The Contribution of Common and Rare Variants to the Complex Genetics of Psychiatric Disorders

Attention deficit/hyperactivity disorder (ADHD), one of the most frequent childhood-onset, chronic and lifelong neurodevelopmental diseases, affects 5 - 10% of school – aged children and adolescents, and 4% of adults. The classified basic symptoms are - according to the diagnostic system DSM-VI - inattentiveness, impulsivity and hyperactivity. Also daily life of patients is impaired by learning problems, relationship crises, conflicts with authority and unemployment, but also comorbidities like sleep - and eating problems, mood - and anxiety disorders, depression and substance abuse disorders are frequently observed. Although several twin and family studies have suggested heritability of ADHD, the likely involvement of multiple genes and environmental factors has hampered the elucidation of its etiology and pathogenesis. Due to the successful medication of ADHD with dopaminergic drugs like methylphenidate, up to now, the search for candidate genes has mainly focused on the dopaminergic and - because of strong interactions - the serotonergic system, including the already analyzed candidate genes DAT1, DRD4 and 5, DBH or 5-HTTLPR. Recently, DNA copy number changes have been implicated in the development of a number of neurodevelopmental diseases and the analysis of chromosomal gains and losses by Array Comparative Genomic Hybridization (Array CGH) has turned out a successful strategy to identify disease associated genes. Here we present the first systematic screen for chromosomal imbalances in ADHD using sub-megabase resolution Array CGH. To detect micro-deletions and -duplications which may play a role in the pathogenesis of ADHD, we carried out a genome-wide screen for copy number variations (CNVs) in a cohort of 99 children and adolescents with severe ADHD. Using high-resolution aCGH, a total of 17 potentially syndrome-associated CNVs were identified. The aberrations comprise four deletions and 13 duplications with approximate sizes ranging from 110 kb to 3 Mb. Two CNVs occurred de novo and nine were inherited from a parent with ADHD, whereas five are transmitted by an unaffected parent. Candidates include genes expressing acetylcholine-metabolising butyrylcholinesterase (BCHE), contained in a de novo chromosome 3q26.1 deletion, and a brain-specific pleckstrin homology domain-containing protein (PLEKHB1), with an established function in primary sensory neurons, in two siblings carrying a 11q13.4 duplication inherited from their affected mother. Other genes potentially influencing ADHD-related psychopathology and involved in aberrations inherited from affected parents are the genes for the mitochondrial NADH dehydrogenase 1 alpha subcomplex assembly factor 2 (NDUFAF2), the brain-specific phosphodiesterase 4D isoform 6 (PDE4D6), and the neuronal glucose transporter 3 (SLC2A3). The gene encoding neuropeptide Y (NPY) was included in a ~3 Mb duplication on chromosome 7p15.2-15.3, and investigation of additional family members showed a nominally significant association of this 7p15 duplication with increased NPY plasma concentrations (empirical FBAT, p = 0.023). Lower activation of the left ventral striatum and left posterior insula during anticipation of large rewards or losses elicited by fMRI links gene dose-dependent increases in NPY to reward and emotion processing in duplication carriers. Additionally, further candidate genes were examined via Matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS). This method enables the analysis of SNPs directly from human genomic DNA without the need for initial target amplification by PCR. All these findings implicate CNVs of behavior-related genes in the pathogenesis of ADHD and are consistent with the notion that both frequent and rare variants influence the development of this common multifactorial syndrome. The second part of this work concentrates on MLC1, a gene associated with Megalencephalic leukoencephalopathy with subcortical cysts, located on chromosome 22q13.33. To get more insight in the disease itself, a targeting vector for a conditional knockout mouse was constructed using homologous recombination. Furthermore, MLC1 has been suggested as a risk gene for schizophrenia, especially the periodic catatonia subtype. An initially identified missense mutation was found to be extremely rare in other patient cohorts; however, a recent report again argued for an association of two intronic MLC1 SNPs with schizophrenia and bipolar disorder. A case-control study of these polymorphisms as well as SNPs in the transcriptional control region of MLC1 was conducted in 212 chronic schizophrenic patients, 56 of which suffered from periodic catatonia, 106 bipolar patients, and 284 controls. Both intronic and promoter polymorphisms were specifically and significantly associated with periodic catatonia but not schizophrenia or bipolar disorder in general. A haplotype constructed from all polymorphisms was also associated with periodic catatonia. The MLC1 variation is associated with periodic catatonia; whether it constitutes a susceptibility or a modifier gene has to be determined.
Das Glioblastom ist mit einem Anteil von 20% an allen hirneigenen Tumoren der häufigste und bösartigste primäre intrakranielle Tumor. Trotz multimodalem Therapiekonzept, das operative Resektion, Strahlen- und Chemotherapie verbindet, haben Patienten eine Prognose von im Mittel nur 14,6 Monaten. Sein aggressives Wachstum zieht eine Vaskularisierung nach sich, die jedoch nicht in ausreichendem Maße die Sauerstoffversorgung des Tumorgewebes sicherstellen kann. Studien mit Messelektroden zeigten einen deutlich reduzierten Sauerstoffpartialdruck im Tumor im Vergleich zum umliegenden Hirngewebe. Dieses hypoxische Milieu löst genetische Alterationen und adaptive Veränderungen der Proteinexpression aus, die eine Selektion besonders aggressiver Tumorzellen bewirkt. Für eine bessere prognostische Einschätzung sowie als zukünftige therapeutische Ziele zur Erhöhung der Response auf Chemo- und Strahlentherapie ist es von großem Interesse, solche Faktoren als mögliche Hypoxiemarker aufzufinden. In dieser Arbeit wurden die Proteine HIF-1α, CAIX, VEGF, EPO und NDRG1 auf mRNA- und Proteinebene in den Glioblastomzelllinien GaMG, U251 und U373 auf eine Veränderung ihrer Expression unter hypoxischen Bedingungen untersucht. Ausmaß (5% O2, 1% O2 und 0,1% O2) und Dauer (1 h, 6 h und 24 h) der Hypoxie wurde variiert. Anschließend wurde über 24 h und 48 h eine Reoxygenierung durchgeführt. Auch wurden Expressionsuntersuchungen an Gewebeproben von Normalhirnen, Astrozytomen WHO Grad II und Glioblastomen vorgenommen. Die Verwendbarkeit von GAPDH als Marker für diese Analysen wurde durch Experimente sichergestellt, die dessen mRNA und das Protein als nicht durch Hypoxie oder Malignisierung reguliert nachwiesen. Wir bestätigten die Rolle von HIF-1α als Mediator der hypoxischen Zellantwort. Während die mRNA konstant blieb, wurde das Protein unter hypoxischen Bedingungen hochreguliert. Dies zeigte auch, dass unser experimentelles Setting funktionierte. NDRG1, CA-IX sowie EPO wurden unter Hypoxie sowohl auf mRNA-, als auch Proteinebene hochreguliert und blieben unter Reooxygenierung stabil. In Glioblastomen waren diese Gene auf mRNA-Ebene bedeutend stärker exprimiert als in niedriggradigen Astrozytomen. HIF-1α, NDRG1, CA-IX sowie EPO können also in humanen Glioblastomzellen als Hypoxiemarker dienen und möglicherweise auch eine prognostische und therapeutische Bedeutung haben.
Characterization of allosteric mechanisms on the M2 and M4 mACh receptor using the FRET-technique

Allosteric modulators have been proposed as promising new compounds to modify protein function. Allosteric binding sites have been discovered for several G-protein-coupled receptors, including M1-5 muscarinic receptors. Since these receptors play a pivotal role in the regulation of a plethora of organ functions, it is particularly important to investigate the mechanisms of allosteric modulation. To study molecular mechanisms of allosteric modulation in the M2 muscarinic receptor, a new FRET-based sensor was designed. CFP fused to the C-terminus of the receptor and a small fluorescent compound FlAsH, which labels a specific binding sequence in the third intracellular loop, were used as donor and acceptor fluorophores, respectively. The first part of the study was to design a functional FRET receptor sensor. After several optimization steps the constructs FLAG-M2-sl3-FlAsH-GSGEG-CFP and HA-FLAG-M2-sl3-FlAsH-GSGEG-CFP were generated which showed good cell-surface expression, robust changes in FRET and the ability to deliver reproducible data. The second part of this thesis sought to elucidate the mechanisms of the allosteric ligand binding and their effects on the receptor conformation. The described modifications, which were introduced in the wild type M2 mAChR to create the FRET sensor can alter receptor functionality and influence receptor expression. Radioligand binding studies revealed that the used transfection method provided sufficient receptor expression but, unfortunately, about 60 % of the FLAG-M2-sl3-FlAsH-GSGEG-CFP receptor remains in the cytosol. However, this was sufficient to perform FRET experiments. Patch clamp GIRK-measurements with acetylcholine evinced that the new M2-sensor was able to activate Gi-proteins. Also, radioligand-binding assays with the second construct HA-FLAG-M2-sl3-FlAsH-GSGEG-CFP showed ligand affinity comparable to the wildtype receptor. Furthermore inhibition of forskolin-stimulated cAMP production was indistinguishable from the behaviour of the wildtype receptor. According to that, the full functionality of both receptor constructs could be confirmed. FRET measurements with the full muscarinic receptor agonists carbachol and acetylcholine confirmed that the FLAG-M2-sl3-FlAsH-GSGEG-CFP receptor construct showed rapid changes in FRET upon addition of both ligands, which were concentration-dependent. Concentration response curves and the resulting EC50 values of both agonists were similar to those already published in literature. In addition, the orthosteric antagonists atropine and methoctramine inhibited the FRET changes induced by the agonists. This inhibition was significantly faster than the washout kinetics, pointing to an active displacement of the agonists by the antagonists. Allosteric ligands gallamine, tacrine and dimethyl-W84 did not alter receptor conformation when added without an orthosteric ligand. However, when applied in addition to muscarinic agonists, all three substances inhibited the FRET-signal. The extent of this inhibition was dependent on the used concentration of the allosteric ligands. These results reveal that conformational changes brought about by allosteric ligands can be measured with the FRET technique. Furthermore real-time FRET-based kinetic measurements could be performed in living cells and showed that the allosteric ligands gallamine and dimethyl-W84 alter receptor conformation significantly faster than the antagonists atropine and methoctramine. This data indicate that allosteric ligands actively induce the conformational changes in the receptor.
Der Prozess von der Entdeckung und Entwicklung eines potentiellen Arzneistoffs bis zu dessen Zulassung ist extrem kosten‐ und zeitintensiv und eine Vielzahl dieser Stoffe kann aufgrund toxischer Nebenwirkungen in präklinischen Studien nicht weiterentwickelt werden. Dabei ist die Niere eines der Hauptziele von Xenobiotika‐induzierter Organtoxizität, jedoch ist eine frühe Detektion von Nierenschäden schwierig. Den derzeitig verwendeten klinischen Parameter, wie Blutharnstoff (BUN) und Serumkreatinin fehlt es an Sensitivität und Spezifität, da sie Fremdstoff‐induzierte Toxizität meist erst aufzeigen, wenn schon ein erheblicher Teil der Nierenfunktion beeinträchtigt ist. Daher ist es notwendig, empfindlichere und zuverlässigere Biomarker zu identifizieren und zu validieren, welche kleinste Nierenschädigungen früher als traditionelle Parameter erkennen. In den letzten Jahren wurden in der Literatur aber auch von verschiedenen Projekten eine Reihe neuer gen‐basierender und Urinbiomarker (Kim‐1, Clusterin, Lipocalin‐2, Timp‐1) identifiziert. Ziel dieser Dissertation war es die Aussagekraft dieser Marker im Vergleich zu traditionellen Endpunkten, einschließlich klinische Chemie und Histopathologie an Gewebe‐, Urin‐ und Serumproben von männlichen Ratten, welche mit Modellsubstanzen für Nephrotoxizität (Aristolochiasäure und Gentamicin) oder nephrotoxischen Arzneistoffkandidaten (PredTox Projekt) behandelt wurden, mittels qRT‐PCR, Immunhistochemie und ELISA zu untersuchen. Zusammenfassend kann man sagen, dass die Effekte auf Ebene der Gen‐ und Proteinexpression generell sehr gut mit den histopathologischen Veränderungen korrelieren. Sie konnten meist früher oder in niedrigeren Dosierungen als die traditionellen Nierenmarker BUN und Serumkreatinin detektiert werden. Eine erhöhte Expression und Exkretion von Kim‐1 zeigte sich in allen Studien als eine der frühesten Antworten auf Schädigung der proximalen Tubuli und stellt somit den empfindlichsten Biomarker dar. Die erhöhte Ausscheidung von Clusterin konnte teilweise vor einer veränderten Gen‐ und Proteinexpression im Gewebe detektiert werden und unterstützen die Verwendung von Clusterin als nicht‐invasiven Biomarker. Obwohl eine gesteigerte Exkretion von Lipocalin‐2 sehr früh nach Schädigung des proximalen Tubulus detektiert werden konnte, ist diese nicht spezifisch für einen Nierenschaden. Dennoch könnte die vermehrte Expression/Ausscheidung von Lipocalin‐2 als frühe Antwort auf eine Entzündung oder einen Gewebeschaden eine sinnvolle Ergänzung der routinemäßigen Testung auf Toxizität darstellen. Ebenfalls konnte ein dosis‐ und zeitabhängiger Konzentrationsanstieg von einem Großteil der potentiellen Biomarker des „WideScreen™ Rat Kidney Toxicity Panels 1 and 2“ im Urin beobachtet werden. Da jedoch die potentiellen Biomarker unterschiedliche Empfindlichkeiten besitzen und unter Umständen auch vom Mechanismus der Toxizität von Verbindungen abhängen, erscheint eine Kombination von verschiedenen Biomarkern zur frühzeitigen Erkennung von proximalen Nierenschäden sowie zur Verlaufskontrolle von Nierenerkrankungen sinnvoll. Durch die einfache Probenahme und leichte Bestimmung ist die Messung der neuen potentiellen Nierenbiomarker im Urin neben der Bestimmung der traditionellen Parameter der klinischen Chemie sowie der Histopathologie sinnvoll für die Identifizierung von Nierenschädigungen in präklinischen Studien.
Der Parathormonrezeptor Typ 1 (PTHR) ist ein G-Protein-gekoppelter Rezeptor der Gruppe 2 und wichtigster Regulator des Kalziumstoffwechsels. Im ersten Teil der Arbeit wurde eine neuartige posttranslationale Modifikation des PTHR in Form einer proteolytischen Spaltung der Ektodomäne identifiziert, charakterisiert und deren Regulation beschrieben. Nach langanhaltender Stimulation des Rezeptors mit Agonisten – aber nicht mit Antagonisten – wurde eine Massen- und Mengenzunahme des Rezeptorproteins beobachtet. Es konnte gezeigt werden, dass der Rezeptor unter basalen Bedingungen einer Spaltung unterliegt. Der Massenunterschied entsteht durch die proteolytische Spaltung der Ektodomäne des PTHR, was nachfolgend die Stabilität des Rezeptors beeinträchtigt. Die Spaltung erfolgte innerhalb einer unstrukturierten Schleife der Ektodomäne, welche die Bereiche für die Ligandenbindung miteinander verbindet. Hierbei handelt es sich um eine Region, die im Vergleich zu anderen Gruppe 2-Rezeptoren spezifisch für den PTHR ist. Das durch die Spaltung entstandene N-terminale Fragment bleibt durch eine Disulfidbrücke mit dem Transmembranteil des Rezeptors verbunden. Durch Versuche mit verschiedenen Proteaseinhibitoren konnte die verantwortliche Protease der Familie der zinkabhängigen extrazellulären Proteasen zugeordnet werden. Diese Ergebnisse beschreiben einen Mechanismus wie die Homoöstase des PTHR reguliert sein könnte. In einem zweiten Abschnitt wurde die Interaktion der Adapterproteine NHERF1 und beta-Arrestin2 mit dem PTHR untersucht. Beide Proteine interagierten unabhängig mit dem Rezeptor, wobei NHERF1 über eine PDZ-Domäne konstitutiv an den C-Terminus des Rezeptors bindet. beta-Arrestin2 hingegen bindet nach Aktivierung des Rezeptors und führt zur Desensitisierung des Rezeptors. Mittels biochemischer und mikroskopischer Methoden konnte gezeigt werden, dass beide Proteine gemeinsam einen ternären Komplex mit dem PTHR bilden, welcher durch die direkte Interaktion zwischen NHERF1 und beta-Arrestin2 vermittelt wird. Dies hat zur Folge, dass beta-Arrestin im basalen Zustand durch NHERF1 an den Rezeptor gekoppelt wird. Durch Analyse der Assoziationskinetik mittels Fluoreszenz-Resonanz-Energietransfer-Messungen zeigte sich, dass diese Kopplung zu einer zweifach erhöhten Rekrutierungsgeschwindigkeit von beta-Arrestin2 an den PTHR führt. Somit stellt unterstützt NHERF1 die beta-Arrestin2-vermittelte Desensitisierung des PTHR.
In this study poplar trees have been examined under different stress conditions. Apart from the detailed descriptions above two main conclusions might be drawn: i) A small plant like Arabidopsis thaliana is highly susceptible to stress situations that might become life-threatening compared to a tree that has extremely more biomass at its disposal. Such an organism might be able to compensate severe stress much longer than a smaller one. It seems therefore reasonable that a crop like Arabidopsis reacts earlier and faster to a massive threat. ii) In poplar both tested stress responses seemed to be regulated by hormones. The reactions to abiotic salt stress are mainly controlled by ABA, which also has a strong impact upon cold and drought stress situations. The term commonly used for ABA is “stress hormone” and is at least applicable to all abiotic stresses. In case of herbivory (biotic stress), jasmonic acid appears to be the key-player that coordinates the defence mechanism underlying extrafloral nectary and nectar production. Thus the presented work has gained a few more insights into the complex network of general stress induced processes of poplar trees. Future studies will help to understand the particular role of the intriguing indirect defence system of the extrafloral nectaries in more detail.
Hochporöse Kohlenstoffaerogele, die über den Sol-Gel-Prozeß auf der Basis von Resorzin und Formaldehyd hergestellt werden, sind Werkstoffe mit beeindruckenden physikalischen Eigenschaften. Leider werden bisher nur geringe Mengen an Kohlenstoffaerogelen produziert und aus Kostengründen auf günstigere Materialien mit vergleichsweise schlechteren Eigenschaften zurückgegriffen. Um diesen Nachteil zu nivellieren lag die Motivation der vorliegenden Arbeit in der Entwicklung neuer Syntheserouten für Kohlenstoffmaterialien mit nanoskaliger Morphologie, wobei insbesondere auf kostengünstige Edukte und/oder einfache Prozessierung zurückgegriffen werden sollte. Als in Frage kommende Eduktsysteme wurden Zucker, sowie Hydroxybenzol-Formaldehyd-Derivate ausgewählt. Die hergestellten Kohlenstoffe wurden hauptsächlich mit Elektronenmikroskopie, Gassorption und Röntgenkleinwinkelstreuung (SAXS) charakterisiert. Um Fehlinterpretationen der experimentellen Daten für das neue Materialsystem zu vermeiden, war ein umfangreiches Wissen zu den Charakterisierungsmethoden und den diesen zugrundeliegenden physikalischen Prinzipien notwendig. Kohlenstoffpulver basierend auf sphärischen Resorzin-Formaldehyd Suspensionen und Sedimenten bilden eine völlig neue Möglichkeit zur Erzeugung von Kohlenstoffnanokugeln. Im Rahmen dieser Arbeit wurde deshalb systematisch der Bereich der Syntheseparameter im RF-System zu den nicht-monolithischen Parametersätzen hin vervollständigt. Anhand der bestimmten Daten konnte diese Stoffklasse umfassend und detailliert charakterisiert und interpretiert werden. Die Partikelgröße hängt im Wesentlichen von der Katalysatorkonzentration und in geringerem Maße von der Eduktmenge in der Startlösung ab. Die ermittelte untere Grenze der Partikelgröße aus stabilen kolloidalen Dispersionen beträgt ca. 30 nm. Größere Partikel als 5 µm konnten trotz Modifikation der Syntheseroute nicht erzeugt werden. Eine Abschätzung über den Aggregationsgrad der Kohlenstoffpulver wurde durchgeführt. Eine Beimischung von Phenol verringert in diesem System zum einen die Partikelgröße und erzeugt zunehmend nicht-sphärische Strukturen. Die aus Gassorption, SAXS und dynamischer Lichtstreuung (DLS) ermittelten Partikelgrößen stimmen gut überein. Bei der Pyrolyse schrumpfen die Partikel auf 84% des Ausgangswerts (Partikeldurchmesser). Ein Fokus dieser Arbeit lag in der Herstellung poröser Kohlenstoffe mit Phenol und Formaldehyd (PF) als Eduktbasis und unterkritischer Trocknung (Kohlenstoffxerogele). Um die Bandbreite der Eigenschaften der resultierenden Kohlenstoffxerogele zu erweitern, wurden zahlreiche Modifikationen der Syntheseparameter und im Herstellungsprozeß durchgeführt. Die Ergebnisse zeigen, daß im Eduktsystem Phenol-Formaldehyd in wäßriger Lösung mit Na2CO3 als basischem Katalysator prinzipiell poröse Xerogele herstellbar sind; allerdings verhindert eine ungewöhnliche Gelierkinetik (Flockenbildung statt Sol-Gel-Übergang) eine umfassende Interpretation des Systems, da die Reproduzierbarkeit der Ergebnisse nicht gewährleistet ist. Bei Phenol-Formaldehyd in wäßriger Lösung und NaOH als Katalysator kommt es meist zu einem Kollabieren des Gelnetzwerks während der Trocknung. Lediglich bei hohem Formaldehydüberschuß zeigt sich ein enger Bereich, in dem Xerogele mit geringer Dichte (rhomin = 0,22 g/cm3) und relevantem Mesoporenvolumen von bis zu 0,59 cm3/g synthetisierbar sind. Die interessanteste Kombination im PF-System ergibt sich mit HCl als Katalysator und n-Propanol als Lösungsmittel. Hier sind hochporöse Kohlenstoffxerogele mit geringen Dichten (rhomin = 0,23 g/cm3) und für Xerogele sehr hoher Mesoporosität von bis zu Vmeso = 0,85 cm3/g möglich. Damit ist es im Rahmen dieser Arbeit erstmals gelungen über konvektive Trocknung homogene hochporöse Xerogel-Formkörper auf PF-Basis zu synthetisieren. Aus der Überwachung des Sol-Gel-Prozesses mit Detektion der Soltemperatur konnten wichtige Erkenntnisse über exo- und endotherme Vorgänge gewonnen werden. Zudem zeigt die Zeitabhängigkeit der Soltemperatur Gemeinsamkeiten für alle untersuchten Hydroxybenzol-Formaldehyd-Systeme. So kann der Gelpunkt der Ansätze zuverlässig und auch reproduzierbar anhand eines zweiten lokalen Temperaturmaximums ermittelt werden, welches mit einer Gelpunktsenthalpie korreliert wird. Damit ist auch eine Prozeßkontrolle, z.B. für die Kombination mit Partikeltechnologien, möglich. Die zugrundeliegenden Strukturbildungsmechanismen, Sol-Gel-Prozeß einerseits und Trocknung andererseits, wurden in-situ mittels SAXS beobachtet und anhand der gewonnenen Daten diskutiert und bewertet. Eine vollständige Adaption des etablierten und akzeptierten Bildungsmechanismus von RF basierten Aerogelen (Partikelbildung aus Kondensationskeimen und Partikelwachstum) für das PF-System wird ausgeschlossen. Vielmehr scheint bei den untersuchten PF-Systemen auch eine Mikrophasenseparation als konkurrierender Prozeß zur Partikelbildung von Relevanz zu sein.
Perylene bisimide (PBI) dyes are a widely used class of industrial pigments, and currently have gained significant importance for organic-based electronic and optical devices. Structural modification at the PBI core results in changes of the optical and electronic properties, which enable tailored functions. Moreover, the aggregation behavior of PBIs is alterable and controllable to achieve new materials, among which organogels are of particular interest because of their potential for applications as supramolecular soft materials. In this work, new PBI-based organic gelators were designed, synthesized, and characterized, and the aggregation behaviors under different conditions were intensively studied by various spectroscopic and microscopic methods. In chapter 2, a brief overview is given on the structural and functional features of organogel systems. The definition, formation and reversibility of organogels are introduced. Some examples on dye based organogel are selected, among which PBI-based organogelators reported so far are especially emphasized. Some basic knowledges of supramolecular chirality are also overviewed such as characterization, amplification, and symmetry breaking of the chiral aggregates. According to our former experiences, PBIs tend to form aggregates because the planer aromatic cores interact with one another by pi-pi interaction. In chapter 3, a new PBI molecule is introduced which possesses amide groups between the conjugated core and periphery alkyl chains. It is found that well oriented aggregates are formed by hydrogen bonding and the pi-pi interaction of the cores. These interactions enable the aggregates to grow in one-dimension forming very long fibers, and these fibers further intercross to 3D network structures, e.g., organogels. In comparison to the very few PBI-based gelators reported before, one advantage of this gelator is that, it is more versatile and can gelate a wide range of organic solvents. Moreover, the well-organized fibers that are composed of extended π-stacks provide efficient pathways for n-type charge carriers. Interestingly, AFM studies reveal that the PBI molecules form well-defined helical fibers in toluene. Both left-handed (M) and right-handed (P) helicities can be observed without any preference for one handedness because the building block is intrinsically achiral. In chapter 4, we tried to influence the M/P enantiomeric ratio by applying external forces. For example, we utilized chiral solvents to generate chiral aggregates with a preferential handedness. AFM analysis of the helices showed that a enantiomeric ratio of about 60: 40 can be achieved by aggregation in chiral solvents R- or S-limonene. Moreover, the long aggregated fibres can align at macroscopic level in vortex flows upon rotary stirring In chapter 5, bulky tetra-phenoxy groups are introduced in the bay area of the PBI gelator. The conjugated core of the new molecule is now distorted because of the steric hindrance. UV/Vis studies reveal a J-type aggregation in apolar solvents like MCH due to intermolecular pi-pi-stacking and hydrogen-bonding interactions. Microscopic studies reveal formation of columnar aggregates in apolar solvent MCH, thus this molecule lacks the ability to form gels in this solvent, but form highly fluorescent lyotropic mesophases at higher concentration. On the other hand, in polar solvents like acetone and dioxane, participation of the solvent molecules in hydrogen bonding significantly reduced the aggregation propensity but enforced the gel formation. The outstanding fluorescence properties of the dye in both J-aggregated viscous lyotropic mesophases and bulk gel phases suggest very promising applications in photonics, photovoltaics, security printing, or as fluorescent sensors. In chapter 6, we did some studies on combining PBI molecules with inorganic gold nanorods. Gold nanorods were synthesized photochemically. By virtue of the thioacetate functionalized PBIs, the rods were connected end to end to form gold nanochains, which were characterized by absorption spectra and TEM measurement. Such chromophore-nanorod hybrids might be applied to guide electromagnetic radiation based on optical antenna technology.
trans-1,1,1,3-Tetrafluoropropene (HFO-1234ze) and 2,3,3,3-tetrafluoropropene (HFO-1234yf) are non-ozone-depleting fluorocarbon replacements with low global warming potentials and short atmospheric lifetimes. They are developed as foam blowing agent and refrigerant, respectively. Investigations on biotransformation in different test species and in vitro systems are required to assess possible health risks of human exposure and needed for commercial development. The biotransformation of HFO-1234ze and HFO-1234yf was therefore investigated after inhalation exposure. Male Sprague-Dawley rats were exposed to air containing 2 000; 10,000; or 50,000 ppm (n=5/concentration) HFO-1234ze or HFO-1234yf. Male B6C3F1 mice were only exposed to 50,000 ppm HFO-1234ze or HFO-1234yf. Due to lethality observed in a developmental study with rabbits after exposure to high concentrations of HFO-1234yf, the metabolic fate of the compound was tested by whole body inhalation exposure of female New Zealand White rabbits to air containing 2 000; 10,000; or 50,000 ppm (n=3/concentration) HFO-1234yf. All inhalation exposures were conducted for 6 h in a dynamic exposure chamber. After the end of the exposures, animals were individually housed in metabolic cages and urines were collected at 6 or 12 h intervals for 48 h (rats and mice) or 60 h (rabbits). For metabolite identification, urine samples were analyzed by 1H-coupled and 1H-decoupled 19F-NMR and by LC/MS-MS or GC/MS. Metabolites were identified by 19F-NMR chemical shifts, signal multiplicity, 1H-19F coupling constants and by comparison with synthetic reference compounds. Biotransformation of HFO-1234ze in rats exposed to 50,000 ppm yielded S-(3,3,3-trifluoro-trans-propenyl)mercaptolactic acid as the predominant metabolite which accounted for 66% of all integrated 19F-NMR signals in urines. No 19F-NMR signals were found in spectra of rat urine samples collected after inhalation exposure to 2 000 or 10,000 ppm HFO-1234ze likely due to insufficient sensitivity. S-(3,3,3-Trifluoro-trans-propenyl)-L-cysteine, N-acetyl-S-(3,3,3-trifluoro-trans-propenyl)-L-cysteine, 3,3,3-trifluoropropionic acid and 3,3,3-trifluorolactic acid were also present as metabolites in urine samples of rats and mice at the 50,000 ppm level. A presumed amino acid conjugate of 3,3,3-trifluoropropionic acid was the major metabolite of HFO-1234ze in urine samples of mice exposed to 50,000 ppm and related to 18% of total integrated 19F-NMR signals. Quantitation of three metabolites in urines of rats and mice was performed, using LC/MS-MS or GC/MS. The quantified amounts of the metabolites excreted with urine in both mice and rats, suggest only a low extent (<<1% of dose received) of biotransformation of HFO-1234ze and 95% of all metabolites were excreted within 18 h after the end of the exposures (t1/2 approx. 6 h). Due to its low boiling point of −22 °C, most of the inhaled HFO-1234ze is expected to be readily exhaled. Moreover, steric and electronic factors may decrease the reactivity of the parent compound with soft nucleophiles such as glutathione. The obtained results suggest that HFO-1234ze is subjected to an addition-elimination reaction with glutathione and to a cytochrome P450-mediated epoxidation at low rates. The extent of a direct addition reaction of HFO-1234ze with glutathione is negligible, compared to that of the observed addition-elimination reaction. The results of in vivo testing of HFO-1234ze could not be supported by in vitro investigations, since HFO-1234ze was not metabolized in incubations with either liver microsomes or subcellular fractions from rat and human. Regarding the structures delineated in the biotransformation scheme of HFO-1234ze, 1,1,1,3-tetrafluoroepoxypropane and 3,3,3-trifluoropropionic acid are toxic intermediates which, however, are not supposed to display toxicity in the species after exposure to HFO-1234ze, due to the low extent of formation and an efficient detoxification of the epoxide by hydrolysis and glutathione conjugation. The findings of biotransformation of HFO-1234ze in rats and mice correlate with the absence of adverse effects in the toxicity testings and indicate their innocuousness to a human exposure. Biotransformation of HFO-1234yf yielded N-acetyl-S-(3,3,3-trifluoro-2-hydroxypropanyl)-L-cysteine as predominat metabolite which accounted for approx. 44, 90 and 32% (50,000 ppm) of total 19F-NMR signal intensities in urine samples from rabbits, rats and mice, respectively. S-(3,3,3-Trifluoro-2-hydroxypropanyl)mercaptolactic acid and the sulfoxides of mercapturic acid and mercaptolactic acid S-conjugate were identified as minor metabolites of HFO-1234yf in urine samples from rabbits, rats and mice, whereas trifluoroacetic acid, 3,3,3-trifluorolactic acid and 3,3,3-trifluoro-1-hydroxyacetone were present as minor metabolites only in urine samples from rats and mice. In dieser Arbeit konnte gezeigt werden, wie verschiedene Techniken zur Verstärkung der Raman-Streuung eingesetzt werden können, um selektiv und sensitiv Wirkstoffe zu charakterisieren und Proteine zu lokalisieren. Die UV-Resonanz-Raman-Spektroskopie wurde zur selektiven Verfolgung der Wirkstoff-Substrat-Wechselwirkung zwischen einem Guanidiniocarbonyl-basierten Peptidrezeptor und seinem Substrat eingesetzt. Durch die enorme Resonanzverstärkung der Ramanstreuung konnten in einer Bindungsstudie die spektralen Änderung bei der Komplexierung des Rezeptors mit einem Tetrapeptid bei submillimolarer Konzentration in Wasser verfolgt werden. Die oberflächenverstärkte Raman-Streuung (surface-enhanced Raman scattering, SERS) wurde zur ultrasensitiven Detektion von festphasengebundenen Substanzen eingesetzt. Die selektive Verstärkung der auf dem Harz gebundenen Substanz wurde durch aggregierte Silber-Nanopartikeln auf der Oberfläche der Harzkügelchen ermöglicht. So konnte auf einem einzigen Harzkügelchen in wenigen Sekunden das SERS-Spektrum einer Substanzmenge von nur 50 fmol aufgenommen werden. In einem SERS-mikroskopischen Raster-Experiment an der Oberfläche eines einzelnen Harzpartikels konnte die hohe Reproduzierbarkeit dieser Technik demonstriert werden. Zwei neue SERS-Marker zur Detektion von Biomolekülen werden abschließend vorgestellt. Bei beiden Marker-Typen werden die Raman-Signale von selbstorganisierenden Monolagen (self-assembled monolayer, SAM) aus Raman-Markern durch Gold/Silber-Nanoschalen als SERS-Substrat verstärkt. Der erste neue SERS-Marker-Typ wurde mit einer SAM aus Raman-Markern mit zwei unterschiedlich langen hydrophilen Abstandshalter-Gruppen synthetisiert. Über das Verhältnis von kurzem zu langem Abstandshalter kann die sterische Hinderung der Bindungsstellen in der SAM zur kontrollierten Konjugation an Antikörper minimiert werden. Der zweite SERS-Marker-Typ beruht auf der Silicaverkapselung einer SAM auf den Gold/Silber-Nanoschalen. Die silicaverkapselten SERS-Marker konnten nach der Oberflächenfunktionalisierung mit Amino-Gruppen über heterobifunktionelle Verknüpfungsreagenzien an Antikörper gekoppelt werden. Beide SERS-Marker wurden zur immunhistochemischen Lokalisierung des prostataspezifischen Antigens in Prostatagewebeschnitten eingesetzt.
Several lines of evidence implicate a dysregulation of tryptophan hydroxylase (TPH)-dependent serotonin (5-HT) synthesis in emotions and stress and point to their potential relevance to the etiology and pathogenesis of various neuropsychiatric disorders. However, the differential expression pattern of the two isoforms TPH1 and TPH2 which encode two forms of the rate-limiting enzyme of 5-HT synthesis is controversial. Here, a comprehensive spatio-temporal analysis clarifies TPH1 and TPH2 expression during pre- and postnatal development of the mouse brain and in adult human brain as well as in peripheral organs including the pineal gland. Four different methods (real time PCR, in situ hybridization, immunohistochemistry and Western blot analysis) were performed to systematically control for tissue-, species- and isoform-specific expression on both the pre- and posttranslational level. TPH2 expression was consistently detected in the raphe nuclei, as well as in fibres in the deep pineal gland and in the gastrointestinal tract. Although TPH1 expression was found in these peripheral tissues, no significant TPH1 expression was detected in the brain, neither during murine development, nor in mouse and human adult brain. Also under conditions like stress and clearing the tissue from blood cells, no changes in expression levels were detectable. Furthermore, the reuptake of 5-HT into the presynaptic neuron by the serotonin transporter (SERT) is the major mechanism terminating the neurotransmitter signal. Thus, mice with a deletion in the Sert gene (Sert KO mice) provide an adequate model for human affective disorders to study lifelong modified 5-HT homeostasis in interaction with stressful life events. To further explore the role of TPH isoforms, Tph1 and Tph2 expression was studied in the raphe nuclei of Sert deficient mice under normal conditions as well as following exposure to acute immobilization stress. Interestingly, no statistically significant changes in expression were detected. Moreover, in comparison to Tph2, no relevant Tph1 expression was detected in the brain independent from genotype, gender and treatment confirming expression in data from native animals. Raphe neurons of a brain-specific Tph2 conditional knockout (cKO) model were completely devoid of Tph2-positive neurons and consequently 5-HT in the brain, with no compensatory activation of Tph1 expression. In addition, a time-specific Tph2 inducible (i) KO mouse provides a brain-specific knockdown model during adult life, resulting in a highly reduced number of Tph2-positive cells and 5-HT in the brain. Intriguingly, expression studies detected no obvious alteration in expression of 5-HT system-associated genes in these brain-specific Tph2 knockout and knockdown models. The findings on the one hand confirm the specificity of Tph2 in brain 5-HT synthesis across the lifespan and on the other hand indicate that neither developmental nor adult Tph2-dependent 5-HT synthesis is required for normal formation of the serotonergic system, although Tph1 does not compensate for the lack of 5-HT in the brain of Tph2 KO models. A further aim of this thesis was to investigate the expression of the neuropeptide oxytocin, which is primarily produced in the hypothalamus and released for instance in response to stimulation of 5-HT and selective serotonin reuptake inhibitors (SSRIs). Oxytocin acts as a neuromodulator within the central nervous system (CNS) and is critically involved in mediating pain modulation, anxiolytic-like effects and decrease of stress response, thereby reducing the risk for emotional disorders. In this study, the expression levels of oxytocin in different brain regions of interest (cortex, hippocampus, amygdala, hypothalamus and raphe nuclei) from female and male wildtype (WT) and Sert KO mice with or without exposure to acute immobilization stress were investigated. Results showed significantly higher expression levels of oxytocin in brain regions which are involved in the regulation of emotional stimuli (amygdala and hippocampus) of stressed male WT mice, whereas male Sert KO as well as female WT and Sert KO mice lack these stress-induced changes. These findings are in accordance with the hypothesis of oxytocin being necessary for protection against stress, depressive mood and anxiety but suggest gender-dependent differences. The lack of altered oxytocin expression in Sert KO mice also indicates a modulation of the oxytocin response by the serotonergic system and provides novel research perspectives with respect to altered response of Sert KO mice to stress and anxiety inducing stimuli.
Der ‚Liber de arte distillandi de simplicibus’, genannt das ‚Kleine Destillierbuch’ des Straßburger Chirurgen Hieronymus Brunschwig (ca. 1450 bis 1512/13) wurde im Jahr 1500 erstmals veröffentlicht. In der vorliegenden Untersuchung sollen die darin genannten humoralpathologischen medizinischen Indikationen der destillierten Pflanzenwässer mit den nach derzeitigem wissenschaftlichem Erkenntnisstand belegten Indikationen verglichen werden und auf Übereinstimmungen und Abweichungen hin untersucht werden. Das ‚Kleine Destillierbuch’ befasst sich mit der Destillation von Arzneimitteln vorwiegend pflanzlicher Herkunft und den medizinischen Anwendungsbereichen dieser Destillate. Das erfolgreiche Werk, das in schriftlicher Form die damalige gängige Praxis der Medizinkundigen in Straßburg fixiert, entwickelte sich zum heilkundlichen Volksbuch des 16. Jahrhunderts, galt aber auch bei Berufs-Destillierern als wichtiges Technik-Lehrbuch für Destillationsverfahren. Daneben waren Brunschwigs Pflanzenbeobachtungen im ‚Kleinen Destillierbuch’ eine wertvolle Grundlage für die ältere deutsche Botanik. Für die Untersuchung wurden Druckausgaben aus den Jahren 1528 und 1610 verwendet. In einem ersten Schritt wurde durch Klärung heute ungebräuchlicher Begriffe der frühneuzeitliche Text erschlossen. Im zweiten Schritt wurden die Zutaten identifiziert, die Brunschwig zur Destillation der 269 pflanzlichen und 36 nicht-pflanzlichen, meist tierischen gebrannten Wässer verwendet. Im dritten Schritt wurden alle Indikationen der Destillate, die Brunschwig in seinen Planzenmonographien nennt, gesammelt und verschiedenen Körperregionen zugeordnet. Anschließend wurden den historischen Indikationsbezeichnungen heute gebräuchliche Bezeichnungen gegenübergestellt, um einen direkten Vergleich der Daten, die zwei grundsätzlich verschiedenen Wissenschaftsystemen entspringen, zu ermöglichen. Im vierten Schritt wurden alle von Brunschwig genannten Anwendungsbereiche der einzelnen „Wässer“-Monographien jeweils in Tabellenform aufgenommen. Diesen historischen Anwendungsbereichen wurden heutige naturwissenschaftlich belegte Indikationen gegenübergestellt. Hierfür wurde v.a. auf W. Blaschek, S. Ebel, E. Hackenthal u. a., Hagers Handbuch der Drogen und Arzneistoffe, HagerROM 2004, Programmversion 5.1, Berlin, Heidelberg 2005 zurückgegriffen. Ein Vergleich der historischen mit den aktuellen Anwendungen erfolgte anhand abgestufter Kategorien (I, II, III, IV). Für eine Interpretation der untersuchten Daten wurden diejenigen von Brunschwig verwendeten Pflanzen ausgewählt, die ein nach heutigem Wissensstand belegtes Anwendungsgebiet besitzen. Diese wurden zunächst nach den für ihre Wirkung relevanten Inhaltsstoffen geordnet. Die Übereinstimmungen bzw. Abweichungen der Brunschwigschen Anwendungsgebiete von den heute definierten Anwendungsgebieten der untersuchten Pflanzen wurden diskutiert. Die relativen Trefferhäufigkeiten wurden ermittelt. Sie liegen zwischen 33 % und 100 %, im Durchschnitt bei 65 %. Für eine weitere Diskussion wurden die gleichen Daten bezüglich der Destillat-Anwendung in den oben genannten Körperregionen geordnet. Die relativen Trefferhäufigkeiten liegen hier in einem Bereich zwischen 43 % und 86 %, im Durchschnitt bei 57 %. Die vorliegende Untersuchung ist Teil eines größeren Forschungsvorhabens. In gleicher Weise sollen mehrere Kräuterbücher des Mittelalters bzw. der frühen Neuzeit untersucht und anschließend mittels eines statistischen Verfahrens einzeln und im Vergleich umfassend ausgewertet werden.
Die vorliegende Dissertation beschreibt detaillierte biochemische und biophysikalische Untersuchungen von rekombinanten Porinen aus den beiden pathogenen Bakterien Nocardia farcinica und Vibrio cholerae sowie von nativen Porinen aus drei Streptomyces Arten. Für die beiden pathogenen Vertreter sind bereits impermebable Zellwandstrukturen beschrieben worden (Daffe et al., 1993; Rodriguez-Torres et al., 1993). Für Streptomyces Arten ist keine vergeichbare, definierte äußere Zellwandbarriere bekannt. Im Fall von S. griseus konnten dennoch undefinierte, kovalente Lipidverknüpfungen mit der Peptidoglykanschicht nachgewiesen werden (Kim et al., 2001). Daher besteht die Notwendigkeit des Transports von Nährstoffen und anderer Moleküle auch bei Streptomyces Arten durch porenformende Proteine, so genannte Porine. Unter Porinen versteht man wassergefüllte Kanäle, die in zwei Klassen unterteilt werden können: allgemeine Diffusionsporen und substratspezifische Porine. Allgemeine Diffusionsporen filtern entsprechend der molekularen Masse der gelösten Substrate und weisen ein lineares Verhältnis zwischen Translokationsrate und Substratkonzentrationsgradient auf. Dagegen kann der Transport bestimmter Substanzen durch spezifische Porine mit einer Substrat-Bindestelle im Kanal durch die Michaelis-Menten-ähnliche Kinetik beschrieben werden. Diese Kanäle ermöglichen den schnellen Influx bestimmter Klassen von Substraten.
Aus früheren Untersuchungen ist bekannt, dass Fleischerzeugnisse höhere Gehalte an polychlorierten Biphenylen (PCB) und polychlorierten Dibenzo-p-dioxinen bzw. polychlorierten Dibenzofuranen (PCDD/F) aufweisen als das unverarbeitete Fleisch selbst. Aufgrund dessen war es Ziel dieser Arbeit, mögliche Kontaminationsquellen für diese Substanzen in Fleischerzeugnissen zu finden. Als Einstieg in die Thematik wurden Bratwurstproben auf ihren WHO-PCB- und WHO-PCDD/F-TEQ untersucht. Dabei hat man auch Produktionsgeräte für Fleischerzeugnisse auf ihr Potential geprüft, diese mit den genannten Substanzen zu kontaminieren. Bei den weiteren Arbeiten wurde besonderes Augenmerk auf die Untersuchung von verschiedenen Gewürzen und Gewürzextrakten gelegt. Wiesen Gewürze Kontaminationen mit den genannten Stoffen auf, wurde möglichen Ursachen nachgegangen. Eine weitere, wichtige Fragestellung war in diesem Zusammenhang, ob und in welchem Ausmass PCBs und Dioxine durch die Extraktion der Gewürze mit organischen Lösemitteln bzw. überkritischem CO2 in den Extrakt übergehen können. Die Bestimmung der WHO-PCDD/F- bzw. WHO-PCB-TEQ in Gewürzen erfolgte nach einem von uns speziell für Gewürze entwickeltem, aufwendigem Clean-Up mittels Kapillargaschromatographie und anschließender hochauflösender Massenspektrometrie (HRGC-HRMS). Die Abtrenn- und Aufreinigungsschritte umfassten (i) Gefriertrocknung, (ii) Homogenisation mit Zusatz an internem Standard, (iii) Separierung mittels beschleunigter Lösemittelextraktion; (iv) verschiedene nacheinander geschaltete Flüssigchromatographie-Schritte (Gelpermeation; Florisil; schwefelsaures Kieselgel; Aktivkohle). Zusammenfassend lässt sich an Einzelergebnissen festhalten: - Aufgrund der Untersuchung von Bratwurstproben (aus einem Zeitraum von zwei Jahren) waren betriebsspezifische Ursachen wie Betriebseinrichtungen - z.B. veraltete Geräte oder ähnliches - für eine Kontamination von Fleischerzeugnissen mit PCB oder Dioxinen nicht grundsätzlich auszuschließen. Sie sind aber wenig wahrscheinlich. - Lediglich bei der Verwendung von über 40 Jahre alten Geräten ergab sich eine Tendenz zur Kontamination von Fleischerzeugnissen mit PCB, aber nicht mit Dioxinen. - Bei der Untersuchung von Gewürzen wurde festgestellt, dass Blattgewürze tendenziell höhere PCB- und Dioxingehalte aufweisen als andere Gewürzarten. - Weder das Erntejahr noch der Erntezeitpunkt (unterschiedliche Wachstumsstadien) sind ausschlaggebend für eine stark variierende Kontamination mit den Analyten. - Bei Aufschlüsselung der einzelnen Proben nach deren Herkunft zeigte sich, dass eine Kontamination standortspezifisch hervorgerufen werden kann. - Die Trocknung (in verschiedenen Varianten) und weitere bei Blattgewürzen gängige Verarbeitungsschritte sind aufgrund der erzielten Ergebnisse als Ursache für die höheren PCB- bzw. Dioxingehalte in Blattgewürzen auszuschließen. - Die Untersuchung von Gewürzextrakten zeigte, dass bei einer CO2-Extraktion PCB und Dioxine zu einem geringeren Grad im Extrakt angereichert werden, als dies bei einer Fest-Flüssigextraktion mit organischen Lösemitteln der Fall ist. Insgesamt betrachtet, kann die Verwendung von naturbelassenen, höchstens getrockneten Gewürzen zu keiner derartigen Kontamination von Fleischerzeugnissen mit PCB oder Dioxinen führen, dass die damit hergestellten Produkte aufgrund einer Überschreitung der gesetzlichen Höchstwerte beanstan¬det werden würden. In einem Rechenmodell ließ sich zeigen, dass bei der Herstellung von Fleisch-erzeugnissen die Verwendung von Gewürzextrakten, die mit organischen Lösungsmitteln hergestellt wurden, einen höheren Eintrag an PCB und Dioxinen im fertigen Fleischerzeugnis hervorruft als dies bei entsprechender Produktion mit naturbelassenen Gewürzen (bei üblichen Einsatzmengen) der Fall ist. Bei Flüssig-CO2-Extrakten hingegen fand im Vergleich zu naturbelassenen Gewürzen ein geringerr Eintrag an PCB und Dioxinen statt.
The widely used chemical acrylamide (AA) has been classified as a probable human carcinogen. This classification was based on positive results in rodent carcinogenicity studies as well as on a number of in vitro mutagenicity assays. In 2002, AA was discovered to be formed during the preparation of starch-containing foods. According to the latest FDA exposure assessment (2006), the average daily intake has been estimated from AA levels in foodstuffs and from nutritional habits to be around 0.4 µg/kg b.w. with a 90th percentile of 0.95 µg/kg b.w.. In children and adolescents however, the daily AA intake is about 1.5 times higher, due to lower body weight and differing consumption patterns. Apart from the diet, humans may be exposed to AA during the production or handling of monomeric AA, from AA residues in polyacrylamides, and from cigarette smoke. After oral administration, AA is readily absorbed and distributed throughout the organism. AA is metabolized to the reactive epoxide glycidamide (GA) via the CYP 450 isoenzyme CYP 2E1. Both, AA and GA are conjugated with glutathione. After enzymatic processing, the mercapturic acids N-Acetyl-S-(2-carbamoylethyl)-L-cysteine (AAMA) as well as the regioisomers N-Acetyl-S-(2-carbamoyl-2-hydroxyethyl)-L-cysteine (GAMA) and N-Acetyl-S-(1-carbamoyl-2-hydroxy-ethyl)-L-cysteine (iso-GAMA) are excreted with urine. An additional pathway for the metabolic conversion of GA is the epoxide hydrolase mediated hydrolysis to the diol compound glyceramide. Following administration of AA at doses exceeding the daily dietary intake by a factor of 1000 - 6000 to human subjects, a new urinary metabolite was found, which could be identified as the S-oxide of AAMA (AAMA-sulfoxide). In general, data from animal studies are used for risk assessment of (potential) human carcinogens. However, inter-species differences in toxicodynamics or toxicokinetics, e.g. in biotransformation may lead to under- or overestimation of human risk. The objective of this work was to establish a highly specific and sensitive analytical method to quantify the major urinary metabolites of AA. Other aims apart from measurements concerning the human background exposure were the evaluation of biotransformation and toxicokinetics of AA in humans and rats after oral administration of 13C3-AA. The obtained data was intended to help avoid linear extrapolation from animal models for future risk assessments of AA carcinogenicity.
Untersuchung des dielektrischen Verhaltens polymerbasierter elektrorheologischer Flüssigkeiten

Auf dem Forschungsgebiet der elektrorheologischen Fluide wurden verstärkt Modelle auf der Basis statischer Systeme entwickelt. In diesen Modellen wird angenommen, dass die Par-tikel der ER-Suspension Ketten von einer Elektrode zur anderen ausbilden. Über die elektro-statische Wechselwirkung der Partikel untereinander in Verbindung mit dem nicht-ohmschen Verhalten des Trägeröls wurde dabei auf die Schubspannung und die Stromdich-te der ERF geschlossen. Diese Vorhersagen waren aufgrund der Vernachlässigung der Dy-namik nur bedingt aussagefähig. In experimentellen Untersuchungen der Schubspannung und Stromdichte wurden die Abhängigkeiten von Scherrate, Feldstärke und Spaltgeometrie näher betrachtet. Für ein besseres Verständnis der ER-Eigenschaften wurden zudem die-lektrische Messungen (Impedanzmessungen) durchgeführt. Als Ergebnis dieser Messungen wurde eine dielektrische Aktivität der ERF im Frequenzbereich von 102 Hz bis 105 Hz für einen hohen ER-Effekt ermittelt. Der Realteil der Permittivität führt in diesem Frequenz-fenster einen großen Sprung durch – dies ist äquivalent mit einem großen Imaginärteil der Permittivität (dielektrischer Verlust ) oder einem großen tan . In dieser Arbeit wurde für die Untersuchungen eine ERF mit Silikonöl als Trägermedium und salzdotiertes Polyurethan als Partikelmaterial verwendet. Im ersten Teil der Arbeit steht die Identifikation der auftretenden Relaxationen – ermittelt durch die dielektrische Spektro-skopie – im Vordergrund. Dabei konnte eine Relaxation aufgrund der Salzdotierung, eine durch Kohlendioxid und Wasser und eine aufgrund des Polyurethans der Partikel nachge-wiesen werden. Da die Dotiersalzrelaxation den größten Beitrag des ER-Effektes verursacht, wurde diese im Rahmen der vorliegenden Arbeit näher betrachtet. Sowohl Lage als auch Stärke der Relaxa-tion lassen sich durch die Partikelkonzentration, den Salzgehalt, die Salzart und durch eine Modifikation der Polymermatrix variieren. In Übereinstimmung mit Messungen am Rheo-meter lassen sich daraus die gewünschten Eigenschaften, im Besonderen das Temperatur-verhalten und die Stärke der ERF, einstellen. Im Weiteren wurde aus den gewonnenen Ergebnissen der dielektrischen Spektroskopie in Verbindung mit rheologischen Messungen ein Schema entwickelt, mit dem es möglich ist, aus der Lage und der Stärke der Salzrelaxation im Vergleich mit bekannten ERF auf die Schubspannung und die Stromdichte zu schließen. Somit ist zum ersten Mal eine Qualitäts-kontrolle aufgrund der Basiseigenschaften der ERF möglich. Im letzten Teil dieser Arbeit wurden die Unterschiede der Messungen in Scher- bzw. Fließ-modus und deren Ursachen beleuchtet. Hierbei konnte aufgezeigt werden, dass die Rotation der Partikel aufgrund der Scherbelastung in Kombination mit dem Strömungsprofil für die unterschiedlichen Messergebnisse verantwortlich ist. Die Unterschiede sind so groß, dass sich kein konstanter Faktor ermitteln lässt, um beide Messmodi miteinander zu vergleichen. Somit muss eine ERF immer in dem Modus charakterisiert werden, der der späteren Belas-tungsart entspricht, um so die korrekten Wert für die Schubspannung und die Stromdichte ermitteln zu können.
Mechanisms and adaptive significance of interspecific associations between tropical ant species

Aggression between ants from different colonies or species is ubiquitous. Exceptions to this rule exist in the form of supercolonies (within a species) and interspecific associations (between species). Probably the most intimate interspecific association is the parabiosis, where two ant species live together in a common nest. They keep their brood separate but jointly use trails and often share food resources. Parabioses are restricted to few species pairings and occur in South American and Southeast Asian rainforests. While the South American parabioses have been studied, albeit poorly, almost nothing is known about their Southeast Asian counterparts. My PhD project focuses on Southeast Asian parabioses between the myrmicine Crematogaster modiglianii Emery 1900 and the considerably larger formicine Camponotus rufifemur Emery 1900. The two species frequently nest together in hollow trees in the tropical lowland rainforest of Borneo. The basic question of my PhD project is why these two species live together. I investigated both proximate and ultimate aspects of this question. For comparative purposes, I included studies on a trail-sharing association in the same habitat. On the proximate level, I investigated which mechanisms facilitate tolerance towards hetero-spe¬ci¬fic nestmates. Ants generally discriminate nestmates from non-nestmates via cuticular hydro¬carbons that function as colony recognition cues. I studied the specificity of nestmate recognition within and between the two parabiotic species. Using gas chromatography-mass spectrometry (GC-MS), I analyzed the cuticular substances in both ant species to find potential differences to non-parabiotic species, and to estimate the substance overlap among the two species. A high substance overlap would e.g. suggest that interspecific tolerance is caused by chemical mimicry. Finally, bioassays were conducted to evaluate the function of different cuticular compounds. Interspecific tolerance in the two parabiotic species was species-specific but not colony-specific. Ca. rufifemur tolerated all Cr. modiglianii individuals, even those from foreign colonies, but strongly attacked workers of other Crematogaster species. Cr. modiglianii, in turn, tolerated Ca. rufifemur workers of certain foreign colonies but attacked those of others. Chemical analyses revealed two sympatric, chemically distinct Ca. rufifemur varieties (‘red’ and ‘black’) with almost no hydrocarbon overlap. Cr. modiglianii only tolerated foreign Ca. rufifemur workers if they belonged to the same chemical variety as their own Ca. rufifemur partner. It also attacked other, non-parabiotic Camponotus species. Thus, reciprocal interspecific tolerance was restricted to the species Cr. modiglianii and Ca. rufifemur. Ca. rufifemur frequently tolerated conspecific non-nestmates of the same chemical variety. Minor workers were more often tolerated than majors, possibly because they possess two to three times lower hydrocarbon quantities per body surface than majors. In contrast, Cr. modiglianii nearly always attacked conspecific non-nestmates. Both species possessed hydrocarbons with considerably higher chain lengths than congeneric, non-parabiotic ant species. Long-chain hydrocarbons are less volatile than shorter ones and thus harder to perceive. They may thus considerably facilitate interspecific tolerance. Moreover, up to 98% of the cuticular hydrocarbons in Ca. rufifemur were methylbranched alkenes, which are highly unusual among insect cuticular hydrocarbons. Cr. modiglianii and Ca. rufifemur had almost no hydrocarbons in common, refuting chemical mimicry as a possible cause of interspecific tolerance. The only hydrocarbons common to both species were two methylbranched alkenes, which constituted 89% of the ‘red’ Ca. rufifemur hydrocarbon profile and also occurred in those Cr. modiglianii colonies that lived together with this Ca. rufifemur variety. Cr. modiglianii presumably acquired these two compounds from its red Ca. rufifemur partner. Cr. modiglianii was significantly less aggressive towards foreign Cr. modiglianii workers that were associated with the same Ca. rufifemur variety than to those associated with the respective other one. Hence, this species seemed to use recognition cues acquired from its parabiotic partner. Apart from hydrocarbons, both species possessed a set of hitherto unknown substances on their cuticle. The quantitative composition of the unknown compounds varied between parabiotic nests but was similar among the two species of a nest. They are probably produced in the Dufour glanf of Cr. modiglianii and transferred to their Ca. rufifemur partner. Possible transfer mechanisms include interspecific trophallaxis and ‘mounting behaviour’, where Cr. modiglianii climbed onto Ca. rufifemur workers without being displaced. Although the composition of the unknown compounds greatly varied between nests, they did not function as nestmate recognition cues since both species used hydrocarbons for nestmate recognition. However, the unknown compounds significantly reduced aggression in Ca. rufifemur. The ultimate, i.e. ecological and evolutionary aspects of my PhD research deal with potential costs and benefits that Cr. modiglianii and Ca. rufifemur may derive from the parabiotic association, their interactions with other species, and population genetic analyses. Additional studies on a trail-sharing association between three other ant species deal with two possible mechanisms that may cause or facilitate trail-sharing. Whether parabioses are parasitic, commensalistic, or mutualistic, is largely unknown and depends on the costs and benefits each party derives from the association. I therefore investigated food competition (as one of the most probable costs), differentiation of foraging niches (which can reduce competition), and several potential benefits of the parabiotic way of life. Besides, I studied interactions between the ant species and the hemiepiphyte Poikilospermum cordifolium. The foraging niches of the two species differed regarding foraging range, daily activity pattern, and food preferences. None of the two species aggressively displaced its partner species from baits. Thus, interference competition for food seemed to be low or absent. For both ant species, a number of benefits from the parabiotic lifestyle seem possible. They include interspecific trail-following, joint nest defence, provision of nest space by the partner species, food exchange via trophallaxis, and mutual brood care. If an ant species follows another species’ pheromone trails, it can reach food resources found by the other species. As shown by artificial extract trails, Ca. rufifemur workers indeed followed trails of Cr. modiglianii but not vice versa. Thus, Ca. rufifemur benefited from Cr. modiglianii’s knowledge on food sources (informational parasitism). In turn, Cr. modiglianii seemed to profit from nest defence by Ca. rufifemur. Ca. rufifemur majors are substantially larger than Cr. modiglianii workers. Although Cr. modiglianii often effectively defended the nest as well, it seemed likely that this species derived a benefit from its partner’s defensive abilities. In neotropical parabioses (ant-gardens), mutualistic epiphytes play an important role in providing nest space. The neotropical Camponotus benefits its Crematogaster partner by planting epiphyte seeds, for which Crematogaster is too small. Similarly, the Bornean parabioses often were inhabited by the hemiepiphyte Poikilospermum cordifolium (Barg.-Petr.) Merr (Cecropiaceae). P. cordifolium seedlings, saplings and sometimes larger indivi¬duals abundantly grew at the entrances of parabiotic nests. However, P. cordifolium provides no additional nest space and, apart from nutritive elaiosomes, perianths, and extrafloral nectar probably plays a less important role for the ants than the neotropical epiphytes. In conclusion, the parabiosis is probably beneficial to both species. The main benefits seem to be nest defence (for Cr. modiglianii) and interspecific trail-following (for Ca. rufifemur). However, Ca. rufifemur seems to be more dependent on its partner than vice versa. For both parabiotic species, I analyzed mitochondrial DNA of ants from different regions in Borneo. My data suggest that there are four genetically and chemically distinct, but closely related varieties of Camponotus rufifemur. In contrast, Crematogaster modiglianii showed high genetic differentiation between distant populations but was not differentiated into genetic or chemical varieties. This argues against variety-specific cocladogenesis between Cr. modiglianii and Ca. rufifemur, although a less specific coevolution of the two species is highly likely. In Bornean rainforests, trail-sharing associations of Polyrhachis (Polyrhachis) ypsilon Emery 1887 and Camponotus (Colobopsis) saundersi Emery 1889 are common and often include further species such as Dolichoderus cuspidatus Smith 1857. I investigated a trail-sharing association between these three species and studied two mechanisms that may cause or facilitate these associations: interspecific trail-following, i.e. workers following another species’ pheromone trail, and differential inter¬specific aggression. In trail-following assays, D. cuspidatus regularly followed extract trails of the other two species, thus probably parasitizing on their information on food sources. In contrast, only few P. ypsilon and Ca. saundersi workers followed hetero¬speci¬fic extract trails. Hence, the association between P. ypsilon and Ca. saundersi cannot be ex¬plained by foragers following heterospecific trails. In this case, trail-sharing may originate from few scout ants that do follow heterospecific pheromone trails and then lay their own trails. Interspecific aggression among P. ypsilon, Ca. saundersi and D. cuspidatus was strongly asymmetric, Ca. saundersi being submissive to the other two species. All three species discriminated between heterospecific workers from the same and a distant trail-sharing site. Thus, it seems likely that the species of a given trail-sharing site habituate to one another. Differential tolerance by dominant ant species may be mediated by selective habituation towards submissive species, and thereby influence the assembly of trail-sharing associations.
Erste tumorassoziierte Veränderungen finden im Glykosilierungsmuster von Glykoproteinen und Glykolipiden statt. Die dabei entstehenden tumorassoziierten Carbohydrat-Antigene sind prominente Zielstrukturen der natürlichen Tumorimmunität (Immune Surveillance) und gewinnen in der Onkologie als immunogene Epitope zunehmend an Bedeutung. Der humane monoklonale IgM-Antikörper SAM-6 ist Teil der tumorspezifischen Immunität. Er wurde mit Hilfe der konventionellen Hybridomatechnologie direkt aus einem an Magenkarzinom erkrankten Menschen isoliert. Neben der Erforschung seines außergewöhnlichen Apoptose-mechanismus konnte innerhalb dieser Arbeit eine Zielstruktur des Antikörpers identifiziert und charakterisiert werden. Der humane monoklonale IgM-Antikörper SAM-6 bindet an eine neue Isoform des Hitzeschockproteins GRP78 (GRP78SAM-6). Das Antigen wurde über mehrstufige chromatographische Verfahren aus Tumorzellmembranextrakten aufgereinigt und nach tryptischen Verdau über die Methode des Peptidmassen-Fingerprinting eindeutig als humanes GRP78 identifiziert. Die auf der Zellmembran lokalisierte Variante des GRP78 besitzt ein Molekulargewicht von 82 kD und wird auf vielfältigen Tumorgeweben stabil exprimiert. GRP78SAM-6 liegt parallel zur 78 kD-Wildtyp-Variante co-exprimiert vor und konnte im Gegensatz zur Wildtyp-Variante nicht auf gesundem Gewebe nachgewiesen werden. Bei der SAM-6-spezifischen Variante des GRP78 handelt es sich um eine posttranslational modifizierte Form des GRP78, die spezifisch auf der Zellmembran lokalisiert ist, nicht jedoch intrazellulär zu finden ist. Sie unterscheidet sich durch zusätzliche Glykosilierungen vom GRP78-Wildtypen, wobei O-glykosidisch verknüpfte Glykane für die Bindung und die Reaktion mit dem SAM-6 Antikörper essentiell sind. Der SAM-6-Rezeptor stellt eine tumorspezifische Isoform des Hitzeschockproteins GRP78 dar, deren O-glykosilierte Carbohydrat-Regionen als Epitop fungieren. Durch die Bindung an GRP78SAM-6 hemmt der Antikörper SAM-6 in vitro als auch in vivo konzentrationsabhängig das Wachstum von Magen- und Pankreaskarzinomzellen und induziert eine neue Art des apoptotischen Zelltodes, die sog. Lipoptose. Es handelt sich um einen durch den Antikörper vermittelten direkten Effekt, der sich ausschließlich auf malignes Gewebe beschränkt. Schlüsselpunkt der apoptotischen Wirkung ist die Akkumulation zytotoxischer Mengen an Cholesterol und Triglyceridestern, die nach Bindung an den Antikörper in Form von Lipoprotein-Partikeln in die Tumorzelle gelangen. Der pentamere IgM-Antikörper bindet neben membranständigem GRP78SAM-6 der Tumorzelle, die ApoB 100-haltigen Lipoproteine VLDL und LDL. Insbesondere oxidativ modifizierte Formen des LDL (oxLDL) zeigten dabei die höchste Bindungsaffinität zum SAM-6 Antikörper. In deren Anwesenheit war ein maximaler lipotoxischer Effekt zu beobachten. Im Rahmen dieser Arbeit ist es gelungen, weite Teile des Lipoptose-Mechanismus aufzuklären. Eigenen Immunfluoreszenzstudien zufolge wird der SAM-6 Antikörper über rezeptorvermittelte Endozytose internalisiert. Die GRP78-vermittelte Internalisierung von oxLDL-beladenem Antikörper scheint daher plausibel und für die tödliche Anhäufung der Lipide verantwortlich zu sein. Die SAM-6-induzierte Apoptose verläuft anschließend über einen spezifischen Signalweg, der Gemeinsamkeiten mit dem intrinsischen Signalweg aufweist, jedoch wie beim extrinsischen Signalweg über externe pro-apoptotische Liganden angeregt wird. Infolge der unphysiologisch hohen intrazellulären Konzentration an oxLDL kommt es zur Induktion einer Caspasenkaskade, die nach der initialen Freisetzung von Cytochrom C aus den Mitochondrien über die Initiatorcaspasen 8 und 9 verläuft und letztendlich durch die Aktivierung der terminalen Caspasen 3 und 6 den apoptotischen Zelltod einleitet. Die Entdeckung von extrazellulär exprimiertem GRP78 auf Tumorzellen bietet die Möglichkeit neuer Therapieansätze in der Onkologie. Die SAM-6-spezifische Variante des GRP78 bietet insbesondere die Möglichkeit eines gezielten Angriffs auf die Tumorzelle, ohne gesunde Zellen zu tangieren. Sie wird auf Tumorgeweben verschiedenster Ätiologie stabil exprimiert und infolge ihres tumorspezifischen Auftretens zur optimalen Zielstruktur der natürlichen körpereigenen Immunantwort gegen Tumore. Der natürliche IgM-Antikörper ist Teil der natürlichen Immunität. Diese verfügt über ein breites Repertoire an Rezeptoren und garantiert die permanente Überwachung und Reaktion gegen modifizierte körpereigene Zellen. Sie ist dafür verantwortlich, dass Tumore sich nur in Ausnahmefällen manifestieren.
CD9 und andere Mitglieder der Tetraspaninfamilie sind an der strukturellen Organisation und der Plastizität der Plasmamembran beteiligt. Dabei inhibiert mAK K41, ein spezifischer CD9-Antikörper, die Hundestaupe-induzierte Zell-Zellfusion und die Virusfreisetzung, während die MV-induzierte Zell-Zellfusion nicht beeinflusst wird. So ist die extrazelluläre Domäne des Hämagglutinin-Proteins von CDV diejenige, die die Empfindlichkeit der Zell-Zellfusion gegenüber dem CD9-Antikörper verursacht, die aber selbst nicht an das CD9-Molekül bindet. Diese Erkenntnisse ließen vermuten, dass strukturelle Veränderungen an der Plasmamembran der Grund für die Hemmung sind bzw. räumliche Expressionsmuster des Rezeptors involviert sein könnten. In der vorliegenden Arbeit konnte gezeigt werden, dass mAK K41 das Konformationsepitop der großen extrazellulären Domäne (LEL) von CD9 erkennt, die nachweislich über ß1-Integrin mit verschiedenen Signalwegen im Innern in Kontakt steht. Die Bindung dieser Domäne induziert folglich eine schnelle Umlagerung und ein Clustern der CD9-Moleküle bis zur Bildung von netzähnlichen Strukturen an den Kontaktstellen zweier Zellen. Durch konfokale und rasterelektronenmikroskopische Untersuchungen konnten mikrovilli-ähnliche Ausstülpungen aufgedeckt werden, die von beiden Seiten aneinander liegender Zellen gebildet werden. Nach einer Zeitspanne von 2 h bis 20 h bildeten diese CD9-haltigen Ausstülpungen feine, mehrere µm lange Mikrovilli aus, die sich in einer Art Geflecht miteinander vernetzten und mikrovilli-artige Reißverschlussstrukturen bildeten. Weiterhin konnte eine starke Kolokalisierung des Ewi-F-Proteins vor und nach der Antikörperinkubation gezeigt werden, sowie eine partielle Kolokalisierung mit ß1-Integrin. Im Vergleich konnten MV-Proteine innerhalb der CD9-haltigen Netzstrukturen beobachtet werden, während CDV-Proteine komplett aus diesen ausgeschlossen wurden. Somit ist die Ausgrenzung der viralen Fusionsmaschinerie von CDV von den CD9-Clustern sowie die physikalische Trennung von den Zellkontakten wohl die Erklärung für die Inhibition der Virus-induzierten Zell-Zellfusion durch mAK K41. Da experimentell keine kausale Verbindung zwischen der Induktion der CD9-haltigen Cluster und bestimmten Signalwegen gezeigt werden und auch kein Beweis hervorgebracht werden konnte, dass die Grundlage der Netzstrukturen durch die Umlagerung des Zytoskeletts entsteht, scheint die Interaktion des Antikörper selbst die treibende Kraft für die Strukturbildung zu sein (Singethan et al., 2008). Zusammenfassend ist zu sagen, dass die Ergebnisse die Relevanz des CD9-Moleküls in gesunden und pathogenen Zell-Zellfusionsprozessen unterstreichen und eindeutig zeigen, dass CD9 die Zellfusion von CDV steuern kann, indem es den Zugang der Fusionsmaschinerie zu den Zellgrenzen reguliert. Das Nipah-Virus (NiV) ist ein hochpathogenes Paramyxovirus, das in Schweinen eine Erkrankung des Respirationstrakts und in Menschen eine schwere fiebrige Enzephalitis mit hohen Mortalitätsraten verursacht. Da es noch keine Vakzine bzw. antivirale Medikamente gegen dies Erkrankung gibt, war die Entwicklung kleiner inhibitorischer Moleküle notwendig. Für das Masern-Virus (MV) wurden bereits kleine Inhibitoren, wie Ox-1, AM-2 und AS-48 entwickelt, die in die Bindungstasche des MV F-Proteins passen und so die Membranfusion verhindern. Basierend auf struktureller Ähnlichkeiten der Paramyxovirus F-Proteine konnte ein Testsystem mit F- und H- oder G-exprimierenden Vektoren entwickelt werden, indem eine Gruppe von Chinolon-Derivaten sowie mehrere andere Substanzen auf ihre Fähigkeit untersucht wurden, die eine MV-, CDV- oder NiV-spezifische Fusion zu hemmen. Dazu wurde zuerst die Zytotoxizität aller Substanzen bewertet, um anschließend ihre Hemmungsaktivität in Zell-Zellfusion-Assays zu untersuchen. So inhibierten zwei Substanzen, QED15B - 12 und QED15A - 12, aus der Gruppe der Chinolon-Derivate die NiV-induzierte Synzytienbildung in Hüllprotein-Transfektions- und Infektions-Assays. Bei molekularen Untersuchungen der Bindungstasche des NiV F-Proteins wurde die hemmende Aktivität beider Chinolon-Derivate bestätigt. So konnte eine hervorragende Wechselwirkung und strukturelle Paßform für die Protein-Bindetasche identifiziert werden und deren Interaktion bewiesen werden. Somit konnte die Substanzklasse der Chinolone als Inhibitoren gegen die NiV-Fusion identifiziert und der Mechanismus der Interaktion mit der Bindetasche als Grund für die inhibierende Wirkung aufgeklärt werden (Niedermeier S. und Singethan K. et al., 2008 zur Veröffentlichung eingereicht). Dabei sind diese Chinolon-Derivate mit ihrer Struktur, die völlig verschieden von den meisten aktiven Molekülen gegen die Masern-induzierte Zellfusion sind, eine vielversprechende neue Verbindungsstruktur, auf der weitere Entwicklungen neuer Inhibitoren und antiviraler Agentien aufgebaut werden können.
