Plants exposed to herbivory may defend themselves by attracting the “enemies of their enemies”, a phenomenon called induced indirect defense (IID). In this process, the de novo production and emission of volatile organic compounds (VOC) by the affected plant is activated via a jasmonic acid (JA) dependent signaling cascade. VOC can be very specific for the inducing herbivore as well as for the emitting plant. Carnivores as predatory mites and parasitoid wasps use these substances as prey- or host-finding cues. If the herbivore is parasitized successfully, its development is slowed and thus the damage of the plant is decreased. Additional abiotic stress may modulate the plant’s ability to produce and/or emit herbivore induced VOC. Ultraviolet (UV) radiation can have multiple physiological effects on plants, amongst others the activation of the expression of genes that are also activated during anti-herbivore defense. To investigate UV effects, foils with different UV transmittance were used to manipulate ambient solar radiation. One foil was permeable for the whole solar spectrum including UV radiation whereas the other excluded radiation below a wavelength of 400 nm. Soybean exposed to UV increased concentrations of isorhamnetin- and quercetin-based flavonoids as effective photo-protective compounds in the leaves and showed a reduced growth compared to plants exposed to ambient radiation lacking UV. The altered chemical composition of the leaves had no effect on food choice and performance of herbivorous Spodoptera frugiperda larvae. Photo-protection by flavonoids seems to be efficient to prevent further UV effects on IID as plants of both treatments emitted the same blend of induced VOC and hence females of the parasitoid Cotesia marginiventris did not prefer plants from on of the treatments in the olfactometer. Nitrogen is one important macronutrient for all trophic levels and thus deficiency of this nutrient was expected to affect IID of soybean profoundly. To manipulate N availability for soybean plants hydroponic culture was used. One treatment was cultured in a standard hydroponic solution whereas in the N deficiency treatment in the solution all salts containing N were replaced with N-free salts. In N deficient plants root biomass was increased to allow the plant to forage more efficiently for the nutrient. Despite this morphological adaptation, photosynthetic efficiency as well as leaf N and soluble protein content were reduced significantly in N deficient soybean. The N deficiency was passed on to the third trophic level as herbivores fed with the affected leaves had a reduced body N content on her part and showed a decreased growth but no feeding preference for the superior food. Parasitoids reared in such N deficient herbivores had significant lower pupal weight compared to parasitoids reared in hosts fed with fully fertilized soybean. N deficient plants emitted a quantitatively altered herbivore induced blend. The two terpenes β-Bergamotene and (E,E)-α-Farnesene were emitted in higher amounts whereas (Z)-3-Hexenyl-α-methylbutyrate was emitted in significantly lower amount. Despite this quantitatively modified VOC blend the parasitoids host-searching behavior was not affected. Heavy metals (HM) are proposed to affect various biochemical pathways in plants including defense pathways by production of reactive oxygen species (ROS) in the tissue. The ROS on its part may affect production and release of endogenous JA, an important messenger in defense signaling. In this study maize plants were grown hydroponically and exposed to different increased concentrations of copper and cadmium. Maize seems to be able to exclude the excess HM from the leaves because the HM were found mainly in the roots and only to a minor degree in the shoots of the plants. Despite this exclusion the HM significantly affected uptake of other metal ions into the plant. The excess of the HM in combination with the attenuated uptake of other ions led to a reduced growth of roots and shoots as well as to reduced photosynthetic efficiency. Thus the nutritional value of the plants for the herbivore was lowered either by direct toxic effects of the HM or indirectly by altering plant chemical composition. S. frugiperda larvae fed with leaves exposed to high HM concentrations showed a significantly reduced growth but they did prefer neither control nor HM treated plants in a food-choice assay. Cu had a transient priming effect on JA as pre-exposure to a high excess of Cu led to higher amounts of herbivore induced JA compared to control plants exposed only to standard concentration of Cu. As anticipated the increased JA was followed by an increase in herbivore induced VOC in high-Cu treated plants caused by a increase of the green leaf volatiles (E)-3-Hexenal, (Z)-3-Hexenol and (Z)-3-Hexenylacetat and the terpenes Linalool, (E)-α-Bergamotene, (E)-β-Farnesene, and β-Sesquiphellandrene. Despite these profound changes in herbivore induced VOC the parasitoids host searching behavior was not affected. As described, the abiotic stresses UV, N deficiency and excess HM affected the morphology and physiology of soybean and maize, the performance of the herbivore S. frugiperda and even the performance of the parasitoid C. marginiventris. However the host searching behavior of the parasitoid was not affected even if the herbivore induced VOC blend was altered. Thus parasitoids seem to be a very reliable defender for plants and IID a very robust way of herbivore defense.
Arabidopsis thaliana (A.th.) mesophyll cells play a pivotal role in the regulation of the drought stress response. The signaling & transport components involved in drought stress regulation within lipid rafts of the plasma membrane were investigated by DRM isolation from highly purified plasma membranes. Detergent treatment with Brij-98 and Triton X-100 resulted in a total of 246 DRM proteins which were identified by nano HPLC-MS/MS. The majority of these proteins could be isolated by Triton X-100 treatment (78.5 %) which remains the ”golden” standard for the isolation of DRMs. Comparing in-gel and in-solution digestion approaches disclosed additional protein identifications for each method but the in-gel approach clearly delivered the majority of the identified proteins (81.8 %). Functionally, a clear bias on signaling proteins was visible – almost 1/3 of the detected DRM proteins belonged to the group of kinases, phosphatases and other signaling proteins. Especially leucine-rich repeat receptor-like protein kinases and calcium-dependent protein kinases were present in Brij-98 & Triton X-100 DRMs, for instance the calcium-dependent protein kinase CPK21. Another prominent member of DRMs was the protein phosphatase 2C 56, ABI1, which is a key regulator of the ABA-mediated drought stress response in A.th. The lipid raft localization of the identified DRM proteins was confirmed by sterol-depletion with the chemical drug MCD. Proteins which depend upon a sterol-rich environment are depleted from DRMs by MCD application. Especially signaling proteins exhibited a strong sterol-dependency. They represented the vast majority (41.5 %) among the Triton X-100 DRM proteins which were no longer detected following MCD treatment. AtRem 1.2 & 1.3 could be shown to be sterol-dependent in mesophyll cells as well as two CPKs (CPK10 & CPK21) and the protein phosphatase ABI1. AtRem 1.2 & 1.3 could be proven to represent ideal plant lipid raft marker proteins due to their strong presence in Triton X-100 DRMs and dependency upon a sterol-rich environment. When fluorescence labeled AtRem 1.2 & 1.3 were transiently expressed in A.th. leaves, they localized to small, patchy structures at the plasma membrane. CPK21 was an intrinsic member of Triton X-100 DRMs and displayed extreme susceptibility to sterol-depletion by MCD in immunological and proteomic assays. Calcium-dependent protein kinases (CPKs) have already been studied to be involved in drought stress regulation, for instance at the regulation of S-type anion channels in guard cells. Hence, further transient expression studies with the anion channel SLAH3, protein kinase CPK21 and its counterpart, protein phosphatase ABI1 were performed in Nicotiana benthamiana. Transient co-expression of CPK21 and the anion channel SLAH3, a highly mesophyll- specific homologue of the guard cell anion channel SLAC1, resulted in a combined, sterol-dependent localization of both proteins in DRMs. Supplementary co-expression of the counterpart protein phosphatase ABI1 induced dislocation of SLAH3 from DRMs, probably by inactivation of the protein kinase CPK21. CPK21 is known to regulate the anion channel SLAH3 by phosphorylation. ABI1 dephosphorylates CPK21 thus leading to deactivation and dislocation of SLAH3 from DRMs. All this regulative events are taking place in DRMs of A.th. mesophyll cells. This study presents the first evidence for a lipid raft-resident protein complex combining signaling and transport functions in A.th. Future perspectives for lipid raft research might target investigations on the lipid raft localization of candidate DRM proteins under presence of abiotic and biotic stress factors. For instance, which alterations in the DRM protein composition are detectable upon exogenous application of the plant hormone ABA? Quantitative proteomics approaches will surely increase our knowledge of the post-transcriptional regulation of gene activity under drought stress conditions.
Jasmonsäure und verwandte Oxylipine wurden bisher als Substanzen, die an der Regulation von Initialisierung und Progression der Blattseneszenz beteiligt sein sollen, kontrovers diskutiert. Bisherige Studien haben sich dabei auf die exogene Applikation von Jasmonaten oder die Messung endogener Spiegel beschränkt. Um die Funktion von Jasmonaten in der Seneszenz-Regulation zu klären, wurden in dieser Arbeit die Profile freier und membranveresterter Oxylipine sowie die Auswirkungen verminderter Oxylipinbildung während der natürlichen Seneszenz und Seneszenz-ähnlicher Prozesse induziert durch Dunkel- und Sorbitol-Inkubation in Blättern von Arabidopsis thaliana untersucht. Jasmonsäure sowie freie 12-Oxo-Phytodiensäure steigen während dieser drei Prozesse an, mit dem stärksten Anstieg von Jasmonsäure nach Dunkelinkubation. Eine deutliche Akkumulation membranveresterter Oxylipine (Arabidopside) konnte lediglich nach Flottierung auf Sorbitol festgestellt werden. Die Mengen an plastidären Mono- und Digalaktosyl-Diacylglycerolen verringerten sich jedoch während der Behandlungen bzw. im Verlauf der Alterung. Zur Untersuchung möglicher Funktionen ansteigender Jasmonat-Konzentrationen wurden Lipoxygenase 2 RNAi-Pflanzen konstruiert, welche basal Jasmonsäure und 12-Oxo-Phytodiensäure produzieren können, jedoch keinen Anstieg während Seneszenz- bzw. Stress-Prozessen zeigen. Die Gehalte an Chlorophyll und Membranlipiden sowie die Genexpression entwicklungsspezifischer Seneszenzmarker waren während der natürlichen und der dunkelinduzierten Seneszenz in diesen Pflanzen nicht verändert. Dies legt nahe, dass diese Oxylipine im Verhältnis zu anderen endogenen Faktoren keine bzw. nur geringe Wirkungen auf die Seneszenz-Progression haben. Aus den gemachten Beobachtungen kann vielmehr geschlossen werden, dass bei diesen Prozessen die Akkumulation von Jasmonaten eher die Folge eines veränderten Lipid-Metabolismus als ein Auslöser der Seneszenz ist. Im Gegensatz dazu zeigen die Lipoxygenase 2 RNAi-Linien eine verlangsamte Seneszenz nach Sorbitol-Behandlung. Ähnlich verhält sich die Allenoxid-Synthase Mutante dde2-2, die zwar 13-Lipoxygenase-Produkte aber keine Jasmonate bilden kann. Dies bedeutet, dass die Jasmonate und nicht andere 13-Lipoxygenase-Produkte für die Seneszenz-ähnlichen Symptome unter diesen Bedingungen verantwortlich sind. Dabei stellt die Sorbitol-induzierte Seneszenz einen Stress-Prozess dar, der sich in vielen Punkten von der natürlichen Seneszenz unterscheidet aber große Ähnlichkeiten zur Seneszenz-Induktion nach exogener Jasmonat-Applikation aufweist. Lipoxygenase 2 ist also durch die Bereitstellung von Oxylipinen weniger in Entwicklungs- als vielmehr in Stress-Prozesse involviert.
Inoculation with plant pathogens induces a diverse range of plant responses which potentially contribute to disease resistance or susceptibility. Plant responses occuring in consequence of pathogen infection include activation of classical defence pathways and changes in metabolic activity. The main defence route against hemibiotrophic bacterial pathogens such as Pseudomonas syringae is based on the phytohormone salicylic acid (SA). SA-mediated responses are strictly regulated and have also been shown to depend on external factors, e.g. the presence of light. A major goal of this work was to provide a better understanding of the light dependency of plant defence responses mediated through SA. The second part of the project focussed on the influence of plant sterols on plant resistance. I analyzed leaf lipid composition and found that accumulation of the phytosterol stigmasterol in leaves and in isolated (plasma) membranes is a significant plant metabolic process occurring upon pathogen infection.
In the humid tropics of SE Asia there are some 14 myrmecophytic species of the pioneer tree genus Macaranga (Euphorbiaceae). In Peninsular Malaysia a close association exists between the trees and the small, non-stinging myrmicine Crema togas ter borneensis. These ants feed mainly on food bodies provided by the plants and have their colonies inside the hollow intemodes. In a ten months field study we were able to demonstrate for four Macaranga species (M. triloba, M. hypoleuca, M. hosei, M. hulletti) that host plants also benefit considerably from ant-occupation. Ants do not contribute to the nutrient demands of their host plant, they do, however, protect it against herbivores and plant competition. Cleaning behaviour of the ants results in the removal of potential herbivores already in their earliest developmental stages. Strong aggressiveness and a mass recruiting system enable the ants to defend the host plant against many herbivorous insects. This results in a significant decrease in leaf damage due to herbivores on ant-occupied compared to ant-free myrmecophytes as well as compared to non-myrmecophytic Macaranga species. Most important is the ants' defense of the host plant against plant competitors, especially vines, which are abundant in the well-lit pioneer habitats where Macaranga grows. Ants bite off any foreign plant part coming into contact with their host plant. Both ant-free myrmecophytes and non-myrmecophytic Macaranga species had a significantly higher incidence of vine growth than specimens with active ant colonies. This may be a factor of considerable importance allowing Macaranga plants to grow at sites of strongest competition.
The woody climber Millettia niuewenhuisii (Fabaceae) and the shrub Myrmeconauclea strigosa (Rubiaceae) in Sabah, Borneo are associated with ants. The hollow stems of Millettia nieuwenhuisii are regularly inhabited by an aggressive Cladomyrma sp., which keeps pseudococcids inside the stem. On Myrmeconauclea strigosa the ants live in hollow internodal swellings near the end of the branches. In this plant many different ant species use the nesting space in an opportunistic manner.

Ficus obscura var. borneensis is a true myrmecophyte. It spontaneously forms cavities (domatia) in parts of its twigs which open by slits, These occur in the internodes and are usually not swollen. The domatia are inhabited by a variety of non-specific tree-living ants including Crematogaster spp., Cataulacus sp., Tetramorium sp., Cardio condyla sp. and Camponotus sp.. Additionally the plant providL a su~ar-containing secretion from extrafloral nectaries on the lower surfaces of the leaves. Examination of herbarium specimens of 37 other South-east Asian Ficus species did not reveal a single specimen with domatia.
An der pflanzlichen Plasmamembran geschieht die erste Wahrnehmung von mikrobiellen Molekülen, die MAMPs genannt werden. MAMP/PAMP Rezeptoren leiten frühe Abwehrantworten, wie die Produktion von reaktiven Sauerstoffspezies (ROS), externe Alkalisierung oder Ethylen, ein. Die Arabidopsis FLS2 rezeptorartige Kinase (RLK) stellt einen plasmamembran-lokalisierten MAMP Rezeptor dar, der über die Detektion des Flagellum von Pseudomonas species, eine basale Immunität in Arabidopsis thaliana vermittelt. Flg22, der kürzeste aktive Teil des bakteriellen Flagellins besteht aus 22 Aminosäuren und ist der bestuntersuchte bakterielle Elizitor. In der vorliegenden Arbeit zeigen wir eine starke Beteiligung von Ionenflüssen in der Initiationsphase der basalen Immunität. Unsere Messungen an intakten Arabidopsis Pflanzen und Pflanzengeweben sind in höchstem Masse reproduzierbar und öffnen eine neue Sicht, über die Natur von Ionentransporten in der Pflanzen - Mikroben Interaktion. Als Antwort auf die Applikation von flg22, haben wir nach einer Verzögerungsphase von etwa 2 Minuten eine transiente, dosis-abhängige Depolarisation (EC50=0,2 nM) in Mesophyll- und Wurzelhaarzellen von A. thaliana messen können. Das um 2 Aminsäuren kürzere Peptid flg22 Δ2 oder das Flagellin anderer Bakterien (Agrobacterium or Azospirillum) führten zu keiner Membrandepolarisation. Ebenso konnten keine Membranspannungsänderungen in dem Arabidopsis Ökotypen Ws-0, dem der funktionelle FLS2 Rezeptor fehlt, detektiert werden. Die Komplementation von Ws-0 Pflanzen mit dem intakten FLS2 Rezeptorgen rief eine Resensibilisierung für flg22 hervor. Mit dem EF-Tu Elizitor Peptid aus E.coli, welches durch den Arabidopsis MAMP Rezeptor EFR detektiert wird, wurden ähnliche Ergebnisse erzielt. Auf der Basis von Aequorin wurden Kalzium-induzierte Lumineszenzmessungen durchgeführt, in denen ein transienter Anstieg der zytosolischen Kalziumkonzentration als Antwort auf die Applikation von flg22 gemessen werden konnte. Dosis-Abhängigkeitsmessungen von flg22 und [Ca2+]cyt wiesen zwei unterschiedliche EC50 Werte, von 43 ± 2 pM und 67 ± 42 nM, auf. Möglicherweise wird auf zwei verschiedene Kalziumpools zugegriffen oder es werden zwei verschiedene Kalziumleitfähigkeiten aktiviert. Die Ionenkanalaktivierung und folgende Depolarisation benötigt die aktive Rezeptorkinase. In bak1-4 Arabidopsis Pflanzen, in denen die FLS2 Untereinheit BAK1 – eine weitverbreitete RLK, die auch mit dem Brassinosteroid Rezeptor assoziiert ist – fehlt, konnte keine Depolarisation als Antwort auf flg22 gemessen werden. Arabidopsis Mesophyllzellen zeigten die typische Alkalisierung des Apoplasten als Antwort auf flg22. Nicht-invasive MIFETM Experimente mit Ionen-selektiven Elektroden ergaben, dass der pH-Anstieg durch einen Einstrom von Protonen hervorgerufen wurde. Zusätzlich wurde ein Ausstrom von Chlorid und Kalium aufgezeichnet. Ähnlich wie das Kalziumsignal waren alle detektierten Ionenströme von transienter Natur. Im zweiten Ansatz wurden Membranpotential-Messungen durchgeführt, während in der externen Lösung die Konzentrationen von Protonen, Kalzium, Kalium oder Anionen variiert wurden. Nur eine Änderung des Anionengradienten hatte einen entscheidenden Einfluss auf die flg22-induzierte Depolarisation, was die Wichtigkeit der Anionenkanalaktivierung unterstreicht. Exudat Analysen ergaben, dass Nitrat das bevorzugt transportierte Ion ist. Unter zahlreichen getesteten Ionenkanalblockern erwies sich lediglich Lanthan als effektiver Blocker des flg22-induzierten zytosolichen Kalziumanstiegs, des Protoneneinstroms und der Membrandepolarisation. Da Lanthan bekanntlich unspezifische Kationenkanäle blockt, kann man an diesem Punkt davon ausgehen, dass Kalzium-aktivierte Anionenkanäle die Membrandepolarisation vermitteln und darauf eine Aktivierung von auswärtsgerichteten Kaliumkanälen folgt. Zukünftige Studien mit Doppelläufigen-Mikroelektroden Spannungsklemmexperimenten oder externen ionenselektiven Elektroden an intakten Schliesszellen werden helfen weitere Informationen über die Natur der Ionenkanäle in der basalen Immunität oder generell in der Pflanzen-Mikroben Interaktion zu erhalten. Über die elektrophysiologische Charakterisierung der multiplen Ionenströme in der basalen Immunität hinaus, ist natürlich der nächste wichtige Schritt das oder die Gene zu finden, die für die Ionenkanäle oder Transporter kodieren, die durch nicht nekrotisierende Elizitoren wie flg22 in der basalen Immunantwort in Pflanzen aktiviert werden.
Regulation of pathogen-inducible volatile compounds in Arabidopsis and their role in plant defense

Plants are constantly attacked by pathogenic microbes. As a result, they have evolved a plethora of constitutive and inducible defense responses to defend against attempted pathogen infection. Although volatile organic compounds have been implicated in plant defense, direct evidence of their function in plant resistance is still lacking. I have examined the role of VOCs in Arabidopsis defense against the hemibiotrophic bacterial pathogen Pseudomonas syringae pv. maculicola. The obtained results show that the vegetative parts of Arabidopsis produces and emits the volatile phenylpropanoid MeSA and three kinds of terpenoids, (E,E)-4,8,12-trimethyltrideca-1,3,7,11-tetraene (TMTT), alpha-ionon and beta-farnesen, upon avirulent and virulent P. syringae inoculation. Whereas the most abundant volatiles, MeSA and TMTT, are already produced at early stages of infection in the compatible and incompatible interaction, enhanced emission of alpha-ionon and beta-farnesen can only be detected in later stages of the compatible interaction. It was revealed that pathogen-induced synthesis of TMTT in Arabidopsis requires the JA signaling pathway but occurs independently of SA defense signaling. Similarly, the production of MeSA is dependent on JA signaling but not on the SA defense signaling pathway. Furthermore, production of MeSA is dependent on the function of ISOCHORISMATE SYNTHASE1, which produces its precursor SA. Upon inoculation with avirulent P. syringae, endogenously produced JA activates the JA signalling pathway to mediate MeSA and TMTT synthesis. By contrast, in the compatible Arabidopsis-Psm interaction, production of MeSA predominantly depends on the P. syringea the virulence factor coronatine, which activates JA downstream signaling. To learn more about the role of inducible VOCs in plant defense responses, I have identified an Arabidopsis T-DNA insertions line with a defect in the TERPENE SYNTHASE4 (TPS4) gene. Emission profiles from this mutant revealed that the induced production of TMTT but not of alpha-ionone, beta-farnesene or MeSA are abolished, demonstrating that TPS4 specifically regulates the P. syringae-induced synthesis of TMTT in Arabidopsis. The lack of TMTT in tps4 mutants, however, does not affect plant defense responses and resistance induction against P. syringae. This excludes a role of the terpenoid as an effective phytoalexin in Arabidopsis leaves against the bacterial pathogen. Moreover, tps4 mutant plants are still able to mount a SAR response, excluding a signaling function of TMTT during SAR. An important aim of our studies was to address the defensive role of MeSA, the major VOC emitted from P. syringae-inoculated Arabidopsis leaves. MeSA has been recently proposed as a critical long distance signal in the development of SAR. I found that two independent T-DNA insertions lines with defects in expression of the pathogen-inducible SA methyl transferase gene BSMT1 are completely devoid of pathogen-induced production of MeSA. However, bsmt1 mutant plants are capable to increase the level of SA in systemic, non-infected leaves of Arabodopsis and develop SAR like wild-type plants upon local P. syringae-inoculation. Thus, MeSA does not function as a critical SAR signal in Arabidopsis. Further experiments showed that SA accumulation in distant leaves occurs due to de novo synthesis through isochorismate synthase. In addition, we also ruled out a critical defensive role of MeSA at inoculation sites, because bsmt1 mutants are able to build up SA-dependent defense responses and local resistance in a wild-type-like manner. The conversion of SA to MeSA and subsequently emission of MeSA from the plant might help the plant to detoxify an excess of SA. This process is regulated by the JA pathway and might be one means to mediate negative crosstalk between JA and SA signaling. Moreover, the COR-triggered conversion of SA to MeSA and emission of the volatile methyl ester could be a way by which virulent P. syringae is able to attenuate the SA-defense pathway.

Ernst Koch (1843-1926), Gymnasiallehrer in Meiningen (Südthüringen), hat sich neben seinen Forschungen zu Geschichte und Landeskunde des Herzogtums Meiningen in den Jahren zwischen 1895 und 1906 auch intensiv mit der Flora von Südthüringen und angrenzender Gebiete Unterfrankens beschäftigt. Seine floristischen Aktivitäten werden dargestellt und seine Beziehungen zum Thüringischen Botanischen Verein anhand des noch erhaltenen Briefwechsels beschrieben.
Invertasen sind Schlüsselenzyme in der Kohlenhydratverteilung und haben möglicherweise auch während einer Pathogeninfektion eine zentrale Bedeutung. In vorliegender Arbeit wurde zunächst die Regulation verschiedener Stoffwechselwege in Arabidopsis thaliana nach Infektionen mit einem virulenten oder avirulenten Stamm von Pseudomonas syringae untersucht. Mit Hilfe der Chlorophyllfluoreszenz-Bildgebung konnten räumliche und zeitliche Veränderungen der Photosynthese verfolgt werden. Verschiedene Parameter waren unterschiedlich reguliert. In beiden Interaktionen waren Effekte nur lokal um die Infektionsstellen erkennbar und qualitativ ähnlich. Unterschiede waren im zeitlichen Eintreten und Verlauf sichtbar. Die Methode schien geeignet für die sensitive, nicht-invasive Pathogenfrüherkennung vor dem Auftreten sichtbarer Symptome. Die Regulation verschiedener Gene innerhalb von Source-Sink-Übergängen und die Aktivität von Invertasen war in den beiden Interaktionen qualitativ unterschiedlich. Die Infektion mit virulenten Bakterien resultierte in einer Repression photosynthetischer Gene. Die Aktivität vakuolärer Invertasen stieg vorübergehend nach Infektion mit virulenten Bakterien an, während sie nach Infektion mit avirulenten Bakterien sank. Die Aktivität extrazellulärer Invertasen war in beiden Interaktionen reprimiert. Die erfolgreiche Generierung verschiedener Bakterienstämme von P. syringae, die das grün fluoreszierende Protein exprimieren, kann bei der weiteren Charakterisierung von Pflanze-Pathogen-Interaktionen helfen. Die Regulation von Invertasen erfolgt auf transkriptioneller und posttranslationaler Ebene. Um die Funktion von Invertasen in Pflanze-Pathogen-Interaktionen zu verstehen, wurde zunächst die Regulation von Invertasen durch endogene proteinogene Invertaseinhibitoren untersucht. In Übereinstimmung mit in silico Expressionsdaten konnte durch Untersuchung von Reportergenlinien und in Northern Blot Analysen eine starke Expression von Invertaseinhibitoren in Blättern von A. thaliana festgestellt werden. Nach Applikation biotischer und abiotischer Stressfaktoren wurde diese Expression nahezu vollständig reprimiert. Die indirekte Bestimmung der Invertaseinhibitoraktivität durch Messung der Invertaseaktivität in Mischextrakten zeigte, dass diese nach einer Pathogeninfektion vollständig reprimiert war. In funktionellen Ansätzen wurden transgene Pflanzen generiert, die Invertaseinhibitoren unter Kontrolle induzierbarer Promotoren exprimieren. Die Induktion der Invertaseinhibitorexpression änderte die Sensitivität gegenüber verschiedenen Pathogenen nicht signifikant. In einem pharmakologischen Ansatz wurde der chemische Inhibitor Acarbose zur Hemmung der Invertaseaktivität in A. thaliana verwendet. Eine Behandlung von Blättern bei gleichzeitiger Infektion mit Bakterien verursachte eine erhöhte Sensitivität der Pflanzen gegenüber der Infektion, eine stärkere Repression verschiedener Chlorophyllfluoreszenzparameter sowie ein erhöhtes Bakterienwachstum im Vergleich zu einer Infektion mit den Bakterien allein. Keine Effekte wurden auf transkriptioneller Ebene bei der Untersuchung von Genen verschiedener Stoffwechselwege gefunden. Die Invertaseaktivität nach zusätzlicher Behandlung mit Acarbose war tendenziell niedriger als die Aktivität nach einer Pathogeninfektion alleine. Acarbose erhöhte die Spiegel an Salicylsäure unabhängig von einer Pathogeninfektion. Da das Bakterienwachstum in Mutanten des Salicylsäure-vermittelten Abwehrweges bei zusätzlicher Behandlung mit Acarbose ebenfalls erhöht war, kann eine Beteiligung dieses Abwehrweges am Acarboseeffekt bisher ausgeschlossen werden. Invertasen sind neben ihrer Beteiligung an der Abwehr für die Regulation von Entwicklungsprozessen wichtig. In einem funktionellen Ansatz mit Pflanzen, die Invertaseinhibitoren induzierbar produzieren, wurde die Funktion von Invertasen getestet. Zur Generierung spezifischer Effekte wurden die Inhibitoren unter Kontrolle synthetischer Promotoren in A. thaliana exprimiert. Unerwarteterweise war das Wachstum putativ transgener Keimlinge jedoch im 4-Blatt-Stadium arretiert. Eine Analyse der Aktivität der ß-Glucuronidase in den entsprechenden Reporterlinien zeigte eine Korrelation zwischen der Wachstumsarretierung und einer hohen Aktivität dieser Promotoren unter verschiedenen in vitro Bedingungen. Dieser negative Effekt der Invertaseinhibition auf das Keimlingswachstum wurde in transgenen Tabakpflanzen bekräftigt, die Invertaseinhibitoren unter Kontrolle eines Tetracyclin-induzierbaren Promotors exprimierten. Eine erfolgreiche Induktion des Promotors resultierte in einer Reduktion des Frischgewichtes der Keimlinge. Mittels in silico Expressionsdaten und Northern Blot Analysen konnte für A. thaliana eine spezifische und starke Expression verschiedener Invertaseisoformen in Keimlingen nachgewiesen werden. Diese komplementären Ergebnisse zeigen die Notwendigkeit der Invertaseaktivität für eine normale Keimlingsentwicklung.

Hieracium fallax Willd. und weitere Hieracium echioides-Zwischenarten im nordwestlichen Bayern

Die Grenze des riesigen eurasiatisch-kontinentalen Areals von Hieracium echioides Lumn. verläuft durch Mitteldeutschland, Zwischenarten aus der Hieracium echioides-Verwandtschaft (sect. Echinina) dringen westlich bis in die Oberrheinebene vor, sind aber im übrigen Süden und Südosten Deutschlands sehr selten oder fehlen. In den letzten Jahren wurden im Nordwesten Bayerns neue Wuchsorte von Hieracium auriculoides Láng (MTB 5526.31, 5924.44, 6125.13, 6223.22), H. calodon Tausch ex Peter (6123.21, 6125.13) und H. fallax Willd. (6223.21) nachgewiesen. Dies stellt den zweiten aktuellen Nachweis von H. fallax in Bayern dar, bemerkenswert ist ein Nachweis von H. auriculoides in der Rhön in ca. 700 m Meereshöhe.
Ein pflanzensoziologisches Modell der Schattentoleranz von Baumarten in den Bayerischen Alpen

The ecological niche, as a summary of the environment in which a tree species can live, is a central concept in vegetation ecology and its application in silviculture. While the fundamental niche focusses on the physiological constraints of survival and growth, the realised niche takes competition in real communities into account. To understand realised niches in a causal fashion requires knowledge of the life cycle of plant species. The concept of regeneration niche is based on the notion that establishment and juvenile growth are particularly sensitive stages. Obviously, silviculturalists must be particularly interested in regeneration niches. The database BERGWALD contains 4,934 phytosociological plots from mountain forests and related vegetation types of the Bavarian Alps. The detailed information on plant species composition (trees, tree regeneration, shrubs, herbs and bryophytes) and cover has so far been used extensively for deriving vegetation units, site types and groups of indicator species. In the present study the database content was analysed with regard to the ecology of tree species in general and their regeneration niche in particular. The availability of light as a crucial resource that changes during forest succession was estimated by calculating average Ellenberg indicator values (mL) based on total field and bryophyte layer composition. The relative frequency of plots across the mL gradient in the total database was juxtaposed to the occurrence of the 16 most common tree species in the tree and in the regeneration layer, respectively. [...] As expected, the realised niches of tree species on the light gradient corresponded broadly to Ellenberg's L-value of tree regeneration. As the regional climax, Abies alba and Fagus sylvatica have coincident optima of tree layer and juvenile occurrences in closed, mature stands. Ulmus glabra and Fraxinus excelsior, as species of lower altitude, exhibit niches most similar to these climax species, followed by Acer pseudoplatanus and Picea abies, two of the most frequent species, that occur from low elevations to the timber line. The intermediate role of these four species is confirmed, as regeneration occurs mostly at light levels higher than those found under adult trees of the same species. Against expectations, Taxus baccata clearly prefers stands with moderate to high light, as do Sorbus aria, Sorbus aucuparia and Alnus incana. While Larix decidua and Pinus cembra occupy very similar overall niches, tree layer and regeneration niches of Larix differ markedly, whereas coincident layer niches in Pinus cembra underpin its status as a climax species at tree line. Pinus sylvestris and Salix eleagnos are typical shade-intolerant pioneers, of which regeneration is practically restricted to non-forest vegetation. Pinus rotundata and Juniperus communis are small trees that are entirely restricted to open stands subject to geomorphological activity. The results demonstrate the potential of phytosociological databases for studying the niches of tree species. To be sure, such analyses are no replacement for physiological and experimental studies. The research community is invited to use this source as a reference framework and an empirical validation for more specialised research.
Iconography of the Genus Hieracium in central Europe - Part 1: General Description and Morphotypes

The genus Hieracium comprises more than one thousand sexual and apomictic species in Europe, with numerous intermediates and microspecies. Only a small fraction of the members of the genus Hieracium has been illustrated or photo-documented in the literature. Since many of these publications are difficult to obtain, only a few specialists are familiar with most of the species and subspecies described in the literature. In order to overcome this problem and encourage geobotanical research on the genus Hieracium, we decided to edit an iconography of central and southern European Hieracia in an electronical journal (Forum geobotanicum) with free international access through the internet. Part I of this endeavour contains descriptions and photographs of the morphological spectrum of the genus Hieracium. Here, we categorize the genus into 15 basic morphotypes. These types conform partly to the sections and subsections of the genus Hieracium, but are in some cases informal and may even include members of different sections. Classification of morphotypes is considered helpful to obtain a first rough picture of an unknown species that then can be traced to the species and subspecies level by using keys or, after completion of this iconography, simply by screening the relevant images. One particularly novel aspect of the present endeavour will be the regular inclusion of magnified images and scanning electron micrographs.
Zur Kenntnis des Ranunculus auricomus-Komplexes in Deutschland: Ranunculus suborbicularis spec. nova

Die bei Schwarz (1949) als R. vertumnalis abgebildeten Pflanzen entsprechen nicht dem Typusmaterial, sondern weichen durch fast kreisförmige Blattspreiten der Schlussblätter und geringe Blattteilung ab. Sie werden hier als R. suborbicularis spec. nov. beschrieben und mit ihrem Blattzyklus und weiteren Belegen abgebildet. Die bekannte Verbreitung erstreckt sich auf Berlin (wohl erloschen), Thüringen und Bayern. Insgesamt ist aufgrund der kleinen Zahl der Populationen eine starke Gefährdung anzunehmen.
In August 2004 the authors of this report undertook a survey on the Genus Oenothera (evening-primroses) in the valley of the river Main between the cities of Schweinfurt and Bamberg (Bavaria, Lower and Upper Franconia). 22 different taxa were identified, most of which are new to Bavaria, one new for Europe (Oenothera hazelae Gates) and another one new for Middle Europe (Oenothera stucchii Soldano). In this report a list of all Oenothera species found in Franconia is given and Oenothera stucchii, a particular tall (up to > 2 m) species with extremely long Hypanthia of 50-70 mm and fruit capsules with truncated teeth, is described and depicted in more detail.
Das Naturschutzgebiet "Garchinger Heide" ist ein Relikt der ehemals ausgedehnten Kalkmagerrasen auf Pararendzinen über Niederterrassenschotter im Alpenvorland. Bei jahrhundertelanger extensiver Landnutzung hat sich auf trockenen nährstoffarmen Böden eine artenreiche Vegetation mit zahlreichen seltenen und gefährdeten Arten entwickelt. Die "Altheide", welche den größten Teil des Naturschutzgebiets einnimmt, ist geprägt durch flachgründige, nährstoffarme Böden, welche eine geschlossene Vegetation mit einem hohen Anteil an Arten der Halbtrockenrasen (hauptsächlich Mesobromion erecti und Cirsio-Brachypodion) tragen. Das 1945 durch Oberbodenabtrag begonnene, aber nicht mehr ausgebaute und verwendete „Rollfeld“ weist einen niedrigen Feinbodenanteil, welcher auf eine geringe Wasserverfügbarkeit schließen lässt, sowie niedrige Gehalte an Gesamtstickstoff und CAL-austauschbaren P2O5 auf. Die Vegetation ist lückig mit einem hohen Anteil an Trockenrasenarten (Xerobromion, Sedo-Scleranthetea, Sesleritalia albicantis). Das heutige Vorkommen zahlreicher Arten, welche in früheren Untersuchungen (1956 und 1986) nicht gefunden wurden, lässt auf eine fortschreitende Sukzession der Vegetation des Rollfeldes hin zu der der Altheide schließen. Der 1959 zum Naturschutzgebiet hinzu gekaufte ehemalige Acker unterscheidet sich immer noch deutlich von der Altheide. Eine allmähliche Annäherung der Standortbedingungen ist jedoch zu beobachten. So sanken die Gehalte an CAL-austauschbarem P_ 2 O_5 und K_2 O im Vergleich zu Werten aus dem Jahr 1993 deutlich ab. Trotz des immer noch hohen Anteils an Grünlandarten (Molinio-Arrhenatheretea) konnten sich im Vergleich zu 1986 mehr Magerrasenarten etablieren.
Prunus spinosa L. (Rosaceae) is one of the most widespread members of the genus Prunus in middle europe. Its morphological plasticity resulted in a number of described taxa at subspecific level. Since the early neolithic times, drupes of the plum family are recorded and exhibit already a remarkable diversity in size and form. Here we present a short historical account to the use of P. spinosa and an overview of the different taxonomic treatments. We examined distribution patterns in general and in particular in the central valley of the river Saale (Thuringia) with respect to ecological, edaphic and climatic factors. We assessed within 16 populations the variability of 22 metric and 10 qualitative morphological characters at 7 different locations. Population sites included forest-, way- and fieldsides, as well as lightish pine forests. Pollen fertility did not increase during the flowering period, all flowers were directly fully fertile from the beginning. In contrast, glucose content varied significantly depending of the status of fertilization. Epicuticular wax structure was without variation amongst the populations. P. spinosa leaves are covered with a smooth layer of slightly striated wax. Morphological characters were scored on 270 branches and 506 fruits. Most of the characters showed enormous variability among and within populations such as metrics of leaves, thorns and character states of flower morphology. The lowest variability among populations and therefore not dependend of modificatory factors was found in fruit characters. Since kernel morphology seems to be genetically rather than modificatory controlled, we applied the 3 taxonomical concepts of Werneck, Kühn and Scholz u. Scholz to identify evolutionary units at subspecific levels. However, population variability was still so high, that from our study here we can not support an infraspecific classification of Prunus spinosa L.
Based on queries of the phytosociological databank BERGWALD, a compilation of 3.504 forest vegetation plots from the Bavarian Alps, the ecological niche of Abies alba is re-assessed. The tree species occurs mostly admixed in mountain forests with Fagus and Picea rather than forming distinctive communities of its own. Climatically, Abies is widely distributed to the upper limit of the montane belt, but occurs only sparsely in subalpine forest. Analysis of Ellenberg indicator values based on total species composition yielded the following results: As a tolerant species Abies regeneration has a marked preference for shady forest, which in turn Abies tree layers themselves help create. It also has a clear preference for acidic topsoil conditions. Sites with low N-supply, such as early successional stages on raw carbonate soils, are rarely colonised by Abies. Also, dry and markedly wet forest sites in the region are avoided by Abies. Permutation-based indicator species analysis found a large number of common forest species as being significantly associated with Abies and its frequent companion Fagus sylvatica, whereas there is a negative relationship with more specialised Seslerion and Erico-Pinion species. As Abies alba has very few specific companion species not shared with either Fagus or Picea, the delimitation of Abietetum-syntaxa appears mostly motivated by ecological rather than phytosociological considerations. As a result of its susceptibility towards game browsing, Abies regeneration is an indicator of high woody species richness. The study broadly confirms Abies alba's status as a climax species intermediate between Fagus and Picea, and demonstrates the potential of large phytosociological databanks for niche modelling.
Activation of mitogen-activated protein (MAP) kinases is a common reaction of plant cells in defense-related signal transduction pathways. Since the downstream events after the activation of MAP kinases are largely unknown in plants, the role of MAP kinases in the co-ordinate regulation of defense reactions and primary carbon metabolism by stress related stimuli has been analyzed in tomato. Thus, the relationship between mitogen activated protein kinases (LpMPK2 and LpMPK3) and extracellular invertases Lin6, as the key enzyme of an apoplasmic phloem unloading pathway, has been analyzed. The results showed that the mRNAs of LpMPK3 and Lin6 are sequentially induced by the same set of stress related stimuli (E-Fol, PGA,wounding, and KCl). The induction of the Lin6 promotor, as revealed by an increase in β-glucuronidase activity after 2 hours, was dependent both on the expression and activation of LpMPK3. These data suggest that the induction of extracellular invertase Lin6 by stress related stimuli requires LpMPK3. Glucose, metabolic molecule, was shown to result in the simultaneous induction of AtMPK4 and AtMPK6 activities that could be separated by anion-exchange chromatography, and characterized by differential cross-reaction with MAP kinase antibodies. Taken together, these data suggest that the activation of MAP inases play central roles in the regulation of sugar signaling. Stomatal movement is controlled by environmental signals including light intensity,humidity and atmospheric CO2 level. In Arabidopsis, a complete MAP kinase signaling cascade regulates stomatal development and patterning. However, the movement of stomata mediated by CO2 induced signaling pathways is not fully studied in higher plants. Here, we show that elevated levels of CO2 induce rapid and transient activation of SIPK and NtMPK4. The activation of both MAP kinases may regulate the anion channel activation for stomatal movement by the elevated level CO2. Up to now, the non-antioxidant function of tocopherol is not clear in higher plant,whereas the ability of tocopherol to modulate the stress tolerance mediated by function of antioxidant has been described in numerous studies. Thus, the function of α-tocopherol in stimuli-induced signal transduction pathways mediated by MAP kinase has been analyzed in tobacco. It has been shown that the activation of MAP kinase was induced by treatment of fungal elicitor and α-tocopherol phosphate but not α-tocopherol. Interestingly, α-tocopherol showed the transient inhibitory effect on the activation of stimuli-induced MAP Kinases in BY2 cells and tobacco plants, whereas ascorbate did not inhibit the activation of MAP kinases. The inhibitory activity test indicated that current application may indirectly affect the activity of MAP kinases. These results suggest that α-tocopherol can negatively regulate stimuliinduced signal transduction pathways via inactivation of MAP kinases. The purine-analogues have been tested and reported to be specific inhibitors of protein kinases mediated by structural-based selectivity in mammalian. Here, we tested C2, N6, N9-trisubstituted purines to determine basic relationship between their chemical structure and inhibitory activity using a particular plant MAP kinase. The modification of substitution in position C2 and N9 caused the increased inhibitory activity of 6-(benzylamino) purine analogue. In addition, 6-(isopentenylamino) purine analogues suggested that addition of a methyl group to position N9 caused at least 2-fold increased inhibitory activity compared with the addition of isopropyl group.Taken together, our study suggests that the selectivity and potency of inhibitors can be improved by structure modification. In addition, we have characterized the physiological function of Arabidopsis thaliana PLAT domain protein 1 (AtPDP1) in modulating the interaction of defense pathways mediated by biotic and abiotic factors. Interestingly, overexpression of AtPDP1 resulted in increasing susceptibility of virulent pathogens and necrotrophic fungus, and developing necrosis induced by unknown biotic factors. However, these overexperssion lines showed the significantly delayed senescence and higher level of phosystem II quantum yield compared with control plants against high salt stress. Our results strongly indicate that AtPDP1 positively regulate with salt tolerance, and enhances the sensitivity to biotic stresses. We propose that the AtPDP1 might be regulated with the complex pathways of interplay among various signaling during stress adaptation.
The island of Rügen (Rugia), located in the Baltic sea, is the most northeastern (NE) part of Germany. Due to its particular geographic position at the border between scandinavian, middle european and continental european floral elements, Rügen harbours several hawkweed species (Hieracia) of the scandinavian area such as Hieracium fuscocinererum, H. subramosum, H. subrigidum and H. diaphanoides subsp. neoornatum and, at the same time, is the most northwestern location of H. echioides. Two endemic Hieracium species have been identified recently, i. e. H. muorum subsp. rugianum and H. caesium subsp. zabelianum (Gottschlich et al. 1998, Bot. Rundbr. Mecklenburg-Vorpommern 31:1-94). In the present communication, two further novel endemic Hieracium taxa will be described, which are restricted to the chalk cliffs of Cape Arkona and Jasmund, i. e. H. swantevitii and H. lachenalii subsp. litocretaceum. H. swantevitii (Swantevit’s hawkweed) is intermediate between H. caesium/H. bifidum and H. lachenalii with hairy, modestly glandular involucre and slightly serrated elongated leaves . This view of an intermediate position of H. swantevitii between these species was further supported by the ultrastructure of epidermal papillae of the outer bracts of the involucre visualized by scanning electron microscopy. H. lachenalii subsp. litocretaceum (chalk cliff hawkweed) is characterized by its narrow anguste to almost linear denticulate leaves in combination with mode rately glandular heads. In addition to the description of these two new hawkweed taxa , the rediscovery of three further species will be reported for Rügen, i. e. H. echioides (W. Gager in SE Rügen), H. cymosum subsp. cymosum (close to Göhren in SE Rügen) and H. subrigidum E Glowe in N Rügen. The locality of H. echioides appears to be most north-western site in middle Europe, the locality of H. cymosum is one of the last growth sites in the northern German lowlands and H. subrigidum (so far only known as a single herbarium specimen, collected 1858 in Rügen) has so far not been recorded in other localities of middle Europe.
• Die Modellpflanze der Pflanzenphysiologen, Arabidopsis thaliana, besitzt mindestens 15 verschiedene kaliumselektive Kanäle, von denen 5 der Strukturklasse der Tandemporen-Kaliumkanäle angehören und daher TPK-Kanäle genannt werden. • Tandemporenkanäle findet man nur bei eukaryontischen Organismen. Die pflanzlichen Tandemporen Kaliumkanäle haben einen gemeinsamen phylogenetischen Ursprung und unterscheiden sich von den Tierischen und denen der Pilze und Einzeller. Die pflanzlichen TPK-Kanäle lassen sich wiederum in die TPK1-Unterfamilie und die TPK2-Unterfamilie unterteilen. Die weitere Evolution der TPK2-Unterfamilie von A. thaliana, TPK2, TPK3, TPK4 und TPK5, lässt sich eindeutig auf bestimmte Duplikationsereignisse im Genom von A. thaliana und dessen Ahnen zurückführen. Auch der Ein-Poren Kaliumkanal KCO3 geht sehr wahrscheinlich auf die Duplikation des TPK2 und einer anschließenden Deletion und nicht auf einen der prokaryontischen Ein-Poren-Kaliumkanal-Prototypen zurück. • Vier der A. thaliana TPK-Kanäle (TPK1, 2, 3 und 5) lokalisieren in der Vakuolenmembran, während einer, TPK4, zum großen Teil im ER, aber auch in der Plasmamembran zu finden ist. Die Translokation des TPK1 folgt dem sekretorischen Pfad vom ER, durch den Golgi und möglichen intermediären Kompartimenten hin zur Membran der lytischen Vakuole. Von entscheidender Bedeutung ist dabei der zytoplasmatische Carboxy-Terminus (CT) des TPK1. Deletionsmutanten des TPK1 CT zeigen, dass die Translokation mindestens zwei Sortierungsschritten, am Ausgang des ER und des Golgi, unterliegt. Fehlt der CT komplett bleibt der Kanal im ER. Die Sortierungssignale des TPK1 CT konnten auf die EF-Hand Domäne I eingegrenzt werden. Anschließende Punktmutationen in diesem Bereich konnten zeigen, dass TPK1 in der eigentlich für die Ca2+ Bindung zuständigen Domäne ein di-azidisches ER-Export Motiv bestehend aus Asparaginsäure, Leucin und Glutaminsäure enthält. Andere Arbeiten legen nahe, dass der Mechanismus des ER-exports von TPK1 auf der Interaktion mit COPII Vesikelhüllproteinen beruht; TPK1 also in Vesikel sortiert wird, die sich am ER abschnüren und mit dem cis-Golgi fusionieren. Der Vergleich mit anderen pflanzlichen TPK Kanälen lässt vermuten, dass TPK1 Orthologe, nicht aber die A. thaliana Homologen ein di-azidisches ER-Exportmotiv besitzen. Die Translokation des TPK3 erwies sich dementsprechend als unabhängig von dessen CT. Weitere Experimente schließen außerdem eine Beteiligung der 14-3-3 Bindung an der Translokation aus. • TPK4 ist der einzige TPK der heterolog in Xenopus Oozyten funktionell exprimiert werden kann. Wie Mutationen an einem essentiellen Aspartat (Asp86, Asp200) in der Pore zeigten, sind beide tandem repetierten Porendomänen einer Kanaluntereinheiten an der Porenbildung beteiligt. Somit formt sich TPK4 ähnlich wie die tierischen TPK-Kanäle voraussichtlich aus zwei Untereinheiten. Ein Austausch der zweiten Porendomäne von TPK4 konnte zeigen, dass TPK2, TPK3 und TPK5, mit ihrer zweiten Porendomäne und TPK4 mit seiner ersten Porendomäne den TPK4 zu einem funktionellen Kaliumkanal komplementieren können. Da keine der TPK4 Eigenschaften, außer geringfügig die relative Permeabilität für Rb+, verändert wurde, kann man absehen, dass die homologen TPK2, TPK3 und TPK5 als instantan aktivierte, spannungsunabhängige Kaliumkanäle der Vakuolenmembran fungieren. Dazu kommt wahrscheinlich ähnlich wie bei TPK1 ein 14-3-3 und Ca2+ abhängiges Öffnen und Schließen. • Weiterführende elektrophysiologische Untersuchungen am TPK4 zeigten eine Beteiligung einer transmembranen Asparaginsäure (Asp110) an der Kaliumpermeation und der schwachen Einwärtsgleichrichtung. Der Aspartatrest ist in die wassergefüllte Aussparung der zytoplasmatischen Porenhälfte orientiert. Damit kann er über ionische Wechselwirkungen sowohl Kalium in der Pore konzentrieren als auch potentielle Kanalblocker wie Mg2+ oder Polyamine binden. Die Konservierung des Aspartats unter anderem bei TPK2, TPK3 und TPK5 deutet daraufhin, dass auch die vakuolären TPK-Kanäle eine Einwärtsgleichrichtung vermitteln, die auf einem spannungsabhängigen Block von zytoplasmatischer Seite basiert. • Im Gegensatz zum zytoplasmatischen Block ist das Schließen des TPK4 durch zytoplasmatische Ansäuerung spannungsunabhängig und ist daher von einer Protonierungsreaktion abhängig. Über zahlreiche Deletionen und Chimären des TPK4 wurde der Bereich, in dem sich pH-Sensor und pH-Tor befinden, auf den Bereich zwischen transmembranen und zytoplasmatischen Domänen eingegrenzt. Darüber hinaus fungieren Histidine nicht als pH-Sensor.
Die Stomata in der Epidermis von Pflanzen sind Poren, die den Gasaustausch mit der Atmosphäre regulieren. Die Öffnungsweite der Stomata kann verändert werden, was eine Optimierung der CO2 Aufnahme für die Photosynthese ermöglicht und gleichzeitig den Wasserverlust durch Transpiration minimiert. Um diese Funktion zu erfüllen, können Stomata verschiedene Stimuli wie Wasserstress (durch Abscisinsäure), Licht und CO2 wahrnehmen. Die oben genannten Reize führen dann zu einer Aufnahme oder Abgabe von osmotisch aktiven Substanzen in zwei Schließzellen, welche die Stomaöffnung kontrollieren. Die Rezeptoren zur Wahrnehmung dieser Stimuli, die intrazellulären Signalwege und die beteiligten Ionentransportproteine in den Schließzellen sind nur lückenhaft bekannt. In dieser Arbeit lag ein Hauptaugenmerk auf der Rolle von Anionenkanälen der Plasmamembran bei Stomabewegungen, sowie auf den Signalwegen welche diese Kanäle steuern. Die Aktivität der Anionenkanäle wurde mit der DEVC (Double Electrode Voltage Clamp) Einstich-Methode in Schließzellen in der intakten Pflanze gemessen, kombiniert mit Calcium Imaging durch den Ca2+ Indikator Farbstoff FURA2. Stomaschlussreaktionen werden durch Abscisinsäure (ABA), CO2 und Dunkelheit induziert und bei allen drei Stimuli konnten wir in Nicotiana tabacum eine Aktivierung von Anionenkanälen beobachten. Das führt zu Anionenefflux aus den Schließzellen und einer Depolarisation der Plasmamembran, was wiederum Kalium-Efflux-Kanäle spannungsabhängig aktiviert. Der resultierende Verlust osmotisch aktiver Teilchen führt dann zu Turgorabnahme der Schließzellen und Stomaschluss. Das zeitliche Muster der Anionenkanalaktivität bei dem Stomaschluss, ausgelöst durch CO2, Dunkelheit und ABA war bei allen Reizen ähnlich. Es zeigte sich eine charakteristische transiente starke und darauf folgende schwächere Anionenkanalaktivität. Dieses konservierte Muster lässt Überschneidungen bei der Signaltransduktion der verschiedenen Stimuli vermuten. Die gesteigerte Aktivität der Anionenkanäle während der Reaktion auf ABA und Dunkelheit wurde in ungefähr der Hälfte der Antworten von einem Anstieg der zytosolischen Ca2+ Konzentration begleitet. Bei beiden Stimuli scheinen somit Ca2+ abhängig und unabhängig Signale intrazellulär weitergeleitet zu werden. Allerdings war der Effekt der Ca2+ Signale auf die Aktivität der Anionenkanäle bei den beiden Stimuli unterschiedlich. Eine zytosolisch erhöhte Ca2+ Konzentration konnte bei Antworten auf ABA nicht mit einer erhöhten Anionenkanalaktivität in Verbindung gebracht werden, bei Dunkelheit hingegen wurde die Aktivität der Anionenkanäle in Anwesenheit von Ca2+ gesteigert. Die wichtige Rolle von Anionenkanälen beim Stomaschluss lässt vermuten, dass ihre Deaktivierung eine Vorraussetzung für eine Stomaöffnung ist. Blaulicht führt bei niedrigen Photonen-Fluss Raten zu Stomaöffnung und sollte daher Anionenkanäle inhibieren. Übereinstimmend damit konnten wir tatsächlich zeigen, dass Blaulicht in Schließzellen von Vicia faba und Arabidopsis thaliana Anionenkanäle deaktiviert. Diese Deaktivierung ist von den Phototropin-Blaulichtrezeptoren abhängig, da die Deaktivierung der Anionenkanäle in Arabidopsis thaliana phot1/phot2 Doppelmutanten nicht beobachtet werden konnte. Neben einer Blaulicht spezifischen Antwort öffnen Stomata auch in Antwort auf photosynthetisch aktive Strahlung (PAR). Die PAR Wahrnehmung scheint zu einem wesentlichen Teil über Veränderungen der interzellulären CO2 Konzentration, ausgelöst durch die Photosyntheseaktivität des Mesophylls, stattzufinden (Roelfsema et al., 2002). In Übereinstimmung mit dieser Hypothese konnten wir in Schließzellen in Albino Blattarealen von Chlorophytum comosum und gebleichten Vicia faba keine Reaktion auf PAR beobachten, obwohl Schließzellen von Chlorophytum comosum in Albino Bereichen funktionierende Chloroplasten besitzen. Die Rolle von CO2 bei der PAR Antwort haben wir des Weiteren in NtMPK4 antisense Pflanzen untersucht. Stomata von NtMPK4 antisense Pflanzen haben nicht auf Änderungen in der atmosphärischen CO2 Konzentration reagiert und zeigten eine stark reduzierte Antwort auf PAR. Diese Ergebnisse bestätigen die wichtige Rolle der intrazellulären CO2 Konzentration bei der PAR Antwort, sie zeigen aber auch, dass es anscheinend zusätzlich zu CO2 noch ein weiteres PAR abhängiges Signal für Stomaöffnung gibt.
Untersuchung zu den frühen ABA-induzierten elektrischen Reaktionen in Schließzellen von Vicia faba

Im Rahmen der vorliegenden Arbeit wurde die Perzeption und frühe Signaltransduktion des Phytohormons ABA in Schließzellprotoplasten von Vicia faba mittels der Patch-Clamp-Technik untersucht. Es wurde entdeckt, dass der ABA-Signaltransduktionskette zur Aktivierung von Plasmamembran-ständigen Anionenkanälen voraussichtlich eine Proteinkinase beinhaltet und durch eine cytosolische ABA-Perzeption ausgelöst wird. Die durch ABA-bewirkte Anionenkanal-Aktivierung verursacht in Schließzellen eine Plasmamembran-Depolarisation. Basierend auf der ABA-induzierten Schließzellen-Depolarisation wurde zudem eine Methode etabliert, um mit dem Spannungs-sensitiven Farbstoff DiBAC4(3) in Populationen von intakten Vicia faba-Schließzellprotoplasten Membranpotential-Änderungen zu quantifizieren.
In order to test the effects of environmental factors on different characteristics of plant leaf waxes, barley plants (Hordeum vulgare) were abiotically stress treated (exposure to darkness, heavy metal, high salt concentrations and drought), and biotically stressed by the infection with powdery mildew (Blumeria graminis f.sp. hordei; Bgh). Different wax parameters like amount, chemical composition, and micromorphology of epicuticular wax crystals, were investigated. Etiolated leaves of barley showed distinctly reduced wax amounts and modifications in their relative composition. The alterations of these wax parameters might be a result of a developmental delay, which could have been caused by a decreased availability of energy for cellular processes, due to lack of light. Cadmium exposure led to a 1.5-fold increase of wax amount, while chemical composition was unaffected. In drought- and salt-stressed plants, all investigated leaf wax parameters remained unaltered. In each of the abiotic treatments, the microstructure of epicuticular wax crystals, formed as typical platelets, was not modified. Even after 6d infection with powdery mildew (Bgh), neither locally nor systemically enforced modifications of wax features were revealed.
The analyzed leave surfaces, resulting from these four abiotic and the biotic treatment (phenotypic approach), were compared to altered leaf surfaces’ characteristics of 18 analyzed eceriferum (cer-) wax mutants (genotypic approach). Within the screening, 5 mutants were selected which distinctly differed from the wild-type in wax amount, portions of epi- and intracuticular wax fraction, relative chemical composition, crystal morphology, and surface wettability (hydrophobicity).
Apart from quantitative and qualitative effects on the leaf waxes, environmentally enforced modifications in cuticular waxes might be reflected in molecular processes of wax biogenesis. Therefore, a barley wax-microarray was established. 254 genes were selected, which are putatively involved in processes of de novo fatty acid biosynthesis, fatty acid elongation, and modification, and which are supposed to take part in lipid-trafficking between cell compartments, and transport of wax components to the outer cell surface. The regulations within the expression pattern evoked by the respective treatments were correlated with the corresponding analytical wax data, and the observed molecular effects of a 3d powdery mildew infection were compared with succeeding fungal morphogenesis. Etiolation and cadmium exposition pointed to transcriptional modifications in the de novo fatty acid synthesis, and in the screened, transport-related mechanisms, which correlate with respective alterations in surface wax characteristics. Moderate changes in the gene expression pattern, evoked by drought- and salinity-stress, might give hints for evolved adaptations in barley to such common habitat stresses. Theinvasion of powdery mildew into the epidermal host cells was reflected in the regulation of several genes. Beside other functions, these genes take part in pathogen defense, and intracellular component transport, or they encode transcription factors. The different modifications within the molecular responses evoked by the investigated abiotic treatments, and the effects of powdery mildew infection representing a biotic stressor, were compared between the different treatments.
In order to test the potential impact of different wax parameters on Bgh, conidia germination and differentiation was comparably investigated on leaf surfaces of abiotically stressed wild-type and cer-mutants, isolated cuticles, and further artificial surfaces. The rates of conidial development were similar on each of the leaf surfaces resulting from the abiotic treatments, while a significant reduction of the germination and differentiation success was revealed for the wax mutant cer-yp.949. Compared to the wild-type, developmental rates on isolated cuticles and extracted leaf waxes of the mutant cer-yp.949 indicated a modified embedding of cuticular waxes, and a possibly changed three-dimensional structure of the cer-yp.949 cuticle, which might explain the reduced conidial developmental rates on leaf surfaces of this particular mutant.
Experiments with Bgh conidia on mechanically de-waxed leaf surfaces (selective mechanical removal of the epicuticular leaf waxes with glue-like gum arabic, followed by an extraction of the intracuticular wax portion with chloroform) demonstrated the importance of the wax coverage for the germination and differentiation of the fungal conidia. On all dewaxed leaf surfaces, except those of cer-yp.949, the differentiation success of the germlings was significantly reduced, by about 20% (“wax-effect”). This result was verified through an artificial system with increased conidia developmental rates on glass slides covered with extracted leaf waxes. Further comparative tests with the major components of barley leaf wax, hexacosanol and hexacosanal, showed that the germination and differentiation of powdery mildew conidia not only depends on the different chemistry, but is also influenced by the respective surface hydrophobicity. Compared to hexacosanol, on hexacosanal coated glass surfaces, higher germination and differentiation rates were achieved, which correlated with increased levels of surface hydrophobicity. Developmental rates of conidia on hydrophobic foils demonstrated that hydrophobicity, as a sole surface factor, may stimulate the conidial germination and differentiation processes. Moreover, the survival of conidia on artificial surfaces is determined by additional surface derived factors, e.g. the availability of water, and a pervadable matrix.
During the last few years an increasing number of physiological processes in plants have been shown to be regulated by NO. NO plays important roles in growth and development, plant disease resistance, abiotic stress, and in above and underground plant organs. In recent years several enzymatic pathways and few non-enzymatic pathways were proposed for nitric oxide production in plants. The major goal of this work was to quantify NO production by plants and especially by roots, and to identify the enzymes responsible for NO production. As a major method, NO production by roots was followed through on-line measurement of NO emission into the gas phase by chemiluminescence (= direct chemiluminescence), and also by indirect chemiluminescence where trace amounts of oxidized products like NO2- and NO3- can be easily measured. Plants used were tobacco wild-type (N. tabacum cv Xanthi or cv Gatersleben), NR-free mutants grown on ammonium in order to prevent NR induction, plants grown on tungstate to inhibit synthesis of functional MoCo-enzymes, and a NO-overproducing nitrite reductase (NiR)-deficient transformant as well as barley, rice and pea. Induction of a hypersensitive response (HR) in tobacco leaves was achieved by using avirulent Pseudomonas syringae pv phaseolicola. At oxygen concentrations of <1%, even completely nitrate reductase (NR)-free root tissues reduced added nitrite to NO, indicating that in roots, NR was not the only source for nitrite-dependent NO formation. By contrast, NR-free leaf slices were not able to reduce nitrite to NO. Root NO formation was blocked by inhibitors of mitochondrial electron transport (Myxothiazol and SHAM), whereas NO formation by NR containing leaf slices was insensitive to the inhibitors. Consistent with that, mitochondria purified from roots, but not those from leaves, reduced nitrite to NO at the expense of NADH. The inhibitor studies suggest that, in root mitochondria, both terminal oxidases participate in NO formation, and they also suggest that even in NR-containing roots, a large part of the reduction of nitrite to NO was catalysed by mitochondria, and less by NR. The differential capacity of root and leaf mitochondria to reduce nitrite to NO appears to be common among higher plants, since it was observed with Arabidopsis, barley, pea, and tobacco. Nitrite and NADH consumption by mitochondria were also measured. Anaerobic, nitrite-dependent NO emission was exclusively associated with the membrane fraction, without participation of matrix components. It was also examined whether root mitochondria and mitochondrial membranes produce nitric oxide (NO) exclusively by reduction of nitrite or also via a nitric oxide synthase (NOS),- and to what extent direct NO measurements could be falsified by NO oxidation. In addition to chemiluminescence, Diaminofluoresceins (DAF) were used as an NO indicators for comparison. In air, mitochondria apparently produced no nitrite-dependent NO, and no NOS activity was detected by direct or indirect chemiluminescence. In contrast, with DAF-2 and DAR-4M an L-arginine-dependent fluorescence increase took place. However, the response of this apparent NOS activity to inhibitors, substrates and cofactors was untypical when compared with commercial iNOS and is considered an artefact. With iNOS, about 2/3 of the NO were oxidized to (nitrite + nitrate). Mitochondria also appear to consume NO without increasing oxidation to (nitrite+ nitrate). We therefore assume formation of NO to a volatile intermediate (eventually N2O3). It was recently shown that the hypersensitive response (HR) of tobacco triggered by the fungal elicitor cryptogein occurred independent of the presence or absence of nitrate reductase (NR). One conclusion was that NR-dependent NO formation played no role in the HR. Here we present evidence that the described scenario may be specific for cryptogein. Pseudomonas syringae pv. phaseolicola was infiltrated into tobacco leaves from WT plant and from the NiR-deficient NO-overproducing clone 271, grown either on nitrate or ammonium. Lesion development as well as bacterial growth and sugar concentrations in leaves and in the leaf apoplast was monitored. Lesion development was positively and bacterial growth was negatively correlated with nitrate nutrition and eventually with NO formation. Bacterial growth was positively correlated with ammonium nutrition and apoplastic sugar concentrations. Total (free and conjugated) SA content were always drastically increased by bacterial infection, but there was no clear correlation with NO production. In the presence of cryptogein, Pseudomonas growth was drastically reduced. This shows that the assumed interdependence of bacterial growth, NO production and the HR is complex and not unifactorial.
Brassicaceae and a few related plant families are characterized by possession of the glucosinolate-myrosinase system. Glucosinolates are amino-acid derived allelochemicals which are hydrolysed upon tissue damage by myrosinase enzymes to produce various degradation products which can be toxic for generalist insects. The larvae of the crucifer-specialist Athalia rosae, the turnip sawfly, sequester glucosinolates into their haemolymph. The role of the glucosinolate-myrosinase system for the interaction of the turnip sawfly with Brassicaceae was examined in this study from two different perspectives: variation within individual plants and between plant species. The plant responses to the feeding by herbivores and the short-term effects this induction had on insect behaviour were investigated in white mustard. Furthermore, plants can use multiple defences. Hence correlations of glucosinolates and myrosinase activities with other defences and nutritional quality and their long-term effects on the development of the insects were investigated in seven different plant species.
Lokalisation, Funktion und Regulation pflanzlicher Tandem-Poren-Kaliumkanäle in Arabidopsis thaliana

Lokalisation - Alle TPKs bis auf TPK4, der in der Plasmamembran lokalisiert ist, sind im Tonoplasten lokalisiert. - Das 14-3-3-Bindemotiv bzw. der komplette N-Terminus spielt im Gegensatz zu den tierischen TPK´s keine Rolle beim Targeting (und evtl. auch beim Assembly), da ein Austausch der N-Termini bzw. Mutationen im 14-3-3- Bindemotiv keinen Einfluss auf die subzelluläre Lokalisation hat. - Im C-Terminus ist möglicherweise ein strukturelles Motiv bzw. eine Erkennungssequenz für das Targeting in unterschiedliche Zielmembranen lokalisiert. Eventuell ist hier auch eine Assembly-Domäne für den Zusammenbau der unterschiedlichen Kanaluntereinheiten vorhanden. TPK4 - Der Kaliumkanal TPK4 wird nach Agro-Infiltration in dem pflanzlichen Expressionssystem Nicotiana benthamiana exprimiert. - TPK4 ist auch in diesem Expressionssystem in der Plasmamembran der Zelle lokalisiert. - Die Ströme, welche aus Mesophyllzellen von TPK4 infiltrierten Blättern abgeleitet wurden, gleichen denen, von TPK4 exprimierenden Oocyten von Xenopus laevis. Somit hat TPK4 in beiden Expressionssystemen die gleichen elektrophysiologischen Eigenschaften. TPK1 - TPK1 bindet über die C-terminalen EF-Hände Calcium und wird durch diese Interaktion aktiviert. - TPK1 interagiert phosphospezifisch und isotypspezifisch mit dem 14-3-3- Protein GRF6. Diese Interaktion führt zur Aktivierung des Kanals. - Die Kinasen CPK3 und CPK29, welche das 14-3-3-Bindemotiv von TPK1 phosphorylieren um eine Interaktion mit 14-3-3-Proteinen zu ermöglichen, gehören zur Familie der CDPKs - Diese Kinasen sind selbst Calcium aktiviert und aller Wahrscheinlichkeit nach unter physiologischen Bedingungen inaktiv. Erst ein Anstieg der freien Calciumkonzentration führt zur Aktivierung der Kinase in der Zelle und damit zur Aktivierung des Kanals. - Das 14-3-3-Bindemotiv ist das einzige Target der CDPK´s im N-Terminus von TPK1 - Die Phosphatase, welche das 14-3-3-Bindemotiv von TPK1 dephosphoryliert gehört zur Familie der PP2A-Proteinphosphatasen. - Es ist möglich, dass die Kinase und damit auch der Kanal durch Salzstress und durch Kaliumunterversorgung aktiviert werden und somit die Signalkaskade für die Aktivierung von TPK1 über Kinasen/14-3-3/Calcium in einen stressphysiologischen Kontext involviert ist. - tpk1.3- und cpk3.1-Verlustmutanten zeigen eine Reduktion in der Keimungsrate unter Salzstress und limitierten Kaliumangebot. Es kann über einen funktionalen Komplex bestehend aus TPK1 und TPC1 zur Aufrechterhaltung der Na+/K+-Homeostase und der elektroneutralen Aufnahme von Na+ in die Vakuole unter Salzstressbedingungen spekuliert werden.
NO has been described as an important component involved in the development of the hypersensitive reaction (Delledonne et.al., 1998). Furthermore, NO induces expression of a set of defence gene, such as PR-1, PAL1 and chalcone synthase (CHS), and accumulation of SA (Durner et al., 1998). In this study, transgenic plants with altered NO levels were used to study the role of NO in plant defence. Arabidopsis plants which, due to expression of a bacterial NO dioxygenase, exhibit lower levels of NO than wild-type plants, show several weakened defence response, including the oxidative burst and expression of phenylpropanoid pathway genes. By contrast, constitutive expression of a bacterial NO synthase in Arabisopsis results in increased levels of endogenous NO. However, these plants do not show constitutively activated defence responses, but suffer from increased susceptibility to various strains of P. syringae. This might indicate that a gradient in NO production rather than constitutive elevation of NO is necessary to trigger plant defence responses. Nevertheless, NO seems to be important for regulation of the oxidative state in plant cells. This function of NO is important during leaf senescence. The data of the present work indicate that NO acts as senescence-delaying factor during plant development. The molecular action of NO in plants and signalling cascades in which NO is involved as second messenger are still poorly understood. Experiments addressing the selective quantification of NO in intact plant tissue, the identification of NO-target proteins as well as the function of NO-modified biomolecules might help to understand the role of NO in plants. Non-host resistance consists of several layers of defence that include preformed compounds existing in plants before pathogen infection and induced defences which the plant activates after recognition of a pathogen. The role of inducible defences in preventing multiplication of non-adapted bacteria is not clear. Our experiments suggest that to restrict non-adapted bacterial growth, pre-formed antimicrobial compounds and an early inducible cell wall-based defence might play an important role in Arabidopsis leaves. Upon inoculation with non-adapted bacteria, we have observed early, TTSS-independent up-regulation of PAL1 and BCB, two lignin biosynthesis genes which might be involved in papilla formation or other kinds of cell wall fortification. Moreover, Arabidopsis pal1 knockout lines permit significantly higher survival of non-adapted bacteria in leaves than wild-type plants, suggesting a functional importance of PAL1 up-regulation. Although non-host bacteria, like host bacteria, induce accumulation of SA and PR gene expression in a TTSS-dependent manner, SA-dependent or JA/ET-dependent defences do not directly contribute to non-host resistance. Moreover, non-adapted bacteria activate similar defence signalling pathways as do host bacteria. However, because of varieties in effector protein composition between different non-adapted bacterial strains, the activated signalling pathways might also include different compounds. The Arabidopsis ecotype Ler 0 is more susceptible to a non-adapted strain of P. syringae than ecotype Col-0. Although differences in glucosinolate content and composition between those ecotypes exist, they are probably not a major reason for the observed difference in non-host resistance. To further understand the mechanisms underlying non-host resistance, the generation of double or triple mutants with deficits in both cell wall-based defences and SA-dependent signal cascades is necessary. Moreover, the study of genome polymorphism and composition of secondary metabolites between Ler-0 and Col-0 can shed new light into the mechanisms of non-host resistance against bacterial pathogens. Additionally, experiments addressing papilla formation and callose biosynthesis in Ler-0 and Col-0 could help to further elucidate bacterial non-host resistance. Our data indicate that localized contact of Arabidopsis leaves with non-adapted bacteria, type III secretion-defective P. syringae strains and bacterial pathogen-associated molecular patterns (PAMPs) induce systemic acquired resistance (SAR) at the whole plant level. This finding contrasts the general belief that an HR or other leaf necroses are required for SAR induction. The observed symptomless systemic response was abolished in all SAR-deficient mutants tested in this study, but was intact in the jar1 mutant, which is compromised in induction of ISR, indicating that non-host bacteria and PAMPs induce SAR in a mechanistically similar way than host bacteria. In addition, our data show that the extent of SA accumulation or PR gene expression induced at sites of virulent or avirulent P. syringae inoculation rather than the amount of tissue necroses or jasmonate accumulation determine the magnitude of SAR. The fact that systemic responses were also triggered after local treatment with type III secretion-defective P. syringae strains and bacterial PAMPs indicate that induction of SAR is TTSS-independent. Instead, recognition of general elicitors like flagellin and LPS play an important role in activation of the SAR process. To broaden the concept of PAMP-based SAR initiation, further general elicitors from bacteria and fungal pathogens should be tested for their capability to induce SAR. Screens for mutants with deficiency in SAR activation by individual PAMPs can help to identify new components involved in the SAR signalling cascade. Possible functions of PAMPs as mobile systemic signals should be tested in future experiments. By selection of candidate genes whose expression is up-regulated in Arabidopsis leaves infected with avirulent and virulent P. syringae and pathophysiological analyses of corresponding T-DNA knockout lines, FLAVIN-DEPENDENT MONOOXYGENASE1 (FMO1) was identified as a key SAR regulator. SAR triggered by P. syringae is completely abolished in fmo1 mutant plants, and pathogen-induced expression of FMO1 in systemic leaves is closely correlated with the capability of different Arabidopsis lines to develop SAR. According to our findings, we have proposed that the FMO1 acts in signal amplification in non-inoculated, systemic leaves to trigger SAR. Experimental verification of the postulated potential amplification cycle underlying SAR should be tested in future experiments. The generation of transgenic lines expressing FMO1::GFP will provide useful information about the cellular localization of the FMO1 protein. Moreover, a comparative metabolomic analysis using SAR-induced wild-type, fmo1 knockout and FMO1 overexpressing lines can be used to identify substrates and reaction products of the FMO1 monooxygenase. As the single yeast FMO (yFMO) provides oxidizing equivalents at the ER for correct protein folding, expression of FMO1 in yfmo mutant yeast combined with protein activity assays might indicate whether FMO1 exhibits functional similarities with yeast FMO, e.g. in assuring proper folding of ER-targeted proteins essential for SAR establishment. Identification of further genes involved in activation of systemic resistance and biochemical characterization of the corresponding proteins can help to understand the SAR process in more detail.

Zusammenfassung: Untersucht wurden die Pflanzenbemalungen in drei unterfränkischen Kirchen, die in ihrer Naturnähe und damit botanischen Korrektheit sowie in ihrer Intention differieren. Die Aufgabe der Arbeit war, soweit möglich eine botanische Bestimmung durchzuführen, nach Gründen für das Auftauchen von floralen Dekorationen in Sakralräumen allgemein und speziell für das Auftauchen einer bestimmten Species im speziellen zu suchen. Die drei betrachteten Kirchen unterscheiden sich zum einen in ihrer ursprünglichen Nutzung: "normale" Pfarrkirche, Hofkapelle eines Domherrenhofs und Betort einer Rosenkranzbruderschaft, die im Zuge der Gegenreformation unter Julius Echter gegründet wurde. Letztere diente wohl auch als repräsentative Schloßkapelle zum Schloß Büchold. Weitere Unterschiede sind in der Qualität der Ausmalung zu erkennen: Die Gotteshäuser in Rothenfels und im Seebacher Hof verfügen über Pflanzendarstellungen, die stark schematisiert sind, wobei die der Allendorfkapelle noch Ansätze von Naturbeobachtung erkennen lassen. Demgegenüber erscheinen die Fresken im Chor der Bücholder Pfarrkirche zwar auch leicht schematisiert, aber doch so nah an der Natur, daß wenigstens zum Teil eine Bestimmung bis auf die Art gelungen ist; bei einigen Exemplaren war dies jedoch nicht möglich. In letzterem Gotteshaus ist die Bemalung ein existentieller Teil des ikonographischen Konzeptes; Rothenfels läßt ein solches nicht erkennen; die Kapelle im Hof Luden entzieht sich einer Beurteilung diesbezüglich, da ihre Innenausstattung kriegsbedingt verbrannt ist. Das dortige Vorkommen von Ruta graveolens L., Rosa spec. und Tulipa spec. als bekannten Marienpflanzen macht ein früher erkennbares Konzept jedoch wahrscheinlich. Zur Kirche St. Nikolaus und Mariae Heimsuchung in Arnstein-Büchold: Der Chor ist in seiner Gesamtheit Teil des Ausstattungsprogramms der Kirche: er symbolisiert den hortus conclusus, den Garten, der Sinnbild nicht nur für die Gottesmutter ist. Innerhalb dieses Chorgartens lassen sich an den liturgisch wichtigen Stellen - Chorbogen, Chorhaupt und Schlußstein - durchweg bekannte Symbolpflanzen finden. Das Chorgewölbe ist in sich gegliedert in 50 Teile und korrespondiert direkt mit der Anzahl der Rosenkränze im Rosenkranzgebet; diese 50 Deckenteile bilden zusammen zwei vierzählige Blüten, die das Garten- oder Blumenmotiv verstärken. An Stellen, die ohne besondere Wichtigkeit sind, hat der Maler Wolfgang Ritterlein eine bunte Mischung aus einheimischen, fremdländischen und phantastischen Pflänzlein gestaltet, so daß in der Gesamtheit nicht nur ein umschlossener Garten, sondern auch eine Wunderkammer, ein Kuriositätenkabinett entsteht. - Damit erweist sich die Bemalung dieses Chors als eine schöne Symbiose aus symbolhafter Ausstattung und Repräsentation wie sie zu Beginn des Barocks nicht selten begegnet.
RS1 ist ein 67-68 kD großes, ubiquitär exprimiertes Protein, das sich an der Innenseite der Plasmamembran befindet und in den Zellkern wandern kann. Durch immunhistochemischen Untersuchungen an Dünndarmschnitten der Maus konnte RS1 das erste Mal in dieser Arbeit im Kern und an der Membran von Enterozyten gezeigt werden. RS1 wird von einem intronlosen Single Copy Gen kodiert und ist fähig Ubiquitin über eine Ubiquitin-assoziierte (UBA) Domäne zu binden. Es reduziert die Konzentration einiger Proteine in der Plasmamembran. Durch Expressionsversuche in Xenopus Oozyten wurde gezeigt, dass RS1 die Menge des Na+-D-Glukosekotransporters SGLT1 in der Plasmamembran transkriptionsunabhängig reduziert. Entsprechend seiner dualen Lokalisation beteiligt sich RS1 aber auch an der Transkriptionsregulation im Zellkern. In der vorliegenden Arbeit konnten Informationen über die physiologische Funktion des membranassoziierten Regulatorproteins RS1 gewonnen werden. Nach Erstellung einer RS1-knock-out Maus wurde sichergestellt, dass ein erfolgreiches Rekombinationsereignis stattgefunden hatte und RS1 tatsächlich nicht mehr exprimiert wurde. Die RS1-knock-out Mäuse waren postnatal lebensfähig, vermehrten sich gut und entwickelten eine Fettsucht mit 30 % mehr Körpergewicht, 80 % mehr Fett und um 40 % vergrößerten Fettzellen. Bei den transgenen Mäusen war weder die Nahrungsaufnahme gesteigert, noch die motorische Aktivität verringert. In der Bürstensaummembran des Dünndarmepithels konnte bei den RS1-knock-out Mäusen die siebenfache Menge an Protein des Na+-abhängigen D-Glukosekotransporters SGLT1 detektiert werden, während die Konzentration des passiven Glukosetransporters GLUT2 in der basolateralen Membran nicht verändert war. Die Zunahme der SGLT1-Proteinmenge war posttranskriptional bedingt. Bei der RS1-knock-out Maus wirkt sich der in Oozyten beobachtete Effekt an der Plasmamembran aus, während der an konfluenten LLCPK1 Zellen gezeigte Effekt im Zellkern nicht zum Tragen kommt. Die transgenen Tiere resorbierten die doppelte Menge an D-Glukose im Dünndarm. Das spricht dafür, dass bei der RS1-knock-out Maus der „turnover“ des SGLT1 beeinflusst sein muss, da die siebenfache SGLT1-Proteinmenge einem verdoppelten Transport über den SGLT1 gegenübersteht. Die RS1-knock-out Mäuse zeigten normale Insulinspiegel und reguläre oralen Glukosebelastungstests. Bei gefütterten Mäusen waren die Serumleptinspiegel ähnlich wie bei Wildtypmäusen, die typische Reduzierung des Serumleptinspiegel konnte bei den Mäusen ohne RS1 aber nicht beobachtet werden. Untersuchungen an Fettzellexplantaten ergaben, dass die Sekretion von Leptin bei RS1- knock-out-Explantaten erhöht war, während die Leptinsynthese und die insulinabhängige Regulation der Leptinsekretion nicht verändert waren. Mit der RS1-knock-out Maus wurde ein neues Fettsuchtmodell geschaffen. RS1 spielt eine physiologisch wichtige Rolle bei der Regulation der D-Glukoseaufnahme im Darm. Der visceralen Adipositas liegt wahrscheinlich eine gesteigerte Nahrungsutilisation durch die verbesserte Glukoseaufnahme über den SGLT1 im Darm zugrunde. Die gesteigerte Glukoseabsorption ist ursächlich für den Anstieg der Fettmasse. Die Fettzellen vergrößern sich und sezernieren dann mehr Leptin. Es ist davon auszugehen, dass die RS1-knock-out Mäuse eine veränderte Nahrungsutilisation aufgrund der verbesserten Glukoseaufnahme im Dünndarm aufweisen. Die Adipositas demzufolge ein sekundärer Effekt. Gleichzeitig kann aber nicht ausgeschlossen werden, dass RS1 direkt auf die Zellen des weißen Fettgewebes wirkt und bei Wildtypmäusen die Sekretion des Leptins aus Vesikeln hemmt.
