Diese Arbeit beschäftigt sich mit der Kompatibilität in der medizinischen Bildgebung unter zwei verschiedenen Aspekten: (A) Beeinflussung von Gradientenfeldern durch das Magnetsystem eines Magnetresonanztomographen. (B) Beeinflussung elektronischer Bauteile durch ionisierende Strahlung.
Imperfektionen in der Gradientenhardware (7–13) führen dazu, dass nicht die ideale zeitliche Gradientenform ausgespielt wird, sondern eine verzerrte Version der Gradienten (6,14). In der nicht-kartesischen Bildgebung führen diese resultierenden Abweichungen in den k-Raum Trajektorien zu Bildartefakten, die sich negativ auf die Diagnosestellung auswirken können. Die linearen und zeitinvarianten Eigenschaften des Gradientensystems ermöglichen die Bestimmung der Übertragungsfunktion (GSTF) (20). Diese Übertragungsfunktion kann innerhalb der Bildrekonstruktion zur Trajektorienkorrektur verwendet werden (14,15,70). In dieser Arbeit wurden mit der Feldkamera (Skope Magnetic Resonance Technologies, Zürich, Schweiz) (22,23) und der schichtselektiven Phantommethode (5,6) zwei etablierte GSTF-Messverfahren verglichen. Dabei wurde die Notwendigkeit einer Abtastzeitkompensation festgestellt, um die GSTF-Informationen entsprechend der gewählten Abtastzeit zu korrigieren (s. Abbildung 16) und die Trajektorien hinreichend zu korrigieren und damit Bildartefakte zu reduzieren. Die Langzeit- und Temperaturanalyse der GSTF zeigte für zwei verschiedene Siemens-Tomographen (Siemens Healthcare, Erlangen, Germany) eine Langzeit und Temperaturstabilität, auch bei extensiven Duty-Cyclen. Damit lässt sich auch einfach eine Pre-emphasis-Korrektur der Gradienten realisieren, was exemplarisch mit einer Zig-Zag- und einer Spiral-Sequenz gezeigt werden konnte. Die GSTF-Pre-emphasis-Korrektur lieferte dabei ähnliche Ergebnisse wie die GSTF-Post-Processing-Technik (s. Abbildung 44 und 47).
In Bezug auf die Kompatibilität in der medizinischen Bildgebung wurde in dieser Arbeit auch die Beeinflussung von medizinischen Implantaten durch ionisierende Strahlung untersucht. Herzschrittmacher, Kardioverter-Defibrillatoren oder andere aktive medizini- sche Implantate können in ihrer Funktion durch ionisierende Strahlung, die bei verschiedenen diagnostischen und therapeutischen Anwendungen appliziert wird, beeinträchtigt werden (28,97,111). In dieser Studie wurden verschiedene elektronische Bauteile, wie Kondensatoren, Transistoren, Batterien und Speicherkarten in einer gewebeäquivalenten Messumgebung bestrahlt und dabei auf ihre Funktionalität überprüft. Die Messumgebung simuliert dabei die Wechselwirkungseigenschaften von menschlichem Gewebe mit ionisierender Strahlung in einem Energiebereich von 10 keV – 6 MeV. Zudem ermöglicht sie mit der Einschubeinheit die Integration von Implantaten/elektronischen Bauteilen, sowie eine realistische Bestrahlungsplanung und Dosisverifikation (35,77). Bei den Kondensatoren zeigten sich während der Bestrahlung ein verändertes Funktionsverhalten, mit signifikant abweichenden Spannungen und Zeitkonstanten gegenüber dem unbestrahlten Zustand. Auch die Batterien haben sich während der Bestrahlung signifikant schneller entladen, als ohne Strahlungsapplikation. Nach der Bestrahlung konnten bei den untersuchten SD-Speicherkarten auch Veränderungen in den Speicherzellen festgestellt werden. Bei den Transistoren war aufgrund von Fehlern im Messsetup und dem Schaltungsdesign keine genauere teststatistische Auswertung möglich. Zusammenfassend lässt sich sagen, dass sich charakteristische Kenngrößen der untersuchten Bauteile bei Strahlungsapplikation signifikant veränderten.
Purpose:
The gradient system transfer function (GSTF) characterizes the frequency transfer behavior of a dynamic gradient system and can be used to correct non‐Cartesian k‐space trajectories. This study analyzes the impact of the gradient coil temperature of a 3T scanner on the GSTF.
Methods:
GSTF self‐ and B\(_0\)‐cross‐terms were acquired for a 3T Siemens scanner (Siemens Healthcare, Erlangen, Germany) using a phantom‐based measurement technique. The GSTF terms were measured for various temperature states up to 45°C. The gradient coil temperatures were measured continuously utilizing 12 temperature sensors which are integrated by the vendor. Different modeling approaches were applied and compared.
Results:
The self‐terms depend linearly on temperature, whereas the B0‐cross‐term does not. Effects induced by thermal variation are negligible for the phase response. The self‐terms are best represented by a linear model including the three gradient coil sensors that showed the maximum temperature dependence for the three axes. The use of time derivatives of the temperature did not lead to an improvement of the model. The B\(_0\)‐cross‐terms can be modeled by a convolution model which considers coil‐specific heat transportation.
Conclusion:
The temperature dependency of the GSTF was analyzed for a 3T Siemens scanner. The self‐ and B0‐cross‐terms can be modeled using a linear and convolution modeling approach based on the three main temperature sensor elements.
Carrion plays an essential role in shaping the structure and functioning of ecosystems and has far‐reaching implications for biodiversity conservation. The change in availability and type of carcasses throughout ecosystems can involve negative effects for scavenging communities. To address this issue, there have been recent conservation management measures of carrion provision in natural systems. However, the optimal conditions under which exposing carcasses to optimize conservation outcomes are still limited. Here, we used camera traps throughout elevational and vegetational gradients to monitor the consumption of 48 deer carcasses over a study period of six years by evaluating 270,279 photographs resulting out of 15,373 trap nights. We detected 17 species visiting carcass deployments, including five endangered species. Our results show that large carcasses, the winter season, and a heterogeneous surrounding habitat enhanced the frequency of carcass visits and the species richness of scavenger assemblages. Contrary to our expectations, carcass species, condition (fresh/frozen), and provision schedule (continuous vs single exposure) did not influence scavenging frequency or diversity. The carcass visitation frequency increased with carcass mass and lower temperatures. The effect of large carcasses was especially pronounced for mesopredators and the Eurasian lynx (Lynx lynx). Lynx were not too influenced in its carrion acquisition by the season, but exclusively preferred remote habitats containing higher forest cover. Birds of prey, mesopredators, and top predators were also positively influenced by the visiting rate of ravens (Corvus corax), whereas no biotic or abiotic preferences were found for wild boars (Sus scrofa). This study provides evidence that any ungulate species of carrion, either in a fresh or in previously frozen condition, attracts a high diversity of scavengers especially during winter, thereby supporting earlier work that carcass provisions may support scavenger communities and endangered species.
Über 30 Jahre Rechtsprechung durch den EuGH haben die Systeme der Besteuerung am Binnenmarkt nachhaltig geprägt. Während sich die Mitgliedstaaten bei der Errichtung ihrer Steuersysteme jedoch zumeist positiv definierter Prinzipien bedienen, basiert die Rechtsprechung des Gerichtshofs in weiten Teilen auf den prohibitiv formulierten Grundverkehrsfreiheiten. In Abwesenheit einer sekundärrechtlichen Harmonisierungslösung forcierte der Gerichtshof in den vergangenen Jahren zunehmend den Integrationsprozess im direkten Steuerrecht. Insbesondere in der jüngeren Vergangenheit fühlten sich die Mitgliedstaaten hierdurch zunehmend in ihren Souveränitätsrechten verletzt. Vorliegende Abhandlung gibt eine Antwort auf die Fragestellung, ob und wie eine unionsrechtskonforme und gleichzeitig ökonomisch-systematische Ausgestaltung der europäischen Steuerrechtsordnungen durch den EuGH noch möglich ist.
Comprehensive bioinformatics identifies key microRNA players in ATG7-deficient lung fibroblasts

Background: Deficient autophagy has been recently implicated as a driver of pulmonary fibrosis, yet bioinformatics approaches to study this cellular process are lacking. Autophagy-related 5 and 7 (ATG5/ATG7) are critical elements of macro-autophagy. However, an alternative ATG5/ATG7-independent macro-autophagy pathway was recently discovered, its regulation being unknown. Using a bioinformatics proteome profiling analysis of ATG7-deficient human fibroblasts, we aimed to identify key microRNA (miR) regulators in autophagy. Method: We have generated ATG7-knockout MRC-5 fibroblasts and performed mass spectrometry to generate a large-scale proteomics dataset. We further quantified the interactions between various proteins combining bioinformatics molecular network reconstruction and functional enrichment analysis. The predicted key regulatory miRs were validated via quantitative polymerase chain reaction. Results: The functional enrichment analysis of the 26 deregulated proteins showed decreased cellular trafficking, increased mitophagy and senescence as the major overarching processes in ATG7-deficient lung fibroblasts. The 26 proteins reconstitute a protein interactome of 46 nodes and miR-regulated interactome of 834 nodes. The miR network shows three functional cluster modules around miR-16-5p, miR-17-5p and let-7a-5p related to multiple deregulated proteins. Confirming these results in a biological setting, serially passaged wild-type and autophagy-deficient fibroblasts displayed senescence-dependent expression profiles of miR-16-5p and miR-17-5p. Conclusions: We have developed a bioinformatics proteome profiling approach that successfully identifies biologically relevant miR regulators from a proteomics dataset of the ATG-7-deficient milieu in lung fibroblasts, and thus may be used to elucidate key molecular players in complex fibrotic pathological processes. The approach is not limited to a specific cell-type and disease, thus highlighting its high relevance in proteome and non-coding RNA research.
Single crystals of three at bay area tetraphenoxy‐substituted perylene bisimide dyes are grown by vacuum sublimation. X‐ray analysis reveals the self‐assembly of these highly twisted perylene bisimides (PBIs) in the solid state via imide–imide hydrogen bonding into hydrogen‐bonded PBI chains. The crystallographic insights disclose that the conformation and sterical congestion imparted by the phenoxy substituents can be controlled by ortho‐substituents. Accordingly, whilst sterically less demanding methyl and isopropyl substituents afford double‐stranded PBI chains of complementary P and M atropo‐enantiomers, single hydrogen‐bonded chains of homochiral PBIs are observed for the sterically more demanding ortho‐phenyl substituents. Investigation of the absorption and fluorescence properties of microcrystals and thin films of these PBIs allow for an unambiguous interpretation of these exciton systems. Thus, the J‐aggregates of the double‐stranded crystals exhibit a much larger (negative) exciton coupling than the single‐stranded one, which in contrast has the higher solid‐state fluorescence quantum yield.
Depressionen gehören zu den häufigsten psychischen Erkrankungen. Neben Symptomen wie Niedergeschlagenheit, Interessenlosigkeit oder Schlafstörungen sind Depressionen auch durch Defizite in kognitiven Funktionen, wie z.B. Aufmerksamkeitsprozessen oder der Wahrnehmung, und eine negativ verzerrte Informationsverarbeitung gekennzeichnet.
Aufgrund der hohen Prävalenz, der starken psychosozialen Funktionseinschränkungen durch depressive Erkrankungen und deren rezidivierenden Charakter besteht die Notwendigkeit, die therapeutischen Interventionen zur Behandlung affektiver Störungen zu verbessern, dadurch die Krankheitsphase der Patienten zu verkürzen und letztendlich auch die Kosten für das Gesundheitssystem zu reduzieren. In diesem Zusammenhang werden in den letzten Jahren verstärkt mögliche Prädiktoren und Korrelate des Therapieerfolgs untersucht. Hierfür könnten negativ verzerrte Informationsverarbeitungsprozesse und Defizite in kognitiven Funktionen objektive Marker darstellen.
Im ersten Teil dieser Arbeit wurde der Covariation Bias, der als Überschätzung des Zusammenhangs zwischen einem krankheitsrelevanten Stimulus und einer aversiven Konsequenz definiert wird, in einem Querschnittsdesign bei schwer depressiven Patienten zu Behandlungsbeginn im Vergleich zu einer Gruppe von Patienten nach einer sechswöchigen Behandlung sowie einer gesunden Kontrollgruppe untersucht. Diese kognitive Verzerrung war bei Patienten mit schwererer Symptomatik unabhängig vom Behandlungszeitpunkt stärker ausgeprägt. Zudem prädizierte der Covariation Bias zu Behandlungsbeginn das Therapieoutcome nach sechs Behandlungswochen dahingehend, dass Patienten mit einer stärkeren kognitiven Verzerrung ein schlechteres Ansprechen auf die Therapie zeigten.
Im zweiten Teil dieser Arbeit wurde das Emotional Processing Paradigma, das aus Aufgaben zur Emotionserkennung und zur Aufmerksamkeitslenkung besteht, zum ersten Mal bei schwer depressiven Patienten im intraindividuellen Verlauf der Behandlung eingesetzt und in Zusammenhang mit dem Therapieerfolg gestellt. Neben Hinweisen darauf, dass Patienten, bei denen sich in den ersten Behandlungswochen unter anderem die Salienz negativer Emotionen verringerte, mit höherer Wahrscheinlichkeit remittierten, zeigten sich vor allem zeitlich stabile Unterschiede im Sinne einer Trait-Variablen zwischen Patienten, die auf die initiale Therapie ansprachen, und Patienten, die keine bedeutsame Verbesserung erfuhren, in den globalen kognitiven Funktionen: Patienten, bei denen es zu keiner klinisch relevanten Verbesserung durch die Therapie kam, wiesen stärkere Defizite auf.
Zusammengenommen weisen die Ergebnisse der vorliegenden Arbeit auf ein stabiles Muster von Defiziten in globalen kognitiven Funktionen bei Patienten mit Depressionen hin. Diese Abweichungen liegen jedoch nicht bei allen schwer depressiven Patienten gleichermaßen vor. Bei Patienten mit Defiziten scheint das Therapieoutcome schlechter zu sein. Somit könnten diese Prozesse der Informationsverarbeitung und kognitive Defizite auf neuropsychologischer Ebene Prädiktoren und Korrelate des Therapieoutcomes darstellen. Im Sinne der personalisierten Medizin könnte in Zukunft die Diagnostik um die Parameter der Informationsverarbeitungsprozesse ergänzt werden und so die Prognose des Therapieerfolgs verbessert und die Behandlung der Patienten individualisiert werden.
Mangelernährung und Appetitlosigkeit werden trotz zunehmender Forschung noch in ihren Auswirkungen auf Patienten und Gesundheitssystem unterschätzt. Um dem entgegenzuwirken, sind zusätzliche Informationen zu Risikofaktoren, Auswirkungen und zu einem adäquaten Screening beider Symptome vonnöten.
Diese Untersuchung geht der Frage nach, ob die Erfassung der Appetitlosigkeit auch ein geeignetes Verfahren zur Einschätzung des Mangelernährungsrisikos darstellt. Zusätzlich wurden die Patienten auf Risikofaktoren und Auswirkungen von Mangelernährung und Appetitlosigkeit hin untersucht. Dazu wurden bei den Patienten unter anderem soziodemographische Daten, ein NRS-2002 Vorscreening und die per ESAS erfasste Symptomlast statistisch ausgewertet. Diese waren zuvor im Rahmen des BUKA Projekts an der Universität Würzburg ermittelt worden.
Von den 486 geeigneten Patienten konnten 477 in die Untersuchung eingeschlossen werden. Davon zeigten 65.6% ein erhöhtes Mangelernährungsrisiko und 31.2% eine manifeste Appetitlosigkeit.
Der Allgemeinzustand und die Lebensqualität beider Gruppen waren signifikant eingeschränkt. Zusätzlich wiesen sie eine deutlich erhöhte Symptombelastung im ESAS auf. Als unabhängige Risikofaktoren für Mangelernährung konnten der BMI, der Allgemeinzustand (ECOG Status), die Tumorlokalisation und der Appetitstatus eruiert werden. Bei der Appetitlosigkeit traf dies auf den ECOG Status, das Alter und das NRS Vorscreening zu.
Die Bestimmung der Appetitlosigkeit erwies sich als nicht ausreichend präzise, um auch das Mangelernährungsrisiko zu erfassen. Insgesamt wurden 61.4% der Patienten übereinstimmend getestet. Die Wahrscheinlichkeit bei erhöhtem Mangelernährungsrisiko (NRS Vorscreening positiv) auch eine manifeste Appetitlosigkeit (ESAS Appetitverlust>3) aufzuweisen betrug nur 44%. Andererseits aber zeigten Patienten mit manifester Appetitlosigkeit in 94% auch ein erhöhtes Mangelernährungsrisiko. Der Einfluss von BMI, Tumorlokalisation und Appetitverlust zeigte sich hierbei signifikant auf die Chance übereinstimmend getestet zu werden.
In der Zusammenschau betonen die Ergebnisse unserer Untersuchung die Wichtigkeit einer präzisen Erfassung von Appetitlosigkeit und Mangelernährung bei Tumorpatienten, auch in der Palliativmedizin. Besonders vor dem Hintergrund einer alternden Gesellschaft mit wahrscheinlich zunehmender Inzidenz und Prävalenz von Tumorerkrankungen gewinnen Erkennung und Therapie von Mangelernährung und Appetitlosigkeit zusätzlich an Bedeutung.
Einleitung:
TRPC-Kanäle spielen eine wichtige Rolle in der Pathologie der Herzinsuffizienz und kardialen Hypertrophie. Diese Effekte werden unter anderem über den Calcineurin-NFAT-Signalweg vermittelt. Ein wichtiger Interaktionspartner und Regulator von TRPC-Kanälen ist das Protein FKBP52. Mittels eines Yeast Two-Hybrid Systems wurde in einer kardialen cDNA library eine Interaktion zwischen einem C-terminalen Fragment von TRPC3 (AS 742-848), welches außerhalb der bekannten FKBP-Bindungsdomäne (AS 703-714) liegt, und FKBP52 beobachtet. Da dies eine weitere Bindungsstelle in FKBP52 vermuten ließ, erzeugten wir ein Fragment von FKBP52, welches FKBP52s genannt wurde und dem die funktionell relevante PPIase I-Domäne mit der bekannten Bindungsstelle fehlt. Eine erste Co-IP zwischen diesem Fragment und TRPC3 war erfolgreich.
Ziel:
Die Bestimmung, ob die Anwesenheit des verkürzten FKBP52 in vivo die Komplexbildung aus TRPC3 bzw. TRPC4 und dem Wildtyp-FKBP52 unterdrückt. Zusätzlich, ob FKBP52s die Interaktion zwischen TRPC3 bzw. TRPC4 und Calcineurin in vivo unterbricht und damit die Aktivierung des Calcineurin-NFAT-Signalweges hemmt.
Methoden:
Co-Immunopräzipitationen (Co-IP) wurden mit HEK-293-Zellen durchgeführt, die mit cDNA transfiziert wurden, welche Gene für TRPC3, TRPC4, Calcineurin A und FKBP52s enthielt. Zur Bestimmung der nukleären Translokation von NFATc1 mittels Fluoreszenzmikroskopie wurden HEK-293-Zellen mit TRPC3, TRPC4, GFP-NFATc1 ± FKBP52s transfiziert. Die statistische Analyse erfolgte mit einer One-Way ANOVA.
Ergebnisse:
In dieser Arbeit konnte gezeigt werden, dass FKBP52 sowohl mit TRPC3 als auch mit TRPC4 interagiert. Ebenso wurde festgestellt, dass FKBP52 auch ohne seine katalytische PPIase I-Domäne Bindungen mit TRPC3 bzw. TRPC4 eingeht. Dieses FKBP52-Konstrukt nimmt ebenso an der Komplexbildung mit TRPC3 bzw. TRPC4 und Calcineurin teil. Des Weiteren ließ sich für TRPC3 zeigen, dass unter Stimulation mit Carbachol (GPCR-Agonist) bei Anwesenheit dieses gekürzten FKBP52 eine signifikant geringere Aktivierung und Wanderung des Transkriptionsfaktors NFAT in den Nucleus erfolgte.
Schlussfolgerung:
FKBP52 spielt daher eine wichtige Rolle in dieser Signalkaskade, indem es entscheidend an der Aktivierung von Calcineurin und dessen Rekrutierung zum TRPC-Kanalkomplex beteiligt ist und damit auch an der Aktivierung des Calcineurin-NFAT-Signalweges.
Bei Tumoren von Kopf und Hals kann primär oder adjuvant durch Bestrahlung therapiert werden. Die Folgen dieser Behandlung können Xerostomie, Karies, Infektionen, Dysphagie oder Mundgeruch sein. Diese Nebenwirkungen vermindern die Lebensqualität des Patienten. Unterschiedliche Behandlungsansätze haben aufgrund von therapiebedingten Einschränkungen nicht den Weg in den klinischen Alltag gefunden. Eine Alternative zu den vorhandenen Behandlungsansätzen kann das Tissue Engineering sein. Das Ziel einer Normalisierung der Speichelproduktion nach Behandlung soll durch eine implantierbare, künstliche Speicheldrüse erreicht werden. Kann humanes natives Speicheldrüsengewebe der Parotis auf gradientenfreiem dreidimensional aufgebauten Polyurethan wachsen und seine Funktionalität beibehalten?
Humane Parotiszellen wurden von 20 Patienten im Alter von 42 - 90 Jahren durch Operation entnommenen und in Polystyrol-Zellkulturflaschen mit dem Nährmedium BEGM herangezüchtet. Es erfolgte eine 2D-Zellverteilung der reinen Parotiskultur. Zur Kontrolle der Vitalität zwischen den Passagen wurde eine Trypan-Blau Färbung verwendet. Als Trägermaterial der Zellen wurde eine biokompatible, abbaubare Matrix aus ε-Polycaprolacton verarbeitet. Die Übertragung der humanen Parotiszellen wurde mit einer Kleberproteinlösung, bestehend aus den Hauptbestandteilen Aprotinin, Fibrinogen und der Thrombinlösung durchgeführt. 7,14 und 21 Tage nach Aufbringung wurde der Überstand der zeitgleich entnommenen Konstrukte zur Überprüfung des α-Amylase konserviert. Zusätzlich wurden an den 3 Untersuchungstagen Konstrukte für die Anfertigung von histologischen Schnitten, quantitativer PCR, indirekter Immunfluoreszenz und zur Elektronenmikroskopie entnommen. Zur Überprüfung der Funktionalität der angezüchteten Speicheldrüsenzellen wurde das Enzym α-Amylase und das Wasserkanalprotein Aquaporin 5 herangezogen.
Bei der Kultivierung der humanen Speicheldrüsenzellen konnte durch den Vitalitätstest Trypan-Blau Färbung in Kombination mit einer Neubauerzählkammer eine konstant hohe Anzahl an vitalen Zellen bis zur 4. Passage nachgewiesen werden. Durch die Lebend/Tot Färbung auf FDA/EB Basis der Konstrukte über die Untersuchungszeit von 14 Tagen konnte keine Vermehrung von avitalen Zellen mikroskopisch festgestellt werden. Die statistische Auswertung mittels Boxplots des ELISA berechnete für den ersten Untersuchungstag einen Median auf niedrigem Niveau von 4,4 U/l und sank im weiteren Zeitverlauf am Untersuchungstag 21 auf die niedrigsten Median von 2,2 U/l ab. α-Amylase konnte an allen 3 Tagen mittels quantitativer PCR und indirekter Immunfluoreszenz belegt werden. Aquaporin 5 als Funktionsnachweis war in der vorliegenden Studie nicht signifikant durch quantitative PCR beweisbar. Die Rasterelektronenmikroskopie bildete adhärente Zellen in kugeliger Form aus den besiedelten Matrices nach 7 Tagen Kultivierung ab. Durch die Transmissionselektronenmikroskopie konnten Zellen, die Zellfortsätze ausgebildet hatten nach 14 Tagen beobachtet werden. Der Versuch, histologische Schnitte auf Grundlage der Paraffineinbettung oder Kryo-Konservierung zu erzeugen, musste frustran abgeschlossen werden.
Eine Kultivierung von Speicheldrüsenzellen auf einer Matrix aus ε-Polycaprolacton ohne Gradienten ist eingeschränkt umsetzbar. Die Studie konnte zeigen, dass das Wachstum der Zellen auf konstant niedrigem Niveau über den Untersuchungszeitraum von 21 Tagen lag. Der Funktionsnachweis von α-Amylase auf absinkendem niedrigem Niveau sowie fehlender Bestätigung von Aquaporin 5 kann als stationäre Phase des Wachstums interpretiert werden. Zur Verbesserung der Zellentwicklung sollte die besiedelte Matrix zu einem 3D-Zellwachstum anregen. Bei sequenziell entstehender Polarität der Zellen käme es zu einer Verbesserung der Vitalität sowie der vermehrten Ausbildung von α-Amylase und Aquaporin 5. Dies könnte in einer Kombination der Zellkultur aus Parotiszellen mit Kokulturen aus humanen Myoepithelzellen und Parenchymzellen erreicht werden. Sehr gute Ergebnisse des Zellwachstums und der Zellfunktion konnten aktuell in anderen Studien auf der Trägersubstanz Matrigel oder durch Rebesiedelung von dezellularisierten Organen beobachtet werden.
Die Patientenbestrahlung stellt eine wichtige Therapiesäule in der onkologischen Behandlung von Hirntumoren dar. Das Hauptaugenmerk wird dabei auf die Erreichung einer vorgegebenen Zieldosis im Tumorgebiet und die ausreichende Schonung von sensiblen Strukturen gerichtet. Wir verglichen insgesamt 4 Bestrahlungstechniken untereinander, welche in ihrer Segmentierung und Feldzahl variiert werden können: KoPlanar (Komplettbestrahlung in einer Ebene), KoPlanar+1 (Bestrahlung in einer Ebene mit einem Zusatzfeld in einer anderen Ebene), 2-Ebenen (Bestrahlung auf 2 unterschiedliche Ebenen verteilt), Quasi-Isotrop (Bestrahlung mit Zentralstrahlen in mehreren unterschiedlichen Ebenen). Die Feldzahl kann zwischen wenigen Feldern (9F oder 10F) und vielen Feldern (15F oder 16F) gewählt werden. Die Segmentanzahl wird entweder bei 64 oder 120 Segmenten festgelegt, alternativ wurde eine freie Optimierung der Feldfluenz ermöglicht.
Dabei zeigte die Quasi-Isotrope Technik eindeutige und signifikante Vorteile gegenüber allen anderen Techniken sowohl bei niedrigen als auch hohen Feldzahlen. Die koplanare Bestrahlung schnitt bei unserer Auswertung am schlechtesten ab. Die 2-Ebenen Technik und KoPlanar+1 Technik können bei hohen Feldzahlen als gleichwertig betrachtet werden, bei niedrigen Feldzahlen zeigt die KoPlanar+1 Technik Vorteile. Aus unserer Sicht sollten die unentschiedenen Vergleiche in weiteren Studien untersucht werden, die das Patientengut weiter einengen. Weiterhin wäre eine Erweiterung der Untersuchungen auf die schneller applizierbaren nonkoplanaren Volumetric Arc –Techniken (VMAT) wünschenswert.
Azole resistance of the fungal pathogen Aspergillus fumigatus is an emerging problem. To identify novel mechanisms that could mediate azole resistance in A. fumigatus, we analyzed the transcriptome of a mitochondrial fission/fusion mutant that exhibits increased azole tolerance. Approximately 12% of the annotated genes are differentially regulated in this strain. This comprises upregulation of Cyp51A, the azole target structure, upregulation of ATP-binding cassette (ABC) superfamily and major facilitator superfamily (MFS) transporters and differential regulation of transcription factors. To study their impact on azole tolerance, conditional mutants were constructed of seven ABC transporters and 17 transcription factors. Under repressed conditions, growth rates and azole susceptibility of the mutants were similar to wild type. Under induced conditions, several transcription factor mutants showed growth phenotypes. In addition, four ABC transporter mutants and seven transcription factor mutants exhibited altered azole susceptibility. However, deletion of individual identified ABC transporters and transcription factors did not affect the increased azole tolerance of the fission/fusion mutant. Our results revealed the ability of multiple ABC transporters and transcription factors to modulate the azole susceptibility of A. fumigatus and support a model where mitochondrial dysfunctions trigger a drug resistance network that mediates azole tolerance of this mold.
Ziel dieser Arbeit war es, die antiinflammatorischen Effekte von Anthocyanen und Anthocyanidinen mit besonderem Augenmerk auf Delphinidin zu vergleichen. Desweiteren sollten im statistischen Vergleich Dosierungen gefunden werden, bei welchen die größte antiinflammatorische Wirksamkeit von Anthocyanen und Anthocyanidinen zu erwarten wäre und ein eventueller Unterschied in der Wirkung der verschiedenen Stoffe berechnet werden.
Die Literaturrecherche mittels der Datenbanken „Pubmed“ und „The Cochrane Library“ lieferte 24 Studien, die die Einschlusskriterien erfüllten. Für Delphinidin allein war die Datenlage zu diesem Zeitpunkt noch zu dürftig, weswegen die Suche auf die Begriffe „Anthocyanins“ in Kombination mit „Antiinflammatory Agents“ ausgedehnt wurde.
Die statistische Auswertung der Daten erfolgte unter der Fragestellung: „ Haben die Variablen Wirkstoff, Wirkdauer, Menge an Wirkstoff und Art (m-RNA oder Protein“ jeweils unter Kontrolle der anderen Variablen einen Einfluss auf die Reduktion von inflammatorischen Markern?“ Die wichtigsten Ergebnisse, die unter dieser Fragestellung errechnet werden konnten, lauten: 1. PDG zeigt eine bessere antiinflammatorische Wirksamkeit als DP und C3G, 2. Luteolin zeigt eine bessere antiinflammatorische Wirksamkeit als Cyanidin.
Gründe für eine höhere antiinflammatorische Wirksamkeit von PDG im Vergleich zu DP und C3G sind derzeit noch offen. Zu diskutieren ist unter anderem, ob DP und C3G biochemisch instabilere Komplexe sind und schneller zerfallen. Grund für eine höhere antiinflammatorische Wirksamkeit von Luteolin im Vergleich zu Cyanidin könnte eine Doppelbindung zwischen C2 und C3 sein, die bei Luteolin, nicht aber bei Cyanidin zu finden ist. In der Zusammenschau dieser Ergebnisse ist festzustellen, dass weitere Studien für eine möglichst genaue statistische Analyse nötig sind, vor allem in Bezug auf die antiinflammatorischen Effekte von Delphinidin.
The CXCL12-CXCR4 axis plays a vital role in many steps of breast cancer metastasis, but the molecular mechanisms have not been fully elucidated. We previously reported that activation of CXCR4 by CXCL12 promotes the nuclear localization of LASP1 (LIM and SH3 protein 1). The nuclear LASP1 then interacts with Snail1 in triple-negative breast cancer (TNBC) cell lines. In this study, we report that the nuclear accumulation and retention of Snail1 was dependent on an increase in nuclear LASP1 levels driven by active CXCR4. The CXCR4-LASP1 axis may directly regulate the stabilization of nuclear Snail1, by upregulating nuclear levels of pS473-Akt, pS9-GSK-3β, A20, and LSD1. Furthermore, the activation of CXCR4 induced association of LASP1 with Snail1, A20, GSK-3β, and LSD1 endogenously. Thus, nuclear LASP1 may also regulate protein-protein interactions that facilitate the stability of Snail1. Genetic ablation of LASP1 resulted in the mislocalization of nuclear Snail1, loss of the ability of TNBC cells to invade Matrigel and a dysregulated expression of both epithelial and mesenchymal markers, including an increased expression of ALDH1A1, a marker for epithelial breast cancer stem-like cells. Our findings reveal a novel role for the CXCR4-LASP1 axis in facilitating the stability of nuclear localized Snail1.
The Concealed Information Test (CIT) enables the detection of certain (e.g., crime-relevant or personal) information, even if participants aim to conceal their knowledge. The current preregistered study investigated whether previously observed impairing effects of alcohol intoxication on participants' performance in a reaction time CIT (RT CIT) field study also translate to a laboratory environment. In contrast to the previous study of Suchotzki and Gamer (Sci Rep 8:7825, 2018) in which alcohol consumption was voluntary and self-administered, the current study used a randomized assignment of participants to either an alcohol group (n=88; receiving a drink with 3 cl alcohol) or a sober control group (n=89; receiving a drink with just some alcohol drops to hide group assignment). After drink administration, participants completed an RT CIT, in which they were instructed to hide knowledge of their own identity. Blood alcohol concentration (BAC) was estimated via breath alcohol ratio. In contrast to the previous field study, results revealed no differences in CIT-performance between intoxicated and sober participants. Aside from questioning the robustness of the result of the previous field study, our results also point to a number of interesting theoretical explanations for the discrepancy between both results, which are elaborated in the discussion.
The bioprinting roadmap

This bioprinting roadmap features salient advances in selected applications of the technique and highlights the status of current developments and challenges, as well as envisioned advances in science and technology, to address the challenges to the young and evolving technique. The topics covered in this roadmap encompass the broad spectrum of bioprinting; from cell expansion and novel bioink development to cell/stem cell printing, from organoid-based tissue organization to bioprinting of human-scale tissue structures, and from building cell/tissue/organ-on-a-chip to biomanufacturing of multicellular engineered living systems. The emerging application of printing-in-space and an overview of bioprinting technologies are also included in this roadmap. Due to the rapid pace of methodological advancements in bioprinting techniques and wide-ranging applications, the direction in which the field should advance is not immediately clear. This bioprinting roadmap addresses this unmet need by providing a comprehensive summary and recommendations useful to experienced researchers and newcomers to the field.
Spinal muscular atrophy with respiratory distress type 1 (SMARD1) is a fatal monogenic motoneuron disease in children with unknown etiology caused by mutations in the immunoglobulin μ-binding protein 2 (IGHMBP2) gene coding for DNA/RNA ATPase/helicase. Despite detailed knowledge of the underlying genetic changes, the cellular mechanisms leading to this disease are not well understood. In the Nmd2J ("neuromuscular disorder") mouse, the mouse model for the juvenile form of SMARD1 patients, in which similar pathological features as diaphragmatic paralysis and skeletal muscle atrophy are observed. Ex vivo studies in Nmd2J mice showed that loss of the motor axon precedes atrophy of the gastrocnemius muscle and does not correlate with neurotransmission defects in the motor endplate. The already described independent myogenic anomalies in the diaphragm and heart of the Nmd2J mouse raised the question whether spinal motoneuron degeneration develops cell autonomously. Ighmbp2 is predominantly localized in the cytoplasm and seems to bind to ribosomes and polysomes, suggesting a role in mRNA metabolism.
In this Ph.D. thesis, morphological and functional analyses of isolated Ighmbp2-deficient (Ighmbp2-def.) motoneurons were performed to answer the question whether the SMARD1 phenotype results from dysregulation of protein biosynthesis. Ighmbp2-deficient motoneurons show only negligible morphological alterations with respect to a slight increase in axonal branches. This observation is consistent with only minor changes of transcriptome based on RNA sequencing data from Ighmbp2-deficient motoneurons. Only the mRNA of fibroblast growth factor receptor 1 (Fgfr1) showed significant up-regulation in Ighmbp2-deficient motoneurons. Furthermore, no global aberrations at the translational level could be detected using pulsed SILAC (Stable Isotope Labeling by Amino acids in cell culture), AHA (L-azidohomoalanine) labeling and SUnSET (SUrface SEnsing of Translation) methods. However, a reduced β-actin protein level was observed at the growth cones of Ighmbp2-deficient motoneurons, which was accompanied with a reduced level of Imp1 protein, a known β-actin mRNA interactor. Live-cell imaging studies using fluorescence recovery after photobleaching (FRAP) showed translational down-regulation of eGFPmyr-β-actin 3'UTR mRNA in the growth cones and the cell bodies, although the amount of β-actin mRNA and the total protein amount in Ighmbp2-deficient motoneurons showed no aberrations. This compartment-specific reduction of β-actin protein occurred independently of a non-existent direct IGHMBPF2 binding to β-actin mRNA. Fgfr1, which was upregulated on the RNA level, did not show an increased protein amount in Ighmbp2-deficient motoneurons, whereas a reduced amount could be detected. Interestingly, a correlation could be found between the reduced amount of the Imp1 protein and the increased Fgfr1 mRNA, since the IMP1 protein binds the FGFR1 mRNA and thus could influence the transport and translation of FGFR1 mRNA. In summary, all data suggest that Ighmbp2 deficiency leads to a local but modest disturbance of protein biosynthesis, which might contribute to the motoneuron defects of SMARD1.
According to theories on moral balancing, a prosocial act can decrease people’s motivation to engage in subsequent prosocial behavior, because people feel that they have already achieved a positive moral self-perception. However, there is also empirical evidence showing that people actually need to be recognized by others in order to establish and affirm their self-perception through their prosocial actions. Without social recognition, moral balancing could possibly fail. In this paper, we investigate in two laboratory experiments how social recognition of prosocial behavior influences subsequent moral striving. Building on self-completion theory, we hypothesize that social recognition of prosocial behavior (self-serving behavior) weakens (strengthens) subsequent moral striving. In Study 1, we show that a prosocial act leads to less subsequent helpfulness when it was socially recognized as compared to a situation without social recognition. Conversely, when a self-serving act is socially recognized, it encourages subsequent helpfulness. In Study 2, we replicate the effect of social recognition on moral striving in a more elaborated experimental setting and with a larger participant sample. We again find that a socially recognized prosocial act leads to less subsequent helpfulness compared to an unrecognized prosocial act. Our results shed new light on the boundary conditions of moral balancing effects and underscore the view that these effects can be conceptualized as a dynamic of self-completion.
Die Juvenile idiopathische Arthritis (JIA) ist ein chronisch entzündliches Krankheitsbild des Kindesalters mit ungeklärter Ursache. Eine ätiopathogenetische Beteiligung des Immunsystems und im speziellen der T-Zellen ist wahrscheinlich.
Die vorliegende durchflusszytometrische Untersuchung umfasst die Daten von 84 JIA Patienten und 40 altersgleichen Kontrollprobanden und vergleicht die Ergebnisse im transversalen sowie longitudinalen Studiendesign.
Die Phänotypisierung der Lymphozyten zeigte im Vergleich zu gesunden Kontrollprobanden eine abweichende Verteilung der T-Helferzell-Subpopulationen. Die CD4 positiven T-Helferzellen der JIA Patienten setzen sich aus einer Anzahl proportional verminderter differenzierter Zellen und vermehrter naiver Zellen zusammen. Im Zytokinprofil wurde eine reduzierte Th1- sowie Th2-Antwort beobachtet. In allen T-Zell-Subpopulationen der JIA Patienten fand sich RORуt, der Transkiptionsfaktor der Th17-Differenzierung, hochreguliert. Im klinischen Schub der Arthritis zeigte sich eine signifikant erhöhte RORуt-Expression sowie eine vermehrte IL-17 Produktion. Die IL-17 produzierenden Zellen zeigten u.a. einen regulatorischen Phänotyp.
Bei den Stimulationsversuchen von CCR6+ Zellen, die wir als IL-17 produzierende Zellen definieren konnten, wurde eine ausgeprägte Plastizität beobachtet. Mit spezifischen Zytokinstimuli wurden die IL-17 produzierenden Zellen vermehrt zu Th17, Th1 oder regulatorischen T-Zellen polarisiert. Die Polarisierbarkeit unterschied sich zwischen JIA Patienten und Kontrollprobanden. Die CCR6+ Zellen der JIA Patienten zeigten im Vergleich zu den Kontrollprobanden ausgeprägte Polarisierungstendenzen zum Th17-Phänotyp und ließen sich nur gering in Th1 oder Treg konvertieren.
Die Ergebnisse unserer Untersuchung lassen auf eine Dominanz im Th17-Differenzierungsweg schließen. Der Transkriptionsfaktor RORуt scheint vor allem im akuten Krankheitsschub zu prädominieren und die Th17-Differenzierung zu steuern. Eine Gegenregulation von Th1- und Th2-Zellen fehlt.
Zusammenfassend ergeben sich Hinweise darauf, dass Th17-Zellen eine bedeutende Rolle in der Pathogenese der Juvenilen idiopathischen Arthritis spielen. In wieweit man IL-17, den CCR6-Rezeptor oder den Transkriptionsfaktor RORуt als Angriffsziel von krankheitsmodifizierenden Therapeutika nutzen kann, muss in weiteren Studien untersucht werden. Ebenso gilt es herauszufinden, ob die forcierte Polarisierung in Richtung regulatorische T-Zellen oder gegenregulatorisch aktive Th1 und Th2 Zellen eine Therapiestrategie darstellt.
Die zervikale spondylotische Myelopathie kann zu schweren neurologischen Funktionsstörungen führen. Vor allem durch die Störung der Feinmotorik und des Gangbilds kann es zu einer Beeinträchtigung im alltäglichen Leben kommen. Eine operative Dekompression des Rückenmarks kann ein Fortschreiten der neurologischen Symptome verhindern oder abmildern.
Ziel dieser Studie war die Erfassung unterschiedlicher neurologischer Dysfunktionen bei zervikaler Myelopathie präoperativ sowie die Evaluierung der funktionellen Veränderung nach operativer Dekompression. Hierbei wurden verschiedene - objektive und subjektive - Messmethoden angewandt. Die Erfassung der feinmotorischen Dysfunktion lag dabei im Vordergrund, da diese in der Literatur bis jetzt nur unzureichend untersucht und beschrieben wurden. Daneben wurde die Regeneration der kernspintomographisch gemessenen Schädigung des Myelons nach operativer Dekompression untersucht.
In dieser Studie wurde ein digitales Graphiktablett zur quantitativen und objektiven Erfassung der feinmotorischen Dysfunktion bei zervikaler Myelopathie benutzt. Eine Beeinträchtigung der Feinmotorik wurde vor allem bei komplexen Schreibaufgaben festgestellt, welche am ehesten Schreibaufgaben im alltäglichen Leben entsprechen. Im Einklang mit früheren Studien wurde auch in dieser Studie eine Störung des Gangbilds bei Patienten mit zervikaler Myelopathie festgestellt.
Die Dauer der Symptome bis zur Operation konnte als einziger signifikanter Einflussfaktor für den Schweregrad der feinmotorischen Dysfunktion ausgemacht werden. Das Geschlecht, der BMI, der Beruf und Sport hatten in dieser Analyse weder einen Einfluss auf den klinischen noch auf den neuroradiologischen Schweregrad der zervikalen Myelopathie.
Nach operativer Dekompression verbesserten sich die feinmotorische Funktion sowie das Gangbild der Patienten. Auch die subjektiven Scores zur Erfassung der neurologischen Funktion (European Myelopathy Score, Nurick-Score) sowie die Schmerzen (Numerische Rating-Skala) verbesserten sich postoperativ. Zudem nahm die Länge des pathologischen Signals im MRT ab.
Faktoren, die einen Trend zur Korrelation mit den postoperativen Veränderungen der Funktion zeigten, waren das Alter der Patienten und die Symptomdauer. In dieser Studie konnte kein Einfluss des BMIs, der Anzahl der operierten Höhen oder der Art des Operationszugangs festgestellt werden.
Obwohl die Anzahl der untersuchten Patienten begrenzt war, gestaltete sich die Gruppe heterogen (Alter, Geschlecht, BMI, Dauer der Symptome). So konnte ein möglichst breites Spektrum an Patienten mit zervikaler Myelopathie untersucht werden.
Die wichtigste Limitation dieser Studie war die hohe drop-out Rate in den postoperativen Verlaufsuntersuchungen. Daher konnten die Ergebnisse der Längsschnittanalyse nur an einer kleinen Patientengruppe ermittelt werden und bei der Untersuchung von Einflussfaktoren für das postoperative Ergebnis nur Tendenzen erkannt werden. Ob diese Faktoren einen prognostischen Wert für das postoperative Ergebnis haben, müsste in weiteren Studien an einem größeren Kollektiv untersucht werden.
In this dissertation, we develop and analyze novel optimizing feedback laws for control-affine systems with real-valued state-dependent output (or objective) functions. Given a control-affine system, our goal is to derive an output-feedback law that asymptotically stabilizes the closed-loop system around states at which the output function attains a minimum value. The control strategy has to be designed in such a way that an implementation only requires real-time measurements of the output value. Additional information, like the current system state or the gradient vector of the output function, is not assumed to be known. A method that meets all these criteria is called an extremum seeking control law. We follow a recently established approach to extremum seeking control, which is based on approximations of Lie brackets. For this purpose, the measured output is modulated by suitable highly oscillatory signals and is then fed back into the system. Averaging techniques for control-affine systems with highly oscillatory inputs reveal that the closed-loop system is driven, at least approximately, into the directions of certain Lie brackets. A suitable design of the control law ensures that these Lie brackets point into descent directions of the output function. Under suitable assumptions, this method leads to the effect that minima of the output function are practically uniformly asymptotically stable for the closed-loop system. The present document extends and improves this approach in various ways.
One of the novelties is a control strategy that does not only lead to practical asymptotic stability, but in fact to asymptotic and even exponential stability. In this context, we focus on the application of distance-based formation control in autonomous multi-agent system in which only distance measurements are available. This means that the target formations as well as the sensed variables are determined by distances. We propose a fully distributed control law, which only involves distance measurements for each individual agent to stabilize a desired formation shape, while a storage of measured data is not required. The approach is applicable to point agents in the Euclidean space of arbitrary (but finite) dimension. Under the assumption of infinitesimal rigidity of the target formations, we show that the proposed control law induces local uniform asymptotic (and even exponential) stability. A similar statement is also derived for nonholonomic unicycle agents with all-to-all communication. We also show how the findings can be used to solve extremum seeking control problems.
Another contribution is an extremum seeking control law with an adaptive dither signal. We present an output-feedback law that steers a fully actuated control-affine system with general drift vector field to a minimum of the output function. A key novelty of the approach is an adaptive choice of the frequency parameter. In this way, the task of determining a sufficiently large frequency parameter becomes obsolete. The adaptive choice of the frequency parameter also prevents finite escape times in the presence of a drift. The proposed control law does not only lead to convergence into a neighborhood of a minimum, but leads to exact convergence. For the case of an output function with a global minimum and no other critical point, we prove global convergence.
Finally, we present an extremum seeking control law for a class of nonholonomic systems. A detailed averaging analysis reveals that the closed-loop system is driven approximately into descent directions of the output function along Lie brackets of the control vector fields. Those descent directions also originate from an approximation of suitably chosen Lie brackets. This requires a two-fold approximation of Lie brackets on different time scales. The proposed method can lead to practical asymptotic stability even if the control vector fields do not span the entire tangent space. It suffices instead that the tangent space is spanned by the elements in the Lie algebra generated by the control vector fields. This novel feature extends extremum seeking by Lie bracket approximations from the class of fully actuated systems to a larger class of nonholonomic systems.
Eine retrospektive Studie anhand von Patientenunterlagen zum Blutverlust, Transfusionshäufigkeit und postoperativen Komplikationen bei Knie-Endo-Prothesen mit systemischer Gabe von 500 mg Tranexamsäure kurz vor Eröffnung der Blutleere! 229 Patienten in der Studiengruppe, von denen 9 eine erlaubte Wiederholungsdosis erhalten hatten, wurden verglichen mit 231 Patienten, die keine Tranexamsäure erhalten hatten. 3 Patienten der Studiengruppe gegenüber 14 Patienten der Kontrollgruppe benötigten eine Bluttransfusion. In der Studiengruppe war der niedrigste postoperative Hb bei 71,2% der Patienten 10 g/dl oder höher. In der Kontrollgruppe war es nur bei 49,4% der Patienten der Fall. Durch die Gabe von Tranexamsäure konnte der durchschnittliche, berechnete Blutverlust bei Männern von etwa 1850 ml auf 1450 ml und bei Frauen von etwa 1300 ml auf etwa 1000 ml gesenkt werden. Die Unterschiede waren signifikant! Im beobachteten Kollektiv konnte keine Steigerung der Rate an Thrombosen oder thrombembolischen Ereignissen festgestellt werden. Das Antifibrinolytikum Tranexamsäure ist in dieser niedrigen Dosierung effektiv und scheint sicher zu sein.
Depending on the connectivity of solubilizing oligoethylene glycol (OEG) side chains to the π‐cores of amphiphilic naphthalene and perylene bisimide dyes, self‐assembly in water occurs either upon heating or cooling. Herein, we show that this effect originates from differences in the enwrapping capability of the π‐cores by the OEG chains. Rylene bisimides bearing phenyl substituents with three OEG chains attached directly to the hydrophobic π‐cores are strongly sequestered by the OEG chains. These molecules self‐assemble at elevated temperatures in an entropy‐driven process according to temperature‐ and concentration‐dependent UV/Vis spectroscopy and calorimetric dilution studies. In contrast, for rylene bisimides in which phenyl substituents with three OEG chains are attached via a methylene spacer, leading to much weaker sequestration, self‐assembly originates upon cooling in an enthalpy‐driven process. Our explanation for this controversial behavior is that the aggregation in the latter case is dictated by the release of “high energy water” from the hydrophobic π‐surfaces as well as dispersion interactions between the π‐scaffolds which drive the self‐assembly in an enthalpically driven process. In contrast, for the former case we suggest that in addition to the conventional explanation of a dehydration of hydrogen‐bonded water molecules from OEG units it is in particular the increase in conformational entropy of back‐folded OEG side chains upon aggregation that provides the pronounced gain in entropy that drives the aggregation process. Thus, our studies revealed that a subtle change in the attachment of solubilizing substituents can switch the thermodynamic signature for the self‐assembly of amphiphilic dyes in water from enthalpy‐ to entropy‐driven.
Here, we assessed whether 36 single nucleotide polymorphisms (SNPs) within the TNFSF4 and MAPKAPK2 loci influence the risk of developing invasive aspergillosis (IA). We conducted a two-stage case control study including 911 high-risk patients diagnosed with hematological malignancies that were ascertained through the aspBIOmics consortium. The meta-analysis of the discovery and replication populations revealed that carriers of the TNFSF4\(_{rs7526628T/T}\) genotype had a significantly increased risk of developing IA (p = 0.00022). We also found that carriers of the TNFSF4\(_{rs7526628T}\) allele showed decreased serum levels of TNFSF14 protein (p = 0.0027), and that their macrophages had a decreased fungicidal activity (p = 0.048). In addition, we observed that each copy of the MAPKAPK2\(_{rs12137965G}\) allele increased the risk of IA by 60% (p = 0.0017), whereas each copy of the MAPKAPK2\(_{rs17013271T}\) allele was estimated to decrease the risk of developing the disease (p = 0.0029). Mechanistically, we found that carriers of the risk MAPKAPK2\(_{rs12137965G}\) allele showed increased numbers of CD38+IgM-IgD- plasmablasts in blood (p = 0.00086), whereas those harboring two copies of the allele had decreased serum concentrations of thymic stromal lymphopoietin (p = 0.00097). Finally, we also found that carriers of the protective MAPKAPK2\(_{rs17013271T}\) allele had decreased numbers of CD27-IgM-IgD- B cells (p = 0.00087) and significantly lower numbers of CD14+ and CD14+CD16- cells (p = 0.00018 and 0.00023). Altogether, these results suggest a role of the TNFSF4 and MAPKAPK2 genes in determining IA risk.

Die Behandlung der distalen Radiusfraktur machte mit der Zeit eine zunehmende
Entwicklung, sodass heute eine Vielzahl von verschiedenen Behandlungsverfahren beschrieben wurden. Dabei befasst sich die vorliegende retrospektive Studie vor allem mit der objektiven, subjektiven, funktionellen und radiologischen Bewertung des Behandlungsergebnisses beim Einsatz der palmaren winkelstabilen Aptus 2,5 mm Rahmenplatte der Firma Medartis.
Patienten und Methoden:
Es wurden 87 Patienten mit 88 distalen Radiusfrakturen im untersuchten Zeitraum vom 01.01.2009 bis 31.12.2011 mit der palmaren winkelstabilen Aptus 2,5mm Rahmenplatte der Firma Medartis in die Studie aufgenommen. Alle Frakturen wurden nach AO klassifiziert. Während der Nachuntersuchung wurde die Patientenanamnese, der Unfallhergang, das funktionelle Outcome im Sinne der Kraft und der Bewegung am Handgelenk und die radiologischen Parameter erhoben. Eine Einordnung in den Gartland und Werley Score, eine Erhebung des subjektiven Wohlempfindens mittels DASH-Score sowie die Einteilung in die Soong Klassifikation erfolgte ebenfalls.
Ergebnis:
Die durchschnittliche Follow Up Zeit lag bei 57±10,9 Monaten. Das Durchschnittsalter der Patienten lag bei 58±17,2 Jahren. Es wurden zehn A2, 21 A3, eine B1, zwei B2, eine B3, sieben C1, 18 C2 und 28 C3 Frakturen erfasst. Das Ergebnis der radiologischen Untersuchung zeigt, dass alle präoperativ gemessenen Parameter höchstsignifikante Unterschiede zu postoperativen Verhältnissen aufwiesen. Nach radiologischer Sicht zeigen 68% des Patientenkollektivs ein einwandfreies Ergebnis mit einem Radiusbasiswinkel von 23°, Dorso-Palmaren Kippwinkel von 3°, einer Ulnar Varianz von +1 mm und einer Radiushöhe von 11 mm. Die Patienten erreichten 98% des Bewegungsausmaß verglichen zu ihrer gesunden Gegenseite. Zusammenfassend konnte bei 79% des Patientenkollektivs ein exzellentes bis gutes Kraftergebnis zur intakten Gegenseite ermittelt werden. Mit einem durchschnittlichen Punktewert von 10 erreichten die Patienten ein exzellentes bis gutes Ergebnis im DASH Score. Auch beim Gartland und Werley Score zeigten sich mit einem durchschnittlichen Punktewert von 1, 85% mit einem exzellenten und 15% mit einem guten Ergebnis. Insgesamt wurden 43 Platten in Klasse 0, 37 Implantate in Klasse 1 und lediglich 2 Platten in Klasse 2 nach Soong klassifiziert. Insgesamt ließ sich bei einem Patienten eine frühe Infektion erkennen, welches sich mit oralen Antibiotika gut behandeln ließ. Drei weitere Patienten wiesen eine intraartikuläre Schraubenlage auf.
Schlussfolgerung:
Bei der Aptus 2,5 mm palmaren winkelstabilen Rahmenplatte von MedArtis handelt es sich um ein sicheres Verfahren in der Behandlung distaler Radiusfrakturen mit sehr guten objektiven, subjektiven, radiologischen und funktionellen Ergebnissen. Die palmare Rahmenplatte kann somit als Standardimplantat empfohlen werden.
Autonomous cars and artificial intelligence that beats humans in Jeopardy or Go are glamorous examples of the so-called Second Machine Age that involves the automation of cognitive tasks [Brynjolfsson and McAfee, 2014]. However, the larger impact in terms of increasing the efficiency of industry and the productivity of society might come from computers that improve or take over business decisions by using large amounts of available data. This impact may even exceed that of the First Machine Age, the industrial revolution that started with James Watt’s invention of an efficient steam engine in the late eighteenth century. Indeed, the prevalent phrase that calls data “the new oil” indicates the growing awareness of data’s importance. However, many companies, especially those in the manufacturing and traditional service industries, still struggle to increase productivity using the vast amounts of
data [for Economic Co-operation and Development, 2018].
One reason for this struggle is that companies stick with a traditional way of using data for decision support in operations management that is not well suited to automated decision-making. In traditional inventory and capacity management, some data – typically just historical demand data – is used to estimate a model that makes predictions about uncertain planning parameters, such as customer demand. The planner then has two tasks: to adjust the prediction with respect to additional information that was not part of the data but still might influence demand and to take the remaining uncertainty into account and determine a safety buffer based on the underage and overage costs. In the best case, the planner determines the safety buffer based on an optimization model that takes the costs and the distribution of historical forecast errors into account; however, these decisions are usually based on a planner’s experience and intuition, rather than on solid data analysis.
This two-step approach is referred to as separated estimation and optimization (SEO). With SEO, using more data and better models for making the predictions would improve only the first step, which would still improve decisions but would not automize (and, hence, revolutionize) decision-making. Using SEO is like using a stronger horse to pull the plow: one still has to walk behind.
The real potential for increasing productivity lies in moving from predictive to prescriptive approaches, that is, from the two-step SEO approach, which uses predictive models in the estimation step, to a prescriptive approach, which integrates the optimization problem with the estimation of a model that then provides a direct functional relationship between the data and the decision. Following Akcay et al. [2011], we refer to this integrated approach as joint estimation-optimization (JEO). JEO approaches prescribe decisions, so they can automate the decision-making process. Just as the steam engine replaced manual work, JEO approaches replace cognitive work.
The overarching objective of this dissertation is to analyze, develop, and evaluate new ways for how data can be used in making planning decisions in operations management to unlock the potential for increasing productivity. In doing so, the thesis comprises five self-contained research articles that forge the bridge from predictive to prescriptive approaches. While the first article focuses on how sensitive data like condition data from machinery can be used to make predictions of spare-parts demand, the remaining articles introduce, analyze, and discuss prescriptive approaches to inventory and capacity management.
All five articles consider approach that use machine learning and data in innovative ways to improve current approaches to solving inventory or capacity management problems. The articles show that, by moving from predictive to prescriptive approaches, we can improve data-driven operations management in two ways: by making decisions more accurate and by automating decision-making. Thus, this dissertation provides examples of how digitization and the Second Machine Age can change decision-making in companies to increase efficiency and productivity.
Die kardialen Hormone Atriales (ANP) und B-Typ (BNP) Natriuretisches Peptid üben bekannte renale und kardiovaskuläre Effekte aus, welche durch ihren gemeinsamen, cGMP-bildenden Guanylatzyklase-Rezeptor A (GC-A) vermittelt werden. Diese Effekte sind entscheidend an der physiologischen Aufrechterhaltung des arteriellen Blutdrucks sowie des intravaskulären Blutvolumens beteiligt. Darüber hinaus zeigen aktuelle Studien, dass NPs die Mobilisierung von Fettsäuren aus dem Fettgewebe und deren Oxidation durch die Skelettmuskulatur steigern sowie die Thermogenese in braunem und weißem Fettgewebe aktivieren können. Dadurch können NPs den Energieverbrauch erhöhen und die Insulinsensitivität verbessern. Desweiteren ist Übergewicht mit einer gestörten NP/GC-A/cGMP-Signalübertragung verbunden, die möglicherweise zur Entwicklung von Diabetes Typ 2 und dessen kardio-metabolischen Folgeerkrankungen beiträgt. In vitro stimuliert synthetisches ANP über GC-A die Glukose-stimulierte Insulinsekretion aus kultivierten pankreatischen Inseln und die β-Zellproliferation. Die Bedeutung für die systemische Insulin/Glukosehomöostase in vivo ist jedoch unklar. Um zu untersuchen, ob die endogenen Herzhormone die sekretorische Funktion und/oder die Proliferation von β-Zellen unter (patho)physiologischen Bedingungen in vivo modulieren, haben wir ein neues genetisches Mausmodell mit selektiver Deletion des GC-A-Rezeptors in β-Zellen (ß GC-A KO) generiert.
In kultivierten Inseln von β GC-A KO-Mäusen waren die insulinotropen und proliferativen Effekte von ANP aufgehoben. Übereinstimmend damit führte die Infusion von BNP bei Kontroll-Tieren in vivo zu leicht erhöhten basalen Plasma-Insulinspiegeln und verbesserter Glukose-induzierter Insulinsekretion. Dieser Effekt von exogenem BNP konnte bei β GC-A KO-Mäusen nicht beobachtet werden, was die effiziente Deletion des GC-A-Rezeptors in β-Zellen bestätigt.
Interessanterweise hatte die Ablation des GC-A-Rezeptors auf ß-Zellen unter basalen Bedingungen keinen Einfluss auf physiologische und metabolische Parameter in vivo. Sowohl männliche als auch weibliche ß GC-A KO-Tiere zeigten keine Unterschiede in der basalen Insulin- und Glukosehomöostase, da sie ähnliche Nüchtern-Blutzucker- und Insulinspiegel (nach Fasten über Nacht) aufwiesen wie die Kontroll-Mäuse. Allerdings zeigten die mit HFD gefütterten β GC-A KO-Tiere frühzeitiger Glukose-Intoleranz sowie eine verminderte adaptive β-Zellproliferation. Abgesehen davon war das konsistenteste Ergebnis der in vivo-Studien der geschlechtsabhängige Unterschied in der Auswirkung der ß-Zellspezifischen GC-A-Deletion auf die Glukose-stimulierte Insulinsekretion. Weibliche, aber nicht männliche ß GC-A KO-Mäuse zeigten erhöhte Nüchtern-Insulinspiegel und eine signifikant erhöhte Glukose-stimulierte Insulinsekretion, was zu einer deutlich verbesserten Glukosetoleranz führte. Der postulierte und untersuchte Mechanismus beinhaltet eine Interaktion von Östrogenen und NPs, welche die Expression des mitochondrialen Uncoupling Protein 2 beeinflussen.
Diese Arbeit erweitert das derzeitige Wissen über die metabolischen Effekte des NP/GC-A-Systems. Insbesondere zeigen die Ergebnisse, dass Natriuretische Peptide zu einer gesteigerten ß-Zellfunktion und Vitalität in frühen Stadien eines erhöhten Insulinbedarfs, d.h. bei Diabetes Typ 2, beitragen. Da die Studien eine wesentliche Rolle dieser kardialen Hormone im endokrinen Pankreas aufdecken, ist es umso wichtiger die pleiotropen Eigenschaften von NPs und ihre möglichen therapeutischen Anwendungen bei kardio-metabolischen Erkrankungen weiter zu untersuchen.
Investigations concerning the reactivity of Ni(0) complexes [Ni(NHC)\(_{2}\)] of NHCs (N‐heterocyclic carbene) of different steric demand, Mes\(_{2}\)Im (= 1,3‐dimesitylimidazoline‐2‐ylidene) and iPr\(_{2}\)Im (= 1,3‐diisopropyl‐imidazoline‐2‐ylidene), with olefins, ketones and aldehydes are reported. The reaction of [Ni(Mes\(_{2}\)Im)\(_{2}\)] 1 with ethylene or methyl acrylate afforded the complexes [Ni(Mes\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐C\(_{2}\)H\(_{4}\))] 3 and [Ni(Mes\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐(C,C)‐H\(_{2}\)C=CHCOOMe)] 4, as it was previously reported for [Ni\(_{2}\)(iPr\(_{2}\)Im)\(_{4}\)(µ‐(η\(^{2}\):η\(^{2}\))‐COD)] 2 as a source for [Ni(iPr\(_{2}\)Im)\(_{2}\)]. In contrast to 2, complex 1 does not react with sterically more demanding olefins such as tetramethylethylene, 1,1‐diphenylethylene and cyclohexene. The reaction of [Ni(NHC)\(_{2}\)] with more π‐acidic ketones or aldehydes led to formation of complexes with side‐on η\(^{2}\)‐(C,O)‐coordinating ligands: [Ni(iPr\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐O=CH\(^{t}\)Bu)] 5, [Ni(iPr\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐O=CHPh)] 6, [Ni(iPr\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐O=CMePh)] 7, [Ni(iPr\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐O=CPh\(_{2}\))] 8, [Ni(iPr\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐O=C(4‐F‐C\(_{6}\)H\(_{4}\))\(_{2}\))] 9, [Ni(iPr\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐O=C(OMe)(CF\(_{3}\)))] 10 and [Ni(Mes\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐O=CHPh)] 11, [Ni(Mes\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐O=CH(CH(CH\(_{3}\))\(_{2}\)))] 12, [Ni(Mes\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐O=CH(4‐NMe\(_{2}\)‐C\(_{6}\)H\(_{4}\)))] 13, [Ni(Mes\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐O=CH(4‐OMe‐C\(_{6}\)H\(_{4}\)))] 14, [Ni(Mes\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐O=CPh\(_{2}\))] 15 and [Ni(Mes\(_{2}\)Im)\(_{2}\)(η\(^{2}\)‐O=C(4‐F‐C\(_{6}\)H\(_{4}\))\(_{2}\))] 16. The reaction of 1 and 2 with these simple aldehydes and ketones does not lead to a significantly different outcome, but NHC ligand rotation is hindered for the Mes\(_{2}\)Im complexes 3, 4 and 11–16 according to NMR spectroscopy. The solid‐state structures of 3, 4, 11 and 12 reveal significantly larger C\(_{NHC}\)‐Ni‐C\(_{NHC}\) angles in the Mes\(_{2}\)Im complexes compared to the iPr\(_{2}\)Im complexes. As electron transfer in d\(^{8}\)‐ (or d\(^{10}\)‐) ML\(_{2}\) complexes to π‐acidic ligands depends on the L–M–L bite angle, the different NHCs lead thus to a different degree of electron transfer and activation of the olefin, aldehyde or ketone ligand, i.e., [Ni(iPr\(_{2}\)Im)\(_{2}\)] is the better donor to these π‐acidic ligands. Furthermore, we identified two different side products from the reaction of 1 with benzaldehyde, trans‐[Ni(Mes\(_{2}\)Im)\(_{2}\)H(OOCPh)] 17 and [Ni\(_{2}\)(Mes\(_{2}\)Im)\(_{2}\)(µ\(_{2}\)‐CO)(µ\(_{2}\)‐η\(^{2}\)‐C,O‐PhCOCOPh)] 18, which indicate that radical intermediates and electron transfer processes might be of importance in the reaction of 1 with aldehydes and ketones.
Plants have evolved many mechanisms to defend against herbivores and pathogens. In many cases, these mechanisms took other duties. One example of such a neofunction- alisation would be carnivory. Carnivory evolved from the defence against herbivores. Instead of repelling the predator with a bitter taste, the plant kills it and absorbs its nutrients. A second example can be found in the pollination process. Many of the genes involved here were originally part of defence mechanisms against pathogens. In this thesis, I study these two examples on a genomic and transcriptomic level. The first project, Genomics of carnivorous Droseraceae, aims at obtaining annotated genome sequences of three carnivorous plants. I assembled the genome of Aldrovanda vesiculosa, annotated those of A. vesiculosa, Drosera spatulata and Dionaea muscipula and com- pared their genomic contents. Because of the high repetitiveness of the D. muscipula genome, I also developed reper, an assembly free method for detection, classification and quantification of repeats. With that method, we were able to study the repeats without the need of incorporating them into a genome assembly. The second large project investigates the role of DEFL (defensin-like) genes in pollen tube guidance in tobacco flowers. We sequenced the transcriptome of the SR1 strain in different stages of the pollination process. I assembled and annotated the transcriptome and searched for differentially expressed genes. We also used a method based on Hidden- Markov-Models (HMM) to find DEFLs, which I then analysed regarding their expression during the different stages of fertilisation. In total, this thesis results in annotated genome assemblies of three carnivorous Droser- aceae, which are used as a foundation for various analyses investigating the roots of car- nivory, insights into the role of DEFLs on a transcriptomic level in tobacco pollination and a new method for repeat identification in complex genomes.
Erstellung eines genregulatorischen Netzwerkes zur Simulation der Entstehung von Zahnhartsubstanz

In dieser Dissertation beschreibt der Autor die Erstellung eines grundlegenden bioinformatischen Modelles der menschlichen Zahnschmelzreifung. Mithilfe der KEGG Pathway-Datenbank wurde ein genregulatorisches Netzwerk (GRN) erstellt, welches maßgeblich auf den Signaltransduktionswegen Apoptose, Zellzyklus, Hedgehog-Signalweg, MAP-Kinase-Weg, mTOR-Signalweg Notch-Signalweg Signalweg, TGF-β-Signalweg und Wnt-Signalweg basiert. Im Weiteren wurde dieses Netzwerk durch zahlreiche verifizierte Wechselwirkungen erweitert und die zahnspezifischen Gene AMELX, AMELY, AMBN, ENAM und DSPP implementiert. In der anschließenden Simulation des Netzwerks mit dem Simulations-Tool Jimena konnten sechs stabile Zustände identifiziert werden. Diese wurden genauer untersucht und den Erkenntnissen eines GEO-Datensatzes gegenübergestellt. Langfristiges Ziel ist es, durch konsequente Optimierung des bioinformatischen Netzwerks Rückschlüsse auf die Odontogenese des Menschen zu ziehen.
Several bis(dimethylamino)‐substituted 1,4‐diaza‐2,3‐diborinines (DADBs) were synthesized with variable substituents at the backbone nitrogen atoms. By reaction with HCl or BX\(_{3}\) (X=Br, I), these species were successfully converted into their synthetically more useful halide congeners. The high versatility of the generated B−X bonds in further functionalization reactions at the boron centers was demonstrated by means of salt elimination (MeLi) and commutation (NMe\(_{2}\) DADBs) reactions, thus making the DADB system a general structural motif in diborane(4) chemistry. A total of 18 DADB derivatives were characterized in the solid state by X‐ray diffraction, revealing a strong dependence of the heterocyclic bonding parameters from the exocyclic substitution pattern at boron. According to our experiments towards the realization of a Dipp‐substituted, sterically encumbered DADB, the mechanism of DADB formation proceeds via a transient four‐membered azadiboretidine intermediate that subsequently undergoes ring expansion to afford the six‐membered DADB heterocycle.
Information about land use/land cover (LULC) and their changes is useful for different stakeholders to assess future pathways of sustainable land use for food production as well as for nature conservation. In this study, we assess LULC changes in the Kilombero catchment in Tanzania, an important area of recent development in East Africa. LULC change is assessed in two ways: first, post-classification comparison (PCC) which allows us to directly assess changes from one LULC class to another, and second, spectral change detection. We perform LULC classification by applying random forests (RF) on sets of multitemporal metrics that account for seasonal within-class dynamics. For the spectral change detection, we make use of the robust change vector analysis (RCVA) and determine those changes that do not necessarily lead to another class. The combination of the two approaches enables us to distinguish areas that show (a) only PCC changes, (b) only spectral changes that do not affect the classification of a pixel, (c) both types of change, or (d) no changes at all. Our results reveal that only one-quarter of the catchment has not experienced any change. One-third shows both, spectral changes and LULC conversion. Changes detected with both methods predominantly occur in two major regions, one in the West of the catchment, one in the Kilombero floodplain. Both regions are important areas of food production and economic development in Tanzania. The Kilombero floodplain is a Ramsar protected area, half of which was converted to agricultural land in the past decades. Therefore, LULC monitoring is required to support sustainable land management. Relatively poor classification performances revealed several challenges during the classification process. The combined approach of PCC and RCVA allows us to detect spatial patterns of LULC change at distinct dimensions and intensities. With the assessment of additional classifier output, namely class-specific per-pixel classification probabilities and derived parameters, we account for classification uncertainty across space. We overlay the LULC change results and the spatial assessment of classification reliability to provide a thorough picture of the LULC changes taking place in the Kilombero catchment.
Following natural disturbances, additional anthropogenic disturbance may alter community recovery by affecting the occurrences of species, functional groups, and evolutionary lineages. However, our understanding of whether rare, common, or dominant species, functional groups, or evolutionary lineages are most strongly affected by an additional disturbance, particularly across multiple taxa, is limited. Here, we used a generalized diversity concept based on Hill numbers to quantify the community differences of vascular plants, bryophytes, lichens, wood‐inhabiting fungi, saproxylic beetles, and birds in a storm‐disturbed, experimentally salvage logged forest. Communities of all investigated species groups showed dissimilarities between logged and unlogged plots. Most species groups showed no significant changes in dissimilarities between logged and unlogged plots over the first seven years of succession, indicating a lack of community recovery. In general, the dissimilarities of communities were mainly driven by rare species. Convergence of dissimilarities occurred more often than divergence during the early stages of succession for rare species, indicating a major role in driving decreasing taxonomic dissimilarities between logged and unlogged plots over time. Trends in species dissimilarities only partially match the trends in dissimilarities of functional groups and evolutionary lineages, with little significant changes in successional trajectories. Nevertheless, common and dominant species contributed to a convergence of dissimilarities over time in the case of the functional dissimilarities of wood‐inhabiting fungi. Our study shows that salvage logging following disturbances can alter successional trajectories in early stages of forest succession following natural disturbances. However, community changes over time may differ remarkably in different taxonomic groups and are best detected based on taxonomic, rather than functional or phylogenetic dissimilarities.
Forests are increasingly affected by natural disturbances. Subsequent salvage logging, a widespread management practice conducted predominantly to recover economic capital, produces further disturbance and impacts biodiversity worldwide. Hence, naturally disturbed forests are among the most threatened habitats in the world, with consequences for their associated biodiversity. However, there are no evidence-based benchmarks for the proportion of area of naturally disturbed forests to be excluded from salvage logging to conserve biodiversity. We apply a mixed rarefaction/extrapolation approach to a global multi-taxa dataset from disturbed forests, including birds, plants, insects and fungi, to close this gap. We find that 757% (mean +/- SD) of a naturally disturbed area of a forest needs to be left unlogged to maintain 90% richness of its unique species, whereas retaining 50% of a naturally disturbed forest unlogged maintains 73 +/- 12% of its unique species richness. These values do not change with the time elapsed since disturbance but vary considerably among taxonomic groups. Salvage logging has become a common practice to gain economic returns from naturally disturbed forests, but it could have considerable negative effects on biodiversity. Here the authors use a recently developed statistical method to estimate that ca. 75% of the naturally disturbed forest should be left unlogged to maintain 90% of the species unique to the area.
Global sustainability agendas focus primarily on halting deforestation, yet the biodiversity crisis resulting from the degradation of remaining forests is going largely unnoticed. Forest degradation occurs through the loss of key ecological structures, such as dying trees and deadwood, even in the absence of deforestation. One of the main drivers of forest degradation is limited awareness by policy makers and the public on the importance of these structures for supporting forest biodiversity and ecosystem function. Here, we outline management strategies to protect forest health and biodiversity by maintaining and promoting deadwood, and propose environmental education initiatives to improve the general awareness of the importance of deadwood. Finally, we call for major reforms to forest management to maintain and restore deadwood; large, old trees; and other key ecological structures.
Die hypertherme intraperitoneale Chemoperfusion (hyperthermic intraperitoneal chemotherapy, HIPEC) ist ein vielversprechendes Verfahren bei Patienten mit limitierter Peritonealkarzinose. Möglicherweise wirken Hitzeschockprotein (HSP)-vermittelte Reparaturmechanismen und proliferative Effekte dem Therapieerfolg jedoch entgegen. Um solche möglichen limitierenden Faktoren zu untersuchen, wurden in einem in vitro-HIPEC-Modell die Auswirkungen von Hyperthermie und Chemotherapie auf die Expression von HSPs und die Proliferationsmarker Ki-67 und PCNA im Kolonkarzinom mittels RT-qPCR und Western Blot untersucht.
Es zeigten sich hochregulierte HSPs, die mit zunehmender Temperatur und durch die Zugabe von Chemotherapie zunahmen. Man kann davon ausgehen, dass hierdurch intrazelluläre antiapoptotische Effekte und Reparaturmechanismen induziert werden. Die Inhibition von HSPs könnte daher zu einer Verbesserung der HIPEC-Therapie führen.
Zum anderen fand sich eine Erhöhung der Proliferationsmarker Ki-67 und PCNA nach der in vitro-HIPEC, was auf ein aggressives Wachstumsverhalten der überlebenden Zellen hindeutet.
The plant hormone jasmonoyl-isoleucine (JA-Ile) is an important regulator of plant growth and defense in response to various biotic and abiotic stress cues. Under our experimental conditions, JA-Ile levels increased approximately seven-fold in NaCl-treated Arabidopsis thaliana roots. Although these levels were around 1000-fold lower than in wounded leaves, genes of the JA-Ile signaling pathway were induced by a factor of 100 or more. Induction was severely compromised in plants lacking the JA-Ile receptor CORONATINE INSENSITIVE 1 or enzymes required for JA-Ile biosynthesis. To explain efficient gene expression at very low JA-Ile levels, we hypothesized that salt-induced expression of the JA/JA-Ile transporter JAT1/AtABCG16 would lead to increased nuclear levels of JA-Ile. However, mutant plants with different jat1 alleles were similar to wild-type ones with respect to salt-induced gene expression. The mechanism that allows COI1-dependent gene expression at very low JA-Ile levels remains to be elucidated.
The second messengers, cyclic adenosine 3′-5′-monophosphate (cAMP) and cyclic guanosine 3′-5′-monophosphate (cGMP), play important roles in many animal cells by regulating intracellular signaling pathways and modulating cell physiology. Environmental cues like temperature, light, and chemical compounds can stimulate cell surface receptors and trigger the generation of second messengers and the following regulations. The spread of cAMP and cGMP is further shaped by cyclic nucleotide phosphodiesterases (PDEs) for orchestration of intracellular microdomain signaling. However, localized intracellular cAMP and cGMP signaling requires further investigation. Optogenetic manipulation of cAMP and cGMP offers new opportunities for spatio-temporally precise study of their signaling mechanism. Light-gated nucleotide cyclases are well developed and applied for cAMP/cGMP manipulation. Recently discovered rhodopsin phosphodiesterase genes from protists established a new and direct biological connection between light and PDEs. Light-regulated PDEs are under development, and of demand to complete the toolkit for cAMP/cGMP manipulation. In this review, we summarize the state of the art, pros and cons of artificial and natural light-regulated PDEs, and discuss potential new strategies of developing light-gated PDEs for optogenetic manipulation.
Phonologische Bewusstheit stellt die Kompetenz dar, Sprache in kleinere Einheiten wie einzelne Silben und Phoneme untergliedern zu können. Sie ist damit eine wichtige Vorläuferfertigkeit für den Schriftspracherwerb.
Die vorliegende Arbeit untersucht die Entwicklung der phonologischen Bewusstheit während des letzten Kindergartenjahres. Zum einen werden die faktorielle Struktur und die Messinvarianz der phonologischen Bewusstheit im Verlauf des letzten Kindergartenjahres analysiert. Als zweiter Aspekt werden die wechselseitigen Zusammenhänge der phonologischen Bewusstheit mit weiteren phonologischen, frühen schriftsprachlichen und sprachlichen Kompetenzen im Vorschulalter untersucht. Der dritte Aspekt bezieht sich auf die institutionelle Förderung phonologischer Bewusstheit bei Vorschulkindern im Kindergarten. Hier werden die Effekte einer expliziten Förderung durch das Trainingsprogramm „Hören, lauschen, lernen“ (Küspert & Schneider, 2018) und einer impliziten Förderung im Kindergartenalltag sowie inzidentelle Einflüsse durch Erwartungseffekte untersucht.
Zur Untersuchung der Fragestellungen wurde ein längsschnittliches Design mit zwei Messzeitpunkten zu Beginn und Ende des letzten Kindergartenjahres verwendet. Die Stichprobe umfasste 390 Vorschulkinder. Für die statistischen Analysen wurden im Wesentlichen Strukturgleichungsmodelle verwendet.
Die Ergebnisse zur Struktur der phonologischen Bewusstheit zeigten Schwierigkeiten bei der längsschnittlichen Abbildung des Konstrukts im Vorschulalter. Da die Tests zur Erfassung der phonologischen Bewusstheit im weiteren Sinne (auf Reim- und Silbenebene) Deckeneffekte aufwiesen und sich im Vorschulalter insbesondere die phonologische Bewusstheit im engeren Sinne (auf Phonemebene) entwickelt, wurde das latente Konstrukt der phonologischen Bewusstheit im Weiteren ausschließlich durch Messverfahren zur Erfassung der phonologischen Bewusstheit im engeren Sinne modelliert. Zudem ließ sich lediglich schwache Messinvarianz etablieren, sodass die Befunde auf einen qualitativen Wandel des Konstrukts während des letzten Kindergartenjahres hindeuten.
Die Befunde zu Zusammenhängen der phonologischen Bewusstheit mit weiteren phonologischen, frühen schriftsprachlichen und sprachlichen Kompetenzen ergaben komplexe wechselseitige Effekte. Die phonologische Bewusstheit sagte dabei die Entwicklung früher schriftsprachlicher Kompetenzen vorher, während sich die Entwicklung der phonologischen Bewusstheit selbst auch durch grammatikalische Kompetenzen erklären ließ.
Bei den Analysen zur Förderung der phonologischen Bewusstheit im Vorschulalter war insbesondere die explizite Förderung durch „Hören, lauschen, lernen“ (Küspert & Schneider, 2018) effektiv. Für die Effektivität des Programms waren zudem die Implementationsbedingungen im Kindergarten relevant. Hier erwies sich vor allem eine vorherige Schulung der Erzieher(innen) als positiv sowie auch eine Abweichung vom Manual in organisatorischer Hinsicht, sodass das Training nicht täglich, sondern mehrmals pro Woche in größeren Abschnitten stattfand. Auf die implizite Förderung der phonologischen Bewusstheit konnte lediglich indirekt über das Wissen der Erzieher(innen) über Sprach- und frühe Schriftsprachförderung geschlossen werden. Das Wissen der Erzieher(innen) über die Förderung phonologischer Bewusstheit war dabei nicht von Bedeutung für die Kompetenzentwicklung der Kinder. Stattdessen wirkte sich das Wissen über sprachliche Fördermaßnahmen, insbesondere bezüglich Maßnahmen bei Aussprachstörungen, günstig auf die Entwicklung der phonologischen Bewusstheit aus. Neben Effekten einer expliziten und impliziten Förderung der phonologischen Bewusstheit waren auch inzidentelle Effekte aufgrund der Urteile der Erzieher(innen) über die Kompetenzen der Kinder nachweisbar. Auch hier wirkten sich die Einschätzungen der sprachlichen Kompetenzen in den Bereichen Aussprache, Wortschatz und Grammatik auf die Entwicklung der phonologischen Bewusstheit aus, während die Einschätzung der phonologischen Bewusstheit selbst durch die Erzieher(innen) die weitere Entwicklung nicht vorhersagen konnte.
Insgesamt sprechen die Befunde für komplexe Zusammenhänge der phonologischen Bewusstheit mit weiteren phonologischen, frühen schriftsprachlichen und sprachlichen Kompetenzen, die auch bei der Konzeption von Fördermaßnahmen berücksichtigt werden sollten.
Media have become omnipresent in children’s and youths’ everyday lives, and they also offer rich chances and challenges for educational contexts. On the one hand, media can, for example, support students’ learning effectively, enhance lessons with innovative tools and methods and help individualize teaching and learning processes. On the other hand, students need to learn, e.g., how to use these media, how to select and evaluate them and how to act responsibly in a digitalized and mediatized world. Teachers are a core stakeholder in this context. To take advantage of the benefits media offer for teaching and learning processes, to support students in the acquisition of respective competencies and to fulfill numerous other media-related tasks and challenges, teachers need to acquire respective competencies in their initial teacher education, which can be summarized as media-related educational competencies.
The relevance of these competencies is evident on different levels. In related research, respective competency models are developed, and in practices of teacher education, competencies are measured and efforts are taken to advance the competencies of preservice teachers. Against this background, this semi-cumulative dissertation presents a theory-based and empirical analysis of the competencies in question from a comprehensive and multidimensional perspective. In accordance with the central aspects outlined, the three systematic main fields focused on are models of media-related educational competencies, their measurement and practices of advancement in teacher education, as well as the interplay of these three fields. The dissertation takes on an international comparative perspective and focuses on the examples of initial teacher education in Germany and the USA.
The article-based dissertation comprises three main parts, framed by introduction and conclusion. The introduction provides a basis for the following work with regards to terminology, scope of research and overall methodology. The first main part is concerned with models of media-related educational competencies and includes a theory-based systematic comparison of three relevant models. This part explicates the varieties between competency models, and it discusses central aspects of selection and application. In Part II, methods and varieties of competency measurement are focused on, and an article is presented which shares results of an exploratory quantitative measurement of the respective competencies of German and US preservice teachers. Overall, this part reveals the potential and limitations of competency measurement and transfers these conclusions to the competency models introduced in Part I. Part III is concerned with an analysis of current practices of advancing media-related educational competencies in Germany and the USA. In this context, stakeholders influencing these practices will be systemized and analyzed in their role and impact. The article included in Part III introduces interviews which were conducted to achieve insights into the perspectives of selected experts, regarding relevant models, practices and outcomes of media-related teacher education in Germany and the USA.
Finally, the Conclusion of the dissertation will draw together the different strands, clarify the close connection between the domains of modeling, measuring and advancing the competencies in question and discuss the interdependencies of these three dimensions. These perspectives help both to contextualize and bring together important facets which have often been treated separately in related research and will add new facets to ultimately achieve a comprehensive and multifaceted viewpoint.
Against the background of the intercultural comparative perspective, the results and findings will ultimately achieve an enhanced and deep analysis and reflection on the complex field of media-related educational competencies in Germany and the USA and beyond.
Die Häufigkeit der endovaskulären Interventionen (EI) an den unteren Extremitäten zur Behandlung der peripheren arteriellen Verschlusskrankheit (pAVK) hat in den letzten Jahren zugenommen. Bei steigender Inzidenz der pAVK und bei gleichzeitiger Reduzierung der Rate mit operativen Interventionen (OI) profitieren die Patienten von der EI. Außerdem stieg die Anzahl der älteren Patienten, die aufgrund einer pAVK im Krankenhaus einer EI zugeführt wurden. Es zeigt sich eine Erweiterung der Indikationen für eine EI von den Claudicanten hin zu den Patienten mit den Stadien der kritischen Ischämie. Hierdurch werden gerade ältere und morbidere Patienten mit komplexeren endovaskulären Prozeduren konfrontiert. Insbesondere im Krankenhaus ergibt sich die Möglichkeit, den pAVK-Patienten, die bisher eine OI mit einer anästhesiologischen Begleitung erhielten, eine EI in SPA anzubieten. In der Regel werden EI in Lokalanästhesie durchgeführt. Die Anwendung der SPA bei EI ist interessant, da es möglicherweise neben der kompletten Schmerzausschaltung der unteren Extremitäten bei erhaltener Kommunikation zu Synergieeffekten durch die periphere Vasodilatation kommen kann. Als ein seltenes Verfahren zur Verbesserung der peripheren Durchblutung hat dies die CT-gesteuerte temporäre Sympathikolyse nach erfolgter EI gezeigt. Um prospektiv zu untersuchen, ob eine EI in SPA eine Verbesserung des Outcome und des Behandlungsablaufs mit sich bringt, ist zunächst wichtig zu zeigen, dass die Anwendung von SPA bei EI nicht gefährlich ist. Hierzu stehen Daten aus der Allgemein-, Viszeral- und Gefäßchirurgischen Abteilung des Klinikums Main-Spessart zur Verfügung. Im Zeitraum vom 15.12.2009 bis 22.01.2015 wurden bei 59 Patienten EI in SPA durchgeführt.
Bei keinem der 59 Patienten kam es durch die Anwendung der SPA zu anästhesieabhängigen Komplikationen. Als zu erwartende Nebenwirkung trat der durch temporäre Sympathikolyse ausgelöste Blutdruckabfall bei allen Patienten auf. Im Mittel sank der Wert um 24,2 % des Ausgangswerts, bei 13 Patienten (22,0 %) wurde eine Korrektur des Blutdruckabfalls mit Akrinor® durchgeführt und gut beherrscht. Für die Stärke des Blutdruckabfalls konnte keine Signifikanz bei der ASA-Einteilung, der Geschlechterverteilung und dem Alter nachgewiesen werden, somit besteht für eine EI in SPA keine Kontraindikation für den älteren und kränkeren pAVK-Patienten.
Die Ergebnisse dieser Dissertation zeigen, dass die Anwendung der SPA bei EI ein sicheres Verfahren ist. Die SPA kann, gerade bei zu erwartenden komplexen peripheren EI eine für den Patienten und Interventionisten angenehme und sichere Alternative zur Lokalanästhesie sein. Der personelle und materialbedingte Aufwand einer EI in SPA, im Vergleich mit einer EI in Lokalanästhesie, ist dabei deutlich erhöht und scheint übertrieben. Dass die EI in SPA Synergieeffekte mit sich bringt und deshalb sinnvoll ist, kann nur vermutet werden. Dieser Zusammenhang könnte im Rahmen einer prospektiven Studie, die beide Anästhesieverfahren für eine EI vergleicht, gezeigt werden.
Comparison of moving and fixed basis sets for nonadiabatic quantum dynamics at conical intersections

We assess the performance of two different types of basis sets for nonadiabatic quantum dynamics at conical intersections. The basis sets of both types are generated using Ehrenfest trajectories of nuclear coherent states. These trajectories can either serve as a moving (time-dependent) basis or be employed to sample a fixed (time-independent) basis. We demonstrate on the example of two-state two-dimensional and three-state five-dimensional models that both basis set types can yield highly accurate results for population transfer at intersections, as compared with reference quantum dynamics. The details of wave packet evolutions are discussed for the case of the two-dimensional model. The fixed basis is found to be superior to the moving one in reproducing nonlocal spreading and maintaining correct shape of the wave packet upon time evolution. Moreover, for the models considered, the fixed basis set outperforms the moving one in terms of computational efficiency.

The fixed basis is found to be superior to the moving one in reproducing true nonlocal spreading and maintaining correct shape of the wave packet upon time evolution. Moreover, for the models considered, the fixed basis set outperforms the moving one in terms of computational efficiency.
Die Mitogen-aktivierten Proteinkinasen ERK1/2 (extrazellulär Signal-regulierte Kinase 1 und 2) sind die Effektorkinasen der Raf/MEK/ERK-Kaskade und verknüpfen externe Stimuli mit der intrazellulä-ren Antwort, wodurch sie wichtige Schlüsselmoleküle der zellulären Signaltransduktion darstellen. Zahlreiche Studien belegen die Beteiligung von ERK1/2 an der Entstehung pathologischer kardialer Hypertrophie. Genauso ist bekannt, dass ERK1/2 anti-apoptotische, kardioprotektive Eigenschaften besitzen.
So führte, wie in dieser Arbeit gezeigt, eine Hemmung der katalytischen ERK1/2-Aktivität durch den MEK-Inhibitor PD98059 zu einer signifikanten Reduktion der hypertrophen Antwort von Kardiomy-ozyten auf den Stimulus Phenylephrin. Dies war allerdings mit einem Anstieg der Apoptoserate in diesen Zellen verbunden, wodurch sich eine Hemmung der totalen ERK-Aktivität als nicht praktika-bel für die Behandlung pathologischer kardialer Hypertrophie herauskristallisierte. In früheren Un-tersuchungen wurde eine Autophosphorylierung von ERK an Threonin 188 (murines ERK2) entdeckt und als Trigger für ERK1/2-vermitteltes hypertrophes Wachstum identifiziert. Diese Autophospho-rylierung steuert die nukleäre Lokalisation von ERK1/2 und ermöglicht so die Aktivierung nukleärer ERK-Zielproteine sowie hypertrophes Wachstum. Eine Interferenz mit der ERKThr188-Phosphorylierung konnte schon in vitro und in vivo erfolgreich einer pathologischen Hypertrophie entgegenwirken, ohne Einfluss auf physiologisches Herzwachstum oder die zytosolischen, anti-apoptotischen Effekte von ERK1/2 zu nehmen. Einen initialen Schritt für das Zustandekommen dieser Autophosphorylierung an Threonin 188 stellt dabei die Dimerisierung von ERK dar. In der vorliegenden Arbeit wurde daher die Inhibition der ERK-Dimerisierung im Hinblick auf die Behand-lung ERKThr188-vermittelter pathologischer Hypertrophie untersucht. Dabei sollte die endogene ERK-Dimerisierung mithilfe eines selbst generierten Peptids unterbunden werden. In Übereinstimmung mit den Ergebnissen zu einer dimerisierungsdefizienten ERK2-Mutante (ERK2-Δ4) konnte das Peptid in vitro und in vivo erfolgreich pathologisch hypertrophes Herzwachstum mindern. Dabei führte es sogar zu einem Rückgang des apoptotischen Zelltodes, ausgelöst durch eine Aortenligation, füh-ren. Es zeigte sich, dass das Peptid die nukleäre Translokation von ERK2 verhindert und dadurch nukleäre ERK-Substrate geringer aktiviert werden. Da eine Dysregulation in der Raf/MEK/ERK-Kaskade auch die Entstehung von Tumoren begünstigen kann, sollte schließlich untersucht wer-den, ob das Prinzip der Hemmung nukleärer ERK-Effekte auch die Proliferation von Krebszellen beeinflussen kann. Es stellte sich heraus, dass die Peptid-vermittelte Hemmung der ERK-Dimerisierung auch die Proliferation von Kolonkarzinomzelllinien mit unterschiedlichen Mutations-stadien der Raf/MEK/ERK-Kaskade reduziert.
In der vorliegenden Arbeit konnte somit die Intervention mit der ERK-Dimerisierung als Target der ERKThr188-Autophosphorylierung als translationale Strategie zur Reduktion nukleärer ERK-Effekte herausgearbeitet werden. Dies bietet die Möglichkeit ERK-vermittelte pathologische kardialer Hy-pertrophie und ERK-vermittelte Tumor-Proliferation zu behandeln, ohne kardiotoxische Nebenwir-kungen zu verursachen.
Der Hodenhochstand ist die häufigste kongenitale Anomalie des männlichen Urogenitaltraktes und steht in Zusammenhang mit einer verminderten Fertilität sowie einer erhöhten Malignität. Seit dem Jahr 2009 liegt das definierte Behandlungsziel der AWMF-Leitlinie für Hodenhochstand bei einer abgeschlossenen Therapie innerhalb der ersten zwölf Lebensmonate. Nationale sowie internationale Studien zeigen bisher eine mangelnde Umsetzung der Therapievorgaben.
Die vorliegende retrospektive Studie untersucht die Umsetzung der Therapieleitlinie an der kinderchirurgischen Abteilung des Uniklinikum Würzburgs im Zeitraum von 2010 bis 2015.
Von den insgesamt 578 Patienten, die aufgrund eines uni- bzw. bilateralem Hodenhochstandes operativ versorgt wurden, sind 21,1 % leitliniengerecht therapiert worden. Das durchschnittliche Operationsalter lag bei 40 Monaten, wobei kein Trend zu einer früheren Operation in dem betrachteten Zeitraum festzustellen ist. Die Erstvorstellung aufgrund eines Hodenhochstandes erfolgte in 30 % der Fälle innerhalb des ersten Lebensjahres in der kinderchirurgischen Abteilung, womit 70 % der Patienten den Zeitpunkt einer empfohlenen Therapie nicht erfüllt haben. Neben dem später resultierenden Operationszeitpunkt bei sekundär aszendierten Hoden zeigte eine Analyse der behandlungsbeeinflussenden Faktoren, dass sich das Vorliegen chronischer Begleiterkrankungen ebenfalls negativ auf den Zeitpunkt der Therapie auswirkt. Vor allem die engmaschige Betreuung verschiedener Fachdisziplinen wirkt sich positiv auf den Therapiezeitpunkt aus.
Zusammenfassend ist festzustellen, dass der grundlegende Baustein einer frühzeitigen Diagnose und Therapie in der fortlaufenden Kontrolle der Hodenlokalisation, der Aufklärung der Eltern bezüglich des Krankheitsbildes und möglicher Spätkomplikation sowie der Anleitung zur Selbstuntersuchung besteht.
To evaluate the effects of Ramadan observance on dietary intake, body mass and body composition of adolescent athletes (design: systematic review and meta-analysis; data sources: PubMed and Web of Science; eligibility criteria for selecting studies: single-group, pre-post, with or without control-group studies, conducted in athletes aged <19 years, training at least 3 times/week, and published in any language before 12 February 2020). Studies assessing body mass and/or body composition and/or dietary intake were deemed eligible. The methodological quality was assessed using ‘QualSyst’. Of the twelve selected articles evaluating body mass and/or body composition, one was of strong quality and eleven were rated as moderate. Ten articles evaluated dietary intake; four were rated as strong and the remaining moderate in quality. Continuation of training during Ramadan did not change body mass from before to the first week (trivial effect size (ES) = −0.011, p = 0.899) or from before to the fourth week of Ramadan (trivial ES = 0.069, p = 0.277). Additionally, Ramadan observance did not change body fat content from before to the first week (trivial ES = −0.005, p = 0.947) and from before to the fourth week of Ramadan (trivial ES = -0.057, p = 0.947). Lean body mass remained unchanged from before to the fourth week of Ramadan (trivial ES = −0.025, p = 0.876). Dietary data showed the intake of energy (small ES = -0.272, p = 0.182), fat (trivial ES = 0.044, p = 0.842), protein (trivial ES = 0.069, p = 0.720), carbohydrate (trivial ES = 0.075, p = 0.606) and water (trivial ES = −0.115, p = 0.624) remained essentially unchanged during as compared to before Ramadan. Continued training of adolescent athletes at least three times/week during Ramadan observance has no effect on body mass, body composition or dietary intake.
Lithium ist noch immer der Goldstandard in der Behandlung der bipolaren Störung und kommt auch in der Behandlung der unipolaren Depression zur Anwendung. Die therapeutische Breite von Lithium ist jedoch gering. Bei zu hohen Spiegeln kann es zu schweren Nebenwirkungen bis hin zu Intoxikationen mit letalem Verlauf kommen. Mit zunehmendem Lebensalter nimmt die renale Lithiumclearance ab. Hinzu kommen somatische Komorbiditäten, welche die renale Lithiumcelarance ebenfalls beeinträchtigen können. Darüber hinaus gibt es eine Vielzahl von Arzneimitteln, welche ebenfalls Einfluss auf den Lithiumspiegel nehmen können. Zu diesen zählen Diuretika, ACE-Hemmer und AT1-Rezeptor-Antagonisten sowie nicht-steroidale Antirheumatika, welche den am häufigsten rezeptierten Medikamente gehören.
In einer retrospektiven naturalistischen Studie wurde der Einfluss einer Komedikation aus einem Schleifen- (Furosemid/Torasemid), Thiazid- (HCT) oder kaliumsparenden Diuretikum (Amilorid, Spironolacton, Triamteren), einem ACE-Inhibitor (Captopril, Enalapril, Lisinopril, Ramipril) oder AT1-Rezeptor-Antagonisten (Candesartan, Losartan, Irbesartan, Olmesartan, Valsartan) oder einem nicht-steroidalen Antirheumatikum (Acetylsalicylsäure, Diclofenac, Ibuprofen) auf den dosisbezogenen Lithiumspiegel untersucht. Als Stichprobe dienten 501 Lithiumpatienten, welche stationär im Zentrum für Psychische Gesundheit der Universitätsklinik Würzburg behandelt worden waren (01/2008 – 12/2015). 92 Patienten (18,4 %) nahmen nur eines der aufgeführten Medikamente ein, während 76 (15,1 %) eine Kombination von bis zu 5 Medikamenten erhielten – somit beinhaltete die Komedikation von 33,5 % der Patienten mindestens eines der aufgeführten Medikamente. Als Kontrollgruppe diente eine Stichprobe von 333 Lithiumpatienten ohne entsprechende Komedikation.
Altersintrinsische Faktoren (p < 0,001; R2=0,289), die GFR (p < 0,001; R2=0,377) sowie das Geschlecht (p < 0,001; R2=0,406) scheinen den größten Einfluss auf den Lithiumspiegel zu nehmen (ca. 41 %), während für die Komedikation ein geringerer Effekt anzunehmen ist (ca. 4%). Die Ergebnisse sprechen für ein signifikantes Interaktionspotential von Diclofenac und Ibuprofen (p = 0,001). Es ergeben sich auch Hinweise auf ein relevantes Interaktionspotential von Hydrochlorothiazid (p = 0,020). Patienten, welche Acetylsalicylsäure (p < 0,001) oder Allopurinol (p = 0,003) erhalten, scheinen ein Risikokollektiv für erhöhte Lithiumspiegel darzustellen. Die dosisbezogenen Lithiumspiegel der Stichproben mit Einnahme eines ACE-Hemmers/AT1-Rezeptor-Antagonisten und eines Schleifendiuretikums unterschieden sich dagegen nicht signifikant von denen der Kontrollstichprobe ohne Komedikation. Es ist zudem davon auszugehen, dass eine Kombination mehrerer Pharmaka mit Interaktionspotential (p < 0,001) ein höheres Risiko für erhöhte dosisbezogene Lithiumspiegel birgt als eine Monotherapie (p = 0,026) und die Indikation einer solchen sollte daher kritisch geprüft werden. Eine zusätzliche Analyse von 32 Fällen von supratherapeutischen Lithiumserumkonzentrationen von ≥ 1,3 mmol/l (1,3–4,1mmol/l) legt nahe, dass sich ein Großteil von Lithiumintoxikationen durch regelmäßige Spiegelkontrollen und Dosisanpassungen unter Berücksichtigung von Komedikation, Alter und Komorbiditäten sowie Psychoedukation der Patienten vermeiden ließen.
Mithilfe eines frühzeitigen und umfassenden Entlassungsmanagements sollen Schnittstellen im Behandlungsprozess überwunden und der Übergang des Patienten in die poststationäre Versorgung nach einem Krankenhausaufenthalt optimal vorbereitet werden. Trotz der allgemein anerkannten Bedeutung des Entlassungsmanagements aus der stationären medizinischen Versorgung erfolgt die Umsetzung bei den Rehabilitationskliniken sehr unterschiedlich. Im Rahmen dieser Arbeit wird ein kurzes Fragebogeninstrument für Patienten systematisch entwickelt und erprobt, welches im Rahmen einer postalischen Nachbefragung ehemaliger Rehabilitanden eingesetzt werden soll. Es soll die Qualität des Entlassungsmanagements der medizinischen Rehabilitation systematisch erfassen und Impulse für das Qualitätsmanagement der Kliniken bieten. Die Ergebnisse der Erprobung in acht Kliniken mit über 900 auswertbaren Fragebögen weisen darauf hin, dass beides möglich ist. Sie werden mit dem Ziel der Weiterentwicklung des Instruments diskutiert und es werden Vorschläge zur weiteren Nutzung im Alltag der Rehabilitation erarbeitet.
Bees need food of appropriate nutritional quality to maintain their metabolic functions. They largely obtain all required nutrients from floral resources, i.e., pollen and nectar. However, the diversity, composition and nutritional quality of floral resources varies with the surrounding environment and can be strongly altered in human-impacted habitats. We investigated whether differences in plant species richness as found in the surrounding environment correlated with variation in the floral diversity and nutritional quality of larval provisions (i.e., mixtures of pollen, nectar and salivary secretions) composed by the mass-provisioning stingless bee Tetragonula carbonaria (Apidae: Meliponini). We found that the floral diversity of larval provisions increased with increasing plant species richness. The sucrose and fat (total fatty acid) content and the proportion and concentration of the omega-6 fatty acid linoleic acid decreased, whereas the proportion of the omega-3 fatty acid linolenic acid increased with increasing plant species richness. Protein (total amino acid) content and amino acid composition did not change. The protein to fat (P:F) ratio, known to affect bee foraging, increased on average by more than 40% from plantations to forests and gardens, while the omega-6:3 ratio, known to negatively affect cognitive performance, decreased with increasing plant species richness. Our results suggest that plant species richness may support T. carbonaria colonies by providing not only a continuous resource supply (as shown in a previous study), but also floral resources of high nutritional quality.
Es wurden die Daten von 119 Meningeompatienten hinsichtlich der lokalen Kontrolle sowie des Gesamtüberlebens und deren Einflussfaktoren untersucht, um dieses Kollektiv hinsichtlich des Therapieerfolges an der Klinik und Poliklinik für Strahlentherapie der Universität Würzburg auszuwerten. Ferner wurden die akuten und chronischen Nebenwirkungen der Strahlenbehandlung erfasst. Darüber hinaus wurden die Fragebogen QLQ-C30 sowie -BN20 zur Evaluation der Lebensqualität nach der Strahlenbehandlung analysiert.
Die Strahlenbehandlung zeigte sich im Rahmen des Follow-Ups von 5,4 Jahren als größtenteils effektive und sichere Behandlungsmethode. Aufgeschlüsselt nach den WHO-Graden lagen die lokalen Kontrollraten bei 93,3% (ohne Histologie), 91,2% (WHO-Grad I), 66,7% (WHO-Grad II) sowie 53,1% (WHO-Grad III) nach fünf Jahren. Nach zehn Jahren lagen die Raten bei 82,0% (ohne Histologie), 91,2% (WHO-Grad I), 46,7% (WHO-Grad II) sowie 42,5% (WHO-Grad III). In der multivariaten Analyse zeigte sich ein grenzwertig signifikanter Unterschied der lokalen Kontrolle für die Histologie (p = 0,050), sowie deutliche Signifikanz zwischen benignen und malignen Meningeomen. Das Gesamtüberleben lag nach fünf Jahren bei 89,6% sowie nach zehn Jahren bei 75,9%. Signifikante Einflussfaktoren in der multivariaten Analyse waren die Histologie sowie die Größe des GTV. Die Rate an medizinisch signifikanten (CTCAE ≥3) akuten und chronischen Nebenwirkungen war niedrig (5,9% bzw. 4,2%) und vergleichbar mit den Daten der Literatur.
Die Arbeit liefert erstmalig Lebensqualitätsdaten mittels der Fragebogen der EORTC an einem reinen Meningeomkollektiv, bei dem alle Patienten eine Strahlenbehandlung erhielten. Diese zeigen schlechtere Werte im Vergleich zur Normalbevölkerung. Ebenso weisen sie auf eine Verschlechterung der Lebensqualität im Laufe der Tumorbehandlung hin. Dies ist allerdings aufgrund des einmaligen Messzeitpunktes und des fehlenden Vergleichskollektivs weder auf die Operation, noch auf die Bestrahlung kausal zurückführbar.
Um den genauen Stellenwert der Strahlentherapie im Therapieregime der Meningeome abhängig von der Histologie beurteilen zu können sowie diese Ergebnisse zu verifizieren und zu festigen, sind aufwendige prospektive Studien nötig. Diese werden zum Teil aktuell durchgeführt. Regelmäßige Kontrollen der Lebensqualität sowie der Nebenwirkungen sollen erhoben und ausgewertet werden, um die Auswirkungen der Therapie möglichst allumfassend darstellen zu können.
Mit fortschreitender chronischer Niereninsuffizienz kommt es zur Akkumulation von Urämietoxinen und im Endstadium unbehandelt zum Tod im sogenannten Urämischen Syndrom. Die Blutreinigung erfolgt bei der am häufigsten verwendeten Form der Nierenersatztherapie, der Hämodialyse, nur unzureichend. Die Folge ist eine erhöhte Morbidität und Mortalität der betroffenen Patienten. Bei der Hämodialyse werden nur Urämietoxine bis zu einer Größe von 20 kDa über die im Dialysator eingesetzten Hohlfaserdialysemembranen diffusiv und konvektiv semiselektiv nach Größenausschluss entfernt. Proteingebundene Urämietoxine, deren effektive Größe durch die Bindung an Transportproteine wie beispielsweise Albumin die Trennschärfe der Dialysemembranen übersteigt, werden retiniert. In-vivo werden proteingebundene Urämietoxine im proximalen Tubulus, einem Teil des tubulären Systems des Nephrons, sekretorisch eliminiert.
Im Rahmen der vorliegenden Promotionsarbeit wurden die ersten Entwicklungsschritte auf dem Weg zu einem sogenannten bio-artifiziellen Tubulus evaluiert. Der angedachte biohybride Filter sollte aus einer Ko-Kultur funktionaler humaner proximaler Tubuluszellen und humaner Endothelzellen (HUVEC) auf synthetischen Hohlfasermembranen bestehen und könnte während der Hämodialyse als zusätzlicher Reinigungsschritt angewendet werden, um unter anderem proteingebundene Urämietoxine effektiv durch aktiven Transport aus dem Blut der Patienten zu entfernen. Die Differenzierung der proximalen Tubuluszellen erfolgte dabei aus adulten adipozytären mesenchymalen Stammzellen (ASC), deren Herkunft eine spätere autologe Behandlung ermöglicht. Die Ko-Kultur mit Endothelzellen wurde zur potentiellen Steigerung der Sekretion proteingebundener Urämietoxine verwendet.
In der vorliegenden Arbeit konnten ASCs durch eine Kombination der löslichen Differenzierungsfaktoren All-Trans-Retinoinsäure (ATRA), Aktivin A und BMP-7 erfolgreich in Zytokeratin 18-exprimierende Zellen differenziert werden, wodurch die erwünschte epitheliale Differenzierung bestätigt wurde. Die Expression funktionaler Proteine, wie das für den Wassertransport relevante Aquaporin 1 oder auch der Na+-/K+-ATPase, konnte in dieser Arbeit bereits vor der Differenzierung nachgewiesen werden. Im nächsten Schritt wurde erfolgreich gezeigt, dass eine simultane, qualitativ hochwertige Ko-Kultur von ASCs und HUVECs auf der mit dem extrazellulären Matrixprotein Fibronektin modifizierten Innen- bzw. Außenseite von synthetischen Hohlfasermembranen aus Polypropylen bzw. Polyethersulfon möglich ist. Die Viabilität beider Zelltypen wurde dabei durch die Verwendung eines für die Ko-Kultur entwickelten Nährmediums erreicht, in welchem die Proliferation von ASCs bei gleichzeitiger Aufrechterhaltung ihrer Stammzelleigenschaften deutlich erhöht war.
Die in dieser Arbeit erzielten Ergebnisse stellen eine aussichtsreiche Basis für einen bio-artifiziellen renalen Tubulus dar. Weitere Entwicklungsschritte, wie die Differenzierung der ASCs zu proximalen Tubuluszellen im 3D-Bioreaktor einschließlich ihrer funktionalen Charakterisierung anhand Tubulusepithel-spezifischer Transporter, sind erforderlich, be-vor erste funktionale Experimente vor dem „Upscaling“ auf klinisch verwendbare Module möglich sind.
Macrophages are key players of the innate immune system that can roughly be divided into the pro-inflammatory M1 type and the anti-inflammatory, pro-healing M2 type. While a transient initial pro-inflammatory state is helpful, a prolonged inflammation deteriorates a proper healing and subsequent regeneration. One promising strategy to drive macrophage polarization by biomaterials is precise control over biomaterial geometry. For regenerative approaches, it is of particular interest to identify geometrical parameters that direct human macrophage polarization. For this purpose, we advanced melt electrowriting (MEW) towards the fabrication of fibrous scaffolds with box-shaped pores and precise inter-fiber spacing from 100 μm down to only 40 μm. These scaffolds facilitate primary human macrophage elongation accompanied by differentiation towards the M2 type, which was most pronounced for the smallest pore size of 40 μm. These new findings can be important in helping to design new biomaterials with an enhanced positive impact on tissue regeneration.
Die aneurysmatische SAB ist trotz etablierter Therapieverfahren (Coiling und Clipping) weiterhin ein Krankheitsbild mit hoher Mortalität. In unserer Arbeit haben wir retrospektiv die Patientenakten der Patienten, die mit der Diagnose aneurysmatische SAB am Universitätsklinikum Würzburg zwischen dem 01.01.1999 und dem 31.12.2009 aufgenommen wurden, ausgewertet. Es konnte dargestellt werden das als Hauptrisikofaktoren für ein schlechtes Therapieergebnis ein schlechter Aufnahmestatus des Patienten und das Auftreten von Komplikationen im Verlauf verantwortlich sind.
We here present the case of a 67-year-old woman with a history of a slowly progressive, polypous nodule on her left wrist. The lesion was excised, and the histological analysis revealed a clear cell tumour that was relatively sharply demarked from the surrounding tissue extending into the subcutaneous tissue. The tumour showed a characteristic trabecular pattern in which the tumour cells were arranged around numerous vessels. The neoplastic cells had a predominantly epithelioid shape, granular eosinophilic to clear cytoplasm and prominent centrally located nucleoli. The histological differential diagnosis included a metastatic clear-cell renal cell carcinoma and a primary cutaneous perivascular epithelioid cell tumour (PEComa). Immunohistochemically, the tumour cells revealed homogenous expression of HMB-45, MiTF and CD10, whereas MART-1 and S100 were negative. Antibodies against actin marked the trabecularly arranged vessels, and the neoplastic cells yielded a patchy positivity against actin and desmin. Additional immunohistochemical stains against pan-cytokeratin, CAIX, PAX-8 and EMA were negative. Based on the morphologic and immunophenotypic findings, the histological diagnosis of a CD10-positive cutaneous PEComa was made.
Clustered Regularly Interspaced Short Palindromic Repeat (CRISPR) loci are found in bacterial and archaeal genomes where they provide the molecular machinery for acquisition of immunity against foreign DNA. In addition to the cas genes fundamentally required for CRISPR activity, a second class of genes is associated with the CRISPR loci, of which many have no reported function in CRISPR-mediated immunity. Here, we characterize MM_0565 associated to the type I-B CRISPR-locus of Methanosarcina mazei Gö1. We show that purified MM_0565 composed of a CRISPR-Cas Associated Rossmann Fold (CARF) and a winged helix-turn-helix domain forms a dimer in solution; in vivo, the dimeric MM_0565 is strongly stabilized under high salt stress. While direct effects on CRISPR-Cas transcription were not detected by genetic approaches, specific binding of MM_0565 to the leader region of both CRISPR-Cas systems was observed by microscale thermophoresis and electromobility shift assays. Moreover, overexpression of MM_0565 strongly induced transcription of the cas1-solo gene located in the recently reported casposon, the gene product of which shows high similarity to classical Cas1 proteins. Based on our findings, and taking the absence of the expressed CRISPR locus-encoded Cas1 protein into account, we hypothesize that MM_0565 might modulate the activity of the CRISPR systems on different levels.

The contribution highlights the use of Landsat spectral-temporal metrics (STMs) for the detection of surface anomalies that are potentially related to buried near-surface paleogeomorphological deposits in the Nile Delta (Egypt), in particular for a buried river branch close to Buto. The processing was completed in the Google Earth Engine (GEE) for the entire Nile Delta and for selected seasons of the year (summer/winter) using Landsat data from 1985 to 2019. We derived the STMs of the tasseled cap transformation (TC), the Normalized Difference Wetness Index (NDWI), and the Normalized Difference Vegetation Index (NDVI). These features were compared to historical topographic maps of the Survey of Egypt, CORONA imagery, the digital elevation model of the TanDEM-X mission, and modern high-resolution satellite imagery. The results suggest that the extent of channels is best revealed when differencing the median NDWI between summer (July/August) and winter (January/February) seasons (ΔNDWI). The observed difference is likely due to lower soil/plant moisture during summer, which is potentially caused by coarser-grained deposits and the morphology of the former levee. Similar anomalies were found in the immediate surroundings of several Pleistocene sand hills (“geziras”) and settlement mounds (“tells”) of the eastern delta, which allowed some mapping of the potential near-surface continuation. Such anomalies were not observed for the surroundings of tells of the western Nile Delta. Additional linear and meandering ΔNDWI anomalies were found in the eastern Nile Delta in the immediate surroundings of the ancient site of Bubastis (Tell Basta), as well as several kilometers north of Zagazig. These anomalies might indicate former courses of Nile river branches. However, the ΔNDWI does not provide an unambiguous delineation.
The monitoring of land cover and land use change is critical for assessing the provision of ecosystem services. One of the sources for long-term land cover change quantification is through the classification of historical and/or current maps. Little research has been done on historical maps using Object-Based Image Analysis (OBIA). This study applied an object-based classification using eCognition tool for analyzing the land cover based on historical maps in the Main river catchment, Upper Franconia, Germany. This allowed land use change analysis between the 1850s and 2015, a time span which covers the phase of industrialization of landscapes in central Europe. The results show a strong increase in urban area by 2600%, a severe loss of cropland (−24%), a moderate reduction in meadows (−4%), and a small gain in forests (+4%). The method proved useful for the application on historical maps due to the ability of the software to create semantic objects. The confusion matrix shows an overall accuracy of 82% for the automatic classification compared to manual reclassification considering all 17 sample tiles. The minimum overall accuracy was 65% for historical maps of poor quality and the maximum was 91% for very high-quality ones. Although accuracy is between high and moderate, coarse land cover patterns in the past and trends in land cover change can be analyzed. We conclude that such long-term analysis of land cover is a prerequisite for quantifying long-term changes in ecosystem services.
Multidrug‐resistant bacteria represent one of the biggest challenges facing modern medicine. The increasing prevalence of glycopeptide resistance compromises the efficacy of vancomycin, for a long time considered as the last resort for the treatment of resistant bacteria. To reestablish its activity, polycationic peptides were conjugated to vancomycin. By site‐specific conjugation, derivatives that bear the peptide moiety at four different sites of the antibiotic were synthesized. The most potent compounds exhibited an approximately 1000‐fold increased antimicrobial activity and were able to overcome the most important types of vancomycin resistance. Additional blocking experiments using d‐Ala‐d‐Ala revealed a mode of action beyond inhibition of cell‐wall formation. The antimicrobial potential of the lead candidate FU002 for bacterial infection treatments could be demonstrated in an in vivo study. Molecular imaging and biodistribution studies revealed that conjugation engenders superior pharmacokinetics.
The stability of Trp in pure solutions and in parenteral AA formulations was evaluated with regard to typically used manufacturing processes, storage conditions and primary packaging. Therefore, thorough stability studies on Trp solutions were conducted beforehand. The applied stressing method, i.e. steam sterilization by autoclave, are chemically seen relatively mild but showed to be efficient to induce Trp degradation in the presence of oxygen. Subsequent identification, separation and characterization were challenging due to similar substance properties, numerous stereoisomers and pairs of diastereomers found amongst them. However, the identified o-aminoacetophenone compounds, Kyn and NFK, are associated with photo reactivity and have photo-oxidizing properties. Thus, best possible protection from UV-light, together with strict oxygen expulsion, are the most important criteria to impede Trp degradation after autoclaving.
The identification of Trp degradation products was assisted by the compilation of a substance library, which included manifold reported and chemically plausible Trp degradation substances. The substances were classified for priority and their early or late-stage occurrence. The large number of possible substances and stereoisomers was narrowed down with the information retrieved from LC-UV/MS experiments. However, final identification was achieved by the synthesis of proposed substances as references. The following eight substances were characterized as Trp degradation substances: Kyn, NFK and three pairs of diastereomers R,R/R,S DiOia, R,R/R,S Oia and cis/trans PIC. Fig. 33 shows the proposed degradation pathway and demonstrates the close chemical relationship, which may be an explanation for the conversion of some substances into each other during the storage period. The proposed pathway brings together the results of different Trp stability and stressing studies, respectively [89, 94, 97, 98, 103, 133]. To our knowledge, the simultaneous formation of the identified degradation substances has not been reported before and especially not under the stressing conditions applied.
The application of a traditional RP-HPLC method was compared to two developed IP-HPLC methods and a RP-HPLC methods using a modified perfluorinated column. Orthogonal analyses methods and especially the combination of UV and MS detection are necessary in order to indicate potentially undetected degradation substances. Main evaluation criteria were the separation performance, analyses time, reproducibility and feasibility. The best results upon assessment of all Trp degradation products, in both; pure Trp solutions and pharmaceutical formulations, were obtained by a traditional RP-HPLC. The optimized method was validated according to ICH guidelines Q2(R1) and meets the criteria of a stability-indicating HPLC-UV method. The validated method has a sufficient separation performance with an adequate selectivity indicating the Trp degradation substances next to each other and next to other AAs in finished pharmaceutical formulations.
The detailed knowledge of Trp degradation and the method presented may be transferred practically to the pharmaceutical industry processing Trp-containing products. In general, the findings might contribute to the quality management of such pharmaceutical products during
manufacturing and storage. Additionally, the study results provide basic information for the establishment of an impurity consideration following the ICH guidelines Q3B (R2) (impurities in new drug products) for products containing Trp. However, further development of the method applying more sophisticated detectors or more potent HPLC techniques like e.g. UHPLC and the implication of more sensitive (MS) detectors like ToF-MS would be advantageous with regard to economic and practical aspects.
The effect of non-personalised tips on the continued use of self-monitoring mHealth applications

Chronic tinnitus, the perception of a phantom sound in the absence of corresponding stimulus, is a condition known to affect patients' quality of life. Recent advances in mHealth have enabled patients to maintain a ‘disease journal’ of ecologically-valid momentary assessments, improving patients' own awareness of their disease while also providing clinicians valuable data for research. In this study, we investigate the effect of non-personalised tips on patients' perception of tinnitus, and on their continued use of the application. The data collected from the study involved three groups of patients that used the app for 16 weeks. Groups A & Y were exposed to feedback from the start of the study, while group B only received tips for the second half of the study. Groups A and Y were run by different supervisors and also differed in the number of hospital visits during the study. Users of Group A and B underwent assessment at baseline, mid-study, post-study and follow-up, while users of group Y were only assessed at baseline and post-study. It is seen that the users in group B use the app for longer, and also more often during the day. The answers of the users to the Ecological Momentary Assessments are seen to form clusters where the degree to which the tinnitus distress depends on tinnitus loudness varies. Additionally, cluster-level models were able to predict new unseen data with better accuracy than a single global model. This strengthens the argument that the discovered clusters really do reflect underlying patterns in disease expression.
This paper is concerned with the rights of refugees. The refugee issue has been an acutely charged item on the political agenda for several years. Although the great waves of influx have flattened out, people are continually venturing into Europe. Europe’s handling of refugees has been subject to strong criticism, and the accusation that various actions contradict internationally agreed law is particularly serious. It remains a question of how to respond appropriately to the influx of people fearing for their lives. This paper examines empirically how young people from different denominations in Germany (n = 2022) and how Roman Catholics from 10 countries (n = 5363) evaluate refugee rights. It also investigates whether individual religiosity moderates the influence of denomination or national context. The results show that there are no significant differences between respondents from different denominations, but there are significant differences between respondents from different countries. However, religiosity was not found to moderate the influence of denomination or national context. These findings suggest that attitudes towards refugee rights depend more on the national context in which people live rather than on their religious affiliation or individual religiosity.
Objective
The biological interpretation of gene expression measurements is a challenging task. While ordination methods are routinely used to identify clusters of samples or co-expressed genes, these methods do not take sample or gene annotations into account. We aim to provide a tool that allows users of all backgrounds to assess and visualize the intrinsic correlation structure of complex annotated gene expression data and discover the covariates that jointly affect expression patterns.
Results
The Bioconductor package covRNA provides a convenient and fast interface for testing and visualizing complex relationships between sample and gene covariates mediated by gene expression data in an entirely unsupervised setting. The relationships between sample and gene covariates are tested by statistical permutation tests and visualized by ordination. The methods are inspired by the fourthcorner and RLQ analyses used in ecological research for the analysis of species abundance data, that we modified to make them suitable for the distributional characteristics of both, RNA-Seq read counts and microarray intensities, and to provide a high-performance parallelized implementation for the analysis of large-scale gene expression data on multi-core computational systems. CovRNA provides additional modules for unsupervised gene filtering and plotting functions to ensure a smooth and coherent analysis workflow.
Water crises are becoming severe in recent times, further fueled by population increase and climate change. They result in complex and unsustainable water management. Spatial estimation of consumptive water use is vital for performance assessment of the irrigation system using Remote Sensing (RS). For this study, its estimation is done using the Soil Energy Balance Algorithm for Land (SEBAL) approach. Performance indicators including equity, adequacy, and reliability were worked out at various spatiotemporal scales. Moreover, optimization and sustainable use of water resources are not possible without knowing the factors mainly influencing consumptive water use of major crops. For that purpose, random forest regression modelling was employed using various sets of factors for site-specific, proximity, and cropping system. The results show that the system is underperforming both for Kharif (i.e., summer) and Rabi (i.e., winter) seasons. Performance indicators highlight poor water distribution in the system, a shortage of water supply, and unreliability. The results are relatively good for Rabi as compared to Kharif, with an overall poor situation for both seasons. Factors importance varies for different crops. Overall, distance from canal, road density, canal density, and farm approachability are the most important factors for explaining consumptive water use. Auditing of consumptive water use shows the potential for resource optimization through on-farm water management by the targeted approach. The results are based on the present situation without considering future changes in canal water supply and consumptive water use under climate change.
Frühe entzündliche Vorgänge scheinen eine große Rolle in der Entstehung der globalen Hirnschädigung in der Frühphase nach einer Subarachnoidalblutung (SAB) zu spielen.
Ziel der vorliegenden Arbeit war es den Effekt der anti-inflammatorischen Medikamente Methylprednisolon und Minozyklin auf die Gehirndurchblutung und frühe Hirnschädigung nach SAB zu untersuchen.
Hierzu wurde ein randomisiertes und kontrolliertes Tierexperiment durchgeführt. Mit Hilfe des endovaskulären Perforationsmodells wurde bei männlichen Sprague-Dawley-Ratten eine SAB ausgelöst. Den Tieren wurde 30 Minuten nach Auftreten der SAB Methylprednisolon, Minozyklin oder Kochsalzlösung intraperitoneal verabreicht.
Sowohl Methylprednisolon als auch Minozyklin verminderten den Anteil Caspase 3 positiver Zellen in immunhistochemischen Färbungen der Hippocampie der Versuchstiere signifikant. In Bezug auf die klinische Untersuchung, den intrakraniellen Druck und die Hirndurchblutung der Ratten ergaben sich keine signifikanten Unterschiede zwischen den Versuchsgruppen. Die Ergebnisse suggerieren, dass Methylprednisolon und Minozyklin den akuten Zellschaden nach SAB reduzieren. Daher könnten sich beide Mittel als geeignet für die Therapie der „Early Brain Injury“ nach SAB erweisen. Weitere Studien zum besseren Verständnis der zugrunde liegenden Wirkmechanismen von Methylprednisolon und Minozyklin auf die Frühphase der SAB sind nötig.
The prevalence of cardiovascular diseases (CVD) increases dramatically with age. Nevertheless, most of the basic research in cardiology has been conducted on young healthy animals which may not necessarily reflect the situation observed in the clinic. The heart undergoes profound changes in elderly, including molecular alterations, myocardial hypertrophy, interstitial fibrosis and functional decline. To date, numerous approaches exist to explain mechanisms of the cardiac aging process whereupon inflammation and immune activity are of increasing interest. Myocardial aging is temporally associated with chronic low-grade systemic inflammation and accumulation of memory T-cells. However, a possible causal relationship between these two phenomena has not yet been investigated. Thus, aim of the present study was to assess how immunological mechanisms contribute to the myocardial aging process.
Herein, the healthy murine heart was found to harbor all major resident leukocyte populations, including macrophages (CD45+CD11b+Ly6G-), granulocytes (CD45+ CD11b+Ly6G+), T-cells (CD45+CD11b-CD3e+), B-cells (CD45+CD11b-B220+) at frequencies that largely surpass those found in skeletal muscles. Age-related structural alterations and functional impairment occur simultaneously with significant shifts of the tissue resident leukocyte composition. Gene expression analyses performed on bulk myocardial samples revealed higher expression levels of TNF and INF- suggesting that in situ inflammation plays a role in the myocardial aging process. Aging was furthermore accompanied by a significant increase in size and cellularity of mediastinal, heart draining lymph nodes (med LN). Moreover, the med LNs harvested from aged mice showed a strong accumulation of effector-memory T-cells (CD44+CD62L-), mainly exhibiting a pro-inflammatory phenotype (Foxp3-, TNF+, IFN- γ+). None of these alterations were observed in popliteal lymph nodes of aged mice, indicating that they might be site-specific.
Next, to go beyond mere associative evidence and examine underlying mechanisms, the myocardial aging process was comprehensively characterized in mice lacking B- (µMT) or CD4+ T-cells (CD4ko). Our analyses revealed that aged CD4+ T-cell-deficient, but not B-cell-deficient mice, exhibit a lower in situ inflammatory tone and preserved ventricular function, as compared to age-matched wild type controls. No differences in the expression levels of genes related to fibrosis were observed in the groups.
Taken together, the results of this study indicate that heart-directed immune responses may spontaneously arise in the elderly, even in the absence of a clear tissue damage or concomitant infection. The T-cell-mediated immunosenescence profile might be particularly associated with age-related myocardial inflammation and functional decline, but not with tissue remodeling. These observations might shed new light on the emerging role of T cells in myocardial diseases, which primarily affect the elderly population.
Fabry Disease (FD) is a rare, X-linked, lysosomal storage disease that mainly causes renal, cardiac and cerebral complications. Enzyme replacement therapy (ERT) with recombinant alpha-galactosidase A is available, but approximately 50% of male patients with classical FD develop inhibiting anti-drug antibodies (iADAs) that lead to reduced biochemical responses and an accelerated loss of renal function. Once immunization has occurred, iADAs tend to persist and tolerization is hard to achieve. Here we developed a pre-treatment prediction model for iADA development in FD using existing data from 120 classical male FD patients from three European centers, treated with ERT. We found that nonsense and frameshift mutations in the α-galactosidase A gene (p = 0.05), higher plasma lysoGb3 at baseline (p < 0.001) and agalsidase beta as first treatment (p = 0.006) were significantly associated with iADA development. Prediction performance of a Random Forest model, using multiple variables (AUC-ROC: 0.77) was compared to a logistic regression (LR) model using the three significantly associated variables (AUC-ROC: 0.77). The LR model can be used to determine iADA risk in individual FD patients prior to treatment initiation. This helps to determine in which patients adjusted treatment and/or immunomodulatory regimes may be considered to minimize iADA development risk.
S. stercoralis is a helminthic parasite which is common in tropical and subtropical regions. It causes a persistent but often inapparent infection in humans. In the state of a protracted immunosuppression this parasite can cause a life-threatening hyperinfection syndrome. Most often the hyperinfection syndrome was found after prolonged high dose corticosteroid treatment. In HIV-infected individuals high dose corticosteroids are used for the treatment of the immune reconstitution inflammatory syndrome (IRIS) or as adjunct treatment in the treatment of meningeal or pericardial tuberculosis. Case reports from Tanzania demonstrate that Strongyloidiasis is prevalent not only in coastal regions but also in the Lake province of Tanzania. However, data on the local prevalence of S. stercoralis infection based on sensitive techniques are scanty, especially in HIV-infected individuals.
The main objective of this study is to provide data on the prevalence of S. stercoralis infections in the adult HIV-infected population attending the Bugando Medical Centre for medical care. Specific objectives of the study are the comparison of the sensitivities and specificities of five different methods in detecting S. stercoralis. Four methods to detect S. stercoralis larvae used stool samples; one method to detect S. stercoralis antibodies required blood samples. The study used the Agar-plate-culture-technique and a modified Harada-Mori-culture-technique for the direct detection of helminthic larvae in the collected faecal samples. In addition, a recently described PCR-assay from faecal specimens and an ELISA for S. stercoralis antibodies have been applied. The Faecal Parasite Concentrator (FPC) stool concentration technique was used for the differential diagnosis of other intestinal helminthic parasites. The results of the study may influence the current treatment guidelines for HIV-infected patients in case that a relevant prevalence of S. stercoralis infection is found. Then, prior to a prolonged iatrogenic immunosuppression -like the high dose corticosteroid treatment for IRIS- a prophylactic anthelminthic treatment capable to eradicate a S. stercoralis infection could be recommendable. The prevalence of a current S. stercoralis infection using the PCR as a gold standard was 5.4%. The Agar plate method showed positive results in 19 out of 278 cases (6.1%), the modified Harada Mori technique in 13 of 278 (4.7%) cases. With PCR as gold standard the sensitivity of the agar plate method was 60%, the positive predictive value 47.4%, the specificity 96.2% and the negative predictive value 97.7 %. The sensitivity of the Harada Mori technique was 36.4%, the positive predictive value 30.7% with a specificity of 96.4% and negative predictive value 97.1%. The modified Harada Mori technique allowed in principal the morphological identification of nematode larvae. Microscopic analysis showed a specificity of 100% and a sensitivity of 46.7%. Antibodies were detected in 45 of 278 cases 16.2% by ELISA, with a sensitivity of 92.9% and a specificity of 87.8%. The findings of this study show that none of the diagnostic tests can be implemented as a routine diagnostic procedure to diagnose a current infection. This leads to the conclusion that it is high time to consider the provision of a prophylactic treatment within patients who are either HIV positive patients who could develop an IRIS after receiving ART, patients with a HTLV-1 infection and the growing number of patients under iatrogenic immunosuppression for various reasons.
Elementary building blocks for quantum repeaters based on fiber channels and memory stations are analyzed. Implementations are considered for three different physical platforms, for which suitable components are available: quantum dots, trapped atoms and ions, and color centers in diamond. The performances of basic quantum repeater links for these platforms are evaluated and compared, both for present‐day, state‐of‐the‐art experimental parameters as well as for parameters that can in principle be reached in the future. The ultimate goal is to experimentally explore regimes at intermediate distances—up to a few 100 km—in which the repeater‐assisted secret key transmission rates exceed the maximal rate achievable via direct transmission. Two different protocols are considered, one of which is better adapted to the higher source clock rate and lower memory coherence time of the quantum dot platform, while the other circumvents the need of writing photonic quantum states into the memories in a heralded, nondestructive fashion. The elementary building blocks and protocols can be connected in a modular form to construct a quantum repeater system that is potentially scalable to large distances.
Adenosine receptor ligands: coumarin−chalcone hybrids as modulating agents on the activity of hARs

Adenosine receptors (ARs) play an important role in neurological and psychiatric disorders such as Alzheimer's disease, Parkinson's disease, epilepsy and schizophrenia. The different subtypes of ARs and the knowledge on their densities and status are important for understanding the mechanisms underlying the pathogenesis of diseases and for developing new therapeutics. Looking for new scaffolds for selective AR ligands, coumarin–chalcone hybrids were synthesized (compounds 1–8) and screened in radioligand binding (hA\(_1\), hA\(_{2A}\) and hA\(_3\)) and adenylyl cyclase (hA\(_{2B}\)) assays in order to evaluate their affinity for the four human AR subtypes (hARs). Coumarin–chalcone hybrid has been established as a new scaffold suitable for the development of potent and selective ligands for hA\(_1\) or hA\(_3\) subtypes. In general, hydroxy-substituted hybrids showed some affinity for the hA\(_1\), while the methoxy counterparts were selective for the hA\(_3\). The most potent hA\(_1\) ligand was compound 7 (K\(_i\) = 17.7 µM), whereas compound 4 was the most potent ligand for hA\(_3\) (K\(_i\) = 2.49 µM). In addition, docking studies with hA\(_1\) and hA\(_3\) homology models were established to analyze the structure–function relationships. Results showed that the different residues located on the protein binding pocket could play an important role in ligand selectivity.
In vorliegender Dissertation wurden die Langzeitergebnisse hinsichtlich der Lebensqualität der Patienten, die sich im Zeitraum 2002-2010 eine proximale Humerusfraktur zuzogen und mittels intramedullärer Drahtosteosynthese nach Kapandji im König Ludwig Haus in Würzburg operiert wurden, untersucht. Die Nachuntersuchung erfolgte mittels klinischer, radiologischer und sonografischer Untersuchung sowie durch Verwendung des DASH-Fragebogens, Constant Murley Score und der Visuellen Analogskala.
Reduced function of the serotonin transporter (SERT) is associated with increased susceptibility to anxiety and depression and with type-2 diabetes, which is especially true in older women. Preference for a “Western diet” (WD), enriched with saturated fat, cholesterol, and sugars, may aggravate these conditions. In previous studies, decreased glucose tolerance, central and peripheral inflammation, dyslipidemia, emotional, cognitive, and social abnormalities were reported in WD-fed young female mice. We investigated the metabolic, molecular, and behavioral changes associated with a 3-week-long dietary regime of either the WD or control diet in 12-month-old female mice with three different Sert genotypes: homozygous (Slc6a4) gene knockout (Sert\(^{−/−}\): KO), heterozygous (Sert\(^{+/−}\): HET), or wild-type mice (Sert\(^{+/+}\): WT). In the WT-WD and KO-WD groups, but not in HET-WD-fed mice, most of changes induced by the WD paralleled those found in the younger mice, including brain overexpression of inflammatory marker Toll-like receptor 4 (Tlr4) and impaired hippocampus-dependent performance in the marble test. However, the 12-month-old female mice became obese. Control diet KO mice exhibited impaired hippocampal-dependent behaviors, increased brain expression of the serotonin receptors Htr2c and Htr1b, as well as increased Tlr4 and mitochondrial regulator, peroxisome proliferator-activated receptor gamma-coactivator-1a (Ppargc1a). Paradoxically, these, and other changes, were reversed in KO-WD mutants, suggesting a complex interplay between Sert deficiency and metabolic factors as well as potential compensatory molecular mechanisms that might be disrupted by the WD exposure. Most, but not all, of the changes in gene expression in the brain and liver of KO mice were not exhibited by the HET mice fed with either diet. Some of the WD-induced changes were similar in the KO-WD and HET-WD-fed mice, but the latter displayed a “rescued” phenotype in terms of diet-induced abnormalities in glucose tolerance, neuroinflammation, and hippocampus-dependent performance. Thus, complete versus partial Sert inactivation in aged mice results in distinct metabolic, molecular, and behavioral consequences in response to the WD. Our findings show that Sert\(^{+/−}\) mice are resilient to certain environmental challenges and support the concept of heterosis as evolutionary adaptive mechanism.
Nectar is crucial to maintain plant-pollinator mutualism. Nectar quality (nutritional composition) can vary strongly between individuals of the same plant species. The factors driving such inter-individual variation have however not been investigated closer. We investigated nectar quality of field scabious, Knautia arvensis in different grassland plant communities varying in species composition and richness to assess whether nectar quality can be affected by the surrounding plant community. We analyzed (with high performance liquid chromatography) the content of carbohydrates, overall amino acids, and essential amino acids. Amino acid and carbohydrate concentrations and proportions varied among plant individuals and with the surrounding plant community but were not related to the surrounding plant species richness. Total and individual carbohydrate concentrations were lowest, while proportions of the essential amino acids, valine, isoleucine, leucine (all phagostimulatory), and lysine were highest in plant species communities of the highest diversity. Our results show that K. arvensis nectar chemistry varies with the composition of the surrounding plant community, which may alter the taste and nutritional value and thus affect the plant’s visitor spectrum and visitation rate. However, the strong inter-individual variation in nectar quality requires additional studies (e.g., in semi-field studies) to disentangle different biotic and abiotic factors contributing to inter-individual nectar chemistry in a plant-community context.
Background and Objectives
To analyze the impact of humidity and temperature on excimer laser ablation of polyethylene terephthalate (PET), polymethylmethacrylate (PMMA) and porcine corneal tissue, and an ablation model to compensate for the temperature and humidity changes on ablation efficiency.
Study Design/Materials and Methods
The study was conducted using an AMARIS 1050RS (Schwind eye‐tech‐solutions) placed inside a climate chamber at ACTS. Ablations were performed on PET, PMMA, and porcine cornea. The impact of a wide range of temperature (~18°C to ~30°C) and relative humidity (~25% to ~80%) on laser ablation outcomes was tested using nine climate test settings. For porcine eyes, change in defocus was calculated from the difference of post‐ablation to pre‐ablation average keratometry readings. Laser scanning deflectometry was performed to measure refractive change achieved in PMMA. Multiple linear regression was performed using the least square method with predictive factors: temperature, relative humidity, time stamp. Influence of climate settings was modeled for pulse energy, pulse fluence, ablation efficiency on PMMA and porcine cornea tissue.
Results
Temperature changes did not affect laser pulse energy, pulse fluence (PET), and ablation efficiency (on PMMA or porcine corneal tissue) significantly. Changes in relative humidity were critical and significantly affected laser pulse energy, high fluence and low fluence. The opposite trend was observed between the ablation performance on PMMA and porcine cornea.
Conclusions
The proposed well‐fitting multi‐linear model can be utilized for compensation of temperature and humidity changes on ablation efficiency. Based on this model, a working window for optimum operation has been found (temperature 18°C to 28°C and relative humidity 25% to 65%) for a maximum deviation of ±2.5% in ablation efficiency in PMMA and porcine corneal tissue.
Endophytes live in partial symbiosis inside a plant and have been detected in all tested plants. They belong to the group of fungi or bacteria and their ecological function is mostly unknown. The fungal endophytes of the genus Epichloë belong to a special group of endophytes. Epichloë endophytes live symbiotically inside cool season grass species and some of them are able to produce alkaloids toxic to vertebrates and insects. Their symbiosis is seen as mutualistic for the following reasons: the fungus provides the plant herbivore resistance by producing alkaloids, and it increases the plant’s drought tolerance as well as its biomass production. In return, the grass provides the fungus shelter, nutrients and dispersal. Epichloë endophytes are host specific and the ability to produce alkaloids differs between species. In order to estimate intoxication risks in grasslands, it is necessary to detect infection rates of different grass species with Epichloë endophytes, and to determine the genotypes and chemotypes of the Epichloë species as well as the produced alkaloid concentrations. Factors like land-use intensity or season may have an influence on infection rates and alkaloid concentrations. Also, different methodological approaches may lead to different results. In this doctoral thesis my general aim was to evaluate intoxication risks in German grasslands caused by Epichloë endophytes. For that I investigated infection rates of different grass species and the genotypes and chemotypes of their Epichloë endophytes in German grasslands (Chapter II). Furthermore, I compared alkaloid concentrations detected with dry and fresh plant weight and different analytical methods. I also detected possible changes on the influence of season or land-use intensity (Chapter III). Additionally, I examined infections with Epichloë endophytes and alkaloid concentrations in commercially available grass seed mixtures and determined how that influences the intoxication risk of grazing animals in Europe (Chapter IV).
It is of agricultural interest to estimate intoxication risks for grazing livestock on German grasslands due to Epichloë infected grass species. Therefore, it is important to investigate which grasses are infected with the Epichloë endophyte, if the endophytes have the ability to produce vertebrate and invertebrate toxic alkaloids and if the alkaloids are indeed produced. I showed that Epichloë festucae var. lolii infecting agriculturally important Lolium perenne lacked the starting gene for ergovaline biosynthesis. Hence, vertebrate toxic ergovaline was not detected in the majority of the collected L. perenne plants. The detection of alkaloid concentrations is an important tool to estimate intoxication risk for vertebrates, but also invertebrates. My studies showed that the usage of dry plant material is crucial to quantify the correct alkaloid concentrations, and that alkaloid concentrations can vary depending on the detection method. Hence, the usage of validated, similar detection methods is important to be able to compare alkaloid concentrations from different studies. Nevertheless, the trends of seasonal changes and the influence of land-use intensity stayed the same, regardless if dry or fresh plant weight was used. Also, alkaloid concentrations were below toxicity thresholds on population level, regardless of the method used. Two commercially available forage grass and two commercially available turf grass seed mixtures were infected with Epichloë endopyhtes and alkaloids were detected. This might contribute to the spreading of Epichloë endopyhtes in Germany, therefore seed mixtures should be tested for Epichloë infections. My results indicate that the intoxication risk is generally low in Germany at the moment, although that might change due to climate change, an increase of monocultural land-use, or the seeding of Epichloë infected grass seeds.
Mycotoxins in agriculturally used plants can cause intoxication in animals and can lead to severe financial losses for farmers. The endophytic fungus Epichloë festucae var. lolii living symbiotically within the cool season grass species Lolium perenne can produce vertebrate and invertebrate toxic alkaloids. Hence, an exact quantitation of alkaloid concentrations is essential to determine intoxication risk for animals. Many studies use different methods to detect alkaloid concentrations, which complicates the comparability. In this study, we showed that alkaloid concentrations of individual plants exceeded toxicity thresholds on real world grasslands in Germany, but not on the population level. Alkaloid concentrations on five German grasslands with high alkaloid levels peaked in summer but were also below toxicity thresholds on population level. Furthermore, we showed that alkaloid concentrations follow the same seasonal trend, regardless of whether plant fresh or dry weight was used, in the field and in a common garden study. However, alkaloid concentrations were around three times higher when detected with dry weight. Finally, we showed that alkaloid concentrations can additionally be biased to different alkaloid detection methods. We highlight that toxicity risks should be analyzed using plant dry weight, but concentration trends of fresh weight are reliable.
Our body is colonized by a vast array of bacteria the sum of which forms our microbiota. The gut alone harbors >1,000 bacterial species. An understanding of their individual or synergistic contributions to human health and disease demands means to interfere with their functions on the species level. Most of the currently available antibiotics are broad‐spectrum, thus too unspecific for a selective depletion of a single species of interest from the microbiota. Programmable RNA antibiotics in the form of short antisense oligonucleotides (ASOs) promise to achieve precision manipulation of bacterial communities. These ASOs are coupled to small peptides that carry them inside the bacteria to silence mRNAs of essential genes, for example, to target antibiotic‐resistant pathogens as an alternative to standard antibiotics. There is already proof‐of‐principle with diverse bacteria, but many open questions remain with respect to true species specificity, potential off‐targeting, choice of peptides for delivery, bacterial resistance mechanisms and the host response. While there is unlikely a one‐fits‐all solution for all microbiome species, I will discuss how recent progress in bacterial RNA biology may help to accelerate the development of programmable RNA antibiotics for microbiome editing and other applications.
The enrichment of deadwood is essential for the conservation of saproxylic biodiversity in managed forests. However, existing strategies focus on a cost‐intensive increase of deadwood amount, while largely neglecting increasing deadwood diversity.
Deadwood objects, that is logs and branches, from six tree species were experimentally sun exposed, canopy shaded and artificially shaded for 4 years, after which the alpha‐, beta‐ and gamma‐diversity of saproxylic beetles, wood‐inhabiting fungi and spiders were analysed. Analyses of beta‐diversity included the spatial distance between exposed deadwood objects. A random‐drawing procedure was used to identify the combination of tree species and sun exposure that yielded the highest gamma‐diversity at a minimum of exposed deadwood amount.
In sun‐exposed plots, species numbers in logs were higher than in shaded plots for all taxa, while in branches we observed the opposite for saproxylic beetles. Tree species affected the species numbers only of saproxylic beetles and wood‐inhabiting fungi. The beta‐diversity of saproxylic beetles and wood‐inhabiting fungi among logs was influenced by sun exposure and tree species, but beta‐diversity of spiders by sun exposure only. For all saproxylic taxa recorded in logs, differences between communities increased with increasing spatial distance.
A combination of canopy‐shaded Carpinus logs and sun‐exposed Populus logs resulted in the highest species numbers of all investigated saproxylic taxa among all possible combinations of tree species and sun‐exposure treatments.
Synthesis and applications. We recommend incorporating the enrichment of different tree species and particularly the variation in sun exposure into existing strategies of deadwood enrichment. Based on the results of our study, we suggest to combine the logs of softwood broadleaf tree species (e.g. Carpinus, Populus), hardwood broadleaf tree species (e.g. Quercus) and coniferous tree species (e.g. Pinus) under different conditions of sun exposure and distribute them spatially in a landscape to maximize the beneficial effects on overall diversity.
Background
Hypophosphatasia (HPP) is a rare, inherited metabolic disorder caused by loss-of-function mutations in the ALPL gene that encodes the tissue-nonspecific alkaline phosphatase TNAP (ORPHA 436). Its clinical presentation is highly heterogeneous with a remarkably wide-ranging severity. HPP affects patients of all ages. In children HPP-related musculoskeletal symptoms may mimic rheumatologic conditions and diagnosis is often difficult and delayed. To improve the understanding of HPP in children and in order to shorten the diagnostic time span in the future we studied the natural history of the disease in our large cohort of pediatric patients. This single centre retrospective chart review included longitudinal data from 50 patients with HPP diagnosed and followed at the University Children's Hospital Wuerzburg, Germany over the last 25 years.
Results
The cohort comprises 4 (8%) perinatal, 17 (34%) infantile and 29 (58%) childhood onset HPP patients. Two patients were deceased at the time of data collection. Diagnosis was based on available characteristic clinical symptoms (in 88%), low alkaline phosphatase (AP) activity (in 96%), accumulating substrates of AP (in 58%) and X-ray findings (in 48%). Genetic analysis was performed in 48 patients (31 compound heterozygous, 15 heterozygous, 2 homozygous mutations per patient), allowing investigations on genotype-phenotype correlations. Based on anamnestic data, median age at first clinical symptoms was 3.5 months (min. 0, max. 107), while median time to diagnosis was 13 months (min. 0, max. 103). Common symptoms included: impairment of motor skills (78%), impairment of mineralization (72%), premature loss of teeth (64%), musculoskeletal pain and craniosynostosis (each 64%) and failure to thrive (62%). Up to now 20 patients started medical treatment with Asfotase alfa.
Conclusions
Reported findings support the clinical perception of HPP being a chronic multi-systemic disease with often delayed diagnosis. Our natural history information provides detailed insights into the prevalence of different symptoms, which can help to improve and shorten diagnostics and thereby lead to an optimised medical care, especially with promising therapeutic options such as enzyme-replacement-therapy with Asfotase alfa in mind.
Die Hypophosphatasie (HPP) ist eine seltene, angeborene Knochen- und Systemerkrankung, welche Patienten allen Alters betrifft. Verursacht wird die Erkrankung durch Mutationen im ALPL-Gen, welches für die gewebeunspezifische Alkalische Phosphatase codiert und mit einem Funktionsverlust des Enzyms einhergeht. Die Ausprägung der klinischen Symptomatik ist sehr heterogen und reicht von milden und unspezifischen bis hin zu potenziell lebensbedrohlichen Symptomen, was die korrekte Diagnose zusätzlich erschwert und verzögert. Um das Verständnis der pädiatrischen HPP zu verbessern und die Dauer von Symptombeginn bis zur korrekten Diagnose zu verkürzen, haben wir den Verlauf der Erkrankung anhand einer retrospektiven Aufarbeitung der Daten von 50 pädiatrischen HPP Patienten, die in den letzten 25 Jahren an der Universitäts-Kinderklinik in Würzburg vorstellig waren, untersucht.
Diese Ergebnisse bestätigen den klinischen Eindruck der HPP als chronische Systemerkrankung, welche aufgrund ihrer unspezifischen klinischen Präsentation oftmals nur mit zeitlicher Verzögerung diagnostiziert wird. Dieser Verzögerung kommt insbesondere im Hinblick auf die 2015 zur Behandlung der pädiatrischen HPP zugelassenen Enzymersatztherapie mit dem Wirkstoff Asfotase alfa eine besondere Bedeutung zu, da die Patienten von einer frühzeitigen Diagnose und einem damit einhergehenden frühzeitigen Beginn der Behandlung profitieren können.
Diese Ergebnisse tragen einen Teil dazu bei, das Bewusstsein und die Kenntnis der Erkrankung zu verbessern, um so die die Zeitspanne zwischen Symptombeginn und Diagnosestellung zu verkürzen und die medizinische Versorgung der Patienten zu verbessern.
Im Rahmen dieser Arbeit wurden die elektronischen Eigenschaften von Graphen auf Metalloberflächen mittels Rastertunnelmikroskopie und Quasiteilcheninterferenz (englisch quasiparticle interference, QPI)-Messungen untersucht. Durch das Verwenden schwerer Substrate sollte die Spin-Bahn-Wechselwirkung des Graphen verstärkt werden und damit eine Bandlücke am K-Punkt der Bandstruktur mittels QPI beobachtet werden. Um das Messen von QPI auf Graphen zu testen, wurde auf der Oberfläche eines SiC(0001)-Kristalls durch Erhitzen Graphen erzeugt und mit dem Rastertunnelmikroskop untersucht. Dieses System wurde schon ausführlich in der Literatur beschrieben und bereits bekannte QPI-Messungen von Streuringen, die auf den Dirac-Kegeln des Graphen am K-Punkt basieren, konnte ich auf gr/SiC(0001) in guter Qualität erfolgreich reproduzieren. Anschließend wurde Graphen nach einem wohlbekannten Verfahren durch Aufbringen von Ethylen auf ein erhitztes Ir(111)-Substrat erzeugt. Dieses gr/Ir(111)-System diente auch als Grundlage für Interkalationsversuche von Bismut (gr/Bi/Ir(111)) und Gadolinium (gr/Gd/Ir(111)) zwischen das Graphen und das Substrat. Auf gr/Bi/Ir(111) wurde ein schon aus der Literatur bekanntes Netzwerk aus Versetzungslinien beobachtet, dem zusätzlich eine Temperaturabhängigkeit nachgewiesen werden konnte. Beim Versuch, Gadolinium zu interkalieren, wurden zwei verschieden Oberflächenstrukturen beobachtet, die auf eine unterschiedlich Anordnung bzw. Menge des interkalierten Gadoliniums zurückzuführen sein könnten. Auf keinem dieser drei Systeme konnten allerdings Streuringe mittels QPI beobachtet werden. Als Vorbereitung der Interkalation von Gadolinium wurden dessen Wachstum und magnetische Eigenschaften auf einem W(110)-Kristall untersucht. Dabei konnte eine aus der Literatur bekannte temperaturabhängige Austauschaufspaltung reproduziert werden. Darüber hinaus konnten sechs verschieden magnetische Domänen beobachtet werden. Zusätzlich sind auf der Oberfläche magnetische Streifen auszumachen, die möglicherweise auf einer Spinspirale basieren. Als Grundlage für die mögliche zukünftige Erzeugung Graphen-artiger Molekülgitter wurde das Wachstum von H-TBTQ und Me-TBTQ auf Ag(111) untersucht. Die Moleküle richten sich dabei nach der Oberflächenstruktur des Silber aus und bilden längliche Inseln, deren Kanten in drei Vorzugsrichtungen verlaufen. Auf H-TBTQ wurde zudem eine zweite, Windmühlen-artige Ausrichtung der Moleküle auf der Oberfläche beobachtet. Auf den mit den Molekülen bedeckten Stellen der Oberfläche wurde eine Verschiebung des Ag-Oberflächenzustands beobachtet, die mit einem Ladungstransfer vom Ag(111)-Substrat auf die TBTQ-Moleküle zu erklären sein könnte.
The muscarinic M\(_1\) acetylcholine receptor is an important drug target for the treatment of various neurological disorders. Designing M\(_1\) receptor-selective drugs has proven challenging, mainly due to the high conservation of the acetylcholine binding site among muscarinic receptor subtypes. Therefore, less conserved and topographically distinct allosteric binding sites have been explored to increase M\(_1\) receptor selectivity. In this line, bitopic ligands, which target orthosteric and allosteric binding sites simultaneously, may provide a promising strategy. Here, we explore the allosteric, M1-selective BQCAd scaffold derived from BQCA as a starting point for the design, synthesis, and pharmacological evaluation of a series of novel bitopic ligands in which the orthosteric moieties and linker lengths are systematically varied. Since β-arrestin recruitment seems to be favorable to therapeutic implication, all the compounds were investigated by G protein and β-arrestin assays. Some bitopic ligands are partial to full agonists for G protein activation, some activate β-arrestin recruitment, and the degree of β-arrestin recruitment varies according to the respective modification. The allosteric BQCAd scaffold controls the positioning of the orthosteric ammonium group of all ligands, suggesting that this interaction is essential for stimulating G protein activation. However, β-arrestin recruitment is not affected. The novel set of bitopic ligands may constitute a toolbox to study the requirements of β-arrestin recruitment during ligand design for therapeutic usage.
Ellagitannins are signature constituents of oak wood and their consumption has been associated with various health benefits. In vivo, they undergo metabolic degradation including gut microbial metabolism yielding urolithins. Only limited data is available about compounds being present in blood after intake of an extract from French oak wood, Robuvit®. In the course of a randomized, double-blind, controlled clinical investigation, 66 patients undergoing hysterectomy received placebo or 300 mg Robuvit® per day before and over 8 weeks after surgery. Serum and blood cell samples were analyzed by liquid chromatography electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS). The number of urolithin producers and the urolithin levels increased after intake of Robuvit®. In serum samples, the median concentration of urolithin A was 14.0 ng/ml [interquartile range (IQR) 57.4] after 8 weeks. Urolithin B was determined at 22.3 ng/ml (IQR 12.6), urolithin C at 2.66 ng/ml (IQR 2.08). In blood cells, lower concentrations and only urolithins A and B were detected. A statistically significant association of lower post-surgical pain scores with metabotype A was detected (p < 0.05). To conclude, supplementation with French oak wood extract raised urolithin generation in patients and suggested health advantages for urolithin-producers.
Maintenance of tumor vasculature integrity is indispensable for tumor growth and thus affects tumor progression. Previous studies have identified platelets as major regulators of tumor vascular integrity, as their depletion selectively renders tumor vessels highly permeable, causing massive intratumoral hemorrhage. While these results establish platelets as potential targets for anti-tumor therapy, depletion is not a treatment option due to the essential role of platelets for hemostasis. This thesis demonstrates for the first time that functional inhibition of glycoprotein (GP) VI on the platelet surface rapidly induces tumor hemorrhage and diminishes tumor growth similar to complete platelet depletion but without inducing systemic bleeding complications. Both, the intratumoral bleeding and tumor growth arrest could be reverted by depletion of Ly6G+ cells confirming them to be responsible for the induction of bleeding and necrosis within the tumor. In addition, GPVI inhibition increased intra-tumoral accumulation of co-administered chemotherapeutic agents, thereby resulting in a profound anti-tumor effect. In summary, this thesis manifests platelet GPVI as a key regulator of vascular integrity specifically in growing tumors, serving as a potential basis for the development of anti-tumor strategies.
In the second part of this thesis, light is shed on the modulating role of bridging integrator 2 (BIN2) in platelet Ca2+ signaling. Stromal interaction molecule 1 (STIM1) mediated store-operated calcium entry (SOCE) is the major route of Ca2+ influx in platelets, triggered by inositol trisphosphate receptor (IP3R)-dependent Ca2+ store release. In this thesis, the BAR domain superfamily member BIN2 was identified as the first Ca2+ signaling modulator, interacting with both, STIM1 and IP3R in platelets. Deletion of BIN2 resulted in reduced Ca2+ store release and Ca2+ influx in response to all tested platelet agonists. These defects were a consequence of impaired IP3R function in combination with defective STIM1-mediated SOC channel activation, while Ca2+ store content and agonist-induced IP3 production were unaltered. These results establish BIN2 as a central regulator of platelet Ca2+ signaling.
The third part of this thesis focuses on the effect of the soluble neuronal guidance protein Sema7A on platelet function. Rosenberger et al. discovered that Sema7A cleavage from red blood cells increases the formation of platelet-neutrophil complexes, thereby reinforcing thrombo-inflammation in myocardial ischemia-reperfusion injury (MIRI). This thesis establishes soluble Sema7A as a stimulator of platelet thrombus formation via its interaction with platelet GPIbα, thereby reinforcing PNC formation. Thus, interfering with the GPIb-Sema7A interaction during MIRI represents a potential strategy to reduce cardiac damage and improve clinical outcome following MI.
Insect microbiota plays an essential role on the hosts’ health and fitness, regulating their development, nutrition and immunity. The natural microbiota of bees, in particular, has been given much attention, largely because of the globally reported bee population declines. However, although the worker honey bee has been associated with distinctive and specialized microbiota, the microbiota of solitary bees has not been examined in detail, despite their enormous ecological importance. The main objectives of the present thesis were a) the bacterial community description for various solitary bee species, b) the association of the solitary bee microbiota with ecological factors such as landscape type, c) the relation of the bee foraging preferences with their nest bacterial microbiota, d) the examination of the nest building material contribution to the nest microbiota, e) the isolation of bacterial strains with beneficial or harmful properties for the solitary bee larvae and f) the pathological investigation of bacteria found in deceased solitary bee larvae.
The findings of the present study revealed a high bacterial biodiversity in the solitary bee nests. At the same time, the bacterial communities were different for each bee host species. Furthermore, it was shown that the pollen bacterial communities underwent compositional shifts reflecting a reduction in floral bacteria with progressing larval development, while a clear landscape effect was absent. The examination of the nest pollen provisions showed different foraging preferences for each included bee species. Both the pollen composition and the host species identity had a strong effect on the pollen bacteria, indicating that the pollen bacterial communities are the result of a combinatory process. The introduced environmental material also contributed to the nest natural microbiome. However, although the larval microbiota was significantly influenced by the pollen microbiota, it was not much associated with that of the nest material.
Two Paenibacillus strains isolated from O. bicornis nests showed strong antifungal activities, while several isolated strains were able to metabolize various oligosaccharides which are common in pollen and nectar. Screening for potential pathogenic bacteria in the nests of O. bicornis unveiled bacterial taxa, which dominated the bacterial community in deceased larvae, while at the same time they were undetectable in the healthy individuals.

Finally, larvae which were raised in vitro developed distinct bacterial microbiomes according to their diet, while their life span was affected.
The present thesis described aspects of the microbiota dynamics in the nests of seven megachilid solitary bee nests, by suggesting which transmission pathways shape the established bacterial communities and how these are altered with larval development. Furthermore, specific bacterial taxa were associated with possible services they might provide to the larvae, while others were related with possible harmful effects. Future studies should integrate microbiota examination of different bee generations and parallel investigation of the microbiota of the nests and their surrounding environment (plant community, soil) to elucidate the bacterial transmission paths which establish the nest microbiota of solitary bees. Functional assays will also allow future studies to characterize specific nest bacteria as beneficial or harmful and describe how they assist the development of healthy bees and the fitness of bee populations.
Solitary bees are subject to a variety of pressures that cause severe population declines. Currently, habitat loss, temperature shifts, agrochemical exposure, and new parasites are identified as major threats. However, knowledge about detrimental bacteria is scarce, although they may disturb natural microbiomes, disturb nest environments, or harm the larvae directly. To address this gap, we investigated 12 Osmia bicornis nests with deceased larvae and 31 nests with healthy larvae from the same localities in a 16S ribosomal RNA (rRNA) gene metabarcoding study. We sampled larvae, pollen provisions, and nest material and then contrasted bacterial community composition and diversity in healthy and deceased nests. Microbiomes of pollen provisions and larvae showed similarities for healthy larvae, whilst this was not the case for deceased individuals. We identified three bacterial taxa assigned to Paenibacillus sp. (closely related to P. pabuli/amylolyticus/xylanexedens), Sporosarcina sp., and Bacillus sp. as indicative for bacterial communities of deceased larvae, as well as Lactobacillus for corresponding pollen provisions. Furthermore, we performed a provisioning experiment, where we fed larvae with untreated and sterilized pollens, as well as sterilized pollens inoculated with a Bacillus sp. isolate from a deceased larva. Untreated larval microbiomes were consistent with that of the pollen provided. Sterilized pollen alone did not lead to acute mortality, while no microbiome was recoverable from the larvae. In the inoculation treatment, we observed that larval microbiomes were dominated by the seeded bacterium, which resulted in enhanced mortality. These results support that larval microbiomes are strongly determined by the pollen provisions. Further, they underline the need for further investigation of the impact of detrimental bacterial acquired via pollens and potential buffering by a diverse pollen provision microbiome in solitary bees.

Dazu werden zum einen bereits bekannte Anbindungsmöglichkeiten an Nanodiamant, wie zum Beispiel die Klick-Reaktion, sowie die Ausbildung von Amidbrücken verwendet. Zum anderen werden neuartige Funktionalisierungsmöglichkeiten wie Protein Ligation und Thioharnstoffbrücken verwendet und somit das Repertoire an bekannten Anbindungsreaktion erweitert.
Des weiteren wurde ein multifunktionales Nanodiamantsystem synthetisiert. Dieses ist in der Lage, zwei verschiedene Moleküle auf einem Partikel zu immobilisieren. Die verwendeten Methoden ermöglichen die Anbindung verschiedener Substanzen aus unterschiedlichen Molekülgruppen an Nanodiamanten und sind somit universell einsetzbar.
Purpose
To assess the impact of isocenter shifts due to linac gantry and table rotation during cranial stereotactic radiosurgery on D\(_{98}\), target volume coverage (TVC), conformity (CI), and gradient index (GI).
Methods
Winston‐Lutz (WL) checks were performed on two Elekta Synergy linacs. A stereotactic quality assurance (QA) plan was applied to the ArcCHECK phantom to assess the impact of isocenter shift corrections on Gamma pass rates. These corrections included gantry sag, distance of collimator and couch axes to the gantry axis, and distance between cone‐beam computed tomography (CBCT) isocenter and treatment beam (MV) isocenter. We applied the shifts via script to the treatment plan in Pinnacle 16.2. In a planning study, isocenter and mechanical rotation axis shifts of 0.25 to 2 mm were applied to stereotactic plans of spherical planning target volumes (PTVs) of various volumes. The shifts determined via WL measurements were applied to 16 patient plans with PTV sizes between 0.22 and 10.4 cm3.
Results
ArcCHECK measurements of a stereotactic treatment showed significant increases in Gamma pass rate for all three measurements (up to 3.8 percentage points) after correction of measured isocenter deviations. For spherical targets of 1 cm3, CI was most severely affected by increasing the distance of the CBCT isocenter (1.22 to 1.62). Gradient index increased with an isocenter‐collimator axis distance of 1.5 mm (3.84 vs 4.62). D98 (normalized to reference) dropped to 0.85 (CBCT), 0.92 (table axis), 0.95 (collimator axis), and 0.98 (gantry sag), with similar but smaller changes for larger targets. Applying measured shifts to patient plans lead to relevant drops in D\(_{98}\) and TVC (7%) for targets below 2 cm\(^3\) treated on linac 1.
Conclusion
Mechanical deviations during gantry, collimator, and table rotation may adversely affect the treatment of small stereotactic lesions. Adjustments of beam isocenters in the treatment planning system (TPS) can be used to both quantify their impact and for prospective correction of treatment plans.
The topical application of tranexamic acid (TXA) helps to prevent post-operative blood loss in total joint replacements. Despite these findings, the effects on articular and periarticular tissues remain unclear. Therefore, this in vitro study examined the effects of varying exposure times and concentrations of TXA on proliferation rates, gene expression and differentiation capacity of chondrocytes and human mesenchymal stromal cells (hMSCs), which underwent osteogenic differentiation. Chondrocytes and hMSCs were isolated and multiplied in monolayer cell cultures. Osteogenic differentiation of hMSCs was induced for 21 days using a differentiation medium containing specific growth factors. Cell proliferation was analyzed using ATP assays. Effects of TXA on cell morphology were examined via light microscopy and histological staining, while expression levels of tissue-specific genes were measured using semiquantitative RT-PCR. After treatment with 50 mg/mL of TXA, a decrease in cell proliferation rates was observed. Furthermore, treatment with concentrations of 20 mg/mL of TXA for at least 48 h led to a visible detachment of chondrocytes. TXA treatment with 50 mg/mL for at least 24 h led to a decrease in the expression of specific marker genes in chondrocytes and osteogenically differentiated hMSCs. No significant effects were observed for concentrations beyond 20 mg/mL of TXA combined with exposure times of less than 24 h. This might therefore represent a safe limit for topical application in vivo. Further research regarding in vivo conditions and effects on hMSC functionality are necessary to fully determine the effects of TXA on articular and periarticular tissues.
Background
While multiple in vitro studies examined mesenchymal stromal cells (MSCs) derived from bone marrow or hyaline cartilage, there is little to no data about the presence of MSCs in the joint capsule or the ligamentum capitis femoris (LCF) of the hip joint. Therefore, this in vitro study examined the presence and differentiation potential of MSCs isolated from the bone marrow, arthritic hyaline cartilage, the LCF and full-thickness samples of the anterior joint capsule of the hip joint.
Methods
MSCs were isolated and multiplied in adherent monolayer cell cultures. Osteogenesis and adipogenesis were induced in monolayer cell cultures for 21 days using a differentiation medium containing specific growth factors, while chondrogenesis in the presence of TGF-ss1 was performed using pellet-culture for 27 days. Control cultures were maintained for comparison over the same duration of time. The differentiation process was analyzed using histological and immunohistochemical stainings as well as semiquantitative RT-PCR for measuring the mean expression levels of tissue-specific genes.
Results
This in vitro research showed that the isolated cells from all four donor tissues grew plastic-adherent and showed similar adipogenic and osteogenic differentiation capacity as proven by the histological detection of lipid droplets or deposits of extracellular calcium and collagen type I. After 27 days of chondrogenesis proteoglycans accumulated in the differentiated MSC-pellets from all donor tissues. Immunohistochemical staining revealed vast amounts of collagen type II in all differentiated MSC-pellets, except for those from the LCF. Interestingly, all differentiated MSCs still showed a clear increase in mean expression of adipogenic, osteogenic and chondrogenic marker genes. In addition, the examination of an exemplary selected donor sample revealed that cells from all four donor tissues were clearly positive for the surface markers CD44, CD73, CD90 and CD105 by flow cytometric analysis.
Conclusions
This study proved the presence of MSC-like cells in all four examined donor tissues of the hip joint. No significant differences were observed during osteogenic or adipogenic differentiation depending on the source of MSCs used. Further research is necessary to fully determine the tripotent differentiation potential of cells isolated from the LCF and capsule tissue of the hip joint.
Zinc is an essential trace element for all living organisms. In mammals, including humans and mice, it is required for normal growth, development, hematopoiesis and immune defense. This thesis investigates the influence of zinc on the development of megakaryocytes (MKs), the cells responsible for bone marrow-derived platelet production. Furthermore, a detailed analysis of the expression of zinc import and export transporters (Slc39a/Slc30a genes) is carried out, firstly over the course of MK differentiation and secondly dependent on extracellular zinc.
In heart failure, a functional block of complex I of the respiratory chain provokes superoxide generation, which is transformed to H\(_2\)O\(_2\) by dismutation. The Krebs cycle produces NADH, which delivers electrons to complex I, and NADPH for H\(_2\)O\(_2\) elimination via isocitrate dehydrogenase and nicotinamide nucleotide transhydrogenase (NNT). At high NADH levels, α-ketoglutarate dehydrogenase (α-KGDH) is a major source of superoxide in skeletal muscle mitochondria with low NNT activity. Here, we analyzed how α-KGDH and NNT control H\(_2\)O\(_2\) emission in cardiac mitochondria. In cardiac mitochondria from NNT-competent BL/6N mice, H\(_2\)O\(_2\) emission is equally low with pyruvate/malate (P/M) or α-ketoglutarate (α-KG) as substrates. Complex I inhibition with rotenone increases H2O2 emission from P/M, but not α-KG respiring mitochondria, which is potentiated by depleting H\(_2\)O\(_2\)-eliminating capacity. Conversely, in NNT-deficient BL/6J mitochondria, H2O2 emission is higher with α-KG than with P/M as substrate, and further potentiated by complex I blockade. Prior depletion of H\(_2\)O\(_2\)-eliminating capacity increases H\(_2\)O\(_2\) emission from P/M, but not α-KG respiring mitochondria. In cardiac myocytes, downregulation of α-KGDH activity impaired dynamic mitochondrial redox adaptation during workload transitions, without increasing H\(_2\)O\(_2\) emission. In conclusion, NADH from α-KGDH selectively shuttles to NNT for NADPH formation rather than to complex I of the respiratory chain for ATP production. Therefore, α-KGDH plays a key role for H\(_2\)O\(_2\) elimination, but is not a relevant source of superoxide in heart. In heart failure, α-KGDH/NNT-dependent NADPH formation ameliorates oxidative stress imposed by complex I blockade. Downregulation of α-KGDH may, therefore, predispose to oxidative stress in heart failure.
Die koronare Herzkrankheit ist weltweit die häufigste Todesursache und belastet die Gesellschaft durch Therapien und Arbeitsausfälle mit hohen Kosten. Die Europäischen Gesellschaft für Kardiologie empfiehlt in den Leitlinien zur Sekundärprävention der koronaren Herzkrankheit verschiedene Lebensstiländerungen. In der vorliegenden Arbeit wurde untersucht, wie häufig diese Empfehlungen tatsächlich gegeben werden, ob diese von den Patienten umgesetzt werden und welche Faktoren den Erhalt der Empfehlung bzw. die Umsetzung beeinflussen. Hierzu wurden 536 Probanden mit bekannter KHK aus dem Raum Würzburg im Rahmen der multizentrischen Querschnittsstudie EUROASPIRE IV befragt und untersucht.
Es konnte gezeigt werden, dass die Empfehlungen insgesamt viel zu selten gegeben werden. Als positiver Einflussfaktor für den Erhalt einer Empfehlung konnte die Teilnahme an einem Rehabilitationsprogramm identifiziert werden. Die Wahrscheinlichkeit, die Empfehlungen zu erhalten, sank zudem mit zunehmendem Alter bei Erstdiagnose der KHK. Diese Erkenntnisse können in Zukunft dabei helfen, die Aufklärung über die Risikofaktoren insgesamt und besonders bei älteren Menschen zu intensivieren. Sie untermauern zudem die große Bedeutung eines Rehabilitationsprogrammes bei der Informationsvermittlung.
Auch die Umsetzung der empfohlenen Lebensstilveränderungen erfolgte nicht in zufriedenstellender Häufigkeit. Für die Beendigung des Rauchens konnte die Teilnahme an einem Rehabilitationsprogramm als größter Einflussfaktor ermittelt werden. Die Wahrscheinlichkeit, dass die verschiedenen Maßnahmen zu Lebensstilveränderungen umgesetzt werden, war beim weiblichen Geschlecht deutlich höher als beim männlichen. Die Umsetzung wurde zudem dadurch beeinflusst, ob der Patient vorher die entsprechende Empfehlung erhalten hatte. Die Daten zeigen somit auch bei der Umsetzung der Maßnahmen die Bedeutung von Rehabilitationsprogrammen, besonders für Raucher, und verdeutlichen, dass vor allem bei Männern weitere Anreize geschaffen werden müssen, damit diese die Empfehlungen zur Lebensstiländerung auch umsetzen. Dass viel mehr Probanden die Maßnahmen umgesetzt haben, wenn sie vorher die entsprechende Empfehlung erhalten haben, unterstreicht die außerordentliche Wichtigkeit der umfassenden Aufklärung des Patienten über die existentiellen Chancen eines Lebensstilwandels.
Graft-versus-Host Disease (GvHD) stellt einen häufigen, den Gesamterfolg einer allogenen hämatopoetischen Stammzelltransplantation limitierenden Faktor dar. Bei dieser Komplikation attackieren vor allem alloreaktive T-Lymphozyten des Stammzellspenders gesunde Körperzellen des Patienten. Infolgedessen kommt es zu Gewebeschäden in den Zielorganen Haut, Leber und Darm. Die Behandlung der GvHD erfordert eine effektive Immunsuppression, was wiederum Graft-versusTumor-Effekte kompromittiert und den Rückfall der malignen Grunderkrankung bedingen kann. Viele Patienten sprechen aus bisher ungeklärten Gründen nicht auf die klassische immunsuppressive Therapie mit Steroiden oder second-line Therapien an. Neue zelluläre Therapien zur Behandlung der refraktären GvHD sind auf dem Vormarsch, bedürfen aber einer weiterführenden klinischen Testung, auch um die exakten Wirkungsmechanismen zu verstehen. Idealerweise könnten neue Testsysteme das GvHD-Potential von allogenen Stammzellpräparaten oder aber das immunsuppressive Potential von neuen GvHD-Therapien vorhersagen, bevor diese in klinischen Studien eingesetzt werden. Ziel der vorliegenden Arbeit war es, ein erstes, in multiplen Replikaten einsetzbares, humanes organotypisches Gewebemodell zur Simulation einer GvHD-Reaktion am Beispiel der Haut zu etablieren.
Zu diesem Zweck wurden artifizielle humane Hautmodelle unter statischen (KollagenHautmodelle) und dynamischen Kulturbedingungen (vaskularisierte Hautmodelle) generiert. Die Injektion unstimulierter PBMCs (engl. peripheral blood mononuclear cells) führte zu keinen histomorphologischen Veränderungen in den KollagenHautmodellen. Im Gegensatz dazu hatte die Injektion vorstimulierter allogener PBMCs eine Zerstörung der epidermalen Strukturen der Kollagen-Hautmodelle zur Folge, welche vergleichbar waren mit Gewebeschäden bei einer akuten GvHD der Haut. Dieselben Schädigungen der Epidermis wurden durch die Injektion von Mediumüberständen vorstimulierter PBMCs in die Kollagen-Hautmodelle erreicht. Im Kulturmedium der Kollagen-Hautmodelle wurden hohe Konzentrationen von Interleukin 2 und 17, Interferon gamma sowie Tumornekrosefaktor alpha gemessen, wodurch auf die Beteiligung von Zytokinen an der inflammatorischen Reaktion geschlossen werden konnte. Auch im komplexeren vaskularisierten Hautmodell verursachte die Injektion vorstimulierter PBMCs histomorphologische Veränderungen entsprechend einer akuten Haut-GvHD sowie einen zeitabhängigen Anstieg proinflammatorischer Zytokine.
Zusammenfassend zeigen die Resultate dieser Arbeit, dass die Induktion einer starken Inflammations- und Immunreaktion in artifiziellen humanen Hautmodellen, welche histomorphologisch eine GvHD imitiert, möglich ist. Dieses Modell könnte als Grundlage für die Entwicklung eines klinisch relevanten Testsystems zur Bestimmung des GvHD-Restpotentials oder zur Festlegung der immunsuppressiven Kapazität innovativer Zellpräparate dienen. Somit könnten humane artifizielle GvHDModelle in klinischen Studien eingesetzt werden und die Erfahrungen aus Tiermodellen ergänzen sowie erste in vitro Ergebnisse im humanen System liefern, welche dann mit dem tatsächlichen klinischen Resultat verglichen werden könnten.
Due to the rapidly increasing development and use of cellular products, there is a rising demand for non-animal-based test platforms to predict, study and treat undesired immunity. Here, we generated human organotypic skin models from human biopsies by isolating and expanding keratinocytes, fibroblasts and microvascular endothelial cells and seeding these components on a collagen matrix or a biological vascularized scaffold matrix in a bioreactor. We then were able to induce inflammation-mediated tissue damage by adding pre-stimulated, mismatched allogeneic lymphocytes and/or inflammatory cytokine-containing supernatants histomorphologically mimicking severe graft versus host disease (GvHD) of the skin. This could be prevented by the addition of immunosuppressants to the models. Consequently, these models harbor a promising potential to serve as a test platform for the prediction, prevention and treatment of GvHD. They also allow functional studies of immune effectors and suppressors including but not limited to allodepleted lymphocytes, gamma-delta T cells, regulatory T cells and mesenchymal stromal cells, which would otherwise be limited to animal models. Thus, the current test platform, developed with the limitation that no professional antigen presenting cells are in place, could greatly reduce animal testing for investigation of novel immune therapies.
Oxylipine werden in der Pflanze unter Stressbedingungen gebildet. Die dafür notwendige Oxidation von Fettsäuren wird entweder nicht-enzymatisch über Radikale wie reaktive Sauerstoffspezies (ROS) oder enzymatisch über Lipoxygenasen katalysiert. Abhängig von der Position der Oxidation in der Fettsäure entstehen dabei C13- oder C9-Oxylipine. Sehr gut erforscht sind C13-Oxylipine wie Jasmonsäure (JA), die bei biotischem Stress und Verwundung gebildet werden und bei exogener Gabe das Wurzelwachstum von Arabidopsis thaliana hemmen. Die C9-Oxylipine wie 9-Hydroxyoktadekatriensäure (9-HOT) sind erst wenig erforscht. Ziel dieser Arbeit war die Charakterisierung von Transkriptionsfaktoren, mit dem Fokus auf 9-HOT-vermittelte Signalwegen in Arabidopsis thaliana. Da bekannt ist, dass auch sie zu einer Hemmung des Wurzelwachstums führen, wurde dazu die Untersuchung des Wurzelwachstums von 10 Tage alten Keimlingen etabliert. Funktionsgewinn-Mutanten des Transkriptionsfaktors TGA5 sowie des TGA5-Zielgens CYTOCHROM P450 MONOOXYGENASE CYP81D11 zeigten auf 9-HOT ein verglichen mit Col-0 deutlich besseres Wurzelwachstum. Die AtTORF-Ex-Kollektion, eine große Sammlung an Überexpressions-Linien verschiedener Transkriptionsfaktoren, wurde hinsichtlich Wurzelwachstums auf dem Oxylipin 9-HOT analysiert. Die Gesamtheit der untersuchten Pflanzen enthielt 263 unabhängige TF-Expressions-Konstrukte. Von 6087 untersuchten Pflanzen zeigten 201 Pflanzen keine Hemmung des Wurzelwachstums auf 9-HOT. Dabei konnten 80 verschiedene Transkriptionsfaktoren identifiziert werden, deren Überexpression die Wurzelwachstums-hemmende Wirkung von 9-HOT kompensiert. Es zeigte sich eine Häufung von Transkriptionsfaktoren der ERF- (ethylene responsive factor) Familie. Die verstärkte Expression der nahe verwandten Transkriptionsfaktoren ERF106 und ERF107 ermöglichte sowohl auf 9-HOT als auch auf 9-KOT ein längeres Wurzelwachstum im Vergleich zum Wildtyp. Die Genexpression von ERF106 und ERF107 wird durch Überflutung aktiviert. Durch Überflutung wird im Wildtyp die Expression von Hypoxia-Antwort-Genen wie HRE1, SUS4 oder PDC1 induziert. In den Funktionsverlust-Mutanten sind diese Gene in der Expression aber nicht beeinflusst. Auch ist nach Überflutung im normalen Tag / Nacht-Rhythmus kein signifikanter Unterschied im Überleben zwischen Col-0 und den Mutanten erf106, erf107 und erf106xerf107 nachweisbar. Zur Identifikation möglicher Ziel-Gene von ERF106 und ERF107 wurden Transkriptom-Analysen durchgeführt. Die Funktionsverlust-Mutanten erf106, erf107 und erf106xerf107 zeigten weder im Grundzustand noch nach 4 Stunden Überflutung Veränderungen in den bekannten Hypoxia-Antwort-Genen. Die Funktionsgewinn-Mutanten von ERF106 und ERF107 zeigten in der Transkriptom-Analyse eine deutliche Aktivierung von Genen, die wichtig für Entgiftung und Stressabwehr sind. Ebenso wurden wichtige Biosynthese-Gene aus der Camalexin- und Glukosinolat-Synthese in den Funktionsgewinn-Mutanten verstärkt exprimiert. Des Weiteren konnte eine verringerte Expression von Genen beobachtet werden, die wichtig für die Regulation der Eisen-Aufnahme sind, darunter bHLH-Transkriptionsfaktoren, der Eisen-Transporter IRON REGULATED TRANSPORTER 1 (IRT1) und die Eisen-Reduktase FERRIC REDUCTION OXIDASE 2 (FRO2). Zusammenfassend wurden in dieser Arbeit durch die Untersuchung der AtTORF-Ex-Kollektion mehrere TF identifiziert, die wichtige Abwehr-Gene gegen Stress- und Vergiftung sowie bedeutende Gene im Bereich der Biosynthese und Eisenaufnahme regulieren können, um so die Antwort auf C9-Oxylipine zu beeinflussen.
Background and Aims: Colonoscopy as standard procedure in endoscopy is often perceived as uncomfortable for patients. Patient's anxiety is therefore a significant issue, which often lead to avoidance of participation of relevant examinations as CRC-screening. Non-pharmacological anxiety management interventions such as music might contribute to relaxation in the phase prior and during endoscopy. Although music's anxiolytic effects have been reported previously, no objective measurement of stress level reduction has been reported yet. Focus of this study was to evaluate the objective measurement of the state of relaxation in patients undergoing colonoscopy.
Methods: Prospective study (n = 196) performed at one endoscopic high-volume center. Standard colonoscopy was performed in control group. Interventional group received additionally self-chosen music over earphones. Facial Electromyography (fEMG) activity was obtained. Clinician Satisfaction with Sedation Instrument (CSSI) and Patients Satisfaction with Sedation Instrument (PSSI) was answered by colonoscopists and patients, respectively. Overall satisfaction with music accompanied colonoscopy was obtained if applicable.
Results: Mean difference measured by fEMG via musculus zygomaticus major indicated a significantly lower stress level in the music group [7.700(±5.560) μV vs. 4.820(±3.330) μV; p = 0.001]. Clinician satisfaction was significantly higher with patients listening to music [82.69(±15.04) vs. 87.3(±15.02) pts.; p = 0.001]. Patient's satisfaction was higher but did not differ significantly.
Conclusions: We conclude that self-chosen music contributes objectively to a reduced stress level for patients and therefore subjectively perceived satisfaction for endoscopists. Therefore, music should be considered as a non-pharmacological treatment method of distress reduction especially in the beginning of endoscopic procedures.
A method is reported for making hollow channels within hydrogels decorated with cell‐adhesion peptides exclusively at the channel surface. Sacrificial fibers of different diameters are used to introduce channels within poly(ethylene glycol) hydrogels crosslinked with maleimide‐thiol chemistry, which are backfilled with a cysteine‐containing peptide solution which is conjugated to the lumen with good spatial efficiency. This allows for peptide patterning in only the areas of the hydrogel where they are needed when used as cell‐guides, reducing the amount of required peptide 20‐fold when compared to bulk functionalization. The power of this approach is highlighted by successfully using these patterned hydrogels without active perfusion to guide fibroblasts and olfactory ensheathing cells—the latter having unique potential in neural repair therapies.
Long-term treatment effect studies in large female Fabry patient groups are challenging to design because of phenotype heterogeneity and lack of appropriate comparison groups, and have not been reported. We compared long-term cardiomyopathy and kidney function outcomes after agalsidase beta treatment with preceding treatment-naive outcomes.
Methods and results Self-controlled pretreatment and post-treatment comparison (piecewise mixed linear modelling) included Fabry female patients ≥18 years at treatment initiation who received agalsidase beta (0.9–1.1 mg/kg every other week) for ≥2 years, with ≥2 pretreatment and ≥2 post-treatment outcome measurements during 10-year follow-up. Left ventricular posterior wall thickness (LVPWT)/interventricular septal thickness (IVST) and estimated glomerular filtration rate (eGFR, Chronic Kidney Disease Epidemiology Collaboration creatinine equation) analyses included 42 and 86 patients, respectively, aged 50.0 and 46.3 years at treatment initiation, respectively. LVPWT and IVST increased pretreatment (follow-up 3.5 years) but stabilized during 3.6 years of treatment (LVPWT: n = 38, slope difference [95% confidence interval (CI)] = - 0.41 [ - 0.68, - 0.15] mm/year, P\(_{pre–post difference}\)<0.01; IVST: n = 38, slope difference =-0.32 [-0.67, 0.02] mm/year, P\(_{pre–post difference}\) = 0.07). These findings were not modified by renal involvement or antiproteinuric agent use. Compared with the treatment-naive period (follow-up 3.6 years), eGFR decline remained modest and stabilized within normal ranges during 4.1 years of treatment (slope difference, 95% CI: -0.13 [-1.15, 0.89] mL/min/1.73m\(^2\)/year, P\(_{pre–post difference}\) = 0.80).
Conclusions Cardiac hypertrophy, progressing during pretreatment follow-up, appeared to stabilize during sustained agalsidase beta treatment. eGFR decline remained within normal ranges. This suggests that treatment may prevent further Fabry-related progression of cardiomyopathy in female patients and maintain normal kidney function.
