Die Bedeutung des Zwei-Partner-Sekretionssystems für die Adhärenz von Meningokokken an Epithelzellen
(2009)
Das two-partner secretion-system (TPS-System) ist ein unter Gram-negativen Bakterien weit verbreiteter Weg der Proteinsekretion. Die als TpsA bezeichneten Exoproteine des TPS Systems benötigen ein spezifisches Partnerprotein (genannt TpsB) in Form eines kanalbildenden Transporters. Im sequenzierten Genom des Meningokokkenstammes MC58 finden sich fünf putative tpsA Gene, die als hemagglutinin/hemolysin-related protein (hrps) bezeichnete werden. Neben MC58 finden sich auch in den anderen sequenzierten Meningokokkenstämmen (FAM18, Z2491, alpha14) hrps. Diese weisen N-terminal Homologien zum filamentösen Hämagglutinin (FHA) von B. pertussis auf, das als TpsA-Protein des two-partner-secretion-system (TPS) aus der Zelle transportiert wird. In dieser Arbeit werden die hrps als hrpA Gene bzw. HrpA-Proteine bezeichnet. Alle sequenzierten Meningokokkenstämme verfügen über tpsB homologe Gene (hrpB), die jeweils in enger Nachbarschaft zu den hrpA Genen zu finden sind. Das Vorhandensein von hrpA und hrpB Genen deutet darauf hin, dass auch Meningokokken über ein funktionales TPS-System verfügen. Bei einer Dot-Blot-Analyse von 830 Meningokokkenstämmen aus einer bayerischen Trägerstudie mit Sonden spezifisch für die C-terminalen Bereiche der im Stamm MC58 gefundenen hrpA Gene hybridisierten 80% der ausgewerteten Stämme mit mindestens einer der Sonden. Stämme der hypervirulenten klonalen Komplexen (ST-8, ST-11, ST32, ST-44) zeigten sogar in über 99% eine positive Reaktion. Dagegen wiesen die nicht-hypervirulenten klonalen Komplexe zu 29% im Dot Blot kein hrpA auf, das homolog zu den hrpA Genen von Stamm MC58 ist, wobei es sich hierbei mehrheitlich (82%) um cnl Stämme handelte, so dass sich nur in 10% der untersuchten Kapsel-null-locus-Stämme (cnl) ein zu den hrpA Genen von MC58 homologes Gen nachweisen ließ. Mit der Hypothese, dass auch diese Stämme ein hrpA besitzen, welches sich im C-terimalen Anteil von denen des MC58 unterscheidet wurden in dieser Arbeit Dot Blots durchgeführt, deren Sonde spezifisch für das hrpB NMC0443 war. 97,6% der mit dieser Sonde untersuchten Stämme zeigten die Anwesenheit eines hrpB Homologs. Um die Vermutung zu bestätigen, dass allen hrpB Genen ein zugehöriges hrpA Gen benachbart liegt, wurden repräsentativ PCRs von häufigen klonalen Komplexen durchgeführt. Dabei konnte gezeigt werden, dass ein TPS-System sowohl in den hypervirulenten als auch den nicht-hypervirulenten klonalen Komplexen der Meningokokken vorkommt. Die vielfältigen Funktionen von bereits untersuchten TpsA Proteinen sind zumeist mit der Pathogenität der Bakterien assoziiert. In dieser Arbeit wurde ein möglicher Einfluss der HrpA Proteine auf die Adhäsion der Bakterien an humane Zellen untersucht. Es konnte gezeigt werden, dass sowohl eine kapsellose, als auch eine kapsellose, LPS-trunkierte hrpA Deletionsmutante signifikant schlechter an Epithelzellen adhäriert als die parentalen Vergleichsstämme. Ebenso zeigten die analog durchgeführten Infektionsversuche mit der hrpB Deletionsmutante einen Adhärenzverlust, der jedoch nur für die unbekapselte und LPS trunkierte hrpB Deletionsmutante signifikant war. In dieser Arbeit ist es gelungen das HrpB Protein des Stammes 2120 in E. coli zu exprimieren und aufzureinigen, sodass die Entwicklung eines gegen HrpB gerichteten Antikörpers in Auftrag gegeben werden konnte. Mit Hilfe dieses Antikörpers sollen noch offene Fragen zur Synthese und dem Transport des HrpB Transportproteins beantwortet werden. Außerdem können weitere Untersuchungen zur Lage und Verteilung der HrpBs in der Meningokokkenmembran dazu beitragen, weiteren Aufschluss über die Komplexität von Pathogenität und Virulenz von N. meningitidis zu geben.
Auf dem Weg vom Primärtumor zur systemischen Metastasierung, der Haupttodesursache von Krebserkrankungen, ist die Einzelzellmigration von Tumorzellen durch dreidimensionales Bindegewebe ein entscheidender Schritt. Die vorliegende Arbeit zeigt Untersuchungen zur Tumorzellmigration und –plastizität in einem 3D-Migrationsmodell. Kleine G-Proteine kontrollieren Zytoskelettfunktionen, insbesondere Aktinpolymerisation und die Bildung von Zellprotrusionen durch Rac sowie Actomyosinkontraktion durch Rho. Durch pharmakologische Inhibitoren von Rac und dem Rho-Effektor ROCK soll deren Bedeutung für Einzelzellmigration in einem dreidimensionalen Modell und vor allem der Effekt auf Morphologie, Plastizität und Migration von Tumorzellen geklärt werden. Nach Inhibition von ROCK zeigen hochinvasive HT1080 Fibrosarkomzellen einen multipolar-dendritischen und sessilen Phänotyp. Nach Hemmung von Rac wird hingegen ein rundlicher, aber ebenfalls apolarer und sessiler Phänotyp induziert. Bei simultaner Inhibition von Rac und ROCK entstehen rundliche, apolare, sessile Zellen mit abortiven Pseudopodien. Wird das Gleichgewicht von Rac und ROCK durch konstitutive Aktivierung von ROCK gestört, so entsteht eine zweigeteilte Population, bestehend aus rundlichen Zellen, die Blebs bilden, und langgezogenen Zellen. Nach Sortierung nach ihrem ß1-Integrinexpressionsniveau zeigten Zellen mit niedriger Integrin-Expression einen rundlichen Migrationstyp mit blasenartigen dynamischen Protrusionen, während Zellen mit hoher Integrin-Expression langgezogen-mesenchymal migrierten. Somit steuern ROCK und Rac gemeinsam und zeitgleich die mesenchymale Einzelzellmigration. Während Rac Protrusion vermittelt, ist ROCK für Kontraktilität und Retraktion verantwortlich. Erst durch Koordination von Rac und Rho/ROCK entsteht somit Polarität und 3D mesenchymale Migration.
Ausgehend von einer potentiell anti-Tumor-aktiven B-Zellen des menschlichen Immunsystem haben sich durch die Untersuchung des peripheren Blutes auf das Vorliegen von lambda3r-positiven, CD19+B-Zellen bei Magenkarzinompatienten und Probanden unterschiedlichen Alters einige sehr interessante Ergebnisse im Bereich der B-Zellimmunität ergeben. Es scheint dabei eine Art B-Zell-Immunosurveillance in Form dieser B-Zellen, sowohl bei Karzinompatienten, als auch bei Gesunden von früher Kindheit an zu geben. Die relative Verteilung dieser Zellen ändert sich dabei im Laufe des Lebens ensprechend den Veränderungen des gesamten B-Zellkompartiments. Es findet eine Abnahme mit dem Alter statt. Im Falle des Auftretens eines Magenkarzinoms kommt es dann zu einer relativen Expansion der in dieser Arbeit beschriebenen lambda3r-positiven CD19+B-Zellen trotz einer gleichzeitig stattfindenden bisher nicht erklärlichen Involution des restlichen B-Zellsystems. Bei der relativen Zunahme dieser Zellen handelt es sich um eine Art Boosterung. Das expandierte Zellkompartiment zeigt dabei Reifungstendenzen, sichtbar im Verlust des Oberflächenmoleküls IgD sowie der Expression von CD27 und IgG. Dem Oberflächenmarker CD5 scheint im Gegensatz zur initialen Hypothese bei der Erst-beschreibung der SC-1-positiven B-Zelle keine zentrale Rolle zuzukommen.
Das Ausmaß der Proteinbindung eines Arzneistoffs wirkt sich auf viele unterschiedliche pharmakokinetische Parameter aus. So wird beispielsweise das Verteilungsvolumen, die Metabolisierung oder die Elimination des entsprechenden Stoffes durch die Höhe seiner Proteinbindung beeinflusst. Da nur der im Plasma frei vorliegende Anteil eines Arzneistoffs in der Lage ist biologische Membranen zu überwinden, können auch nur die freien Arzneistoffmoleküle eine pharmakologische Wirkung an Rezeptoren oder Enzymen auslösen. Dementsprechend ist auch die Intensität der hervorgerufenen Wirkung von der Größe des ungebundenen Anteils eines Arzneistoffs abhängig. Aufgrund dieser Zusammenhänge ist klar, dass die Proteinbindung eines Arzneistoffes letztendlich Einfluss auf die Dosisfindung hat. Zur Ermittlung der Proteinbindung stehen viele unterschiedliche Methoden, wie beispielsweise die HPLC, Kapillarelektrophorese, Ultrazentrifugation, Gleichgewichtsdialyse und Ultrafiltration zur Verfügung. In der vorliegenden Arbeit wurde die kontinuierliche Ultrafiltration zur Ermittlung der Proteinbindung von Arzneistoffen angewendet. Hier wird die Proteinbindung nicht nur anhand einer bestimmten Arzneistoff- bzw. Albuminkonzentration gemessen, sondern über einen weiteren Bereich von Wirkstoff-Protein-Verhältnissen beobachtet. Des Weiteren ist der apparative Aufwand im Vergleich zu vielen anderen Methoden als geringer einzustufen. Im Rahmen dieser Arbeit wurde, die auf der von Heinze[122] entwickelte Messanlage weiter optimiert und eine zweite Anlage mit einem Diodenarraydetektor aufgebaut. Für letztere musste eine Software-Anpassung vorgenommen werden. Folgende Projekte wurden durchgeführt: 1) Um den In-vivo-Bedingungen nahe zu kommen, wurde bei der Bestimmung der Proteinbindung der Sartane nicht nur BSA und HSA verwendet, sondern erstmals auch humanes Plasma. Die Plasmamessungen der Sartane verliefen insgesamt problemlos, allerdings ist eine erfolgreiche Messung stark von der Qualität des eingesetzten Plasmas abhängig, wie Messungen der Naphthylisochinoline gezeigt haben. Im Vergleich mit HSA und Plasma ergaben die Messungen der Sartane mit bovinem Serumalbumin geringfügig erniedrigte Proteinbindungswerte. Insgesamt sind alle Ergebnisse sehr gut mit den Literaturwerten vergleichbar. 2) Das Ausmaß der Proteinbindung von Naphthylisochinolinen war bislang unbekannt und lag im Bereich von ca. 30-70%. Erneut waren die Resultate aus den Messungen von BSA und HSA nahezu gleich. 3) Am Beispiel der Interaktion zwischen Phenprocoumon und Phenylbutazon wurden zwei unterschiedliche Ansätze getestet, um die Verdrängung aus der Proteinbindung zu simulieren. Die erste Methode entsprach hierbei einer Konkurrenz der beiden interagierenden Stoffe um die Proteinbindungsstellen. Durch den Einfluss des Phenylbutazon verringerte sich die Proteinbindung des Phenprocoumon um 1%, was allerdings als statistisch nicht signifikant betrachtet werden kann. Im zweiten Ansatz, der eine direktere Verdrängung aus der Proteinbindung simulieren sollte, fiel die Proteinbindung des Phenprocoumon gegenüber den Einzelmessungen um 2,5% ab. Unter physiologischen Konzentrationsverhältnissen sank sich die Proteinbindung des Phenprocoumon auf 93,3%. Der freie Anteil erhöhte sich dementsprechend von 1% auf 6,7%. Somit konnte der Einfluss des Phenylbutazon auf die Proteinbindung des Phenprocoumon erfolgreich nachgewiesen werden. Die unveränderte Proteinbindung des Phenylbutazon im inversen Ansatz und die ermittelten pK-Werte bestätigen diese Interaktion. Grundsätzlich ist es also möglich mit der kontinuierlichen Ultrafiltration solche Interaktionen zu simulieren. 4) Zuletzt sollte der Frage nachgegangen werden, ob es mit der kontinuierlichen Ultrafiltration auch möglich ist die Proteinbindung von wasserunlöslichen Stoffen, nämlich den Aziridinen, in Gegenwart steigender Mengen DMSO, zu bestimmen. Die erhaltenen Ergebnisse wurden mit Literaturwerten ohne DMSO-Zusatz verglichen. Abgesehen von Candesartan, das eine lineare Korrelation zwischen DMSO-Gehalt der Wirkstofflösung und Absinken der Proteinbindung zeigte, konnte kein Zusammenhang zwischen der DMSO-Konzentration und der gemessenen Proteinbindung festgestellt werden. Die Mittelwerte lagen im Bereich der Literaturwerte. Insgesamt zeigten alle Versuchsreihen, dass die kontinuierliche Ultrafiltration eine ausgezeichnete, schnelle und robuste Screeningmethode zur Bestimmung des Ausmaßes der Proteinbindung bekannter und neuer Wirkstoffe darstellt.
The bacterial pathogen Legionella pneumophila replicates intracellularly in protozoa, but can also cause severe pneumonia, called Legionnaires' disease. The bacteria invade and proliferate in the alveolar macrophages of the human lung. L. pneumophila bacteria exhibit a biphasic life cycle: replicative bacteria are avirulent; in contrast, transmissive bacteria express virulence traits and flagella. Primarily aim of this thesis was to evaluate the impact of the regulatory proteins FleQ, FleR, and RpoN in flagellar gene regulation. Phenotypic analysis, Western blot and electron microscopy of regulatory mutants in the genes coding for FleQ, RpoN and FleR demonstrated that flagellin expression is strongly repressed and that these mutants are non-flagellated in transmissive phase. Transcriptomic studies of these putative flagellar gene expression regulators demonstrated that fleQ controls the expression of numerous flagellar biosynthetic genes. Together with RpoN, FleQ controls transcription of 14 out of 31 flagellar class II genes, coding for the basal body, hook, and regulatory proteins. Unexpectedly, 7 out of 15 late flagellar genes class III and IV) are expressed dependent on FleQ but independent of RpoN. Thus, in contrast to the commonly accepted view that enhancer binding proteins as FleQ always interact with RpoN to initiate transcription, our results strongly indicate that FleQ of L. pneumophila regulates gene expression RpoN-dependent as well as RpoN-independent. Moreover, transcriptome analysis of a fleR mutant strain elucidated that FleR does not regulate the flagellar class III genes as previously suggested. Instead FleR regulates together with RpoN numerous protein biosynthesis and metabolic genes. Based on these experimental results our modified model for the transcriptional regulation of flagellar genes in L. pneumophila is that flagellar class II genes are controlled by FleQ and RpoN, while flagellar class III and IV genes are controlled in a fleQ-dependent but rpoN-independent manner. Although all L. pneumophila strains share the same complex life style, various pathotypes have evolved. This is reflected by the genomes, which contain e.g. genomic islands. The genomic island Trb-1 of L. pneumophila Corby, carries all genes necessary for a type-IV conjugation system, an integrase gene and a putative oriT site. The second aim of this thesis was to investigate the implication of this genomic island in conjugative DNA transfer. Using conjugation assays we showed that the oriT site located on Trb-1 is functional and contributes to conjugation between different L. pneumophila strains. As this is the first oriT site of L. pneumophila known to be functional our results provide evidence that conjugation is a major mechanism for the evolution of new pathotypes in L. pneumophila.
Circa 1% der Weltbevölkerung ist an schizophrenen Psychosen erkrankt. Durch Störung kognitiver und exekutiver Funktionen bedürfen diese Patienten regelmäßiger Untersuchung und Betreuung, was nicht nur für den einzelnen Betroffenen, sondern auch sozioökonomisch bedeutsam ist. Die Einteilung der endogenen Psychosen nach Karl Leonhard stellt eine hoch differenzierte, nosologisch orientierte Krankheitsklassifikation dar, die sich durch eine exakte Darstellung der diagnostischen Kriterien und durch eine Vielzahl von präzise voneinander abgegrenzten Krankheitsbildern mit spezifischer Verlaufscharakteristik auszeichnet. Der in dieser Arbeit vertretene Ansatz geht davon aus, dass es sich bei den schizophrenen Psychosen nicht um eine einzelne Erkrankung, sondern um verschiedene Krankheitsentitäten handelt, die wiederum unterschiedlichen pathogenentischen Prinzipien unterliegen. Ziel war es darzustellen, dass sich die zykloiden Psychosen mit immer wiederkehrenden Manifestationen im Vergleich zu den unsystematischen Schizophrenien mit überwiegend hereditärer Genese und im Vergleich zu den monomorph und monophasisch ablaufenden systematischen Schizophrenien hinsichtlich der Immunparameter deutlich unterschieden. Methode: Um eine mögliche Immunpathogenese bestimmter Formen endogener Psychosen belegen zu können, wurden in einer retrospektiven Untersuchung 61 Patienten aus dem schizophrenen Formenkreis nach Karl Leonhard (32 zykloide Psychosen, 21 unsystematische und 12 systematische Schizophrenien) gegenübergestellt und hinsichtlich ausgewählter Immunparameter aus Serum und Liquor, klinischer Verlaufsparameter und soziodemographischer Variablen untersucht. Ergebnisse: Die Analyse immunologischer Parameter aus Serum und Liquor erbrachte keine signifikanten Unterschiede zwischen den einzelnen Erkrankungsgruppen. Ebenso wurden bei der Verteilung auf beide Geschlechter, bei der Anzahl von Allergikern und bei der Anzahl der Patienten mit Gefäßrisikofaktoren keine signifikanten Unterschiede zwischen den Erkrankungsgruppen nach der Leonhard-Klassifikation ermittelt. Auch die Untersuchung peripherer Parameter und Serum- Liquorparametern bei Patienten mit Erstdiagnose ergab keine signifikanten Unterschiede. Im Rahmen einzelner klinischer Verlaufsparameter unterschieden sich jedoch die zykloiden Psychosen signifikant von den schizophrenen Psychosen. Konklusion: In einer Folgestudie könnte die Analyse speziellerer Immunparameter, wie beispielsweise Zytokine, wichtige Hinweise erbringen, um die zykloiden Psychosen auch auf paraklinischem Wege von chronisch schizophrenen Psychosen zu differenzieren und um neue, auf mögliche immunologische Prozesse abgestimmte Behandlungsalternativen prüfen zu können.
The taphonomic and paleoecologic aspects of the Upper Hauterivian to Lower Barremian Agua de la Mula Member of the Agrio Formation (Neuquén Basin, Argentina) were studied in the frame of the sequence stratigraphic paradigm. The Agua de la Mula Member, a ca. 600 m thick succession of highly cyclic marine sediments was surveyed at two localities. Detailed bed-by-bed sedimentologic, stratigraphic, ichnologic, taphonomic and paleoecologic data collection allowed a precise paleoenvironmental, stratigraphic, taphonomic and synecologic interpretation, in a controlled sequence stratigraphic framework. The main architectural stratigraphic component is the Starvation-Dilution Sequence, interpreted as a the effect of a sixth-order, Milankovitch precession-driven cycle. Dilution hemisequences are siliciclastic-dominated and show evidence of depth changes. Starvation hemisequences show a diverse variation of mixed carbonate-siliciclastic facies that is linked to sequence stratigraphy. Ammonite-based biostratigraphy was revised and new knowledge proposed. The stratigraphic framework was improved by combining biostratigraphy, sequence stratigraphy and event stratigraphy. Nine main sequences were described, linked to other stratigraphic markers and correlated with other sequence stratigraphic charts. Several orders of cyclicity were inferred. Third- and fourth-order sequences are the major sequences, not subordinated to higher hierarchies (lower order). Precession, obliquity, and short and long eccentricity cycles of the Milankovitch band are proposed. Among the different sequence stratigraphic models the transgression-regression model fits the majority of the sequences described in this work. The depositional-sequence model could be applied only to the first third-order sequence, in which the true sequence boundary is identifiable. Starvation-dilution sequences, however, are composed by to components that are not completely explained by those models. Starvation hemisequences developed in intermediate to deep settings record the transgressive phase as well as the earLy regressive one without visible stratigraphic boundaries. 112 samples with 22,572 individuals were grouped into fifteen fossil associations and one assemblage that reflect the interaction of different factors: age, position in major, medium and starvation dilution sequences and, linked to sequence stratigraphy, depth, oxygen availability, rate of terrigenous input, water agitation, and substrate conditions. Temporary possible reduction in oxygen content is inferred based on all sources of available evidence. Organic buildups are briefly described and their development interpreted in terms of the sequence stratigraphic framework. Vertical patterns of replacement of fossil associations are described and related to sequence stratigraphy. Five types of skeletal concentrations represent the diversity of coquinas decribed in this study. Type 1, 2, 4 and 5 correspond to starvation hemisequences deposited in progressively shallower settings, from basin to inner ramp. Type 3 is embedded into dilution hemisequences and inferred to be linked to shell bed type I of Kidwell (1985). Types 1 and 2 correspond to transgression, maximum flooding and early regression without distinction. Type 4A as well as Type 5 are interpreted as onlap shell beds (Kidwell 1991a) or early TST shell beds (Fürsich and Pandey 2003). Type 4B corresponds to the MFZ shell bed (Fürsich and Pandey 2003) or mid-cycle shell bed (Abbott 1997), while Type 4C to the downlap shell bed (Kidwell 1991a). Time-averaging of shell beds was assessed with precision as the time involved in the deposition of the starvation hemisequences could be inferred. All shell beds comprise within-habitat assemblages forming within a few thousand years, with little environmental condensation. The fossilization of the marine calcareous shells is modelled as a series of steps called windows: environmental, destructional, burial and diagenetic. The “diagenetic window” is the most relevant. Connected to this it is proposed that carbonate dissolution is the primary control on the development of shell beds, as has been proposed before (Fürsich 1982; Fürsich and Pandey 2003). The interpretative power resulting from combining several lines of evidence, e.g., facies analysis, sequence stratigraphy, biostratigraphy, trace fossil analysis, paleoecology and taphonomy, and unravelling their multiple relationships, are the most relevant conclusions of this study.
Auch nach jahrelanger Forschung ist eine Transplantation von Langerhans-Inseln noch keine Standard-Therapie des Insulin-abhängigen Diabetes mellitus. Ein Problem ist die grosse Anzahl benötigter Pankreata, mögliche Lösung die Verwendung porziner Langerhans-Inseln. Damit die späteren Spender-Langerhans-Inseln ein gutes In-Vivo-Ergebnis erzielen können, muss bereits eine gute Funktion in vitro gewährleistet sein. In dieser Arbeit wurde untersucht, ob sich eine verbesserte Funktion der Langerhans-Inseln durch eine Exposition von extrazellulären Matrixproteinen (ECMP) erreichen lässt. Weiterhin wurde ein Einfluss von Polyelektrolytschichten auf die Funktion der Langerhans-Inseln und ihre mikroskopische Darstellbarkeit untersucht. Mit der sogenannten Nanoverkapselung soll bei der Transplantation eine Immunisolation der Spender-Inseln als Schutz vor dem Immunsystem des Empfängers erreicht werden. Es wurden zwei verschiedene Kapseln untersucht: Chitosan MMW/PEGPPGPEG/Chitosan MMW (CPC) und Chitosan MMW/PAAMA/PAH 15 kDa (CPP). Wir konnten zeigen, dass die Funktion der Langerhans-Inseln generell am Tag +6 besser ist als am Tag +1, weil erst dann eine Erholung vom Stress der Isolierung stattgefunden hat. Eine Verbesserung der Inselzell-Funktion durch die ECMP konnte bestätigt werden, allerdings hängt sie von den jeweils verwendeten Komponenten der extrazellulären Matrix ab. Durchweg gute Perifusionsergebnisse am Tag +6 werden nach Exposition mit Kollagen IV, Laminin und Fibronektin erreicht. Bei den Versuchen mit den Polyelektrolyt-Nanokapseln ergaben sich deutliche Unterschiede zwischen den beiden untersuchten Nanokapseln. Bei allen verwendeten Polyelektrolyten war auf gute Biokompatibilität geachtet worden. Dennoch scheinen die Schichten CPC einen negativen Einfluss auf die Langerhans-Inseln zu haben: Hier kam es zu einer verschlechterten Funktion der Langerhans-Inseln am Tag +6. Die Langerhans-Inseln mit der Nanokapsel CPP dagegen zeigten gute Ergebnisse. Interessanterweise sahen wir aber bei CPP-Nanokapseln eine verstärkte Autofluoreszenz, ein Zeichen für zellulären Stress. Durch die Verwendung von FITC-gekoppelten Polyelektrolyten lässt sich mikroskopisch eine verstärkte Fluoreszenz der Langerhans-Inseln nachweisen. Der Nachweis der Vollständigkeit der Polyelektrolytschichten muss jedoch in weiteren Studien erbracht werden. Die hier gewonnenen Erkenntnisse über die extrazellulären Matrixproteine sind zunächst für zukünftige Versuche wichtig: Die Funktion der Langerhans-Inseln kann so verbessert werden, dies ist eine Voraussetzung für erfolgreiche Versuche. Ausserdem können die ECMP für die spätere Vorbereitung von Langerhans-Inseln vor der Transplantation verwendet werden. Entscheidend ist, dass die Langerhans-Inseln für mehrere Tage mit den ECMP inkubiert werden, um so ihre Funktion zu verbessern. Bei der Polyelektrolyt-Nanoverkapselung besteht noch viel Forschungsbedarf: Anhand des Erfolg versprechenden Ergebnisses mit der CPP-Nanokapsel muss weitergearbeitet werden. Offensichtlich führt – bei Auswahl der richtigen Polyelektrolyte – die Nanoverkapselung nicht zu einer verschlechterten Funktion der Langerhans-Inseln. Bei einer grossen Anzahl in Frage kommender Polyelektrolyte muss die optimale Kombination von Schichten gefunden und ihre Fähigkeit zur Immunisolation untersucht werden. Bezüglich der Darstellung der Nanokapseln bietet sich die konfokale Fluoreszenzmikroskopie an.
In mammals, the RAF family of serine/threonine kinases consists of three members, A-, B- and C-RAF. Activation of RAF kinases involves a complex series of phosphorylations. Although the most prominent phosphorylation sites of B- and C-RAF are well characterized, little is known about regulatory phosphorylation of A-RAF. Using mass spectrometry, we identified here a number of novel in vivo phosphorylation sites in A-RAF. The physiological role and the function of these sites were investigated subsequently by amino acid exchange at the relevant positions. In particular, we found that S432 participates in MEK binding and is indispensable for A-RAF signaling. On the other hand, phosphorylation within the activation segment does not contribute to epidermal growth factor-mediated activation. Regarding regulation of A-RAF activity by 14-3-3 proteins, we show that A-RAF activity is regulated differentially by its C-terminal and internal 14-3-3 binding domain. Furthermore, by use of SPR technique, we found that 14-3-3 proteins associate with RAF in an isoform-specific manner. Of importance, we identified a novel regulatory domain in A-RAF (referred to as IH-segment) positioned between amino acids 248 and 267, which contains seven putative phosphorylation sites. Three of these sites, serines 257, 262 and 264, regulate A-RAF activation in a stimulatory manner. The spatial model of the A-RAF fragment including residues between S246 and E277 revealed a “switch of charge” at the molecular surface of the IH-region upon phosphorylation, suggesting a mechanism in which the high accumulation of negative charges may lead to an electrostatic destabilization of protein/membrane interaction resulting in depletion of A-RAF from the plasma membrane. Activation of B- and C-RAF is regulated by phosphorylation at conserved residues within the negative-charge regulatory region (N-region). Identification of phosphopeptides covering the sequence of the N-region led to the conclusion that, similar to B- and C-RAF, kinase activity of A-RAF is regulated by phosphorylation of the N-region. Abrogation of A-RAF activity by S299A substitution and elevated activity of the A-RAF-Y301D-Y302D mutant confirmed this conclusion. In addition, we studied the role of the non-conserved residues within the N-region in the activation process of RAF kinases. The non-conserved amino acids in positions –3 and +1 relative to the highly conserved S299 in A-RAF and S338 in C-RAF have so far not been considered as regulatory residues. Here, we demonstrate that Y296R substitution in A-RAF led to a constitutively active kinase. In contrast, G300S substitution (mimicking B- and C-RAF) acts in an inhibitory manner. These data were confirmed by analogous mutations in C-RAF. Based on the three-dimensional structure of the catalytic domain of B-RAF, a tight interaction between the N-region residue S339 and the catalytic domain residue R398 was identified in C-RAF and proposed to inhibit the kinase activity of RAF proteins. Furthermore, Y296 in A-RAF favors a spatial orientation of the N-region segment, which enables a tighter contact to the catalytic domain, whereas a glutamine residue at this position in C-RAF abrogates this interaction. Considering this observation, we suggest that Y296, which is unique for A-RAF, is a major determinant of the low activating potency of this RAF isoform. Finally, the residues R359 in A-RAF and R398 in C-RAF, which interact with the N-region, are also involved in binding of phosphatidic acid. Substitution of this conserved arginine by alanine resulted in accumulation of hyper-phosphorylated form of RAF, suggesting that this residue play a crucial role in phosphorylation-mediated feedback regulation of A- and C-RAF. Collectively, we provide here for the first time a detailed analysis of in vivo A-RAF phosphorylation status and demonstrate that regulation of A-RAF by phosphorylation exhibits unique features compared with B- and C-RAF.
