In dieser Studie wurden 280 Patienten, die im Zeitraum vom 01.01.2008 bis
30.06.2012 am Universitätsklinikum Würzburg aufgrund einer Raumforderung der
Leber operiert wurden, analysiert. Davon konnten 150 Patienten mit insgesamt 385
Tumoren bewertet werden. Die zentrale Fragestellung der Arbeit beschäftigte sich
damit, die Sensitivität der MR-DWI mit anderen präoperativen bildgebenden
Untersuchungsmethoden zu bewerten. Bisherige Studien beschäftigten sich bereits
mit dem Vergleich der MR-DWI mit anderen diagnostischen Verfahren. Jedoch
wurden dabei nicht konkret die aus Sicht eines Chirurgen wichtigen Punkte detailliert
beleuchtet. In dieser Arbeit wurden daher die Ergebnisse der Befunde der MR-DWI,
CT und konventionellem MRT dem intraoperativen und histopathologisch
gesichertem Endbefund gegenübergestellt und bezüglich der Parameter:
Tumordignität, Tumoranzahl, Tumorgröße und Tumorlokalisation verglichen.
Während die Sensitivität der MR-DWI zur Tumorlokalisation etwas unter der
Sensitivität der CT und MRT lag, konnten für die Parameter Tumoranzahl und
Tumordignität bessere Ergebnisse der MR-DWI als für die CT herausgestellt werden.
Die Genauigkeit der DWI für diese beiden Punkte liegt nah bei der hohen Sensitivität
der MRT. Jedoch müssen die sehr guten Ergebnisse der MRT in dieser Arbeit durch
die geringe Anzahl der MRT Untersuchungen relativiert werden.
Bezüglich der Tumoranzahl ergibt sich bei der MR-DWI die höchste Sensitivität,
gefolgt von MRT und CT.
Zusammenfassend kann gesagt werden, dass die MR-DWI wie auch die CT und
MRT ein sehr sensitives Verfahren zur Detektion von Leberläsionen darstellt. Dabei
müssen gewisse Vor- und Nachteile gegenüber den anderen Methoden abgewogen
werden. Eine als ungünstig zu wertende Strahlenexposition besteht nicht, ebenso
kein zusätzlicher zeitlicher Aufwand.
In nahezu allen hier untersuchten Bereichen zeigte die MR-DWI vorrangig bessere
Resultate als die CT. Im Vergleich mit der MRT konnten geringfügig niedrigere oder
ähnlich gute Werte ermittelt werden. In Bezug auf die Tumoranzahl war die MR-DWI
das sensitivste Verfahren.
Um die hier erbrachten Ergebnisse zu sichern, sollten zukünftige Studien die
Homogenität der Patienten sowie die Anzahl der CT, MRT und MR-DWI
Untersuchungen erhöhen. Weiterhin könnten die Geräteeinstellungen bei der MRDWI
an bestimmte Fragestellungen angepasst und so genauere Ergebnisse erzielt
werden. Bruegel et al konnte für unterschiedliche Leberläsionen bereits die
charakteristischen ADC-Werte ermitteln. In einer weiteren Studie von
Kilickesmez wurden ebenfalls maligne und benigne Leberläsionen über die
entsprechenden ADC-Werte voneinander abgegrenzt. Daraus ergab sich die
Empfehlung, zur Diagnostik von Leberläsionen die konventionelle MRT mit der MRDWI
zu ergänzen.
Ein weiterer Ansatzpunkt ist die Reduktion von Störfaktoren für die MR-DWI und
damit die Verbesserung der Bildbeurteilung. In einer Arbeit von Yu-Chun wurde
beispielsweise die optimale Temperatur für die MR-DWI Schnittbildgebung am
Tiermodell angewandt und so auf molekularer Ebene die Artefaktbildung reduziert.
Eine Verbesserung konnte auch durch Ergänzung der MR-DWI mit anderen
Verfahren nachgewiesen werden. MR-DWI in Verbindung mit Kontrastmittel ergab
eine erhöhte Sensitivität für kleine HCC bis 2cm als Kontrastmittel-MRT ohne DWI.
Es ist zu erwarten, dass die MR-DWI in Zukunft durch exaktere Einstellungen der b-
Werte und ADC-Messungen in Abhängigkeit der zu untersuchenden Organregion,
durch Reduktion störender Faktoren oder durch Kombination mit anderen
bildgebenden Verfahren zur Optimierung der präoperativen Diagnostik bei
Lebertumoren beiträgt.
In this dissertation the electronic and high-energy optical properties of thin nanoscale
films of the magnetic topological insulator (MTI) (V,Cr)y(BixSb1-x)2-yTe3 are studied
by means of X-ray photoelectron spectroscopy (XPS) and electron energy-loss
spectroscopy (EELS). Magnetic topological insulators are presently of broad interest
as the combination of ferromagnetism and spin-orbit coupling in these materials
leads to a new topological phase, the quantum anomalous Hall state (QAHS), with
dissipation less conduction channels. Determining and controlling the physical
properties of these complex materials is therefore desirable for a fundamental understanding
of the QAHS and for their possible application in spintronics. EELS can
directly probe the electron energy-loss function of a material from which one can
obtain the complex dynamic dielectric function by means of the Kramers-Kronig
transformation and the Drude-Lindhard model of plasmon oscillations.
The XPS core-level spectra in (V,Cr)y(BixSb1-x)2-yTe3 are analyzed in detail with
regards to inelastic background contributions. It is shown that the spectra can be
accurately described based on the electron energy-loss function obtained from an
independent EELS measurement. This allows for a comprehensive and quantitative
analysis of the XPS data, which will facilitate future core-level spectroscopy studies
in this class of topological materials. From the EELS data, furthermore, the bulk and
surface optical properties were estimated, and compared to ab initio calculations
based on density functional theory (DFT) performed in the GW approximation
for Sb2Te3. The experimental results show a good agreement with the calculated
complex dielectric function and the calculated energy-loss function. The positions of
the main plasmon modes reported here are expected to be generally similar in other
materials in this class of nanoscale TI films. Hence, the present work introduces
EELS as a powerful method to access the high-energy optical properties of TI
thin films. Based on the presented results it will be interesting to explore more
systematically the effects of stoichiometry, magnetic doping, film thickness and
surface morphology on the electron-loss function, potentially leading to a better
understanding of the complex interplay of structural, electronic, magnetic and
optical properties in MTI nanostructures.
Neurotrophine beeinflussen durch die Modulation von Prozessen wie Zellproliferation, -migration, Apoptose und Synapsenbildung entscheidend die neuronale Plastizität. Sie gelten deshalb als Kandidatengene neuronaler Entwicklungsstörungen wie Autismus-Spektrum-Störungen (ASS). Die vorgelegte Arbeit zielt auf die weitere Klärung der Rolle von Brain Derived Neurotrophic Factor (BDNF) bei der Ätiopathophysiologie der ASS durch Expressionsanalysen im Blut als potenziellem Surrogat zentralnervöser Prozesse.
In gut charakterisierten ASS-Stichproben und - neben gesunden Kontrollprobanden - einer klinischen Kontrollgruppe von Patienten mit Aufmerksamkeitsdefizit-/ Hyperaktivitätsstörung (ADHS) wurde die BDNF-mRNA-Expression in Vollblut sowie BDNF-Proteinserumkonzentrationen untersucht. Zusätzlich wurden mögliche Einflussfaktoren auf die BDNF-Werte wie Alter, IQ, autismusspezifische Symptomatik, Komorbidität und Medikation analysiert.
In einer ersten Stichprobe (ASS-Patienten versus gesunde Kontrollen) wurden
signifikant erniedrigte BDNF-Serumkonzentrationen in der Patientengruppe mittels
Enzyme-Linked-Immunosorbent-Assay gemessen (p = 0,040). In einer zweiten unabhängigen Stichprobe (Patienten mit ASS, Patienten mit ADHS und gesunde Kontrollen) wurde auf mRNA-Ebene mittels quantitativer Real-Time-Polymerasekettenreaktion ebenfalls ein signifikanter Gruppenunterschied ermittelt mit erniedrigter BDNF-Expression in der ASS-Gruppe im Vergleich zu gesunder Kontrollgruppe (p = 0,011), sowie einem Trend zu erniedrigten BDNF-Werten bei ADHS-Patienten im Vergleich zu gesunden Probanden (p = 0,097). Des Weiteren wurde eine signifikante negative Korrelation zwischen Alter und BDNF-mRNA-Expression bei Patienten mit ASS sowie eine positive Korrelation von Alter und BDNF-Serumkonzentrationen bei gesunden Kontrollen gemessen. Auch korrelierten die BDNF-Werte im Serum mit der Ausprägung des autistischen Phänotyps. In einer Subgruppe der ADHS-Patienten wurde kein Einfluss von Psychostimulanzien auf die BDNF-mRNA-Expression gemessen.
Der Einbezug größerer Stichproben sowie die systematische Erfassung weiterer potenzieller Einflussfaktoren auf die BDNF-Expression (wie pubertärer Entwicklungsstand bzw. Geschlechtshormonkonzentrationen) könnten in zukünftigen Studien zu einer weiteren Klärung der pathophysiologischen Rolle von BDNF bei Kindern und Jugendlichen mit ASS beitragen.
In the context of quantum mechanical calculations, the properties of non-adiabatic coupling in a small system, the Shin-Metiu model, is investigated.
The transition from adiabatic to non-adiabatic dynamics is elucidated in modifying the electron-nuclear interaction. This allows the comparison of weakly correlated electron-nuclear motion with the case where the strong correlations determine the dynamics.
The studies of the model are extended to include spectroscopical transitions being present in two-dimensional and degenerate four-wave mixing spectroscopy.
Furthermore, the quantum and classical time-evolution of the coupled motion in the complete electron-nuclear phase space is compared for the two coupling cases.
Additionally, the numerically exact electron flux within the weak coupling case is compared to the Born-Oppenheimer treatment.
In the last part of the thesis, the model is extended to two dimensions.
The system then possesses potential energy surfaces which exhibit a typical 'Mexican hat'-like structure and a conical intersection in the adiabatic representation.
Thus, it is possible to map properties of the system onto a vibronic coupling (Jahn-Teller) hamiltonian. Exact wave-packet propagations as well as nuclear wave-packet dynamics in the adiabatic and diabatic representation are performed.
Fremdschämen or Fremdscham, a negative emotion which arises while observing someone behave inappropriately, comes to fame after the turn of the millennium in german speaking countries. There, they name it literally „other‘s shame“ and it becomes obvious that this emotion happens most commonly while watching TV: reality shows, talent shows and bad comedies. The word even makes it to the dictionaries starting 2009, as its use increases unstoppably in everyday language, starting to get used in more and more situations, seemingly as a synonym of embarrassing or shameful. Still, a look in the emotional research on the subject returns exactly zero results as of 2011, leaving open the question as of what this emotion might be, and what it is not. The present wort aims at explaining not only the phenomenon of Fremdschämen, but also the Emotion behind it - Embarrassment -, at a process level.
New experimental methods have drastically accelerated the pace and quantity at which biological data is generated. High-throughput DNA sequencing is one of the pivotal new technologies. It offers a number of novel applications in various fields of biology, including ecology, evolution, and genomics. However, together with those opportunities many new challenges arise. Specialized algorithms and software are required to cope with the amount of data, often requiring substantial training in bioinformatic methods. Another way to make those data accessible to non-bioinformaticians is the development of programs with intuitive user interfaces.
In my thesis I developed analyses and programs to tackle current problems with high-throughput data in biology. In the field of ecology this covers the establishment of the bioinformatic workflow for pollen DNA meta-barcoding. Furthermore, I developed an application that facilitates the analysis of ecological communities in the context of their traits. Information from multiple public databases have been aggregated and can now be mapped automatically to existing community tables for interactive inspection. In evolution the new data are used to reconstruct phylogenetic trees from multiple genes. I developed the tool bcgTree to automate this process for bacteria. Many plant genomes have been sequenced in current years. Sequencing reads of those projects also contain data from the chloroplasts. The tool chloroExtractor supports the targeted extraction and analysis of the chloroplast genome. To compare the structure of multiple genomes specialized software is required for calculation and visualization of the relationships. I developed AliTV to address this. In contrast to existing programs for this task it allows interactive adjustments of produced graphics. Thus, facilitating the discovery of biologically relevant information. Another application I developed helps to analyze transcriptomes even if no reference genome is present. This is achieved by aggregating the different pieces of information, like functional annotation and expression level, for each transcript in a web platform. Scientists can then search, filter, subset, and visualize the transcriptome.
Together the methods and tools expedite insights into biological systems that were not possible before.
Hintergrund. Die gesetzlich vorgeschriebene Gefährdungsbeurteilung psychischer Belastung gewinnt zunehmend an Bedeutung. Ein Standardinstrument, das in diesem Rahmen seit einigen Jahren zur Anwendung kommt, ist der Kurzfragebogens zur Arbeitsanalyse (KFZA), von Prümper et al. (1995). Dieser Fragebogen wurde ursprünglich für die Beurteilung von Bildschirmarbeitsplätzen konzipiert und für diese Berufsgruppe validiert. Ziel der vorliegenden Arbeit war es, die faktorielle Validität des KFZA bei einem Einsatz im Gesundheitswesen mittels einer explorativen Faktorenanalyse zu überprüfen. Da eine Fragebogenversion zum Einsatz kam, die zusätzlich spezifische Ergänzungsfragen für das Gesundheitswesen enthielt, sollte in einem zweiten Schritt auch dieser erweiterte KFZA einer Faktorenanalyse unterzogen werden.
Methodik. Insgesamt 1731 Datensätze waren über einen Zeitraum von zehn Jahren in verschiedenen norddeutschen Krankenhäusern als Routinedaten erhoben worden. Nach listenweisem Fallausschluss in Folge des Einsatzes unterschiedlicher Fragebogenvarianten standen für den KFZA 1163 Datensätze und davon 1095 Datensätze für den erweiterten KFZA zur faktorenanalytischen Auswertung zur Verfügung. Die 26 Items des KFZA bzw. die 37 Items der erweiterten Version wurden einer explorativen Faktorenanalyse nach der Hauptkomponentenmethode unterzogen. Die Zahl der Faktoren wurde sowohl mittels Kaiser- als auch Scree-Kriterium bestimmt. Für die Interpretation der Faktoren wurden diese sowohl orthogonal nach der Varimax-Methode als auch direct-oblimin rotiert. Zur Abschätzung der Reliabilität wurde die interne Konsistenz anhand des Cronbach-α-Koeffizienten berechnet.
Ergebnisse. Für die 26 Items des KFZA führte das Kaiser-Kriterium zu einer 7-Faktoren-Lösung mit einer Gesamtvarianzaufklärung von 62,0%, der Scree-Plot dagegen deutete auf vier Faktoren hin. Orthogonale und oblique Rotation brachten vergleichbare Ergebnisse. Die inhaltliche Interpretation unterstützte die Anzahl von sieben Faktoren, die wie folgt benannt wurden: „Soziale Beziehungen“, „Handlungsspielraum“, „Partizipations- und Entwicklungs-möglichkeiten“, „Quantitative Arbeitsbelastungen“, „Umgebungsbelastungen“, „Vielseitigkeit“ und „Qualitative Arbeitsbelastungen“. Für diese Skalen, die jeweils 2 bis 6 Items umfassten, konnten Cronbach-α-Koeffizienten zwischen 0,63 und 0,80 ermittelt werden. Die Faktorenanalyse des erweiterten KFZA mit insgesamt 37 Items führte nach Bestimmung des Kaiser-Kriteriums und Betrachtung der inhaltlichen Plausibilität zu einer 9-Faktoren-Lösung mit einer Gesamtvarianzaufklärung von 59,5%. Die beiden zusätzlichen Faktoren wurden mit „Fehlbeanspruchungsfolgen“ und „Emotionale Belastungen“ benannt. Die Werte des Cronbach-α-Koeffizienten lagen für diese Skalen zwischen 0,63 und 0,87.
Diskussion. Statt der von den Autoren des KFZA beschriebenen elf Faktoren wurden bei einem Einsatz im Gesundheitswesen sieben Faktoren ermittelt. Auch wenn sich die Anzahl der Faktoren reduzierte, ließ sich die Struktur inhaltlich relativ gut replizieren. Besonders die Items des KFZA-Faktors „Ganzheitlichkeit“ erwiesen sich jedoch für den Einsatz im Gesundheitswesen als nicht passgenau. Die Ergänzungsitems des erweiterten KFZA bildeten zwei zusätzliche Faktoren bzw. ließen sich den zuvor ermittelten Faktoren sinnvoll zuordnen.
Die vorliegende Arbeit liefert somit einen Beitrag zur Einschätzung der Validität dieses in der Praxis häufig eingesetzten Instruments. Die psychometrische Prüfung kann jedoch noch nicht als vollständig erachtet werden und sollte in nachfolgenden Studien fortgeführt werden.
Kürzlich wurden bei immunvermittelten Neuropathien Autoantikörper gegen Proteine
des paranodalen axoglialen Komplexes beschrieben. Deren Charakteristika,
Prävalenzen, pathophysiologische Relevanz sowie Bedeutung für Diagnostik
und Therapie sind jedoch noch nicht abschließend erforscht.
In dieser Studie wurden daher Seren und Plasmapheresematerial (PE-Material)
von 150 Patienten mit inflammatorischen Neuropathien, nämlich 105 mit chronisch
inflammatorischer demyelinisierender Polyneuropathie (CIDP), 21 mit Guillain-
Barré-Syndrom (GBS) und 24 mit multifokaler motorischer Neuropathie
(MMN), welche etablierte diagnostische Kriterien der jeweiligen Krankheit erfüllen,
sowie 74 Kontrollen mittels immunhistochemischen Färbungen an murinen
Zupfnervenpräparaten und/oder ELISA (Enzyme-linked Immunosorbent Assay)
auf Autoantikörper gegen die paranodalen Proteine Caspr, Contactin-1 und Neurofascin-
155 untersucht. Bei positivem Ergebnis wurde deren Spezifität mittels
immunhistochemischen Färbungen an transfizierten HEK (Human embryonic kidney)-
293-Zellen und Präinkubationsversuchen bestätigt. Es wurden die IgG-Subklassen
und die Antikörpertiter bestimmt und das Komplementbindungsverhalten
unter Zugabe von intravenösen Immunglobulinen (IVIG) mit zellbasierten und
ELISA-basierten Methoden analysiert. Klinische Merkmale und das Therapieansprechen
Antikörper-positiver Patienten wurden ermittelt und mit den experimentellen
Ergebnissen in Zusammenhang gesetzt.
IgG-Autoantikörper gegen Contactin-1 konnten bei vier Patienten mit CIDP nachgewiesen
werden, IgG-Autoantikörper gegen Caspr bei einem Patienten mit
CIDP und einer Patientin mit GBS. Es konnten keine weiteren Autoantikörper bei
CIDP-Patienten, GBS-Patienten, MMN-Patienten oder bei den Kontrollen detektiert
werden. Die Prävalenz von Autoantikörpern gegen axogliale paranodale Proteine
liegt somit in dieser Studie bei jeweils 4,76% bei CIDP und GBS und 0%
bei MMN. Die Antikörper gehörten bei Patienten in der akuten Erkrankungsphase
(zwei der CIDP-Patienten mit Anti-Contactin-1-Autoantikörpern und eine GBS-Patientin mit Anti-Caspr-Autoantikörpern) hauptsächlich den Subklassen IgG1
und IgG3 an, bei Patienten in der chronischen Phase (zwei der CIDP-Patienten
mit Anti-Contactin-1-Autoantikörpern, ein CIDP-Patient mit Anti-Caspr-Autoantikörpern)
überwog die Subklasse IgG4. Experimentell kam es zur Komplementbindung
und -aktivierung abhängig vom Gehalt der Subklassen IgG1-3, nicht
aber IgG4; diese konnte durch die Zugabe von IVIG dosisabhängig gemindert
werden. Alle Autoantikörper-positiven CIDP-Patienten zeigten einen GBS-artigen
Beginn mit einer schweren motorischen Beteiligung. Anti-Contactin-1-positive
Patienten kennzeichnete klinisch zusätzlich das Vorkommen einer Ataxie und eines
Tremors, Anti-Caspr-positive Patienten das Vorkommen starker neuropathischer
Schmerzen. Elektrophysiologisch standen neben Hinweisen auf eine Leitungsstörung
Zeichen einer axonalen Schädigung im Vordergrund. Als histopathologisches
Korrelat lagen eine nodale Architekturstörung und ein Axonverlust
vor. Die Patienten zeigten nur in der Anfangsphase der Erkrankung ein Ansprechen
auf IVIG. Bei drei CIDP-Patienten mit IgG4-Autoantikörpern (zwei Patienten
mit Anti-Contactin-1-Antikörpern und ein Patient mit Anti-Caspr-Antikörpern)
wurde eine Therapie mit Rituximab durchgeführt. Diese führte zu einer Titerreduktion
und zur zeitgleichen klinischen und elektrophysiologischen Befundbesserung
bei zwei Patienten.
Die in dieser Arbeit angewandten Screeningmethoden führten zum erfolgreichen
Nachweis von Autoantikörpern gegen paranodale axogliale Proteine. Die Patienten
mit positivem Autoantikörpernachweis definieren eine kleine Untergruppe mit
ähnlichen klinischen Merkmalen im Kollektiv der Patienten mit inflammatorischen
Polyneuropathien. Histopathologische Merkmale sowie das Therapieansprechen
auf antikörperdepletierende Therapie sprechen in Kombination mit den Ergebnissen
weiterer Studien zu paranodalen Autoantikörpern für eine pathogenetische
Relevanz der Autoantikörper. Mit einem charakteristischen, am Schnürring ansetzenden
Pathomechanismus könnten Neuropathien mit Nachweis von paranodalen
Autoantikörpern der kürzlich eingeführten Entität der Nodo-Paranodopathien
angehören. Die Komplementaktivierung und das Therapieansprechen der Patienten auf IVIG stehen möglicherweise in Zusammenhang mit der prädominanten
IgG-Subklasse. Diese könnte auch in Bezug auf die Chronifizierung eine
Rolle spielen. Der Nachweis von Autoantikörpern gegen paranodale Proteine hat
wohlmöglich in Zukunft direkte Konsequenzen auf das diagnostische und therapeutische
Prozedere bei Patienten mit CIDP und GBS; weitere klinische und experimentelle
Daten aus größeren, prospektiven Studien sind jedoch zum weiteren
Verständnis und zur Charakterisierung dieser Entität notwendig.
Kapitel 1 Darstellung und Reaktivität des Cyanoborylens (3)
Im Rahmen dieser Arbeit ist es gelungen, in einer dreistufigen Synthese das erste basenstabilisierte Cyanoborylen [(cAAC)B(CN)]4 (3) in hohen Ausbeuten darzustellen (Schema 64). Hervorzuheben ist hierbei, dass dieser Ansatz keine „klassische“ Metallborylen- Vorstufe benötigt, weshalb wenig Synthesestufen und bessere Ausbeuten erreicht werden konnten. Schema 64. Eine erste Besonderheit von [(cAAC)B(CN)]4 (3) ist, dass dieses das einzige bislang bekannte Borylen darstellt, welches eine Stabilisierung durch Oligomerisierung erfährt und somit in Folgereaktionen nicht erst in situ generiert werden muss. Die elektronische Untersuchung von 3 durch Cyclovoltammetrie hat zudem gezeigt, dass 3 ein Redoxpotential von E1/2 = −0.83 V besitzt und somit eine chemische Oxidation zu neuen Verbindungen führen könnte, was durch Umsetzung mit AgCN demonstriert wurde (Schema 65). Hierdurch konnte [(cAAC)B(CN)3] (4) erfolgreich dargestellt und vollständig charakterisiert werden. [(cAAC)B(CN)3] (4) ist erst das zweite strukturell untersuchte basenstabilisierte Tricyanoboran. Zudem wurde die Reaktivität von [(cAAC)B(CN)]4 (3) gegenüber verschiedenen Lewis-Basen untersucht. Ziel hierbei war es, das oligomere Strukturmotiv aufzubrechen und gemischte zweifach basenstabilisierte Borylene zu realisieren. Hierbei konnte eine deutliche Abhängigkeit von der Basenstärke und dem sterischen Anspruch der Lewis-Base aufgedeckt werden. So hat sich gezeigt, dass Lewis-Basen wie THF, MeCN, Pyridin und PEt3 zu schwach sind, um die oligomere Struktur aufzubrechen. Im Gegensatz dazu führten die Umsetzungen von [(cAAC)B(CN)]4 (3) mit den starken Lewis-Basen cAAC bzw. IPr zu keinerlei Umsatz, was vermutlich auf einen zu großen sterischen Anspruch zurückzuführen ist. Dementsprechend verlief die Umsetzung von [(cAAC)B(CN)]4 (3) mit der starken und sterisch nicht anspruchsvollen Base IMeMe erfolgreich und lieferte [(cAAC)B(CN)(IMeMe)] (5) in guten Ausbeuten (Schema 65). Während [(cAAC)B(CN)(PEt3)] (6) nicht durch Umsetzung von [(cAAC)B(CN)]4 (3) mit PEt3 zugänglich ist, konnte dieses jedoch auch durch Reduktion von [(cAAC)BBr2(CN)] (2) in Gegenwart von PEt3 erhalten werden (Schema 66). [(cAAC)B(CN)(PEt3)] (6) stellt hierbei das das bislang erste bekannte Phosphan-stabilisierte Borylen dar. Schema 66. Kristallstruktur und Synthese von [(cAAC)B(CN)(PEt3)] 6.
Kapitel 2 Reaktivität von 3 gegenüber Chalcogenen und Chalcogeniden
In weiterführenden Studien wurde zudem die Reaktivität von 3 gegenüber Chalcogenen und Chalcogeniden im Detail untersucht. Durch Verwendung der entsprechenden Stöchiometrie konnte 3 hierbei selektiv zu den Bor-Chalcogen-Heterocyclen 9, 10, 13-15 umgesetzt werden (Schema 67). Schema 67. Darstellung von 9, 10, 13-15. Diese Ergebnisse wurden anschließend mit der Reaktivität des Konstitutionsisomers LII verglichen. In diesem Zusammenhang konnten 11 und 12 durch stöchiometrische Reaktionsführung dargestellt werden (Schema 68), welche nachfolgend in die bereits erwähnten Verbindungen 9 und 10 überführt werden konnten (Schema 69). Des Weiteren konnte 3 erfolgreich mit Ph2Se2, Me2Se2 und Ph2S2 zu 16-18 umgesetzt werden (Schema 70), wobei 16 und 18 auch durch Umsetzung von LII mit Ph2Se2 bzw. Ph2S2 zugänglich sind (Schema 70). Schema 70. Synthese von 16-18. Das tetramere Borylen 3 und das Diboren LII zeigen ähnliche Reaktivitäten gegenüber elementaren Chalcogenen sowie Dichalcogeniden. Lediglich die Darstellung der dreigliedrigen B2E-Heterocyclen 11 und 12 gelingt selektiv nur ausgehend von LII.
Kapitel 3 Darstellung und Reaktivität des Borylanions (19)
Ein weiterer Aspekt dieser Arbeit beschäftigte sich mit der Synthese und Reaktivität des Borylanions 19, eines der wenigen bekannten nukleophilen Borspezies. Der Zugang zu 19 durch Deprotonierung von 1 (Schema 71) ist hierbei besonders bemerkenswert, da es eine bis dato kaum bekannte bzw. verwendete Methode ist, da borgebundene Wasserstoffatome in der Regel hydridischer Natur sind, weshalb eine Deprotonierung normalerweise nicht möglich ist und nur für zwei weitere Systeme beschrieben ist. Hierzu zählen die Synthese des Dianions XLVII[6a, 6b] und die Synthese des Borylanions XLVIII[45]. Eine Gemeinsamkeit dieser drei Spezies ist die Gegenwart elektronenziehender Cyanidsubstituenten welche eine Umpolung der B‒H-Bindung bedingen, wodurch eine Deprotonierung erst ermöglicht wird. Schema 71. Synthese von 19. Um diesen Sachverhalt genauer zu untersuchen, wurden Rechnungen durchgeführt und die partiellen Ladungen (NBO) des borgebunden Wasserstoff an BH3, [(cAAC)BH3] und 1 auf dem BP86/def2-SVP-Niveau berechnet (Abbildung 53). Abbildung 53. Teilladungen (NBO) von BH3, [(cAAC)BH3] und 1 (BP86/def2-SVP). Durch Austausch eines der Hydride in [(cAAC)BH3] durch eine Cyanogruppe werden die borgebunden Wasserstoffe in 1 deutlich protischer (+0.038, +0.080), wobei schon durch Koordination des cAAC-Liganden an BH3 zwei der vorher hydridischen Wasserstoffe (BH3: partielle Ladung: –0.101) erheblich positiver geladen wird (+0.050). Der nukleophile Charakter von 19 wurde anschließend durch Reaktivitätsstudien untersucht. So führte die Umsetzung von 19 mit [(PPh3)AuCl] zur Bildung von [(cAAC)BH(CN)(AuPPh3)] (20) (Schema 72). Während die Umsetzung von 19 mit Tritylderivaten keine isolierbare Verbindung lieferte, konnte durch Umsetzung mit den schweren, weichen Homologen R3ECl (R = Ph, E = Ge, Sn und Pb; R = Me, E = Sn) eine ganze Reihe von Boranen dargestellt werden (Schema 72). Schema 72. Synthese von 20-24. Die Umsetzung der entsprechenden Silylderivate R3SiCl war hingegen mit einem anderen Reaktionsverlauf verbunden (Schema 73). Schema 73. Synthese von 25-28. Demnach erfolgt die Reaktion von 19, im Gegensatz zu den höheren Homologen, mit den Silylderivaten nicht am weichen, nukleophilen Borzentrum sondern am härteren Cyanostickstoffatom. Demzufolge wurden hierbei zunächst die Silylisonitrilverbindungen 25 und 26 gebildet, wobei 25 labil ist und innerhalb kürzester Zeit in 27 übergeht. Im Gegensatz dazu konnte 28 nur durch Bestrahlung von 26 dargestellt werden. Die Bindungsverhältnisse in 26 wurden zudem auch durch DFT-Rechnungen auf dem BP86/def2-SVP-Niveau untersucht. Die Analyse der Kohn–Sham MOs offenbarte hierbei ein HOMO mit π-Bindungscharakter über die gesamte CcAAC‒B‒CCN-Einheit mit angrenzendem π-Antibindungscharakter über die C‒NEinheiten beider Donorliganden (Abbildung 54). Während die Umsetzung von 26 mit Cu(I)Cl dessen hohes Reduktionsvermögen verdeutlichte, führte die Umsetzung mit Lithium in THF zur Bildung des Borylanions 19 und LiSiPh3. Die Reaktion von 26 mit BH3∙SMe2 lieferte hingegen quantitativ [(cAAC)BH3] (29), während bei Umsetzung mit Ph3SnCl quantitativ 22 gebildet wurde (Schema 74). Dieses sehr unterschiedliche Reaktionsverhalten rechtfertigt eine Beschreibung von 26 sowohl als ein Silylisonitrilborylen, als auch eine zwitterionische Silyliumboryl-Spezies. Schema 74. Kapitel 4 Darstellung und Reaktivität von [(cAAC)BH3] (29)
Da 1 selektiv deprotoniert werden kann und [(cAAC)BH3] (29) Rechnungen zufolge ebenfalls borgebundene Wasserstoffe mit protischem Charakter besitzt, wurde versucht, diese Reaktivität auf 29 zu übertragen. Demzufolge wurde im Rahmen dieser Arbeit [(cAAC)BH3] (29) dargestellt und dessen Reaktivität gegenüber anionischen (Schema 75) und neutralen (Schema 76) Nukleophilen untersucht. Es hat sich jedoch gezeigt, dass die Umsetzung von [(cAAC)BH3] (29) mit Lithiumorganylen nicht zur Deprotonierung führt, sondern zur Bildung der Lithiumborate 30, 32 und 34, unabhängig von der Hybridisierung des Lithiumorganyls (sp3: LiNp, sp2: LiMes, sp: LiCCPh). Der Reaktionsmechanismus wurde durch DFT-Rechnungen untersucht (Abbildung 47). Diese zeigen eindeutig, das [(cAAC)BH3] (29) in einem Gleichgewicht mit dem entsprechenden Boran [(cAAC‒H)BH2] steht. Bei der stark exergonischen nukleophilen Addition der entsprechenden Basen wird [(cAAC‒H)BH2] aus dem Gleichgewicht entfernt (30: −29.6 kcal∙mol‒1; 32: ‒12.4 kcal∙mol‒1) und die Lithiumborate 30 und 32 gebildet. Diese Lithiumborate gehen dann durch Reaktion mit Me3SiCl in die entsprechenden cAACBoranaddukten 31, 33 und 35 über (Schema 75). Schema 75. Synthese von 30-35. Diese zweistufige Synthese ist deshalb bemerkenswert, da dies einer ungewöhnlichen Substitution an einem sp3-Boran gleichkommt. Des Weiteren wurde die Reaktivität von [(cAAC)BH3] (29) gegenüber neutralen Lewis-Basen untersucht. So konnte bei der Umsetzung mit cAAC Verbindung 36 und bei der Umsetzung mit Pyridin Verbindung 37 erhalten werden (Schema 76). Schema 76. Synthese von 36 und 37. Der Mechanismus der Bildung von 36 und 37 wurde ebenfalls durch DFT-Rechnungen untersucht, welche auf eine reversible Reaktion des Pyridin-Addukts 37 hindeutet. Dies konnte auch experimentell bestätigt werden. Im Gegensatz dazu ist die Bildung von 36 irreversibel.
Kapitel 5 Darstellung und Vergleich neuer Diborene
Im Rahmen dieser Arbeit ist es zudem gelungen, eine Reihe an NHC-Boranaddukten (42-50) darzustellen und diese zum Großteil in die entsprechenden Diborene (51-58) zu überführen (Schema 77). Schema 77. Synthese der NHC-Boranaddukte 42-50 sowie deren Umsetzung zu den Diborenen 51-58. Die meisten Verbindungen konnten hierbei vollständig charakterisiert und somit die NMR-spektroskopischen und strukturellen Daten miteinander verglichen werden. Die 11B-NMRSignale von 51-58 wurden in einem engen Bereich (20.2 bis 22.5 ppm) beobachtet, welcher sich mit dem von X und XI (21.3 und 22.4 ppm)[17] deckt. Im Festkörper weisen die Diborene einen B‒B-Abstand zwischen 1.576(4) Å (51) und 1.603(4) Å (54) auf, ohne dass ein Trend
erkennbar ist. Dieser Bereich ist zudem nahezu identisch mit bereits bekannten IMe-stabilisierten 1,2-Diaryldiborenen (1.585(4) bis 1.593(5) Å).[16-17] Einige dieser Diborene sind durch die entsprechende Wahl des Substitutionsmusters sehr labil und konnten deshalb nicht isoliert werden. Es ist dennoch gelungen UV-vis-spektroskopische Daten von 51, 52, 57 und 58 zu erhalten (Abbildung 55). Abbildung 55. UV-vis-Absorptionsspektren von 51, 52, 57 und 58. Die genaue Analyse der UV-vis-Spektren von 51, 52, 57 und 58 offenbart eine gewisse Abhängigkeit der Maxima vom Substitutionsmuster. Der Vergleich der Diborene 51-58 hat gezeigt, dass das Substitutionsmuster einen entscheidenden Einfluss auf die Lage der Grenzorbitale hat, was die Eigenschaften der Diborene deutlich verändert. So führte die Einführung einer Diphenylaminogruppe am Thienylrest zur Aufhebung der Koplanarität der Th‒B=B‒Th-Ebene, weshalb die entsprechenden Spezies durch die fehlende π-Konjugation sehr labil sind. Diese Beeinflussung der Koplanarität konnte bereits in kleinem Ausmaß bei der Substitution durch eine Me3Si-Gruppe beobachtet werden. Auch der Einfluss unterschiedlicher NHCs wurde untersucht. Während die Einführung von IMeMe kaum einen Einfluss auf die Absorptionsmaxima zeigt, führt die Verwendung von IPr zu einer deutlichen Verschiebung. Als das stabilste Diboren erwies sich im Rahmen dieser Untersuchung das [(IMe)BTh)]2 (X).
In this work the energy transfer and excitonic coupling in different chromophore arrangements were investigated. A difference in the coupling strength was introduced by varring the connecting unit and the spacial orientation relative to each other.
The synthesis of the 2,7-substituted pyrene compounds could be optimised and good yields of HAB 1 and HAB 2 and small amounts of HAB 2 could be achieved by cobalt-catalysed trimerisation or Diels Alder reaction in the end. Absorption and fluorescence spectra reveal strong intramolecular interactions between the pyrene molecules in the HAB 1. Excitation spectra recorded at the high and low energy fluorescence suggest the contribution of two components to the spectra. One being similar to the ground state aggregate and a second species similar to undisturbed pyrene. All these feature can be accounted to two different fluorescent states which are due to electronical decoupling in the excited state. Due to the strong intramolecular coupling already in the ground state of the molecule, no energy transfer could be studied, as the six pyrene units cannot be seen as separate spectroscopic entities between which energy could be transferred.
In the second part of this thesis dye conjugates of different size and alignment were synthesised to study the interaction of the transition-dipole moments. Therefore a systematic investigation of Sonogashira conditions was performed in order to obtain good yields of the desired compounds and keep dehalogenation at a minimum level. Nevertheless only the symmetrical triads could be purified as the asymmeric triads and pentades proved to decompose during purification.
The pyrene containing triads Py2B and Py2SQB show small interactions already in the ground state represented by red shifts of the spectra and a broadening of the bands. Nevertheless, these interactions are in the weak coupling regime and energy transfer between the constituents is possible. On the contrary in the TA spectra it is obvious that always the whole triad, at least to some extend is excited. To question if the excitation of the high energy state is deactivated by energy transfer or rather IC in a superchromophore could not be distinguished in the course of this work. At present additional time-dependent calculations of the dynamics are in progress to get a deeper understanding of the photophysical processes taking place in the triads.
The dye conjugates B2SQB-3 and (SQB)2B-4 can be assigned to the strong interaction range and hence are describable by exciton theory. The transition-dipole moments proved to be more than additive and increase for both compounds from absorption to fluorescence. This can be explained by an enhancement of the coupling in the relaxed excited state compared to the absorption into the Franck-Condon state due to a more steep potential energy surface in the excited state and hence smaller fluctuations.
In the last part of this thesis the influence of disrupting electronical communication by implementing a rigid non-conjugated bridge in a bichromophoric trans-squaraine system was tested. While the flexible linked squaraines show complex spectra due to different conformers the SQA2Anth compound is rigified and no rotation is possible. This change in flexibility is represented in the steady-state spectra where just one main absorption and fluorescence band is present due to a single allowed excitonic state. The system proves to own an excited state that is completely delocalised over the whole molecule.
The interaction of synaptic proteins orchestrate the function of one of the most complex organs, the brain. The multitude of molecular elements influencing neurological correlations makes imaging processes complicated since conventional fluorescence microscopy methods are unable to resolve structures beyond the diffraction-limit.
The implementation of super-resolution fluorescence microscopy into the field of neuroscience allows the visualisation of the fine details of neural connectivity. The key element of my thesis is the super-resolution technique dSTORM (direct Stochastic Optical Reconstruction Microscopy) and its optimisation as a multi-colour approach. Capturing more than one target, I aim to unravel the distribution of synaptic proteins with nanometer precision and set them into a structural and quantitative context with one another. Therefore dSTORM specific protocols are optimized to serve the peculiarities of particular neural samples.
In one project the brain derived neurotrophic factor (BDNF) is investigated in primary, hippocampal neurons. With a precision beyond 15 nm, preand post-synaptic sites can be identified by staining the active zone proteins bassoon and homer. As a result, hallmarks of mature synapses can be exhibited. The single molecule sensitivity of dSTORM enables the measurement of endogenous BDNF and locates BDNF granules aligned with glutamatergic pre-synapses. This data proofs that hippocampal neurons are capable of enriching BDNF within the mature glutamatergic pre-synapse, possibly influencing synaptic plasticity.
The distribution of the metabotropic glutamate receptor mGlu4 is investigated in physiological brain slices enabling the analysis of the receptor in its natural environment. With dual-colour dSTORM, the spatial arrangement of the mGlu4 receptor in the pre-synaptic sites of parallel fibres in the molecular layer of the mouse cerebellum is visualized, as well as a four to six-fold increase in the density of the receptor in the active zone compared to the nearby environment. Prior functional measurements show that metabotropic glutamate receptors influence voltage-gated calcium channels and proteins that are involved in synaptic vesicle priming. Corresponding dSTORM data indeed suggests that a subset of the mGlu4 receptor is correlated with the voltage-gated calcium channel Cav2.1 on distances around 60 nm.
These results are based on the improvement of the direct analysis of localisation data. Tools like coordinated based correlation analysis and nearest neighbour analysis of clusters centroids are used complementary to map protein connections of the synapse. Limits and possible improvements of these tools are discussed to foster the quantitative analysis of single molecule localisation microscopy data.
Performing super-resolution microscopy on complex samples like brain slices benefits from a maximised field of view in combination with the visualisation of more than two targets to set the protein of interest in a cellular context. This challenge served as a motivation to establish a workflow for correlated structured illumination microscopy (SIM) and dSTORM. The development of the visualisation software coSIdSTORM promotes the combination of these powerful super-resolution techniques even on separated setups. As an example, synapses in the cerebellum that are affiliated to the parallel fibres and the dendrites of the Purkinje cells are identified by SIM and the protein bassoon of those pre-synapses is visualised threedimensionally with nanoscopic precision by dSTORM.
In this work I placed emphasis on the improvement of multi-colour super-resolution imaging and its analysing tools to enable the investigation of synaptic proteins. The unravelling of the structural arrangement of investigated proteins supports the building of a synapse model and therefore helps to understand the relation between structure and function in neural transmission processes.
SPRED2 ist ein Inhibitor des Ras/ERK-MAPK-Signalwegs. Um die Folgen einer SPRED2-Defizienz zu erforschen, wurden im Rahmen vorheriger von Ullrich et al. durchgeführter Untersuchungen mittels Gene-Trap-Methode bereits mannigfaltige Auffälligkeiten im Phänotyp der SPRED2-Mäuse festgestellt. So zeigten die Tiere einen Hypochondroplasie-ähnlichen Zwergenwuchs, Verhaltensauffälligkeiten, einen krankhaft gesteigerten Wasserkonsum und nicht zuletzt eine deutlich reduzierte Lebenserwartung im Vergleich mit den WT-Tieren. Des Weiteren fielen erhöhte Aldosteronspiegel auf, die bei näheren Untersuchungen nicht einer erhöhten Aktivität des RAAS geschuldet zu sein schienen. Vielmehr zeigte sich eine deutlich erhöhte Aldosteron-Synthase-Expression in der Nebennierenrinde. Erste Hinweise darauf, dass die SPRED2-Defizienz auch Auswirkungen auf den kardiologischen Phänotyp haben könnte, ergaben sich bereits bei initialen Untersuchungen von Ullrich et al. So konnte bei den SPRED2-KO-Tieren neben hämodynamischer Auffälligkeiten eine gesteigerte Herz-Körpergewicht-Ratio festgestellt werden.
Die im Rahmen dieses Folgeprojekts durchgeführten Untersuchungen sollten die Frage klären, ob die Defizienz des SPRED2-Gens Auswirkungen auf die Herzleistung hat und hierüber die verkürzte Lebenserwartung der KO-Tiere verschulden könnte. Hierfür wurden zunächst Untersuchungen der elektrischen kardialen Aktivität mittels EKG und Elektrophysiologischer Untersuchung durchgeführt. Die Ermittlung von Herzrhythmusstörung und die Quantifizierung derselben spielte hierbei eine besondere Rolle. Des Weiteren sollte mit der Durchführung von PSR-Färbungen zur Bestimmung des kardialen Kollagengehaltes histologischen Fragestellungen Rechnung getragen werden.
Aufgrund des bereits aus den vorherigen Studien bekannten Hyperaldosteronismus der KO-Tiere stellte sich darüber hinaus die Frage, ob die im Rahmen der Studie feststellbaren kardiologischen Auffälligkeiten als Konsequenz der gesteigerten Aldosteronwerte, oder aber als direkte Folge des Genotyps gewertet werden müssen. Aus diesem Grund wurden alle oben genannten Untersuchungen mit Tieren, welche einer Behandlung mit dem Aldosteronantagonisten Eplerenon zugeführt worden waren, wiederholt.
Bei der Auswertung der basalen Ruhe- und Stress-EKGs zeigten sich einige Parameter bei den KO-Tieren pathologisch verändert. So war das QRS-Intervall, als Korrelat zur intraventrikulären Überleitungszeit, bei den KO-Mäusen verlängert, im Stress-EKG waren darüber hinaus sowohl die Dauer der P-Welle als auch des PQ-Intervalls erhöht. Durch die Behandlung mit Aldosteron waren diese Unterschiede zwischen WT- und KO-Gruppe teilweise nicht mehr feststellbar. Das die atrioventrikuläre Überleitungszeit abbildende PQ-Intervall war sowohl im Vergleich mit dem behandelten WT, als auch mit dem unbehandelten WT nicht mehr signifikant erhöht. Auch die Länge des QRS-Komplexes näherte sich unter Eplerenon-Behandlung dem der unbehandelten WT-Tiere an und sank bei der Stress-EKG-Auswertung sogar unterhalb des Signifkanzniveaus.
Bei der EKG-Analyse in Bezug auf Arrhythmien ergab sich bei Gegenüberstellung der basalen WT- und KO-Gruppe eine deutlich gesteigerte Vulnerabilität für Herzrhythmusstörungen bei den KO-Tieren. Durch die Behandlung mit Eplerenon konnte hierbei ein deutlicher Erfolg erzielt werden mit signifikanter Reduktion der Arrhythmieereignisse.
Die elektrophysiologische Untersuchung ergab neben unauffälligen Parametern der Funktion des Sinusknotens und der AV-Überleitung ebenfalls Hinweise für eine gesteigerte Empfindlichkeit für Arrhythmien. Die durch EPU induzierten Arrhythmien zeigten sich durch Eplerenon-Behandlung gleichermaßen rückgängig.
Mittels Kollagenfärbung konnte der initiale Verdacht, dass die SPRED2-KO-Tiere zu einer vermehrten kardialen Fibrosierung neigen, bestätigt werden. Dabei zeigte sich durch die Behandlung mit Eplerenon eine deutliche Beeinflussung und Reduktion des kardialen Kollagengehaltes.
Insgesamt lässt sich schlussfolgern, dass die mannigfaltigen phänotypischen Effekte, die die SPRED2-Defizienz bedingt, nur teilweise dem Hyperaldosteronismus der Tiere geschuldet sind und durch therapeutische Einflussnahme auf diesen auch nur partiell kompensiert werden können.
Atomic nanowires formed by self-assembled growth on semiconducting surfaces represent a feasible physical realization of quasi-1D electron systems and can be used to study fascinating 1D quantum phenomena. The system in the focus of this thesis, Si(553)-Au, is generated by Au adsorption onto a stepped silicon surface. It features two different chain types, interspersed with each other: A Au chain on the terrace, and a honeycomb chain of graphitic silicon located at the step edge. The silicon atoms at the exposed edges of the latter are predicted to be spin-polarized and charge-ordered [1], leading to an ordered array of local magnetic moments referred to as ``spin chains''.
The present thesis puts this spin chain proposal to an experimental test.
A detailed scanning tunneling microscopy (STM) and scanning tunneling spectroscopy (STS) scrutiny reveals a distinct unoccupied density of states (DOS) feature localized at every third Si step-edge atom, which aligns perfectly with the density functional theory (DFT) prediction.
This finding provides strong evidence for the formation of spin chains at the Si(553)-Au step edges, and simultaneously rules out the interpretation of previous studies which attributed the x3 step-edge superstructure to a Peierls instability.
To study the formation of spin chains in further detail, an additional member of the so-called Si(hhk)-Au family -- Si(775)-Au -- is analyzed.
Based on DFT modeling (performed by S.C. Erwin, Naval Research Laboratory, USA) and detailed STM and STS experiments, a new structure model for this surface is developed, and the absence of spin chains at the Si(775)-Au step edges is demonstrated.
The different step-edge charge distributions of all known Si(hhk)-Au surfaces are traced back to an electron transfer between the terrace and the step edge. Accordingly, an unintentional structure defect should create a localized spin at the Si(775)-Au step edge. This prediction is verified experimentally, and suggest that surface chemistry can be used to create and destroy Si spin chains.
Having clarified why spin chains form on some Si(hhk)-Au surfaces but not on others, various interaction effects of the Si(553)-Au spin chains are inspected.
A collaborative analysis by SPA-LEED (M. Horn-von Hoegen group, University of Duisburg-Essen, Germany), DFT (S.C. Erwin), and STM reveals strong lateral coupling between adjacent spin chains, bearing interesting implications for their magnetic ordering. The centered geometry uncovered leads to magnetic frustration, and may stabilize a 2D quantum spin liquid.
Moreover, a complex interplay between neighboring Au and Si chains is detected.
Specifically, the interaction is found effectively ``one-way'', i.e., the Si step edges respond to the Au chains but not vice versa.
This unidirectional effect breaks the parity of the Si chains, and creates two different configurations of step edges with opposite directionality.
In addition to the static properties of the Si(553)-Au surface mentioned above, the occurrence of solitons in both wire types is witnessed in real space by means of high-resolution STM imaging. The solitons are found to interact with one another such that both move in a coupled fashion along the chains. Likewise, STM experiments as a function of the tunneling current suggest an excitation of solitons along the step edge by the STM tunneling tip.
Solitons are also found to play an essential role in the temperature-dependent behavior of the Si(553)-Au step edges.
It is an accepted fact that the distinct x3 superstructure of the Si(553)-Au step edges vanishes upon heating to room temperature. As a first step in exploring this transition in detail over a large temperature range, a previously undetected, occupied electronic state associated with the localized step-edge spins is identified by means of angle-resolved photoemission spectroscopy (ARPES).
A tracking of this state as a function of temperature reveals an order-disorder-type transition. Complementary STM experiments attribute the origin of this transition to local, thermally activated spin site hops, which correspond to soliton-anitsoliton pairs.
Finally, a manipulation of the Si(553)-Au atomic wire array is achieved by the stepwise adsorption of potassium atoms. This does not only increase the filling of the Au-induced surface bands culminating in a metal-insulator transition (MIT), but also modifies the Si step-edge charge distribution, as indicated by STM and ARPES experiments.

Das Opc-Protein ist ein Außenmembranprotein von Meningokokken, das über extrazelluläre Matrixproteine mit Integrinen der Wirtszelle interagiert. Opc ist in Menschen immunogen und induziert bakterizide Antikörper. Das Opc-Protein wurde daher als aussichtsreicher Impfstoff-Kandidat angesehen, da es außerdem relativ gut konserviert ist. Allerdings wird das Opc-Protein nicht von allen Meningokokkenstämmen exprimiert. Einerseits fehlt das opc-Gen in einigen klonalen Komplexen (z.B. ST-8, ST-11, ST-53), andererseits ist die Opc-Expression nicht konstitutiv wegen einer phasenvariablen Transkription, die auf einem Poly-Cytidin-Bereich im Promotor des opc-Gens beruht.
In dieser Arbeit wurde die Präsenz des opc-Gens und die Opc-Expression in zwei großen Sammlungen deutscher Meningokokkenisolate von invasiven Erkrankungen (n=1141) und gesunden Trägern (n=792) untersucht.
Das opc-Gen war bei 71% der invasiven und 77% der Trägerstämme nachweisbar. Der größte Teil der opc-Gen negativen Stämme gehörte zu den klonalen Komplexen ST-8, ST-11, ST-213, ST-231, ST-334 und ST-53.
Der Anteil opc-positiver Stämme, die Opc in vitro exprimieren, war bei den invasiven Stämmen kleiner als bei den Trägerstämmen (13% vs. 29%, p<0,001, Chi-square-Test).
Der größere Anteil Opc-exprimierender Trägerstämme ist u.a. am ehesten mit der Überrepräsentation von wenig pathogenen klonalen Komplexen (ST-23, ST-35, ST-198) mit einer hohen Opc-Expressionsrate zu erklären.
24 von den 176 invasiven Stämmen mit einer Anzahl von 11 - 14 Cs in der Promotor-Region, die die Opc-Expression begünstigt, zeigten weder im ELISA noch im Westernblot eine Opc-Expression. Bei 14 dieser 24 Stämme wurde als Ursache ein phasenvariabler, intragenischer Poly-Adenin-Bereich identifiziert, der zu einer Leserasterverschiebung führte.
Die Vermutung mehrerer Autoren, dass die Opc-Expression mit dem klinischen Bild der Meningitis verknüpft ist, konnte mit der hier genutzten großen Stammsammlung nicht bestätigt werden. Invasive Stämme, die das Opc-Protein exprimierten, wurden genauso häufig von Patienten mit dem klinischen Bild der Meningitis isoliert wie Stämme, die das Opc-Protein nicht exprimierten (46% vs. 47%, Chi-square-Test: p<0,9). Allerdings gibt es eine starke Assoziation der Gegenwart des opc-Gens mit dem klinischen Merkmal Meningitis. Dieser Befund gibt Anlass zu der Hypothese, dass in vitro und in vivo Expression von Opc sich unterscheiden.
Zusammenfassend lässt sich festhalten, dass das Opc-Protein nur in 19,8% aller Isolate (invasive und Trägerstämme zusammengenommen) exprimiert wurde. Es zeigte sich eine Tendenz zu häufigerer Opc-Expression in apathogenen Trägerisolaten. Das Vorhandensein des opc-Gens, nicht aber die in vitro Expression konnten mit dem klinischen Merkmal Meningitis assoziiert werden. Zusätzlich wurde ein weiterer Mechanismus der intragenischen Phasenvariation beschrieben.
Cadherin-13 (CDH13) is an atypical member of the cadherin superfamily, a group of membrane proteins mediating calcium-dependent cellular adhesion. Although CDH13 shows the classical extracellular cadherin structure, the typical transmembrane and cytoplasmic domains are absent. Instead, CDH13 is attached to the cell membrane via a glycosylphosphatidylinositol (GPI) anchor. These findings and many studies from different fields suggest that CDH13 also plays a role as a cellular receptor. Interestingly, many genome-wide association studies (GWAS) have found CDH13 as a risk gene for attention-deficit/hyperactivity disorder (ADHD) and other neurodevelopmental disorders. In previous work from our research group, strong expression of Cdh13 mRNA in interneurons of the hippocampal stratum oriens (SO) was detected. Therefore, double-immunofluorescence studies were used to evaluate the degree of co-expression of CDH13 with seven markers of GABAergic interneuron subtypes. For this purpose, murine brains were double stained against CDH13 and the respective marker and the degree of colocalization in the SO of the hippocampus was assessed. Based on the result of this immunofluorescence study, quantitative differences in interneuron subtypes of the SO between Cdh13 knockout (ko), heterozygote (het) and wildtype (wt) mice were investigated in this dissertation using stereological methods. In addition, genotype- dependent differences in the expression of genes involved in GABAergic and glutamatergic neurotransmission were analyzed by quantitative real-time PCR (qRT-PCR). Primers targeting different GABA receptor subunits, vesicular GABA and glutamate transporter, GABA synthesizing enzymes and their interaction partners were used for this purpose.
The results of the stereological quantification of the interneuron subtypes show no significant differences in cell number, cell density or volume of the SO between Cdh13 ko, het and wt mice. On the other hand, qRT-PCR results indicate significant differences in the expression of tropomyosin-related kinase B gene (TrkB), which encodes the receptor of brain-derived neurotrophic factor (BDNF), a regulator of GABAergic neurons. This finding supports a role for CDH13 in the regulation of BDNF signaling in the hippocampus.
A complete simulation system is proposed that can be used as an educational tool by physicians in training basic skills of Minimally Invasive Vascular Interventions. In the first part, a surface model is developed to assemble arteries having a planar segmentation. It is based on Sweep Surfaces and can be extended to T- and Y-like bifurcations. A continuous force vector field is described, representing the interaction between the catheter and the surface. The computation time of the force field is almost unaffected when the resolution of the artery is increased.
The mechanical properties of arteries play an essential role in the study of the circulatory system dynamics, which has been becoming increasingly important in the treatment of cardiovascular diseases. In Virtual Reality Simulators, it is crucial to have a tissue model that responds in real time. In this work, the arteries are discretized by a two dimensional mesh and the nodes are connected by three kinds of linear springs. Three tissue layers (Intima, Media, Adventitia) are considered and, starting from the stretch-energy density, some of the elasticity tensor components are calculated. The physical model linearizes and homogenizes the material response, but it still contemplates the geometric nonlinearity. In general, if the arterial stretch varies by 1% or less, then the agreement between the linear and nonlinear models is trustworthy.
In the last part, the physical model of the wire proposed by Konings is improved. As a result, a simpler and more stable method is obtained to calculate the equilibrium configuration of the wire. In addition, a geometrical method is developed to perform relaxations. It is particularly useful when the wire is hindered in the physical method because of the boundary conditions. The physical and the geometrical methods are merged, resulting in efficient relaxations. Tests show that the shape of the virtual wire agrees with the experiment. The proposed algorithm allows real-time executions and the hardware to assemble the simulator has a low cost.
Marine sponge-associated actinomycetes are reservoirs of diverse natural products with novel biological activities. Their antibiotic potential has been well explored against a range of Gram positive and negative bacteria. However, not much is known about their anti-infective or anti-virulence potential against human pathogens. This Ph.D. project aimed to investigate the anti-infective (anti-Shiga toxin and anti-biofilm) potential of sponge-derived actinobacteria through identification and isolation of their bioactive metabolites produced and characterizing their mechanism of action by transcriptomics. This thesis is divided into three studies with the overall objective of exploring the anti-infective efficacy of actinomycetes-derived extracts and compound(s) that could possibly be used as future therapeutics.
The first study deals with investigation on the anti-Shiga toxin effects of sponge-associated actinomycetes. Diarrheal infections pose a huge burden in several developing and developed countries. Diarrheal outbreaks caused by Enterohemorrhagic Escherichia coli (EHEC) could lead to life-threatening complications like gastroenteritis and haemolytic uremic syndrome (HUS) if left untreated. Shiga toxin (Stx) produced by EHEC is a major virulence factor that negatively affects the human cells, leading them to death via apoptosis. Antibiotics are not prescribed against EHEC infections since they may enhance the risk of development of HUS by inducing the production and release of Stx from disintegrating bacteria and thereby, worsening the complications. Therefore, an effective drug that blocks the Stx production without affecting the growth needs to be urgently developed. In this study, the inhibitory effects of 194 extracts and several compounds originating from a collection of marine sponge-derived actinomycetes were evaluated against the Stx production in EHEC strain EDL933 with the aid of Ridascreen® Verotoxin ELISA assay kit. It was found that treatment with the extracts did not lead to significant reduction in Stx production. However, strepthonium A isolated from the culture of Streptomyces sp. SBT345 (previously cultivated from the Mediterranean sponge Agelas oroides) reduced the Stx production (at 80 μM concentration) in EHEC strain EDL933 without affecting the bacterial growth. The structure of strepthonium A was resolved by spectroscopic analyses including 1D and 2D-NMR, as well as ESI-HRMS and ESI-HRMS2 experiments. This demonstrated the possible application of strepthonium A in restraining EHEC infections.

In the second study, the effect of marine sponge-associated actinomycetes on biofilm formation of staphylococci was assessed. Medical devices such as contact lenses, metallic implants, catheters, pacemakers etc. are ideal ecological niches for formation of bacterial biofilms, which thereby lead to device-related infections. Bacteria in biofilms are multiple fold more tolerant to the host immune responses and conventional antibiotics, and hence are hard-to-treat. Here, the anti-biofilm potential of an organic extract derived from liquid fermentation of Streptomyces sp. SBT343 (previously cultivated from the Mediterranean sponge Petrosia ficiformis) was reported. Results obtained in vitro demonstrated its anti-biofilm (against staphylococci) and non-toxic nature (against mouse macrophage (J774.1), fibroblast (NIH/3T3) and human corneal epithelial cell lines). Interestingly, SBT343 extract could inhibit staphylococcal biofilm formation on polystyrene, glass and contact lens surfaces without affecting the bacterial growth. High Resolution Fourier Transform Mass Spectrometry (HR-MS) analysis indicated the complexity and the chemical diversity of components present in the extract. Preliminary physio-chemical characterization unmasked the heat stable and non-proteinaceous nature of the active component(s) in the extract. Finally, fractionation experiments revealed that the biological activity was due to synergistic effects of multiple components present in the extract.
In the third study, anti-biofilm screening of 50 organic extracts generated from solid and liquid fermentation of 25 different previously characterized sponge-derived actinomycetes was carried out. This led to identification of the anti-biofilm organic extract derived from the solid culture of Streptomyces sp. SBT348 (previously cultivated from the Mediterranean sponge Petrosia ficiformis). Bioassay-guided fractionation was employed to identify the active fraction Fr 7 in the SBT348 crude extract. Further purification with semi-preparative HPLC led to isolation of the bioactive SKC1, SKC2, SKC3, SKC4 and SKC5 sub-fractions. The most active sub-fraction SKC3 was found to be a pure compound having BIC90 and MIC values of 3.95 μg/ml and 31.25 μg/ml against S. epidermidis RP62A. SKC3 had no apparent toxicity in vitro on cell lines and in vivo on the greater wax moth Galleria melonella larvae. SKC3 was stable to heat and enzymatic treatments indicating its non-proteinaceous nature. HR-MS analysis revealed the mass of SKC3 to be 1258.3 Da. Structure elucidation of SKC3 with the aid of 1D and 2D-NMR data is currently under investigation. Further, to obtain insights into the mode of action of SKC3 on S. epidermidis RP62A, RNA sequencing was done. Transcriptome data revealed that SKC3 was recognized by RP62A at 20 min and SKC3 negatively interfered with the central metabolism of staphylococci at 3 h. Taken

together, these findings suggest that SKC3 could be a lead structure for development of new anti-staphylococcal drugs.
Overall, the results obtained from this work underscore the anti-infective attributes of actinomycetes consortia associated with marine sponges, and their applications in natural product drug discovery programs.
Ziel dieser Arbeit war es Veränderungen mitochondrialer Atmungskettenfunktionen von murinem Herz und muriner Niere unter einer inhalativen Anästhesie mit Sevofluran in Abhängigkeit von der inspiratorischen Sauerstoffkonzentration zu untersuchen.
In unserer in vivo Studie wurden männliche Black Six Mäuse 6 Stunden mit 1.0 MAC Sevofluran anästhesiert. Je nach Versuchsgruppe wurde mit einer inspiratorischen Sauerstoffkonzentration von 21%, 50% oder 100% ventiliert. Am Ende des Versuchsprotokolls wurden Herz und Nieren entnommen und direkt weiterverarbeitet. Es erfolgten photometrische Analysen und einer Blue Native Polyacrylamid Gel Elektrophorese zur Darstellung der mitochondrialen Komplexaktivitäten, der Proteincarbonylierung mitochondrialer Proteine und der Empfindlichkeit der isolierten Mitochondrien gegenüber der Ca2+ induzierten Schwellung.
Zusammenfassend zeigen unsere Ergebnisse, dass eine sechsstündige volatile Anästhesie zu einer deutlichen Änderung der mitochondrialen respiratorischen Komplexaktivitäten, sowohl in der Niere als auch im Herzen führt. Insbesondere wies Komplex I eine erhöhte enzymatische Aktivität auf. Die Veränderungen der mitochondrialen Respiration waren größtenteils unabhängig von der inspiratorischen Sauerstoffkonzentration und führten zu keiner mitochondrialen Schädigung. Sowohl die Superoxid Produktion als auch die Proteincarbonylierung blieben unverändert. Die mitochondriale Empfindlichkeit gegenüber der Ca2+ induzierten Schwellung zeigte keine Veränderung gegenüber den Kontrolltieren. Somit ergaben sich keine toxischen Auswirkungen hoher Sauerstoffkonzentrationen im Sinne eines erhöhten oxidativen Schadens des mitochondrialen Proteoms oder einer Membranschädigung.
The goal of this work is to improve the understanding of adsorption-induced deformation in nanoporous (and in particular microporous) materials in order to explore its potential for material characterization and provide guidelines for related technical applications such as adsorption-driven actuation. For this purpose this work combines in-situ dilatometry measurements with in-depth modeling of the obtained adsorption-induced strains. A major advantage with respect to previous studies is the combination of the dilatometric setup and a commercial sorption instrument resulting in high quality adsorption and strain isotherms. The considered model materials are (activated and thermally annealed) carbon xerogels, a sintered silica aerogel, a sintered hierarchical structured porous silica and binderless zeolites of type LTA and FAU; this selection covers micro-, meso- and macroporous as well as ordered and disordered model materials.
All sample materials were characterized by scanning electron microscopy, gas adsorption and sound velocity measurements. In-situ dilatometry measurements on mesoporous model materials were performed for the adsorption of N2 at 77 K, while microporous model materials were also investigated for CO2 adsorption at 273 K, Ar adsorption at 77 K and H2O adsorption at 298 K. Within this work the available in-situ dilatometry setup was revised to improve resolution and reproducibility of measurements of small strains at low relative pressures, which are of particular relevance for microporous materials.
The obtained experimental adsorption and strain isotherms of the hierarchical structured porous silica and a micro-macroporous carbon xerogel were quantitatively analyzed based on the adsorption stress model; this approach, originally proposed by Ravikovitch and Neimark, was extended for anisotropic pore geometries within this work. While the adsorption in silica mesopores could be well described by the classical and analytical theory of Derjaguin, Broekhoff and de Boer, the adsorption in carbon micropores required for comprehensive nonlocal density functional theory calculations. To connect adsorption-induced stresses and strains, furthermore mechanical models for the respective model materials were derived. The resulting theoretical framework of adsorption, adsorption stress and mechanical model was applied to the experimental data yielding structural and mechanical information about the model materials investigated, i.e., pore size or pore size distribution, respectively, and mechanical moduli of the porous matrix and the nonporous solid skeleton. The derived structural and mechanical properties of the model materials were found to be consistent with independent measurements and/or literature values. Noteworthy, the proposed extension of the adsorption stress model proved to be crucial for the correct description of the experimental data.
Furthermore, it could be shown that the adsorption-induced deformation of disordered mesoporous aero-/xerogel structures follows qualitatively the same mechanisms obtained for the ordered hierarchical structured porous silica. However, respective quantitative modeling proved to be challenging due to the ill-shaped pore geometry of aero-/xerogels; good agreement between model and experiment could only be achieved for the filled pore regime of the adsorption isotherm and the relative pressure range of monolayer formation. In the intermediate regime of multilayer formation a more complex model than the one proposed here is required to correctly describe stress related to the curved adsorbate-adsorptive interface. Notably, for micro-mesoporous carbon xerogels it could be shown that micro- and mesopore related strain mechanisms superimpose one another.
The strain isotherms of the zeolites were only qualitatively evaluated. The result for the FAU type zeolite is in good agreement with other experiments reported in literature and the theoretical understanding derived from the adsorption stress model. On the contrary, the strain isotherm of the LTA type zeolite is rather exceptional as it shows monotonic expansion over the whole relative pressure range. Qualitatively this type of strain isotherm can also be explained by the adsorption stress model, but a respective quantitative analysis is beyond the scope of this work.
In summary, the analysis of the model materials' adsorption-induced strains proved to be a suitable tool to obtain information on their structural and mechanical properties including the stiffness of the nonporous solid skeleton. Investigations on the carbon xerogels modified by activation and thermal annealing revealed that adsorption-induced deformation is particularly suited to analyze even small changes of carbon micropore structures.
Seit Jahren wird trotz der stetigen Modernisierung der medizinischen Versorgungsleistungen ein Anstieg der stationären Einweisungen in den psychiatrischen Kliniken registriert. Dabei nutzt eine kleine Gruppe von Patienten, die sogenannten Heavy User, den Großteil der zur Verfügung stehenden therapeutischen Ressourcen. Diese explorative Studie beschreibt eine Gruppe von erwachsenen, weiblichen Heavy Usern (n=23) mit restriktiver bzw. bulimischer Anorexie oder Bulimie, die mindestens drei stationäre Aufnahmen in einer psychiatrischen Klinik aufwiesen. Als Vergleich dient eine Kontrollgruppe von weiblichen Nicht-Heavy Usern (n=13) mit maximal einem stationären Voraufenthalt.
Die Ergebnisse zeigen, dass Heavy User mit der Hauptdiagnose einer Essstörung spezifische soziodemographische, diagnostische und psychopathologische als auch Persönlichkeitsmerkmale aufweisen, die eine rechtzeitige Identifizierung dieser Patientengruppe ermöglichen könnten. Der Heavy Use wird als multifaktorielles Geschehen verdeutlicht und die Notwendigkeit einer frühen Identifikation und Intervention betont. Da die bisherigen Studienergebnisse sehr uneinheitliche Resultate lieferten, sind weitere Untersuchungen dieser Patientengruppe unerlässlich. Die Entwicklung alternativer und individueller Therapieansätze ist angezeigt, um passende Versorgungsangebote für diese therapieresistenten Patienten zu schaffen.
