In der vorliegenden Arbeit sollte ein Interview auf die Gütekriterien Objektivität, Validität und Reliabilität überprüft werden. Es sollte untersucht werden, ob das Interview geeignet ist, aktuell vorhandene Symptome der ADHS und der ODD dimensional zu erfassen. Dabei wurden 61 Patienten der Klinik und Poliklinik für Kinder- und Jugendpsychiatrie und Psychotherapie der Universität Würzburg mit ihren Müttern befragt. Die Objektivität wurde überprüft, indem das Interview auf Video aufgezeichnet und nochmals von einem zweiten Beurteiler ausgewertet wurde. Die Summenwerte der beiden Interviewer wurden miteinander korreliert. Die Korrelation war signifikant und ergab einen Wert von rk= ,98. Die Objektivität im Sinne der Beurteilerübereinstimmung kann somit als hoch angesehen werden. Es konnte gezeigt werden, dass die zusammenfassende Beurteilung des Interviewers höher mit der Einschätzung aufgrund der Angaben der Mütter korreliert als mit der Einschätzung aufgrund der Angaben der Kinder. Die Korrelation zwischen der zusammenfassenden Beurteilung und der Einschätzung aufgrund der Angaben der Müttern ergab einen Wert von r= ,98, die Korrelation zwischen der zusammenfassenden Beurteilung und der Einschätzung aufgrund der Angaben der Kindern einen Wert von r= ,57. Die zusammenfassende Beurteilung des Interviewers gründet demnach im Wesentlichen auf den Angaben der Mütter. Die Konstruktvalidität wurde ermittelt, indem das Interview mit anderen diagnostischen Verfahren verglichen wurde. Die Korrelation des Interviews mit ADHS-nahen Konstrukten war signifikant und ergab Werte zwischen rtc= ,48 und rtc= ,70. Die diskriminante Validität wurde durch Korrelation mit ADHS-fernen Konstrukten ermittelt. Der Korrelationskoeffizient betrug rtc= ,27. Die Validität liegt somit im mittleren bis oberen Bereich. Ebenfalls wurde belegt, dass Kinder mit zusätzlicher Störung des Sozialverhaltens einen höheren Gesamtscore im Interview erreichen. Das Interview diskriminiert demnach erwartungsgemäß zwischen ADHS-Patienten mit zusätzlicher Störung des Sozialverhaltens und ADHS-Patienten ohne zusätzliche Störung. Die Überprüfung der Gütekriterien erzielte gute Ergebnisse für Objektivität und Validität. Demnach werden Symptome der ADHS und der oppositionellen Störung des Sozialverhaltens mit dem untersuchten Verfahren in hinreichender Güte erfasst.
Theories of attention deficit hyperactivity disorder (ADHD) aetiology have placed a focus on impaired behavioural inhibition presumably leading to executive function (EF) deficits. Neuroimaging studies report neurophysiological findings consistent with these hypothesised impairments, and investigations of functional brain activation from a network perspective report hypoactivation in the frontoparietal network as well as hyperactivation in the dorsal attention network. Studies investigating the acute effects of stimulant medication on EF show an improvement on behavioural EF measures including working memory. In addition, methylphenidate (MPH) was shown to up-regulate the task-positive/ frontoparietal network in children and adolescents with ADHD. So far, there are only few studies investigating the impact of ADHD on behavioural and neurophysiological EF measures as well as the effect of several weeks of stimulant medication in adult patients.
The importance of the catechol-O-methyltransferase (COMT) enzyme for subcortical and cortical dopaminergic and noradrenergic functioning furthermore led to studies investigating a potential interactive impact of COMT genotype and ADHD on neuropsychological functioning, with a particular focus on working memory. The results of these studies were very heterogeneous. In addition, as none of the studies compared the results of ADHD patients to those of a healthy control group, possible differential effects of COMT in patients and healthy controls could not be examined.
The aim of this dissertation was to investigate selective attention properties of the central executive component during a working memory task and to transfer this task to fMRI. A third study then aimed to investigate the effects of adult ADHD (aADHD), MPH, and COMT genotype on working memory with a particular focus on activation of the task-positive network during the analysis of the fMRI data.
The first study (EEG) could replicate and extend the results from previous research. This study could furthermore connect the overall activation in frontal areas to suppression efficiency in posterior visual areas as well as establish the impact of hyperactive/ impulsive ADHD symptoms on task performance. The second study (fMRI) allowed the successful transfer of the paradigm to fMRI, and the further replication and extension of previous findings. In addition, this study showed the sensitivity of the task to the effects of the COMT genotype. The third study (fMRI) was one of the first studies that exploratorily investigated the effects COMT in a sample of aADHD patients and a comparable healthy control group. This study showed an interactive effect of these two factors on neuropsychological measures as well as on fMRI activation during a classic n-back working memory task. In addition, this task led to more activation in the task-positive network of the aADHD group compared to a healthy control group in the absence of performance differences, pointing towards compensatory activation in the aADHD group. Furthermore, activation in the frontal cortex was increased in patients taking MPH compared to a placebo. The fMRI data from the selective attention task moreover showed decreased activation in the right DLPFC of the patient group, which was associated with reduced suppression efficiency across all participants. The clinical effect of MPH in the third study was visible but did not reach significance, which is probably attributable to a lack of experimental power.
The studies in this dissertation could successfully replicate and extend previous findings. A goal for future studies should be the further investigation of the interactive effects of COMT genotype and aADHD on neuropsychological test results and fMRI activation, but also on medication response and adverse effects. In this context, the adaptation of a network perspective during the analysis of fMRI data seems to be the best way to detect existing between-group differences.
The Contribution of Common and Rare Variants to the Complex Genetics of Psychiatric Disorders

Attention deficit/hyperactivity disorder (ADHD), one of the most frequent childhood-onset, chronic and lifelong neurodevelopmental diseases, affects 5 - 10% of school – aged children and adolescents, and 4% of adults. The classified basic symptoms are - according to the diagnostic system DSM-VI - inattentiveness, impulsivity and hyperactivity. Also daily life of patients is impaired by learning problems, relationship crises, conflicts with authority and unemployment, but also comorbidities like sleep - and eating problems, mood - and anxiety disorders, depression and substance abuse disorders are frequently observed. Although several twin and family studies have suggested heritability of ADHD, the likely involvement of multiple genes and environmental factors has hampered the elucidation of its etiology and pathogenesis. Due to the successful medication of ADHD with dopaminergic drugs like methylphenidate, up to now, the search for candidate genes has mainly focused on the dopaminergic and - because of strong interactions - the serotonergic system, including the already analyzed candidate genes DAT1, DRD4 and 5, DBH or 5-HTTLPR. Recently, DNA copy number changes have been implicated in the development of a number of neurodevelopmental diseases and the analysis of chromosomal gains and losses by Array Comparative Genomic Hybridization (Array CGH) has turned out a successful strategy to identify disease associated genes. Here we present the first systematic screen for chromosomal imbalances in ADHD using sub-megabase resolution Array CGH. To detect micro-deletions and -duplications which may play a role in the pathogenesis of ADHD, we carried out a genome-wide screen for copy number variations (CNVs) in a cohort of 99 children and adolescents with severe ADHD. Using high-resolution aCGH, a total of 17 potentially syndrome-associated CNVs were identified. The aberrations comprise four deletions and 13 duplications with approximate sizes ranging from 110 kb to 3 Mb. Two CNVs occurred de novo and nine were inherited from a parent with ADHD, whereas five are transmitted by an unaffected parent. Candidates include genes expressing acetylcholine-metabolising butyrylcholinesterase (BCHE), contained in a de novo chromosome 3q26.1 deletion, and a brain-specific pleckstrin homology domain-containing protein (PLEKHB1), with an established function in primary sensory neurons, in two siblings carrying a 11q13.4 duplication inherited from their affected mother. Other genes potentially influencing ADHD-related psychopathology and involved in aberrations inherited from affected parents are the genes for the mitochondrial NADH dehydrogenase 1 alpha subcomplex assembly factor 2 (NDUFAF2), the brain-specific phosphodiesterase 4D isoform 6 (PDE4D6), and the neuronal glucose transporter 3 (SLC2A3). The gene encoding neuropeptide Y (NPY) was included in a ~3 Mb duplication on chromosome 7p15.2-15.3, and investigation of additional family members showed a nominally significant association of this 7p15 duplication with increased NPY plasma concentrations (empirical FBAT, p = 0.023). Lower activation of the left ventral striatum and left posterior insula during anticipation of large rewards or losses elicited by fMRI links gene dose-dependent increases in NPY to reward and emotion processing in duplication carriers. Additionally, further candidate genes were examined via Matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS). This method enables the analysis of SNPs directly from human genomic DNA without the need for initial target amplification by PCR. All these findings implicate CNVs of behavior-related genes in the pathogenesis of ADHD and are consistent with the notion that both frequent and rare variants influence the development of this common multifactorial syndrome. The second part of this work concentrates on MLC1, a gene associated with Megalencephalic leukoencephalopathy with subcortical cysts, located on chromosome 22q13.33. To get more insight in the disease itself, a targeting vector for a conditional knockout mouse was constructed using homologous recombination. Furthermore, MLC1 has been suggested as a risk gene for schizophrenia, especially the periodic catatonia subtype. An initially identified missense mutation was found to be extremely rare in other patient cohorts; however, a recent report again argued for an association of two intronic MLC1 SNPs with schizophrenia and bipolar disorder. A case-control study of these polymorphisms as well as SNPs in the transcriptional control region of MLC1 was conducted in 212 chronic schizophrenic patients, 56 of which suffered from periodic catatonia, 106 bipolar patients, and 284 controls. Both intronic and promoter polymorphisms were specifically and significantly associated with periodic catatonia but not schizophrenia or bipolar disorder in general. A haplotype constructed from all polymorphisms was also associated with periodic catatonia. The MLC1 variation is associated with periodic catatonia; whether it constitutes a susceptibility or a modifier gene has to be determined.
Bei ADHS (Aufmerksamkeitsdefizit- / Hyperaktivitätsstörung) handelt es sich um eine meist im Kindesalter beginnende psychiatrische Erkrankung, die die betroffenen Menschen meist bis ins erwachsene Alter begleitet. In den letzten Jahrzehnten wurden viele Untersuchungen an Kindern, welche an ADHS erkrankt waren, durchgeführt. Es zeigten sich Veränderungen in der Power verschiedener Frequenzbereiche. Im Vergleich mit gesunden Kindern zeigten die jungen ADHS Patienten eine Zunahme an langsamer Wellenaktivität, sowie eine Abnahme der schnellen Wellenaktivität. Ähnliche Studien mit erwachsenen ADHS- Patienten sind dagegen seltener durchgeführt worden. Dabei wurde, analog zu den Kinderstudien, eine Zunahme der langsamen Wellenaktivität festgestellt. Im Gegensatz zu den Kindern, wurde bei den Erwachsenen zusätzlich eine Zunahme der Alphaaktivität beobachtet. Des Weiteren stimmten die Ergebnisse bezüglich der zunehmenden Aktivität des Betabandes ebenso nicht überein, wie die fronto- zentral normale und parietal verminderte Betaaktivität bei erwachsenen ADHS- Patienten. Aus diesem Grund verfolgt die vorliegende Studie das Ziel, Unterschiede in den EEG- Frequenzbändern zwischen erwachsenen ADHS- Patienten und einer Kontrollgruppe herauszuarbeiten. Dazu wurden EEG- Frequenzen von 31 an ADHS erkrankten Erwachsenen, mit denen von 31 gesunden Kontrollpersonen verglichen. Die beiden Gruppen unterschieden sich im Durchschnitt nicht hinsichtlich Alter, Geschlecht und Händigkeit. Ein Ruhe- EEG wurde, für jeweils zweimal 5 min pro Proband, mit Hilfe von 21 Elektroden abgeleitet. Die Elektroden wurden gemäß des international anerkannten 10-20 Systems angebracht. Die auf diese Weise aufgezeichneten Daten wurden zunächst mit dem Programm Brain Vision Analyzer bearbeitet. Anschließend wurde die Summe der einzelnen Frequenzbänder mit Hilfe von FFT (Fast Fourier Transformation) in ihre Einzelbänder zerlegt. Jedes Frequenzband wurde einzeln hinsichtlich absoluter Power analysiert. Die ADHS Patienten zeigten eine signifikante Zunahme der Power im Alpha- und Thetaband. Keine Unterschiede wurden für das Betaband ausgemacht. Unsere Studie zeigt, dass die Abnormalitäten innerhalb des EEG-Power-Spektrums bei an ADHS erkrankten Erwachsenen nicht mit denen der an ADHS erkrankten Kinder übereinstimmen. Die power density des Alpha- und des Thetabandes könnte ein zuverlässiges Kriterium darstellen, um gesunde und an ADHS erkrankte Erwachsene zu unterscheiden. In einem weiteren Ansatz sollte ein Zusammenhang zwischen der ADHS- Symptomatik und EEG- Power bei 163 Gesunden untersucht werden. Die eventuell vorhandene ADHS- Symptomatik wurde anhand des ASRS- Fragebogens (Adult Self- Report Scale) ermittelt. Über einen Auswertungsschlüssel kann zum einen die Neigung zu Unaufmerksamkeit, zum anderen zu Hyperaktivität/ Impulsivität aufgezeigt werden. Die Ableitung des EEG erfolgte nach der oben erwähnten Methode. Die Ergebnisse, die die Korrelation zwischen Kontrollgruppe und Fragebogen betreffen, müssen als wenig aussagekräftig beurteilt werden. Lässt man innerhalb dieser Studie das Geschlecht außen vor, so wird deutlich, dass nahezu keine signifikanten Zusammenhänge vorliegen. Unter Berücksichtigung des Geschlechts konnten wir bei gesunden Frauen, ähnlich wie Morgan et al. (2005), mehr Delta- und Betaaktivität als bei den gesunden, männlichen Probanden messen. Im Vergleich zwischen Kontrollgruppe und Patienten konnten wir keine geschlechtsspezifischen Unterschiede ausmachen. Deswegen vermuten wir, dass sich geschlechtsspezifische Unterschiede zwischen Erkrankten und Gesunden im Erwachsenenalter vermindern (Clarke et al. 2001b) Basierend auf unseren Ergebnissen scheinen Langzeitstudien für ADHS Patienten sinnvoll, um die Unterschiede von EEG- Abnormitäten während des Heranwachens zu erforschen und zu berücksichtigen.
Bei Kindern mit Aufmerksamkeitsdefizit-/Hyperaktivitätsstörung (ADHS) konnten Defizite in der Leistung des Arbeitsgedächtnisses nachgewiesen werden. In zahlreichen funktionell bildgebenden Studien zeigte sich außerdem ein mit dem Arbeitsgedächtnis assoziierter Unterschied in der präfrontalen Aktivierung im Vergleich zu Gesunden. Bisher wurde jedoch noch nicht näher analysiert, ob diese Defizite von der Komponente (visuell-räumliches (SWM) oder visuell-bildliches Arbeitsgedächtnis (OWM)) oder dem Prozess des Arbeitsgedächtnisses (untergeordnete Speichersysteme oder zentrale Exekutive) abhängig sind. Diese Arbeit ist die erste, die mit der funktionellen Nah-Infrarot Spektroskopie (fNIRS) das Arbeitsgedächtnis bei Kindern mit ADHS untersucht. Dabei war das Hauptziel zum einen die Replikation von Gruppenunterschieden in Verhalten und präfrontaler Aktivierung und zum anderen die Gewinnung näherer Informationen über die differentielle Ausprägung der präfrontalen Defizite bei ADHS. Die Stichprobe bestand aus 19 Kindern mit ADHS vom kombinierten Typ (DSM-IV) und 19 Kontrollkindern, die vergleichbar waren nach Alter (8-15 Jahre), Geschlecht, Händigkeit und Intelligenz. Das von uns gewählte Paradigma differenzierte zwischen Aufgaben zum OWM und SWM, innerhalb derer wiederum zwei prozessabhängige Phasen (Speicherung und Abgleich) unterschieden wurden, und enthielt zusätzlich eine Kontrollaufgabe (CON). Die präfrontale Aktivierung wurde mittels fNIRS über die Konzentrationsänderungen der Chromophoren oxygeniertes Hämoglobin (O2Hb) und deoxygeniertes Hämoglobin (HHb) bestimmt. Es zeigte sich ein signifikant unterschiedliches Verhalten abhängig von der Bedingung (CON > SWM > OWM). Eine typische präfrontale Aktivierung des Arbeitsgedächtnisses mit der höchsten Aktivierung im dorsolateralen präfrontalen Kortex (DLPFC) beschränkte sich auf die Abgleichphase. Weder in Hinblick auf das Verhalten noch auf die präfrontale Aktivierung konnten signifikante Gruppenunterschiede nachgewiesen werden. Für das Nichtauftreten von Gruppenunterschieden können verschiedene mögliche Gründe angeführt werden. Unter anderem legten explorative Analysen nahe, dass Kinder mit ADHS und gesunde Kinder unterschiedlichen Reifungsprozessen unterliegen. Auch scheint die chronische Einnahme der ADHS-Standardmedikation einen Einfluss auf die Hirnaktivität zu nehmen. Die Diskussion unserer Ergebnisse stellt zusammen mit der Offenlegung von Kritikpunkten an unserer sowie an vorhergehenden Studien einen wichtigen Ansatzpunkt zur Versuchsoptimierung in zukünftigen Arbeiten dar. Darüber hinaus ist weitere Forschung nötig, um detailliertere Kenntnisse über den Einfluss von Alter, Reifungsprozessen, chronischer MPH-Medikation, ADHS-Subtypen und dem Aufgabendesign zu erlangen.
Mit der vorliegenden Arbeit soll die Ausprägung der Delinquenz und deren Vorhersagemöglichkeiten bei ehemaligen Patienten der Kinder- und Jugendpsychiatrie Würzburgs mit der Diagnose einer ADHS untersucht werden. Anhand von schrittweisen Diskriminanzanalysen wurden verschiedene Faktoren auf deren Vorhersagekraft für eine spätere Delinquenz untersucht. Dabei konnte mit sechs Variablen bei 74% der Probanden richtig zwíschen später vorhandener oder nicht vorhandener Delinquenz unterschieden werden. Noch bessere Ergebnisse wurden bei dem Versuch erzielt, Vergehen in bestimmten Deliktuntergurppen vorherzusagen.
Attention-Deficit/Hyperactivity Disorder (ADHD) endophenotypes as a link between phenotype and genotype were the focus of the present work. Candidate endophenotypes were investigated via neuropsychological tasks during the simultaneous recording of a 21-channel electroencephalogram. Since endophenotypes are assumed to more closely reflect genetic variation, the influence of ADHD-associated genes Catechol-O-methyl transferase (COMT), the dopamine transporter (DAT, SLC6A3) and Latrophilin-3 (LPHN3) was analysed. Response inhibition was assessed with a cued Continuous Performance Test, for working memory we used an n-back task, sensory gating was measured via the paired clicks paradigm and response time variability (RTV) was quantified by the standard deviation of reaction times. The sample comprised medicated (N=36) and unmedicated (N=42) ADHD patients and matched control children and adolescents (N=41). The electrophysiological correlate of response inhibition was the centroid location during response execution and inhibition, and the degree of anteriorization (NGA). Sensory gating reflects the attenuation of the P50 response to the second of two auditory stimuli presented in short succession. Working memory was examined during target and non-target trials, reflecting specific information processing stages: early sensory processing (P100 and N100), selection of material (P150), memory retrieval (N300), event categorization (P300) and updating of working memory content (P450). Performance was quantified in terms of omission errors reflecting inattention and false alarms reflecting impulsivity, as well as speed and variability of reactions. Unmedicated ADHD patients had more omission errors and more variable reaction times, pointing to difficulties with attention and state regulation. NGA did not prove an optimal endophenotype candidate, since it was not yet developed in approximately half of the examined children and adolescents. It was independent of diagnosis; however ADHD risk alleles for DAT conferred lower NGA as well as more variable reaction times across groups. DAT genotype interacted with diagnosis on the level of centroid location, however, it did not manifest in performance deficits. In the case of sensory gating, homozygosity for the DAT allele associated with ADHD (10R) conferred impairment. ADHD was only relevant in participants without genetic risk, where patients without medication struggled most with suppression. In the working memory task, DAT modulated the timing of material selection in interaction with cognitive load and diagnosis: under high load unmedicated patients showed delayed responses, while under low load risk carriers on medication had faster responses than controls. Early processing and event-categorization were stronger in unmedicated ADHD with risk genotype, but dampened without risk. An interesting trend emerged for LPHN3, where carrying all risk variants was associated with higher NGA in ADHD patients irrespective of medication. This warrants further study, as the haplotype also exerts a positive influence on sensory gating specifically in patients. At the same time within the genetic risk group, unmedicated patients had the weakest NGA. However, the LPHN3 risk haplotype effected more posterior Go centroids, putatively facilitating response execution, which is supported by a higher number of false alarms. When inhibition was required, the risk variants led to more posterior centroids in unmedicated compared to medicated patients as well as controls, speaking to differences in inhibition-related brain activation. While as expected the risk haplotype led to compromised gating in unmedicated ADHD, this was reversed in healthy controls where the haplotype was acting in a protective manner with enhanced filtering. During working memory operations, the risk haplotype showed stronger N300 responses suggesting investment of more resources. While COMT did not exert an influence on NGA directly, carriers of the risk allele (met) had more posterior centroids both during response execution and inhibition, and displayed more variable responses in addition to being more prone to false alarms. Unmedicated patients produced smaller P300 during successful execution of responses than controls in absence of the risk allele, while with risk they had shorter latencies and presumably tend towards premature reactions. Additionally, it brought out impairments in sensory gating, thus making unmedicated patients less able to filter out irrelevant information, while they were able to compensate with the protective genotype. The influence of COMT on sensory gating seems to be specific for ADHD, as this gene was of no consequence in healthy controls. In the working memory task, met was beneficial for updating as reflected by P450 amplitude. In ADHD irrespective of medication COMT did not change P450 strength, but for controls this effect was observed.
EEG-Studie mit 34 erwachsenen ADHS-Patienten und 34 Kontrollpersonen mit einem modifizierten Eriksen-Flanker-Test. Dabei zeigten sich bei den ADHS Patienten verglichen mit der Kontrollgruppe verlängerte Reaktionszeiten, aber eine normale Fehleranzahl und ein normales post error slowing. Die ERN und die Pe waren vermindert. Dies weist auf Defizite in der Handlungsüberwachung bei erwachsenen ADHS-Patienten hin.
Humans and other animals share choice preference for smaller-but-sooner over later-but-larger rewards, indicating that the subjective value of a reward is discounted as a function of time. This phenomenon referred to as delay discounting (DD), represents one facet of impulsivity which is inherently connected with reward processing and, within a certain range, adaptive. Maladaptive levels, however, can lead to suboptimal decision-making and represent important characteristics of psychopathologies such as attention-deficit/hyperactivity disorder (ADHD). In line with a proposed influence of dysregulated dopamine (DA) levels on impulsivity, neural structures involved in DD (the ventral-striatum [VS]; orbitofrontal cortex [OFC]) are highly innervated by dopaminergic neurons. However, studies explicitly testing the triadic interplay of dopaminergic neurotransmission, impulsivity and brain activation during intertemporal choice are missing. Therefore, the first study of the thesis examined the effect of different DA-bioavailability levels, indicated by a genetic polymorphism (Val158Met) in the gene of the catechol-O-methyltransferase, on the association of delay discounting and OFC activation. OFC response to monetary rewards that varied by delay-to-delivery was recorded with functional near-infrared spectroscopy (fNIRS) in a sample of 49 healthy human subjects. The results suggest a DA-related enhancement in OFC function from low (low DA level) to partial (intermediate DA level) and full (high DA level) reward delay sensitivity. Furthermore, DA-bioavailability was shown to moderate the association of neural reward delay sensitivity and impulsivity: OFC reward delay sensitivity was strongly correlated with impulsivity at intermediate DA-levels, but not at low or high DA-levels where impulsivity was related to delay-independent OFC amplitudes. It is concluded that DA-level should be considered as a crucial factor whenever impulsivity-related brain activation, in particular to reward delay, is examined in healthy subjects. Dysfunctional reward processing, accompanied by a limited ability to tolerate reward delays (delay aversion), has been proposed as an important feature in ADHD putatively caused by striatal hypo-dopaminergia. Therefore, the aim of the second study of this thesis was to examine subcortical processing of reward delays and to test for neural indicators of a negative emotional response to delay periods. Using functional magnetic resonance imaging (fMRI), brain activation in adult patients with ADHD (n=14) and healthy control subjects (n=12) was recorded during the processing of immediate and delayed rewards. Compared with healthy control subjects, hyporesponsiveness of the VS reward system was evident in patients with ADHD for both immediate and delayed rewards. In contrast, delayed rewards evoked hyperactivation in the dorsal caudate nucleus and the amygdala of ADHD patients, corroborating the central predictions of the delay aversion hypothesis. In combination both studies support the conception of a close link between delay discounting, brain activation and dopaminergic neurotransmission. The results implicate that studies on neural correlates of DD have to account for the DA-bioavailability level and for a negative emotional response to reward delays.
Attention-deficit/hyperactivity disorder (ADHD) is a genetically complex childhood onset neurodevelopmental disorder which is highly persistent into adulthood. Several chromo-somal regions associated with this disorder were identified previously in genome-wide linkage scans, association (GWA) and copy number variation (CNV) studies. In this work the results of case-control and family-based association studies using a can-didate gene approach are presented. For this purpose, possible candidate genes for ADHD have been finemapped using mass array-based SNP genotyping. The genes KCNIP4, CDH13 and DIRAS2 have been found to be associated with ADHD and, in addition, with cluster B and cluster C personality disorders (PD) which are known to be related to ADHD. Most of the associations found in this work would not withstand correction for multiple testing. However, a replication in several independent populations has been achieved and in conjunction with previous evidence from linkage, GWA and CNV studies, it is assumed that there are true associations between those genes and ADHD. Further investigation of DIRAS2 by quantitative real-time PCR (qPCR) revealed expression in the hippocampus, cerebral cortex and cerebellum of the human brain and a significant increase in Diras2 expression in the mouse brain during early development. In situ hybrid-izations on murine brain slices confirmed the results gained by qPCR in the human brain. Moreover, Diras2 is expressed in the basolateral amygdala, structures of the olfactory system and several other brain regions which have been implicated in the psychopatholo-gy of ADHD. In conclusion, the results of this work provide further support to the existence of a strong genetic component in the pathophysiology of ADHD and related disorders. KCNIP4, CDH13 and DIRAS2 are promising candidates and need to be further examined to get more knowledge about the neurobiological basis of this common disease. This knowledge is essential for understanding the molecular mechanisms underlying the emergence of this disorder and for the development of new treatment strategies.
