Tbe alkylating potency of unstable N-nitrosamino acids and N-nitrosopeptides was investigated in vitro using 4-(para-nitrobenzyl)pyridine (NBP) as nucleophile. Of the amino acids, Met and those with an aromatic side chain were the most potent. The relative overall alkylating potency was 23:10:5:4:2:1: for Trp, Met, His, 1)rr, Phe and Gly, respectively. The homo-dipeptides were much more potent than the amino acids, with relative potencies of 400:110:100:8:3:1, for Trp-Trp, l)T-'I)T, Met-Met, Asp-Asp, Phe-Phe and Gly, respectively. In the one-phase reaction system (in which NBP is already present durlog the nitrosation reaction at acidic pH), all amino acids tested showed a second-order reaction for nitrite. In the two-phase system (in which NBP is added only after bringing the nitrosation reaction mixture to neutrality), all amino acids tested except one again showed a second-order reaction for nitrite (Phe, His, Asp and the dipeptide artiticial sweetener aspartame); only Met under these conditions bad a reaction order of one for nitrite. This could mean that nitrosation of the side chain of Metproduces a second N-nitroso product which is relatively stable in acid but reacts with NBP under neutral conditions. In the human stomach, this side-chain nitrosation might become more important than the reactions at the primary amino group, firstly because of the greater stability of the product(s) in acid and secondly because of the tirst-order reaction rate for nitrite. A decrease in nitrite concentration from the millimolar concentrations ofthe in-vitro assay to the micromolar concentrations in the stomach reduces the reaction rate by a factor of 1000 for the side-chain nitrosation, whereas a million-fold reduction will be observed for nitrosation of the amino group.
Um sich vor dem Austrocknen zu schützen, haben Pflanzen eine Transpirationsbarriere entwickelt, die als Membran alle primären, oberirdischen Pflanzenteile überzieht. Diese so genannte Kutikula besteht hauptsächlich aus den lipophilen Komponenten Kutin und Wachs und reduziert so effektiv den Verlust von Wasser und wasserlöslichen Nährstoffen aus dem Blattinneren. Trotzdem ist sie nicht vollständig undurchlässig, und so können Wasser und gelöste Substanzen wie organische und anorganische Nährstoffe, Pestizide oder Umweltchemikalien die Kutikula in beiden Richtungen permeieren. Dabei ist offensichtlich, dass die zu Grunde liegenden Transportmechanismen den Ernährungszustand der Pflanzen, die Effizienz von Pestiziden und die Wirkung von Umweltchemikalien beeinflussen. Ein genaues Verständnis der Transportprozesse auf denen die kutikuläre Permeation basiert, kann helfen die Wirkweise von blattapplizierten Dünge- und Pflanzenschutzmitteln zu optimieren, indem gezielt Wirk- oder Zusatzstoffe modelliert werden können, welche die Aufnahme steigern. In der vorliegenden Arbeit sollte deshalb der Einfluss physiko-chemischer Eigenschaften von hydrophilen Verbindungen auf die kutikuläre Permeation untersucht werden. Nicht zuletzt wegen ihrer strukturellen Ähnlichkeit mit den blattapplizierten Herbiziden Glufosinat und Glyphosat wurden Aminosäuren als Modellsubstenzen ausgewählt. Die verwendeten Aminosäuren sind gut wasserlöslich, wobei alle Oktanol/Wasser Verteilungskoeffizienten kleiner als 1 sind. Zusätzlich liegen alle Aminosäuren in gelöster Form als Ionen vor, was zu einer Hydratisierung der Moleküle führt. Es wird spekuliert, dass hydratisierte Moleküle keinen Zugang zur lipophilen Phase der Kutikula haben. Welche Rolle die Hydrathülle bei der Permeation tatsächlich spielt, ist allerdings noch unklar. Viele Aktivwirkstoffe liegen nur unter ganz bestimmten Bedingungen in geladener Form vor, während die Richtung der kontinuierlichen Nettoladung der Aminosäuren durch den pH Wert modifiziert wird. Damit kann der Einfluss verschiedener Ladungszustände auf die kutikuläre Permeation unter Verwendung eines einheitlichen Sets von Modellsubstanzen untersucht werden. Unter natürlichen Bedingungen sind Aminosäuren unter anderem auf Blattoberflächen zu finden, wo sie blattassoziierten Mikroorganismen eine profitable Nahrungsquelle bieten. Ob äußere Faktoren für die Deposition dieser Recourcen verantwortlich sind, oder ob der Ursprung innerhalb des Blattgewebes liegt, wird kontrovers diskutiert. Die Sorption von Aminosäuren in isolierte Kutikularmembranen ist sehr gering, und korreliert - anders als bei lipophilen Substanzen - nicht mit dem Oktanol/Wasser Verteilungskoeffizienten. Das zeigt, dass der Verteilung von lipophilen und hydrophilen Substanzen innerhalb der Kutikula verschiedene Mechanismen zu Grunde liegen. Unter einer gegebenen Bedingung werden die kutikulären Leitwerte der Aminosäuren negativ vom Molvolumen beeinflusst. Zudem übersteigt die Länge des Permeationswegs die eigentliche Dicke der Membran um ein Vielfaches. Diese Zusammenhänge kennzeichnen eine gehinderte Diffusion innerhalb einer engporigen und weit verzweigten Umgebung. Eine Änderung des pH Wertes wirkt sich in unterschiedlicher Form auf die Leitwerte von Wasser und Aminosäuren aus. Mit steigendem pH Wert erhöht sich die Wasserpermeabilität isolierter Kutikularmembranen, was durch eine zunehmende, messbare Wassersorption in die Kutikula erklärt werden kann. Eine pH abhängige Dissoziation funktioneller Gruppen bewirkt eine Schwellung des polaren Weges, weshalb auch für die anionischen Aminosäuren bei pH 11 die höchsten Leitwerte gemessen wurden. Die zwitterionischen Aminosäuren bei pH 6 wiesen hingegen die geringsten Leitwerte auf, was im Widerspruch zu der Beobachtung steht, dass bei pH 1 die geringste Wassersorption in die Kutikula stattfindet. Eine Erklärung hierfür liefern die Hydrathüllen, die bei den zwitterionischen Aminosäuren am stärksten und bei den anionischen Species am geringsten ausgeprägt sind. Eine negative Korrelation aller gemessenen Aminosäureleitwerte mit den entsprechenden hydratisierten Molvolumen zeigt eindeutig, dass die Hydrathülle eine wichtige Größe für die Permeation durch die Kutikula darstellt. Dabei nimmt der Leitwert einer hydrophilen Substanz mit definiertem Molvolumen mit kleiner werdender Hydrathülle zu. Intakte Blätter wurden in flüssiges Wasser als Rezeptorlösung getaucht, um steady-state Bedingungen aufrecht zu erhalten. Dabei konnte gezeigt werden, dass die Permeabilitäten von intakten Kutikularmembranen, die anhand der natürlichen Aminosäurekonzentration innerhalb der Blätter bestimmt wurden, in derselben Größenordnung liegen, wie die für isolierte Membranen gemessenen. Außerdem konnte ein Vergleich der Flussraten auf der Ober- und Unterseite der Blätter zeigen, dass die stomatären Poren nicht direkt in den Leachingprozess involviert sind.
The charged aerosol detector (CAD) is an aerosol-based detector employed in liquid chromatography which has become established in the field of pharmaceutical analysis due to its outstanding performance characteristics, e.g. the almost uniform response for nonvolatile analytes. Owing to its principle of detection, the response of the CAD depends on the volatility of a compound and is inherently nonlinear. However, the newly implemented instrumental settings evaporation temperature and power function value (PFV) are valuable tools to overcome some of these drawbacks and can even enhance the detector’s capabilities when adjusted properly.
This thesis aimed to evaluate the impact of the new instrumental settings on the CAD performance. Additionally, the influence of modern separation techniques for small polar compounds on the CAD was assessed and the applicability of hyphenated UV-CAD techniques explored. The optimization strategies derived from the evaluation procedures and the conjunction of the instrumental and chromatographic techniques investigated were utilized for the challenging impurity profiling of amino acids and amino acid-like drugs.
The results of the method validation procedures confirmed the broad applicability of the CAD in the pharmaceutical analysis of nonvolatile compounds, supported by satisfactory sensitivity and reproducibility for meeting the regulatory requirements with respect to the ICH guidelines Q2(R1) and Q3A(R2). The limits of applicability include the analysis of semivolatile compounds, and the method transfer between current and legacy CAD models. Further advances in the definition and standardization of allowed ranges for the instrumental settings and the establishment of general optimization procedures in the method development could lead to a more widespread use of the detection technique in compendial methods.
The main aim of this thesis was the synthesis and structural characterization of penta and hexacoordinate silicon(IV) complexes. In the course of these studies, the neutral pentacoordinate silicon(IV) complexes 38, 39, 43−48, 54 and 55 were prepared. Furthermore, the neutral hexacoordinate silicon(IV) complexes 33−36, 49, 50, 52, 53, 56−62, 63, 64 and 65 were synthesized. All compounds were characterized by elemental analyses, NMR spectroscopy in solution (1H, 13C, 15N, 29Si) and in the solid-state (13C, 15N, 29Si VACP/MAS NMR), as well as single-crystal X-ray diffraction (except 45, 47−49, 52, 53 and 63).
The requirements for the impurity profiling of substances for pharmaceutical use have become greater over time. They can be accomplished by the use of modern instrumental analysis techniques, which have been evolved in the last decades. New types of columns with HILIC, mixed-mode and chiral stationary phases are suitable for the separation of all kinds of substances mixtures, that were previously hardly possible with the use of common reversed phase columns. Modern, almost universal detectors like CAD, ELSD and CNLSD can be applied for a sensitive detection of substances without a chromophore. However, in addition to some small individual disadvantages to these methods, the costs are high and applications are still kind of rare. Thus, the introduction of these devices at a broader level has not yet taken place. While this presumably will change over time, there is a need for methods that enable the impurity profiling of challenging substances with widespread analytics devices.
Methionine is a substance with hydrophobic and hydrophilic impurities. With the help of a mixed-mode stationary phase, which is a combination of a reversed phase and a strong cationic exchanger, the separation of all putative impurities was found possible with good sensitivity and selectivity. The method requires apart from the column only standard isocratic HPLC equipment and was successfully validated.
The evaluation of the enantiomeric purity of amino acids is challenging. Two approaches were made. The first method utilizes CE by means of in-capillary derivation with OPA and the subsequent separation with a cyclodextrin. With the use of OPA/NAC and γ-cyclodextrin, a simple and cost-effective method for the indirect enantioseparation of 16 amino acids was developed. With the second approach, racemic amino acids can be analyzed with HPLC and in-needle derivatization. For this, different columns and chiral thiols were evaluated and the chromatographic parameters were optimized. A method with OPA/NIBLC, a pentafluorophenyl column made the enantioseparation of 17 amino acids feasible. A LOQ of the minor enantiomer down to 0.04 % can be achieved with UV spectrophotometric detection. A similar method was developed for impurity profiling of L-amino acids. This can be used alternatively for the amino acid analysis performed by the European Pharmacopoeia.
A simple, robust, precise and accurate method for the evaluation of impurities in glyceryl trinitrate solution was developed and validated. The four impurities of glyceryl trinitrate are separated by means of an acetonitrile-water gradient and the assay for this substance is also possible.
This thesis was dedicated to the studies of the electronic and chemical properties of liquids and solutions using soft x-ray spectroscopies. The used photon-in-photon-out methods namely x-ray absorption spectroscopy (XAS), x-ray emission spectroscopy (XES), and resonant inelastic x-ray scattering (RIXS) appeared to be an excellent choice for these studies. In the framework of this thesis, the necessary experimental setup for using the above mentioned experimental techniques on liquids was developed. Hereby, a new flow-through liquid cell was introduced which simplifies the studies of liquids and solutions. The cell design is very flexible and thus can be modified for gases and liquid/solid interfaces. With this cell it is possible to study the samples under well-controlled conditions (temperature and flow rate). The novel flow-through liquid cell is part of the new SALSA synchrotron endstation including an electron analyzer and a novel high-resolution, high-transmission soft x-ray spectrometer. The latter makes it possible to measure two-dimensional RIXS maps in a very short time, which include the full excitation and emission information in one plot. Making use of the new instrumentation, a variety of different liquids and solutions were investigated. As first system, aqueous solutions of sodium hydroxide (NaOH) and sodium deuteroxide (NaOD) were investigated. In the XAS as well as in the XES spectra a pronounced concentration dependence was found. At non-resonant energies, the spectra are dominated by the solvent and thus look similar to water. Making use of the pre-pre-edge in the absorption spectra which can exclusively be attributed to OH- / OD- it was possible to extract the resonant emission spectra of the ions which show an indication for proton dynamics during the core-hole lifetime. For the solid state NaOH XES spectra it was possible to reveal a high energetic shoulder and a low energetic shoulder at the high energy emission feature. These shoulders can be assigned to self-dissociation processes where OH- forms O2- ions and H2O. The study of NaOH was also of interest for the studies of the amino acids, which were in the focus of the next part, since the pH-values of the respective solutions were controlled by NaOH. In the next part of this thesis, amino acid solutions were investigated. Amino acids are the building blocks of peptides and proteins and thus important for life science. The investigated representatives were glycine, the simplest amino acid, and lysine, an amino acid with two amine groups. Both amino acids react on pH-value changes at the amine group where the local environment at the nitrogen atom changes (NH2 ↔ NH3+). A strong change of the spectra induced by this protonation/deprotonation could be found. Furthermore, for low pH-values (protonated amine groups) the amine groups are influenced by strong proton dynamics. First DFT calculations confirm the dissociation model of the amino acids. Qualitatively the high energy peak in the N K XES spectra can be attributed to the deprotonated amine group and the low energy area for the protonated amine group. Besides amino acids, alcohols and acids are important in biological processes. Therefore, the smallest alcohol (methanol) and the smallest carboxylic acid (acetic acid) were under investigation. For the liquid methanol XES spectra a very good agreement with DFT calculations of gas phase methanol could be found. This observation suggests that the influence of the environment (hydrogen bonding) on the spectra is small. The achieved spectra are in good agreement with DFT calculations found in literature. It was possible to selectively excite the two non-equivalent oxygen atoms in acetic acid and to reveal the carboxyl specific C K XES. The carbon XAS spectra showed strong differences compared to gas phase measurements which might be a hint for the influence of the hydrogen bond network. The investigation of the electronic and chemical properties of liquids and solutions is a very young field of research and the results presented in this thesis show that it is a very interesting topic. The presented results can be seen as the fundamental frame work for all following studies. With the understanding of basic, i.e., simple, systems as shown in this work it will be possible to understand complex biological systems in their native environment, e.g., peptides and proteins, which are the building blocks of life.
Ziel der Dissertation „Entwicklung von Cysteinproteaseinhibitoren – ein klassischer und ein kombinatorischer Ansatz zur Inhibitoroptimierung“ war die Optimierung von neuen Inhibitoren von Falcipain-2 und Rhodesain als neue potentielle Wirkstoffe gegen Malaria bzw. die Schlafkrankheit über zwei verschiedene Methoden. Es handelt sich hierbei um einen klassischen und einen kombinatorischen Ansatz. Der klassische Ansatz basiert auf einer Struktur, deren Aktivität per Zufall entdeckt wurde. In Screenings von synthetisierten Strukturanaloga, gestützt durch virtuelle Docking-Experimente am aktiven Zentrum der Cysteinproteasen, wurden Struktur-Aktivitäts-Beziehungen erarbeitet. Bei der kombinatorischen Methode wurde zunächst ein peptidischer Inhibitor entworfen, der durch Festphasensynthese an einem geeigneten Harz synthetisiert wurde. Durch den kombinatorischen Einsatz von Aminosäuren konnte auf diese Weise unter enormer Zeitersparnis eine Bibliothek von 150 Inhibitoren synthetisiert werden. In einem Screening dieser harzgebundenen Inhibitoren wurden anschließend die potentesten Inhibitoren identifiziert. Die Aktivität der gefundenen Inhibitoren aus beiden Ansätzen an den protozoischen Erregern wurde durch in-vitro-Experimente an Plasmodien und Trypanosomen untersucht. Beim klassischen Ansatz wurde eine neue Substanzklasse entwickelt, die sehr gute Hemmeigenschaften an beiden Cysteinproteasen mit IC50-Werten im niedrigen mikromolaren Bereich zeigten. Außerdem besaßen sie eine hohe in-vitro-Aktivität gegenüber den Erregern im gleichen Konzentrationsbereich. Einige der Inhibitoren zeigten keine Zytotoxizität an Makrophagen. Aus dem klassischen Ansatz konnten also hochaktive Substanzen mit geringer Zytotoxizität entwickelt werden, deren Einsatz als Wirkstoffe gegen Malaria oder der Schlafkrankheit denkbar ist. Für den kombinatorischen Ansatz wurde zur Inhibitoroptimierung eine Screeningmethode für Falcipain-2 und Rhodesain direkt an einem geeigneten Harz zur Festphasensynthese entwickelt. Neu bei dieser Screeningmethode war es, dass erstmals ein quantitatives Screening einer Inhibitorbibliothek möglich sein sollte, und nicht nur die besten Inhibitoren identifiziert werden können. Aus den Ergebnissen der Festphasenscreenings an beiden Proteasen wurden 14 besonders interessante Inhibitoren der Bibliothek ausgewählt und synthetisiert. Diese Verbindungsklasse zeigte ebenfalls sehr gute Ergebnisse an den isolierten Enzymen, in den mikrobiologischen Tests an den Erregern jedoch fielen alle Ergebnisse vergleichsweise schlechter aus. Die schlechte Löslichkeit, die Bioverfügbarkeit und der Metabolismus durch den Erreger der peptidischen Inhibitoren schienen von großer Bedeutung zu sein. Der bearbeitete kombinatorische Ansatz lieferte eine neuartige Screeningmethode, die auch auf andere Targets anwendbar ist.
The polyspecific organic cation transporters (OCT) are involved in the elimination and distribution of drugs, environmental toxins, and endogenous organic cations including monoamine neurotransmitters. Steroid hormones inhibit organic cation transport by the three OCT subtypes with different affinities showing distinct species difference; for example, the IC50 values for corticosterone inhibition of cation uptake by transporters rOCT1 and rOCT2 are ~150μM and ~4 μM, respectively. By introducing domains and amino acids from rOCT2 into rOCT1, we identified three amino acids in the presumed 10th TMD of rOCT2 which are responsible for the higher affinity of corticosterone in comparison to rOCT1. This is the first study which revealed the components of the binding site for corticosterone in OCTs. The evidence is presented that these amino acids (alanine 443, leucine 447, and glutamine 448 in rOCT1 and isoleucine 443, tyrosine 447, and glutamate 448 in rOCT2) are probably located within the substrate binding region of OCTs since the affinity of transported cations was increased together with the affinity of corticosterone. In the double mutant rOCT1(L447Y/Q448E) the IC50 value for the inhibition of [3H]MPP (0.1 μM) uptake by corticosterone (24 ± 4 μM) was significantly higher compared to the IC50 value for inhibition of [14C]TEA (10 μM) uptake (5.3 ± 1.7 μM), indicating an allosteric interaction between transported substrate and corticosterone. The data suggest that more than one compound can bind simultaneously to the substrate binding region. These results confirm previous suggestion that binding of substrates and inhibitors to OCTs involves interaction with a comparatively large surface that may include multiple binding domains rather than with a structurally restricted single binding site.
The impurity profiling of pharmaceutical ingredients can oppose many challenges. The best part of active pharmaceutical ingredients (APIs) and the related substances are detectable by UV detection, a very common detection principle. However, if an API lacks a suitable chromophore other means of detection are necessary. The corona charged aerosol detector (CAD) is a detector capable of detecting substances independent of their chemical structure. This “universal” detector has only one limitation: The analyte has to have a sufficiently low vapor pressure. Another important challenge that comes often together with the lack of a chromophore concerns the separation. These substances (e.g. most amino acids and derivatives) often contain structures that make them difficult to retain on conventional reversed phase columns.
Possible solutions to overcome these challenges, like the application of the CAD and the benefit of so-called mixed-mode stationary phases in impurity profiling for pharmacopoeial purposes were explored in this work. The related substances analyzed in this thesis comprise amino acids, inorganic ions, bisphosphonic acids, basic and acidic derivatives of amino acids (esters and amides).
The successful development and validation of mixed-mode liquid chromatography methods with CAD detection for carbocisteine and ibandronate sodium might help to increase the acceptance of this versatile detector in the pharmaceutical industry and in official authorities dealing with the determination of related substances.
The combination of UV and CAD detection proved very useful during the analysis of Bicisate. Most of the related substances and some unidentified impurities were detectable by CAD whereas a synthesis by-product, a semi-volatile ester, was only detectable in the UV trace. The simple combination covers all relevant impurities in a single analysis.
Two truly orthogonal methods regarding separation and detection for the enantiomeric purity of magnesium-L-aspartate helped to find the reason for elevated D aspartic acid content in the drug substance. A very quick and sensitive indirect separation using the OPA derivatization with NAC was developed as a powerful screening tool, whereas the direct separation of D- and L-CBQCA-Asp derivatives confirmed the results. Both methods were optimized in order to do without substances mentioned on the REACH list, like sodium tetraborate which is very frequently applied in standard derivatization protocols and CE separations.
The importance of orthogonal detection principles in the determination of related substances of amino acids was discussed in a review article dealing with the revision of amino acid monographs in the Ph. Eur..
Kapillarelektrophoretische Reinheitsanalytik verschiedener Arzneistoffe des Europäischen Arzneibuchs

Die Kapillarelektophorese (CE), deren Trennprinzip auf der Wanderung geladener Teilchen im elektrischen Feld basiert, ist eine Methode, die in verschiedenen Techniken angewandt werden kann. Sowohl die wässrige Kapillarzonenelektrophorese (CZE) als auch die wasserfreie CE (NACE), aber auch die elektrokinetische Chromatographie mittels Mikroemulsion (MEEKC) wurden in dieser Arbeit für die Reinheitsanalytik der im Europäischen Arzneibuch beschriebenen Wirkstoffe Ethambutol, Quetiapin, Ephedrin sowie Levodopa und deren jeweils strukturverwandter Substanzen benutzt. Der Wirkstoff Ethambutol wird in der (S,S)-Form verwendet, die im Ph. Eur. 7 als Dihydrochlorid aufgeführt ist. Um eine Trennmethode für (S,S)-Ethambutol, sein Enantiomer und die achirale meso-Verbindung entwickeln zu können, wurden die beiden stereoisomeren Verunreinigungen aus 2-Amino-1-butanol und Diethyloxalat synthetisiert. Zur Trennung dieser drei Ethambutol-Isomere wurde CZE als Methode gewählt. In saurem Phosphatpuffer musste eine hohe Probenkonzentration von 1 mg/ml verwendet werden, um die Substanzen mit UV detektieren zu können (λ: 200 nm). In alkalischem Tetraboratpuffer war das Chromophor dank der freien Elektronenpaare der Stickstoff-Moleküle besser ausgeprägt und die Intensität der Peaks deutlich intensiver. Als chirale Selektoren wurden die nativen α-, β- und γ-Cyclodextine (CDs) und verschiedene derivatisierte β-CDs eingesetzt. Die Methode wurde vielfach in Bezug auf Molarität und pH-Wert der Puffer, Konzentration der verschiedenen chiralen Selektoren, Spannung und Temperatur modifiziert. Jedoch konnte keine Trennung der Stereoisomere erreicht werden. Eine CD-modifizierte MEEKC-Methode wurde herangezogen, um die Racemate der Aminosäuren Dopa, Methyldopa, Tyrosin und Phenylalanin voneinander zu trennen. Dazu wurde eine Mikroemulsion (ME) aus Ethylacetat, SDS, 1-Butanol, Phosphatpuffer, sulf. β-CD und, wenn nötig, aus dem organischen Modifier 2-Propanol eingesetzt. Für jede DL-Aminosäure wurde die Zusammensetzung der ME als auch die Geräteeinstellungen (Spannung, Temperatur) optimiert. Die Trennung von DL-Dopa konnte ohne Zugabe eines organischen Modifiers durchgeführt werden. Auf Grundlage dieser individuellen Methoden wurden zwei CD-modifizierte MEEKC-Methoden entwickelt, mit denen alle vier untersuchten Racemate getrennt werden konnten. Die abschließende Validierung in Bezug auf Wiederholpräzision (Auflösung, Migrationszeiten, Verhältnis der korrigierten Peakflächen und Anzahl der theoretischen Böden) und Detektionsgrenzen zeigte, dass die Methoden präzise Ergebnisse liefern. Die Technik der MEEKC wurde auch zur Trennung von Ephedrin-Derivaten genutzt. Wedig et al. konnten die Racemate von Ephedrin, Pseudoephedrin, N-Methylephedrin und Norephedrin mit einer HDAS-β-CD-modifizierten CZE-Methode in einem Lauf basislinientrennen, indem ein 50 mM Phosphatpuffer, pH 3,0 als HGE eingesetzt wurde. Aus diesem HGE und den organischen Bestandteilen, die zur Trennung der Aminosäuren führten, wurde eine ME hergestellt. Entgegen der Methode von Wedig et al. konnte mittels HDAS-β-CD keine zufriedenstellende Trennleistung erreicht werden. Durch Austausch des chiralen Selektors gegen sulf. β-CD und Modifizierung des Phosphatpuffers in Ionenstärke und pH-Wert konnte für alle vier Epedrin-Derivate eine Basislinientrennung erzielt werden. Diese MEEKC-Methode wurde auf weitere Ephedrin-Derivate angewandt, wodurch das racemische 2-(Dibutylamino)-1-phenyl-1-propanol partiell, die Racemate von Adrenalin, 2-Amino-1-phenylethanol und Diethylnorephedrin vollständig voneinander getrennt werden konnten. Während mit der HDAS-β-CD-modifizierten CZE-Methode alle vier Ephedrin-Derivaten in einem Lauf getrennt werden konnten, hat die MEEKC-Methode den Vorteil mit dem kostengünstigeren sulf. β-CD auszukommen. Schlussendlich wurde eine Reinheitsanalytik von Quetiapin und seinen verwandten Substanzen Quetiapindesethanol, Quetiapin-N-Oxid und Quetiapinlactam entwickelt. Da Quetiapinlactam fast ausschließlich in organischen Lösungsmitteln löslich ist, sollte eine wasserfreie CE-Methode (NACE) eingesetzt werden. Zwar konnte eine Methode entwickelt werden, deren HGE aus Methanol, Acetonitril, Ammoniumacetat und Essigsäure bestand, und mit der Quetiapin und seine drei verwandten Substanzen sehr gut getrennt werden konnten. Allerdings konnte sie aufgrund von Stromabbrüchen nicht validiert werden. Alternativ wurde eine wässrige, gut reproduzierbare CZE-Methode gefunden, deren Elektrolytlösung aus einem 80 mM Phosphatpuffer, pH 4.0 bestand. Aufgrund der Wasserunlöslichkeit von Quetiapinlactam konnten so nur Quetiapin und die Verunreinigungen Quetiapindesethanol und Quetiapin-N-Oxid erfasst werden. Abschließend wurde die CZE-Methode validiert, wodurch die hohe Präzision der ermittelten Werte gezeigt werden konnte.
The main focus of this thesis was the synthesis and analysis of multifunctional oligopeptides. The study of their non-covalent interactions with various counterparts revealed interesting new results, leading to both methodological and application related progress. The first project of this thesis concentrated on the in-depth analysis of the peptide receptor CBS-Lys-Lys-Phe-NH2 to acquire a better understanding of its binding mode upon complexation with a substrate. In this context it was possible to develop—in cooperation with the group of Prof. Sebastian Schlücker—a direct and label free spectroscopic detection of immobilized compounds which are often found in combinatorial libraries. This new screening method utilizes the advantages of the surface enhanced Raman spectroscopy and allowed for the first time a surface mapping of a single polystyrene bead for the identification of peptides in femtomolar concentrations. Hence, this method allows a very fast and sensitive detection of resin bound compounds. The development of this promising new approach set the starting point for future experiments to enable on-bead library screenings and to investigate the complex formation of immobilized compounds. After the comprehensive analysis of the basic structural features of small peptide receptors in the first part of this thesis, the second big block focused on its in vitro evaluation using biological relevant targets. Therefore, several different modifications of the initial peptide structures were synthesized. These modifications provided a molecular toolkit for the tailor made synthesis of structures individually designed for the respective target. The first tests addressed the interaction with Alzheimer’s related amyloid fibrils. During these experiments, the successful SPPS syntheses of tri- and tetravalent systems were achieved. The comparison of the multivalent form with the corresponding monovalent version was then under special investigations. These concentrated mainly on the interaction with various bacteria strains, as well as with different parasites. To localize the compounds within the organisms, the synthesis of fluorescence labelled versions was achieved. In addition, several compounds were tested by the Institute for Molecular Infection Biology of the University of Würzburg for their antibacterial activity. This thorough evaluation of the biological activity generated precious information about the influence of small structural changes in the peptide receptors. Especially the distinct influence of the multivalency effect and the acquired synthetic skills led to the development of an advanced non-covalent recognition event, as described in the final project of this thesis. The last part of this thesis discussed the development of a novel inhibitor for the serine protease beta-tryptase based on a tailor-made surface recognition event. It was possible to study and analyze the complex interaction with the unique structure of tryptase, that features a tetrameric frame and four catalytic cleavage sites buried deep inside of the hollow structure. However, the point of attack were not the four binding pockets, as mostly described in the literature, but rather the acidic areas around the cleavage sites and at the two circular openings. These should attract peptides with basic residues, which then can block the accessibility to the active sites. A combinatorial library of 216 tetravalent peptide compounds was synthesized to find the best structural composition for the non-covalent inhibition of beta-tryptase. For the screening of the library a new on-bead assay was applied. With this method a simultaneous readout of the total inhibition of all library members was possible, thus allowing a fast and direct investigation of the still resin bound inhibitors. Several additional experiments in solution unveiled the kinetics of the inhibition process. In conclusion, both mono- and multivalent inhibitors interact in a non-destructive and reversible way with the tryptase.

The specifically labeled subunit was cleaved with cyanogen bromide and N-bromosuccinimide, and the resultant fragments were separated by gel chromatography in the presence of 80% (vol/vol) formic acid. The N,N'-dicyclohexylcarbodiimide label was recovered in each organism exclusively in a 17-residue fragment. Further analysis by automated solid-phase Edman degrada.ti.on revealed tha~ the bound label was present at only one positIOn, correspondmg to a glutamyl residue. Das Ziel vorliegender Arbeit war die Entwicklung einer HPLC-MS(/MS)-Methode, die im Rahmen der pharmazeutischen Qualitätskontrolle zur direkten Quantifizierung von Aminosäuren (AS) in Infusionslösungen angewendet werden kann. Die Zielset-zung schloss eine Validierung innerhalb der für die Zweckbestimmung vorgesehenen Grenzen ein. Im Rahmen der Methodenentwicklung wurde das ESI-MS/MS-Fragmentierungs-muster von 21 Aminosäuren, von 20 stabil-isotopenmarkierten Aminosäuren, die als interne Standards verwendet wurden, sowie von einigen weiteren Substanzen bestimmt. Nach Kenntnis von Precursor- und Produktionen erstellte man eine SRM-Methode zur spezifischen MS/MS-Analyse. Dabei wurden durch das jeweilige Frag-mentierungsmuster bedingte Interferenzen bei den zu untersuchenden Aminosäuren bestimmt, die bei der zu erarbeitenden HPLC-MS-Methode beachtet werden mussten. Die Methodenentwicklung zur HPLC-MS-Analytik von underivatisierten AS umfasste mit der RP-HPLC unter Verwendung eines Ionenpaarreagenzes (IP) und der hydrophilen Interaktionschromatographie (HILIC) zwei verschiedene chromatographi-sche Ansätze. Bei der Anwendung der RP-HPLC ergaben sich Probleme. Die Verwendung eines IP, im vorliegenden Fall TDFHA (Tridecafluorheptansäure), führte zu langen Equilibrierungs-, Re-Equilibrierungs- und Spülzeiten und damit bei zwar relativ kurzer HPLC-Laufzeit zu einem aber insgesamt hohen Zeitaufwand. Gleich-zeitig war die LC-MS-Anlage auf diese Anwendung fixiert, da das Ionenpaarreagenz das Gerät stark verschmutzte und dadurch andere Analysen erheblich störte. Zudem waren die Retentionszeiten der Analyten trotz langer Equilibrierungszeiten schlecht reproduzierbar, so dass eine solche Methode im Rahmen der pharmazeutischen Qualitätskontrolle schwer validierbar wäre. Weiterführende Untersuchungen erfolgten daher nicht. In nachfolgenden Studien mit der HILIC wurden verschiedene Einflussparameter (Anteil organischer Phase im Fließmittel, pH-Wert des Fließmittels, Temperatur der Säule, Pufferkonzentration im Fließmittel, Gradientenelution) auf die Trennung der AS an einer ZIC®-HILIC-Säule untersucht. Durch Optimierung der Parameter wurde so eine HILIC-HPLC-Methode entwickelt, bei der 21 AS und 20 ihrer isotopen-markierten Referenz-AS innerhalb von 20 min eluierten. Diejenigen AS, bei denen im Rahmen der Fragmentierungsstudien Interferenzen aufgrund gleicher bzw. ähnlicher Massen der Precursor- bzw. Produktionen aufgetreten waren, wurden chroma-tographisch getrennt. Gleichzeitig hat sich die SIM-Analyse als anwendbar erwiesen. Die Anwendung des spezifischeren SRM-Modus und damit der Tandem-Massenspektrometrie war nicht erforderlich. Im Rahmen der nachfolgenden Studien zur Validierung ergab sich, dass die entwickelte Methode über einen weiten Bereich eine lineare Abhängigkeit zwischen Konzentrations- und Messwerten zeigte. Für drei der 21 AS (NAcCys, NAcTyr, Pro) wurde die quadratische Regression mit dem Anpassungstest nach Mandel als geeig-neteres Regressionsmodell ermittelt. Bei Untersuchungen zur Wiederfindung wurde ein Einfluss der Matrix-Lösung der Infusionslösung festgestellt, der zu Abweichungen hinsichtlich des Quotienten AreaAS / AreaIS führte, so dass eine Quantifizierung innerhalb der geforderten Grenzen bei Kalibrierung über reine Standardlösungen nicht möglich war. Die Validierung wurde daher nachfolgend in der Matrixlösung durchgeführt. Dabei wurde gezeigt, dass mit der entwickelten HILIC-HPLC-MS-Methode Aminosäuren in Infusionslösungen mit hoher Präzision und Richtigkeit bestimmt werden können. Neun der 21 untersuchten AS konnten im Bereich von 30% - 350%, zehn weitere im Bereich von 50% - 350% innerhalb der zur Gehaltsbestimmung von pharmazeutischen Formulierungen vorgeschriebenen Grenzen (Wiederfindung Einzelbestimmung: 98% -102.0%, Mittelwert einer Dreifachbestimmung: 98.5% – 101.5%) quantifiziert werden. Für His und Phe gelang allerdings keine Quantifizierung innerhalb der Akzeptanzkriterien, wobei der Grund hierfür in weiteren Studien geklärt werden müsste. Mit der entwickelten Methode ist damit eine gleichzeitige Quantifizierung verschiedener AS-Infusionslösungsformulierungen möglich, die sich bei gleicher Matrix in der Konzentration an AS unterscheiden. Beispielsweise seien hier die Formulierungen „Aminoplasmal® E 5% / 10% /15%“ genannt, die mit der validierten Methode erfassbar sind. Die Probenvorbereitung beschränkt sich dabei auf den Zusatz der IS-Formulierung zur Infusionslösung und einen Verdünnungsschritt. Die Quanti-fizierung erfolgt über eine 5-Punkt-Kalibriergerade, die aus einer AS- und IS-Standardmischung, nach Zusatz der einfach herzustellenden Elektrolyt-Matrix, erstellt wird. Die Analysenzeit der HPLC-MS-Methode beträgt einschließlich Equilibrie-rungszeit 35 min und ist damit deutlich kürzer als die 120 min, die bei der nach wie vor zur AS-Analytik allgemein gebräuchlichen Ionenaustauschchromatographie mit Ninhydrin-Nachsäulenderivatisierung anzusetzen sind.
Das Ziel der vorliegenden Arbeit war die Klärung der Fragestellung, ob sich die quantitative NMR-Spektroskopie zur Bestimmung von Identität, Reinheit und Gehalt von Arzneistoffen eignet, und wie sich Präzision und Empfindlichkeit dieser Methode im Vergleich zu etablierten chromatographischen Verfahren verhalten. Die quantitative Untersuchung der drei strukturell jeweils verwandten Mehrkomponentengemische Codergocrinmesilat, Clomifencitrat und Flupentixoldihydrochlorid bewies eindrucksvoll die Eignung der 1H-NMR-Spektroskopie als orthogonale, analytische Messmethode im Vergleich zu validierten HPLC-Arzneibuchmethoden. Die im Rahmen einer Validierung der 1H-NMR-Methode ermittelten Ergebnisse erfüllten bezüglich Präzision und Richtigkeit die an eine im pharmazeutischen Bereich eingesetzte analytische Methode gestellten Anforderungen; zudem wurden weitere Prüfparameter wie Selektivität, Linearität, Robustheit und Stabilität verifiziert. Externe-Standard-Experimente wie "Zwei-Röhrchen-Methode" und ERETIC-Verfahren bestätigten die quantitativen Ergebnisse der Internen Standardisierung; jedoch wurde hier -- insbesondere für die ERETIC-Technik -- eine höhere Fehleranfälligkeit und somit eine größere Streuung der Einzelergebnisse beobachtet. Am Beispiel von Codergocrinmesilat und Flupentixoldihydrochlorid konnte zudem die Eignung anderer NMR-aktiver Kerne wie 13C und 19F für die quantitative Analyse von komplexen Substanzgemischen aufgezeigt werden. Das Potential der 1H-NMR-Spektroskopie für die Reinheitsprüfung von Arzneistoffen wurde am Beispiel der Aminosäure L-Alanin aufgezeigt. Die zu erwartenden Verunreinigungen Glutamin-, Asparagin-, Äpfel- und Fumarsäure konnten im Gegensatz zu den "veralteten" Prüfmethoden des Europäischen Arzneibuches eindeutig identifiziert und quantifiziert werden; mit einer Bestimmungsgrenze von <= 0.03% wurden die Vorgaben der ICH-Richtlinie Q3A(R2) erfüllt. Die deutliche Übereinstimmung der NMR-spektroskopisch ermittelten Ergebnisse einer quantitativ untersuchten Alanin-Modellmischung mit einer für den Routinebetrieb geeigneten HPLC-Methode unter Einsatz verschiedener Detektoren wie CAD, NQAD, ELSD und MS, sowie der Vergleich wichtiger Prüfparameter wie Linearität und Nachweisgrenze bestätigten die Eignung der 1H-NMR-Spektroskopie im Rahmen der routinemäßig durchgeführten Qualitätskontrolle. Die Aufdeckung von Arzneimittelfälschungen mit Hilfe der NMR-Spektroskopie wurde im Rahmen dieser Arbeit anhand der zwei aktuellen Fallbeispiele Heparin und Glycerin näher untersucht. Die in Zusammenhang mit dem Heparin-Skandal verantwortliche Kontaminante OSCS konnte neben Dermatansulfat und weiteren natürlich vorkommenden Glykosaminoglykan-Verunreinigungen im 1H-NMR-Spektrum eindeutig identifiziert und auf 0.1% OSCS bzw. 0.5% Dermatansulfat begrenzt werden. Eine präzise und richtige quantitative Bestimmung der beiden Glykosaminoglykane wurde über die N-Acetyl-Resonanzen mit Hilfe der Signalhöhenbestimmung und dem Standard-Additionsverfahren ermöglicht; deutliche Abweichungen vom "wahren" Gehalt wurden hingegen, bedingt durch starke Signalüberlagerungen, nach Flächenvergleich beobachtet. Weitere Verunreinigungen, insbesondere Lösungsmittelrückstände, die während des Extraktions- und Reinigungsprozesses des Heparins eingesetzt werden, konnten ebenfalls über charakteristische Resonanzen identifiziert und mit Hilfe der Internen-Standard-Methode quantitativ erfasst werden. Eine umfangreiche Untersuchung von 145 Heparin-API-Mustern mittels NMR-Spektroskopie und weiteren, neuentwickelten Verfahren wie HPLC, CE, IR- und Raman-Spektroskopie konnte die Eignung der entwickelten 1H-NMR-Methode bestätigen. Potentielle Glycerin-Kontaminanten wie Diethylenglycol und Ethylenglycol konnten ebenso wie eine weitere, natürlich vorkommende Verunreinigung, Propylenglycol, mittels 1H- und 13C-NMR-Spektroskopie identifiziert und quantifiziert werden. Beide Methoden erfüllten die in der USP beschriebenen Anforderungen, die für pharmazeutisch eingesetztes Glycerin jeweils höchstens 0.1% Diethylenglycol bzw. Ethylenglycol erlaubt. Während die quantitative Reinheitsprüfung beim Einsatz der 1H-NMR-Spektroskopie mit einer Messdauer im Bereich von etwa 30 min für den Routineeinsatz geeignet ist, ist die entwickelte quantitative 13C-NMR-Methode beim Einsatz von Spektrometern geringer Magnetfeldstärke aufgrund einer geringen Nachweisempfindlichkeit und der NOE-Problematik für den Routinebetrieb nur bedingt anwendbar. Abschließend kann zusammengefasst werden, dass die untersuchten Beispiele die NMR-Spektroskopie als in hohem Maße geeignet für die quantitative Analyse von Arzneimitteln ausweisen.
Neuste Studien haben ergeben, dass Asc-1 Knock-out Mäuse aufgrund einer verminderten intrazellulären Glycinkonzentration in synaptischen Boutons im Gehirn, einen Hyperekplexie-ähnlichen Phänotyp entwickeln. Aufgrund nicht vollständig geklärter Ursachen für die Entstehung des Krankheitsbildes der Hyperekplexie beim Menschen, wurde eine Kohorte von 51 Patienten zusammengetragen, um vor dem Hintergrund der Forschungsergebnisse zu Asc-1 im Tiermodell, das kodierende Gen beim Menschen SLC7A10 als mögliches Kandidatengen auf Sequenzalterationen zu untersuchen. Hierfür wurde aus Vollblut der an Hyperekplexie erkrankten Patienten genomische DNA isoliert, um mittels PCR und anschließendem Screening der Sequenzen, Mutationen innerhalb funktionell wichtiger Bereiche des Gens zu eruieren. Neben weiteren Sequenzunterschieden, die meist in Introns gefunden wurden, wurde die codierende Mutation G307R innerhalb von Exon 7 identifiziert, die letztendlich der Grund für eine Versuchsreihe war, um zu hinterfragen, ob dieser Aminosäureaustausch in der Proteinsequenz funktionelle Konsequenzen zur Folge hat. HEK293-Zellen wurden mit dem zuvor hergestellten Klon G307R transfiziert, um über Biotinylierung, immuncytochemische Färbungen und funktionelle Untersuchungen die Aktivität des Transporters zu beurteilen. Hier zeigte sich ein Funktionsverlust von über 95 %, bei uneingeschränkter Oberflächenexpression. ASC-1 bestätigt sich damit als neue Ursache in der Ausprägung von Hyperekplexie. Ferner können Zusammenhänge mit geistiger Retardierung und eingeschränkter neuronaler Plastizität bestehen.
This thesis focuses on the investigation of the electronic structure of amino acids and
salts in aqueous solution using X-ray spectroscopic methods. Both material groups are
of fundamental importance with regards to many physiological reactions, especially
for the Hofmeister effect which describes the solubility of proteins in salt solutions.
Hence, the investigation of the electronic structure of amino acids and the influence of
ions on the hydrogen bonding network of liquid water are important milestones to a
deeper understanding of the Hofmeister series.
Besides investigating the electronic structure of amino acids in aqueous solution,
the spectra were used to develop a building block model of the spectral fingerprints of
the functional groups and were compared to spectral signatures of suitable reference
molecules. In the framework of this thesis, it is shown that the building block approach
is a useful tool with allows the interpretation of spectral signatures of considerably
more complex molecules
In this work, the focus lies on the investigation of the occupied and unoccupied
electronic states of molecules in solid state, as well as in aqueous solution. Hereby,
different X-ray spectroscopic methods were applied. X-ray emission spectroscopy
(XES) was used to probe the occupied electronic structure of the solution, while the
unoccupied electronic structure was addressed by using X-ray absorption spectroscopy
(XAS). Finally, resonant inelastic X-ray scattering (RIXS) as a combination of XAS
and XES measurements provides the combined information about the unoccupied and
occupied molecular levels. The element specific character of the three measurement
methods is a feature which allows the investigation of the local electronic structure of
a single functional group. With RIXS, also non-equivalent atoms of the same element
can be addressed separately.
Within this thesis firstly, a library of the XE spectra of all 20 proteinogenic amino
acids in zwitterionic form is presented. From this sample-set XES fingerprints of
the protonated alpha-amino group NH3+ and the deprotonated carboxylic group COO- were evaluated and used to identify the XES fingerprints of the nitrogen and oxygen
containing functional groups of the side chains of the amino acids. The data is discussed
based on a building block approach. Furthermore, the XE spectra of the functional
groups of lysine and histidine, namely the NH2 group and the C3N2H4 ring structure,
are both compared to XE spectra of suitable reference molecules (imidazole, ammonia
and methylamine). It is found that the XE and RIXS spectra of the side chains of lysine
and histidine show large similarities to the XE spectra of the reference molecules. This
agreement in the XE and RIXS spectra allows a qualitative investigation of XE and
RIXS spectra of more complex amino acids using the XE and RIXS spectra of suitable
reference molecules.
The chemical structure of histidine and proline is quite different from the structures
of the other proteinogenic amino acids. Due to the unique chemical structure of
the side chain which in both cases consists of a heterocyclic ring structure, these two
amino acids were investigated in more detail. Zubavichus et al. [1] have shown that
amino acids are decomposing while exposed to X-ray radiation of the experiment. The
damage is irreversible and molecular fragments can adsorb on the membrane of the
experimental setup. This contamination can also create a spectral signature which
then overlaps with the signal of the solution and which complicates the interpretation
of the data. To record spectra which are free from contributions of adsorbed molecular
fragments on the membrane, the adsorption behavior was investigated.
In contrast to the solid phase in which the amino acids are present as salts in one
electronic conformation, the charge state of the amino acids can be manipulated in
aqueous solution by tuning the pH-value. By doing this, all possible charge states are
accessible (cation, anion, zwitterion). In this work it is shown that also the spectra
of the different charge states can be modeled by the spectra of suitable reference
molecules using the building block approach. The spectral changes occurring upon
protonation and deprotonation of the functional groups are explored and verified by
comparing them to theoretical calculations.
The comparison with measurements of pyrrolidine show that the electronic structure
which surrounds the nitrogen atom of proline is strongly influenced by the
ring structure of the side chain. Furthermore, the proline, pyrrolidine, and histidine
molecules are also degrading during the liquid sample measurements. This can be
observed by the detection of a new spectral component which increases with the
measurement time originating from the window membrane. In all cases, the speed of
the agglomeration of molecular fragments at the membrane was observed to be highly
sensitive to the pH value of the solution.
To understand the Hofmeister series, also the impact of the salt ions have to be
investigated. In this study the influence of potassium chloride (KCl) on the hydrogen
bond network of water was studied by using non-resonantly excited XES as well as
RIXS. A decreased dissociation of hydrogen molecules and changes in the molecular
vibrations could be detected. These changes were interpreted with a molecular
reorganization of the water molecules and a decreased number of hydrogen bonds.
The stability of Trp in pure solutions and in parenteral AA formulations was evaluated with regard to typically used manufacturing processes, storage conditions and primary packaging. Therefore, thorough stability studies on Trp solutions were conducted beforehand. The applied stressing method, i.e. steam sterilization by autoclave, are chemically seen relatively mild but showed to be efficient to induce Trp degradation in the presence of oxygen. Subsequent identification, separation and characterization were challenging due to similar substance properties, numerous stereoisomers and pairs of diastereomers found amongst them. However, the identified o-aminoacetophenone compounds, Kyn and NFK, are associated with photo reactivity and have photo-oxidizing properties. Thus, best possible protection from UV-light, together with strict oxygen expulsion, are the most important criteria to impede Trp degradation after autoclaving.
The identification of Trp degradation products was assisted by the compilation of a substance library, which included manifold reported and chemically plausible Trp degradation substances. The substances were classified for priority and their early or late-stage occurrence. The large number of possible substances and stereoisomers was narrowed down with the information retrieved from LC-UV/MS experiments. However, final identification was achieved by the synthesis of proposed substances as references. The following eight substances were characterized as Trp degradation substances: Kyn, NFK and three pairs of diastereomers R,R/R,S DiOia, R,R/R,S Oia and cis/trans PIC. Fig. 33 shows the proposed degradation pathway and demonstrates the close chemical relationship, which may be an explanation for the conversion of some substances into each other during the storage period. The proposed pathway brings together the results of different Trp stability and stressing studies, respectively [89, 94, 97, 98, 103, 133]. To our knowledge, the simultaneous formation of the identified degradation substances has not been reported before and especially not under the stressing conditions applied.
The application of a traditional RP-HPLC method was compared to two developed IP-HPLC methods and a RP-HPLC methods using a modified perfluorinated column. Orthogonal analyses methods and especially the combination of UV and MS detection are necessary in order to indicate potentially undetected degradation substances. Main evaluation criteria were the separation performance, analyses time, reproducibility and feasibility. The best results upon assessment of all Trp degradation products, in both; pure Trp solutions and pharmaceutical formulations, were obtained by a traditional RP-HPLC. The optimized method was validated according to ICH guidelines Q2(R1) and meets the criteria of a stability-indicating HPLC-UV method. The validated method has a sufficient separation performance with an adequate selectivity indicating the Trp degradation substances next to each other and next to other AAs in finished pharmaceutical formulations.
The detailed knowledge of Trp degradation and the method presented may be transferred practically to the pharmaceutical industry processing Trp-containing products. In general, the findings might contribute to the quality management of such pharmaceutical products during
manufacturing and storage. Additionally, the study results provide basic information for the establishment of an impurity consideration following the ICH guidelines Q3B (R2) (impurities in new drug products) for products containing Trp. However, further development of the method applying more sophisticated detectors or more potent HPLC techniques like e.g. UHPLC and the implication of more sensitive (MS) detectors like ToF-MS would be advantageous with regard to economic and practical aspects.
Im Rahmen der vorliegenden Arbeit wurden Beiträge zur C/Si-Bioisosterie und zur asymmetrischen Synthese neuer siliciumhaltiger alpha-Aminosäuren geleistet. Im Vordergrund der Untersuchungen stand die Entwicklung neuer präparativer Methoden zur Darstellung siliciumhaltiger Wirkstoffe und die Untersuchung ihrer pharmakologischen Wirksamkeit sowie die Synthese racemischer und enantiomerenreiner siliciumhaltiger alpha-Aminosäuren des beta-(Trimethylsilyl)alanin-Typs.
Der erste Teil der vorliegenden Arbeit beschreibt neuartige Synthesen siliciumhaltiger Aminosäuren, ausgehend von Malonsäurediethylester. Weitere Teilschritte dieser Synthesen sind eine biokatalytische enantioselektive Esterspaltung von Malonsäurediethylester-Derivaten mittels Schweineleberesterase und ein Curtius-Abbau. Es wurde in allen Schritten darauf geachtet, kostengünstige und leicht handhabbare Chemikalien zu verwenden, um eine etwaige Produktion der Aminosäuren in einem größeren Maßstab zu ermöglichen. Der zweite Teil der vorliegenden Arbeit widmet sich Versuchen, aus den im vorangehenden Abschnitt beschriebenen Aminosäuren potentielle dipeptidische, siliciumorganische Süßstoffe herzustellen. Diese werden am besten als Aspartam-Analoga beschrieben. Die Charakterisierung der Verbindungen erfolgte durch NMR-Spektroskopie (1H, 13C, 15N, 29Si), Elementaranalysen und gegebenenfalls durch Kristallstrukturnalysen.
Synthese einer Bibliothek von Aminosäure-basierten Oligopeptid-Amphiphilen mittels Festphasensynthese, deren kovalente Knüpfung an einen nukleophilen Kern zu C3-symmetrischen Sternmesogenen und die Analyse der Einflüsse der verwendeten Aminosäuren auf die Sekundärstruktur des synthetisierten Moleküls.
The work presented in this thesis was mainly targeted at exploring the capabilities of evaporation based LC detectors as well as further alternatives for the control of impurities in substances not exhibiting a suitable chromophore for UV-detection. In the course of the work carried out, several new methods for the identification, impurities control and composition testing of APIs were elaborated. An evaporation based detector that entered into the field of pharmaceutical analysis in the recent years was the Evaporative Light Scattering Detector (ELSD). However, non-reproducible spikes were reported when injecting concentrated test solutions as they are usually required for the control of impurities. The reasons, for the appearance of these spikes as well as possibilities for their avoidance were explored in a systematic study. Moreover, the dependence of the detector sensitivity on different eluent composition, eluent flow-rate and ELSD settings was investigated. In the course of the revision of the Ph.Eur. monographs for aspartic acid and alanine, a C18 reversed phase ion-pair LC method using 1 mmol/L of perfluoroheptanoic acid as an ion-pair reagent and a charged aerosol detector (CAD) was developed and fully validated for the purity control of Asp. The method was capable of separating the organic acids and major amino acids known to occur as process related impurities. With a slight modification, the method was also applicable for the purity control of Ala. Based on the developed LC-CAD method for the impurity control of alanine, a comparative study of the performance characteristics of different evaporation based LC detectors, i.e. ELSD, CAD and the recently developed Nano Quantity Analyte Detector (NQAD) was carried out. Additionally, an MS detector and qNMR were included in this study. It was found that the control of impurities in Alanine at an ICH conform level could be ensured using LC coupled to CAD, MSD and NQAD detection as well as by the use of qNMR. In terms of performance, prize and ease of use CAD and NQAD were found to be the most suitable alternatives. In terms of repeatability and sensitivity, the CAD appeared slightly superior to the NQAD. The quality of streptomycin sulfate is not sufficiently controlled by the current Ph.Eur. monograph in that an appropriate test for the control of the related substances is missing. A study was carried out to develop a C18 reversed phase ion-pair LC method using pentafluoropropionic acid as an ion-pair reagent and a CAD for the identification and control of the related substances. The developed method allowed the separation of 21 impurities from streptomycin. Moreover, coupling of the method to MS allowed the identification of the separated impurities. The method was shown to be sufficiently sensitive to control the related substances with a disregard limit of 0.1% as it is normally applied in the Ph.Eur. for products derived from fermentation. Currently, the aescin content of horse-chestnut standardized dry extract is determined using a complex and laborious photometric determination. A more selective LC-UV assay determination for beta-aescin has been proposed for the Ph.Eur. draft monograph of horse-chestnut standardized dry extract. Possibilities were explored to further improve the LC-method using detection by CAD. It was demonstrated that by the use of a modified LC-CAD method several problems related to the differences in the UV-response of the various components contained in the active aescin fraction could be eliminated. Moreover the proposed reference standard strategy was reviewed. Eventually, it was demonstrated on the example of two different clusters of pharmacologically active peptides how low energy collision induced dissociation mass spectrometry (low energy CID-MS) can successfully be used for identification testing in pharmacopoeial monographs. In this respect, the combination of a direct confirmation of the molecular mass via the m/z-ratio of the molecule ions with structural sequence information obtained by low energy CID-MS experiments was found to deliver a higher degree of certainty of the identity of a given substance than the set of tests currently described in the monographs. A significant gain in efficiency and throughput and important reduction of the amount of sample consumed during testing were identified as being additional advantages of this approach. Taken together, it could be demonstrated on various examples how recent technological advancements in the field of analytical chemistry can contribute to improve the quality control of APIs.
Die Qualitätskontrolle von pharmazeutisch verwendeten Substanzen ist eine Voraussetzung für die sichere Anwendung von Arzneimitteln. Sie ist eng mit der Bestimmung der Verunreinigungen einer Substanz im Rahmen der Reinheitsanalytik verknüpft. Dabei spielt das Europäische Arzneibuch (Ph. Eur.) eine wichtige Rolle. Die in der vorliegenden Arbeit durchgeführten Untersuchungen zur Bestimmung des Verunreinigungsprofils waren auf die Neuerarbeitung oder Überarbeitung der im Ph. Eur. beschriebenen Prüfungen auf „Verwandte Substanzen“ ausgerichtet. Im Rahmen der Neuerarbeitung einer Monographie für Trimebutin und Trimebutin-Maleat wurde eine RP-HPLC-Methode entwickelt. Neben den bekannten Verunreinigungen war ein weiterer Peak nachweisbar, der N-Desmethyltrimebutin zugeordnet wurde. Die Trennung zwischen N-Desmethyltrimebutin und dem Hauptpeak sowie zwischen zwei bekannten Verunreinigungen ist kritisch. Für beide Peakpaare wurden Systemeignungskriterien festgelegt. Zur Definition von Akzeptanzkriterien in den erarbeiteten Monographieentwürfen wurden Chargen untersucht. Die Quantifizierung erfolgte über die „Externer-Standard“-Methode mit einer Verdünnung der Untersuchungslösung als Referenz. Falls erforderlich, wurden die Korrekturfaktoren in die Berechnung des Gehaltes einbezogen. Die DC-Methode zur Prüfung auf „Verwandte Substanzen“ von Adenosin sollte gegen eine HPLC-Methode ausgetauscht werden. In Übereinstimmung mit Ergebnissen des EDQM-Labors haben die Untersuchungen bestätigt, dass mit einer Ionenpaar-HPLC-Methode die Anwesenheit von Inosin, Guanosin, Uridin und Adenin in Adenosin kontrolliert werden kann. Durch die Festlegung einer Auflösung > 1.5 zwischen Adenin und Inosin als Systemeignungskriterium werden nicht geeignete HPLC-Säulen erkannt. Durch die DC-Prüfung des Ph. Eur. können verschiedene Nucleotide nachgewiesen werden. Die vorgeschlagene HPLC-Methode wurde durch Einführung eines Gradienten so geändert, dass Nucleotide, Nucleoside und Adenin gleichzeitig nachgewiesen werden konnten. Die Analyse der Proben bestätigte die Ergebnisse der DC, dass kleine Mengen Adenosin-5’-monophosphat in den Chargen vorhanden waren. Eine CE-Methode zur Reinheitsprüfung von Glutathion wurde entsprechend der Kommentare zum in Pharmeuropa veröffentlichten Monographievorschlag überarbeitet. Die kritischen Trennungen zwischen dem internen Standard und Cystein sowie zwischen L-γ-Glutamyl-L-cystein und dem Hauptpeak, die durch den Systemeignungstest überprüft werden, waren durch den pH-Wert des Trennpuffers kontrollierbar. Glutathion zeigt beispielhaft, dass das Verunreinigungsprofil fermentativ gewonnener Produkte komplex ist und von den Herstellungs- und Aufreinigungsprozessen abhängt. Gleiches gilt für Aminosäuren, die vermehrt biotechnologisch gewonnen werden. In den Aminosäure-Monographien ist zur Reinheitsprüfung eine DC auf „Ninhydrin-positive Substanzen“ vorgeschrieben. Diese Methode ist auf den Nachweis anderer Aminosäuren, die durch unvollständige Abtrennung bei der Extraktion aus Proteinhydrolysaten stammen können, ausgelegt. Die Analyse von Aminosäuren mittels CE nach Derivatisierung mit FMOC-Cl ermöglicht einen sensitiven und selektiven Nachweis anderer Aminosäuren. Durch den Einsatz von CBQCA als Derivatisierungsreagenz können auch Aminozucker und kleine Peptide erfasst werden. Mit beiden Methoden wurde das Verunreinigungsprofil von Histidin, Isoleucin, Phenylalanin und Serin bestimmt. Während in den Phe- und Ser-Proben keine Verunreinigungen erfasst wurden, wurden in den Ile-Proben nach Derivatisierung mit FMOC-Cl andere Aminosäuren gefunden. Alle untersuchten Aminosäuren zeigten nach Derivatisierung mit CBQCA viele Verunreinigungen. Wegen Peaküberlagerungen konnten Aminosäuren mit dem verwendeten Trennpuffer (25 mM Boratpuffer pH 9.20; 25 mM SDS) nicht bestimmt werden. Durch eine Variation des Puffers (20 mM Tetraboratpuffer pH 9.3; 100 mM SDS) war es möglich, die CBQCA-derivatisierten Aminosäuren zu trennen und zuzuordnen. Im Hinblick auf die Anwesenheit anderer Aminosäuren waren die Ergebnisse der beiden Verfahren vergleichbar. Insbesondere wurden in den Ile-, Phe- und Ser-Proben keine basischen Aminosäuren sowie Cystein gefunden. Deren FMOC-Derivate comigrierten mit einem Peak, der nach Strukturaufklärung mittels NMR-Spektroskopie als Nebenprodukt der Derivatisierungsreaktion mit FMOC-Cl identifiziert wurde. Als Verunreinigungen von biotechnologisch hergestellten Aminosäuren kommen organische Säuren in Frage. Durch eine RP-HPLC unter Zusatz von Heptafluorbuttersäure als Ionenpaarreagenz wurden Aminosäuren und organische Säuren getrennt. Die Detektion erfolgte mittels ELSD. Nach dem Hauptpeak wurden Störsignale detektiert, weshalb sich die Anwendung der Methode zur Reinheitsprüfung als schwierig herausstellte. Die durchgeführten Experimente deuten darauf hin, dass der ELSD mit der großen Substanzmenge überlastet ist und es zur Verschleppung von Substanz kommt.
Gegenstand der vorliegenden Arbeit war die Untersuchungen von Verunreinigungsprofilen von Aminosäuren aus biotechnologischer Herstellung. Dazu sollten die Aminosäuren Arg, His, Ile, Lys, Phe, Pro, Ser und Trp von verschiedenen Herstellern und aus unterschiedlichen Batches genauer unter die Lupe genommen werden. Mit der im Europäischen Arzneibuch beschriebenen dünnschichtchromatographischen Methode (DC-Methode) für „mit Ninhydrin nachweisbare Substanzen“ können ausschließlich Fremdaminosäuren nachgewiesen werden und dies nur, wenn der jeweilige Gehalt relativ hoch ist. Andere Stoffgruppen, die aus der biotechnologischen Herstellung stammen, werden aufgrund der Trennbedingungen sowie der Detektionsart der DC-Methode nicht erfasst. Deshalb mussten neue Methoden entwickelt werden. Laut Vorschriften der International Conference of Harmonisation (ICH) müssen unbekannte Verunreinigungen in Wirkstoffen für orale Therapeutika auf 0,1 % w/w begrenzt werden können. In die Untersuchungen wurden neben den Fremdaminosäuren auch Peptide, Aminozucker und Nucleinsäuren als potentielle Verunreinigungen, die aus der Herstellung bzw. aus dem Reinigungsprozess kommen, einbezogen. Diese zusätzlichen Stoffgruppen können aus den „Nebenaktivitäten“ der Mikroorganismen entstehen. Aminosäuren, Peptide und Aminozucker wurden versucht, kapillarelektrophoretisch zu quantifizieren. Für die Bestimmung von Nucleinsäuren wurde zusätzlich die UV-Spektroskopie eingesetzt.
Die Cyclodextrin-modifizierte Kapillarelektrophorese (CE) ist eine wichtige chirale analytische Technik geworden, die zur HPLC und zur Gaschromatographie komplementär ist und sich deshalb für die Analyse der Abbauprodukte von Aspartam gut eignet. Ausgehend von diesen Abbauprodukten wurden im Arbeitskreis Scriba an der Universität Jena systematische Studien über die Trennung von Enantiomeren verschiedener Dipeptide mit einer Vielzahl von nativen und derivatisierten Cyclodextrinen bei verschiedenen pH-Werten durchgeführt. Bei der Trennung der Enantiomere von z. B. Ala-Phe oder Ala-Tyr mit beta-Cyclodextrin wurde eine Umkehr der Migrationsreihenfolge bei Erhöhung des Puffer-pH-Werts von 2,5 auf 3,5 festgestellt. Mit Heptakis-(6-sulfato)-beta-cyclodextrin (HS-beta-CD), Heptakis-(2,3-O-diacetyl)-beta-cyclodextrin (Diac-beta-CD) und Heptakis-(2,3-diacetyl-6-sulfato)-beta-cyclodextrin (HDAS-beta-CD) wurde diese Umkehr der Migrationsreihenfolge nicht beobachtet. Das Ziel der vorliegenden Arbeit war, den Mechanismus der Wechselwirkungen zwischen Cyclodextrinen und Aminosäuren bzw. Dipeptiden gründlich und umfassend zu untersuchen. Dabei ging es v. a. um die Untersuchung der Mechanismen der chiralen Erkennung durch Cyclodextrine, die mit Hilfe von verschiedensten Analysenmethoden, vor allem potentiometrische Titrationen und spektroskopische Methoden, wie der NMR-, UV- und CD-Spektroskopie durchgeführt werden sollten. Damit sollte auch die beobachtete Umkehr der Migrationsreihenfolge bei Erhöhung des pH-Wertes des Laufpuffers in der CE erklärt werden. Die potentiometrische Titrationsmethode lieferte sinnvolle Bindungskonstanten für Cyclodextrin-Einschlusskomplexe mit Aminosäuren. Eine Analyse der Struktur-Aktivitätsbeziehungen für Aminosäuren und Cyclodextrine ergab, dass ein gewisses Volumen der Aminosäure-Seitenkette und damit ein gutes Ausfüllen der Cyclodextrin-Kavität nötig ist, um den vollen Nutzen aus den hydrophoben Wechselwirkungen zwischen der Aminosäure-Seitenkette und der Cyclodextrin-Kavität zu ziehen. Eine Verlängerung des hydrophilen Restes, der aus der Kavität herausragt, wie bei den untersuchten Dipeptiden Ala-Phe und Ala-Tyr der Fall, führt zu der Möglichkeit der Ausbildung von Wasserstoff-Brücken mit den Hydroxylgruppen am breiteren Rand der Cyclodextrin-Kavität und damit zu einer stärkeren Bindung an das Cyclodextrin. Um den chiralen Erkennungsprozess von beta-CD und einigen seiner Derivate, nämlich HS-beta-CD, Diac-beta-CD und HDAS-beta-CD, zu verstehen, wurden NMR-Experimente durchgeführt, und zwar wurden „durch Komplexbildung induzierte Verschiebungen der chemischen Verschiebungen” (complexation induced chemical shifts, CICS) gemessen und mittels ROESY-Experimenten die Komplexgeometrie untersucht. Betrachtet man die CICS, die für die Paare Diac-beta-CD/Ala-Phe, HDAS-beta-CD/Ala-Phe, Diac-beta-CD/Ala-Tyr und HDAS-beta-CD/Ala-Tyr auftreten, dann zeigt sich, dass sie relativ klein sind und demnach auf eine eher schwache Wechselwirkung des jeweiligen Gastmoleküls mit dem Wirt hindeuten. Die CICS für beta-CD- und HS-beta-CD-Dipeptid-Komplexe bestätigten einen Einschluss des aromatischen Restes in die Cyclodextrin-Kavität. Es konnte gezeigt werden, dass bei pH 2.5 das DD-Enantiomer von Ala-Tyr tiefer in die Kavität von beta-CD eintaucht als das LL-Enantiomer. Außerdem ist bei pH 3.5 die Eintauchtiefe in die Kavität geringer als bei pH 2.5, was durch die Ergebnisse der ROESY-Experimente bestätigt werden konnte. Um einen besseren Einblick in die Bindungsmodi der Enantiomere von Ala-Phe und Ala-Tyr mit beta-CD bei unterschiedlichen pH-Werten zu erhalten, wurden Moleküldynamik-(MD-)-Simulationen durchgeführt. Die Simulationen wurden mit jedem Enantiomer von Ala-Phe und Ala-Tyr in jedem möglichen Protonierungszustand durchgeführt, d. h. Kation, Zwitterion und Anion. Zum ersten Mal wurden MD-Simulationen für eine größere Serie von unterschiedlichen Komplexen von Enantiomeren in verschiedenen Protonierungszuständen systematisch über den langen Zeitraum von 1 ns (=1000 ps) ausgeführt. Die Eintauchtiefe der untersuchten Dipeptide wurde mit Hilfe einer in die Cyclodextrin-Kavität eingepassten Ebene berechnet. Auf diese Weise konnten Informationen über das unterschiedliche Einschlussverhalten der untersuchten Dipeptide erhalten werden. Die angewendete Methode lässt sich leicht auf andere Wirt-Gast-Komplexe übertragen und erleichtert die Datenerfassung auch in anderen Fällen. Es konnte gezeigt werden, dass bei pH 2.5 das DD-Enantiomer von Ala-Phe tiefer in die Kavität von beta-Cyclodextrin eintaucht als das LL-Enantiomer, wohingegen bei pH 3.5 der umgekehrte Fall vorliegt. Betrachtet man das Dipeptid Ala-Tyr, dann dringt bei pH 2.5 das DD-Enantiomer tiefer ein, wohingegen keine klare Aussage über das Eindringverhalten der Enantiomere bei pH 3.5 gemacht werden kann. Die CICS und die Ergebnisse der Kapillarelektrophorese weisen jedoch auf ein tieferes Eindringen des LL-Enantiomers hin.
