Two-Dimensional Electron Systems at Surfaces — Spin-Orbit Interaction and Electronic Correlations

This thesis addresses three different realizations of a truly two-dimensional electron system (2DES), established at the surface of elemental semiconductors, i.e., Pt/Si(111), Au/Ge(111), and Sn/Si(111). Characteristic features of atomic structures at surfaces have been studied using scanning tunneling microscopy and low energy electron diffraction with special emphasis on Pt deposition onto Si(111). Topographic inspection reveals that Pt atoms agglomerate as trimers, which represent the structural building block of phase-slip domains. Surprisingly, each trimer is rotated by 30° with respect to the substrate, which results in an unexpected symmetry breaking. In turn, this represents a unique example of a chiral structure at a semiconductor surface, and marks Pt/Si(111) as a promising candidate for catalytic processes at the atomic scale. Spin-orbit interactions (SOIs) play a significant role at surfaces involving heavy adatoms. As a result, a lift of the spin degeneracy in the electronic states, termed as Rashba effect, may be observed. A candidate system to exhibit such physics is Au/Ge(111). Its large hexagonal Fermi sheet is suggested to be spin-split by calculations within the density functional theory. Experimental clarification is obtained by exploiting the unique capabilities of three-dimensional spin detection in spin- and angle-resolved photoelectron spectroscopy. Besides verification of the spin splitting, the in-plane components of the spin are shown to possess helical character, while also a prominent rotation out of this plane is observed along straight sections of the Fermi surface. Surprisingly and for the first time in a 2DES, additional in-plane rotations of the spin are revealed close to high symmetry directions. This complex spin pattern must originate from crystalline anisotropies, and it is best described by augmenting the original Rashba model with higher order Dresselhaus-like SOI terms. The alternative use of group-IV adatoms at a significantly reduced coverage drastically changes the basic properties of a 2DES. Electron localization is strongly enhanced, and the ground state characteristics will be dominated by correlation effects then. Sn/Si(111) is scrutinized with this regard. It serves as an ideal realization of a triangular lattice, that inherently suffers from spin frustration. Consequently, long-range magnetic order is prohibited, and the ground state is assumed to be either a spiral antiferromagnetic (AFM) insulator or a spin liquid. Here, the single-particle spectral function is utilized as a fundamental quantity to address the complex interplay of geometric frustration and electronic correlations. In particular, this is achieved by combining the complementary strengths of ab initio local density approximation (LDA) calculations, state-of-the-art angle-resolved photoelectron spectroscopy, and the sophisticated many-body LDA+DCA. In this way, the evolution of a shadow band and a band backfolding incompatible with a spiral AFM order are unveiled. Moreover, beyond nearest-neighbor hopping processes are crucial here, and the spectral features must be attributed to a collinear AFM ground state, contrary to common expectation for a frustrated spin lattice.
Obgleich das erzählerische Syntagma der Tristansage sich ohne weitere Zugaben zu entfalten vermag, ist die Liebesgeschichte von Tristan und Isolde schon in der Mitte des 12. Jahrhunderts eng mit den Legenden von König Artus und seinen Rittern verwoben. Die Frage, wie sich der keltische Tristanstoff zum Artusroman verhält, ist in der mediävistischen Forschung bislang jedoch nur in Ansätzen und zumeist wenig kontrovers diskutiert worden. Dabei bietet gerade die kontingente Verbindung beider literarischer Welten bei Eilhart und Béroul zu einem arthurisierten Tristanroman die Möglichkeit, die zentralen Scharnierstellen zweier Erzähltradition zu untersuchen und nach den Möglichkeiten und Grenzen des Erzählens von der Liebe um 1200 zu fragen.
Ziel der vorliegenden, experimentellen Arbeit war die Untersuchung der Persönlichkeitseigenschaft der Impulsivität bei adulten Patienten mit ADHS und vergleichbaren Versuchspersonen. Da die Ätiopathogenese erhöhter Impulsivität bei Patienten mit ADHS bislang ungeklärt ist, eine Beeinflussung des dopaminergen Systems durch vorausgehende Studien allerdings nahe liegt, wurden die verwendeten Impulsivitätsmessungen zusätzlich bezüglich einer möglichen Beeinflussung durch den Val158Met-COMT-Polymorphismus analysiert. Die Untersuchung beinhaltet mit 71 adulten Patienten mit ADHS und 54 gesunden Kontrollpersonen, die nach Alter, Geschlecht, IQ, Kopfumfang und Händigkeit vergleichbar waren, eine der aktuell größten Stichproben adulter Patienten mit ADHS. Während einer Stoppsignal-Aufgabe zur Erfassung der Antworthemmungsfähigkeit als Korrelat der Impulsivität wurden die Verhaltensdaten der Stichproben sowie die hirnphysiologischen Veränderungen mittels funktioneller Nahinfrarotspektroskopie aufgezeichnet und ausgewertet. Die erhobenen experimentellen Daten wurden anschließend mit selbstbewerteter Impulsivität des I7-Impulsivitätsfragebogens nach Eysenck verglichen und auf mögliche Zusammenhänge überprüft. Zudem wurden die beobachteten Ergebnisse der vorliegenden Arbeit auf einen möglichen Einfluss durch den Val158Met-COMT-Polymorphismus untersucht. Auf der Verhaltensebene zeigten Patienten mit ADHS im Vergleich zu gesunden Kontrollpersonen sowohl für die SSRZ als auch für die Go-RZ signifikant langsamere Reaktionszeiten. Im Vergleich der funktionellen Daten konnten für Patienten mit ADHS während der erfolgreichen Stopp-Trials signifikant bzw. tendenziell verminderte Aktivierungen im Bereich des IFC und DLPFC in beiden Hemisphären festgestellt werden. Die Untersuchung der selbstbewerteten Impulsivität anhand des I7-Fragebogens ergab ebenfalls einen signifikanten Unterschied zwischen den beiden Stichproben. Bezüglich der Zusammenhänge zwischen den einzelnen Impulsivitätsmessungen konnten für Patienten mit ADHS signifikant bzw. tendenziell negative Zusammenhänge zwischen SSRZ und Oxygenierung im rechten IFC sowie zwischen SSRZ bzw. Go-RZ und I7-Impulsivitätswerten festgestellt werden. In der Untersuchung des Einflusses durch den Val158Met-COMT-Polymorphismus ergab sich ein sehr heterogenes Bild, in dem sich keine eindeutig systematischen Genotyp- oder Interaktionseffekte zeigten. Während die beobachteten Befunde auf Verhaltensebene auf eine generelle Verlangsamung adulter Patienten mit ADHS im Vergleich zu gesunden Kontrollpersonen hindeuten, könnte das verminderte Aktivierungsmuster im Bereich des IFC und DLPFC während der erfolgreichen Stopp-Trials bei Patienten mit ADHS möglicherweise das zugrundeliegende funktionelle Korrelat zu den beobachteten Reaktionsunterschieden zwischen den beiden Stichproben darstellen. Obwohl Patienten mit ADHS im Vergleich zu gesunden Kontrollen aufgrund ihrer erhöhten Impulsivität defizitär erscheinen, deuten die Befunde bezüglich des Zusammenhangs zwischen selbstbewerteter und experimentell erhobener Impulsivität innerhalb der Patientengruppe einen möglichen Vorteil erhöhter I7-Impulsivitätswerte für die Reaktionsfähigkeit in der Stoppsignal-Aufgabe an. Bezüglich der Beeinflussung der erhobenen Daten durch den Val158Met-COMT-Polymorphismus lassen sich anhand der Befunde dieser Arbeit keine klaren Schlussfolgerungen ziehen.
Das Airtraq ist ein neues Videolaryngoskop, für das eine Version für die Doppellumenintubation existiert. Die Doppellumenintubation gilt aufgrund spezifischer Charakteristika als schwierige Intubation. In dieser Studie wurde der Nutzen des Airtraq-Laryngoskops im Vergleich zum Macintosh-Laryngoskop untersucht.
Evaluation of a pathophysiological role of the interleukin-6-type cytokine oncostatin M (OSM) for human diseases has been complicated by the fact that mouse models of diseases targeting either OSM or the OSM receptor (OSMR) complex cannot fully reflect the human situation. This is due to earlier findings that human OSM utilizes two receptor complexes, glycoprotein 130 (gp130)/leukemia inhibitory factor receptor (LIFR) (type I) and gp130/OSMR (type II), both with wide expression profiles. Murine OSM on the other hand only binds to the gp130/OSMR (type II) receptor complex with high affinity. Here, we characterize the receptor usage for rat OSM. Using different experimental approaches (knock-down of the OSMR expression by RNA interference, blocking of the LIFR by LIF-05, an antagonistic LIF variant and stably transfected Ba/F3 cells) we can clearly show that rat OSM surprisingly utilizes both, the type I and type II receptor complex, therefore mimicking the human situation. Furthermore, it displays cross-species activities and stimulates cells of human as well as murine origin. Its signaling capacities closely mimic those of human OSM in cell types of different origin in the way that strong activation of the Jak/STAT, the MAP kinase as well as the PI3K/Akt pathways can be observed. Therefore, rat disease models would allow evaluation of the relevance of OSM for human biology.
Cover contact graphs

We study problems that arise in the context of covering certain geometric objects called seeds (e.g., points or disks) by a set of other geometric objects called cover (e.g., a set of disks or homothetic triangles). We insist that the interiors of the seeds and the cover elements are pairwise disjoint, respectively, but they can touch. We call the contact graph of a cover a cover contact graph (CCG). We are interested in three types of tasks, both in the general case and in the special case of seeds on a line: (a) deciding whether a given seed set has a connected CCG, (b) deciding whether a given graph has a realization as a CCG on a given seed set, and (c) bounding the sizes of certain classes of CCG’s. Concerning (a) we give efficient algorithms for the case that seeds are points and show that the problem becomes hard if seeds and covers are disks. Concerning (b) we show that this problem is hard even for point seeds and disk covers (given a fixed correspondence between graph vertices and seeds). Concerning (c) we obtain upper and lower bounds on the number of CCG’s for point seeds.
Optimal open-loop control, i.e. the application of an analytically derived control rule, is demonstrated for nanooptical excitations using polarization-shaped laser pulses. Optimal spatial near-field localization in gold nanoprisms and excitation switching is realized by applying a shift to the relative phase of the two polarization components. The achieved near-field switching confirms theoretical predictions, proves the applicability of predefined control rules in nanooptical light–matter interaction and reveals local mode interference to be an important control mechanism.
Ziel der Studie ist die Optimierung der Messinstrumente zur Ermittlung neurokognitiver Defizite vor und nach Herzbypassoperationen, um einen Beitrag zur Etablierung eines einheitlichen Standards zu leisten. Hierfür findet die TAP als computergestütztes Verfahren im Vergleich zu konventionellen Papier-Bleistift-Tests Einsatz. Dass Patienten nach einer ACVB-Operation neurokognitive Defizite aufweisen, hat sich in dieser Studie bewahrheitet. Aufgrund der in dieser Studie erhobenen Daten setzt sich eine suffiziente Testbatterie zur Überprüfung neurokognitiver Leistungseinschränkungen nach Herzbypassoperationen aus dem Benton-Test und dem Test „Zahlennachsprechen“ zur Überprüfung der Gedächtnisfunktion sowie den TAP-Tests „Alertness“ mit Warnton, „Alertness“ ohne Warnton und „Geteilte Aufmerksamkeit“, zusammen.
The superconducting properties of complex materials like the recently discovered iron-pnictides or strontium-ruthenate are often governed by multi-orbital effects. In order to unravel the superconductivity of those materials, we develop a multi-orbital implementation of the functional renormalization group and study the pairing states of several characteristic material systems. Starting with the iron-pnictides, we find competing spin-fluctuation channels that become attractive if the superconducting gap changes sign between the nested portions of the Fermi surface. Depending on material details like doping or pnictogen height, these spin fluctuations then give rise to $s_{\pm}$-wave pairing with or without gap nodes and, in some cases, also change the symmetry to $d$-wave. Near the transition from nodal $s_{\pm}$-wave to $d$-wave pairing, we predict the occurrence of a time-reversal symmetry-broken $(s+id)$-pairing state which avoids gap nodes and is therefore energetically favored. We further study the electronic instabilities of doped graphene, another fascinating material which has recently become accessible and which can effectively be regarded as multi-orbital system. Here, the hexagonal lattice structure assures the degeneracy of two $d$-wave pairing channels, and the system then realizes a chiral $(d+id)$-pairing state in a wide doping range around van-Hove filling. In addition, we also find spin-triplet pairing as well as an exotic spin-density wave phase which both become leading if the long-ranged hopping or interaction parameters are slightly modified, for example, by choosing different substrate materials. Finally, we consider the superconducting state of strontium-ruthenate, a possible candidate for chiral spin-triplet pairing with fascinating properties like the existence of half-quantum vortices obeying non-Abelian statistics. Using a microscopic three orbital description including spin-orbit coupling, we demonstrate that ferromagnetic fluctuations are still sufficient to induce this $\bs{\hat{z}}(p_x\pm ip_y)$-pairing state. The resulting superconducting gap reveals strong anisotropies on the $d_{xy}$-dominated Fermi-surface pocket and nearly vanishes on the other remaining two pockets.
The "Large Hadron Collider" (LHC) is currently the most powerful particle accelerator. It provides particle collisions at a center of mass energy in the Tera-electronvolt range, which had never been reached in a laboratory before. Thereby a new era in high energy particle physics has began. Now it is possible to test one of the most precise theories in physics, the Standard Model of particle physics, at these high energies. The purpose is particularly served by four large experiments installed at the LHC, namely "A Toroidal LHC ApparatuS" (ATLAS), the "Compact-Muon-Solenoid" (CMS), the "Large Hadron Collider beauty" (LHCb) and "A Large Ion Collider Experiment" (ALICE). Besides exploring the high energy behavior of the well-established portions of the Standard Model, one of the main objectives is to find the Higgs boson included in the model, but not discovered by any preceding effort. It is of tremendous importance since fermions and heavy electroweak gauge bosons acquire mass because of this boson. Although the success of the Standard Model in describing nature is already undisputed, there are some flaws due to observations inexplicable within this theory only. Therefore searches for physics beyond the Standard Model are promoted at the LHC experiments as well. In order to achieve the defined goals, crucial aspects are firstly precise measurements, to verify Standard Model predictions in detail, and secondly an evaluation of as much information as accessible by the detectors, to recognize new phenomena as soon as possible for subsequent optimizations. Both challenges are only possible with a superior understanding of the detectors. An inevitable contribution to attain this knowledge is a realistic simulation, partially requiring new implementation techniques to describe the very complex instrumentation. The research presented here is performed under the patronage of the ATLAS collaboration with a special focus on measurements done with muon spectrometer. Thus a first central issue is the performance of the spectrometer in terms of physics objects that are recognized by the device, the compatibility of data and the existing simulation as well as its improvement and finally the extension of the acceptance region. Once the excellent behavior and comprehension of the muon spectrometer is demonstrated, a second part addresses one physics use case of reconstructed muons. The electroweak force is part of the Standard Model and causes the interaction of heavy electroweak gauge bosons with fermions as well as their self-interaction. In proton-proton collisions such gauge bosons are produced. However, they decay immediately into a pair of fermions. In case of the Z boson, which is one of the gauge bosons, oppositely charged fermions of the same generation, including muons, emerge. The various decay modes are determined precisely at particle accelerators other than the LHC. However, the associated production of two Z bosons is measured less exactly at those facilities because of a very low cross section. The corresponding results acquired with the ATLAS experiment exceed all previous measurements in terms of statistics and accuracy. They are reported in this thesis as obtained from the observation of events with four charged leptons. The enhancement of the signal yield based on the extension of the muon spectrometer acceptance is especially emphasized as well as alternative methods to estimate background events. Furthermore, the impact on the probing of couplings of three Z bosons and intersection with the search for the Standard Model Higgs boson are pointed out.
The present thesis is devoted to the spectroscopic study of rare earth manganites RMnO3 (R = Gd, Dy, Tb, Eu(1 - x)Y(x)) in the submillimeter frequency range. A dynamic manifestation of a strong magnetoelectric coupling in these systems is the existence of electromagnons - spin waves excited by the electric component of the electromagnetic wave. The exact analytical solution of the Landau-Lifshitz equations obtained for cycloidal antiferromagnets builds the bridge between the inelastic neutron scattering and the optical experiments. A semi-quantitative agreement is achieved between the theory and the results by these two experimental techniques. Two suggested mechanisms of the magnetoelectric coupling, the inverse Dzyaloshinskii-Moriya (IDM) interaction and the symmetric Heisenberg exchange (HE) striction, are introduced in a perturbative manner. The qualitative conclusions regarding both static and dynamic electric properties are also in agreement with the experiment. GdMnO3 is the system in which the electromagnons were first detected at low frequencies. Far infrared measurements in GdMnO3 presented here have confirmed the existence of a second high frequency electromagnon at 75 reciprocal centimeter. The detection of an additional mode suggests the existence of at least short range ferroelectric order. Such order has not been observed in static experiments so far. The electromagnons in Eu(1 - x)Y(x)MnO3 helped to clarify the role of the rare earth magnetism. As the Y(3+) ions are diamagnetic and Eu(3+) ions possess Van Vleck paramagnetism only, it is the Mn subsystem that is primarily responsible for the magnetoelectric properties of rare earth manganites. The electromagnons in DyMnO3 and TbMnO3 do not change their excitation conditions upon the flop of the spin cycloid in external magnetic fields. This fact still lacks consistent theoretical explanation. Detailed measurements on TbMnO3 of different orientations have allowed to prove the existence of the IDM electromagnon. The study of DyMnO3 in external magnetic fields has shown that, depending on the Dy ordering, the electromagnons and static electric polarization can be either enhanced or suppressed. Thus, the magnetic order of rare earth moments still plays an important role. As a general result of the present work, the IDM interaction is capable to describe the static electric polarization and the weak electro-active excitation in the high-field phase of TbMnO3. The HE model is successful in explaining the high frequency electromagnon, including its excitation conditions and the spectral weight. However, both models are still unable to describe the energy and the spectral weight of the low frequency electromagnon. Further theoretical and experimental efforts are required in this direction.
In der vorliegenden Arbeit wurde die Eignung einer im 3D-Pulverdruckverfahren fabrizierten Trägerstruktur auf Calciumphosphat-Basis (Bruschit) als Zellkultur-Scaffold untersucht. Dazu wurden die Konstrukte in vitro mit osteoblastären Zellen besiedelt und deren Proliferations- und Differenzierungsverhalten über eine Kultivierungsdauer von 12 Tagen analysiert. Als Parameter dienten hierbei die Zellviabilität, die Aktivität des osteoblastären Enzyms Alkalische Phosphatase sowie die Mediumkonzentration von Osteocalcin. Des Weiteren wurde der pH-Wert des Kulturmediums sowie die Konzentrationen der freien Elektrolyte Calcium und Phosphat untersucht. Die Ergebnisse belegen eine gute Zytokompatibilität des Trägermaterials. Diese äußerte sich in einer progredienten Proliferation phänotypisch osteoblastärer Zellen (gemäß Rasterelektronenmikroskopie). Die Zellen exprimierten das ostoblastentypische Enzym Alkalische Phosphatase, welches als früher Differenzierungsmarker gilt. Die Analyse der Osteocalcinproduktion führte aufgrund methodischer Probleme nicht zu verwertbaren Ergebnissen. Die Untersuchung des verbrauchten Zellkulturmediums ergab keine unphysiologischen Schwankungen des pH-Wertes. Jedoch konnten signifikante Veränderungen der Konzentration an freien Calcium und Phosphat-Ionen im Medium festgestellt werden. Diese sind auf die Löslichkeit des Trägermaterials im physiologischen Milieu zurückzuführen. Zusammenfassend konnte mittels vorliegender in vitro Versuche eine geeignete Zytokompatibilität des untersuchten Materials herausgearbeitet werden. Für mögliche klinische Anwendungen zum Knochenersatz sind weitergehende Untersuchungen, insbesondere osteokonduktiver Eigenschaften im orthotopen Implantatlager im Rahmen von in vivo Untersuchungen, erforderlich.
Klinische Ergebnisse und Lebensqualität nach neoadjuvanter Radiochemotherapie von Rektumkarzinomen

Die derzeitige Standardtherapie bei fortgeschrittenen Rektumkarzinomen der UICC Stadien II und III besteht aus der neoadjuvanten Radio(chemo)therapie mit nachfolgender chirurgischer Intervention. Hierbei werden die beiden Therapiemodalitäten, der Kurzzeit-Radiotherapie(5x5Gy) und unmittelbare Operation von der Langzeit-Radiochemotherpaie (28x1,8Gy) mit einem Intervall von 4-6 Wochen bis zur Operation, unterschieden. Im Hinblick auf das Auftreten von Lokalrezidiven sowie auf das Gesamtüberleben sprechen die Ergebnisse für eine bessere Wirksamkeit der LZ-RChT. Dennoch gibt es klinische Situation, bei denen eine KZ-Radiotherapie sinnvoller sein kann. Somit kann als Konsequenz eine differenzierte Indikationsstellung für diese beiden Therapiemodalitäten abgeleitet werden.
The Mediterranean area reveals a strong vulnerability to future climate change due to a high exposure to projected impacts and a low capacity for adaptation highlighting the need for robust regional or local climate change projections, especially for extreme events strongly affecting the Mediterranean environment. The prevailing study investigates two major topics of the Mediterranean climate variability: the analysis of dynamical downscaling of present-day and future temperature and precipitation means and extremes from global to regional scale and the comprehensive investigation of temperature and rainfall extremes including the estimation of uncertainties and the comparison of different statistical methods for precipitation extremes. For these investigations, several observational datasets of CRU, E-OBS and original stations are used as well as ensemble simulations of the regional climate model REMO driven by the coupled global general circulation model ECHAM5/MPI-OM and applying future greenhouse gas (GHG) emission and land degradation scenarios.
Rezidive eines Nebennierenkarzinoms sind trotz vermeintlich kompletter Tumorresektion im Verlauf der Tumorerkrankung häufig. In der Literatur ist der Stellenwert einer Rezidivoperation bis dato jedoch nicht gut untersucht. Daher wurde in dieser retrospektiven Studie aus den Daten des Deutschen Nebennierenkarzinom-Registers der Einfluss der unterschiedlichen Behandlungen eines Rezidivs auf den weiteren Erkrankungsverlauf bei 154 Patienten untersucht, die nach makroskopisch kompletter Resektion des Primarius im Verlauf ein Rezidiv entwickelt hatten. Das progressionsfreie Überleben und das Gesamtüberleben nach dem Rezidiv wurden mittels Kaplan-Meier-Methode ermittelt. Prognosefaktoren wurden mit Hilfe von der Cox-Regressionsanalyse berechnet. Insgesamt wurden 101 Patienten am Rezidiv operiert und 99 Patienten haben (teils zusätzlich) eine medikamentöse Therapie erhalten. Im Laufe der Nachsorge kam es bei 144 (94%) Patienten zu einem Rezidiv bzw. Tumorprogress (im Median nach einer Zeitspanne von 6 Monaten (1-144 Monate)). In der multivariaten Cox-Regressionsanalyse wurden als Prognosefaktoren Alter, Zeitspanne bis zum Rezidiv, Lokalisation, Anzahl der Läsionen, der Resektionsstatus bei der Rezidivoperation und zusätzliche Therapien einbezogen. Hierbei zeigte sich, dass nur zwei Faktoren unabhängig von den anderen einen signifikanten Einfluss auf die Prognose hatten: die Zeitspanne bis zum ersten Rezidiv und der Resektionsstatus der Rezidivoperation. So hatten Patienten, deren Rezidiv mehr als 12 Monate nach der Erstoperation auftrat ein deutlich niedrigeres adjustiertes Risiko für ein erneutes Rezidiv bzw. Progress als Patienten mit einem früheren Rezidiv (HR 0,56 (95% CI 0,39-0,79); p<0.001). Ähnlich war das Rezidivrisiko bei den Patienten, bei denen eine komplette Resektion erzielt werden konnte deutlich geringer als bei den nicht operierten nur medikamentös behandelten Patienten (HR 0,40 (95% CI 0,17-0,92); p=0,031). Bezüglich des Überlebens nach dem Rezidiv war die Risikoreduktion dieser zwei Prognosefaktoren noch deutlicher: War die Zeitspanne bis zum ersten Rezidiv über 12 Monate, lag die Hazard Ratio für den Nebennierenkarzinom-bedingten Tod bei 0,31 (95% CI 0,20-0,47; p<0,001) und bei der R0-Resektion bei 0,33 (95%CI 0,11-0,96; p=0,042), so dass hier jeweils das Risiko, am NN-Ca zu versterben, um ca. 70% reduziert war. In der homogeneren Subgruppenanalyse aller potentiell resektablen Patienten (n=68) zeigte sich in der multivariaten Auswertung ein ähnliches Ergebnis. Eine RX/R1-Resektion wies im Vergleich zur R0-Resektion ein 2-fach und eine R2-Resektion ein 3-fach höheres Risiko eines erneuten Rezidivs auf. Eine R2-Resektion erhöhte das Sterberisiko durch das Tumorleiden um das 2,8-fache. Die mit Abstand beste Prognose hatten die Patienten, die ihr erstes Rezidiv später als 12 Monate nach der Erstoperation entwickelten und dann komplett reseziert (R0-Resektion) werden konnten. Diese 22 Patienten hatten ein medianes progressionsfreies Überleben von 24 Monate (3-220 Monate) und ein medianes Gesamtüberleben von 58 Monaten (18-220 Monaten). 5 Patienten davon waren zum Zeitpunkt der aktuellen Analyse sogar noch rezidivfrei. Schlussfolgernd lässt sich sagen, dass in der vorliegenden Arbeit die beiden aussagekräftigsten Prädiktoren für die Prognose nach Rezidiv die Zeitspanne bis zum Rezidiv und die komplette Resektabilität sind. Unsere Daten legen nahe, dass Patienten mit Spätrezidiv eine Rezidiv-Operation erhalten sollten, wenn präoperativ eine vollständige Resektion möglich erscheint. Wenn sich ein Frührezidiv (<12 Monate) entwickelt oder eine in-sano-Resektion präoperativ nicht möglich erscheint, profitieren diese Patienten von einer Rezidiv-Operation wahrscheinlich eher nicht.
This dissertation focuses on selected novels written by contemporary indigenous authors from Aotearoa/New Zealand and examines the fictional imagination of the human body as a medium of cultural identity and memory. The novels discussed are Keri Hulme’s »The Bone People« (1984), »Nights in the Gardens of Spain« (1995) and »The Uncle’s Story« (2000) by Witi Ihimaera as well as James George’s »Hummingbird« (2003). In order to further decolonisation processes and to come to terms with the colonial past and the complexity of present realities, the fictional works position the human body as an active entity in the negotiation of specific cultural epistemologies. This project explores the narrative translation of corporeality that is used to locate alternative concepts of identity and cultural memory. Taking into account indigenous perspectives, this thesis makes use of the current theoretical approaches presented by pragmatism and affect theory in order to analyse the investment of the novels in feeling and the reciprocal relationship between text and corporeality depicted by the narratives. On the one hand, the novels aim to undermine oppressive and marginalising categories by placing particular emphasis on »sensuous gaps« in the text. On the other hand, the narratives intend to construct alternative identities and evoke specific aspects of indigenous histories and knowledge by imagining the human body in terms of »sensuous inscription«. The novels portray individuals who act from a place in-between different cultures, and articulate a desire to dissolve polarities and emphasise individual and cultural transformation as a formative element in the creation of complex identities and new perspectives.
Acute graft-versus-host disease (aGvHD) is an immune syndrome associated with allogeneic hematopoietic cell transplantation (allo-HCT) that is mediated by alloreactive donor T cells attacking the gastrointestinal tract, liver, and skin of the host. Early diagnosis remains problematic and to date mainly relies on clinical symptoms and histopathology. Previously, different groups demonstrated that in order to cause aGvHD, alloreactive T cells require the expression of appropriate homing receptors to efficiently migrate from their priming sites to their target tissues. Therefore, the development of a predictive test based on the homing receptor expression profile of peripheral blood T cells seems attractive to identify patients at risk before the onset of aGvHD. The aim of this study was to analyze migrating alloreactive donor T cell kinetics in the peripheral blood early after allo-HCT in a murine model across minor histocompatibility antigens (miHAg) followed by a precise characterization of the homing receptor expression profile of migrating donor lymphocytes in order to identify suitable predictive markers. Combining daily bioluminescence imaging (BLI) and flow cytometry (FC) allowed defining two weeks of massive alloreactive donor T cell migration before clinical aGvHD symptoms became apparent. Peripheral blood donor T lymphocytes highly up-regulated the homing markers α4β7 integrin, and P- and E-selectin-ligand at peak time points of cell migration. The combination with the activation markers CD25 and CD69 and low expression levels of L-selectin allowed alloreactive donor T cell definition. Based on this migration phase we postulated a potential diagnostic window to precisely identify alloreactive donor T cells upon their homing receptor expression profile. Consequently, targeted pre-emptive treatment with rapamycin starting at the earliest detection time point of alloreactive donor T cells in the peripheral blood (day+6) significantly prolonged survival of treated mice. Based on this data, we propose a potential diagnostic window for alloreactive cell detection based on their homing receptor expression profile for a timely and effective therapeutic intervention before the clinical manifestation of aGvHD.
Attention-deficit/hyperactivity disorder (ADHD) is the most frequent psychiatric disorder in children and adolescents and is often treated with methylphenidate (MPH), resulting in MPH exposure in more than 1% of all children in many countries. A 2005 report on cytogenetic effects in peripheral lymphocytes from 12 ADHD children treated for 3 months with MPH raised questions about its genetic toxicity and potential carcinogenicity. A healthy control group (23 individuals), a chronically MPH-treated (>12 months) group (21 patients), and a drug naïve group of ADHD-affected children (26 patients), which was analyzed again after 3 months (17 patients) and 6 months (11 patients), provided samples for analysis of micronucleated lymphocytes. No significant alteration in genomic damage as seen as micronucleus frequency in peripheral lypmphocytes was detected after MPH treatment. No indication for genomic damage induced by MPH was obtained in this study. Ongoing studies in the USA, as well as continuation of recently published epidemiological cancer incidence analysis should provide additional reassurance for MPH-treated ADHD patients.
Trypanosome Motion Represents an Adaptation to the Crowded Environment ofthe Vertebrate Bloodstream

Blood is a remarkable habitat: it is highly viscous, contains a dense packaging of cells and perpetually flows at velocities varying over three orders of magnitude. Only few pathogens endure the harsh physical conditions within the vertebrate bloodstream and prosper despite being constantly attacked by host antibodies. African trypanosomes are strictly extracellular blood parasites, which evade the immune response through a system of antigenic variation and incessant motility. How the flagellates actually swim in blood remains to be elucidated. Here, we show that the mode and dynamics of trypanosome locomotion are a trait of life within a crowded environment. Using high-speed fluorescence microscopy and ordered micro-pillar arrays we show that the parasites mode of motility is adapted to the density of cells in blood. Trypanosomes are pulled forward by the planar beat of the single flagellum. Hydrodynamic flow across the asymmetrically shaped cell body translates into its rotational movement. Importantly, the presence of particles with the shape, size and spacing of blood cells is required and sufficient for trypanosomes to reach maximum forward velocity. If the density of obstacles, however, is further increased to resemble collagen networks or tissue spaces, the parasites reverse their flagellar beat and consequently swim backwards, in this way avoiding getting trapped. In the absence of obstacles, this flagellar beat reversal occurs randomly resulting in irregular waveforms and apparent cell tumbling. Thus, the swimming behavior of trypanosomes is a surprising example of micro-adaptation to life at low Reynolds numbers. For a precise physical interpretation, we compare our high-resolution microscopic data to results from a simulation technique that combines the method of multi-particle collision dynamics with a triangulated surface model. The simulation produces a rotating cell body and a helical swimming path, providing a functioning simulation method for a microorganism with a complex swimming strategy
Ziel dieser retrospektiven Arbeit war es, die Daten von Patienten, die stereotaktisch aufgrund eines Lungentumors (NSCLC, Lungenmetastasen und Rezidiven) bestrahlt wurden, hinsichtlich des Therapieerfolges auszuwerten. Hierfür wurden die Unterlagen von 148 Patienten der Klinik und Poliklinik für Strahlentherapie der Universität Würzburg bezüglich der lokalen und systemischen Kontrolle, des Überlebens und der strahlenbedingten Nebenwirkungen untersucht. Für die Analyse wurden die Patienten in zwei Gruppen unterteilt. Die erste Gruppe bestand aus 40 Patienten mit 41 NSCLC im frühen Stadium (Stadium I und T3N0M0). In der zweiten Gruppe wurden 108 Patienten mit 25 NSCLC im fortgeschrittenen (Stadium III) und metastasierten Stadium sowie Patienten mit 111 Lungenmetastasen und zehn Rezidiven zusammengefasst. Die Bestrahlung erfolgte je nach Lage und Größe des Tumors mit Bestrahlungsdosen zwischen 6 und 26 Gy in einer bis acht Fraktionen, wobei die biologisch effektive Dosis appliziert auf das PTV zwischen 24 und 94 Gy lag. Die mediane Nachsorgedauer betrug 14 Monate. Bei der Betrachtung des lokalen Tumoransprechens zeigten sich in beiden Gruppen gute Ergebnisse mit lokalen Kontrollraten nach drei Jahren von jeweils 84%. In der statistischen Analyse wurde deutlich, dass eine lokale Tumorkontrolle sehr stark mit der Höhe der verabreichten Bestrahlungsdosis korreliert. So wurden mit BED > 80 Gy signifikant bessere Ergebnisse erzielt als mit BED < 80 Gy. Dementsprechend konnte auch mit den hochdosierten Bestrahlungsschemata (1 x 26 Gy auf 80% und 3 x 12,5 Gy auf 65%) eine bessere Kontrolle erreicht werden. In der ersten Gruppe der NSCLC im frühen Stadium waren die CTV und PTV der Tumoren, die lokal kontrolliert blieben signifikant kleiner, als die CTV und PTV der Tumoren, die lokal rezidivierten. Die systemische Kontrollrate fiel in der ersten Gruppe besser aus als in der zweiten Gruppe. Nach drei Jahren lagen die Werte entsprechend bei 36% und 16%. In der Gruppe der NSCLC im frühen Stadium erwies sich die Größe des bestrahlten Volumens als Faktor, der die systemische Kontrolle beeinflusst, wobei systemisch kontrollierte Patienten ein signifikant kleineres CTV und PTV hatten. Die Überlebensraten der Patienten lagen in der Gruppe 1 und 2 nach drei Jahren bei 32% und 17%. Sowohl der Leistungszustand vor Behandlungsbeginn als auch die Größe des bestrahlten Volumens beeinflussten das Überleben der Patienten. In der Gruppe 2 zeigte sich außerdem, dass ein hochdosiertes Bestrahlungsschema vorteilhaft für die Überlebensdauer eines Patienten ist. Da 46% der Todesursachen in der Gruppe 1 auf Begleiterkrankungen zurückzuführen waren, fiel folglich auch das tumorspezifische Überleben mit 51% nach drei Jahren deutlich besser aus als das Gesamtüberleben. Die Faktoren, die ein tumorspezifisches Überleben begünstigten, waren in Gruppe 1 das Alter und die Größe des bestrahlten Volumens. In der Gruppe 2 wirkte sich noch zusätzlich die Höhe der Bestrahlungsdosis signifikant auf das CSS aus. Das krankheitsfreie Überleben in der Gruppe 1 lag nach drei Jahren bei 23% und unterschied sich signifikant zwischen den einzelnen Tumorhistologien, wobei Patienten mit einem Adeno-CA am längsten krankheitsfrei überlebten. Außerdem hatten Patienten mit einem krankheitsfreien Überleben im Median kleinere CTV und PTV als Patienten, die während der Nachsorge verstarben oder nicht kontrolliert blieben. Die Gruppe 2 erreichte nach drei Jahren eine DFS-Rate von 8%. Die Rate an strahleninduzierten Nebenwirkungen fiel insgesamt gering aus. Schwere Nebenwirkungen ≥ Grad 3 traten in der Gesamtgruppe zu einem Anteil von 1,4% auf. Es zeigten sich jeweils eine Grad 3 und eine Grad 5 Nebenwirkung in der Gruppe 2 nach mehr als sechs Monaten nach Bestrahlung. Leichte akute und späte Nebenwirkungen ≤ Grad 2 konnten bei 42,5% der Patienten in der Gruppe 1 und bei 39,7% der Patienten der Gruppe 2 beobachtet werden. Weder die Höhe der BED noch die Größe des bestrahlten Volumens konnten mit dem Auftreten einer Nebenwirkung und deren Schweregrad in Verbindung gebracht werden. Insgesamt lässt sich festhalten, dass die stereotaktische Bestrahlung eine effektive und sichere Therapiemethode zur Bestrahlung von Lungenherden darstellt. Wird die stereotaktische Bestrahlung in kurativer Intention eingesetzt, sollte darauf geachtet werden, dass eine ausreichend hohe Bestrahlungsdosis Anwendung findet, um eine lokale Tumorkontrolle zu erzielen.
Background: Melatonin (MLT) has many health implications, therefore it is of valuable importance to develop specific analytical methods for determination of MLT in the presence of its main contaminant, N-{2-[1-({3-[2-(acetylamino)ethyl]-5-methoxy-1H-indol-2-yl}methyl)-5-methoxy-1H-indol-3-yl]ethyl}acetamide (10). For development of these analytical methods, compound 10 had to be prepared in an adequate amount. Results: Compound 10 was synthesized in six steps starting from 5-methoxyindole-2-carboxylic acid (1). Analytical performance of the proposed spectrofluorimetric methods was statistically validated with respect to linearity, accuracy, precision and specificity. The proposed methods were successfully applied for the assay of MLT in laboratory prepared mixtures containing up to 60 % of compound 10 and in commercial MLT tablets with recoveries not less than 99.00 %. No interference was observed from common pharmaceutical additives and the results were favorably compared with those obtained by a reference method. Conclusions: This work describes simple, sensitive, and reliable second derivative spectrofluorimetric method in addition to two multivariate calibration methods, principal component regression (PCR) and partial least square (PLS), for the determination of MLT in the presence of compound 10.
Background: Oncolytic viruses, including vaccinia virus (VACV), are a promising alternative to classical mono-cancer treatment methods such as surgery, chemo- or radiotherapy. However, combined therapeutic modalities may be more effective than mono-therapies. In this study, we enhanced the effectiveness of oncolytic virotherapy by matrix metalloproteinase (MMP-9)-mediated degradation of proteins of the tumoral extracellular matrix (ECM), leading to increased viral distribution within the tumors. Methods: For this study, the oncolytic vaccinia virus GLV-1h255, containing the mmp-9 gene, was constructed and used to treat PC-3 tumor-bearing mice, achieving an intra-tumoral over-expression of MMP-9. The intra-tumoral MMP-9 content was quantified by immunohistochemistry in tumor sections. Therapeutic efficacy of GLV-1h255 was evaluated by monitoring tumor growth kinetics and intra-tumoral virus titers. Microenvironmental changes mediated by the intra-tumoral MMP-9 over-expression were investigated by microscopic quantification of the collagen IV content, the blood vessel density (BVD) and the analysis of lymph node metastasis formation. Results: GLV-1h255-treatment of PC-3 tumors led to a significant over-expression of intra-tumoral MMP-9, accompanied by a marked decrease in collagen IV content in infected tumor areas, when compared to GLV-1h68-infected tumor areas. This led to considerably elevated virus titers in GLV-1h255 infected tumors, and to enhanced tumor regression. The analysis of the BVD, as well as the lumbar and renal lymph node volumes, revealed lower BVD and significantly smaller lymph nodes in both GLV-1h68- and GLV-1h255- injected mice compared to those injected with PBS, indicating that MMP-9 over-expression does not alter the metastasis-reducing effect of oncolytic VACV. Conclusions: Taken together, these results indicate that a GLV-1h255-mediated intra-tumoral over-expression of MMP-9 leads to a degradation of collagen IV, facilitating intra-tumoral viral dissemination, and resulting in accelerated tumor regression. We propose that approaches which enhance the oncolytic effect by increasing the intra-tumoral viral load, may be an effective way to improve therapeutic outcome.
EINLEITUNG: Neben der potentiellen Heilung können systematische Lymphadenektomien der Axilla und Leiste erhebliche Komplikationen, wie die Ausbildung von Lymphozelen und Lymphfisteln, nach sich ziehen. METHODEN: Es wurden alle Patienten erfasst, bei denen im Zeitraum zwischen 3/2002 und 12/2008 in der Abteilung der Allgemein-, Viszeral-, Gefäß- und Kinderchirurgie der Bayerischen Julius-Maximilians-Universität Würzburg eine systematische Lymphknotendissektion der Axilla oder Leiste erhielten. Nach Sichtung der OP-Bücher wurden die jeweiligen Verläufe anhand der Aktenlage analysiert. ERGEBNISSE: 146 Patienten wurden lymphadenektomiert, davon 69 axillär und 77 inguinal. Mit 93,1 % stellte die Hauptindikation für diese Operationen das maligne Melanom der Haut dar. Während die 30-Tage-Mortalität 0 % betrug, entwickelten 83 von 146 Patienten (56,8 %) eine Lymphfistel, 58 Patienten (39,7 %) eine Lymphozele. Die Entstehungsrate von Lymphfisteln und/oder Lymphozelen war nach inguinaler Lymphadenektomie mit 80,5 % signifikant höher als nach axillärer mit 62,3%. In der weiterführenden Statistik konnte festgestellt werden, dass sich mit Hilfe von ROC-Kurven Patienten mit und ohne Lymphfistel über die postoperative Drainagemenge trennen lassen. Die Rezidivraten für die Rotlicht-, Röntgenreizbestrahlung, sowie die operative Revision lagen bei 76,4 %, 22,4 % und 16,7 %. SCHLUSSFOLGERUNG: Trotz der niedrigen Mortalität waren Lymphadenektomien der Axilla und Leiste mit erheblicher Morbidität behaftet. Die Inzidenz lymphatischer Komplikationen war auch im Literaturvergleich hoch. Inguinale Dissektionen zogen statistisch signifikant mehr Lymphfisteln und Lymphozelen nach sich. ROC-Kurven können für einen frühzeitigen Therapiebeginn sowie Drainagezug herangezogen werden. Die Therapie der Lymphfisteln/Lymphozelen ist bislang nicht standardisiert. Eine alleinige Rotlichttherapie war zur Behandlung nicht erfolgreich, für die Röntgenreizbestrahlung und operative Revision konnten jedoch gute Behandlungsergebnisse erzielt werden.
Gegenstand dieser Arbeit ist die Erstellung eines sogenannten "Targeting Vektors" zur gezielten Ausschaltung des Gens für Plasmakallikrein in der Maus, als Vorbereitung zur Schaffung einer Plasmakallikrein-defizienten Mauslinie. Plasmakallikrein ist eine im Blut zirkulierende Serinprotease, die Funktionen in Hämostase, Thrombusbildung und Fibrinolyse hat sowie sowohl direkt als auch indirekt mittels Bradykinin an Entzündungsvorgängen beteiligt ist. Zwei 5836 und 3834 bp lange Abschnitte aus dem murinen Plasmakallikrein-Gen wurden durch PCR isoliert und in ein Plasmid kloniert, das neben Resistenzgenen gegen Ampicillin und Neomycin auch das β-Galaktosidase-Gen zum Nachweis einer erfolgreichen Transfektion enthält. Der so entstandene "Targeting Vektor" hat eine Gesamtgröße von 18072 bp, die Basensequenz wurde durch Sequenzierung verifiziert. Der Vektor soll im Plasmakallikrein-Gen einen Teil der Exons 2 und 3 und damit einen Großteil des Signalpeptids und der ersten Proteindomäne funktionsunfähig machen. An den mit dieser Methode erstellten Knockout-Mäusen können die Funktionen von Plasmakallikrein genauer untersucht werden.
Background: Septic acute liver and intestinal failure is associated with a high mortality. We therefore investigated the influence of volume resuscitation with different crystalloid or colloid solutions on liver and intestine injury and microcirculation in septic rodents. Methods: Sepsis was induced by cecal ligation and puncture (CLP) in 77 male rats. Animals were treated with different crystalloids (NaCl 0.9% (NaCl), Ringer’s acetate (RA)) or colloids (Gelafundin 4% (Gel), 6% HES 130/0.4 (HES)). After 24 h animals were re-anesthetized and intestinal (n = 6/group) and liver microcirculation (n = 6/group) were obtained using intravital microscopy, as well as macrohemodynamic parameters were measured. Blood assays and organs were harvested to determine organ function and injury. Results: HES improved liver microcirculation, cardiac index and DO2-I, but significantly increased IL-1β, IL-6 and TNF-α levels and resulted in a mortality rate of 33%. Gel infused animals revealed significant reduction of liver and intestine microcirculation with severe side effects on coagulation (significantly increased PTT and INR, decreased haemoglobin and platelet count). Furthermore Gel showed severe hypoglycemia, acidosis and significantly increased ALT and IL-6 with a lethality of 29%. RA exhibited no derangements in liver microcirculation when compared to sham and HES. RA showed no intestinal microcirculation disturbance compared to sham, but significantly improved the number of intestinal capillaries with flow compared to HES. All RA treated animals survided and showed no severe side effects on coagulation, liver, macrohemodynamic or metabolic state. Conclusions: Gelatine 4% revealed devastated hepatic and intestinal microcirculation and severe side effects in CLP induced septic rats, whereas the balanced crystalloid solution showed stabilization of macro- and microhemodynamics with improved survival. HES improved liver microcirculation, but exhibited significantly increased pro-inflammatory cytokine levels. Crystalloid infusion revealed best results in mortality and microcirculation, when compared with colloid infusion.
Es handelt sich um eine experimentelle Arbeit zur Untersuchung der Auswirkungen verschiedener Volumensubstitutionslösungen auf die Integrität der Leber in der CLP-induzierten Sepsis der Ratte. 40 Ratten wurden in 5 Grp. eingeteilt, anästhesiert und median laparotomiert. Während das Coecum der Sham-Tiere im ursprünglichen Zustand verblieb, erhielten alle anderen Tiere eine CLP. Die Tiere wurden im Anschluss entpsprechend ihrer Gruppe entweder nur mit dem Grundbedarf an NaCl (Sham) oder mit dem Grundbedarf an NaCl und dem jeweiligen Substitutionsmittel NaCl , SteroIso, Gelafundin, 6%HES 130/0,4 infundiert. Der Versuch lief über 24 h. Danach wurde die Tiere reanästhesiert, laparotomiert und eine in-vivo-Mikroskopie der Leber durchgeführt. Im Anschluss wurden sowohl hämodynamische Werte, Serumparameter und Zytokinwerte als auch histopathologische Daten ermittelt.
Diese Arbeit beschäftigt sich mit der Herstellung von positionierten In(Ga)As-Quantenpunkten zur Integration in Halbleiter-Mikroresonatoren. Dazu wurden systematisch die optischen Eigenschaften - insbesondere die Linienbreite und die Feinstrukturaufspaltung der Emission einzelner Quantenpunkte - optimiert. Diese Optimierung erfolgt im Hinblick auf die Verwendung der Quantenpunkte in Lichtquellen zur Realisierung einer Datenübertragung, die durch Quantenkryptographie abhörsicher verschlüsselt wird. Ein gekoppeltes Halbleitersystem aus einem Mikroresonator und einem Quantenpunkt kann zur Herstellung von Einzelphotonenquellen oder Quellen verschränkter Photonen verwendet werden. In dieser Arbeit konnten positionierte Quantenpunkte skalierbar in Halbleiter-Mikroresonatoren integriert werden. In(Ga)As-Quantenpunkte auf GaAs sind ein häufig untersuchtes System und können heutzutage mit hoher Kristallqualität durch Molekularstrahlepitaxie hergestellt werden. Um die Emission der Quantenpunkte gerichtet erfolgen zu lassen und die Auskoppeleffizienz der Bauteile zu erhöhen, wurden Mikrosäulenresonatoren oder photonische Kristallresonatoren eingesetzt. Die Integration in diese Resonatoren erfolgt durch Ausrichtung an Referenzstrukturen, wodurch dieses Verfahren skalierbar. Die Strukturierung der Substrate für die Herstellung von positionierten Quantenpunkten wurde durch optische Lithographie und Elektronenstrahllithographie in Kombination mit unterschiedlichen Ätztechniken erreicht. Für den praktischen Einsatz solcher Strukturen wurde ein Kontaktierungsschema für den elektrischen Betrieb entwickelt. Zur Verbesserung der optischen Eigenschaften der positionierten Quantenpunkte wurde ein Wachstumsschema verwendet, das aus einer optisch nicht aktiven In(Ga)As-Schicht und einer optisch aktiven Quantenpunktschicht besteht. Für die Integration einzelner Quantenpunkte in Halbleiter-Mikroresonatoren wurden positionierte Quantenpunkte auf einem quadratischen Gitter mit einer Periode von 200 nm bis zu 10 mum realisiert. Eine wichtige Kenngröße der Emission einzelner Quantenpunkte ist deren Linienbreite. Bei positionierten Quantenpunkten ist diese häufig aufgrund spektraler Diffusion größer als bei selbstorganisierten Quantenpunkten. Im Verlauf dieser Arbeit wurden unterschiedliche Ansätze und Strategien zur Überwindung dieses Effekts verfolgt. Dabei konnte ein minimaler Wert von 25 mueV für die Linienbreite eines positionierten Quantenpunktes auf einem quadratischen Gitter mit einer Periode von 2 μm erzielt werden. Die statistische Auswertung vieler Quantenpunktlinien ergab eine mittlere Linienbreite von 133 mueV. Die beiden Ergebnisse zeugen davon, dass diese Quantenpunkte eine hohe optische Qualität besitzen. Die FSS der Emission eines Quantenpunktes sollte für die direkte Erzeugung polarisationsverschränkter Photonen möglichst klein sein. Deswegen wurden unterschiedliche Ansätze diskutiert, um die FSS einer möglichst großen Zahl von Quantenpunkten systematisch zu reduzieren. Die FSS der Emission von positionierten In(Ga)As-Quantenpunkten auf (100)-orientiertem Galliumarsenid konnte auf einen minimalen Wert von 9.8 mueV optimiert werden. Ein anderes Konzept zur Herstellung positionierter Quantenpunkte stellt das Wachstum von InAs in geätzten, invertierten Pyramiden in (111)-GaAs dar In (111)- und (211)-In(Ga)As sollte aufgrund der speziellen Symmetrie des Kristalls bzw. der piezoelektrischen Felder die FSS verschwinden. Mit Hilfe von Quantenpunkten auf solchen Hochindex-Substraten konnten FSS von weniger als 5 mueV gemessen werden. Bis zu einem gewissen Grad kann die Emission einzelner Quantenpunkte durch laterale elektrische Felder beeinflusst werden. Besonders die beobachtete Reduzierung der FSS positionierter In(Ga)As-Quantenpunkte auf (100)-orientiertem GaAs von ca. 25 mueV auf 15 mueV durch ein laterales, elektrisches Feld ist viel versprechend für den künftigen Einsatz solcher Quantenpunkte in Quellen für verschränkte Photonen. Durch die Messung der Korrelationsfunktion wurde die zeitliche Korrelation der Emission von Exziton und Biexziton nachgewiesen und das Grundprinzip zum Nachweis eines polarisationsverschränkten Zustandes gezeigt. In Zusammenarbeit mit der Universität Tokyo wurde ein Konzept entwickelt, mit dem künftig Einzelquantenpunktlaser skalierbar durch Kopplung positionierter Quantenpunkte und photonischer Kristallkavitäten hergestellt werden können. Weiterhin konnte mit Hilfe eines elektrisch kontaktierten Mikrosäulenresonators bei spektraler Resonanz von Quantenpunktemission und Kavitätsmode eine Steigerung der spontanen Emission nachgewiesen werden. Dieses System ließ sich bei geeigneten Anregungsbedingungen auch als Einzelphotonenquelle betreiben, was durch den experimentell bestimmten Wert der Autokorrelationsfunktion für verschwindende Zeitdifferenzen nachgewiesen wurde.
Fanconi anemia (FA) is a rare genomic instability syndrome. Disease-causing are biallelic mutations in any one of at least 15 genes encoding members of the FA/BRCA pathway of DNA-interstrand crosslink repair. Patients are diagnosed based upon phenotypical manifestationsand the diagnosis of FA is confirmed by the hypersensitivity of cells to DNA interstrand crosslinking agents. Customary molecular diagnostics has become increasingly cumbersome, time-consuming and expensive the more FA genes have been identified. We performed Whole Exome Sequencing (WES) in four FA patients in order to investigate the potential of this method for FA genotyping. In search of an optimal WES methodology we explored different enrichment and sequencing techniques. In each case we were able to identify the pathogenic mutations so that WES provided both, complementation group assignment and mutation detection in a single approach. The mutations included homozygous and heterozygous single base pair substitutions and a two-base-pair duplication in FANCJ, -D1, or - D2. Different WES strategies had no critical influence on the individual outcome. However, database errors and in particular pseudogenes impose obstacles that may prevent correct data perception and interpretation, and thus cause pitfalls. With these difficulties in mind, our results show that WES is a valuable tool for the molecular diagnosis of FA and a sufficiently safe technique, capable of engaging increasingly in competition with classical genetic approaches.
Background: Multiple myeloma (MM) is a B-cell malignancy, where malignant plasma cells clonally expand in the bone marrow of older people, causing significant morbidity and mortality. Typical clinical symptoms include increased serum calcium levels, renal insufficiency, anemia, and bone lesions. With standard therapies, MM remains incurable; therefore, the development of new drugs or immune cell-based therapies is desirable. To advance the goal of finding a more effective treatment for MM, we aimed to develop a reliable preclinical MM mouse model applying sensitive and reproducible methods for monitoring of tumor growth and metastasis in response to therapy. Material and Methods: A mouse model was created by intravenously injecting bone marrow-homing mouse myeloma cells (MOPC-315.BM) that expressed luciferase into BALB/c wild type mice. The luciferase in the myeloma cells allowed in vivo tracking before and after melphalan treatment with bioluminescence imaging (BLI). Homing of MOPC-315.BM luciferase+ myeloma cells to specific tissues was examined by flow cytometry. Idiotype-specific myeloma protein serum levels were measured by ELISA. In vivo measurements were validated with histopathology. Results: Strong bone marrow tropism and subsequent dissemination of MOPC-315.BM luciferase+ cells in vivo closely mimicked the human disease. In vivo BLI and later histopathological analysis revealed that 12 days of melphalan treatment slowed tumor progression and reduced MM dissemination compared to untreated controls. MOPC-315.BM luciferase+ cells expressed CXCR4 and high levels of CD44 and a4b1 in vitro which could explain the strong bone marrow tropism. The results showed that MOPC-315.BM cells dynamically regulated homing receptor expression and depended on interactions with surrounding cells. Conclusions: This study described a novel MM mouse model that facilitated convenient, reliable, and sensitive tracking of myeloma cells with whole body BLI in living animals. This model is highly suitable for monitoring the effects of different treatment regimens.
Retinitis pigmentosa (RP) ist eine vererbte Form der Erblindung, die durch eine progressive Degeneration von Photorezeptorzellen in der Retina verursacht wird. Neben „klassischen“ RP-Krankheitsgenen, die direkt oder indirekt mit dem Sehprozess und der Aufrechterhaltung der Photorezeptoren in Verbindung stehen, können auch Mutationen in Genen für konstitutive Spleißfaktoren zur Photorezeptordegeneration führen. RP kann daher als Paradebeispiel einer Erkrankung mit paradoxer Gewebespezifität angesehen werden: Defekte in essentiellen und ubiquitär exprimierten Genen führen zu einem Phänotyp, der nur wenige Zelltypen betrifft. Um Einblicke in diesen außergewöhnlichen Pathomechanismus zu erhalten, wurde im Rahmen der vorliegenden Arbeit ein Tiermodell für Spleißfaktor-vermittelte RP im Zebrafisch Danio rerio etabliert. Zunächst wurde gezeigt, dass eine RP verursachende Punktmutation des Spleißfaktors Prpf31 auch in dessen Zebrafisch-Homolog zu einem Verlust der physiologischen Aktivität führt. Als Modell für die Prpf31-Mangelsituation diente dann die durch ein Antisense-Morpholino induzierte partielle Reduktion der Prpf31-Expression in Zebrafischlarven. Konsistent mit einem RP-Phänotyp zeigte sich in diesen Larven eine starke Beeinträchtigung des Sehvermögens. Sie wurde – ebenfalls analog zu RP – durch defekte Photorezeptoren verursacht, die bei ansonsten normal entwickelter Retina eine deutlich veränderte Morphologie aufwiesen. Daraufhin konnten in einer genomweiten Transkriptomanalyse der Augen von Prpf31-defizienten Larven erstmals in vivo photorezeptorspezifische Gene identifiziert werden, deren Expression durch den Mangel an Prpf31 beeinträchtigt war. Im zweiten Teil der Arbeit wurde untersucht, ob es neben den bereits bekannten RP-Krankheitsgenen weitere Spleißfaktoren gibt, deren Defekt die Degeneration von Photorezeptoren auslösen kann. Dazu wurde in Zebrafischlarven ein Mangel an Prpf4 erzeugt, einem Spleißfaktor, der bislang nicht mit RP in Verbindung gebracht worden war. Der Phänotyp dieser Fische war nicht von dem des Prpf31 RP-Modells zu unterscheiden. Dies lieferte einen Hinweis darauf, dass auch Defekte in Prpf4 in der Lage sein könnten, RP auszulösen. Tatsächlich konnte durch genetisches Screening ein RP-Patient mit einer Punktmutation in Prpf4 identifiziert werden (Kollaboration mit Hanno Bolz, Universität Köln). Die biochemische Analyse dieser Mutation zeigte, dass sie zu einem Defekt der Integration von Prpf4 in spleißosomale Untereinheiten und zu dessen Funktionsverlust in vivo führt. Mit dem in dieser Arbeit etablierten Tiermodell konnte zum ersten Mal in vivo ein von Spleißfaktor-Mutationen verursachter Pathomechanismus von Retinitis pigmentosa nachvollzogen werden. Die vom Prpf31-Mangel betroffenen Photorezeptortranskripte stellen vielversprechende Kandidaten für die Vermittlung der Gewebespezifität dar und unterstützen die Hypothese, dass ihre ineffiziente Prozessierung den RP-Phänotyp auslöst. Die Entdeckung eines weiteren Spleißfaktors, dessen Defizienz ebenfalls zu defekten Photorezeptoren führt, zeigt, dass offenbar der Funktionsverlust des Spleißosoms generell in der Lage ist, die Degeneration dieser Zellen zu verursachen. Dies ist nicht zuletzt auch von klinischer Relevanz, da vermutet werden kann, dass sich unter den vielen bisher nicht identifizierten RP-Krankheitsgenen weitere Spleißfaktoren befinden.
Die heute in der Krebstherapie vorherrschenden konventionellen Therapiemethoden weisen Defizite bezüglich ihrer Wirksamkeit auf und rufen oftmals gravierende Nebenwirkungen hervor. Eine Alternative für die Behandlung von Tumoren ist der Einsatz onkolytischer Viren. Um einen erfolgreichen klinischen Einsatz onkolytischer Viren zu ermöglichen, ist eine Verstärkung von deren Wirksamkeit durch die Insertion therapeutischer Gene wünschenswert. Im Rahmen der vorliegenden Arbeit sollte der Abbau von Proteinen der extrazellulären Matrix durch die Insertion des Relaxin- oder Matrixmetalloproteinase 9-Gens (MMP-9) in das Vaccinia-Virus Genom erreicht und dadurch die Virusausbreitung im Tumorgewebe erleichtert werden. Hierfür wurden die rekombinanten Vaccinia-Viren GLV-1h169, codierend für das Hormon Relaxin und GLV-1h255, codierend für das Enzym MMP-9, eingesetzt. Es wurde analysiert, ob die Expression dieser Proteine zu einem Abbau von Matrixproteinen führt, dies die Virusausbreitung erleichtert und die Lyse infizierter Tumorzellen gegenüber dem parentalen Virus GLV-1h68 verstärkt. GLV-1h169 wurde in DU145-, PC3- und C33A-Tumor-tragende Mäuse injiziert und die Wirkung des viral-codierten Relaxins auf die extrazelluläre Matrix und die virale Ausbreitung im Tumorgewebe analysiert. In Zellkultur-Experimenten wurde ermittelt, dass die Insertion des Relaxin-Gens in das GLV-1h169-Genom das Replikationsverhalten in DU145-Zellen gegenüber dem des parentalen Virus GLV-1h68 nicht negativ beeinflusst. In DU145-, PC3- und C33A-Tumorschnitten konnte eine Expression von Relaxin in GLV-1h169-infizierten Bereichen nachgewiesen werden. Die Expression von Relaxin soll durch die Aktivierung des Relaxin-Signalweges zur Translation von MMP-9 führen. Das Enzym wird von infizierten Zellen sezerniert und spaltet Proteine der extrazellulären Matrix. Der Gehalt der MMP-9 Substrate Collagen IV und Laminin in GLV-1h169 behandelten DU145- und C33A-Tumoren wurde analysiert und mit jenem in GLV-1h68- und PBS- behandelten Tumoren verglichen. In Virus-behandelten DU145-Tumoren zeigte sich im Vergleich mit PBS-behandelten Tumoren ein signifikant verringerter Collagen IV- und Laminingehalt. Weiterhin war der Collagen IV-Gehalt in GLV-1h169 infizierten Tumoren signifikant niedriger als in GLV-1h68 infizierten. Dies führte jedoch nicht zu einer Erhöhung des Virustiters und nicht zu einer verbesserten Virusausbreitung. GLV-1h68- und GLV-1h169-infizierte Tumore zeigten gegenüber PBS-behandelten Tumoren eine starke Regression. Die GLV-1h169-vermittelte Relaxin-Expression führte jedoch nicht zu einer weiteren Verstärkung der Tumorregression. In Virus-behandelten C33A-Tumoren wurde eine signifikante Erhöhung des Collagen IV- und Laminingehalts gegenüber PBS-behandelten Tumoren nachgewiesen. Dies könnte durch eine Virus-induzierte Inflammationsreaktion hervorgerufen werden, die eine Fibroblasten-vermittelte Collagenablagerung nach sich zieht. Das MMP-9 Expressionsle-vel war in Virus-behandelten Tumoren gegenüber PBS-behandelten signifikant erhöht, jedoch bewirkte die GLV-1h169-vermittelte Expression von Relaxin keine zusätzliche MMP-9 Expression. In Tumorrandbereichen erfolgte eine Expression von Relaxin und MMP-9, im Tumorinneren jedoch nur eine Expression von Relaxin. Hingegen wurde eine Korrelation zwischen der MMP-9-Expression und der Präsenz MHC II-positiver Zellen beobachtet. Diese Zellen migrieren von außen in das Tumorgewebe und exprimieren dort MMP-9. Bei der Analyse der Virustiter und –ausbreitung im Tumorgewebe zeigten sich keine signifikanten Unterschiede zwischen GLV-1h68- und GLV-1h169-injizierten Tieren. Die Injektion von beiden onkolytischen Viren in C33A-Tumor-tragende Mäuse führte zu einer starken Tumorregression. Diese wurde jedoch nicht durch die GLV-1h169-vermittelte Relaxin-Expression beeinflusst. Da die Aktivierung des Relaxin-Signalweges zu einer Expression des vascular endothelial growth factors (VEGF) führen kann, welcher die Angiogenese stimuliert, wurde die Blutgefäßdichte in C33A-Tumoren ermittelt. Die Expression von Relaxin führte nicht zu einer erhöhten Blutgefäßdichte. Die Basalmembran von Blutgefäßen enthält Collagen IV, deshalb wurde untersucht, ob die Relaxin-Expression eine erhöhte Permeabilität der Gefäße bewirkt. In den Virus-behandelten Tumoren zeigte sich eine gegenüber PBS-behandelten Tumoren signifikant erhöhte Gefäß-Permeabilität, jedoch bewirkte die Expression von Relaxin keine weitere Erhöhung der Gefäß-Permeabilität...
Protection of healthy tissues from infection with systemically administered vaccinia virus strains

Oncolytic virotherapy using recombinant vaccinia virus strains is a promising approach for the treatment of cancer. To further improve the safety of oncolytic vaccinia viruses, the cellular microRNA machinery can be applied as the host’s own security mechanism to avoid unwanted viral replication in healthy tissues. MicroRNAs are a class of small single-stranded RNAs which due to their ability to mediate post-transcriptional gene-silencing, play a crucial role in almost every regulatory process in cellular metabolism. Different cancers display unique microRNA expression patterns, showing significant up- or downregulation of endogenously expressed microRNAs. Furthermore, the behavior of cancer cells can be altered by either adding microRNAs known to inhibit cancer cell spread and proliferation or suppressing cancer promoting microRNAs (oncomirs) making microRNAs promising targets for cancer gene therapy. The cell’s own RNAi machinery can also be utilized to control viral replication due to the virus dependence on the host cell replication machinery, a process controlled by microRNAs. GLV-1h68 is a replication-competent recombinant oncolytic vaccinia virus constructed and generated by Genelux Corp., San Diego, CA, USA which carries insertions of three reporter gene cassettes for detection and attenuation purposes and is currently being evaluated for cancer treatment in clinical trials. Though there are hardly any side effects found in GLV-1h68 mediated oncolytic therapy an increased tropism for replication exclusively in cancer cells is desirable. Therefore it was investigated whether or not further cancer cell specificity of a recombinant vaccinia virus strain could be obtained without compromising its oncolytic activity using microRNA interference. Let-7a is a well characterized microRNA known to be expressed in high levels in healthy tissues and strongly downregulated in most cancers. To control vaccinia virus replication rates, four copies of the mature human microRNA let-7a target sequence were cloned behind the stop codon in the 3’end of the vaccinia virus D4R gene, using a GLV-1h68 derivative, GLV-1h190, as parental strain yielding the new recombinant virus strain GLV-1h250. The D4R gene belongs to the group of early transcribed vaccinia genes and encodes an essential enzyme, uracil DNA glycosylase, which catalyzes the removal of uracil residues from double-stranded DNA. A defect in D4R prevents vaccinia virus from entering into the intermediate and late phase of replication, leading to an aborted virus replication. After expression of the microRNA target sequence from the vaccinia virus genome, the endogenously expressed microRNA-let-7a should recognize its target structure within the viral mRNA transcript, thereby binding and degrading the viral mRNA which should lead to a strong inhibition of the virus replication in healthy cells. GLV-1h250 replication rates in cancerous A549 lung adenocarcinoma cells, which show a strong down-regulation of microRNA let-7a, was comparable to the replication rates of its parental strain GLV-1h190 and the control strain GLV-1h68. In contrast, GLV-1h250 displayed a 10-fold decrease in viral replication in non-cancerous ERC cells when compared to GLV-1h190 and GLV-1h68. In A549 tumor bearing nude mice GLV-1h250 replicated exclusively in the tumorous tissue and resulted in efficient tumor regression without adverse effects leading to the conclusion that GLV-1h250 replicates preferentially in cancerous cells and tissues, which display low endogenous let-7a expression levels.
Plant communities in the European Alps are assumed to be highly affected by climate change since temperature rise in this region is above the global average. It is predicted that higher temperatures will lead to advanced snowmelt dates and that the number of extreme weather events will increase. The aims of this study were to determine the impacts of extreme climatic events on flower phenology and to assess whether those impacts differed between lower and higher altitudes. In 2010 an experiment simulating advanced and delayed snowmelt as well as drought event was conducted along an altitudinal transect ca. every 250m (600-2000 m a.s.l.) in the Berchtesgaden National Park, Germany. The study showed that flower phenology is strongly affected by altitude; however there were few effects of the manipulative treatments on flowering. The effects of advanced snowmelt were significantly greater at higher than at lower sites, but no significant difference was found between both altitudinal bands for the other treatments. The response of flower phenology to temperature declined through the season and the length of flowering duration was not significantly influenced by treatments. The stronger effect of advanced snowmelt at higher altitudes might be a response to differences in treatment intensity across the gradient. Consequently, shifts in the date of snowmelt due to global warming may affect species more at higher than at lower altitudes since changes may be more pronounced at higher altitudes. Our data indicate a rather low risk of drought events on flowering phenology in the Bavarian Alps.
This thesis deals with the chaotic dynamics of nonlinear networks consisting of semiconductor lasers which have time-delayed self-feedbacks or mutual couplings. These semiconductor lasers are simulated numerically by the Lang-Kobayashi equations. The central issue is how the chaoticity of the lasers, measured by the maximal Lyapunov exponent, changes when the delay time is changed. It is analysed how this change of chaoticity with increasing delay time depends on the reflectivity of the mirror for the self-feedback or the strength of the mutal coupling, respectively. The consequences of the different types of chaos for the effect of chaos synchronization of mutually coupled semiconductor lasers are deduced and discussed. At the beginning of this thesis, the master stability formalism for the stability analysis of nonlinear networks with delay is explained. After the description of the Lang-Kobayashi equations and their linearizations as a model for the numerical simulation of semiconductor lasers with time-delayed couplings, the artificial sub-Lyapunov exponent $\lambda_{0}$ is introduced. It is explained how the sign of the sub-Lyapunov exponent can be determined by experiments. The notions of "strong chaos" and "weak chaos" are introduced and distinguished by their different scaling properties of the maximal Lyapunov exponent with the delay time. The sign of the sub-Lyapunov exponent $\lambda_{0}$ is shown to determine the occurence of strong or weak chaos. The transition sequence "weak to strong chaos and back to weak chaos" upon monotonically increasing the coupling strength $\sigma$ of a single laser's self-feedback is shown for numerical calculations of the Lang-Kobayashi equations. At the transition between strong and weak chaos, the sub-Lyapunov exponent vanishes, $\lambda_{0}=0$, resulting in a special scaling behaviour of the maximal Lyapunov exponent with the delay time. Transitions between strong and weak chaos by changing $\sigma$ can also be found for the Rössler and Lorenz dynamics. The connection between the sub-Lyapunov exponent and the time-dependent eigenvalues of the Jacobian for the internal laser dynamics is analysed. Counterintuitively, the difference between strong and weak chaos is not directly visible from the trajectory although the difference of the trajectories induces the transitions between the two types of chaos. In addition, it is shown that a linear measure like the auto-correlation function cannot unambiguously reveal the difference between strong and weak chaos either. Although the auto-correlations after one delay time are significantly higher for weak chaos than for strong chaos, it is not possible to detect a qualitative difference. If two time-scale separated self-feedbacks are present, the shorter feedback has to be taken into account for the definition of a new sub-Lyapunov exponent $\lambda_{0,s}$, which in this case determines the occurence of strong or weak chaos. If the two self-feedbacks have comparable delay times, the sub-Lyapunov exponent $\lambda_{0}$ remains the criterion for strong or weak chaos. It is shown that the sub-Lyapunov exponent scales with the square root of the effective pump current $\sqrt{p-1}$, both in its magnitude and in the position of the critical coupling strengths. For networks with several distinct sub-Lyapunov exponents, it is shown that the maximal sub-Lyapunov exponent of the network determines whether the network's maximal Lyapunov exponent scales strongly or weakly with increasing delay time. As a consequence, complete synchronization of a network is excluded for arbitrary networks which contain at least one strongly chaotic laser. Furthermore, it is demonstrated that the sub-Lyapunov exponent of a driven laser depends on the number of the incoherently superimposed inputs from unsynchronized input lasers. For networks of delay-coupled lasers operating in weak chaos, the condition $|\gamma_{2}|<\mathrm{e}^{-\lambda_{\mathrm{m}}\,\tau}$ for stable chaos synchronization is deduced using the master stability formalism. Hence, synchronization of any network depends only on the properties of a single laser with self-feedback and the eigenvalue gap of the coupling matrix. The characteristics of the master stability function for the Lang-Kobayashi dynamics is described, and consequently, the master stability function is refined to allow for precise practical prediction of synchronization. The prediction of synchronization with the master stability function is demonstrated for bidirectional and unidirectional networks. Furthermore, the master stability function is extended for two distinct delay times. Finally, symmetries and resonances for certain values of the ratio of the delay times are shown for the master stability function of the Lang-Kobyashi equations.
Ab dem letzten Schwangerschaftsdrittel können Föten Geräusche, Stimmen und Gespräche hören (Vince et al. 1985, Querleu et al. 1988, Damstra-Wijmenga et al. 1991, Johansson et al. 1992, Hepper/Shahidullah 1994). Die pränatalen Hörerfahrungen werden in einem fötalen Gedächtnis gespeichert und bleiben so bis ins Neugeborenenalter bestehen (u.a. Hepper 1988, Hepper et al. 1993). Neben einer generellen pränatalen Prägung ist für Neugeborene eine muttersprachspezifische Prägung sowohl perzeptiver als auch produktiver Leistungen belegt, die sich in der Differenzierungsfähigkeit der Muttersprache von einer unbekannten Sprache (z.B. Mehler et al. 1986, Byers-Heinlein et al. 2010) sowie in einer deutlichen perzeptiven (z.B. Mehler et al. 1986) und produktiven Präferenz (Mampe et al. 2009) für ihre Muttersprache zeigt. In der vorliegenden Arbeit wurden 1744 Schreimelodien von insgesamt 60 gesunden Neugeborenen untersucht, deren Mütter während der Schwangerschaft entweder eine oder zwei Sprachen gesprochen hatten. 40 Neugeborene hatten monolingual französische Mütter. 20 Neugeborene hatten bilinguale Mütter (französisch und eine weitere Sprache). Neben dem Vergleich des Einflusses einer pränatal monolingualen und einer pränatal bilingualen Sprachumgebung auf die Schreimelodiekonturproduktion Neugeborener wurde untersucht, ob das quantitative Verhältnis mit dem beide Sprachen pränatal gehört wurden die Schreimelodiekonturproduktion der bilingualen Neugeborenen beeinflusst. Ein objektiver Vergleich geeigneter einfachbögiger Schreimelodiekonturen erfolgte mithilfe des EF-Modells. Das Modell lieferte für jede Melodiekontur genau einen Maximalwert (in Form des Formparameters α), der zwischen 0 und 1 variierte. Die von den bilingualen Neugeborenen produzierten Melodiekonturen unterschieden sich statistisch signifikant von denen der monolingualen Neugeborenen (T-Test, p=0,031). Die monolingual französischen Neugeborenen produzierten gehäuft steigende Melodiekonturen (α>0,5). Für die bilingualen Neugeborenen konnte keine Präferenz für die Produktion einer bestimmten Melodiekonturform festgestellt werden. Für die bilingualen Neugeborenen wurde zudem gezeigt, dass zwischen dem quantitativen Verhältnis der pränatal gehörten Umgebungssprachen und der Melodieproduktion im Neugeborenenalter kein statistisch signifikanter Zusammenhang (H-Test nach Kruskal-Wallis, p=0,273) besteht. Der hinreichende Unterschied der α-Verteilungen der mono- und bilingualen Neugeborenen belegt, dass pränatale auditive Erfahrungen bereits die Melodieproduktion Neugeborener prägen.
Multi-Wavelength Observations of the high-peaked BL Lacertae objects 1ES 1011+496 and 1ES 2344+514

BL Lacertae objects belong to the most luminous sources in the Universe. They represent a subclass of active galactic nuclei with a spectrum that is dominated by non-thermal emission, extending from radio wavelengths to tera electronvolt (TeV) energies. The emission is strongly variable on time scales of years down to minutes, and arises from relativistic jets pointing at small angles to the line of sight of the observer, which is the reason for naming them “blazars”. Blazars are the dominant extragalactic source class in the radio, microwave and gamma-ray regime, are prime candidates for the origin of the Cosmic Rays and excellent laboratories to study black hole and jet physics as well as relativistic effects. Despite more than 20 years of observational efforts, the physical mechanisms driving their emission are not yet fully understood. So far, studies of their broad-band continuum emission were mostly concentrated on bright, flaring states. However, for a better understanding of the central engine powering the jets, the bias from flux-limited observations of the past must be overcome and their long-term average continuum spectral energy distributions (SEDs) must be determined. This work presents the first simultaneous multi-wavelength campaigns from the radio to the TeV regime of two high-frequency peaked BL Lacertae objects known to emit at TeV energies. The first source, 1ES 1011+496, was observed between February and May 2008, the second one, 1ES 2344+514, between September 2008 and February 2009. The extensive observational campaigns were organised independently from an external trigger for the presence of a flaring state. Since the duty cycle of major flux outbursts is known to be rather low, the campaigns were expected to yield SEDs representative of the long-term average emission. Central for this thesis is the analysis of data obtained with the MAGIC Cherenkov telescope, measuring energy spectra and light curves from ~0.1 to ~10 TeV. For the remaining instruments, observation time was proposed and additional data was organised by collaboration with the instrument teams by the author of this work. Such data was obtained mostly in a fully reduced state. Individual light curves are investigated as well as combined in a search for inter-band correlations. The data of both sources reveal a notable lack of a correlation between the emission at radio and optical wavelengths, indicating that the radio and short-wavelength emission arise in different regions of the jet. Quasi-simultaneous SEDs of two different flux states are observationally determined and described by a one-zone as well as a self-consistent two-zone synchrotron self-Compton model. First approaches to model the SEDs by means of a Chi2 minimisation technique are briefly discussed. The SEDs and the resulting model parameters, characterising the physical conditions in the emission regions, are compared to archival data. Though the models can describe the data well, for 1ES 1011+496 the model parameters indicate that in addition to the synchrotron and inverse-Compton emission of relativistic electrons, emission due to accelerated protons seems to be required. The SEDs of 1ES 2344+514 reveal one of the lowest activity states ever detected from the source. Despite that, the model parameters are not indicative of a distinct quiescent state, which may be caused by the degeneracy of the different parameters in one-zone models. Moreover, indications accumulate that the radiation can not be attributed to a single emission region. The results disfavour some of the current blazar classification schemes and the so-called “blazar sequence”, emphasising the need for a more realistic explanation of the systematics of the blazar SEDs in terms of fundamental parameters.
Synergistic Effect of Caffeine and Glucocorticoids on Expression of Surfactant Protein B (SP-B) mRNA

Administration of glucocorticoids and caffeine is a common therapeutic intervention in the neonatal period, but possible interactions between these substances are still unclear. The present study investigated the effect of caffeine and different glucocorticoids on expression of surfactant protein (SP)-B, crucial for the physiological function of pulmonary surfactant. We measured expression levels of SP-B, various SP-B transcription factors including erythroblastic leukemia viral oncogene homolog 4 (ErbB4) and thyroid transcription factor-1 (TTF-1), as well as the glucocorticoid receptor (GR) after administering different doses of glucocorticoids, caffeine, cAMP, or the phosphodiesterase-4 inhibitor rolipram in the human airway epithelial cell line NCI-H441. Administration of dexamethasone (1 mM) or caffeine (5 mM) stimulated SP-B mRNA expression with a maximal of 38.8611.1-fold and 5.261.4-fold increase, respectively. Synergistic induction was achieved after coadministration of dexamethasone (1 mM) in combination with caffeine (10 mM) (206659.7-fold increase, p,0.0001) or cAMP (1 mM) (2136111-fold increase, p = 0.0108). SP-B mRNA was synergistically induced also by administration of caffeine with hydrocortisone (87.9639.0), prednisolone (154666.8), and betamethasone (12366.4). Rolipram also induced SP-B mRNA (64.9621.0-fold increase). We detected a higher expression of ErbB4 and GR mRNA (7.0- and 1.7-fold increase, respectively), whereas TTF-1, Jun B, c-Jun, SP1, SP3, and HNF-3a mRNA expression was predominantly unchanged. In accordance with mRNA data, mature SP-B was induced significantly by dexamethasone with caffeine (13.869.0-fold increase, p = 0.0134). We found a synergistic upregulation of SP-B mRNA expression induced by co-administration of various glucocorticoids and caffeine, achieved by accumulation of intracellular cAMP. This effect was mediated by a caffeinedependent phosphodiesterase inhibition and by upregulation of both ErbB4 and the GR. These results suggested that caffeine is able to induce the expression of SP-transcription factors and affects the signaling pathways of glucocorticoids, amplifying their effects. Co-administration of caffeine and corticosteroids may therefore be of benefit in surfactant homeostasis.
Natalizumab is a recombinant monoclonal antibody raised against integrin alpha-4 (CD49d). It is approved for the treatment of patients with multiple sclerosis (MS), a chronic inflammatory autoimmune disease of the CNS. While having shown high therapeutic efficacy, treatment by natalizumab has been linked to progressive multifocal leukoencephalopathy (PML) as a serious adverse effect. Furthermore, drug cessation sometimes induces rebound disease activity of unknown etiology. Here we investigated whether binding of this adhesion-blocking antibody to T lymphocytes could modulate their phenotype by direct induction of intracellular signaling events. Primary CD4+ T lymphocytes either from healthy donors and treated with natalizumab in vitro or from MS patients receiving their very first dose of natalizumab were analyzed. Natalizumab induced a mild upregulation of IL-2, IFN-c and IL-17 expression in activated primary human CD4+ T cells propagated ex vivo from healthy donors, consistent with a pro-inflammatory costimulatory effect on lymphokine expression. Along with this, natalizumab binding triggered rapid MAPK/ERK phosphorylation. Furthermore, it decreased CD49d surface expression on effector cells within a few hours. Sustained CD49d downregulation could be attributed to integrin internalization and degradation. Importantly, also CD4+ T cells from some MS patients receiving their very first dose of natalizumab produced more IL-2, IFN-c and IL-17 already 24 h after infusion. Together these data indicate that in addition to its adhesion-blocking mode of action natalizumab possesses mild direct signaling capacities, which can support a pro-inflammatory phenotype of peripheral blood T lymphocytes. This might explain why a rebound of disease activity or IRIS is observed in some MS patients after natalizumab cessation.
From November 2nd - 4th 2012, the 4th NEUROWIND e.V. meeting was held in Motzen, Brandenburg, Germany. Again more than 60 participants, predominantly at the doctoral student or postdoc level, gathered to share their latest findings in the fields of neurovascular research, neurodegeneration and neuroinflammation. Like in the previous years, the symposium provided an excellent platform for scientific exchange and the presentation of innovative projects in the stimulating surroundings of the Brandenburg outback. This year’s keynote lecture on the pathophysiological relevance of neuronal networks was given by Christian Gerloff, Head of the Department of Neurology at the University Clinic of Hamburg-Eppendorf. Another highlight of the meeting was the awarding of the NEUROWIND e.V. prize for young scientists working in the field of experimental neurology. The award is donated by the Merck Serono GmbH, Darmstadt, Germany and is endowed with 20.000 Euro. This year the jury decided unanimously to adjudge the award to Michael Gliem from the Department of Neurology at the University Clinic of Düsseldorf (group of Sebastian Jander), Germany, for his outstanding work on different macrophage subsets in the pathogenesis of ischemic stroke published in the Annals of Neurology in 2012.
Unter dem Einfluss von M-CSF und GM-CSF entwickeln sich CD14-positive periphere humane Blutmonozyten zu CD68-positiven M-CSF- bzw. GM-CSF-Makrophagen. M-CSF-Makrophagen lassen sich mit INFg und LPS zu klassisch aktivierten M1-Makrophagen, oder mit IL-4 und IL-10 zu alternativ aktivierten M2-Makrophagen differenzieren. Durch GM-CSF werden aus Monozyten GM-CSF-Makrophagen induziert. Im Gegensatz zu M1-Makrophagen sind GM1-Makrophagen bisher noch wenig untersucht. Mit INFg und LPS werden GM-CSF-Makrophagen zu GM1-Makrophagen aktivert. In der vorliegenden Arbeit wurde überprüft, wie groß die Übereinstimmung zwischen M-CSF- und M2-Makrophagen sowie zwischen GM-CSF- und M1-Makrophagen / GM1-Makrophagen ist. Im Gegensatz zu M-CSF- und GM-CSF stellt Laktat aber keinen Differenzierungsfaktor für Monozyten dar. Jedoch beeinflusst Laktat den Phänotyp von M2-Makrophagen und hemmt die Ausschüttung von IL-12 und NO durch M1- und GM1-Makrophagen.
Ziel dieser Arbeit war es, strukturelle Veränderungen präsynaptischer Aktiver Zonen als mögliches Korrelat synaptischer Plastizität zu detektieren. Damit soll die Hypothese getestet werden, dass strukturelle Plastizität Aktiver Zonen eine zentrale Rolle bei der Informationsverarbeitung im Gehirn und bei Lern- und Gedächtnisprozessen spielt. Dazu war es notwendig Methoden zu etablieren, die die strukturelle Analyse Aktiver Zonen und deren Veränderung in vitalem Gewebe ermöglichen. Um die Untersuchungen in einem Gewebe mit plastischen Eigenschaften durchzuführen, wurden Methoden zur Herstellung organotypischer hippocampaler Hirnschnittkulturen etabliert, da hippokampale Moosfasersynapsen ausgeprägte präsynaptische Plastizität aufweisen (Bliss und Collingridge, 1993). Durch Einzelzellelektroporation wurde es möglich, individuelle Neurone mit Transgenen zur Markierung der gesamten Zelle (DsRed) und synaptischer Substrukturen wie Aktive Zonen (z.B.: GFP-CAST, einem Fluorophor-markierten AZ-Protein) zu transfizieren. Mit konfokaler Bildgebung transfizierter Zellen konnten strukturierte Anreicherungen von GFP-CAST in Moosfaserboutons dargestellt werden. Konfokale Bildgebung von Doppelimmunfluoreszenzfärbungen zur detaillierten Analyse der Proteinlokalisation zeigte ein diffraktionsbedingtes Auflösungsdefizit, das auch durch die Anwendung von STED-Mikroskopie nicht zufriedenstellend gelöst werden konnte. Um eine präzise Karte synaptischer Proteine zu erstellen, wurde hochauflösende Mikroskopie (dSTORM) mit einer lateralen räumlichen Auflösung von 20 nm etabliert. Dabei erwiesen sich die ausgeprägte Plastizität, die hohe Dichte an Aktiven Zonen und die variable Gestalt der Boutons im hippokampalen Präparat als problematisch. Aus diesem Grund wurde die elektronenmikroskopisch gut charakterisierte neuromuskuläre Endplatte mit ihrer symmetrischen molekularen Struktur als Präparat für dSTORM verwendet. An der Endplatte konnte die molekulare Organisation der Aktiven-Zonen-Proteine Piccolo und Bassoon dargestellt werden. Zudem konnten erstmals die Mündungen postsynaptischer Falten lichtmikroskopisch aufgelöst werden. So gelang es Werkzeuge zu etablieren, die mit lichtmikroskopischen Methoden die Darstellung der Architektur Aktiver Zonen mit molekularer Auflösung ermöglichen. Die Herausforderung wird es sein, diese neue Dimension in funktionellem Kontext zu nutzen. Die experimentellen Grundlagen dazu wurden durch eine spezielle Badkammer und die Etablierung von Rollertubekulturen bereits gelegt. Dabei ermöglicht dSTORM die Adressierung quantitativer Fragestellungen bis hin zur Bestimmung der Molekülanzahl.
Background It is well known that carbohydrates play fundamental roles in cell signaling and infection processes as well as tumor formation and progression. However, the interaction pathways and cellular receptors targeted by carbohydrates and glycoconjugates remain poorly examined and understood. This lack of research stems, at least to a major part, from accessibility problems of large, branched oligosaccharides. Results To test glycan - cell interactions in vitro, a variety of tailored oligosaccharides was synthesized chemo-enzymatically. Glycosyltransferases from the GRAS organisms Bacillus megaterium (SacB) and Aspergillus niger (Suc1) were used in this study. Substrate engineering of these glycosyltransferases generally acting on sucrose leads to the controlled formation of novel tailored di-, tri- and tetrasaccharides. Already industrially used as prebiotics in functional food, the immunogenic potential of novel oligosaccharides was characterized in this study. A differential secretion of CXCL8 and CCL2 was observed upon oligosaccharide co-cultivation with colorectal epithelial Caco-2 cells. Conclusion Pure carbohydrates are able to stimulate a cytokine response in human endothelial cells in vitro. The type and amount of cytokine secretion depends on the type of co-cultivated oligosaccharide.
In recent years, Ideomotor Theory has regained widespread attention and sparked the development of a number of theories on goal-directed behavior and learning. However, there are two issues with previous studies’ use of Ideomotor Theory. Although Ideomotor Theory is seen as very general, it is often studied in settings that are considerably more simplistic than most natural situations. Moreover, Ideomotor Theory’s claim that effect anticipations directly trigger actions and that action-effect learning is based on the formation of direct action-effect associations is hard to address empirically. We address these points from a computational perspective. A simple computational model of Ideomotor Theory was tested in tasks with different degrees of complexity.The model evaluation showed that Ideomotor Theory is a computationally feasible approach for understanding efficient action-effect learning for goal-directed behavior if the following preconditions are met: (1) The range of potential actions and effects has to be restricted. (2) Effects have to follow actions within a short time window. (3) Actions have to be simple and may not require sequencing. The first two preconditions also limit human performance and thus support Ideomotor Theory. The last precondition can be circumvented by extending the model with more complex, indirect action generation processes. In conclusion, we suggest that IdeomotorTheory offers a comprehensive framework to understand action-effect learning. However, we also suggest that additional processes may mediate the conversion of effect anticipations into actions in many situations.
Pars plana vitrectomy for malignant glaucoma in non-glaucomatous and in filtered glaucomatous eyes

Purpose: To assess the outcomes of pars plana vitrectomy for the treatment of malignant glaucoma in patients with and without previous filtration surgery. Patients and methods: Data of 15 patients developing malignant glaucoma after trabeculectomy (60%) or following ophthalmic interventions other than filtration surgery (40%) were recorded retrospectively. Pars plana vitrectomy was performed in case of failed medical or laser treatment recreating the normal pathway of aqueous humor. The main outcome measures were the postoperative intraocular pressure (IOP), the frequency of complications, and success rate based on the following criteria: IOP reduction by $20% and to #21 mmHg (definition one) or an IOP , 18 mmHg (definition two) with (qualified success) and without (complete success) glaucoma medication. Results: Vitrectomy reduced IOP from baseline in eyes with and without previous trabeculectomy during a median follow-up of 16.4 months (range 7 days to 58 months); although the majority of patients required glaucoma medication to reach desired IOP. The complete success rates were 11% (both definitions) for patients with filtering blebs and none of the patients without previous trabeculectomy had complete success at the 12-month visit. Complications were few and included transient shallowing of the anterior chamber, choroidal detachment, corneal decompensation, filtering bleb failure, and need for further IOP-lowering procedures. Conclusion: Pars plana vitrectomy is equally effective for malignant glaucoma caused by trabeculectomy or interventions other than filtration surgery, although IOP-lowering medication is necessary in nearly all cases to maintain target IOP.
Onkologische Patienten mit Läsionen an der Zunge zeigen bei einem Mere-Exposure-Versuch mit verbalen Stimuli einen stabilen Mere-Exposure-Effekt. Dies hibt Hinweise daruaf, dass stimulusspezifische-sensomotorische Simulationen im wesentlichen unabhängig von der körperlichen Peripherie sind und sich vor allem in zentralen Strukturen abspielen (Gehirn).
Many microRNAs (miRNAs) are co-regulated during the same physiological process but the underlying cellular logic is often little understood. The conserved, immunomodulatory miRNAs miR-146 and miR-155, for instance, are co-induced in many cell types in response to microbial lipopolysaccharide (LPS) to feedback-repress LPS signalling through Toll-like receptor TLR4. Here, we report that these seemingly co-induced regulatory RNAs dramatically differ in their induction behaviour under various stimuli strengths and act non-redundantly through functional specialization; although miR-146 expression saturates at sub-inflammatory doses of LPS that do not trigger the messengers of inflammation markers, miR-155 remains tightly associated with the pro-inflammatory transcriptional programmes. Consequently, we found that both miRNAs control distinct mRNA target profiles; although miR-146 targets the messengers of LPS signal transduction components and thus downregulates cellular LPS sensitivity, miR-155 targets the mRNAs of genes pervasively involved in pro-inflammatory transcriptional programmes. Thus, miR-155 acts as a broad limiter of pro-inflammatory gene expression once the miR-146 dependent barrier to LPS triggered inflammation has been breached. Importantly, we also report alternative miR-155 activation by the sensing of bacterial peptidoglycan through cytoplasmic NOD-like receptor, NOD2. We predict that dosedependent responses to environmental stimuli may involve functional specialization of seemingly coinduced miRNAs in other cellular circuitries as well.
Faces in context: A review and systematization of contextual influences on affective face processing

Facial expressions are of eminent importance for social interaction as they convey information about other individuals’ emotions and social intentions. According to the predominant “basic emotion” approach, the perception of emotion in faces is based on the rapid, auto- matic categorization of prototypical, universal expressions. Consequently, the perception of facial expressions has typically been investigated using isolated, de-contextualized, static pictures of facial expressions that maximize the distinction between categories. However, in everyday life, an individual’s face is not perceived in isolation, but almost always appears within a situational context, which may arise from other people, the physical environment surrounding the face, as well as multichannel information from the sender. Furthermore, situational context may be provided by the perceiver, including already present social infor- mation gained from affective learning and implicit processing biases such as race bias.Thus, the perception of facial expressions is presumably always influenced by contextual vari- ables. In this comprehensive review, we aim at (1) systematizing the contextual variables that may influence the perception of facial expressions and (2) summarizing experimental paradigms and findings that have been used to investigate these influences. The studies reviewed here demonstrate that perception and neural processing of facial expressions are substantially modified by contextual information, including verbal, visual, and auditory information presented together with the face as well as knowledge or processing biases already present in the observer. These findings further challenge the assumption of auto- matic, hardwired categorical emotion extraction mechanisms predicted by basic emotion theories. Taking into account a recent model on face processing, we discuss where and when these different contextual influences may take place, thus outlining potential avenues in future research.
Background: Compensation of brain injury in multiple sclerosis (MS) may in part work through mechanisms involving neuronal plasticity on local and interregional scales. Mechanisms limiting excessive neuronal activity may have special significance for retention and (re-)acquisition of lost motor skills in brain injury. However, previous neurophysiological studies of plasticity in MS have investigated only excitability enhancing plasticity and results from neuroimaging are ambiguous. Thus, the aim of this study was to probe long-term depression-like central motor plasticity utilizing continuous theta-burst stimulation (cTBS), a non-invasive brain stimulation protocol. Because cTBS also may trigger behavioral effects through local interference with neuronal circuits, this approach also permitted investigating the functional role of the primary motor cortex (M1) in force control in patients with MS. Methods: We used cTBS and force recordings to examine long-term depression-like central motor plasticity and behavioral consequences of a M1 lesion in 14 patients with stable mild-to-moderate MS (median EDSS 1.5, range 0 to 3.5) and 14 age-matched healthy controls. cTBS consisted of bursts (50 Hz) of three subthreshold biphasic magnetic stimuli repeated at 5 Hz for 40 s over the hand area of the left M1. Corticospinal excitability was probed via motor-evoked potentials (MEP) in the abductor pollicis brevis muscle over M1 before and after cTBS. Force production performance was assessed in an isometric right thumb abduction task by recording the number of hits into a predefined force window. Results: cTBS reduced MEP amplitudes in the contralateral abductor pollicis brevis muscle to a comparable extent in control subjects (69 ± 22% of baseline amplitude, p < 0.001) and in MS patients (69 ± 18%, p < 0.001). In contrast, postcTBS force production performance was only impaired in controls (2.2 ± 2.8, p = 0.011), but not in MS patients (2.0 ± 4.4, p = 0.108). The decline in force production performance following cTBS correlated with corticomuscular latencies (CML) in MS patients, but did not correlate with MEP amplitude reduction in patients or controls. Conclusions: Long-term depression-like plasticity remains largely intact in mild-to-moderate MS. Increasing brain injury may render the neuronal networks less responsive toward lesion-induction by cTBS.
It is of interest to define bacterial toxin biochemical properties to use them as molecular-syringe devices in order to deliver enzymatic activities into host cells. Binary toxins of the AB7/8-type are among the most potent and specialized bacterial protein toxins. The B subunits oligomerize to form a pore that binds with high affinity host cell receptors and the enzymatic A subunit. This allows the endocytosis of the complex and subsequent injection of the A subunit into the cytosol of the host cells. Here we report that the addition of an N-terminal His6-tag to different proteins increased their binding affinity to the protective antigen (PA) PA63-channels, irrespective if they are related (C2I) or unrelated (gpJ, EDIN) to the AB7/8-family of toxins. His6-EDIN exhibited voltage-dependent increase of the stability constant for binding by a factor of about 25 when the trans-side corresponding to the cell interior was set to 270 mV. Surprisingly, the C. botulinum toxin C2II-channel did not share this feature of PA63. Cell-based experiments demonstrated that addition of an N-terminal His6-tag promoted also intoxication of endothelial cells by C2I or EDIN via PA63. Our results revealed that addition of His6-tags to several factors increase their binding properties to PA63 and enhance the property to intoxicate cells.
Ziel dieser klinisch prospektiven Studie ist es bei über 80 jährigen Patienten mit moderater und hochgradiger AS echokardiographische Parameter zu identifizieren, die die langfristige Prognose für diese Patienten quantifizieren können. Methodik: Es wurde bei 118 Patienten eine konventionelle echokardiographische Standarduntersuchung und eine Gewebedoppleruntersuchung durchgeführt. Zudem wurden die aktuellen klinischen Daten erhoben. Nach durchschnittlich 641 ± 224.2 wurde eine telefonische Follow Up Untersuchung durchgeführt. Ergebnisse: Die Patienten wurden anhand der konventionellen echokardiographischen Parameter in 5 Gruppen eingeteilt. Eine hochgradige Aortenstenose mit reduziertem Gradienten und reduzierter EF ist mittels der Standardechokardiographiedaten von einer moderaten AS signifikant unterscheidbar. Besser gelingt es jedoch eine Niedrig Gradient AS mit der mittels Gewebedoppler ermittelten Strainrate und der Ringmotion von einer mittelgradigen AS zu unterscheiden. Die Patienten aus der Gruppe mit einer hochgradigen AS und einem hohen Gradienten wiesen das höchste Risiko eines kardialen Todes auf. Es stellte sich heraus, dass diese Patienten mittels Ringmotion und Strainrate signifikant von den Patienten mit einem niedrigeren Risiko unterschieden werden können. Zusammenfassung: Die Detektion des Grades der AS mit zusätzlich der Ringmotion und der Strainrate hat signifikante Vorteile gegenüber der Risikoquantifizierung über die EF und den Druckgradienten über der Aortenklappe alleine. Dies liefert wiederum eine bessere Einschätzung bezüglich der Dringlichkeit und Planung des Zeitpunktes für einen operativen Aortenklappenersatz, da alleine mit den Parametern der EF und des Gradienten die eigentlichen Hochrisikopatienten nicht ausreichend erfasst werden.
Vergleich der retropubischen und perinealen radikalen Prostatektomie im Hinblick auf die gesundheitsbezogene Lebensqualität. Die Patienten wurden zu vier Zeitpunkten (präoperativ, nach 3, 6 und 12 Monaten) mit den Lebensqualitätsfragebögen SF-36, EORTC QLQ-C30 und QLQ-PR25 sowie einem selbst gestalteten zusätzlichen Fragebogen untersucht. Insgesamt zeigten sich bei geringer Teststärke kaum statistisch signifikante Unterschiede in den Operationsmethoden bei doch deutlich unterschiedlichen postoperativen Verläufen.
Die hier vorliegende Arbeit behandelt die Darstellung und Charakterisierung von Koordinationspolymeren und MOFs der Lanthanide und des Bariums mit verschiedenen, azolischen, N-Heterozyklen. Es können insgesamt 18 neue organisch-anorganische Hybridmaterialien sowie eine Reihe von co-dotierten Verbindungen präsentiert werden. Neben der strukturellen Charakterisierung dieser Materialien anhand von Röntgenpulverbeugungsdaten steht die Betrachtung der thermischen und photolumineszenzspektroskopischen Eigenschaften im Fokus. Die Lanthanide von La bis Lu, mit den Ausnahmen Eu und Pm ergeben durch Reaktion mit 1H-1,2,3-Triazol die Reihe der isotypen dichten 3D-MOFs 3{infinite}[Ln(Tz*)3]. Die photolumineszenzspektroskopische Untersuchung dieser Verbindungen offenbart eine große Bandbreite verschiedener Lumineszenzphänomene inklusive eines so erstmals beobachteten intrinsischen inner-filter-Effektes der Ln3+-Ionen. Die Strukturen dieser isotypen Reihe von Verbindungen wurden anhand von Röntgenpulverbeugungsdaten gelöst und verfeinert. Ein 2D-Polymorph dieser Verbindungen, 2{infinite}[Ln(Tz*)3], wird für Ln = Sm, Tb beschrieben und anhand von Einkristallbeugungsdaten charakterisiert. Die Reaktion von Eu mit 1H-Benzotriazol ergibt das 1D-Koodinationspolymer 1{infinite}[Eu(Btz)2(BtzH)2], welches das erste Beispiel eines zweiwertigen Seltenerdbenzotriazolats darstellt. Die Analyse der thermischen Eigenschaften dieser Verbindung weist auf die Umwandlung in das 3D-MOF 3{infinite}[Eu(Btz)2] hin, dessen Struktur ebenfalls aus Röntgenpulverbeugungsdaten bestimmt wurde. Die Untersuchung der Photolumineszenzeigenschaften der Verbindungen der Mischkristallreihe 3{infinite}[Ba1-xEux(Im)2], welche aus Reaktionen der salzartigen Hydride BaH2 und EuH2 mit Imidazol dargestellt wurden, zeigen, dass die Synthese lumineszenter MOFs durch Co-Dotierung nicht lumineszenter Netzwerke mit geeigneten Lumineszenzzentren möglich ist. Die Struktur dieser Materialien wurde anhand der undotierten Verbindung 3{infinite}[BaEu(Im)2] aus Röntgenpulverbeugungsdaten bestimmt. Die strukturelle Charakterisierung von Materialien anhand von Röntgenpulverbeugungsdaten nimmt einen besonderen Stellenwert innerhalb dieser Arbeit an. Es wird ein Verfahren präsentiert, das über Indizierung des Diffraktogramms, Intensitätsextraktion und Strukturlösung über charge-flipping-Methoden eine Möglichkeit darstellt ab initio die Struktur eines unbekannten Materials ausschließlich anhand von Röntgenpulverbeugungsdaten zu bestimmten. Des Weiteren wird aufgezeigt, wie die Strukturen dieser Materialien, mit einem hohen Anteil schwach streuender Leichtatome, durch die Verwendung von rigid body constraints verfeinert werden können. Die Strukturlösungen von 3{infinite}[Ga(Tz)2], 3{infinite}[Sr(Im)2]:Ce und 3{infinite}[Yb(Im)3]ImH aus Röntgenpulverbeugungsdaten stellen weitere Beispiele für die erfolgreiche Anwendung der im Rahmen dieser Arbeit präsentierten Methode dar.
Both specific stimulus valence and unspecific processing dynamics can influence evaluative responses. Eight experiments investigated their respective influence on evaluative judgments in the domain of stereotyping. Valence of stereotypic information and consistency-driven fluency were manipulated in an impression formation paradigm. When information about the to-be-evaluated target person was strongly valenced, no effects of consistency-driven fluency were observed. Higher cognitive processes, valence of inconsistent attributes, processing priority of category information, and impression formation instructions were ruled out as possible factors responsible for the non-occurrence of fluency effects. However, consistency-driven fluency did influence the evaluative judgment, if the information about a target person was not strongly valenced. It is therefore concluded that both stimulus valence and consistency-driven processing fluency play a role in evaluative judgments in the domain of stereotyping. The respective impact of stimulus valence is much stronger than the impact of unspecific processing dynamics, however. Implications for fluency research and the applied field of stereotype change are discussed.
Diazepinomicin is a dibenzodiazepine alkaloid with an unusual structure among the known microbial metabolites discovered so far. Diazepinomicin was isolated from the marine sponge-associated strain Micromonospora sp. RV115 and was identified by spectroscopic analysis and by comparison to literature data. In addition to its interesting preclinical broad-spectrum antitumor potential, we report here new antioxidant and anti-protease activities for this compound. Using the ferric reducing antioxidant power (FRAP) assay, a strong antioxidant potential of diazepinomicin was demonstrated. Moreover, diazepinomicin showed a significant antioxidant and protective capacity from genomic damage induced by the reactive oxygen species hydrogen peroxide in human kidney (HK-2) and human promyelocytic (HL-60) cell lines. Additionally, diazepinomicin inhibited the proteases rhodesain and cathepsin L at an IC50 of 70–90 μM. It also showed antiparasitic activity against trypomastigote forms of Trypanosoma brucei with an IC50 of 13.5 μM. These results showed unprecedented antioxidant and anti-protease activities of diazepinomicin, thus further highlighting its potential as a future drug candidate.
Terahertz electromagnetic fields are non-ionizing electromagnetic fields in the frequency range from 0.1 to 10 THz. Potential applications of these electromagnetic fields include the whole body scanners, which currently apply millimeter waves just below the terahertz range, but future scanners will use higher frequencies in the terahertz range. These and other applications will bring along human exposure to these fields. Up to now, only a limited number of investigations on biological effects of terahertz electromagnetic fields have been performed. Therefore, research is strongly needed to enable reliable risk assessment. Cells were exposed for 2 h, 8 h, and 24 h with different power intensities ranging from 0.04 mW/cm2 to 2 mW/cm2, representing levels below, at, and above current safety limits. Genomic damage on the chromosomal level was measured as micronucleus formation. DNA strand breaks and alkali-labile sites were quantified with the comet assay. No DNA strand breaks or alkali-labile sites were observed as a consequence of exposure to terahertz electromagnetic fields in the comet assay. The fields did not cause chromosomal damage in the form of micronucleus induction.
Connective tissue growth factor (CTGF/CCN2) is an angiogenetic and profibrotic factor, acting downstream of TGF-b, involved in both airway- and vascular remodeling. While the T-helper 1 (Th1) cytokine interferon-gamma (IFN-c) is well characterized as immune-modulatory and anti-fibrotic cytokine, the role of IFN-c in lung endothelial cells (LEC) is less defined. Tumour necrosis factor alpha (TNF-a) is another mediator that drives vascular remodeling in inflammation by influencing CTGF expression. In the present study we investigated the influence of IFN-c and TNF-a on CTGF expression in human LEC (HPMEC-ST1.6R) and the effect of CTGF knock down on human LEC. IFN-c and TNF-a down-regulated CTGF in human LEC at the promoter-, transcriptional- and translational-level in a dose- and time-dependent manner. The inhibitory effect of IFN-c on CTGF-expression could be almost completely compensated by the Jak inhibitor AG-490, showing the involvement of the Jak-Stat signaling pathway. Besides the inhibitory effect of IFN-c and TNF-a alone on CTGF expression and LEC proliferation, these cytokines had an additive inhibitory effect on proliferation as well as on CTGF expression when administered together. To study the functional role of CTGF in LEC, endogenous CTGF expression was down-regulated by a lentiviral system. CTGF silencing in LEC by transduction of CTGF shRNA reduced cell proliferation, but did not influence the anti-proliferative effect of IFN-c and TNF-a. In conclusion, our data demonstrated that CTGF was negatively regulated by IFN-c in LEC in a Jak/Stat signaling pathway-dependent manner. In addition, an additive effect of IFN-c and TNF-a on inhibition of CTGF expression and cell proliferation could be found. The inverse correlation between IFN-c and CTGF expression in LEC could mean that screwing the Th2 response to a Th1 response with an additional IFN-c production might be beneficial to avoid airway remodeling in asthma.
Das Renin-Angiotensin-Aldosteron-System (RAAS) reguliert den Blutdruck und den Wasser- und Elektrolythaushalt des Körpers. Angiotensin II (Ang II), das aktive Peptid des RAAS, bewirkt eine Vasokonstriktion und in höheren Konzentrationen Bluthochdruck. Epidemiologische Studien haben gezeigt, dass eine Verbindung zwischen Hypertonie und dem gehäuften Auftreten von Krebs besteht. Eine Metaanalyse von 13 Fall-Kontroll-Studien konnte einen Zusammenhang zwischen Hypertonie und einem erhöhten Risiko, an einem Nierenzellkarzinom zu erkranken nachweisen. In vitro-Studien und Studien an der isolierten Niere konnten bereits genotoxische Effekte des blutdruckregulierenden Hormons Ang II zeigen. Zielsetzung dieser Arbeit war es, zunächst in vivo zu prüfen, ob steigende Ang II-Konzentrationen einen Einfluss auf die genomische Stabilität von Nieren- und Herzzellen besitzen. Hierzu wurden im Dosisversuch männliche C57BL/6-Mäuse mit osmotischen Minipumpen ausgestattet, die Ang II in vier verschiedenen Konzentrationen zwischen 60 ng/kg min und 1 µg/kg min über einen Zeitraum von 28 Tagen abgeben sollten. Während des Versuchszeitraums fanden regelmäßige, nicht-invasive Blutdruckmessungen an der Maus statt. Die Behandlung mit Ang II führte zu einem signifikanten Anstieg des Blutdrucks und zu histopathologischen Veränderungen der Glomeruli und des Tubulussystems, was sich in einer verschlechterten Albumin-Ausscheidung wiederspiegelte. Außerdem induzierte die Behandlung mit Ang II die dosisabhängige Bildung von reaktiven Sauerstoffspezies, DNA-Doppelstrangbrüchen und oxidativer DNA-Schäden. Diese Parameter waren bereits in Tieren erhöht, die keinen Bluthochdruck entwickelten und stiegen mit der höchsten Ang II-Konzentration noch an, obwohl hier im Vergleich zur Vorgängergruppe, die eine geringere Ang II-Konzentration erhielt, kein höherer Blutdruck vorlag. Diese Beobachtung deutet auf eine mögliche Unabhängigkeit des entstandenen Schadens vom Bluthochdruck hin und lenkt die Aufmerksamkeit auf Ang II als genomschädigenden Faktor. Der folgende Interventionsversuch sollte Aufschluss über die mögliche blutdruckunabhängige genomschädigende Wirkung von Ang II geben. Dazu wurden C57BL/6-Mäuse neben der Ang II-Behandlung in einer Konzentration von 600 ng/kg min zusätzlich über einen Zeitraum von 28 Tagen mit 5 verschiedenen Substanzen behandelt: Candesartan, Ramipril, Hydralazin, Eplerenon und Tempol. Candesartan ist ein Ang II-Rezeptor-Antagonist, der selektiv den AT1-Rezeptor blockiert. Ramipril wirkt als Hemmer des Angiotensin-Konversions-Enzyms und verhindert die Bildung von endogenem Ang II aus Ang I. Hydralazin, als Vasodilatator, greift nicht in das Renin-Angiotensin-Aldosteron-System ein. Eplerenon blockiert als selektiver Aldosteronantagonist den Mineralkortikoidrezeptor. Tempol wirkt als Antioxidans. Die Behandlung mit Ang II in einer Konzentration von 600 ng/kg min im Interventionsversuch führte zur Hochregulierung der NADPH-Oxidase 4 und zur Produktion reaktiver Sauerstoffspezies in der Niere und im kardiovaskulären Gewebe. Der entstandene oxidative Stress führte wiederum zu DNA-Schäden und einer Aktivierung der Transkriptionsfaktoren Nrf2 und NF-B. Nrf2-vermittelt wurde die Induktion antioxidativer Gene ausgelöst, was allerdings nicht ausreichend war, um vor Ang II-induzierten ROS und DNA-Schäden zu schützen. Eine längerfristige NF-B-Aktivierung durch hohe Ang II-Spiegel kann das Überleben und die Proliferation von Zellen, die DNA-Schäden in Form von Doppelstrangbrüchen tragen, fördern, was eine Tumor-initiierende Wirkung haben könnte. Die beschriebenen Effekte erhöhter Ang II-Spiegel konnten durch die Intervention mit dem AT1-Rezeptorblocker Candesartan verhindert werden, was die Beteiligung des Rezeptors nachweist. Eine blutdruckunabhängige, genomschädigende Wirkung von Ang II konnte leider durch die Intervention mit Hydralazin nicht verdeutlicht werden, da die erwünschte langfristige Blutdrucksenkung ausblieb. Allerdings zeigte die Intervention mit Tempol eine Abnahme an oxidativem Stress und DNA-Schäden trotz ausbleibender Blutdrucksenkung. Die Bedeutung von ROS in der Bildung von DNA-Schäden und die Unabhängigkeit dieser Schäden vom Blutdruck konnten somit hervorgehoben werden. Die Tatsache, dass die Intervention mit Ramipril den Blutdruck nicht senken konnte, der oxidative Stress und die DNA-Schäden durch mögliche antioxidative Eigenschaften aber vermindert wurden, unterstützt diese Beobachtung. Die Intervention mit Eplerenon führte zum Teil zu einer Verminderung an ROS und DNA-Schäden, brachte diese Parameter aber nicht auf Kontrollniveau zurück. Somit ist eine Beteiligung von Aldosteron nicht auszuschließen.
Background If detected in time, delayed cerebral vasospasm after aneurysmal subarachnoid hemorrhage (SAH) may be treated by balloon angioplasty or chemical vasospasmolysis in order to enhance cerebral blood flow (CBF) and protect the brain from ischemic damage. This study was conceived to compare the diagnostic accuracy of detailed neurological examination, Transcranial Doppler Sonography (TCD), and Perfusion-CT (PCT) to detect angiographic vasospasm. Methods The sensitivity, specificity, positive and negative predictive values of delayed ischemic neurological deterioration (DIND), pathological findings on PCT- maps, and accelerations of the mean flow velocity (MVF) were calculated. Results The accuracy of DIND to predict angiographic vasospasm was 0.88. An acceleration of MFV in TCD (>140 cm/s) had an accuracy of 0.64, positive PCT-findings of 0.69 with a higher sensitivity, and negative predictive value than TCD. Interpretation Neurological assessment at close intervals is the most sensitive and specific parameter for cerebral vasospasm. PCT has a higher accuracy, sensitivity and negative predictive value than TCD. If detailed neurological evaluation is possible, it should be the leading parameter in the management and treatment decisions. If patients are not amenable to detailed neurological examination, PCT at regular intervals is a helpful tool to diagnose secondary vasospasm after aneurysmal SAH.
Background: Ga-[1,4,7,10-tetraazacyclododecane-N,N0,N00,N000-tetraacetic acid]-d-Phe1,Tyr3-octreotate (DOTATATE) positron emission tomography (PET) is commonly used for the visualization of somatostatin receptor (SSTR)-positive neuroendocrine tumors. SSTR is also known to be expressed on macrophages, which play a major role in inflammatory processes in the walls of coronary arteries and large vessels. Therefore, imaging SSTR expression has the potential to visualize vulnerable plaques. We assessed 68Ga-DOTATATE accumulation in large vessels in comparison to 18F-2-fluorodeoxyglucose (FDG) uptake, calcified plaques (CPs), and cardiovascular risk factors. Methods: Sixteen consecutive patients with neuroendocrine tumors or thyroid cancer underwent both 68Ga-DOTATATE and 18F-FDG PET/CT for staging or restaging purposes. Detailed clinical data, including common cardiovascular risk factors, were recorded. For a separate assessment, they were divided into a high-risk and a low-risk group. In each patient, we calculated the maximum target-to-background ratio (TBR) of eight arterial segments. The correlation of the TBRmean of both tracers with risk factors including plaque burden was assessed. Results: The mean TBR of 68Ga-DOTATATE in all large arteries correlated significantly with the presence of CPs (r = 0.52; p < 0.05), hypertension (r = 0.60; p < 0.05), age (r = 0.56; p < 0.05), and uptake of 18F-FDG (r = 0.64; p < 0.01). There was one significant correlation between 18F-FDG uptake and hypertension (0.58; p < 0.05). Out of the 37 sites with the highest focal 68Ga-DOTATATE uptake, 16 (43.2%) also had focal 18F-FDG uptake. Of 39 sites with the highest 18F-FDG uptake, only 11 (28.2%) had a colocalized 68Ga-DOTATATE accumulation. Conclusions: In this series of cancer patients, we found a stronger association of increased 68Ga-DOTATATE uptake with known risk factors of cardiovascular disease as compared to 18F-FDG, suggesting a potential role for plaque imaging in large arteries. Strikingly, we found that focal uptake of 68Ga-DOTATATE and 18F-FDG does not colocalize in a significant number of lesions.
When a key press causes a stimulus, the key press is perceived later and the stimulus earlier than key presses and stimuli presented independently. This bias in time perception has been linked to the intention to produce the effect and thus been called intentional binding (IB). In recent studies it has been shown that the IB effect is stronger when participants believed that they caused the effect stimulus compared to when they believed that another person caused the effect (Desantis et al., 2011). In this experiment we ask whether causal beliefs influence the perceived time of an effect when the putative effect occurs temporally close to another stimulus that is also an effect. In our study two participants performed the same task on connected computers with separate screens. Each trial started synchro- nously on both computers. When a participant pressed a key, a red and a yellow stimulus appeared as action effects simultaneously or with a slight delay of up to 50 ms. The partic- ipants’ task was to judge the temporal order of these two effect stimuli. Participants were either told that one participant caused one of the two stimuli while the other participant seated at the other computer caused the other stimulus, or each participant was told that he/she caused both stimuli. The different causal beliefs changed the perceived time of the effects’ appearance relative to each other. When participants believed they each caused one effect, their “own” effect was perceived earlier than the other participant’s effect. When the participants believed each caused both effects, no difference in the perceived temporal order of the red and yellow effect was found. These results confirm that higher order causal beliefs change the perceived time of an action effect even in a setting in which the occurrence of the putative effect can be directly compared to a reference stimulus.

Genetische und erworbene thrombophile Gerinnungsstörungen als Quelle chronischer Schmerzsyndrome

Anhand einer umfassenden Falldarstellung einer jungen Patientin mit einem lebensbedrohlichen Gesichtsschmerzsyndrom, das nach septischer Thrombose der periorbitalen venösen und arteriellen Gefäße aufgetreten war, wurde die Bedeutung einer medikamentösen Antikoagulation für die erfolgreiche Schmerztherapie herausgearbeitet. An diesem Fallbeispiel konnte aber auch gezeigt werden, dass keine sicheren Parameter für die Indikation einer solchen Gerinnungstherapie vorlagen. Die Bedeutung dieses Falls lag unzweifelhaft in der Erkenntnis, dass in einer anhaltenden Aktivierung des Kontaktsystems der Gerinnung ein bislang unterschätztes Potential für die Entstehung und Unterhaltung ungeklärter Schmerzen liegen könnte und nicht zuletzt auch daran, dass sich diese ätiologische Komponente in der Komplexität der Erkrankung diagnostisch nicht eindeutig sichern ließ. Mit der Translokation von LPS aus der intestinalen Mukosa in endothelial vorgeschädigte Gefäßabschnitte wurde eine Hypothese vorgetragen, die neben einer schwer detektierbaren inflammatorischen Komponente auch das prokoagulatorische Potential der Schmerzentstehung erklären könnte. Die prokoagulatorische Komponente dieses hypothetischen Entstehungs-mechanismus chronischer Schmerzen müsste, so die Arbeitshypothese, umso dominanter sein, wenn prokoagulatorisch wirksame genetische Faktoren bei den Patienten hinzukommen. Unter der Annahme, dass eine solche zusätzliche Diathese nicht nur eine Schrittmacherfunktion haben, sondern auch einen diagnostischen Beitrag liefern könnte, wurde dieses diagnostische Pilotprojekt mit der empirisch begründeten Heparintherapie von 97 Schmerzpatienten verbunden. Alle Pa-tienten wurden mit dem niedermolekularen Heparin Enoxaparin behandelt und nach zehn Behandlungstagen in vier verschiedene Respondergruppen (Gruppe 1 bis 4) eingeteilt. Diese Gruppen wurden auf fünf prothrombotische Parameter untersucht. Dazu wurden die Allelprävalenzen des Plasminogen Aktivator Inhibitor-(PAI-1 4G/5G) Polymorphismus, der Faktor V-Leiden-Mutation, der Prothrombin (G20210A) Genmutation sowie die Prävalenzen der Hyperfibrinogenämie und des Protein S-Mangels ermittelt. Mit Hilfe des exakten Fisher Tests wurden jeweils die Allelprävalenzen und Parameter sowohl der Respondergruppen 1 bis 3 mit einem Kollektiv der Allgemeinbevölkerung als auch mit dem Kollektiv der Non-Responder (Gruppe 4) verglichen. Die Prävalenz des Allels A der Faktor V-Leiden-Mutation G1691A war im Enoxaparin-Kollektiv bei den Respondern der Gruppen 1 bis 3 im Vergleich zur Allgemeinbevölkerung und zur Non-Respondergruppe (Gruppe 4) signifikant erhöht. Die Allelprävalenzen und Parameter der übrigen prokoagulatorischen Faktoren unterschieden sich von denen der Kontrollgruppen nicht. Anhand des Kallikrein-Kinin-Systems als möglichem Effektor des Hämosta-sesystems konnten Hinweise auf die kausale Wirksamkeit des nieder-molekularen Heparins Enoxaparin bei der Behandlung chronischer Schmerzen gegeben werden.
Both human herpes viruses and Chlamydia are highly prevalent in the human population and are detected together in different human disorders. Here, we demonstrate that co-infection with human herpes virus 6 (HHV6) interferes with the developmental cycle of C. trachomatis and induces persistence. Induction of chlamydial persistence by HHV6 is independent of productive virus infection, but requires the interaction and uptake of the virus by the host cell. On the other hand, viral uptake is strongly promoted under co-infection conditions. Host cell glutathione reductase activity was suppressed by HHV6 causing NADPH accumulation, decreased formation of reduced glutathione and increased oxidative stress. Prevention of oxidative stress restored infectivity of Chlamydia after HHV6-induced persistence. We show that co-infection with Herpes simplex virus 1 or human Cytomegalovirus also induces chlamydial persistence by a similar mechanism suggesting that Chlamydia -human herpes virus co-infections are evolutionary shaped interactions with a thus far unrecognized broad significance.
Flexible behavior is only possible if contingencies between own actions and following environmental effects are acquired as quickly as possible; and recent findings indeed point toward an immediate formation of action-effect bindings already after a single coupling of an action and its effect. The present study explored whether these short-term bindings occur for both, stimulus- and goal-driven actions (“forced-choice actions” vs. “free-choice actions”). Two experiments confirmed that immediate action-effect bindings are formed for both types of actions and affect upcoming behavior. These findings support the view that action-effect binding is a ubiquitous phenomenon which occurs for any type of action.
Delayed cerebral vasospasm following subarachnoid hemorrhage (SAH) is a serious medical complication, characterized by constriction of cerebral arteries leading to varying degrees of cerebral ischemia. Numerous clinical and experimental studies have been performed in the last decades; however, the pathophysiologic mechanism of cerebral vasospasm after SAH still remains unclear. Among a variety of experimental SAH models, the double hemorrhage rat model involving direct injection of autologous arterial blood into the cisterna magna has been used most frequently for the study of delayed cerebral vasospasm following SAH in last years. Despite the simplicity of the technique, the second blood injection into the cisterna magna may result in brainstem injury leading to high mortality. Therefore, a modified double hemorrhage model of cisterna magna has been developed in rat recently. We describe here step by step the surgical technique to induce double SAH and compare the degree of vasospasm with other cisterna magna rat models using histological assessment of the diameter and cross-sectional area of the basilar artery
Previous research using neuroimaging methods proposed a link between mechanisms controlling motor response inhibition and suppression of unwanted memories.The present study investigated this hypothesis behaviorally by combining the think/no-think paradigm (TNT) with a go/no-go motor inhibition task. Participants first learned unpleasant cue-target pairs. Cue words were then presented as go or no-go items in the TNT. Participants’ task was to respond to the cues and think of the target word aloud or to inhibit their response to the cue and the target word from coming to mind. Cued recall assessed immediately after the TNT revealed reduced recall performance for no-go targets compared to go targets or baseline cues not presented in the TNT. The results demonstrate that doing the no-think and no-go task concurrently leads to memory suppression of unpleasant items during later recall. Results are discussed in line with recent empirical research and theoretical positions.
The ability of CD4+Foxp3+ regulatory T-cells (Treg) to produce interleukin (IL)-10 is important for the limitation of inflammation at environmental interfaces like colon or lung. Under steady state conditions, however, few Tregs produce IL-10 ex vivo. To investigate the origin and fate of IL-10 producing Tregs we used a superagonistic mouse anti-mouse CD28 mAb (CD28SA) for polyclonal in vivo stimulation of Tregs, which not only led to their numeric expansion but also to a dramatic increase in IL-10 production. IL-10 secreting Tregs strongly upregulated surface receptors associated with suppressive function as compared to non-producing Tregs. Furthermore, polyclonally expanding Tregs shifted their migration receptor pattern after activation from a CCR7+CCR52 lymph node-seeking to a CCR72CCR5+ inflammationseeking phenotype, explaining the preferential recruitment of IL-10 producers to sites of ongoing immune responses. Finally, we observed that IL-10 producing Tregs from CD28SA stimulated mice were more apoptosis-prone in vitro than their IL-10 negative counterparts. These findings support a model where prolonged activation of Tregs results in terminal differentiation towards an IL-10 producing effector phenotype associated with a limited lifespan, implicating built-in termination of immunosuppression.
Elektrophysiologische Korrelate der Konfliktverarbeitung bei hierarchischen Stimuli mit drei Ebenen

Diese Studie untersucht den Einfluss des Aufmerksamkeitsfokus und des Interferenzgrades dreidimensionaler Stimuli auf Verhaltensdaten (Anzahl korrekter Antworten, Reaktionszeiten) und elektrophysiologische Daten. Wir orientierten uns hierbei an einem von Blasi gestalteten dreidimensionalen Pfeilbild- Paradigma (Blasi et al., 2005, 2007). Blasis Paradigma darf man insofern als sehr innovativ bezeichnen, da er der einer der wenigen war, der im Bereich der Interferenzforschung dreidimensionales Stimulusmaterial zum Einsatz brachte. Jedoch sind wir der Meinung, dass Blasis Entwurf des Paradigmas sich trotz aller Innovativität nicht in vollem Ausmaß dazu eignet, valide Aussagen über Interferenz und Aufmerksamkeit zu treffen. Der mögliche Einfluss des visuellen Erscheinungsbildes eines Stimulus, die Möglichkeit von Lateralitätseffekten, die Beschränkung auf nur einzelne Pfeilstimuli, sowie die mangelnde Trennung von Interferenz- und Aufmerksamkeitsprozessen fanden dort keine kritische Beachtung. Im Gegensatz zu Blasi führten wir unsere Untersuchungen nicht mit dem fMRI, sondern mittels ereigniskorrelierter Potentiale (P1, P2, N2, P3) durch. Zahlreiche andere Studien vor uns untersuchten bereits die Auswirkungen von Interferenz und Aufmerksamkeitsfokus auf ebendiese Potentiale, da EKPs eine genauere zeitliche Information liefern. Das Besondere an unserem Vorgehen war, dass – soweit in der Literatur bekannt – zum ersten Mal der Einfluss von Aufmerksamkeit und Interferenz völlig unabhängig von dem optischen Erscheinungsbild des Stimulus, ohne einen zu erwartetem Lateralitätseffet und ohne Konfundation von Interferenz- und Aufmerksamkeitsprozessen diskutiert werden konnte.
In der vorliegenden Arbeit wurden erstmals im 3D-Pulerdruckverfahren hergestellte Struvit-Matrizes auf ihre Eignung als Trägermaterial für Knochenzellen in vitro untersucht. Hierzu wurde die Zytokompatibilität sowie die chemische Löslichkeit von gedruckten Struvit-Strukturen betrachtet. In einem zweiten Schritt wurde untersucht, ob die biologische Funktion von BMP-2-Lösungen nach Durchlaufen des Druckprozesses erhalten bleibt und ob es möglich ist, BMP-2 unter Beibehaltung seiner biologischen Wirksamkeit direkt in Struvit-Matrizes zu drucken. Als Reaktanten zur Herstellung der Struvit-Matrizes wurde modifiziertes Farringtonit-Pulver mit definierter Körnung und eine äquimolare Binder-Lösung aus DAHP und ADHP verwendet. Die untersuchten Zellkulturträger mit Magnesiumammoniumphosphatchemie zeigten eine ausreichende Zytokompatibilität in vitro. Außerdem wurde gezeigt, dass thermolabile Proteine wie BMP-2 im 3D-Pulverdruckverfahren unter weitgehender Beibehaltung ihrer biologischen Wirksamkeit in vitro grundsätzlich prozessierbar sind. Die Freisetzung direkt eingedruckter Proteine aus den Struvit-Matrizes blieb jedoch hinter den Erwartungen zurück. Mit Struvit steht ein alternatives Zementsystem für den 3D-Pulverdruck zur Verfügung, welches spezifische Vorteile gegenüber den etablierten Calciumphosphaten bietet. Weitere Untersuchungen sind erforderlich, um die Ursache für die geringe BMP-Freisetzung aus den Struvit-Matrizes zu ermitteln und die Vorteile der neutralen Abbindereaktion voll nutzen zu können.
Diversity of Nonribosomal Peptide Synthetase Genes in the Microbial Metagenomes of Marine Sponges

Genomic mining revealed one major nonribosomal peptide synthetase (NRPS) phylogenetic cluster in 12 marine sponge species, one ascidian, an actinobacterial isolate and seawater. Phylogenetic analysis predicts its taxonomic affiliation to the actinomycetes and hydroxy-phenyl-glycine as a likely substrate. Additionally, a phylogenetically distinct NRPS gene cluster was discovered in the microbial metagenome of the sponge Aplysina aerophoba, which shows highest similarities to NRPS genes that were previously assigned, by ways of single cell genomics, to a Chloroflexi sponge symbiont. Genomic mining studies such as the one presented here for NRPS genes, contribute to on-going efforts to characterize the genomic potential of sponge-associated microbiota for secondary metabolite biosynthesis.
Molecular characterization of antimicrobial peptide genes of the carpenter ant Camponotus floridanus

The production of antimicrobial peptides (AMPs) is a major defense mechanism against pathogen infestation and of particular importance for insects relying exclusively on an innate immune system. Here, we report on the characterization of three AMPs from the carpenter ant Camponotus floridanus. Due to sequence similarities and amino acid composition these peptides can be classified into the cysteine-rich (e.g. defensin) and glycine-rich (e.g. hymenoptaecin) AMP groups, respectively. The gene and cDNA sequences of these AMPs were established and their expression was shown to be induced by microbial challenge. We characterized two different defensin genes. The defensin-2 gene has a single intron, whereas the defensin-1 gene has two introns. The deduced amino acid sequence of the C. floridanus defensins is very similar to other known ant defensins with the exception of a short C-terminal extension of defensin-1. The hymenoptaecin gene has a single intron and a very peculiar domain structure. The corresponding precursor protein consists of a signal- and a pro-sequence followed by a hymenoptaecin-like domain and six directly repeated hymenoptaecin domains. Each of the hymenoptaecin domains is flanked by an EAEP-spacer sequence and a RR-site known to be a proteolytic processing site. Thus, proteolytic processing of the multipeptide precursor may generate several mature AMPs leading to an amplification of the immune response. Bioinformatical analyses revealed the presence of hymenoptaecin genes with similar multipeptide precursor structure in genomes of other ant species suggesting an evolutionary conserved important role of this gene in ant immunity.
Introduction: Sleep disturbances are common in adolescents and adversely affect performance, social contact, and susceptibility to stress. We investigated the hypothesis of a relationship between sleep and health-related quality of life (HRQoL), and applied self- and proxy ratings. Materials and Methods: The sample comprised 92 adolescents aged 11–17 years. All participants and their parents completed a HRQoL measure and the Sleep Disturbance Scale for Children (SDSC ). Children with SDSC T -scores above the normal range (above 60) were classified as poor sleepers. Results: According to self- and proxy ratings, good sleepers reported significantly higher HRQoL than poor sleep- ers. Sleep disturbances were significantly higher and HRQoL significantly lower in self- as compared to parental ratings. Parent-child agreement was higher for subscales measuring observable aspects. Girls experienced significantly stronger sleep disturbances and lower self-rated HRQoL than boys. Discussion: Our findings support the positive relationship of sleep and HRQoL. Furthermore, parents significantly underestimate sleep disturbances and overestimate HRQoL in their children.
A brain-computer interface (BCI) enables communication without movement based on brain signals measured with electroencephalography (EEG). BCIs usually rely on one of three types of signals: the P300 and other components of the event-related potential (ERP), steady state visual evoked potential (SSVEP), or event related desynchronization (ERD). Although P300 BCIs were introduced over twenty years ago, the past few years have seen a strong increase in P300 BCI research. This closed-loop BCI approach relies on the P300 and other components of the ERP, based on an oddball paradigm presented to the subject. In this paper, we overview the current status of P300 BCI technology, and then discuss new directions: paradigms for eliciting P300s; signal processing methods; applications; and hybrid BCIs. We conclude that P300 BCIs are quite promising, as several emerging directions have not yet been fully explored and could lead to improvements in bit rate, reliability, usability, and flexibility.
The antidepressant fluoxetine has been under discussion because of its potential influence on cancer risk. It was found to inhibit the development of carcinogen-induced preneoplastic lesions in colon tissue, but the mechanisms of action are not well understood. Therefore, we investigated anti-proliferative effects, and used HT29 colon tumor cells in vitro, as well as C57BL/6 mice exposed to intra-rectal treatment with the carcinogen N-methyl-N’-nitro-N-nitrosoguanidine (MNNG) as models. Fluoxetine increased the percentage of HT29 cells in the G0/G1 phase of cell-cycle, and the expression of p27 protein. This was not related to an induction of apoptosis, reactive oxygen species or DNA damage. In vivo, fluoxetine reduced the development of MNNG-induced dysplasia and vascularization-related dysplasia in colon tissue, which was analyzed by histopathological techniques. An anti-proliferative potential of fluoxetine was observed in epithelial and stromal areas. It was accompanied by a reduction of VEGF expression and of the number of cells with angiogenic potential, such as CD133, CD34, and CD31-positive cell clusters. Taken together, our findings suggest that fluoxetine treatment targets steps of early colon carcinogenesis. This confirms its protective potential, explaining at least partially the lower colon cancer risk under antidepressant therapy.
Die vorliegende Arbeit untersucht die positiven Auswirkungen des Angiotensin-II-Rezeptor-Antagonisten Telmisartan auf die endotheliale Funktion und Thrombozytenaktivierung bei Ratten mit Streptozotocin-induziertem Diabetes mellitus. In Gefäßreaktivitätsstudien, Luminometer- und Fluoreszenzmessungen und mit Hilfe der Durchflusszytometrie wurden die Wirkungen des Medikamentes überprüft. Es konnte gezeigt werden, dass sich durch Telmisartan die NO-Bioverfügbarkeit verbessert, welche maßgeblich für die endotheliale Funktion verantwortlich ist und durch Ca2+-abhängige Aktivierung der eNOS und dehnungsinduzierte, Ca2+-unabhängigen NO-Bildung beeinflusst wird. Positiv wird des Weiteren die Sensitivität der glatten Gefäßmuskelzellen gegenüber NO beeinflusst, was zur Vasodilatation führt. Die atherosklerosefördernde Superoxidbildung wird zusätzlich reduziert. Es erfolgten außerdem Messungen von thrombozytengebundenem Fibrinogen, dementsprechend der GP IIb/IIIa-Aktivität, und der VASP-Phosphorylierung, demzufolge dem NO/cGMP-Signalweg, in Thrombozyten durch FITC-markierte Antikörper mit Hilfe der Durchflusszytometrie. Es wurde gezeigt, dass die Thrombozytenaktivierung, die für den initialen Schritt der Atherosklerose verantwortlich gemacht wird, durch Telmisartan verringert wird. Alle Messungen wurden vergleichend in einer Kontroll-, Placebo- und Telmisartangruppe durchgeführt. Die beobachtete Blutdrucksenkung ist, nach früheren Betrachtungen, nicht alleine verantwortlich für die verbesserte endotheliale Funktion, welche bei dem Einsatz von AT-II-Antagonisten beobachtet wird. Telmisartan wirkt, laut einer Studie, als einziger AT-II-Antagonist als partieller PPAR-Rezeptor, so dass Insulinresistenz und metabolische Parameter verbessert werden. Über diese Wirkungen beeinflusst Telmisartan auch die endotheliale Funktion und die Thrombozytenaktivierung. Zur Reduktion von vaskulären Komplikationen bei Diabetes mellitus erscheint Telmisartan aufgrund der vorliegenden Ergebnisse als sinnvolle medikamentöse Therapie.
Recent studies show that combinations of defined key developmental transcription factors (TFs) can reprogram somatic cells to pluripotency or induce cell conversion of one somatic cell type to another. However, it is not clear if single genes can define a cells identity and if the cell fate defining potential of TFs is also operative in pluripotent stem cells in vitro. Here, we show that ectopic expression of the neural TF Neurogenin2 (Ngn2) is sufficient to induce rapid and efficient differentiation of embryonic stem cells (ESCs) into mature glutamatergic neurons. Ngn2-induced neuronal differentiation did not require any additional external or internal factors and occurred even under pluripotency-promoting conditions. Differentiated cells displayed neuron-specific morphology, protein expression, and functional features, most importantly the generation of action potentials and contacts with hippocampal neurons. Gene expression analyses revealed that Ngn2-induced in vitro differentiation partially resembled neurogenesis in vivo, as it included specific activation of Ngn2 target genes and interaction partners. These findings demonstrate that a single gene is sufficient to determine cell fate decisions of uncommitted stem cells thus giving insights into the role of key developmental genes during lineage commitment. Furthermore, we present a promising tool to improve directed differentiation strategies for applications in both stem cell research and regenerative medicine.
Background One third of all cancer patients will develop bone metastases and the vertebral column is involved in approximately 70 % of these patients. Conventional radiotherapy with of 1–10 fractions and total doses of 8-30 Gy is the current standard for painful vertebral metastases; however, the median pain response is short with 3–6 months and local tumor control is limited with these rather low irradiation doses. Recent advances in radiotherapy technology – intensity modulated radiotherapy for generation of highly conformal dose distributions and image-guidance for precise treatment delivery – have made dose-escalated radiosurgery of spinal metastases possible and early results of pain and local tumor control are promising. The current study will investigate efficacy and safety of radiosurgery for painful vertebral metastases and three characteristics will distinguish this study. 1) A prognostic score for overall survival will be used for selection of patients with longer life expectancy to allow for analysis of long-term efficacy and safety. 2) Fractionated radiosurgery will be performed with the number of treatment fractions adjusted to either good (10 fractions) or intermediate (5 fractions) life expectancy. Fractionation will allow inclusion of tumors immediately abutting the spinal cord due to higher biological effective doses at the tumor - spinal cord interface compared to single fraction treatment. 3) Dose intensification will be performed in the involved parts of the vertebrae only, while uninvolved parts are treated with conventional doses using the simultaneous integrated boost concept. Methods / Design It is the study hypothesis that hypo-fractionated image-guided radiosurgery significantly improves pain relief compared to historic data of conventionally fractionated radiotherapy. Primary endpoint is pain response 3 months after radiosurgery, which is defined as pain reduction of ≥2 points at the treated vertebral site on the 0 to 10 Visual Analogue Scale. 60 patients will be included into this two-centre phase II trial. Conclusions Results of this study will refine the methods of patient selection, target volume definition, treatment planning and delivery as well as quality assurance for radiosurgery. It is the intention of this study to form the basis for a future randomized controlled trial comparing conventional radiotherapy with fractionated radiosurgery for palliation of painful vertebral metastases. Aberrations in gene expression are a hallmark of cancer cells. Differential tumor-specific transcript levels of single genes or whole sets of genes may be critical for the neoplastic phenotype and important for therapeutic considerations or useful as biomarkers. As an approach to filter out such relevant expression differences from the plethora of changes noted in global expression profiling studies, we searched for changes of gene expression levels that are conserved. Transcriptomes from massive parallel sequencing of different types of melanoma from medaka were generated and compared to microarray datasets from zebrafish and human melanoma. This revealed molecular conservation at various levels between fish models and human tumors providing a useful strategy for identifying expression signatures strongly associated with disease phenotypes and uncovering new melanoma molecules.
In this study, a double-donor concept is used to improve the performance of thermally evaporated merocyanine(s)/C60 bulk heterojunction (BHJ) solar cells. It is shown that the co-evaporation of two merocyanine dyes with absorption bands at ~ 500 nm (SW dye) and ~ 650 nm (LW dye), respectively, together with C60 fullerene results in an improvement of open-circuit voltage (VOC), short-circuit current (JSC) as well as total power conversion efficiency (PCE) compared to the best single-donor cell. The enhancement of JSC is attributed to a higher photon harvesting efficiency of the mixed-donor devices due to a better spectral coverage.
Zusammenfassend lässt sich festhalten, dass die in dieser Abreit vorgestellten Squaraine herausragend gute NIR-Absorptions- und NIR-Emissionseigenschaften aufweisen, die sie für zahlreiche Anwendungen interessant machen. Darüber hinaus konnte gezeigt werden, dass ihre besondere cis-Konfiguration und ihr daraus resultierendes Dipolmoment zu vorteilhaften Anordnungen in dünnen Filmen und in Blends mit PCBM führen. Diese Strukturen zeigen für dipolare Moleküle beeindruckende Exzitonen- und Ladungstransporteigenschaften, die vielversprechende Anwendungen in der organischen Elektronik wie in hier untersuchten lösungsprozessierten BHJ-Solarzellen oder auch in OFETs erwarten lassen.
Das Ziel der Studie war es, den vorbeschriebenen Befund des P50-Gating-Defizits bei Schizophrenie, insbesondere die Unterschiede zwischen den verschiedenen Subgruppen nach Leonhard zu replizieren und darüber hinaus diejenigen kortikalen Areale zu detektieren, die während Bedingungen gesteigerten sensorischen Gatings mit signifikanter Aktivierung reagieren. Ferner sollten mögliche Differenzen im Muster kortikaler Aktivierung zwischen gesunden Kontrollen und Patienten aufgedeckt werden, um das kortikale Substrat defizitären sensorischen Gatings zu ermitteln.
Background: Panic disorder is common (5% prevalence) and females are twice as likely to be affected as males. The heritable component of panic disorder is estimated at 48%. Glutamic acid dehydrogenase GAD1, the key enzyme for the synthesis of the inhibitory and anxiolytic neurotransmitter GABA, is supposed to influence various mental disorders, including mood and anxiety disorders. In a recent association study in depression, which is highly comorbid with panic disorder, GAD1 risk allele associations were restricted to females. Methodology/Principal Findings: Nineteen single nucleotide polymorphisms (SNPs) tagging the common variation in GAD1 were genotyped in two independent gender and age matched case-control samples (discovery sample n = 478; replication sample n = 584). Thirteen SNPs passed quality control and were examined for gender-specific enrichment of risk alleles associated with panic disorder by using logistic regression including a genotype6gender interaction term. The latter was found to be nominally significant for four SNPs (rs1978340, rs3762555, rs3749034, rs2241165) in the discovery sample; of note, the respective minor/risk alleles were associated with panic disorder only in females. These findings were not confirmed in the replication sample; however, the genotype6gender interaction of rs3749034 remained significant in the combined sample. Furthermore, this polymorphism showed a nominally significant association with the Agoraphobic Cognitions Questionnaire sum score. Conclusions/Significance: The present study represents the first systematic evaluation of gender-specific enrichment of risk alleles of the common SNP variation in the panic disorder candidate gene GAD1. Our tentative results provide a possible explanation for the higher susceptibility of females to panic disorder.
Numerous studies have shown that humans automatically react with congruent facial reactions, i.e., facial mimicry, when seeing a vis-á-vis’ facial expressions. The current experiment is the first investigating the neuronal structures responsible for differences in the occurrence of such facial mimicry reactions by simultaneously measuring BOLD and facial EMG in an MRI scanner. Therefore, 20 female students viewed emotional facial expressions (happy, sad, and angry) of male and female avatar characters. During picture presentation, the BOLD signal as well as M. zygomaticus major and M. corrugator supercilii activity were recorded simultaneously. Results show prototypical patterns of facial mimicry after correction for MR-related artifacts: enhanced M. zygomaticus major activity in response to happy and enhanced M. corrugator supercilii activity in response to sad and angry expressions. Regression analyses show that these congruent facial reactions correlate significantly with activations in the IFG, SMA, and cerebellum. Stronger zygomaticus reactions to happy faces were further associated to increased activities in the caudate, MTG, and PCC. Corrugator reactions to angry expressions were further correlated with the hippocampus, insula, and STS. Results are discussed in relation to core and extended models of the mirror neuron system (MNS).
Lyme disease Borreliae are highly dependent on the uptake of nutrients provided by their hosts. Our study describes the identification of a 36 kDa protein that functions as putative dicarboxylate-specific porin in the outer membrane of Lyme disease Borrelia. The protein was purified by hydroxyapatite chromatography from Borrelia burgdorferi B31 and designated as DipA, for dicarboxylate-specific porin A. DipA was partially sequenced, and corresponding genes were identified in the genomes of B. burgdorferi B31, Borrelia garinii PBi and Borrelia afzelii PKo. DipA exhibits high homology to the Oms38 porins of relapsing fever Borreliae. B. burgdorferi DipA was characterized using the black lipid bilayer assay. The protein has a singlechannel conductance of 50 pS in 1 M KCl, is slightly selective for anions with a permeability ratio for cations over anions of 0.57 in KCl and is not voltage-dependent. The channel could be partly blocked by different di- and tricarboxylic anions. Particular high stability constants up to about 28,000 l/mol (in 0.1 M KCl) were obtained among the 11 tested anions for oxaloacetate, 2-oxoglutarate and citrate. The results imply that DipA forms a porin specific for dicarboxylates which may play an important role for the uptake of specific nutrients in different Borrelia species.
Background: Severe brain edema is observed in a number of patients suffering from subarachnoid hemorrhage (SAH). Little is known about its pathogenesis and time-course in the first hours after SAH. This study was performed to investigate the development of brain edema and its correlation with brain perfusion after experimental SAH. Methods: Male Sprague–Dawley rats, randomly assigned to one of six groups (n = 8), were subjected to SAH using the endovascular filament model or underwent a sham operation. Animals were sacrificed 15, 30, 60, 180 or 360 minutes after SAH. Intracranial pressure (ICP), mean arterial blood pressure (MABP), cerebral perfusion pressure (CPP) and bilateral local cerebral blood flow (LCBF) were continuously measured. Brain water content (BWC) was determined by the wet/dry-weight method. Results: After SAH, CPP and LCBF rapidly decreased. The decline of LCBF markedly exceeded the decline of CPP and persisted until the end of the observation period. BWC continuously increased. A significant correlation was observed between the BWC and the extent of the perfusion deficit in animals sacrificed after 180 and 360 minutes. Conclusions: The significant correlation with the perfusion deficit after SAH suggests that the development of brain edema is related to the extent of ischemia and acute vasoconstriction in the first hours after SAH.
In effector T and B cells immune receptor signals induce within minutes a rise of intracellular Ca++, the activation of the phosphatase calcineurin and the translocation of NFAT transcription factors from cytosol to nucleus. In addition to this first wave of NFAT activation, in a second step the occurrence of NFATc1/αA, a short isoform of NFATc1, is strongly induced. Upon primary stimulation of lymphocytes the induction of NFATc1/αA takes place during the G1 phase of cell cycle. Due to an auto-regulatory feedback circuit high levels of NFATc1/αA are kept constant during persistent immune receptor stimulation. Contrary to NFATc2 and further NFATc proteins which dampen lymphocyte proliferation, induce anergy and enhance activation induced cell death (AICD), NFATc1/αA supports antigenmediated proliferation and protects lymphocytes against rapid AICD. Whereas high concentrations of NFATc1/αA can also lead to apoptosis, in collaboration with NF-κB-inducing co-stimulatory signals they support the survival of mature lymphocytes in late phases after their activation. However, if dysregulated, NFATc1/αA appears to contribute to lymphoma genesis and – as we assume – to further disorders of the lymphoid system. While the molecular details of NFATc1/αA action and its contribution to lymphoid disorders have to be investigated, NFATc1/αA differs in its generation and function markedly from all the other NFAT proteins which are expressed in lymphoid cells. Therefore, it represents a prime target for causal therapies of immune disorders in future.
Brain–computer interfaces (BCI) based on event-related potentials (ERP) allow for selection of characters from a visually presented character-matrix and thus provide a communica- tion channel for users with neurodegenerative disease. Although they have been topic of research for more than 20 years and were multiply proven to be a reliable communication method, BCIs are almost exclusively used in experimental settings, handled by qualified experts. This study investigates if ERP–BCIs can be handled independently by laymen without expert support, which is inevitable for establishing BCIs in end-user’s daily life situations. Furthermore we compared the classic character-by-character text entry against a predictive text entry (PTE) that directly incorporates predictive text into the character- matrix. N = 19 BCI novices handled a user-centered ERP–BCI application on their own without expert support. The software individually adjusted classifier weights and control parameters in the background, invisible to the user (auto-calibration). All participants were able to operate the software on their own and to twice correctly spell a sentence with the auto-calibrated classifier (once with PTE, once without). Our PTE increased spelling speed and, importantly, did not reduce accuracy. In sum, this study demonstrates feasi- bility of auto-calibrating ERP–BCI use, independently by laymen and the strong benefit of integrating predictive text directly into the character-matrix.
Background: A solid diagnosis of sleep disorders in children should include both self-ratings and parent ratings. However, there are few standardized self-assessment instruments to meet this need. The Children’s Sleep Comic is an adapted version of the unpublished German questionnaire “Freiburger Kinderschlafcomic” and provides pictures for items and responses. Because the drawings were outdated and allowed only for qualitative analysis, we revised the comic, tested its applicability in a target sample, and suggest a procedure for quantitative analysis. Methods: All items were updated and pictures were newly drawn. We used a sample of 201 children aged 5–10 years to test the applicability of the Children’s Sleep Comic in young children and to run a preliminary analysis. Results: The Children’s Sleep Comic comprises 37 items covering relevant aspects of sleep disorders in children. Application took on average 30 minutes. The procedure was well accepted by the children, as reflected by the absence of any dropouts. First comparisons with established questionnaires indicated moderate correlations. Conclusion: The Children’s Sleep Comic is appropriate for screening sleep behavior and sleep problems in children. The interactive procedure can foster a good relationship between the investigator and the child, and thus establish the basis for successful intervention if necessary.
Background: To analyze long-term results of radiotherapy alone for stage I-III low grade follicular lymphoma and to compare outcome after extended field irradiation (EFI) and total nodal irradiation (TNI). Methods and materials: Between 1982 and 2007, 107 patients were treated with radiotherapy alone for low grade follicular lymphoma at Ann Arbor stage I (n = 50), II (n = 36) and III (n = 21); 48 and 59 patients were treated with EFI and TNI, respectively. The median total dose in the first treatment series of the diaphragmatic side with larger lymphoma burden was 38 Gy (25 Gy – 50 Gy) and after an interval of median 30 days, a total dose of 28 Gy (12.6 Gy – 45 Gy) was given in the second treatment series completing TNI. Results: After a median follow-up of 14 years for living patients, 10-years and 15-years overall survival (OS) were 64% and 50%, respectively. Survival was not significantly different between stages I, II and III. TNI and EFI resulted in 15-years OS of 65% and 34% but patients treated with TNI were younger, had better performance status and higher stage of disease compared to patients treated with EFI. In multivariate analysis, only age at diagnosis (p<0.001, relative risk [RR] 1.06) and Karnofsky performance status (p = 0.04, RR = 0.96) were significantly correlated with OS. Freedom from progression (FFP) was 58% and 56% after 10-years and 15-years, respectively. Recurrences outside the irradiated volume were significantly reduced after TNI compared to EFI; however, increased rates of in-field recurrences and extra-nodal out-of-field recurrence counterbalanced this effect resulting in no significant difference in FFP between TNI and EFI. In univariate analysis, FFP was significantly improved in stage I compared to stage II but no differences were observed between stages I/II and stage III. In multivariate analysis no patient or treatment parameter was correlated with FFP. Acute toxicity was significantly increased after TNI compared to EFI with a trend to increased late toxicity as well. Conclusions: Radiotherapy alone for stage I and II follicular lymphoma resulted in long-term OS with high rates of disease control; no benefit of TNI over EFI was observed. For stage III follicular lymphoma, TNI achieved promising OS and FFP and should be considered as a potentially curative treatment option.
Upon synthesis, nascent polypeptide chains are subject to major rearrangements of their side chains to obtain an energetically more favorable conformation in a process called folding. About one third of all cellular proteins pass through the secretory pathway and undergo oxidative folding in the endoplasmic reticulum (ER). During oxidative folding, the conformational rearrangements are accompanied by the formation of disulfide bonds – covalent bonds between cysteine side chains that form upon oxidation. Protein disulfide isomerase (PDI) assists in the folding of substrates by catalyzing the oxidation of pairs of cysteine residues and the isomerization of disulfide bonds as well as by acting as chaperones. In addition to PDI itself, a family of related ER-resident proteins has formed. All PDI family members share the thioredoxin fold in at least one of their domains and exhibit a subset of the PDI activities. Despite many studies, the role of most PDI family members remains unclear. The project presented in this thesis was aimed to establish tools for the biochemical characterization of single members of the PDI family and their role in the folding process. A combination of fluorescence based assays was developed to selectively study single functions of PDI family members and relate their properties of either catalysis of oxidation or catalysis of isomerization or chaperone activity to the rest of the protein family. A binding assay using isothermal titration calorimetry (ITC) was established to complement the activity assays. Using ITC we could show for the first time that members of the PDI family can distinguish between folded and unfolded proteins selectively binding the latter. The unique information provided by this method also revealed a two-site binding of unfolded proteins by PDI itself. In addition to the functional characterization, experiments were conducted to further investigate the oligomeric state of PDI. We could show that the equilibrium between structurally different states of PDI is heavily influenced by the redox state of the protein and its environment. This new data could help to further our understanding of the interplay between oxidases like PDI and their regenerative enzymes like Ero1, which may be governed by structural changes in response to the change in redox status. Another structural approach was the screening of all investigated PDI family members for suitable crystallization conditions. As a result of this screening we could obtain protein crystals of human ERp27 and were able to solve the structure of this protein with X-ray crystallography. The structure gives insight into the mechanisms of substrate binding domains within the PDI family and helps to understand the interaction of ERp27 with the redox active ERp57. In collaboration with the group of Heike Hermanns we could further show the physiological importance of this interaction under oxidative stress. In conclusion, the project presented in this thesis provides novel tools for an extensive analysis of the activities of single PDI family members as well as a useful set of methods to characterize novel oxidoreductases and chaperones. The initial results obtained with the our novel methods are very promising. At the same time, the structural approach of this project could successfully solve the structure of a PDI family member and give information about the interplay within the PDI family.
Intrazerebrale stereotaktische Eingriffe werden zu einem großen Teil ohne direkte Sichtkontrolle durchgeführt. Ein Operateur muss sich deshalb bei der räumlichen Festlegung von Strukturen und beim Anfahren dieser Strukturen auf Hilfsmittel wie stereotaktische Geräte und auf Atlanten, über welche die stereotaktischen Geräte gesteuert werden, verlassen. Trotz großer Fortschritte bei den bildgebenden Verfahren während der letzten dreißig Jahre, ist es gegenwärtig noch nicht möglich, zuverlässig alle subkortikalen Strukturen mit computertomographischen (CT) oder magnetresonanztomographischen (MRT) zu identifizieren oder begrenzen. Eine ganze Reihe zytoarchitektonischer beziehungsweise immunhistochemischer Atlanten wurde veröffentlicht. Dennoch ist es nicht gelungen, die Ergebnisse und Abbildungen dieser Atlanten mit bildgebenden Verfahren bis in die gewünschten Details zu kombinieren, um auf diese Weise das immer noch geringe Auflösungsvermögen radiologischer Methoden zu erhöhen. Deformationen bei der Gewebsentnahme des Gehirns, bei der anschließenden Einbettung, bei der alkoholischen Dehydrierung des Gewebes, Verformungen beim Schneiden und Färben der Schnitte überfordern selbst hoch komplexe mathematische Verfahren und Algorithmen beim Versuch, zytoarchitektonische und immunhistochemische Schnitte mit der gewünschten Präzision den radiologischen Ergebnissen und Bildern und damit indirekt auch den Verhältnissen in vivo anzupassen. Als Alternative verwendeten wir ungewöhnlich dicke (350 – 440 µm) Gallozyanin- (Nissl) gefärbte Serienschnitte durch die Gehirne (ZNS) von drei Personen im Alter von 56, 68 und 36 Jahren. Bei einem Fall wurde das ZNS post mortem mit einem Kernspintomographen vor der Entnahme gescannt. Die Serienschnitte durch dieses Gehirn und das eines zweiten und dritten nicht-gescannten Falles wurden mit Gallozyanin gefärbt, die zytoarchitektonischen Grenzen des Thalamuskomplexes und seiner Unterkerne wurden nach Hassler (1982) identifiziert, jede ihrer Grenzen mit dem Cursor eines Graphiktabletts umfahren und die Gestalt des Thalamuskomplexes und seiner Unterkerne mit Hilfe von Photoshop CS5® und eines computergestützten 3D-Rekonstruktionsprogramms (Amira®) dargestellt. Im Fall 3 ließen sich nach Dunkelfeldbeleuchten die Verteilung markhaltiger Fasern studieren und die zytoarchitektonischen mit myeloarchitektonischen Befunden erweitern und ergänzen. Zusätzlich konnten im Fall 1 die histologischen Serienschnitte und ihre 3D Rekonstruktion mit dem post mortem in cranio MRT registriert werden. Insgesamt kann dieser methodische Ansatz als eine robuste und relativ einfache wenn auch mit umfangreicherer manueller Tätigkeit verbundene Technik zur sehr detailreichen unverformten Korrelation zytoarchitektonischer und kernspinotomographsicher Darstellung des Thalamuskomplexus und seiner Unterkerne angesehen werden. Sie könnte als Grundlage für die Herausgabe eines multimedialen 3D stereotaktischen Atlas des menschlichen Gehirns dienen.
Spin-Bahn-Kopplung in Grenzschichten: Mikroskopische Zusammenhänge und Strategien zur Manipulation

Die vorliegende Arbeit befasst sich mit dem Einfluss der Spin-Bahn-Kopplung (SBK) auf die zweidimensionale elektronische Struktur von Festkörperoberflächen und -grenzflächen. Aufgrund der strukturellen Inversionsasymmetrie kann die SBK in derartigen Systemen eine Spinaufspaltung der elektronischen Zustände herbeiführen und eine charakteristische impulsabhängige Spinstruktur induzieren (Rashba-Effekt). Die Studien in dieser Arbeit sind zum einen darauf gerichtet, das physikalische Verständnis der mikroskopischen Zusammenhänge, die die Spinaufspaltung und die Spinorientierung elektronischer Zustände an Grenzflächen bestimmen, zu verbessern. Des Weiteren sollen Möglichkeiten zur Manipulation der SBK durch kontrollierte Variationen chemischer und struktureller Grenzflächenparameter erforscht werden. Als Modellsysteme für diese Fragestellungen dienen die isostrukturellen Oberflächenlegierungen BiCu2 und BiAg2, deren elektronische Struktur mittels winkelaufgelöster Photoelektronenspektroskopie (ARPES) und spinaufgelöster ARPES untersucht wird. Die Resultate der Experimente werden mithilfe von ab initio-Rechnungen und einfacheren Modellbetrachtungen interpretiert. Die Arbeit schließt mit einer ausblickenden Präsentation von Experimenten zu dem topologischen Isolator Bi2Se3(0001). Vergleichende ARPES-Messungen zu BiAg2/Ag(111) und BiCu2/Cu(111) zeigen, dass bereits geringe Unterschiede in der Grenzschichtmorphologie die Größe der Spinaufspaltung in der elektronischen Struktur um ein Vielfaches verändern können. Zudem belegen spinaufgelöste Experimente eine invertierte Spinorientierung der elektronischen Zustände in BiCu2 im Vergleich mit dem Referenzsystem Au(111). Beide Resultate können durch eine theoretische Analyse des Potentialprofils und der elektronischen Ladungsverteilung senkrecht zu der Grenzfläche in Kombination mit einfachen Modellbetrachtungen verstanden werden. Es stellt sich heraus, dass Asymmetrien in der Ladungsverteilung das direkte mikroskopische Bindeglied zwischen der Spinstruktur des elektronischen Systems und den strukturellen und chemischen Parametern der Grenzschicht bilden. Weitergehende ARPES-Experimente zeigen, dass die spinabhängige elektronische Struktur zudem signifikant durch die Symmetrie des Potentials parallel zu der Grenzflächenebene beeinflusst wird. Eine Manipulation der SBK wird in BiCu2 durch die Deposition von Adatomen erreicht. Hierdurch gelingt es, die Spinaufspaltung sowohl zu vergrößern (Na-Adsorption) als auch zu verringern (Xe-Adsorption). ARPES-Experimente an dem ternären Schichtsystem BiAg2/Ag/Au(111) belegen erstmalig eine Kopplung zwischen elektronischen Bändern mit entgegengesetztem Spincharakter in einem zweidimensionalen System mit Spinaufspaltung (Interband-Spin-Bahn-Kopplung). Der zugrundeliegende Kopplungsmechanismus steht in bemerkenswerter Analogie zu den Auswirkungen der SBK auf die spinpolarisierte elektronische Struktur in ferromagnetischen Systemen. Variationen in der Schichtdicke des Ag-Substratfilms erlauben es, die Stärke der Interband-SBK zu manipulieren.
Background: To analyze the accuracy and inter-observer variability of image-guidance (IG) using 3D or 4D cone-beam CT (CBCT) technology in stereotactic body radiotherapy (SBRT) for lung tumors. Materials and methods: Twenty-one consecutive patients treated with image-guided SBRT for primary and secondary lung tumors were basis for this study. A respiration correlated 4D-CT and planning contours served as reference for all IG techniques. Three IG techniques were performed independently by three radiation oncologists (ROs) and three radiotherapy technicians (RTTs). Image-guidance using respiration correlated 4D-CBCT (IG-4D) with automatic registration of the planning 4D-CT and the verification 4D-CBCT was considered gold-standard. Results were compared with two IG techniques using 3D-CBCT: 1) manual registration of the planning internal target volume (ITV) contour and the motion blurred tumor in the 3D-CBCT (IG-ITV); 2) automatic registration of the planning reference CT image and the verification 3D-CBCT (IG-3D). Image quality of 3D-CBCT and 4D-CBCT images was scored on a scale of 1–3, with 1 being best and 3 being worst quality for visual verification of the IGRT results. Results: Image quality was scored significantly worse for 3D-CBCT compared to 4D-CBCT: the worst score of 3 was given in 19 % and 7.1 % observations, respectively. Significant differences in target localization were observed between 4D-CBCT and 3D-CBCT based IG: compared to the reference of IG-4D, tumor positions differed by 1.9 mm± 0.9 mm (3D vector) on average using IG-ITV and by 3.6 mm± 3.2 mm using IG-3D; results of IG-ITV were significantly closer to the reference IG-4D compared to IG-3D. Differences between the 4D-CBCT and 3D-CBCT techniques increased significantly with larger motion amplitude of the tumor; analogously, differences increased with worse 3D-CBCT image quality scores. Inter-observer variability was largest in SI direction and was significantly larger in IG using 3D-CBCT compared to 4D-CBCT: 0.6 mm versus 1.5 mm (one standard deviation). Inter-observer variability was not different between the three ROs compared to the three RTTs. Conclusions: Respiration correlated 4D-CBCT improves the accuracy of image-guidance by more precise target localization in the presence of breathing induced target motion and by reduced inter-observer variability.
Background: To investigate geometric and dosimetric accuracy of frame-less image-guided radiosurgery (IG-RS) for brain metastases. Methods and materials: Single fraction IG-RS was practiced in 72 patients with 98 brain metastases. Patient positioning and immobilization used either double- (n = 71) or single-layer (n = 27) thermoplastic masks. Pre-treatment set-up errors (n = 98) were evaluated with cone-beam CT (CBCT) based image-guidance (IG) and were corrected in six degrees of freedom without an action level. CBCT imaging after treatment measured intra-fractional errors (n = 64). Pre- and posttreatment errors were simulated in the treatment planning system and target coverage and dose conformity were evaluated. Three scenarios of 0 mm, 1 mm and 2 mm GTV-to-PTV (gross tumor volume, planning target volume) safety margins (SM) were simulated. Results: Errors prior to IG were 3.9 mm± 1.7 mm (3D vector) and the maximum rotational error was 1.7° ± 0.8° on average. The post-treatment 3D error was 0.9 mm± 0.6 mm. No differences between double- and single-layer masks were observed. Intra-fractional errors were significantly correlated with the total treatment time with 0.7mm±0.5mm and 1.2mm±0.7mm for treatment times ≤23 minutes and >23 minutes (p<0.01), respectively. Simulation of RS without image-guidance reduced target coverage and conformity to 75% ± 19% and 60% ± 25% of planned values. Each 3D set-up error of 1 mm decreased target coverage and dose conformity by 6% and 10% on average, respectively, with a large inter-patient variability. Pre-treatment correction of translations only but not rotations did not affect target coverage and conformity. Post-treatment errors reduced target coverage by >5% in 14% of the patients. A 1 mm safety margin fully compensated intra-fractional patient motion. Conclusions: IG-RS with online correction of translational errors achieves high geometric and dosimetric accuracy. Intra-fractional errors decrease target coverage and conformity unless compensated with appropriate safety margins.
Der fast 1200 Jahre bestehende Dom Würzburgs wurde am 16. März 1945 durch den Angriff der alliierten britischen Streitkräfte sehr stark beschädigt. Der Holzdachstuhl sowie viele Einrichtungsgegenstände und Kunstwerke verbrannten, doch die Architektur der Wände und der Gewölbe blieben zunächst erhalten, so dass der Wiederaufbau eine Frage der Denkmalpflege zu sein schien. Durch den Einfluss verschiedener architektonischer und externer Faktoren wurde die Bausubstanz der nördlichen Hochschiffwand derart geschwächt, dass sie am 19. Februar 1946 einstürzte und den Dom endgültig zu einer Ruine werden ließ. Der Wiederaufbau zog sich aufgrund der unterschiedlichen Ansichten der beteiligten Institutionen über mehr als 20 Jahre hin. Die verschiedenen Perspektiven und diskutierten Alternativen werden im Verlauf der vorliegenden Arbeit dargestellt.
Um eine mögliche elektrophysiologische, kardiale Ursache für den plötzlichen Tod von STIM2 Knock-Out Mäusen zu prüfen, wurde eine elektrophysiologische Charakterisierung mittels Ruhe- und Stress-EKG, telemetrischem Langzeit-EKG sowie Elektrophysiologischer Untersuchung durchgeführt. Hierbei konnte keine kardial-elektrophysiologische Grundlage für den plötzlichen Tod dieser Tiere gefunden werden.
Synthese von Dextran-umhüllten Eisenoxid-Nanopartikeln als Kontrastmittel für die MR-Tomographie

Durch Fällung von Eisen(II)- und Eisen(III)-salzen wurden Dextran-umhüllte Eisenoxid-Nanopartikel (SPIOs) und durch anschließende Umsetzung mit Epichlorhydrin und Ammoniak CLIOs gewonnen. An diesen Kolloiden wurden niedermolekulare Moleküle wie Diamine oder Bernsteinsäureanhydrid als Linker angebracht. Ein weiterer Aspekt dieser Arbeit stellt die Anbindung von Fluoreszenzmarkern und Antikörpern an der Partikeloberfläche sowie deren spektroskopische Untersuchung dar.
Background: The 2009 pandemic influenza A (H1N1) (PIA) virus infected large parts of the pediatric population with a wide clinical spectrum and an initially unknown complication rate. The aims of our study were to define clinical characteristics and outcome of pandemic influenza A (H1N1) 2009-associated hospitalizations (PIAH) in children <18 years of age. All hospitalized cases of children <18 years of age with laboratory-confirmed pandemic influenza A (H1N1) 2009 in the region of Wuerzburg (Northern Bavaria, Germany) between July 2009 and March 2010 were identified. For these children a medical chart review was performed to determine their clinical characteristics and complications. Results: Between July 2009 and March 2010, 94 PIAH (62% males) occurred in children <18 years of age, with a median age of 7 years (IQR: 3–12 years). Underlying diseases and predisposing factors were documented in 40 (43%) children; obesity (n = 12, 30%), asthma (n = 10, 25%) and neurologic disorders (n = 8, 20%) were most frequently reported. Sixteen (17%) children received oxygen supplementation; three (3%) children required mechanical ventilation. Six (6%) children were admitted to an intensive care unit, four of them with underlying chronic diseases. Conclusions: Most PIAH demonstrated a benign course of disease. However, six children (6%) needed treatment at an intensive care unit for severe complications.
Background: External beam radiotherapy (EBRT) is the treatment of choice for irresectable meningioma. Due to the strong expression of somatostatin receptors, peptide receptor radionuclide therapy (PRRT) has been used in advanced cases. We assessed the feasibility and tolerability of a combination of both treatment modalities in advanced symptomatic meningioma. Methods: 10 patients with irresectable meningioma were treated with PRRT (177Lu-DOTA0,Tyr3 octreotate or - DOTA0,Tyr3 octreotide) followed by external beam radiotherapy (EBRT). EBRT performed after PRRT was continued over 5–6 weeks in IMRT technique (median dose: 53.0 Gy). All patients were assessed morphologically and by positron emission tomography (PET) before therapy and were restaged after 3–6 months. Side effects were evaluated according to CTCAE 4.0. Results: Median tumor dose achieved by PRRT was 7.2 Gy. During PRRT and EBRT, no side effects>CTCAE grade 2 were noted. All patients reported stabilization or improvement of tumor-associated symptoms, no morphologic tumor progression was observed in MR-imaging (median follow-up: 13.4 months). The median pre-therapeutic SUVmax in the meningiomas was 14.2 (range: 4.3–68.7). All patients with a second PET after combined PRRT + EBRT showed an increase in SUVmax (median: 37%; range: 15%–46%) to a median value of 23.7 (range: 8.0–119.0; 7 patients) while PET-estimated volume generally decreased to 81 ± 21% of the initial volume. Conclusions: The combination of PRRT and EBRT is feasible and well tolerated. This approach represents an attractive strategy for the treatment of recurring or progressive symptomatic meningioma, which should be further evaluated.
Dendritic cells (DCs) are major players in the control of adaptive tolerance and immunity. Therefore, their specific generation and adoptive transfer into patients or their in vivo targeting is attractive for clinical applications. While injections of mature immunogenic DCs are tested in clinical trials, tolerogenic DCs still are awaiting this step. Besides the tolerogenic potential of immature DCs, also semi-mature DCs can show tolerogenic activity but both types also bear unfavorable features. Optimal tolerogenic DCs, their molecular tool bar, and their use for specific diseases still have to be defined. Here, the usefulness of in vitro generated and adoptively transferred semi-mature DCs for tolerance induction is outlined. The in vivo targeting of semi-mature DCs as represented by steady state migratory DCs are discussed for treatment of autoimmune diseases and allergies. First clinical trials with transcutaneous allergen application may point to their therapeutic use in the future.
