N-Glykosylierungen spielen beim Env-Gen eine wichtige Rolle. Sie dienen nicht nur als „Escape-Phänomen“ zur Verhinderung einer Elimination des Virus durch neutralisierende Antikörper. Es hat sich gezeigt, dass bestimmte Menschen sich mit HIV infizieren können, aber es zu keinem Zeitpunkt zu AIDS-typischen Symptomen kommt, ohne die Einnahme antiretroviraler Therapie (ART). Solche Menschen werden als Elite Controller bezeichnet. Ihr Organismus kann selbst die Viruslast in sehr geringen Grenzen halten (< 50 Kopien/ml). Ziel dieser Arbeit ist es, den Einfluss von N-Glykosylierungen in der Entstehung von Elite Controller zu untersuchen und prozentuell eine Tendenz zu schaffen, inwieweit die Glykosylierungsdichte des Env-Proteins entscheidend ist. Es konnte gezeigt werden, dass eine immunologische Kontrolle auch auf der B-Zellebene stattfinden kann. Als Hinweis dient die geringe Glykosylierungsdichte im Bereich CD4bs und MPER, die indirekt über MHC Klasse II zu einer erhöhten Produktion von Antikörpern führen kann. Bisher wurde bei Elite Controller die T-Zellebene als mögliche immunologische Kontrolle beschrieben, jedoch gibt diese Arbeit hinweise, dass auch eine immunologische Kontrolle mittels Antikörper möglich ist. Die glykosylierten Zielepitope können eine große Hilfe sein für das Aussehen eines späteren Impfstoffs.
The present thesis concerns the molecular imaging of opioid receptors and human butyrylcholinesterase with the aid of tailored probes, which are suitable for the respective applied imaging techniques. The first part focusses on imaging of opioid receptors with selective probes using total internal reflection- and single molecule fluorescence microscopy. Design and synthesis of the ligands are presented and their pharmacological characterization and application in microscopy experiments are shown. The second part of this thesis focused on the development of 18F-labeled, selective radiotracers for imaging of butyrylcholinesterase via positron emission tomography. The design and synthesis of each a reversible and pseudoirreversible 18F-labeled tracer are presented. After evaluation of the binding properties of each tracer, their initial application in ex vivo autoradiography- and preliminary in vivo microPET studies is described and analyzed.
Basierend auf die vorangegangene Studie der Abteilung für Neuropathologie des Pathologischen Instituts der Universität Würzburg, in der genetische Aberrationen auf dem Chromosom 9 in Ependymome beschrieben wurden, sollte eine Screeninguntersuchung des gesamten Chromosoms sowie eine verfeinerte Analyse der zuvor auffälligen Regionen erfolgen. Ziel der vorliegenden Arbeit war es, mögliche Tumorsuppressorgene oder Onkogene, die mit der Entstehung oder Progression von Ependymome verbunden sind, zu definieren. Weiterhin sollte eine Korrelation zu den klinischen Daten erfolgen. Dabei konnte bei erwachsenen Patienten gezeigt werden, dass Aberrationen auf Chromosom 9 die Prognose verbessern. Speziell der Marker D9S1872 bei DCR1 (DBCCR1) zeigte einen Trend zu einem Überlebensvorteil bei Erwachsenen. Deshalb könnte er in Zukunft zur prognostischen Einteilung von Ependymompatienten in der Klinik beitragen. Des Weiteren konnten bei Kindern in supratentoriellen Tumoren signifikant mehr Aberrationen gefunden wurden als in infratentoriellen. Dies deutet darauf hin, dass Ependymome zumindest bei Kindern abhängig von der Lokalisation unterschiedliche Entstehungswege haben. Es konnten zwei Regionen identifiziert werden, in denen sich häufig Aberrationen fanden: 9p21.1-22.3 und 9q31.3-33.3. Weiterhin konnten Lokalisation und Resektionsausmaß als unabhängige Prognosefaktoren bestätigt werden.
Von Januar 1981 bis Juni 2005 erfolgte bei 87 Patienten in der Klinik und Poliklinik für Mund-, Kiefer-, Gesichtschirurgie der Universität Würzburg eine Rekonstruktion der Nase nach ablativer Chirurgie und Trauma. Ziel der Untersuchung war eine deskriptive Darstellung der Behandlungsmethoden ausgedehnter Defekte an der Nase. Das Patientengut wurde retrospektiv erfasst. Dabei handelt es sich um 81 Tumor- und 6 Traumapatienten. Im Rahmen der Untersuchung wurden 23 (26,4%) Patienten anhand einer visuellen Analogskala im Bezug auf Lebensqualität, Ästhetik und Funktion befragt; gleichzeitig erfolgte eine Beurteilung der Ästhetik durch den Zweituntersucher und meine Person. Die ausgedehnten Nasendefekte des vorgestellten Patientengutes wurden am häufigsten durch gefäßgestielte paramediane Stirnlappen (17 Patienten, 19,5%) versorgt. Der paramediane Stirnlappen stellt in der Klinik und Poliklinik für Mund-, Kiefer-,Gesichtschirurgie der Universität Würzburg eine bevorzugte Operationsmethode dar. Zur Rekonstruktion von ausgedehnten Nasendefekten stehen neben dem paramedianen Stirnlappen, je nach Indikationsstellung, weitere häufig angewandte operative Wiederherstellungsverfahren zur Verfügung. In der Klinik und Poliklinik für Mund-, Kiefer-,Gesichtschirurgie der Universität Würzburg wurden 15 (17,2%) Patienten mit Vollhauttransplantaten von supraclaviculär, 14 (16,1%) Patienten mit Schwenklappen aus der Nasolabial- und Wangenregion oder Rotationslappen und 9 (10,3%) Patienten mit mikrochirurgisch reanastomosierten Radialislappen versorgt. Die Literaturübersicht bestätigt die vorliegenden Untersuchungsergebnisse.
Am Ende ihres stationären Aufenthaltes wurden die Patienten eines psychiatrischen Krankenhauses gebeten in einer freiwillig und anonym durchgeführten Fragebogenaktion den Klinikaufenthalt zu beurteilen. Es sollten neben allgemeinen Angaben u.a. die Behandlung, Betreuung, Information und Aufklärung sowie die Unterbringung beuteilt werden. Die Bewertung erfolgte mittels einer Skala. Zusätzlich hatten die Patienten die Möglichkeit frei formulierte Kommentare abgeben. In der vorliegenden Arbeit wird Validität dieser Fragebogenaktion anhand der freien Kommentare überprüft und bestätigt.
Additive manufacturing processes such as 3D printing are booming in the industry due to their high degree of freedom in terms of geometric shapes and available materials. Focusing on patient-specific medicine, 3D printing has also proven useful in the Life Sciences, where it exploits the shape fidelity for individualized tissues in the field of bioprinting. In parallel, the current systems of bioreactor technology have adapted to the new manufacturing technology as well and 3D-printed bioreactors are increasingly being developed. For the first time, this work combines the manufacturing of the tissue and a tailored bioreactor, significantly streamlining the overall process and optimally merging the two processes. This way the production of the tissues can be individualized by customizing the reactor to the tissue and the patient-specific wound geometry. For this reason, a common basis and guideline for the cross-device and cross-material use of 3D printers was created initially. Their applicability was demonstrated by the iterative development of a perfusable bioreactor system, made from polydimethylsiloxane (PDMS) and a lignin-based filament, into which a biological tissue of flexible shape can be bioprinted. Cost-effective bioink-replacements and in silico computational fluid dynamics simulations were used for material sustainability and shape development. Also, nutrient distribution and shear stress could be predicted in this way pre-experimentally.
As a proof of functionality and adaptability of the reactor, tissues made from a nanocellulose-based Cellink® Bioink, as well as an alginate-based ink mixed with Me-PMeOx100-b-PnPrOzi100-EIP (POx) (Alginate-POx bioink) were successfully cultured dynamically in the bioreactor together with C2C12 cell line. Tissue maturation was further demonstrated using hMSC which were successfully induced to adipocyte differentiation. For further standardization, a mobile electrical device for automated media exchange was developed, improving handling in the laboratory and thus reduces the probability of contamination.
Entwicklung klinischer Methoden zur Quantifizierung der longitudinalen Relaxationszeit T1 in der MRT

Die Aufgabenstellung in der vorliegenden Arbeit bestand in der Entwicklung und Umsetzung neuer T1-Quantifizierungsverfahren, die zuverlässig in der klinischen Routine angewendet werden können. Die ausgearbeiteten Techniken umfassten dabei zwei Hauptarbeitsschwerpunkte.
Zum einen die Implementierung einer neuartigen dynamischen T1- Thermometriemethode für MR-Sicherheitsuntersuchungen medizinischer Geräte und Implantate, wie beispielsweise Kathetern oder Herzschrittmachern, und zum anderen die Entwicklung eines robusten kardialen T1-Mapping-Verfahrens, welches auch bei stärker erkrankten Patienten mit eingeschränkter Atemanhaltefähigkeit stabil anwendbar ist.
Mit der entwickelten kombinierten Heiz- und T1-Thermometriesequenz konnte ein neues Verfahren präsentiert werden, mit dem ein zu untersuchendes medizinisches Gerät oder Implantat kontrolliert erwärmt und die Temperaturänderung zeitgleich präzise erfasst werden kann. Dabei war es möglich, die HF-induzierte Erwärmung der metallischen Beispielimplantate sowohl in homogenem Gel als auch in inhomogenem Muskelgewebe exakt und ortsaufgelöst zu quantifizieren. Die MR-technisch errechneten Temperaturwerte zeigten dabei eine sehr gute Übereinstimmung zu den ermittelten Referenzwerten mit einer Temperaturabweichung von meist weniger als 1K. Die Ergebnisse zeigen, dass es mit der präsentierten Methode möglich ist, die räumliche Temperaturverteilung in einem großen Bereich mit einer einzigen Messung quantitativ zu erfassen. Dies ist neben der Nichtinvasivität der Methode der größte Vorteil im Vergleich zu der Einzelpunktmessung mittels eines bei solchen Messungen sonst zumeist verwendeten fluoroptischen Temperatursensors. Bei gestreckten Implantaten kann demnach idealerweise das gesamte Objekt während einer einzigen Messung auf potentielle Temperaturänderungen oder sogenannte Hotspots untersucht werden, was bei der Verwendung von Temperatursensoren lediglich mit großem Zeitaufwand möglich ist, da hier die Temperatur jeweils nur punktuell erfasst werden kann. Im Vergleich zu anderen publizierten MR-Thermometrieverfahren, welche auf der PRF-Technik basieren, bietet die hier präsentierte Methode vor allem den Vorteil, dass hiermit auch eine präzise Temperaturquantifizierung in inhomogenem biologischem Gewebe mit starken Suszeptibilitätsunterschieden wie beispielsweise zwischen Herz und Lunge möglich ist. Somit stellt die Methode ein leistungsstarkes Hilfsmittel für nicht-invasive MR-Sicherheitsuntersuchungen nicht nur an medizinischen Implantaten sondern beispielsweise auch für MR-geführte Interventionen dar.
Mit der entwickelten kardialen T1-Mapping-Sequenz TRASSI wurde eine leistungsstarke Methode zur exakten und hoch aufgelösten Generierung kardialer T1-Karten in äußerst kurzer Messzeit (< 6 s) vorgestellt. Durch ihre außerordentliche Robustheit sowohl gegenüber Bildartefakten als auch Herzrhythmusstörungen während der Datenakquisition bietet die Sequenz deutlich verbesserte Möglichkeiten für die Diagnostik verschiedener Herzerkrankungen. Aufgrund der sehr kurzen Akquisitionszeit wird insbesondere auch die Generierung von T1-Karten bei schwer erkrankten Patienten mit kurzer Atemanhaltefähigkeit ermöglicht. Im Vergleich zu derzeit üblicherweise verwendeten alternativen Verfahren wie etwa MOLLI, konnten die T1-Karten mit vergleichbarer Bildauflösung in bis zu 70% kürzerer Messzeit akquiriert werden. Die Ergebnisse der durchgeführten Phantommessungen belegen außerdem, dass die Methode exaktere T1-Werte liefert als dies beispielsweise mit MOLLI möglich ist. Des Weiteren weist TRASSI im Gegensatz zu MOLLI keine T1-Abhängigkeit von der Herzrate auf, wodurch die vorgestellte Technik besonders für diagnostische Studien geeignet ist, welche eine sehr hohe Genauigkeit und Reproduzierbarkeit im Zeitverlauf oder zwischen verschiedenen Patienten erfordern.
Mit TRASSI konnten die Strukturen des Herzens bei den durchgeführten in vivo Untersuchungen durchweg mit scharfen Kanten und ohne Bewegungsartefakte dargestellt werden. Dabei wurde unabhängig von der Herzrate und der Bildebene stets eine sehr gute Bildqualität erreicht. Der Hauptgrund hierfür ist vermutlich in der sehr kurzen Akquisitionszeit und der radialen Datenaufnahme zu sehen. Beide Verfahren reduzieren Artefakte aufgrund von Bewegungen wie beispielsweise Herzschlag und Atmung erheblich. Die aufgenommenen T1-Karten zeigen bei allen Probanden und Patienten eine gute diagnostische Bildqualität. So konnten auch die infarzierten Bereiche bei Patienten mit Myokardinfarkt deutlich visualisiert und quantitativ erfasst werden. Nochmals hervorzuheben ist die beobachtete besondere Robustheit der TRASSI Methode gegenüber
Artefakten beziehungsweise T1-Quantifizierungsfehlern bei Patienten mit Herzrhythmusstörungen. Auch bei untersuchten Patienten mit starken Arrhythmien während der Bildgebung konnte eine sehr gute Bildqualität und Genauigkeit der errechneten T1-Karten erreicht werden. Die Ergebnisse der Extrazellularvolumen-Quantifizierung zeigen zudem, dass mittels TRASSI auch weiterführende diagnostische Methoden entwickelt und angewandt werden können. Dabei konnten durch Rückrechnung hochaufgelöster und präziser Extrazellularvolumen-Karten beispielsweise Infarktbereiche deutlich visualisiert und signifikante Unterschiede zwischen akut und chronisch infarziertem Herzmuskelgewebe nicht nur identifiziert sondern auch quantitativ charakterisiert werden. Somit ist diese Methode insbesondere für eine potentielle Differenzierung zwischen reversibel und irreversibel geschädigten Herzarealen interessant.
Für die Zukunft ist es wünschenswert, weitergehende Untersuchungen an verschiedenen spezifischen Herzerkrankungen vorzunehmen. Zu solchen Erkrankungen gehören beispielsweise die Herzmuskelentzündung (Myokarditis) oder Herzklappenerkrankungen. Diese Krankheitsbilder sind hinsichtlich einer möglichen transienten oder permanenten Schädigung des Herzmuskels mit den bisher verfügbaren Verfahren nur sehr schwer oder lediglich im weit fortgeschrittenen Stadium exakt diagnostizierbar. Die vorgestellte TRASSI-Sequenz bietet hier eine gute Möglichkeit für eine frühzeitige Erkennung der Auswirkungen solcher Erkrankungen auf den Herzmuskel. Weiterführende Untersuchungen der TRASSI-Methode zu deren Robustheit gegenüber spezifischen Herzrhythmusstörungen und ein umfassender Vergleich zum bereits etablierten MOLLI-Verfahren könnten darüber hinaus die Alltagstauglichkeit von TRASSI weiter spezifizieren und den Weg in die klinische Routine ebnen. Die bereits dargelegten positiven Ergebnisse des Verfahrens lassen vermuten, dass TRASSI potentiell ein sehr gutes nicht-invasives Diagnoseverfahren für verschiedenste Herzerkrankungen darstellt. Im Vergleich zu bereits bestehenden Techniken liegen die Vorteile der TRASSI-Methode nach den bisher vorliegenden Ergebnissen zusammenfassend vor allem in der Generierung diagnostisch verlässlicherer T1-Werte bei gleichzeitig verringerter Messzeit, wodurch das Verfahren insbesondere auch für schwer erkrankte Patienten mit starken Arrhythmien und eingeschränkter Atemanhaltefähigkeit geeignet ist.
TRASSI ist darüber hinaus aber auch für MR-Untersuchungen im Hochfeld besser geeignet als entsprechende bSSFP-basierende Verfahren wie beispielsweise MOLLI. Dies liegt vor allem daran, dass TRASSI eine Gradientenecho-basierte Bildgebungsmethode ist und somit eine niedrige spezifische Absorptionsrate aufweist. Zudem sind Gradientenecho-Sequenzen allgemein weniger empfindlich gegenüber Suszeptibilitätsartefakten, so dass beispielsweise metallische Implantate bei Patienten sich weniger störend auf die erreichbare Bildqualität auswirken.
In der vorliegenden Arbeit wurde sowohl eine exakte T1-Thermometriesequenz als auch eine sehr schnelle und präzise kardiale T1-Mapping-Methode vorgestellt. Für zukünftige Arbeiten ist es wünschenswert, beide Sequenzen bzw. deren Mechanismen zu vereinen und eine Temperaturquantifizierung am Herzen praktisch durchzuführen. Dies wäre zum einen für MR-Sicherheitsuntersuchungen von Schrittmacherelektroden in vivo vorteilhaft, und zum anderen wäre hiermit eine direkte Erfolgskontrolle während einer Katheterablation realisierbar. Eine solche Ablationsbehandlung könnte durch eine genaue Lokalisierung des behandelten - also erhitzten - Herzareals sehr viel präziser durchgeführt werden, wodurch auch bei komplexeren Ablationen die Behandlungserfolge erhöht werden könnten. In einer ersten Veröffentlichung hierzu konnte bereits gezeigt werden, dass eine MR-gestützte Katheterablation die Heilungs- und Erfolgsaussichten des Eingriffes steigern kann. Dieses Verfahren könnte potentiell mit Hilfe einer Echtzeittemperaturüberwachung basierend auf dem TRASSI-Verfahren noch weiter verbessert werden.
In Zusammenfassung wurden in dieser Arbeit zwei neue T1-Quantifizierungsverfahren entwickelt und vorgestellt, die voraussichtlich zuverlässig im klinischen Alltag angewendet werden können und neue nicht-invasive diagnostische Möglichkeiten eröffnen. Die implementierten Sequenzen ermöglichen dabei zum einen eine exakte Temperaturquantifizierung und zum anderen ein präzises kardiales T1-Mapping. Beide Verfahren versprechen dabei robuste und reproduzierbare Ergebnisse und könnten in Zukunft den Weg in die klinische Routine finden und so bei einer fundierten Diagnostik verschiedenster Herzerkrankungen behilflich sein.
Schultererkrankungen nehmen in sozioökonomischer Hinsicht einen Spitzenplatz unter den Erkrankungen des Bewegungsapparates ein.58 Die häufigen „klassischen Schultererkrankungen“ wie Schulterinstabilitäten, Rotatorenmanschettenrupturen und Omarthrosen sind schon sehr lange bekannt und durch viele Studien gut untersucht. Demgegenüber stellen Läsionen des Aufhängeapparates der langen Bizepssehne (LBS) eine sehr junge Schulterpathologie dar, über die es noch wenige Daten gibt. Im Jahre 1994 formten Walch et al.62 für diese komplexe intraartikuläre Halteschlinge der LBS den Begriff des „stabilizing pulley“, oder Bizepspulley. Im Folgenden sollen Anatomie, Funktion und Pathologie des Bizepspulleys mit allen wichtigen anatomischen Strukturen kurz erläutert werden.
Die Arbeit untersucht den Bezug der Artemis zu Frauen. Neben den meist in der Forschungsliteratur erörterten Aspekten der Artemis als Geburtsgöttin und Beschützerin der Kindheit und der Jugend leistet Artemis bei entscheidenden biologischen Ereignissen im Leben von Frauen Beistand, die den gynäkologischen Aspekt vor Augen stellen. Es handelt sich dabei um die Menarche, die eheliche Vereinigung, die Entbindung und das Wochenbett. Zu diesem Zweck wurden im ersten Teil der Arbeit einschlägige Funktionsepiklesen zusammengestellt, die Zeugnis von diesem Wirkungskreis der Artemis ablegen. Mit Bezug auf die aus dem ersten Teil gewonnenen Ergebnisse untersucht der zweite Teil der Arbeit diese Hilfe der Artemis anhand von Kulten auf der Südwestpeloponnes.
This PhD thesis addresses the photophysics of selected small organic molecules with the purpose of using them for efficient and even novel light sources. In particular, the studies presented focused on revealing the underlying exciton dynamics and determining the transition rates between different molecular states. It was shown how the specific properties and mechanisms of light emission in fluorescent molecules, molecules with phosphorescence or thermally activated delayed fluorescence (TADF), biradicals, and multichromophores can be utilized to build novel light-emitting devices. The main tool employed here was the analysis of the emitters’ photon statistics, i.e. the analysis of the temporal distribution of emitted photons, during electrical or optical excitation. In the introduction of this work, the working principle of an organic light-emitting diode (OLED) was introduced, while Chapter 2 provided the physical background of the relevant properties of organic molecules and their interaction with light. In particular, the occurrence of discrete energy levels in organic semiconductors and the process of spontaneous light emission were discussed. Furthermore, in this chapter a mathematical formalism was elaborated with the goal to find out what kind of information about the studied molecule can be obtained by analyzing its photon statistics. It was deduced that the intensity correlation function g (2)(t) contains information about the first two factorial moments of the photon statistics and that higher order factorial moments do not contain any additional information about the system under study if the system is always in the same state after the emission of a photon. To conclude the introductory part, Chapter 3 introduced the utilized characterization methods including confocal microscopy of single molecules, time correlated single photon counting and temperature dependent photoluminescence measurements. To provide the background necessary for an understanding of for the following result chapters, in Section 4.1 a closer look was taken at the phenomenon of blinking and photobleaching of individual molecules. For a squaraine-based fluorescent emitter rapid switching between a bright and dark state was observed during photoexcitation. Using literature transition rates between the molecular states, a consistent model was developed that is able to explain the distribution of the residence times of the molecule in the bright and dark states. In particular, an exponential and a power-law probability distribution was measured for the time the molecule resides in tis bright and dark state, respectively. This behavior as well as the change in photoluminescence intensity between the two states was conclusively explained by diffusion of residual oxygen within the sample, which had been prepared in a nitrogen-filled glovebox. For subsequent samples of this work, thin strips of atomic aluminum were deposited on the matrices to serve as oxygen getter material. This not only suppressed the efficiency of photobleaching, but also noticeably prolonged the time prior to photobleaching, which made many of the following investigations possible in the first place. For emitters used in displays, emission properties such as narrow-band luminescence and short fluorescence lifetimes are desired. These properties can be influenced not only by the emitter molecule itself, but also by the interaction with the chosen environment. Therefore, before focusing on the photophysics of individual small organic molecules, Section 4.2 highlighted the interaction of a perylene bisimide-based molecular species with its local environment in a disordered polymethyl methacrylate matrix. In a statistical approach, individual photophysical properties were measured for 32 single molecules and correlations in the variation of the properties were analyzed. This revealed how the local polarity of the molecules’ environment influences their photophysics. In particular, it was shown how an increase in local polarity leads to a red-shifted emission, narrower emission lines, broader vibronic splitting between different emission lines in combination with a smaller Huang-Rhys parameter, and a longer fluorescence lifetime. In the future, these results may help to embed individual chromophores into larger macromolecules to provide the chromophore with the optimal local environment to exhibit the desired emission properties. The next two sections focused on a novel and promising class of chromophores, namely linear coordinated copper complexes, synthesized in the group of Dr. Andreas Steffen at the Institute of Inorganic Chemistry at the University of Würzburg. In copper atoms, the d-orbitals are fully occupied, which prevents undesirable metal-centered d-d⋆ states, which tend to lie low in energy and recombine non-radiatively. Simultaneously, the copper atom provides a flexible coordination geometry, while complexes in their linear form are expected to exhibit the least amount of excited state distortions. Depending on the chosen ligands, these copper complexes can exhibit phosphorescence as well as temperature activated delayed fluorescence. In Section 4.3, a phosphorescent copper complex with a chlorine atom and a 1-(2,6-diisopropylphenyl)-3,3,5,5-tetramethyl-2-pyrrolidine-ylidene- ligand was tested for its suitability as an optically active material in an OLED. For this purpose, an OLED with a polyspirobifluorene-based copolymer matrix and the dopant at a concentration of 20 wt% was electrically excited. Deconvolution of the emission spectrum in contributions from the matrix and the dopant revealed that 60 % of the OLEDs emission was due to the copper complex. It was also shown that the shape of the emission spectrum of the copper complex remains unchanged upon incorporation into the OLED, but is red-shifted by about 233 meV. In Section 4.4, a second copper complex exhibiting thermally activated delayed fluorescence was analyzed. This complex comprised a carbazolate as well as a 2-(2,6- diisopropyl)-phenyl-1,1-diphenyl-isoindol-2-ium-3-ide ligand and was examined in the solid state and at the single-molecule level, where single photon emission was recorded up to an intensity of 78’000 counts per second. The evaluation of the second-order autocorrelation function of the emitted light proved an efficient transition between singlet and triplet excited states on the picosecond time scale. In the solid state, the temperature- dependent fluorescence decay of the complex was analyzed after pulsed photoexcitation in the temperature range between 300 K and 5 K. From these measurements, a small singlet-triplet energy gap of only 65 meV and a triplet sublevel splitting of 3.0 meV were derived. The transition rates between molecular states could also be determined. Here, the fast singlet decay time of τS1 = 9.8ns proved the efficient thermally activated delayed fluorescence process, which was demonstrated for the first time for this new class of copper(I) complexes thus. While the use of thermally activated delayed fluorescence is a potential way to harness otherwise long-living dark triplet states, radicals completely avoid dark triplet states. However, this usually comes with the huge drawback of the molecules being chemically unstable. Therefore, two chemically stable biradical species were synthesized in the framework of the DFG research training school GRK 2112 on Molecular biradicals: structure, properties and reactivity, by Yohei Hattori in the group of Prof. Dr. Christoph Lambert and Rodger Rausch in the group of Prof. Dr. Frank Würthner at the Institute of Organic Chemistry at the University of Würzburg, respectively. In Section 4.5, it was investigated how these molecules can be used in OLEDs. In the first isoindigo based biradical (6,6’-bis(3,5-di-tert-butyl-4-phenoxyl)-1,1’-bis(2- ethylhexyl)-[3,3’-biindolinyl-idene]-2,2’-dione) two tert-butyl moieties kinetically block chemical reactions at the place of the lone electrons and an electron-withdrawing core shifts the electron density into the center of the chromophore. With these properties, it was possible to realize a poly(p-phenylene vinylene) copolymer based OLED doped with the biradical and to observe luminescence during optical as well as electrical excitation. Analyzing shapes of the photo- and electroluminescence spectra at different doping concentrations, Förster resonance energy transfer was determined to be the dominant transition mechanism for excitons from the matrix to the biradical dopants. Likewise, OLEDs could be realized with the second diphenylmethylpyridine based birad- ical (4-(5-(bis(2,4,6-trichlorophenyl)methyl)-4,6-dichloropyridin-2-yl)-N-(4-(5-(bis(2,4,6- -trichlorophenyl)methyl)-4,6-dichloropyridin-2-yl)phenyl)-N-(4-methoxyphenyl)aniline) as dopant. In this biradical, chlorinated diphenylmethyl groups protect the two unpaired electrons. Photo- and electroluminescence spectra showed an emission in the near in- frared spectral range between 750 nm and 1000 nm. Also, Förster resonance energy trans- fer was the dominant energy transfer mechanism with an transfer efficiency close to 100 % even at doping concentrations of only 5 wt%. In addition to demonstrating the working OLEDs based in biradicals, the detection of luminescence of the two biradical species in devices also constitutes an important step toward making use of experimental techniques such as optically detected electron spin resonance, which could provide information about the electronic states of the emitter and their spin manifold during OLED operation. Another class of emitters studied are molecules in which several chromophores are co- valently linked to form a macrocyclic system. The properties of these multichromophores were highlighted in Section 4.6. Here, it was analyzed how the photophysical behavior of the molecules is affected by the covalent linking, which determines the interaction be- tween the chromophores. The first multichromophore, 2,2’-ditetracene, was synthesized by Lena Ross in the group of Prof. Dr. Anke Krüger at the Institute of Organic Chemistry at the University of Würzburg and was analyzed in this work both at the single-molecule level and in its aggregated crystalline form. While the single crystals were purified and grown in a vertical sublimation oven, the samples for the single molecule studies were prepared in matrices of amorphous polymethyl methacrylate and crystalline anthracene. Tetracene was analyzed concurrently to evaluate the effects of covalent linking. In samples where the distance between two molecules is sufficiently large, tetracene and 2,2’-ditracene show matching emission profiles with the only difference in the Franck-Condon factors and a de- creased photoluminescence decay time constant from 14 ns for tetracene to 5 ns for 2,2’- ditracene, which can be attributed to the increased density of the vibrational modes in 2,2’-ditracene. Evaluation of the photon statistics of individual 2,2’-ditracene molecules however showed that the system does not behave as two individual chromophores but as a collective state, preserving the spectral properties of the two tetracene chromophores. Complementary calculations performed by Marian Deutsch in the group of Prof. Dr. Bernd Engels at the Institute of Physical and Theoretical Chemistry at the University of Würzburg helped to understand the processes in the materials and could show that the electronic and vibronic modes of 2,2’-ditracene are superpositions of the modes occurring in tetracene. In contrast, single-crystalline 2,2’-ditetracene behaves significantly different than tetracene, namely exhibiting a red shift in photoluminescence of 150 meV, caused by an altered crys- talline packing that lowers the S1-state energy level. Temperature-dependent photolu- minescence measurements revealed a rich emission pattern from 2,2’-ditetracene single crystals. The mechanisms behind this were unraveled using photoluminescence lifetime density analysis in different spectral regions of the emission spectrum and at different tem- peratures. An excimer state was identified that is located about 5 meV below the S1-state, separated by a 1 meV barrier, and which can decay to the ground state with a time constant of 9 ns. Also, as the S1-state energy level is lowered below the E(S1) ≥ 2 ×E(T1) threshold, singlet fission is suppressed in 2,2’-ditetracene in contrast to tetracene. Therefore, at low temperatures, photoluminescence is enhanced by a factor of 46, which could make 2,2’- ditetracene a useful material for future applications in devices such as OLEDs or lasers. The second multichromophore species, para-xylylene bridged perylene bisimide macrocycles, were synthesized by Peter Spenst in the group of Prof. Dr. Frank Würthner at the Institute of Organic Chemistry at the University of Würzburg, by linking three and four perylene bisimides, respectively. To reveal the exciton dynamics in these macrocycles, highly diluted monomers as well as trimers and tetramers were doped into matrices of polymethyl methacrylate to create thin films in which individual macrocycles could be analyzed. The emission spectra of the macrocycles remained identical to those of the monomers, indicating weak coupling between the chromophores. Single photon emission could be verified for monomers as well as macrocycles, as exciton-exciton annihilation processes suppress the simultaneous emission of two photons from one macrocycle. Nevertheless, the proof of the occurrence of a doubly excited state was obtained by excitation power dependent photon statistics measurements. The formalism developed in the theory part of this thesis for calculating the photon statistics of multichromophore systems was used here to find a theoretical model that matches the experimental results. The main features of this model are a doubly excited state, fast singlet-singlet annihilation, and an efficient transition from the doubly excited state to a dark triplet state. The occurrence of triplet-triplet annihilation was demonstrated in a subsequent experiment in which the macrocycles were excited at a laser intensity well above the saturation intensity of the monomer species. In contrast to the monomers, the trimers and tetramers exhibited neither a complete dark state nor saturation of photoluminescence. Both processes, efficient singlet-singlet and triplet-triplet annihilation make perylene bisimide macrocycles exceptionally bright single photon emitters. These advantages were utilized to realize a room temperature electrically driven fluorescent single photon source. For this purpose, OLEDs were fabricated using polyvinylcarbazole and 2-tert-butylphenyl-5-biphenyl-1,3,4-oxadiazol blends as a host material for perylene bisimide trimers. Photon antibunching could be observed in both optically and electrically driven devices, representing the first demonstration of electrically driven single photon sources using fluorescent emitters at room temperature. As expected from the previous optical experiments, the electroluminescence of the molecules was exceptionally bright, emitting about 105 photons per second, which could be seen even by eye under the microscope. Finally, in the last section 4.7 of this thesis, two additional measurement schemes were proposed as an alternative to the measurement of the second-order correlation function g (2)(t) of single molecules, which only provides information about the first two factorial moments of the molecules’ photon statistics. In the first scheme, the g (3)(t) function was measured with three photodiodes, which is a consequential extension of the Hanbury Brown and Twiss measurement with two photodiodes. It was demonstrated how measuring the g (3)(t) function is able to identify interfering emitters with non-Poisson statistics in the experiment. The second setup was designed with an electro-optic modulator that repeatedly gen- erates photoexcitation in the form of a step function. The recording of luminescence transients for different excitation intensities yields the same results as the correspond- ing g (2)-functions measured on single emitters, both in their shape and in their depen- dence on excitation power. To demonstrate this concept, the TADF emitter TXO-TPA (2- [4-(diphenylamino)phenyl]-10,10-dioxide-9H-thioxanthen-9-one) was doped at a concen- tration of 10−4 wt% in a mCP (1,3-Bis(N-carbazolyl)benzene) matrix. This concentration was low enough that TXO-TPA molecules did not interact with each other, but an ensem- ble of molecules was still present in the detection volume. The intramolecular transition rates between singlet and triplet states of TXO-TPA could be derived with an error of at most 5 %. Other experimental techniques designed to obtain this information require ei- ther lengthy measurements on single molecules, where sample preparation is also often a challenge, or temperature-dependent fluorescence lifetime measurements, which require a cryostat, which in turn places constraints on the sample design used. In future, this ap- proach could establish a powerful method to study external factors influencing molecular transition rates. Overall, this thesis has introduced new molecular materials, revealed their photophys- ical properties, and demonstrated how they can be used to fabricate efficient and even novel light sources.
This thesis aims for a better understanding of the mechanisms underlying anxiety as well as trauma- and stressor-related disorders and the development of new therapeutic approaches. I was first interested in the associative learning mechanisms involved in the etiology of anxiety disorders. Second, I explored the therapeutic effects of transcutaneous vagus nerve stimulation (tVNS) as a promising new method to accelerate and stabilize extinction learning in humans.
For these purposes, I applied differential anxiety conditioning protocols realized by the implementation of virtual reality (VR). Here, a formerly neutral virtual context (anxiety context, CTX+) is presented whereby the participants unpredictably receive mildly aversive electric stimuli (unconditioned stimulus, US). Another virtual context (safety context, CTX-) is never associated with the US. Moreover, extinction of conditioned anxiety can be modeled by presenting the same contexts without US delivery. When unannounced USs were administered after extinction, i.e. reinstatement, the strength of the “returned” conditioned anxiety can provide information on the stability of the extinction memory.
In Study 1, I disentangled the role of elemental and conjunctive context representations in the acquisition of conditioned anxiety. Sequential screenshots of two virtual offices were presented like a flip-book so that I elicited the impression of walking through the contexts. Some pictures of CTX+ were paired with an US (threat elements), but not some other screenshots of the same context (non-threat elements), nor the screenshots depicting CTX- (safety elements). Higher contingency ratings for threat compared to non-threat elements revealed elemental representation. Electro-cortical responses showed larger P100 and early posterior negativity amplitudes elicited by screenshots depicting CTX+ compared to CTX- and suggested conjunctive representation. These results support the dual context representation in anxiety acquisition in healthy individuals.
Study 2 addressed the effects of tVNS on the stabilization of extinction learning by using a context conditioning paradigm. Potentiated startle responses as well as higher aversive ratings in CTX+ compared to CTX- indicate successful anxiety conditioning. Complete extinction was found in startle responses and valence ratings as no differentiation between CTX+ and CTX- suggested. TVNS did not affect extinction or reinstatement of anxiety which may be related to the inappropriate transferability of successful stimulation parameters from epilepsy patients to healthy participants during anxiety extinction.
Therefore, in Study 3 I wanted to replicate the modulatory effects of tVNS on heart rate and pain perception by the previously used parameters. However, no effects of tVNS were observed on subjective pain ratings, on pain tolerance, or on heart rate. This led to the conclusion that the modification of stimulation parameters is necessary for a successful acceleration of anxiety extinction in humans.
In Study 4, I prolonged the tVNS and, considering previous tVNS studies, I applied a cue conditioning paradigm in VR. Therefore, during acquisition a cue (CS+) presented in CTX+ predicted the US, but not another cue (CS-). Both cues were presented in a second context (CTX-) and never paired with the US. Afterward, participants received either tVNS or sham stimulation and underwent extinction learning. I found context-dependent cue conditioning only in valence ratings, which was indicated by lower valence for CS+ compared to CS- in CTX+, but no differential ratings in CTX-. Successful extinction was indicated by equal responses to CS+ and CS-. Interestingly, I found reinstatement of conditioned fear in a context-dependent manner, meaning startle response was potentiated for CS+ compared to CS- only in the anxiety context. Importantly, even the prolonged tVNS had no effect, neither on extinction nor on reinstatement of context-dependent cue conditioning. However, I found first evidence for accelerated physiological contextual extinction due to less differentiation between startles in CTX+ compared to CTX- in the tVNS than in the sham stimulated group.
In sum, this thesis first confirms the dual representation of a context in an elemental and a conjunctive manner. Second, though anxiety conditioning and context-dependent cue conditioning paradigms worked well, the translation of tVNS accelerated extinction from rats to humans needs to be further developed, especially the stimulation parameters. Nevertheless, tVNS remains a very promising approach of memory enhancement, which can be particularly auspicious in clinical settings.
Echinococcus multilocularis is the causative agent of alveolar echinococcosis (AE), a life-threatening disease with limited options of chemotherapeutic treatment. Anti-AE chemotherapy is currently based on a single class of drugs, the benzimidazoles. Although acting parasitocidic in vitro, benzimidazoles are merely parasitostatic during in vivo treatment of AE and cause severe site effects. In the case of operable lesions, the resection of parasite tissue needs to be supported by a prolonged chemotherapy. Thus, the current treatment options for AE are inadequate and require alternatives. In the present work, the flatworm signaling pathways were analyzed to establish potential targets for novel therapeutic approaches. I focused on factors that are involved in development and proliferation of E. multilocularis using molecular, biochemical and cell biological methods. Among the analysed factors were three MAP kinases of the parasite, EmMPK1, an Erk-1/2 orthologue, EmMPK2, a p38 orthologue and EmMPK3, an Erk7/8 orthologue. Further, I identified and characterized EmMKK2, a MEK1/2 orthologue of the parasite, which, together with the known kinases EmRaf and EmMPK1, forms an Erk1/2-like MAPK module. Moreover, I was able to demonstrate several influences of host growth factors such as EGF (epidermal growth factor) and insulin on worm signaling mechanisms and larval growth, including the phosphorylation of Elp, an ezrin-radixin-moesin like protein, EmMPK1, EmMPK3 and increased mitotic activity of Echinococcus cells. In addition, several substances were examined for their efficacy against the parasite including (i) general tyrosine kinase inhibitors (PP2, leflunamide), (ii) compounds designed to inhibit the activity of receptor tyrosine kinases, (iii) anti-neoplastic agents (miltefosine, perifosine), (iv) serine/threonine kinase inhibitors that have been designed to block the Erk1/2 MAPK cascade and (v) inhibitors of p38 MAPKs. In these studies, EmMPK2 proved to be a promising drug target for the following reasons. Amino acid sequence analysis disclosed several differences to human p38 MAPKs, which is likely to be the reason for the observed enhanced basal activity of recombinant EmMPK2 towards myelin basic protein in comparison to human recombinant p38 MAPK-α. In addition, the prominent auto-phosphorylation activity of the recombinant EmMPK2 protein together with the absence of an interaction with the Echinococcus MKKs suggest a different mechanism of regulation compared to the human enzyme. EmMPK2 activity could be effectively inhibited in vitro and in cultivated metacestode vesicles by treatment with SB202190 and ML3403, two ATP-competitive pyridinyl imidazole inhibitors of p38 MAPKs, in a concentration-dependent manner. Moreover, both compounds, in particular ML3403, caused parasite vesicle inactivation at concentrations which did not affect cultured mammalian cells. Likewise, during the cultivation of Echinococcus primary cells, the presence of ML3403 prevented the generation of new vesicles. Targeting members of the EGF signaling pathway, particulary of the Erk1/2-like MAPK cascade, with Raf and MEK inhibitors prevented the phosphorylation of EmMPK1 in metacestodes cultivated in vitro. However, although parasite growth was prevented under these conditions, the structural integrity of the metacestode vesicles maintained during long-term cultivation in the presence of the MAPK cascade inhibitors. Similar results were obtained when studying the effects of other drugs mentioned above. Taken together, several targets could be identified that reacted with high sensitivity to the presence of inhibitory substances, but did not cause the parasite’s death with one exception, the pyridinyl imidazoles. Based on the presented data, I suggest pyridinyl imidazoles as a novel class of anti-Echinococcus drugs and imply EmMPK2 as survival signal mediating factor, the inhibition of which could be used for the treatment of AE.
Das grampositive Bakterium Staphylococcus epidermidis ist ein wesentlicher Bestandteil der kommensalen Flora der Haut und der Schleimhäute des Menschen. Jedoch stellen diese Bakterien eine häufige Ursache nosokomialer Katheter-assozierter Infektionen bei immunsupprimierten Patienten dar. Dies liegt zum einen an der Fähigkeit von S. epidermidis, Biofilm zu bilden. Diese physikalische Barriere schützt die Bakterien vor dem Immunsystem sowie vor Antibiotika. Dabei zählen sie zu den häufigsten Erregern von Infektionen an implantierten Fremdkörpern mit Plastikoberflächen, wie z. B. Venenkathetern, künstlichen Herzklappen oder Gefäßprothesen. Zum anderen stellt die Antibiotikaresistenzentwicklung unter S. epidermidis ein zunehmendes Problem dar. Vor allem die late-onset Sepsis, die durch S. epidermidis als Erreger verursacht werden kann, stellt für Frühgeborene eine Gefahr dar. Ziel der vorliegenden Arbeit war, für S. epidermidis als häufigsten und klinisch bedeutsamen KoNS zu eruieren, ob die zunehmende Dauer des stationären Krankenhausaufenthaltes von sehr kleinen Frühgeborenen mit einer höheren Rate an ica-Präsenz, Biofilmbildung und Antibiotikaresistenz assoziiert ist, sowie die Verbreitungswege und das Reservoir für diese S. epidermidis-Stämme zu identifizieren. Hierzu wurden sequenzielle Isolate von S. epidermidis bei Müttern, Kindern und vom Krankenhauspersonal gewonnen und mittels MLST (Multilocus-Sequence-Typing) klonal typisiert. Sie wurden auf Antibiotikaresistenzen, Biofilmbildung und Präsenz des icaA-Gens, das eine Rolle bei der Biofilmbildung spielt, sowie des mecA-Gens untersucht und mit Isolaten, die aus Blutkulturen oder Venenkatheter des Kindes isoliert wurden, verglichen. Es fiel auf, dass die Isolate der sehr kleinen Frühgeborenen deutlich mehr Virulenzfaktoren, wie z.B. Biofilmbildung, hohe Antibiotikaresistenzraten sowie die Präsenz des mecA- und des icaA- Gens, als die maternalen Stämme besaßen. Im Vergleich mit den Ergebnissen der untersuchten Personalstämme liegt der Verdacht nahe, dass oftmals auch das Personal als Transmitter, vor allem von Klonen mit mehreren Virulenzfaktoren, dient. Das Krankenhaus-Milieu scheint dabei ein ideales Reservoir für die Ausbreitung solcher gefährlichen S. epidermidis-Stämme zu sein.
In this doctoral thesis we cover the performance evaluation of next generation data plane architectures, comprised of complex software as well as programmable hardware components that allow fine granular configuration. In the scope of the thesis we propose mechanisms to monitor the performance of singular components and model key performance indicators of software based packet processing solutions. We present novel approaches towards network abstraction that allow the integration of heterogeneous data plane technologies into a singular network while maintaining total transparency between control and data plane. Finally, we investigate a full, complex system consisting of multiple software-based solutions and perform a detailed performance analysis. We employ simulative approaches to investigate overload control mechanisms that allow efficient operation under adversary conditions. The contributions of this work build the foundation for future research in the areas of network softwarization and network function virtualization.
Im Rahmen dieser Arbeit wurde eine neuartige Methode entwickelt, mit der es möglich ist, Magnetisierungsverläufe ausgewählter Schichten und Grenzflächen in dünnen Schichtsystemen zu bestimmen. Diese Resonante Magnetische Röntgenreflektometrie (XRMR: X-ray Resonant Magnetic Reflectometry) kombiniert die Methode der konventionellen Röntgenreflektometrie mit resonanten magnetischen Effekten, die an Absorptionskanten magnetischer Atome auftreten. Analog zur herkömmlichen Reflektometrie, die Aussagen über Schichtdicken und vertikale Grenzflächenrauhigkeiten zulässt, liefert die XRMR das tiefenabhängige magneto-optische Profil der untersuchten magnetischen Schicht. Durch die Aufnahme zweier Reflexionsspektren bei invertierter Helizität des einfallenden Röntgenstrahls oder Umkehr der Magnetisierungsrichtung der Probe in der Nähe der Absorptionskante eines magnetischen Elements erhält man als Messsignal das Asymmetrieverhältnis, das die Information über das tiefenabhängige Magnetisierungsprofil der untersuchten Schicht enthält. Zur Anpassung an die gemessene Asymmetrie über ein optisches Näherungsverfahren ist die Modellierung der optischen Konstanten der magnetischen Schicht oder Grenzfläche notwendig, die hierzu in viele dünne Einzelschichten künstlich aufgeteilt wird. Wichtig hierbei ist die korrekte Bestimmung der dispersiven und absorptiven Ladungsanteilen des komplexen Brechungsindex durch vorherige Messung des Absorptionskoeffizienten und der Berechnung der Dispersion über die Kramers-Kronig-Relation. XRMR-Experimente wurden an Pt/Co-Schichtsystemen an den Synchrotronstrahlungsquellen HASYLAB/Hamburg und BESSYII/Berlin durchgeführt, um die Anwendbarkeit der Messmethodik im harten und weichen Röntgenbereich zu demonstrieren. Durch die intrinsische Elementselektivität resonanter Streuung und die Verstärkung magnetischer Effekte durch Interferenzerscheinungen ist es möglich, Informationen über sehr kleine induzierte magnetische Momente an der Grenzfläche zu einer ferromagnetischen Schicht zu erhalten. Dies konnte bei der Untersuchung einer einzelnen Pt/Co-Bilage gezeigt werden, bei der das Magnetisierungsprofil der Pt-Schicht an der Pt/Co-Grenzfläche bestimmt wurde. Im Weiteren konnte durch XRMR-Messungen an einer Serie von einzelnen Pt/Co-Grenzübergängen das Zusammenspiel von chemischer Grenzflächenrauhigkeit und induziertem Pt-Magnetisierungsprofil untersucht werden. Wichtig war es, die Einsetzbarkeit der Methode im weichen Röntgenbereich zu zeigen, in dem die L2,3 Kanten der 3d-Übergangsmetalle liegen, die für den Magnetismus eine herausragende Rolle spielen. Hierbei konnte durch Messung an der Co-L3 Kante das Magnetisierungsprofil einer einzelnen Co-Schicht in einer Pt/Co/Cu-Trilage extrahiert werden. Des Weiteren erlaubt die Methode die Aufnahme elementspezifischer Hysteresekurven vergrabener dünner Schichten in Schichtsystemen mit hoher Qualität. Das Verfahren ist daher prädestiniert zur quantitativen Untersuchung von modernen neuen magnetoelektronischen Komponenten wie GMR- und TMR-Sensoren, MRAM’s oder Halbleiterstrukturen der viel versprechenden „Spintronic“. Es können bei derartigen Systemen Grenzflächenphänomene vergrabener Schichten zerstörungsfrei untersucht werden und im Weiteren auch Themen, die eher der Grundlagenforschung zuzuordnen sind, wie induzierter Grenzflächenmagnetismus oder auch oszillatorische Austauschkopplung in Zukunft quantitativ und elementselektiv behandelt werden.
The prediction and the experimental discovery of topological insulators has set the stage for a novel type of electronic devices. In contrast to conventional metals or semiconductors, this new class of materials exhibits peculiar transport properties at the sample surface, as conduction channels emerge at the topological boundaries of the system.
In specific materials with strong spin-orbit coupling, a particular form of a two-dimensional topological insulator, the quantum spin Hall state, can be observed.
Here, the respective one-dimensional edge channels are helical in nature, meaning that there is a locking of the spin orientation of an electron and its direction of motion.
Due to the symmetry of time-reversal, elastic backscattering off interspersed impurities is suppressed in such a helical system, and transport is approximately ballistic.
This allows in principle for the realization of novel energy-efficient devices, ``spintronic`` applications, or the formation of exotic bound states with non-Abelian statistics, which could be used for quantum computing.
The present work is concerned with the general transport properties of one-dimensional helical states. Beyond the topological protection mentioned above, inelastic backscattering can arise from various microscopic sources, of which the most prominent ones will be discussed in this Thesis. As it is characteristic for one-dimensional systems, the role of electron-electron interactions can be of major importance in this context.
First, we review well-established techniques of many-body physics in one dimension such as perturbative renormalization group analysis, (Abelian) bosonization, and Luttinger liquid theory. The latter allow us to treat electron interactions in an exact way.
Those methods then are employed to derive the corrections to the conductance in a helical transport channel, that arise from various types of perturbations.
Particularly, we focus on the interplay of Rashba spin-orbit coupling and electron interactions as a source of inelastic single-particle and two-particle backscattering. It is demonstrated, that microscopic details of the system, such as the existence of a momentum cutoff, that restricts the energy spectrum, or the presence of non-interacting leads attached to the system, can fundamentally alter the transport signature.
By comparison of the predicted corrections to the conductance to a transport experiment, one can gain insight about the microscopic processes and the structure of a quantum spin Hall sample.
Another important mechanism we analyze is backscattering induced by magnetic moments. Those findings provide an alternative interpretation of recent transport measurements in InAs/GaSb quantum wells.
In the first part of this work we presented the synthesis and photophysical properties of a series of transition metal donor-acceptor Ir(III)complexes of the type [(C^N)2Ir(N^N)][PF6]. The Ir(III) was connected with hole conducting donor-moieties like carbazole (CZ) and triarylamine (TAA) linked via a methylene and ethylene bridge to the cyclometalating C^N ligands phenylpyrazole (ppz) and phenylpyridine (ppy). Bidentate N^N and P^P ligands like 2,2’-bipyridyl (bpy), 3,4,7,8-tetramethyl-1,10-phenanthroline (tmp) and cis-1,2-bis(diphenylphosphino)ethylene (bdppe) were used as acceptor units. In order to analyse the influence of the electron density of the bpy ligand, TAA-complexes with acceptor- and donor-substituted bpy acceptor units were synthesised. Therefore, 4,4’-dinitro-2,2’-bipyridyl, 4,4’-dichloro-2,2’-bipyridyl, 4,4’-dimethoxy-2,2’-bipyridyl and 4,4’-dimethylamino-2,2’bipyridyl were used as neutral N^N ligands. In order to compare the photophysical properties, all reference compounds without hole conducting component were syntesised. All the carbazole compounds, except the bdppe complexes, exhibit emission and transient absorption properties similar to their reference compounds that make them interesting for OLED (organic light emitting device) applications. LEC (light emitting electrochemical cell) studies show a red shifted luminescence. The triarylamine compounds do not luminesce at RT but they exhibit an intense, blue-shifted and long-lived luminescence at 77 K in a rigid matrix. The transient absorption spectra differ strongly from that of their reference compounds. The spectra display characteristic features of the spectra of the isolated radical anions and cations supported by spectroelectrochemical measurements. Thus, it can be assumed that the transient states are charge separated (CS) states in which the positive charge is localised at the TAA donor units and the negative charge at the N^N acceptor units. The decays of the transient states are biexponentially what indicates the presence of two transient states, the 1CS and the 3CS state. To understand this behaviour the differently substituted bipyridyl-complexes were synthesised and analysed. Temperature dependent transient absorption measurements showed that all rate constants are indepentend of the temperature, except for the complex with OMe subsituents at the bpy ligand. The equilibrium constant K = k1 / k2 is nearly one for all complexes. For the OMe-compound it decreases with increasing temperature. Plotting the rate constants vs. the free energy differences (determined by cyclovoltammetry measurements) shows that all constants are decreasing with increasing donor strength of the bpy ligand. DFT calculations on the OMe-compound are already in work. In the second part of this work, neutral Ir(III) and Pt(II) complexes of the type [(O^O)Ir(N^N)2] and [(O^O)Pt(N^N)] were introduced. There, TTA was connected directly or via a CH2 bridge to acectylacetonate (acac = O^O) in order to probe the influence of the different kinds of connection on the photophysics of the complexes. As the bidentate N^N ligand 2,2’-bipyridyl (bpy) was chosen. All the corresponding reference compounds without triarylamine were obtained in order to compare with the TAA substituted analoga. Furthermore, the homoleptic fac Ir(N^N)3 complex with triarylamine connected via a methylene and ethylene bridge to phenylpyrazole as introduced in the first part of this work was synthesised. The synthesis of the Ir(III) compound with the TAA substituted acac ligand connected via the CH2 group was not successful. All the neutral triarylamine-substituted -diketonato Pt(II) and Ir(III) complexes do not luminesce at RT, except the Pt(II)-complex with CH2 bridge. This compound shows transient state characteristics that are in good agreement with the luminescence lifetimes at RT and that are similar to the reference compound, what suggests to a 3Pt(N^N)(O^O) state. The complexes without the CH2 bridging unit show no transient signals what may be caused by charge-transfer quenching due to the direct linkage between donor and acceptor unit. The homoleptic fac Ir(N^N)3 complex exhibits no emission at RT and no transient signals. At 77 K it shows a highly structured emission with 14 s lifetime. Compared to the literature-known reference compound this emission is caused by the population of a 3Ir(ppz)3 state. Our findings are important for designing complexes with stronger acceptor units (i.e. naphthaleneimide) for long CS states lifetimes to be used as photosynthesisers in solar cells and other optoelectronic devices. Besides, LEC and OLED studies on the carbazole complexes are still of interest to analyse the degree of triplet-triplet-annihiliation in these devices.
Koordinationspolymere auf der Basis von Terpyridin und Dipyridyltriazin: Synthese und Anwendung

Der erste Teil der Arbeit untersucht den Einsatzes von 4,6-Di-(pyrid-2´-yl)-1,3,5-triazin als Baustein für Metallo-supramolekulare Polyelektrolyte. Die dafür nötigen ditopen Liganden werden mittels Stille Kreuzkupplungen dargestellt. Die Absorptions- und Fluoreszenzeigenschaften können durch den Einbau von Oligothiophenen eingestellt werden.
Im zweiten Teil der Arbeit werden die elektrorheologischen Eigenschaften von Metallo-supramolekularen Polyelektrolyten untersucht. Zu diesem Zweck werden die Koordinationspolymere in das Schichtsilikat Montmorillonit interkaliert. Die Interkalation wird mittels verschiedener analytischer Methoden wie Pulverdiffraktometrie, Thermoanalyse oder Infrarotspektroskopie untersucht. Die entstehenden Nanokomposite zeigen einen elektrorheologischen Effekt bei einer geringen Stromdichte.
Die komplexen Aktivitätsmuster während der Futtersuche bei Ameisen sind kein Resultat einer einfachen Selbstorganisation mit starren Regeln sind, sondern diese Regeln werden vielmehr permanent durch den Informationsaustausch zwischen den Arbeiterinnen modifiziert. Die Furagierökologie hat vor allem einen Einfluss auf die Rekrutierungsstrategie der Tiere. Blattschneiderameisen furagieren an großen und stabilen Nahrungsressourcen auf diese sie nach dem Auffinden sofort stark rekrutieren. Camponotus rufipes besucht hingegen Futterquellen, die in ihrer Ergiebigkeit schlecht vorhersagbar sind. Daher steigern die Tiere ihre Rekrutierungsintensität erst nachdem sie sich durch mehrmaliges Aufsuchen der Futterquelle von deren Beständigkeit überzeugt haben.
