Synthesis of (RS)-5-amino-3-aryl (methyl)-pentanoic acid hydrochlorides, 3 aminomethyl-5-chloro-benzoic acid hydrochloride and (RS)-4-amino-3-(4`-ethynyl(iodo)-phenyl)-butanoic acid hydrochlorides have been accomplished. The aim of their synthesis was to evaluate their GABABR agonist activity and to derive a model which will correlate their structure with the observed pEC50. The GABABR agonist activity of the prepared compounds has been determined in functional assay based on calcium measurement in vitro using tsA cells transfected with GABAB1b/GABAB2/Gαq-z5. Reviews on the neurotransmitter receptors (ligand-gated ion channel receptors and G protein-coupled receptors), their agonists and antagonists have been given in the general part of this work. A detailed discussion on the strategy followed for the synthesis of the designed compounds as well as the starting materials and intermediates has been described and illustrated in Schemes 2-6. The synthesized compounds were evaluated for their GABABR agonist activity. Furthermore, these compounds were docked in the available 3D homology model of GABABR using the program FlexiDock implemented in SYBYL software. Subsequently, we derived a predictive model which correlates the experimentally determined pEC50 with the calculated binding energy of certain baclofen analogues and homologues. In addition, we used the program DISCO (DIStance COmparisons) implemented in SYBYL software to find the pharmacophore features of GABAB agonists.
Baclofen (1) is a potent and selective agonist for bicuculline-insensitive GABAB receptors and is used clinically as an antispastic and muscle relaxant agent. In the search for new bioactive chemical entities that bind specifically to GABAB receptors, we report here the synthesis of certain baclofen homologues, namely (R,S)-5-amino-3-arylpentanoic acid hydrochlorides (R,S)-1a–h as well as (R,S)-5-amino-3-methylpentanoic acid [(RS)-1i] to be evaluated as GABABR agonists. Compound 1a is an agonist to GABAB receptors with an EC50 value of 46 μM on tsA201 cells transfected with GABAB1b/GABAB2/Gqz5, being the most active congener among all the synthesized compounds.
Cholinesterases are important biological targets responsible for regulation of cholinergic transmission, and their inhibitors are used for the treatment of Alzheimer’s disease. To design new cholinesterase inhibitors, of different structure-based design strategies was followed, including the modification of compounds from a previously developed library and a fragment-based design approach. This led to the selection of heterodimeric structures as potential inhibitors. Synthesis and biological evaluation of selected candidates confirmed that the designed compounds were acetylcholinesterase inhibitors with \(IC_{50}\) values in the mid-nanomolar to low micromolar range, and some of them were also butyrylcholinesterase inhibitors.
Starting in the late 1990s ionic liquids (ILs) gained momentum both in academia as well as industry. ILs are defined as organic salts with a melting point below 100 °C. Active pharmaceutical ingredients (APIs) may be transferred into ILs by creating salts with a bulky counterion with a soft electron density. ILs have demonstrated the potential to tune important pharmaceutical features such as the solubility and the dissolution rate, particularly addressing the challenge of poor water soluble drugs (PWSD). Due to the tunability of ILs, modification of physico-chemical properties of APIs may be envisioned without any modifications of the chemical structure.
In the first chapter the potential as well as the limitation of ILs are discussed. The chapter commences with an overview of preparation and characterization of API-ILs. Moreover, examples for pharmaceutical parameters are presented which may be affected by IL formation, including the dissolution rate, kinetic solubility or hygroscopicity as well as biopharmaceutical performance and toxicology. The impact of IL formation on those pharmaceutically relevant features is highlighted, resulting in a blueprint for a novel formulation concept to overcome PWSD challenges without the need for structural changes of the API.
Within the second chapter the IL concept is detailed for one specific API - counterion combination. A poorly water soluble acidic API against migraine attacks was transformed into an IL in an effort to minimize the time to maximum plasma concentration (tmax) and optimize the overall bioavailability. These studies were conducted in parallel to a prodrug of the API for comparison of the IL strategy versus a strategy involving modification of the API’s structure. A significantly longer duration of API supersaturation and a 700 fold faster dissolution rate of the IL in comparison to the free acid were obtained and the underlying mechanism was elucidated. The transepithelial absorption was determined using Caco-2 cell layers. For the IL about 3 times more substance was transported in comparison to the prodrug when substances were applied as suspensions, despite the higher permeability of the prodrug, as increased solubility of the IL exceeded this effect. Cytotoxicity of the counterion was assessed in hepatic, renal and macrophage cell lines, respectively, and IC50 values were in the upper µM / lower mM range. The outcome of the study suggested the IL approach instrumental for tuning biopharmaceutical properties, without structural changes of the API as required for preparation of prodrugs. Thus the toolbox for formulation strategies of poorly water soluble drugs could be extended by an efficient concept.
The third chapter focuses on the effect of different counterions on the physico-chemical properties of an API-IL, in particular to overcome the challenge of poor water solubility. Therefore, the same poorly water soluble acidic API against migraine attacks mentioned above was combined with 36 counterions resulting in ILs and low lattice enthalpy salts (LLES). Depending on the counterions, different dissolution rates, durations of supersaturation and hygroscopicities were obtained and release profiles could be tailored from immediate to sustained release. Besides, in vitro the cytotoxicity of the counterions was assessed in three cell lines. Using molecular descriptors such as the number of hydrophobic atoms, the graph theoretical diameter and the number of positive charges of the counterion, the dissolution rate, supersaturation and hygroscopicity as well as the cytotoxicity of counterions could be adequately modeled, rendering it possible to predict properties of new LLESs.
Within the forth chapter different poorly water soluble APIs were combined with the counterion tetrabutylphosphonium (TBP) studying the impact on the pharmaceutical and physical properties of the APIs. TBP-ILs and low lattice enthalpy salts were prepared of the acidic APIs Diclofenac, Ibuprofen, Ketoprofen, Naproxen, Sulfadiazine, Sulfamethoxazole and Tolbutamide. NMR and IR spectroscopy, DSC, XRPD, DVS and dissolution rate measurements, release profiles and saturation concentration measurements were used to characterize the free acids and TBP salts as compared to the corresponding sodium salts. The TBP salts as compared to the free acids displayed lower melting points and glass transition temperatures and up to 1000 times higher dissolution rates. The increase in the dissolution rate directly correlated with the salts’ hygroscopicity, an aspect which is critically discussed in terms of pharmaceutical translation challenges. In summary TBP ILs of solid salts were proved instrumental to approach the challenge of poor water solubility. The outcome profiled tailor-made counterions as a powerful formulation strategy to address poor water solubility, hence bioavailability and ultimately therapeutic potential of challenging APIs.
In summary, a plethora of ILs and LLESs were prepared by combination of different acidic APIs and counterions. The IL and LLESs concept was compared to conventional salt and prodrug strategies. By choice of the counterion, biopharmaceutical relevant parameters were deliberately modified and release profiles were tuned ranging from immediate to prolonged release. The impact of distinct structural counterion features controlling the dissolution, supersaturation, hygroscopicity and counterion cytotoxicity were identified, correlations were presented and predictive models were built. ILs and LLESs could be proven to be a powerful concept for the formulation of poorly water soluble acidic APIs.
LC-ESI und MALDI-Massenspektrometrische Analyse nativer und derivatisierter Zucker und Glykane

Glykane sind weitverbreitete Biomoleküle, die meist in Form von Glykokonjugaten, wie beispielsweise als Glykoproteine oder Glykolipide, vorliegen. Durch die Interaktion von Glykanen mit Glykan-bindenden Proteinen wird eine Vielzahl an biochemischen Prozessen ausgelöst, sowohl physiologischer, als auch pathologischer Art. Die Aufklärung der beteiligten Glykanstrukturen ist daher nicht nur wichtig für das Verständnis dieser Prozesse, sondern kann auch Hinweise auf verschiedene Erkrankungen geben.
Die Identifizierung von Glykanstrukturen kann über verschiedene Wege erfolgen. In der instrumentellen Analytik spielt dabei vor allem die ESI- und MALDI Massenspektrometrie eine wichtige Rolle, da diese sowohl für Detektion, als auch Fragmentierung großer Biomoleküle geeignet sind. Um die Analyse von Zuckern mittels chromatographischer und massenspektrometrischer Methoden zu erleichtern, werden häufig Derivatisierungsreagenzien eingesetzt. Diese verringern die Polarität der Zucker und erleichtern die Detektion durch das Einbringen von Chromo- oder Fluorophoren. Zur Derivatisierung am reduzierenden Terminus von Glykanen und Zuckern eignen sich vor allem Aminierungsreagenzien oder Hydrazide. Hydrazide haben gegenüber anderen Derivatisierungsreagenzien den Vorteil einer einfachen, salzfreien Umsetzung, aus der ein stabiles Derivat mit geschlossenem terminalen Zuckerring hervorgeht.
Für die vorliegende Arbeit wurde die Derivatisierung mit den neuen Hydrazid Reagenzien INH und BINH, sowie dem bereits von Dr. P. Kapková bearbeiteten BACH untersucht. Als Vergleich dienten die underivatisierten Kohlenhydrate, wie auch das standardmäßig eingesetzte Aminierungsreagenz 2-AB. Dabei sollte das Ver-halten verschiedener Zucker und Glykane in Bezug auf chromatographische Trennung, Signalintensität und Fragmentierung analysiert werden.
Zunächst wurde die Umsetzung von Mono-, Di- und Trisacchariden mit den neuen Derivatisierungsreagenzien INH und BINH optimiert. Dadurch konnte bei beiden Substanzen die komplette Umsetzung der Zucker in ihre Derivate gewährleistet werden. Auch die Derivatisierung mit Hilfe der Mikrowelle konnte bei INH erfolgreich durchgeführt werden. Auf diese Weise ließ sich die Reaktionszeit, im Vergleich zu den im Thermo-mixer® benötigten 90 Minuten, auf 20 Minuten verkürzen. Aufgrund der großen Men-gen an Zucker und Derivatisierungsreagenz, die für die Umsetzung in der Mikrowelle nötig sind, war der Versuch jedoch nur für INH geeignet.
Im nächsten Schritt wurde das Trennverhalten der verschiedenen Mono-, Di- und Tri-saccharid-Derivate auf RP-C18- und HILIC-Phasen untersucht. Bei den Monosaccha-riden konnte durch keines der Derivate eine vollständige Trennung auf einer der Pha-sen erreicht werden. Das beste Ergebnis wurde durch INH auf der HILIC-Säule erzielt, doch auch dort konnten die Epimere Glucose, Mannose und Galactose nicht vollstän-dig separiert werden. Die Trennung der Disaccharide Maltose, Cellobiose und Lactose konnte auf der HILIC-Phase mit allen Derivaten außer BACH erfolgreich durchgeführt werden, auf der RP-C18 erwies sich dagegen nur 2-AB als geeignet. Bei den Trisac-chariden 3'SLN und 6'SLN konnten sowohl underivatisierte Zucker, als auch sämtliche Derivate mittels HILIC getrennt werden. Auch auf der C18-Phase war eine Trennung der BINH, BACH und 2-AB-Derivate möglich. Des Weiteren konnte durch die Derivati-sierungen die Signalintensität gegenüber den underivatisierten Zuckern deutlich gesteigert werden.
Nach ihrer Trennung lassen sich massegleiche Di- und Trisaccharide anhand des Fragmentierungsmusters unterscheiden. Während bei den underivatisierten Disaccha-riden Maltose, Cellobiose und Lactose die charakteristischen Fragmente nur schwach sichtbar waren, konnte mit Hilfe der Hydrazide INH, BINH und BACH die Differenzie-rung deutlich erleichtert werden. Die 2-AB-Derivatisierung zeigte dagegen keine Ver-besserung der Fragmentierungseigenschaften. Bei der Unterscheidung der Trisaccharide 3’SLN und 6’SLN waren ebenfalls sowohl underivatisierte, als auch Hydrazid-derivatisierte Zucker im Vorteil gegenüber den 2-AB-Derivaten.
Die Derivatisierung der N-Glykane von Ribonuclease B und Ovalbumin führte bei der Analyse mittels MALDI-TOF zu einer deutlichen Steigerung der Sensitivität. Beispiels-weise ließen sich bei den Glykanen des Ovalbumins durch die Derivatisierungen drei zusätzliche Strukturen im Vergleich zu den nativen Glykanen detektieren. Auch das Fragmentierungsverhalten der Glykane am MALDI-TOF/TOF konnte mit Hilfe der Derivatisierungen erheblich verbessert werden. Besonders die Umsetzung mit BINH führte zu einer Vielzahl charakteristischer Ringfragmente, wodurch die Aufklärung der verschiedenen Glykanstrukturen deutlich vereinfacht wurde. Auch im Vergleich zu 2 AB zeigten die Hydrazid-Derivate sowohl bessere Fragmentierungseigenschaften, als auch eine einfachere Handhabung für die Messung mittels MALDI-MS.
Eine weitere Möglichkeit zur Identifikation von Glykanstrukturen liegt in der spezifischen Bindung durch Lektine. Diese Untersuchung gibt des Weiteren auch einen Hinweis auf funktionelle Eigenschaften der Glykane. Dafür wird die hohe Affinität von Biotin-haltigen Derivatisierungsreagenzien zu Avidin und Streptavidin genutzt. Nach der auf diese Weise erfolgten Immobilisierung der Glykane können diese mittels spezifischer Lektine nachgewiesen werden. Die Eignung des neuen Derivatisierungsreagen-zes BINH für diese Zwecke wurde anhand eines Glykan-Arrays getestet. Dadurch ließ sich bestätigen, dass BINH-derivatisierte Glykane und Zucker sowohl in der Lage sind an Streptavidin zu binden, als auch durch Lektine nachgewiesen werden können. Daher kann davon ausgegangen werden, dass BINH grundsätzlich für den Einsatz in bio-chemischen Methoden geeignet ist.
Zusammenfassend lässt sich sagen, dass die Derivatisierung von Kohlenhydraten mit INH, BINH und BACH zu einer deutlichen Verbesserung der Trenn- und Fragmentierungseigenschaften führten. Dadurch konnten Identifizierung und Strukturanalyse sowohl von kleinen Zuckern, als auch von Glykanen erleichtert werden. Im Vergleich zu dem Standard-Derivatisierungsreagenz 2-AB zeigten die Hydrazide nicht nur im Bereich der Fragmentierungen, sondern auch durch die einfachere Derivatisierungsreaktion wesentliche Vorteile.
Ziel dieser Arbeit war es die intranasalen pharmakokinetischen Abläufe nach topischer Applikation mittels in vitro und ex vivo Experimenten zu charakterisieren und ihre Auswirkungen auf die Pharmakodynamik zu beschreiben. Es wurden hierbei die nasal angewendeten Glucocorticoide Triamcinolonacetonid (TCA), Budesonid (Bud), Beclomethasondipropionat (BDP), Fluticasonpropionat (FP), Mometasonfuroat (MF) und Fluticasonfuroat (FF) sowie ihre handelsüblichen Präparate Nasacort® (TCA), Budes® (Bud), ratioAllerg® (BDP), Flutide® Nasal (FP), Nasonex® (MF) und Avamys® (FF) untersucht. Zum Aufbringen der Wirkstoffsuspension auf die nasale Mukosa durch den Patienten kommen Dosierpumpsprays zum Einsatz, deren Dosiervorrichtung das Applikationsvolumen festlegen. Die Bestimmung des Sprühstoßvolumens unterschiedlicher handelsüblicher Präparate ergab Werte zwischen 56 und 147 µL/Sprühstoß. Zum ersten Mal wurde eine Methode entwickelt, die es ermöglichte die Glucocorticoidkonzentration in den wässrigen Überständen handelsüblicher Präparate zu bestimmen. Die Wasserlöslichkeit der unterschiedlichen Glucocorticoide sowie die galenische Zusammensetzung der Formulierungen konnten als mögliche Einflussfaktoren für den bereits gelösten Wirkstoffanteil ermittelt werden. Es konnte erstmals gezeigt werden, dass die Löslichkeit der Wirkstoffkristalle der lipophileren Substanzen wie BDP, FP, MF und FF durch das KNS signifikant verbessert wurde. Die Löslichkeit der hydrophileren Wirkstoffe wie TCA und Bud wurde durch das KNS dagegen nur leicht beeinflusst. Nach der Auflösung der Wirkstoffkristalle können die gelösten Moleküle zur cytoplasmatischen Zielstruktur, dem Glucocorticoidrezeptor diffundieren und spezifisch binden. Neben der Rezeptorbindung erfolgt auch eine unspezifische Bindung an Nasengewebe. So wurde zunächst mit einem einfachen, bereits etablierten Versuchsaufbau erstmalig die Bindung von FF an Nasengewebe im Vergleich zu anderen Glucocorticoiden in vitro untersucht. Die lipophileren Substanzen wie FF, MF und FP grenzten sich hierbei durch eine höhere Gewebebindung von den hydrophileren TCA und Bud ab. Es wurde ein neues Modell entwickelt, welches die pharmakokinetische Untersuchung zur Gewebebindung mit der Pharmakodynamik der Wirkstoffe in Form ihres antiinflammatorischen Effektes im Zellkulturmodell verknüpfen sollte. So wurde wirkstoffgesättigtes Gewebe einer extensiven Auswaschphase in Humanplasma ausgesetzt, um es anschließend mit humanen Lungenepithelzellen zu inkubieren. Es konnte gezeigt werden, dass alle Gewebeproben den Wirkstoff in das Zellkulturmedium freisetzten, was eine Hemmung der IL-8 Sekretion aus Lungenepithelzellen zur Folge hatte. Die Stärke des antiinflammatorischen Effektes der IL-8 Hemmung durch FF, FP, Bud und TCA entsprach der Kombination aus Geweberetention und intrinsischer Aktivität (Rezeptoraffinität) der Wirkstoffe. Gewebeproben des MF führten zur geringsten IL-8 Hemmung, was durch die chemische Instabilität der Substanz erklärt werden konnte. Um eine weitere Annäherung an physiologische Verhältnisse zu erreichen, wurde erfolgreich ein Modell entwickelt, welches nach Applikation der Suspension auf die Mukosa die pharmakokinetischen Abläufe simulieren sollte. Dafür entscheidend war die Einbettung respiratorischen Gewebes in eine Gel-Matrix, wodurch eine zusammenhängende Mukosa-ähnliche Oberfläche erreicht werden konnte. Als Modellsubstanzen wurden Bud aus Budes®-Nasenspray und FP aus Flutide® Nasal sowie als Vertreter der Antihistaminika Azelastin-HCl (AZ-HCl) aus Vividrin® akut eingesetzt. Es konnte gezeigt werden, dass die Gewebebindung des AZ-HCl im Vergleich zu den Glucocorticoiden am höchsten war. Die gute Korrelation der Gewebebindung der Substanzen mit ihrem Verteilungsvolumen und der Vergleich von FP mit Bud zeigte die erfolgreiche Etablierung des Modells. Beide Glucocorticoide zeigten nach Applikation der Wirkstoffsuspension zunächst eine ähnliche Assoziation an das Gewebe. Bei der Inkubation der Gewebe-Gel-Matrix mit Humanplasma wurde schließlich Fluticasonpropionat aus der Gewebe-Gel-Bindung im Vergleich zu Budesonid langsam freigesetzt. Weiterhin wurde im Gewebe-Gel-Modell die Bedeutung der Pharmakokinetik der Wirkstoffe auf die Pharmakodynamik ermittelt. Es wurden Freisetzungsproben der Gewebe-Gel-Matrices von AZ-HCl und FP auf ihre antiinflammatorische Aktivität untersucht. Die deutlich höhere Bindung des AZ-HCl an die Gewebe-Gel-Matrix äußerte sich auch in höheren Plasmakonzentrationen bei der Freisetzung im Vergleich zu FP. Jedoch konnte im Zellkulturmodell gezeigt werden, dass die Hemmung der IL-8 Freisetzung des FP, trotz der geringeren Konzentration in der Freisetzungsmatrix, das antiinflammatorische Potential des AZ-HCl übertraf.
Most drugs are no longer produced in their own countries by the pharmaceutical companies, but by contract manufacturers or at manufacturing sites in countries that can produce more cheaply. This not only makes it difficult to trace them back but also leaves room for criminal organizations to fake them unnoticed. For these reasons, it is becoming increasingly difficult to determine the exact origin of drugs. The goal of this work was to investigate how exactly this is possible by using different spectroscopic methods like nuclear magnetic resonance and near- and mid-infrared spectroscopy in combination with multivariate data analysis. As an example, 56 out of 64 different paracetamol preparations, collected from 19 countries around the world, were chosen to investigate whether it is possible to determine the pharmaceutical company, manufacturing site, or country of origin. By means of suitable pre-processing of the spectra and the different information contained in each method, principal component analysis was able to evaluate manufacturing relationships between individual companies and to differentiate between production sites or formulations. Linear discriminant analysis showed different results depending on the spectral method and purpose. For all spectroscopic methods, it was found that the classification of the preparations to their manufacturer achieves better results than the classification to their pharmaceutical company. The best results were obtained with nuclear magnetic resonance and near-infrared data, with 94.6%/99.6% and 98.7/100% of the spectra of the preparations correctly assigned to their pharmaceutical company or manufacturer.
Spectroscopic methods were established decades ago in a wide variety of fields. This also applies to the pharmaceutical field, although they initially were mostly used for identity testing or structure elucidation only. Technical developments, such as miniaturization (NMR benchtop devices), Fourier transformations (for NMR, MIR spectroscopy) or the combination with chemometric evaluation (e.g., in Process Analytical Technology, PAT), have further increased their importance and opened up new applications. The aim of this work was to investigate further new approaches and to find new applications for already established methods and to show their benefits.
By means of MIR, NIR and NMR data and their chemometric evaluation (principal component analysis, PCA; hierarchical cluster analysis, HCA; linear discriminant analysis, LDA), possibilities were presented to successfully determine the manufacturer or the pharmaceutical company of various paracetamol preparations. In the course of this, various similarities and correlations between the preparations of individual companies could also be identified. For this purpose, a suitable sample preparation was developed for each spectroscopic method, and suitable measurement parameters in order to obtain reproducible spectra for the chemometric evaluation were determined. Furthermore, the results of the two unsupervised methods (HCA, PCA) were compared with each other. The HCA was able to confirm those of the PCA for the very most part. Additionally, through these methods it was possible to characterize many of the preparations based on clusters formed by comparable tablet compositions.
In order to be able to measure unmortared, whole tablets using the NIR spectrometer, an attachment was developed and manufactured using 3D printing. Its functionality was demonstrated by measuring and analyzing the tablets of two different batches of nine paracetamol preparations. The batches were clearly distinguished on the basis of a PCA and a significant difference was also demonstrated by means of statistical tests.
For NMR spectroscopy, a method was developed to obtain optimized "fingerprint" spectra of drug formulations. For this purpose, a 1D DOSY measurement was elaborated, in which the signals of the active ingredient could be filtered out by the appropriate choice of measurement parameters. The chemometric evaluation can thus focus on the remaining signals of the excipients, on the basis of which the preparations of the same API can be distinguished. Especially in the case of formulations that consist largely of active ingredient, data pre processing of the spectra can thus be simplified and greater importance can be assigned to the originally very small excipient signals.
A quantitative 1H NMR method was developed for the comparison of a high field spectrometer (400 MHz) with a benchtop spectrometer (80 MHz) for two finished drugs. It was shown that it is possible to obtain comparable results with both instruments, but that the influence of the excipients on the signals and the lower resolution of the benchtop instrument must be taken into account. Therefore, it was not possible to obtain comparable results without further optimization of the method for one of the active ingredients.
In the investigation of various reactions between APIs and excipients using DOSY, its usefulness as a screening method in stability testing was demonstrated. For this purpose, three different APIs and excipients were stressed together and the reaction mixtures were subsequently measured using DOSY. Based on the translational diffusion coefficient, the reaction products could be identified and distinguished from the active ingredients and the excipients used. The importance of thoughtful processing could also be demonstrated. If all peak heights are selected when evaluating signals split by direct spin spin coupling, this allows the detection of hidden signals as long as not all signals have the same diffusion coefficient. The selective selection of individual peak heights in the case of split signals also enables the evaluation of signals that overlap slightly. However, the limitations of this method were also shown when two signals overlap too much and differ too little in their diffusion coefficients.
Hence, it has been successfully demonstrated in the various projects that the new chemometric approaches, as well as the new applications of already established methods, enable in depth findings and thus have a clear added value.
Der aus der in Frankreich kultivierten Meeres-Kiefer (Pinus pinaster) gewonnene und standardisierte Rindenextrakt Pycnogenol enthält neben Procyanidinen auch weitere polyphenolische sekundäre Naturstoffe und ist zudem weltweit als USP-gelistetes Nahrungsergänzungsmittel kommerziell erhältlich. Der Konsum von polyphenolreichen Lebensmitteln ist ebenso wie die Einnahme von Pycnogenol mit einer Vielzahl von positiven Effekten bei verschiedenen pathophysiologischen Prozessen assoziiert. Dazu zählen beispielsweise antioxidative oder antiinflammatorische Wirkungen, welche sowohl in vitro als auch in vivo beobachtet werden konnten. Bislang gelang es nach der Einnahme des Extraktes nicht alle in Humanserum oder -plasma detektierten Substanzen zu identifizieren; zudem ist nicht geklärt, von welchen Stoffen konkret eine Bioaktivität ausgeht oder ob diese durch synergistische Effekte zustande kommt. Aus diesen Gründen sollten in der vorliegenden Arbeit im Rahmen einer Klinischen Studie bislang nicht beschriebene Analyten in Humanserum mittels UHPLC-qTOF-MS charakterisiert werden. Hierbei wurde ein ungerichteter, metabolomischer Ansatz gewählt. Die Studienproben der Proband*innen wurden dabei also ohne etwaige Restriktionen analysiert, beispielsweise hinsichtlich möglicher Molekülstrukturen oder der Retentionszeiten der detektierten Analyten. Näher betrachtet werden sollten Analyten, die in einer individuellen Serumprobe nach Beginn der viertägigen Pycnogenol-Einnahme neu auftraten.
In Anschluss an eine Probenvorbereitung mittels methanolischer Proteinpräzipitation im sauren Milieu konnten in dem Humanserum der Proband*innen im ESI-Positiv-Modus fünf und im ESI-Negativ-Modus 23 interessante Analyten nachgewiesen werden, die auf die Einnahme von Pycnogenol zurückzuführen waren. Elf dieser Substanzen konnte eine Struktur zugeordnet werden, wobei alle ausschließlich als Sulfatkonjugate vorlagen. Zu diesen zählten neben Zimtsäure-Derivaten wie Ferulasäure-Sulfat zudem Flavonoide, z. B.
Taxifolin-Sulfat, aber auch Phenylvaleriansäure-Abkömmlinge, beispielsweise Hydroxydihydroxyphenylvaleriansäure-Sulfat, sowie Vertreter aus der Gruppe der Benzoesäuren und weitere Aromaten wie z. B. Pyrogallol-Sulfat oder Protocatechusäure-Sulfat.
Nach unserem besten Wissen war der Aspekt der ausschließlichen Sulfatierung neuartig. Wie aufgrund des interindividuell variablen Metabolismus zu erwarten, insbesondere durch das enterale Mikrobiom, war die Verteilung dieser sogenannten Marker innerhalb der 15 Studienteilnehmenden sehr heterogen. Nicht jeder Marker wurde bei jeder Person erfasst; die Spannweite reichte dabei von einem Teilnehmenden im Falle des mikrobiellen Metaboliten Hydroxyphenylvaleriansäure-Sulfat bis hin zu 14 Proband*innen bei einer nicht-identifizierbaren, jedoch wahrscheinlich endogenen Substanz im ESI-Positiv-Modus. Am häufigsten wurden die elf zuordenbaren Analyten vier Stunden nach der Einnahme von Pycnogenol über einen Zeitraum von vier Tagen bestimmt.
Im Anschluss sollte die Bioaktivität dieser Substanzen in einem endothelialen Zellkulturmodell untersucht werden. Das Endothel wurde als Zielstruktur gewählt, da eine endotheliale Dysfunktion in der Pathogenese einer Reihe von Krankheiten mit ausgeprägter Mortalität und Morbidität eine bedeutende Rolle spielt. Zudem wurde bereits eine positive Wirkung auf die Endothelfunktion nach der Einnahme von Pycnogenol beschrieben, wobei bis dato der Mechanismus auf molekularer Ebene unklar war.
Die Charakterisierung der Sulfatkonjugate bezüglich ihrer Bioaktivität ex vivo mit humanen Endothelzellen aus der Nabelschnurvene (HUVEC) gestaltete sich herausfordernd. Initial sollte untersucht werden, inwiefern diese Substanzen einer durch einen Entzündungsstimulus hervorgerufenen Schädigung der endothelialen Glycocalyx entgegenwirken oder diese vermeiden können. Allerdings ließen sich mit den verschiedenen inflammatorischen Stimuli Lipopolysaccharid (LPS), Tumornekrosefaktor-α (TNF-α) und Wasserstoffperoxid bezüglich Konzentration und Inkubationsdauer keine reproduzierbaren Kulturbedingungen für eine valide ELISA-Quantifizierung des endothelialen Markers Heparansulfat etablieren. Im Anschluss erfolgte unter dem Einfluss einer TNF-α-Stimulation ein orientierendes Screening mit den Monosubstanzen Ferulasäure und Protocatechusäure bzw. mit deren Sulfatkonjugaten in Konzentrationen von 0,1 und 0,5 µM. Dabei zeigten die Konjugate beider Analyten bei der niedrigeren Konzentration tendenziell eine glycocalyx-protektive Wirkung, welche bei der höheren Konzentration jedoch nicht mehr beobachtet werden konnte. Die endotheliale Permeabilität wurde mittels eines FITC-Dextran-Permeabilitäts-Assays untersucht. Hiermit sollte ebenfalls ein möglicher endothel-protektiver Einfluss der sulfatierten Substanzen unter entzündlichen Bedingungen (TNF-α-Stimulation) beleuchtet werden. Jedoch konnte weder bei Ferulasäure oder Protocatechusäure noch bei deren Sulfatkonjugate oder Taxifolin in diesem Modell ein Einfluss auf die endotheliale Barrierefunktion erfasst werden.
Ursprünglich war abschließend geplant in einem ex vivo-Modell die Humanserum-Proben mit dem darin enthaltenen Gemisch aus möglicherweise bioaktiven Metaboliten direkt im Zellkulturmodell auf ihre Wirkung zu testen. Dies hat den Vorteil, dass simultan synergistische Effekte und Einflüsse der Matrix untersucht werden können und ausschließlich in vivo erreichbare Konzentrationen eingesetzt werden. Aufgrund der limitierten Verfügbarkeit der Studienproben und der oben geschilderten heterogenen Ergebnisse wurde auf eine weitere Analyse im Rahmen eines ex vivo-Modells verzichtet.
Mit der vorliegenden Arbeit konnte gezeigt werden, dass nach der Einnahme von Pycnogenol resorbierte Bestandteile und Metabolite in Humanserum ausschließlich als Sulfatkonjugate vorlagen. Zudem wurde bezüglich der Evaluation der endothelialen Bioaktivität durch Polyphenole eine Grundlage für weitere Untersuchungen geschaffen. Damit konnte ein Beitrag zur pharmakokinetischen und -dynamischen Charakterisierung von Pycnogenol geleistet werden.
Charakterisierung der Mikrostruktur und der Permeationseigenschaften von Polysiloxan-Netzwerken

Ziel dieser Arbeit war es, die Permeationseigenschaften von Polysiloxan-Membranen im Hinblick auf ihre definierte Mikrostruktur näher zu analysieren. Es konnte gezeigt werden, dass die Mikrostruktur der Membranen einen statistisch signifikanten Einfluss auf die Permeationsgeschwindigkeit der untersuchten Substanzen hat. Zum besseren Verständnis der Permeation wurden auch die Diffusionsvorgänge innerhalb der Membran untersucht. Durch den Einsatz der Konfokalen Raman-Spektroskopie ist es gelungen, den Aufbau eines Konzentrationsgradienten innerhalb der Membran zu zeigen. Weiterhin konnte der Einfluss der Mikrostruktur der Membranen auf die Geschwindigkeit des Aufbaus dieses Gradienten nachgewiesen werden.
