In multizellulären Organismen ist die Apoptose, eine Art des programmierten Zelltods, ein hoch spezifischer, natürlicher Prozess um unerwünschte, überschüssige oder beschädigte Zellen auf einem geordneten, nicht entzündlichen, Weg zu beseitigen. Auch im menschlichen Körper werden täglich Milliarden an Zellen durch Apoptose abgebaut. Dadurch kann der Organismus die Zellzahl und Gewebegröße regulieren. Eine Fehlregulation der Apoptose kann weitreichende Konsequenzen haben. Während es bei einer unzureichenden Apoptose zu Autoimmunität und Tumoren kommen kann, führt ein gesteigerter Zelltod zu Immunschwäche oder akuten und chronischen degenerativen Erkrankungen. Aus diesem Grund ist die Erforschung der genauen Regulation des programmierten Zelltods von großer Bedeutung. Die Mitochondrien spielen eine Schlüsselrolle bei der Regulation des programmierten Zelltods. Während des Ablaufs der Apoptose kommt es zur Freisetzung von Mediatoren der Apoptose aus diesen Organellen. Diese Moleküle sind dann an der Aktivierung von Caspasen beteiligt, welche die Degradation von Proteinen und als Folge davon der DNA bewerkstelligen. Die Proteine der Bcl-2 Familie, zu der pro- und anti-apoptotische Mitglieder gehören, kontrollieren die Integrität der Mitochondrien hauptsächlich durch Protein-Protein und Protein-Membran Interaktionen. Viele anti-apoptotische Mitglieder der Bcl-2 Familie sind mittels einer C-terminalen Transmembrandomäne in der Lage an die äußere Mitochondrienmembran zu binden. A1, ein anti-apoptotisches Mitglied dieser Proteinfamilie, besitzt allerdings keine typische Transmembrandomäne. Die Funktion und Lokalisation des Proteins werden sehr kontrovers diskutiert. Daher ist das Ziel dieser Arbeit, die Lokalisation und Funktion von A1 zu analysieren. Um ein umfassendes Bild zu bekommen, wandten wir gentechnische Methoden zur Modifizierung des A1 C-Terminus an. Die intrazelluläre Lokalisation des Proteins wurde mittels konfokaler Mikroskopie untersucht. Die Bedeutung des proteasomalen Abbaus von A1 wurde schließlich durch Inhibitionsexperimente charakterisiert. Durch eine Fusion des C-terminalen Endes von A1 mit dem „enhanced green fluorescent protein“ haben wir die Funktion und Lokalisierungseigenschaften des A1 C-terminus mittels Durchflusszytometrie und konfokaler Mikroskopie untersucht. Wir konnten zeigen, dass das C-terminale Ende das so entstandene chimäre Protein deutlich destabilisieren konnte. Eine Blockade des proteasomalen Abbaus führte zu einer Stabilisierung des Fusionsproteins. Des Weiteren konnte in der konfokalen Mikroskopie eine diffuse Verteilung des chimären Proteins in 293T Zellen nachgewiesen werden. Diese Ergebnisse weisen darauf hin, dass das C-terminale Ende von A1 keine Lokalisation an spezifische intrazelluläre Organellen vermitteln kann, jedoch für die Instabilität und den proteasomalen Abbau des Proteins verantwortlich ist. Eine Analyse der Lokalisation des Gesamtproteins A1 in 293T Zellen mittels konfokaler Mikroskopie konnte eine Verteilung von A1 zugunsten des Zytoplasmas mit Anreicherung an den Mitochondrien nachweisen. Obwohl das C-terminale Ende von A1 für sich keine spezifische intrazelluläre Lokalisation vermittelt, zeigte das Protein eine Kolokalisation an den Mitochondrien. Somit scheinen noch andere N-terminale Bereiche des Proteins an der Lokalisation von A1 beteiligt zu sein. Zusammenfassend konnte mit dieser Arbeit gezeigt werden, dass der C-Terminus von A1 eine wichtige Rolle für die Stabilität des anti-apoptotischen Bcl-2 Familienmitglieds spielt.
Aging is known to be a risk factor for structural abnormalities and functional decline in the nervous system. Characterizing age-related changes is important to identify putative pathways to overcome deleterious effects and improve life quality for the elderly. In this study, the peripheral nervous system of 24-month-old aged C57BL/6 mice has been investigated and compared to 12-month-old adult mice. Aged mice showed pathological alterations in their peripheral nerves similar to nerve biopsies from elderly human individuals, with nerve fibers showing demyelination and axonal damage. Such changes were lacking in nerves of adult 12-month-old mice and adult, non-aged humans. Moreover, neuromuscular junctions of 24-month-old mice showed increased denervation compared to adult mice. These alterations were accompanied by elevated numbers of macrophages in the peripheral nerves of aged mice. The neuroinflammatory conditions were associated with impaired myelin integrity and with a decline of nerve conduction properties and muscle strength in aged mice.
To determine the pathological impact of macrophages in the aging mice, macrophage depletion was performed in mice by oral administration of CSF-1R specific kinase (c-FMS) inhibitor PLX5622 (300 mg/kg body weight), which reduced the number of macrophages in the peripheral nerves by 70%. The treated mice showed attenuated demyelination, less muscle denervation and preserved muscle strength. This indicates that macrophage-driven inflammation in the peripheral nerves is partially responsible for the age-related neuropathy in mice.
Based on previous observations that systemic inflammation can accelerate disease progression in mouse models of neurodegenerative diseases, it was hypothesized that systemic inflammation can exacerbate the peripheral neuropathy found in aged mice. To investigate this hypothesis, aged C57BL/6 mice were intraperitoneally injected with a single dose of lipopolysaccharide (LPS; 500 μg/kg body weight) to induce systemic inflammation by mimicking bacterial infection, mostly via activation of Toll-like receptors (TLRs). Altered endoneurial macrophage activation, highlighted by Trem2 downregulation, was found in LPS injected aged mice one month after injection. This was accompanied by a so far rarely observed form of axonal perturbation, i.e., the occurrence of “dark axons” characterized by a damaged cytoskeleton and an increased overall electron density of the axoplasm. At the same time, however, LPS injection reduced demyelination and muscle denervation in aged mice. Interestingly, TREM2 deficiency in aged mice led to similar changes to LPS injection. This suggests that LPS injection likely mitigates aging-related demyelination and muscle denervation via Trem2 downregulation.
Taken together, this study reveals the role of macrophage-driven inflammation as a pathogenic mediator in age-related peripheral neuropathy, and that targeting macrophages might be an option to mitigate peripheral neuropathies in aging individuals. Furthermore, this study shows that systemic inflammation may be an ambivalent modifier of age-related nerve damage, leading to a distinct type of axonal perturbation, but in addition to functionally counteracting, dampened demyelination and muscle denervation. Translationally, it is plausible to assume that tipping the balance of macrophage polarization to one direction or the other may determine the functional outcome in the aging peripheral nervous system of the elderly.
Charcot-Marie-Tooth (CMT) Neuropathien stellen als häufigste erblich bedingte neurologische Erkrankungen eine Gruppe genetisch heterogener, chronisch progredienter peripherer Polyneuropathien dar. Die Lebensqualität der Patienten ist bei fehlender kurativer Therapieoption vor allem durch motorische und sensorische Defizite deutlich eingeschränkt. In verschiedenen Studien konnte die pathophysiologische Relevanz einer sekundären Entzündungsreaktion, insbesondere durch Makrophagen und Lymphozyten vermittelt, in Mausmodellen dreier CMT1 Subtypen (CMT1A, CMT1B, CMT1X) aufgezeigt werden. Auch in Folge einer Läsion peripherer Nerven ist eine akute Entzündungsreaktion von entscheidender Bedeutung, wobei sich bereits Gemeinsamkeiten zwischen der postläsionalen Waller´schen Degeneration (WD) und CMT1 Neuropathien identifizieren ließen. Während die aktive Beteiligung der Autophagie Schwann´scher Zellen (hier kurz SZ Autophagie genannt) an der Myelindegradation im Falle einer WD jedoch vielfach beschrieben wurde, ist Ähnliches in CMT1 Neuropathien bisher nur unzureichend untersucht. Da in einer Studie in Cx32def Mausmodellen der CMT1X Erkrankung auch nach Reduktion endoneuraler Makrophagen anhaltende Demyelinisierung beobachtet werden konnte, sollte das Vorkommen von SZ Autophagie sowie deren mögliche Beeinflussung durch Makrophagen in diesen Myelinmutanten untersucht werden.
In der vorliegenden Arbeit wurden sowohl Wildtyp (Wt) Mäuse in ex vivo und in vivo Modellen einer WD als auch Cx32def Myelinmutanten zweier Altersstufen (4 und 12 Monate) mit einem niedermolekularen CSF1-Rezeptor-Inhibitor (CSF1RI) zur Reduktion endoneuraler Makrophagen behandelt, wobei sich vergleichende histochemische bzw. immunhistochemische Analysen peripherer Nerven behandelter und unbehandelter Tiere anschlossen.
Im Rahmen der Etablierung immunhistochemischer Methodik zeigte sich hierbei unter den kontrollierten Bedingungen einer ex vivo Ischiasnervenkultur eine vermehrte Aktivierung der SZ Autophagie in behandelten Wt Mäusen. Auch 4 Monate alte behandelte Cx32def Tiere wiesen, verglichen mit unbehandelten Myelinmutanten bzw. Wt Mäusen derselben Altersstufe, eine vermehrte autophagische Aktivität in SZ auf. Diese scheint sich jedoch im weiteren Verlauf der Erkrankung zu reduzieren, da im Falle der 12 Monate alten Cx32def Modelltiere weniger autophagisch aktive SZ Profile bzw. kaum Unterschiede zwischen behandelten und unbehandelten Tieren beobachtet werden konnten.
Die Ergebnisse lassen somit eine mögliche aktive Beteiligung von SZ Autophagie insbesondere in der Pathophysiologie der frühen Phase einer CMT1X Erkrankung sowie deren Beeinflussung durch endoneurale Makrophagen vermuten. Dies sollte vornehmlich in der Entwicklung von Therapiestrategien der CMT1X bedacht werden, da sich eine frühe Reduktion pathophysiologisch relevanter endoneuraler Makrophagen somit auch nachteilig auf die Myelinintegrität auswirken könnte.
The prevalence of cardiovascular diseases (CVD) increases dramatically with age. Nevertheless, most of the basic research in cardiology has been conducted on young healthy animals which may not necessarily reflect the situation observed in the clinic. The heart undergoes profound changes in elderly, including molecular alterations, myocardial hypertrophy, interstitial fibrosis and functional decline. To date, numerous approaches exist to explain mechanisms of the cardiac aging process whereupon inflammation and immune activity are of increasing interest. Myocardial aging is temporally associated with chronic low-grade systemic inflammation and accumulation of memory T-cells. However, a possible causal relationship between these two phenomena has not yet been investigated. Thus, aim of the present study was to assess how immunological mechanisms contribute to the myocardial aging process.
Herein, the healthy murine heart was found to harbor all major resident leukocyte populations, including macrophages (CD45+CD11b+Ly6G-), granulocytes (CD45+ CD11b+Ly6G+), T-cells (CD45+CD11b-CD3e+), B-cells (CD45+CD11b-B220+) at frequencies that largely surpass those found in skeletal muscles. Age-related structural alterations and functional impairment occur simultaneously with significant shifts of the tissue resident leukocyte composition. Gene expression analyses performed on bulk myocardial samples revealed higher expression levels of TNF and INF- suggesting that in situ inflammation plays a role in the myocardial aging process. Aging was furthermore accompanied by a significant increase in size and cellularity of mediastinal, heart draining lymph nodes (med LN). Moreover, the med LNs harvested from aged mice showed a strong accumulation of effector-memory T-cells (CD44+CD62L-), mainly exhibiting a pro-inflammatory phenotype (Foxp3-, TNF+, IFN- γ+). None of these alterations were observed in popliteal lymph nodes of aged mice, indicating that they might be site-specific.
Next, to go beyond mere associative evidence and examine underlying mechanisms, the myocardial aging process was comprehensively characterized in mice lacking B- (µMT) or CD4+ T-cells (CD4ko). Our analyses revealed that aged CD4+ T-cell-deficient, but not B-cell-deficient mice, exhibit a lower in situ inflammatory tone and preserved ventricular function, as compared to age-matched wild type controls. No differences in the expression levels of genes related to fibrosis were observed in the groups.
Taken together, the results of this study indicate that heart-directed immune responses may spontaneously arise in the elderly, even in the absence of a clear tissue damage or concomitant infection. The T-cell-mediated immunosenescence profile might be particularly associated with age-related myocardial inflammation and functional decline, but not with tissue remodeling. These observations might shed new light on the emerging role of T cells in myocardial diseases, which primarily affect the elderly population.
Hintergrund: Die Juvenile Idiopathische Arthritis (JIA) ist eine heterogene Erkrankung mit unbekannter Ätiologie der Arthritis bei Patientinnen und Patienten unter 16 Jahren für mehr als sechs Wochen. Das Humane Zytomegalievirus (HCMV) ist ein lymphotropes Betaherpesvirus, das im menschlichen Körper persistiert und eine anhaltende Stimulation des Effektor-T-Zell-Systems bewirkt. Sowohl für JIA als auch für HCMV zeigt sich eine vorzeitige Immunoseneszenz.
Ziel: Untersuchung des potenziellen Einflusses von HCMV auf das vorzeitig gealterte Immunsystem von JIA-Patientinnen und Patienten.
Methoden: T-Zell-Phänotyp, intrazelluläre Zytokinproduktion und die Expression von Chemokinrezeptoren wurden mittels Durchflusszytometrie (FACS) gemessen. Der HCMV-Serostatus wurde mittels Enzyme-Linked Immunosorbent Assay (ELISA) gemessen. Phänotyp und Zytokinproduktion von Lymphozyten von JIA-Patientinnen und Patienten und Gesundkontrollen wurden hinsichtlich ihres HCMV-Serostatus verglichen.
Ergebnisse: Sowohl JIA-Patientinnen und Patienten als auch Gesundkontrollen zeigten einen Zusammenhang zwischen HCMV-Seropositivität und Immunseneszenz, nämlich geringere Anteile naiver T-Zellen und relativ höhere Anteile an differenzierten T-Zellen. Bei den JIA-Patientinnen und Patienten war die HCMV-Seropositivität mit einer höheren intrazellulären IFNγ-Produktion verbunden. T-Zellen bei JIA-Patientinnen und Patienten zeigten eine höhere CCR5-Expression in Verbindung mit HCMV-Seropositivität. Diese Assoziation wurde bei Gesundkontrollen nicht beobachtet.
Schlussfolgerung: Der T-Zell-Phänotyp war bei JIA-Patientinnen und Patienten und Gesundkontrollen ähnlich mit HCMV assoziiert. Im Gegensatz dazu zeigten JIA-Patientinnen und Patienten Hinweise auf eine TH1-Dominanz in Verbindung mit HCMV-Seropositivität. Bei CCR5 ist dieser Effekt bei JIA-Patientinnen und Patienten signifikant stärker als bei Gesundkontrollen. Die vorliegende Studie legt nahe, dass HCMV-assoziierte Veränderungen der T-Zell-Differenzierung bei JIA-Patientinnen und Patienten bestätigt werden können.

Multiple Sklerose ist eine der häufigsten neurologischen Erkrankungen, die zu motorischen, sensiblen und vegetativen Einschränkungen führt. Häufig beginnt die Erkrankung mit einem schubförmigen Verlauf, dem eine progrediente Verschlechterung folgt. Trotzdem leiden ca. 15% der Patienten bereits von Beginn an, an einer primär progressiven Variante der MS, die bereits mit der progredienten Phase beginnt. Bis jetzt ist die Pathophysiologie nicht vollständig verstanden. Lange Zeit dachte man, dass MS primär eine reine Autoimmun-Erkrankung darstellt, aber in den letzten Jahren ergab sich die Frage, ob es vor allem in den progressiven Formen, eine zytodegenerative Komponente gibt, auf die eine sekundäre Inflammation folgt. Eine mögliche Ursache stellen hier Mutationen des PLP 1 Gens dar, die normalerweise mit leukodystrophischen Erkrankungen assoziiert sind. Es gab zwei Fallberichte, in denen von Patienten berichtet wurde, die unterschiedliche Mutationen in diesem Gen hatten und den klinischen Phänotyp einer MS zeigten. Diese Arbeit sollte nun die Auswirkungen der Mutationen, beziehungsweise einer Nullmutation des PLP 1 Gens in 18- und zum Teil 12-Monate alten Mausmutanten untersuchen. Hier konnten Myelin-Veränderungen und axonaler Schaden in immunhistochemischen Untersuchungen sowie mittels Elektronenmikroskopie und optischer Kohärenztomographie gezeigt werden. Weiter konnte eine Neuroinflammation und damit einhergehend eine zunehmende Anzahl CD8+ T-Zellen sowie einer erhöhten Anzahl an Mikroglia/Makrophagen gefunden werden. Dies ging mit vergleichsweise reduzierten Leistungen der Mutanten bei der motorischen Rotarod-Analyse einher. Interessanterweise wurde weniger neuraler Schaden in den heterozygoten Varianten gefunden, obwohl das Ausmaß der Entzündung gleichblieb. Dies könnte für eine zielgerichtete immunvermittelte Schädigung der Oligodendrozyten sprechen, die zu neuronalem Schaden führt. So konnte gezeigt werden, dass es durch Punktmutationen in einem Myelinprotein-codierendem Gen zu einer sekundären Entzündung kommen kann, die mit dem klinischen Phänotyp einer progressiven MS einhergeht. Weiter sind diese Mausmodelle ein Beispiel für eine genetische Erkrankung des ZNS, in denen die Klinik maßgeblich durch die begleitende Inflammation und nicht allein durch den genetischen Schaden verursacht wird.
Leishmanien besitzen eine Vielzahl von Mechanismen, die humorale und zelluläre Immunabwehr effektiv zu unterlaufen. Diese hängen eng mit der Expression von hauptsächlich zwei Glykokonjugaten auf der Parasitenoberfläche zusammen, dem gp63 und dem Lipophosphoglykan. Die Parasiten sind einerseits schlechte Aktivatoren des alternativen Komplementweges und umgehen damit ihre eigene extrazelluläre Lyse. Oberflächengebundene Komplementfaktoren fördern andererseits die Aufnahme der Leishmanien durch Makrophagen. Solange diese nicht durch T-Zellen aktiviert sind, dienen sie den Parasiten als "Refugium". Dies gilt insbesondere, als Leishmanien in der Lage sind, 1. den "oxidative burst" zu hemmen; 2. toxische Sauerstoffmetaboliten zu entgiften; 3. abbauende lysosomale Enzyme zu hemmen und 4. das saure Milieu in den Lysosomen für ihren eigenen Metabolismus auszunutzen. Schließlich unterlaufen Leishmanien die zelluläre Immunabwehr des Wirts, indem sie die Aktivierung von T-Lymphozyten hemmen und die Expansion von T-Zell-Sub-populationen bewirken, die für ihr eigenes Überleben nützlich sind.
The bidirectional influence of parenchymal cells and cells of the immune system, especially of antigen-presenting and CD8\(^+\) T cells, in situations of putative auto- immune pathogenicity and degeneration was the main topic of this thesis. In the first part, the influence of human muscle cells on antigen-presenting cells was investigated. In inflammatory myopathies prominent infiltrates of immune cells containing T cells and antigen-presenting cells like macrophages and dendritic cells are present. The hypothesis was that human myoblasts have an inhibiting influence on these antigen-presenting cells under homeostatic conditions. A dysfunction or impairment under inflammatory circumstances might contribute to the development of myopathic conditions. The surface analysis of dendritic cells cocultured with myoblasts showed that immature dendritic cells could be driven into a reversible semi- mature state with significantly elevated levels of CD80. These dendritic cells were additionally characterized by their inhibiting function on T-cell proliferation. It was also shown that the lysates of healthy myoblasts could strongly enhance the phagocytic ability of macrophages, which could help with muscle regeneration and which might be disturbed in myositis patients. The second part of this thesis was about the clonal specificity of CD8\(^+\) T cells in a mouse model with genetically induced over-expression of PLP in oligodendrocytes. Here, we could show that the cytotoxic T lymphocytes, which had previously been shown to be pathogenic, were clonally expanded in the CNS of the transgenic mice. The amino acid sequences of the corresponding receptor chains were not identical, yet showed some similarities, which could mean that these clones recognize similar antigens (or epitopes of the same antigen). The knockout of PD-1 in this setting allowed for an analysis of the importance of tissue immune regulation. It became evident that the absence of PD-1 induced a larger number of clonal expansions in the CNS, hinting towards a reduced threshold for clonal disturbance and activation in these T cells. The expansions were, however, not pathogenic by themselves. Only in the presence of tissue damage and an antigenic stimulus (in our case the overexpression of PLP), the PD-1 limitation exacerbated the immune pathogenicity. Therefore, only in the presence of a “tissue damage signal”, the dyshomeostasis of T cells lacking PD-1 achieved high pathogenetic relevance. Finally, we investigated the pathogenetic role of CD8 T cells in Rasmussen encephalitis, a rare and chronic neurological disease mainly affecting children. The analysis of the T-cell receptor repertoire in Rasmussen encephalitis patients in the peripheral CD4\(^+\) and CD8\(^+\) T-cell compartments as well as the brain revealed the involvement of T cells in the pathogenicity of this disease. Many clonal expansions in the brain matched CD8\(^+\) T-cell expansions in the periphery on the sequence level. These putatively pathogenic clones could be visualized by immunohistochemistry in the brain and were found in close proximity to astrocytes and neurons. Additionally, the expanded clones could be found in the periphery of patients for at least one year.
Charcot-Marie-Tooth Neuropathien sind die häufigsten hereditären Erkrankungen des peripheren Nervensystems und dennoch bis heute nicht therapierbar. Die Lebensqualität der Patienten ist durch motorische und sensorische Defizite der Extremitäten häufig stark eingeschränkt. Ursache können unter anderem Mutationen in Schwann-Zellen sein, die zu dem typischen Bild von Demyelinisierung und axonalem Schaden führen. In den letzten Jahren konnte in Mausmodellen das Immunsystem als wichtiger Mediator in der Pathogenese der CMT 1 Subtypen A, B und X identifiziert werden. Insbesondere Makrophagen spielen eine tragende Rolle bei dem Verlust der axonalen Integrität, bei der Schädigung der Myelinscheiden, sowie bei der Dedifferenzierung von Schwann-Zellen. Entscheidender Faktor für Proliferation und Aktivierung der Makrophagen ist hierbei das Zytokin CSF-1, dessen korrespondierender Rezeptor auf Makrophagen exprimiert wird. Der CSF-1/CSF1R Signalweg bietet somit einen vielversprechenden Angriffspunkt.
In der vorliegenden Arbeit wurden Mausmodelle der CMT 1 Subtypen A, B und X mit einem niedermolekularen CSF-1-Rezeptor Inhibitor behandelt. Anschließend erfolgte eine funktionelle und strukturelle Auswertung der peripheren Nerven.
Das beste Ansprechen auf die Therapie zeigten Cx32def Mutanten. Strukturell fielen ein verringerter axonaler Schaden und eine verbesserte axonale Regenerationsfähigkeit sowie erhaltene neuromuskuläre Synapsen auf. Funktionell äußerte sich dies in verbesserten elektrophysiologischen Parametern und einem Krafterhalt, welcher als klinischer Parameter die größte Relevanz für betroffene Patienten hat und somit besonders hervorzuheben ist.
Auch P0het Mutanten zeigten Verbesserungen nach der CSF1RI Behandlung. Anders als bei Cx32def Tieren zeigte sich hier jedoch vor allem ein Erhalt der Myelinintegrität. Weiterhin wirkte sich die Therapie positiv auf elektrophysiologische Parameter und Krafttests aus. Vor allem besonders stark betroffene Individuen schienen hierbei von der CSF1RI Behandlung zu profitieren.
Bei PMP22tg Mutanten hingegen konnten keine positiven Effekte der CSF1RI Behandlung nachgewiesen werden. Strukturelle und funktionelle Parameter behandelter Tiere unterschieden sich nicht von unbehandelten.
Diese Ergebnisse unterstreichen die Relevanz der sekundären Entzündungsreaktion in CMT 1 Neuropathien als wichtigen Mediator in der Pathogenese. Weiterhin konnte gezeigt werden, dass eine Intervention im CSF-1/CSF1R Signalweg einen vielversprechenden möglichen Ansatz für die Therapie der bisher nicht behandelbaren CMT 1 Subypen X und B darstellt. Unausweichlich ist hierbei ein möglichst früher Therapiestart vor Ausprägung der ersten molekularen und histologischen Veränderungen. Im Hinblick auf die nicht die Lebenserwartung reduzierende Erkrankung muss ferner eine Minimierung der Nebenwirkungen der Therapie gewährleistet sein. Besonders hervorzuheben ist hier die Verwendung eines Inhibitors, welcher nicht in das zentrale Nervensystem vordringen kann und somit die Funktion der Mikroglia nicht beeinträchtigt.
Dendritic cell-based vaccination is a well established technique for preventive and therapeutic instruction of the immune system where conservative vaccine formulations fail to cure or prevent diseases, respectively. Efficiency of this technique already was demonstrated in infectious diseases as well as for cancer in animal or human studies. Well controlled manipulation and antigen-loading of immature DC is most beneficial to this technique. But, time-consuming and cost-extensive procedures for preparation of DC precursors, expansion and stimulation of DC and inpatient administration are big disadvantages regarding vaccine development for pandemic infectious diseases that occur mainly in underdeveloped countries. Therefore vaccines are needed that are pathogen-tailored and able to induce equal immune responses as their DC-based vaccine models. For vaccination against Leishmania parasites such a DC-based vaccine is feasible and its efficacy to induce protective Th1-based immune responses was already demonstrated in several animal studies. But, one of our own studies indicated supportive activity of host cells exceeding the allocation of T cells to become activated by transferred DC. IL-12, an important cytokine for the induction of Th1-related immune responses, has to be produced by host cells. Therefore, the aim of this study was to investigate the mechanism of BMDC-based vaccination with regard to simplification of the vaccine formulation. Key questions that have been addressed are: Which cells process the information that is transferred by the injected DC and what are the key components of this information? Further more, it was looked at whether altered vaccine formulations are able to induce protective immunity and whether they share equal molecular mechanisms. The current paradigm of BMDC-based vaccination proposes direct interaction of transferred BMDC with host T cells. These BMDC have to be antigen-loaded for stimulation via antigen-peptide-MHC molecule-complexes and they have to be activated for proper co-stimulation of T cells. Here, this study demonstrates that neither activation for co-stimulation nor direct interaction with adequate MHC molecules is needed for the induction of protective immunity against infection with Leishmania-parasites. Disrupted antigen-loaded BMDC are able to induce protective immunity in BALB/c mice without pre-stimulation via CpG ODN. Beyond, if BMDC were used with a different MHC-background than recipient mice then the vaccine still would be efficient in terms of reduction of footpad swelling and parasite load in draining lymph nodes. Even more, DC-specific features are no key component that leads to protective immunity as vaccination with disrupted antigen-loaded MΦ shows equal properties than before mentioned vaccine formulations. Further more, it was found that host DC play a major role in transforming the incoming signal, received from transferred antigen-loaded DC, into Th1-related stimuli and Leishmania-antigen-specific T cell activation. Suspensions of disrupted antigen-loaded DC resemble a combination of laid off soluble molecules together with exosome-like vesicles that formed after disruption of membranes. Here it was shown that separation of the membranous and soluble fractions and subsequent transfer into BALB/c mice will lead to protection of these mice against infection with L. major promastigotes only if the membranous fraction is used as vaccine. More, this vaccine formulation takes advantage of easy storage at -80°C with no need of fresh production. This clearly demonstrates that the immunity-inducing principle of disrupted DC-based vaccination lies within the membrane enclosed fraction. On a molecular level, disrupted antigen-loaded DC induce Th1-related cytokines during vaccination and as response on pathogen encounter. In vivo assays revealed IL-12 production and antigen-specific T cell proliferation among splenocytes that were stimulated with disrupted antigen-loaded DC. Splenocytes of accordingly vaccinated mice produce tremendous amounts of IFNγ after stimulation with Leishmania parasites. In summary, disrupted antigen-loaded BMDC fulfil all characteristics of DC-based vaccination against Leishmania major. But, while purification of membranes of antigen-loaded DC and subsequent transfer to BALB/c mice leads to control of the disease in the animal model, only slight levels of Th1-related cytokines are seen in the in vivo assays. Whether this points towards a loss of vaccine activity on unseen levels or unknown sites where Th1-related immunity is induced by both, complete solution and purified membranes, still has to be determined.
Präeklampsie ist einer der Hauptgründe für maternale und fetale Mortalität und Morbidität. Obwohl die Aetiologie weitgehend unklar ist, wird angenommen, dass eine mangelnde Toleranz von Seiten des mütterlichen Organismus gegenüber dem Fetus und dadurch dessen sukzessive Abstossung ursächlich sein könnte. T regulatorischen Zellen wurde in der Vergangenheit eine mögliche Rolle in der Aufrechterhaltung einer gesunden Schwangerschaft mit erfolgreicher Integration des Fetus, eines durch die paternalen Antigene charakterisierten semi-haploiden Allografts, zugesprochen. In dieser Arbeit wurden T regulatorische Zellen am Ende des dritten Trimesters im peripheren Blut nicht-Schwangerer, gesunder Schwangerer sowie Schwangerer mit Präeklampsie untersucht. Zusätzlich wurden T-regulatorische Zellen in decidualem Gewebe bestimmt. Aufgrund der verschiedenen Ergebnisse vorangegangener Studien und der Uneinigkeit über die Identifizierung T regulatorischer Zellen wurden T regulatorische Zellen in dieser Arbeit mittels der drei gängigsten Markerkombinationen (foxp3+, CD4+CD25hi sowie CD4+CD127loCD25+) charakterisiert. Im peripheren Blut gesunder Schwangerer gab es signifikant mehr T regulatorische Zellen als im peripheren Blut nicht-Schwangerer gemessen mit den Markerkombinationen CD4+CD25hi sowie CD4+CD127loCD25+. In Schwangerschaften mit Präeklampsie zeigten sich eine verminderte Frequenz von T regulatorischer Zellen verglichen mit gesunden Schwangerschaften für die Markerkombinationen foxp3+ und CD4+CD127loCD25+. Auf lokaler Ebene, der Decidua, wurde bei gesunden Schwangerschaften sowie Schwangerschaften mit Präeklampsie eine erhöhte Frequenz gegenüber dem peripheren Blut gemessen. T-regulatorische Zellen scheinen also eine Rolle in der Aufrechterhaltung gesunder Schwangerschaften zu spielen, während sie in Schwangerschaften mit Präeklampsie vermindert sind.
With the progress in sequencing of the honey bee genome new data become available which allows the search and identification of genes coding for homologous proteins found in other organism. Two genes coding for c-type lysozymes were identified in the genome of A. mellifera through an online-based BLAST search. Expression of both intron-less genes seems not to be under the regulatory control of either of the two pathways involved in humoral insect immunity, i.e. Toll and Imd, since no NF-κB transcription factor binding sites are found upstream of the genes. The encoded Lys-1 and Lys-2 are 157 and 143 amino acid long, respectively, and share a sequence similarity of 90%. Further in silico analysis revealed a signal peptidase cleavage site at the N-terminus of each amino acid sequence, strongly suggesting a secretion of the enzymes into the surrounding environment of the producing cells. Sequence alignments of both amino acid sequences with other c-type lysozymes identified the highly conserved active site glutamic acid (Glu32) as well as eight highly conserved cysteine residues. However, an important aspartic acid (Asp50) in the active site that helps to stabilize a substrate intermediate during catalysis is replaced by a serine residue in the lysozymes of A. mellifera. The replacement of the active site aspartic acid in the honey bee lysozymes suggests a different catalytic mechanism and/or a different substrate-specificity in respect to other c-type lysozymes. Furthermore, 3D-models of Lys-1 and Lys-2 were generated based on the sequence similarity of A. mellifera lysozymes with other c-type lysozymes. The published 3D structure of the lysozyme from the silkmoth Bombyx mori, which shares the highest sequence similarity of all available structures with A. mellifera lysozymes, was used as template for the construction of the 3D-models. The models of Lys-1 and Lys-2 suggest that both enzymes resemble, in large part, the structure of B. mori lysozyme. In order to identify the set of AMPs in the hemolymph of A. mellifera, hemolymph of immunized bees was analyzed. Applying SDS-polyacrylamide gel electrophoresis and mass spectrometry on hemolymph from immunized bees, three out of the four peptides were identified, i.e. abaecin, defensin 1 and hymenoptaecin. Furthermore, Lys-2 was identified in the hemolymph by mass spectrometry, conclusively demonstrating the presence of a lysozyme in the hemolymph of A. mellifera for the first time. However, the protein levels of Lys-2 were not affected by bacterial injection, suggesting that the gene expression of the putative antibacterial protein is not under the regulatory control of the Imd and/or Toll pathway. Besides the abovementioned antimicrobial peptides, the 76 kDa large transferrin was also identified. Transferrin is an iron-binding protein that has been implicated in innate immunity in the honey bee. Furthermore, the effect of pathogenic dose, the timeline of peptide induction and the age-related accumulation of the aforementioned AMPs were studied. The intensity of expression of the antimicrobial peptides, abaecin, defensin 1, and hymenoptaecin as well as transferrin increased proportionally with the amount of bacteria injected into the hemocoel. No such effect was observed for the protein levels of Lys-2. Furthermore, up-regulation of the three antibacterial peptides and transferrin was observed within the first 24 h following infection with E. coli (gram-). Infection with the gram+ bacterium Micrococcus flavus resulted in high and moderate protein levels for transferrin and abaecin, respectively, whereas hardly any accumulation of hymenoptaecin was observed, indicating that the gene expression of abaecin and transferrin is somehow positively correlated, and would suggest a shared regulatory pathway that differs from that of hymenoptaecin. Although bacterial infections didn’t seem to stimulate the production of Lys-2, different concentrations in the hemolymph were observed in bees of different ages, suggesting a correlation between the expression of Lys-2 and the age-related division of labor of adult worker honey bees, also known as age polyethism. The results further allow a proposed causal connection between the age-dependent accumulation of Lys-2 and the hemolymph titer of the gonotrophic hormone juvenile hormone, which is the “behavioral pacemaker” in adult honey bees.
While TGF-β is able to regulate miRNA expression in numerous cell types, TGF-β-dependent changes in the miRNA profile of CD8+ T cells had not been studied before. Considering that TGF-β suppresses CD8+ T cell effector functions in numerous ways, we wondered whether induction of immune-regulatory miRNAs could add to the known transcriptional effects of TGF-β on immune effector molecules. In this study, we used miRNA arrays, deep sequencing and qRT-PCR to identify miRNAs that are modulated by TGF-β in human CD8+ T cells. Having found that the TGF-β-dependent downregulation of NKG2D surface expression in NK cells and CD8+ T cells does not go along with a corresponding reduction in mRNA levels, this pathway appeared to be a possible target of TGF-β-inducible miRNAs. However, this hypothesis could not be confirmed by miRNA reporter assays. Instead, we observed that DAP10 transcription is suppressed by TGF-β which in turn negatively affects NKG2D surface expression. In spite of promising preliminary experiments, technical difficulties associated with the transfection of primary NK cells and NK cell lines unfortunately precluded the final proof of this hypothesis.
Instead, we focused on the TGF-β-induced changes in the miRNome of CD8+ T cells and confirmed the induction of the miR-23a cluster members, namely miR-23a, miR-27a and miR-24 by three different techniques. Searching for potential targets of these miRNAs which could contribute to the immunosuppressive action of TGF-β in T cells, we identified and confirmed a previously unknown regulation of IFN-γ mRNA by miR-27a and miR-24. Newly generated miRNA reporter constructs further revealed that LAMP1 mRNA is a target of miR-23a. Upon modulation of the miR-23a cluster in CD8+ T cells by the respective miRNA antagomirs and mimics, significant changes in IFN-γ expression confirmed the functional relevance of our findings. Effects on CD107a/LAMP1 expression were, in contrast, rather minimal. Still, overexpression of the miR-23a cluster attenuated the cytotoxic activity of antigen-specific CD8+ T cells. Taken together, these functional data reveal that the miR-23a cluster not only is induced by TGF-β, but also exerts a suppressive effect on CD8+ T-cell effector functions, even in the absence of TGF-β signaling.
Einfluss einer Prednisolon-Behandlung auf B-Zell-Populationen bei Patienten mit HIV-Infektion

Die HIV-Infektion wird von einer Hyperimmunaktivierung begleitet, die vermutlich eine treibende Kraft in der Pathogenese der Entwicklung von AIDS darstellt. Die Rolle der B-Zellen im Rahmen dieser Veränderungen und dem Einfluss einer Prednisolontherapie wird durch die ProCort-Studie genauer beschrieben. Diese stellt eine zweijährige, Placebo-kontrollierte, randomisierte Doppelblindstudie dar, bei der die Patienten 5mg Prednisolon täglich einnahmen. Mittels durchflusszytometrischen Messungen wurden bestimmte B-Zellpopulationen differenziert und quantifiziert. Die resultierenden Ergebnisse wurden weiterhin mit bekannten Immunaktivierungsmarkern korreliert.
In der Arbeit konnte gezeigt werden, dass Studienteilnehmerinnen durch eine Prednisolontherapie signifikant mehr Memory-B-Zellen bzw. resting Memory-B-Zellen im Vergleich zur Placebogruppe aufweisen.
Ferner konnte die Bedeutung der B-Zellen als prognostischer Marker der HIV-Infektion dadurch unterstützt werden, dass die genannten B-Zellreihen signifikant negative Korrelationen zu anderen, bereits etablierten Progressionsmarkern (CD4/CD8-Ratio, CD8/CD38/HLADR-Aktivierung, suPAR, sCD14) vorlagen.
Zusammenfassend zeigt die Arbeit, dass die Veränderungen im B-Zellkompartment Teil des Immunaktiverungsprozesses im Rahmen der HIV- Infektion sind und Prednisolon modulierende Einflüsse darauf hat.
Shigellosis, or bacillary dysentery, is a rectocolitis caused by the gram-negative, enteroinvasive bacteria of the genus Shigella. Shigellosis still remains a major public health burden with an estimated 80 million cases of bloody diarrhoea and 700.000 deaths per year, primarily in children under the age of 5. Shigella disrupts, invades, and causes inflammatory destruction of the colonic epithelium in humans through virulence effectors secreted by the type III secretion apparatus (TTSA). In contrast to the Shigella-induced manipulation of the host innate immune response, the impact of Shigella on the adaptive immunity has been poorly studied thus far. In order to understand why the naturally induced protective humoral response requires several infections to be primed and is of short duration, the work presented here investigates if Shigella is able to directly interact with T cells. Indeed, it has been shown that Shigella was able to invade and proliferate inside T cells. Furthermore, Shigella was able to inhibit T cell migration through a TTSA effector. Moreover, the Shigella effector IpgD, a phosphoinositide 4-phosphatase that specifically dephosphorylates phosphatidylinositol-(4,5)-bisphosphate (PIP2) into phosphatidylinositol-(5)-monophosphate (PI(5)P), was identified as the effector responsible for the observed inhibition. It could be demonstrated that IpgD was responsible for a reduction of intracellular PIP2 levels in T cells. Further experiments showed a reduced level of phosphorylated ezrin, radixin and moesin (ERM) proteins in infected, as well as with IpgD transfected, T cells. The ERM protein family plays an imported role in signal transduction and motility and their activity is closely related to the binding of PIP2. Therefore, the low level of PIP2 leads to a dephosphorylation of the ERM proteins which inhibits T cells response to chemokine stimulation. Indeed, IpgD transfected T cells show a reduced ability to re-localise the ERM proteins upon chemokine stimulation. Targeting T cell motility, via TTSA effectors, could explain the low level of specific T cell priming during Shigella infection. This is the first report of Shigella induced manipulation of T cell function and on the inhibition of T cell migration by a bacterial effector.
Invasive Aspergillosen zählen auch heute noch zu den potentiell lebensbedrohlichen Infektionen, die gemeinsam mit anderen invasiven Pilzinfektionen für die hohe Mortalität bei immunsupprimierten Patienten verantwortlich sind (Lin et al. 2001). Die Entwicklung und Erforschung spezifischer diagnostischer Methoden und antimykotischer Medikamente konnten die Behandlungschancen einer IA zwar verbessern, bringen aber weiterhin keine befriedigenden Erfolge. So ist es dringend erforderlich, alternative Therapieoptionen zu erforschen und zu entwickeln. Da sich seit einigen Jahren das Augenmerk vermehrt in Richtung Immuntherapie konzentriert und diese Therapieform auch bei der Behandlung invasiver Aspergillosen Anwendung findet, wurden in diesem Zusammenhang die Immunmodulatoren Resiquimod und Alum auf ihre Wirkung auf dendritische Zellen bei einer Aspergillus-Infektion analysiert. Dendritische Zellen besitzen in der Immunabwehr gegen Aspergillus eine Schlüsselrolle, indem sie als Bindeglied zwischen adaptivem und angeborenem Immunsystem fungieren und somit essentiell für eine effektive T-Zell gesteuerte Immunantwort sind. Der mögliche Einfluss der beiden Modulatoren auf die Sekretion inflammatorischer Zytokine dendritischer Zellen wurde auf Protein-Ebene untersucht und die Modifikation der Expression bestimmter Oberflächenmarker als Reaktion auf Resiquimod analysiert. Es zeigte sich, dass das Adjuvans Alum dendritische Zellen in ihrer Immunantwort gegen Aspergillus nicht beeinflusst und zu keiner gesteigerten Sekretion inflammatorischer Zytokine führt. Aus diesem Grund wurde auf die Bestimmung des Expressionsmuster der Oberflächenmoleküle auf dendritischen Zellen in Abhängigkeit von Alum verzichtet. Hingegen konnte Resiquimod einen positiven Trend in der verstärkten Zytokinsekretion aufweisen. So ließ sich in Anwesenheit von Resiquimod eine verstärkte pro-inflammatorische Immunantwort gegen Aspergillus fumigatus erkennen. Dieser additive Effekt von R848 zeigte sich auch bei der Expression kostimulatorischer Moleküle dendritischer Zellen. Es zeigte sich eine gesteigerte Reifung pilzstimulierter dendritischer Zellen in Anwesenheit von Resiquimod durch Zunahme der Level von CD40, CD80 und CD86. In der Expression des Markers CD83 konnte keine einheitliche Aussage getroffen werden, da es spenderabhängig sowohl zu einer Zu-, als auch Abnahme der Fluoreszenzintensität von CD83 als Reaktion auf eine Ko-Stimulation mit Aspergillus und R848 kam. Es war festzustellen, dass die Zellen auf die eingesetzten Stimulantien stark spenderabhängig reagieren.
Auf Grundlage dieser Ergebnisse könnte sich ein möglicher Nutzen des Immunmodulators Resiquimod für die Therapie invasiver Aspergillosen ergeben. Gerade immunsupprimierte Patienten mit einer invasiven Aspergillose könnten von einer DC-basierten Immuntherapie in Verbindung mit Resiquimod profitieren.
Dies gilt es jedoch nur, wenn es durch weitere Analysen und Versuchsreihen bestätigt werden kann.
The pathogenic role of endogenous antibodies in a mouse model for Charcot-Marie-Tooth 1B neuropathy

Charcot-Marie-Tooth (CMT) type 1 neuropathies are a genetically heterogeneous group of non-treatable inherited disorders affecting the peripheral nervous system that lead to sensory and motor dysfunction. Secondary low grade inflammation, implicating the innate and adaptive immune system, could previously be identified as a substantial disease modifier in two mouse models for CMT1, CMT1B and 1X, respectively. However, the exact mechanism how the adaptive immune system contributes to disease pathogenesis is not completely understood. Based on observations that the accumulation of endogenous antibodies to myelin components is important for rapid myelin clearance after nerve injury during Wallerian degeneration, a possibly similar mechanism was considered for endogenous antibodies as disease amplifier in mice heterozygously deficient for P0 (P0het), mimicking some typical features of CMT1B.
In this study an increased antibody deposition was detected in the affected peripheral nerves of P0het myelin mutant mice. By crossbreeding P0het mutants with mice specifically lacking B-lymphocytes, and therefore antibodies (JHD-/-), a decline of endoneurial macrophages together with a substantially ameliorated demyelination could be demonstrated in 6-month-old mutant mice. Moreover, reconstitution with murine IgGs reverted the neuropathic phenotype, substantiating that endogenous antibodies are potentially pathogenic at this early stage of disease. Unexpectedly, in 12-months-old P0het mutants, JHD deficiency resulted in disease aggravation accompanied by an increased inflammatory reaction and M2-polarized macrophage response.
These observations suggest that in a mouse model for CMT1B, the lack of endogenous antibodies has a dichotomous effect: ameliorating early macrophage-mediated demyelination, as opposed to increasing inflammatory reactions leading to disease aggravation at older ages.
Rolle der gammadelta T-Zellen in der Immunantwort bei Patienten mit gastrointestinalen Tumoren

Zusammenfassend konnte im Rahmen der vorliegenden Arbeit die Frage nach der Fähigkeit der selektiven Stimulierung mittels des Phosphorantigens HMBPP und den beiden BTN3 Antikörpern bestätigt werden. Es konnte zudem wie erwartet hierbei ein Unterschied zwischen den beiden Kohorten detektiert werden. Dabei zeigte die Kohorte der Normalspender erwartungsgemäß eine stärkere Aktivierungs- sowie Proliferations-fähigkeit. Normalspender ließen sich signifikant besser mit HMBPP aktivieren und bei bestimmter Konzentration signifikant besser proliferieren, bei BTN3A und sc20.1 konnten keine signifikanten Unterschiede ermittelt werden, allerdings anhand der Mittelwerte eine deutlich stärkere Aktivierung und Proliferation aufgezeigt werden. Außerdem konnten interessante interindividuelle Unterschiede detektiert werden, die neue Erkenntnisse brachten. Mit Hilfe der untersuchten Oberflächenmoleküle CD45RA und CD27 und der Einteilung der gammadelta T-Zellen in unterschiedliche Subgruppen konnten so mögliche Erklärungen für die Unterschiede zwischen den Kohorten aufgezeigt werden. Normalspender zeigten signifikant höhere Anteile an naiven gammadelta T-Zellen und nicht signifikant höhere Anteile an central memory T-Zellen, demnach eine deutliche Verschiebung in Richtung nicht differenzierter Subsets, wohingegen die Tumorkohorte signifikant höhere effector memory T-Zellen aufwiesen und somit eine deutliche Verschiebung in Richtung differenzierter Subsets. Dadurch kann erklärt werden, weshalb Normalspender besser aktiviert werden und besser proliferieren können. Auch die Einteilung in unterschiedliche Profile 1-6 anhand CD28, CD27 und CD16 lieferte Gründe für den Unterschied zwischen den Kohorten, wobei Normalspender der Gruppe 1 und 2, Tumorpatienten der Gruppe 3 und 4 angehörten. Durch Ermittlung weiterer signifikanter Änderungen einiger exprimierter Oberflächenmoleküle CD39, CD161 und PD1 wurde mit Hilfe der vorliegenden Arbeit bekräftigt, dass einige Faktoren betrachtet werden müssen, die die Proliferation und Aktivierung der gammadelta T-Zellen positiv und negativ beeinflussen können. Es konnte jedoch auch erneut verdeutlicht werden, wie komplex und weitgreifend der Aktivierungsmechanismus, die damit verbundene Expansion und die Auslösung der einzelnen Effektorfunktionen ist.
