In this article we offer initial insights into the fairly new interdisciplinary and international domain of robotics in Christian religious practice. We are a group of scholars in media ethics, practical theology/religious education, and human computer interaction, who have been engaged in this discourse since 2017.
A natural starting point is our study of BlessU2, a “blessing robot,” a device which received considerable recognition from the global public at the Wittenberg 500th reformation anniversary in 2017. We thus begin with the results of this study. Secondly, we will briefly address the relevant theses from Gabriele Trovato et al., as presented in their 2019 article on so-called theomorphic robots – followed by our interdisciplinary discussion of their approach. Finally, we draw conclusions for further work on the field of “religious robots.”
Somewhat more carefully: Section 1 offers starting points within the perspectives of Christian religious practice: here, the blessing robot is both cause and occasion for doing religion and theologizing in the context of existential questions (1.1). We continue with perceptions in the field of religion regarding “Discursive Design Theory” (1.2). The interaction of humans with computers as posing questions for theological standardization of religious practice is focused upon in 1.3. Section 2 reconstructs the HRI/HCI-initiative to develop theomorphic robots in a twofold manner, i.e., the idea of developing theomorphic robots (2.1) and the concept of theomorphic robots: Questions and objections (2.2). In this part of the article we raise discussion points concerning the relationship between technology and religion and the need for sharpening the understanding of religion within the research field. Section 3 closes with propositions and alternatives.
Neurodegeneration plays an essential role in Parkinson’s disease (PD). Several crucial neuronal pro-and antidegeneration markers were described to be altered in disease models accompanied by neurodegeneration. In the AAV1/2-A53T-aSyn PD rat model progressive time-dependent motor impairment and neurodegeneration in the nigrostriatal tract starting from 2 weeks after PD model induction could be found. Downregulation of Nrf2 in SN and nigrostriatal axon localization, a trend of Tau downregulation in SN and upregulation in axon localization in the AAV1/2-A53T-aSyn PD rat model were observed, indicating potential therapeutic value of these two molecular targets in PD. No alterations of SARM1 and NMNAT2 could be detected, indicating little relevance of these two molecules with our AAV1/2-A53T-aSyn rat model.
Plexus injury often occurs after motor vehicle accidents and results in lifelong disability with severe neuropathic pain. Surgical treatment can partially restore motor functions, but sensory loss and neuropathic pain persist. Regenerative medicine concepts, such as cell replacement therapies for restoring dorsal root ganglia (DRG) function, set high expectations. However, up to now, it is unclear which DRG cell types are affected by nerve injury and can be targeted in regenerative medicine approaches.
This study followed the hypothesis that satellite glial cells (SGCs) might be a suitable endogenous cell source for regenerative medicine concepts in the DRG. SGCs originate from the same neural crest-derived cell lineage as sensory neurons, making them attractive for neural repair strategies in the peripheral nervous system. Our hypothesis was investigated on three levels of experimentation. First, we asked whether adult SGCs have the potential of sensory neuron precursors and can be reprogrammed into sensory neurons in vitro. We found that adult mouse DRG harbor SGC-like cells that can still dedifferentiate into progenitor-like cells. Surprisingly, expression of the early developmental transcription factors Neurog1 and Neurog2 was sufficient to induce neuronal and glial cell phenotypes. In the presence of nerve growth factor, induced neurons developed a nociceptor-like phenotype expressing functional nociceptor markers, such as the ion channels TrpA1, TrpV1 and NaV1.9. In a second set of experiments, we used a rat model for peripheral nerve injury to look for changes in the DRG cell composition. Using an unbiased deep learning-based approach for cell analysis, we found that cellular plasticity responses after nerve injury activate SGCs in the whole DRG. However, neither injury-induced neuronal death nor gliosis was observed. Finally, we asked whether a severe nerve injury changed the cell composition in the human DRG. For this, a cohort of 13 patients with brachial plexus injury was investigated. Surprisingly, in about half of all patients, the injury-affected DRG showed no characteristic DRG tissue. The complete entity of neurons, satellite cells, and axons was lost and fully replaced by mesodermal/connective tissue. In the other half of the patients, the basic cellular entity of the DRG was well preserved. Objective deep learning-based analysis of large-scale bioimages of the “intact” DRG showed no loss of neurons and no signs of gliosis.
This study suggests that concepts for regenerative medicine for restoring DRG function need at least two translational research directions: reafferentation of existing DRG units or full replacement of the entire multicellular DRG structure. For DRG replacement, SGCs of the adult DRG are an attractive endogenous cell source, as the multicellular DRG units could possibly be rebuilt by transdifferentiating neural crest-derived sensory progenitor cells into peripheral sensory neurons and glial cells using Neurog1 and Neurog2.
In dieser Arbeit wurde die Krankheitsprogression im Parkinson-Mausmodell hm2α-SYN-39 mit zunehmendem Alter charakterisiert. Die Mäuse wurden in 4 Altersgruppen (2-3, 7-8, 11-12, 16-17 Monate) mit motorischen Verhaltenstests auf einen Parkinson-Phänotyp untersucht. Zudem erfolgten Untersuchungen des dopaminergen Systems zur Detektion von neurochemischen Veränderungen und einer Neurodegeneration im nigrostriatalen Trakt. Weiterhin wurden neuroinflammatorische Prozesse des adaptiven und angeborenen IS in der SN und im Striatum mittels immunhistochemischer Färbungen beurteilt.
Ein Parkinson-Phänotyp in diesem Mausmodell zeigte sich nur leicht ausgeprägt, sodass der Rotarod- und Zylinder-Test lediglich den Hinweis auf eine nicht-signifikante Einschränkung der Motorik erbrachte. Dennoch ergab die stereologische Quantifizierung TH- und Nissl-positiver Zellen in der SNpc der hm2α-SYN-39 Mäuse eine altersabhängige, signifikant-progrediente Reduktion der dopaminergen Neurone mit zunehmendem Alter. Eine signifikant niedrigere TH-positive Zellzahl dieser tg Mäuse zeigte sich ab einem Alter von 16-17 Monaten verglichen zu gleichaltrigen wt Tieren. Dagegen war die Neurodegeneration im Striatum etwas weniger ausgeprägt. Die tg Mäuse präsentierten im Alter von 16-17 Monaten eine nicht-signifikante Erniedrigung der dopaminergen Terminalen verglichen zu gleichaltrigen wt Tieren. Ein DA-Mangel im Striatum der tg Mäuse konnte mittels HPLC bestätigt werden. Bis zum Alter von 16-17 Monaten wurde eine signifikante Reduktion der DA-Level von 23,2 % verglichen zu gleichaltrigen wt Mäusen gezeigt. Außerdem erniedrigt waren die striatalen Level von NA und 5-HAT bei tg Mäusen, passend zu den bisherigen Ergebnissen bei Parkinson-Patienten.
Immunhistochemische Untersuchungen einer Neuroinflammation im nigrostriatalen Trakt ergaben eine tendenziell erhöhte Infiltration von CD4- und CD8-positiven T-Zellen bei hm2α-SYN-39 Mäusen mit zunehmendem Alter, wobei die Infiltration CD8-positiver Zellen ausgeprägter war als bei CD4-positiven Zellen. Eine noch deutlichere neuroinflammatorische Reaktion zeigte das angeborene IS. Hierbei ergab die immunhistologische Quantifizierung CD11b-positiver mikroglialer Zellen einen hochsignifikanten Anstieg im nigrostriatalen Trakt bei hm2α-SYN-39 Mäusen schon im jungen Alter.
Zusammenfassend präsentierte dieses Parkinson-Mausmodell eine langsam-progrediente Parkinson-Pathologie mit begleitender Neuroinflammation im nigrostriatalen Trakt während des Alterns, wobei die Immunantwort der mikroglialen Zellen zu einem früheren Zeitpunkt einsetzte als die T-Zellinfiltration und Neurodegeneration. Dieses Mausmodell bietet zahlreiche Möglichkeiten zur zukünftigen Erforschung der Pathophysiologie beim MP. Generell weist diese Arbeit auf eine bedeutende Rolle neuroinflammatorischer Prozesse in der Krankheitsprogression der Parkinsonerkrankung hin und soll dazu ermutigen Neuroinflammation durchaus intensiver in tg Tiermodellen zu untersuchen.
Kanurennsport ist in Deutschland eine der erfolgreichsten olympischen
Sommersportarten und hat mit 12 potenziellen Goldmedaillenchancen eine hohe
Bedeutung für den deutschen Spitzensport. In der nationalen als auch
internationalen wissenschaftlichen Forschung ist Kanurennsport jedoch bis dato
unzureichend untersucht. Dabei stellt Kanurennsport als eine der wenigen vorrangig
durch die Oberkörpermuskulatur angetriebenen Sportarten eine Besonderheit dar.
Ein zentraler Forschungsschwerpunkt ist seit einigen Jahrzehnten die Erforschung
der optimalen Verteilung der Trainingsintensität (engl. training intensity distribution;
TID) für die Leistungsentwicklung von Ausdauerathlet:innen. Häufig wird die
Trainingsintensität hierzu in einem Drei-Zonen-Modell kategorisiert, bei dem Zone
(Z) 1 einer Intensität unterhalb der aeroben Schwelle, Z2 der Intensität zwischen
der aeroben und anaeroben Schwelle und Z3 Intensitäten oberhalb der anaeroben
Schwelle entspricht. Forschungsergebnisse weisen darauf hin, dass sich die TID
nicht nur in Abhängigkeit von Sportart, Belastungsform, Trainingsstatus und
Saisonphase unterscheidet, sondern auch in Abhängigkeit von der eingesetzten
Quantifizierungsmethode (z.B. Herzfrequenz, Geschwindigkeit, Wattleistung, etc.).
Für die Sportart Kanurennsport besteht bezüglich TID-Forschung großer
Nachholbedarf, da bisherige Untersuchungen ausschließlich in Ausdauerportarten
stattfanden, die hauptsächlich den Unterkörper (z.B. Radfahren, Laufen) oder Oberund
Unterkörper (Schwimmen, Rudern) in die Vortriebsgenerierung einbinden.
Bislang fehlen Informationen zu rein aus dem Oberkörper angetriebenen
Sportarten.
Als Grundlage für die Bestimmung der Trainingsintensitätszonen werden in
Trainingspraxis und Forschung Stufentests zur Bestimmung der maximalen
Sauerstoffaufnahme sowie der Leistung an der aeroben und anaeroben
ventilatorisch- und/oder laktatbasierten Schwelle angewandt. Die Stufentest werden
im Kanurennsport aktuell vorrangig mittels Labordiagnostik auf dem Kanu-
Ergometer durchgeführt, da diese weniger stark durch die diversen Umwelteinflüsse
(Wind, Wellen, Temperatur, Strömung, etc.) beeinträchtigt wird. Jedoch gibt es
Hinweise, dass die Belastung auf dem Ergometer biomechanisch und physiologisch
von der auf dem Wasser im Kanurennsport abweicht, sodass deren Mehrwert für
die Diagnostik und die Trainingsplanung in Frage zu stellen ist.
Ziel der vorliegenden kumulativen Dissertation war es
(1) zu untersuchen, inwiefern eine laborbasierte Leistungsdiagnostik einer
feldbasierten im Kanurennsport entspricht (Studie 1) und daraufhin die
Methoden der Leistungsdiagnostik für die Studien 2 und 3 zu wählen; und
(2) erste wissenschaftliche Erkenntnisse zur TID und deren
Quantifizierungsmethodik in der Sportart Kanurennsport zu gewinnen
(Studie 2 & 3).
Diese sollten dann mit dem Wissensstand aus Sportarten, die obere und untere
Extremitäten (z.B. Biathlon, Rudern) bzw. primär die unteren Extremitäten (z.B.
Radsport, Laufen) für den Vortrieb einsetzen, abgeglichen werden.
Zusammenfassend konnte zunächst in Studie 1 aufgrund von Unterschieden in der
VO2, der Muskeloxygenierung im Musculus biceps brachii sowie im subjektiven
Belastungsempfinden dargestellt werden, dass sich eine Belastung auf dem
Wasser von der auf dem Ergometer unterscheidet und somit eine wasserbasierte
Leistungsdiagnostik im Kanurennsport vorzuziehen ist.
Die Ergebnisse aus den Studien 2 und 3 zeigten, dass die TID im Saisonverlauf
variiert und im Mittel einen hohen Anteil (80–90%) niedrigintensiven Trainings (Z1)
aufwies, wobei in der Vorbereitungsphase eine pyramidale TID Struktur (Z1>Z2>Z3)
und in der Wettkampfvorbereitung die Tendenz zu einer vermehrt polarisierten
Struktur (Z1>Z3>Z2) gefunden wurde. Somit weisen die Ergebnisse trotz der
physiologischen sowie biomechanischen Unterschiede zu Sportarten, die Oberbzw.
Ober- und Unterkörper bei der Vortriebsgenerierung einsetzen, eine
vergleichbare TID Struktur im Kanurennsport auf. Es ist zu vermuten, dass der
geringe Impact auf das Skelettmuskelsystem und die damit einhergehende
Möglichkeit, sehr hohe Trainingsvolumen mit der vergleichsweise kleinen
Oberkörpermuskulatur zu verwirklichen, diese TID-Struktur bedingen. Zudem
konnte dargestellt werden, dass die Wahl der Quantifizierungsmethode (extern vs.
intern; basierend auf physiologischen Parametern vs. Wettkampftempo) die
Darstellung der TID beeinflusst. Für eine adäquate Vergleichbarkeit und den
gezielten Einsatz muss insofern in der Forschung wie auch in den Sportarten ein
Konsens über die Wahl der Quantifizierungsmethode erarbeitet werden. Es scheint
zudem empfehlenswert die TID-Quantifizierungsmethode anhand der
Trainingsphase auszuwählen, wobei sich in der allgemeinen und spezifischen
Vorbereitungsperiode vorzugsweise eine TID Quantifizierung nach physiologischen
Kenngrößen empfiehlt. Hierbei erscheint ein Mix aus HF-basierter Analyse für Z1
sowie für längere Belastungen in Z2 und geschwindigkeitsbasierter Analyse für Z3
sowie kürzere Belastungen der Z2 zweckmäßig. In der Wettkampfvorbereitung stellt
sich dann zusätzlich eine Zoneneinteilung basierend auf dem Wettkampftempo als
sinnvoll dar.
Aufgrund der starken intra- und interindividuellen Variation der TID ist der
individuelle Mehrwert der auf dem Gruppenmittelwert basierenden Ergebnisse
jedoch zu hinterfragen und weist auf den Bedarf nach einer individuelleren
Betrachtung der TID und ihrer Effekte hin. Genauso stellt sich ein starker Einfluss
der allgemeinen physischen Aktivität sowie psychischer Belastungen auf die TID
und ihre Effekte dar, der wiederrum die Notwendigkeit eines holistischen
Betrachtungsansatzes für zukünftige Forschung aufzeigt. Außerdem gibt es im
Allgemeinen eine große Wissenslücke in Bezug auf Athletinnen in der TIDForschung,
weshalb die bisherigen Erkenntnisse für die Trainingsgestaltung
weiblicher Athleten mit Vorsicht behandelt werden müssen.
Various types of cancer involve aberrant cell cycle regulation. Among the pathways responsible for tumor growth, the YAP oncogene, a key downstream effector of the Hippo pathway, is responsible for oncogenic processes including cell proliferation, and metastasis by controlling the expression of cell cycle genes. In turn, the MMB multiprotein complex (which is formed when B-MYB binds to the MuvB core) is a master regulator of mitotic gene expression, which has also been associated with cancer. Previously, our laboratory identified a novel crosstalk between the MMB-complex and YAP. By binding to enhancers of MMB target genes and promoting B-MYB binding to promoters, YAP and MMB co-regulate a set of mitotic and cytokinetic target genes which promote cell proliferation. This doctoral thesis addresses the mechanisms of YAP and MMB mediated transcription, and it characterizes the role of YAP regulated enhancers in transcription of cell cycle genes.
The results reported in this thesis indicate that expression of constitutively active, oncogenic YAP5SA leads to widespread changes in chromatin accessibility in untransformed human MCF10A cells. ATAC-seq identified that newly accessible and active regions include YAP-bound enhancers, while the MMB-bound promoters were found to be already accessible and remain open during YAP induction. By means of CRISPR-interference (CRISPRi) and chromatin immuniprecipitation (ChIP), we identified a role of YAP-bound enhancers in recruitment of CDK7 to MMB-regulated promoters and in RNA Pol II driven transcriptional initiation and elongation of G2/M genes. Moreover, by interfering with the YAP-B-MYB protein interaction, we can show that binding of YAP to B-MYB is also critical for the initiation of transcription at MMB-regulated genes. Unexpectedly, overexpression of YAP5SA also leads to less accessible chromatin regions or chromatin closing. Motif analysis revealed that the newly closed regions contain binding motifs for the p53 family of transcription factors. Interestingly, chromatin closing by YAP is linked to the reduced expression and loss of chromatin-binding of the p53 family member Np63. Furthermore, I demonstrate that downregulation of Np63 following expression of YAP is a key step in driving cellular migration.
Together, the findings of this thesis provide insights into the role of YAP in the chromatin changes that contribute to the oncogenic activities of YAP. The overexpression of YAP5SA not only leads to the opening of chromatin at YAP-bound enhancers which together with the MMB complex stimulate the expression of G2/M genes, but also promotes the closing of chromatin at ∆Np63 -bound regions in order to lead to cell migration.
Companies are expected to act as international players and to use their capabilities to provide customized products and services quickly and efficiently. Today, consumers expect their requirements to be met within a short time and at a favorable price. Order-to-delivery lead time has steadily gained in importance for consumers. Furthermore, governments can use various emissions policies to force companies and customers to reduce their greenhouse gas emissions. This thesis investigates the influence of order-to-delivery lead time and different emission policies on the design of a supply chain. Within this work different supply chain design models are developed to examine these different influences. The first model incorporates lead times and total costs, and various emission policies are implemented to illustrate the trade-off between the different measures. The second model reflects the influence of order-to-delivery lead time sensitive consumers, and different emission policies are implemented to study their impacts. The analysis shows that the share of order-to-delivery lead time sensitive consumers has a significant impact on the design of a supply chain. Demand uncertainty and uncertainty in the design of different emission policies are investigated by developing an appropriate robust mathematical optimization model. Results show that especially uncertainties on the design of an emission policy can significantly impact the total cost of a supply chain. The effects of differently designed emission policies in various countries are investigated in the fourth model. The analyses highlight that both lead times and emission policies can strongly influence companies' offshoring and nearshoring strategies.
Fear and anxiety disorders – interaction of AVP and OXT brain systems with the serotonergic system

Anxiety disorders pose a great burden onto society and economy and can have devastating consequences for affected individuals. Treatment options are still limited to psychopharmacotherapy originally developed for the treatment of depression and behavioral therapy. A combination of genetic traits together with aversive events is most likely the cause of these diseases. Gene x environment studies are trying to find a link between genetic traits and specific negative circumstances. In a first study, we focused on social anxiety disorder (SAD), which is the second most-common anxiety disorder after specific phobias. We used a social fear conditioning (SFC) paradigm, which is able to mimic the disease in a mouse model. We wanted to investigate protein levels, as well as mRNA expression of immediate early genes (IEGs), to determine brain areas affected by the paradigm. We also included genes of the vasopressin (AVP)-, oxytocin (OXT)-, neuropeptide Y (NPY)-, and the serotonin system, to investigate the effects of SFC on neurotransmitter gene expression levels in brain regions related to social as well as fear-related behavior. AVP and OXT regulate a lot of different social and anxiety-related behaviors, both positive and negative. Finding a link between different neurotransmitter systems in the development of anxiety disorders could help to identify potential targets for new treatment approaches, which are desperately needed, because the rate of patients not responding to available treatment is very high.
We were able to show altered gene expression of the IEGs cFos and Fosl2, as well as a change in number and density of cFOS-positive cells in the dorsal hippocampus, indicating an influence of SFC on neuronal activity. Our results reveal a possible involvement of anterior dentate gyrus (DG), as well as cornu ammonis area 1 (CA1) and CA3 in the dorsal hippocampus during the expression of social fear. Contrary to our hypothesis, we were not able to see changes in neuronal activity through expression changes of IEGs in the amygdala. Significant higher IEG immunoreactivity and gene expression in the dorsal hippocampus of animals without fear conditioning (SFC-), compared to animals with fear conditioning (SFC+), indicate an involvement of different hippocampal regions in two possible scenarios. Either as elevated gene expression in SFC- animals compared to SFC+ animals or as reduction in SFC+ animals compared to SFC- animals. However, this question cannot be answered without an additional control of basal IEG-activity without social interaction. The NPY system in general and the neuropeptide y receptor type 2 in particular seem to be involved in regulating the response to social fear, mostly through the septum region. In addition to that, a possible role for the induction of social fear response could be identified in the serotonergic system and especially the serotonin receptor 2a of the PVN.
In a second study we focused on changes in the serotonergic system. A polymorphism in the human serotonin transporter (5-HTT) gene is associated with higher risks for the development of anxiety disorders. This makes the 5-HTT a widely used target to study possible causes and the development of anxiety disorders. In mice, a genetically induced knockout of the 5-Htt gene is associated with increased anxiety-like behavior. High amounts of stress during pregnancy, also known as prenatal stress, significantly increase the risk to develop psychiatric disorders for the unborn child. We utilized a prenatal stress paradigm in mice heterozygous for the 5-Htt gene. Some of the animals which had been subjected to prenatal stress showed noticeably “unsocial” interaction behavior towards conspecifics. Again, we were searching for links between the serotonergic system and AVP- and OXT systems. Through quantitative gene expression analysis, we were able to show that both AVP and OXT neuromodulator systems are affected through prenatal stress in female mice, but not in male mice. The 5-Htt genotype seems to be only slightly influential to AVP, OXT or any other neurotransmitter system investigated. Gene expression of AVP and OXT brain systems is highly influenced through the estrous cycle stages of female mice. Additionally, we analyzed the AVP and OXT neuropeptide levels of mice with different 5-Htt genotypes and in both sexes, in order to see whether the production of AVP and OXT is influenced by 5-Htt genotype. On neuropeptide level, we were able to identify a sex difference for vasopressin-immunoreactive (ir) cells in the PVN, with male mice harboring significantly more positive cells than female mice.
All presented studies aimed on the improvement of the quality analysis of already monographed drugs. Thereby different LC methods were applied and coupled to i.e., the UV/VIS detector, the CAD or a hyphenation of these detectors, respectively. The choice of the chromatographic system including the detector was largely dependent on the physicochemical properties of the respective analytes.
With the risk-assessment report on the API cetirizine we presented an exemplary tool, that can help to minimize the risk of the occurrence of unexpected impurities. An in- deep analysis of each step within synthesis pathway by means of reaction matrices of all compounds was performed. It is essential to understand the complete impurity profile of all reactants, solvents, and catalysts and to include them in the matrix. Finally, the API of this synthesis was checked if all impurities are identified by this tool. Of note, a shortcoming of such a targeted approach is that impurities can still occur, but they are not captured. This disadvantage can be partially compensated by non-targeted approaches if they are performed in parallel with the other studies that represent most of the impurities. However, this work also shows that even in a supposedly simple synthesis, potentially hundreds of by-products can be formed. For each of them, it must be decided individually whether their formation is probable or how their quantity can be minimized in order to obtain APIs, that are as pure as possible.
In the dapsone project it was aimed to replace the existing old Ph. Eur. TLC method with a modern RP-HPLC method. This was successful and since Ph. Eur. 10.6, the method developed in this work, became a valid monograph. Within the revision process of the monograph, the individual limits for impurities were tightened. However, this new method needs HPLC instrumentation, suitable to perform gradients. As this is not always available in all control laboratories, we also developed an alternative, more simple method using two different isocratic runs for the impurity analysis. The obtained batch results of both, the new pharmacopoeial method and the more simple one, were in a comparable order of magnitude. Furthermore, within the method development stage of the Ph. Eur. method, we could identify one unknown impurity of the impurity reference by high-resolution MS/MS analysis.
Also, in the baclofen project it was aimed to replace the existing Ph. Eur. method with the introduction of an additional impurity to be quantified. A corresponding method was developed and validated. However, due to the harmonization process of the pharmacopoeias, it is currently not used. In addition, we tried to find further, non- 116

chromophoric impurities by means of the CAD. However, except for one counterion of an impurity, no further impurities were found. Also, the aforementioned new impurity could not be detected above the reporting threshold in the batches analyzed. As the only individually specified impurity A is also present at a low level, it can be concluded that the examined batches of baclofen are very pure.
The use of universal detectors, such as the CAD can be particularly interesting for compounds with no chromophore or those with only a weak chromophore. Therefore, we decided to take a closer look at the impurity profile of acarbose. Currently, acarbose and its impurities are being studied by low wavelength UV detection at 210 nm. Therefore, the question arose whether there are no other impurities in the API that do not show absorption at this wavelength. CAD, which offers consistent detection properties for all non-volatile compounds, is ideally suited for this purpose. However, it was not so easy to use the CAD together with the UV detector, for example, as a hyphenated detection technique, because the Ph. Eur. method uses phosphate buffers. However, this is non-volatile and therefore inappropriate for the CAD. Therefore, an attempt was made to replace the buffer with a volatile one. However, since this did not lead to satisfactory results and rather the self-degradation process of the stationary phase used could be observed by means of the CAD, it was decided to switch to alternative stationary phases. A column screening also revealed further difficulties with acarbose and its impurities: they show an epimerization reaction at the end of the sugar chain. However, since one wanted to have uniform peaks in the corresponding chromatograms, one had to accelerate this reaction significantly to obtain only one peak for each component. This was best achieved by using two stationary phases: PGC and Amide-HILIC. Impurity-profiling methods could be developed on each of the two phases. In addition, as expected, new impurities could be detected, albeit at a low level. Two of them could even be identified by spiking experiments as the sugar fragments maltose and maltotriose.
Taken together, it can be concluded, that this work has contributed significantly to the improvement of the quality analysis of monographed drugs. In addition to the presented general tool for the identification of potential impurities, one of the methods developed, had already been implemented to the Ph. Eur. In an effort to improve the CAD's universal detection capabilities, additional methods have also been developed. Further, new improved methods for the impurity profiling are ready to use.
In Nervensystemen bedürfen Informationsweitergabe und Gedächtnisformation eines präzisen Zusammenspiels von Synapsen in Zeit und Raum. Synaptische Transmission basiert strukturell auf mesoskopischen cytosolischen Kompartimenten an der präsynaptischen Membran, sogenannten Aktiven Zonen (AZ). Ihre Cytomatrix, bestehend aus zentralen Gerüstproteinen wie Rab3 interacting molecule (RIM), ermöglicht eine schnelle Freisetzung synaptischer Vesikel. Die Defizienz der lokal häufigsten Isoform RIM1α resultiert an einer komplexen zentralen Säugersynapse, die des hippocampalen Moosfaserboutons (MFB) zu im Cornu ammonis (CA)3 befindlichen Pyramidalzellen, in einer dezimierten Langzeitplastizität. Auf Verhaltensebene zeigen diese Mäuse eine reduzierte Lernfähigkeit.
Die vorliegende Dissertation widmet sich grundlegend der bisher unbekannten dreidimensionalen (3D) AZ-Ultrastruktur des MFB in akuten Hippocampusschnitten der adulten Wildtyp- und RIM1α-Knock-Out-Maus (RIM1α\(^{-/-}\)). In einer methodischen Entwicklungsphase wurde ein neuartiges, anspruchsvolles Protokoll der nahezu artefaktfreien (near to native) Synapsenpräparation am MFB mittels Hochdruckgefrierung und Gefriersubstitution sowie der 3D-Modellierung mittels Elektronentomographie etabliert. In einer zweiten Experimentier- und Analysephase ermöglichte die hochwertige synaptische Gewebeerhaltung in beiden Genotypen eine standardisierte, auf Programmierskripten basierte Quantifizierung der AZ-Ultrastruktur bis auf die Ebene eines individuell gedockten synaptischen Vesikels.
Dieser Dissertation gelingt der Nachweis, dass eine Defizienz von RIM1α zu einer multidimensionalen ultrastrukturellen Veränderung der AZ und ihres Vesikelpools am MFB führt. Neben einer Reduktion, Dezentralisierung und strukturellen Veränderung (eng) gedockter Vesikel – der ultrastrukturellen Messgrößen von unmittelbar freisetzungsfähigen Vesikeln – verdichtet sich der distaler lokalisierte Vesikelpool auf zugleich größeren, heterogenen AZ-Flächen mit erweitertem synaptischem Spalt. Vorliegende Untersuchungen tragen zum Verständnisgewinn über eine zentrale Rolle von RIM1α für das Docking und die Organisation von Vesikeln der AZ im MFB bei. Darüber hinaus stellen die präzisen ultrastrukturellen Analysen eine morphologische Grundlage für weiterführende Studien mit Hilfe modernster Techniken dar, beispielsweise über die Auswirkungen der geänderten RIM1α\(^{-/-}\) AZ-Ultrastruktur auf die präsynaptische Plastizität sowie in Korrelation zum Gedächtnis und Lernen der Tiere.
Metallic nanostructures possess the ability to support resonances in the visible wavelength regime which are related to localized surface plasmons. These create highly enhanced electric fields in the immediate vicinity of metal surfaces. Nanoparticles with dipolar resonance also radiate efficiently into the far-field and hence serve as antennas for light. Such optical antennas have been explored during the last two decades, however, mainly as standalone units illuminated by external laser beams and more recently as electrically driven point sources, yet merely with basic antenna properties. This work advances the state of the art of locally driven optical antenna systems. As a first instance, the electric driving scheme including inelastic electron tunneling over a nanometer gap is merged with Yagi-Uda theory. The resulting antenna system consists of a suitably wired feed antenna, incorporating a tunnel junction, as well as several nearby parasitic elements whose geometry is optimized using analytical and numerical methods. Experimental evidence of unprecedented directionality of light emission from a nanoantenna is provided. Parallels in the performance between radiofrequency and optical Yagi-Uda arrays are drawn. Secondly, a pair of electrically connected antennas with dissimilar resonances is harnessed as electrodes in an organic light emitting nanodiode prototype. The organic material zinc phthalocyanine, exhibiting asymmetric injection barriers for electrons and holes, in conjunction with the electrode resonances, allows switching and controlling the emitted peak wavelength and directionality as the polarity of the applied voltage is inverted. In a final study, the near-field based transmission-line driving of rod antenna systems is thoroughly explored. Perfect impedance matching, corresponding to zero back-reflection, is achieved when the antenna acts as a generalized coherent perfect absorber at a specific frequency. It thus collects all guided, surface-plasmon mediated input power and transduces it to other nonradiative and radiative dissipation channels. The coherent interplay of losses and interference effects turns out to be of paramount importance for this delicate scenario, which is systematically obtained for various antenna resonances. By means of the here developed semi-analytical toolbox, even more complex nanorod chains, supporting topologically nontrivial localized edge states, are studied. The results presented in this work facilitate the design of complex locally driven antenna systems for optical wireless on-chip communication, subwavelength pixels, and loss-compensated integrated plasmonic nanocircuitry which extends to the realm of topological plasmonics.
Background
Ischemic stroke immediately evokes a strong neuro-inflammatory response within the vascular compartment, which contributes to primary infarct development under vessel occlusion as well as further infarct growth despite recanalization, referred to as ischemia/reperfusion injury. Later, in the subacute phase of stroke (beyond day 1 after recanalization), further inflammatory processes within the brain parenchyma follow. Whether this second wave of parenchymal inflammation contributes to an additional/secondary increase in infarct volumes and bears the potential to be pharmacologically targeted remains elusive. We addressed the role of the NLR-family pyrin domain-containing protein 3 (NLRP3) inflammasome in the subacute phase of ischemic stroke.
Methods
Focal cerebral ischemia was induced in C57Bl/6 mice by a 30-min transient middle cerebral artery occlusion (tMCAO). Animals were treated with the NLRP3 inhibitor MCC950 therapeutically 24 h after or prophylactically before tMCAO. Stroke outcome, including infarct size and functional deficits as well as the local inflammatory response, was assessed on day 7 after tMCAO.
Results
Infarct sizes on day 7 after tMCAO decreased about 35% after delayed and about 60% after prophylactic NLRP3 inhibition compared to vehicle. Functionally, pharmacological inhibition of NLRP3 mitigated the local inflammatory response in the ischemic brain as indicated by reduction of infiltrating immune cells and reactive astrogliosis.
Conclusions
Our results demonstrate that the NLRP3 inflammasome continues to drive neuroinflammation within the subacute stroke phase. NLRP3 inflammasome inhibition leads to a better long-term outcome—even when administered with a delay of 1 day after stroke induction, indicating ongoing inflammation-driven infarct progression. These findings may pave the way for eagerly awaited delayed treatment options in ischemic stroke.
This decade saw the development of new high-end light microscopy approaches. These technologies are increasingly used to expand our understanding of cellular function and the molecular mechanisms of life and disease. The precision of state-of-the-art super resolution microscopy is limited by the properties of the applied fluorescent label. Here I describe the synthesis and evaluation of new functional fluorescent probes that specifically stain gephyrin, universal marker of the neuronal inhibitory post-synapse. Selected probe precursor peptides were synthesised using solid phase peptide synthesis and conjugated with selected super resolution capable fluorescent dyes. Identity and purity were defined using chromatography and mass spectrometric methods. To probe the target specificity of the resulting probe variants in cellular context, a high-throughput assay was established. The established semi-automated and parallel workflow was used for the evaluation of three selected probes by defining their co-localization with the expressed fluorescent target protein. My work provided NN1Dc and established the probe as a visualisation tool for essentially background-free visualisation of the synaptic marker protein gephyrin in a cellular context. Furthermore, NN1DA became part of a toolbox for studying the inhibitory synapse ultrastructure and brain connectivity and turned out useful for the development of a label-free, high-throughput protein interaction quantification assay.
This work deals with the acceleration of cardiovascular MRI for the assessment
of functional information in steady-state contrast and for viability assessment
during the inversion recovery of the magnetization. Two approaches
are introduced and discussed in detail. MOCO-MAP uses an exponential
model to recover dynamic image data, IR-CRISPI, with its low-rank plus
sparse reconstruction, is related to compressed sensing.
MOCO-MAP is a successor to model-based acceleration of parametermapping
(MAP) for the application in the myocardial region. To this end, it
was augmented with a motion correction (MOCO) step to allow exponential
fitting the signal of a still object in temporal direction. Iteratively, this
introduction of prior physical knowledge together with the enforcement of
consistency with the measured data can be used to reconstruct an image
series from distinctly shorter sampling time than the standard exam (< 3 s
opposed to about 10 s). Results show feasibility of the method as well as
detectability of delayed enhancement in the myocardium, but also significant
discrepancies when imaging cardiac function and artifacts caused already by
minor inaccuracy of the motion correction.
IR-CRISPI was developed from CRISPI, which is a real-time protocol
specifically designed for functional evaluation of image data in steady-state
contrast. With a reconstruction based on the separate calculation of low-rank
and sparse part, it employs a softer constraint than the strict exponential
model, which was possible due to sufficient temporal sampling density via
spiral acquisition. The low-rank plus sparse reconstruction is fit for the use on
dynamic and on inversion recovery data. Thus, motion correction is rendered
unnecessary with it.
IR-CRISPI was equipped with noise suppression via spatial wavelet filtering.
A study comprising 10 patients with cardiac disease show medical
applicability. A comparison with performed traditional reference exams offer
insight into diagnostic benefits. Especially regarding patients with difficulty
to hold their breath, the real-time manner of the IR-CRISPI acquisition provides
a valuable alternative and an increase in robustness.
In conclusion, especially with IR-CRISPI in free breathing, a major acceleration
of the cardiovascular MR exam could be realized. In an acquisition
of less than 100 s, it not only includes the information of two traditional
protocols (cine and LGE), which take up more than 9.6 min, but also allows
adjustment of TI in retrospect and yields lower artifact level with similar
image quality.
Diese Studie evaluiert die Mundgesundheit von Hypophosphatasie (HPP)-Patienten und beinhaltet die Untersuchungen folgender Kenngrößen: erster Milchzahnverlust, Anzahl fehlender Zähne, DMF/T-Index (Kariös/Fehlend/Gefüllt), Blutung auf Sondierung (BOP), Sondierungstiefen (PPD), klinisches Attachmentniveau (CAL), Zahnlockerungen und kombinierter CDC/AAP Parodontal-Index. Die Daten wurden zwischen HHP-Patienten mit einer und zwei Mutationen auf dem für die alkalische Phosphatase kodierenden Genabschnitt (ALPL-Gen), sowie mit den Ergebnissen der DMS V verglichen, welche die Mundgesundheit der deutschen Allgemeinbevölkerung repräsentiert. 80 Patienten wurden untersucht; 64 von ihnen waren weiblich, 18 männlich. Das Mittlere Alter lag bei 46,4 Jahren. 55 Studienteilnehmer wiesen eine Mutation und 18 zwei Mutationen des ALPL-Gens auf (n=7 keine Testung). Patienten, die von zwei Mutationen des für die alkalische Phosphatase kodierenden Genabschnitts betroffen waren, zeigten ein höheres Risiko für Parodontitis und Zahnverlust als HPP-Patienten mit nur einer Mutation, sowie im Vergleich mit der deutschen Allgemeinbevölkerung. Allerdings stiegen auch bei Patienten mit nur einer Mutation mit zunehmendem Lebensalter die Ausprägungen parodontaler Erkrankungen, sowie die Zahnverlustrate im Vergleich zur Durchschnittsbevölkerung in Deutschland.
Humane mesenchymale Stromazellen (hMSCs) sind Interessengebiet der Forschung im Bereich des Tissue Engineering und werden häufig in Bezug auf Knorpelregeneration untersucht. Hierbei sind bereits mehrere potentielle Quellen nachgewiesen worden. Fokus dieser Disseration war die Vergleichende in vitro-Charakterisierung des Differenzierungspotentials von hMSCs von sechs verschiedenen Geweben des Kniegelenkes bei Patientinnen mit Gonarthrose um zu erforschen, welches Gewebe das meiste Potential für eine mögliche Extraktion von hMSCs birgt.
Hierfür wurden Zellen aus der Spongiose, dem Knorpelgewebe, des vorderen Kreuzbandes, der Menisken, der Synovialmebran sowie des Hoffa’schen Fettkörpers von fünf verschiedenen Spenderinnen isoliert und apidogen, osteogen sowie chondrogen differenziert sowie anschließend histologisch, immunhistochemisch und molekularbiologisch untersucht und die Ergebnisse miteinander verglichen. Hierbei wurde die zunächst der Nachweis erbracht, dass es sich bei allen Zellen um hMSCs handelt sowie anschließend gezeigt, dass alle Zellen ein multipotentes Differenzierungspotential aufweisen. Während kein statistisch relevanter Nachweis erbracht werden konnte, dass eine Zellquelle hierbei überlegen ist, scheinen die Zellen der Spongiosa sowie der Synovialmembran das vielversprechendste Potential zu bieten und eigenen sich somit als Quelle für weitere Forschung.
The field of giftedness and gifted education has long been characterized by internal fragmentation and inconsistent definitions of core concepts (e.g., Ambrose et al., 2010; Coleman, 2006; McBee et al., 2012). It was only in recent years that increased efforts have been made to organize available research findings and thereby bring back greater uniformity to the field of giftedness and gifted education. For example, Preckel et al.’s (2020) Talent Development in Achievement Domains (TAD) framework integrates theoretical perspectives and empirical knowledge from different parts of the field. It is general in concept and can be applied to a wide range of achievement domains. By specifically focusing on measurable psychological constructs as well as their relevance at different stages of the talent development process, Preckel et al.’s (2020) TAD framework is well suited as a starting point for generating more domain-specific talent development models.
The present thesis represents one of the first attempts to empirically test the validity of Preckel et al.’s (2020) TAD framework in academic domains using longitudinal data. The longitudinal data came from a sample of former junior students at Julius-Maximilians-Universität (JMU) Würzburg who showed high academic achievement potential.
There were two related research issues: Research Issue 1 first aimed to document in detail how the educational trajectories of former junior students unfold in the years following their Abitur. To this end, a follow-up was conducted among 208 young adults who had participated in the junior study program at JMU Würzburg between the winter semester of 2004/2005 and the summer semester of 2011. The design of the follow-up questionnaire was based on a series of research questions that had emerged from the relevant literature on junior study programs in Germany. The follow-up ran from October 2019 to February 2020. The data were analyzed descriptively and documented as a detailed report. The results of Research Issue 1 revealed that the former junior students continued to be academically (and later professionally) successful long after their school years. For example, at the time of the follow-up, almost all former junior students had earned a bachelor’s and a master’s degree, most often with notable academic successes (e.g., scholarships, awards/prizes). In addition, more than half of those who responded had begun or already completed a doctoral degree, also recording special academic accomplishments (e.g., scientific publications, scholarships). A significant proportion of the former junior students had already entered the workforce at the time of their response. A look at their current professional situation revealed an above-average expression of success indicators (e.g., income, professional status). The clear majority of the former junior students reported that, even in retrospect, they would choose to take part in the junior study program at JMU Würzburg again.
Research Issue 2 aimed to determine the extent to which the structure of Preckel et al.’s (2020) TAD framework could be empirically validated in academic domains. The educational trajectories of 84 former junior students at JMU Würzburg who had chosen a subject from the same subject field in their regular studies as in their junior studies served as the data basis. The educational trajectories were compiled from the former junior students’ follow-up data and from their data on the selection process for the junior study program at JMU Würzburg. Combining the structural assumptions of Preckel et al.’s (2020) TAD framework with relevant insights from individual academic disciplines made it possible to derive hypotheses regarding potential predictors and indicators of the talent development stages aptitude, competence, and expertise in academic domains. Structural equation models were used for data analysis. The results of Research Issue 2 suggested that the talent development stages aptitude, competence, and expertise, while being predictive of each other in their chronological order, could be satisfactorily modeled using framework-compliant indicators in academic domains. In comparison, the talent development stage transformational achievement could not (yet) be modeled based on the longitudinal data. Among the hypothesized predictors, former junior students’ investigative interests and their metacognitive abilities reliably determined the talent development stages competence and expertise, whereas the remaining predictors did not make significant contributions.
Taken together, the results of the present thesis suggest that the validity of Preckel et al.’s (2020) TAD framework can only be partially confirmed in academic domains. Unlike the postulated indicators, the predictors in Preckel et al.’s (2020) TAD framework do not seem to be easily generalizable to academic domains but to be highly specific with regard to the talent domain under consideration. Therefore, a natural progression of the present thesis would be to examine the structure of Preckel et al.’s (2020) TAD framework at the subordinate level of subject fields or even at the level of individual academic disciplines, for example.
Attention-deficit/hyperactivity disorder (ADHD) is the most prevalent neurodevelopmental disorder described in psychiatry today. ADHD arises during early childhood and is characterized by an age-inappropriate level of inattention, hyperactivity, impulsivity, and partially emotional dysregulation. Besides, substantial psychiatric comorbidity further broadens the symptomatic spectrum. Despite advances in ADHD research by genetic- and imaging studies, the etiopathogenesis of ADHD remains largely unclear. Twin studies suggest a heritability of 70-80 % that, based on genome-wide investigations, is assumed to be polygenic and a mixed composite of small and large, common and rare genetic variants. In recent years the number of genetic risk candidates is continuously increased. However, for most, a biological link to neuropathology and symptomatology of the patient is still missing. Uncovering this link is vital for a better understanding of the disorder, the identification of new treatment targets, and therefore the development of a more targeted and possibly personalized therapy.
The present thesis addresses the issue for the ADHD risk candidates GRM8, FOXP2, and GAD1. By establishing loss of function zebrafish models, using CRISPR/Cas9 derived mutagenesis and antisense oligonucleotides, and studying them for morphological, functional, and behavioral alterations, it provides novel insights into the candidate's contribution to neuropathology and ADHD associated phenotypes. Using locomotor activity as behavioral read-out, the present work identified a genetic and functional implication of Grm8a, Grm8b, Foxp2, and Gad1b in ADHD associated hyperactivity. Further, it provides substantial evidence that the function of Grm8a, Grm8b, Foxp2, and Gad1b in activity regulation involves GABAergic signaling. Preliminary indications suggest that the three candidates interfere with GABAergic signaling in the ventral forebrain/striatum. However, according to present and previous data, via different biological mechanisms such as GABA synthesis, transmitter release regulation, synapse formation and/or transcriptional regulation of synaptic components. Intriguingly, this work further demonstrates that the activity regulating circuit, affected upon Foxp2 and Gad1b loss of function, is involved in the therapeutic effect mechanism of methylphenidate. Altogether, the present thesis identified altered GABAergic signaling in activity regulating circuits in, presumably, the ventral forebrain as neuropathological underpinning of ADHD associated hyperactivity. Further, it demonstrates altered GABAergic signaling as mechanistic link between the genetic disruption of Grm8a, Grm8b, Foxp2, and Gad1b and ADHD symptomatology like hyperactivity. Thus, this thesis highlights GABAergic signaling in activity regulating circuits and, in this context, Grm8a, Grm8b, Foxp2, and Gad1b as exciting targets for future investigations on ADHD etiopathogenesis and the development of novel therapeutic interventions for ADHD related hyperactivity. Additionally, thigmotaxis measurements suggest Grm8a, Grm8b, and Gad1b as interesting candidates for prospective studies on comorbid anxiety in ADHD. Furthermore, expression analysis in foxp2 mutants demonstrates Foxp2 as regulator of ADHD associated gene sets and neurodevelopmental disorder (NDD) overarching genetic and functional networks with possible implications for ADHD polygenicity and comorbidity. Finally, with the characterization of gene expression patterns and the generation and validation of genetic zebrafish models for Grm8a, Grm8b, Foxp2, and Gad1b, the present thesis laid the groundwork for future research efforts, for instance, the identification of the functional circuit(s) and biological mechanism(s) by which Grm8a, Grm8b, Foxp2, and Gad1b loss of function interfere with GABAergic signaling and ultimately induce hyperactivity.
Schädel-Hirn Trauma ist die führende Ursache von Tod und Behinderung unter jungen Erwachsenen in den USA und Europa. Darüber hinaus steigert Schädel-Hirn Trauma das Risiko eine Demenzerkrankung oder andere neurodegenerative Erkrankung zu erleiden. Aus diesem Grund stellt eine bessere Erkenntnis der subakuten und chronischen pathophysiologischen Prozesse eine wichtige Grundlage für eine mögliche zukünftige neuroprotektive Therapie dar. Ziel dieser Arbeit war es daher eine Übersicht von funktionellen Einschränkungen und zellulären Veränderungen in der subakuten Phase innerhalb der ersten drei Monate darzustellen. Dazu wurden Verhaltensexperimente zu kognitiven Leistungen wie räumliches Lernen, kognitive Plastizität, episodisches Gedächtnis, Angstverhalten und allgemeine Lokomotion durchgeführt. Dabei konnten funktionale Einschränkungen der Tiere im Bereich der kognitiven Flexibilität, dem räumlichen Lernen, dem belohnungsmotivierten Verhalten, sowie Hyperaktivität beobachtet werden. Weiterführend erfolgten histologische und immunhistologische Untersuchungen an den Mäusegehirnen. So konnten in unserem Tiermodell sowohl lokale neuroinflammatorische Veränderungen nachgewiesen werden, also auch generalisierte Veränderungen, welche sich auf Isocortex und Hippocampus erstreckten und beide Hemisphären gleichermaßen betrafen. Ebenso konnten demyelinisierende Prozesse im Bereich der Läsion beobachtet werden. Im Bereich des Cortex zeigte sich außerdem eine axonale Schädigung mit begleitender Neuroinflammation, sowie eine Infiltration von B-Zellen. Anschließend wurde eruiert, ob eine Korrelation von funktionalem Outcome und histologischen Veränderungen besteht. Dabei zeigte sich eine signifikante Korrelation neuroinflammatorischer Prozesse mit Einschränkungen im räumlichen Lernen und Umlernen, sowie Auffälligkeiten im Bereich des belohnungsmotivierten Verhaltens. Damit ordnet sich diese Arbeit in die bestehenden Erkenntnisse zur Pathophysiologie des SHTs ein und ergänzt diese weiter.
In highly polarized neurons, endoplasmic reticulum (ER) forms a dynamic and continuous network in axons that plays important roles in lipid synthesis, Ca2+ homeostasis and the maintenance of synapses. However, the mechanisms underlying the regulation of axonal ER dynamics and its function in regulation of local translation still remain elusive. In the course of my thesis, I investigated the fast dynamic movements of ER and ribosomes in the growth cone of wildtype motoneurons as well as motoneurons from a mouse model of Spinal Muscular Atrophy (SMA), in response to Brain-derived neurotrophic factor (BDNF) stimulation. Live cell imaging data show that ER extends into axonal growth cone filopodia along actin filaments and disruption of actin cytoskeleton by cytochalasin D treatment impairs the dynamic movement of ER in the axonal filopodia. In contrast to filopodia, ER movements in the growth cone core seem to depend on coordinated actions of the actin and microtubule cytoskeleton. Myosin VI is especially required for ER movements into filopodia and drebrin A mediates actin/microtubule coordinated ER dynamics. Furthermore, we found that BDNF/TrkB signaling induces assembly of 80S ribosomes in growth cones on a time scale of seconds. Activated ribosomes relocate to the presynaptic ER and undergo local translation. These findings describe the dynamic interaction between ER and ribosomes during local translation and identify a novel potential function for the presynaptic ER in intra-axonal synthesis of transmembrane proteins such as the α-1β subunit of N-type Ca2+ channels in motoneurons. In addition, we demonstrate that in Smn-deficient motoneurons, ER dynamic movements are impaired in axonal growth cones that seems to be due to impaired actin cytoskeleton. Interestingly, ribosomes fail to undergo rapid structural changes in Smn-deficient growth cones and do not associate to ER in response to BDNF. Thus, aberrant ER dynamics and ribosome response to extracellular stimuli could affect axonal growth and presynaptic function and maintenance, thereby contributing to the pathology of SMA.
Ziel dieser Studie war es, zu untersuchen, ob dendritische Zellen eine Rolle beim ischämischen Schlaganfall spielen. Zur Beantwortung dieser Fragestellung wurde ein Mausmodell gewählt, in dem es nach Administration von Diphterietoxin zur selektiven Depletion CD11c positiver Zellen kommt (C.FVB-Tg(Itgax-DTR/EGFP)57Lan/J). Hierbei wird der Diphterietoxinrezeptor unter dem CD11c Promotor (ITGAX) exprimiert. Aufgrund der Wiederherstellung dendritischer Zellen nach ca. 24 Stunden waren wiederholte Applikationen von Diphterietoxin notwendig. Die Zusammensetzung anderer Immunzellen wurde dabei im Wesentlichen nicht geändert.
Für eine Schlaganfallinduktion wurde eine tMCAO (transient middle cerebral artery occlusion) durchgeführt. Hierbei wird durch Okklusion der A. cerebri media mittels Verschlussfilament für 30 oder 60 Minuten ein Schlaganfall im Mediastromgebiet induziert.
Es wurden unterschiedliche Verschlusszeiten, Zeitpunkte und Depletionsraten untersucht. In keinem der Versuchsansätze kam es zu einer signifikanten Veränderung des Schlaganfallvolumens nach Depletion CD11c positiver Zellen.
Mittels quantitativer real-time PCR wurde die Expression unterschiedlicher Zytokine nach tMCAO und CD11c-Depletion untersucht. An Tag 1 nach Schlaganfallinduktion und hoher Depletionsrate ergab sich eine Verminderung der Expression von IL-1β und IL-6, während an Tag 3 und niedriger Depletionsrate die Expression dieser Zytokine nach CD11c-Depletion zunahm. Grund hierfür könnte die Expression dieser Zytokine durch andere Zellen des Immunsystems, wie etwa neutrophile Granulozyten oder Mikroglia/Makrophagen sein, die möglicherweise einer regulatorischen Funktion durch die Interaktion von Dendritischen Zellen und regulatorischen T-Zellen unterliegen. Weitere experimentelle Ansätze sind notwendig, um diese Fragestellung beantworten zu können.
TGF-β zeigte durchgehend in allen Versuchsanordnungen eine verminderte Expression nach der Depletion dendritischer Zellen. Es ist naheliegend, dass dieses neuroprotektiv-regulatorische Zytokin direkt einer Produktion durch dendritische Zellen oder von nachfolgend aktivierten T-Zellen unterliegt.
In immunhistochemischen Studien konnte des Weiteren keine Änderung des Immigrationsverhaltens von CD11b+ Zellen ins Gehirn gesehen werden.
Diese Studie unterliegt jedoch einigen Limitationen. So stellte sich im Laufe der Experimente heraus, dass die wiederholte Applikation von Diphterietoxin zu einer erhöhten Mortalität der Versuchstiere führte. Nach Fertigstellung der Experimente erschien hierzu eine Publikation, welche die wiederholte Administration von DTX und die Entwicklung einer Myokarditis im gewählten Mausmodell in Zusammenhang brachte.
Die Arbeit untersucht die Leistungsrelevanz allgemeiner psychischer Leistungsfaktoren im Kontext der Talentförderung im weiblichen Nachwuchsfußball. Hierbei werden die Zusam- menhänge zwischen drei psychischen Dispositionen sowie einem psychischen Zustand und der aktuellen Spielkompetenz untersucht, wobei folgende Leistungsfaktoren fokussiert werden: die dispositionelle Handlungsorientierung, die dispositionelle Lageorientierung, die grundsätzliche Entscheidungskompetenz sowie das situative Flow-Erleben.
Auf Grundlage der Flow-Theorie und der Theorie der Handlungskontrolle sowie dem aktu- ellen Forschungsstand wird angenommen, dass mit steigendem Flow-Erleben eine Zunahme der aktuellen Spielkompetenz einhergeht und dieser psychische Zustand zudem den Zusam- menhang zwischen den ausgewählten Dispositionen und der aktuellen Spielkompetenz voll- ständig vermittelt. Die beiden Theorien und der aktuelle Forschungsstand lassen für eine aus- geprägte dispositionelle Handlungsorientierung sowie eine hohe Entscheidungskompetenz eine Zunahme und für eine ausgeprägte dispositionelle Lageorientierung eine Abnahme des Flow- Erlebens vermuten. Die Zusammenhänge werden mit und ohne vorheriger Stressinduktion und unter Berücksichtigung theoretisch abgeleiteter Kontrollvariablen untersucht.
Der Arbeit liegt eine quantitative Querschnittsstudie zugrunde, die bei 139 Nachwuchsspie- lerinnen aus acht verschiedenen Vereinen in Deutschland durchgeführt wurde. Die Datenaus- wertung erfolgt mittels multipler linearer Regressionsanalysen und linearer Strukturgleichungs- modelle.
Das Flow-Erleben erweist sich als leistungsförderlicher Zustand und verlässlicher Mediator des Zusammenhangs zwischen beiden dispositionellen Arten der Handlungskontrolle und der Spielkompetenz. Die dispositionelle Handlungsorientierung stellt einen positiven und die dis- positionelle Lageorientierung einen negativen Prädiktor für das Flow-Erleben dar. In Bezug auf die Entscheidungskompetenz können die theoretischen Annahmen nicht bestätigt werden. Auf dieser Grundlage erfolgen Implikationen für die sportpädagogische Praxis und die zukünftige Forschungsarbeit zum Flow-Erleben im weiblichen Nachwuchsfußball.
Development, Simulation and Evaluation of Mobile Wireless Networks in Industrial Applications

highly congested and guaranteeing the availability of wireless channels is becoming
increasingly difficult. In this work we show, that ad-hoc networking solutions can be
used to provide new communication channels and improve the performance of mobile
automation systems. These ad-hoc networking solutions describe different communi-
cation strategies, but avoid relying on network infrastructure by utilizing the Peer-to-
Peer (P2P) channel between communicating entities.
This work is a step towards the effective implementation of low-range communication
technologies(e.g. VisibleLightCommunication(VLC), radarcommunication, mmWave
communication) to the industrial application. Implementing infrastructure networks
with these technologies is unrealistic, since the low communication range would neces-
sitate a high number of Access Points (APs) to yield full coverage. However, ad-hoc
networks do not require any network infrastructure. In this work different ad-hoc net-
working solutions for the industrial use case are presented and tools and models for
their examination are proposed.
The main use case investigated in this work are Automated Guided Vehicles (AGVs)
for industrial applications. These mobile devices drive throughout the factory trans-
porting crates, goods or tools or assisting workers. In most implementations they must
exchange data with a Central Control Unit (CCU) and between one another. Predicting
if a certain communication technology is suitable for an application is very challenging
since the applications and the resulting requirements are very heterogeneous.
The proposed models and simulation tools enable the simulation of the complex inter-
action of mobile robotic clients and a wireless communication network. The goal is to
predict the characteristics of a networked AGV fleet.
Theproposedtoolswereusedtoimplement, testandexaminedifferentad-hocnetwork-
ing solutions for industrial applications using AGVs. These communication solutions
handle time-critical and delay-tolerant communication. Additionally a control method
for the AGVs is proposed, which optimizes the communication and in turn increases the
transport performance of the AGV fleet. Therefore, this work provides not only tools
for the further research of industrial ad-hoc system, but also first implementations of
ad-hoc systems which address many of the most pressing issues in industrial applica-
tions.
Die hier vorliegende Untersuchung beschäftigt sich mit dem Vergleich zweier minimalinvasiver operativer Therapiestrategien der distalen Unterarmfraktur als häufigster Fraktur im Kindesalter.
Für die vorliegende Studie erfolgte eine umfassende retrospektive Auswertung von 122 Fällen distaler kindlicher Unterarmfrakturen aus einem Zeitraum von elf Jahren, die in der kinderchirurgischen Abteilung des Universitätsklinikums Würzburg osteosynthetisch versorgt wurden. Ziel der Untersuchung war die Betrachtung der Epidemiologie des Verletzungsbildes und des Therapieerfolges in Abhängigkeit von der gewählten Osteosynthesetechnik. Die Kinder wurden dabei entweder allein durch eine Kirschner-Draht-Osteosynthese des Radius oder durch eine zusätzliche Fixierung der Ulna mittels elastisch stabiler intramedullärer Nagelung versorgt.
Durch beide Operationsstrategien konnte für die meisten Patientinnen und Patienten ein gutes Therapieergebnis erreicht werden. Hinsichtlich der Operations- und Hospitalisierungsdauer zeigte sich ein Vorteil für Kinder, die osteosynthetisch lediglich durch Kirschner-Drähte versorgt wurden. In anderen Studien beobachtete Komplikationen in Form sekundärer Dislokationen der Fraktur auch nach Osteosynthese oder Infektionen im Bereich der Drahtenden konnten im hier untersuchten Kollektiv seltener festgestellt werden.
Die vorliegende Untersuchung zeigt, dass, sofern die Indikation zur operativen Versorgung besteht, die Fixierung des Radius allein mittels Kirschner-Drähten eine adäquate Therapie distaler Frakturen von Ulna und Radius im Kindesalter darstellt. Von einer kombinierten Osteosynthese mittels Kirschner-Drähten und ESIN könnten besonders Patientinnen und Patienten profitieren, bei denen initial große Dislokationen der Fraktur, Begleitverletzungen des Arms oder eine Lokalisation im Bereich des diametaphysären Übergangs bestehen. Um diese Beobachtungen auch in größeren Kollektiven und prospektiv sowie randomisiert zu untersuchen, sind weitere Studien wünschenswert.
Cardiovascular disease and the acute consequence of myocardial infarc- tion remain one of the most important causes of morbidity and mortality in all western societies. While much progress has been made in mitigating the acute, life-threatening ischemia caused by infarction, heart failure of the damaged my- ocardium remains prevalent. There is mounting evidence for the role of T cells in the healing process after myocardial infarction, but relevant autoantigens, which might trigger and regulate adaptive immune involvement have not been discov- ered in patients.
In this work, we discovered an autoantigenic epitope in the adrenergic receptor beta 1, which is highly expressed in the heart. This autoantigenic epitope causes a pro-inflammatory immune reaction in T cells isolated from pa- tients after myocardial infarction (MI) but not in control patients. This immune reaction was only observed in a subset of MI patients, which carry at least one allele of the HLA-DRB1*13 family. Interestingly, HLA-DRB1*13 was more com- monly expressed in patients in the MI group than in the control group.
Taken together, our data suggests antigen-specific priming of T cells in MI patients, which leads to a pro-inflammatory phenotype. The primed T cells react to a cardiac derived autoantigen ex vivo and are likely to exhibit a similar phenotype in vivo. This immune phenotype was only observed in a certain sub- set of patients sharing a common HLA-allele, which was more commonly ex- pressed in MI patients, suggesting a possible role as a risk factor for cardiovas- cular disease.
While our results are observational and do not have enough power to show strong clinical associations, our discoveries provide an essential tool to further our understanding of involvement of the immune system in cardiovascu- lar disease. We describe the first cardiac autoantigen in the clinical context of MI and provide an important basis for further translational and clinical research in cardiac autoimmunity.
A disturbance in the symbiotic mutualism between the intestinal microbiome and the human host’s organism (syn. dysbiosis) accompanies the development of a variety of inflammatory and metabolic diseases that comprise the Metabolic Syndrome, chronic inflammatory gut diseases like Crohn’s disease, Non-alcoholic fatty liver disease (NAFLD) and cardiovascular diseases, among others. The changed uptake and effectiveness of short chain fatty acids (SCFAs) as well as an increase of the intestinal permeability are common, interdependent disease elements in this regard. Short chain fatty acids are end-products of intestinal bacterial fermentation and affect the mucosal barrier integrity via numerous molecular mechanisms.
There is evidence to suggest, that SCFAs have a modulating influence on Signal transducer and activator of transcription 3 (STAT3) in intestinal epithelial cells. STAT3 is a central gene-transcription factor in signaling pathways of proliferation and inflammation. It can be activated by growth factors and other intercellular signaling molecules like the cytokine Oncostatin M (OSM). The mode of STAT3’s activation exhibits, finally, a decisive influence on the immunological balance at the intestinal mucosa. Therefore, the posttranslational modification of STAT3 under the influence of SCFAs is likely to be a very important factor within the development and -progression of dysbiosis-associated diseases.
In this study, a clear positive in vitro-effect of the short chain fatty acid butyrate on the posttranslational serine727-phosphorylation of STAT3 and its total protein amount in the human adenocarcinoma cell line CACO2 is verified. Moreover, an increased gene expression of the OSM-receptor subunit OSMRβ can be observed after butyrate incubation. Histone deacetylase inhibition is shown to have a predominant role in these effects. Furthermore, a subsequent p38 MAPK-activation by Butyrate is found to be a key molecular mechanism regarding the STAT3-phosphorylation at serine727-residues. To consider the portion of butyrate receptor signaling in this context in future assays, a CACO-2 cell 3D-culture model is introduced in which an improvement of the GPR109A-receptor expression in CACO-2 cells is accomplished.
Sepsis ist eine dysregulierte Reaktion des Organismus auf eine Infektion. Bei Sepsis werden oft Blutungs- und Thromboseereignisse beobachtet, welche in einer Disseminierten Intravasalen Gerinnung (DIG) gipfeln können. Thrombozyten sind die Schlüsselzellen von Thrombose und Hämostase. Bei Sepsis und DIG kommt es häufig zu einem Abfall der Thrombozytenzahl, doch Blutungs- und Thromboseereignisse können unabhängig von der Thrombozytenzahl auftreten, was zusätzlich eine Veränderung der Thrombozytenfunktion nahelegt.
In dieser Arbeit wurde deshalb die Thrombozytenfunktion bei 15 Patienten mit Sepsis zu drei Zeitpunkten im Krankheitsverlauf untersucht. Es konnte bei unauffälliger Rezeptorexpression keine Voraktivierung der Thrombozyten mittels Durchflusszytometrie festgestellt werden. Jedoch war die Aktivierung nach Stimulation mit multiplen Agonisten signifikant reduziert. Besonders ausgeprägt war die Hyporeaktivität bei Stimulation des Kollagen-Rezeptors GPVI mit dem Agonisten CRP-XL. Es wurde gezeigt, dass nach GPVI-Stimulation eine reduzierte Phosphorylierung der nachgeschalteten Proteine Syk und LAT im Vergleich zum Gesundspender induziert wird. In Kreuzinkubationsexperimenten hatte die (Co )Inkubation von Thrombozyten in Plasma von Sepsispatienten oder mit Bakterienisolaten aus Sepsis-Blutkulturen keinen Effekt auf die Thrombozytenreaktivität. Allerdings konnte durch Sepsis-Vollblut eine signifikante GPVI-Hyporeaktivität in Thrombozyten von gesunden Probanden induziert werden, was einen zellulären Mediator als Ursache des Defekts nahelegt. In dieser Arbeit wurde gezeigt, dass insbesondere die GPVI-Signalkaskade bei Sepsis massiv beeinträchtigt ist. Der Immunorezeptor GPVI ist ein vielversprechendes Zielmolekül, um die Pathogenese der Sepsis, des Capillary Leak und die immunregulatorische Rolle von Thrombozyten besser zu verstehen. Die GPVI-Hyporeaktivität könnte als zukünftiger Biomarker für die Sepsis-Frühdiagnose genutzt werden.
In der vorliegenden Arbeit werden die Konzeption und Realisierung eines Computertomographen zur Materialanalyse auf Basis eines Rasterelektronenmikroskops mit einem räumlichen Auflösungsvermögen im Nanometerbereich diskutiert. Durch einen fokussierten Elektronenstrahl, der mit einer Beschleunigungsspannung von 30 kV auf eine mikrostrukturierte Wolframnadel mit einem Spitzenradius von bis zu 50 nm gezielt wird, entsteht ein kleiner Röntgenbrennfleck über den mit geometrischer Vergrößerung hochauflösende Projektionen eines zu untersuchenden Objekts erzeugt werden. Durch Rotation des Testobjekts werden Projektionen aus verschiedenen Blickwinkeln aufgenommen und über einen speziellen Rekonstruktionsalgorithmus zu einem 3-dimensionalen Bild zusammengefügt.
Bei der Beurteilung der Einzelkomponenten des Geräts wird insbesondere auf Struktur, Form und den elektrochemischen Herstellungsprozess der Röntgenquelle eingegangen. Eine ausreichend genaue Positionierung von Messobjekt und Röntgenbrennfleck wird über Piezoachsen realisiert, während die Stabilität des Röntgenbrennflecks über die Elektronenoptik des Rasterelektronenmikroskops und die Form der Quellnadel optimiert wird.
Das räumliche Auflösungsvermögen wird über die Linienspreizfunktion an Materialkanten abgeschätzt. Für eine Wolfram-Block-Quelle ergibt sich dabei ein Auflösungsvermögen von 325 nm – 400 nm in 3D, während der Quellfleck einer Wolframnadel das Auflösungsvermögen der Anlage auf 65 nm – 90 nm in 2D und 170 nm – 300 nm in 3D bei Messungen an einem AlCu29-Testobjekt anhebt. Außerdem werden die Auswirkungen der Phasenkontrastcharakteristik der Röntgenquelle auf die rekonstruierten Bilder nach Anwendung eines Paganin-Filters diskutiert. Dabei zeigt sich, dass durch Anwendung des Filters ein verbessertes Signal-zu-Rausch-Verhältnis auf Kosten der räumlichen Bildauflösung erzielt werden kann.
Eine Vergleichsmessung mit einem kommerziell verfügbaren Röntgenmikroskop zeigt die Stärken des vorgestellten Systems bei Untersuchung von stark absorbierenden Messobjekten. Das kompakte Design erlaubt eine Weiterentwicklung in Richtung eines nanoCT-Moduls als Upgrade Option für Rasterelektronenmikroskope im Gegensatz zu den weitaus teureren bisher verbreiteten nanoCT-Geräten.
Mammalian embryonic development is subject to complex biological relationships that need to be understood. However, before the whole structure of development can be put together, the individual building blocks must first be understood in more detail. One of these building blocks is the second cell fate decision and describes the differentiation of cells of the inner cell mass of the embryo into epiblast and primitive endoderm cells. These cells then spatially segregate and form the subsequent bases for the embryo and yolk sac, respectively. In organoids of the inner cell mass, these two types of progenitor cells are also observed to form, and to some extent to spatially separate. This work has been devoted to these phenomena over the past three years. Plenty of studies already provide some insights into the basic mechanics of this cell differentiation, such that the first signs of epiblast and primitive endoderm differentiation, are the expression levels of transcription factors NANOG and GATA6. Here, cells with low expression of GATA6 and high expression of NANOG adopt the epiblast fate. If the expressions are reversed, a primitive endoderm cell is formed. Regarding the spatial segregation of the two cell types, it is not yet clear what mechanism leads to this. A common hypothesis suggests the differential adhesion of cell as the cause for the spatial rearrangement of cells. In this thesis however, the possibility of a global cell-cell communication is investigated. The approach chosen to study these phenomena follows the motto "mathematics is biology's next microscope". Mathematical modeling is used to transform the central gene regulatory network at the heart of this work into a system of equations that allows us to describe the temporal evolution of NANOG and GATA6 under the influence of an external signal. Special attention is paid to the derivation of new models using methods of statistical mechanics, as well as the comparison with existing models. After a detailed stability analysis the advantages of the derived model become clear by the fact that an exact relationship of the model parameters and the formation of heterogeneous mixtures of two cell types was found. Thus, the model can be easily controlled and the proportions of the resulting cell types can be estimated in advance. This mathematical model is also combined with a mechanism for global cell-cell communication, as well as a model for the growth of an organoid. It is shown that the global cell-cell communication is able to unify the formation of checkerboard patterns as well as engulfing patterns based on differently propagating signals. In addition, the influence of cell division and thus organoid growth on pattern formation is studied in detail. It is shown that this is able to contribute to the formation of clusters and, as a consequence, to breathe some randomness into otherwise perfectly sorted patterns.
The aim of this study was to provide a comprehensive overview of the frontal bone in the forensic context with special emphasis on its shape. Analyses on 19th and 20th century Euro-American and German crania were performed in terms of population differences, sexual dimorphism, secular change, and metopism.
It could clearly be seen that the frontal bone on its own already provides a lot of information toward the biological profile of an individual. Overall, using size and shape combined for analyses would always produce the best results, followed by shape only and then size only. Nevertheless, Log_Centroid_Size was the best sex-discriminating variable in the size-shape combined analyses for both populations. Population differences as well as sexual dimorphism could both be assessed (with varying accuracy) using size only and shape only respectively.
Very little secular change between the 19th and 20th century was found for the frontal in both groups respectively, with the secular change that could be seen mostly being shape variation.
Metopism analysis was only performed on German crania, because the presence or absence of a metopic suture was not documented for the Euro-American crania. Unfortunately, the results of these analyses were very limited due to too small sample sizes for the overall low percentage of metopism. The metopic frontal was once again found to be short in relation to its width and presenting a more rounded frontal curvature. The attempt of creating a formula to morphometrically assess the presence of metopism was not successful.
The results of this thesis suggest that forensic case work on skeletal remains would greatly benefit from a broader application of Geometric Morphometrics and consequently from larger databases containing shape data as well as more advanced and user-friendly software for this type of analyses.

Ziel ist es, Schüler:innen im inklusiven Anfangsunterricht in multimodale Erzählmöglichkeiten der App Book Creator einzuführen (Kapitel eins und zwei), mit denen sie anschließend digital eine Geschichte weitererzählen können (Kapitel drei und vier). In Kapitel fünf wird den Schüler:innen ein mögliches Ende der Geschichte angeboten. Alle Materialien wurden mit Blick auf die oben genannten spezifischen Zielgruppen entwickelt und erprobt. Sie sind im Rahmen des BMBF-geförderten Projekts CoTeach-Arbeitspaket 4 „Medienkompetenzen in inklusiven Grundschulklassen im Bereich Digital Storytelling“ entstanden, das von Prof Dr. Sanna Pohlmann-Rother und Prof. Dr. Christoph Ratz geleitet wird.
Obwohl die europäische Verwaltung und ihr Verwaltungsverfahren zunehmend an Bedeutung gewinnt, fehlt eine Generalkodifikation des Verwaltungsverfahrensrechts auf EU-Ebene. Die vorliegende Arbeit zielt darauf ab, den aktuellen Kodifikationsstand und die Möglichkeiten einer zukünftigen Kodifikation zu erörtern. Insbesondere werden die rechtlichen Rahmenbedingungen einer Kodifikation untersucht, wobei der Schwerpunkt auf der Frage liegt, inwieweit die EU eine Kompetenz für eine solche besitzt. Daneben werden die rechtlichen Grenzen aus mitgliedstaatlicher Sicht am Beispiel Deutschland erörtert. Auf dieser Grundlage wird anschließend dargelegt, in welcher Form eine Kodifikation theoretisch umgesetzt werden kann. Außerdem werden drei aktuelle Kodifikationsentwürfe in ihren Grundzügen beschrieben, miteinander verglichen und kritisch beleuchtet. Anschließend werden die wesentlichen Punkte im Bereich Kodifikationsfähigkeit und -bedürftigkeit dargestellt, bevor als Abschluss die generellen Verwirklichungschancen einer Kodifikation sowie die Verwirklichungschancen der drei Entwürfe beurteilt werden.
The fast and accurate yield estimates with the increasing availability and variety of global satellite products and the rapid development of new algorithms remain a goal for precision agriculture and food security. However, the consistency and reliability of suitable methodologies that provide accurate crop yield outcomes still need to be explored. The study investigates the coupling of crop modeling and machine learning (ML) to improve the yield prediction of winter wheat (WW) and oil seed rape (OSR) and provides examples for the Free State of Bavaria (70,550 km2), Germany, in 2019. The main objectives are to find whether a coupling approach [Light Use Efficiency (LUE) + Random Forest (RF)] would result in better and more accurate yield predictions compared to results provided with other models not using the LUE. Four different RF models [RF1 (input: Normalized Difference Vegetation Index (NDVI)), RF2 (input: climate variables), RF3 (input: NDVI + climate variables), RF4 (input: LUE generated biomass + climate variables)], and one semi-empiric LUE model were designed with different input requirements to find the best predictors of crop monitoring. The results indicate that the individual use of the NDVI (in RF1) and the climate variables (in RF2) could not be the most accurate, reliable, and precise solution for crop monitoring; however, their combined use (in RF3) resulted in higher accuracies. Notably, the study suggested the coupling of the LUE model variables to the RF4 model can reduce the relative root mean square error (RRMSE) from −8% (WW) and −1.6% (OSR) and increase the R
2 by 14.3% (for both WW and OSR), compared to results just relying on LUE. Moreover, the research compares models yield outputs by inputting three different spatial inputs: Sentinel-2(S)-MOD13Q1 (10 m), Landsat (L)-MOD13Q1 (30 m), and MOD13Q1 (MODIS) (250 m). The S-MOD13Q1 data has relatively improved the performance of models with higher mean R
2 [0.80 (WW), 0.69 (OSR)], and lower RRMSE (%) (9.18, 10.21) compared to L-MOD13Q1 (30 m) and MOD13Q1 (250 m). Satellite-based crop biomass, solar radiation, and temperature are found to be the most influential variables in the yield prediction of both crops.
The landscape of today’s programming languages is manifold. With the diversity of applications, the difficulty of adequately addressing and specifying the used programs increases. This often leads to newly designed and implemented domain-specific languages. They enable domain experts to express knowledge in their preferred format, resulting in more readable and concise programs. Due to its flexible and declarative syntax without reserved keywords, the logic programming language Prolog is particularly suitable for defining and embedding domain-specific languages.
This thesis addresses the questions and challenges that arise when integrating domain-specific languages into Prolog. We compare the two approaches to define them either externally or internally, and provide assisting tools for each. The grammar of a formal language is usually defined in the extended Backus–Naur form. In this work, we handle this formalism as a domain-specific language in Prolog, and define term expansions that allow to translate it into equivalent definite clause grammars. We present the package library(dcg4pt) for SWI-Prolog, which enriches them by an additional argument to automatically process the term’s corresponding parse tree. To simplify the work with definite clause grammars, we visualise their application by a web-based tracer.
The external integration of domain-specific languages requires the programmer to keep the grammar, parser, and interpreter in sync. In many cases, domain-specific languages can instead be directly embedded into Prolog by providing appropriate operator definitions. In addition, we propose syntactic extensions for Prolog to expand its expressiveness, for instance to state logic formulas with their connectives verbatim. This allows to use all tools that were originally written for Prolog, for instance code linters and editors with syntax highlighting. We present the package library(plammar), a standard-compliant parser for Prolog source code, written in Prolog. It is able to automatically infer from example sentences the required operator definitions with their classes and precedences as well as the required Prolog language extensions. As a result, we can automatically answer the question: Is it possible to model these example sentences as valid Prolog clauses, and how?
We discuss and apply the two approaches to internal and external integrations for several domain-specific languages, namely the extended Backus–Naur form, GraphQL, XPath, and a controlled natural language to represent expert rules in if-then form. The created toolchain with library(dcg4pt) and library(plammar) yields new application opportunities for static Prolog source code analysis, which we also present.
Einleitung: Multiple Sklerose gilt als häufigste neurologische Erkrankung der Gruppe junger Erwachsener. Die chronisch-entzündliche Erkrankung befällt das zentrale Nervensystem in Form von Demyelinisierungen. In der Altersgruppe der 20-40 Jährigen tritt die Erstmanifestation der Multiplen Sklerose am häufigsten auf. Diese Altersklasse ist besonders stark im Internet vertreten. Die vorliegende Studie untersuchte sozio-demografische Faktoren und das Internetverhalten von MS Patienten. Im Rahmen einer dreimonatigen Pilotstudie wurde eine geheime Facebookgruppe eingerichtet und mit aktuellen sowie grundlegenden Inhalten rund um Multiple Sklerose unterhalten.
Methoden: Im Rahmen einer postalischen Fragebogenaktion wurden 1437 Multiple Sklerose Patienten kontaktiert. Der Eingangsfragebogen, der als Selbstausfüllungs-instrument gestaltet wurde, ermittelte soziodemografische Patientencharakteristiken und das Nutzungsverhalten der sozialen Medien. Die Rücksenderate entsprach 32 % (n=486 Patienten). Aus dieser Studiengesamtpopulation wurden 39 Patienten in eine Pilotstudie eingeschlossen. Das Patientenkollektiv bestand aus 21 Studienteilnehmern der Interven-tionsgruppe und 18 Studienteilnehmern der Kontrollgruppe. Vor und nach der Interven-tion kamen ein Adhärenzfragebogen, die MFIS-D und das BDI zum Einsatz.
Ergebnisse: Das Durchschnittsalter der Gesamtpopulation lag bei 41 Jahren und 77,2 % gehörten dem weiblichen Geschlecht an. Keine erschwerte Medikamenteneinnahme gaben 56,9 % der befragten Personen an. Soziale Netzwerke wurden von 53,5 % der Untersuchten genutzt, wobei davon 61,6 % täglich soziale Medien aufriefen. Facebook konnte mit 78,9 % der Patienten als meistgenutzte Social Media Plattform ermittelt werden. Der häufigste Beitrittsgrund zu Facebook war der Kontakt zu Freunden (79,5 %). Die Lebensqualität der Gesamtstudienpopulation wurde am meisten durch den Multiple Sklerose Kenntnisstand und den EDSS Grad beeinflusst. Die soziodemo-grafischen Faktoren Alter, Berufstätigkeit und Kinder konnten als Variablen, die mit der Nutzung sozialer Medien im Zusammenhang stehen, identifiziert werden. Geschlecht und Schulabschluss nahmen Einfluss auf die subjektive Einschätzung der Multiple Sklerose Grundlagenkenntnisse. Das Facebookpilotprojekt untersuchte vor (Z1) und nach (Z2) der Intervention den Adhärenztyp, den Fatiguegrad und den Depressionsgrad. Durch die Intervention konnte keinerlei Veränderung bezüglich Adhärenz, Fatigue und Depression festgestellt werden.
Diskussion: Soziale Medien dienen vorrangig dem sozialen Austausch als dem Transport gesundheitsrelevanter Informationen. Ziel in der Zukunft müssen maßgeschneiderte E-Health Plattformen sein, die auf ärztlichem Background basieren. Wichtig ist es dabei, dass bestimmte Rahmenbedingungen sichergestellt werden. Hierzu gehören unter anderem Datenschutz, Autorentransparenz und Aktualität. In anderen Bereichen mit speziellen Schwerpunkten wurden bereits Versuche der onlinebasierten Patientenbetreuung durchgeführt. Trotz der positiven Einflüsse, die soziale Medien auf das Gesundheitsverhalten der Patienten haben können, ist der physische Kontakt zum Arzt derzeit noch nicht aus dem Behandlungskonzept wegzudenken.
Mit steigender Nachfrage an minimal-invasiven Therapieoptionen wächst auch das Interesse an innovativen Alternativen im Bereich des arteriellen Gefäßverschlusses nach PVI. Ziel der vorliegenden Arbeit war es, die Effektivität und Sicherheit eines neu auf dem Markt befindlichen, Doppelclip-basierten aktiven VVS zu prüfen. Eine hohe technische Erfolgsrate von 98,8 % bei einer geringen Komplikationsrate von 3,6 % wurde verzeichnet. Bei Anwesenheit der Komorbiditäten Dm und CNI 5 zeigte sich eine signifikante Assoziation zu einem vermehrten Auftreten von Komplikationen. Ein nachgewiesener signifikanter Zusammenhang bestand zudem zwischen einem erhöhten Kalzifikationsgrad der Punktionsstelle bei Vorliegen einer pAVK und eines Dm. Eine erhöhte Gefäßrigidität aufgrund von Komorbiditäten und vaskulären Kalzifikationen, intrinsische Fremdkörperreaktionen, vasoregulatorische Reaktionen oder Produktversagen bieten hypothetische Erklärungsansätze für die einzelnen komplikativen Fälle. Die Nutzung des VVS in spezifischen, bisher nicht beschriebenen Situationen (Unterdimensionierung, Anwendung nach Gefäßpunktionen mit Zugangsschleusen bis 9F, antegrade Punktionsrichtung, anspruchsvolle Eingriffe multimorbider Patienten mit komplexem vaskulärem Status) erwies sich als suffizient. Zur Prävention schwerwiegender Komplikationen während zukünftiger Interventionen wurden die Empfehlung ausgesprochen, eine Durchleuchtungsaufnahme zur Lagekontrolle vor Implantation des proximalen Clips anzufertigen. Eine speziell für Gefäßverschlüsse nach antegrader Punktion konzipierte Zugangsschleuse könnte das Abknicken von Zugangsschleusen bei VVS Applikation verhindern. Zusammenfassend kann das untersuchte aktive VVS mit einzigartigem Wirkmechanismus und spezifischen Design als effektiv und sicher angesehen werden.
Facing Enemies. Modulation of Revenge Interactions based on Opponent State Indicators of Suffering

Research on revenge often treats vengeful acts as singular one-way experiences, an approach which fails to account for the social nature and functions of revenge. This dissertation aims to integrate emotional punishment reactions into dynamic revenge sequences to investigate the affective and cognitive consequences of revenge within a social interaction.
Exacting revenge can evoke intense affective consequences, from feelings of guilt to the genuine enjoyment of the suffering of others. In Chapter 2, affective responses towards suffering opponents and the regulation of aggression based on the appraisal of distinct suffering indicators were investigated. Results indicate that the observation of opponent pain evokes positive affect (measured via facial muscle contractions during the observation), which is followed by a downregulation of subsequent punishment. Both, positive affective reactions and the downregulation of punishment, were only observed following pain and not sadness expressions. Empathic distress, indexed by negative affective reactions, was only present following the observation of pain in non-provoking opponents. Showcasing the modulation of empathy related processes due to provocation and competition.
In Chapter 3, a significant escalation of punishment, when being confronted with Schadenfreude, was observed. Results are interpreted as supporting the assumption that opponent monitoring processes inform subsequent action selection. The observation of opponent smiles led to imitation behavior (facial mimicry), which was partially attenuated due to previous provocation. The different functions of smile mimicry in the context of the aggressive competitive setting are discussed as containing simulation aspects (to aid in opponent understanding) and as a potential mirroring of dominance gestures, to avoid submission.
In an additional series of studies, which are presented in Chapter 4, changes in memory of opponent faces following vengeful encounters were measured. Based on provocation, and punishment outcomes (pain & anger), face memory was distorted, resulting in more positive representations of opponents that expressed pain. These results are discussed as evidence of the impact of outcome appraisals in the formation of opponent representations and are theorized to aid empathy avoidance in future interactions.
The comparison of desired and observed opponent states, is theorized to result in appraisals of the punishment outcomes, which evoke affective states, inform the action selection of subsequent punishments, and are integrated into the representation of the opponent in memory.
Overall, the results indicate that suffering cues that are congruent with the chosen punishment action are appraised as positive, evoking an increase in positive affect. The emergence of positive affect during the observation of successful aggressive actions supports recent theories about the chronification of aggressive behavior based on reinforcement learning. To allow positive affect to emerge, affective empathic responses, such as distress, are theorized to be suppressed to facilitate the goal attainment process. The suffering of the opponent constitutes the proximate goal during revenge taking, which highlights the importance of a theoretical differentiation of proximate and ultimate goals in revenge to allow for a deeper understanding of the underlying motives of complex revenge behavior.
Mammalian phoshoglycolate phosphatase (PGP, also known as AUM) belongs to the ubiquitous HAD superfamily of phosphatases. As several other members of HAD phosphatases, the Mg2+-dependent dephosphorylation is conducted via a nucleophilic attack from a conserved aspartate residue in the catalytic cleft. The protein structure of PGP could not yet be solved entirely. Only a hybrid consisting of the PGP cap and the PDXP core (pyridoxal phosphatase, closest enzyme paralog) was crystallizable so far. PGP is able to efficiently dephosphorylate 2-phosphoglycolate, 2-phospho-L-lactate, 4-phospho-D-erythronate, and glycerol-3-phosphate in vitro which makes them likely physiological substrates. The first three substrates can be derived from metabolic side reactions (during glycolysis) and inhibit key enzymes in glycolysis and pentose phosphate pathway, the latter is situated at the intersection between glycolysis and lipogenesis. 2-phosphoglycolate can also be released in the context of repair of oxidative DNA damage. The activity of purified PGP can be reversibly inhibited by oxidation - physiologically likely in association with epidermal growth factor (EGF) signal transduction. In fact, an association between persistently lacking PGP activity (via downregulation) and the presence of hyperphosphorylated proteins after EGF stimulation has been identified. Reversible oxidation and transient inactivation of PGP may be particularly important for short-term and feedback regulatory mechanisms (as part of the EGF signaling). Furthermore, cellular proliferation in PGP downregulated cells is constantly reduced. Whole-body PGP inactivation in mice is embryonically lethal. Despite the many well-known features and functions, the knowledge about PGP is still incomplete.
In the present work the influence of reactive oxygen species (ROS) on PGP activity in cells und a possible connection between oxidative stress and the proliferation deficit of PGP downregulated cells was investigated. For the experiments, a spermatogonial cell line was used (due to the high PGP expression in testis). PGP activity can be reversibly inhibited in cellular lysates by H2O2 (as a ROS representative). Reversible oxidation could thus indeed be physiologically important. More oxidative DNA damage (by bleomycin) showed no PGP-dependent effects here. EGF stimulation (as an inducer of transient and well-controlled ROS production), low concentrations of menadione (as an oxidant) and N-acetylcysteine (as an antioxidant) were able to approximate the proliferation rate in PGP downregulated cells to that of control cells. The redox regulation of PGP could thus have an influence on cellular proliferation as a feedback mechanism - a mechanism that could not take place in PGP downregulated cells. However, the connections are probably even more complex and cannot be elucidated by a sole examination of the proliferation rate. The present results can thus only be regarded as preliminary experiments.
For a better understanding of the features and functions of PGP, this work then focused on specific regulation of enzyme activity by pharmacologically applicable small molecules. Four potent inhibitors had previously been identified in a screening campaign. In this work, three of these four inhibiting compounds could be further characterized in experiments with highly purified, recombinant murine and human PGP. Compounds #2 and #9 showed competitive inhibition properties with a markedly rising KM value with little or no change in vmax. The results were consistent for all tested protein variants: the murine and the human PGP as well as a PGP/PDXP hybrid protein. Compound #1 was the most potent and interesting PGP-inhibitory molecule: less change in KM and a constant decrease in vmax as well as a lower impact on the PGP/PDXP hybrid hint at a mixed mode of inhibition as a combination of competitive and non-competitive inhibition. The characterization of the potential inhibitors can serve as a basis for further structural analysis and studies on the complex physiological role of PGP.
Emotional-associative learning processes such as fear conditioning and extinction are highly relevant to not only the development and maintenance of anxiety disorders (ADs), but also to their treatment. Extinction, as the laboratory analogue to behavioral exposure, is assumed a core process underlying the treatment of ADs. Although exposure-based treatments are highly effective for the average patient suffering from an AD, there remains a gap in treatment efficacy with over one third of patients failing to achieve clinically significant symptom relief. There is ergo a pressing need for intensified research regarding the underlying neural mechanisms of aberrant emotional-associative learning processes and the neurobiological moderators of treatment (non-)response in ADs.
The current thesis focuses on different applications of the fundamental principles of fear conditioning and extinction by using two example cases of ADs from two different multicenter trials. First, we targeted alterations in fear acquisition, extinction, and its recall as a function of psychopathology in panic disorder (PD) patients compared to healthy subjects using fMRI. Second, exposure-based therapy and pre-treatment patient characteristics exerting a moderating influence on this essential learning process later on (i.e. treatment outcome) were examined using multimodal functional and structural neuroimaging in spider phobia.
We observed aberrations in emotional-associative learning processes in PD patients compared to healthy subjects indicated by an accelerated fear acquisition and an attenuated extinction recall. Furthermore, pre-treatment differences related to defensive, regulatory, attentional, and perceptual processes may exert a moderating influence on treatment outcome to behavioral exposure in spider phobia. Although the current results need further replication, on an integrative meta level, results point to a hyperactive defensive network system and deficient emotion regulation processes (including extinction processes) and top-down control in ADs. This speaks in favor of transdiagnostic deficits in important functional domains in ADs.
Deficits in transdiagnostic domains such as emotion regulation processes could be targeted by enhancing extinction learning or by means of promising tools like neurofeedback. The detection of pre-treatment clinical response moderators, for instance via machine learning frameworks, may help in supporting clinical decision making on individually tailored treatment approaches or, respectively, to avoid ineffective treatment and its related financial costs. In the long run, the identification of neurobiological markers which are capable of detecting non-responders a priori represents an ultimate goal.
Einflüsse der Photophysik und Photochemie von Cyaninfarbstoffen auf die Lokalisationsmikroskopie

In den letzten Jahren haben sich hochauflösende Fluoreszenzmikroskopiemethoden, basierend auf der Lokalisation einzelner Fluorophore, zu einem leistungsstarken Werkzeug etabliert, um Fluoreszenzbilder weit unterhalb der Auflösungsgrenze zu generieren. Hiermit können räumliche Auflösungen von ~ 20 nm erzielt werden, was weit unterhalb der Beugungsgrenze liegt. Dabei haben zahlreiche Optimierungen und Entwicklungen neuer Methoden in der Einzelmolekül-Lokalisationsmikroskopie die Genauigkeit der orstspezifischen Bestimmung einzelner Fluorophore auf bis zu ~ 1 – 3 nm erhöht. Eine Auflösung im molekularen Bereich, weit unterhalb von ~ 10 nm bleibt allerdings herausfordernd, da die Lokalisationsgenauigkeit nur ein Kriterium hierfür ist. Allerdings wurde sich in den letzten Jahren überwiegend auf die Verbesserung dieses Parameters konzentriert. Weitere Kriterien für die fluoreszenzmikroskopische Auflösung sind dabei unter anderem die Markierungsdichte und die Kopplungseffizienz der Zielstruktur, sowie der Kopplungsfehler (Abstand zur Zielstruktur nach Farbstoffkopplung), die sich herausfordernd für eine molekulare Auflösung darstellen. Auch wenn die Kopplungseffizienz und -dichte hoch und der Kopplungsfehler gering ist, steigt bei Interfluorophordistanzen < 5nm, abhängig von den Farbstoffen, die Wahrscheinlichkeit von starken und schwachen Farbstoffwechselwirkungen und damit von Energieübertragungsprozessen zwischen den Farbstoffen, stark an. Daneben sollten Farbstoffe, abhänging von der Lokalisationsmikroskopiemethode, spezifische Kriterien, wie beispielsweise die Photoschaltbarkeit bei dSTORM, erfüllen, was dazu führt, dass diese Methoden häufig nur auf einzelne Farbstoffe beschränkt sind. In dieser Arbeit konnte mithilfe von definierten DNA-Origami Konstrukten gezeigt werden, dass das Blinkverhalten von Cyaninfarbstoffen unter dSTORM-Bedingungen einer Abstandsabhängigkeit aufgrund von spezifischen Energieübertragungsprozessen folgt, womit Farbstoffabstände im sub-10 nm Bereich charakterisiert werden konnten. Darüber hinaus konnte diese Abstandsabhängigkeit an biologischen Proben gezeigt werden. Hierbei konnten verschiedene zelluläre Rezeptoren effizient und mit geringem Abstandsfehler zur Zielstruktur mit Cyaninfarbstoffen gekoppelt werden. Diese abstandsabhänigen Prozesse und damit Charakterisierungen könnten dabei nicht nur spezifisch für die häufig unter dSTORM-Bedingungen verwendeten Cyaninfarbstoffen gültig sein, sondern auch auf andere Farbstoffklassen, die einen Auszustand zeigen, übertragbar sein. Darüber hinaus konnte gezeigt werden, dass hochauflösende dSTORM Aufnahmen unabhängig vom Farbstoffkopplungsgrad der Antikörpern sind, welche häufig für Standardfärbungen von zellulären Strukturen verwendet werden. Dabei konnte durch Photonenkoinzidenzmessungen dargelegt werden, dass aufgrund komplexer Farbstoffwechselwirkungen im Mittel nur ein Farbstoff aktiv ist, wobei höhere Kopplungsgrade ein komplexes Blinkverhalten zu Beginn der Messung zeigen. Durch die undefinierten Farbstoffabstände an Antikörpern konnte hier kein eindeutiger Energieübertragungsmechanismus entschlüsselt werden. Dennoch konnte gezeigt werden, dass Farbstoffaggregate bzw. H-Dimere unter dSTORM-Bedingungen destabilisiert werden. Durch die zuvor erwähnten DNA-Origami Konstrukte definierter Interfluorophordistanzen konnten Energieübertragungsmechanismen entschlüsselt werden, die auch für die Antikörper diverser Kopplungsgrade gültig sind. Des Weiteren konnten, ausgelöst durch komplexe Energieübertragungsprozesse höherer Kopplungsgrade am Antikörper, Mehrfarbenaufnahmen zellulärer Strukturen generiert werden, die über die spezifische Fluoreszenzlebenszeit separiert werden konnten. Dies stellt hier eine weitere Möglichkeit dar, unter einfachen Bedingungen, schnelle Mehrfarbenaufnahmen zellulärer Strukturen zu generieren. Durch die Verwendung des selben Farbstoffes unterschiedlicher Kopplungsgrade kann hier nur mit einer Anregungswellenlänge und frei von chromatischer Aberration gearbeitet werden. Neben den photophysikalischen Untersuchungen der Cyaninfarbstoffe Cy5 und Alexa Fluor 647 wurden diese ebenso photochemisch näher betrachtet. Dabei konnte ein neuartiger chemischer Mechanismus entschlüsselt werden. Dieser Mechanismus führt, ausgelöst durch Singulett-Sauerstoff (1O2), zu einer Photozerschneidung des konjugierten Doppelbindungssystems um zwei Kohlenstoffatome, was zu strukturellen und spektroskopischen Veränderungen dieser Farbstoffe führt. Auf Grundlage dieses Mechanismus konnte eine neue DNA-PAINT Methode entwickelt werden, die zu einer Beschleunigung der Aufnahmezeit führt.
To study coisotropic reduction in the context of deformation quantization we introduce constraint manifolds and constraint algebras as the basic objects encoding the additional information needed to define a reduction. General properties of various categories of constraint objects and their compatiblity with reduction are examined. A constraint Serre-Swan theorem, identifying constraint vector bundles with certain finitely generated projective constraint modules, as well as a constraint symbol calculus are proved. After developing the general deformation theory of constraint algebras, including constraint Hochschild cohomology and constraint differential graded Lie algebras, the second constraint Hochschild cohomology for the constraint algebra of functions on a constraint flat space is computed.
Der postoperative Hypoparathyreoidismus (PH) stellt eine der häufigsten Komplikationen nach Schilddrüsenoperationen dar. Ziel dieses systematischen Reviews und Metaanalyse ist die Erarbeitung einer einheitlichen Definition sowie die Ermittlung des bestmöglichen Ansatzes für eine frühzeitige Detektion des PH.
Nach Durchführung einer systematischen Literaturrecherche gemäß der PICo-Systematik unter Verwendung der Datenbanken Embase, Pubmed und der Cochrane Library, erfolgte die themenbezogene Aufarbeitung der eingeschlossenen Studien, sowie eine Bias-Bewertung und Metaanalyse geeigneter Arbeiten.
Von 13.704 Artikeln konnten 188 in die weitere Analyse eingeschlossen werden. In diesen fanden sich sehr heterogene Definitionen des PH. Sowohl in der systematischen Analyse als auch in der Metaanalyse zeigte sich eine genauere Vorhersagekraft des PH durch eine postoperative im Vergleich zu einer intraoperativen PTH-Messung. Keiner der analysierten Zeiträume innerhalb des ersten postoperativen Tages (POD1) zeigte eine signifikante Überlegenheit in der Vorhersage eines PH. Die PTH- Schwellenwerte 10 bzw. 15 pg/ml können einen PH zuverlässig detektieren. Als Entscheidungsgrundlage zwischen den beiden Werten kann die untere Normwertgrenze des angewendeten Testverfahrens herangezogen werden. Bei präoperativer PTH-Abnahme nach Anästhesieeinleitung ist ein relativer PTH-Abfall von prä- nach postoperativ von 73 ± 11% prädiktiv für die Entwicklung eines PH. Die Bestimmung des Calciumspiegels an POD1 ist obligat und optimiert insbesondere die Erkennung einer biochemischen Hypokalzämie.
Ein nicht nachweisbarer oder inadäquat niedriger postoperativer PTH-Spiegel im Zusammenhang mit einer biochemischen oder symptomatischen Hypokalzämie kann als einheitliche Definition des postoperativen Hypoparathyreoidismus vorgeschlagen werden. Die Messung des Parathormons sollte zwischen einer und sechs Stunden postoperativ, spätestens aber innerhalb von 24 Stunden erfolgen. Sowohl der Schwellenwert ≤ 15 pg/ml als auch ein relativer PTH-Abfall von prä- nach postoperativ sind zuverlässig in der Detektion gefährdeter Patienten.
Despite accounting for only a small proportion of all skin cancers, malignant melanoma
displays a serious health risk with increasing incidence and high mortality rate. Fortunately,
advances in the treatment of malignant melanoma now prolong survival and enhance response
and treatment efficacy. Established biomarkers help evaluate disease progression and
facilitate choosing appropriate and individual treatment options. However, the need for easily
accessible and reliable biomarkers is rising to predict patient-specific clinical outcome.
Eosinophil infiltration into the tumor and high peripheral eosinophil counts prior and during
treatment have been associated with better response in patients for various cancer entities,
including melanoma. An analysis of a heterogeneous study cohort reported high serum ECP
levels in non-responders. Hence, eosinophil frequency and serum ECP as a soluble
eosinophil-secreted mediator were suggested as prognostic biomarkers in melanoma. We
examined whether melanoma patients treated with first-line targeted therapy could also benefit
from the effects of eosinophils. In total, 243 blood and serum samples from patients with
advanced melanoma were prospectively and retrospectively collected before and after drug
initiation. To link eosinophil function to improved clinical outcome, soluble serum markers and
peripheral blood counts were used for correlative studies using a homogeneous study cohort.
In addition, functional and phenotypical characterizations provided insights into the expression
profile and activity of freshly isolated eosinophils, including comparisons between patients and
healthy donors.
Our data showed a significant correlation between high pre-treatment blood eosinophil counts
and improved response to targeted therapy and by trend to combinatorial immunotherapy in
patients with metastatic melanoma. In accordance with previous studies our results links
eosinophil blood counts to better response in melanoma patients. High pre-treatment ECP
serum concentration correlated with response to immunotherapy but not to targeted therapy.
Eosinophils from healthy donors and patients showed functional and phenotypical similarities.
Functional assays revealed a strong cytotoxic potential of blood eosinophils towards
melanoma cells in vitro, inducing apoptosis and necrosis. In addition, in vitro cytotoxicity was
an active process of peripheral eosinophils and melanoma cells with bidirectional features and
required close cell-cell interaction. The extent of cytotoxicity was dose-dependent and showed
susceptibility to changes in physical factors like adherence. Importantly, we provide evidence
of an additive tumoricidal function of eosinophils and combinatorial targeted therapy in vitro. In
summary, we give valuable insights into the complex and treatment-dependent role of
eosinophils in melanoma. As a result, our data support the suggestion of eosinophils and their
secreted mediators as potential prognostic biomarkers. It will take additional studies to
examine the molecular mechanisms that underlie our findings.
Das kolorektale Karzinom zählt zu den häufigsten Tumoren in den westlichen Nationen. Für die Heilung ist neben der frühen Diagnose und der korrekten Klassifikation in UICC-Stadien, die Auswahl einer effektiven individuellen Therapiestrategie von Bedeutung. Der Schlüssel für die erfolgreiche Verbesserung dieser und weiterer Schritte in der Behandlung eines kolorektalen Karzinom ist die Analyse der Interaktion zwischen kolorektalen Karzinomzellen und Immunzellen in einem 3D-Zellkulturmodell, das es ermöglicht, die in vivo Situation möglichst realitätsnah zu imitieren.
Im Vergleich zwischen einem 2D- und 3D-Zellkulturmodell konnten in dieser Arbeit bedeutende und reproduzierbare Unterschiede sowohl im Wachstumsverhalten und in der Zellmorphologie von Kolonkarzinomzellen (SW480-Zellen), Fibroblasten und Immunzellen (PBMC) als auch in deren Interaktion untereinander aufgezeigt werden.
PBMC induzieren im 2D-System sowohl bei der MK der SW480-Zellen als auch bei der KK mit Fibroblasten im Vergleich zum 3D-System deutlich mehr Apoptose. So gibt es bei der MK mit 1, 2 und 3 Millionen PBMC eine durchschnittliche Apoptosezunahme von 88 % nach 24 h und von 95 % nach 48 h. Bei der KK mit ebenfalls 1, 2 und 3 Millionen PBMC gibt es mit durchschnittlich 89 % nach 24 h und 92 % nach 48 h vergleichbare Apoptosezunahmen.
Zellkulturen auf azellularisierter Darmmatrix bieten den Raum für die bereits erwähnten wichtigen 3D-Zell-ECM- und Zell-Zellinteraktionen, realitätsnahe extrazelluläre Matrizen und soluble Substanzen, die eine sehr große Bedeutung für die Ausbildung tumorspezifischer Charakteristika haben wie z. B. Karzinogenese, Gewebsdifferenzierung Proliferationsgeschwindigkeit, Metastasierungspotential, Malignität, Angiogeneseeigenschaften und Mikroumgebungsbedingungen wie sie im System Mensch vorkommen.
Sie besitzen durch ihre viel komplexere histologische Struktur im Vergleich zu den zweidimensionalen Monolayer-Systemen eine höhere Aussagekraft, bieten eine bessere Vergleichbarkeit mit Tumorsystemen in lebenden Organismen und mindern die experimentelle Fehlinterpretationsraten im Vergleich zu 2D-Modellen. So ist es auch nicht verwunderlich, dass die Ergebnisse bezüglich der Apoptoserate der SW480-Zellen in beiden Zellkulturmodellen so unterschiedlich ausfallen. Durch eine gute strategische Planung der Durchführung weiterer Experimente und der Quantifizierung von weiteren möglichen Zytokinen könnte der Pool an möglichen Wegen hin zur Apoptose der SW480-Zelle eingeengt werden.
Unsere Ergebnisse zeigen, wie wichtig es ist, auch zukünftig weiter an dieser Thematik zu forschen, um so die Therapie des kolorektalen Karzinoms entscheidend zu verbessern. So wäre es durchaus vorstellbar, innerhalb dieses 3D-Modells durch Experimente, die beispielsweise die Tumorzellumgebung oder die Immunzelleigenschaften verändern, dem tumorzellbiologischen Verhalten und der immunologischen Tumor-Wirtsbeziehung in vivo entscheidend näher zu kommen.
Georg Sticker war Arzt, Hochschullehrer, Hygieniker und Medizinhistoriker. Das Themenspektrum seiner Arbeiten ist sehr umfangreich. Der Schwerpunkt dieser Arbeit liegt auf seinen Hauptforschungsgebieten, der Medizingeschichte und hier vor allem auf der Seuchengeschichte zu Pest, Lepra und Syphilis und der Bedeutung der Geschichte der Epidemien für die Epidemiologie; sowie seinen großen ärztlichen Vorbildern Hippokrates und Paracelsus. Auch über Naturheilkunst und Gesundheit und Erziehung berichtet er mit großem Engagement.Nach seiner Berufung an die Universität Würzburg beendet er seine ärztliche Tätigkeit, gründet hier im Jahr 1921 das Institut für Geschichte der Medizin und widmet sich ganz der Medizingeschichte
