihydroisochinolin-Alkaloide, Ancistrocladinium A und B, entdeckt. Diese Verbindungen weisen als strukturelle Besonderheit eine außergewöhnliche Iminium-Aryl-Achse auf und besitzen zudem exzellente anti-infektive Aktivitäten, insbesondere gegen den Erreger der Orientbeule, Leishmania major. Ziel der vorliegenden Dissertation war die Synthese neuartiger sterisch gehinderter und strukturell vereinfachter Naphthylisochinoline für weiterführende Struktur-Aktivitäts-Beziehungen (SAR-Studien) und die stereochemische Analyse dieser Verbindungen. Zudem sollte eine Syntheseroute zu den neuartigen dimeren C,C-gekuppelten Naphthylisochinolin-Alkaloiden Shuangancistrotectorin A und B entwickelt werden.
The technology of organic photovoltaics offers the possibility of low-cost devices due to easy fabrication procedures and low material consumption and at the same time high flexibility concerning the applied substrates or design features such as the color palette. Owing to these benefits, this research field is highly active, being reflected by the continuously rising number of publications. Chapter 1 gives an extensive overview of a part of these reports, namely the field of solution-processed BHJ organic solar cells using small molecules as electron-donating materials. In the early years of this research area (2006-2008), well known hole transporting materials such as triphenylamine based chromophores, oligothiophenes and polyaromatic hydrocarbons were applied. However, many of these dyes lacked absorption at longer wavelengths and were therefore limited in their light harvesting qualities. Later, chromophores based on low band gap systems consisting of electron-donating and electron-accepting units showing internal charge transfer overcame this handicap. Today, donor-substituted diketopyrrolopyrroles (D-A-D chromophores), squaraines (D-A-D chromophores) and acceptor substituted oligothiophenes (A-D-A chromophores) are among the most promising dyes for small molecule based organic solar cells with PCEs of 4-5%. This work is based on the findings of the groups of Würthner and Meerholz, which tested merocyanine dyes for the first time in organic BHJ solar cells.4 According to the Bässler theory85, the high dipolarity of these dyes should hamper the charge transport, but the obtained first results with PCE of 1.7% proved the potenital of this class of dyes for this application. Merocyanine dyes offer the advantages of facile synthesis and purification, high tinctorial strength and monodispersity. Additionally, the electronic structure of the dyes, namely the absorption as well as the electrochemical properties, can be adjusted by using the right combination of donor and acceptor units. For these reasons, this class of dye is highly interesting for the application in organic solar cells. It was the aim of the thesis to build more knowledge about the potential and limitations of merocyanines in BHJ photovoltaic devices. By screening a variety of donor and acceptor groups a comprehensive data set both for the molecular materials as well as for the respective solar devices was generated and analyzed. As one focus, the arrangement of the chromophores in the solid state was investigated to gain insight about the packing in the solar cells and its relevance for the performance of the latter. To do so, X-ray single crystal analyses were performed for selected molecules. By means of correlations between molecular properties and the characteristics of the corresponding solar cells, several design rules to generate efficient chromophores for organic photovoltaics were developed. This thesis concerned the quantification of cell adhesion molecules (CAM) in and on thin hydrogel films as surface modification of biomaterials. The established and well characterized, per se inert NCO-sP(EO-stat-PO) hydrogel system which allows the easy and reproducible bioactivation with peptides was used as basis for this thesis. Two methods can be used to functionalize the coatings. Ligands can either be mixed into the prepolymer solution in prior to layer formation (mix-in method), or freshly prepared coatings can be incubated with ligand solution (incubation method). Divided into three major parts, the first part of the thesis dealt with the concentration of ligands in the bulk hydrogel, whereas the second part of the thesis focused on the surface sensitive quantification of CAMs at the biointerface. The results were correlated with cell adhesion kinetics. The third part of this thesis investigated the biochemical and the structural mimicry of the extracellular matrix (ECM). ECM proteins were presented via sugar-lectin mediated binding and cell behavior on these surfaces was analyzed. Cell behavior on three-dimensional fibers with identical surface chemistry as the coatings in the previous sections of the thesis was analyzed and correlated with the amount of peptide used for bioactivation. Overall, the main question of this work was ‘How much?’ regarding maximal as well as optimal ligand concentrations for controlled cell-hydrogel interactions. The focus in the first practical part of this thesis was to analyze the amount of ligands in NCO-sP(EO-stat-PO) hydrogels using classical quantification methods. Coatings in 96-well plates as well as on glass were functionalized with GRGDS and 125I-YRGDS for radioisotopic detection (Chapter 3). Using the incubation method for functionalization, a maximal ligand binding using peptide concentrations of 600 µg/mL could be determined. When functionalization was introduced via the mix-in method, a clear tendency for higher ligand concentrations with increasing ligand to prepolymer ratio was observed, but no maximal ligand binding could be detected with a ligand to prepolymer ratio of 2/1 being the highest ratio investigated. This ratio of 2/1 was not exceeded to ensure that complete crosslinking of the hydrogel was not affected. In Chapter 4, a fluorinated amino acid and an iodinated peptide were immobilized to the hydrogels using the mix-in method and were detected by X-ray photoelectron spectroscopy (XPS) and time-of-flight secondary ion mass spectrometry (TOF-SIMS). In these measurements, maximal ligand binding was detected for a ligand to prepolymer ratio of 1/1. Higher ligand to prepolymer ratios did not result in any significant increase in ligand concentrations in the surface near regions of the crosslinked hydrogels. To address the question of how many ligands were actually accessible for cell interaction at the interface, surface sensitive quantification methods were applied in the second part of this thesis. For the quantification with surface plasmon resonance (SPR) and surface acoustic wave technology (SAW) (Chapter 5), the hydrogel coating procedure needed to be transferred onto cystamine functionalized gold surfaces. Characterization with ellipsometry and atomic force microscopy (AFM) revealed inhomogeneous cystamine binding to the activated surfaces, which resulted in inhomogeneous coatings. Nevertheless, it could be shown that SPR as well as SAW were suitable methods for the surface sensitive quantification of the ligand concentration on NCO-sP(EO-stat-PO) hydrogels. Non-functionalized coatings resisted non-specific serum as well as streptavidin (SA) adsorption. Coatings functionalized with biocytin and GRGDSK-biotin introduced specific SA binding that was dependent on the biotin concentration at the surface. Additionally, enzyme linked immunosorbent assay (ELISA) and enzyme linked lectin assay (ELLA) (Chapter 6) were applied to coatings in 96-well plates and on glass. Coatings were functionalized with the model molecule biocytin, the biotinylated peptide GRGDSK-biotin, the ECM protein fibronectin (FN), as well as the carbohydrates N-acetylglucosamine (GlcNAc) and N-acetyllactosamine (LacNAc). All ligands could be successfully detected with antibodies or SA via ELISA or ELLA. Maximal GRGDSK-biotin binding to the hydrogel coatings on glass was achieved at a peptide to prepolymer ratio of 1/5, which was used as reference value in Chapter 8. Last but not least, cell adhesion (Chapter 7) was quantified depending on the GRGDS concentration on hydrogel coatings on glass. Maximal adhesion of primary human dermal fibroblast (HDF) was observed at GRGDS to prepolymer ratios of 1/5, when adherent cells were counted on life cell images. Quantification of adherent cells using the CASY® cell counter revealed maximal HDF adhesion at molar ligand to prepolymer ratios of 1/2. However, cell vitality detected by intracellular enzyme activities was not dependent on the GRGDS concentration. Cells which managed to adhere were vital regardless of the amount of ligands present. Additionally, adhesion of fibroblasts from the murine cell line NIH L929 was analyzed by counting on life cell images. These cells, being much smaller than the HDF cells, needed higher GRGDS to prepolymer ratios (2/1) for proper cell adhesion. All quantification methods applied to analyze hydrogels which were functionalized by the mix-in method in Chapter 3, 4, 6 and 7, were compared in Chapter 8. Radiodetection gave information about the ligand concentrations throughout the whole hydrogel and no maximal amount of ligands could be detected when increasing the peptide to prepolymer ratio. In contrast, XPS and TOF-SIMS which only penetrated the surface near regions of the coating, a maximal ligand binding to the hydrogel was detected for 1/1 ratios. SPR and SAW were not included in this comparison, as the coatings on gold need to be optimized first. The two surface sensitive quantification methods (ELISA and HDF adhesion) could give information about the quantity of peptide which was sterically available for SA or cell binding. With these methods, maximal SA and cell binding was detected at ratios of 1/5. These results underline the importance of carefully compare the different methods. Beside ligand quantification on hydrogels, the third part of this thesis was concerned with the biochemical and structural mimicry of the ECM by advanced ECM engineering to design biomimetic biomaterials that are better accepted by cells and tissue. The subject of Chapter 9 was the biomimetic and flexible presentation of the ECM protein FN. FN was attached via sugar-lectin mediated binding to NCO-sP(EO-stat-PO) hydrogels. The build-up of the covalently immobilized sugar poly-N-acetyllactosamine (polyLacNAc), the subsequent non-covalent binding of the fungal galectin His6CGL2, and FN could be elegantly proven by fluorescent staining on coatings which were functionalized with the sugar by micro contact printing (MCP). Further experiments were carried out on build-ups, where polyLacNAc was immobilized on the hydrogel by incubation. Optimal parameters for the layer build-up were determined by ELLA/ELISA. Only the complete build-up induced proper adhesion of HDFs. Compared to tissue culture polystyrene (TCPS), cells adhered and spread faster on the biomimetic surfaces. The flexible presentation of FN allowed HDFs to rearrange homogenously immobilized FN into fibrillar structures, which seemed not to be possible when FN was adsorbed on glass or covalently bound directly to the hydrogel coatings. This new approach of a flexible and biomimetic presentation of an ECM protein allows new ways to design biomaterials with best possible cell-material interactions. The work described in Chapter 10 focused on the structural mimicry of the fibrous ECM structures by electrospinning of synthetic, bioactive, and degradable fibers. Poly(D,L-lactide-co-glycolide) (PLGA) and NCO-sP(EO-stat-PO) were electrospun out of one solution in an easy one-step preparation resulting in fibers with an ultrathin inert hydrogel layer at the surface. By adding GRGDS to the solution prior to electrospinning, specifically interacting fibers could be obtained. In comparison to PLGA, the adsorption of bovine serum albumin (BSA) could be reduced by 99.2%. As a control, the non-active peptide GRGES was immobilized to the fiber. These fibers did not allow cell adhesion, showing that the integrity of the hydrogel coated fibers was not affected by the immobilization of peptides. HDF adhesion was obtained by functionalization with GRGDS, leading to the adhesion, spreading, and proliferation of HDFs. Also mesenchymal stem cells (MSC) could adhere to GRGDS functionalized fibers. Additionally, for ligand quantification, the ELISA technique was successfully transferred to fiber substrates. To highlight the potential of the approaches for the biochemical and structural mimicry of the ECM, the sugar polyLacNAc was immobilized on the PLGA/sP(EO-stat-PO) fibers followed by the subsequent layer build-up with His6CGL2 and FN. These fibers triggered HDF adhesion.
The thesis enhances the strategy of non-destructive fluorescence read-out in rylene bisimide-diarylethene containing photochromic systems. The fluorescence of the emitter unit is quenched by a photoinduced electron transfer only to one of the isomeric forms of the photochrome. The driving force of the fuorescence-quenching electron transfer was calculated by the help of the Rehm-Weller equation. The novel photochromic systems satisfy the necessary requirements for non-destructive read-out in write/read/erase fluorescent memory devices.
Biomolecules are difficult to investigate in their native environment. The vast complexity of cellular systems and seldom availability of chemical reactions compatible with the physiological milieu make it a challenging task. Bioorthogonal chemical reactions serve as a key to achieve selective ligation, whose components must react rapidly and selectively with each other under physiological conditions in the presence of the plethora of functionalities necessary to sustain life. In this dissertation, we focused on the synthesis of chemical reporters and probe molecules for bioorthogonal labeling through click reaction. Initially, sialic acid derivatives with a linker containing terminal alkyne functionality were synthesized. After the synthesis of azide derivatives of fluorescent dyes as counter partners, they were conjugated with sialic acids through Cu(I) catalyzed alkyne azide cycloaddition (CuAAC). The successful in vitro conjugation of Sia and fluorescent dyes was followed by metabolic tagging of human larynx carcinoma (HEp-2) and the carcinoma of Chinese hamster ovary (CHOK1) with alkynated Sia that were subsequently ligated with fluorescein azide. Finally, the stained cells were subjected to fluorescent microscopy to obtain their images. To enable the click reaction compatible to in vivo applications, the reactivity of cyclooctyne was enhanced by two different approaches. In a first approach, following the Bertozzi’s strategy, two fluorine atoms were introduced adjacent to the alkyne to lower the LUMO. In a second strategy the ring strain of cyclooctyne was attempted to be enhanced by the introduction of an amide group. In addition, glutarimide derivatives with free amino and carboxylic acid functional groups were synthesized by domino-Michael addition-cyclization-reaction.
Beschrieben ist die Isolierung und Strukturaufklärung von Naturstoffen (Naphthochinone, dimere Naphthaline und Naphthylisochinolin-Alkaloide) aus Zell- und Wurzelkulturen von Triphyophyllum peltatum. Darüber hinaus wurden Experimente zur Biosynthese der Naphthylisochinolin-Alkaloide mit 13C2-markierten Vorstufen durchgeführt. Ebenso Bestandteil der Arbeit war die Etablierung von Kalluskulturen von Aloe saponaria, aus der ebenfalls Sekundärmetabolite isoliert wurden.
Plant-derived natural products and their analogs continue to play an important role in the discovery of new drugs for the treatment of human diseases. Potentially promising representatives of secondary metabolites are the naphthylisoquinoline alkaloids, which show a broad range of activities against protozoan pathogens, such as plasmodia, leishmania, and trypanosoma. Due to the increasing resistance of those pathogens against current therapies, highly potent novel agents are still urgently needed. Thus, it is worthy to discover new naphthylisoquinoline alkaloids hopefully with pronounced bioactivities by isolation from plants or by synthesis. The naphthylisoquinoline alkaloids are biosynthetically related to another class of plant-derived products, the naphthoquinones, some of which have been recently found to display excellent anti-multiple myeloma activities without showing any cytotoxicities on normal blood cells. Multiple myeloma still remains incurable, although remissions may be induced with co-opted therapeutic treatments. Therefore, more potent naphthoquinones are urgently required, and can be obtained by isolation from plants or by synthesis. In detail, the results in this thesis are listed as follows: 1) Isolation and characterization of naphthylisoquinoline alkaloids from the stems of a Chinese Ancistrocladus tectorius species. Nine new naphthylisoquinoline alkaloids, named ancistectorine A1 (60), N-methylancistectorine A1 (61), ancistectorine A2 (62a), 5-epi-ancistectorine A2 (62b), 4'-O-demethylancistectorine A2 (63), ancistectorine A3 (64), ancistectorine B1 (65), ancistectorine C1 (66), and 5-epi-ancistrolikokine D (67) were isolated from the Chinese A. tectorius and fully characterized by chemical, spectroscopic, and chiroptical methods. Furthermore, the in vitro anti-infectious activities of 60-62 and 63-66 have been tested. Three of the metabolites, 61, 62a, and 62b, exhibited strong antiplasmodial activities against the strain K1 of P. falciparum without showing significant cytotoxicities. With IC50 values of 0.08, 0.07, and 0.03 μM, respectively, they were 37 times more active than the standard chloroquine (IC50 = 0.26 μM). Moreover, these three compounds displayed high antiplasmodial selectivity indexes ranging from 100 to 3300. According to the TDR/WHO guidelines, they could be considered as lead compounds. In addition, seven alkaloids, 69-74 (structures not shown here), were isolated from A. tectorius that were known, but new to the plant, together with another fourteen known compounds (of these, only the structures of the three main alkaloids, 5a, 5b, and 78 are shown here), which had been previously found in the plant. The three metabolites ancistrocladine (5a), hamatine (5b), and (+)-ancistrocline (78) were found to show no or moderate activities against the MM cell lines. 2) Isolation and characterization of naphthylisoquinoline alkaloids from the root bark of a new, botanically yet undescribed Congolese Ancistrocladus species. An unprecedented dimeric Dioncophyllaceae-type naphthylisoquinoline alkaloid, jozimine A2 (84), as first recognized by G. Bauckmann from an as yet undescribed Ancistrocladus species, was purified and characterized as part of this thesis. Its full structural assignment was achieved by spectroscopic and chiroptical methods, and further confirmed by an X-ray diffraction analysis, which had never succeeded for any other dimeric naphthylisoquinoline alkaloids before. Structurally, the dimer is composed of two identical 4'-O-demethyldioncophylline A halves, coupled through a sterically hindered central axis at C-3',3'' of the two naphthalene moieties. Pharmacologically, jozimine A2 (84) showed an extraordinary antiplasmodial activity (IC50 = 1.4 nM) against the strain NF54 of P. falciparum. Beside jozimine A2 (85), another new alkaloid, 6-O-demethylancistrobrevine C (84), and four known ones, ancistrocladine (5a), hamatine (5b), ancistrobrevine C (86), and dioncophylline A (6) were isolated from the Ancistrocladus species, the latter in a large quantity (~500 mg), showing that the plant produces Ancistrocladaceae-type, mixed-Ancistrocladaceae/Dioncophyllaceae-type, and Dioncophyllaceae-type naphthyl- isoquinoline alkaloids. Remarkably, it is one of the very few plants, like A. abbreviatus, and A. barteri, that simultaneously contain typical representatives of all the above three classes of alkaloids. 3) Semi-synthesis of jozimine A2 (85), 3'-epi-85, jozimine A3 (93) and other alkaloids from dioncophylline A (6). The dimeric naphthylisoquinoline alkaloids, jozimine A2 (85) and 3'-epi-85, constitute rewarding synthetic targets for a comparative analysis of their antiplasmodial activities and for a further confirmation of the assigned absolute configurations of the isolated natural product of 85. They were semi-synthesized in a four-step reaction sequence from dioncophylline A (6) in cooperation with T. Büttner. The key step was a biomimetic phenol-oxidative dimerization at C-3' of the N,O-dibenzylated derivative of 89 by utilizing Pb(OAc)4. This is the first time that the synthesis of such an extremely sterically hindered (four ortho-substituents) naphthylisoquinoline alkaloid – with three consecutive biaryl axes! – has been successfully achieved. A novel dimeric naphthylisoquinoline, jozimine A3 (93), bearing a 6',6''-central biaryl axis, was semi-synthesized from 5'-O-demethyldioncophylline A (90) by a similar biomimetic phenol-oxidative coupling reaction as a key step, by employing Ag2O. HPLC analysis with synthetic reference material of 3'-epi-85 and 93 for co-elution revealed that these two alkaloids clearly are not present in the crude extract of the Ancistrocladus species from which jozimine A2 (85) was isolated. This evidences that jozimine A2 (85) is very specifically biosynthesized by the plant with a high regio- and stereoslectivity. Remarkably, the two synthetic novel dimeric naphthylisoquinoline alkaloids 3'-epi-85 and 93 were found to display very good antiplasmodial activities, albeit weaker than that of the natural and semi-synthetic product 85. Additionally, the two compounds 3'-epi-85 and 93 possessed high or moderate selectivity indexes, which were much lower than that of 85. However, they can still be considered as new lead structures. Two unprecedented oxidative products of dioncophylline A, the diastereomeric dioncotetralones A (94a) and B (94b), were synthesized from dioncophylline A (6) in a one-step reaction. Remarkably, the aromatic properties in the “naphthalene” and the “isoquinoline” rings of 94a and 94b are partially lost and the “biaryl” axis has become a C,C-double bond, so that the two halves are nearly co-planar to each other, which has never been found among any natural or synthetic naphthylisoquinoline alkaloid. Their full structural characterization was accomplished by spectroscopic methods and quantum-chemical CD calculations (done by Y. Hemberger). The presumed reaction mechanism was proposed in this thesis. In addition, one of the two compounds, 94a, exhibited a highly antiplasmodial activity (IC50 = 0.09 μM) with low cytotoxicity, and thus, can be considered as a new promising lead structure. Its 2'-epi-isomer, 94b, was inactive, evidencing a significant effect of chirality on the bioactivity. Of a number of naphthylisoquinoline alkaloids tested against the multiple-myeloma cell lines, the three compounds, dioncophylline A (6), 4'-O-demethyldioncophylline A (89), and 5'-O-demethyldioncophylline A (90) showed excellent activities, even much stronger than dioncoquinones B (10), C (102), the epoxide 175, or the standard drug melphalan. 4) Isolation and characterization of bioactive naphthoquinones from cell cultures of Triphyophyllum peltatum. Three new naphthoquinones, dioncoquinones C (102), D (103), and E (104), the known 8-hydroxydroserone (105), which is new to this plant, and one new naphthol dimer, triphoquinol A (107), were isolated from cell cultures of T. peltatum in cooperation with A. Irmer. Dioncoquinone C (102) showed an excellent activity against the MM cells, very similar to that of the previously found dioncoquinone B (10), without showing any inhibitory effect on normal cells. The other three naphthoquinones, 103105, were inactive or only weakly active. 5) Establishment of a new strategy for a synthetic access to dioncoquinones B (10) and C (102) on a large scale for in vivo experiments and for the synthesis of their analogs for first SAR studies. Before the synthesis of dioncoquinone B (10) described in this thesis, two synthetic pathways had previously been established in our group. The third approach described here involved the preparation of the joint synthetic intermediate 42 with the previous two routes. The tertiary benzamide 135 was ortho-deprotonated by using s-BuLi/TMEDA, followed by transmetallation with MgBr2▪2Et2O, and reaction with 2-methylallyl bromide (139). It resulted in the formation of ortho-allyl benzamide 140, which was cyclized by using methyl lithium to afford the naphthol 42. This strategy proved to be the best among the established three approaches with regard to its very low number of steps and high yields. By starting with 136, this third strategy yielded the related bioactive natural product, dioncoquinone C (102), which was accessed by total synthesis for the first time. To identify the pharmacophore of the antitumoral naphthoquinones, a library of dioncoquinone B (10) and C (102) analogs were synthesized for in vitro testing. Among the numerous naphthoquinones tested, the synthetic 7-O-demethyldioncoquinone C (or 7-O-hydroxyldioncoquinone B) (145), constitutes another promising basic structure to develop a new anti-MM agent. Furthermore, preliminary SAR results evidence that the three hydroxy functions at C-3, C-5, and C-6 are essential for the biological properties as exemplarily shown through the compounds 10, 102, and 145. All other mixed OH/OMe- or completely OMe-substituted structures were entirely inactive. By a serendipity the expoxide 175 was found to display the best anti-MM activity of all the tested isolated metabolites from T. peltatum, the synthesized naphthoquinones, and their synthetic intermediates. Toxic effects of 175 on normal cells were not observed, in contrast to the high toxicities of all other epoxides. Thus, the anti-MM activity of 175 is of high selectivity. The preliminary SAR studies revealed that the 6-OMe group in 175 is required, thus differed with the above described naphthoquinones (where 6-OH is a requisite in 10, 102, and 145), which evidenced potentially different modes of action for these two classes of compounds. 6) The first attempted total synthesis of the new naturally occurring triphoquinone (187a), which was recently isolated from the root cultures of T. peltatum in our group. A novel naphthoquinone-naphthalene dimer, 187a (structure shown in Chapter 10), was isolated in small quantities from the root cultures of T. peltatum. Thus, its total synthesis was attempted for obtaining sufficient amounts for selected biotestings. The key step was planned to prepare the extremely sterically hindered (four ortho-substituents) binaphthalene 188 by a coupling reaction between the two 2-methylnaphthalene derivatives. Test reactions involving a system of two simplified 2-methylnaphthylboron species and 2-methylnaphthyl bromide proved the Buchwald ligand as most promising. The optimized conditions were then applied to the two true - highly oxygenated - coupling substrates, between the 2-methylnaphthylboron derivatives 210, 211, 213, or 214 and the 2-methylnaphthyl iodides (or bromides) 215 (206), 215 (206), 212 (205), or 212 (205), respectively. Unfortunately, this crucial step failed although various bases and solvent systems were tested. This could be due to the high electron density of the two coupling substrates, both bearing strongly OMOM/OMe-donating function groups. Cyclodysidins A-D (216-219), four new cyclic lipopeptides with a- and ß-amino acids, were isolated from the Streptomyces strain RV15 derived from a marine sponge by Dr. U. Abdelmohsen. Their structures were established as cyclo-(ß-AFA-Ser-Gln-Asn-Tyr-Asn-Ser-Thr) by spectroscopic analysis using 2D NMR techniques and CID-MS/MS in the course of this thesis. In conclusion, the present work contributes to the discovery of novel antiplasmodial naphthylisoquinoline alkaloids and antitumoral naphthoquinones, which will pave the way for future studies on these two classes of compounds.
Eine Reihe von Acetylen-verknüpften Perylenbisimid(PBI)-Makrozyklen mit unterschiedlicher Ringgröße wurde durch Palladium-katalysierte Homokupplung synthetisiert und mit Hilfe von Recycling-GPC getrennt. Diese Makrozyklen wurden durch NMR-Spektroskopie und Massenspektrometrie charakterisiert und weiterhin die photophysikalischen Eigenschaften durch UV/Vis-Absorptions- und Fluoreszenzemissions-Messungen untersucht. Die Selbstorganisation dieser PBI-Makrozyklen zu hochgeordneten Nanostrukturen auf HOPG-Oberflächen wurde mittels Rasterkraftmikroskopie untersucht.
Im Laufe der Evolution entwickelten Pflanzen, Bakterien, Pilze und Tiere eine Vielzahl von Signal-, Boten- und Abwehrstoffen. Diese für die Organismen oft lebensnotwendigen Sekundärmetabolite werden von den jeweiligen Produzenten oft sehr effizient und teilweise auch hoch stereoselektiv produziert. Die Effizienz und Selektivität, mit denen diese Stoffe mit den in der Natur zur Verfügung stehenden biosynthetischen Mitteln hergestellt werden, ist für Chemiker im Labor, trotz eines breiten Methodenrepertoires, eine große Herausforderung und oft gelingt die Laborsynthese nur über viele Stufen sowie in geringen Ausbeuten und/oder mit schlechten Enantio- oder Diastereoselektivitäten. Daher wird in der chemischen Synthese immer wieder die Neuentwicklung und Erweiterung von Methoden vorangetrieben. Neben der Synthese im Labor ist auch die Untersuchung von Biosynthesewegen sowie die Charakterisierung der dafür verantwortlichen Enzyme ein interessantes Forschungsgebiet, um von der Natur zu lernen und diese Erkenntnisse für die Entwicklung neuer Synthesestrategien und Methoden zu nutzen. Viele der bislang isolierten Verbindungen zeigen neben den für die produzierenden Organismen wichtigen Wirkungen auch Aktivitäten gegen für Menschen gefährliche Krankheitserreger. Daher ist die Isolierung und Strukturaufklärung neuer bioaktiver Naturstoffe als Leitstrukturen für neue Medikamente ein lohnendes Ziel für Wissenschaftler. In den letzten Jahren wurden aber immer häufiger bereits bekannte Verbindungen isoliert. Zur Vermeidung solcher Mehrfachisolierung werden auch die analytischen Methoden zur Identifikation und Strukturaufklärung stetig verbessert Einen wichtigen Stellenwert hat hierbei die Kopplung von Messgeräten wie z.B. MS oder NMR mit chromatographischen Anlagen wie GC, HPLC oder UPLC eingenommen. Die Kombination von Flüssigchromatographie-Anlagen mit chiroptischen Detektoren wie CD-Spektrometern erlaubt hierbei manchmal sogar die Zuordnung von Absolutkonfigurationen direkt aus dem Rohextrakt. Ziel der vorliegenden Arbeit war einerseits die Anwendung neuer Methoden bei der Entwicklung von Syntheserouten zu axialchiralen Naturstoffen und andererseits die Aufklärung der Konstitution und Konfiguration von Naturstoffen und synthetischen Verbindungen sowie die Untersuchung von Biosyntheseintermediaten.
