Das Synaptonemalkomplexprotein SYCP1 ist eine Strukturkomponente des Synaptonemalkomplexes (SC) von Saeugern, einer meiosespezifischen Struktur, die wesentlich fuer die Synapse, Rekombination und Segregation homologer Chromosomen ist. Der SC besteht aus zwei lateralen Elementen (LEs) und einer zentralen Region (CR), in deren Mitte das zentrale Element (CE) liegt. Dabei sind die LEs den Achsen der homologen Chromosomen aufgelagert und werden in der CR durch Transversalfilamente (TFs) mit dem CE verbunden. Im Protein SYCP1 (125 kDa) flankieren zwei nicht-helikale terminale Domaenen eine ausgedehnte zentrale „Coiled-Coil“-Domaene. Fuer diese Domaene wird angenommen, dass sie die die Kluft zwischen LEs und CE ueberbrueckt, wobei die C-Termini in den LEs verankert sind und die N-Termini im CE lokalisiert wurden. Um die molekulare Architektur des SC besser zu verstehen und die Bedeutung von SYCP1 für die Zusammenlagerung des SC aufzudecken, wurden die Polymerisationseigenschaften von SYCP1 erforscht. Dazu wurde das Protein in somatischen Zellen exprimiert. In diesem experimentellem Ansatz polymerisierte SYCP1 autonom zu filamentoesen Strukturen, welche sich auf ultrastruktureller Ebene als alternierende elektronendichte Balken offenbarten, die ueber TFs verbunden waren. Dieser Aufbau glich parallel aneinander gereihten Stapeln von SCs, so genannten Polykomplexen (PCs). Die Analyse der Orientierung der SYCP1 Molekuele innerhalb der PCs erwies, dass diese hochorganisiert vorliegen und die Organisation von SYCP1 innerhalb von PCs und SCs identisch ist. Folglich kann sich SYCP1 sogar in Abwesenheit anderer SC-Proteine zu Strukturen zusammenlagern, die der CR entsprechen und muss dementsprechend beim Aufbau der CR des SC den grundlegenden Faktor darstellen. Für eine genauere Analyse wurden ausgewaehlte Mutanten von SYCP1 exprimiert. Moleküle mit modifizierter Laenge der zentralen alpha-helikalen Domaene resultierten in der Bildung von PCs mit veränderter Weite der CR. Dies beweist, dass die „Coiled-Coil“-Domaene den Abstand der CR eines PC bestimmt und impliziert dieselbe Funktion in der SC-Bildung. Darueber hinaus wurde gezeigt, dass SYCP1 Molekuele mit Deletion des nicht-helikalen N-Terminus immer noch in der Lage sind, PCs zu bilden, diese Eigenschaft aber stark eingeschraenkt ist. Das bezeugt die Bedeutung des N-Terminus sowohl in der PC-Bildung als auch im Aufbau des CE von SCs, weist aber dabei auch dem vorderen Teil der „Coiled-Coil“-Domaene eine wichtige Rolle zu. Im Gegensatz dazu war bei Mutanten mit Deletion des nicht-helikalen C-Terminus die PC-Bildung vollstaendig blockiert, was auf eine große Bedeutung dieser Domaene fuer die Polymerisation hinweist. Ein weiterer Hauptgegenstand der Arbeit war die Charakterisierung von Bindungspartnern von SYCP1. Über Immungoldlokalisation auf Maushoden konnten die Proteine Syce1 und Cesc1 als erste ausschliessliche Komponenten des CE des SC bestimmt werden. Zusaetzlich wurde die Interaktion dieser Proteine mit dem N-Terminus von SYCP1 verifiziert. SYCP1 bildet also die Grundstruktur des CE aus und rekrutiert Syce1 und Cesc1.
Results are presented of cloning cDNA of procine growth hormone, analysis of its primary structure, and creation of a construction capable of expression of this cDNA in Esqheriahia coti cells. It is shown that in the population of mRNA coding porcine growth hormone, heterogeneity is noted which is manifested not only at the level of the nucleotide sequence, but also is reflected in the amino acid sequence of the mature hormone.
Besides external characteristics and reading a piece of DNA (barcode), the DNA weight per nucleus (genome size) via flow cytometry is a key value to detect species and hybrids and determine ploidy. In addition, the DNA weight appears to be related to various properties, such as the size of the cell and the nucleus, the duration of mitosis and meiosis and the generation time. Sometimes it is even possible to distinguish between groups or sections, which can lead to new classification of the genera. The variation in DNA weight is also useful to analyze biodiversity, genome evolution and relationships between related taxa. Moreover, it is important to know how large a genome is before one determines the base sequence of the DNA of a plant. Flow cytometry is also important for understanding fundamental processes in plants such as growth and development and recognizing chimeras. In the literature, DNA weight measurements are usually limited to one genus and often only locally (Siljak et al. 2010; Bai et al. 2012). In this study, however, it was decided to investigate all vascular plants from one country. This can also contribute to the protection of rare plants. This study is the first flora in the world whose weight of DNA per nucleus and peak patterns has been determined. More than 6400 plants, representing more than 2350 (sub)species (more than 90%) have been collected, thanks to the help of almost 100 volunteers of Floristisch Onderzoek Nederland (Floron). Multiple specimens of many species have therefore been measured, preferably from different populations, in some cases more than fifty. For 1370 species, these values were not previously published. Moreover, a good number of the remaining 45% are new for The Netherlands. In principle, each species has a fixed weight of DNA per nucleus. It has also been found that, especially between the genera, there are strong differences in the number of peaks that determine the DNA weight, from one to five peaks. This indicates that in a plant or organ there are sometimes nuclei with multiples of its standard DNA weight (multiple ploidy levels). It is impossible to show graphs of more than 2350 species. Therefore, we have chosen to show the peak pattern in a new way in a short formula. Within most genera there are clear differences in the DNA weights per nucleus between the species, in some other genera the DNA weight is hardly variable. Based on about twenty genera that were previously measured completely in most cases (‘t Hart et al. 2003: Veldkamp and Zonneveld 2011; Soes et al. 2012; Dirkse et al. 2014, 2015; Verloove et al. 2017; Zonneveld [et al.] 2000−2018), it can be noted that even if all species of a genus have the same number of chromosomes, there can still be a difference of up to three times in the weight of the DNA. Therefore, a twice larger DNA weight does not have to indicate four sets of chromosomes. Finally, this research has also found clues to examine further the current taxonomy of a number of species or genera.
Untersuchungen zum Scherstress-responsiven Element des PDGF-Promotors Zellen sind im menschlichen Organismus ständig mechanischen Kräften ausgesetzt. Eine dieser Kräfte ist die spezielle Schubkraft, die durch Flüssigkeitsstrom auf Zellen ausgeübt wird und als Scherstress bezeichnet wird. Die Stimulation von Zellen durch Scherstress führt zu diversen Reaktionen, die von Wanderungsvorgängen, über Umbau des Zytoskeletts bis hin zur gesteigerten Expression verschiedener Gene reichen. Für die Induktion des Gens der B-Kette des Plättchen-Wachstumsfaktors (PDGF) wurde von Resnick 1993 ein sechs Basenpaare langes cis-aktives Scherstress-responsives Promotorelement (SSRE) identifiziert, das an der Transkriptionsinduktion des Gens durch Scherstress beteiligt ist. In der hier vorliegenden Arbeit sollten die Eigenschaften dieses SSRE gezeigt werden, indem ein Reportergen-Assay mit dem grün fluoreszierenden Proteins EGFP in Zellen der Linie ECV304 ausgetestet wurde. Eine Fragestellung der Arbeit bestand darin, zu prüfen ob das SSRE notwendig und ausreichend für die Scherstressresponsivität eines Promotors ist und ob die Expressionsstärke des Reporterproteins EGFP mit der Stärke und Dauer der Scherstressstimulation korreliert. Zudem sollte der Effekt verschiedenar-tiger pharmakologischer Substanzen auf den PDGF-Promotor unter Scherstress gezeigt werden. Dazu wurde ein neues arithmetisches Verfahren entwickelt, das erlaubt, Zellen unter Einwirkung von Scherkraft angemessen zu vermessen und statistisch auszuwerten. Durch den Einsatz des EGFP und einer hochsensitiven ICCD-Photonenkamera war es möglich, die Messung der Expressionskinetik des Reportergens in Echtzeit durchzufüh-ren. Dabei konnte gezeigt werden, dass in ECV304-Zellen unter Scherstress von 0,5, 12 und 30 dyn/cm² die Expression des intakten PDGF-Promotors mit der Stärke und Dauer der Scherstressstimulation korreliert. Dieser Effekt ist nicht zu beobachten, wenn das SSRE aus dem PDGF-Promotor entfernt wird. Ebenfalls zu keiner Steigerung der Genexpression kommt es, wenn das Reportergen hinter den Promotor des Cytomegalie-Virus (CMV) geschaltet ist, der kein bekanntes Scherstress-responsives Promotor-Element enthält. Inseriert man in den CMV-Promotor das SSRE, so zeigt der Promotor unter Scherstress-Einwirkung ebenfalls eine gesteiger-te Expression, die allerdings hinter der des PDGF-Vollkonstrukts zurückbleibt. Es konnte damit gezeigt werden, dass das SSRE in einen nicht Scherstress-responsiven Promotor verbracht diesen für Strömungskräfte suszeptibel macht. Der PDGF-Promotor enthält eine bekannte Phorbolester-Bindungsstelle. In der vorlie-genden Arbeit konnte gezeigt werden, dass ECV304-Zellen bei Inkubation in mit Phor-bolester versetztem Medium die Transkription des Reportergens deutlich steigern. Ebenfalls zu einer Steigerung der Expression des PDGF-Vollkonstrukts kommt es bei appliziertem Scherstress von 12 dyn/cm² und gleichzeitiger Hemmung der Proteinkinase C (PK-C). Zwar zeigt die Hemmung mit Chelerythrin eine schwächere Lichtintensitäts-zunahme, wie sie beim Einsatz von Calphostin C, beide Messungen ergaben aber eine Steigerung der Lichtintensität gegenüber der Messung des PDGF-Vollkonstrukts bei 12 dyn/cm² in normalem Kulturmedium.
Polyspezifische organische Kationentransporter (OCTs) der SLC22 Familie transportieren organische Kationen entlang eines elektrochemischen Gradienten und spielen eine entscheidende Rolle bei der Ausscheidung und Gewebeverteilung von endogenen organischen Kationen und bei der Aufnahme, Ausscheidung und Verteilung von kationischen Medikamenten und Toxinen. Zu den endogenen transportierten Substanzen gehört auch der Neurotransmitter Acetylcholin (ACh), der unter anderem in der menschlichen Haut eine wichtige Rolle in der Zelldifferenzierung und –proliferation spielt. Die dermatologischen Antiinfektiva Gentianaviolett (GV) und Brillantgrün (BG) aus der Gruppe der Triphenylmethanfarbstoffe werden in der Behandlung lokaler Wundinfektionen verwendet und zeigen im klinischen Gebrauch Nebenwirkungen wie Wundheilungsstörungen. Es konnte gezeigt werden, dass die Farbstoffe die OCTs konzentrationsabhängig hemmen und es wurden die Werte der halbmaximalen Hemmkonzentration ermittelt. Dabei zeigte sich, dass GV und BG zu den Hemmstoffen mit der höchsten Affinität zu den OCTs gehören. Ein Transport der Farbstoffe durch die OCTs konnte nicht nachgewiesen werden, die toxische Wirkung auf Keratinozyten in in-vitro Versuchen mit menschlichen Zellreihen wurde bestätigt. Ein Zusammenhang zwischen den beobachteten Wundheilungsstörungen unter der Therapie mit Triphenylmethanfarbstoffen und der Hemmung des ACh-Transportes durch die OCTs konnte nicht bestätigt werden.
Neurofilament depletion improves microtubule dynamics via modulation of Stat3/stathmin signaling

In neurons, microtubules form a dense array within axons, and the stability and function of this microtubule network is modulated by neurofilaments. Accumulation of neurofilaments has been observed in several forms of neurodegenerative diseases, but the mechanisms how elevated neurofilament levels destabilize axons are unknown so far. Here, we show that increased neurofilament expression in motor nerves of pmn mutant mice, a model of motoneuron disease, causes disturbed microtubule dynamics. The disease is caused by a point mutation in the tubulin-specific chaperone E (Tbce) gene, leading to an exchange of the most C-terminal amino acid tryptophan to glycine. As a consequence, the TBCE protein becomes instable which then results in destabilization of axonal microtubules and defects in axonal transport, in particular in motoneurons. Depletion of neurofilament increases the number and regrowth of microtubules in pmn mutant motoneurons and restores axon elongation. This effect is mediated by interaction of neurofilament with the stathmin complex. Accumulating neurofilaments associate with stathmin in axons of pmn mutant motoneurons. Depletion of neurofilament by Nefl knockout increases Stat3-stathmin interaction and stabilizes the microtubules in pmn mutant motoneurons. Consequently, counteracting enhanced neurofilament expression improves axonal maintenance and prolongs survival of pmn mutant mice. We propose that this mechanism could also be relevant for other neurodegenerative diseases in which neurofilament accumulation and loss of microtubules are prominent features.
3D cell culture models which closely resemble real human tissues are of high interest for disease modelling, drug screening as well as a deeper understanding of human developmental biology. Such structures are termed organoids. Within the last years, several human organoid models were described. These are usually stem cell derived, arise by self-organization, mimic mechanisms of normal tissue development, show typical organ morphogenesis and recapitulate at least some organ specific functions. Many tissues have been reproduced in vitro such as gut, liver, lung, kidney and brain. The resulting entities can be either derived from an adult stem cell population, or generated from pluripotent stem cells using a specific differentiation protocol. However, many organoid models only recapitulate the organs parenchyma but are devoid of stromal components such as blood vessels, connective tissue and inflammatory cells. Recent studies show that the incorporation of endothelial and mesenchymal cells into organoids improved their maturation and might be required to create fully functional micro-tissues, which will allow deeper insights into human embryogenesis as well as disease development and progression. In this review article, we will summarize and discuss recent works trying to incorporate stromal components into organoids, with a special focus on neural organoid models.
Organoids derived from human pluripotent stem cells are interesting models to study mechanisms of morphogenesis and promising platforms for disease modeling and drug screening. However, they mostly remain incomplete as they lack stroma, tissue resident immune cells and in particular vasculature, which create important niches during development and disease. We propose, that the directed incorporation of mesodermal progenitor cells (MPCs) into organoids will overcome the aforementioned limitations. In order to demonstrate the feasibility of the method, we generated complex human tumor as well as neural organoids. We show that the formed blood vessels display a hierarchic organization and mural cells are assembled into the vessel wall. Moreover, we demonstrate a typical blood vessel ultrastructure including endothelial cell-cell junctions, a basement membrane as well as luminal caveolae and microvesicles. We observe a high plasticity in the endothelial network, which expands, while the organoids grow and is responsive to anti-angiogenic compounds and pro-angiogenic conditions such as hypoxia. We show that vessels within tumor organoids connect to host vessels following transplantation. Remarkably, MPCs also deliver Iba1\(^+\) cells that infiltrate the neural tissue in a microglia-like manner.
Most humans become infected with human cytomegalovirus (HCMV). Typically, the immune system controls the infection, but the virus persists and can reactivate in states of immunodeficiency. While substantial information is available on the contribution of CD8 T cells and antibodies to anti-HCMV immunity, studies of the T\(_{H}\)1, T\(_{H}\)2, and T\(_{H}\)17 subsets have been limited by the low frequency of HCMV-specific CD4 T cells in peripheral blood mononuclear cell (PBMC). Using the enzyme-linked Immunospot\(^{®}\) assay (ELISPOT) that excels in low frequency measurements, we have established these in a sizable cohort of healthy HCMV controllers. Cytokine recall responses were seen in all seropositive donors. Specifically, interferon (IFN)-\({\gamma}\) and/or interleukin (IL)-17 were seen in isolation or with IL-4 in all test subjects. IL-4 recall did not occur in isolation. While the ratios of T\(_{H}\)1, T\(_{H}\)2, and T\(_{H}\)17 cells exhibited substantial variations between different individuals these ratios and the frequencies were relatively stable when tested in samples drawn up to five years apart. IFN-\({\gamma}\) and IL-2 co-expressing polyfunctional cells were seen in most subjects. Around half of the HCMV-specific CD4 cells were in a reversible state of exhaustion. The data provided here established the T\(_{H}\)1, T\(_{H}\)2, and T\(_{H}\)17 characteristic of the CD4 cells that convey immune protection for successful immune surveillance against which reactivity can be compared when the immune surveillance of HCMV fails.
There is a largely divergent body of literature regarding the relationship between Epstein-Barr virus (EBV) infection and brain inflammation in multiple sclerosis (MS). Here, we tested MS patients during relapse (n = 11) and in remission (n = 19) in addition to n = 22 healthy controls to study the correlation between the EBV- and brain-specific B cell response in the blood by enzyme-linked immunospot (ELISPOT) and enzyme-linked immunosorbent assay (ELISA). Cytomegalovirus (CMV) was used as a control antigen tested in n = 16 MS patients during relapse and in n = 35 patients in remission. Over the course of the study, n = 16 patients were untreated, while n = 33 patients received immunomodulatory therapy. The data show that there was a moderate correlation between the frequencies of EBV- and brain-reactive B cells in MS patients in remission. In addition we could detect a correlation between the B cell response to EBV and disease activity. There was no evidence of an EBV reactivation. Interestingly, there was also a correlation between the frequencies of CMV- and brain-specific B cells in MS patients experiencing an acute relapse and an elevated B cell response to CMV was associated with higher disease activity. The trend remained when excluding seronegative subjects but was non-significant. These data underline that viral infections might impact the immunopathology of MS, but the exact link between the two entities remains subject of controversy.
As soon as Peripheral Blood Mononuclear Cells (PBMC) are isolated from whole blood, some cells begin dying. The rate of apoptotic cell death is increased when PBMC are shipped, cryopreserved, or stored under suboptimal conditions. Apoptotic cells secrete cytokines that suppress inflammation while promoting phagocytosis. Increased numbers of apoptotic cells in PBMC may modulate T cell functions in antigen-triggered T cell assays. We assessed the effect of apoptotic bystander cells on a T cell ELISPOT assay by selectively inducing B cell apoptosis using α-CD20 mAbs. The presence of large numbers of apoptotic B cells did not affect T cell functionality. In contrast, when PBMC were stored under unfavorable conditions, leading to damage and apoptosis in the T cells as well as bystander cells, T cell functionality was greatly impaired. We observed that measuring the number of apoptotic cells before plating the PBMC into an ELISPOT assay did not reflect the extent of PBMC injury, but measuring apoptotic cell frequencies at the end of the assay did. Our data suggest that measuring the numbers of apoptotic cells prior to and post T cell assays may provide more stringent PBMC quality acceptance criteria than measurements done only prior to the start of the assay.
Bei der Multiplen Sklerose (MS) handelt es sich um eine Autoimmunerkrankung des zentralen Nervensystems (ZNS). Abhängig von der betroffenen ZNS-Region kann es zu vielfältigen Symptomen kommen. Neben neurologischen Symptomen verursacht durch ZNS-Läsionen leidet ein Großteil der MS-Patienten auch unter gastrointestinalen Funktionsstörungen. Diese gastrointestinalen Symptome wurden bisher eher auf Läsionen im Rückenmark zurückgeführt und nicht direkt in Verbindung mit der autoimmunen Ätiologie der Erkrankung gebracht.
In dieser Studie wurde das enterische Nervensystem (ENS) in einem B-Zell- und Antikörper-abhängigen Mausmodell der MS untersucht. Dafür wurde der Autoimmunprozess durch Immunisierung mit MP4, einem Fusionsprotein aus dem Myelin-Basischen-Protein (MBP) und dem Proteolipid-Protein (PLP), ausgelöst. Das ZNS und ENS wurden in den unterschiedlichen Erkrankungsstadien immunhistochemisch und elektronenmikroskopisch analysiert. Neben der Immunpathologie des ZNS konnte dabei eine Degeneration des ENS schon vor dem Einsetzen der ersten neurologischen Defizite nachgewiesen werden. Die ENS-Pathologie war antikörper-mediiert und ging einher mit einer verringerten gastrointestinalen Motilität sowie mit einer Gliose und Neurodegeneration des ENS.
Mithilfe von Immunpräzipitation und Massenspektrometrie konnten im ENS vier mögliche Zielstrukturen des Autoimmunprozesses identifiziert werden, was auf sog. epitope spreading hindeutet. Auch im Plasma von MS-Patienten konnten Antikörper gegen drei dieser Antigene nachgewiesen werden. Des Weiteren zeigten sich in Kolon-Resektaten von MS-Patienten erste Ansätze einer Neurodegeneration und Gliose des ENS.
In dieser Studie wurde zum ersten Mal ein direkter Zusammenhang zwischen der Autoimmunreaktion gegen das ZNS und einer simultanen Reaktion gegen das ENS gezeigt. Dies kann einen Paradigmenwechsel im Verständnis der Immunpathogenese der MS anstoßen und neue therapeutische und diagnostische Ansätze initiieren.
Clinical prognosis of metastasized colorectal carcinoma (CRC) is still not at desired levels and novel drugs are needed. Here, we focused on the multi-tyrosine kinase inhibitor E7080 (Lenvatinib) and assessed its therapeutic efficacy against human CRC cell lines in vitro and human CRC xenografts in vivo. The effect of E7080 on cell viability was examined on 10 humanCRCcell lines and humanendothelial cells (HUVEC). The inhibitory effect of E7080 on VEGF-induced angiogenesis was studied in an ex vivo mouse aortic ring angiogenesis assay. In addition, the efficacy of E7080 against xenografts derived fromCRC cell lines and CRC patient resection specimenswithmutated KRASwas investigated in vivo. Arelatively low cytotoxic effect of E7080 on CRC cell viabilitywas observed in vitro. Endothelial cells (HUVEC)weremore susceptible to the incubation with E7080. This is in line with the observation that E7080 demonstrated an anti-angiogenic effect in a three-dimensional ex vivo mouse aortic ring angiogenesis assay. E7080 effectively disrupted CRC cell-mediated VEGF-stimulated growth of HUVEC in vitro. Daily in vivo treatment with E7080 (5 mg/kg) significantly delayed the growth of KRAS mutated CRC xenografts with decreased density of tumor-associated vessel formations and without tumor regression. This observation is in line with results that E7080 did not significantly reduce the number of Ki67-positive cells in CRC xenografts. The results suggest antiangiogenic activity of E7080 at a dosage thatwas well tolerated by nudemice. E7080 may provide therapeutic benefits in the treatment of CRC with mutated KRAS.
Die Barriereeigenschaften des Gefäßendothels werden durch Verschluß- und Adhärenskontakte vermittelt. Das Ca2+-abhängige Zelladhäsionsmolekül VE-Cadherin vermittelt in Adhärenskontakten die Adhäsion benachbarter Endothelzellen. Es wurde vermutet, daß die extrazelluläre Ca2+-Konzentration und das intrazelluläre Aktinfilamentsystem die Adhäsionseigenschaften von VE-Cadherin verändern können. Daher wurden diese Einflußfaktoren mit Hilfe der Laserpinzetten-Technik untersucht. Hierzu wurden Latex-Mikroperlen mit rekombinanten VE-Cadherin-Fc-Molekülen beschichtet, die damit an VE-Cadherin-Moleküle von Endothelzellen binden und Zell-Zell-Kontakte simulieren konnten. Es zeigte sich, daß die ausschließlich durch VE-Cadherin vermittelte Interaktion zwischen Mikroperlen und Endothelzellen direkt von der extrazellulären Ca2+-Konzentration abhängig war und sich durch eine s-förmige Titrationskurve beschreiben ließ: Die Bindungshäufigkeit der Mikroperlen war bei Ca2+-Konzentrationen nahe 0,0 mM gering (26-27 %), nahm ab 0,8 mM stark zu (38 %) und erreichte bei 1,8 mM ein Maximum (65 %). Halbmaximale Bindung (KD) wurde bei 1,1 mM Ca2+ erreicht. Die Bindung war hochkooperativ (Hill Koeffizient nH = 4,6). Um die Eigenschaften des Aktinfilamentsystems zu verändern, wurden die Zellen mit Cytochalasin B, Cytochalasin D und dem Ca2+-Ionophor A 23187 inkubiert. Dabei nahm die Bindungshäufigkeit der Mikroperlen deutlich gegenüber Kontrollbedingungen ab. Es wurde gefolgert, daß ein intaktes Aktinfilamentsystem unmittelbar die Interaktion zwischen VE-Cadherin-Molekülen stärkte. Die Ergebnisse dieser Arbeit liefern damit neue Erkenntnisse über die Eigenschaften von VE-Cadherin: Die Adhäsion dieses Moleküls wird im physiologischen Ca2+-Bereich reguliert und ist direkt von einem intakten Aktinfilamentsystem abhängig. Es ist vorstellbar, daß die durch VE-Cadherin vermittelten Barriereeigenschaften des Endothels in vivo durch ähnliche Mechanismen reguliert werden. Ein Abfall der Ca2+-Konzentration im Interzellularspalt unter den für die Adhäsion kritischen Wert von 1,1 mM könnte durch Agonist-vermittelte Öffnung von Ca2+-Kanälen erfolgen. Eingeströmtes Ca2+ könnte seinerseits über Aktivierung von Gelsolin zur Fragmentation von Aktinfilamenten führen und so die Adhäsion weiter schwächen.
Aims: Although mortality rate is very high, diagnosis of acute myocarditis remains challenging with conventional tests. We aimed to elucidate the potential role of longitudinal 2-Deoxy-2-\(^{18}\)F-fluoro-D-glucose (\(^{18}\)F-FDG) positron emission tomography (PET) inflammation monitoring in a rat model of experimental autoimmune myocarditis.
Methods and results: Autoimmune myocarditis was induced in Lewis rats by immunizing with porcine cardiac myosin emulsified in complete Freund’s adjuvant. Time course of disease was assessed by longitudinal \(^{18}\)F-FDG PET imaging. A correlative analysis between in- and ex vivo \(^{18}\)F-FDG signalling and macrophage infiltration using CD68 staining was conducted. Finally, immunohistochemistry analysis of the cell-adhesion markers CD34 and CD44 was performed at different disease stages determined by longitudinal \(^{18}\)F-FDG PET imaging. After immunization, myocarditis rats revealed a temporal increase in 18F-FDG uptake (peaked at week 3), which was followed by a rapid decline thereafter. Localization of CD68 positive cells was well correlated with in vivo \(^{18}\)F-FDG PET signalling (R\(^2\) = 0.92) as well as with ex vivo 18F-FDG autoradiography (R\(^2\) = 0.9, P < 0.001, respectively). CD44 positivity was primarily observed at tissue samples obtained at acute phase (i.e. at peak 18F-FDG uptake), while CD34-positive staining areas were predominantly identified in samples harvested at both sub-acute and chronic phases (i.e. at \(^{18}\)F-FDG decrease).
Conclusion: \(^{18}\)F-FDG PET imaging can provide non-invasive serial monitoring of cardiac inflammation in a rat model of acute myocarditis.
The parotid gland is one of the major salivary glands producing a serous secretion, and it plays an essential role in the digestive and immune systems. Knowledge of peroxisomes in the human parotid gland is minimal; furthermore, the peroxisomal compartment and its enzyme composition in the different cell types of the human parotid gland have never been subjected to a detailed investigation. Therefore, we performed a comprehensive analysis of peroxisomes in the human parotid gland’s striated duct and acinar cells. We combined biochemical techniques with various light and electron microscopy techniques to determine the localization of parotid secretory proteins and different peroxisomal marker proteins in parotid gland tissue. Moreover, we analyzed the mRNA of numerous gene encoding proteins localized in peroxisomes using real-time quantitative PCR. The results confirm the presence of peroxisomes in all striated duct and acinar cells of the human parotid gland. Immunofluorescence analyses for various peroxisomal proteins showed a higher abundance and more intense staining in striated duct cells compared to acinar cells. Moreover, human parotid glands comprise high quantities of catalase and other antioxidative enzymes in discrete subcellular regions, suggesting their role in protection against oxidative stress. This study provides the first thorough description of parotid peroxisomes in different parotid cell types of healthy human tissue.
A growing body of scientific evidence indicates that protein homeostasis, also designated as proteostasis, is causatively linked to chronic diabetic nephropathy (DN). Experimental studies have demonstrated that the insulin signaling in podocytes maintain the homeostatic unfolded protein response (UPR). Insulin signaling via the insulin receptor non-canonically activates the spliced X-box binding protein-1 (sXBP1), a highly conserved endoplasmic reticulum (ER) transcription factor, which regulates the expression of genes that control proteostasis. Defective insulin signaling in mouse models of diabetes or the genetic disruption of the insulin signaling pathway in podocytes propagates hyperglycemia induced maladaptive UPR and DN. Insulin resistance in podocytes specifically promotes activating transcription factor 6 (ATF6) dependent pathogenic UPR. Akin to insulin, recent studies have identified that the cytoprotective effect of anticoagulant serine protease-activated protein C (aPC) in DN is mediated by sXBP1. In mouse models of DN, treatment with chemical chaperones that improve protein folding provides an additional benefit on top of currently used ACE inhibitors. Understanding the molecular mechanisms that transmute renal cell specific adaptive responses and that deteriorate renal function in diabetes will enable researchers to develop new therapeutic regimens for DN. Within this review, we focus on the current understanding of homeostatic mechanisms by which UPR is regulated in DN.
Megakaryocytes (MKs) release platelets into the lumen of bone marrow (BM) sinusoids while remaining to reside within the BM. The morphogenetic events of this complex process are still not fully understood. We combined confocal laser scanning microscopy with transmission and serial block-face scanning electron microscopy followed by 3D-reconstruction on mouse BM tissue sections. These analyses revealed that MKs in close vicinity to BM sinusoid (BMS) wall first induce the lateral retraction of CXCL12-abundant reticular (CAR) cells (CAR), followed by basal lamina (BL) degradation enabling direct MK-sinusoidal endothelial cells (SECs) interaction. Subsequently, an endothelial engulfment starts that contains a large MK protrusion. Then, MK protrusions penetrate the SEC, transmigrate into the BMS lumen and form proplatelets that are in direct contact to the SEC surface. Furthermore, such processes are induced on several sites, as observed by 3D reconstructions. Our data demonstrate that MKs in interaction with CAR-cells actively induce BMS wall alterations, including CAR-cell retraction, BL degradation, and SEC engulfment containing a large MK protrusion. This results in SEC penetration enabling the migration of MK protrusion into the BMS lumen where proplatelets that are adherent to the luminal SEC surface are formed and contribute to platelet release into the blood circulation.
Der epithelialen Präsenz des Carcinoembryonic antigen-related cell adhesion molecule-1 (CEACAM1) in Prostatadrüsen wird eine tumorsupprimierende Funktion zugeschrieben. Maligne Veränderungen des Prostatadrüsenepithels bei einem PCa führen zu einer Abnahme der epithelialen CEACAM1-Expression, zu einem Verlust der Zellpolarität und zu einer erhöhten Zellproliferation (prostatische intraepitheliale Neoplasie (PIN)). Während des PIN-Stadiums exprimieren benachbarte Blut- und Lymphgefäße CEACAM1. CEACAM1 selbst wirkt pro-angiogen und stimuliert die Gefäßneubildung und auch die Neubildung von Lymphgefäßen, Lymphangiogenese. Seine Rolle in der Tumor-Lymphangiogenese und dadurch bedingten Metastasierung von Tumoren wurde bisher nicht ausreichend geklärt. Ziel dieser Arbeit war es, die Rolle von CEACAM1 bei der lymphogenen PCa-Metastasierung anhand von immunhistochemischen (IHC) Analysen am humanem PCa-Prostata- und Lymphknoten-(LN)-Gewebe, sowie im Mausmodell zu analysieren.
Laut den Immunfluoreszenzanalysen traten in den PIN-Arealen signifikant mehr CEACAM1-positive Blut- und Lymphgefäße auf, als in den darauffolgenden Tumorstadien. Weiter wurde eine CEACAM1-Expression in LN-Sinusgewebe bereits bei Niedrig-Risiko-Patienten (pN0) detektiert. Diese frühe CEACAM1-Expression trat auch in den LN im PCa-Mausmodell auf. Weiter wurde im LN-Gewebe von „Hoch-Risiko“-Patienten (pN1) eine luminale CEACAM1-Expression innerhalb der aus Tumorzellen bestehenden Drüsen beobachtet, die mit der CEACAM1-Expression in nativen Prostatadrüsen vergleichbar ist. Auch das angiogen-aktivierte Gefäßendothel von pN0- und pN1-LN war CEACAM1-positiv. Bei Hoch-Risiko-Patienten (pN1) nahmen die CEACAM1-positiven Blut- und Lymphgefäße im Tumorstroma mit zunehmender Dedifferenzierung des Gewebes ab. Die CEACAM1/PSA-Doppelimmun-fluoreszenzanalysen ergaben eine heterogene Expression der beiden Marker bei Intermediate-risk-Patienten und mit zunehmender Dedifferenzierung des Tumorgewebes einen epithelialen Verlust der CEACAM1-Expression in den PSA-positiven G3-Tumordrüsen. Das Fehlen von PSA in pN0-LN und die nachweisbare Expression von PSA in pN1-LN bestätigten PSA als geeigneten PCa-Zellmarker in LN. In pN1-LN ohne Drüsenbildung traten Zellansammlungen mit einer nach außen gerichteten CEACAM1-positiven Front und einem im Zentrum liegenden PSA-positiven Bereich auf. Diese Befunde belegen einen Zusammenhang zwischen der endothelialen CEACAM1-Expression im Sinus und der Mikrometastasierungswahrscheinlichkeit im pN0-LN-Gewebe von PCa-Patienten. Potentiell lässt sich daher im Niedrig-Risiko-PCa-Patientenkollektiv über eine CEACAM1-Bestimmung in LN das Risiko für eine Metastasierung frühzeitig erkennen.
The influence of microsomal (mAHH) and nuclear (nAHH) aryl hydrocarbon hydroxylase activity on the covalent binding of t:titiated benzo(a)pyrene to rat liver DNA was evaluated in vivo. Induction ofmAHH was obtained after phenobarbitone treatment (180% of control), which increased DNA binding to 210%, but left the nAHH unchanged. mAHH and nAHH were slightly indilced with dieldrin (130% and 120%), but the binding remairred unchanged. The increasing effect of mAHlt as weil as the possibly decreasing effect of nAHH induction on the binding became obvious when the data of 11 individual rats were used to solve the equation Binding = aX(mAHH) + bX(nAHH) + c. Multiple linear regression analysis resulted in positive values for a and c, a negative value for b, and a multiple correlation coefficient R = 0.82. An influence of other enzymes involved in the metabolism of benzo(a)pyrene cannot be excluded. The Study shows clearly that the binding of a foreign compound to DNA in vivo is not only dependent on microsomal enzyme activities but also on nuclear activities even if the latter are considerably lower than those of mic'rosomes.
Desmosomen sind Zell-Zell-Kontakte, die eine starke interzelluläre Haftung vermitteln. Sie sind daher besonders wichtig für die Integrität von Geweben wie der Haut, die laufend einer starken mechanischen Beanspruchung ausgesetzt sind. Pemphigus vulgaris ist eine Autoimmundermatose, die zur Ausbildung schlaffer Blasen durch Spaltbildung in der Epidermis führt. Als ursächlich dafür wurden Autoantikörper gegen die desmosomalen Cadherine Dsg1 und 3 herausgestellt, die in Desmosomen vorkommen. cAMP ist ein wichtiger Botenstoff des Zellstoffwechsels und an der Regulierung und Modulation einer Vielzahl von zellulären Prozessen beteiligt, darunter auch die Stabilisierung der Endothelbarriere über Stärkung der Haftung eines klassischen Cadherins, nämlich VE-Cadherin.
In Anknüpfung an die vorliegenden Daten aus Pemphigus- und Endothelforschung beschäftigt sich diese Arbeit mit der Rolle von cAMP bei Pemphigus vulgaris. Es wurde in Keratinozytenkultur sowie im neonatalen Pemphigus-Mausmodell untersucht, ob die Erhöhung der intrazellulären cAMP-Spiegel einen Einfluss auf PV-IgG-induzierte morphologische und funktionelle Veränderungen hat.
Eine Erhöhung des intrazellulären cAMP-Spiegels konnte sowohl in vitro als auch in vivo als protektiv herausgestellt werden. In Keratinozytenkultur konnte gezeigt werden, dass eine Erhöhung des intrazellulären cAMP-Spiegels durch Forskolin/Rolipram oder Isoproterenol in der Lage war, die PV-IgG-induzierten morphologischen Veränderungen, die Dsg3-Depletion, sowie den Adhäsionsverlust zu blockieren. Weiterhin konnte die Blasenbildung in vivo durch cAMP-Erhöhung vollständig verhindert werden.
Im Anschluss wurde untersucht, ob die Inkubation mit PV-IgGs einen Einfluss auf die intrazellulären cAMP-Spiegel in vitro hat. Dabei konnte gezeigt werden, dass die Zellen mit einer Erhöhung der cAMP-Spiegel reagieren, wenn auch in einem geringeren Ausmaß als durch die eingesetzten Mediatoren. Somit kann cAMP als Rettungsmechanismus der Zellen angesehen werden und es wurde daraufhin der Einfluss von cAMP auf die Regeneration von Keratinozyten nach PV-IgG-Inkubation untersucht. Dieser Prozess konnte durch eine cAMP-Erhöhung verbessert werden und erwies sich als partiell abhängig von PKA. Schlussendlich konnte nachgewiesen werden, dass cAMP in vitro wie in vivo über die Blockade der p38MAPK-Aktivierung protektiv wirkt.
Zusammenfassend konnte so ein neuer Einblick in die zelluläre Antwort von Keratinozyten auf Pemphigus-Autoantikörperbindung gewonnen werden. Dieser könnte auch im Hinblick auf die Entwicklung neuer therapeutischer Strategien bei Pemphigus vulgaris wichtig sein.
The RS1 protein, a 67 kDa protein, encoded by an intronless single copy gene that was only detected in mammals, mediates transcriptional and post-transcriptional down-regulation of the sodium-D-glucose co-transporter SGLT1. The short-term post-transcriptional down-regulation of SGTL1 by RS1 has been shown to occur at the trans-Golgi network (TGN). In the present study, two tripeptides from the human RS1 protein (hRS1), GlnCysPro and GlnSerPro, that induce the post-transcriptional down-regulation of SGLT1 at the TGN, were identified. The application of the tripeptides led to 40-50% reduction of the amount of the SGLT1 protein in the plasma membrane, which correlated to the degree of decrease in SGLT1-mediated glucose transport. For the short-term down-regulation of SGLT1 by the tripeptides, the effective intracellular concentrations IC50 values of 2.0 nM (GlnCysPro, QCP) and 0.16 nM (GlnSerPro, QSP) were estimated. The observed down-regulation of SGLT1 by the tripeptides QCP and QSP, similar to hRS1 protein, was attenuated by different intracellular monosaccharides including nonmetabolized methyl-α-D-glucopyranoside and 2-deoxyglucose. On the contrary, the short-term inhibition of the hOCT2 by QCP could only be observed after rising of intracellular concentration of AMG. QCP and QSP are transported by H+-peptide cotransporter PEPT1 that is co-located with SGLT1 in the small intestinal enterocytes and thereafter effectively down-regulate hSGLT1-mediated transport of AMG. The data indicates that orally applied tripeptides QCP or QSP can be used to down-regulate D-glucose absorption in small intestine and used for treatment of obesity and diabetes mellitus.
Contribution of adventitia-derived stem and progenitor cells to new vessel formation in tumors

Blocking tumor vascularization has not yet come to fruition to the extent it was hoped for, as angiogenesis inhibitors have shown only partial success in the clinic. We hypothesized that under- appreciated vascular wall-resident stem and progenitor cells (VW-SPCs) might be involved in tumor vascularization and influence effectiveness of anti-angiogenic therapy. Indeed, in patient samples, we observed that vascular adventitia-resident CD34\(^+\) VW-SPCs are recruited to tumors in situ from co-opted vessels. To elucidate this in detail, we established an ex vivo model using concomitant embedding of multi-cellular tumor spheroids (MCTS) and mouse aortic rings (ARs) into collagen
gels, similar to the so-called aortic ring assay (ARA). Moreover, ARA was modified by removing the ARs’ adventitia that harbors VW-SPCs. Thus, this model enabled distinguishing the contribution of VW-SPCs from that of mature endothelial cells (ECs) to new vessel formation. Our results show that the formation of capillary-like sprouts is considerably delayed, and their number and network formation were significantly reduced by removing the adventitia. Substituting iPSC-derived neural spheroids for MCTS resulted in distinct sprouting patterns that were also strongly influenced by the
presence or absence of VW-SPCs, also underlying the involvement of these cells in non-pathological vascularization. Our data suggest that more comprehensive approaches are needed in order to block all of the mechanisms contributing to tumor vascularization.
Ongoing research to fight cancer, one of the dominant diseases of the 21st century has led to big progress especially when it comes to understanding the tumor growth and metastasis. This includes the discovery of the molecular mechanisms of tumor vascularization, which is critically required for establishment of tumor metastasis.
Formation of new blood vessels is the first step in tumor vascularization. Therefore, understanding the molecular and cellular basis of tumor vascularization attracted a significant effort studying in biomedical research. The blood vessels for supplying tumor can be formed by sprouting from pre-existing vessels, a process called angiogenesis, or by vasculogenesis, that is de novo formation of blood vessels from not fully differentiated progenitor cell populations. Vasculogenic endothelial progenitor cells (EPCs) can either be activated from populations in the bone marrow reaching the pathological region via the circulation or they can be recruited from local reservoirs. Neovessel formation influences tumor progression, hence therapeutic response model systems of angiogenesis/vasculogenesis are necessary to study the underlying mechanisms. Although, initially the research in this area focused more on angiogenesis, it is now well understood that both angiogenesis and postnatal vasculogenesis contribute to neovessel formation in adult under both most pathological as well as physiological conditions. Studies in the last two decades demonstrate that in addition to the intimal layer of fully differentiated mature endothelial cells (ECs) and various smaller supplying vessels (vasa vasorum) that can serve as a source for new vessels by angiogenesis, especially the adventitia of large and medium size blood vessels harbors various vascular wall-resident stem and progenitor cells (VW-SPCs) populations that serve as a source for new vessels by postnatal vasculogenesis. However, little is known about the potential role of VW-SPCs in tumor vascularization.
To this end, the present work started first to establish a modified aortic ring assay (ARA) using mouse aorta in order to study the contribution of vascular adventitia-resident VW-SPCs to neovascularization in general and in presence of tumor cells. ARA is already established an ex vivo model for neovascularization allows to study the morphogenetic events of complex new vessel formation that includes all layers of mature blood vessels, a significant advantage over the assays that employ monolayer endothelial cell cultures. Moreover, in contrast to assays employing endothelial cells monocultures, both angiogenic and vasculogenic events take place during new vessel formation in ARA although the exact contribution of these two processes to new vessel formation cannot be easily distinguished in conventional ARA. Thus, in this study, a modified protocol for the ARA (mdARA) was established by either removing or keeping the aortic adventitia in place. The mdARA allows to distinguish the role of VW-SPCs from those of other aortic layers. The present data show that angiogenic sprouting from mature aortic endothelium was markedly delayed when the adventitial layer was removed. Furthermore, the network between the capillary-like sprouts was significantly reduced in absence of aortic adventitia. Moreover, the stabilization of new sprouts by assembling the NG2+ pericyte-like cells that enwrapped the endothelial sprouts from the outside was improved when the adventitial layer remained in place.
Next, mimicking the tumor-vessel adventitia-interaction, multicellular tumor spheroids (MCTS) and aortic rings (ARs) with or without adventitia of C57BL/6-Tg (UBC-GFP) mice were confronted within the collagen gel and cultured ex vivo. This 3D model enabled analysis of the mobilization, migration and capillary-like sprouts formation by VW-SPCs within tumor-vessel wall-interface in comparison to tumor-free side of the ARs. Interestingly, while MCTS preferred the uptake of single vascular adventitia-derived cells, neural spheroids were directly penetrated by capillary-like structures that were sprouted from the aortic adventitia. In summary, the model established in this work allows to study new vessel formation by both postnatal vasculogenesis and angiogenesis under same conditions. It can be applied in various mouse models including reporter mouse models, e.g. Cxcr1 CreER+/mTmG+/- mice, in which GFP-marked macrophages of the vessel wall were directly observed as they mobilized from their niche and migrated into collagen gel. Another benefit of the model is that it can be used for testing different factors such as small molecules, growth factors, cytokines, and drugs with both pro- and anti-angiogenic/vasculogenic effects.
rOCT1 und hOCT2 sind zwei homologe Transportproteine für organische Kationen, die in Niere und Leber den ersten Schritt der transepithelialen Sekretion von Metaboliten und Xenobiotika vermitteln. Eines ihrer wesentlichen Charakteristika ist neben der Potentialabhängigkeit die Polyspezifität hinsichtlich Transportsubstraten und Hemmstoffen. Beide Transporter können als Uniporter klassifiziert werden, d.h. sie besitzen keine obligate Kopplung an ein Austausch- oder Cosubstrat wie z.B. Natrium, Protonen oder andere organische Kationen. Darüberhinaus zeigen sie das für Transportproteine typische und von Kanälen distinkte Charakteristikum der trans-Stimulierbarkeit. Mit rOCT1 konnten erstmals für einen Prototypen der Transportersuperfamilie SLC22 nähere Aufschlüsse über den Transportmechanismus erhalten werden. Zum einen wurde nachgewiesen, daß rOCT1 eine direktionale Asymmetrie besitzt, d.h. unter Sättigungsbedingungen besteht eine kinetische Bevorzugung der Transportrichtung von extra- nach intrazellulär um den Faktor zwei bis fünf. Zum anderen wurden anhand von rOCT1 neue Erkenntnisse zum Bindungs- und Interaktionsverhalten von Hemmstoffen und Transportsubstraten in Abhängigkeit von der Transportrichtung gewonnen. Hierbei erscheint das bisherige Modell von topologisch festgelegter kompetitiver und allosterischer Hemmung zu stark vereinfacht und nicht zutreffend. Die Bindung von Transportsubstraten und die dadurch induzierten Konformationsänderungen scheinen selbst die Bindungseigenschaften von Hemmstoffen in mehreren Zuständen zu beeinflussen. Auch scheinen Transport- und Ionenleitfähigkeit von OCT zu differenzieren zu sein. Zur Klärung der Fragestellungen wurde das Expressionsmodell Xenopus-Oozyte durch die Etablierung der sogenannten Effluxmethodik funktionell und methodisch wesentlich erweitert.
A recombinant plasmid was constructed containing the gene for bovine growth hormone joinea with the regulatory region and the region coding the signal sequence of the Escherichia coli alkaline phosphatase gene. In conditions of phosphorus starvation, which c~s derepression of alkaline phosphatase, expression was shown of the gene for bovine growth hormone, in addition to partial processing and secretion of protein into periplasm.
Eine Reihe mehrtägiger Suchexkur-sionen / Transekte in verschiedene Regionen Bayerns in den Jahren 2011 bis 2014 waren der Gattung Taraxacum gewidmet. Unter den gesammelten und beobachteten Arten ist Taraxacum broddesonii (sect. Ruderalia / Taraxacum) neu für Deutschland. Neu für Bayern sind Taraxacum fusciflorum, marklundii, spiculatum (sect. Hamata) und Taraxacum acroglossum, atroviride, clarum, floccosum, freticola, glossodon, hemicyclum, homoschistum, infuscatum, intumescens, lacinulatum, leucopodum, lundense, ottonis, pallidipes, praestabile, pseudoretroflexum, pulverulentum, saxonicum, sellandii, sundbergii, uncidentatum, uniforme, violaceinervosum (sect. Ruderalia / Taraxacum). Taraxacum lojoënse wird als ältester und korrekter Name für T. lippertianum und T. matricium und wahrscheinlich auch für T. ampelophytum und T. debrayi angesehen. Seltenere Arten sind abgebildet.
Local axonal function of STAT3 rescues axon degeneration in the pmn model of motoneuron disease

Axonal maintenance, plasticity, and regeneration are influenced by signals from neighboring cells, in particular Schwann cells of the peripheral nervous system. Schwann cells produce neurotrophic factors, but the mechanisms by which ciliary neurotrophic factor (CNTF) and other neurotrophic molecules modify the axonal cytoskeleton are not well understood. In this paper, we show that activated signal transducer and activator of transcription-3 (STAT3), an intracellular mediator of the effects of CNTF and other neurotrophic cytokines, acts locally in axons of motoneurons to modify the tubulin cytoskeleton. Specifically, we show that activated STAT3 interacted with stathmin and inhibited its microtubule-destabilizing activity. Thus, ectopic CNTF-mediated activation of STAT3 restored axon elongation and maintenance in motoneurons from progressive motor neuronopathy mutant mice, a mouse model of motoneuron disease. This mechanism could also be relevant for other neurodegenerative diseases and provide a target for new therapies for axonal degeneration.
Zur SLC22-Genfamilie von Karnitin- und organischen Ionentransportern gehören u. a. auch die organischen Kationentransporter der Ratte rOCT1 und rOCT2 sowie deren humane Analoga. Diese funtionell bereits gut charakterisierten Proteine werden in verschiedenen Geweben exprimiert und transportieren endogene und pharmakologisch relevante Substanzen. Die Aufklärung des Transportmechanismus und der Regulation sind Gegenstand intensiver Forschungsbemühungen. In dieser Arbeit wurde durch elekrophysiologische Methoden, die modifiziert auch zur Messung von Membrankapazitäten und des Strom-Teilchen-Aufnahmeverhältnisses (CFR) eingesetzt wurden, der Effekt des ungeladenen, membranpermeablen SH-Gruppenregenz MMTS auf den in Xenopus laevis-Oozyten exprimierten rOCT1 untersucht. Durch MMTS kam es zu einer Aktivierung von substratinduzierten Strömen und Kapazitätsänderungen, einer unterschiedlichen Veränderung von Substrat- und Inhibitoraffinitäten sowie einer Zunahme der absoluten Oozytenmembrankapazität. Die CFR und die Spannungsabhängigkeit änderten sich nicht. An rOCT2 konnten teils gleich-, teils gegensinnige Veränderungen nach MMTS-Exposition gezeigt werden. Diese Phänomene können am ehesten durch eine Konformationsänderung der polyvalenten Substratbindungsregion, einen verstärken Membraneinbau der Transportermoleküle und eine gesteigerte Transportrate des Einzeltransporters erklärt werden. Durch die Untersuchung von Mutanten konnten die Cysteine 322 und 451 als essentiell für den MMTS-Effekt identifiziert werden. Die Daten deuten darauf hin, dass die Modifikation dieser Cysteine für die Effekte zwingend erforderlich ist. Möglicherweise ist aber ein Xenopus-eigenes Regulatorprotein an der Entstehung der Effekte beteiligt, welches in die entsprechenden Proteinregionen des rOCT1 eingreift. Mit der Identifikation von Cystein 451 konnte ein weiterer Beweis für die wichtige Bedeutung der 10. Transmembrandomäne erbracht werden. Mit dem Cystein 322 wurde eine weitere wichtige Aminosäure für die Funktion und möglicherweise auch die Regulation von rOCT1 identifiziert.
Verschluss- und Adherenskontakte zwischen olfaktorischen Neuronen, olfaktorischen Gliazellen und den epithelialen Zellen des peripheren olfaktorischen Systems bestimmen Barriere- und Adhäsionseigenschaften im olfaktorischen Epithel, sowie die Kompartimentierung und Axonwachstumsprozesse in den Fila olfactoria. Zur Untersuchung der zellulären und subzellulären Lokalisation von Verschlusskontaktproteinen (Occludin, Claudin 1-5, Zonula occludens proteins (ZO) 1-3) und Adherenskontaktproteinen (N-Cadherin, E-Cadherin, alpha-, beta-, p120-Catenin) wurden immunhistochemische und immunelektronenmikroskipsche Verfahren an Gewebeschnitten des olfaktorischen Systems der Ratte durchgeführt. Mit Ausnahme von Claudin 2 waren alle untersuchten Verschlusskontaktproteine in Kontakten des olfaktorischen Epithels kolokalisiert. Unterschiedliche Immunfluoreszenzintensitäten konnten in den Kontakten zwischen olfaktorischen Neuronen und epithelialen Zellen beobachtet werden. Immunreaktivität von Claudin 5 wurde in Kontakten der olfaktorischen Gliazellen in den Fila olfactoria lokalisiert, Claudin 1- Immunreaktivität konnte in peripheren Bereichen der Fila olfactoria beobachtet werden. Hierbei zeigten sich Unterschiede in der Lokalistaion der Claudine mit verschiedenen ZOs. Stützzellen bildeten durch N-Cadherin vermittelte Adherenskontakte mit den olfaktorischen Neuronen, E-Cadherin-vermittelte Adherenskontakte mit Drüsen- und Microvilluszellen, sowie durch N- und E-Cadherin vermittelte Adherenskontakte mit benachbarten Stützzellen aus. Alpha-, beta- und p120-Catenin konnten in allen Adherenskontakten des olfaktorischen Epithels nachgewiesen werden. Olfaktorische Neurone bildeten in Abhängigkeit von deren Reifestadium unterschiedlich häufig Adherenskontakte aus. Adherenskontakte in den Fila olfactoria wurden zwischen Gliazellen, zwischen Gliazellen und Axonen, sowie zwischen Axonen untereinander lokalisiert. In den meisten Adherenskontakten der Fila olfactoria zeigte sich Kolokalisation zwischen N-Cadherin und den verschiedenen untersuchten Cateninen. In der Peripherie der Fila olfactoria konnten durch E-Cadherin vermittelte Adherenskontakte beobachtet werden. Die Funktion von Interzellularkontakten wird maßgeblich durch ihren molekularen Aufbau bestimmt. Charakteristische molekulare Zusammensetzungen der neuronalen, epithelialen und glialen Verschlusskontakte im olfaktorischen Epithel und den Fila olfactoria lässt auf unterschiedliche Eigenschaften dieser Kontakte schließen. Adherenskontakte sind für Neuro- und Axogenesevorgänge von entscheidender Bedeutung, und es liegt nahe, dass diese Kontakte auch bei diesen Prozessen im peripheren olfaktorischen Epithel eine wichtige Rolle spielen. Um Untersuchungen zur Ausbildung und zu möglichen Funktionen von Kontakten zwischen olfaktorischen Gliazellen und olfaktorischen Neuronen auch in vitro zu ermöglichen, wurden Primärkulturen von olfaktorischen Gliazellen etabliert und erste Experimente zur Charakterisierung der Kulturen bezüglich ihrer Homogenität sowie bezüglich verschiedener in vivo gefundener Eigenschaften, wie die Expression von Kontaktproteinen und die Produktion des ciliary neurotrophic factor (CNTF) in Einzelkulturen und in Kokulturen mit olfaktorischen Neuronen durchgeführt.
The Na+-D-glucose cotransporter in small intestine is regulated in response to food composition. Short term regulation of SGLT1 occurs post-transcriptionally in response to changes in luminal glucose. Adaptation to dietary carbohydrate involves long term regulation at the transcriptional level. The intracellular protein RS1 (gene RSC1A1) is involved in transcriptional and post-transcriptional regulation of SGLT1. RS1 contains an N-terminal domain with many putative phosphorylation sites. By Expressing SGLT1 in oocytes of Xenopus laevis it was previously demonstrated that the post-transcriptional down-regulation of SGLT1 by RS1 was dependent on the intracellular glucose concentration and activated by protein kinase C (PKC). The role of RS1 for short term regulation of SGLT1 in mouse small intestine in response to glucose and PKC was investigated comparing effects in RS1-/- mice and wildtype mice. Effects on SGLT1 activity were determined by measuring phlorizin inhibited uptake of α-methylglucoside (AMG). The involvement of RS1 in glucose dependent short term regulation could not be elucidated for technical reasons. However, evidence for RS1 independent short-term downregulation of SGLT1 after stimulation of PKC could be provided. It was shown that this downregulation includes decrease in the amount and/or in turnover of SGLT1 in the brush-border membrane as well as an increase of substrate affinity for AMG transport. Trying to elucidate the role of RS1 in long term regulation of SGLT1 in small intestine in response to glucose and fat content of the diet, wildtype and RS1-/- mice were kept for 2 months on a normo-caloric standard diet with high glucose and low fat content (ND), on a hyper-caloric glucose-galactose reduced diet with high fat content (GGRD) or on a hyper-caloric diet with a high fat and high glucose content (HFHGD). Thereafter the animals were starved overnight and SGLT1 mediated AMG uptake was measured. Independent of diet AMG uptake in ileum was smaller compared to duodenum and jejunum. In jejunum of wildtype and RS1-/- mice kept on the fat rich diets (GGRD and HFHGH) transport activity of SGLT1 was lower compared to mice kept on ND with low fat content. This result suggests an RS1 independent downregulation due to fat content of diet. Different to RS1-/- mice, the duodenum of wildtype mice showed transport activity of SGLT1 smaller in mice kept on glucose galactose reduced diet (GGRD) compared to the glucose galactose rich diets (ND and HFHGG). These data indicate that RS1 is involved in glucose dependent long term regulation in duodenum.
Die Stabiltät und Integrität der Epidermis beruht zu einem großen Teil auf der intakten Funktion der Desmosomen. Diese fleckförmigen Zellkontakte vermitteln extrazellulär die Haftung zwischen den Keratinozyten durch Desmocadherine und sind intrazellulär über Adaptorproteine im Intermediärfilamentsystem des Zellskeletts verankert. Diese Funktion ist bei der Autoimmunerkrankung Pemphigus gestört, die zu intraepidermaler Blasenbildung durch Akantholyse der Keratinozyten führt. Pemphigus vulgaris (PV) und Pemphigus foliaceus (PF) stellen die beiden Hauptvarianten dar, wobei PV durch Autoantikörper gegen die Desmocadherine Desmoglein (Dsg) 3 und oftmals zusätzlich gegen Dsg 1, PF durch Autoantikörper nur gegen Dsg 1 gekennzeichnet ist. Rho-GTPasen sind zelluläre Regulatorproteine, die das Aktinzytoskelett und verschiedene Zellkontakte beeinflussen. Die vorliegende Arbeit beschäftigte sich mit dem Einfluss von Rho-GTPasen bei der Regulation von desmosomal vermittelter Adhäsion. In einem zweiten Teil wurde die Beteiligung von Rho-GTPasen bei den Pemphigusvarianten PV und PF näher charakterisiert. Für den ersten Abschnitt wurden bakterielle Toxine verwendet, die spezifisch Rho GTPasen aktivieren bzw. inhibieren, während für den zweiten Teil IgG-Fraktionen von PV- und PF-Patienten in Kombination mit aktivierenden Toxinen zur Anwendung kamen. Eine Inhibition der drei Hauptvertreter der Rho-GTPasen in kultivierten Keratinozyten und humaner Epidermis führte zu einer Rarefizierung des Aktinfilamentsystems, zu Verlust von membranständig lokalisiertem Dsg 1 und 3 und zu Zelldissoziation sowie zu verminderter Dsg 1 und 3-vermittelter Haftung von Mikroperlen auf der Oberfläche von Keratinozyten. Die Aktivierung der GTPasen resultierte in vermehrter linearisierter Darstellbarkeit von Aktin und Dsg 3 an den Zellgrenzen und einer verstärkten Dsg-vermittelten Haftung. Pemphigus-IgG führten ebenfalls zu Zelldissoziation und Verlust von Dsg-Immunreaktivität in Keratinozytenkulturen, zu Spaltbildung in humaner Epidermis und zum Verlust der durch Dsg 1 und Dsg 3 vermittelten Adhäsion. Dies ging einher mit einer vermehrten Menge an nicht am Zytoskelett verankerten Dsg 3 und wurde durch eine p38MAPK-abhängige Verminderung der Aktivität von Rho A moduliert. Die Aktivierung von Rho A verhinderte die Ausbildung der Pemphigus-induzierten Effekte nahezu vollständig. Zusammenfassend regulieren Rho-GTPasen die desmosomale Haftung in Keratinozyten. Die Daten zeigen weiterhin, dass Pemphigus-IgG durch eine Inhibition von Rho A diese Regulation beeinträchtigt, was zu Schwächung der Zytoskelettverankerung von Desmogleinen und zu Haftungsverlust und Spaltbildung führt. Somit ist Rho A ein wichtiger Faktor der Pemphigus-Pathogenese und stellt einen Erfolg versprechenden Ansatzpunkt zur Entwicklung neuer Therapieoptionen dar.
Um das Transmittersignal von Glutamat zu beenden und eine neurotoxische Anreicherung zu verhindern, muss Glutamat aus dem Extrazellularraum des ZNS entfernt werden. Dafür sind die hochaffinen Glutamattransporter in Gliazellen und Neuronen zuständig, die Glutamat aus dem Extrazellularraum aufnehmen. In der vorliegenden Arbeit wurde die regionale und zelluläre Verteilung der Glutamattransporter GLT1, GLAST und EAAC1 in Hippocampus, Kleinhirn und Rückenmark von Maus und Ratte untersucht. Als Nachweismethoden wurden Western Blot-Analysen und immunhistochemische Nachweise an fixierten Kryostatschnitten und Semidünnschnitten von gefriergetrockneten Geweben eingesetzt.
Background:
Multiple sclerosis (MS) is a chronic autoimmune disease of the central nervous system (CNS) for which several new treatment options were recently introduced. Among them is the monoclonal anti-CD52 antibody alemtuzumab that depletes mainly B cells and T cells in the immune periphery. Considering the ongoing controversy about the involvement of B cells and in particular the formation of B cell aggregates in the brains of progressive MS patients, an in-depth understanding of the effects of anti-CD52 antibody treatment on the B cell compartment in the CNS itself is desirable.
Methods:
We used myelin basic protein (MBP)-proteolipid protein (PLP)-induced experimental autoimmune encephalomyelitis (EAE) in C57BL/6 (B6) mice as B cell-dependent model of MS. Mice were treated intraperitoneally either at the peak of EAE or at 60 days after onset with 200 μg murine anti-CD52 vs. IgG2a isotype control antibody for five consecutive days. Disease was subsequently monitored for 10 days. The antigen-specific B cell/antibody response was measured by ELISPOT and ELISA. Effects on CNS infiltration and B cell aggregation were determined by immunohistochemistry. Neurodegeneration was evaluated by Luxol Fast Blue, SMI-32, and Olig2/APC staining as well as by electron microscopy and phosphorylated heavy neurofilament serum ELISA.
Results:
Treatment with anti-CD52 antibody attenuated EAE only when administered at the peak of disease. While there was no effect on the production of MP4-specific IgG, the treatment almost completely depleted CNS infiltrates and B cell aggregates even when given as late as 60 days after onset. On the ultrastructural level, we observed significantly less axonal damage in the spinal cord and cerebellum in chronic EAE after anti-CD52 treatment.
Conclusion:
Anti-CD52 treatment abrogated B cell infiltration and disrupted existing B cell aggregates in the CNS.
Die Blut-Hirn-Schranke reguliert den Transport von Molekülen aus dem Blut in das Gehirn und aus dem Hirngewebe in das Blut. Die Grundlage dieser für den Erhalt der Homöostase im Gehirn wichtigen Schranke bilden zwischen Endothelzellen der Gehirnkapillaren (BCECs) entwickelte, besonders dichte Zonulae Occludentes (Tight Junctions). Viele Krankheiten, zum Beispiel die Multiple Sklerose, gehen mit einer Dysfunktion der BBB einher, die molekularen Grundlagen verschiedener Störungen und damit die Therapiemöglichkeiten sind bisher jedoch oftmals noch unbekannt. Ein grundlegendes Problem der Forschung an der BBB war bislang das Fehlen eines geeigneten immortalisierten in vitro-Modelles zum Verständnis der Differenzierung und Regulierung der Schrankenfunktion. Es gelang nun erstmals, aus murinen BCECs ein solches in vitro-Modell der BBB zu entwickeln, welches wichtige Charakteristika der BBB in vivo aufweist. Zu den Eigenschaften der BBB in vivo zählen allgemein ein hoher transendothelialer elektrischer Widerstand (TER) von bis zu 2000  x cm², die Expression der TJ-Proteine Occludin, Claudin-1, Claudin-3 und Claudin-5 sowie eine geringe Rate transzellulärer Transportvorgänge. Die Entwicklung einer immortalisierten Zelllinie als in vitro-Modell der BBB beinhaltete das Bereitstellen einer möglichst natürlichen Umgebung für die Endothelzellen. Durch Zugabe von Wachstums- und Differenzierungsfaktoren sowie Serumreduktion im Differenzierungsmedium konnte eine dichte Schrankenfunktion induziert werden, welche sich anhand von TER-Messungen nachweisen ließ. Mittels immuncytochemischen und molekularbiologischen Methoden wurde außerdem die Expression verschiedener TJ-Proteine in den immortalisierten BCECs gezeigt. Die Permeabilität der BBB wird durch eine Reihe von Faktoren beeinflusst. So war zu erkennen, dass Glucocorticoide und Insulin die Barrierenfunktion der BBB induzieren und die Zugabe dieser Faktoren die in vitro-Kultivierung von BCECs ermöglicht, ohne dass diese dabei für die BBB in vivo wesentliche Charakteristika verlieren. Diese Ergebnisse stimmen überein mit anderen Studien, denenzufolge für die Induktion und Aufrechterhaltung komplexer Tight Junctions bei kultivierten Endothelzellen Glucocorticoide förderlich sind. Auch klinisch wird dieser Einfluss von Glucocorticoiden bereits genutzt: so konnten im Falle der Multiplen Sklerose Therapieerfolge durch die Gabe von Corticosteroiden erzielt werden.
Diabetes mellitus is a common disease affecting more than 537 million adults worldwide. The microvascular complications that occur during the course of the disease are widespread and affect a variety of organ systems in the body. Diabetic retinopathy is one of the most common long-term complications, which include, amongst others, endothelial dysfunction, and thus, alterations in the blood-retinal barrier (BRB). This particularly restrictive physiological barrier is important for maintaining the neuroretina as a privileged site in the body by controlling the inflow and outflow of fluid, nutrients, metabolic end products, ions, and proteins. In addition, people with diabetic retinopathy (DR) have been shown to be at increased risk for systemic vascular complications, including subclinical and clinical stroke, coronary heart disease, heart failure, and nephropathy. DR is, therefore, considered an independent predictor of heart failure. In the present review, the effects of diabetes on the retina, heart, and kidneys are described. In addition, a putative common microRNA signature in diabetic retinopathy, nephropathy, and heart failure is discussed, which may be used in the future as a biomarker to better monitor disease progression. Finally, the use of miRNA, targeted neurotrophin delivery, and nanoparticles as novel therapeutic strategies is highlighted.
The polyspecific organic cation transporters (OCT) are involved in the elimination and distribution of drugs, environmental toxins, and endogenous organic cations including monoamine neurotransmitters. Steroid hormones inhibit organic cation transport by the three OCT subtypes with different affinities showing distinct species difference; for example, the IC50 values for corticosterone inhibition of cation uptake by transporters rOCT1 and rOCT2 are ~150μM and ~4 μM, respectively. By introducing domains and amino acids from rOCT2 into rOCT1, we identified three amino acids in the presumed 10th TMD of rOCT2 which are responsible for the higher affinity of corticosterone in comparison to rOCT1. This is the first study which revealed the components of the binding site for corticosterone in OCTs. The evidence is presented that these amino acids (alanine 443, leucine 447, and glutamine 448 in rOCT1 and isoleucine 443, tyrosine 447, and glutamate 448 in rOCT2) are probably located within the substrate binding region of OCTs since the affinity of transported cations was increased together with the affinity of corticosterone. In the double mutant rOCT1(L447Y/Q448E) the IC50 value for the inhibition of [3H]MPP (0.1 μM) uptake by corticosterone (24 ± 4 μM) was significantly higher compared to the IC50 value for inhibition of [14C]TEA (10 μM) uptake (5.3 ± 1.7 μM), indicating an allosteric interaction between transported substrate and corticosterone. The data suggest that more than one compound can bind simultaneously to the substrate binding region. These results confirm previous suggestion that binding of substrates and inhibitors to OCTs involves interaction with a comparatively large surface that may include multiple binding domains rather than with a structurally restricted single binding site.
1994 wurde von Gründemann et al. der erste organische Kationentransporter, der rOCT1 beschrieben. Es wurden bereits einige Aminosäuren identifiziert, die bei der Bindung kationischer Substanzen beteiligt sind. Hierbei handelt es sich um Phenylalanin 160 der zweiten Transmembrandomäne, Tryptophan 218, Tyrosin 222 und Threonin 226 der vierten Transmembrandomäne, um Arginin 440, Leucin 447, Glutamin 448 der zehnten und um Aspartat 475 der elften Transmembrandomäne. Hintergrund der Versuche dieser Arbeit war das im Jahre 2005 von Sturm et al. identifizierte Cystein 451. Es liegt zwischen der zehnten und elften Transmembrandomäne. Cystein 451 ist wahrscheinlich auf Grund seiner Lage im Strukturmodell nicht direkt an der Bindung von Substraten beteiligt. Es wird vermutet, dass die Mutation des Cysteins 451 die Positionen von Aminosäuren in der Bindungsstelle verändert. Daher wurden die Mutante C451M, die Doppelmutanten L447F/C451M, L447Y/C451M und die Dreifachmutante Y222F/L447F/C451M mittels Tracer-Fluxexperimenten hinsichtlich der Hemmung der Tetraethylammonium-Aufnahme durch Kortikosteron und durch Tetrabutylammonium untersucht. Die Mutation C451M steigert verglichen mit dem rOCT1-Wildtyp die Affinität für Kortikosteron, jedoch sinkt bei dieser Mutante die TBuA-Affinität. Man nimmt nun aufgrund dieser Mutageneseversuche und den bereits zuvor generierten Modellen des rOCT1 an, dass aufgrund seiner Lage Cystein 451 nicht direkt an der Bindung von Substraten beteiligt ist, sondern einen indirekten Effekt auf die Substratbindungsregion des Transporters ausübt. Weiterhin wurde festgestellt, dass die Mutanten L447Y/C451M und L447F/C451M gegensätzliche Affinitäten für TBuA und Kotikosteron haben. Tauscht man das Leucin an Position 447 gegen ein Tyrosin aus, so wird der Transporter weniger affin für Kortikosteron, jedoch steigt die TBuA-Affinität. Tauscht man das Leucin gegen ein Phenylalanin aus, verhält es sich gegensätzlich. Die Position 222 scheint weder an der TBuA-Bindung, noch an der Bindung von Kortikosteron maßgeblich beteiligt zu sein.
Mouse embryonic stem cells (ESCs) are maintained in a naive ground state of pluripotency in the presence of MEK and GSK3 inhibitors. Here, we show that ground-state ESCs express low Myc levels. Deletion of both c-myc and N-myc (dKO) or pharmacological inhibition of Myc activity strongly decreases transcription, splicing, and protein synthesis, leading to proliferation arrest. This process is reversible and occurs without affecting pluripotency, suggesting that Myc-depleted stem cells enter a state of dormancy similar to embryonic diapause. Indeed, c-Myc is depleted in diapaused blastocysts, and the differential expression signatures of dKO ESCs and diapaused epiblasts are remarkably similar. Following Myc inhibition, pre-implantation blastocysts enter biosynthetic dormancy but can progress through their normal developmental program after transfer into pseudo-pregnant recipients. Our study shows that Myc controls the biosynthetic machinery of stem cells without affecting their potency, thus regulating their entry and exit from the dormant state.
Tumors are characterized by a rigid, highly cross-linked extracellular matrix (ECM), which impedes homogeneous drug distribution and potentially protects malignant cells from exposure to therapeutics. Lysyl oxidases are major contributors to tissue stiffness and the elevated expression of these enzymes observed in most cancers might influence drug distribution and efficacy. We examined the effect of lysyl oxidases on drug distribution and efficacy in 3D in vitro assay systems. In our experiments elevated lysyl oxidase activity was responsible for reduced drug diffusion under hypoxic conditions and consequently impaired cytotoxicity of various chemotherapeutics. This effect was only observed in 3D settings but not in 2D-cell culture, confirming that lysyl oxidases affect drug efficacy by modification of the ECM and do not confer a direct desensitizing effect. Both drug diffusion and efficacy were strongly enhanced by inhibition of lysyl oxidases. The results from the in vitro experiments correlated with tumor drug distribution in vivo, and predicted response to therapeutics in murine tumor models. Our results demonstrate that lysyl oxidase activity modulates the physical barrier function of ECM for small molecule drugs influencing their therapeutic efficacy. Targeting this process has the potential to significantly enhance therapeutic efficacy in the treatment of malignant diseases.
The mouse gastro-intestinal and biliary tract mucosal epithelia harbor choline acetyltransferase (ChAT)-positive brush cells with taste cell-like traits. With the aid of two transgenic mouse lines that express green fluorescent protein (EGFP) under the control of the ChAT promoter (EGFP\(^{ChAT}\)) and by using in situ hybridization and immunohistochemistry we found that EGFP\(^{ChAT}\) cells were clustered in the epithelium lining the gastric groove. EGFP\(^{ChAT}\) cells were numerous in the gall bladder and bile duct, and found scattered as solitary cells along the small and large intestine. While all EGFP\(^{ChAT}\) cells were also ChAT-positive, expression of the high-affinity choline transporter (ChT1) was never detected. Except for the proximal colon, EGFP\(^{ChAT}\) cells also lacked detectable expression of the vesicular acetylcholine transporter (VAChT). EGFP\(^{ChAT}\) cells were found to be separate from enteroendocrine cells, however they were all immunoreactive for cytokeratin 18 (CK18), transient receptor potential melastatin-like subtype 5 channel (TRPM5), and for cyclooxygenases 1 (COX1) and 2 (COX2). The ex vivo stimulation of colonic EGFP\(^{ChAT}\) cells with the bitter substance denatonium resulted in a strong increase in intracellular calcium, while in other epithelial cells such an increase was significantly weaker and also timely delayed. Subsequent stimulation with cycloheximide was ineffective in both cell populations. Given their chemical coding and chemosensory properties, EGFP\(^{ChAT}\) brush cells thus may have integrative functions and participate in induction of protective reflexes and inflammatory events by utilizing ACh and prostaglandins for paracrine signaling.
Ultra-high field cardiac MRI in large animals and humans for translational cardiovascular research

A key step in translational cardiovascular research is the use of large animal models to better understand normal and abnormal physiology, to test drugs or interventions, or to perform studies which would be considered unethical in human subjects. Ultrahigh field magnetic resonance imaging (UHF-MRI) at 7 T field strength is becoming increasingly available for imaging of the heart and, when compared to clinically established field strengths, promises better image quality and image information content, more precise functional analysis, potentially new image contrasts, and as all in-vivo imaging techniques, a reduction of the number of animals per study because of the possibility to scan every animal repeatedly. We present here a solution to the dual use problem of whole-body UHF-MRI systems, which are typically installed in clinical environments, to both UHF-MRI in large animals and humans. Moreover, we provide evidence that in such a research infrastructure UHF-MRI, and ideally combined with a standard small-bore UHF-MRI system, can contribute to a variety of spatial scales in translational cardiovascular research: from cardiac organoids, Zebra fish and rodent hearts to large animal models such as pigs and humans. We present pilot data from serial CINE, late gadolinium enhancement, and susceptibility weighted UHF-MRI in a myocardial infarction model over eight weeks. In 14 pigs which were delivered from a breeding facility in a national SARS-CoV-2 hotspot, we found no infection in the incoming pigs. Human scanning using CINE and phase contrast flow measurements provided good image quality of the left and right ventricle. Agreement of functional analysis between CINE and phase contrast MRI was excellent. MRI in arrested hearts or excised vascular tissue for MRI-based histologic imaging, structural imaging of myofiber and vascular smooth muscle cell architecture using high-resolution diffusion tensor imaging, and UHF-MRI for monitoring free radicals as a surrogate for MRI of reactive oxygen species in studies of oxidative stress are demonstrated. We conclude that UHF-MRI has the potential to become an important precision imaging modality in translational cardiovascular research.
Adhesion-type G protein-coupled receptors (aGPCRs), a large molecule family with over 30 members in humans, operate in organ development, brain function and govern immunological responses. Correspondingly, this receptor family is linked to a multitude of diverse human diseases. aGPCRs have been suggested to possess mechanosensory properties, though their mechanism of action is fully unknown. Here we show that the Drosophila aGPCR Latrophilin/dCIRL acts in mechanosensory neurons by modulating ionotropic receptor currents, the initiating step of cellular mechanosensation. This process depends on the length of the extended ectodomain and the tethered agonist of the receptor, but not on its autoproteolysis, a characteristic biochemical feature of the aGPCR family. Intracellularly, dCIRL quenches cAMP levels upon mechanical activation thereby specifically increasing the mechanosensitivity of neurons. These results provide direct evidence that the aGPCR dCIRL acts as a molecular sensor and signal transducer that detects and converts mechanical stimuli into a metabotropic response.
Synapse-associated protein 1 (Syap1/BSTA) is the mammalian homologue of Sap47 (synapse-associated protein of 47 kDa) in Drosophila. Sap47 null mutant larvae show reduced short-term synaptic plasticity and a defect in associative behavioral plasticity. In cultured adipocytes, Syap1 functions as part of a complex that phosphorylates protein kinase B alpha/Akt1 (Akt1) at Ser\(^{473}\) and promotes differentiation. The role of Syap1 in the vertebrate nervous system is unknown. Here, we generated a Syap1 knock-out mouse and show that lack of Syap1 is compatible with viability and fertility. Adult knock-out mice show no overt defects in brain morphology. In wild-type brain, Syap1 is found widely distributed in synaptic neuropil, notably in regions rich in glutamatergic synapses, but also in perinuclear structures associated with the Golgi apparatus of specific groups of neuronal cell bodies. In cultured motoneurons, Syap1 is located in axons and growth cones and is enriched in a perinuclear region partially overlapping with Golgi markers. We studied in detail the influence of Syap1 knockdown and knockout on structure and development of these cells. Importantly, Syap1 knockout does not affect motoneuron survival or axon growth. Unexpectedly, neither knockdown nor knockout of Syap1 in cultured motoneurons is associated with reduced Ser\(^{473}\) or Thr\(^{308}\) phosphorylation of Akt. Our findings demonstrate a widespread expression of Syap1 in the mouse central nervous system with regionally specific distribution patterns as illustrated in particular for olfactory bulb, hippocampus, and cerebellum.
Blood vessel organoids are an important in vitro model to understand the underlying mechanisms of human blood vessel development and for toxicity testing or high throughput drug screening. Here we present a novel, cost-effective, and easy to manufacture vascular organoid model. To engineer the organoids, a defined number of human induced pluripotent stem cells are seeded in non-adhesive agarose coated wells of a 96-well plate and directed towards a lateral plate mesoderm fate by activation of Wnt and BMP4 signaling. We observe the formation of a circular layer of angioblasts around days 5–6. Induced by VEGF application, CD31\(^+\) vascular endothelial cells appear within this vasculogenic zone at approximately day 7 of organoid culture. These cells arrange to form a primitive vascular plexus from which angiogenic sprouting is observed after 10 days of culture. The differentiation outcome is highly reproducible, and the size of organoids is scalable depending on the number of starting cells. We observe that the initial vascular ring forms at the interface between two cell populations. The inner cellular compartment can be distinguished from the outer by the expression of GATA6, a marker of lateral plate mesoderm. Finally, 14-days-old organoids were transplanted on the chorioallantois membrane of chicken embryos resulting in a functional connection of the human vascular network to the chicken circulation. Perfusion of the vessels leads to vessel wall maturation and remodeling as indicated by the formation of a continuous layer of smooth muscle actin expressing cells enwrapping the endothelium. In summary, our organoid model recapitulates human vasculogenesis, angiogenesis as well as vessel wall maturation and therefore represents an easy and cost-effective tool to study all steps of blood vessel development and maturation directly in the human setting without animal experimentation.
Glutamat spielt in der Entwicklung des ZNS eine besondere Rolle (z.B. Steuerung der Migration von Proneuronen, Einfluß auf die Ausbildung von Synapsen und Differenzierung von Pyramidenzelldendriten und Proliferation glialer Vorläuferzellen). Durch Regulation der extrazellulären Glutamatkonzentration kommt den Glutamattransportern dabei eine besondere Bedeutung zu. Ziel der vorliegenden Arbeit war deshalb, die postnatale zelluläre und regionale Expression der Glutamattransporter GLT1 und GLAST im Hippocampus der Ratte zu untersuchen, um Rückschlüsse auf ihre Funktion während der postnatalen Ontogenese (Postnataltag 1-60, P1-60) zu ziehen. Dazu wurde nichtradioaktive In-situ-Hybridisierung (ISH) mit Digoxigenin-markierten cRNA-Sonden eingesetzt. Unsere Ergebnisse zeigen, dass beide Transporter vor allem in Gliazellen (besonders Astroblasten/Astrozyten) nachweisbar sind und dass die Expression von GLT1 und GLAST während der Ontogenese der Ratte im Hippocampus unterschiedlich erfolgt. Aus unseren Ergebnissen kann geschlossen werden, dass GLAST und GLT1 eine unterschiedliche Bedeutung während der Ausbildung der hippocampalen Verbindungen haben dürften. Der trisynaptische hippocampale Verbindungsweg entwickelt sich hauptsächlich in der ersten postnatalen Woche. Während dieser Zeit verlagert sich die exzitatorische Funktion in Hippocampusneuronen von GABA auf Glutamat. Diese Transmitterumstellung korreliert zeitlich mit der deutlichen Zunahme der GLT1-Expression im Hippocampus, was auf eine entscheidende Rolle von GLT1 bei der Bildung hippocampaler Verbindungen durch Regulation der extrazellulären Glutamatkonzentration hinweist. Demgegenüber ist kein offensichtlicher Zusammenhang zwischen GLAST-Expression und Transmitterumstellung zu erkennen. Wie in adulten Stadien kommt dem Transporter unter physiologischen Bedingungen auch in der Ontogenese wohl vor allem eine Reservefunktion zu.
Als Therapieversuch bei Plexusläsionen wird die Replantation ausgerissener Vorderwurzelfasern durchgeführt. Voraussetzung für die erfolgreiche Regeneration von Motoneuronaxonen sind 1. Überleben einer ausreichenden Anzahl von Motoneuronen 2. erfolgreiche Wiederherstellung der Kontuität ausgerissener Axone mit dem Rückenmark und 3. funktionelle Hochwertigkeit regenerierter Axone. Neurotrophe Faktoren können Überleben und Regenerationsfähigkeit von Motoneuronen fördern. Gegenstand der vorliegenden Arbeit war die Analyse des Einflusses von CNTF und BDNF auf die Regeneration von Motoneuronaxonen nach Ausriss und Replantation im Segment C7 nach einer Überlebenszeit von 3 Wochen bzw. 6 Monaten. Vervollständigt wurden diese Untersuchungen durch detaillierte morphologische Analysen von Spinalganglien, durchtrennter Hinterwurzel und verletztem Hinterhorn. In verschiedenen Gruppen von adulten Kaninchen wurden CNTF, BDNF, oder beide Faktoren auf die ventrolaterale Replantationsstelle appliziert, Kontrollen wurden ohne Faktor belassen (n>5). Die Überlebenszeit der Versuchstiere lag bei 3 Wochen (n=3 Kontrollen) und 6 Monaten (n=27). Aus dem perfundiertem Gewebe wurden Semidünnschnitte durch Vorderwurzel/Spinalganglien und Kryostatserienschnitte durch das Segment C7 angefertigt. DiI-Fluoreszenztracing, Markscheidenfärbung, eine modifizierte Klüver-Barrera-Färbung der Kryostatschnitte sowie eine Touloidinblaufärbung der Semidünnschnitte ermöglichte die morphologische und morphometrische Analyse des Gewebes. Die Anzahl der überlebenden Motoneurone lag nach sechs Monaten bei allen Versuchsgruppen bei etwa 30%. Fluoreszenz-Tracing und Markscheidenfärbungen von Serienschnitten zeigten, dass Axone sowohl über die ursprünglichen ventralen Austrittstellen als auch über die ventrolaterale Replantationsstelle das Rückenmark verließen und im Bereich des Spinalganglions eine kompakte Vorderwurzel bildeten. Ventral austretende Axone zeigten signifikant größere Durchmesser als lateral austretende. Ausmaß und Art der Regeneration waren interindividuell unterschiedlich, die besten Ergebnisse zeigte die Replantation nah am ursprünglichen Austrittsort der Vorderwurzel. Unterschiede zwischen den Gruppen waren nicht deutlich. In Semidünnschnitten durch die regenerierte Vorderwurzel fanden sich nach drei Wochen kaum intakte, myelinisierte Axone, nach sechs Monaten war die Zahl der Axone auf etwa 45% der Zahl der gesunden Seite angestiegen. Regenerierte Axone waren dünn, typische Motoneuronaxone stellten nur einen kleinen Teil der regenerierten Axone. Gruppenunterschiede fanden sich im Axon-Myelinverhältnis, das bei Kontrollen der replantierten Seiten signifikant erniedrigt war. Diese Erniedrigung war noch vorhanden, jedoch nicht mehr signifikant bei Tieren, die mit CNTF- und BDNF-behandelt wurden. Die replantierten Vorderwurzeln der CNTF+BDNF-Gruppe zeigte überwiegend eine signifikant bessere Myelinisierung als die replantierten Kontrollen. An der früheren Hinterwurzeleintrittszone am Rückenmark wurden in Tieren mit geringem Verletzungsausmaß kleine ZNS-Gewebsprotrusionen beobachtet, in denen sich myelinisierte Axone befanden. Diese Axone zeigten eine Wachstumsrichtung in die Peripherie, was auf eine Sprossung der sensorischen Rückenmarksneurone schließen lässt. Innerhalb des Spinalganglions waren Neuron- und Axondichte auf den verletzten Seiten nicht wesentlich verändert. Eine leichte Abnahme des relativen Anteils großer Neurone und Axone wurde in den verletzten Seiten der Kontrollgruppe beobachtet. Für Axone war diese Abnahme statistisch signifikant. Im Gegensatz dazu war dies in Tieren, die mit neurotrophen Faktoren behandelt wurden, nicht zu beobachten. Bei allen Tieren zeigte sich ein beträchtliches Auswachsen von Hinterwurzelaxonen aus dem Spinalganglion. Diese Axone fanden keine spontane Verbindung mit dem proximalen Rest der Wurzel, sondern waren durch Bindegewebe eingehüllt. Bei etwa der Hälfte der Tiere zeigte sich, dass einer Untergruppe dieser Axone in Richtung des Narbengewebes der replantierten Vorderwurzel gewachsen war und über Defekte in der Bindegewebshülle teilweise sogar in die Vorderwurzel einwuchsen. Ein möglicher Einfluss der applizierten neurotrophen Faktoren auf das quantitative Regenerationsergebnis scheint also in diesem Modell gering zu sein. Auf eine qualitative Verbesserung deutet die Normalisierung des Axon-Myelinverhältnisses großer regenerierter Axone bei Kombinationsbehandlung hin. Die im vorliegenden Modell beträchtliche Regenerationskapazität der Hinterwurzel scheint bisher unterschätzt worden zu sein. Das unerwartete Einwachsen von Hinterwurzelaxonen in die Vorderwurzel könnte mit einer funktionellen Beeinträchtigung der regenerierten Vorderwurzel verbunden sein.
Transcriptional and distributional profiling of microglia in retinal angiomatous proliferation

Macular neovascularization type 3, formerly known as retinal angiomatous proliferation (RAP), is a hallmark of age-related macular degeneration and is associated with an accumulation of myeloid cells, such as microglia (MG) and infiltrating blood-derived macrophages (MAC). However, the contribution of MG and MAC to the myeloid cell pool at RAP sites and their exact functions remain unknown. In this study, we combined a microglia-specific reporter mouse line with a mouse model for RAP to identify the contribution of MG and MAC to myeloid cell accumulation at RAP and determined the transcriptional profile of MG using RNA sequencing. We found that MG are the most abundant myeloid cell population around RAP, whereas MAC are rarely, if ever, associated with late stages of RAP. RNA sequencing of RAP-associated MG showed that differentially expressed genes mainly contribute to immune-associated processes, including chemotaxis and migration in early RAP and proliferative capacity in late RAP, which was confirmed by immunohistochemistry. Interestingly, MG upregulated only a few angiomodulatory factors, suggesting a rather low angiogenic potential. In summary, we showed that MG are the dominant myeloid cell population at RAP sites. Moreover, MG significantly altered their transcriptional profile during RAP formation, activating immune-associated processes and exhibiting enhanced proliferation, however, without showing substantial upregulation of angiomodulatory factors.
Ischemic insults to the heart and brain, i.e., myocardial and cerebral infarction, respectively, are amongst the leading causes of death worldwide. While there are therapeutic options to allow reperfusion of ischemic myocardial and brain tissue by reopening obstructed vessels, mitigating primary tissue damage, post-infarction inflammation and tissue remodeling can lead to secondary tissue damage. Similarly, ischemia in retinal tissue is the driving force in the progression of neovascular eye diseases such as diabetic retinopathy (DR) and age-related macular degeneration (AMD), which eventually lead to functional blindness, if left untreated. Intriguingly, the easily observable retinal blood vessels can be used as a window to the heart and brain to allow judgement of microvascular damages in diseases such as diabetes or hypertension. The complex neuronal and endocrine interactions between heart, retina and brain have also been appreciated in myocardial infarction, ischemic stroke, and retinal diseases. To describe the intimate relationship between the individual tissues, we use the terms heart-brain and brain-retina axis in this review and focus on the role of transforming growth factor β (TGFβ) and neurotrophins in regulation of these axes under physiologic and pathologic conditions. Moreover, we particularly discuss their roles in inflammation and repair following ischemic/neovascular insults. As there is evidence that TGFβ signaling has the potential to regulate expression of neurotrophins, it is tempting to speculate, and is discussed here, that cross-talk between TGFβ and neurotrophin signaling protects cells from harmful and/or damaging events in the heart, retina, and brain.
Immunosenescence is considered a possible factor in the development of age-related macular degeneration and choroidal neovascularization (CNV). However, age-related changes of myeloid cells (MCs), such as microglia and macrophages, in the healthy retina or during CNV formation are ill-defined. In this study, Cx3cr1-positive MCs were isolated by fluorescence-activated cell sorting from six-week (young) and two-year-old (old) Cx3cr1\(^{GFP/+}\) mice, both during physiological aging and laser-induced CNV development. High-throughput RNA-sequencing was performed to define the age-dependent transcriptional differences in MCs during physiological aging and CNV development, complemented by immunohistochemical characterization and the quantification of MCs, as well as CNV size measurements. These analyses revealed that myeloid cells change their transcriptional profile during both aging and CNV development. In the steady state, senescent MCs demonstrated an upregulation of factors contributing to cell proliferation and chemotaxis, such as Cxcl13 and Cxcl14, as well as the downregulation of microglial signature genes. During CNV formation, aged myeloid cells revealed a significant upregulation of angiogenic factors such as Arg1 and Lrg1 concomitant with significantly enlarged CNV and an increased accumulation of MCs in aged mice in comparison to young mice. Future studies need to clarify whether this observation is an epiphenomenon or a causal relationship to determine the role of immunosenescence in CNV formation.
In patients with low-risk breast cancer, intraoperative radiotherapy (IORT) during breast-conserving surgery is a novel and convenient treatment option for delivering a single high dose of irradiation directly to the tumour bed. However, edema and fibrosis can develop after surgery and radiotherapy, which can subsequently impair quality of life. TGF-β is a strong inducer of the extracellular matrix component hyaluronan (HA). TGF-β expression and HA metabolism can be modulated by irradiation experimentally, and are involved in edema and fibrosis. We therefore hypothesized that IORT may regulate these factors.Wound fluid (WF) draining from breast lumpectomy sites was collected and levels of TGF-β1 and HA were determined by ELISA. Proliferation and marker expression was analyzed in primary lymphatic endothelial cells (LECs) treated with recombinant TGF-β or WF. Our results show that IORT does not change TGF-β1 or HA levels in wound fluid draining from breast lumpectomy sites, and does not lead to accumulation of sHA oligosaccharides. Nevertheless, concentrations of TGF-β1 were high in WF from patients regardless of IORT, at concentrations well above those associated with fibrosis and the suppression of LEC identity. Consistently, we found that TGF-β in WF is active and inhibits LEC proliferation. Furthermore, all three TGF-β isoforms inhibited LEC proliferation and suppressed LEC marker expression at pathophysiologically relevant concentrations.
Given that TGF-β contributes to edema and plays a role in the regulation of LEC identity, we suggest that inhibition of TGF-β directly after surgery might prevent the development of side effects such as edema and fibrosis.
Voltage-gated calcium channels (VGCCs) are widely distributed within the central nervous system (CNS) and presumed to play an important role in the pathophysiology of a broad spectrum of CNS disorders including Alzheimer’s and Parkinson’s disease as well as multiple sclerosis. Several calcium channel blockers have been in clinical practice for many years so that their toxicity and side effects are well studied. However, these drugs are primarily used for the treatment of cardiovascular diseases and most if not all effects on brain functions are secondary to peripheral effects on blood pressure and circulation. While the use of calcium channel antagonists for the treatment of CNS diseases therefore still heavily depends on the development of novel strategies to specifically target different channels and channel subunits, this review is meant to provide an impulse to further emphasize the importance of future research towards this goal.
Multiple sclerosis (MS) is the most prevalent neurological disease of the central nervous system (CNS) in young adults and is characterized by inflammation, demyelination and axonal pathology that result in multiple neurological and cognitive deficits. The focus of MS research remains on modulating the immune response, but common therapeutic strategies are only effective in slowing down disease progression and attenuating the symptoms; they cannot cure the disease. Developing an option to prevent neurodegeneration early on would be a valuable addition to the current standard of care for MS. Based on our results we suggest that application of nimodipine could be an effective way to target both neuroinflammation and neurodegeneration. We performed detailed analyses of neurodegeneration in experimental autoimmune encephalomyelitis (EAE), an animal model of MS, and in in vitro experiments regarding the effect of the clinically well-established L-type calcium channel antagonist nimodipine. Nimodipine treatment attenuated the course of EAE and spinal cord histopathology. Furthermore, it promoted remyelination. The latter could be due to the protective effect on oligodendrocytes and oligodendrocyte precursor cells (OPCs) we observed in response to nimodipine treatment. To our surprise, we detected calcium channel-independent effects on microglia, resulting in apoptosis. These effects were cell type-specific and independent of microglia polarization. Apoptosis was accompanied by decreased levels of nitric oxide (NO) and inducible NO synthase (iNOS) in cell culture as well as decreased iNOS expression and reactive oxygen species (ROS) activity in EAE. Overall, application of nimodipine seems to generate a favorable environment for regenerative processes and could therefore be a novel treatment option for MS, combining immunomodulatory effects while promoting neuroregeneration.
Controversy surrounds neutrophil function in cancer because neutrophils were shown to provide both pro-and antitumor functions. We identified a heterogeneous subset of low-density neutrophils (LDNs) that appear transiently in self-resolving inflammation but accumulate continuously with cancer progression. LDNs display impaired neutrophil function and immunosuppressive properties, characteristics that are in stark contrast to those of mature, high-density neutrophils (HDNs). LDNs consist of both immature myeloid-derived suppressor cells (MDSCs) and mature cells that are derived from HDNs in a TGF-beta-dependent mechanism. Our findings identify three distinct populations of circulating neutrophils and challenge the concept that mature neutrophils have limited plasticity. Furthermore, our findings provide a mechanistic explanation to mitigate the controversy surrounding neutrophil function in cancer.
Das Naturschutzgebiet "Garchinger Heide" ist ein Relikt der ehemals ausgedehnten Kalkmagerrasen auf Pararendzinen über Niederterrassenschotter im Alpenvorland. Bei jahrhundertelanger extensiver Landnutzung hat sich auf trockenen nährstoffarmen Böden eine artenreiche Vegetation mit zahlreichen seltenen und gefährdeten Arten entwickelt. Die "Altheide", welche den größten Teil des Naturschutzgebiets einnimmt, ist geprägt durch flachgründige, nährstoffarme Böden, welche eine geschlossene Vegetation mit einem hohen Anteil an Arten der Halbtrockenrasen (hauptsächlich Mesobromion erecti und Cirsio-Brachypodion) tragen. Das 1945 durch Oberbodenabtrag begonnene, aber nicht mehr ausgebaute und verwendete „Rollfeld“ weist einen niedrigen Feinbodenanteil, welcher auf eine geringe Wasserverfügbarkeit schließen lässt, sowie niedrige Gehalte an Gesamtstickstoff und CAL-austauschbaren P2O5 auf. Die Vegetation ist lückig mit einem hohen Anteil an Trockenrasenarten (Xerobromion, Sedo-Scleranthetea, Sesleritalia albicantis). Das heutige Vorkommen zahlreicher Arten, welche in früheren Untersuchungen (1956 und 1986) nicht gefunden wurden, lässt auf eine fortschreitende Sukzession der Vegetation des Rollfeldes hin zu der der Altheide schließen. Der 1959 zum Naturschutzgebiet hinzu gekaufte ehemalige Acker unterscheidet sich immer noch deutlich von der Altheide. Eine allmähliche Annäherung der Standortbedingungen ist jedoch zu beobachten. So sanken die Gehalte an CAL-austauschbarem P_ 2 O_5 und K_2 O im Vergleich zu Werten aus dem Jahr 1993 deutlich ab. Trotz des immer noch hohen Anteils an Grünlandarten (Molinio-Arrhenatheretea) konnten sich im Vergleich zu 1986 mehr Magerrasenarten etablieren.
In dieser Arbeit wurden die histologischen und immunhistologischen Auswirkungen der
Kombination aus Inhibition des PD-1/PD-1L-Checkpoints und PLOD-2-Blockade
untersucht. Es konnte festgestellt werden, dass die Immuntherapie anschlägt und dabei
als Monotherapie die stärksten Tumornekrosen induzierte. Das Ansprechen auf die
Immuntherapie mit BMS-1166 war jedoch sehr unterschiedlich. In der Kombination mit
dem PLOD-2-Inhibitor Minoxidil wurden hingegen einheitlichere, aber auch geringere
Nekroseanteile festgestellt. Dabei muss jedoch beachtet werden, dass die
Kombinationsbehandlung die stärkste Auswirkung auf das Tumorwachstum hatte. So
waren diese Tumore die kleinsten und leichtesten, was in Zusammenhang mit dem
ausgeprägten kollagenen Netzwerk dieser Gruppe stehen könnte. Die Kombination
zeigte keine Auswirkung auf die Tumorvaskularisierung und die Zellteilungsaktivität,
sowie auch keine Auffälligkeiten bezüglich der Infiltration mit Immunzellen.
Lungenmetastasen kamen in allen Behandlungsgruppen vor. Bei der
Kombinationsbehandlung waren jedoch die durchschnittlich größten Lungenmetastasen
festzustellen.
In dieser Arbeit konnte keine klare signifikante Verbesserung der Brustkrebstherapie
durch die Kombination von Inhibition der Kollagensynthese durch PLOD-2-Blockade und
Inhibierung des PD-1/PD-1L-Checkpoints aufgezeigt werden. Das kollagene Netzwerk
war auffällig und sollte genauer untersucht werden. Es lohnt sich weiter an
Kombinationen aus Immuntherapeutikum und EZM-Destabilisierung zu arbeiten. Die
TME muss dabei weiterhin Ansatzpunkt der Forschung bleiben, um eine erleichterte
Penetration der Medikamente in den Tumor zu erzielen. Hier ist der Austausch des
Medikaments zur EZM-Destabilisierung empfehlenswert. Die LOX-Inhibierung hat sich
bereits in Kombination mit Chemotherapie als vorteilhaft erwiesen (Rossow et al., 2018)
und sollte nachfolgend in einem ähnlichen Versuchsaufbau mit dem
Immuntherapeutikum BMS-1166 ausprobiert werden.
Background
The key for successful delivery in minimally-invasive hip replacement lies in the exact knowledge about the surgical anatomy. The minimally-invasive direct anterior approach to the hip joint makes it necessary to clearly identify the tensor fasciae latae muscle in order to enter the Hueter interval without damaging the lateral femoral cutaneous nerve. However, due to the inherently restricted overview in minimally-invasive surgery, this can be difficult even for experienced surgeons.
Methods and Surgical Technique
In this technical note, we demonstrate for the first time how to use the tensor fasciae latae perforator as anatomical landmark to reliably identify the tensor fasciae latae muscle in orthopaedic surgery. Such perforators are used for flaps in plastic surgery as they are constant and can be found at the lateral third of the tensor fasciae latae muscle in a direct line from the anterior superior iliac spine.
Conclusion
As demonstrated in this article, a simple knowledge transfer between surgical disciplines can minimize the complication rate associated with minimally-invasive hip replacement.
Primary structure is determined of an insertion of a clone isolated from the library of hypophyseal cDNA of cattle by hybridization with a probe specific for prolactin. Analysis of nucleotide sequences showed that in the process of cloning, reorganization occurred in structure of preprolactin cDNA, including an inversion of the 5'-terminal and deletion of the central section of cDNA. Nevertheless, from structure of cDNA, nucleotide sequences can be deduced of extended 5'- and 3'-terminal sections of preprolactin mRNA in cattle with lengths of 257 and 551 nucleotide residues, respectively. When these sequences are compared to those established previously, some differences were found in primary structure. The most important of them is the presence of an additional codon which codes alanine at the position (-22) of the signal peptide. It is suggested that heterogeneity of preprolactin mRNA of cattle in the section coding the signal peptide is the result of alternative splicing, as was shown for preprolactin mRNA in rats.
Peptide and polypeptide hormones represent an extensive group of biologically active compounds of important significance for medicine and agriculture. In recent years genetic engineering methods have been used to create strains of microorganisms synthesizing eukaryotic proteins, including hormones and their precursors. The first stage of such developments is the isolation of DNA coding the des~red product. We have accomplished the cloning of the cDNA of a number of polypeptide and peptide hormones of the pituitary of man and domestic animals. The model gene of human calcitonin has also been synthesized and cloned. The obtained genes are being used to develop methods for the microbiological synthesis of human and animal-hormones.
B cell aggregates in the central nervous system (CNS) have been associated with rapid disease progression in patients with multiple sclerosis (MS). Here we demonstrate a key role of carcinoembryogenic antigen-related cell adhesion molecule1 (CEACAM1) in B cell aggregate formation in MS patients and a B cell-dependent mouse model of MS. CEACAM1 expression was increased on peripheral blood B cells and CEACAM1\(^+\) B cells were present in brain infiltrates of MS patients. Administration of the anti-CEACAM1 antibody T84.1 was efficient in blocking aggregation of B cells derived from MS patients. Along these lines, application of the monoclonal anti-CEACAM1 antibody mCC1 was able to inhibit CNS B cell aggregate formation and significantly attenuated established MS-like disease in mice in the absence of any adverse effects. CEACAM1 was co-expressed with the regulator molecule T cell immunoglobulin and mucin domain −3 (TIM-3) on B cells, a novel molecule that has recently been described to induce anergy in T cells. Interestingly, elevated coexpression on B cells coincided with an autoreactive T helper cell phenotype in MS patients. Overall, these data identify CEACAM1 as a clinically highly interesting target in MS pathogenesis and open new therapeutic avenues for the treatment of the disease.
B cells have only recently begun to attract attention in the immunopathology of multiple sclerosis (MS). Suitable markers for the prediction of treatment success with immunomodulatory drugs are still missing. Here we evaluated the B cell response to brain antigens in n = 34 relapsing-remitting MS (RRMS) patients treated with glatiramer acetate (GA) using the enzyme-linked immunospot technique (ELISPOT). Our data demonstrate that patients can be subdivided into responders that show brain-specific B cell reactivity in the blood and patients without this reactivity. Only in patients that classified as B cell responders, there was a significant positive correlation between treatment duration and the time since last relapse in our study. This correlation was GA-specific because it was absent in a control group that consisted of interferon-\(\beta\) (IFN-\(\beta\))-treated RRMS patients (n = 23). These data suggest that GA has an effect on brain-reactive B cells in a subset of patients and that only this subset benefits from treatment. The detection of brain-reactive B cells is likely to be a suitable tool to identify drug responders.
Multiple Sklerose (MS) ist die häufigste neurologische Erkrankung, die bei jungen Erwachsenen zu dauerhaften körperlichen Einschränkungen führt. Ein Kennzeichen der MS sind zeitlich und örtlich disseminierte entzündliche Läsionen im zentralen Nervensystem (ZNS). Die Läsionsart, die am häufigsten auftritt, ist u. a. durch Antikörperablagerungen charakterisiert. Die häufigste Verlaufsform der MS tritt in Schüben auf. Im Laufe der Erkrankung bilden sich die Symptome in der Mehrzahl der Patienten unvollständig zurück und es entwickelt sich ein chronischer Verlauf. Trotz intensiver Forschung ist die Ätiologie der MS bisher unbekannt Bis heute gibt es keine Biomarker, um den Therapieerfolg oder das Therapieversagen der MS-Basistherapeutika (Glatirameracetat und β-Interferon) zu bestimmen. Aktuelle Studien, bei denen B-Zellen depletiert wurden, zeigten eine signifikante Reduktion MS-typischer Läsionen und der Schubrate bei der schubförmigen MS. Man vermutet, dass autoreaktive B-Zellen vielfältige Aufgaben in der Pathogenese der MS übernehmen: sie produzieren Autoantikörper, präsentieren autoreaktiven T-Zellen Autoantigene und sezernieren Mediatoren, die zur Aktivierung anderer Immunzellen führen. Es ist noch unklar, welche B-Zell-Untergruppe bei der MS besondere Relevanz hat. Vor kurzem wurden B1-Zellen beim Menschen beschrieben. Eine Studie zeigte, dass die Anzahl der B1-Zellen in unbehandelten MS-Patienten signifikant erniedrigt war. Des Weiteren wurden im ZNS von chronisch erkrankten MS-Patienten B-Zell-Aggregate nachgewiesen. Diese B-Zell-Aggregate ähneln sekundären lymphatischen Organen und könnten zur Progredienz der Erkrankung beitragen. Eine ex vivo-Studie zeigte, dass die B-Zell-Aggregat-Bildung durch das Adhäsionsmolekül CEACAM1-(carcinoembryogenic antigen-related cell adhesion molecule 1) vermittelt wird. Überdies ist die Koexpression von CEACAM1 und TIM-3 (T-cell immunoglobulin- and mucin-domain containing-3) für immunerschöpfte und tolerante T-Zellen charakteristisch. Schließlich konnte unsere Arbeitsgruppe zeigen, dass ZNS-reaktive B-Zellen nur im Blut von Patienten mit einem klinisch isolierten Syndrom und MS-Patienten nachweisbar waren.
In meiner Studie habe ich den Einfluss von MS-Basistherapeutika und einer MS-Eskalationstherapie auf die B-Zell-Untergruppen untersucht. Dabei habe ich die naive B-Zell-, B-Gedächtniszell-, B1-Zell- und Plasmablasten-Zahl von gesunden Probanden sowie unbehandelten und behandelten MS-Patienten miteinander verglichen. Die B-Zell-Untergruppen wurden durchflusszytometrisch untersucht. Die B1-Zell-Zahl war bei behandelten und unbehandelten MS-Patienten signifikant erniedrigt. In einer weiteren Studie konnte ich zeigen, dass die Anwesenheit von ZNS-reaktiven B-Zellen im Blut von glatirameracetat-behandelten MS-Patienten mit dem Therapieerfolg assoziiert war. Die ZNS-reaktiven B-Zellen wurden durch einen ZNS-Lysat-ELISPOT detektiert. Schließlich habe ich in einer dritten Studie die Expression von CEACAM1 und TIM-3 auf B-Zellen bei natalizumab-behandelten MS-Patienten durchflusszytometrisch untersucht. Im Vergleich zu gesunden Probanden zeigte sich, dass im Blut der MS-Patienten die CEACAM1+- und die CEACAM1\(^+\)TIM-3\(^+\)-B-Zell-Zahl signifikant erhöht war. Im Gegensatz dazu waren CEACAM1\(^+\)TIM-3\(^+\)-T-Helferzellen signifikant erniedrigt in behandelten MS-Patienten.
Meine Arbeit belegt, dass die B1-Zell-Population unabhängig von der MS-Therapie in MS-Patienten erniedrigt ist. Ungeklärt bleibt, ob diese Erniedrigung eine Folge oder eine Ursache der Erkrankung ist. B1-Zellen sind die Quelle von natürlichen Antikörpern in Mensch und Tier. Sie haben protektive Eigenschaften und sind bei der B-Zell-Toleranzinduktion beteiligt. Die protektiven Funktionen der natürlichen Antikörper könnten durch die Erniedrigung der B1-Zell-Zahl ausbleiben. Zusätzlich waren B-Zellen mit einem immunerschöpften Phänotyp im Blut von MS-Patienten erhöht. Trotz Stimulation konnte kein Phänotyp bei T-Helferzellen induziert werden, der für tolerante und immunerschöpfte T-Zellen beschrieben worden ist. In zukünftigen Studien sollte man die B1-Zell-Zahl und die CEACAM1\(^+\)TIM-3\(^+\)-B- und -T-Zell-Zahl bei Patienten mit einem klinisch isolierten Syndrom im Liquor und im Blut untersuchen. Damit könnte man feststellen, ob B1-Zellen aus der Peripherie bei MS-Patienten in das ZNS migrieren. Die Anwesenheit ZNS-reaktiver B-Zellen im Blut von behandelten MS-Patienten zeigte sich in meiner Arbeit als ein Marker, um den Therapieerfolg zu dokumentieren. Eine weiterführende Querschnittstudie (COPSELECT) wird ZNS-reaktive B-Zellen mittels ZNS-Lysat-ELISPOT als zukünftige Therapie-Biomarker ausführlicher untersuchen. MS-Biomarker wären für den einzelnen Betroffenen von großer Bedeutung und hätten ebenfalls gesundheitsökonomisch eine hohe Relevanz.
Multiple sclerosis (MS) is an autoimmune disorder of the central nervous system (CNS) and characterized by the infiltration of immune cells, demyelination and axonal loss. Loss of axons and nerve fiber pathology are widely accepted as correlates of neurological disability. Hence, it is surprising that the development of neuroprotective therapies has been neglected for a long time. A reason for this could be the diversity of the underlying mechanisms, complex changes in nerve fiber pathology and the absence of biomarkers and tools to quantify neuroregenerative processes. Present therapeutic strategies are aimed at modulating or suppressing the immune response, but do not primarily attenuate axonal pathology. Yet, target-oriented neuroprotective strategies are essential for the treatment of MS, especially as severe damage of nerve fibers mostly occurs in the course of disease progression and cannot be impeded by immune modulatory drugs. This review shall depict the need for neuroprotective strategies and elucidate difficulties and opportunities.
In August 2004 the authors of this report undertook a survey on the Genus Oenothera (evening-primroses) in the valley of the river Main between the cities of Schweinfurt and Bamberg (Bavaria, Lower and Upper Franconia). 22 different taxa were identified, most of which are new to Bavaria, one new for Europe (Oenothera hazelae Gates) and another one new for Middle Europe (Oenothera stucchii Soldano). In this report a list of all Oenothera species found in Franconia is given and Oenothera stucchii, a particular tall (up to > 2 m) species with extremely long Hypanthia of 50-70 mm and fruit capsules with truncated teeth, is described and depicted in more detail.
LOX-catalyzed collagen stabilization is a proximal cause for intrinsic resistance to chemotherapy

The potential of altering the tumor ECM to improve drug response remains fairly unexplored. To identify targets for modification of the ECM aiming to improve drug response and overcome resistance, we analyzed expression data sets from pre-treatment patient cohorts. Cross-evaluation identified a subset of chemoresistant tumors characterized by increased expression of collagens and collagen-stabilizing enzymes. We demonstrate that strong collagen expression and stabilization sets off a vicious circle of self-propagating hypoxia, malignant signaling, and aberrant angiogenesis that can be broken by an appropriate auxiliary intervention: Interfering with collagen stabilization by inhibition of lysyl oxidases significantly enhanced response to chemotherapy in various tumor models, even in metastatic disease. Inhibition of collagen stabilization by itself can reduce or enhance tumor growth depending on the tumor type. The mechanistical basis for this behavior is the dependence of the individual tumor on nutritional supply on one hand and on high tissue stiffness for FAK signaling on the other.
Die EZM bildet ein Netzwerk quervernetzter Proteine, welches alle Zellen im Tumor umgibt. Sie übt direkte Effekte auf die Medikamenteneinbringung und -verteilung aus und somit auch auf die therapeutische Effizienz von Chemotherapeutika. Die LOX(L)-Proteinfamilie katalysiert die oxidative Desaminierung von Lysinresten in Elastin und Kollagenfasern und ermöglicht dadurch eine intra- und intermolekulare Quervernetzung. Diese wird für die Reifung und Stabilisierung der Kollagene in der EZM benötigt. Eine erhöhte LOX(L)-Expression steigert durch eine verstärkte EZM-Quervernetzung die Gewebesteifheit im Tumor und bildet so eine physikalische Diffusionsbarriere. Durch diese Barriere wird die Versorgung mit Sauerstoff und Nährstoffen reduziert. Die resultierende Hypoxie im Tumor kann eine fehlgeleitete Angiogenese triggern und zu einer Aktivierung maligner Signalkaskaden führen. In dieser Arbeit wurden durch eine LOX(L)-Inhibierung mittels βAPN einerseits und eine ektopische LOX-/LOXL2-Überexpression andererseits Auswirkungen solcher Eingriffe auf verschiedene Indikatoren wie Zellproliferation und apoptose, Versorgung mit Sauerstoff und Nährstoffen, Angiogenese, Hypoxie, Makrophageninfiltration und die Expression verschiedener Wachstumsfaktoren analysiert. Die Versuche wurden an fünf verschiedenen Tumoren (4T1-, E0771- und EMT6-Brustkarzinome, LLC-Lungenkarzinome und MT6-Fibrosarkome) durchgeführt. Die Ergebnisse dieser Arbeit demonstrieren eine direkte Verbindung zwischen der EZM und einer Therapieresistenz. Nach βAPN-Behandlung konnte eine verbesserte Versorgung mit Sauerstoff und Nährstoffen beobachtet werden, welches in einer Verringerung maligner Signalkaskaden und folglich auch in einer verbesserten Vaskularisierung resultierte. Als Konsequenz wurde die therapeutische Effizienz von Chemotherapeutika verbessert. Im Gegensatz dazu führte eine LOX-/LOXL2-Überexpression zu einer erhöhten Therapieresistenz. Die vorliegende Studie zeigt, dass die Modifizierung der EZM durch eine Hemmung von LOX(L) das Potenzial birgt, das Ansprechen von Chemotherapeutika in der Behandlung von Krebserkrankungen zu verbessern.
Bei Patienten, die an einer speziellen Form der MS erkrankten, konnten Entzündungsinfiltrate in den Meningen nachgewiesen werden, die in ihrem Aufbau lymphoidem Gewebe ähnelten. Das Auftreten dieser Infiltrate war mit einem schwereren Krankheitsverlauf assoziiert. Das Mausmodell der B-Zell-abhängigen MP4-induzierten experimentellen autoimmunen Enzephalomyelitis (EAE) zeigt in den Kleinhirnen der Mäuse Infiltrate, die den Infiltraten beim Menschen ähneln.
Wir nutzten die MP4-induzierte EAE, um die Mechanismen der Erkrankungsentstehung und Progression besser zu verstehen. Ziel dieser Arbeit war es intakte und stabile Ribonukleinsäuren (RNA) aus den Infiltraten der Mäuse zu isolieren. Sowie Identifizierung von Gene, die während verschiedener Stadien der zerebralen Entzündungsreaktion hochreguliert waren.
Wir verglichen die Möglichkeiten der RNA-Isolation bei Paraffin-eingebettetem Gewebe und kryofixiertem Gewebe. Um die Vergleichbarkeit der Qualitäts- und Quantitätsanalyse zu gewährleisten, wurde für jede Probe eine RNA Integritätsnummer (RIN) ermittelt.
Wir führten eine Laser Capture Microdissection (LCM) und anschließende Gensequenzierung der zerebralen Infiltrate bei Mäusen durch, bei denen eine MP4-abhängige EAE ausgelöst wurde. Des Weiteren verglichen wir die Ergebnisse mit den Expressionsprofilen von sekundär lymphatischen Organen (SLOs).
Insgesamt konnten wir 43 Gene herausfiltern, die im Vergleich zu den Kontrollgruppen hochreguliert waren.
Die Entwicklung von ektopen lymphatischen Strukturen (ELS) im zentralen Nervensystem (ZNS) ist ein komplexer und bisher wenig verstandener Prozess. In dieser Studie beobachteten wir 43 Gene, die während der Entwicklung von B- Zell-Infiltraten in den Kleinhirnen von MP4-immunisierten Mäusen signifikant hochreguliert waren. Von diesen Genen wurden bereits 14 im Zusammenhang mit der MS erwähnt und sind zum Teil Gegenstand aktiver Forschung.
Summary
Here we describe a novel neuro-mesodermal assembloid model that recapitulates aspects of peripheral nervous system (PNS) development such as neural crest cell (NCC) induction, migration, and sensory as well as sympathetic ganglion formation. The ganglia send projections to the mesodermal as well as neural compartment. Axons in the mesodermal part are associated with Schwann cells. In addition, peripheral ganglia and nerve fibers interact with a co-developing vascular plexus, forming a neurovascular niche. Finally, developing sensory ganglia show response to capsaicin indicating their functionality.
The presented assembloid model could help to uncover mechanisms of human NCC induction, delamination, migration, and PNS development. Moreover, the model could be used for toxicity screenings or drug testing. The co-development of mesodermal and neuroectodermal tissues and a vascular plexus along with a PNS allows us to investigate the crosstalk between neuroectoderm and mesoderm and between peripheral neurons/neuroblasts and endothelial cells.

We demonstrated previously that phosphocholine and phosphocholine-modified macromolecules efficiently inhibit ATP-dependent release of interleukin-1β from human and murine monocytes by a mechanism involving nicotinic acetylcholine receptors (nAChR). Interleukin-1β is a potent pro-inflammatory cytokine of innate immunity that plays pivotal roles in host defence. Control of interleukin-1β release is vital as excessively high systemic levels cause life threatening inflammatory diseases. In spite of its structural similarity to acetylcholine, there are no other reports on interactions of phosphocholine with nAChR. In this study, we demonstrate that phosphocholine inhibits ion-channel function of ATP receptor P2X7 in monocytic cells via nAChR containing α9 and α10 subunits. In stark contrast to choline, phosphocholine does not evoke ion current responses in Xenopus laevis oocytes, which heterologously express functional homomeric nAChR composed of α9 subunits or heteromeric receptors containing α9 and α10 subunits. Preincubation of these oocytes with phosphocholine, however, attenuated choline-induced ion current changes, suggesting that phosphocholine may act as a silent agonist. We conclude that phophocholine activates immuno-modulatory nAChR expressed by monocytes but does not stimulate canonical ionotropic receptor functions.
Die Verbreitung der Gattung Chamaesyce auf den Friedhöfen des Landkreises Main-Spessart, Bayern

Während der Jahre 2003 bis 2007 wurden Vorkommen und Verbreitung der Gattung Chamaesyce auf 126 Friedhöfen des Landkreises Main-Spessart untersucht. Drei Arten, C. humifusa, C. maculata und C. prostrata, konnten nachgewiesen werden. Bevorzugte Wuchsorte sind neben Bahnhöfen, botanischen Gärten und Gärtnereien vor allem Friedhöfe. Auf den untersuchten 126 Friedhöfen des Main-Spessart-Kreises wuchsen die Chamaesyce-Arten auf Kies- und Sandwegen, in Pflaster- und Plattenfugen, auf Gräbern und Beeten. C. humifusa wurde auf 27, C. maculata auf 46 und C. prostrata auf einem der 126 Friedhöfe des Untersuchungsgebietes gefunden.
Early treatment with glucocorticoids could help reduce both cytotoxic and vasogenic edema, leading to improved clinical outcome after stroke. In our previous study, isosteviol sodium (STVNA) demonstrated neuroprotective effects in an in vitro stroke model, which utilizes oxygen-glucose deprivation (OGD). Herein, we tested the hypothesis that STVNA can activate glucocorticoid receptor (GR) transcriptional activity in brain microvascular endothelial cells (BMECs) as previously published for T cells. STVNA exhibited no effects on transcriptional activation of the glucocorticoid receptor, contrary to previous reports in Jurkat cells. However, similar to dexamethasone, STVNA inhibited inflammatory marker IL-6 as well as granulocyte-macrophage colony-stimulating factor (GM-CSF) secretion. Based on these results, STVNA proves to be beneficial as a possible prevention and treatment modality for brain ischemia-reperfusion injury-induced blood–brain barrier (BBB) dysfunction.
The present study was conducted on the rOCT1, a member of SLC22 family. Structurally, it consists of 12 membrane spanning α-helices with both N- and C-termini intracellular. Studies done so far, through tracer uptake and inhibition, reconstitution of rOCT1 in nanodiscs and proteoliposomes and voltage-clamp fluorometry, have identified the main amino acids in the cleft of rOCT1 that interact in a critical manner with the substrates/inhibitors either directly or indirectly. Homology modeling studies have also supported these observations. In the present study we aimed at measuring the binding of substrates MPP+ and TEA+ to rOCT1 at 0oC in order to establish the amino acids in the cleft region that interact with the substrate when the transporter is frozen in the outward-open conformation. Previously identified crucial amino acids (Asp475, Phe160, Leu447, Arg440, Trp218 and Tyr222) were selected for the study. rOCT1 wild-type and its mutants were stably expressed in HEK293 cells and these cells were used for the binding measurements with the radioactive substrate (MPP+ or TEA+) at 0°C in Mg-Ca-PBS buffer as described in “Materials and Methods” section in detail. rOCT1 wild-type revealed for MPP+-binding a KD which was not significantly different from the corresponding Km value. Also, after addition of 10 nM non-radioactive MPP+, an initial increase of about 20% in bound MPP+ was observed. The results indicate that the Km for transport is dependent on the binding of MPP+ to the outward-open conformation and hints at the possibility of allosteric interaction between the binding sites. Mutations at position Trp218, Phe160 and Asp475 resulted in a change in the KD value. Trp218 mutations also showed an allosteric increase similar to the rOCT1 wild-type. This study suggests that these amino acids are located at a critical position in the outward-open conformation for MPP+ transport. TEA+-binding could not be observed in rOCT1 wild-type, indicating that the binding site is perhaps inaccessible for TEA+ in frozen outward-open state. The mutants D475E, F160A, L447F, R440K and Y222F showed a very low affinity binding with a very high KD value as compared to the corresponding Km values indicating that the transporter might have different affinities for extra-cellular binding alone and for the complete transport process especially if temperature is the limiting factor. Substrate inhibition studies done using both MPP+ and TEA+ have confirmed the existence of overlapping binding sites for these two ligands. This study has confirmed the direct interaction of Trp218, Phe160, Asp475 with MPP+ and Phe160, Asp475, Leu447, Arg440 and Tyr222 with TEA+ in the outward-open conformation.
Die Identifizierung endogener Stammzellen mit kardiogenem Potenzial und die Möglichkeit, deren Differenzierung zu steuern, würde einen Meilenstein in der kardioregenerativen Therapie darstellen. Innerhalb der Gefäßwand konnten unterschiedliche Stamm- und Vorläuferzellen identifiziert werden, die sog. Gefäßwand-residenten Stammzellen (VW-SCs). Zuletzt konnten aus CD34(+) VW-SCs, ohne genetische Manipulation, Kardiomyozyten generiert werden. Zusätzlich fungiert die Gefäßwand als Quelle inflammatorischer Zellen, die essenziell für die kardiogene Differenzierung der VW-SCs zu sein scheinen.
Ziel dieser Arbeit war es, das Verhalten von CD44(+) VW-SCs zu untersuchen, um herauszufinden, inwieweit dieser Stammzelltyp eine endogene Generierung von Kardiomyozyten unterstützen könnte. Dabei wurde mit infarzierten Mäuseherzen, dem Aortenringassay (ARA) und dem kardialen Angiogeneseassay (CAA) gearbeitet.
Sowohl in vivo in ischämischen Arealen infarzierter Mäuseherzen als auch ex vivo im CAA kam es zu einem signifikanten Anstieg von CD44(+) Zellen. Mittels Färbungen auf CD44 und Ki-67 konnte die Teilungsfähigkeit dieser Zellen demonstriert werden.
Ex vivo ließen sich aus CD44(+) Zellen F4/80(+) Makrophagen generieren. Die CD44(+) VW-SCs können sich dabei sowohl zu pro-inflammatorischen iNOS(+) M1- als auch zu anti-inflammatorischen IL-10(+) M2-Makrophagen differenzieren. Eine Modulation der kardialen Inflammation könnte einen entscheidenden Einfluss auf die Kardiomyogenese haben.
Unter VEGF-A kam es im CAA zu einer deutlichen Zunahme von CD44(+) Zellen. Unter Lenvatinib blieb das kardiale Sprouting gänzlich aus, die Anzahl der CD44(+) Zellen stagnierte und die VW-SCs verblieben in ihren physiologischen Nischen innerhalb der Gefäßwand.
Warum es nach einem MI kaum zu einer funktionellen Herzmuskelregeneration kommt, ist weiterhin unklar. Die therapeutische Beeinflussung koronaradventitieller CD44(+) VW-SCs und inflammatorischer Prozesse könnte dabei zukünftig eine wichtige therapeutische Option darstellen.
Eine familiäre Myopathie und Kardiomyopathie, der eine Missense-Mutation des alpha-B-Crystallin-Gens zugrunde liegt, weist auf eine wichtige Bedeutung des Stressproteins alpha-B-Crystallin im Herzen hin. Die chaperone-ähnlichen Eigenschaften von alpha-B-Crystallin und die unter kardialer Ischämie zu beobachtende schnelle Translokation vom Zytosol an das elastische Titin-Filamentsystem lassen eine protektive Rolle von alpha-B-Crystallin unter Stressbedingungen vermuten. Ziel der vorliegenden Arbeit war es, eine eventuelle kardioprotektive Funktion von alphaB-Crystallin durch die Charakterisierung alpha-B-Crystallin gendeletierter Mäuse nachzuweisen. Wir etablierten hierfür ein Versuchssystem zur Untersuchung der Kontraktilität isolierter Papillarmuskeln im Organbad. Im Rahmen des Aufbaus unseres Versuchssystems untersuchten wir zunächst den Einfluss der Ca2+-Konzentration, der Temperatur und der Kontraktionsbedingungen (Auxotonie vs. Isometrie) auf die Kraft-Frequenz-Beziehung von murinem Myokard. Wir konnten zeigen, dass die Kraft-Frequenz-Beziehung von Myokardpräparaten der Maus von den genannten Versuchsbedingungen abhängig ist. Bei niedrigen Ca2+-Konzentrationen und Temperaturen ([Ca2+] = 1,0 mM, Temp. = 27 °C) ist sie positiv, flacht bei zunehmender Ca2+-Konzentration und Temperatur ab und ist für [Ca2+] = 5,0 mM, Temp. = 37 °C negativ. Auxotone Kontraktionsbedingungen führen im Vergleich zu isometrischen bei gleichen Ca2+-Konzentrationen und Temperaturen zu einem flacheren Verlauf der Kraft-Frequenz-Beziehung. Unter annähernd physiologischen Bedingungen verläuft die Kraft-Frequenz-Beziehung des Mäuse-Myokards flach bis leicht positiv. Im Gegensatz zum Menschen scheinen somit bei der Maus für eine Steigerung des Herz-Zeit-Volumens andere Mechanismen als eine positive Kraft-Frequenz-Beziehung von Bedeutung zu sein. Hierbei ist insbesondere der Frank-Starling-Mechanismus und die sympathoadrenerge Innervation des Herzens zu erwähnen. Zur Charakterisierung der kardialen Funktion von alphaB-Crystallin untersuchten wir die Kontraktilität isolierter Papillarmuskeln von Wildtyp- und alpha-B-Crystallin gendeletierten Mäusen unter simulierter Ischämie (Glucose- und Sauerstoffentzug) und Reperfusion im Organbad. Unter Kontrollbedingungen zeigten sich zwischen wt- und alpha-B-/- Muskeln keine Unterschiede in der Zuckungskraft, der Geschwindigkeit der Kraftentwicklung und der Relaxationszeit. Die während der 20-minütigen simulierten Ischämie entwickelte Kontraktur setzte jedoch bei den alpha-B-/- Muskeln signifikant früher ein und verlief signifikant stärker als bei wt-Muskeln. Nach einer 60-minütigen Reperfusionsphase blieb die Kontraktur der alpha-B-/- Muskeln im Vergleich zu wt-Muskeln signifikant erhöht. Bezüglich Zuckungskraft, Geschwindigkeit der Kraftentwicklung und Relaxationszeit zeigten sich weder während noch nach simulierter Ischämie deutliche Unterschiede zwischen den Muskeln beider Mäusestämme. Diese Ergebnisse lassen darauf schließen, dass das Fehlen von alpha-B-Crystallin am Gesamtherz nicht zu einer Störung der systolischen Herzfunktion, sondern zu einer eingeschränkten myokardialen Relaxationsfähigkeit unter Ischämie und Reperfusion führen würde. Da alpha-B-Crystallin unter kardialer Ischämie an das elastische Titin-Filamentsystem bindet, könnten die elastischen Eigenschaften des Myokards unter Ischämie durch einen Mangel an alpha-B-Crystallin derart beeinträchtigt werden, dass es zu einer höheren Rigidität der Muskulatur kommt. Eine Funktion von alpha-B-Crystallin im Herzen ist somit möglicherweise die Aufrechterhaltung der elastischen Eigenschaften des Myokards unter kardialer Ischämie und Reperfusion.
Recombinant beta interferons-1 (IFNβ-1) are used as first line therapies in patients with relapsing multiple sclerosis (MS), a chronic inflammatory and neurodegenerative disease of the CNS. IFNβ-1a/b has moderate effects on the prevention of relapses and slowing of disease progression. Fibroblast growth factors (FGFs) and FGF receptors (FGFRs) are known to play a key role in the pathology of MS and its model EAE. To investigate the effects of short-term treatment with s.c. IFNβ-1a versus the combined application of s.c. IFNβ-1a and oligodendrocyte-specific deletion of FGFR1 (Fgfr1\(^{ind−/−}\) mice) in MOG\(_{35-55}\)-induced EAE. IFNβ-1a (30 mg/kg) was applied s.c. from days 0–7 p.i. of EAE in controls and Fgfr1\(^{ind−/−}\) mice. FGFR signaling proteins associated with inflammation/degeneration in MS/EAE were analyzed by western blot in the spinal cord. Further, FGFR1 in Oli-neu oligodendrocytes were inhibited by PD166866 and treated with IFNβ-1a (400 ng/mL). Application of IFNβ-1a over 8 days resulted in less symptoms only at the peak of disease (days 9–11) compared to controls. Application of IFNβ-1a in Fgfr1\(^{ind−/−}\) mice resulted in less symptoms primarily in the chronic phase of EAE. Fgfr1\(^{ind−/−}\) mice treated with IFNβ-1a showed increased expression of pERK and BDNF. In Oli-neu oligodendrocytes, treatment with PD166866 and IFNβ-1a also showed an increased expression of pERK and BDNF/TrkB. These data suggest that the beneficial effects in the chronic phase of EAE and on signaling molecules associated with ERK and BDNF expression are caused by the modulation of FGFR1 and not by interferon beta-1a. FGFR may be a potential target for therapy in MS.
Multiple sclerosis (MS) is a chronic inflammatory and degenerative disease of the central nervous system (CNS). MS commonly affects the cerebellum causing acute and chronic symptoms. Cerebellar signs significantly contribute to clinical disability, and symptoms such as tremor, ataxia, and dysarthria are difficult to treat. Fibroblast growth factors (FGFs) and their receptors (FGFRs) are involved in demyelinating pathologies such as MS. In autopsy tissue from patients with MS, increased expression of FGF1, FGF2, FGF9, and FGFR1 was found in lesion areas. Recent research using mouse models has focused on regions such as the spinal cord, and data on the expression of FGF/FGFR in the cerebellum are not available. In recent EAE studies, we detected that oligodendrocyte-specific deletion of FGFRs results in a milder disease course, less cellular infiltrates, and reduced neurodegeneration in the spinal cord. The objective of this study was to characterize the role of FGFR1 in oligodendrocytes in the cerebellum. Conditional deletion of FGFR1 in oligodendrocytes (Fgfr1\(^{ind−/−}\) was achieved by tamoxifen application, EAE was induced using the MOG\(_{35-55}\) peptide. The cerebellum was analyzed by histology, immunohistochemistry, and western blot. At day 62 p.i., Fgfr1\(^{ind−/−}\) mice showed less myelin and axonal degeneration compared to FGFR1-competent mice. Infiltration of CD3(+) T cells, Mac3(+) cells, B220(+) B cells and IgG(+) plasma cells in cerebellar white matter lesions (WML) was less in Fgfr1\(^{ind−/−}\)mice. There were no effects on the number of OPC or mature oligodendrocytes in white matter lesion (WML). Expression of FGF2 and FGF9 associated with less myelin and axonal degeneration, and of the pro-inflammatory cytokines IL-1β, IL-6, and CD200 was downregulated in Fgfr1\(^{ind−/−}\) mice. The FGF/FGFR signaling protein pAkt, BDNF, and TrkB were increased in Fgfr1\(^{ind−/−}\) mice. These data suggest that cell-specific deletion of FGFR1 in oligodendrocytes has anti-inflammatory and neuroprotective effects in the cerebellum in the EAE disease model of MS.
Multiple sclerosis (MS) is a chronic inflammatory and neurodegenerative disease of the central nervous system (CNS) affecting more than two million people worldwide. In MS, oligodendrocytes and myelin sheaths are destroyed by autoimmune-mediated inflammation, while remyelination is impaired. Recent investigations of post-mortem tissue suggest that Fibroblast growth factor (FGF) signaling may regulate inflammation and myelination in MS. FGF2 expression seems to correlate positively with macrophages/microglia and negatively with myelination; FGF1 was suggested to promote remyelination. In myelin oligodendrocyte glycoprotein (MOG)\(_{35–55}\)-induced experimental autoimmune encephalomyelitis (EAE), systemic deletion of FGF2 suggested that FGF2 may promote remyelination. Specific deletion of FGF receptors (FGFRs) in oligodendrocytes in this EAE model resulted in a decrease of lymphocyte and macrophage/microglia infiltration as well as myelin and axon degeneration. These effects were mediated by ERK/Akt phosphorylation, a brain-derived neurotrophic factor, and downregulation of inhibitors of remyelination. In the first part of this review, the most important pharmacotherapeutic principles for MS will be illustrated, and then we will review recent advances made on FGF signaling in MS. Thus, we will suggest application of FGFR inhibitors, which are currently used in Phase II and III cancer trials, as a therapeutic option to reduce inflammation and induce remyelination in EAE and eventually MS.
Fibroblast growth factor (FGF) signaling is involved in the pathogenesis of multiple sclerosis (MS). Data from neuropathology studies suggest that FGF signaling contributes to the failure of remyelination in MS. In MOG\(_{35–55}\)-induced EAE, oligodendrocyte-specific deletion of FGFR1 and FGFR2 resulted in a less severe disease course, reduced inflammation, myelin and axon degeneration and changed FGF/FGFR and BDNF/TrkB signaling. Since signaling cascades in oligodendrocytes could not be investigated in the EAE studies, we here aimed to characterize FGFR-dependent oligodendrocyte-specific signaling in vitro. FGFR inhibition was achieved by application of the multi-kinase-inhibitor dovitinib and the FGFR1/2/3-inhibitor AZD4547. Both substances are potent inhibitors of FGF signaling; they are effective in experimental tumor models and patients with malignancies. Effects of FGFR inhibition in oligodendrocytes were studied by immunofluorescence microscopy, protein and gene analyses. Application of the tyrosine kinase inhibitors reduced FGFR1, phosphorylated ERK and Akt expression, and it enhanced BDNF and TrkB expression. Furthermore, the myelin proteins CNPase and PLP were upregulated by FGFR inhibition. In summary, inhibition of FGFR signaling in oligodendrocytes can be achieved by application of tyrosine kinase inhibitors. Decreased phosphorylation of ERK and Akt is associated with an upregulation of BDNF/TrkB signaling, which may be responsible for the increased production of myelin proteins. Furthermore, these data suggest that application of FGFR inhibitors may have the potential to promote remyelination in the CNS.
Taxane (wie Paclitaxel oder Cabazitaxel) sind bewährte Arzneimittel in den systemischen Therapieschemata vieler bösartiger Erkrankungen, einschließlich Brust- und Eierstockkrebs. Sie fördern die Stabilisierung der Mikrotubuli, was zu einem Stillstand des Zellzyklus während der Mitose führt, auf den die Apoptose folgt. Neben dieser antimitotischen Wirkung von Taxanen ist seit einiger Zeit auch eine gefäßverändernde Wirkung von Taxanen bekannt. Kürzlich wurde gezeigt, dass Taxane tatsächlich Störungen in der Gefäßarchitektur verursachen, indem sie den Kalziumeinstrom über TRPC6, einen unselektiven Kationenkanal, auslösen. Der erhöhte intrazelluläre Ca2+-Spiegel bewirkt eine Rundung der Endothelzellen, was zu einer Störung des endothelialen Monolayers, Serumausfluss und Gefäßkollaps führt.
In dieser Arbeit konzentrierten wir uns auf die Gefäßbetten von peripheren Organen wie dem Herzen oder der Niere in Abhängigkeit vom Tumorstadium und der Taxol-Behandlung. Die Organe wurden mit immunhistochemischen Techniken angefärbt, um Veränderungen in der Architektur und Morphologie der Blutgefäße zu untersuchen.
Wir fanden Veränderungen in der Morphologie der Kapillaren des Herzens und darüber hinaus Veränderungen in der Expression endothelialer Antigene in Abhängigkeit vom Tumorstadium, insbesondere eine zunehmende endotheliale Expression von TRPC6 in Abhängigkeit vom Tumorstadium.
Diese Ergebnisse liefern neue Erkenntnisse für das Verständnis der systemischen Auswirkungen maligner Erkrankungen und tragen dazu bei, Folgeerkrankungen bei Patienten mit fortgeschrittenem Krebs zu verhindern.
Background
Multiple sclerosis (MS) is an autoimmune disease of the central nervous system (CNS) characterized by inflammation, demyelination and axonal pathology. Myelin basic protein/proteolipid protein (MBP-PLP) fusion protein MP4 is capable of inducing chronic experimental autoimmune encephalomyelitis (EAE) in susceptible mouse strains mirroring diverse histopathological and immunological hallmarks of MS. Limited availability of human tissue underscores the importance of animal models to study the pathology of MS.
Methods
Twenty-two female C57BL/6 (B6) mice were immunized with MP4 and the clinical development of experimental autoimmune encephalomyelitis (EAE) was observed. Methylene blue-stained semi-thin and ultra-thin sections of the lumbar spinal cord were assessed at the peak of acute EAE, three months (chronic EAE) and six months after onset of EAE (long-term EAE). The extent of lesional area and inflammation were analyzed in semi-thin sections on a light microscopic level. The magnitude of demyelination and axonal damage were determined using electron microscopy. Emphasis was put on the ventrolateral tract (VLT) of the spinal cord.
Results
B6 mice demonstrated increasing demyelination and severe axonal pathology in the course of MP4-induced EAE. In addition, mitochondrial swelling and a decrease in the nearest neighbor neurofilament distance (NNND) as early signs of axonal damage were evident with the onset of EAE. In semi-thin sections we observed the maximum of lesional area in the chronic state of EAE while inflammation was found to a similar extent in acute and chronic EAE. In contrast to the well-established myelin oligodendrocyte glycoprotein (MOG) model, disease stages of MP4-induced EAE could not be distinguished by assessing the extent of parenchymal edema or the grade of inflammation.
Conclusions
Our results complement our previous ultrastructural studies of B6 EAE models and suggest that B6 mice immunized with different antigens constitute useful instruments to study the diverse histopathological aspects of MS.
The pathophysiology of tremor in Parkinson’s disease (PD) is evolving towards a complex alteration to monoaminergic innervation, and increasing evidence suggests a key role of the locus coeruleus noradrenergic system (LC-NA). However, the difficulties in imaging LC-NA in patients challenge its direct investigation. To this end, we studied the development of tremor in a reserpinized rat model of PD, with or without a selective lesioning of LC-NA innervation with the neurotoxin DSP-4. Eight male rats (Sprague Dawley) received DSP-4 (50 mg/kg) two weeks prior to reserpine injection (10 mg/kg) (DR-group), while seven male animals received only reserpine treatment (R-group). Tremor, rigidity, hypokinesia, postural flexion and postural immobility were scored before and after 20, 40, 60, 80, 120 and 180 min of reserpine injection. Tremor was assessed visually and with accelerometers. The injection of DSP-4 induced a severe reduction in LC-NA terminal axons (DR-group: 0.024 ± 0.01 vs. R-group: 0.27 ± 0.04 axons/um\(^2\), p < 0.001) and was associated with significantly less tremor, as compared to the R-group (peak tremor score, DR-group: 0.5 ± 0.8 vs. R-group: 1.6 ± 0.5; p < 0.01). Kinematic measurement confirmed the clinical data (tremor consistency (% of tremor during 180 s recording), DR-group: 37.9 ± 35.8 vs. R-group: 69.3 ± 29.6; p < 0.05). Akinetic–rigid symptoms did not differ between the DR- and R-groups. Our results provide preliminary causal evidence for a critical role of LC-NA innervation in the development of PD tremor and foster the development of targeted therapies for PD patients.
Zusammenfassungen 83 5 Zusammenfassungen 5.1 Zusammenfassung Durch Expressionsklonierung wurde 1994 der erste organische Kationentransporter, rOCT1, aus der Ratte isoliert (Gründemann et al., 1994). 1999 wurde eine Aminosäure in der 11. Transmembrandomäne von rOCT1 entdeckt, welche Teil der Substratbindungstasche dieses Transporters war (Gorboulev et al., 1999). Zielsetzung der vorliegenden Arbeit war es weitere funktionell relevante Aminosäuren zu identifizieren. Ein Vergleich der „Helical Wheels“ aller 12 hypothetischen Transmembrandomänen zeigte eine Akkumulation von 5 OCT und OCTN spezifischen Aminosäuren in der vierten Transmembrandomäne auf einer Seite. Bei diesen Aminosäuren handelte es sich um K215, W218, Y222, T226 und V229. Es wurden verschiedene Punktmutationen an diesen Positionen eingeführt. Es zeigte sich mit Hilfe radioaktiv markierter Substrate von rOCT1, dass selbst Substitutionen durch strukturell verwandte Aminosäuren bei den flankieren Aminosäuren zu einem Ausfall des rOCT1 vermittelten Substrattransport führte. Weiterhin schien an Position 218 für den rOCT1- vermittelten Transport von TEA eine aromatische Aminosäure von großer funktioneller Relevanz zu sein. Wir vermuten hier eine Kationen p-Elektronen Interaktion des aromatischen Ringsystems des Tryptophans mit der positiven Ladung des TEA. Versuche mit den Mutanten des Tyrosins 222 zeigten ebenfalls Änderungen bei den Transportraten und Affinitäten verschiedener Substrate. Eine Kationen-p Interaktion konnte ausgeschlossen werden, jedoch war die Affinität der Mutante Y222F zum TPeA um einen Faktor 20 gegenüber dem Wildtyp erhöht. Weiterführende Untersuchungen mit der Zwei- Elektrodenspannungsklemme zeigten unterschiedliche Affinitäten des TPeA zum Wildtyp im Vergleich zum mutierten Protein in seiner nach außen bzw. nach innen gerichteten Konformation. War die Form der Inhibierung des TEA-induzierten Stromes durch TPeA beim Wildtyp kompetitiv, so zeigte sie bei der Mutante einen nicht-kompetitiven Charakter. Die Mutante T226A zeigte ebenfalls Änderungen in Affinität und Selektivität. Bei allen transportierenden Mutanten zeigte sich, dass der Transport von MPP nicht oder kaum verändert war, hingegen wurden sehr starke Änderungen der Transportcharakteristika von TEA gefunden, was auf verschiedene Substratbindungsstellen in rOCT1 hinweist. Diese Versuche zeigen deutlich die funktionelle Relevanz und die Beteiligung der mutierten Aminosäurepositionen an der Substratbindetasche von rOCT1. In Versuchen, in welchen Chinin als Inhibitor des rOCT2 vermittelten Transports genutzt wurde, passierte Chinin mittels Diffusion in seiner ungeladenen Form die Oozytenmembran und hemmte rOCT2 von der Innenseite. Dies könnte der Grund für die nicht-kompetitive Form der Inhibition der TEA-Aufnahme durch Chinin sein. Diese Versuche wurden dadurch bestätigt, dass die protonierte Form des Chinins eine kompetitive Form der Inhibition zeigte und den Transporter von außen hemmte.
Reorganisation der Zellkontakte der Endothelbarriere bei der Stabilisierung durch cAMP und Rac1

Zwischen Blutkompartiment und umliegenden Interstitium besteht eine Barriere, die durch eine einzelne Schicht aus Endothelzellen gebildet wird. Essentiell für diese Barriere, deren Funktion in der Begrenzung des Austausches von Flüssigkeit und gelösten Stoffen liegt, sind interzelluläre Junktionen, welche die Endothelzellen miteinander verbinden. Durch eine gestörte Funktion und Regulation der Endothelbarriere entstehen beim Menschen verschiedene Pathologien wie zum Beispiel Ödeme, hämorrhagischer Schlaganfall und vaskuläre Malformationen.
Es ist bekannt, dass cAMP die Endothelbarriere zum Teil durch Aktivierung der kleinen GTPase Rac1 stabilisiert. Trotz der großen medizinischen Relevanz dieses Signalweges, sind die damit einhergehenden Effekte auf die interzellulären Kontakte auf ultrastruktureller Ebene weitgehend unbekannt.
In mikrovaskulären Endothelzellkulturen kam es ähnlich wie in intakten Mikrogefäßen zur Stärkung der Barrierefunktion. So resultierte sowohl nach Behandlung mit Forskolin und Rolipram (F/R), welche zur Steigerung der intrazellulären cAMP-Spiegel führen, als auch nach Zugabe von 8-(4-chlorophenylthio)-2´-O-methyladenosin-3´,5´-cyclic monophosphorothioate (O-Me-cAMP), einem selektiven Aktivator des cAMP nachgeschalteten Epac/Rap1-Signalweges, ein Anstieg des TER; außerdem konnte durch beide Substanzen (F/R und O-Me-cAMP) die Aktivierung von Rac1 induziert werden. Desweiteren wurde eine verstärkte Intensität und Linearisierung des Immunfluoreszenzsignals der Zelljunktionsproteine VE-Cadherin und Claudin5 entlang der Zellgrenzen beobachtet.
In der ultrastrukturellen Analyse der interzellulären Kontaktzonen-Architektur zeigte sich unter F/R- oder O-Me-cAMP-Exposition ein signifikanter Anstieg an komplexen Interdigitationen. Diese komplexen Strukturen waren dadurch charakterisiert, dass sich die Membranen benachbarter Zellen, die durch zahlreiche endotheliale Junktionen stabilisiert wurden, über vergleichsweise lange Distanzen eng aneinanderlegten, so dass ein deutlich verlängerter Interzellularspalt resultierte. Die Inhibition der Rac1-Aktivierung durch NSC-23766 verminderte die Barrierefunktion und blockierte effektiv die O-Me-cAMP-vermittelte Barrierestabilisierung und Reorganisation der Kontaktzone einschließlich der Junktionsproteine.
Demgegenüber konnte die F/R-vermittelte Barrierestabilisierung durch NSC-23766 nicht beeinträchtigt werden.
Parallel dazu durchgeführte Experimente mit makrovaskulären Endothelien zeigten, dass es in diesem Zelltyp unter Bedingungen erhöhter cAMP-Konzentrationen weder zur Rac1-Aktivierung noch zur Barrierestärkung oder Kontaktzonen-Reorganisation kam.
Diese Ergebnisse deuten darauf hin, dass in mikrovaskulären Endothelien Rac1-vermittelte Änderungen der Kontaktzonen-Morphologie zur cAMP-induzierten Barrierestabilisierung beitragen.
Cell-permeant recombinant Nanog protein promotes pluripotency by inhibiting endodermal specification

A comprehensive understanding of the functional network of transcription factors establishing and maintaining pluripotency is key for the development of biomedical applications of stem cells. Nanog plays an important role in early development and is essential to induce natural pluripotency in embryonic stem cells (ESCs). Inducible gain-of-function systems allowing a precise control over time and dosage of Nanog activity would be highly desirable to study its vital role in the establishment and maintenance of pluripotency at molecular level. Here we engineered a recombinant cell permeable version of Nanog by fusing it with the cell penetrating peptide TAT. Nanog-TAT can be readily expressed in and purified from E. coli and binds to a consensus Nanog DNA sequence. At cellular level it enhances proliferation and self-renewal of ESCs in the absence of leukemia inhibitory factor (LIF). Nanog-TAT together with LIF acts synergistically as judged by enhanced clonogenicity and activation of an Oct4-promoter-driven GFP reporter gene. Furthermore Nanog-TAT, in the absence of LIF, promotes pluripotency by inhibiting endodermal specification in a Stat3-independent manner. Our results demonstrate that Nanog protein transduction is an attractive tool allowing control over dose and time of addition to the cells for studying the molecular control of pluripotency without genetic manipulation.
Photon-counting detector (PCD) CT allows for ultra-high-resolution (UHR) examinations of the shoulder without requiring an additional post-patient comb filter to narrow the detector aperture. This study was designed to compare the PCD performance with a high-end energy-integrating detector (EID) CT. Sixteen cadaveric shoulders were examined with both scanners using dose-matched 120 kVp acquisition protocols (low-dose/full-dose: CTDI\(_{vol}\) = 5.0/10.0 mGy). Specimens were scanned in UHR mode with the PCD-CT, whereas EID-CT examinations were conducted in accordance with the clinical standard as “non-UHR”. Reconstruction of EID data employed the sharpest kernel available for standard-resolution scans (ρ\(_{50}\) = 12.3 lp/cm), while PCD data were reconstructed with both a comparable kernel (11.8 lp/cm) and a sharper dedicated bone kernel (16.5 lp/cm). Six radiologists with 2–9 years of experience in musculoskeletal imaging rated image quality subjectively. Interrater agreement was analyzed by calculation of the intraclass correlation coefficient in a two-way random effects model. Quantitative analyses comprised noise recording and calculating signal-to-noise ratios based on attenuation measurements in bone and soft tissue. Subjective image quality was higher in UHR-PCD-CT than in EID-CT and non-UHR-PCD-CT datasets (all p < 0.001). While low-dose UHR-PCD-CT was considered superior to full-dose non-UHR studies on either scanner (all p < 0.001), ratings of low-dose non-UHR-PCD-CT and full-dose EID-CT examinations did not differ (p > 0.99). Interrater reliability was moderate, indicated by a single measures intraclass correlation coefficient of 0.66 (95% confidence interval: 0.58–0.73; p < 0.001). Image noise was lowest and signal-to-noise ratios were highest in non-UHR-PCD-CT reconstructions at either dose level (p < 0.001). This investigation demonstrates that superior depiction of trabecular microstructure and considerable denoising can be realized without additional radiation dose by employing a PCD for shoulder CT imaging. Allowing for UHR scans without dose penalty, PCD-CT appears as a promising alternative to EID-CT for shoulder trauma assessment in clinical routine.
In this study, the impact of reconstruction sharpness on the visualization of the appendicular skeleton in ultrahigh-resolution (UHR) photon-counting detector (PCD) CT was investigated. Sixteen cadaveric extremities (eight fractured) were examined with a standardized 120 kVp scan protocol (CTDI\(_{vol}\) 10 mGy). Images were reconstructed with the sharpest non-UHR kernel (Br76) and all available UHR kernels (Br80 to Br96). Seven radiologists evaluated image quality and fracture assessability. Interrater agreement was assessed with the intraclass correlation coefficient. For quantitative comparisons, signal-to-noise-ratios (SNRs) were calculated. Subjective image quality was best for Br84 (median 1, interquartile range 1–3; p ≤ 0.003). Regarding fracture assessability, no significant difference was ascertained between Br76, Br80 and Br84 (p > 0.999), with inferior ratings for all sharper kernels (p < 0.001). Interrater agreement for image quality (0.795, 0.732–0.848; p < 0.001) and fracture assessability (0.880; 0.842–0.911; p < 0.001) was good. SNR was highest for Br76 (3.4, 3.0–3.9) with no significant difference to Br80 and Br84 (p > 0.999). Br76 and Br80 produced higher SNRs than all kernels sharper than Br84 (p ≤ 0.026). In conclusion, PCD-CT reconstructions with a moderate UHR kernel offer superior image quality for visualizing the appendicular skeleton. Fracture assessability benefits from sharp non-UHR and moderate UHR kernels, while ultra-sharp reconstructions incur augmented image noise.
Aims/hypothesis
Several glucose-sensing pathways have been implicated in glucose-triggered secretion of glucagon-like peptide-1 (GLP-1) from intestinal L cells. One involves glucose metabolism and closure of ATP-sensitive K\(^+\) channels, and another exploits the electrogenic nature of Na\(^+\)-coupled glucose transporters (SGLTs). This study aimed to elucidate the role of these distinct mechanisms in glucose-stimulated GLP-1 secretion.
Methods
Glucose uptake into L cells (either GLUTag cells or cells in primary cultures, using a new transgenic mouse model combining proglucagon promoter-driven Cre recombinase with a ROSA26tdRFP reporter) was monitored with the FLII\(_{12}\)Pglu-700μδ6 glucose sensor. Effects of pharmacological and genetic interference with SGLT1 or facilitative glucose transport (GLUT) on intracellular glucose accumulation and metabolism (measured by NAD(P)H autofluorescence), cytosolic Ca\(^{2+}\) (monitored with Fura2) and GLP-1 secretion (assayed by ELISA) were assessed.
Results
L cell glucose uptake was dominated by GLUT-mediated transport, being abolished by phloretin but not phloridzin. NAD(P)H autofluorescence was glucose dependent and enhanced by a glucokinase activator. In GLUTag cells, but not primary L cells, phloretin partially impaired glucose-dependent secretion, and suppressed an amplifying effect of glucose under depolarising high K\(^+\) conditions. The key importance of SGLT1 in GLUTag and primary cells was evident from the impairment of secretion by phloridzin or Sglt1 knockdown and failure of glucose to trigger cytosolic Ca\(^{2+}\) elevation in primary L cells from Sglt1 knockout mice.
Conclusions/interpretation
SGLT1 acts as the luminal glucose sensor in L cells, but intracellular glucose concentrations are largely determined by GLUT activity. Although L cell glucose metabolism depends partially on glucokinase activity, this plays only a minor role in glucose-stimulated GLP-1 secretion.
RS1 ist ein 67-68 kD großes, ubiquitär exprimiertes Protein, das sich an der Innenseite der Plasmamembran befindet und in den Zellkern wandern kann. Durch immunhistochemischen Untersuchungen an Dünndarmschnitten der Maus konnte RS1 das erste Mal in dieser Arbeit im Kern und an der Membran von Enterozyten gezeigt werden. RS1 wird von einem intronlosen Single Copy Gen kodiert und ist fähig Ubiquitin über eine Ubiquitin-assoziierte (UBA) Domäne zu binden. Es reduziert die Konzentration einiger Proteine in der Plasmamembran. Durch Expressionsversuche in Xenopus Oozyten wurde gezeigt, dass RS1 die Menge des Na+-D-Glukosekotransporters SGLT1 in der Plasmamembran transkriptionsunabhängig reduziert. Entsprechend seiner dualen Lokalisation beteiligt sich RS1 aber auch an der Transkriptionsregulation im Zellkern. In der vorliegenden Arbeit konnten Informationen über die physiologische Funktion des membranassoziierten Regulatorproteins RS1 gewonnen werden. Nach Erstellung einer RS1-knock-out Maus wurde sichergestellt, dass ein erfolgreiches Rekombinationsereignis stattgefunden hatte und RS1 tatsächlich nicht mehr exprimiert wurde. Die RS1-knock-out Mäuse waren postnatal lebensfähig, vermehrten sich gut und entwickelten eine Fettsucht mit 30 % mehr Körpergewicht, 80 % mehr Fett und um 40 % vergrößerten Fettzellen. Bei den transgenen Mäusen war weder die Nahrungsaufnahme gesteigert, noch die motorische Aktivität verringert. In der Bürstensaummembran des Dünndarmepithels konnte bei den RS1-knock-out Mäusen die siebenfache Menge an Protein des Na+-abhängigen D-Glukosekotransporters SGLT1 detektiert werden, während die Konzentration des passiven Glukosetransporters GLUT2 in der basolateralen Membran nicht verändert war. Die Zunahme der SGLT1-Proteinmenge war posttranskriptional bedingt. Bei der RS1-knock-out Maus wirkt sich der in Oozyten beobachtete Effekt an der Plasmamembran aus, während der an konfluenten LLCPK1 Zellen gezeigte Effekt im Zellkern nicht zum Tragen kommt. Die transgenen Tiere resorbierten die doppelte Menge an D-Glukose im Dünndarm. Das spricht dafür, dass bei der RS1-knock-out Maus der „turnover“ des SGLT1 beeinflusst sein muss, da die siebenfache SGLT1-Proteinmenge einem verdoppelten Transport über den SGLT1 gegenübersteht. Die RS1-knock-out Mäuse zeigten normale Insulinspiegel und reguläre oralen Glukosebelastungstests. Bei gefütterten Mäusen waren die Serumleptinspiegel ähnlich wie bei Wildtypmäusen, die typische Reduzierung des Serumleptinspiegel konnte bei den Mäusen ohne RS1 aber nicht beobachtet werden. Untersuchungen an Fettzellexplantaten ergaben, dass die Sekretion von Leptin bei RS1- knock-out-Explantaten erhöht war, während die Leptinsynthese und die insulinabhängige Regulation der Leptinsekretion nicht verändert waren. Mit der RS1-knock-out Maus wurde ein neues Fettsuchtmodell geschaffen. RS1 spielt eine physiologisch wichtige Rolle bei der Regulation der D-Glukoseaufnahme im Darm. Der visceralen Adipositas liegt wahrscheinlich eine gesteigerte Nahrungsutilisation durch die verbesserte Glukoseaufnahme über den SGLT1 im Darm zugrunde. Die gesteigerte Glukoseabsorption ist ursächlich für den Anstieg der Fettmasse. Die Fettzellen vergrößern sich und sezernieren dann mehr Leptin. Es ist davon auszugehen, dass die RS1-knock-out Mäuse eine veränderte Nahrungsutilisation aufgrund der verbesserten Glukoseaufnahme im Dünndarm aufweisen. Die Adipositas demzufolge ein sekundärer Effekt. Gleichzeitig kann aber nicht ausgeschlossen werden, dass RS1 direkt auf die Zellen des weißen Fettgewebes wirkt und bei Wildtypmäusen die Sekretion des Leptins aus Vesikeln hemmt.
In enterocytes, protein RS1 (RSC1A1) mediates an increase of glucose absorption after ingestion of glucose-rich food via upregulation of Na+-D-glucose cotransporter SGLT1 in the brush-border membrane (BBM). Whereas RS1 decelerates the exocytotic pathway of vesicles containing SGLT1 at low glucose levels between meals, RS1-mediated deceleration is relieved after ingestion of glucose-rich food. Regulation of SGLT1 is mediated by RS1 domain RS1-Reg, in which Gln-Ser-Pro (QSP) is effective. In contrast to QSP and RS1-Reg, Gln-Glu-Pro (QEP) and RS1-Reg with a serine to glutamate exchange in the QSP motif downregulate the abundance of SGLT1 in the BBM at high intracellular glucose concentrations by about 50%. We investigated whether oral application of QEP improves diabetes in db/db mice and affects the induction of diabetes in New Zealand obese (NZO) mice under glucolipotoxic conditions. After 6-day administration of drinking water containing 5 mM QEP to db/db mice, fasting glucose was decreased, increase of blood glucose in the oral glucose tolerance test was blunted, and insulin sensitivity was increased. When QEP was added for several days to a high fat/high carbohydrate diet that induced diabetes in NZO mice, the increase of random plasma glucose was prevented, accompanied by lower plasma insulin levels. QEP is considered a lead compound for development of new antidiabetic drugs with more rapid cellular uptake. In contrast to SGLT1 inhibitors, QEP-based drugs may be applied in combination with insulin for the treatment of type 1 and type 2 diabetes, decreasing the required insulin amount, and thereby may reduce the risk of hypoglycemia.
Die MP4-induzierte experimentelle autoimmune Encephalomyelitis (EAE) erlaubt eine fokussierte Betrachtung von B-Zellen, die eine wichtige Rolle bei der Pathogenese der Multiplen Sklerose (MS) spielen. Es konnte zum Beispiel gezeigt werden, dass das Vorhandensein von B-Zell-Aggregaten im zentralen Nervensystem (ZNS) von MS-Patienten mit einem aggravierten Krankheitsverlauf assoziiert war. Diese Follikel könnten dabei als ektope lymphatische Strukturen den Immunprozess aktiv gestalten und somit ein therapeutisches Ziel darstellen. In der vorliegenden Studie wurde der Effekt des Sphingosin-1-Phosphat-Rezeptor-Modulators Fingolimod (FTY720) auf die autoreaktive B-Zell-Antwort und speziell die Bildung von B-Zell-Aggregaten im Kleinhirn der MP4-EAE-Mäuse untersucht.

During stroke the blood–brain barrier (BBB) is damaged which can result in vasogenic brain edema and inflammation. The reduced blood supply leads to decreased delivery of oxygen and glucose to affected areas of the brain. Oxygen and glucose deprivation (OGD) can cause upregulation of glucose uptake of brain endothelial cells. In this letter, we investigated the influence of MK801, a non-competitive inhibitor of the NMDA-receptor, on the regulation of the glucose uptake and of the main glucose transporters glut1 and sglt1 in murine BBB cell line cerebEND during OGD. mRNA expression of glut1 was upregulated 68.7- fold after 6 h OGD, which was significantly reduced by 10 μM MK801 to 28.9-fold. Sglt1 mRNA expression decreased during OGD which was further reduced by MK801. Glucose uptake was significantly increased up to 907% after 6 h OGD and was still higher (210%) after the 20 h reoxygenation phase compared to normoxia. Ten micromolar MK801 during OGD was able to reduce upregulated glucose uptake after OGD and reoxygenation significantly. Presence of several NMDAR subunits was proven on the mRNA level in cerebEND cells. Furthermore, it was shown that NMDAR subunit NR1 was upregulated during OGD and that this was inhibitable by MK801. In conclusion, the addition of MK801 during the OGD phase reduced significantly the glucose uptake after the subsequent reoxygenation phase in brain endothelial cells.
Ziel dieser Arbeit war die Untersuchung des Einflusses von ACh und vom cholinergen Signalling in der Angiogenese. Neben der klassischen Rolle des AChs im neuronalen System, konnten es in den letzten Jahren auch in nicht-neuronal innerviertem Gewebe festgestellt werden. In dieser Arbeit konzentrierte man sich auf beobachtete cholinerge Zellen in der Aortenwand einer ChAT-eGFP-Maus und in davon ausgehenden neu gebildeten Gefäßen. Nach Durchführung des Aortenringassays nach Baker konnte zunächst nachgewiesen werden, dass es sich bei ACh um einen Angiogenese-Stimulator handelt. Eine Carbacholkonzentration von 1 µM erwies sich als bester Angiogenesestimulator. Der Inhibitorversuch mit Lenvatinib, einem Hemmer des VEGFR, zeigte, dass die cholinerge Wirkung zur Aktivierung ähnlicher angiogeneseseinhibierenden Mechanismen wie bei einer VEGF-Stimulation kommt. Bei der zeitlichen Weiterverfolgung des Versuchs konnte man feststellen, dass sich die Gefäßformation mit Dauer des Versuches ändert.
Bei der immunhistologischen Analyse der neu gebildeten Gefäße wurde am ehesten eine Überlappung der ChAT-Zellen mit Zellen gefunden, die sich positiv für die Perizytenmarker NG2 und Desmin zeigten. Es ist anzunehmen, dass das ACh, exprimiert von einigen Perizyten, eine Leitschiene für das sich neu bildende Gefäß bietet und zu einem gerichteten Wachstum führt. Vor Kurzem wurde in der Wand erwachsener Blutgefäße eine Nische für Stammzellen identifiziert. Um zu untersuchen ob die ChAT-Zellen dort ihren Ursprung haben, wurde mit Stammzellmarkern gefärbt. Diese Arbeit hat mit ihren Ergebnissen eine signifikante Grundlage für weiterführende Studien geliefert.
A comprehensive analysis of the molecular network of cellular factors establishing and maintaining pluripotency as well as self renewal of pluripotent stem cells is key for further progress in understanding basic stem cell biology. Nanog is necessary for the natural induction of pluripotency in early mammalian development but dispensable for both its maintenance and its artificial induction. To gain further insight into the molecular activity of Nanog, we analyzed the outcomes of Nanog gain-of-function in various cell models employing a recently developed biologically active recombinant cell-permeant protein, Nanog-TAT. We found that Nanog enhances the proliferation of both NIH 3T3 and primary fibroblast cells. Nanog transduction into primary fibroblasts results in suppression of senescence-associated beta-galactosidase activity. Investigation of cell cycle factors revealed that transient activation of Nanog correlates with consistent downregulation of the cell cycle inhibitor p27\(^{KIP1}\) (also known as CDKN1B). By performing chromatin immunoprecipitation analysis, we confirmed bona fide Nanog-binding sites upstream of the p27\(^{KIP1}\) gene, establishing a direct link between physical occupancy and functional regulation. Our data demonstrates that Nanog enhances proliferation of fibroblasts through transcriptional regulation of cell cycle inhibitor p27 gene.
Endothelzellen kleiden als einschichtiger Zellverband die Blutgefäße aus und bilden so eine Barriere zwischen Blut und Interstitium. Der Austausch von Flüssigkeit und Makromolekülen über diese Barriere wird durch die transzelluläre und parazelluläre Permeabilität reguliert. Die parazelluläre Permeabilität ist von der Integrität der interzellulären endothelialen Junktionen abhängig. Eine Schwächung der Adhäsion und Öffnung der Tight Junctions bedingt unweigerlich einen Anstieg der Permeabilität, die bei verschiedenen pathologischen Bedingungen, z.B. inflammatorischen Ödemen und allergischem Schock, lebensbedrohlich werden kann. Unter physiologischen Be-dingungen ist das Endothel verschiedenen mechanischen Stimuli wie Scherstress durch den Blutfluß, zyklischer Dehnung durch die Wandspannung und hydrostatischem Druck durch den Blutdruck ausgesetzt. Da die Effekte des hydrostatischen Drucks auf die Biologie der Endothelzelle weitgehend unverstanden sind, sollte in der vorliegenden Arbeit der Einfluss des physiologischen hydrostatischen Drucks auf die Integrität des endothelialen Zellverbands näher untersucht werden. Sowohl in mikrovaskulären Endothelzellen als auch in makro-vaskulären Endothelzellen wurde gefunden, dass hydrostatischer Druck von 5-15 cmH2O, wie er typischerweise in Blutkapillaren in vivo herrscht, einen protektiven Einfluss auf die Endothelbarriere gegenüber permeabilitätssteigernden Einflüssen vermittelt. Es konnte gezeigt werden, dass eine extrazelluläre Depletion von Ca2+ durch EGTA zu einem Verlust von VE-Cadherin aus den endothelialen Junktionen mit Lückenbildung zwischen den Zellen führt (dargestellt durch Immunfluoreszenz) und dass dieser Effekt durch die gleichzeitige Applikation eines hydrostatischen Drucks von 15 cmH2O weitgehend verhindert werden konnte. Auch die durch Cytochalasin D induzierte Actindepolymerisation und interzelluläre Lückenbildung sowie die Dissoziation der Zellkontakte und Zellablösung nach Zugabe des Ca2+/Calmodulin-Antagonisten Trifluperazin und die Thrombin-induzierte Zelldissoziation konnten durch gleichzeitige Druckexposition von 15 cmH2O inhibiert werden. Darüberhinaus konnte mit Hilfe der Laserpinzetten-Technik gezeigt werden, dass hydrostatischer Druck die Haftung von mit VE-Cadherin beschichteten Mikroperlen an der endothelialen Zelloberfläche sowohl in Abwesenheit von extrazellulärem Ca2+ als auch unter Einfluss von Cytochalasin D und Trifluperazin nahezu unvermindert ermöglichte, während ohne hydrostatischen Druck die Mikroperlen unter diesen Bedingungen (Ca2+-Depletion, Cytochalasin D, Trifluperazin) nicht mehr hafteten. Im weiteren Verlauf der Arbeit wurde untersucht, welche Mechanismen an den druckvermittelten Signalwegen beteiligt sein könnten. Es ist bekannt, dass cAMP und auch die Mitglieder der Rho-GTPasen-Familie Endothelbarriere-stabilisierende Funktionen haben. Es konnten jedoch keine signifikanten Veränderungen der cAMP-Konzentrationen sowie der Rho A- und Rac 1-Aktivität in makrovaskulären Endothelzellen unter hydrostatischem Druck von 15 cmH2O innerhalb von 45 Minuten nachgewiesen werden. Da Caveolin-1 in der Literatur eine Rolle in der Mechanotransduktion von zyklischer Dehnung und Scherstress zugesprochen wird, wurden im Labor generierte Endothelzellen aus Caveolin-1-defizienten Mäusen untersucht. Caveolin-1 stabilisiert plasmalemmale Invaginationen, die Caveolae, die eine Vielzahl an Molekülen mit signalgebenden und -weiterleitenden Funktionen beherbergen. In Caveolin-1-defizienten Endothelzellen war hydrostatischer Druck nicht in der Lage eine Destabilisierung des endothelialen Zellrasens durch Cytochalsin D, Trifluperazin und EGTA zu unterdrücken. Die Ergebnisse dieser Arbeit haben gezeigt, dass ein physiologischer hydrostatischer Druck zur Erhaltung der endothelialen Integrität und ihrer Barrierefunktion beiträgt und Caveolin-1-vermittelte Mechanismen bei der Mechanotransduktion des hydrostatischen Drucks eine Rolle spielen.
