Malaria ist eine schwerwiegende Krankheit, die jährlich über eine Million Menschen tötet. Die zunehmende Resistenzbildung gegenüber den verwendeten Medikamenten macht die Entwicklung neuer Antimalariamittel dringend notwendig. Daher sind die Vitamin B6 Synthese und der Shikimatweg von besonderem Interesse, da diese beiden Synthesewege nur im Parasiten und nicht im Menschen vorkommen. Unter der Voraussetzung, dass diese essentiell für den Parasiten sind, böten sie ideale Ansatzpunkte zur Entwicklung neuer Antimalariamittel. Voraus gegangene Studien haben gezeigt, dass Plasmodium falciparum in der Lage ist, PLP de novo mittels eines bifunktionalen Enzymkomplex, bestehend aus den Proteinen Pdx1 und Pdx2, zu synthetisieren. Pdx1 stellt dabei die eigentliche Synthase dar, während Pdx2 als Glutaminase-Partner das benötigte Ammoniumion für den heterocyclen Ring bereitstellt. Zusätzlich dazu verfügt der Parasit auch über einen salvage pathway um PLP zu „recyclen“, in dem der Pyridoxalkinase PdxK eine Schlüsselfunktion zufällt. Knockout Studien der pdx1 im Mausmalariasystem P. berghei haben gezeigt, dass PbPdx1 für eine optimale Entwicklung der Blutstadien benötigt wird, nicht jedoch für deren Überleben. Im Rahmen dieser Arbeit habe ich die Effekte eines pbpdxK(-) Knockouts in demselben System untersucht. Es konnte eine monoklonale Knockoutlinie generiert werden, was zeigte, dass PbPdxK nicht essentiell für das Überleben des Parasiten in den Blutstadien ist. Die Entwicklung während des Blutstadiums war von dem pbpdxK(-) Knockout nicht betroffen. Allerdings zeigte sich im Moskitostadium eine drastische Reduktion der Sporozoitenzahl sowohl in den Mitteldärmen als auch in den Speicheldrüsen. Dieses Ergebnis legt nahe, dass PbPdxK essentiell für das Überleben der Sporozoiten ist. Daneben wurde versucht, die Gene pfpdx1, pfpdx2 sowie pfpdxK in P. falciparum 3D7 durch Verwendung der single cross over Strategie auszuschalten. Es konnte jedoch für keines der genannten Konstrukte eine Integration in die jeweiligen Genloci anhand von PCR-Analysen nachgewiesen werden. Ebenso scheiterte der Versuch, durch Rekombination eines komplementären Genabschnitts die Funktion des Gens zu rekonstituieren. Daher bleibt es unklar, ob pfpdx1, pfpdx2 und pfpdxK durch Knockout Strategien auszuschalten sind oder nur für Genmanipulationen nicht zugänglich sind. Die Kultivierung von P. falciparum 3D7 Parasiten in Vitamin B6 depletiertem Medium hatte keinen Effekt auf deren Wachstum. Eine anschließende Analyse der Proteinextrakte zeigte eine erhöhte Expression der PfPdxK, während sich das Expressionslevel der PfPdx1 nicht veränderte. Es scheint, dass der Parasit in der Lage ist Vitamin B6 Mangel durch vermehrte Nutzung des salvage pathways vollständig zu kompensieren. Frühere Arbeiten zeigten, dass der C-Terminus der Pdx1 in die Aktivität des PLP Synthasekomplexes involviert ist. Aus diesem Grund wurden verschiedene C-terminale Deletionsmutanten der PfPdx1 konstruiert und dabei bis zu 30 Aminosäuren entfernt. Diese Analysen ergaben, dass der C-Terminus vier verschiedene Funktionen besitzt: das Assembly der Pdx1 Untereinheiten zum Dodekamer, die Bindung des Pentosesubstrats Ribose 5-Phosphat, die Bildung des Intermediats I320 und schließlich die PLP Synthese. Diese unterschiedlichen Funktionen wurden durch verschiedene Deletionsvarianten identifiziert. Darüber hinaus waren alle Deletionsvarianten in der Lage, die Glutaminase Pdx2 zu aktivieren, was zeigt, dass das Dodekamer nicht Vorraussetzung für die Glutaminaseaktivität ist. Aufgrund der geringen PLP Syntheseaktivität in vitro wurde vermutet, dass der PfPdx1/PfPdx2 Komplex durch einen zusätzlichen Faktor aktiviert wird. Daher wurde versucht, mittels Yeast 2-Hybrid, basierend auf einer PCR-amplifizierten P. falciparum 3D7 cDNA-Bibliothek als bait und PfPdx1 als prey, einen Interaktionspartner zu identifizieren. Mehrere Klone wurden gewonnen, die alle einen Bereich des Mal13P1.540, einem putativen Hsp70 Proteins, enthielten. Jedoch scheiterten alle Versuche, die Protein-Protein-Interaktion mit rekombinant exprimierten Protein zu bestätigen. Ebenso war es nicht möglich, das vollständige Mal13P1.540 rekombinant zu exprimieren sowie dessen Lokalisation in vivo zu bestimmen. Daher bleibt die Interaktion von PfPdx1 und Mal13P1.540 ungeklärt. Neben der Vitamin B6 Biosynthese konnten auch einige Gene des Shikimatweges in Plasmodium identifiziert werden. In P. berghei konnten der C-terminale Teil der 3-Dehydroquinatsynthase (2) sowie die Shikimatkinase (5) und die 5-Enoylpyruvylshikimat 3-Phosphatsynthase (6) in einem open reading frame (ORF) identifiziert werden, der dieselbe genetische Organisation aufweisen wie der Arom-Komplex der Hefen. Mit Hilfe eines Komplementationsassay wurde die Funktionalität dieses ORFs überprüft. Dazu wurden S. cerevisiae BY4741Δaro1, ein Hefestamm ohne funktionalen Arom-Komplex, mit dem Pb2_6_5_ABC Fragment transformiert. Die so transformierten Hefen waren nicht in der Lage, auf Mangelplatten ohne aromatische Aminosäuren zu wachsen, was zeigte, dass das Pb2_6_5_ABC Konstrukt den BY4741Δaro1 Phänotyp nicht komplementieren konnte. Der Versuch, mit Hilfe des Baculovirussytems rekombiant exprimiertes Protein zu erhalten, verlief erfolglos. Ebenso war es nicht möglich, Teile des Proteins für Immunisierungen zu exprimieren. Daher bleibt die Funktionalität des Pb2_6_5_ABC Konstruktes ungeklärt.
In this thesis we have used Drosophila melanogaster as a model organism to investigate proteins and their putative interacting partners that are directly or indirectly involved in the release of neurotransmitters at the synapse. We have used molecular techniques to investigate conserved synaptic proteins, synapsin and synapse associated protein of 47 kD (SAP47), and a putative interaction partner of SAP47, tubulin binding chaperone E-like (TBCEL). SAP47 and synapsins are highly conserved synaptic vesicle associated proteins in Drosophila melanogaster. To further investigate the role and function of Sap47 and Syn genes, we had earlier generated the null mutants by P-element mutagenesis (Funk et al., 2004; Godenschwege et al., 2004). Western blots and ELISA of brain homogenates from Sap47156 null mutants showed the presence of up-regulated phospho-synapsin in comparison to wild-type (CS) and the presence of up-regulated phospho-synapsin was partially abolished when a pan-neuronal rescue of SAP47 was performed by the Gal4- UAS technique. Thus, the results suggest a qualitative and quantitative modulation of synapsin by SAP47. At the transcript level, we did not observe any difference in content of Syn transcript in Sap47156 and wild-type CS flies. The question of a direct molecular interaction between SAP47 and synapsin was investigated by co-immunoprecipitation (Co-IP) experiments and we did not find any stable interactions under the several IP conditions we tested. The possibility of Sap47 as a modifier of Syn at the genetic level was investigated by generating and testing homozygous double null mutants of Sap47 and Syn. The Syn97, Sap47156 double mutants are viable but have a reduced life span and decreased locomotion when compared to CS. In 2D-PAGE analysis of synapsins we identified trains of spots corresponding to synapsins, suggesting that synapsin has several isoforms and each one of them is posttranslationally modified. In an analysis by Blue native-SDS-PAGE (BN-SDS-2D- PAGE) and Western blot we observed synapsin and SAP47 signals to be present at 700-900 kDa and 200-250 kDa, respectively, suggesting that they are part of large but different complexes. We also report the possibility of Drosophila synapsin forming homo- and heteromultimers, which has also been reported for synapsins of vertebrates. In parallel to the above experiments, phosphorylation of synapsins in Drosophila was studied by IP techniques followed by 1D-SDS gel electrophoresis and mass spectrometry (in collaboration with S. Heo and G. Lubec). We identified and verified 5 unique phosphorylation sites in Drosophila synapsin from our MS analysis. Apart from phosphorylation modifications we identified several other PTMs which have not been verified. The significance of these phosphorylations and other identified PTMs needs to be investigated further and their implications for synapsin function and Drosophila behavior has to be elucidated by further experiments. In a collaborative project with S. Kneitz and N. Nuwal, we investigated the effects of Sap156 and Syn97 mutations by performing a whole Drosophila transcriptome microarray analysis of the individual null mutants and the double mutants (V2 and V3). We obtained several candidates which were significantly altered in the mutants. These genes need to be investigated further to elucidate their interactions with Sap47 and Syn. In another project, we investigated the role and function of Drosophila tubulin- binding chaperone E-like (Tbcel, CG12214). The TBCEL protein has high homology to vertebrate TBCE-like (or E-like) which has high sequence similarity to tubulin-binding chaperone E (TBCE) (hence the name TBCE-Like). We generated an anti-TBCEL polyclonal antiserum (in collaboration with G. Krohne). According to flybase, the Tbcel gene has only one exon and codes for two different transcripts by alternative transcription start sites. The longer transcript RB is present only in males whereas the shorter transcript RA is present only in females. In order to study the gene function we performed P- element jump-out mutagenesis to generate deletion mutants. We used the NP4786 (NP) stock which has a P(GawB) insertion in the 5’ UTR of the Tbcel gene. NP4786 flies are homozygous lethal due to a second-site lethality as the flies are viable over a deficiency (Df) chromosome (a deletion of genomic region spanning the Tbcel gene and other upstream and downstream genes). We performed the P-element mutagenesis twice. In the first trial we obtained only revertants and the second experiment is still in progress. In the second attempt, jump-out was performed over the deficiency chromosome to prevent homologous chromosome mediated double stranded DNA repair. During the second mutagenesis an insertion stock G18151 became available. These flies had a P-element insertion in the open reading frame (ORF) of the Tbcel gene but was homozygous viable. Western blots of fresh tissue homogenates of NP/Df and G18151 flies probed with anti-TBCEL antiserum showed no TBCEL signal, suggesting that these flies are Tbcel null mutants. We used these flies for further immunohistochemical analyses and found that TBCEL is specifically expressed in the cytoplasm of cyst cells of the testes and is associated with the tubulin of spermatid tails in wild-type CS, whereas in NP/Df and G18151 flies the TBCEL staining in the cyst cells was absent and there was a disruption of actin investment cones. We also found enrichment of TBCEL staining around the actin investment cone. These results are also supported by the observation that the enhancer trap expression of the NP4786 line is localised to the cyst cells, similar to TBCEL expression. Also, male fertility of NP/Df and G18151 flies was tested and they were found to be sterile with few escapers. Thus, these results suggest that TBCEL is involved in Drosophila spermatogenesis with a possible role in the spermatid elongation and individualisation process.
PART I Animals need to constantly evaluate their external environment in order to survive. In some cases the internal state of the animal changes to cope with it’s surrounding. In our study we wanted to investigate the role of amines in modulating internal states of Drosophila. We have designed a behavioral paradigm where the flies are fixed in space but can walk on a small styrofoam ball suspended by a gentle stream of air. The walking activity of flies was used as behavioral readout. PART I Animals need to constantly evaluate their external environment in order to survive. In some cases the internal state of the animal changes to cope with it’s surrounding. In our study we wanted to investigate the role of amines in modulating internal states of Drosophila. We have designed a behavioral paradigm where the flies are fixed in space but can walk on a small styrofoam ball suspended by a gentle stream of air. The walking activity of flies was used as behavioral readout. An operant training paradigm was established by coupling one of the walking directions to incidence of heat punishment. We observed that animals quickly realized the contingency of punishment with walking direction and avoided walking in the punished direction in the presence of punishment, but did not continue walking in the unpunished direction in the absence of the punishment. This would indicate that the flies do not form a memory for the punished direction or rapidly erase it under new conditions. On having established the paradigm with heat punishment we have attempted to activate selected subsets of neuronal populations of Drosophila while they were walking on the ball. The selective activation of neurons was achieved by expressing the light-activated ion channel channelrhodopsin-2 (ChR2) using the Gal4-UAS system and coupling the unidirectional walking of the animals on the ball with the incidence of blue light required to activate the channels and depolarize the neurons. The feasibility of this approach was tested by light-activating sugar sensitive gustatory receptor neurons expressing ChR2, we found that when the light was actuated the flies preferred to turn in one direction the optically “rewarded” direction. Next we similarly activated different subsets of aminergic neurons. We observed that in our setup animals avoided to turn in the direction which was coupled to activation of dopaminergic neurons indicating that release of dopamine is disliked by the animals. This is in accordance with associative learning experiments where dopamine is believed to underlie the formation of an association between a neutral conditioned stimulus with the aversive unconditioned stimulus. However, when we activated tyraminergic/octopaminergic neurons we did not observe any directional preference. The activation of dopaminergic and tyraminergic/octopaminergic neurons led to arousal of the animals indicating that we were indeed successful in activating those neurons. Also, the activation of serotonergic neurons did not have any effect on directional preference of the animals. With this newly established paradigm it will be interesting to find out if in insects like in mammals a reward mediating system exists and to test subsets of aminergic or peptidergic neurons that could possibly be involved in a reward signaling system which has not been detected in our study. Also, it would be interesting to localize neuropile regions that would be involved in mediating choice behavior in our paradigm. PART II In collaboration with S. Kneitz (IZKF Wuerzburg) and T. Nuwal we performed genome-wide expression analysis of two pre-synaptic mutants - Synapsin (Syn97) and Synapse associated protein of 47 kDa (Sap47156). The rationale behind these experiments was to identify genes that were up- or down-regulated due to these mutations. The microarray experiments provided us with several candidate genes some of which we have verified by qPCR. From our qPCR analysis we can conclude that out of the verified genes only Cirl transcripts seem to be reproducibly down regulated in Synapsin mutants. The Cirl gene codes for a calcium independent receptor for latrotoxin. Further qPCR experiments need to be performed to verify other candidate genes. The molecular interactions between CIRL and SYN or their genes should now be investigated in detail.
Fish of the genus Xiphophorus belong to the oldest animal models in cancer research. The oncogene responsible for the generation of spontaneous aggressive melanoma encodes for a mutated epidermal growth factor receptor (Egfr) and is called xmrk for Xiphophorus melanoma receptor kinase. Xmrk constitutive activation mechanisms and subsequent signaling pathways have already been investigated and charaterized but it is still unknown if Egfr ligands may also play a role in Xmrk-driven melanoma formation. To investigate the potential role of Egfr ligands in Xmrk-driven melanoma, I firstly analyzed the evolution of teleost and tetrapod Egfr/Egfr ligand systems. I especially focused on the analysis on the medaka fish, a closely related species to Xiphophorus, for which the whole genome has been sequenced. I could identify all seven Egfr ligands in medaka and could show that the two teleost-specific Egfr copies of medaka display dissimilar expression patterns in adult tissues together with differential expression of Egfr ligand subsets, arguing for subfunctionalization of receptor functions in this fish. Our phylogenetic and synteny analyses supported the hypothesis that only one gene in the chordate ancestor gave rise to the diversity of Egfr ligands found in vertebrate genomes today. I also could show that the Egfr extracellular subdomains implicated in ligand binding are not evolutionary conserved between tetrapods and teleosts, making the use of heterologous ligands in experiments with fish cells debatable. Despite its well understood and straight-forward process, Xmrk-driven melanomagenesis in Xiphophorus is problematic to further investigate in vivo. Our laboratory recently established a new melanoma animal model by generating transgenic mitf::xmrk medaka fishes, a Xiphophorus closely related species offering many more advantages. These fishes express xmrk under the control of the pigment-cell specific Mitf promoter. During my PhD thesis, I participated in the molecular analysis of the stably transgenic medaka and could show that the Xmrk-induced signaling pathways are similar when comparing Xiphophorus with transgenic mitf::xmrk medaka. These data together with additional RNA expression, protein, and histology analyses showed that Xmrk expression under the control of a pigment cell-specific promoter is sufficient to induce melanoma in the transgenic medaka, which develop very stereotyped tumors, including uveal and extracutaneous melanoma, with early onset during larval stages. To further investigate the potential role of Egfr ligands in Xmrk-driven melanoma, I made use of two model systems. One of them was the above mentioned mitf::xmrk medaka, the other was an in-vitro cell culture system, where the EGF-inducible Xmrk chimera HERmrk is stably expressed in murine melanocytes. Here I could show that HERmrk activation strongly induced expression of amphiregulin (Areg) and heparin-binding EGF-like growth factor (Hbegf) in melanocytes. This regulation was dependent on the MAPK and SRC signaling pathways. Moreover, upregulation of Adam10 and Adam17, the two major sheddases of Egfr ligands, was observed. I also could demonstrate the functionality of the growth factors by invitro analyses. Using the mitf::xmrk medaka model I could also show the upregulation of a subset of ligand genes, namely egf, areg, betacellulin (btc) and epigen (epgn) as well as upregulation of medaka egfrb in tumors from fish with metastatic melanoma. All these results converge to support an Xmrk-induced autocrine Egfr ligand loop. Interestingly, my in-vitro experiments with conditioned supernatant from medaka Egf- and Hbegf-producing cells revealed that not only Xiphophorus Egfrb, but also the pre-activated Xmrk could be further stimulated by the ligands. Altogether, I could show with in-vitro and in-vivo experiments that Xmrk is capable of inducing a functional autocrine Egfr ligand loop. These data confirm the importance of autocrine loops in receptor tyrosine kinase (RTK)-dependent cancer development and show the possibility for a constitutively active RTK to strengthen its oncogenic signaling by ligand binding.
In our analysis I was interested in the gene duplications, with focus on in-paralogs. In-paralogs are gene duplicates which arose after species split. Here I analysed the in-paralogs quantitatively, as well as qualitatively. For quantitative analysis genomes of 21 species were taken. Most of them have vastly different lifestyles with maximum evolutionary distance between them 1100 million years. Species included mammals, fish, insects and worm, plus some other chordates. All the species were pairwised analysed by the Inparanoid software, and in-paralogs matrix were built representing number of in-paralogs in all vs. all manner. Based on the in-paralogs matrix I tried to reconstruct the evolutionary tree using in-paralog numbers as evolutionary distance. If all 21 species were used the resulting tree was very far from real one: a lot of species were misplaced. However if the number was reduced to 12, all of the species were placed correctly with only difference being wrong insect and fish clusters switched. Then to in-paralogs matrix the neighbour-net algorithm was applied. The resulting "net" tree showed the species with fast or slow duplications rates compared to the others. We could identify species with very high or very low duplications frequencies and it correlates with known occurrences of the whole genome duplications. As the next step I built the graphs for every single species showing the correlation between their in-paralogs number and evolutionary distance. As we have 21 species, graph for every species is built using 20 points. Coordinates of the points are set using the evolutionary distance to that particular species and in-paralogs number. In mammals with increasing the distance from speciation the in-paralogs number also increased, however not in linear fashion. In fish and insects the graph close to zero is just the same in mammals' case. However, after reaching the evolutionary distances more than 800 million years the number of inparalogs is beginning to decrease. We also made a simulation of gene duplications for all 21 species and all the splits according to the fossil and molecular clock data from literature. In our simulation duplication frequency was minimal closer to the past and maximum in the near-present time. Resulting curves had the same shape the experimental data ones. In case of fish and insect for simulation the duplication rate coefficient even had to be set negative in order to repeat experimental curve shape. To the duplication rate coefficient in our simulation contribute 2 criteria: gene duplications and gene losses. As gene duplication is stochastical process it should always be a constant. So the changing in the coefficient should be solely explained by the increasing gene loss of old genes. The processes are explained by the evolution model with high gene duplication and loss ratio. The drop in number of in-paralogs is probably due to the BLAST algorithm. It is observed in comparing highly divergent species and BLAST cannot find the orthologs so precisely anymore. In the second part of my work I concentrated more on the specific function of inparalogs. Because such analysis is time-consuming it could be done on the limited number species. Here I used three insects: Drosophila melanogaster (fruit y), Anopheles gambiae (mosquito) and Apis mellifera (honeybee). After Inparnoid analyses and I listed the cluster of orthologs. Functional analyses of all listed genes were done using GO annotations and also KEGG PATHWAY database. We found, that the gene duplication pattern is unique for each species and that this uniqueness is rejected through the differences in functional classes of duplicated genes. The preferences for some classes reject the evolutionary trends of the last 350 million years and allow assumptions on the role of those genes duplications in the lifestyle of species. Furthermore, the observed gene duplications allowed me to find connections between genomic changes and their phenotypic manifestations. For example I found duplications within carbohydrate metabolism rejecting feed pattern adaptation, within photo- and olfactory-receptors indicating sensing adaptation and within troponin indicating adaptations in the development. Despite these species specific differences, found high correlations between the independently duplicated genes between the species. This might hint for a "pool" of genes preferentially duplicated. Taken together, the observed duplication patterns reject the adaptational process and provide us another link to the field of genomic zoology.
In die Kernmembran von Eukaryoten sind Kernporenkomplexe eingelagert. Diese stellen die einzige Verbindung zwischen dem Nukleo- und Zytoplasma dar und vermitteln den gerichteten Transport von Proteinen und Ribonukleoproteinpartikeln über die Kernhülle. Durch vorangehende Versuche unserer Arbeitsgruppe konnte gezeigt werden, dass es experimentell möglich ist, die Bildung einer kontinuierlichen Doppelmembran von der Insertion der Kernporenkomplexe zu trennen (Ewald et al., 1997). Dabei spielen verschiedene im Extrakt enthaltene Membranfraktionen eine Rolle. Erst kürzlich wurden in unserer Arbeitsgruppe zwei unterschiedliche Membranfraktionen aus Xenopus Extrakt isoliert, die aufgrund ihrer Dichte als 40% und 30% Membranfraktion benannt wurden. Massenspektrometrische Untersuchungen zeigten, dass sich in der 30% Membranfraktion, welche für die Kernporenkomplexbildung verantwortlich zu sein scheint, das Major Vault Protein (MVP) befindet. MVP ist Hauptbestandteil der Vault-Komplexe, großer tonnenförmiger Ribonukleoproteinpartikel, denen bislang eine Vielzahl von zellulären Funktionen zugeordnet wurden, die meisten davon jedoch noch stark debattiert. Vaults könnten womöglich eine Rolle als Transporter über die Kernporenkomplexe spielen und wurden schon mehrfach mit dem Aufbau einer multiplen Arzneimittelresistenz in Verbindung gebracht. Die Beteiligung von MVP bei der Bildung der Kernporenkomplexe ist eine neue zelluläre Funktion und sollte deshalb in dieser Arbeit näher untersucht werden. In dieser Arbeit wurden zunächst die 40% und 30% Membranfraktionen auf ihr unterschiedliches Verhalten bei der Bildung der Kernhülle separat und in Kombination genauer untersucht. Dabei zeigte sich, dass die 40% Membranfraktion an Chromatin bindet und eine kontinuierliche Doppelmembran aufbaut. Die 30% Membranfraktion konnte alleine nicht an Chromatin binden, induzierte aber in der durch die 40% Membranfraktion gebildeten Doppelmembran den Aufbau von Kernporenkomplexen. Durch Immunfluoreszenzaufnahmen und ultrastrukturelle Untersuchungen wurde belegt, dass das an der 30% Membranfraktion assoziierte MVP für die Bildung von Kernporenkomplexen verantwortlich war. Ferner konnten wir zeigen, dass sowohl MVP als auch Vault-Partikel die de novo Insertion von Kernporenkomplexen in kontinuierliche Doppelmembranen induzieren konnten. Die molekularen Mechanismen der Kernporenkomplexbildung durch MVP wurden mit Hilfe von artifiziellen Lipidmembranen analysiert. Anhand von unilamellaren Liposomen und elektronenmikroskopischen Aufnahmen konnte gezeigt werden, dass MVP die Lipidstruktur beeinflussen und perforieren kann. Zudem löste MVP die Bildung von Poren in schwarzen Lipidmembranen aus und führte zur Messung von Strömen durch Einzelkanalmessungen über die entstandenen Poren. Um die bei dem Prozess der Kernporenkomplexbildung beteiligten Bindungspartner von MVP zu identifizieren, wurden mehrere Protein-Protein-Bindungsstudien durchgeführt. Unter den ermittelten MVP-Bindungspartnern ließen sich keine Nukleoporine mit dem Sequenzmotiv FXFG identifizieren, es ist jedoch nicht auszuschließen, dass MVP bei der Bildung der Kernporenkomplexe mit anderen Nukleoporinen interagiert. Da eine frühere Arbeit die Bedeutung von Mikrotubuli bei der Bildung der Kernporenkomplexe aufzeigte (Ewald et al., 2001), wurden in dieser Arbeit die Interaktionen der isolierten 40% und 30% Membranfraktionen und von MVP mit dem Mikrotubulinetzwerk näher analysiert. Dabei zeigte sich, dass nur die 30% Membranfraktion mit Mikrotubuli interagierte und eine Inhibition der Mikrotubulipolymerisation durch Colchizin den Einbau von Kernporenkomplexen verhinderte. Im Gegensatz dazu interagierten die 40% Membranvesikel nicht mit Mikrotubuli und daher hat eine Colchizin-induzierte Inhibition der Mikrotubulipolymerisation keinen Effekt auf den Aufbau einer kontinuierlichen Doppelmembran. Durch immunfluoreszenzmikroskopische Untersuchungen konnte zudem gezeigt werden, dass die Lokalisation von MVP an der Kernhülle ebenfalls Abhängig von Mikrotubuli ist. Um zu demonstrieren, dass die MVP-induzierte Kernporenkomplexbildung im zellfreien System abhängig vom Transport von MVP zur Kernhülle ist, wurde die Zugabe von MVP zu porenlosen Kernen nach einer Colchizin-Behandlung analysiert. Hierbei konnte belegt werden, dass MVP Mikrotubuli auch benötigt, um die Bildung von Kernporenkomplexen in der Kernmembran zu initiieren. Da Mikrotubulifilamente im zellfreien System mit ihren Plus-Enden gegen die Chromatinoberfläche gerichtet sind, sollten für den gerichteten Transport zum Chromatin Motorproteine der Kinesin-Familie eine Rolle spielen. Durch die Inhibition von Mklp2, einem mitotischen Kinesin, konnte der Aufbau der Kernporenkomplexe durch MVP in porenlosen Kernen blockiert werden.
Die Rolle der Hirnrinde als Zentrum komplexer Funktionen wie Lernen und Ge-dächtnis wird nicht zuletzt durch deren komplexe, in Schichten organisierte Architek-tur ermöglicht. Von entscheidender Bedeutung ist die präzise Positionierung von Nervenzellen, die im Laufe der Embryonalentwicklung in der Ventrikularzone (VZ) geboren werden und anschließend in radialer Richtung zu ihrem Bestimmungsort wandern. Die Funktion des Neurotrophin-Rezeptors TrkB an der Entwicklung des zerebralen Kortex war Gegenstand dieser Arbeit. Am Tag 12,5 der Embryonalentwicklung konnte die Expression von TrkB so-wohl in den Zellen der VZ als auch in neu geborenen Neuronen der Präplatte nach-gewiesen werden. Die Phosphorylierung des Rezeptors erfolgte dabei unabhängig von den beiden Liganden BDNF und NT-3. Ebenso führten BDNF oder NT-3 zu keiner zellulären Antwort in isolierten kortikalen Vorläuferzellen, wohingegen die Stimulation mit EGF eine Phosphorylierung von TrkB an der PLCγ- und der Shc-Bindungsstelle hervorrief. Durch pharmakologische Inhibition und die Überexpression dominant negativer Src-Mutanten konnte die Beteiligung des EGF-Rezeptors und zweier neuronal exprimierter Src-Kinasen, cSrc und Fyn, an dieser Transaktivierung von TrkB durch EGF gezeigt werden. Durch die Zugabe von EGF kam es im Zuge der Aktivierung von TrkB auch zur Umverteilung des Rezeptors von intrazellulären Kompartimenten zur Zellmem-bran. Die Retention des Rezeptors im Zytoplasma wurde über post-translationelle Modifikation reguliert. Die Verhinderung von N-Glykosylierung durch Tunicamycin-Behandlung kortikaler Vorläuferzellen führte zur Exposition von TrkB an der Zellober-fläche und konnte so Responsivität gegenüber BDNF herstellen. Die physiologische Bedeutung einer Transaktivierung von TrkB durch EGF wurde durch das Fehlen der TrkB-Aktivierung in EGFR KO-Mäusen am Embryonal-tag 12,5 gezeigt. Dies hatte eine fehlerhafte Positionierung kortikaler Nervenzellen zum Zeitpunkt E15,5 zur Folge. Anhand eines Migrationsassays konnte schließlich gezeigt werden, dass die EGF-induzierte Wanderung kortikaler Vorläuferzellen in vitro mit einer asymmetrischen Translokation von TrkB einhergeht. Über die Transaktivierung von TrkB in frühen Phasen der Kortexentwicklung spielt EGF eine wichtige Rolle bei der Induktion neuronaler Differenzierung und ist an der Regulation der Wanderung postmitotischer Neurone in der Hirnrinde beteiligt.
Macrophages are important effector cells of the innate and adaptive immune response and exert a wide variety of immunological functions which necessitates a high level of plasticity on the chromatin level. In response to pathogen-associated molecular patterns (PAMPs) or inflammatory signals macrophages undergo a process of cellular activation which is associated with morphologic, functional and biochemical changes. Toll-like receptors (TLR) are able to sense many different PAMPs. TLR4 is an important sensor for lipopolysaccharide (LPS) which elicits a major portion of the host’s inflammatory response through the activation of many different signaling pathways such as the NF-κB and the MAPK protein kinase pathways RASRAF- MEK-ERK, p38 and JNK. Polycomb group (PcG) proteins are well known chromatin modifiers which function in large complexes and are required to maintain chromatin structure in a transcriptionally repressed state. It has previously been shown that the PcG protein Bmi1 is phosphorylated by 3pK, a downstream effector kinase of the MAPK protein kinase pathways RAS-RAF-MEK-ERK, p38 and JNK. In this work I analyzed the role of Bmi1 as a downstream effector of MAPK signaling during macrophage activation. Unexpectedly a rapid up-regulation on the Bmi1 protein level was observed in bone marrow derived macrophages (BMDMs) after LPS treatment. The Bmi1 induction was associated with transient protein phosphorylation that occured downstream of MAPK signaling. LPS treatment of BMDMs in the absence of Bmi1 resulted in a pronounced increase of IL-10 secretion. This secretion of the anti-inflammatory cytokine IL-10 was associated with increased IL-10 mRNA levels. Furthermore, siRNA mediated knock down of Bmi1 in J774A.1 macrophages also resulted in elevated IL-10 mRNA levels in response to LPS. ChIP analysis revealed that Bmi1 binds to throughout the il-10 locus. Alternative activation of wild type BMDMs via concomitant TLR4 and FcγR activation which triggers high IL-10 expression is paralleled by an attenuated Bmi1 protein expression. These results identify Bmi1 as a repressor of IL-10 expression during activation of macrophages.
Um sich vor dem Austrocknen zu schützen, haben Pflanzen eine Transpirationsbarriere entwickelt, die als Membran alle primären, oberirdischen Pflanzenteile überzieht. Diese so genannte Kutikula besteht hauptsächlich aus den lipophilen Komponenten Kutin und Wachs und reduziert so effektiv den Verlust von Wasser und wasserlöslichen Nährstoffen aus dem Blattinneren. Trotzdem ist sie nicht vollständig undurchlässig, und so können Wasser und gelöste Substanzen wie organische und anorganische Nährstoffe, Pestizide oder Umweltchemikalien die Kutikula in beiden Richtungen permeieren. Dabei ist offensichtlich, dass die zu Grunde liegenden Transportmechanismen den Ernährungszustand der Pflanzen, die Effizienz von Pestiziden und die Wirkung von Umweltchemikalien beeinflussen. Ein genaues Verständnis der Transportprozesse auf denen die kutikuläre Permeation basiert, kann helfen die Wirkweise von blattapplizierten Dünge- und Pflanzenschutzmitteln zu optimieren, indem gezielt Wirk- oder Zusatzstoffe modelliert werden können, welche die Aufnahme steigern. In der vorliegenden Arbeit sollte deshalb der Einfluss physiko-chemischer Eigenschaften von hydrophilen Verbindungen auf die kutikuläre Permeation untersucht werden. Nicht zuletzt wegen ihrer strukturellen Ähnlichkeit mit den blattapplizierten Herbiziden Glufosinat und Glyphosat wurden Aminosäuren als Modellsubstenzen ausgewählt. Die verwendeten Aminosäuren sind gut wasserlöslich, wobei alle Oktanol/Wasser Verteilungskoeffizienten kleiner als 1 sind. Zusätzlich liegen alle Aminosäuren in gelöster Form als Ionen vor, was zu einer Hydratisierung der Moleküle führt. Es wird spekuliert, dass hydratisierte Moleküle keinen Zugang zur lipophilen Phase der Kutikula haben. Welche Rolle die Hydrathülle bei der Permeation tatsächlich spielt, ist allerdings noch unklar. Viele Aktivwirkstoffe liegen nur unter ganz bestimmten Bedingungen in geladener Form vor, während die Richtung der kontinuierlichen Nettoladung der Aminosäuren durch den pH Wert modifiziert wird. Damit kann der Einfluss verschiedener Ladungszustände auf die kutikuläre Permeation unter Verwendung eines einheitlichen Sets von Modellsubstanzen untersucht werden. Unter natürlichen Bedingungen sind Aminosäuren unter anderem auf Blattoberflächen zu finden, wo sie blattassoziierten Mikroorganismen eine profitable Nahrungsquelle bieten. Ob äußere Faktoren für die Deposition dieser Recourcen verantwortlich sind, oder ob der Ursprung innerhalb des Blattgewebes liegt, wird kontrovers diskutiert. Die Sorption von Aminosäuren in isolierte Kutikularmembranen ist sehr gering, und korreliert - anders als bei lipophilen Substanzen - nicht mit dem Oktanol/Wasser Verteilungskoeffizienten. Das zeigt, dass der Verteilung von lipophilen und hydrophilen Substanzen innerhalb der Kutikula verschiedene Mechanismen zu Grunde liegen. Unter einer gegebenen Bedingung werden die kutikulären Leitwerte der Aminosäuren negativ vom Molvolumen beeinflusst. Zudem übersteigt die Länge des Permeationswegs die eigentliche Dicke der Membran um ein Vielfaches. Diese Zusammenhänge kennzeichnen eine gehinderte Diffusion innerhalb einer engporigen und weit verzweigten Umgebung. Eine Änderung des pH Wertes wirkt sich in unterschiedlicher Form auf die Leitwerte von Wasser und Aminosäuren aus. Mit steigendem pH Wert erhöht sich die Wasserpermeabilität isolierter Kutikularmembranen, was durch eine zunehmende, messbare Wassersorption in die Kutikula erklärt werden kann. Eine pH abhängige Dissoziation funktioneller Gruppen bewirkt eine Schwellung des polaren Weges, weshalb auch für die anionischen Aminosäuren bei pH 11 die höchsten Leitwerte gemessen wurden. Die zwitterionischen Aminosäuren bei pH 6 wiesen hingegen die geringsten Leitwerte auf, was im Widerspruch zu der Beobachtung steht, dass bei pH 1 die geringste Wassersorption in die Kutikula stattfindet. Eine Erklärung hierfür liefern die Hydrathüllen, die bei den zwitterionischen Aminosäuren am stärksten und bei den anionischen Species am geringsten ausgeprägt sind. Eine negative Korrelation aller gemessenen Aminosäureleitwerte mit den entsprechenden hydratisierten Molvolumen zeigt eindeutig, dass die Hydrathülle eine wichtige Größe für die Permeation durch die Kutikula darstellt. Dabei nimmt der Leitwert einer hydrophilen Substanz mit definiertem Molvolumen mit kleiner werdender Hydrathülle zu. Intakte Blätter wurden in flüssiges Wasser als Rezeptorlösung getaucht, um steady-state Bedingungen aufrecht zu erhalten. Dabei konnte gezeigt werden, dass die Permeabilitäten von intakten Kutikularmembranen, die anhand der natürlichen Aminosäurekonzentration innerhalb der Blätter bestimmt wurden, in derselben Größenordnung liegen, wie die für isolierte Membranen gemessenen. Außerdem konnte ein Vergleich der Flussraten auf der Ober- und Unterseite der Blätter zeigen, dass die stomatären Poren nicht direkt in den Leachingprozess involviert sind.
Memory is dynamic: shortly after acquisition it is susceptible to amnesic treatments, gets gradually consolidated, and becomes resistant to retrograde amnesia (McGaugh, 2000). Associative olfactory memory of the fruit fly Drosophila melanogaster also shows these features. After a single associative training where an odor is paired with electric shock (Quinn et al., 1974; Tully and Quinn, 1985), flies form an aversive odor memory that lasts for several hours, consisting of qualitatively different components. These components can be dissociated by mutations, their underlying neuronal circuitry and susceptibility to amnesic treatments (Dubnau and Tully, 1998; Isabel et al., 2004; Keene and Waddell, 2007; Masek and Heisenberg, 2008; Xia and Tully, 2007). A component that is susceptible to an amnesic treatment, i.e. anesthesia-sensitive memory (ASM), dominates early memory, but decays rapidly (Margulies et al., 2005; Quinn and Dudai, 1976). A consolidated anesthesia-resistant memory component (ARM) is built gradually within the following hours and lasts significantly longer (Margulies et al., 2005; Quinn and Dudai, 1976). I showed here that the establishment of ARM requires less intensity of shock reinforcement than ASM. ARM and ASM rely on different molecular and/or neuronal processes: ARM is selectively impaired in the radish mutant, whereas for example the amnesiac and rutabaga genes are specifically required for ASM (Dudai et al., 1988; Folkers et al., 1993; Isabel et al., 2004; Quinn and Dudai, 1976; Schwaerzel et al., 2007; Tully et al., 1994). The latter comprise the cAMP signaling pathway in the fly, with the PKA being its supposed major target (Levin et al., 1992). Here I showed that a synapsin null-mutant encoding the evolutionary conserved phosphoprotein Synapsin is selectively impaired in the labile ASM. Further experiments suggested Synapsin as a potential downstream effector of the cAMP/PKA cascade. Similar to my results, Synapsin plays a role for different learning tasks in vertebrates (Gitler et al., 2004; Silva et al., 1996). Also in Aplysia, PKA-dependent phosphorylation of Synapsin has been proposed to be involved in regulation of neurotransmitter release and short-term plasticity (Angers et al., 2002; Fiumara et al., 2004). Synapsin is associated with a reserve pool of vesicles at the presynapse and is required to maintain vesicle release specifically under sustained high frequency nerve stimulation (Akbergenova and Bykhovskaia, 2007; Li et al., 1995; Pieribone et al., 1995; Sun et al., 2006). In contrast, the requirement of Bruchpilot, which is homologous to the mammalian active zone proteins ELKS/CAST (Wagh et al., 2006), is most pronounced in immediate vesicle release (Kittel et al., 2006). Under repeated stimulation of a bruchpilot mutant motor neuron, immediate vesicle release is severely impaired whereas the following steady-state release is still possible (Kittel et al., 2006). In line with that, knockdown of the Bruchpilot protein causes impairment in clustering of Ca2+ channels to the active zones and a lack of electron-dense projections at presynaptic terminals (T-bars). Thus, less synaptic vesicles of the readily-releasable pool are accumulated to the release sites and their release probability is severely impaired (Kittel et al., 2006; Wagh et al., 2006). First, I showed that Bruchpilot is required for aversive olfactory memory and localized the requirement of Bruchpilot to the Kenyon cells of the mushroom body, the second-order olfactory interneurons in Drosophila. Furthermore, I demonstrated that Bruchpilot selectively functions for the consolidated anesthesia-resistant memory. Since Synapsin is specifically required for the labile anesthesia sensitive memory, different synaptic proteins can dissociate consolidated and labile components of olfactory memory and two different modes of neurotransmission (high- vs. low frequency dependent) might differentiate ASM and ARM.
Aus Vorarbeiten in der Arbeitsgruppe war bekannt, dass Mäuse durch Injektion von tolerogenen, TNF-gereiften und MOG-beladenen DZ vor EAE geschützt werden können. Eines der Ziele dieser Arbeit war es zu untersuchen, ob das koinhibitorische Molekül B7-H1 auf der Oberfläche der DZ einen Einfluss auf das tolerogene Potential der DZ hat. Dazu wurden B7-H1-defiziente DZ generiert, mit TNF gereift, mit MOG-Peptid beladen und intravenös in Mäuse injiziert, bevor die EAE induziert wurde. Es zeigte sich, dass diese DZ die Tiere sogar noch besser vor der Krankheit schützen konnten als die WT DZ. Die Injektion der B7-H1-/- DZ bewirkte eine verstärkte Produktion von IL-10 and IL-13 nach Restimulation der Milz-Zellen in vitro und eine erhöhte Menge von protektiven Serum-Zytokinen (IL-4 und IL-13), welche von NKT-Zellen produziert wurden. Versuche mit CD1d-/- und Jα281-/- Mäusen haben ergeben, dass diese Zytokine von Typ II NKT-Zellen produziert wurden. Weitere Versuche mit Typ I und II NKT-Zell-Linien haben bestätigt, dass nur die Typ II NKT-Zellen von einem endogenen CD1d-Ligand der DZ stimuliert werden können. Außerdem wird dies über B7-H1 reguliert, da die NKT-Zell-Reaktion in Abwesenheit von B7-H1 stärker ist. Des Weiteren konnte gezeigt werden, dass neben den NKT-Zellen MZ B-Zellen nötig sind, um die Mäuse vor der EAE-Entwicklung zu schützen. Versuche mit CD22-/- Mäusen, welche eine Reduktion in der MZ B-Zell-Population zeigen, haben ergeben, dass in Abwesenheit von MZ B-Zellen keine EAE-Protektion mehr möglich ist. In dieser Arbeit wurde auch der Effekt von Glykolipid-Antigenen, welche einen Großteil der Myelinscheide des ZNS ausmachen, auf DZ untersucht. Ausgewählte Lipide führen zu einer Reifung der DZ, welche sich in der verstärkten MHC II- und CD86-Expression, jedoch nicht in der Produktion von Zytokinen äußert. Außerdem sind diese Lipid-gereiften DZ in der Lage T-Zellen zu stimulieren. Als letzter Punkt wurde in dieser Arbeit der Zusammenhang zwischen Masern-Virus-Infektionen und EAE untersucht. Es konnte gezeigt werden, dass eine cerebrale Masern-Infektion zusammen mit einer EAE-Induktion einen dramatischen Effekt auf die Tiere hat. Für diese Versuche wurde ein Maus-Modell einer persistierenden Masern-Infektion im Gehirn verwendet. Diese Tiere leben nach der Virus-Behandlung ohne Symptome. Nach der EAE-Induktion starben diese Tiere jedoch bereits wenige Tage später aufgrund einer MOG-Peptid-spezifischen Reaktion.
Bone Morphogenetic Proteins (BMPs) bilden zusammen mit den Activinen, Growth and Differentiation Factors (GDFs) und Transforming Growth Factor β (TGF-β) die Transforming Growth Factor β-Superfamilie von sekretierten Signalproteinen. Sie spielen eine wichtige Rolle in der Entwicklung, Erhaltung und Regeneration von Geweben und Organen. Die Signalvermittlung dieser Proteine erfolgt durch die Bindung von zwei verschiedenen Typen von Serin-/Threonin-Kinaserezeptoren, die als Typ-I- und Typ-II-Rezeptoren bezeichnet werden. Im ersten Schritt erfolgt die Bindung an den hochaffinen Rezeptor (im Fall von BMP-2 der Typ-I-Rezeptor), im nächsten Schritt wird der niederaffine Rezeptor in den Komplex rekrutiert. Bis heute sind lediglich sieben Typ-I- und fünf Typ-II-Rezeptoren bekannt, was auf eine Promiskuität in der Liganden-Rezeptor-Interaktion schließen lässt. Die Architektur beider Rezeptorsubtypen ist dabei relativ ähnlich. Beide bestehen aus einer ligandenbindenden extrazellulären Domäne, einer Transmembrandomäne sowie einer intrazellulären Kinasedomäne. Eine nacheinander ablaufende Transphosphorylierung der intrazellulären Domänen führt zu einer Phosphorylierung von SMAD-Proteinen, die dann als nachgeschaltete Vermittler fungieren und die Transkription regulierter Gene auslösen. Im Hauptteil dieser Arbeit wurden die initialen Schritte der Rezeptorkomplexformierung sowie die Mobilität der Rezeptoren mit Hilfe von fluoreszenzmikroskopischen Methoden untersucht. Dabei konnte festgestellt werden, dass für die Bildung eines Signalkomplexes eine bestimmte Schwellenkonzentration des Liganden nötig ist und dass der Mechanismus nach einem Alles-oder-Nichts-Prinzip wie ein Schalter funktioniert. Außerdem konnten Unterschiede in der Nutzung der gleichen Rezeptoren durch verschiedene Liganden festgestellt werden. Die anderen Teile der Arbeit befassen sich mit der Funktionalität der verschiedenen Rezeptordomänen in der Signalübermittlung, der Analyse von hoch- und niederaffinen Ligandenbindestellen auf ganzen Zellen sowie dem Einfluss des SMAD- und des MAPK-Signalwegs auf die Induktion der Alkalischen Phosphatase. Dabei konnte gezeigt werden, dass die Art der SMAD-Phosphorylierung allein vom Typ der Kinasedomäne abhängig ist, dass auf einer Zelle verschiedene Rezeptorpopulationen existieren, welche von unterschiedlichen Ligandenkonzentrationen angesprochen werden, und dass die Induktion der Alkalischen Phosphatase stark vom zeitlichen Verlauf der SMAD- und MAPK-Aktivierung abhängig ist.
GDF-15 ist ein atypisches Mitglied der TGF-b-Superfamilie. Unter physiologischen Bedingungen kommt es nur in der Plazenta in größeren Mengen vor, während es in zahlreichen Tumoren überexprimiert gefunden wurde. Die genaue Funktion von GDF-15 im Tumorkontext ist nicht genau geklärt. Aufgrund der häufigen und hohen Expression in Tumoren scheint GDF-15 eine wesentliche Funktion im Tumorprogress auszuüben. Das Ovarialkarzinom (OvCA) nimmt die Stellung als tödlichste gynäkologische Erkrankung ein. Da der Tumor meist erst in fortgeschrittenen Stadien diagnostiziert wird, sind bis heute die Heilungschancen schlecht. Häufig kommt es zum Rezidiv nach zunächst erfolgreicher Chemotherapie und mit 30% ist die 5-Jahres-Überlebenschance gering. Für die chemoresistenten Fälle gibt es bis zum heutigen Zeitpunkt keine effektive Therapie. Dies verdeutlicht die Notwendigkeit, neue innovative Therapiestrategien zu entwickeln. Günstige immunologische Parameter korrelieren mit der Überlebensdauer von OvCA-Patientinnen, was die Immuntherapie beim OvCA in den Fokus der experimentellen klinischen Therapie rückt. Doch um neue immuntherapeutische Strategien entwickeln zu können, müssen zunächst immunologisch relevante Angriffspunkte identifiziert werden. Das in vielen Tumoren exprimierte GDF-15 ist mit einem der stärksten immunsuppressiven Faktoren verwandt, was die Vermutung nahe legt, dass auch GDF-15 eine immunologisch relevante Funktion im Tumorkontext ausüben könnte. Daher wurden die Expression und die mögliche Funktion von GDF-15 als Immunmodulator im Ovarialkarzinom untersucht. Expressionsanalysen von OvCA-Gewebe und primären OvCA-Zellen zeigten, dass GDF-15 das am stärksten überexprimierte Gen der untersuchten TGF-b-Familienmitglieder im OvCA ist. Auch als sezerniertes Protein wird GDF-15 in vivo und in vitro im OvCA detektiert, was auf eine funktionale Rolle von GDF-15 im OvCA hindeutet. Normalerweise eliminiert das Immunsystem entartete körpereigene Zellen. Manchmal gelingt es Tumorzellen jedoch, sich dieser Immunüberwachung zu entziehen und dem Immunsystem zu „entwischen“. Inwieweit GDF-15 bei der Koordination des „immune escape“ des OvCA eine Rolle spielt, sollte im Fokus dieser Arbeit stehen. Das Hauptaugenmerk lag dabei auf der Wirkung von GDF-15 auf NK-Zellen, da diese als frühe Effektoren und wichtige Mediatoren zwischen angeborenem und adaptivem Immunsystem nicht nur eine Schlüsselrolle bei der immunologische Überwachung spielen, sondern sich dadurch auch als ideale Werkzeuge für die Tumorimmuntherapie auszeichnen. Exogenes GDF-15 hemmt in vitro die Lyseaktivität von NK-Zellen gegenüber OvCA-Zellen. Endogene GDF-15-Defizienz der OvCA-Zellen sensitiviert diese für NK-Zell-Lyse und endogene GDF-15-Überexpression mindert die NK-Lyseaktivität. Die Hemmung der NK-Lyseaktivität kann durch verschiedene synergistisch wirkende Mechanismen erfolgen: durch Rezeptormodulation, durch direkte Modulation des Lysemechanismus und durch Apoptoseregulation. Wie TGF-b1 reguliert GDF-15 die Expression des aktivierenden NK-Rezeptors NKG2D von der Zelloberfläche herunter und induziert zusätzlich die Expression des inhibierenden Rezeptors CD305 und die des mit NKG2A- und NKG2C-assoziierten Rezeptors CD94. Daneben greift GDF-15 direkt in den Lysemechanismus der NK-Zellen ein, indem es die Granzym B-Expression beeinflusst. Darüber hinaus sensitiviert GDF-15 Immunzellen für die Apoptose durch die Induktion von Fas/CD95. Signaltransduktionsanalysen zeigen, dass GDF-15 in Immunzellen die SMAD-Proteine zeitverzögert zu TGF-b aktiviert, was auf eine indirekte Wirkung schließen lässt. Zusätzlich kann GDF-15 auch die die p38/MAPK in Immunzellen aktivieren. Die Genregulation von GDF-15 und TGF-b1 in NK-Zellen ist sehr verschieden. Beide Zytokine regulieren überwiegend Gene aus gleichen Funktionalitätsclustern, allerdings sind die einzelnen von TGF-b1 und GDF-15 regulierten Gene verschieden. Nur drei Gene (CD55, Caspase-8 und Apolipoprotein 6) sind durch GDF-15 und TGF-b1 gleich reguliert. Zusammengefasst zeigt sich eine funktionale Analogie von GDF-15 und TGF-b1 in NK-Zellen. TGF-b1 scheint eine stärkere Wirkung zu induzieren, dafür zeigt GDF-15 hier ein breiteres Funktionalitätsspektrum. Durch die Charakterisierung der funktionalen Rolle von GDF-15 als Immunmodulator in Tumoren ist hier ein neuer potentieller Angriffspunkt identifiziert worden, welcher Grundlage für neue Tumortherapiestrategien, nicht nur für das OvCA, sondern auch für andere GDF-15-exprimierende Tumore sein kann.
Avian pathogenic Escherichia coli (APEC) represent a subset of the so-called extraintestinal pathogenic Escherichia coli (ExPEC) pathotype that can cause various extraintestinal infections in humans and animals. APEC are the causative agent of localized colibacillosis or systemic infection in poultry. In this latter case, the syndrome starts as an infection of the upper respiratory tract and develops into a systemic infection. Generally, ExPEC are characterized by a broad variety of virulence-associated factors that may contribute to pathogenesis. Major virulence factors, however, that clearly define this pathotype, have not been identified. Instead, virulence-associated genes of ExPEC and thus also of APEC could be used in a mix-and-match-fashion. Both pathotypes could not be clearly distinguished by molecular epidemiology, and this suggested a hypothetical zoonotic risk caused by APEC. Accordingly, the main scientific question of this study was to characterize common traits as well as differences of APEC and human ExPEC variants that could either support the possible zoonotic risk posed by these pathogenic E. coli strains or indicate factors involved in host specificity. Comparative genomic analysis of selected APEC and human ExPEC isolates of the same serotype indicated that these variants could not be clearly distinguished on the basis of (i) general phenotypes, (ii) phylogeny, (iii) the presence of typical ExPEC virulence genes, and (iv) the presence of pathoadaptive mutations. Allelic variations in genes coding for adhesins such as MatB and CsgA or their regulators MatA and CsgD have been observed, but further studies are required to analyze their impact on pathogenicity. On this background, the second part of this thesis focused on the analysis of differences between human ExPEC and APEC isolates at the gene expression level. The analysis of gene expression of APEC and human ExPEC under growth conditions that mimick their hosts should answer the question whether these bacterial variants may express factors required for their host-specificity. The transcriptomes of APEC strain BEN374 and human ExPEC isolate IHE3034 were compared to decipher whether there was a specific or common behavior of APEC and human ExPEC, in response to the different body temperatures of man (37°C) or poultry (41°C). Only a few genes were induced at 41 °C in each strain relative to growth at 37 °C. The group of down-regulated genes in both strains was markedly bigger and mainly included motility and chemotaxis genes. The results obtained from the transcriptome, genomic as well as phenotypic comparison of human ExPEC and APEC, supports the idea of a potential zoonotic risk of APEC and certain human ExPEC variants. In the third part of the thesis, the focus was set on the characterization of Mat fimbriae, and their potential role during ExPEC infection. Comparison of the mat gene cluster in K-12 strain MG1655 and O18:K1 isolate IHE3034 led to the discovery of differences in (i) DNA sequence, (ii) the presence of transcriptional start and transcription factor binding sites as well as (iii) the structure of the matA upstream region that account for the different regulation of Mat fimbriae expression in these strains. A negative role of the H-NS protein on Mat fimbriae expression was also proven at 20 °C and 37 °C by real-time PCR. A major role of this fimbrial adhesin was demonstrated for biofilm formation, but a significant role of Mat fimbriae for APEC in vivo virulence could not yet be determined. Interestingly, the absence of either a functional matA gene or that of the structural genes matBCDEF independently resulted in upregulation of motility in E. coli strains MG1655 and IHE3034 by a so far unknown mechanism. In conclusion, the results of this thesis indicate a considerable overlap between human and animal ExPEC strains in terms of genome content and phenotypes. It becomes more and more apparent that the presence of a common set of virulence-associated genes among ExPEC strains as well as similar virulence gene expression patterns and phylogenetic backgrounds indicate a significant zoonotic risk of avian-derived E. coli isolates. In addition, new virulence factors identified in human ExPEC may also play a role in the pathogenesis of avian ExPEC.
In dieser Doktorarbeit habe ich die Regulation der Expression des zuckerbelohnten Verhaltens durch den Fütterungszustand bei Drosophila melanogaster untersucht. Die Fliegen können während einer Trainingsphase mit Hilfe einer Zuckerbelohnung auf einen bestimmten Duft konditioniert werden. Nach dem Training können die Fliegen dann auf das olfaktorische Gedächtnis getestet werden. Die Bereitschaft das zuckerkonditionierte Gedächtnis im Test zu zeigen wird vom Fütterungszustand kontrolliert, wie ich in Übereinstimmung mit den Ergebnissen früherer Arbeiten demonstrierte (Tempel et al. 1983; Gruber 2006; Krashes et al. 2008). Nur nicht gefütterte Fliegen exprimieren das Gedächtnis, während Fütterungen bis kurz vor dem Test eine reversibel supprimierende Wirkung haben. Einen ähnlichen regulatorischen Einfluss übt der Futterentzug auch auf die Expression anderer futterbezogener Verhaltensweisen, wie z.B. die naive Zuckerpräferenz, aus. Nachdem ich den drastischen Einfluss des Fütterungszustands auf die Ausprägung des zuckerkonditionierten Verhaltens gezeigt bzw. bestätigt hatte, habe ich nach verhaltensregulierenden Faktoren gesucht, die bei einer Fütterung die Gedächtnisexpression unterdrücken. Als mögliche Kandidaten untersuchte ich Parameter, die zum Teil bereits bei verschiedenen futterbezogenen Verhaltensweisen unterschiedlicher Tierarten als „Sättigungssignale“ identifiziert worden waren (Marty et al. 2007; Powley and Phillips 2004; Havel 2001; Bernays and Chapman 1974; Simpson and Bernays 1983; Gelperin 1971a). Dabei stellte sich heraus, dass weder die „ernährende“ Eigenschaft des Futters, noch ein durch Futteraufnahme bedingter Anstieg der internen Glukosekonzentration für die Suppression des zuckerkonditionierten Gedächtnisses notwendig sind. Die Unterdrückung der Gedächtnisexpression kann auch nicht durch Unterschiede in den aufgenommenen Futtermengen, die als verhaltensinhibitorische Dehnungssignale des Verdauungstrakts wirken könnten, oder mit der Stärke des süßen Geschmacks erklärt werden. Die Suppression des zuckerbelohnten Verhaltens folgte den Konzentrationen der gefütterten Substanzen und war unabhängig von deren chemischen Spezifität. Deshalb wird die Osmolarität des aufgenommenen Futters als ein entscheidender Faktor für die Unterdrückung der zuckerkonditionierten Gedächtnisexpression angenommen. Weil nur inkorporierte Substanzen einen Unterdrückungseffekt hatten, wird ein osmolaritätsdetektierender Mechanismus im Körper 67 postuliert, wahrscheinlich im Verdauungstrakt und/oder der Hämolymphe. Die Hämolymphosmolarität ist als „Sättigungssignal“ bei einigen wirbellosen Tieren bereits nachgewiesen worden (Bernays and Chapman 1974; Simpson and Raubenheimer 1993; Gelperin 1971a; Phifer and Prior 1985). Deshalb habe ich mit Hilfe genetischer Methoden und ohne die Fliegen zu füttern, versucht über einen künstlich induzierten Anstieg der Trehaloseund Lipidkonzentrationen die Osmolarität der Hämolymphe in Drosophila zu erhöhen. Eine solche konzentrationserhöhende Wirkung für Lipide und die Trehalose, dem Hauptblutzucker der Insekten, ist bereits für das adipokinetische Hormon (AKH), das von Zellen der Corpora cardiaca exprimiert wird, nachgewiesen worden (Kim and Rulifson 2004; Lee and Park 2004; Isabel et al. 2005). Es stellte sich heraus, dass die künstliche Stimulierung AKH-produzierender Neurone das zuckerkonditionierten Verhalten temporär, reversible und selektiv unterdrückt. Gleiche Behandlungen hatten keinen Effekt auf ein aversiv konditioniertes olfaktorisches Gedächtnis oder ein naives Zuckerpräferenzverhalten. Wie aus dieser Arbeit hervorgeht, stellt wahrscheinlich die Osmolarität des Verdauungstrakts und der Hämolymphe oder nur der Hämolymphe ein physiologisches Korrelat zum Fütterungszustand dar und wirkt als unterdrückendes Signal. Dass Fütterungen das zuckerkonditionierte Verhalten und die Zuckerpräferenz supprimieren, die künstliche Stimulation AKH-produzierender Zellen aber selektiv nur die zuckerbelohnte Gedächtnisexpression unterdrückt, deutet auf mindestens zwei unterschiedliche „Sättigungssignalwege“ hin. Außerdem macht es deutlich wie uneinheitlich futterbezogene Verhaltensweisen, wie das zuckerbelohnte Verhalten und die naive Zuckerpräferenz, reguliert werden.
Pluripotente embryonale Stammzellen (ES Zellen) sind aufgrund ihrer Selbsterneuerung- und ihrer Multiliniendifferenzierungs-Fähigkeiten interessante Zelltypen sowohl für die Grundlagenforschung als auch für die regenerative Medizin. Uniparentale Zygoten mit zwei väterlichen (androgenetisch: AG) oder zwei mütterlichen (gynogenetisch: GG; parthenogenetisch: PG) Genomen sind nicht in der Lage, lebensfähige Nachkommen zu entwickeln. Sie entwickeln sich jedoch erfolgreich bis zu Blastozysten, aus denen pluripotente ES Zellen abgeleitet werden können. Mit uniparentalen ES Zellen können zum Einen parent-of-origin-spezifische Einflüsse auf die Gewebeentwicklung untersucht und zum Anderen histokompatible und somit therapeutisch relevante Zellpopulationen generiert werden. Obwohl viele Aspekte des in vitro und in vivo Differenzierungspotenzials von PG ES Zellen aus mehreren Spezies in den zurückliegenden Jahren untersucht worden sind, ist das volle Differenzierungspotenzial von AG ES Zellen bisher nicht erschöpfend analysiert worden. Zellen der Inneren Zellmasse (ICM) von PG und AG Embryonen zeigten nach Blastozysteninjektion ortsspezifische Kontribution zur Gehirnentwicklung, wobei PG Zellen bevorzugt im Cortex und im Striatum lokalisierten, während sich AG Zellen verstärkt im Hypothalamus nachzuweisen waren. Aus AG und GG ES Zellen konnten zudem in vitro hämatopoetische Stammzellen differenziert werden, die nach Transplantation im Mausmodell tumorfrei das gesamte hämatopoetische System repopulierten. Weiterhin konnte gezeigt werden, dass AG ES Zellen ein mit N ES Zellen vergleichbares in vitro und in vivo Differenzierungspotential in der frühen neuralen Entwicklung besitzen. Das Ziel meiner Arbeit war es zu untersuchen, ob murine AG ES Zellen sich zu verschiedenen neuronalen Subtypen entwickeln können und ob sie tumorfrei neurale Zelltypen nach Transplantation bilden können. In dieser Studie wurden AG ES Zellen im Vergleich zu biparentalen (N) ES Zellen in vitro über Embryoid Bodies (EBs) zunächst zu pan-neuronalen Vorläuferzellen (pNPCs) und weiter zu Neuron- und Glialzell-Marker (ß-III Tubulin (Tuj-1), NeuN, TH und GFAP) positiven Zellen differenziert.. Weiterhin wurde das dopaminerge (DA) Differenzierungspotential von AG ES Zellen näher untersucht, indem sie in einem Ko-Kultursystem mit Stromazellen gerichtet differenziert wurden. Diese DA Neurone wurden durch semiquantitative RT-PCR Analysen und immunhistochemische Färbungen für DA Neuronen-spezifische Marker (TH, PITX3, Nurr1) charakterisiert. Darüber hinaus wurde der Imprinting-Status von neun ausgesuchten Loci in AG und N ES, pNPC und DA Zellkulturen durch real-time RT-PCR Analysen untersucht. Die hier analysierten Gene, die im Gehirn allelspezifisch exprimiert werden, zeigten in pNPCs eine parent-of-origin-spezifische Genexpression mit Ausnahme von Ube3a. Nach Blastozysteninjektion wurde die Bildung von DA Neuronen in AG und N fötalen chimären Gehirnen untersucht. Hier zeigte sich, dass TH- and PITX3-positive AG DA Neurone abgeleitet aus ES Zellen im Mittelhirn von E12.5 und E16.5 Chimären detektiert werden konnten. Diese fötalen chimären Gehirne zeigten eine verbreitete und gleichmäßige Verteilung der AG Donorzellen in den Arealen Cortex, Striatum und Hypothalamus. Stereotaktische Transplantationen von AG und N pNPCs in ein „Traumatic Brain Injury (TBI) Model“ zeigten zudem, dass frühe Differenzierungsstufen von AG und N pNPC-Kulturen häufig Teratome generierten. Durch die Transplantation von langzeitdifferenzierten AG oder N pNPC-Kulturen konnte jedoch ein tumorfreies Anwachsen neuronaler und glialer Zellen erreicht werden. Die immunhistochemische Auswertung von Transplantaten bezüglich der Donorzellkontribution im Gehirn erfolgten bis zu drei Monaten nach der Injektion. Die vorliegenden Ergebnisse zeigen, dass AG ES Zellen neurales Differenzierungspotential, speziell zur Bildung von DA Neuronen, besitzen. Darüber hinaus konnte gezeigt werden, dass langzeitdifferenzierte AG und N pNPCs nach Transplantation im traumatisierte Mausgehirnmodell tumorfrei anwachsen und anschließend zu neuralen Zellen differenzieren können. Trotz unbalancierter Genexpression von imprinted Genen lässt sich feststellen, dass AG ES Zellen therapeutisch relevant für zukünftige zelluläre Ersatzstrategien von Nervengewebe sein können.
Metastasis is the cause of death in 90% of cancer-related deaths in men. Melanoma and Non-Small-Cell Lung Cancer (NSCLC) are both tumour types with poor prognosis, lacking appropriate therapeutic possibilities, not least because of their high rate of metastasis. Thus understanding the process of metastasis might unravel therapeutic targets for developing further therapeutic strategies. The generation of a transgenic mouse model expressing B-RafV600E in melanocytes, a mutation that is found in about 60% of all melanoma, would result in an ideal tool to study melanoma progression and metastasis. In this work, a doxycycline-inducible system was constructed for expression of B-RafV600E and transgenic animals were generated, but the expression system has to be improved, since this strategy didn’t give rise to any viable, transgene carrying mice. Furthermore, since it was shown in the work of others that the metastatic behavior of tumour cell lines could be reversed by an embryonic microenvironment and the influence of a tumourigenic microenvironment on melanocytes lead to the acquisition of tumour cell-like characteristics, the question arose, whether B-Raf is as important in melanocyte development as it is in melanoma progression. In this work, the embryonal melanocyte development in B-Raf-deficient and wildtype mouse embryos was examined and there were no differences observed in the localization and number of neural crest stem cells as well as in the localization of the dopachrome-tautomerase positive melanoblasts in the embryos and in cultured neural tube explants. The expression of oncogenic C-Raf in lung epithelial cells has yielded a model for NSCLC giving rise to adenomas lacking spontaneous progression or metastasis. The co-expression of c-Myc in the same cells accelerates the tumour development and gives rise to liver and lymphnode metastases. The expression of c-Myc alone in lung epithelial cells leads to late tumour development with incomplete penetrance. A mutation screen in this work resulted in the observation that a secondary mutation in KRas or LKB1 is necessary for tumour formation in the c-Myc single transgenic animals and suggested metastasis as an early event, since the corresponding metastases of the mutation-prone primary lung tumours were negative for the observed mutations. Furthermore, in this work it was shown that the expression of chicken c-Myc in a non-metastatic NSCLC cell line leads to metastatic clones, showing that c-Myc is sufficient to induce metastasis. Additionally a panel of metastasis markers was identified, that might serve as diagnostic markers in the future.
In dieser Arbeit sollte die Funktion der Ribosomalen S6 Kinase 2 (RSK2) auf neuronaler Ebene untersucht werden. Dahingehend gab es, z.B. auf Grund der Phänotypen von Fliegen und Mäusen mit Mutationen im entsprechenden Gen oder von Patienten mit Coffin-Lowry-Syndrom (CLS) nur Vermutungen. Es bestand letztlich die Hoffnung, einen Beitrag zur Aufklärung der Pathophysiologie des CLS zu leisten. Es stellte sich auf Grund von Experimenten sowohl in vivo als auch in vitro in verschiedenen Modellsystemen in dieser Arbeit heraus, daß RSK2 einen negativen Einfluß auf das Neuriten- und Synapsenwachstum hat. In kultivierten Motoneuronen führte der KO von RSK2 zu längeren Axonen und die Überexpression eines konstitutiv aktiven RSK2-Konstrukts zu kürzeren Axonen. In PC12-Zellen führte die Expression von konstitutiv aktiven RSK2 Konstrukten zur Verkürzung der Neuriten und die Expression eines Kinase-inaktiven RSK2 Konstrukts zu längeren Neuriten. In vivo war die neuromuskuläre Synapse bei RSK2-KO Mäusen vergrößert und hatte bei Drosophila rsk Mutanten mehr Boutons. Das RSK2-Protein ist in Motoneuronen der Maus und in überexprimierter Form in den Boutons der neuromuskulären Synapse bei Drosophila nachweisbar. Damit wurde zum ersten Mal die Funktion von RSK2 auf neuronaler Ebene beschrieben. Bezüglich des Mechanismus, wie RSK2 das Nervenwachstum beeinflußt gab es deutliche Hinweise, die dafür sprechen, daß RSK2 dies über eine in der Literatur schon häufiger beschriebene Hemmung der MAPK ERK1/2 erreicht. Für diese Hypothese spricht die Tatsache, daß die ERK-Phosphorylierung in murinen Motoneuronen und im Rückenmark embryonaler Mäuse der RSK2-Mutante erhöht ist und der Axonwachstumsdefekt durch eine Hemmung von MEK/ERK behoben werden kann. Auch ist die ERK-Phosphorylierung an der murinen Muskel-Endplatte in der Mutante erhöht. Zudem zeigen genetische Epistasis-Experimente in Drosophila, daß RSK die Bouton-Zahl über ERK/RL hemmt. RSK scheint also in Drosophila von der Funktion her der RSK2-Isoform in Wirbeltieren sehr ähnlich zu sein. Ein weiteres wichtiges Ergebnis ist die Beobachtung, daß RSK2 bei Motoneuronen keinen wesentlichen Einfluß auf das Überleben der Zellen in Gegenwart neurotropher Faktoren hat. Möglicherweise spielen hier redundante Funktionen der RSK Familienmitglieder eine Rolle. Ein bislang unerklärter Befund ist die reduzierte Frequenz spontaner Depolarisationen bzw. damit einhergehender Ca2+ Einströme bei RSK2-KO Motoneuronen in Zellkultur. Die Häufigkeit und Dichte von Ca2+-Kanälen und aktive Zonen Proteinen war in Motoneuronen nicht von der Anwesenheit des RSK2-Proteins abhängig. Im Hippocampus konnte außerdem das RSK2-Protein präsynaptisch in den Moosfaser-Boutons der CA3 Region nachgewiesen werden. Es befindet sich auch in den Pyramidenzellen, aber nicht in den Pyramidenzell-Dendriten in CA3. Bezüglich der Bedeutung dieser Befunde für die Aufklärung der Pathologie des CLS ist zu folgern, daß der neuro-psychologische Phänotyp bei CLS Patienten wahrscheinlich nicht durch reduziertes Überleben von Neuronen, sondern eher durch disinhibiertes Axonwachstum oder Synapsenwachstum bedingt ist. Dies kann grob sowohl für die peripheren als auch die zentralen Defekte gelten, denn die Synapsen im ZNS und am Muskel sind in ihrer molekularen Ausstattung z.B. im Bereich der Vesikel, der aktiven Zonen oder der Transmitterausschüttung sehr ähnlich. Weiterhin könnte eine veränderte synaptische Plastizität u.a. an der Moosfaser-Pyramidenzell-Synapse in der CA3 Region des Hippocampus eine Rolle bei den kognitiven und mnestischen Einschränkungen der Patienten spielen. Die Entdeckung, daß aktiviertes ERK bei den beobachteten Effekten eine Rolle spielt kann für die Entwicklung von Therapiestrategien eine wertvolle Erkenntnis sein.
Members of the RAF protein kinase family are key regulators of diverse cellular processes. The need for isoform-specific regulation is reflected by the fact that all RAFs not only display a different degree of activity but also perform isoform-specific functions at diverse cellular compartments. Protein-protein-interactions and phosphorylation events are essential for the signal propagation along the Ras-RAF-MEK-ERK cascade. More than 40 interaction partners of RAF kinases have been described so far. Two of the most important regulators of RAF activity, namely Ras and 14-3-3 proteins, are subject of this work. So far, coupling of RAF with its upstream modulator protein Ras has only been investigated using truncated versions of RAF and regardless of the lipidation status of Ras. We quantitatively analyzed the binding properties of full-length B- and C-RAF to farnesylated H-Ras in presence and absence of membrane lipids. While the isolated Ras-binding domain of RAF exhibit a high binding affinity to both, farnesylated and nonfarnesylated H-Ras, the full-length RAF kinases demonstrate crucial differences in their affinity to Ras. In contrast to C-RAF that requires carboxyterminal farnesylated H-Ras for interaction at the plasma membrane, B-RAF also binds to nonfarnesylated H-Ras in the cytosol. For identification of the potential farnesyl binding site we used several fragments of the regulatory domain of C-RAF and found that the binding of farnesylated H-Ras is considerably increased in the presence of the cysteine-rich domain of RAF. In B-RAF a sequence of 98 amino acids at the extreme N terminus enables binding of Ras independent of its farnesylation status. The deletion of this region altered Ras binding as well as kinase properties of B-RAF to resemble C-RAF. Immunofluorescence studies in mammalian cells revealed essential differences between B- and C-RAF regarding the colocalization with Ras. In conclusion, our data suggest that that B-RAF, in contrast to C-RAF, is also accessible for nonfarnesylated Ras in the cytosolic environment due to its prolonged N terminus. Therefore, the activation of B-RAF may take place both at the plasma membrane and in the cytosolic environment. Furthermore, the interaction of RAF isoforms with Ras at different subcellular sites may also be governed by the complex formation with 14-3-3 proteins. 14-3-3 adapter proteins play a crucial role in the activation of RAF kinases, but so far no information about the selectivity of the seven mammalian isoforms concerning RAF association and activation is available. We analyzed the composition of in vivo RAF/14-3-3 complexes isolated from mammalian cells with mass spectrometry and found that B-RAF associates with a greater variety of 14-3-3 proteins than C- and A-RAF. In vitro binding assays with purified proteins supported this observation since B-RAF showed highest affinity to all seven 14-3-3 isoforms, whereas C-RAF exhibited reduced affinity to some and A-RAF did not bind to the 14-3-3 isoforms epsilon, sigma, and tau. To further examine this isoform specificity we addressed the question of whether both homo- and heterodimeric forms of 14-3-3 proteins participate in RAF signaling. By deleting one of the two 14-3-3 isoforms in Saccharomyces cerevisiae we were able to show that homodimeric 14-3-3 proteins are sufficient for functional activation of B- and C-RAF. In this context, the diverging effect of the internal, inhibiting and the activating C-terminal 14-3-3 binding domain in RAF could be demonstrated. Furthermore, we unveil that prohibitin stimulates C-RAF activity by interfering with 14-3-3 at the internal binding site. This region of C-RAF is also target of phosphorylation as part of a negative feedback loop. Using tandem MS we were able to identify so far unknown phosphorylation sites at serines 296 and 301. Phosphorylation of these sites in vivo, mediated by activated ERK, leads to inhibition of C-RAF kinase activity. The relationship of prohibitin interference with 14-3-3 binding and phosphorylation of adjacent sites has to be further elucidated. Taken together, our results provide important new information on the isoform-specific regulation of RAF kinases by differential interaction with Ras and 14-3-3 proteins and shed more light on the complex mechanism of RAF kinase activation.
Die Matrix-Proteine von Vertretern der Ordnung Mononegavirales sind essentiell für späte Schritte im viralen Lebenszyklus, insbesondere der Knospung und Partikelmorphogenese. Die Abschnürung und Freisetzung umhüllter RNA-Viren ist dabei abhängig von dem Transport des viralen Matrix-Proteins und seiner Interaktion mit Wirtsproteinen wie dem ESCRT-System (endosomal sorting complex required for transport), welches in die Sortierung zellulärer Proteine involviert ist. Im Verlauf der Masernvirus (MV)-Infektion interagiert das M-Protein einerseits mit dem viralen Nukleoproteinkomplex und andererseits mit den viralen Glykoproteinen an der Oberfläche. Die Bedeutung des MV-M-Proteins für die Partikelproduktion und sein intrazellulärer Transport wurden bislang kaum untersucht. Bisher ist nur bekannt, dass das M-Protein oligomerisiert, teilweise monoubiquitiniert vorliegt und in Zellen, wenn alleine exprimiert, die Produktion von Virus-like-particle (VLP) vermittelt (Pohl et al., 2007). In dieser Studie wird gezeigt, dass das MV-M-Protein ähnlich wie das VP40-Protein des Ebolavirus (EBOV) mit Lipid Rafts und TEMs (tetraspanin enriched microdomain) assoziiert ist, wobei aber das M-Protein weniger effizient an der Plasmamembran akkumuliert. Beide Proteine unterscheiden sich nicht wesentlich in ihrer Assoziation mit Kompartimentmarkern. Interessant ist jedoch, dass das VP40-Protein und das M-Protein an der Plasmamembran mit dem Adaptor Protein-3 (AP-3) kolokalisieren. Die Kolokalisation des M-Proteins mit AP-3 wird aber nur in infizierten Zellen und nicht in Zellen, in denen das M-Protein allein exprimiert wird, beobachtet. Im Gegensatz zum VP40-Protein, welches die ESCRT-Komponenten über seine N-terminale L-Domäne rekrutiert und diese für die Partikelproduktion benutzt, geschieht dies beim M-Protein ESCRT-unabhängig, da die Mutation der Motive, die Ähnlichkeiten zu den bekannten L-Domänen zeigen, keine Auswirkungen auf die VLP-Produktion haben. Zudem rekrutierte das M-Protein weder Tsg101, Aip-1 oder Vps4 an die Plasmamembran, noch wird die VLP- oder die Virusproduktion durch dominant negatives Vps4 inhibiert. Der Transfer der VP40 L-Domäne in das MV-M-Protein hatte weder einen Einfluss auf die Assoziation mit Tetraspaninen noch auf die ESCRT-Abhängigkeit der VLP-Produktion. Damit wurde gezeigt, dass die VLP-Freisetzung des MV-M-Proteins ESCRT-unabhängig ist. Die Freisetzung erfolgt beim MV durch einen grundsätzlich anderen Weg, der noch untersucht werden muss.
Die Regulation des Tonus glatter Muskelzellen wird entscheidend von den beiden antagonistisch wirkenden second messengern cAMP und Ca2+ beeinflusst. Ein Ziel dieser Arbeit war herauszufinden, ob diese beiden Botenstoffe auch direkten Einfluss aufeinander haben können und welche Enzyme in diesem Fall an den Prozessen beteiligt sind. cAMP-Signale in intakten Zellen konnten wir in Echtzeit mit Hilfe des FRET-basierten cAMP-Sensors Epac1-camps beobachten; Ca2+-Signale durch Markieren der Zellen mit Fura-2. Anstiege der intrazellulären Ca2+-Konzentration in VSMCs wurden durch Aktivierung von endogen exprimierten, Gq-gekoppelten P2Y6-Rezeptoren mit Uridindiphosphat (UDP) ausgelöst. Durch eine zusätzliche in-vitro Kalibrierung des Epac1-camps konnten darüber hinaus absolute cAMP-Konzentrationen in einzelnen lebenden Zellen berechnet werden. Während ein Anstieg der Ca2+-Konzentration auf nicht vorstimulierte VSMCs keinen signifikante Einfluss auf die intrazellulären cAMP-Konzentrationen hatte, bewirkte die Aktivierung der purinergen Rezeptoren einen deutlichen Rückgang der intrazellulären cAMP-Konzentration in mit Isoproterenol vorstimulierten VSMCs. Dieser Effekt konnte sowohl durch die Komplexierung von Ca2+ mit BAPTA-AM als auch durch die Überexpression der Ca2+-insensitiven AC4 antagonisiert werden. Adenylatcyclase-Aktivitäts-Assays in VSMC-Membranen zeigten ebenfalls einen Rückgang der Cyclaseaktivität nach Zugabe von 2 und 5 μM freiem Ca2+. Die Hemmung der einzigen Ca2+-regulierbaren PDE1 mit dem selektiven PDE1-Inhibitor 8-Methoxymethyl-IBMX (8-MM-IBMX) hatte im Gegensatz dazu keinen Einfluss auf die durch UDP verursachte Änderung der cAMP-Konzentration in vorstimulierten VSMCs. Schließlich bewirkte die Herunterregulation der Ca2+-inhibierbaren AC5 und 6 mit siRNA einen signifikante Hemmung des durch UDP verursachten Effekts. Fasst man alle diese Ergebnisse zusammen, so lässt sich folgende Schlussfolgerung ziehen: Der durch purinerge Stimulation verursachte Rückgang der cAMP-Konzentration in mit Isoproterenol vorstimulierten VSMCs wird durch eine Hemmung der Ca2+-hemmbaren AC5 und 6 vermittelt. Dadurch sind zwei für die Regulation des Tonus wichtige Signalwege in VSMCs miteinander verbunden, die sich somit gegenseitig entscheidend beeinflussen können. Ein weiterer Bestandteil dieser Arbeit war die Entwicklung eines transgenen Mausmodells, das glattmuskelspezifisch den cAMP-Sensor Epac1-camps exprimiert. Mit Hilfe eines solchen Tiermodells könnten in Zukunft cAMP-Änderungen in intakten Geweben und vielleicht sogar in lebenden Tieren beobachtet werden. Durch Anwendung des Cre-loxP-Rekombinationssystems gelang es eine glatt¬muskelspezifische, für den Epac1-camps transgene Mauslinie zu generieren. Mit isolierten VSMCs dieser Tiere konnten bereits erste FRET-Messungen durchgeführt und agonistinduzierte cAMP-Änderungen beobachtet werden.
Cysteines play important roles in the biochemistry of many proteins. The high reactivity, redox properties, and ability of the free thiol group to coordinate metal ions designate cysteines as the amino acids of choice to form key catalytic components of many enzymes. Also, cysteines readily react with reactive oxygen and nitrogen species to form reversible oxidative thiol modifications. Over the last few years, an increasing number of proteins have been identified that use redox-mediated thiol modifications to modulate their function, activity, or localization. These redox-regulated proteins are central players in numerous important cellular processes. First aim of this study was to discover nitric oxide (NO) sensitive proteins in E. coli, whose redox-mediated functional changes might explain the physiological alterations observed in E. coli cells suffering from NO-stress. To identify E. coli proteins that undergo reversible thiol modifications upon NO-treatment in vivo, I applied a differential thiol trapping technique combined with two-dimensional gel analysis. 10 proteins were found to contain thiol groups sensitive to NO-treatment. Subsequent genetic studies revealed that the oxidative modifications of AceF & IlvC are, in part, responsible for the observed NO-induced growth inhibition. Noteworthy, the majority of identified protein targets turned out to be specifically sensitive towards reactive nitrogen species. This oxidant specificity was tested on one NO-sensitive protein, the small subunit of glutamate synthase. In vivo and in vitro activity studies demonstrated that glutamate synthase rapidly inactivates upon nitric oxide treatment but is resistant towards other oxidative stressors. These results imply that reactive oxygen and nitrogen species affect distinct physiological processes in bacteria. The second aim of my study was to identify redox-sensitive proteins in S. cerevisiae and to use their redox state as in vivo read-out to assess the role of oxidative stress during the eukaryotic aging process. I first determined the precise in vivo thiol status of almost 300 yeast proteins located in the cytosol and sub-cellular compartments of yeast cells using a highly quantitative mass spectrometry based thiol trapping technique, called OxICAT. The identified proteins can be clustered in four groups: 1) proteins, whose cysteine residues are oxidation resistant; 2) proteins with structurally or functionally important cysteine modifications 3) proteins with highly oxidation-sensitive active site cysteines, which are partially oxidized in exponentially growing yeast cells due to their exquisite sensitivity towards low amounts of ROS; 4) proteins that are reduced in exponentially growing cells but harbor redox-sensitive cysteine(s) that affect the catalytic function of the protein during oxidative stress. These oxidative stress sensitive proteins were identified by exposure of yeast cells to sublethal concentrations of H2O2 or superoxide. It was shown that the major targets of peroxide- and superoxide-mediated stress in the cell are proteins involved in translation, glycolysis, TCA cycle and amino acid biosynthesis. These targets indicate that cells rapidly redirect the metabolic flux and energy towards the pentose phosphate pathway in an attempt to ensure the production of the reducing equivalent NADPH to counterattack oxidative stress. These results reveal that the quantitative assessment of a protein’s oxidation state is a valuable tool to identify catalytically active and redox-sensitive cysteine residues. The OxICAT technology was then used to precisely determine extent and onset of oxidative stress in chronologically aging S. cerevisiae cells by utilizing the redox status of proteins as physiological read-out. I found that chronological aging yeast cells undergo a global collapse of the cellular redox homeostasis, which precedes cell death. The onset of this collapse appears to correlate with the yeast life span, as caloric restriction increases the life span and delays the redox collapse. These results suggest that maintenance of the redox balance might contribute to the life expanding benefits of regulating the caloric intake of yeast. Clustering analysis of all oxidatively modified proteins in chronological aging yeast revealed a subset of proteins whose oxidative thiol modifications significantly precede the general redox collapse. Oxidation of these early target proteins, which most likely results in a loss of their activity, might contribute to or even cause the observed loss of redox homeostasis (i.e., thioredoxin reductase) in chronologically aging yeast. These studies in aging yeast expand our understanding how changes in redox homeostasis affect the life span of yeast cells and confirm the importance of oxidative thiol modifications as key posttranslational modifications in pro- and eukaryotic organisms.
Die Knochenhomöostase erfolgt durch das Zusammenspiel mehrerer Zelltypen. Während die Osteoblasten für den Knochenaufbau verantwortlich sind, resorbieren die Osteoklasten Knochengewebe. Beide Vorgänge werden durch die Osteozyten streng reguliert. Eine Störung im strikt regulierten Gleichgewicht zwischen Knochenabbau und Knochenaufbau kann daher zu Knochenkrankheiten wie Osteoporose führen. Auf molekularer Ebene erfolgt die Kommunikation zwischen den einzelnen Zelltypen über zwei wichtige Signalwege, den der „Bone Morphogenetic Protein“-Superfamilie (BMPs) und den der Wnt-Proteine. Die Signalübertragung wird hierbei durch sekretierte Faktoren induziert, die an Rezeptoren auf der Zelloberfläche binden. Deren Aktivierung führt zu einem intrazellulären Signal, welches letztlich die Expression von Zielgenen reguliert. Beide Signalwege werden auf mehreren Ebenen, extrazellulär, membranständig und intrazellulär reguliert. Das 2003 identifizierte Sclerostin ist ein Vertreter der extrazellulären Regulatorproteine und wurde aufgrund seiner Zugehörigkeit zur DAN-Familie zunächst fälschlicherweise als direkter Inhibitor des BMP-Signalwegs eingestuft. Mittlerweile wird allerdings davon ausgegangen, dass Sclerostin den Wnt-Signalweg negativ reguliert, indem es die Wnt Ko-Rezeptoren LRP5 und LRP6 bindet, die beide zu der Familie der „Low-density lipoprotein receptors“ gehören. Über den molekularen Inhibitionsmechanismus von Sclerostin war jedoch zum Startpunkt dieser Dissertationsarbeit wenig bekannt. Daher wurde Sclerostin im Rahmen dieser Arbeit biophysikalisch und biochemisch charakterisiert. Die Aufklärung mittels NMR-Spektroskopie ergab für Sclerostin eine Struktur, die sich in drei Regionen gliedert: den Cystinknoten, sowie einen „Loop“-Bereich und die Fingerregion. Vom zentralen Cystinknoten gehen drei Peptid-Schleifen in zwei entgegengesetzte Richtungen aus. Schleife eins und drei bilden eine definierte ß-Faltblattstruktur und ähneln zwei Fingern einer Hand. Die zweite Schleife, welche vom Cystinknoten isoliert in die entgegengesetzte Richtung verläuft („Loop“), ist wie die beiden langen N- und C-Termini flexibel und unstrukturiert. Die in Zusammenarbeit mit der Firma AbD-Serotec entstandenen Fab-Fragmente ermöglichten die Bestimmung des Bindeepitops der Sclerostin/LRP5-Interaktion im Bereich der unstrukturierten dritten Schleife von Sclerostin. Die Struktur von Sclerostin und die Identifikation des Bindeepitops auf Sclerostinseite geben nun erste Einblicke in den molekularen Mechanismus der Sclerostin/LRP5-Interaktion. Diese Kenntnis kann für die Entwicklung von Kleinmolekülinhibitoren mittels rationalem Drugdesign genutzt werden, welche, wie auch der in Kooperation entwickelte die Sclerostinaktivität neutralisierende Antikörper AbD09097, hochinteressante Ansätze für neuartige anabole Therapien von Krankheiten mit Knochenschwund darstellen.
This study focuses on phosphoantigen specific Vg9Vd2 T cells which only exist in human and non-human primates. This population accounts for 1%-5% of peripheral blood T-lymphocytes but their frequency can rise to 50% of total blood T cells upon infection. Vg9Vd2 T cells can be activated by nonpeptide compounds with critical phosphate moieties which are termed as phosphoantigens. These include isopentenyl pyrophosphate (IPP), a key compound of isoprenoid synthesis in all organisms, and (E)-4-Hydroxy-3-methyl-but-2-enyl pyrophosphate (HMBPP), a direct precursor of IPP in DOXP pathway which only exist in eubacteria, plants, apicomplexaen parasites. Its activity as phosphoantigen is at least 1000 fold higher than that of IPP. However, direct structural evidence of phosphoantigen binding to the TCR is missing so far. Moreover, Vg9Vd2 T cells have potent anti-tumor activity e.g. against the B-cell lymphoma Daudi, whose Vg9Vd2 T cell activating properties have been suggested to result from sensing of abnormal intracellular IPP levels by the Vg9Vd2 TCR or Vg9Vd2 TCR binding to other postulated ligands such as an ectopically expressed F1-ATPase or UL-16 binding protein 4 (ULBP4). Aminobisphosphonates and alkymines were hypothesized to activate Vg9Vd2 T cells indirectly by inhibiting the IPP consuming enzyme farnysyl pyrophosphates synthesis (FPPS) although off target effects of these drugs or a direct interaction with the Vg9Vd2 TCR could not be excluded. This thesis presents new approaches for the mechanistic analysis of Vg9Vd2 T cell activation. By employing retroviral transduction of FPPS specific shRNA, it shows that specific shRNA reduces expression of FPPS and is sufficient to convert hematopoietic and non-hematopoietic tumor cell lines into Vg9Vd2 T cell activators. FPPS knockdown cells activated Vg9Vd2 T cells as measured by increased levels of CD69 and CD107a, kill of FPPS knockdown cells and induction of IFN-γ secretion. The IPP-synthesis-inhibiting drug mevastatin reduced Vg9Vd2 T cell activation by FPPS knockdown cells or aminobisphosphonate treated cells but not activation by the phosphoantigen bromohydrin pyrophosphate (BrHPP). A reduced growth of the FPPS knockdown cells has not been observed which is different to what has been reported for aminobisphosphonate treated cells. Finally, the human B-cell lymphoma RAJI has been transduced with Tetracyclin-inducible FPPS specific shRNA and proven to gain and loose the capacity to activate Vg9Vd2 TCR transductants upon doxycylin provision or removal. Another approach for the analysis of Vg9Vd2 T cell activation is Vg9Vd2 TCR transduced mouse cell lines with specificity for phosphoantigens. In contrast to the previously used Vg9Vd2 TCR transduced Jurkat cells, these cells do not present phosphoantigens, and are therefore specially suited for analysis of phosphoantigen presentation. The response of the new TCR transductants to presumed Vg9Vd2 TCR ligands/activators such as phosphoantigens, aminobisphosphonates or FPPS knockdown cells, depended strongly on the expression of a rat/mouse CD28 molecule by the transductants and its ligation by the (CD80) counter receptor on the ligand-presenting cell. The response is likely to reflect recognition of cognate Vg9Vd2 TCR antigens since mutations in the TCR-δ chain CDR2 and 3 abolished this response but activation by TCR or CD3 specific antibodies. A major difference between TCR transductants and primary gd T cells, was the lacking response of TCR transductants to Daudi or IPP. In addition their sensitivity to other soluble phosphoantigens was about 100 fold weaker than that of primary cells, stimulation of both cell type to CD80 expressing FPPS knock down or aminobisphosphonates was similar. Finally, the transductants have also been used to analyze effects of over-expression or knockdown of enzymes of isoprenoid synthesis such as 3-hydroxy-3-methyl-glutaryl-CoA reductase (HMG-CoA reductase or HMGR), mevalonate-5-pyrophosphate decarboxylase (MVD), isopentenyl pyrophosphate isomerase (IDI), geranyl-geranyl pyrophosphate synthase (GGPPS) but no clear effects have been found. In conclusion, this thesis supports the concept of Vg9Vd2 T cells being sensors of a dysregulated isoprenoid metabolism and established new tools to study ligand recognition and TCR mediated activation of this T cell population. These tools will be most useful to address following questions: 1) How does the dysregulation of isoprenoid metabolism affect tumor growth? 2) What is the correlation between the modulation of IPP levels and the Vg9Vd2 TCR binding or expression of other postulated ligands? 3) Are there any mevalonate pathway enzymes other than FPPS and HMGR, which play an important role in Vg9Vd2 T cells activation? 4) What is/are the putative phosphoantigen-presenting molecule(s)?
Platelets play a central role in thrombosis, hemostasis, and inflammation. Here, we show that activated platelets release inorganic polyphosphate (polyP), a polymer of 60- 100 phosphate residues that directly bound to and activated the plasma protease factor XII. PolyP-driven factor XII-activation triggered release of the inflammatory mediator bradykinin by plasma kallikrein-mediated kininogen processing. PolyP increased vascular permeability and induced fluid extravasation in skin microvessels of mice. Mice deficient in factor XII or bradykinin receptors were resistant to polyP-induced leakage. PolyP initiated clotting of plasma via the contact pathway. Ablation of intrinsic coagulation pathway proteases factor XII and factor XI protected mice from polyPtriggered lethal pulmonary embolism. Targeting polyP with phosphatases interfered with procoagulant activity of activated platelets and blocked platelet-induced thrombosis in mice. Infusion of polyP restored defective plasma clotting of Hermansky- Pudlak Syndrome patients, which lack platelet polyP. The data identify polyP as a new class of mediator having fundamental roles in platelet-driven proinflammatory and procoagulant disorders.
Aim of this thesis was to study the contribution of the hosts immune system during tumor regression. A wild-type rejection model was studied in which tumor regression is mediated through an adaptive, T cell host response (Research article 1). Additionally, the relationship between VACV infection and cancer rejection was assessed by applying organism-specific microarray platforms to infected and non-infected xenografts. It could be shown that tumor rejection in this nude mouse model was orchestrated solely by the hosts innate immune system without help of the adaptive immunity. In a third study the inflammatory baseline status of 75 human cancer cell lines was tested in vitro which was correlated with the susceptibility to VACV and Adenovirus 5 (Ad5) replication of the respective cell line (Manuscript for Research article 3). Although xenografts by themselves lack the ability to signal danger and do not provide sufficient proinflammatory signals to induce acute inflammation, the presence of viral replication in the oncolytic xenograft model provides the "tissue-specific trigger" that activates the immune response and in concordance with the hypothesis, the ICR is activated when chronic inflammation is switched into an acute one. Thus, in conditions in which a switch from a chronic to an acute inflammatory process can be induced by other factors like the immune-stimulation induced by the presence of a virus in the target tissue, adaptive immune responses may not be necessary and immune-mediated rejection can occur without the assistance of T or B cells. However, in the regression study using neu expressing MMC in absence of a stimulus such as a virus and infected cancer cells thereafter, adaptive immunity is needed to provoke the switch into an acute inflammation and initiate tissue rejection. Taken together, this work is supportive of the hypothesis that the mechanisms prompting TSD differ among immune pathologies but the effect phase converges and central molecules can be detected over and over every time TSD occurs. It could be shown that in presence of a trigger such as infection with VACV and functional danger signaling pathways of the infected tumor cells, innate immunity is sufficient to orchestrate rejection of manifested tumors.
