A search for the Standard Model Higgs boson in the H→WW(⋆)→ℓνℓνH→WW(⋆)→ℓνℓν (ℓ=e,μℓ=e,μ) decay mode is presented. The search is performed using proton–proton collision data corresponding to an integrated luminosity of 4.7 fb\(^{−1}\) at a centre-of-mass energy of 7 TeV collected during 2011 with the ATLAS detector at the Large Hadron Collider. No significant excess of events over the expected background is observed. An upper bound is placed on the Higgs boson production cross section as a function of its mass. A Standard Model Higgs boson with mass in the range between 133 GeV and 261 GeV is excluded at 95% confidence level, while the expected exclusion range is from 127 GeV to 233 GeV.
Detailed measurements of the electron performance of the ATLAS detector at the LHC are reported, using decays of the Z, W and J/ψ particles. Data collected in 2010 at s√=7 TeV are used, corresponding to an integrated luminosity of almost 40 pb\(^{−1}\). The inter-alignment of the inner detector and the electromagnetic calorimeter, the determination of the electron energy scale and resolution, and the performance in terms of response uniformity and linearity are discussed. The electron identification, reconstruction and trigger efficiencies, as well as the charge misidentification probability, are also presented.
Proton–proton collisions at √s=7 TeV and heavy ion collisions at \(\sqrt{sNN}\)=2.76 TeV were produced by the LHC and recorded using the ATLAS experiment’s trigger system in 2010. The LHC is designed with a maximum bunch crossing rate of 40 MHz and the ATLAS trigger system is designed to record approximately 200 of these per second. The trigger system selects events by rapidly identifying signatures of muon, electron, photon, tau lepton, jet, and B meson candidates, as well as using global event signatures, such as missing transverse energy. An overview of the ATLAS trigger system, the evolution of the system during 2010 and the performance of the trigger system components and selections based on the 2010 collision data are shown. A brief outline of plans for the trigger system in 2011 is presented.
Using inelastic proton-proton interactions at s√=900 GeV and 7 TeV, recorded by the ATLAS detector at the LHC, measurements have been made of the correlations between forward and backward charged-particle multiplicities and, for the first time, between forward and backward charged-particle summed transverse momentum. In addition, jet-like structure in the events is studied by means of azimuthal distributions of charged particles relative to the charged particle with highest transverse momentum in a selected kinematic region of the event. The results are compared with predictions from tunes of the pythia and herwig++ Monte Carlo generators, which in most cases are found to provide a reasonable description of the data.
Diazepinomicin is a dibenzodiazepine alkaloid with an unusual structure among the known microbial metabolites discovered so far. Diazepinomicin was isolated from the marine sponge-associated strain Micromonospora sp. RV115 and was identified by spectroscopic analysis and by comparison to literature data. In addition to its interesting preclinical broad-spectrum antitumor potential, we report here new antioxidant and anti-protease activities for this compound. Using the ferric reducing antioxidant power (FRAP) assay, a strong antioxidant potential of diazepinomicin was demonstrated. Moreover, diazepinomicin showed a significant antioxidant and protective capacity from genomic damage induced by the reactive oxygen species hydrogen peroxide in human kidney (HK-2) and human promyelocytic (HL-60) cell lines. Additionally, diazepinomicin inhibited the proteases rhodesain and cathepsin L at an IC50 of 70–90 μM. It also showed antiparasitic activity against trypomastigote forms of Trypanosoma brucei with an IC50 of 13.5 μM. These results showed unprecedented antioxidant and anti-protease activities of diazepinomicin, thus further highlighting its potential as a future drug candidate.
Nach Einschätzung der Weltgesundheitsorganisation WHO wird Krebs im Jahr 2013 die weltweit häufigste Todesursache bei Menschen und Haustieren sein. Diese Situation erfordert die Entwicklung neuer therapeutischer Ansätze. Hauptziel einer Tumortherapie ist es, sowohl den Primärtumor als auch die Metastasen möglichst vollständig zu entfernen. Dabei wird nach Methoden gesucht, die im Gegensatz zu den meisten gegenwärtigen therapeutischen Einsätzen, wie der chirurgischen Entfernung bösartiger Neubildungen, Chemotherapie und Strahlentherapie, selektiv die bösartigen Zellen erkennen und zerstören können. Eine faszinierende Möglichkeit in dieser Hinsicht ist die Verwendung von onkolytischen Viren, die die Fähigkeit besitzen, sich selektiv sowohl in Primärtumoren als auch in Metastasen anzusiedeln und die Krebszellen dort zu zerstören. Das Konzept, dass Viren nützlich für die Bekämpfung von Krebs sein könnten, ist nicht neu. Allerdings konnte erst in den letzten Jahren durch zahlreiche Studien bestätigt werden, dass verschiedene Viren in der Lage sind, eine signifikante Antitumorwirkung in vivo auszuüben. Zu den erfolgversprechenden onkolytischen Viren zählen insbesondere Adenovirus, Herpes simplex Virus, Reovirus und Vaccinia-Virus, die sich bereits in Phase III der klinischen Studien befinden oder kurz davor sind. Die therapeutische Nutzung von tumorspezifischen onkolytischen Viren beim Menschen hat bereits begonnen. Im Rahmen der vorliegenden Doktorarbeit wurden verschiedene Aspekte der Wirkungsweise von Vaccinia-Virus-Stämmen bei der Therapie verschiedener Tumore aus Mensch und Hund im Xenotransplantat-Mausmodell bearbeitet: die Onkolyse der Krebszellen und Inhibition des Tumorwachstums sowie die Effekte der Virusinfektion auf das Tumormikromilieu und die Mitwirkung des angeborenen Immunsystems bei der Virotherapie. Das Tumormikromilieu (Stroma) setzt sich aus einer Vielzahl verschiedener Zellen und Komponenten der extrazellulären Matrix zusammen. Die Krebszellen bilden unter anderem mit Endothelzellen des Blut- und Lymphsystems und verschiedenen Immunzellen eine komplexe Organ-ähnliche Struktur. Weitere wichtige Bestandteile des Stromas sind Wachstumsfaktoren, Chemokine und Zytokine und die Tumorvaskulatur. Diese ist durch zahlreiche strukturelle und funktionelle Abnormalitäten charakterisiert, wodurch die Effektivität von Strahlen- und Chemotherapie herabgesetzt wird. Weiterhin ist das Tumormikromilieu durch seine Ähnlichkeit mit einer chronischen Entzündungsreaktion gekennzeichnet und wirkt immunsupprimierend auf rekrutierte Leukozyten, die wiederum die Inflammation verstärken und die Angiogenese und das Tumorwachstum weiter fördern. Aufgrund dieser vielen Komponenten ist die Zusammensetzung jedes Tumors einzigartig, weswegen Standardtherapien häufig nicht zu einer Heilung führen. Die Wirkung der Viren bei der Virotherapie beruht vermutlich auf 4 Mechanismen, die einzeln oder in Kombination auftreten können: die direkte Onkolyse der Krebszellen, die Zerstörung des Tumorblutgefäßsystems, die Aktivierung des Immunsystems des Wirts und die Suppression der microRNA-Expression des Wirtes. Zusätzlich kann die Expression therapeutischer Gene die onkolytische Wirkung verstärken. Zum Nachweis der Onkolyse der Krebszellen und Inhibition des Tumorwachstums wurde zuerst das Virus GLV-1h68 in einem autologen humanen Melanomzellpaar, 888-MEL und 1936-MEL, eingesetzt. Das GLV-1h68-Virus wurde auf Basis des Wildtyp Vaccinia-Virus LIVP durch die Insertion von 3 Expressionskassetten in den drei Genloci F14.5L, J2R und A56 genetisch konstruiert. 888-MEL, eine zu einem frühen Zeitpunkt der Krebserkrankung aus einer Metastase isolierte Zelllinie, zeigt nach Infektion mit GLV-1h68 im Mausmodell Tumornekrose („Responder“), während 1936-MEL aus einer späten Metastasierungsphase kaum mit Onkolyse auf eine Virusinfektion reagiert („Poor-Responder“). Die onkolytische Wirkung konnte mittels Durchflusszytometrie in Tumoren beider Zelllinien zu einem frühen Zeitpunkt nach Virusinfektion nachgewiesen werden. In 888-MEL-Tumoren wurde hierbei eine große Zahl infizierter und toter Zellen nach Virusinfektion gefunden. Gleichzeitig wurde eine hohe Zahl an Immunzellen detektiert, die nach Virusinfektion reduziert war. In den schwächer reagierenden 1936-MEL-Tumoren konnte eine Onkolyse bei Infektion mit höherer Virusmenge und zu einem früheren Zeitpunkt demonstriert werden, wodurch mehr Zellen infiziert wurden. Zusätzlich wurde eine Steigerung der nur in geringer Zahl vorhandenen Immunzellen nachgewiesen. Trotz des unterschiedlichen Tumormikromilieus konnte somit ein onkolytischer Effekt in beiden Tumormodellen erzielt werden. ...
The high failure rate of new drug candidates in preclinical or clinical studies due to hepatotoxicity represents a considerable problem in the drug development. Hence, there is an urgent need to develop new approaches for early and reliable prediction of drug-induced hepatotoxicity that enables a better identification of drug candidates with high potential for toxicity at early stages of drug development. Therefore, the aim of this work was to improve the prediction of drug-induced liver injury in preclinical studies through evaluation of more reliable and sensitive biomarkers of hepatotoxicity and a better understanding of the underlying mechanistic basis for drug-induced toxicity. First, the ability of a set of potential markers (NGAL, thiostatin, clusterin, PON1) to detect early signs of liver injury was assessed in rats treated with drug candidates that were dropped from further development, in part due to toxic adverse effects in the liver. In summary, PON1 and clusterin were not consistently altered in response to liver injury and thus provide no additive information to the traditional liver enzymes in detecting drug-induced hepatotoxicity. In contrast, thiostatin and NGAL were increased in serum and urine of treated animals in a time- and dose-dependent manner. These changes correlated well with mRNA expression in the target organ and generally reflected the onset and degree of drug-induced liver injury. Receiver-operating characteristics analyses supported serum thiostatin, but not NGAL, as a better indicator of drug-induced hepatobiliary injury than conventional clinical chemistry parameters, such as ALP, ALT and AST. Although thiostatin, an acute phase protein expressed in a range of tissues, may not be specific for liver injury, our results indicate that thiostatin may serve as a sensitive, minimally-invasive diagnostic marker of inflammation and tissue damage in preclinical safety assessment. In the second part of this work, combined application of genomics profiling technology and RNAi to inhibit the pharmacological target of a drug candidate BAY16, a glucagon receptor (GCGR) antagonist, was used to determine if interference with the pharmacological target plays a role in the toxic response to BAY16, and to narrow down those molecular changes that are associated with toxicity, and not the pharmacological action of BAY16. In contrast to Bay 16, which was found to be cytotoxic at concentrations of 75 µM, silencing of the glucagon receptor did not affect cell viability in primary rat hepatocytes. Thus, it can be concluded that hepatotoxicity of Bay 16 was not related to the drugs inhibitory effect on the glucagon receptor in vitro and in vivo. These findings were supported by the fact that most of BAY16-induced changes in gene expression occurred independently of the pharmacological modulation of GCGR. These off-target effects include altered xenobiotic metabolism, oxidative stress, increased fatty acid synthesis, and alterations in cholesterol and bile acid metabolic processes. Although it was not possible to draw a final conclusion about the mechanism of BAY16 hepatotoxicity, changes in these molecular mechanisms appear contribute to progression of hepatic injury. With regard to drug safety assessment in preclinical studies, the utilization of siRNA technology in vitro represents a new approach to improve mechanistic understanding of the nature of drug’s toxicity, being either chemically mediated or due to primary or secondary pharmacological mode of action.
Optimal open-loop control, i.e. the application of an analytically derived control rule, is demonstrated for nanooptical excitations using polarization-shaped laser pulses. Optimal spatial near-field localization in gold nanoprisms and excitation switching is realized by applying a shift to the relative phase of the two polarization components. The achieved near-field switching confirms theoretical predictions, proves the applicability of predefined control rules in nanooptical light–matter interaction and reveals local mode interference to be an important control mechanism.
Background: During early stages of brain development, secreted molecules, components of intracellular signaling pathways and transcriptional regulators act in positive and negative feed-back or feed-forward loops at the mid-hindbrain boundary. These genetic interactions are of central importance for the specification and subsequent development of the adjacent mid-and hindbrain. Much less, however, is known about the regulatory relationship and functional interaction of molecules that are expressed in the tectal anlage after tectal fate specification has taken place and tectal development has commenced.
Results: Here, we provide experimental evidence for reciprocal regulation and subsequent cooperation of the paired-type transcription factors Pax3, Pax7 and the TALE-homeodomain protein Meis2 in the tectal anlage. Using in ovo electroporation of the mesencephalic vesicle of chick embryos we show that (i) Pax3 and Pax7 mutually regulate each other's expression in the mesencephalic vesicle, (ii) Meis2 acts downstream of Pax3/7 and requires balanced expression levels of both proteins, and (iii) Meis2 physically interacts with Pax3 and Pax7. These results extend our previous observation that Meis2 cooperates with Otx2 in tectal development to include Pax3 and Pax7 as Meis2 interacting proteins in the tectal anlage.
Conclusion: The results described here suggest a model in which interdependent regulatory loops involving Pax3 and Pax7 in the dorsal mesencephalic vesicle modulate Meis2 expression. Physical interaction with Meis2 may then confer tectal specificity to a wide range of otherwise broadly expressed transcriptional regulators, including Otx2, Pax3 and Pax7.
Imprinted genes play important roles in brain development. As the neural developmental capabilities of human parthenogenetic embryonic stem cells (hpESCs) with only a maternal genome were not assessed in great detail, hence here the potential of hpESCs to differentiate into various neural subtypes was determined. In addition DNA methylation and expression of imprinted genes upon neural differentiation was also investigated. The results demonstrated that hpESC-derived neural stem cells (hpNSCs) showed expression of NSC markers Sox1, Nestin, Pax6, and Musashi1 (MS1), the silencing of pluripotency genes (Oct4, Nanog) and the absence of activation of neural crest (Snai2, FoxD3) and mesodermal (Acta1) markers. Moreover, confocal images of hpNSC cultures exhibited ubiquitous expression of NSC markers Nestin, Sox1, Sox2 and Vimentin. Differentiating hpNSCs for 28 days generated neural subtypes with neural cell type-specific morphology and expression of neuronal and glial markers, including Tuj1, NeuN, Map2, GFAP, O4, Tau, Synapsin1 and GABA. hpNSCs also responded to region-specific differentiation signals and differentiated into regional phenotypes such as midbrain dopaminergic- and motoneuron-type cells. hpESC-derived neurons showed typical neuronal Na+/K+ currents in voltage clamp mode, elicited multiple action potentials with a maximum frequency of 30 Hz. Cell depicted a typical neuron-like current pattern that responded to selective pharmacological blockers of sodium (tetrodotoxin) and potassium (tetraethylammonium) channels. Furthermore, in hpESCs and hpNSCs the majority of CpGs of the differentially methylated regions (DMRs) KvDMR1 were methylated whereas DMR1 (H19/Igf2 locus) showed partial or complete absence of CpG methylation, which is consistent with a parthenogenetic (PG) origin. Upon differentiation parent-of-origin-specific gene expression was maintained in hpESCs and hpNSCs as demonstrated by imprinted gene expression analyses. Together this shows that despite the lack of a paternal genome, hpNSCs are proficient in differentiating into glial- and neuron-type cells, which exhibit electrical activity similar to newly formed neurons. Moreover, maternal-specific gene expression and imprinting-specific DNA-methylation are largely maintained upon neural differentiation. hpESCs are a means to generate histocompatible and disease allele-free ESCs. Additionally, hpESCs are a unique model to study the influence of imprinting on neurogenesis.
Parent of origin imprints on the genome have been implicated in the regulation of neural cell type differentiation. The ability of human parthenogenetic (PG) embryonic stem cells (hpESCs) to undergo neural lineage and cell type-specific differentiation is undefined. We determined the potential of hpESCs to differentiate into various neural subtypes. Concurrently, we examined DNA methylation and expression status of imprinted genes. Under culture conditions promoting neural differentiation, hpESC-derived neural stem cells (hpNSCs) gave rise to glia and neuron-like cells that expressed subtype-specific markers and generated action potentials. Analysis of imprinting in hpESCs and in hpNSCs revealed that maternal-specific gene expression patterns and imprinting marks were generally maintained in PG cells upon differentiation. Our results demonstrate that despite the lack of a paternal genome, hpESCs generate proliferating NSCs that are capable of differentiation into physiologically functional neuron-like cells and maintain allele-specific expression of imprinted genes. Thus, hpESCs can serve as a model to study the role of maternal and paternal genomes in neural development and to better understand imprinting-associated brain diseases.
This thesis is devoted to numerical verification of optimality conditions for non-convex optimal control problems. In the first part, we are concerned with a-posteriori verification of sufficient optimality conditions. It is a common knowledge that verification of such conditions for general non-convex PDE-constrained optimization problems is very challenging. We propose a method to verify second-order sufficient conditions for a general class of optimal control problem. If the proposed verification method confirms the fulfillment of the sufficient condition then a-posteriori error estimates can be computed. A special ingredient of our method is an error analysis for the Hessian of the underlying optimization problem. We derive conditions under which positive definiteness of the Hessian of the discrete problem implies positive definiteness of the Hessian of the continuous problem. The results are complemented with numerical experiments. In the second part, we investigate adaptive methods for optimal control problems with finitely many control parameters. We analyze a-posteriori error estimates based on verification of second-order sufficient optimality conditions using the method developed in the first part. Reliability and efficiency of the error estimator are shown. We illustrate through numerical experiments, the use of the estimator in guiding adaptive mesh refinement.
5.1 Immuntherapie mit vom Tumorstroma abgeleiteten Peptiden Tumore bestehen nicht nur aus Tumorzellen, sondern auch aus der sie umgebenden extrazellulären Matrix (EZM), und Stromazellen wie Fibroblasten (cancer-associated fibroblast; CAF) und Endothelzellen (tumor endothelial cell; TEC). Diese Stromazellen haben durch die Ausschüttung von Zytokinen, proteolytischen Enzymen, Wachstums- und Angiogenesefaktoren einen entscheidenden Einfluss auf die Tumorprogression. Sie unterscheiden sich von den Stromazellen der normalen Gewebe durch die Expression von sogenannten Tumorstroma-assoziierten Antigenen (TSAA). Damit sollten Therapien, die auf TSAA abzielen, universell einsetzbar und weniger anfällig gegenüber Resistenzentwicklungen (immune escape Mechanismen) sein, da Stromazellen im Gegensatz zu neoplastischen Zellen genetisch relativ stabil sind. Für eine Immuntherapie mit vom Tumorstroma abgeleiteten Peptiden wählten wir die TSAA Endoglin und Fap, welche während der Wundheilung und im Tumorstroma induziert werden. Dabei sollte überprüft werden, ob prophylaktische Vakzinierungen in C57Bl/6j Mäusen Peptid-reaktive T-Zellen induzieren können, und das Wachstum von transplantieren Grm1-transgenen Tumoren reduziert werden kann. In der Tat konnten wir sowohl bei Endoglin- als auch bei Fap Peptid vakzinierten Tieren in vivo Peptid-reaktive Lymphozyten im Blut und zu einem geringeren Anteil auch in der Milz nachweisen, welche Peptid-gepulste syngene Milzzellen lysieren konnten. Allerdings konnte in beiden Fällen keine Reduktion des Tumorwachstums gegenüber der Kontrollgruppe beobachtet werden. Bei der Fap-Peptid-vakzinierten Gruppe war das Tumorwachstum gegenüber der Kontrollgruppe sogar gesteigert. Dies könnte darauf hindeuten, dass die Induktion Fap-Peptid-reaktiver T-Zellen tumorpromovierend wirkt. Möglicherweise könnte aber durch eine Modifikation des Vakzinierungsprotokolls bzw. durch eine Kombination mit anderen Immuntherapeutika ein verbessertes Ansprechen auf eine Endoglin bzw. Fap basierte Immuntherapie erzielt werden. 5.2 Immunsuppressive Mechanismen im Grm1-transgenen Melanom-Modell Grm1-transgene Mäuse entwickeln spontan kutane Melanome. Dieses Modell erlaubte es uns in der vorliegenden Arbeit spontane Immunantworten im Laufe der Melanomentstehung zu untersuchen. Hierfür analysierten wir sowohl ex vivo als auch in vitro aus Milz und Lymphknoten gewonnene Lymphozyten von Mäusen, welche keine Tumorläsionen bzw. eine niedrige oder hohe Tumorlast aufwiesen. Dabei konnten wir ex vivo einen Anstieg der Frequenz aktivierter CD4+ und CD8+ Lymphozyten mit zunehmender Tumorlast zeigen. Bei tumortragenden Tieren exprimierten jedoch hauptsächlich CD4+ T-Zellen Aktivierungsmarker nach in vitro Stimulation. Interessanterweise waren diese Zellen tumortragender Tiere auch funktionell beeinträchtigt, was sich in einer verminderten Proliferationskapazität nach in vitro Stimulation zeigte. Weitere Analysen ergaben, dass die erhöhte Frequenz regulatorischer T Zellen bei tumortragenden Tieren ein frühes Ereignis im Laufe der Tumorentstehung ist. Gleichzeitig konnte auch ein starker Anstieg der immunsupprimierenden Zytokine Tgf-β1 und Il-10 sowohl in den Lymphknoten als auch im Tumorgewebe beobachtet werden. Dabei war die Tgf-β1-Expression sowohl im Tumor als auch im tumor-drainierenden Lymphknoten erhöht, während Il-10 im Tumor nur moderat exprimiert wurde, was eine komplexere Regulation der Il-10-Expression nahe legt. Dies bedeutet, dass in Grm1-transgenen Mäusen ähnlich wie auch bei Melanompatienten zelluläre und zytokinabhängige Mechanismen zur Tumorentstehung beitragen und dieses Modell daher geeignet ist, um präklinisch immunmodulierende Therapieansätze zu testen.
Cellular and cytokine-dependent immunosuppressive mechanisms of grm1-transgenic murine melanoma

Grm1-transgenic mice spontaneously develop cutaneous melanoma. This model allowed us to scrutinize the generic immune responses over the course of melanoma development. To this end, lymphocytes obtained from spleens, unrelated lymph nodes and tumor-draining lymph nodes of mice with no evidence of disease, and low or high tumor burden were analyzed ex vivo and in vitro. Thereby, we could demonstrate an increase in the number of activated CD4\(^+\) and CD8+ lymphocytes in the respective organs with increasing tumor burden. However, mainly CD4\(^+\) T cells, which could constitute both T helper as well as immunosuppressive regulatory T cells, but not CD8\(^+\) T cells, expressed activation markers upon in vitro stimulation when obtained from tumor-bearing mice. Interestingly, these cells from tumor-burdened animals were also functionally hampered in their proliferative response even when subjected to strong in vitro stimulation. Further analyses revealed that the increased frequency of regulatory T cells in tumor-bearing mice is an early event present in all lymphoid organs. Additionally, expression of the immunosuppressive cytokines TGF-β1 and IL-10 became more evident with increased tumor burden. Notably, TGF-β1 is strongly expressed in both the tumor and the tumor-draining lymph node, whereas IL-10 expression is more pronounced in the lymph node, suggesting a more complex regulation of IL-10. Thus, similar to the situation in melanoma patients, both cytokines as well as cellular immune escape mechanisms seem to contribute to the observed immunosuppressed state of tumor-bearing grm1-transgenic mice, suggesting that this model is suitable for preclinical testing of immunomodulatory therapeutics.
Hintergrund: Über die psychische Gesundheit von Asylsuchenden in Deutschland ist wenig bekannt. Ziel dieser Studie ist, (1) die psychische Gesundheit der Asylsuchenden in der Würzburger Gemeinschaftsunterkunft zu beschreiben, (2) ihre Wahrnehmung der aktuellen Lebensbedingungen zu erfassen, sowie (3) mögliche Zusammenhänge zwischen beiden Bereichen zu untersuchen. Methoden: Alle Bewohner der Würzburger Gemeinschaftsunterkunft, welche zum Zeitpunkt der Befragung mindestens 16 Jahre alt waren und den Studienfragebogen in einer der Sprachen Arabisch, Amharisch, Farsi, Russich, Somali, Deutsch oder Englisch ausfüllen konnten, konnten an dieser Querschnittbefragung teilnehmen. Das Vorhandensein von psychischen Erkrankungen (Somatoformes Syndrom, Depressive Syndrome, Angstsyndrome, Alkoholsyndrom und eine Screeningfrage für PTBS), sowie das Ausmaß an psychosozialem Stress wurden mittels des PRIME-MD Patient Health Questionnaire (PHQ) gemessen. Die subjektive Einschätzung der Lebensbedingungen durch die Teilnehmer wurde mit einem für die spezifischen Bedingungen entwickelten Fragebogen erfasst. Die Ergebnisse wurden deskriptiv dargestellt und Korrelationen zwischen der Bewertung der Lebensbedingungen und ausgewählten Parametern der psychischen Gesundheit wurden mittels Chi-Quadrat-Tests und des Spearman Rangkorrelationskoeffizienten berechnet. Ergebnisse: Insgesamt nahmen 183 Bewohner an der Befragung teil. Der PHQ konnte für 140 Teilnehmer ausgewertet werden, der Fragebogen zu aktuellen Lebensbedingungen für 113 Teilnehmer. Die häufigsten PHQ-Syndrome waren das Somatoforme Syndrom (38,6%; n=54), Depressive Syndrome (25,7% (n=36) Major Depressives Syndrom; 22,8% (n=32) andere depressive Syndrome) und Angstsyndrome (11,4% (n=16) Paniksyndrom, 9,3% (n=13) andere Angstsyndrome). Bei 38,6% (n=54) ergab der PHQ für mehr als ein Syndrom ein positives Ergebnis. Die Lebensbedingungen in der Gemeinschaftsunterkunft wurden größtenteils negativ bewertet und ihre Auswirkungen auf die eigene Gesundheit wurden im Mittel als „ziemlich stark“ beurteilt. Eine schlechtere Bewertung der Lebensbedingungen und eine längere Aufenthaltsdauer in der Gemeinschaftsunterkunft waren in univariaten Analysen mit einem schlechteren Ergebnis bezüglich verschiedener Parameter der psychischen Gesundheit assoziiert (z.B. depressive Syndrome, psychosoziale Belastung). Schlussfolgerung: Verschiedene Limitationen der Studie müssen berücksichtigt werden (z.B. Querschnittdesign, mangelnde Validierung der Fragebogenübersetzungen). Dennoch zeigen diese Ergebnisse eine deutliche Unzufriedenheit der Studienteilnehmer mit den Lebensbedingungen in der Gemeinschaftsunterkunft auf und lassen eine hohe Prävalenz psychischer Erkrankungen in der Studienpopulation vermuten.
Tropische Infektionskrankheiten wie Malaria, Leishmaniose oder auch die Afrikanische Trypanosomiase sind aufgrund von zunehmenden Resistenzen der Erreger, globaler Erwärmung, aber auch von Versäumnissen in der Vergangenheit bei der kontinuierlichen Weiterentwicklung bestehender sowie der Erforschung neuer Medikamente auch im 21. Jahrhundert noch eine große Bedrohung für Millionen von Menschen. Die Suche nach neuartigen Wirkstoffen und deren Weiterentwicklung zu potenziellen Medikamenten ist daher zwingend erforderlich. Insbesondere Produkte des Sekundärstoffwechsels wie etwa die Alkaloide bilden wichtige Grundlagen als Leitstrukturen für pharmazeutische Wirkstoffe. Eine solche Klasse phytochemischen Ursprungs sind die Naphthylisochinolin-Alkaloide mit interessanten strukturellen Eigenschaften sowie pharmakologischen Wirksamkeiten. Einige Vertreter zeigen ausgeprägte In-vitro-Aktivitäten gegen protozoische Erreger wie Plasmodien, Leishmanien und Trypanosomen. Besonders die neuartige Unterklasse ionischer N,C-verknüpfter Naphthylisochinolin-Alkaloide, wie z.B. Ancistrocladinium A und Ancistrocladinium B, zeichnen sich durch gute antileishmaniale Wirkungen aus. In Vorarbeiten zeigten erste Studien zu Struktur-Aktivitäts-Beziehungen (SAR-Studien) mit vereinfachten N,C-gekuppelten Arylisochinolinen, dass sich durch gezielte Strukturvariation die Aktivität gegen einen Erreger verbessern lässt. Zusätzlich wurde mit ersten Untersuchungen zum Wirkmechanismus dieser interessanten Verbindungen begonnen. Darüber hinaus ermöglicht die kontinuierliche Verbesserung der analytischen Methoden inzwischen die schnelle und gezielte Suche nach neuen Verbindungen aus der Natur. Durch die Anwendung von Online-Analyse-Verfahren, wie z.B. die Kopplung von HPLC mit NMR und MS, gelingt die Aufklärung der Konstitution von Substanzen direkt aus Extrakten. Ziel der vorliegenden Arbeit war die Verbesserung der biologischen Aktivitäten der N,C-verknüpften Arylisochinoline durch strukturelle Derivatisierung sowie Beiträge zur Aufklärung des Wirkmechanismus mittels markierter Verbindungen. Zusätzlich sollten Naturstoffe unter Verwendung moderner HPLC-Kopplungstechniken untersucht und strukturell aufgeklärt werden.
Die vorliegende Arbeit beschäftigt sich mit der Licht-Materie-Wechselwirkung in Quantenpunkt-Mikroresonatoren und deren vertikalen und lateralen Emissionseigenschaften. Quantenpunkte sind nanoskopische Strukturen, in denen die Beweglichkeit der Ladungsträger unterhalb der de-Broglie-Wellenlänge eingeschränkt ist, wodurch die elektronische Zustandsdichte diskrete Werte annimmt. Sie werden daher auch als künstliche Atome bezeichnet. Um die Emissionseigenschaften der Quantenpunkte zu modifizieren, werden sie im Rahmen dieser Arbeit als aktive Schicht in Mikrosäulenresonatoren eingebracht. Diese bestehen aus einer GaAs lambda-Kavität, die zwischen zwei Braggspiegeln aus alternierenden GaAs und AlAs Schichten eingefasst ist. Diese Resonatoren bieten sowohl eine vertikale Emission über Fabry-Perot Moden, als auch eine laterale Emission über Flustergaleriemoden. Die Licht-Materie-Wechselwirkung zwischen den Resonatormoden und lokalisierten Ladungsträgern in den Quantenpunkten, genannt Exzitonen, kann in zwei Regime unterteilt werden. Im Regime der starken Kopplung wird der spontane Emissionsprozess in einem Quantenpunkt reversibel und das emittierte Photon kann wieder durch den Quantenpunkt absorbiert werden. Die theoretische Beschreibung der Kopplung eines Exzitons an die Resonatormode erfolgt über das Jaynes-Cummings Modell und kann im Tavis-Cummings Modell auf mehrere Emitter erweitert werden. Ist die Dämpfung des Systems zu gross, so befindet man sich im Regime der schwachen Kopplung, in dem die Emissionsrate des Quantenpunkts durch den Purcell-Effekt erhöht werden kann. In diesem Regime können Mikrolaser mit hohen Einkopplungsraten der spontanen Emission in die Resonatormode und niedrigen Schwellpumpströmen realisiert werden. Zur Charakterisierung der Proben werden vor allem die Methoden der Mikro-Elektrolumineszenz und der Photonenkorrelationsmessungen eingesetzt.
The two bradykinin receptors B1R and B2R are central components of the kallikrein–kinin system with different expression kinetics and binding characteristics. Activation of these receptors by kinins triggers inflammatory responses in the target organ and in most situations enhances tissue damage. We could recently show that blocking of B1R, but not B2R, protects from cortical cryolesion by reducing inflammation and edema formation. In the present study, we investigated the role of B1R and B2R in a closed head model of focal traumatic brain injury (TBI; weight drop). Increased expression of B1R in the injured hemispheres of wild-type mice was restricted to the later stages after brain trauma, i.e. day 7 (P<0.05), whereas no significant induction could be observed for the B2R (P>0.05). Mice lacking the B1R, but not the B2R, showed less functional deficits on day 3 (P<0.001) and day 7 (P<0.001) compared with controls. Pharmacological blocking of B1R in wild-type mice had similar effects. Reduced axonal injury and astroglia activation could be identified as underlying mechanisms, while inhibition of B1R had only little influence on the local inflammatory response in this model. Inhibition of B1R may become a novel strategy to counteract trauma-induced neurodegeneration.
Traumatic brain injury (TBI) is a result of an outside force causing immediate mechanical disruption of brain tissue and delayed pathogenic events. In order to examine injury processes associated with TBI, a number of rodent models to induce brain trauma have been described. However, none of these models covers the entire spectrum of events that might occur in TBI. Here we provide a thorough methodological description of a straightforward closed head weight drop mouse model to assess brain injuries close to the clinical conditions of human TBI.
Melanoma arises from the malignant transformation of melanocytes and is one of the most aggressive forms of human cancer. In fish of the genus Xiphophorus, melanoma development, although very rarely, happens spontaneously in nature and can be induced by interspecific crossing. The oncogenic receptor tyrosine kinase, Xmrk, is responsible for melanoma formation in these fishes. Since Xiphophorus are live-bearing fishes and therefore not compatible with embryonic manipulation and transgenesis, the Xmrk melanoma model was brought to the medaka (Oryzias latipes) system. Xmrk expression under the control of the pigment cell specific mitf promoter leads to melanoma formation with 100% penetrance in medaka. Xmrk is an orthologue of the human epidermal growth factor receptor (EGFR) and activates several downstream signaling pathways. Examples of these pathways are the direct phosphorylation of BRAF and Stat5, as well as the enhanced transcription of C-myc. BRAF is a serine-threonine kinase which is found mutated at high frequencies in malignant melanomas. Stat5 is a transcription factor known to be constitutively activated in fish melanoma. C-myc is a transcription factor that is thought to regulate the expression of approximately 15% of all human genes and is involved in cancer progression of a large number of different tumors. To gain new in vivo information on candidate factors known to be involved in melanoma progression, I identified and analysed BRAF, Stat5 and C-myc in the laboratory fish model system medaka. BRAF protein motifs are highly conserved among vertebrates and the results of this work indicate that its function in the MAPK signaling is maintained in medaka. Transgenic medaka lines carrying a constitutive active version of BRAF (V614E) showed more pigmented skin when compared to wild type. Also, some transiently expressing BRAF V614E fishes showed a disrupted eye phenotype. In addition, I was able to identify two Stat5 copies in medaka, named Stat5ab/a and Stat5ab/b. Sequence analysis revealed a higher similarity between both Stat5 sequences when compared to either human Stat5a or Stat5b. This suggests that the two Stat5 copies in medaka arose by an independent duplication processes. I cloned these two Stat5 present in medaka, produced constitutive active and dominant negative gene versions and successfully established transgenic lines carrying each version under the control of the MITF promoter. These lines will help to elucidate questions that are still remaining in Stat5 biology and its function in melanoma progression, like the role of Stat5 phosphorylation on tumor invasiveness. In a third project during my PhD work, I analysed medaka C-myc function and indentified two copies of this gene in medaka, named c-myc17 and c-myc20, according to the chromosome where they are located. I produced conditional transgenic medaka lines carrying the c-myc17 gene coupled to the hormone binding domain of the estrogen receptor to enable specific transgene activation at a given time point. Comparable to human C-myc, medaka C-myc17 is able to induce proliferation and apoptosis in vivo after induction. Besides that, C-myc17 long-term activation led to liver hyperplasia. In summary, the medaka models generated in this work will be important to bring new in vivo information on genes involved in cancer development. Also, the generated transgenic lines can be easily crossed to the melanoma developing Xmrk medaka lines, thereby opening up the possibility to investigate their function in melanoma progression. Besides that, the generated medaka fishes make it possible to follow the whole development of melanocytes, since the embryos are transparent and can be used for high throughput chemical screens.
Intrazerebrale stereotaktische Eingriffe werden zu einem großen Teil ohne direkte Sichtkontrolle durchgeführt. Ein Operateur muss sich deshalb bei der räumlichen Festlegung von Strukturen und beim Anfahren dieser Strukturen auf Hilfsmittel wie stereotaktische Geräte und auf Atlanten, über welche die stereotaktischen Geräte gesteuert werden, verlassen. Trotz großer Fortschritte bei den bildgebenden Verfahren während der letzten dreißig Jahre, ist es gegenwärtig noch nicht möglich, zuverlässig alle subkortikalen Strukturen mit computertomographischen (CT) oder magnetresonanztomographischen (MRT) zu identifizieren oder begrenzen. Eine ganze Reihe zytoarchitektonischer beziehungsweise immunhistochemischer Atlanten wurde veröffentlicht. Dennoch ist es nicht gelungen, die Ergebnisse und Abbildungen dieser Atlanten mit bildgebenden Verfahren bis in die gewünschten Details zu kombinieren, um auf diese Weise das immer noch geringe Auflösungsvermögen radiologischer Methoden zu erhöhen. Deformationen bei der Gewebsentnahme des Gehirns, bei der anschließenden Einbettung, bei der alkoholischen Dehydrierung des Gewebes, Verformungen beim Schneiden und Färben der Schnitte überfordern selbst hoch komplexe mathematische Verfahren und Algorithmen beim Versuch, zytoarchitektonische und immunhistochemische Schnitte mit der gewünschten Präzision den radiologischen Ergebnissen und Bildern und damit indirekt auch den Verhältnissen in vivo anzupassen. Als Alternative verwendeten wir ungewöhnlich dicke (350 – 440 µm) Gallozyanin- (Nissl) gefärbte Serienschnitte durch die Gehirne (ZNS) von drei Personen im Alter von 56, 68 und 36 Jahren. Bei einem Fall wurde das ZNS post mortem mit einem Kernspintomographen vor der Entnahme gescannt. Die Serienschnitte durch dieses Gehirn und das eines zweiten und dritten nicht-gescannten Falles wurden mit Gallozyanin gefärbt, die zytoarchitektonischen Grenzen des Thalamuskomplexes und seiner Unterkerne wurden nach Hassler (1982) identifiziert, jede ihrer Grenzen mit dem Cursor eines Graphiktabletts umfahren und die Gestalt des Thalamuskomplexes und seiner Unterkerne mit Hilfe von Photoshop CS5® und eines computergestützten 3D-Rekonstruktionsprogramms (Amira®) dargestellt. Im Fall 3 ließen sich nach Dunkelfeldbeleuchten die Verteilung markhaltiger Fasern studieren und die zytoarchitektonischen mit myeloarchitektonischen Befunden erweitern und ergänzen. Zusätzlich konnten im Fall 1 die histologischen Serienschnitte und ihre 3D Rekonstruktion mit dem post mortem in cranio MRT registriert werden. Insgesamt kann dieser methodische Ansatz als eine robuste und relativ einfache wenn auch mit umfangreicherer manueller Tätigkeit verbundene Technik zur sehr detailreichen unverformten Korrelation zytoarchitektonischer und kernspinotomographsicher Darstellung des Thalamuskomplexus und seiner Unterkerne angesehen werden. Sie könnte als Grundlage für die Herausgabe eines multimedialen 3D stereotaktischen Atlas des menschlichen Gehirns dienen.
Incidence rates of infections caused by environmental opportunistic fungi have risen over recent decades. Aspergillus species have emerged as serious threat for the immunecompromised, and detailed knowledge about virulence-determining traits is crucial for drug target identification. As a prime saprobe, A. fumigatus has evolved to efficiently adapt to various stresses and to sustain nutritional supply by osmotrophy, which is characterized by extracellular substrate digestion followed by efficient uptake of breakdown products that are then fed into the fungal primary metabolism. These intrinsic metabolic features are believed to be related with its virulence ability. The plethora of genes that encode underlying effectors has hampered their in-depth analysis with respect to pathogenesis. Recent developments in Aspergillus molecular biology allow conditional gene expression or comprehensive targeting of gene families to cope with redundancy. Furthermore, identification of essential genes that are intrinsically connected to virulence opens accurate perspectives for novel targets in antifungal therapy.
Malaria and HIV are among the most important global health problems of our time and together are responsible for approximately 3 million deaths annually. These two diseases overlap in many regions of the world including sub-Saharan Africa, Southeast Asia and South America, leading to a higher risk of co-infection. In this study, we generated and characterized hybrid molecules to target P. falciparum and HIV simultaneously for a potential HIV/malaria combination therapy. Hybrid molecules were synthesized by covalent fusion between azidothymidine (AZT) and dihydroartemisinin (DHA), tetraoxane or chloroquine (CQ); and a small library was generated and tested for antiviral and antimalarial activity. Our data suggest that dihyate is the most potent molecule in vitro, with antiplasmodial activity comparable to that of DHA (IC50 = 26 nM, SI > 3000), a moderate activity against HIV (IC50 = 2.9 µM; SI > 35) and safe to HeLa cells at concentrations used in the assay (CC50 > 100 µM). Pharmacokinetic studies further revealed that dihyate is metabolically unstable and is cleaved following an O-dealkylation once in contact with cytochrome P450 enzymes. The later further explains the uneffectiveness of dihyate against the CQ-sensitive P. berghei N strain in mice when administered by oral route at 20 mg/kg. Here, we report on a first approach to develop antimalarial/anti-HIV hybrid molecules and future optimization efforts will aim at producing second generation hybrid molecules to improve activity against HIV as well as compound bioavailability. With the emergence of resistant parasites against all the counterpart drugs of artemisinin derivatives used in artemisinin based combination therapies (ACTs), the introduction of antibiotics in the treatment of malaria has renewed interest on the identification of antibiotics with potent antimalarial properties. In this study we also investigated the antiplasmodial potential of thiostrepton and derivatives, synthesized using combinations of tail truncation, oxidation, and addition of lipophilic thiols to the terminal dehydroamino acid. We showed that derivatives SS231 and SS234 exhibit a better antiplasmodial activity (IC50 = 1 µM SI > 59 and SI > 77 respectively) than thiostrepton (IC50 = 8.95 µM, SI = 1.7). The antiplasmodial activity of these derivatives was observed at concentrations which are not hemolytic and non-toxic to human cell lines. Thiostrepton and derivatives appeared to exhibit transmission blocking properties when administered at their IC50 or IC90 concentrations and our data also showed that they attenuate proteasome activity of Plasmodium, which resulted in an accumulation of ubiquitinated proteins after incubation with their IC80 concentrations. Our results indicate that the parasite’s proteasome could be an attractive target for therapeutic intervention. In this regard, thiostrepton derivatives are promising candidates by dually acting on two independent targets, the proteasome and the apicoplast, with the capacity to eliminate both intraerythrocytic asexual and transmission stages of the parasite. To further support our findings, we evaluated the activity of a new class of antimalarial and proteasome inhibitors namely peptidyl sulfonyl fluorides on gametocyte maturation and analogues AJ34 and AJ38 were able to completely suppress gametocytogenesis at IC50 concentrations (0.23 µM and 0.17 µM respectively) suggesting a strong transmission blocking potential. The proteasome, a major proteolytic complex, responsible for the degradation and re-cycling of non-functional proteins has been studied only indirectly in P. falciparum. In addition, an apparent proteasome-like protein with similarity to bacterial ClpQ/hslV threonine-peptidases was predicted in the parasite. Antibodies were generated against the proteasome subunits alpha type 5 (α5-SU), beta type 5 (β5-SU) and pfhslV in mice and we showed that the proteasome is expressed in both sexual and asexual blood stages of P. falciparum, where they localize in the nucleus and in the cytoplasm. However, expression of PfhslV was only observed in trophozoites and shizonts. The trafficking of the studied proteasome subunits was further investigated by generating parasites expressing GFP tagged proteins. The expression of α5-SU-GFP in transgenic parasite appeared to localize abundantly in the cytoplasm of all blood stages, and no additional information was obtained from this parasite line. In conclusion, our data highlight two new tools towards combination therapy. Hybrid molecules represent promising tools for the cure of co-infected individuals, while very potent antibiotics with a wide scope of activities could be useful in ACTs by eliminating resistant parasites and limiting transmission of both, resistances and disease.
Nach wie vor ist die Zahl der Malaria-Neuerkrankungen mit ca. 500 Millionen Menschen weltweit sehr besorgniserregend. Durch zunehmende Resistenzen der Erreger gegen zahlreiche Arzneimittel wird die Situation zusätzlich verschärft. Daher ist die Suche und Entwicklung neuartiger Medikamente wichtiger denn je. Die Natur ist immer noch das größte Reservoir an neuen Wirkstoffen, welche als Leitstrukturen für Arzneistoffe fungieren. In den letzten Jahren wurde eine große Zahl neuartiger, biologisch aktiver Naturstoffe identifiziert und hinsichtlich ihres Potenzials für eine pharmazeutische Weiterentwicklung analysiert. Die Naphthylisochinolin-Alkaloide gehören zu solch einer vielversprechenden Wirkstoffklasse, da sie sich v.a. durch ihre hervorragenden pharmakologischen Eigenschaften auszeichnen. Kürzlich wurden die ersten N,C-gekuppelten Naphthyldihydroisochinolin-Alkaloide, Ancistrocladinium A und B, entdeckt. Diese Verbindungen weisen als strukturelle Besonderheit eine außergewöhnliche Iminium-Aryl-Achse auf und besitzen zudem exzellente anti-infektive Aktivitäten, insbesondere gegen den Erreger der Orientbeule, Leishmania major. Ziel der vorliegenden Dissertation war die Synthese neuartiger sterisch gehinderter und strukturell vereinfachter Naphthylisochinoline für weiterführende Struktur-Aktivitäts-Beziehungen (SAR-Studien) und die stereochemische Analyse dieser Verbindungen. Zudem sollte eine Syntheseroute zu den neuartigen dimeren C,C-gekuppelten Naphthylisochinolin-Alkaloiden Shuangancistrotectorin A und B entwickelt werden.
Background: Sorbents have been shown to adsorb iodinated radiocontrast media. Objective: In this study we describe a simple method to compare various sorbents in terms of capacity to adsorb radiocontrast media. Methods: Iodixanol solution was injected into columns filled with three types of sorbent at filtration velocities of increasing magnitude. Two variables of interest – contrast removal rate and matched iodine retention (MIR) – were calculated to measure the adsorption efficiency and the mass of contrast iodine adsorbed versus sorbent used, respectively. Results: The highest contrast removal and MIR for Porapak Q, CST 401 and Amberlite XAD4 were 41, 38 and 16% (p = 0.22 and 0.0005 for comparisons between Porapak Q-CST 401 and CST 401-Amberlite XAD4) and 0.060, 0.055 and 0.024, respectively (p = 0.18 and 0.0008). Extrapolation to a clinical scenario may suggest that removal of 8 ml iodixanol could be achieved by masses of sorbents of 43, 47 and 107 g, respectively. Conclusion: In this study we set a benchmark for comparing the radiocontrast-adsorbing efficiency of polymer sorbents during first-pass experiments, using a readily available methodology.
Introduction: Several common alleles have been shown to be associated with breast and/or ovarian cancer risk for BRCA1 and BRCA2 mutation carriers. Recent genome-wide association studies of breast cancer have identified eight additional breast cancer susceptibility loci: rs1011970 (9p21, CDKN2A/B), rs10995190 (ZNF365), rs704010 (ZMIZ1), rs2380205 (10p15), rs614367 (11q13), rs1292011 (12q24), rs10771399 (12p11 near PTHLH) and rs865686 (9q31.2).
Methods: To evaluate whether these single nucleotide polymorphisms (SNPs) are associated with breast cancer risk for BRCA1 and BRCA2 carriers, we genotyped these SNPs in 12,599 BRCA1 and 7,132 BRCA2 mutation carriers and analysed the associations with breast cancer risk within a retrospective likelihood framework.
Results: Only SNP rs10771399 near PTHLH was associated with breast cancer risk for BRCA1 mutation carriers (per-allele hazard ratio (HR) = 0.87, 95% CI: 0.81 to 0.94, P-trend = 3 x 10\(^{-4}\)). The association was restricted to mutations proven or predicted to lead to absence of protein expression (HR = 0.82, 95% CI: 0.74 to 0.90, P-trend = 3.1 x 10\(^{-5}\), P-difference = 0.03). Four SNPs were associated with the risk of breast cancer for BRCA2 mutation carriers: rs10995190, P-trend = 0.015; rs1011970, P-trend = 0.048; rs865686, 2df P = 0.007; rs1292011 2df P = 0.03. rs10771399 (PTHLH) was predominantly associated with estrogen receptor (ER)-negative breast cancer for BRCA1 mutation carriers (HR = 0.81, 95% CI: 0.74 to 0.90, P-trend = 4 x 10\(^{-5}\)) and there was marginal evidence of association with ER- negative breast cancer for BRCA2 mutation carriers (HR = 0.78, 95% CI: 0.62 to 1.00, P-trend = 0.049).
Conclusions: The present findings, in combination with previously identified modifiers of risk, will ultimately lead to more accurate risk prediction and an improved understanding of the disease etiology in BRCA1 and BRCA2 mutation carriers.

In the centrosymmetric molecular structure of the title compound \([Pt_2(C_6H_{14}P)_2H_2)(C_9H_{21}P)_2]\), each \(Pt^{II}\) atom is bound on one side to a phosphane ligand \((PiPr_3)\) and a hydrido ligand. On the other side, it is bound to two phosphanide ligands \((μ-PiPr_2)\), which engage a bridging position between the two \(Pt^{II}\) atoms, forming a distorted square-planar structure motif. The PtPt distance is 3.6755(2)Å. A comparable molecular structure was observed for bis-(μ-di-tert-butyl-phosphanido)bis-[hydrido(triethyl-phosphane)platinum(II)] [Itazaki et al. (2004 ). In dieser post-hoc Analyse der Deutschen Diabetes und Dialyse Studie wurde der Einfluss von NT-proBNP und Troponin T auf plötzlichen Herztod, Schlaganfall, Myokardinfarkt und die Gesamtmortalität während vierjähriger Studiendauer bei 1255 Patienten mit Diabetes mellitus Typ 2 an der Hämodialyse analysiert. Des Weiteren wurde die Bedeutung einer longitudinalen Messung der Biomarker nach 6 Monaten auf die Endpunkte untersucht. Patienten mit dem höchsten NT-proBNP respektive Troponin T wiesen die größte Ereignisrate für plötzlichen Herztod, Schlaganfall und die Gesamtmortalität auf. In der multivariaten Regressionsanalyse waren sowohl NT-proBNP als auch Troponin T jeweils starke unabhängige Prädiktoren für plötzlichen Herztod, Schlaganfall und die Gesamtmortalität. Eine Assoziation von NT-proBNP mit dem Auftreten von Myokardinfarkten wurde nicht gesehen. Nicht nur ein hoher Ausgangswert der Biomarker, sondern auch eine Zunahme von NT-proBNP und Troponin T nach 6 Monaten waren assoziiert mit einer schlechteren Langzeitprognose
Animals acquire predictive values of sensory stimuli through reinforcement. In the brain of Drosophila melanogaster, activation of two types of dopamine neurons in the PAM and PPL1 clusters has been shown to induce aversive odor memory. Here, we identified the third cell type and characterized aversive memories induced by these dopamine neurons. These three dopamine pathways all project to the mushroom body but terminate in the spatially segregated subdomains. To understand the functional difference of these dopamine pathways in electric shock reinforcement, we blocked each one of them during memory acquisition. We found that all three pathways partially contribute to electric shock memory. Notably, the memories mediated by these neurons differed in temporal stability. Furthermore, combinatorial activation of two of these pathways revealed significant interaction of individual memory components rather than their simple summation. These results cast light on a cellular mechanism by which a noxious event induces different dopamine signals to a single brain structure to synthesize an aversive memory.
Für die palliative Bestrahlung des NSCLC stehen mehrere, sehr unterschiedliche hypofraktionierte Behandlungsschemata zur Verfügung. Prospektive Studien in der Vergangenheit konnten keine Überlegenheit für eines dieser Regime zeigen. Ziel vorliegender retrospektiver Arbeit war es, die Effektivität der Radiatio mit 13 bis 15 Fraktionen zu 3 Gy zu überprüfen. Hierzu untersuchten wir die Daten von 57 Patienten, die sich in den Jahren 2006 bis 2008 in der Strahlentherapie der Universitätsklinik Würzburg einer solchen Therapie unterzogen. Das Patientengut unterteilten wir für die Untersuchung in zwei Gruppen M0 und M1, deren Prognose wir unterschiedlich einschätzten. Der Einteilung lag das Vorhandensein von Fernmetastasen zu Behandlungsbeginn zugrunde.
Das Gesamtüberleben war für Patienten der M0-Gruppe signifikant besser und lag für M1-Patienten in einem zu erwartenden Bereich. 17,5% unserer Patienten lebten 18 Monate oder länger. Welche Ursachen hinter diesem prolongierten Überleben stehen könnten, blieb jedoch weitgehend unklar.
Für das Gesamtüberleben zeigten sich verschiedene u.a. aus der Literatur bekannte Prognosefaktoren wie das UICC-Stadium, der Allgemeinzustand und eine chemotherapeutische Behandlung. Andere Faktoren, deren Einfluss wir vermuteten, führten zu keinen signifikanten bzw. widersprüchlichen Ergebnissen. Hierzu zählten insbesondere der Charlson comorbidity score und das Alter. Für die Höhe der Gesamtdosis und die Größe des PTV wurde interessanterweise kein Einfluss auf das Überleben nachgewiesen. Die lokale Kontrolle war von diesen beiden Variablen ebenfalls unabhängig.
Ein systemischer Progress trat bei unseren Patienten tendenziell früher auf als ein lokaler Progress.
Der Allgemeinzustand der Patienten wurde von der Bestrahlung im Wesentlichen nicht negativ beeinflusst, Infektionen traten so gut wie gar nicht auf. Wie bereits aus prospektiven Studien zur hypofraktionierten Bestrahlung bekannt, waren Akuttoxizitäten, insbesondere Ösophagitiden, relativ häufig.

We study problems that arise in the context of covering certain geometric objects called seeds (e.g., points or disks) by a set of other geometric objects called cover (e.g., a set of disks or homothetic triangles). We insist that the interiors of the seeds and the cover elements are pairwise disjoint, respectively, but they can touch. We call the contact graph of a cover a cover contact graph (CCG). We are interested in three types of tasks, both in the general case and in the special case of seeds on a line: (a) deciding whether a given seed set has a connected CCG, (b) deciding whether a given graph has a realization as a CCG on a given seed set, and (c) bounding the sizes of certain classes of CCG’s. Concerning (a) we give efficient algorithms for the case that seeds are points and show that the problem becomes hard if seeds and covers are disks. Concerning (b) we show that this problem is hard even for point seeds and disk covers (given a fixed correspondence between graph vertices and seeds). Concerning (c) we obtain upper and lower bounds on the number of CCG’s for point seeds.
We have studied the responses of honey bees at different life stages (Apis mellifera) to controlled infection with acute bee paralysis virus and have identified the haemolymph of infected larvae and adult worker bees as the compartment where massive propagation of ABPV occurs. Insects respond with a broad spectrum of induced innate immune reactions to bacterial infections, whereas defence mechanisms based on RNA interference play a major role in antiviral immunity. In this study, we have determined that honey bee larvae and adult workers do not produce a humoral immune reaction upon artificial infection with ABPV, in contrast to control individuals challenged with Escherichia coli. ABPV-infected bees produced neither elevated levels of specific antimicrobial peptides (AMPs), such as hymenoptaecin and defensin, nor any general antimicrobial activity, as revealed by inhibition-zone assays. Additionally, adult bees did not generate melanised nodules upon ABPV infection, an important cellular immune function activated by bacteria and viruses in some insects. Challenge of bees with both ABPV and E. coli showed that innate humoral and cellular immune reactions are induced in mixed infections, albeit at a reduced level.
Durch Fortschritte in der Technologie haben interventionelle Eingriffe am Herzen in den letzten Jahrzehnten einen herausragenden Stellenwert entwickelt und zu einer Reduktion von aufwendigen Operationen am Herzen geführt. Die Ausbildung im Herzkatheterlabor, die nach dem konservativen „appreticeship-model“ erfolgt, gerät in Anbetracht der sinkenden finanziellen Mittel, Zeitmangel und der ethischen Fragen bezüglich Patientensicherheit immer mehr in Diskussion. Die Entwicklung der Virtual-Reality-Simulatoren für Kathetereingriffe bietet hier durch die Realitätsnähe einen Ansatzpunkt für die Möglichkeit eines individuell angepassten, repetitiven Trainings ohne die Gefährdung eines Patienten. Standardsituationen als auch seltene Komplikationen können nachgestellt werden. Diese Studie weist nach, dass Training an den Virtual-Reality-Simulatoren CATHI und Immersion zu einer Risikoreduktioin bei der Durchführung einer perkutanen Coronarintervention führt. Zur Untersuchung der Effekte von Virtual-Reality-Training auf die Performance einer perkutanen Coronarintervention wurde an der medizinischen Klinik Wuerzburg eine kontrolliert-radnomisierte Studie mit 33 Anfängern in der interventionellen Kardiologie durchgeführt. 16 Teilnehmer (Simulationsgruppe) erhielten ein intensives acht-stuendiges Simulationstraining an zwei verschiedenen Virtual-Reality-Simulatoren (CATHI und Immersion), 17 Teilnehmer bildeten die Kontrollgruppe, die den konservativen Ausbildungsgang repräsentierte und kein Simulationstraining erhielt. Alle Teilnehmer mussten in Form einer Prä- und Postevaluation unter realitätsnahen Umständen im Herzkatheterlabor der Uniklinik Würzburg innerhalb von 30 Minuten eine perkutane Coronarintervention an einem pulsatilen Herzkreislaufmodell aus Silikon (CoroSim) eigenständig vornehmen. Dabei musste eine an einer Aufteilung lokalisierte hochgradige Stenose ohne Abgänge mit einer Länge von 10mm und einem Gefäßdurchmesser von 4mm eröffnet werden. Die Ergebnisse zeigten für die Präevaluation keine gruppenspezifischen Unterschiede. Nach dem Simulationstraining zeigte sich eine signifikante Verbesserung der Simulationsgruppe bei der Risikominimierung in Bezug auf Sicherheit bei der Anwendung des Führungskatheters, des Koronardrahts, des Ballon/Stents und bei der KM-Injektion, während sich die Kontrollgruppe in diesen Punkten nicht verbessern konnte. Die aktuelle Studie zeigt, dass Training an den Virtual-Reality-Simulatoren, als Ergänzung zur herkömmlichen Ausbildung, ein hohes Potential für die Optimierung von interventionellen Herzkathetereingriffen verfügt.
Die Auswertung des Patientenkollektivs von 2000 bis 2004 an der Chirurgischen Universitätsklinik Würzburg ergab 63 Patienten (Gruppe 1), die einer Minoramputation und
59 Patienten die einer Minor- mit konsekutiver Majoramputation (Gruppe 2) infolge pAVK unterzogen wurden. Eine Abhängigkeit zwischen Alter und Majornachamputationsrate konnte nicht festgestellt werden, jedoch eine Tendenz beim Einfluss von Comorbiditäten wie Diabetes mellitus und dialysepflichtiger Niereninsuffizienz auf die Wundheilung und Liegedauer.
Die Anzahl an durchgeführten gefäßchirurgischen Eingriffen wie PTA und Bypass-Operation lag in beiden Gruppen mit 65% bzw. 72% im Vergleich zur Literatur im Standardbereich.
Eine hohe Rate an gefäßmedizinischer Diagnostik und Therapie scheint auch bei fortgeschrittener pAVK (Grad IV) erforderlich, um die Notwendigkeit von Amputationen
insbesondere die Anzahl an Majoramputationen zu verringern. Bei hoher und mit Gruppe 1 vergleichbarer Interventionsrate in Gruppe 2 lässt sich allerdings auch erkennen, dass trotz Ausschöpfung dieser Massnahmen die Rate an Majoramputationen und damit des
Beinverlustes hoch ist.
Positiv zu werten ist, dass es bei über 50% der Amputierten ausgereicht hat, eine Amputation im Fussbereich (Minoramputation) durchzuführen. Bei diesen 63 Patienten, war bei 58 Patienten sogar nur ein Eingriff nötig. Ferner handelte es sich bei den Majoramputationen in der Mehrzahl um Unterschenkelamputation, und somit um einen nur partiellen Beinverlust . 76% der durchgeführten ersten Majoramputationen erfolgten in den ersten beiden Monaten nach vorausgegangener Minoramputation, die größte Anzahl innerhalb des ersten Monats. Auch die letzte Amputation, die definitive Versorgung, erfolgte
in den meisten Fällen innerhalb der ersten beiden Monate nach Primäreingriff. Somit ist ein nur unwesentlicher Aufschub bis zur definitiven Versorgung ersichtlich.
Der Versuch einer Konsolidierung der Ischämiefolgen (Gangrän) mittels Minoramputation scheint bei fortgeschrittener pAVK im Stadium IV nach Ausschöpfung der
gefäßmedizinischen Diagnostika und Interventionen somit immer gerechtfertigt, und sollte wenn möglich einer Majoramputation vorgezogen werden.
Die durchschnittliche Krankenhausverweildauer in der Gruppe der Minoramputationen lag bei 28 Tagen, in der Gruppe 2 der Majoramputationen bei 39 Tagen. Die Mortalitätsrate ergibt einen deutlich erhöhten Wert in der Gruppe der Majoramputationen. Die Dreijahresmortalität betrug in der Gruppe der Minoramputationen 20% und der Majoramputationen 58%. Es zeigte sich eine Zunahme der Mortalität mit zunehmender
Amputationshöhe und zunehmender Zahl der Amputationen. Diese Daten lassen sich mit 52 aktuellen Literaturangaben durchaus vergleichen und beweisen die schlechte Prognose für AVK-Patienten, bei denen eine Majoramputation unausweichlich ist.
Insgesamt ist es wichtig, dass ein Team aus Chirurgen, Gefäßchirurgen, Radiologen und Angiologen kooperativ zusammen arbeitet, um dem Patienten, eine für ihn
beste Versorgung anbieten zu können. Hier sollte auch nicht vor einem oft höheren Patientenalter zurückgeschreckt werden, denn häufig konnte gerade bei diesen Patienten durch eine Bypass-Operation eine sonst vermutlich unumgängliche Amputation im Unter- bzw.
Oberschenkelbereich verhindert werden.
Numerous small non-coding RNAs (sRNAs) in bacteria modulate rates of translation initiation and degradation of target mRNAs, which they recognize through base-pairing facilitated by the RNA chaperone Hfq. Recent evidence indicates that the ternary complex of Hfq, sRNA and mRNA guides endoribonuclease RNase E to initiate turnover of both the RNAs. We show that a sRNA not only guides RNase E to a defined site in a target RNA, but also allosterically activates the enzyme by presenting a monophosphate group at the 5′-end of the cognate-pairing “seed.” Moreover, in the absence of the target the 5′-monophosphate makes the sRNA seed region vulnerable to an attack by RNase E against which Hfq confers no protection. These results suggest that the chemical signature and pairing status of the sRNA seed region may help to both ‘proofread’ recognition and activate mRNA cleavage, as part of a dynamic process involving cooperation of RNA, Hfq and RNase E.
Background: Alveolar echinococcosis (AE) is caused by the metacestode stage of Echinococcus multilocularis. Differential diagnosis with cystic echinococcosis (CE) caused by E. granulosus and AE is challenging. We aimed at improving diagnosis of AE on paraffin sections of infected human tissue by immunohistochemical testing of a specific antibody.
Methodology/Principal Findings: We have analysed 96 paraffin archived specimens, including 6 cutting needle biopsies and 3 fine needle aspirates, from patients with suspected AE or CE with the monoclonal antibody (mAb) Em2G11 specific for the Em2 antigen of E. multilocularis metacestodes. In human tissue, staining with mAb Em2G11 is highly specific for E. multilocularis metacestodes while no staining is detected in CE lesions. In addition, the antibody detects small particles of E. multilocularis (spems) of less than 1 mm outside the main lesion in necrotic tissue, liver sinusoids and lymphatic tissue most probably caused by shedding of parasitic material. The conventional histological diagnosis based on haematoxylin and eosin and PAS stainings were in accordance with the immunohistological diagnosis using mAb Em2G11 in 90 of 96 samples. In 6 samples conventional subtype diagnosis of echinococcosis had to be adjusted when revised by immunohistology with mAb Em2G11.
Conclusions/Significance: Immunohistochemistry with the mAb Em2G11 is a new, highly specific and sensitive diagnostic tool for AE. The staining of small particles of E. multilocularis (spems) outside the main lesion including immunocompetent tissue, such as lymph nodes, suggests a systemic effect on the host.
The neurodegenerative disorder Alzheimer's disease (AD) is the cause of approximately 60% of the world's 35 million patients suffering from dementia. Current research focuses here are on association with other diseases such as diabetes type 2 (T2DM), possible genetic markers, specific signal transduction pathways within the brain and potential protein modification, because the pathogenesis and etiology of AD are still not fully understood. Specifically association of T2DM with AD came to the focus with the so-called "Rotterdam study" in 1999, indicating that T2DM doubles the risk of developing AD. In the meantime, it is known that the prevalence rate in patients with T2DM is 30%. Drugs commonly used in the treatment of T2DM such as peroxisome proliferator-activated receptors gamma (PPARγ) agonists show improvement of the cognitive abilities in patients with early stage of dementia, with potential therapeutically relevance. Therefore it is important not only to investigate a link between these diseases, but also to investigate the insulin signaling pathway in the brain of AD patients. In order to investigate this complex issue in more details and demonstrate additional links between T2DM and AD, the present study used several basic biological methods to clarify the question: "Is impaired insulin signaling pathway within the brain crucial for the development of AD?" from several points of view. The methods used in this work have been i) an analysis of single nucleotide (SNP) polymorphism of the insulin-degrading enzyme gene (IDE) in relation to risk of AD and / or of T2DM, ii) post-mortem histochemical studies of brain tissue of patients with only AD, with AD combined with T2DM and with only T2DM compared with an age-matched control group, and iii.) investigations of neurochemical pathways and gene/protein expression changes of a human cell culture as a consequences of amyloid β (Aβ) treatment. After analysis of the IDE SNP polymorphism in the selected VITA (Vienna Trans Danube Aging) cohort disease-specific effects were discovered. The upstream polymorphism (IDE2) was found to influence AD risk in a protective manner, while the downstream polymorphism (IDE7) modified the T2DM risk. Based on the SNP results, the presented study delineate the model that IDE promoter and 3‟ untranslated region/downstream variation can have different effects on IDE expression, maybe a relevant endophenotype with disorder-specific effects on AD and T2DM susceptibility. Furthermore, the human post-mortem studies could show that both AD as well as T2DM patients had a significantly lower density of the insulin receptor (IR) in the hippocampus, whereas a significantly increased density of inactive phosphorylated PPARγ has been found and this persisted even in patients with both diseases. Summarizing the histological study, it was possible to reveal common histological features of AD and T2DM, but no direct connection between the two diseases. Although AD is nowadays not only characterized by amyloid-containing plaque deposits and by the hyperphosphorylation of tau protein, the excessive Aβ42 presence in the brains of AD patients is still playing a key role. Up to date it is still not entirely clear which physical form of Aβ42 is responsible for the development of AD. The present work investigated, what impact has the state of aggregation of Aβ42 on genes and proteins of the insulin signaling pathway and the amyloid cascade. It could be shown that the oligomeric variant enhanced specifically the gene and protein expression of glycogen synthase kinase (GSK) 3β and also the enzyme activity was significantly increased, but has in turn strongly inhibited the IR gene and protein expression. Additionally, the effect of Aβ42 on monoamine oxidase B (MAO-B) was examined. An effect of both aggregated forms of Aβ42 had on enzyme activity was discovered. However, the fibrillar variants led to significantly increased activity of MAO-B while the oligomeric variants inhibited the enzyme activity. Several previous studies have demonstrated the involvement of increased MAO-B activity in AD, but the present work provides for the first time a direct link between the states of aggregation of Aβ42 to enzyme activity. Finally the results of the presented thesis can be summarized to following conclusion: Although AD and T2DM sharing some degrees of common features, still there is a lack of direct association, and therefore the diseases must be considered more independent rather than linked. But the impaired cerebral insulin signaling pathway seems to be another manifested hallmark of AD.
This thesis focuses on various aspects and techniques of 19F magnetic resonance (MR). The first chapters provide an overview of the basic physical properties, 19F MR and MR sequences related to this work. Chapter 5 focuses on the application of 19F MR to visualize biological processes in vivo using two different animal models. The dissimilar models underlined the wide applicability of 19F MR in preclinical research. A subsection of Chapter 6 shows the application of compressed sensing (CS) to 19F turbo-spin-echo chemical shift imaging (TSE-CSI), which leads to reduced measurement time. CS, however, can only be successfully applied when a sufficient signal-to-noise ratio (SNR) is available. When the SNR is low, so-called spike artifacts occur with the CS algorithm used in the present work. However, it was shown in an additional subsection that these artifacts can be reduced using a CS-based post processing algorithm. Thus, CS might help overcome limitations with time consuming 19F CSI experiments. Chapter 7 deals with a novel technique to quantify the B+1 profile of an MR coil. It was shown that, using a specific application scheme of off resonant pulses, Bloch-Siegert (BS)-based B+1 mapping can be enabled using a Carr Purcell Meiboom Gill (CPMG)-based TSE sequence. A fast acquisition of the data necessary for B+1 mapping was thus enabled. In the future, the application of BS-CPMG-TSE B+1 mapping to improve quantification using 19F MR could therefore be possible.
Es wurden Übergangsmetallkomplexe der Metalle Rhodium, Palladium und Platin synthetisiert und unter anderem in Bezug auf ihre Reaktivität als Lewis-Basen untersucht. Hierfür wurden Lewis-Addukte mit Aluminiumtrichlorid mit experimentellen und quantenchemischen Methoden umfassend untersucht. Des Weiteren wurde die Reaktivität von verschiedenen Fluorboranen gegnüber Lewis-basischen Übergangsmetallkomplexen untersucht. Auch konnte die Reaktivität von Münzgruppenmetall-Halogen-Bindungen gegenüber Lewis-Basen analysiert werden. So konnten nicht nur Insertionsprodukte, sondern auch kationische Metal-Only Lewis Pairs, unter anderem anhand von Einkristallröntgenstrukturanalysen und Dichte-Funktional-Theorie Rechnungen, untersucht werden.
This work takes a close look at several quite different research areas related to the design of networked embedded sensor/actuator systems. The variety of the topics illustrates the potential complexity of current sensor network applications; especially when enriched with actuators for proactivity and environmental interaction. Besides their conception, development, installation and long-term operation, we'll mainly focus on more "low-level" aspects: Compositional hardware and software design, task cooperation and collaboration, memory management, and real-time operation will be addressed from a local node perspective. In contrast, inter-node synchronization, communication, as well as sensor data acquisition, aggregation, and fusion will be discussed from a rather global network view. The diversity in the concepts was intentionally accepted to finally facilitate the reliable implementation of truly complex systems. In particular, these should go beyond the usual "sense and transmit of sensor data", but show how powerful today's networked sensor/actuator systems can be despite of their low computational performance and constrained hardware: If their resources are only coordinated efficiently!
Background: Cytokines such as interleukin 6 (IL-6) have been implicated in dual functions in neuropsychiatric disorders. Little is known about the genetic predisposition to neurodegenerative and neuroproliferative properties of cytokine genes. In this study the potential dual role of several IL-6 polymorphisms in brain morphology is investigated.
Methodology: In a large sample of healthy individuals (N = 303), associations between genetic variants of IL-6 (rs1800795; rs1800796, rs2069833, rs2069840) and brain volume (gray matter volume) were analyzed using voxel-based morphometry (VBM). Selection of single nucleotide polymorphisms (SNPs) followed a tagging SNP approach (e. g., Stampa algorigthm), yielding a capture 97.08% of the variation in the IL-6 gene using four tagging SNPs. Principal findings/results In a whole-brain analysis, the polymorphism rs1800795 (-174 C/G) showed a strong main effect of genotype (43 CC vs. 150 CG vs. 100 GG; x = 24, y = -10, z = -15; F(2,286) = 8.54, p(uncorrected) = 0.0002; p(AlphaSim-corrected) = 0.002; cluster size k = 577) within the right hippocampus head. Homozygous carriers of the G-allele had significantly larger hippocampus gray matter volumes compared to heterozygous subjects. None of the other investigated SNPs showed a significant association with grey matter volume in whole-brain analyses.
Conclusions/significance: These findings suggest a possible neuroprotective role of the G-allele of the SNP rs1800795 on hippocampal volumes. Studies on the role of this SNP in psychiatric populations and especially in those with an affected hippocampus (e.g., by maltreatment, stress) are warranted.
We perform global fits to the parameters of the Constrained Minimal Super-symmetric Standard Model (CMSSM) and to a variant with non-universal Higgs masses (NUHM1). In addition to constraints from low-energy precision observables and the cosmological dark matter density, we take into account the LHC exclusions from searches in jets plus missing transverse energy signatures with about 5 fb\(^{−1}\) of integrated luminosity. We also include the most recent upper bound on the branching ratio B\(_s\) → μμ from LHCb. Furthermore, constraints from and implications for direct and indirect dark matter searches are discussed. The best fit of the CMSSM prefers a light Higgs boson just above the experimentally excluded mass. We find that the description of the low-energy observables, (g − 2)\(_μ\) in particular, and the non-observation of SUSY at the LHC become more and more incompatible within the CMSSM. A potential SM-like Higgs boson with mass around 126 GeV can barely be accommodated. Values for B(B\(_s\)→μμ) just around the Standard Model prediction are naturally expected in the best fit region. The most-preferred region is not yet affected by limits on direct WIMP searches, but the next generation of experiments will probe this region. Finally, we discuss implications from fine-tuning for the best fit regions.
Background
Therapeutic vaccination directed to induce an anti-tumoral T-cell response is a field of extensive investigation in the treatment of melanoma. However, many vaccination trials in melanoma failed to demonstrate a correlation between the vaccine-specific immune response and therapy outcome. This has been mainly attributed to immune escape by antigen loss, rendering us in the need of new vaccination targets.
Patients and methods
This phase-II trial investigated a peptide vaccination against survivin, an oncogenic inhibitor-of-apoptosis protein crucial for the survival of tumor cells, in HLA-A1/-A2/-B35-positive patients with treatment-refractory stage-IV metastatic melanoma. The study endpoints were survivin-specific T-cell reactivity (SSTR), safety, response, and survival (OS).
Results
Sixty-one patients (ITT) received vaccination therapy using three different regimens. 55 patients (PP) were evaluable for response and survival, and 41/55 for SSTR. Patients achieving progression arrest (CR + PR + SD) more often showed SSTRs than patients with disease progression (p = 0.0008). Patients presenting SSTRs revealed a prolonged OS (median 19.6 vs. 8.6 months; p = 0.0077); multivariate analysis demonstrated SSTR as an independent predictor of survival (p = 0.013). The induction of SSTRs was associated with gender (female vs. male; p = 0.014) and disease stage (M1a/b vs. M1c; p = 0.010), but not with patient age, HLA type, performance status, or vaccination regimen.
Conclusion
Survivin-specific T-cell reactivities strongly correlate with tumor response and patient survival, indicating that vaccination with survivin-derived peptides is a promising treatment strategy in melanoma.

Background: Tardigrades are multicellular organisms, resistant to extreme environmental changes such as heat, drought, radiation and freezing. They outlast these conditions in an inactive form (tun) to escape damage to cellular structures and cell death. Tardigrades are apparently able to prevent or repair such damage and are therefore a crucial model organism for stress tolerance. Cultures of the tardigrade Milnesium tardigradum were dehydrated by removing the surrounding water to induce tun formation. During this process and the subsequent rehydration, metabolites were measured in a time series by GC-MS. Additionally expressed sequence tags are available, especially libraries generated from the active and inactive state. The aim of this integrated analysis is to trace changes in tardigrade metabolism and identify pathways responsible for their extreme resistance against physical stress. Results: In this study we propose a novel integrative approach for the analysis of metabolic networks to identify modules of joint shifts on the transcriptomic and metabolic levels. We derive a tardigrade-specific metabolic network represented as an undirected graph with 3,658 nodes (metabolites) and 4,378 edges (reactions). Time course metabolite profiles are used to score the network nodes showing a significant change over time. The edges are scored according to information on enzymes from the EST data. Using this combined information, we identify a key subnetwork (functional module) of concerted changes in metabolic pathways, specific for de- and rehydration. The module is enriched in reactions showing significant changes in metabolite levels and enzyme abundance during the transition. It resembles the cessation of a measurablemetabolism (e.g. glycolysis and amino acid anabolism) during the tun formation, the production of storage metabolites and bioprotectants, such as DNA stabilizers, and the generation of amino acids and cellular components from monosaccharides as carbon and energy source during rehydration. Conclusions: The functional module identifies relationships among changed metabolites (e.g. spermidine) and reactions and provides first insights into important altered metabolic pathways. With sparse and diverse data available, the presented integrated metabolite network approach is suitable to integrate all existing data and analyse it in a combined manner.
It is of interest to define bacterial toxin biochemical properties to use them as molecular-syringe devices in order to deliver enzymatic activities into host cells. Binary toxins of the AB7/8-type are among the most potent and specialized bacterial protein toxins. The B subunits oligomerize to form a pore that binds with high affinity host cell receptors and the enzymatic A subunit. This allows the endocytosis of the complex and subsequent injection of the A subunit into the cytosol of the host cells. Here we report that the addition of an N-terminal His6-tag to different proteins increased their binding affinity to the protective antigen (PA) PA63-channels, irrespective if they are related (C2I) or unrelated (gpJ, EDIN) to the AB7/8-family of toxins. His6-EDIN exhibited voltage-dependent increase of the stability constant for binding by a factor of about 25 when the trans-side corresponding to the cell interior was set to 270 mV. Surprisingly, the C. botulinum toxin C2II-channel did not share this feature of PA63. Cell-based experiments demonstrated that addition of an N-terminal His6-tag promoted also intoxication of endothelial cells by C2I or EDIN via PA63. Our results revealed that addition of His6-tags to several factors increase their binding properties to PA63 and enhance the property to intoxicate cells.
Background: Hyperactivity is one of the core symptoms in attention deficit hyperactivity disorder (ADHD). However, it remains unclear in which way the motor system itself and its development are affected by the disorder. Movement-related potentials (MRP) can separate different stages of movement execution, from the programming of a movement to motor post-processing and memory traces. Pre-movement MRP are absent or positive during early childhood and display a developmental increase of negativity.
Methods: We examined the influences of response-speed, an indicator of the level of attention, and stimulant medication on lateralized MRP in 16 children with combined type ADHD compared to 20 matched healthy controls.
Results: We detected a significantly diminished lateralisation of MRP over the pre-motor and primary motor cortex during movement execution (initial motor potential peak, iMP) in patients with ADHD. Fast reactions (indicating increased visuo-motor attention) led to increased lateralized negativity during movement execution only in healthy controls, while in children with ADHD faster reaction times were associated with more positive amplitudes. Even though stimulant medication had some effect on attenuating group differences in lateralized MRP, this effect was insufficient to normalize lateralized iMP amplitudes.
Conclusions: A reduced focal (lateralized) motor cortex activation during the command to muscle contraction points towards an immature motor system and a maturation delay of the (pre-) motor cortex in children with ADHD. A delayed maturation of the neuronal circuitry, which involves primary motor cortex, may contribute to ADHD pathophysiology.
Primary osteoporosis is an age-related disease characterized by an imbalance in bone homeostasis. While the resorptive aspect of the disease has been studied intensely, less is known about the anabolic part of the syndrome or presumptive deficiencies in bone regeneration. Multipotent mesenchymal stem cells (MSC) are the primary source of osteogenic regeneration. In the present study we aimed to unravel whether MSC biology is directly involved in the pathophysiology of the disease and therefore performed microarray analyses of hMSC of elderly patients (79-94 years old) suffering from osteoporosis (hMSC-OP). In comparison to age-matched controls we detected profound changes in the transcriptome in hMSC-OP, e.g. enhanced mRNA expression of known osteoporosis-associated genes (LRP5, RUNX2, COL1A1) and of genes involved in osteoclastogenesis (CSF1, PTH1R), but most notably of genes coding for inhibitors of WNT and BMP signaling, such as Sclerostin and MAB21L2. These candidate genes indicate intrinsic deficiencies in self-renewal and differentiation potential in osteoporotic stem cells. We also compared both hMSC-OP and non-osteoporotic hMSC-old of elderly donors to hMSC of similar to 30 years younger donors and found that the transcriptional changes acquired between the sixth and the ninth decade of life differed widely between osteoporotic and non-osteoporotic stem cells. In addition, we compared the osteoporotic transcriptome to long term-cultivated, senescent hMSC and detected some signs for pre-senescence in hMSC-OP. Our results suggest that in primary osteoporosis the transcriptomes of hMSC populations show distinct signatures and little overlap with non-osteoporotic aging, although we detected some hints for senescence-associated changes. While there are remarkable inter-individual variations as expected for polygenetic diseases, we could identify many susceptibility genes for osteoporosis known from genetic studies. We also found new candidates, e.g. MAB21L2, a novel repressor of BMP-induced transcription. Such transcriptional changes may reflect epigenetic changes, which are part of a specific osteoporosis-associated aging process.
Natalizumab is a recombinant monoclonal antibody raised against integrin alpha-4 (CD49d). It is approved for the treatment of patients with multiple sclerosis (MS), a chronic inflammatory autoimmune disease of the CNS. While having shown high therapeutic efficacy, treatment by natalizumab has been linked to progressive multifocal leukoencephalopathy (PML) as a serious adverse effect. Furthermore, drug cessation sometimes induces rebound disease activity of unknown etiology. Here we investigated whether binding of this adhesion-blocking antibody to T lymphocytes could modulate their phenotype by direct induction of intracellular signaling events. Primary CD4+ T lymphocytes either from healthy donors and treated with natalizumab in vitro or from MS patients receiving their very first dose of natalizumab were analyzed. Natalizumab induced a mild upregulation of IL-2, IFN-c and IL-17 expression in activated primary human CD4+ T cells propagated ex vivo from healthy donors, consistent with a pro-inflammatory costimulatory effect on lymphokine expression. Along with this, natalizumab binding triggered rapid MAPK/ERK phosphorylation. Furthermore, it decreased CD49d surface expression on effector cells within a few hours. Sustained CD49d downregulation could be attributed to integrin internalization and degradation. Importantly, also CD4+ T cells from some MS patients receiving their very first dose of natalizumab produced more IL-2, IFN-c and IL-17 already 24 h after infusion. Together these data indicate that in addition to its adhesion-blocking mode of action natalizumab possesses mild direct signaling capacities, which can support a pro-inflammatory phenotype of peripheral blood T lymphocytes. This might explain why a rebound of disease activity or IRIS is observed in some MS patients after natalizumab cessation.
Spin-Bahn-Kopplung in Grenzschichten: Mikroskopische Zusammenhänge und Strategien zur Manipulation

Die vorliegende Arbeit befasst sich mit dem Einfluss der Spin-Bahn-Kopplung (SBK) auf die zweidimensionale elektronische Struktur von Festkörperoberflächen und -grenzflächen. Aufgrund der strukturellen Inversionsasymmetrie kann die SBK in derartigen Systemen eine Spinaufspaltung der elektronischen Zustände herbeiführen und eine charakteristische impulsabhängige Spinstruktur induzieren (Rashba-Effekt). Die Studien in dieser Arbeit sind zum einen darauf gerichtet, das physikalische Verständnis der mikroskopischen Zusammenhänge, die die Spinaufspaltung und die Spinorientierung elektronischer Zustände an Grenzflächen bestimmen, zu verbessern. Des Weiteren sollen Möglichkeiten zur Manipulation der SBK durch kontrollierte Variationen chemischer und struktureller Grenzflächenparameter erforscht werden. Als Modellsysteme für diese Fragestellungen dienen die isostrukturellen Oberflächenlegierungen BiCu2 und BiAg2, deren elektronische Struktur mittels winkelaufgelöster Photoelektronenspektroskopie (ARPES) und spinaufgelöster ARPES untersucht wird. Die Resultate der Experimente werden mithilfe von ab initio-Rechnungen und einfacheren Modellbetrachtungen interpretiert. Die Arbeit schließt mit einer ausblickenden Präsentation von Experimenten zu dem topologischen Isolator Bi2Se3(0001). Vergleichende ARPES-Messungen zu BiAg2/Ag(111) und BiCu2/Cu(111) zeigen, dass bereits geringe Unterschiede in der Grenzschichtmorphologie die Größe der Spinaufspaltung in der elektronischen Struktur um ein Vielfaches verändern können. Zudem belegen spinaufgelöste Experimente eine invertierte Spinorientierung der elektronischen Zustände in BiCu2 im Vergleich mit dem Referenzsystem Au(111). Beide Resultate können durch eine theoretische Analyse des Potentialprofils und der elektronischen Ladungsverteilung senkrecht zu der Grenzfläche in Kombination mit einfachen Modellbetrachtungen verstanden werden. Es stellt sich heraus, dass Asymmetrien in der Ladungsverteilung das direkte mikroskopische Bindeglied zwischen der Spinstruktur des elektronischen Systems und den strukturellen und chemischen Parametern der Grenzschicht bilden. Weitergehende ARPES-Experimente zeigen, dass die spinabhängige elektronische Struktur zudem signifikant durch die Symmetrie des Potentials parallel zu der Grenzflächenebene beeinflusst wird. Eine Manipulation der SBK wird in BiCu2 durch die Deposition von Adatomen erreicht. Hierdurch gelingt es, die Spinaufspaltung sowohl zu vergrößern (Na-Adsorption) als auch zu verringern (Xe-Adsorption). ARPES-Experimente an dem ternären Schichtsystem BiAg2/Ag/Au(111) belegen erstmalig eine Kopplung zwischen elektronischen Bändern mit entgegengesetztem Spincharakter in einem zweidimensionalen System mit Spinaufspaltung (Interband-Spin-Bahn-Kopplung). Der zugrundeliegende Kopplungsmechanismus steht in bemerkenswerter Analogie zu den Auswirkungen der SBK auf die spinpolarisierte elektronische Struktur in ferromagnetischen Systemen. Variationen in der Schichtdicke des Ag-Substratfilms erlauben es, die Stärke der Interband-SBK zu manipulieren.
The thesis enhances the strategy of non-destructive fluorescence read-out in rylene bisimide-diarylethene containing photochromic systems. The fluorescence of the emitter unit is quenched by a photoinduced electron transfer only to one of the isomeric forms of the photochrome. The driving force of the fuorescence-quenching electron transfer was calculated by the help of the Rehm-Weller equation. The novel photochromic systems satisfy the necessary requirements for non-destructive read-out in write/read/erase fluorescent memory devices.
Background: In higher plants, a diverse array of developmental and growth-related processes is regulated by the plant hormone auxin. Recent publications have proposed that besides the well-characterized Auxin Response Factors (ARFs) that bind Auxin Response Elements (AuxREs), also members of the bZIP- and MYB-transcription factor (TF) families participate in transcriptional control of auxin-regulated genes via bZIP Response Elements (ZREs) or Myb Response Elements (MREs), respectively. Results: Applying a novel bioinformatic algorithm, we demonstrate on a genome-wide scale that singular motifs or composite modules of AuxREs, ZREs, MREs but also of MYC2 related elements are significantly enriched in promoters of auxin-inducible genes. Despite considerable, species-specific differences in the genome structure in terms of the GC content, this enrichment is generally conserved in dicot (Arabidopsis thaliana) and monocot (Oryza sativa) model plants. Moreover, an enrichment of defined composite modules has been observed in selected auxin-related gene families. Consistently, a bipartite module, which encompasses a bZIP-associated G-box Related Element (GRE) and an AuxRE motif, has been found to be highly enriched. Making use of transient reporter studies in protoplasts, these findings were experimentally confirmed, demonstrating that GREs functionally interact with AuxREs in regulating auxin-mediated transcription. Conclusions: Using genome-wide bioinformatic analyses, evolutionary conserved motifs have been defined which potentially function as AuxRE-dependent coupling elements to establish auxin-specific expression patterns. Based on these findings, experimental approaches can be designed to broaden our understanding of combinatorial, auxin-controlled gene regulation.
Synthese von Dextran-umhüllten Eisenoxid-Nanopartikeln als Kontrastmittel für die MR-Tomographie

Durch Fällung von Eisen(II)- und Eisen(III)-salzen wurden Dextran-umhüllte Eisenoxid-Nanopartikel (SPIOs) und durch anschließende Umsetzung mit Epichlorhydrin und Ammoniak CLIOs gewonnen. An diesen Kolloiden wurden niedermolekulare Moleküle wie Diamine oder Bernsteinsäureanhydrid als Linker angebracht. Ein weiterer Aspekt dieser Arbeit stellt die Anbindung von Fluoreszenzmarkern und Antikörpern an der Partikeloberfläche sowie deren spektroskopische Untersuchung dar.
Die invasive Aspergillose ist eine schwerwiegende opportunistische systemische Infektion bei immunsupprimierten Patienten. In dieser Arbeit wurden verschiedene PCR-Verfahren und ein ELISA-Verfahren zum Antigennachweis in Hinblick auf Sensitivität, Sensibilität und positiver bzw. negativer Vorhersagewahrscheinlichkeit verglichen. Die konventionelle 18S-PCR ist ein panfungales Assay und wegen zahlreicher falsch-positiver Ergebnisse nicht geeignet zur Frühdiagnose. Die ITS-PCR hat sich aufgrund guter Spezifität als vielversprechend erwiesen, muss aber noch mit größeren Fallzahlen evaluiert werden. Der Antigennachweis (Platelia, Fa. Bio-Rad) weist eine hohe Sensitivität und negativen prädiktiven Wert auf. Als vielversprechend dürfte zukünftig eine Kombination aus PCR und Antigennachweisverfahren gelten.
Background. Nitric oxide (NO)-signal transduction plays an important role in renal ischemia/reperfusion (I/R) injury. NO produced by endothelial NO-synthase (eNOS) has protective functions whereas NO from inducible NO-synthase (iNOS) induces impairment. Rosiglitazone (RGZ), a peroxisome proliferator-activated receptor (PPAR)-gamma agonist exerted beneficial effects after renal I/R injury, so we investigated whether this might be causally linked with NOS imbalance. Methods. RGZ (5 mg/kg) was administered i.p. to SD-rats (f) subjected to bilateral renal ischemia (60 min). Following 24 h of reperfusion, inulin-and PAH-clearance as well as PAH-net secretion were determined. Morphological alterations were graded by histopathological scoring. Plasma NOx-production was measured. eNOS and iNOS expression was analyzed by qPCR. Cleaved caspase 3 (CC3) was determined as an apoptosis indicator and ED1 as a marker of macrophage infiltration in renal tissue. Results. RGZ improves renal function after renal I/R injury (PAH-/inulin-clearance, PAH-net secretion) and reduces histomorphological injury. Additionally, RGZ reduces NOx plasma levels, ED-1 positive cell infiltration and CC3 expression. iNOS-mRNA is reduced whereas eNOS-mRNA is increased by RGZ. Conclusion. RGZ has protective properties after severe renal I/R injury. Alterations of the NO pathway regarding eNOS and iNOS could be an explanation of the underlying mechanism of RGZ protection in renal I/R injury.
Background
Among the modalities for lung imaging, proton magnetic resonance imaging (MRI) has been the latest to be introduced into clinical practice. Its value to replace X-ray and computed tomography (CT) when radiation exposure or iodinated contrast material is contra-indicated is well acknowledged: i.e. for paediatric patients and pregnant women or for scientific use. One of the reasons why MRI of the lung is still rarely used, except in a few centres, is the lack of consistent protocols customised to clinical needs.
Methods
This article makes non-vendor-specific protocol suggestions for general use with state-of-the-art MRI scanners, based on the available literature and a consensus discussion within a panel of experts experienced in lung MRI.
Results
Various sequences have been successfully tested within scientific or clinical environments. MRI of the lung with appropriate combinations of these sequences comprises morphological and functional imaging aspects in a single examination. It serves in difficult clinical problems encountered in daily routine, such as assessment of the mediastinum and chest wall, and even might challenge molecular imaging techniques in the near future.
Conclusion
This article helps new users to implement appropriate protocols on their own MRI platforms.

MRI of the lung is recommended in a number of clinical indications. Having a non-radiation alternative is particularly attractive in children and young subjects, or pregnant women.
Methods
Provided there is sufficient expertise, magnetic resonance imaging (MRI) may be considered as the preferential modality in specific clinical conditions such as cystic fibrosis and acute pulmonary embolism, since additional functional information on respiratory mechanics and regional lung perfusion is provided. In other cases, such as tumours and pneumonia in children, lung MRI may be considered an alternative or adjunct to other modalities with at least similar diagnostic value.
Results
In interstitial lung disease, the clinical utility of MRI remains to be proven, but it could provide additional information that will be beneficial in research, or at some stage in clinical practice. Customised protocols for chest imaging combine fast breath-hold acquisitions from a “buffet” of sequences. Having introduced details of imaging protocols in previous articles, the aim of this manuscript is to discuss the advantages and limitations of lung MRI in current clinical practice.
Conclusion
New developments and future perspectives such as motion-compensated imaging with self-navigated sequences or fast Fourier decomposition MRI for non-contrast enhanced ventilation- and perfusion-weighted imaging of the lung are discussed.

Background: The angiotensin II receptor subtype 2 (AT2 receptor) is ubiquitously and highly expressed in early postnatal life. However, its role in postnatal cardiac development remained unclear.
Methodology/Principal Findings: Hearts from 1, 7, 14 and 56 days old wild-type (WT) and AT2 receptor-deficient (KO) mice were extracted for histomorphometrical analysis as well as analysis of cardiac signaling and gene expression. Furthermore, heart and body weights of examined animals were recorded and echocardiographic analysis of cardiac function as well as telemetric blood pressure measurements were performed. Moreover, gene expression, sarcomere shortening and calcium transients were examined in ventricular cardiomyocytes isolated from both genotypes. KO mice exhibited an accelerated body weight gain and a reduced heart to body weight ratio as compared to WT mice in the postnatal period. However, in adult KO mice the heart to body weight ratio was significantly increased most likely due to elevated systemic blood pressure. At postnatal day 7 ventricular capillarization index and the density of \(\alpha\)-smooth muscle cell actin-positive blood vessels were higher in KO mice as compared to WT mice but normalized during adolescence. Echocardiographic assessment of cardiac systolic function at postnatal day 7 revealed decreased contractility of KO hearts in response to beta-adrenergic stimulation. Moreover, cardiomyocytes from KO mice showed a decreased sarcomere shortening and an increased peak Ca\(^{2+}\) transient in response to isoprenaline when stimulated concomitantly with angiotensin II.
Conclusion: The AT2 receptor affects postnatal cardiac growth possibly via reducing body weight gain and systemic blood pressure. Moreover, it moderately attenuates postnatal vascularization of the heart and modulates the beta adrenergic response of the neonatal heart. These AT2 receptor-mediated effects may be implicated in the physiological maturation process of the heart.
Die Malaria und andere Infektionskrankheiten sind immer noch die Haupttodesursache in Entwicklungsländern. Durch das jahrzehntelange Versäumnis, neue Wirkstoffe zu entwickeln, und durch die rasante Ausbreitung von Resistenzen gegen herkömmliche Medikamente sind in vielen Regionen der Erde besorgniserregende Zahlen über Neuinfektionen und Todesfälle zu beobachten. Die Suche nach neuen Wirkstoffen ist daher dringend erforderlich und die Hauptaufgabe des Sonderforschungsbereichs 630 an der Universität Würzburg. An diesem interdisziplinären Projekt beteiligt sich unsere Forschungsgruppe vor allem mit der Naturstoffklasse der Naphthylisochinolin-Alkaloide. Neben ihren interessanten strukturellen Eigenschaften haben mehrere Vertreter dieser Sekundärmetabolite vielversprechende Aktivitäten gegen Plasmodien, Leishmanien und Trypanosomen. Dioncophyllin C (24), das bisher wirksamste Naphthylisochinolin gegen P. falciparum, zeigt nicht nur eine exzellente Aktivität in vitro, sondern auch in vivo. In Kooperation mit der Forschergruppe von K. Baumann (Braunschweig) führte man QSAR-Studien durch, um die für die biologische Wirkung entscheidenden Strukturmerkmale zu identifizieren und neue vereinfachte Analoga der Leistruktur 24 vorzuschlagen. Ziel der vorliegenden Arbeit war aufbauend auf Vorarbeiten in unserer Gruppe die Darstellung von strukturell vereinfachten Derivaten des Naturstoffs 24. Die Ergebnisse der biologischen Untersuchungen sollten ausgewertet und somit neue Struktur-Wirkungs-Beziehungen aufgestellt werden. Weiterhin sollten auch Chinolinium-Salze, die man als Analoga der N,C-verknüpften Naphthylisochinoline ansehen kann, synthetisiert werden und innerhalb des SFB 630 und bei unseren Partnern am Schweizerischen Tropen- und Public-Health-Institut auf ihre biologische Aktivität untersucht werden. Man erhoffte sich neben möglichen antiinfektiven Eigenschaften auch Rückschlüsse auf Struktur-Wirkungs-Beziehungen. Zusätzlich sollte die synthetische und analytische chemische Expertise unseren Kooperationspartnern in zwei Projekten außerhalb des SFB 630 zur Verfügung gestellt werden. Dabei handelte es sich einerseits um die Strukturaufklärung von Biosyntheseintermediaten mit Hilfe der HPLC-NMR-Kopplung und andererseits um die Darstellung langkettiger Aldehyde für die biologische Untersuchung des Prä-Penetrationsprozesses eines getreideschädigenden Pilzes.
Das ACL ist eine der am häufigsten verletzten Strukturen des menschlichen Körpers bei Sportunfällen und die Anzahl der Rekonstruktionen wird weiterhin zunehmen. In den letzten Jahren wurden große Fortschritte in der Charakterisierung der biomechanischen Eigenschaften von Sehnen und Bändern erzielt und neue Ansätze auf dem Gebiet des Tissue Engineering eröffnen neue Möglichkeiten. In dieser Arbeit wurde ein ACL-Konstrukt aus biomaterialbasiertem Kollagen I entwickelt und getestet. Die gewonnenen Ergebnisse können als Grundlage für die Herstellung eines Kreuzbandkonstruktes verwendet werden.
Unter Verwendung fluoreszenzmarkierter Remorine der taxonomischen Gruppe 1b wurden Nanodomänen in Arabidopsis Plasmamembranen (PM) unter Verwendung hoch auflösender Laser Scanning-Systeme sichtbar gemacht. In diesen kompartimentierten Membranbereichen lagerten sich Sterol-abhängige Remorine aus verschiedenen Pflanzen-familien zusammen und zeigten dort Kolokalisation. Dies wurde statistisch belegt durch hohe Pearson und Spearman Korrelationskoeffizienten. Remorine konnten schließlich als pflanzliche Markerproteine für kompartimentierte Membranbereiche etabliert werden. Die Nanodomänen zeigten zu keinem Zeitpunkt laterale Bewegungen in der PM und scheinen sowohl von zytoskelettären Strukturen als auch von Komponenten der Zellwand stabilisiert zu werden. Möglicherweise spielen transmembrane Tetraspanine sowie GPI-verankerte SKU5-Proteine eine Rolle bei der stabilen Verankerung. Für zwei native Arabidopsis Remorine wurden posttranslationale Modifikationsstellen aufgedeckt, die der Anheftung dieser hydrophilen Proteine an die PM dienen. Weiterhin scheinen gleichartige Remorine miteinander zu interagieren. Beispielsweise waren im Zytosol lokalisierte Remorin-Mutanten bei einer gleichzeitigen Expression der entsprechenden Vollängenproteine erneut an der PM zu finden. Für die Remorine wurde postuliert, dass sie mit anderen Proteinen interagieren und dabei makromolekulare Strukturen ausbilden. Den Remorinen könnte daher eine Aufgabe bei der molekularen Organisation pflanzlicher Membrandomänen zukommen, indem sie ein filamentartiges Netzwerk innerhalb distinkter Domänen ausbilden, das möglicherweise zur Stabilität und Aufrechterhaltung dieser spezialisierten Bereiche beiträgt. Unter Einbeziehung der STED-Mikroskopie wurde eine empirische Größenverteilung von 97±4nm Durchmesser für PM-ständige Domänen in Arabidopsis ermittelt. Hinsichtlich der physiologischen Relevanz konnte gezeigt werden, dass die Domänen eine Rolle bei der ABA-vermittelten, kalziumabhängigen Regulation des Anionenkanals SLAH3 einnehmen. SLAH3 wird durch kalziumabhängige Kinasen aus der CDPK-Familie aktiviert, im Speziellen durch CPK21 und CPK23. Beide Kinasen werden durch die ABA-sensitiven Phosphatasen ABI1 und ABI2 reguliert. Die spezifisch stattfindenden Interaktionen zwischen SLAH3 und CPK21, sowie zwischen CPK21 und ABI1 waren auf Nanodomänen beschränkt und wurden durch die Methodik der bimolekularen Fluoreszenzkomplementation erstmals in planta nachgewiesen, mit Remorinen der taxonomischen Gruppe 1b als etablierte Markerproteine für Membrandomänen.
Background: Sterols and Sphingolipids form lipid clusters in the plasma membranes of cell types throughout the animal and plant kingdoms. These lipid domains provide a medium for protein signaling complexes at the plasma membrane and are also observed to be principal regions of membrane contact at the inception of infection. We visualized different specific fluorescent lipophilic stains of the both sphingolipid enriched and non-sphingolipid enriched regions in the plasma membranes of live protoplasts of Arabidopsis thaliana. Results: Lipid staining protocols for several fluorescent lipid analogues in plants are presented. The most emphasis was placed on successful protocols for the single and dual staining of sphingolipid enriched regions and exclusion of sphingolipid enriched regions on the plasma membrane of Arabidopsis thaliana protoplasts. A secondary focus was placed to ensure that these staining protocols presented still maintain cell viability. Furthermore, the protocols were successfully tested with the spectrally sensitive dye Laurdan. Conclusion: Almost all existing staining procedures of the plasma membrane with fluorescent lipid analogues are specified for animal cells and tissues. In order to develop lipid staining protocols for plants, procedures were established with critical steps for the plasma membrane staining of Arabidopsis leaf tissue and protoplasts. The success of the plasma membrane staining protocols was additionally verified by measurements of lipid dynamics by the fluorescence recovery after photobleaching technique and by the observation of new phenomena such as time dependent lipid polarization events in living protoplasts, for which a putative physiological relevance is suggested.
Lebensqualität bei Fabry-Patienten: Erhebung mit dem SF-36®Fragebogen Elisabeth Blohm Hintergrund: Der Morbus Fabry ist eine X-chromosomal vererbte, lysosomale Speichererkrankung bedingt durch den Mangel an dem Enzym α-Galaktosidase A. Die gesundheitsbezogene Lebensqualität von Fabry-Patienten ist im Vergleich zur Normalbevölkerung oder Patienten anderer chronischer Erkrankungen sowohl bei physischen als auch psychischen Aspekten reduziert. Es ist bekannt, dass für den Morbus Fabry typische Symptome, wie Schmerzen oder Dysfunktionen lebenswichtiger Organe, wie Herz- und Niereninsuffizienz, sowie frühzeitige Schlaganfälle zu einer signifikant verringerten gesundheitsbezogenen Lebensqualität beitragen. Fragestellung: Ziel dieser Arbeit war es, mit Hilfe des SF-36-Fragebogens die gesundheitsbezogene Lebensqualität einer großen Kohorte von Fabry-Patienten zum Zeitpunkt der ersten Vorstellung in einem spezialisierten Fabry Zentrum zu ermitteln. Gleichzeitig sollten begleitende Faktoren identifiziert werden, die mit den verschiedenen Dimensionen der psychischen und physischen Funktionsfähigkeit assoziiert sind. Dabei legten wir einen Schwerpunkt auf die Nierenfunktion. Methoden: Es wurden 99 Patienten mit Morbus Fabry eingeschlossen. Wir untersuchten die Daten des ersten Besuchs in unserem Fabry-Zentrum. Die Patientencharakteristika der Studienteilnehmer und die verschiedenen Skalen der HRQoL wurden über die CKD-Stadien unter Verwendung von ANOVA, Kruskal-Wallis-Test, χ²-Test und Fisher’s-exact-Test verglichen. Die mit den verschiedenen Dimensionen der HRQoL assoziierten Faktoren wurden mittels einer linearen Regressionsanalyse untersucht. Im multivariaten Modell wurden die folgenden Variablen in das Modell aufgenommen: Alter, Geschlecht, Nierenfunktion, Schmerzen, Schmerztherapie und vaskuläres Ereignis. Ergebnisse: Die meisten Patienten, besonders die meisten Frauen, hatten eine erhaltene Nierenfunktion, wohingegen Patienten mit eingeschränkter Nierenfunktion mit höherer Wahrscheinlichkeit männlich waren. Alle Patienten, die einer RRT erhielten, waren männlich; zwei von ihnen erhielten eine Nierentransplantation, sieben waren unter Dialyse. Eine eingeschränkte Nierenfunktion, besonders die Notwendigkeit einer RRT war über allen Skalen mit einer deutlichen Reduktion der HRQoL assoziiert. Desweiteren waren männliches Geschlecht, Schmerzen und Schmerztheapie signifikant und deutlich mit niedrigeren Werten in den SF-36-Skalen assoziiert. Im multivariaten Modell stellten sich eine eingeschränkte Nierenfunktion und Schmerzen als die Hauptfaktoren für reduzierte Werte in den physikalischen Kategorien heraus (körperliche Funktionsfähigkeit, körperliche Rollenfunktion, körperliche Schmerzen, allgemeine Gesundheitswahrnehmung und körperlicher Summenwert). Im Gegensatz dazu stellte sich heraus, dass die Notwendigkeit einer RRT mit reduzierter HRQoL in den psychischen und sozialen Kategorien assoziiert war. Schlussfolgerung: In dieser großen Kohorte von Fabry-Patienten aus einem Zentrum war die chronische Nierenerkrankung, besonders die Notwendigkeit einer RRT, ein entscheidender Faktor mir eine reduzierte HRQoL in physischen und psychisch/sozialen Aspekten des Lebens. Außerdem hatten Schmerzen eine unabhängige Beziehung mit niedrigeren Werten der physischen Skalen. Neben der Enzymersatztherapie könnten eine optimale Behandlung der Nierenerkrankung, sowie eine effektive Schmerztherapie helfen, die HRQoL von Fabry-Patienten zu verbessern.
Self-organized nanowires at semiconductor surfaces offer the unique opportunity to study electrons in reduced dimensions. Notably the dimensionality of the system determines it’s electronic properties, beyond the quasiparticle description. In the quasi-one-dimensional (1D) regime with weak lateral coupling between the chains, a Peierls instability can be realized. A nesting condition in the Fermi surface leads to a backfolding of the 1D electron band and thus to an insulating state. It is accompanied by a charge density wave (CDW) in real space that corresponds to the nesting vector. This effect has been claimed to occur in many surface-defined nanowire systems, such as the In chains on Si(111) or the Au reconstructions on the terraced Si(553) and Si(557) surfaces. Therefore a weak coupling between the nanowires in these systems has to be concluded. However theory proposes another state in the perfect 1D limit, which is completely destroyed upon slight coupling to higher dimensions. In this so-called Tomonaga-Luttinger liquid (TLL) state, the quasiparticle description of the Fermi liquid breaks down. Since the interaction between the electrons is enhanced due to the strong confinement, only collective excitations are allowed. This leads to novel effects like spin charge separation, where spin and charge degrees of freedom are decoupled and allowed to travel independently along the 1D-chain. Such rare state has not been realized at a surface until today. This thesis uses a novel approach to realize nanowires with improved confinement by studying the Au reconstructed Ge(001) surface. A new cleaning procedure using piranha solution is presented, in order to prepare a clean and long-range ordered substrate. To ensure optimal growth of the Au nanowires the phase diagram is extensively studied by scanning tunneling microscopy (STM) and low energy electron diffraction (LEED). The structural elements of the chains are revealed and described in high detail. Remarkably a structural phase transition of the delicate wire structure is found to occur above room temperature. Due to the lack of energy gaps a Peierls transition can be excluded as its origin. The transition is rather determined as 3D Ising type and therefore includes the substrate as well. Two hallmark properties of a TLL are found in the Au/Ge(001) wires by spectroscopic studies: Power-law suppression of the density of states (DOS) and universal scaling. This impressively proves the existence of a TLL in these chains and opens up a gateway to an atomic playground. Local studies and manipulations of a TLL state become possible for the first time. These comprise (i) doping by alkaline atoms, (ii) studies on chain ends and (iii) tunable coupling between the chains by additional Au atoms. Most importantly these manipulations offer input and test for theoretical models and predictions, and are thereby ultimately advancing the field of correlated electrons.
Ziel der vorliegenden Arbeit ist es darzustellen, wie groß das psychosoziale Unterstützungsbedürfnis von Patienten mit maligner Krebsdiagnose ist und welche multivariablen Zusammenhänge mit diesem Bedürfnis bestehen. Die dazu nötigen Daten wurden mit einem Selbsteinschätzungsfragebogen erhoben, der im Rahmen einer multizentrisch angelegten Querschnittsstudie zum Thema „Prävalenz psychosozialer Belastungen, psychischer Störungen und ihr Behandlungsbedarf bei Tumorpatienten“ in fünf Zentren in Deutschland entwickelt wurde. Die Stichprobe umfasste 386 Patienten im Alter von 18 bis 75 Jahren, die deutsch sprachen und an einer malignen Krebserkrankung litten. Das Unterstützungsbedürfnis der Patienten wurde in Bezug auf geschlechtsspezifische Unterschiede, Alter, Setting und vorhandene psychosoziale Unterstützung durch Freunde und Familie untersucht. Die Ergebnisse decken sich weitgehend mit denen der bisherigen Forschung. So gab ungefähr ein Drittel der Patienten einen Wunsch nach psychosozialer Unterstützung an. Weiterhin gaben Patienten, die sich psychisch belastet fühlten, häufiger einen Wunsch nach psychosozialer Unterstützung an als Patienten, die sich nicht belastet fühlten. Patienten mit bekannter Metastasierung oder unklarem Metastasierungsstatus äußerten im Vergleich zu Patienten mit lokalisierter Tumorerkrankung weniger häufig den Wunsch nach psychosozialer Unterstützung. Patienten, die bereits vor dem Zeitpunkt der Befragung psychosoziale Unterstützung in Anspruch genommen hatten, äußerten signifikant häufiger ein Unterstützungsbedürfnis als Patienten, die bislang keine psychosoziale Unterstützung in Anspruch genommen hatten. Alter, Geschlecht, Setting und vorhandene Unterstützung durch Freunde und Familie zeigten keinen signifikanten Zusammenhang mit dem Unterstützungsbedürfnis. Insgesamt äußert mit ungefähr einem Drittel der Patienten ein großer Anteil des Kollektivs ein Unterstützungsbedürfnis. Dies sollte die Konsequenz nach sich ziehen, dass Patienten, die durch ihre Tumorerkrankung nicht nur körperlich, sondern auch seelisch belastet sind, eine entsprechende Behandlung zuteil oder zumindest angeboten wird.
Einfluss von Monozyten auf Heilungsvorgänge und kardiale Thromboembolien nach einem Myokardinfarkt

Beim Myokardinfarkt führt die Unterbrechung der Blutversorgung zur Nekrose von Myozyten. Hierauf werden im Rahmen der Entzündungsreaktion verschiedene Reaktionen angestoßen, die dazu dienen sollen, die Integrität und Funktion des Herzens zu erhalten. Einen wichtigen Anteil an der Inflammationsreaktion haben die Monozyten, deren Funktion in diesem Rahmen bis jetzt nur wenig erforscht ist. In dieser Studie wurde die Rolle von Monozyten auf die Heilungsvorgänge nach einem Infarkt untersucht. Durch die Gabe von Clodronat-Liposom wurden in der Behandlungsgruppe die Monozyten ausgeschaltet. Es zeigte sich, dass die Mortalität in der Behandlungsgruppe aufgrund der Bildung eines linksventrikulären Thrombus deutlich erhöht ist. Dieser war bereits innerhalb der ersten 24h in den echokardiographischen Untersuchungen sichtbar. Veränderungen im Gerinnungssystem konnten als Ursache ausgeschlossen werden. Durch eine CD-31-Färbung wurde deutlich, dass in den infarzierten Herzen der Behandlungsgruppe ein Defekt im Endokard entstanden war, der als Ursache für die Entstehung der Thromben gewertet werden kann. Zudem wurde in der Behandlungsgruppe neben einem verschlechterten Abräumvorgang eine signifikant erniedrigte Kollagen-1-Produktion, ein erhöhtes MMP-9, ein leicht erniedrigtes TGF sowie erhöhtes VEGF gemessen. In dieser Studie wurde somit gezeigt, dass Monozyten ein essentieller Bestandteil der Heilungsvorgänge nach einem Infarkt sind. Durch eine eingeschränkte Monozytenfunktion wird zudem aufgrund von Defekten im Endothel die Bildung eines linksventrikulären Thrombus und folgender thromboembolischer Ereignisse begünstigt.
Werkstattbericht Hybridedition zu Goethes Faust. Es handelt sich dabei um ein Gemeinschaftsprojekt des Freien deutsche Hochstifts, des Goethe- und Schiller Archivs Weimar und der Universität Würzburg. Der Aufsatz befasst sich mit dem derzeitigen Entwicklungsstand der Edition, deren Konzeption als Hybridedition und verschiedenen Aspekten der Datenmodellierung.
Diffusionsgewichtete MRT Bildgebung bei Erkrankungen im Zahn-Mund-Kiefer/Hals-Nasen-Ohren Bereich

Zielsetzung: Die Zielsetzung dieser Arbeit war es zu beurteilen, inwieweit die Diffusionsgewichtete MRT Bildgebung bei der Differenzierung zwischen malignen und benignen Erkrankungen im Kopf-Hals Bereich nützlich sein kann. Außerdem ging es um die Sammlung der mittleren ADC-Werte von verschiedenen Diagnosen. Material und Methoden: Es wurden diffusionsgewichtete Bilder mit einer SE-EPI-Sequenz (b-Wert Paarung von 50 und 800 s/mm2, 1,5T) verglichen. 53 Plattenepithelcarzinome (PeCa), 21 benigne Befunde, 6 Lymphome, 6 Hämangiome, 6 Zysten Ergebnis: Die mittlere ADC- Wert (MW x10-3mm²/s +/-SD) der PeCa betrug 0.967 (0.118), der benignen Läsionen 1.458 (0.371), der Lymphome 0.649 (0.167), der Hämangiome 1.654 (0.254), der Zysten 2.310 (0.655) Der mittlere ADC-Wert der PeCa war signifikant kleiner als der der benignen Läsionen. Ein ADC- Wert von 1.202x10-3mm²/s kann als Grenzwert für die Differenzierung zwischen benignen und malignen Läsionen im Kopf-Hals Bereich angesehen werden. Schlussfolgerung: Die ADC-Werte können für die Charakterisierung von Läsionen im Kopf-Hals Bereich genutzt werden.
Der konventionelle Stift-Stumpfaufbau ist seit Jahrzehnten eine prothetische Standardtherapie bei der Restauration von Zähnen, deren klinische Krone einen erheblichen Substanzverlust erlitten haben. Bei dieser Art der Restauration sind klinisch jedoch wiederkehrende Versagensmechanismen zu beobachten, die negativen Einfluss auf Überlebensraten der restaurierten Zähne haben. Die vorliegende Untersuchung beschäftigt sich mit der Frage, ob diesen Versagensarten durch eine neuartige Verankerung, die sich anstelle einer kraftschlüssigen eine formschlüssige Verbindung zunutze macht, eliminiert werden können und wie ein geeignetes Verankerungselement beschaffen sein muss. Dazu wurde zunächst aus theoretischen Überlegungen heraus ein mechanisches Ersatzmodell erstellt, das die Wirkung der angreifenden Kräfte auf die Einheit aus restaurierter Zahnwurzel und Stift-Stumpfaufbau vereinfacht darstellt. Anschließend wurden die aus dem klinischen Alltag bekannten Versagensarten als Grundlage für die Designänderungen am Verankerungselement verwendet. An die Stelle eines langen und dünnen Wurzelstiftes tritt damit ein kurzer, dicker Stift, an dessen Ende ein invers konisches Verbindungselement angebracht ist. Dieses greift in eine ebenfalls invers konische Kavität, die durch besondere Werkzeuge in die Zahnwurzel präpariert wird. Über eine Einfassung, die den klassischen Fassreifen ersetzt, werden mechanische Kräfte auf das Dentin übertragen. Dieses Design wurde in In-Vitro-Versuchen getestet und optimiert, während die so gewonnenen Ergebnisse (Lastniveaus und Bruchmechanik) als Grundlage für Finite-Elemente-Simulationen dienten, mit deren Hilfe die Spannungsverteilung in der Zahnwurzel unter mechanischer Belastung sichtbar gemacht wurde. Der Zyklus aus FEM und Bruchversuchen erlaubte es, innerhalb weniger Iterationen einen Anker zu entwickeln, der mittlerweile als CE-zertifiziertes Medizinprodukt im klinischen Alltag Anwendung findet. Aus dem geometrischen Ersatzmodell konnte weiterhin die klinische Indikation für einen Stift-Stumpfaufbau verfeinert werden – es liefert auch eine mögliche Erklärung für die Ursache der teilweise großen Diskrepanzen zwischen verschiedenen Ergebnissen in der Literatur.

Daher entschied sich die UB Würzburg für die Produktion eines Imagefilms. Der Artikel bietet einen praxisorientierten Überblick über die verschiedenen Arbeitsschritte, die bei der Erstellung eines Imagefilms notwendig sind und gibt Tipps für die erfolgreiche Produktion.
The 7th International Symposium on Neuroprotection and Neurorepair was held from May 2nd to May 5th, 2012 in Potsdam, Germany. The symposium, which directly continues the successful Magdeburg meeting series, attracted over 330 colleagues from 29 countries to discuss recent findings and advances in the field. The focus of the 2012 symposium was widened from stroke and traumatic brain injury to neurodegenerative diseases, notably dementia, and more generally the ageing brain. Thereby, emphasis was given on neurovascular aspects of neurodegeneration and stroke including the blood–brain barrier, recent findings regarding the pathomechanism of Alzheimer’s disease, and brain imaging approaches. In addition, neurobiochemical aspects of neuroprotection, the role of astrogliosis, the clinical progress of cell-based approaches as well as translational hurdles and opportunities were discussed in-depth. This review summarizes some of the most stimulating discussions and reports from the meeting.
Multiple fluorescence in situ hybridization is the method of choice for studies aimed at determining simultaneous production of signal transduction molecules and neuromodulators in neurons. In our analyses of the monoamine receptor mRNA expression of peptidergic neurons in the rat telencephalon, double tyramide-signal-amplified fluorescence in situ hybridization delivered satisfactory results for coexpression analysis of neuropeptide Y (NPY) and serotonin receptor 2C (5-HT2C) mRNA, a receptor subtype expressed at high-to-moderate abundance in the regions analyzed. We first developed a double labeling protocol combining fluorescence and chromogenic in situ hybridization and subsequently expanded this variation to combine double fluorescence and chromogenic in situ hybridization for triple labelings. With this method, we documented expression of 5-HT2C and/or 5-HT1A in subpopulations of telencephalic NPY-producing neurons. The method developed in the present study appears suitable for conventional light and fluorescence microscopy, combines advantages of fluorescence and chromogenic in situ hybridization protocols and thus provides a reliable non-radioactive alternative to previously published multiple labeling methods for coexpression analyses in which one mRNA species requires highly sensitive detection.
Multiple fluorescence in situ hybridization is the method of choice for studies aimed at determining simultaneous production of signal transduction molecules and neuromodulators in neurons. In our analyses of the monoamine receptor mRNA expression of peptidergic neurons in the rat telencephalon, double tyramide-signal-amplified fluorescence in situ hybridization delivered satisfactory results for coexpression analysis of neuropeptide Y (NPY) and serotonin receptor 2C (5-HT2C) mRNA, a receptor subtype expressed at high-to-moderate abundance in the regions analyzed. Recent development of proteomic approaches and generation of large-scale proteomic datasets calls for new methods for biological interpretation of the obtained results. Systems biological approaches such as integrated network analysis and functional module search have become an essential part of proteomic investigation. Proteomics is especially applied in anucleate cells such as platelets. The underlying molecular mechanisms of platelet activation and their pharmacological modulation are of immense importance for clinical research. Advances in platelet proteomics have provided a large amount of proteomic data, which has not yet been comprehensively investigated in a systems biological perspective. To this end, I assembled platelet specific data from proteomic and transcriptomic studies by detailed manual curation and worked on the generation of a comprehensive human platelet repository for systems biological analysis of platelets in the functional context of integrated networks (PlateletWeb) (http:/PlateletWeb.bioapps.biozentrum.uni-wuerzburg.de). I also added platelet-specific experimentally validated phosphorylation data and generated kinase predictions for 80% of the newly identified platelet phosphosites. The combination of drug, disease and pathway information with phosphorylation and interaction data makes this database the first integrative platelet platform available for platelet research. PlateletWeb contains more than 5000 platelet proteins, which can also be analyzed and visualized in a network context, allowing identification of all major signaling modules involved in platelet activation and inhibition. Using the wealth of integrated data I performed a series of platelet-specific analyses regarding the platelet proteome, pathways, drug targets and novel platelet phosphorylation events involved in crucial signaling events. I analyzed the statistical enrichment of known pathways for platelet proteins and identified endocytosis as a highly represented pathway in platelets. Further results revealed that highly connected platelet proteins are more often targeted by drugs. Using integrated network analysis offered by PlateletWeb, I analyzed the crucial activation signaling pathway of adenosine diphosphate (ADP), visualizing how the signal flow from receptors to effectors is maintained. My work on integrin inside-out signaling was also based on the integrated network approach and examined new platelet-specific phosphorylation sites and their regulation using kinase predictions. I generated hypothesis on integrin signaling, by investigating the regulation of Ser269 phosphorylation site on the docking protein 1 (DOK1). This phosphorylation site may influence the inhibiting effect of DOK1 on integrin a2bb3. Extending the integrated network approach to further cell lines, I used the assembled human interactome information for the analysis of functional modules in cellular networks. The investigation was performed with a previously developed module detection algorithm, which finds maximum-scoring subgraphs in transcriptomic datasets by using assigned values to the network nodes. We extended the algorithm to qualitative proteomic datasets and enhanced the module search by adding functional information to the network edges to concentrate the solution onto modules with high functional similarity. I performed a series of analyses to validate its performance in small-sized (virus-infected gastric cells) and medium-sized networks (human lymphocytes). In both cases the algorithm extracted characteristic modules of sample proteins with high functional similarity. The functional module search is especially useful in site-specific phosphoproteomic datasets, where kinase regulation of the detected sites is often sparse or lacking. Therefore, I used the module detection algorithm in quantitative phosphoproteomic datasets. In a platelet phosphorylation dataset, I presented a pipeline for network analysis of detected phosphorylation sites. In a second approach, the functional module detecting algorithm was used on a phosphoproteome network of human embryonic stem cells, in which nodes represented the maximally changing phosphorylation sites in the experiment. Additional kinases from the human phosphoproteome in PlateletWeb were included to the network to investigate the regulation of the signal flow. Results indicated important phosphorylation sites and their upstream kinases and explained changes observed in embryonic stem cells during differentiation. This work presents novel approaches for integrated network analysis in cells and introduces for the first time a systematic biological investigation of the human platelet proteome based on the platelet-specific knowledge base PlateletWeb. The extended methods for optimized functional module detection offer an invaluable tool for exploring proteomic datasets and covering gaps in complex large-scale data analysis. By combining exact module detection approaches with functional information data between interacting proteins, characteristic functional modules with high functional resemblance can be extracted from complex datasets, thereby focusing on important changes in the observed networks.
Das Renin-Angiotensin-Aldosteron-System (RAAS) reguliert den Blutdruck und den Wasser- und Elektrolythaushalt des Körpers. Angiotensin II (Ang II), das aktive Peptid des RAAS, bewirkt eine Vasokonstriktion und in höheren Konzentrationen Bluthochdruck. Epidemiologische Studien haben gezeigt, dass eine Verbindung zwischen Hypertonie und dem gehäuften Auftreten von Krebs besteht. Eine Metaanalyse von 13 Fall-Kontroll-Studien konnte einen Zusammenhang zwischen Hypertonie und einem erhöhten Risiko, an einem Nierenzellkarzinom zu erkranken nachweisen. In vitro-Studien und Studien an der isolierten Niere konnten bereits genotoxische Effekte des blutdruckregulierenden Hormons Ang II zeigen. Zielsetzung dieser Arbeit war es, zunächst in vivo zu prüfen, ob steigende Ang II-Konzentrationen einen Einfluss auf die genomische Stabilität von Nieren- und Herzzellen besitzen. Hierzu wurden im Dosisversuch männliche C57BL/6-Mäuse mit osmotischen Minipumpen ausgestattet, die Ang II in vier verschiedenen Konzentrationen zwischen 60 ng/kg min und 1 µg/kg min über einen Zeitraum von 28 Tagen abgeben sollten. Während des Versuchszeitraums fanden regelmäßige, nicht-invasive Blutdruckmessungen an der Maus statt. Die Behandlung mit Ang II führte zu einem signifikanten Anstieg des Blutdrucks und zu histopathologischen Veränderungen der Glomeruli und des Tubulussystems, was sich in einer verschlechterten Albumin-Ausscheidung wiederspiegelte. Außerdem induzierte die Behandlung mit Ang II die dosisabhängige Bildung von reaktiven Sauerstoffspezies, DNA-Doppelstrangbrüchen und oxidativer DNA-Schäden. Diese Parameter waren bereits in Tieren erhöht, die keinen Bluthochdruck entwickelten und stiegen mit der höchsten Ang II-Konzentration noch an, obwohl hier im Vergleich zur Vorgängergruppe, die eine geringere Ang II-Konzentration erhielt, kein höherer Blutdruck vorlag. Diese Beobachtung deutet auf eine mögliche Unabhängigkeit des entstandenen Schadens vom Bluthochdruck hin und lenkt die Aufmerksamkeit auf Ang II als genomschädigenden Faktor. Der folgende Interventionsversuch sollte Aufschluss über die mögliche blutdruckunabhängige genomschädigende Wirkung von Ang II geben. Dazu wurden C57BL/6-Mäuse neben der Ang II-Behandlung in einer Konzentration von 600 ng/kg min zusätzlich über einen Zeitraum von 28 Tagen mit 5 verschiedenen Substanzen behandelt: Candesartan, Ramipril, Hydralazin, Eplerenon und Tempol. Candesartan ist ein Ang II-Rezeptor-Antagonist, der selektiv den AT1-Rezeptor blockiert. Ramipril wirkt als Hemmer des Angiotensin-Konversions-Enzyms und verhindert die Bildung von endogenem Ang II aus Ang I. Hydralazin, als Vasodilatator, greift nicht in das Renin-Angiotensin-Aldosteron-System ein. Eplerenon blockiert als selektiver Aldosteronantagonist den Mineralkortikoidrezeptor. Tempol wirkt als Antioxidans. Die Behandlung mit Ang II in einer Konzentration von 600 ng/kg min im Interventionsversuch führte zur Hochregulierung der NADPH-Oxidase 4 und zur Produktion reaktiver Sauerstoffspezies in der Niere und im kardiovaskulären Gewebe. Der entstandene oxidative Stress führte wiederum zu DNA-Schäden und einer Aktivierung der Transkriptionsfaktoren Nrf2 und NF-B. Nrf2-vermittelt wurde die Induktion antioxidativer Gene ausgelöst, was allerdings nicht ausreichend war, um vor Ang II-induzierten ROS und DNA-Schäden zu schützen. Eine längerfristige NF-B-Aktivierung durch hohe Ang II-Spiegel kann das Überleben und die Proliferation von Zellen, die DNA-Schäden in Form von Doppelstrangbrüchen tragen, fördern, was eine Tumor-initiierende Wirkung haben könnte. Die beschriebenen Effekte erhöhter Ang II-Spiegel konnten durch die Intervention mit dem AT1-Rezeptorblocker Candesartan verhindert werden, was die Beteiligung des Rezeptors nachweist. Eine blutdruckunabhängige, genomschädigende Wirkung von Ang II konnte leider durch die Intervention mit Hydralazin nicht verdeutlicht werden, da die erwünschte langfristige Blutdrucksenkung ausblieb. Allerdings zeigte die Intervention mit Tempol eine Abnahme an oxidativem Stress und DNA-Schäden trotz ausbleibender Blutdrucksenkung. Die Bedeutung von ROS in der Bildung von DNA-Schäden und die Unabhängigkeit dieser Schäden vom Blutdruck konnten somit hervorgehoben werden. Die Tatsache, dass die Intervention mit Ramipril den Blutdruck nicht senken konnte, der oxidative Stress und die DNA-Schäden durch mögliche antioxidative Eigenschaften aber vermindert wurden, unterstützt diese Beobachtung. Die Intervention mit Eplerenon führte zum Teil zu einer Verminderung an ROS und DNA-Schäden, brachte diese Parameter aber nicht auf Kontrollniveau zurück. Somit ist eine Beteiligung von Aldosteron nicht auszuschließen.
Susac syndrome, a rare but probably underdiagnosed combination of encephalopathy, hearing loss, and visual deficits due to branch retinal artery occlusion of unknown aetiology has to be considered as differential diagnosis in various conditions. Particularly, differentiation from multiple sclerosis is often challenging since both clinical presentation and diagnostic findings may overlap. Optical coherence tomography is a powerful and easy to perform diagnostic tool to analyse the morphological integrity of retinal structures and is increasingly established to depict characteristic patterns of retinal pathology in multiple sclerosis. Against this background we hypothesised that differential patterns of retinal pathology facilitate a reliable differentiation between Susac syndrome and multiple sclerosis. In this multicenter cross-sectional observational study optical coherence tomography was performed in nine patients with a definite diagnosis of Susac syndrome. Data were compared with age-, sex-, and disease duration-matched relapsing remitting multiple sclerosis patients with and without a history of optic neuritis, and with healthy controls. Using generalised estimating equation models, Susac patients showed a significant reduction in either or both retinal nerve fibre layer thickness and total macular volume in comparison to both healthy controls and relapsing remitting multiple sclerosis patients. However, in contrast to the multiple sclerosis patients this reduction was not distributed over the entire scanning area but showed a distinct sectorial loss especially in the macular measurements. We therefore conclude that patients with Susac syndrome show distinct abnormalities in optical coherence tomography in comparison to multiple sclerosis patients. These findings recommend optical coherence tomography as a promising tool for differentiating Susac syndrome from MS.
Intrathecal, Polyspecific Antiviral Immune Response in Oligoclonal Band Negative Multiple Sclerosis

Background: Oligoclonal bands (OCB) are detected in the cerebrospinal fluid (CSF) in more than 95% of patients with multiple sclerosis (MS) in the Western hemisphere. Here we evaluated the intrathecal, polyspecific antiviral immune response as a potential diagnostic CSF marker for OCB-negative MS patients.
Methodology/Principal Findings: We tested 46 OCB-negative German patients with paraclinically well defined, definite MS. Sixteen OCB-negative patients with a clear diagnosis of other autoimmune CNS disorders and 37 neurological patients without evidence for autoimmune CNS inflammation served as control groups. Antibodies against measles, rubella, varicella zoster and herpes simplex virus in paired serum and CSF samples were determined by ELISA, and virus-specific immunoglobulin G antibody indices were calculated. An intrathecal antibody synthesis against at least one neurotropic virus was detected in 8 of 26 (31%) patients with relapsing-remitting MS, 8 of 12 (67%) with secondary progressive MS and 5 of 8 (63%) with primary progressive MS, in 3 of 16 (19%) CNS autoimmune and 3 of 37 (8%) non-autoimmune control patients. Antibody synthesis against two or more viruses was found in 11 of 46 (24%) MS patients but in neither of the two control groups. On average, MS patients with a positive antiviral immune response were older and had a longer disease duration than those without.
Conclusion: Determination of the intrathecal, polyspecific antiviral immune response may allow to establish a CSF-supported diagnosis of MS in OCB-negative patients when two or more of the four virus antibody indices are elevated.
Die Reaktion der Verbindungen trans-[Pt{B(Br)(R)}Br(PCy3)2] mit Lewis-aciden Bromboranen BBr2(R) liefert Bromo-verbrückte, zweikernige Borylkomplexe. Sowie die jeweiligen Phosphan–Boran-Addukte Cy3P–BBr2(R). Die Reaktion von [Pt{B(X)(R)}(-X)(PCy3)]2 mit 4-Picolin erfolgt unter Koordination der Base am Boratom unter formaler Halogenidverschiebung zur Entstehung der ersten neutralen, basenstabilisierten Borylenkomplexe cis-[Pt{B(R)(4-Pic)}X2(PCy3)]. Durch oxidative Addition der B–Cl-Bindung von BCl3 an [Pt(PCy3)2] ist trans-[Pt(BCl2)Cl(PCy3)2] zugänglich, welches durch Reaktion mit Na[BArf4] zum kationischen Borylkomplex trans-[Pt(BCl2)(PCy3)2][BArf4] umgesetzt wird. Durch die strukturelle Charakterisierung von trans-[Pt{B(Br)(Fc)}Br(PiPr3)2] und trans-[Pt{B(Br)(Fc)}(PiPr3)2][BArf4] kann gezeigt werden, dass der Borylligand {B(Br)(Fc)} durch das {Pt(PiPr3)2}-Fragment in einem neutralen sowie in einem kationischen, T-förmigen Komplex stabilisiert werden kann. Die Reaktion von trans-[Pt{B(Br)(NMe2)}(PCy3)2][BArf4] mit Acetonitril führt zur Bildung des kationischen Acetonitrilkomplexes trans-[Pt{B(Br)(NMe2)}(NCMe)(PCy3)2][BArf4]. Durch die Reaktion von trans-[Pt{B(Br)(NMe2)}Br(PCy3)2] mit Na2[B12Cl12] im Verhältnis 2:1 und Zugabe von Acetonitril wird trans-[Pt{B(Br)(NMe2)}(NCMe)(PCy3)2]2[B12Cl12] als erste kationische, metallorganische Verbindung, die durch [B12Cl12]2− stabilisiert wird, erhalten. Die Abstraktion des Bromoliganden aus trans-[Pt{B(4-Pic)(NMe2)}Br(PCy3)2][BArf4] mittels Na[BArf4] führt zur Bildung des ersten dikationischen 14-Elektronenkomplexes trans- [Pt{B(NMe2)(4-Pic)}(PCy3)2][BArf4]2 mit einer freien Koordinationsstelle. Die Reaktion von trans-[Pt(BCat’)Br(PCy3)2] mit MeLi liefert trans-[Pt(BCat’)Me(PCy3)2]. Die Anwesenheit von Alkinen oder Bisphosphanen (P–P) beschleunigt die Reduktive Eliminierung von CatBMe. Die Reaktion von trans–[Pt(BCat’)Me(PCy3)2] mit Cat2B2 führt zu einem Reaktionsgemisch, welches auf einen komplexen Reaktionsverlauf schließen lässt. Diese Prozesse verlaufen assoziativ. Es werden zwei mögliche Reaktionsmechanismen vorgeschlagen. Dies sind I) die reduktive Eliminierungsreaktion aus einem anfänglich gebildeten, hexakoordinierten Platinkomplex und II) eine -Bindungsmetathese der B–B- mit der Pt–C- Bindung. Die oxidative Addition von Cat2B2 an [Pt(PCy3)3] erfolgt reversibel. Die strukturellen Parameter des Bisborylkomplexes im Kristall deuten auf einen sterisch überfrachteten cis-Bis(boryl)komplex mit relativ schwach gebundenen Borylliganden hin. Das neuartige Phosphan P(CH2Cy)3, welches sich durch einen flexiblen sterischen Anspruch auszeichnet, wird als Ligand in niedervalenten Phosphankomplexen eingesetzt. Der Platinkomplex reagiert mit 1,3,5-(C6H3)(BBr2)3 selektiv zu 1,3,5-trans-[Pt(BBr)Br{P(CH2Cy)3}2]3(C6H3), dem ersten Tris(boryl)komplex. Die Bis- und Tris(phosphan)rhodium(I)-Komplexe, welche im Überschuss mit Phosphan im Gleichgewicht vorliegen, reagieren mit CatBH zu trans-[Rh(BCat)ClH{P(CH2Cy)3}2]. [Pt(PCy3)2] reagiert mit CatBH in einer cis-selektiv verlaufenden Reaktion. Die Reaktion von [Pt{P(CH2Cy)3}2] mit CatBH im Überschuss führt zur Bildung von trans-[Pt(BCat)H{P(CH2Cy)3}2], cis-[Pt(BCat)2{P(CH2Cy)2}2] und H2 im Gleichgewicht. Gemäß quantenchemischen Berechnungen erfolgt die oxidative Addition der B–H-Bindung an [Pt(PR3)2] (R=Me, Cy, CH2Cy) ausgehend von einem -Präkursorkomplex. Durch die oxidative Addition der B–H-Bindung von CatBH an cis-[Pt(BCat)H(PR3)2] wird ein hyperkoordiniertes Platin(IV)-Intermediat gebildet, aus welchem das thermodynamisch stabilere trans-konfigurierte Isomer gebildet werden kann. Dieses Platin(IV)-Intermediat stellt die Schlüsselverbindung für die nachfolgende Dehydrokupplung dar. Durch einen Übergangszustand, in welchem Diwasserstoff abgespalten werden kann, wird ein cis-Bis(boryl)platinkomplex gebildet. Durch eine -Bindungsmetathese mit der B–H-Bindung von CatBH kann die B–B-Bindung geknüpft und Diboran(4) abgespalten werden. Das metallhaltige Produkt dieser Reaktion ist identisch mit dem trans-(Boryl)(hydrido)platinkomplex. Durch die Flexibilität der P(CH2Cy)3-Liganden werden durchweg Intermediate berechnet, welche geringere Deformationskräfte aufweisen als mit den rigiden PCy3-Liganden.
Background: Remote ischemic conditioning is gaining interest as potential method to induce resistance against ischemia reperfusion injury in a variety of clinical settings. We performed a systematic review and meta-analysis to investigate whether remote ischemic conditioning reduces mortality, major adverse cardiovascular events, length of stay in hospital and in the intensive care unit and biomarker release in patients who suffer from or are at risk for ischemia reperfusion injury.
Methods and Results: Medline, EMBASE and Cochrane databases were searched for randomized clinical trials comparing remote ischemic conditioning, regardless of timing, with no conditioning. Two investigators independently selected suitable trials, assessed trial quality and extracted data. 23 studies in patients undergoing cardiac surgery (15 studies), percutaneous coronary intervention (four studies) and vascular surgery (four studies), comprising in total 1878 patients, were included in this review. Compared to no conditioning, remote ischemic conditioning did not reduce mortality (odds ratio 1.22 [95% confidence interval 0.48, 3.07]) or major adverse cardiovascular events (0.65 [0.38, 1.14]). However, the incidence of myocardial infarction was reduced with remote ischemic conditioning (0.50 [0.31, 0.82]), as was peak troponin release (standardized mean difference -0.28 [-0.47, -0.09]).
Conclusion: There is no evidence that remote ischemic conditioning reduces mortality associated with ischemic events; nor does it reduce major adverse cardiovascular events. However, remote ischemic conditioning did reduce the incidence of peri-procedural myocardial infarctions, as well as the release of troponin.
Millionen Menschen weltweit leiden an den verschiedensten Autoimmunerkrankungen. Diese Krankheiten entstehen, wenn das Immunsystem gesundes körpereigenes Gewebe angreift und zerstört. An der Pathogenese sind sowohl Komponenten des angeborenen Immunsystems als auch Bestandteile des adaptiven Immunsystems, wie Lymphozyten und Antikörper, beteiligt. Da die Ursachen und molekularen Mechanismen der Pathogenese dieser Erkrankungen bis heute weitgehend unbekannt sind, wurden in dieser Arbeit autoaggressive Lymphozyten bei den humanen Autoimmunerkrankungen Polymyositis und Multiple Sklerose näher untersucht. Die Polymyositis ist eine chronisch entzündliche Erkrankung der Skelettmuskulatur. Die Muskelfasern werden dabei von zytotoxischen CD8+ gd-T-Lymphozyten infiltriert, attackiert und schließlich zerstört. In einem seltenen Fall der Polymyositis wurden die Muskelzellen hingegen in ähnlicher Weise von CD8- gd-T-Lymphozyten angegriffen. Die gd-T-Lymphozyten waren monoklonal expandiert und ihr Rezeptor, im Folgenden als M88 bezeichnet, wurde als Vg1.3+Vd2+ identifiziert. Frühere Untersuchungen der Antigenspezifität dieser Zellen zeigten, dass M88 mehrere funktionell und strukturell verschiedene Proteine aus unterschiedlichen Spezies erkennt. Die Bindung erfolgt spezifisch durch die Antigenerkennungsregionen beider Rezeptorketten von M88. In dieser Arbeit wurden verschiedene bakterielle und humane Proteine des Translationsapparates als Antigene von M88 identifiziert. Weitere ausführliche Untersuchungen eines paradigmatischen bakteriellen Antigens, dem Translationsinitiationsfaktor EcIF1, zeigten, dass M88 an Oberflächen-exponierte Konformationsepitope von Proteinen bindet. Interessanterweise erkennt M88 mehrere humane Aminoacyl-tRNA-Synthetasen, Antigene, die in anderen Formen der Myositis von Autoantikörpern angegriffen werden. Diese Beobachtung ergibt eine bemerkenswerte Verbindung zwischen T-Zell- und Antikörper-vermittelten B-Zell-Antworten bei der autoimmunen Myositis. Bei der Multiplen Sklerose ist das zentrale Nervensystem betroffen. Autoaggressive Lymphozyten greifen die Myelinschicht der Nervenzellen im Gehirn und Rückenmark an und zerstören sie. Im Liquor cerebrospinalis von Patienten lassen sich klonal expandierte und affinitätsgereifte B-Zellen sowie „oligoklonale Banden“ (OKB) Antikörper nachweisen. Obwohl diese Merkmale auf eine Antigen-induzierte Immunantwort hindeuten, sind die zugrundeliegenden Antigene und die Rolle der OKB bei der Pathogenese bis heute unbekannt. In dieser Arbeit wurde die Antigenspezifität von fünf IgG OKB-Antikörpern aus drei Patienten untersucht. Durch verschiedene proteinbiochemische Methoden konnten intrazelluläre Kandidatenantigene identifiziert werden. Interessanterweise sind darunter mehrere nukleäre Proteine, die an der Transkriptionsregulation oder der RNA-Prozessierung beteiligt sind. Reaktivitäten gegen intrazelluläre Antigene treten auch bei anderen Autoimmunerkrankungen, wie beispielsweise dem systemischen Lupus erythematodes, auf. Diese Ergebnisse könnten auf einen allgemeinen Mechanismus der Entstehung und Funktion von Autoantikörpern bei diesen humanen Autoimmunerkrankungen hindeuten.
Die bronchopulmonale Dysplasie (BPD) ist immer noch die häufigste schwere Komplikation von Frühgeburtlichkeit. Fortschritte in der Therapie, speziell die Einführung von Surfactant in den 1990er Jahren zur Behandlung eines Atemnotsyndroms (RDS), das einen der Hauptrisikofaktoren für die Entstehung einer BPD darstellt, haben Veränderungen der BPD mit sich gebracht. Diese „neue“ BPD unterscheidet sich vor allem hinsichtlich Pathophysiologie und Therapie enorm von der BPD der „Vor-Surfactant-Ära“. Die Datenlage zu langfristigen Einschränkungen der Lungenfunktion und Belastbarkeit von ehemaligen Frühgeborenen, die an der „neuen“ Form der BPD erkrankt sind, ist noch spärlich und uneinheitlich. Ziel dieser Studie war, die Bedeutung der „neuen“ BPD für die langfristige pulmonale Morbidität und die körperliche Leistungsfähigkeit von ehemaligen Extrem-Frühgeborenen zu beleuchten. Mögliche Konsequenzen für die Kinder mit „neuer“ BPD könnten frühe Interventionsmaßnahmen (Trainingsprogramme, medikamentöse Kontrolle einer subklinischen Inflammation) zur Verbesserung der pulmonalen Funktion und der körperlichen Aktivität und Belastbarkeit sein. Der Einschluss der eNO-Messung sollte gleichzeitig der Bewertung dieser nicht-invasiven Untersuchungsmethode zur Einschätzung der pulmonalen Morbidität von ehemaligen Frühgeborenen dienen. Es nahmen insgesamt 40 Kinder zwischen sieben und zwölf Jahren an dieser Studie teil, die in drei Gruppen – Frühgeborene mit BPD, Frühgeborene ohne BPD und Reifgeborene – aufgeteilt wurden. Die Probanden absolvierten Lungen-funktionsuntersuchungen, eine eNO-Messung, kardiale Untersuchungen, einen Belastungstest auf einem Spiroergometer sowie eine Aktivitätsmessung mittels Akzelerometrie. Anschließend wurden die Ergebnisse der Untersuchungen ausgewertet und unter den drei Gruppen verglichen. Frühgeborene mit neuer BPD zeigten auch im Schulalter bleibende Defizite. Diese betrafen neben der psychomotorischen Entwicklung vor allem die pulmonale Funktion, die Aktivität und die Leistungsfähigkeit dieser Kinder. Bei der Lungenfunktionsuntersuchung konnte festgestellt werden, dass frühgeborene Kinder im Schulalter unter anhaltender Atemwegsobstruktion sowie Kinder mit BPD zusätzlich unter einem gestörten Gastransfer leiden. In der Spiroergometrie ergab sich zudem eine für das Geschlecht und die Körpergröße korrigierte verminderte Sauerstoffaufnahme (VO2max) bei maximaler Belastung für Kinder mit BPD. Mit Hilfe einer Regressionsanalyse konnte die gestörte Diffusionskapazität als entscheidendes Kriterium für die Leistungseinschränkung der Kinder mit BPD identifiziert werden. Die Vermutung liegt nahe, dass all diese Einschränkungen auf die in frühester Kindheit entstandenen histologischen Veränderungen im Rahmen der „neuen“ BPD zurückgehen. Außerdem stellte sich heraus, dass frühgeborene Kinder, unabhängig ob mit oder ohne BPD, eine geringere Aktivität aufwiesen. Ob diese funktionalen Einschränkungen auch noch im Heranwachsenden- und Erwachsenenalter bestehen, muss durch weitere Studien bewiesen werden. Die eNO-Messung erbrachte in diesem Zusammenhang keine konsistenten Ergebnisse. Zusammenfassend konnte unsere Studie Anhaltspunkte für langfristige funktionale Konsequenzen der „neuen“ BPD für Lungenfunktion und körperliche Belastbarkeit im Schulalter abbilden. Die verminderte Sauerstoffaufnahme (VO2max) bei maximaler Belastung bei Kindern mit BPD spiegelt die Beeinträchtigung der alveolokapillären Einheit wider.
Derivate von Vinylsulfonen (VS), die zur Klasse der Michael-Akzeptoren gehören, haben sich in den letzten Jahren als potente irreversible Inhibitoren von Cystein-Proteasen etabliert. Durch einen nucleophilen Angriff des Cys-Restes im aktiven Zentrum der Protease auf das beta-Kohlenstoffatom der C-C-Doppelbindung wird die Protease irreversibel alkyliert. Ziel dieser Arbeit war es, einfache theoretische und experimentelle Methoden zu entwickeln, um erste Schlussfolgerungen hinsichtlich der Reaktivität unterschiedlicher Vinylsulfone ziehen zu können, die zur vollständigen Aufklärung der Struktur-Wirkungsbeziehung von Vinylsulfonen mit diversen Cystein-Proteasen dienen. Im ersten Teil der Arbeit wurden quantenmechanische Rechnungen an kleinen Vinylsulfon-Bausteinen angestellt, um den Einfluss unterschiedlicher Substitutionsmuster an der Sulfoneinheit auf die Reaktionskinetik von Vinylsulfonen zu untersuchen. Anhand der jeweiligen Potentialflächen ließen sich die charakteristischen Punkte der Reaktion, wie der Reaktionskomplex, der Übergangszustand (transition state, TS) sowie das Produkt mitsamt ihren Energien und Geometrien bestimmen. Die Höhe der Energiebarriere, die zum Erreichen des TS überwunden werden muss, die sogenannte Aktiverungsenergie, hängt über die Arrhenius-Gleichung mit den kinetischen Parametern der Reaktion zusammen. Es lässt sich also durch die Kenntnis der Aktivierungsenergien die Reaktivitätsreihenfolge unterschiedlich substituierter Vinylsulfone VS vorhersagen. Im zweiten Teil dieser Arbeit wurden Vinylsulfonbausteine synthetisiert und an separat hergestellte Peptide gekuppelt, sodass potentielle Inhibitoren erhalten wurden. So konnten u.a. die peptidischen Inhibitoren Mu-D-Phe-L-HomoPhe-VS-Me und MP-D-Phe-L-HomoPhe-VS-Me hergestellt werden. Ein zweites Syntheseprojekt beschäftigte sich mit der Kupplung von Peptiden an neue Derivate der trans-Aziridin-2,3-dicarbonsäure. Die synthetisierten Inhibitoren waren Z-Phe-Ala-Azi, Boc-Leu-Pro-Azi und Z-Pro-Leu-Azi. Hierfür wurden die Peptide des Vinylsulfonsprojekts in umgekehrter Aminosäure-Reihenfolge synthetisiert, um sie an die Aziridinbausteine kuppeln zu können. Der dritte Teil der Doktorarbeit befasste sich mit der experimentellen Untersuchung der synthetisierten Vinylsulfonbausteine sowie den erhaltenen peptidischen VS- und Aziridin-basierten Inhibitoren. Es wurden einerseits Enzym-Assays durchgeführt, um die prozentuale Hemmung verschiedener Cystein-Proteasen durch die synthetisierten Moleküle zu messen. Keine der Verbindungen wies jedoch eine signifikannte Hemmung der Proteasen Rhodesain, Falcipain 2 und Cathepsin B auf. Andererseits wurden Modellsysteme entwickelt, um die Kinetik der Reaktionen der Vinylsulfon- und Aziridinbausteine mit einem geeigneten Thiol als Enzym-Imitat zu verfolgen. Ein zielführendes Modell konnte mit Phenylethanthiol in deuteriertem Methanol realisiert werden. Durch Zusatz von NaOH, KOH oder KOtBu konnte zusätzlich die Reaktion mit dem Thiolat untersucht werden. Die Reaktionen wurden sowohl mit IR- als auch NMR-Spektroskopie verfolgt und es wurden die Geschwindigkeitskonstanten 2. Ordnung bestimmt. Auf den ersten Blick konnte mit dem theoretischen Modell der experimentell gefundene Trend nicht vorhergesagt werden. Die Reihenfolge der Sulfonderivate aber, die an der Sulfongruppe ein weiteres Heteroatom tragen, Sulfonester und Sulfonamid, wurde richtig abgeschätzt. Der Unterschied in der Aktivierungsenergie zwischen den Sulfonestern beläuft sich auf 0.7 kcal pro mol. Über die Arrheniusgleichung, ergibt sich bei Annahme desselben Arrhenius-Faktors bei einer Temperatur von 25°C, dass OPhVS um einen Faktor 3 schneller als OMeVS reagieren sollte. Tatsächlich wurde im Experiment ein Faktor von 2.6 gefunden. Aufgrund der unterschiedlichen Substituenten am Stickstoffatom, ist das Amid nicht vollständig mit seinem H-substituierten theoretischen Pendant vergleichbar. Dass das Sulfonamid langsamer als die Sulfonester reagieren, wurde vom theoretischen Modell ebenfalls richtig vorhergesagt.
A binary tanglegram is a drawing of a pair of rooted binary trees whose leaf sets are in one-to-one correspondence; matching leaves are connected by inter-tree edges. For applications, for example, in phylogenetics, it is essential that both trees are drawn without edge crossings and that the inter-tree edges have as few crossings as possible. It is known that finding a tanglegram with the minimum number of crossings is NP-hard and that the problem is fixed-parameter tractable with respect to that number.
We prove that under the Unique Games Conjecture there is no constant-factor approximation for binary trees. We show that the problem is NP-hard even if both trees are complete binary trees. For this case we give an O(n 3)-time 2-approximation and a new, simple fixed-parameter algorithm. We show that the maximization version of the dual problem for binary trees can be reduced to a version of MaxCut for which the algorithm of Goemans and Williamson yields a 0.878-approximation.

Untersucht wird die Gesellschaftsgründung unter Beteiligung Minderjähriger, der Eintritt eines Minderjährigen in eine bestehende Gesellschaft durch Rechtsgeschäft unter Lebenden, die Umwandlung einer Gesellschaftsbeteiligung und der Eintritt eines Minderjährigen in eine Gesellschaft durch Erwerb von Todes wegen. Hierbei wird das Schwergewicht der Untersuchung auf die Personengesellschaften gelegt, da bei den Personengesellschaften typischerweise erhebliche Haftungsrisiken für den Minderjährigen bestehen. Bei den Kapitalgesellschaften wird die Rechtslage nur vergleichsweise dargestellt.
Glioblastoma multiforme (GBM) represents the most aggressive form of malignant brain tumors and remains a therapeutically challenge. Intense research in the field has lead to the testing of oncolytic viruses to improve tumor control. Currently, a variety of different oncolytic viruses are being evaluated for their ability to be used in anti-cancer therapy and a few have entered clinical trials. Vaccinia virus, is one of the viruses being studied. GLV-1h68, an oncolytic vaccinia virus engineered by Genelux Corporation, was constructed by insertion of three gene cassettes, RUC-GFP fusion, β-galactosidase and β- glucuronidase into the genome of the LIVP strain. Since focal tumor radiotherapy is a mainstay for cancer treatment, including glioma therapy, it is of clinical relevance to assess how systemically administered oncolytic vaccinia virus could be combined with targeted ionizing radiation for therapeutic gain. In this work we show how focal ionizing radiation (IR) can be combined with multiple systemically delivered oncolytic vaccinia virus strains in murine models of human U-87 glioma. After initial experiments which confirmed that ionizing radiation does not damage viral DNA or alter viral tropism, animal studies were carried out to analyze the interaction of vaccinia virus and ionizing radiation in the in vivo setting. We found that irradiation of the tumor target, prior to systemic administration of oncolytic vaccinia virus GLV-1h68, increased viral replication within the U-87 xenografts as measured by viral reporter gene expression and viral titers. Importantly, while GLV-1h68 alone had minimal effect on U-87 tumor growth delay, IR enhanced GLV-1h68 replication, which translated to increased tumor growth delay and mouse survival in subcutaneous and orthotopic U-87 glioma murine models compared to monotherapy with IR or GLV-1h68. The ability of IR to enhance vaccinia replication was not restricted to the multi-mutated GLV-1h68, but was also seen with the less attenuated oncolytic vaccinia, LIVP 1.1.1. We have demonstrated that in animals treated with combination of ionizing radiation and LIVP 1.1.1 a strong pro-inflammatory tissue response was induced. When IR was given in a more clinically relevant fractionated scheme, we found oncolytic vaccinia virus replication also increased. This indicates that vaccinia virus could be incorporated into either larger hypo-fraction or more conventionally fractionated radiotherapy schemes. The ability of focal IR to mediate selective replication of systemically injected oncolytic vaccinia was demonstrated in a bilateral glioma model. In mice with bilateral U-87 tumors in both hindlimbs, systemically administered oncolytic vaccinia replicated preferentially in the focally irradiated tumor compared to the shielded non- irradiated tumor in the same mouse We demonstrated that tumor control could be further improved when fractionated focal ionizing radiation was combined with a vaccinia virus caring an anti-angiogenic payload targeting vascular endothelial growth factor (VEGF). Our studies showed that following ionizing radiation expression of VEGF is upregulated in U-87 glioma cells in culture. We further showed a concentration dependent increase in radioresistance of human endothelial cells in presence of VEGF. Interestingly, we found effects of vascular endothelial growth factor on endothelial cells were reversible by adding purified GLAF-1 to the cells. GLAF-1 is a single- chain antibody targeting human and murine VEGF and is expressed by oncolytic vaccinia virus GLV-109. In U-87 glioma xenograft murine models the combination of fractionated ionizing radiation with GLV-1h164, a vaccinia virus also targeting VEGF, resulted in the best volumetric tumor response and a drastic decrease in vascular endothelial growth factor. Histological analysis of embedded tumor sections 14 days after viral administration confirmed that blocking VEGF translated into a decrease in vessel number to 30% of vessel number found in control tumors in animals treated with GLV-164 and fractionated IR which was lower than for all other treatment groups. Our experiments with GLV-1h164 and fractionated radiotherapy have shown that in addition to ionizing radiation and viral induced tumor cell destruction we were able to effectively target the tumor vasculature. This was achieved by enhanced viral replication translating in increased levels of GLAF-2 disrupting tumor vessels as well as the radiosensitization of tumor vasculature to IR by blocking VEGF. Our preclinical results have important clinical implications of how focal radiotherapy can be combined with systemic oncolytic viral administration for highly aggressive, locally advanced tumors with the potential, by using a vaccinia virus targeting human vascular endothelial growth factor, to further increase tumor radiation sensitivity by engaging the vascular component in addition to cancer cells.
In this PhD thesis, the fingerprints of geometry and topology on low dimensional mesoscopic systems are investigated. In particular, holographic non-equilibrium transport properties of the quantum spin Hall phase, a two dimensional time reversal symmetric bulk insulating phase featuring one dimensional gapless helical edge modes are studied. In these metallic helical edge states, the spin and the direction of motion of the charge carriers are locked to each other and counter-propagating states at the same energy are conjugated by time reversal symmetry. This phenomenology entails a so called topological protection against elastic single particle backscattering by time reversal symmetry. We investigate the limitations of this topological protection by studying the influence of inelastic processes as induced by the interplay of phonons and extrinsic spin orbit interaction and by taking into account multi electron processes due to electron-electron interaction, respectively. Furthermore, we propose possible spintronics applications that rely on a spin charge duality that is uniquely associated with the quantum spin Hall phase. This duality is present in the composite system of two helical edge states with opposite helicity as realized on the two opposite edges of a quantum spin Hall sample with ribbon geometry. More conceptually speaking, the quantum spin Hall phase is the first experimentally realized example of a symmetry protected topological state of matter, a non-interacting insulating band structure which preserves an anti-unitary symmetry and is topologically distinct from a trivial insulator in the same symmetry class with totally localized and hence independent atomic orbitals. In the first part of this thesis, the reader is provided with a fairly self-contained introduction into the theoretical concepts underlying the timely research field of topological states of matter. In this context, the topological invariants characterizing these novel states are viewed as global analogues of the geometric phase associated with a cyclic adiabatic evolution. Whereas the detailed discussion of the topological invariants is necessary to gain deeper insight into the nature of the quantum spin Hall effect and related physical phenomena, the non-Abelian version of the local geometric phase is employed in a proposal for holonomic quantum computing with spin qubits in quantum dots.
Background: Because most human stroke victims are elderly, studies of experimental stroke in the aged rather than the young rat model may be optimal for identifying clinically relevant cellular responses, as well for pinpointing beneficial interventions.
Methodology/Principal Findings: We employed the Affymetrix platform to analyze the whole-gene transcriptome following temporary ligation of the middle cerebral artery in aged and young rats. The correspondence, heat map, and dendrogram analyses independently suggest a differential, age-group-specific behaviour of major gene clusters after stroke. Overall, the pattern of gene expression strongly suggests that the response of the aged rat brain is qualitatively rather than quantitatively different from the young, i.e. the total number of regulated genes is comparable in the two age groups, but the aged rats had great difficulty in mounting a timely response to stroke. Our study indicates that four genes related to neuropathic syndrome, stress, anxiety disorders and depression (Acvr1c, Cort, Htr2b and Pnoc) may have impaired response to stroke in aged rats. New therapeutic options in aged rats may also include Calcrl, Cyp11b1, Prcp, Cebpa, Cfd, Gpnmb, Fcgr2b, Fcgr3a, Tnfrsf26, Adam 17 and Mmp14. An unexpected target is the enzyme 3-hydroxy-3-methylglutaryl-Coenzyme A synthase 1 in aged rats, a key enzyme in the cholesterol synthesis pathway. Post-stroke axonal growth was compromised in both age groups.
Conclusion/Significance: We suggest that a multi-stage, multimodal treatment in aged animals may be more likely to produce positive results. Such a therapeutic approach should be focused on tissue restoration but should also address other aspects of patient post-stroke therapy such as neuropathic syndrome, stress, anxiety disorders, depression, neurotransmission and blood pressure.

Die Frage, wie sich der keltische Tristanstoff zum Artusroman verhält, ist in der mediävistischen Forschung bislang jedoch nur in Ansätzen und zumeist wenig kontrovers diskutiert worden. Dabei bietet gerade die kontingente Verbindung beider literarischer Welten bei Eilhart und Béroul zu einem arthurisierten Tristanroman die Möglichkeit, die zentralen Scharnierstellen zweier Erzähltradition zu untersuchen und nach den Möglichkeiten und Grenzen des Erzählens von der Liebe um 1200 zu fragen.
Epithelial and endothelial cells (EC) are building paracellular barriers which protect the tissue from the external and internal environment. The blood-brain barrier (BBB) consisting of EC, astrocyte end-feet, pericytes and the basal membrane is responsible for the protection and homeostasis of the brain parenchyma. In vitro BBB models are common tools to study the structure and function of the BBB at the cellular level. A considerable number of different in vitro BBB models have been established for research in different laboratories to date. Usually, the cells are obtained from bovine, porcine, rat or mouse brain tissue (discussed in detail in the review by Wilhelm et al. 1). Human tissue samples are available only in a restricted number of laboratories or companies 2,3. While primary cell preparations are time consuming and the EC cultures can differ from batch to batch, the establishment of immortalized EC lines is the focus of scientific interest.
Here, we present a method for establishing an immortalized brain microvascular EC line from neonatal mouse brain. We describe the procedure step-by-step listing the reagents and solutions used. The method established by our lab allows the isolation of a homogenous immortalized endothelial cell line within four to five weeks. The brain microvascular endothelial cell lines termed cEND 4 (from cerebral cortex) and cerebEND 5 (from cerebellar cortex), were isolated according to this procedure in the Förster laboratory and have been effectively used for explanation of different physiological and pathological processes at the BBB. Using cEND and cerebEND we have demonstrated that these cells respond to glucocorticoid- 4,6-9 and estrogen-treatment 10 as well as to pro-infammatory mediators, such as TNFalpha 5,8. Moreover, we have studied the pathology of multiple sclerosis 11 and hypoxia 12,13 on the EC-level. The cEND and cerebEND lines can be considered as a good tool for studying the structure and function of the BBB, cellular responses of ECs to different stimuli or interaction of the EC with lymphocytes or cancer cells.
Zahlreiche experimentelle Befunde lassen vermuten, dass G-Protein gekoppelte Rezeptoren (GPCR) nach ihrer Aktivierung einer ligandenselektiven Änderung der Rezeptorkonformation unterliegen. Ziel der vorliegenden Arbeit war es dieses Phänomen am Subtyp 2 der muskarinischen Acetylcholinrezeptoren (M2 AChR) zu untersuchen. Muskarinische Acetylcholinrezeptoren (mAChR) können in fünf Subtypen (M1-M5) unterschieden werden. Durch die Beteiligung der mAChR an zahlreichen physiologischen Prozessen stellen sie wichtige Zielstrukturen pharmakologischer Therapien dar. Da die orthosterische Ligandenbindestelle (= Bindestelle des endogenen Liganden) in allen fünf Subtypen hoch konserviert ist, wird ihr pharmakologischer Nutzen derzeit allerdings durch die unselektive Rezeptormodulation und dem damit verbundenen Auftreten unerwünschter Arzneimittelwirkungen stark limitiert. Ein Ansatz zur Erzielung subtypselektiver Effekte besteht in der Verwendung allosterischer Modulatoren. Da die allosterische Bindestelle der mAChR eine geringere Sequenzhomologie aufweist, können so gezielt einzelne Subtypen der mAChR reguliert werden. Der M2 AChR stellt hinsichtlich allosterischer Modulation ein gut charakterisiertes Modellsystem dar. Für ihn wurde bereits eine Vielzahl allosterischer Liganden entwickelt. Auch bitopische Liganden, die sowohl einen allosterischen als auch einen orthosterischen Anteil enthalten, wurden für den M2 AChR bereits beschrieben. Im ersten Teil der vorliegenden Arbeit wurden verschiedene FRET-Sensoren des M2 AChR generiert und charakterisiert. Als FRET-Paar wurden das cyan fluoreszierende Protein (CFP) und der niedermolekulare fluorescein-basierte Fluorophor FlAsH (fluorescein arsenical hairpin binder) gewählt. CFP wurde in den Sensoren am Ende des C-Terminus angefügt. Die zur Markierung mit FlAsH nötige Tetracysteinsequenz wurde in verschiedenen Bereichen der dritten intrazellulären Rezeptorschleife (IL) eingebracht. Die auf diese Weise erstellten Re-zeptorsensoren trugen das Tetracysteinmotiv in der N terminalen (M2i3-N) bzw. in der C terminalen Region (M2i3-C) von IL 3. Die Charakterisierung der Rezeptorsensoren bezüglich Ligandenbindung, Gi-Protein Aktivierung und β-Arrestin2 Translokation ergab keine signifikanten Unterschiede zwischen M2i3-N, M2i3 C und M2CFP oder Wildtyp M2 AChR. Zunächst wurden sowohl unterschiedliche orthosterische, als auch allosterische Liganden hinsichtlich ihrer mittleren effektiven Konzentration und ihrer maximalen Wirkstärke an den Rezeptorsensoren untersucht. Mit Hilfe von FRET-Messungen konnte ein superago-nistisches Verhalten des orthosterischen Testliganden Iperoxo gezeigt werden. Die Eigenschaften der allosterischen Substanzen wurden durch Messung der Rezeptordeakti-vierungskinetik und des maximalen Hemmeffekts auf einen vorstimulierten Rezeptor charakterisiert. Alle allosterischen Liganden deaktivierten den vorstimulierten M2 AChR mit einer schnelleren Kinetik als Atropin. Die EC50-Werte der unterschiedlichen Substanzen waren unabhängig von der Markierungsposition im verwendeten Rezeptorsensor vergleich-bar. Ausnahmen bildeten die allosterischen Liganden JK 289, JK 338, ½ W84 und EHW 477, die liganden- und sensorabhängig unterschiedliche mittlere effektive Konzentrationen aufwie-sen. Bei der Untersuchung der Konformationsänderung des M2 AChR konnte kein liganden-selektiver Unterschied zwischen den FRET-Signalen für 19 getestete orthosterische Liganden beobachtet werden. Dies deutet darauf hin, dass alle orthosterischen Testliganden eine dem Acetylcholin (ACh) vergleichbare Änderung der M2 AChR Konformation induzier-ten. Um zu untersuchen, ob für die orthosterischen Testliganden eine Korrelation zwischen ihrer maximalen Wirkstärke hinsichtlich Rezeptoraktivierung in FRET-Experimenten und der Aktivierung nachgeschalteter Signalwege besteht, wurde die orthosterisch-vermittelte Translokation von β-Arrestin2 mit Hilfe der Konfokalmikroskopie bestimmt. Bis auf 5-Methyl-furmethiodid translozierten alle orthosterischen Liganden β-Arrestin2 in einem Ausmaß, das mit der maximalen Rezeptoraktivierung vergleichbar war. Dagegen rief 5 Methylfurmethiodid verglichen mit dem endogenen Liganden ACh zwar eine ca. halbmaximale Rezeptorakti-vierung, aber nur eine äußerst geringe β-Arrestin2 Translokation hervor. Im zweiten Teil der Arbeit wurde der Einfluss verschiedener Allostere auf eine ligandenselektive Konformationsänderung des M2 AChR untersucht. Die allosterischen Liganden JK 337 und Seminaph beeinflussten den M2i3-C Sensor signifikant stärker, als das M2i3-N Konstrukt. Dagegen zeigte EHW 477 eine stärkere Beeinflussung der Rezeptorkon-formation des M2i3-N-, als des M2i3-C Sensors. Dies erlaubt die Vermutung, dass JK 337 und Seminaph eine stärkere Bewegung unterhalb von Transmembrandomäne (TM) 6, als unterhalb von TM 5 hervorriefen. Die Ergebnisse für EHW 477 legen nahe, dass TM 5 eine größere Bewegung eingeht, als TM 6. FRET-basierte Untersuchungen der Einflüsse der allosterischen Testliganden auf nachgeschaltete Signalwege ergaben, dass sowohl die Akti-vierung des Gi Proteins, als auch die β-Arrestin2 Translokation selektiv durch einzelne allosterische Liganden beeinflusst werden. Auch ein Zusammenhang zwischen Rezeptor-aktivierung und der Regulation nachgeschalteter Signalwege war erkennbar. Allerdings waren auf Grund der Versuchsbedingungen keine quantitativen Aussagen möglich. Im Folgenden wurden die bitopischen Liganden Hybrid 1 und 2 (H 1, H 2) hinsichtlich ihres Effekts auf die Konformationsänderung des M2 AChR untersucht. Während eine Stimulation mit H 1 vergleichbare FRET-Signale an beiden Sensoren ergab, konnte mit H 2 weder am M2i3-N-, noch an M2i3-C Sensor eine FRET-Änderung detektiert werden. Um den mangeln-den Effekt der Hybridsubstanzen in FRET-mikroskopischen Untersuchungen aufzuklären, wurden verschiedene Ansätze gewählt: Mit kettenverlängerten Derivaten der Hybridsubstanzen konnte in FRET-Messungen eine Änderung des FRET-Signals detektiert werden. Die Entfernung des allosterischen Bausteins führte in FRET-Experimenten zu einer verglichen mit dem endogenen Liganden ACh etwa halbmaximalen Aktivierung beider Sensoren. Dagegen resultierte die Mutation der alloste-rischen Bindestelle in nachfolgenden FRET-Untersuchungen mit H 1 und H 2 in keiner Signaländerung des FRET-Ratio. Diese Beobachtungen führten zu der Annahme, dass die Linkerkette, die orthosterischen und allosterischen Baustein der Hybride miteinander verbindet, zu kurz war um eine gleichzeitige Bindung an die allosterische und orthosterische Bindestelle zu ermöglichen. Ein anderer Erklärungsansatz besteht darin, dass nach Bindung des Orthosters der Kanal zwischen orthosterischer und allosterischer Bindestelle durch die Konformationsänderung des Rezeptors verschlossen wird, weshalb keine dauerhafte, dualsterische Bindung der Hybridsubstanzen an den M2 AChR möglich ist. Im Rahmen der vorliegenden Arbeit ist es gelungen mittels FRET-Experimenten die Existenz einer ligandenselektiven Rezeptorkonformation des M2 AChR mit allosterischen Liganden nachzuweisen. Darüber hinaus konnte auch ein Bezug zum Auftreten einer funktionellen Selektivität mit allosterischen Liganden hergestellt werden. Die Untersuchung von 19 orthosterischen Liganden hinsichtlich ihres Einflusses auf die Rezeptorkonformation des M2 AChR ergab keinen Hinweis auf eine ligandenselektive Konformationsänderung. Die Betrachtung der orthosterisch-vermittelten Translokation von β-Arrestin2 zeigte, dass zwischen der Effizienz der orthosterischen Testliganden, den M2 AChR zu aktivieren und dem Ausmaß, in dem sie eine β Arrestin2 Translokation induzierten eine direkte Korrelation besteht. Lediglich 5-Methylfurmethiodid rief eine ungleich geringere β-Arrestin2 Translokation hervor, verglichen mit dem Ausmaß an Rezeptoraktivierung. Diese Beobachtung deutet auf die Existenz eines signaling-bias für diesen Liganden hin. Die Untersuchung der dualsterischen Liganden H 1 und 2 bezüglich ihrer Fähigkeit zur Rezeptoraktivierung ergab, dass erst durch eine Verlängerung der Linkerkette, durch die orthosterischer und alloste-rischer Baustein miteinander verbunden sind eine Konformationsänderung des M2 AChR hin zu einer aktiven Konformation erreicht werden kann. Es kann somit angenommen werden, dass in den ursprünglichen Hybridsubstanzen H 1 und H 2 eine zu kurze Linkerkette, durch die keine dualsterische Bindung der Hybride an die allosterische und orthosterische Bindestelle möglich ist, ursächlich für die mangelnde Rezeptoraktivierung des M2 AChR war.
Acute graft-versus-host disease (aGvHD) is an immune syndrome associated with allogeneic hematopoietic cell transplantation (allo-HCT) that is mediated by alloreactive donor T cells attacking the gastrointestinal tract, liver, and skin of the host. Early diagnosis remains problematic and to date mainly relies on clinical symptoms and histopathology. Previously, different groups demonstrated that in order to cause aGvHD, alloreactive T cells require the expression of appropriate homing receptors to efficiently migrate from their priming sites to their target tissues. Therefore, the development of a predictive test based on the homing receptor expression profile of peripheral blood T cells seems attractive to identify patients at risk before the onset of aGvHD. The aim of this study was to analyze migrating alloreactive donor T cell kinetics in the peripheral blood early after allo-HCT in a murine model across minor histocompatibility antigens (miHAg) followed by a precise characterization of the homing receptor expression profile of migrating donor lymphocytes in order to identify suitable predictive markers. Combining daily bioluminescence imaging (BLI) and flow cytometry (FC) allowed defining two weeks of massive alloreactive donor T cell migration before clinical aGvHD symptoms became apparent. Peripheral blood donor T lymphocytes highly up-regulated the homing markers α4β7 integrin, and P- and E-selectin-ligand at peak time points of cell migration. The combination with the activation markers CD25 and CD69 and low expression levels of L-selectin allowed alloreactive donor T cell definition. Based on this migration phase we postulated a potential diagnostic window to precisely identify alloreactive donor T cells upon their homing receptor expression profile. Consequently, targeted pre-emptive treatment with rapamycin starting at the earliest detection time point of alloreactive donor T cells in the peripheral blood (day+6) significantly prolonged survival of treated mice. Based on this data, we propose a potential diagnostic window for alloreactive cell detection based on their homing receptor expression profile for a timely and effective therapeutic intervention before the clinical manifestation of aGvHD.
Mit dieser Arbeit konnte erstmals gezeigt werden, dass ANP über die Aktivierung der endothelialen Guanylyl Cyklase A (GC-A) akut die Permeabilität für Albumin an postkapillären Venolen stimuliert. Durch diesen Mechanismus ist ANP neben der chronischen auch an der akuten Reduktion des Plasmavolumens und des systolischen arteriellen Blutdrucks beteiligt. Aufgrund der wichtigen extrarenalen endothelialen Effekte stellt ANP das einzige hypovolämische Hormon im Organismus dar und ist somit ein bedeutender physiologischer Regulator der transvaskulären Volumenhomöostase. Jedoch sind diese Effekte deutlich von denen der vasoaktiven Substanzen, wie z. B. Histamin oder Thrombin und anderer diuretisch wirkender Substanzen, wie z. B. Schleifendiuretika, abzugrenzen. Vermutlich wird der permeabilitätssteigernde Effekt von ANP durch die beobachtete gesteigerte PKG- und PKA-abhängige Phosphorylierung von VASP vermittelt, wodurch möglicherweise die parazelluläre Permeabilität selektiv für Albumin gesteigert wird. Den gegenregulatorischen Mechanismus zur endothelialen GC-A stellt der NPR-C dar. Durch diesen erfolgt eine verminderte PKA- und PKG-abhängige Phosphorylierung von VASP, was durch die ANP-Infusion an EC GC-A KO Mäusen beobachtet wurde und zu einer Reduktion der physiologischen Permeabilität für Albumin an den postkapillären Venolen führen könnte. Somit wird wahrscheinlich durch diesen Mechanismus das Plasmavolumen, aber nicht der systolische arterielle Blutdruck in vivo gesteigert und eine übersteigerte Reaktion der GC-A verhindert. Aufgrund der enormen Bedeutung des ANP/GC-A Systems für die Modulation der endothelialen Permeabilität und die akuten sowie chronischen Regulation des Plasmavolumens und arteriellen Blutdrucks besitzt es bei Dysfunktion eine bedeutende klinische Relevanz. Denn durch eine verminderte Sekretion von ANP oder der Dysfunktion des endothelialen GC-A-Rezeptors, durch verschiedene Single Nucleotid Polymorphismen (SNPs) oder Desensitisierung, kann dieses System an der Entstehung der essentiellen Hypertonie oder der Hypervolämie bzw. Hypernatriämie bei herzinsuffizienten Patienten beteiligt sein.
Internationalismen im Grundwortschatz: Untersuchungen zur romanistischen Mehrsprachigkeitsdidaktik

In dieser Arbeit wird an Hand eines Häufigkeitswörterbuches des Italienischen (Lessico di frequenza dell’italiano parlato von De Mauro et al. 1993) untersucht, wie sich Kenntnisse anderer Sprachen, des Deutschen, Englischen und Französischen und/oder Spanischen, auf das Verstehen und Lernen von 2500 der häufigsten Wörter des Italienischen auswirken. Damit soll ein Beitrag geleistet werden zur mehrsprachigkeitsdidaktischen Fragestellung, wie viele Wörter des italienischen Grundwortschatzes aus anderen Sprachen schon bekannt sind und wie viele noch gelernt werden müssen, was mit konkreten Zahlen belegt werden soll.
Within the scope of this thesis two main topics have been investigated: the examination of micromagnetic sensors and transport of massive and massless Dirac fermions in HgTe quantum wells. For the investigation of localized, inhomogeneous magnetic fields, the fabrication and characterization of two different non-invasive and ultra sensitive sensors has been established at the chair ”Experimentelle Physik” of the University of Würzburg. The first sensor is based on the young technique named micro-Hall magnetometry. The necessary semiconductor devices (Hall cross structures) were fabricated by high-resolution electron beam lithography based on two different two dimensional electron gases (2DEGs), namely InAs/(Al,Ga)Sb- and HgTe/(Hg,Cd)Te- heterostructures. The characteristics have been examined in two different ways. Measurements in homogeneous magnetic fields served for characterization of the sensors, whereas the investigation of artificially produced sub-µm magnets substantiates the suitability of the devices for the study of novel nanoscale magnetic materials (e.g. nanowires). Systematic experiments with various magnets are in accordance with the theory of single-domain particles and anisotropic behavior due to shapes with high aspect ratio. The highest sensitivity for strongly localized fields was obtained at T = 4.2 K for a (200x200) nm^2 Hall cross - made from shallow, high mobility HgTe 2DEG. Although the field resolution was merely δB ≈ 100 µT, the nanoscale sensor size yields an outstanding flux resolution of δΦ = 2 10^(−3) Φ0, where Φ0 = h/2e is the flux quantum. Translating this result in terms of magnetic moment, the sensitivity allows for the detection of magnetization changes of a particle centered on top of the sensor as low as δM ≈ 10^2 µB, with the magnetic moment of a single electron µB, the Bohr magneton. The further examination of a permalloy nanomagnet with a cross-section of (100x20) nm^2 confirms the expected resolution ability, extracted from the noise of the sensor. The observed high signal-to-noise ratio validates the detection limit of this sensor in terms of geometry. This would be reached for a magnet (same material) with quadratic cross-section for an edge length of 3.3 nm. Moreover, the feasibility of this sensor for operation in a wide temperature range (T = mK... > 200 K) and high magnetic fields has been confirmed. The second micromagnetic sensor is the micro-SQUID (micro-Superconducting-QUantum-Interference-Device) based on niobium. The typical sensor area of the devices built in this work was (1.0x1.0) µm^2, with constrictions of about 20 nm. The characterization of this device demonstrates an amazing field sensitivity (regarding its size) of δB < 1 µT. Even though the sensor was 25 times larger than the best micro-Hall sensor, it provided an excellent flux resolution in the order of δΦ ≈ 5 10^(−4) Φ0 and a similar magnetic moment resolution of δM ≈ 10^2 µB. Furthermore, the introduction of an ellipsoidal permalloy magnet (axes: 200 nm and 400 nm, thickness 30 nm) substantiates the suitability for the detection of minuscule, localized magnetic fields. The second part of the thesis deals with the peculiar transport properties of HgTe quantum wells. These rely on the linear contribution to the band structure inherent to the heterostructure. Therefore the system can be described by an effective Dirac Hamiltonian, whose Dirac mass is tunable by the variation of the quantum well thickness. By fabrication and characterization of a systematical series of substrates, a system with vanishing Dirac mass (zero energy gap) has been confirmed. This heterostructure therefore resembles graphene (a monolayer of graphite), with the difference of exhibiting only one valley in the energy dispersion of the Brillouin zone. Thus parasitical intervalley scattering cannot occur. The existence of this system has been proven by the agreement of theoretical predictions, based on widely accepted band structure calculations with the experiment (Landau level dispersion, conductivity). Furthermore, another particularity of the band structure - the transition from linear to parabolic character - has been illustrated by the widths of the plateaus in the quantum Hall effect. Finally, the transport of ”massive” Dirac fermions (with finite Dirac mass) is investigated. In particular the describing Dirac Hamiltonian induces weak localization effects depending on the Dirac mass. This mechanism has not been observed to date, and survives in higher temperatures compared to typical localization mechanisms.
Non–Small-Cell Lung Cancer (NSCLC) is the most frequent human lung cancer and a major cause of death due to its high rate of metastasis1. These facts emphasize the urgent need for the investigation of new targets for anti-metastatic therapy. Up to now a number of genes and gene products have been identified that positively or negatively affect the probability of established human tumor cell lines to metastasize2. Previously, together with the group of Professor Ulf Rapp, we have described the first conditional mouse model for metastasis of NSCLC and identified a gene, c-MYC, that is able to orchestrate all steps of this process. We could identify potential markers for detection of metastasis and highlighted GATA4, which is exclusively expressed during lung development, as a target for future therapeutic intervention2. However, the mechanism underlying this metastatic conversion remained to be identified, and was therefore the focus of the present work. Here, GATA4 is identified as a MYC target in the development of metastasis and epigenetic alterations at the GATA4 promoter level are shown after MYC expression in NSCLC in vivo and in vitro. Such alterations include site-specific demethylation that accompanies the displacement of the MYC-associated zinc finger protein (MAZ) from the GATA4 promoter, which leads to GATA4 expression. Histone modification analysis of the GATA4 promoter revealed a switch from repressive histone marks to active histone marks after MYC binding, which corresponds to active GATA4 expression. This work identifies a novel epigenetic mechanism by which MYC activates GATA4 leading to metastasis in NSCLC, suggesting novel potential targets for the development of anti-metastatic therapy.
Durch das Auftreten neuer Stämme resistenter Krankheitserreger ist die Suche nach neuartigen Wirkstoffen gegen diese, sich ständig weiter ausbreitende Bedrohung, dringend notwendig. Der interdisziplinäre Sonderforschungsbereich 630 der Universität Würzburg stellt sich dieser Aufgabe, indem hier neuartige Xenobiotika synthetisiert und auf ihre Wirksamkeit getestet werden. Die hier vorgelegte Dissertation fügt sich hierbei nahtlos in die verschiedenen Fachbereiche des SFB630 ein: Sie stellt eine Schnittstelle zwischen Synthese und Analyse der Effekte der im Rahmen des SFB630 synthetisierten Isochinolinalkaloid-Derivaten. Mit den hier angewandten bioinformatischen Methoden wurden zunächst die wichtigsten Stoffwechselwege von S. epidermidis R62A, S. aureus USA300 und menschlicher Zellen in sogenannten metabolischen Netzwerkmodellen nachgestellt. Basierend auf diesen Modellen konnten Enzymaktivitäten für verschiedene Szenarien an zugesetzten Xenobiotika berechnet werden. Die hierfür benötigten Daten wurden direkt aus Genexpressionsanalysen gewonnen. Die Validierung dieser Methode erfolgte durch Metabolommessungen. Hierfür wurde S. aureus USA300 mit verschiedenen Konzentrationen von IQ-143 behandelt und gemäß dem in dieser Dissertation vorgelegten Ernteprotokoll aufgearbeitet. Die Ergebnisse hieraus lassen darauf schließen, dass IQ-143 starke Effekte auf den Komplex 1 der Atmungskette ausübt – diese Resultate decken sich mit denen der metabolischen Netzwerkanalyse. Für den Wirkstoff IQ-238 ergaben sich trotz der strukturellen Ähnlichkeiten zu IQ-143 deutlich verschiedene Wirkeffekte: Dieser Stoff verursacht einen direkten Abfall der Enzymaktivitäten in der Glykolyse. Dadurch konnte eine unspezifische Toxizität dieser Stoffe basierend auf ihrer chemischen Struktur ausgeschlossen werden. Weiterhin konnten die bereits für IQ-143 und IQ-238 auf Bakterien angewandten Methoden erfolgreich zur Modellierung der Effekte von Methylenblau auf verschiedene resistente Stämme von P. falciparum 3D7 angewandt werden. Dadurch konnte gezeigt werden, dass Methylenblau in einer Kombination mit anderen Präparaten gegen diesen Parasiten zum einen die Wirkung des Primärpräparates verstärkt, zum anderen aber auch in gewissem Maße vorhandene Resistenzen gegen das Primärpräparat zu verringern vermag. Somit konnte durch die vorgelegte Arbeit eine Pipeline zur Identifizierung der metabolischen Effekte verschiedener Wirkstoffe auf unterschiedliche Krankheitserreger erstellt werden. Diese Pipeline kann jederzeit auf andere Organismen ausgeweitet werden und stellt somit einen wichtigen Ansatz um Netzwerkeffekte verschiedener, potentieller Medikamente aufzuklären.
Although progenitor cells of the conducting airway have been spatially localized and some insights have been gained regarding their molecular phenotype, relatively little is known about the mechanisms regulating their maintenance, activation, and differentiation. This study investigates the potential roles of E-cadherin in mouse Clara cells, as these cells were shown to represent the progenitor/stem cells of the conducting airways and have been implicated as the cell of origin of human non-small cell lung cancer. Postnatal inactivation of E-cadherin affected Clara cell differentiation and compromised airway regeneration under injury conditions. In steady-state adult lung, overexpression of the dominant negative E-cadherin led to an expansion of the bronchiolar stem cells and decreased differentiation concomitant with canonical Wnt signaling activation. Expansion of the bronchiolar stem cell pool was associated with an incessant proliferation of neuroepithelial body-associated Clara cells that ultimately gave rise to bronchiolar hyperplasia. Despite progressive hyperplasia, only a minority of the mice developed pulmonary solid tumors, suggesting that the loss of E-cadherin function leads to tumor formation when additional mutations are sustained. The present study reveals that E-cadherin plays a critical role in the regulation of proliferation and homeostasis of the epithelial cells lining the conducting airways.
A metacommunity approach will be a useful framework to assess and predict changes in biodiversity in spatially structured landscapes and changing environments. However, the relationship between two core elements of metacommunity dynamics, dispersal and species interaction are not well understood. Most theoretical studies on dispersal evolution assume that target species are in isolation and do not interact with other species although the species interactions and community structure should have strong interdependence with dispersal. On the one hand, a species interaction can change the cost and benefit structure of dispersing in relation to non-dispersing individuals. On the other hand, with dispersal, an individual can follow respectively avoid species partners. Moreover, it is also important to explore the interdependence between dispersal and species interaction with spatial and temporal heterogeneity of environment because it would allow us to gain more understanding about responses of community to disturbances such as habitat destruction or global climate change, and this aspect is up to now not well-studied. In this thesis, I focus on the interactive and evolutionary feedback effects between dispersal and various types of interspecific interactions in different environmental settings. More specifically, I contrast dispersal evolution in scenarios with different types of interactions (chapter 2), explore the concurrent evolution of dispersal and habitat niche width (specialization) in spatial heterogeneous landscape (chapter 3) and consider (potential) multidimensional evolutionary responses under climate change (chapter 4). Moreover, I investigate consequences of different dispersal probability and group tolerance on group formation respectively group composition and the coexistence of ‘marker types’ (chapter 5). For all studies, I utilize individual-based models of single or multiple species within spatially explicit (grid-based) landscapes. In chapter 5, I also use an analytical model in addition to an individual-based model to predict phenomenon in group recognition and group formation. ...
