The auditory system is an exquisitely complex sensory organ dependent upon the synchronization of numerous processes for proper function. The molecular characterization of hereditary hearing loss is complicated by extreme genetic heterogeneity, wherein hundreds of genes dispersed genome-wide play a central and irreplaceable role in normal hearing function. The present study explores this area on a genome-wide and single gene basis for the detection of genetic mutations playing critical roles in human hearing.
This work initiated with a high resolution SNP array study involving 109 individuals. A 6.9 Mb heterozygous deletion on chromosome 4q35.1q35.2 was identified in a syndromic patient that was in agreement with a chromosome 4q deletion syndrome diagnosis. A 99.9 kb heterozygous deletion of exons 58-64 in USH2A was identified in one patient. Two homozygous deletions and five heterozygous deletions in STRC (DFNB16) were also detected. The homozygous deletions alone were enough to resolve the hearing impairment in the two patients. A Sanger sequencing assay was developed to exclude a pseudogene with a high percentage sequence identity to STRC from the analysis, which further solved three of the six heterozygous deletion patients with the hemizygous, in silico predicted pathogenic mutations c.2726A>T (p.H909L), c.4918C>T (p.L1640F), and c.4402C>T (p.R1468X). A single patient who was copy neutral for STRC and without pathogenic copy number variations had compound heterozygous mutations [c. 2303_2313+1del12 (p.G768Vfs*77) and c.5125A>G (p.T1709A)] in STRC. It has been shown that STRC has been previously underestimated as a hearing loss gene. One additional patient is described who does not have pathogenic copy number variation but is the only affected member of his family having hearing loss with a paternally segregating translocation t(10;15)(q26.13;q21.1).
Twenty-four patients without chromosomal aberrations and the above described patient with an USH2A heterozygous deletion were subjected to a targeted hearing loss gene next generation sequencing panel consisting of either 80 or 129 hearing-relevant genes. The patient having the USH2A heterozygous deletion also disclosed a second mutation in this gene [c.2276G>T (p.C759F)]. This compound heterozygous mutation is the most likely cause of hearing loss in this patient. Nine mutations in genes conferring autosomal dominant hearing loss [ACTG1 (DFNA20/26); CCDC50 (DFNA44); EYA4 (DFNA10); GRHL2 (DFNA28); MYH14 (DFNA4A); MYO6 (DFNA22); TCF21 and twice in MYO1A (DFNA48)] and four genes causing autosomal recessive hearing loss were detected [GJB2 (DFNB1A); MYO7A (DFNB2); MYO15A (DFNB3), and USH2A]. Nine normal hearing controls were also included. Statistical significance was achieved comparing controls and patients that revealed an excess of mutations in the hearing loss patients compared to the control group. The family with the GRHL2 c.1258-1G>A mutation is only the second family published worldwide with a mutation described in this gene to date, supporting the initial claim of this gene causing DFNA28 hearing loss. Audiogram analysis of five affected family members uncovered the progressive nature of DFNA28 hearing impairment. Regression analysis predicted the annual threshold deterioration in each of the five family members with multiple audiograms available over a number of years.
Das Ziel dieser Dissertation besteht darin, die für sein Künstlerleben relevanten Aspekte der Biografie Sándor (Alexander)von Wagners darzustellen. Dabei bewegen wir uns auf einem noch kaum erforschten Gebiet und bieten gleichsam die nötigen Anhaltspunkte für weitere Forschungsbeiträge. Des Weiteren sind die Werke Wagners zu betrachten. Insbesondere die anhand der Biographie belegbaren Einflüsse auf seine Werke und die von seinen Werken ausgehende Wirkung sind dabei zu berücksichtigen, um anhand der Darstellung von Wagners Position, peripher auch diejenige osteuropäischer Künstler, im gesamteuropäischen Kontext aufzudecken.
Im Mittelpunkt der vorliegenden Dissertation steht der künstlerische Werdegang Sándor (Alexander) von Wagners. Er stammte aus Ungarn, ließ sich später in München als professioneller Kunstmaler nieder und arbeitete von 1866 bis 1869 als Hilfslehrer und von 1869 bis 1910 als ordentlicher Professor für Maltechnik an der Münchner Kunstakademie. In dieser Abhandlung werden die für die künstlerische Entwicklung wesentlichen Aspekte seines Lebenslaufes dargestellt:
- die künstlerische Laufbahn Wagners unter besonderer Betrachtung seiner künstlerischen Vorbereitung und Studien sowie seiner Karriere in München
- Grundzüge seiner Werke und Einflüsse, die Wagners Lehrer und Professoren auf sein künstlerisches Schaffen ausübten
- Wagners kunstpädagogische Tätigkeit an der Königlichen Kunstakademie in München
- sein Privatleben in München
Daneben liefert diese Untersuchung eine Liste mit Wagners Studenten und legt ein vorläufiges Werkverzeichnis vor. Vollständigkeit war auf Grund der prekären Forschungslage im Rahmen einer einzelnen Dissertation nicht anzustreben, sondern es galt, eine erste grundlegende Basis zu schaffen, deren Ergänzung weiteren Forschungsbemühungen vorbehalten bleiben muss. Diese Arbeit will damit als Anregung zu weiteren Untersuchungen dienen.
Fragestellung: Allgemeine und mütterspezifische Kontextfaktoren in Zusammenhang mit der Kindererziehung können zu einer mütterspezifischen Problemkonstellation führen, die zur Ausbildung oder Aufrechterhaltung von Gesundheitproblemen beiträgt (vgl. Medizinischer Dienst des Spitzenverbandes Bund der Krankenkassen e.V., 2012). Als Vorsorge- und Rehabilitationsmaßnahmen sind für diesen Bereich Mutter-Kind-Maßnahmen etabliert und eine Qualitätssicherung gesetzlich vorgeschrieben. In dieser Studie wurde untersucht, wie die Prozessqualität von den Müttern wahrgenommen und beschrieben wird.
Material und Methoden: Als Material dienten 147 Erfahrungsberichte aus dem
Internet von Müttern, die an einer Mutter-Kind-Maßnahme teilgenommen hatten.
Die Auswertung erfolgte durch eine qualitative Inhaltsanalyse nach Mayring (2008). Es wurde ein induktives Verfahren gewählt, um eine materialgeleitete Bildung der Kategorien zu erreichen. Kodiert wurden Textstellen, die sich auf Merkmale der Prozessqualität bezogen sowie Bewertungen der Patientinnen von globalen Aspekten und der Maßnahme im Ganzen (Fazit). Insgesamt wurden 2424 Textstellen kodiert, die in 157 Kategorien erfasst und in neun Oberbereiche eingeteilt wurden. Die Strukturierung des Kategoriensystems orientiert sich an dem „idealen Prozess“, der durch eine Expertengruppe unter der Leitung der Universität Würzburg formuliert wurde. Es erfolgte eine Analyse der Nennungshäufigkeiten der Kategorien, der Verteilung auf negative und positive Kategorien, der Bewertungskategorien und der Richtung der Bewertung, sowie des Fazits. Es wurden absolute und relative Häufigkeiten berechnet.
Ergebnisse: Die Mütter beschreiben überdurchschnittlich häufig Merkmale, die in die Phasen Anreise und Hoteleigenschaften kodiert wurden. Besonders häufig werden positive Kategorien in der Phase Anreise genannt, wohingegen viele negative Merkmale in der Phase Hoteleigenschaften beschrieben werden. Die Mütter zeigen sich insgesamt zufrieden mit der Maßnahme (79%), die ärztliche Betreuung und das Freizeitangebot werden allerdings überdurchschnittlich negativ bewertet.
Schlussfolgerung: Diese Studie konnte zeigen, dass Mütter einen anderen Schwerpunkt in der Prozessqualität setzen als Experten. Prozessmerkmale, die sich auf die Hoteleigenschaften beziehen, sind ihnen besonders wichtig. Hier besteht möglicherweise Verbesserungsbedarf. Es sollte geprüft werden, ob eventuell eine vermehrte Berücksichtigung des Bereichs Hoteleigenschaften in zukünftigen Qualitätssicherungsverfahren im Bereich der Prozessqualität sinnvoll wäre, um die Sicht der Mütter zu integrieren. Das Verfahren und das Kategoriensystem sollten an anderen Stichproben validiert werden, um mögliche Schwächen in der Methodik auszuschließen. Eine Überprüfung der Ergebnisse durch andere Erhebungsmethoden ist ebenfalls wünschenswert.
Die vorliegende Dissertation beschäftigt sich mit dem Wesen der Integration im Kontext betrieblicher Informationssysteme. Konkret werden dabei vier Forschungsfragen behandelt:
1. Welche Treiber führen zur Integration betrieblicher Informationssysteme (bzw. verhindern diese)?
2. Welche positiven/negativen Auswirkungen entstehen in Folge der Integration betrieblicher Informationssysteme?
3. Welche Erfolgsfaktoren tragen zum Gelingen/Misslingen der Integration betrieblicher Informationssysteme bei?
4. Wie kann das Konzept der Integration betrieblicher Informationssysteme systematisiert werden?
Zur Beantwortung dieser Forschungsfragen werden zwei wesentliche methodische Ansätze herangezogen:
Im Rahmen einer strukturierten Literaturanalyse werden relevante vorhandene Publikationen gesichtet und systematisch aufgearbeitet, um den Stand der Forschung zu präzisieren und Forschungslücken zu identifizieren.
Die Literaturanalyse bildet die Basis für eine qualitative Querschnittsanalyse, in der 23 Unternehmen aus dem deutschsprachigen Raum (Deutschland, Österreich, Schweiz) mittels semistrukturierten Interviews zu den zuvor identifizierten Forschungslücken befragt wurden.
Der Forschungsbeitrag der Arbeit besteht hauptsächlich in der Konstruktion eines ganzheitlichen Integrationsmodells, das die Forschungsergebnisse in Anlehnung an die vier Forschungsfragen in einem mehrdimensionalen Modell darstellt:
1. Treiber der Integration konnten auf technologischer, organisatorischer, unternehmensinterner und -externer Ebene identifiziert werden. Neuartige Erkenntnisse konnten insb. im Bereich der Hindernisse aufgezeigt werden, die bisher kaum in der Literatur berücksichtigt wurden.
2. Auswirkungen der Integration konnten auf der operativen Ebene, im Management, der Strategie, der IT-Infrastruktur und der Organisation nachgewiesen werden. Neue Einblicke konnten auch hier insb. im Bereich der negativen Auswirkungen geschaffen werden, denen in vorherigen Studien wenig Beachtung geschenkt wurde.
3. Erfolgsfaktoren wurden auf der unternehmensexternen, -internen, organisationalen, technologischen und der Projektmanagement-Ebene gesammelt. Neuigkeitswert besitzen hier insb. die Faktoren im sozio-kulturellen Unternehmensumfeld, die in der Literatur noch kaum bearbeitet wurden.
4. Das facettenreiche Konzept der Integration betrieblicher Informationssysteme wurde nach Sichtung der einschlägigen Literatur als soziotechnisches System modelliert, dessen technische und soziale Subsysteme wechselseitig voneinander abhängen und sich gegenseitig unterstützen. Demzufolge kann auch die Integration im betrieblichen Kontext nur durch eine ganzheitliche Betrachtung der technischen und organisatorischen Teilsysteme eines Unternehmens gelingen.
Die Arbeit endet mit einem kritischen Rückblick auf bestehende Limitationen bzw. einem Ausblick auf zukünftige Forschungsperspektiven, da die gewählte Methodik naturgemäß gewissen Einschränkungen der Validität und Reliabilität unterliegt. Quantitative Forschungsmethoden bieten dahingegen das Potenzial, auf Basis großzahliger Stichproben Ergebnisse mit einer höheren Generalisierbarkeit und Genauigkeit hervorzubringen.
Platelets are important players in haemostasis and their activation is essential to limit post-traumatic blood loss upon vessel injury. On the other hand, pathological platelet activation may lead to thrombosis resulting in myocardial infarction and stroke. Platelet activation and subsequent thrombus formation are, therefore, tightly regulated and require a well-defined interplay of platelet surface receptors, intracellular signalling molecules, cytoskeletal rearrangements and the activation of the coagulation cascade.
In vivo thrombosis and haemostasis models mimic thrombus formation at sites of vascular lesions and are frequently used to assess thrombotic and haemostatic functions of platelets. In this dissertation, different in vivo models were used in mice to address the question at what level a reduced platelet count (PC) compromises stable thrombus formation. To study this, mice were rendered thrombocytopenic by low-dose anti-GPIbα antibody treatment and subjected to a tail bleeding time assay as well as to four different in vivo thrombosis models. Haemostasis and occlusive thrombus formation in small vessels were only mildly affected even at severe reductions of the PC. In contrast, occlusive thrombus formation in larger arteries required higher PCs demonstrating that considerable differences in the sensitivity for PC reductions exist between these models.
In a second part of this study, mice were rendered thrombocytopenic by injection of high-dose anti-GPIbα antibody which led to the complete loss of all platelets from the circulation for several days. During recovery from thrombocytopenia, the newly generated platelet population was characterised and revealed a defect in immunoreceptor tyrosine-based activation motif (ITAM)-signalling. This defect translated into impaired arterial thrombus formation.
To further investigate ITAM-signalling in vivo, genetically modified mice were analysed which display a positive or negative regulation of platelet ITAM-signalling in vitro. Whereas mice lacking the adapter Grb2 in platelets showed a delayed thrombus formation in vivo after acetylsalicylic acid treatment, Clp36ΔLIM bone marrow chimeric mice and SLAP/SLAP2-deficient mice displayed pro-thrombotic properties in vivo. Finally, mice lacking the adapter protein EFhd2 were analysed in vitro and in vivo. However, EFhd2-deficient platelets showed only a minor increase in the procoagulant activity compared to control.
Das Ziel dieser Arbeit war die Entwicklung und die Anfertigung eines 3D Erdfeld-NMR Tomographen, um damit die benötigte Technik der MR eines MR-MPI-Tomographen am Lehrstuhl zu etablieren. Daraufhin wurden alle nötigen Komponenten für ein komplettes 3D Erdfeld-NMR-System entwickelt, gebaut und getestet. Mit diesem Wissen wurde in enger Zusammenarbeit mit der MPI-Arbeitsgruppe am Lehrstuhl ein multimodaler MR-MPI-Tomograph angefertigt und die prinzipielle Machbarkeit der technischen Kombination dieser zwei Modalitäten (MRT/MPI) in einer einzigen Apparatur gezeigt.
Auf diesem Entwicklungsweg sind zusätzlich innovative Systemkomponenten entstanden, wie der Bau eines neuen Präpolarisationssystems, mit dem das Präpolarisationsfeld kontrolliert und optimiert abgeschaltet werden kann. Des Weiteren wurde ein neuartiges 3D Gradientensystem entwickelt, das parallel und senkrecht zum Erdmagnetfeld ausgerichtet werden kann, ohne die Bildgebungseigenschaften zu verlieren. Hierfür wurde ein 3D Standard-Gradientensystem mit nur einer weiteren Spule, auf insgesamt vier Gradientenspulen erweitert. Diese wurden entworfen, gefertigt und anhand von Magnetfeldkarten ausgemessen. Anschließend konnten diese Ergebnisse mit der hier präsentierten Theorie und den Simulationsergebnissen übereinstimmend verglichen werden.
MPI (Magnetic Particle Imaging) ist eine neue Bildgebungstechnik mit der nur Kontrastmittel detektiert werden können. Das hat den Vorteil der direkten und eindeutigen Detektion von Kontrastmitteln, jedoch fehlt die Hintergrundinformation der Probe. Wissenschaftliche Arbeiten prognostizieren großes Potential, die Hintergrundinformationen der MRT mit den hochauflösenden Kontrastmittelinformationen mittels MPI zu kombinieren. Jedoch war es bis jetzt nicht möglich, diese beiden Techniken in einer einzigen Apparatur zu etablieren. Mit diesem Prototyp konnte erstmalig eine MR-MPI-Messung ohne Probentransfer durchgeführt und die empfindliche Lokalisation von Kontrastmittel mit der Überlagerung der notwendigen Hintergrundinformation der Probe gezeigt werden. Dies ist ein Meilenstein in der Entwicklung der Kombination von MRT und MPI und bringt die Vision eines zukünftigen, klinischen, multimodalen MR-MPI-Tomographen ein großes Stück näher.
This dissertation deals with certain business strategies that have become particularly relevant with the spread and development of new information technologies.
The introduction explains the motivation, discusses different ways of defining the term "two-sided market", and briefly summarizes the subsequent essays.
The first essay examines the effects of product information on the pricing and advertising decision of a seller who offers an experience good whose quality is unknown to consumers prior to purchase. It comprises of two theoretical models which differ with respect to their view on advertising. The analysis addresses the question how the availability of additional, potentially misleading information affects the seller's quality-dependent pricing and advertising decision.
In the first model, in which both advertising and product reviews make consumers aware about product existence, the seller's optimal price turns out to be increasing in product quality. However, under certain circumstances, also the seller of a low-quality product prefers setting a high price. Within the given framework, the relationship between product quality and advertising depends on the particular parameter constellation.
In the second model, some consumers are assumed to interpret price as a signal of quality, while others rely on information provided by product reviews. Consequently, and differently from the first part, pricing may indirectly inform consumers about product quality. On the one hand, in spite of asymmetric information on product quality, equilibria exist that feature full information pricing, which is in line with previous results presented by the signaling literature. On the other hand, potentially misleading product reviews may rationalize further pricing patterns. Moreover, assuming that firms can manipulate product reviews by investing in concealed marketing, equilibria can arise in which a high price signals low product quality. However, in these extreme cases, only a few (credulous) consumers consider buying the product.
The second essay deals with trade platforms whose operators not only allow sellers to offer their products to consumers, but also offer products themselves. In this context, the platform operator faces a hold-up problem if he sets classical two-part tariffs (on which previous literature on two-sided markets focussed) as potential competition between the platform operator and sellers reduces platform attractiveness. Since some sellers refuse to join the platform, products whose existence is not known to the platform operator in the first place and which can only be established by better informed sellers may not be offered at all. However, revenue-based fees lower the platform operator's incentives to compete with sellers, increasing platform attractiveness. Therefore, charging such proportional fees can be profitable, what may explain why several trade platforms indeed do charge proportional fees.
The third essay examines settings in which sellers can be active both on an intermediary's trade platform and in other sales channels. It explores the sellers' incentives to set different prices across sales channels within the given setup. Afterwards, it analyzes the intermediary's tariff decision, taking into account the implications on consumers' choice between different sales channels. The analysis particularly focusses on the effects of a no-discrimination rule which several intermediaries impose, but which appears to be controversial from a competition policy view. It identifies under which circumstances the intermediary prefers restricting sellers' pricing decisions by imposing a no-discrimination rule, attaining direct control over the split-up of customers on sales channels. Moreover, it illustrates that such rules can have both positive and negative effects on welfare within the given framework.
Function and regulation of phospholipase D in blood platelets: in vitro and in vivo studies in mice

Summary
Platelet activation and aggregation are crucial for primary hemostasis but can also result in occlusive thrombus formation. Agonist induced platelet activation involves different signaling pathways leading to the activation of phospholipases (PL) which produce second messengers. While the role of PLCs in platelet activation is well established, less is known about the relevance of PLDs. In the current study, the function and regulation of PLD in platelets was investigated using genetic and pharmacological approaches.
In the first part of this thesis, adhesion, activation and aggregation of platelets from mice lacking PLD2 or both PLD1 and PLD2 were analyzed in vitro and in vivo. While the absence of PLD2 resulted in slightly reduced PLD activity in platelets, it had no detectable effect on the platelet function in vitro and in vivo. However, the combined deficiency of both PLD isoforms resulted in defective alpha-granule release and protection in a model of ferric chloride induced arteriolar thrombosis, effects that were not observed in mice lacking only one PLD isoform. These results revealed, for the first time, redundant roles of PLD1 and PLD2 in platelet alpha-granule secretion and indicate that this may be relevant for pathological thrombus formation. Thus, PLD might represent a promising target for antithrombotic therapy.
Thus, this hypothesis was tested more directly in the second part of this thesis. The effects of pharmacological inhibition of PLD activity on hemostasis, thrombosis and thrombo-inflammatory brain infarction in mice were assessed. Treatment of platelets with the reversible, small molecule PLD inhibitor 5-Fluoro-2-indolyl des-chlorohalopemide (FIPI) led to a specific blockade of PLD activity that was associated with reduced -granule release and integrin activation. Mice that received FIPI at a dose of 3 mg/kg displayed reduced occlusive thrombus formation upon chemical injury of carotid arteries or mesenterial arterioles. Similarly, FIPI-treated mice had smaller infarct sizes and significantly better motor and neurological function 24 hours after transient middle cerebral artery occlusion. This protective effect was not associated with major intracerebral hemorrhage or prolonged tail bleeding times. Thus, pharmacological PLD inhibition might represent a safe therapeutic strategy to prevent arterial thrombosis or ischemic stroke.
After revealing a central role for PLD in thrombo-inflammation, the regulation of PLD activity in platelets was analyzed in the last part of the thesis. Up to date, most studies made use of inhibitors potentially exerting off-target effects and consequently PLD regulation is discussed controversially. Therefore, PLD activity in mice genetically lacking potential modulators of PLD activity was determined to address these controversies. These studies revealed that PLD is tightly regulated during initial platelet activation. While integrin outside-in signaling and Gi signaling was dispensable for PLD activation, it was found that PLC dependent pathways were relevant for the regulation of PLD enzyme activity.
The interim reporting process provides decision-useful information to investors and market participants. However the legal circumstances of external interim auditor reviews differ worldwide. A mandatory review rule in the US as opposed to a contrary decision of the German legislator raises the question of the cost-benefit-relation of auditor reviews. Using a German sample of 1,023 firm-year observations from 2007 to 2010, I extract the costs and the benefits of voluntary semi-annual reviews. The unique German legal environment makes it possible to split the cost effect of a review in the price effect (included in audit-related fees) and a possible reduction of audit fees resulting from an improved year-around audit process. I observe a significant increase of audit and audit-related fees of around 14.5% (total fee effect). Additionally, the study provides evidence on declining audit fees for reviewed firms as compared to a matched sample of non-reviewed firms. The effect of an interim review on quarterly earnings quality – using discretionary accruals as an earning management proxy – shows no significant influence.
This study investigates the effect of the error announcement risk on the demand for voluntary interim auditor reviews. Material changes in the German legal environment in 2007 introduced an enforcement system for semi-annual financial reports. The demand for voluntary semi-annual reviews increased significantly from 0.8% in 2006 to 14.6% in 2007 and increased further to 19.5% until 2010 for a sample of 1,278 firm-year observations. This study addresses the question whether the enforcement structure and the resulting error announcement risk exposure have an influence on voluntary external monitoring. After controlling for agency costs, the corporate governance structure, and selected review cost factors, results of a logistic regression analysis show a positive influence of error announcement risk on the likelihood of engaging an auditor to review the semi-annual interim report. The findings contribute to the literature by demonstrating that the quality of the enforcement system and the risk of error findings influence the review decision of the board of directors positively.
In 2004 German legislation established the Financial Reporting Enforcement Panel. In 147 cases since then, the panel has ordered the announcement of errors in previously disclosed and audited financial statements of German firms. We use this unique dataset to evaluate the consequences of increasing earnings management over time on enforcement releases and their recognition in audit fees. Ettredge et al. (2010) provide evidence on a phenomenon called ‘balance sheet bloat’ that is due to income increasing earnings management and later influences the disclosure of misstated financial statements. Thus, the evidence of earnings management recognition in audit fees (Abbott et al. 2006) and the hypothesis of future information content in fees by Stanley (2011) leads us to hypothesize that auditors recognize increasing audit risk in audit fees before the enforcement process starts. We extend related earnings management and audit fee literature by modeling the development of earnings management within the misstatement firms and systematically link it to auditor reactions. We find significant predictive power of different commonly used accrual measures for enforcement releases in the period prior and up to the misstatement period. In this period of time, we also observe an audit fee increase, e.g. the recognition of increased audit risk. We investigate an audit fee effect after the misstatement period but find no significant relation.
Pulse-Sequence Approaches for Multidimensional Electronic Spectroscopy of Ultrafast Photochemistry

Observing chemical reactions in real time with femtosecond laser pulses has evolved into a very popular field of research since it provides fascinating insights into the nature of photochemical transformations. Nevertheless, many photochemical reactions are still too complex for which reason the underlying mechanisms and all engaged species cannot be identified thoroughly. In these cases, conventional time-resolved spectroscopy techniques reach their technical limits and advanced approaches are required to follow the conversion of reactants to their products including all reaction intermediates.
The aim of this work was therefore the development of novel methods for ultrafast spectroscopy of photoreactive systems. Though the concept of coherent multidimensional spectroscopy has so far exclusively been used to explore photophysical phenomena, it also offers great potential for the study of photochemical processes due to its capability of extracting spectroscopic information along several frequency dimensions. This allows resolving the photochemical connectivity between various interconvertible molecular species with ultrafast temporal resolution on the basis of their absorption and emission properties as the spectral correlations are explicitly visualized in the detected spectra.
The ring-open merocyanine form of the photochromic compound 6-nitro BIPS was studied in Chap. 4 of this work. Merocyanines and their associated ring-closed spiropyrans are promising candidates for future applications as, for instance, molecular electronics or optical data storage due to their unique property of being switchable between two stable congurations via light illumination. Transient absorption with sub-50 fs temporal resolution and broadband probing was employed to characterize the photodynamics of this system with variable excitation wavelengths. Using global data analysis, it could be inferred that two different merocyanine isomers with differing excited-state lifetimes exist in solution. These isomers differ in the cis/trans conguration in the last bond of the methine bridge. The minority of isomers exist in the all-trans conguration (TTT) while the isomer with a cis conguration of the third dihedral angle (TTC) is dominant. A characteristic band, detected after long pump-probe delays, was attributed to the unidirectional cis->trans photoisomerization reaction of the TTC to the TTT form. The quantum yield of the reaction was estimated to be (18+-4) %. In addition, pronounced coherent vibrational wave-packet oscillations were observed and it was concluded that these signatures are related to the product formation.
Coherent two-dimensional electronic spectroscopy was successfully implemented using a partially collinear pump-probe beam geometry in combination with a femtosecond pulse shaper. The use of a whitelight probe continuum enabled us to probe contributions far-off the diagonal over the complete visible range. By properly adjusting the relative phase between the first two laser pulses with the pulse shaper, the principle of phase-cycling was explained and it was demonstrated that the measurement can be carried out in the so-called "rotating frame" in which the observed frequencies detected during the coherence time are shifted to lower values. It was shown that these concepts allow the extraction of the desired background-free photon echo while the amount of necessary data points is highly reduced.
In order to put our proposal of multidimensional spectroscopy of photoreactive systems into practice, third-order two- and three-dimensional spectroscopy was then employed for an in-depth analysis of a photoreactive process, in which the photoisomerization of 6-nitro BIPS served as a model system. The measured two-dimensional spectra revealed the cis->trans photoisomerization after long population times. By collecting a large data set of two-dimensional spectra for short population times and by applying a Fourier transform along the population time axis, the third-order three-dimensional spectrum was obtained. The novelty of this approach compared to coherent two-dimensional spectroscopy is the introduction of a third axis associated with the vibrational frequencies of the molecular system. In this way, the formation of the reaction product was evidenced and it was shown that the product is formed in its first excited singlet state within 200 fs after excitation. This method hence visualizes the photochemical connections between different reactive molecular species in an intuitive manner and further exposes the normal modes connecting reactant and product. Such conclusions cannot be drawn with conventional third-order techniques such as transient absorption since they are
not capable of capturing the full third-order response, but only a subset of it. The reaction mechanism and the role of the observed vibrational modes were uncovered by comparing the experimental data with the results of high-level quantum-chemical calculations performed by our collaborators in the group of Prof. B. Engels from the
theoretical chemistry department at the University of Würzburg. Specific calculated molecular normal modes could be assigned to the experimentally observed vibrational frequencies and potential energy surfaces of the electronic ground state and of the first excited state were computed. The technique implemented in this chapter is general and is applicable for the time-resolved analysis of a wide range of chemical reaction networks.
In the first part of Chap. 5, coherent two-dimensional spectroscopy was employed to track the reaction paths of the related 6,8-dinitro BIPS after S1 excitation. Several differences to the photochemical properties of 6-nitro BIPS were found. From the 2D spectra, the cis-trans isomerization between the two merocyanine isomers could be excluded as a major reaction path for this compound. To explore the dynamics after reexcitation to higher-lying electronic states, pump-repump-probe spectroscopy was implemented and the formation of a new species, a radical cation, was observed. To identify the precursor isomer, triggered-exchange two-dimensional spectroscopy, a fifth-order technique previously only available in the infrared regime for vibrational transitions, was implemented for the first time for electronic excitations in the visible. This approach combines the properties of the pump-repump-probe technique with the potential of coherent two-dimensional spectroscopy. It correlates the absorption frequency of a reactive molecular species with the emission signatures of the product formed from this species after an additional absorption of a photon. Using this method, it was unambiguously proven that only the TTC isomer reacts to the radical cation thus forming the precursor species of the reaction. Electronic triggered-exchange two-dimensional spectroscopy is hence another improved technology for time-resolved spectroscopy with applications in the study of multistep photoreactions and higher-lying electronic states. While in the two preceding chapters third- and fifth-order experiments were discussed that neglect the vectorial character of light-matter interactions, Chap. 6 focused on a novel theoretical formalism enabling the description of light fields optimized for polarization-sensitive higher-order nonlinearities. This formalism is based on the von Neumann time-frequency representation of shaped femtosecond laser pulses which permits the definition of multipulse sequences on a discrete time-frequency lattice. Hence, not only the temporal spacing between subpulses is adjustable, but also the center frequencies may be adapted such that they fit the experimental requirements. This method was generalized to the description of pulse sequences with time-varying polarization states. It was shown that by using this description, the polarization ellipticity, orientation angle, relative phase and intensity, and the time-frequency location of each subpulse is explicitly controllable. The accuracy of the transformations from Fourier space to von Neumann domain and vice versa was demonstrated. Moreover, a strict accordance between the von Neumann polarization parameters with the conventional parameters in time domain was found for well separated subpulses. A potential future application of this approach is polarization-sensitive multidimensional spectroscopy in which hidden cross peaks may be isolated by defining the pulses in the von Neumann picture with suitable polarization sequences. This method could also be used in quantum control experiments in which the polarization of the light field is used as a major control knob.
This thesis summarizes our efforts to open the field of femtochemistry to the concept of coherent multidimensional electronic spectroscopy. Making use of femtosecond pulse shaping, sub-50 fs temporal resolution, broadband spectral probing, higher-order nonlinearities, and new types of laser pulse descriptions, the presented methods might stimulate further future advancements in this research area.
Assistierte Reproduktionstechniken (ARTs) zur Behandlung von Infertilität werden mit einer erhöhten Häufigkeit von epigenetischen Aberrationen während der Gametogenese und der frühen Embryonalentwicklung in Verbindung gebracht, speziell durch eine Beeinträchtigung von geprägten Genen. Die in vitro-Maturation (IVM) von Eizellen ist eine ART, die bereits routinemäßig zur Reproduktion von ökonomisch wertvollen Zuchttieren wie dem Hausrind (Bos taurus) eingesetzt wird. IVM-Oozyten weisen jedoch eine verringerte Entwicklungs-kompetenz zum Blastozystenstadium dar, welche möglicherweise auf eine beeinträchtigte epigenetische Regulation zurückzuführen ist.
Von allen bekannten epigenetischen Mechanismen ist die DNA-Methylierung die meist untersuchte DNA-Modifikation. In dieser Arbeit wurden zur Klärung der Frage nach den Auswirkungen der IVM auf die DNA-Methylierung geprägter als auch nicht geprägter Gene Oozyten des Hausrinds analysiert. Diese Tierart weist eine ähnliche Präimplantations-entwicklung und Tragezeit wie der Mensch auf und wird daher zunehmend als Modell zum Studium der humanen Keimzell- und Embryonalentwicklung herangezogen. Im Gegensatz zu Mensch und Maus gibt es bislang nur wenig Information über bovine geprägte Gene. Das erste Ziel der hier dargestellten Forschungsarbeiten war daher die Identifizierung und Charakterisierung der bovinen differenziell methylierten Regionen (DMRs) der drei geprägten Genorte von IGF2/H19, SNRPN und PEG3, welche mit Imprintingdefekten des Menschen und/oder im Mausmodell assoziiert werden. Die hier erstmalig erfolgte Beschreibung von mehreren intergenischen DMRs mittels Bisulfitsequenzierung und Pyrosequenzierung belegt die Existenz und evolutionäre Konservierung der IGF2/H19-Imprintingkontrollregion (ICR) beim Rind. Der geprägte Zustand der IGF2/H19-ICR sowie der bovinen Gene SNRPN und PEG3 wurde durch den Nachweis differenzieller Methylierung in plazentalen und somatischen Geweben sowie in Spermien und parthenogenetischen Embryonen bestätigt. Die beobachteten Methylierungsprofile waren typisch für genomische Prägung.
Die direkte Bisulfitsequenzierung nach vorangegangener Limiting Dilution (LD) erlaubt die Analyse von Methylierungsmustern einzelner Allele (DNA-Moleküle) von einigen wenigen oder auch nur einer einzigen Zelle (El Hajj et al., 2011). In einem ersten LD-Versuch an bovinen Oozyten wurden die drei vorab charakterisierten und geprägten Gene hinsichtlich möglicher epigenetischer Veränderungen untersucht, welche durch verschiedene IVM-Bedingungen und -Medien (TCM und mSOF) hervorgerufen werden könnten. Die Gesamtrate von Methylierungsfehlern einzelner CpG-Stellen sowie die von ganzen Allelen (Imprintingfehlern) unterschied sich nicht wesentlich zwischen den beiden IVM-Gruppen und der in vivo-Gruppe. Dieses Ergebnis weist darauf hin, dass die gängigen IVM-Protokolle keinen oder nur einen geringfügigen Einfluss auf diese entscheidenden epigenetischen Markierungen haben.
IVM-Oozyten präpuberaler Kälber weisen eine herabgesetzte Entwicklungskompetenz im Vergleich zu IVM-Oozyten aus adulten Tieren auf. Aus diesem Grund wurde in einem zweiten LD-Versuchsansatz die Promotormethylierung von drei entwicklungsrelevanten, nicht geprägten Genen (SLC2A1, PRDX1, ZAR1) nach ovarieller Stimulation mit FSH und/oder IGF1 untersucht. Sowohl ungereifte als auch in vitro-gereifte Oozyten präpuberaler und adulter Kühe zeigten eine deutliche, unbeeinträchtige Hypomethylierung der drei Genpromotoren ohne jegliche Unterschiede zwischen den verschiedenen Alterstypen der Spendertiere oder deren Behandlung. Weder das Alter, die hormonelle Stimulation noch die IVM scheinen somit einen Einfluss auf den Methylierungsstatus dieser drei Gene zu haben.
Zusammenfassend spiegelte sich die reduzierte Entwicklungsfähigkeit von IVM-Eizellen aus adulten und präpuberalen Kühen nicht in abnormalen Methylierungsmustern der untersuchten geprägten und ungeprägten Gene wider. Dies lässt auf eine generelle Stabilität der etablierten DNA-Methylierungsprofile in Oozyten schließen. Aus diesem Grund müssen andere epigenetische Mechanismen als die DNA-Methylierung wie beispielsweise ncRNAs oder Histonmodifikationen zur Reduktion der Entwicklungskompetenz von präpuberalen und IVM-Oozyten beitragen. Diese Veränderungen behindern mutmaßlich die zytoplasmatische Reifung der Eizelle, welche wiederum zu einer späteren Beeinträchtigung der Entwicklung der Zygote und des Embryos führt.

The most commonly used assay to quantitate the response of peripheral T~cells upon stimulation with growth factors is determination of incorporated (JH]TdR. We compared thls test to three other methods: 1. direct countlog of cells with a Coulter type counter as reference assay, 2. a colorimetric assay using the tetrazolium dye 3-[ 4,S-dimethylthiazol-l-yl]-2,5diphenyl tetrazolium (MTT), which is a cheap and increasingly popular non-radioactive method and 3. incorporation of the thymidine analog 5-bromo-2'-deoxyuridine detection with a monoclonal antibody on cytospins. Primary human PHA-blasts from >30 healthy individuals were stimulated with IL-2, IL-4 aod IL-7 and assayed with up to four different methods. We discuss the advantages and disadvantages of the assays used and tbe effects of differences between cell preparations. We observed no significant variations between individuals for the dose dependence, but the relative emctency of IL4 compared to IL-2 and IL-7 was variable. This was probably due to the slower response observed upon stimulation with this factor.

The src-gene family in mammals and birds consists of 9 closely related protein tyrosine kinases. We have cloned the c-yes and fyn bomologues of the src-family from the teleost fish Xiphophorus helleri. Both genes show a high degree of sequence conservation and exhibit all structural motifs diagnostic for functional src-like protein tyrosine kinases. Sequence comparisons revealed three domains (exon 2, exons 3--6, exons 7-12) which evolve at different rates. Both genes exhibit an identical expression pattern, with preferential expression in neural tissues. No transcripts of c-yes were found in liver wbich is contrary to the situation in higher vertebrales. In malignant melanoma, elevated Ieveis of c-yes andfyn were detected indicating a possible function during secondary steps of tumor progression for src-related tyrosine kinases.
Autolyzed, antigen-extracted, allogeneic (AAA) bone was prepared from human cortical bone and its morphologic, biomechanical, and osteoinductive properlies were compared with untreated (frozen) as well as lyophilized human bone. Scanning electron microscopy revealed removal of inprganic calcium phosphates and persistence of shrunken collagen fibrils on the surface of AAA bone matrix. Biomechanical testing of differently prepared bone samples showed that lyophilization increased both the modulus of elasticity (P < .00001) and the compressive strength (P < .00001 ). Depending on the depth of decalcification in the preparation of AAA bone, both measured values decreased in rehydrated AAA bone compared with untreated bone {P < .00001 ). Completely demineralized and rehydrated AAA bone was soft, flexible, and showed very little compressive strength. Differences in biomechanical behavior between samples drilled longitudinally or perpendicularly to the diaphyseal bone axis were observed. Xenogeneic human bone samples were implanted in muscle pouches of Sprague-Dawley rats for 6 weeks. AAA bone implants showed chondrogenesis and osteogenesis in 50% of the cases, while untreated or lyophilized bone implants induced no new cartilage or bone formation. As decalcification exposed xenogeneic organic matrix components, AAA bone implants provoked the highest inflammatory reaction. When AAA bone samples were implanted in immunosuppressed rats, the inflammatory reaction was suppressed and 94o/o of the implants showed endochondral bone formation. The chondroinductivity of the bone samples also was tested in vitro using neonatal rat muscle tissue to avoid interference with inflammatory cells and secreted cytokines. In this assay, 68°/o of AAA bone samples induced chondroneogenesis, while untreated as weil as lyophilized bone samples failed to induce any cartilage formation. The results clearly dernonstrafe that AAA bone has osteoinductive properties. Biomechanical stability of AAA bone implants depends on the degree of demineralization. Thus, they can be prepared in an appropriate manner for different indications in oral and maxillofacial surgery.
The expression of the c-src gene in embryonie and adult tissue of the teleost fish Xiphophorus helleri was analyzed by in-situ hybridization. The highly conserved fish c-src gene was found to be expressed at high levels in midterm embryos, where c-src mRNA was localized in developing neurons of the sensory layer of the differentiating retina and in the developing brain. In adult tissues the expression of c-src was found to persist in certain cell types of the brain and the neural retina, especially in the bipolar cells of the inner nuclear layer, which are postmitotic, fully differentiated mature neurons. Thus c-src in Xiphophorus appears to be a developmentally regulated proto-oncogene which is important for neuronal differentiation during organogenesis, but whose persistence of expression in certain terminally differentiated neurons strongly suggests a particular maintenance function for c-src in these cells as well.

Expression of pp60 c-src, the first well defined proto-oncogene product, is developmentally regulated and tissue-specific, with neuronal tissues displaying high amounts of the c-src encoded pp60 c-src kinase activity. In the central nervous system pp60 s-src is preferentially expressed in regions characterized by a high content of grey matter and elevated density of nerve terminals. In this study we show for the first time a direct interaction between pp60 c-src and synaptophysin as a physiological target protein in neurons by demonstrating that endogenous pp60 c-src is able to phosphorylate synaptophysin (p38). p38 is a major constituent of the synaptic vesicle membrane protein and is thought to play a key role in the exocytosis of small synaptic vesicles and possibly small clear vesicles in neuroendocrine cells.

This study addresses the longitudinal relationship among verbal ability, memory capacity, phonological awareness, and reading performance. Data from 92 German children were used to explore the exact relation among these variables. Indicators of verbal ability, memory capacity, and phonological awareness were assessed in kindergarten and again after the first grade. The interrelationships among these factors, and the subsequent influence they have on decoding speed and reading comprehension during the second grade were examined via structural equation modefing procedures. Overall, the results of the longitudinal analyses show that the relationship of memory capacity and phonological awareness remains stable over time, and that memory capacity predicts performance on phonological awareness tasks in both kindergarten and second grade. Phonological awareness proved to be a significant predictor of decoding speed, which in turn considerably influenced reading comprehension.

Junge und erwachsene Schachexperten und -novizen wurden bezüglich ihrer Behaltensleistungen für kurzzeitig dargebotene Schachstellungen und für Anordnungen geometrischer Körper miteinander verglichen. Die Ergebnisse zeigen eine differenzierte Wirksamkeit von Expertise in Abhängigkeit von der Vertrautheit mit dem zu Lernenden Material und von der Art der Aufgabenstellung. Je vorwissensbezogener das zu Lernende Material ist, desto deutlicher ist der Einfluß von Expertise auf Gedächtnisleistungen nachweisbar. Dies gilt in gleicher Weise für unmittelbare wie für längerfristige Behaltensleistungen und für den Lernfortschritt. Im Unterschied dazu zeigt sich weder bei der Vorhersage eigener künftiger noch bei der Bewertung erbrachter Gedächtnisleistungen ein systematischer Einfluß von Expertise.
Im vorliegenden Beitrag werdenneuere nends in der Forschung zur frühen Prognose von Leseleistungen dargestellt. Der Überblick ergibt, daß sich insbesondere solche Forschungsarbeiten bewährt haben, die an kognitiven Informationsverarbeitungsmodellen orientiert sind. Über die Verknüpfung von korrelativen Längsschnitt- und experimentellen Trainingsstudien ließ sich belegen, daß die im Vorschul- bzw. Kindergartenalter beobachtete Informationsverarbeitungskompetenz eine kausale Beziehung zu der schulischen Leseleistung aufwies. Es wurde weiterhin gezeigt, daß phonologische Kompetenzen im Vorschulalter auch effizient trainiert werden können. Trotz dieser insgesamt beeindruckenden Befunde scheinen individuelle Prognosen des Schriftspracherwerbs nach wie vor jedoch problematisch.

In neueren Untersuchungen zur Mathematikerziehung im Elementarbereich wird verstärkt auf die Bedeutung kognitiver Prozesse (Strategien) für die erfolgreiche Bewältigung von Problemlöseaufgaben hingewiesen. Im vorliegenden Beitrag wird insbesondere auf das Wissen um kognitive Prozesse und deren Steuerung, also auf Metakognitionen eingegangen. Es wird zunächst eine Einführung in traditionelle Kategorien von Metakognition gegeben und dann auf eine Weiterentwicklung eingegangen, die als "Modell des kompetenten Strategie-Anwenders" bekanntgeworden ist. Dieses Modell wird dann als Grundlage für Empfehlungen benutzt, die darauf abzielen, den Mathematikunterricht effizienter zu gestalten.
Die Arbeit legt bei den Ausführungen zum theoretischen Hintergrund dar, dass sich hinsichtlich der Rahmenbedingungen für erzieherisches Handeln in den letzten Jahrzehnten viele gesellschaftliche und familiäre Veränderungen ergaben. Befragungen von Eltern zeigten teilweise eine Verunsicherung in Bezug auf die Erziehung ihrer Kinder. Gleichzeitig stellen psychische Störungen und Verhaltensauffälligkeiten im Kindes- und Jugendalter ein gesellschaftlich relevantes Problem dar. Nahezu jedes fünfte Kind weist nach epidemiologischen Studien psychische Probleme auf. Aus Studien zu Risiko- und Schutzfaktoren hinsichtlich der Entstehung solcher Störungen lassen sich verschiedene präventive oder therapeutische Interventionen ableiten. Häufig werden dabei biologische, psychosoziale und familiäre Risikofaktoren unterschieden. Aspekte der Erziehung oder des familiären Umfelds können sowohl förderlich als auch dysfunktional bezüglich der Entwicklung eines Kindes sein. Familiäre Risikofaktoren sind im Vergleich zu vielen biologischen oder psychosozialen Einflüssen potentiell veränderbare Faktoren und sollten unbedingt bei der Behandlung von psychisch kranken Kindern und Jugendlichen mit berücksichtigt werden. Generell erweist sich die Psychotherapie im Kindes- und Jugendalter als ähnlich effektiv wie bei Erwachsenen. Verschiedene Original-, Übersichtsarbeiten und Metaanalysen belegen die Wirksamkeit von Elterntrainings hinsichtlich der Verbesserung von Erziehungsverhalten und Verhaltensauffälligkeiten der Kinder. Wirksamkeitsnachweise finden sich mehrheitlich zu kognitiv-behavioralen Elterntrainings. Sie legen meistens den Schwerpunkt auf konkretes Erziehungsverhalten und versuchen im Sinne des Selbstmanagementansatzes eine Hilfe zu Selbsthilfe bzw. eine Stärkung der elterlichen Ressourcen zu erreichen. Während vor allem randomisiert-kontrollierte Studien aus dem angloamerikanischen und australischen Raum vorliegen, bestanden in Deutschland - im Vergleich zu der Vielzahl der Elterntrainings und der Häufigkeit der Angebote - bis vor einigen Jahren nur relativ wenige Evaluationsstudien.
Mit „Plan E“ stellte sich ein neues Elterntraining in der vorliegenden Studie der Frage nach seinem Wirksamkeitsnachweis. Nach Kenntnis des Autors handelt es sich dabei um das bisher einzige Elterntraining, welches störungsunspezifisch, altersunabhängig, als offene Gruppe und für den Einsatz sowohl im ambulanten als auch im (teil-)stationären Bereich konzipiert wurde.
Nachdem in dieser Studie zunächst das Ausmaß der psychischen Belastung der teilnehmenden Kinder, Jugendlichen und deren Eltern ausführlich dargestellt und entsprechende Zusammenhangsmaße berechnet und präsentiert werden, besteht der Hauptfokus der Arbeit auf der Evaluation des Trainings „Plan E“. Die Untersuchung erfolgte anhand einer klinischen Inanspruchnahmepopulation in einem randomisiert-kontrollierten Design. Diesbezüglich kamen verschiedene Fragebogen- sowie ein standardisiertes Beobachtungsverfahren zum Einsatz.
Durch „Plan E“ ließ sich eine Reduktion dysfunktionaler Erziehungspraktiken sowie eine Verbesserung der emotionalen Befindlichkeit der Eltern erreichen. Auf das elterliche Kompetenzerleben zeigte das Programm keinen Einfluss. Hinsichtlich der Verhaltensauffälligkeiten der Kinder fanden sich bei varianzanalytischen Auswertungen keine Hinweise für einen zusätzlichen Effekt durch die Teilnahme an „Plan E“; dagegen wurde bei Analysen durch non-parametrische Verfahren deutliche Hinweise dafür gefunden, dass durch die Teilnahme am Elterntraining der Anteil von auffällig klassifizierten Kindern deutlicher abnahm. Überprüfungen der Wirksamkeit des Elterntrainings auf die Eltern-Kind-Interaktion anhand einer Beobachtungsskala lieferten keine eindeutigen Befunde.
Die Ergebnisse sowie die Untersuchungsmethodik werden abschließend kritisch diskutiert. Schlussfolgerungen für mögliche weitere Forschungsvorhaben werden dargestellt.
Cystein rich protein 61 (CYR61/CCN1) und Connective tissue growth factor (CTGF/CCN2) stellen aufgrund ihrer Multifunktionalität zwei sehr interessante Vertreter aus der derzeit sechs Mitglieder umfassenden Familie der CCN-Proteine (CCN- CYR61/CCN1, CTGF/CCN2, NOV/CCN3, WISP1-3/CCN4-6) dar. Seit der Entdeckung von CYR61 und CTGF konnten die überlappenden, aber meist nicht redundanten zellspezifischen Effekte in verschiedenen Zellsystemen nachgewiesen werden. Die Einflüsse auf zahlreiche Prozesse wie Proliferation und Migration, aber auch Angiogenese und das Überleben von Zellen lassen eine weitreichende Bedeutung im Zusammenhang mit vielen Entwicklungsprozessen vermuten, so auch der des muskuloskelettalen Systems und der Entwicklung der Lunge.
In der vorliegenden Arbeit wurden für die nähere Charakterisierung von CYR61 und CTGF humane mesenchymale Stammzellen (hMSC) und die humane primäre Lungenendothelzelllinie HPMEC-ST1.6R (human pulmonary microvascular endothelial cells) gewählt. Beide Zellsysteme sind für die Untersuchung der Funktionsfähigkeit in den verschiedenen Kompartimenten bestens geeignet. So ist die Zelllinie HPMEC-ST1.6R den primären Endothelzellen, im Vergleich mit anderen in der Forschung eingesetzten Zelllinien, in Bezug auf spezifische Oberflächenmarker am nächsten. Mesenchymale Stammzellen bilden als multipotente Zellen das Rückrat des muskuloskelettalen Systems und sind an der Homöostase des menschlichen Stütz- und Bewegungsapparates maßgeblich beteiligt.
Um experimentell nutzbare Konzentrationen an rekombinanten Proteinen zu erhalten, wurde ein Baculovirus-Expressionsystems gewählt. Nach der erfolgreichen Klonierung der CTGF/Fc-Tag Sequenz in einen Expressionsvektor konnte dies auch durch Produktion in SF21-Insektenzellen erreicht und erstmalig rekombinantes CTGF/Fc von hoher Reinheit gewonnen werden. Allerdings konnte eine beständige Funktionsfähigkeit der aufgereinigten Proteine mittels eines Proliferationstestes nachfolgend nur bedingt bestätigt werden.
Für die weitere Versuchsplanung, einer Untersuchung der Auswirkung von rekombinantem CTGF (rCTGF) bzw. CYR61 (rCYR61) auf die Zielzellen, musste zunächst die zelleigene ctgf bzw. cyr61 Expression herunterreguliert werden, um einen endogenen Störeffekt auszuschließen. Durch den Einsatz spezifischer shRNAs konnte ctgf/CTGF sowohl in den hMSC-, wie auch den HPMEC-ST1.6R-Zielzellen deutlich herunterreguliert und nachfolgend eine markant reduzierte Proliferation beobachtet werden. Ein Effekt für die Regulation von cyr61 blieb aus.
In dieser Arbeit wurden anschließend erstmals mittels Microarray-Analysen Veränderungen im Genexpressionsmuster der ctgf herunterregulierten hMSC- bzw. Lungenendothelzellen gegenüber Kontrollzellen untersucht. Des Weiteren war die Auswirkung einer Behandlung von ctgf herunterregulierten Zielzellen mit rCTGF gegenüber unbehandelten Kontrollzellen von Interesse. Für beide Zellsysteme konnten signifikante Genregulationen nach der Behandlung mit CTGF spezifischen shRNAs gegenüber den Kontrollzellen detektiert werden, mit interessanten Genclustern im Bereich der TGF-beta (transforming growth factor ß) Signalgebung, sowie der fokalen Adhäsion (z.B. VEGF). Eine Behandlung mit rCTGF hingegen zeigte gegenüber den unbehandelten Kontrollzellen in der Auswertung der Microarray-Analyse keine signifikante Veränderung im Genexpressionsmuster.
In dieser Arbeit wurden, neben einer effektiven Gewinnung von rekombinantem CTGF und der Herunterregulation der endogenen ctgf Expression, wichtige Erkenntnisse zur Biologie von CTGF (und CYR61) in mesenchymalen Stammzellen hMSC und der Lungenendothelzelllinie HPMEC-ST1.6R erlangt. Die erhaltenen Microarray-Daten bieten eine fundierte Grundlage für zahlreiche fortführende Untersuchungen.
Für eine Vielzahl der Frauen mit Blutungsstörungen steht das Erreichen einer „normalen Blutung“ als Therapieziel von Thermachoice® im Vordergrund und weniger ein völliger Blutungsstopp. Im telefonischen Gespräch mit den Studienteilnehmerinnen zeigte sich, dass viele Frauen die Menstruation als einen zyklisch wieder kehrenden Reinigungsprozess des Körpers ansehen, der nach einer inneren Uhr funktioniert, auf den sie ungern verzichten möchten. (Vgl. auch Bitzer et al., 2005, S. 282-287) Auch die Unversehrtheit des Körpers und die Vollkommenheit der Weiblichkeit sind Beweggründe für eine gebärmutter-erhaltende Therapie bei Hypermenorrhoe.
Mit einer signifikanten Reduktion der Blutungsstärke, der Dysmenorrhoe und der seelischen Belastung in dieser Studie, stellt Thermachoice® eine effektive, sichere, kostengünstige und wenig in die körperliche Unversehrtheit eingreifende Option zur Behandlung von Hypermenorrhoe dar.
Im Gegensatz zu hormonellen oder anderen medikamentösen Maßnahmen ist die fehlende systemische Wirkung auf den Organismus bei Thermachoice® ein erwähnenswerter Vorteil. Auch eine langfristige Anwendungsdauer ist bei einer Behandlung mit Thermachoice® hinfällig. Mit der Blutungsreduktion durch die Endometriumablation kann in vielen Fällen das Zeitfenster bis zum Erreichen der Menopause überbrückt werden.
Im Vergleich mit invasiveren Operationsmethoden, wie der Hysterektomie, ist die Komplikationsrate von Thermachoice® geringer, ebenso das Risiko für Nachoperationen. Der Verlust der Fruchtbarkeit durch eine Hysterektomie kann zu einer emotionalen Belastung werden und zu Partnerschaftskonflikten führen. Auch wenn von einer Schwangerschaft nach Thermachoice® aus medizinischer Sicht abzuraten ist, so sind möglicherweise die Tatsache und das Wissen um die erhaltene Gebärmutter ausreichend, um das Auftreten affektiver Störungen zu mindern.
Es ist nicht das Ziel dieser Arbeit festzulegen, ob Thermachoice® besser oder schlechter zur Behandlung von Blutungsstörungen geeignet ist als alternative Therapiemaßnahmen. Es lässt sich sagen, dass Thermachoice® im Rahmen der individuellen Entscheidung eine zufriedenstellende Option zur sanften Behandlung von Blutungsstörungen darstellt. Thermachoice® ist vor allem für diejenigen Frauen geeignet, die ihren Uterus behalten möchten und bei denen keine operationswürdige Senkung des Uterus und der Vagina besteht. Auch Patientinnen mit einem hohen Operationsrisiko und Frauen, bei denen Kontraindikationen für Alternativen bestehen, profitieren von dieser Methode.
Vilos et al. hält fest, dass bei ausgewählten Kandidatinnen nach 10 Jahren klinischer Erfahrung TBEA die bevorzugte chirurgische Methode darstellt, um Menorrhagien zu behandeln.
Für die individuellen Bedürfnisse der Frauen gibt es ein breites Spektrum an Behandlungsangeboten. Letztendlich kann ein ideales Therapieergebnis in Form maximaler Patientinnen-Zufriedenheit nur erzielt werden, indem im Vorfeld alle Vor- und Nachteile der einzelnen Therapieoptionen erörtert werden und gemeinsam mit der Frau eine Entscheidung getroffen wird.
Die Therapie mit Unterkieferprotrusionsschienen sind in der S3-Leitlinie von 2009 und dem Positionspapier der DGZS von 2006 für Patienten mit leichter bis mittelgradiger obstruktiver Schlafapnoe empfohlen.
Diese retrospektive Analyse beschäftigt sich mit dem Therapieverlauf bei Patienten, nachdem sie im Schlaflabor polysomnographisch untersucht worden waren und eine Empfehlung für eine UKPS bekommen hatten.
Hierzu wurden 52 Patienten, die im Schlaflabor nach der PSG in den Jahren 2007 bis 2009 eine Empfehlung für eine Verordnung einer UKPS erhalten hatten, mit Hilfe eines dafür entwickelten Fragebogens und Telefoninterviews nach dem weiteren Therapieablauf befragt.
Von 47 Patienten erhielt man auswertbare Antworten, so dass ihre Daten in die Studie aufgenommen wurden.
Die Charakterisierung der Patientengruppe ergab ein durchschnittliches Alter von 57,3 Jahren; weiterhin nahmen 57,5% Männer und 42,5% Frauen teil. Der durchschnittliche BMI der Gruppe lag bei 30,7. 89,4% der Patienten schnarchen, 44,7% litten unter leichter OSA, 10,6% unter mäßiger OSA und 2,1% unter schwerer OSA. Die ESS-Werte betrugen im Durchschnitt 9,2 von 24 möglichen Punkten.
Der Großteil der Patienten (78,7%) hatte sich nach dem Aufenthalt im Schlaflabor keine UKPS anfertigen lassen.
Als Gründe wurden zu hohe Kosten (18,9%), Zeitmangel (16,2%), geringer Leidensdruck (16,2%), die Bevorzugung anderer Therapien (13,5%), Unwissenheit über die Therapieempfehlung UKPS (13,5%), Abraten durch den Zahnarzt (13,5%) und die Ablehnung des Fremdkörpers im Mund (2,7%) angegeben.
Lediglich 21,3%, also 10 von 47 Patienten, wurden mit einer UKPS versorgt. Bei der Beurteilung der Schlafqualität auf einer Skala von 1 (sehr schlecht) bis 7 (sehr gut) gaben fünf Patienten an, dass sich ihr Schlafqualität seit Nutzung der Schiene deutlich verbessert habe. Sie bewerteten ihre Schlafqualität auf der Skala im Durchschnitt mit 2,2 Punkten höher. Drei Patienten konnten keine Unterschiede feststellen. Lediglich zwei Patienten schliefen mit den UKPS schlechter.
Die Therapie führte weiterhin zu einer Reduktion des subjektiv empfundenen Schnarchens (nicht reduziert bei 0, mäßig bei 4 (40%), stark bei 5 Patienten (50%), vollständig bei einem Patienten (10%)).
Die Punktwerte im ESS lagen vor Therapie bzw. mit UKPS bei 8,4 vs. 5,7 (Verbesserung im Durchschnitt um 2,7 Punkte). 80% der Patienten benutzten die UKPS durchschnittlich sechs bis sieben Nächte, 20% nur vier Nächte pro Woche.
Abschließend lässt sich feststellen, dass die Therapie mit UKPS effektiv ist.
Die Umsetzung der Empfehlung für eine UKPS durch die Patienten ist allerdings unzureichend. Verlaufsuntersuchungen, wie bei der PAP-Therapie gefordert, sind bei den Patienten nicht erfolgt. Sowohl die Aufklärung über die empfohlenen Therapiemaßnahmen, als auch die weitere medizinische Betreuung nach Entlassung aus dem Schlaflabor sind verbesserungswürdig.

Am Beispiel einer semantischen Kategorisierungsaufgabe (sort-recall task) wurde der Frage nachgegangen, in welchen Bestimmungsgrößen sich die Gedächtnisleistungen von Schulkindern, jüngeren sowie älteren Erwachsenen voneinander unterscheiden. Es wurde angenommen, daß für diese drei Altersgruppen Gedächtnisleistungen bei dieser Aufgabe in unterschiedlicher Weise durch Strategie- und Wissensaspekte bestimmt sind. Die im Vergleich zu Schulkindern und älteren Erwachsenen üblicherweise besseren Leistungen jüngerer Erwachsener sollten demnach im wesentlichen auf die konsequentere Nutzung von Gedächtnisstrategien rückführbar sein. Erwartet wurde weiterhin, daß die bei Schulkindern und älteren Erwachsenen oft vorfindbaren "Produktionsdefizite" in der Strategienutzung unterschiedliche Ursachen haben : fehlt es bei den Schülern am notwendigen Gedächtniswissen (Metagedächtnis), so sind die Defizite der älteren Menschen vorwiegend auf die mangelnde Erfahrung mit der Aufgabe zurückzuführen. Diese Annahmen wurden in einer Studie mit je 24 Probanden aus den drei genannten Altersgruppen überprüft. Während sich das erwartete Produktionsdefizit bei den Kindern auf unzureichendes Metagedächtnis zurückführen ließ, gab es wenig Anhaltspunkte dafür, daß das Strategiedefizit älterer Menschen in wesentlichen auf mangelnde Aufgabenerfahrung rückführbar ist. Leistungsunterschiede zwischen jüngeren und älteren Erwachsenen beruhen nicht auf unterschiedlichem Gedächtniswissen, sondern dürften auf dem kombinierten Einfluß von Strategie- und Kapazitätsdefiziten basieren.
To study the activation of HIV by human spumaretrovirus (HSRV) the long terminal repeats (LTRs) of HSRV, HIVl and HIV2 were examined with respect to their ability to function as transcriptional promoters in virus infected and uninfected cells. Transient transfections using plasmids in which the L TRs of the three viruses were coupled to the bacterial chloramphenicol acetyltransferase (CA T) gene revealed (i) the level of cat gene expression directed by the HSRV LTR was markedly increased in HSRV infected cells compared to uninfected cells, (ii) cat gene expression driven by the HIV1 LTR, but not by the HIV2 LTR could be enhanced upon HSRV infection, whereas (iii) neither in HIV1 nor in HIV2 infected cells an effect on HSRV LTR driven cat geneexpression was detected.
Neoplasia in Xiphophorus can be classified into: a) a Jarge group triggered by carcinogens; b) a large group triggered by promoters; and c) a small group that develops "spontaneously" according to Mendelian Jaw. The process leading to susceptibility for neoplasia is represented by the disintegration of gene systems that normally protect the fish from neoplasia. Interpopulational arid interracial hybridization is the most effective process that Ieads to disintegration of the protective gene systems. Environmental factors may complete disintegration in somatic cells and thus may trigger neoplasia. The applications of the findings on Xiphophorus to humans are discussed.
The capacity of Xiphophorus to develop neoplasia can be formally assigned to a "tumor gene" (Tu), which appears to be a normal part of the genome of all individuals. The wild fish have evolved population-specific and cell type-specific systems of regulatory genes (R) for Tu that protect the fish from neoplasia. Hybridization of members of different wild populations in the laborstory followed by treatment of the hybrids with carcinogens led to disintegration of the R systems permitting excessive expression of Tu and thus resulting in neoplasia. Certain hybrids developed neoplasia even spontaneously. Observations on the genuine phenotypic effect of the derepressed Tu in the early embryo indicated an essential normal function of this oncogene in cell differentiation, proliferation and cell-cell communication. Tu appeared to be indispensable in the genome but may also be present in accessory copics. Recently, c-src, the cellular homolog of the Rous sarcoma virus oncogene v-src, was detected in Xiphophorus. The protein product of c-src, pp60c-src, was identified and then examined by its associated kinase activity. This pp60c-src was found in all individuals tested, but, depending on the genotype, its kinase activity was different. The genetic characters of c-src, such as linkage relations, dosage relations, expression, etc., correspond to those of Tu. From a systematic study which showed that pp60c-src was present in all metazoa tested ranging from mammals down to sponges, we concluded that c-src has evolved with the multicellular organization of animals. Neoplasia of animals and humans is a characteristic closely related to this evolution. Our data showed that small aquariurn fish, besides being used successfully because they are time-, space-, and money-saving systems for carcinogenicity testing, are also highly suitable for basic studies on neoplasia at the populational, morphological, developmental, cell biological, and molecular levels.
A panel of simple repetitive oligonucleotide probes has been designed and tested for multilocus DNA fingerprinting in some 200 fungal, plant and animal species as well as man. To date at least one of the probes has been found to be informative in each species. The human genome, however, has been the major target of many fingerprintins studies. Using the probe (CAC)5 or (GTG)5, individualization of all humans is possible except for monozygotic twins. Paternity analyses are now perfonned on a routine basis by the use of multilocus fingerprints, inctuding also cases of deficiency, i.e. where one of the parents is not available for analysis. In forensie science stain analysis is feasible in all tissue remains containing nuc)eated cells. Depending on the degree of DNA degradation a variety of oligonucleotides are informative, and they have been proven useful in actual case work. Advantages in comparison to other methods including enzymatic DNA amplification techniques (PCR) are evident. Fingerprint patterns of tumors may be changed due to the gain or loss of chromosomes and/or intrachromosomal deletion and amplification events. Locus-specific probes were isolated from the human (CAC)5/( GTG)5 fingerprint with a varying degree of informativeness (monomorphic versus truly hypervariable markers). The feasibility of three different approaches. for the isolation of hypervariable mono-locus probes was evaluated. Finally, one particular mixed simple (gt)n(ga)m repeat locus in the second intron of the HLA-DRB genes has been scrutinized to allow comparison of the extent of exon-encoded (protein-) polymorphisms versus intronie bypervariability of simple repeats: adjacent to a single gene sequence (e.g. HLA-DRB1*0401) many different length alleles were found. Group-specific structures of basic repeats were identified within the evolutionarily related DRB alleles. As a further application it is suggested here that due to the ubiquitous interspersion of their targets, short probes for simple repeat sequences are especially useful tools for ordering genomic cosmid, yeast artificial chromosome and phage banks.
1. Thr ecological separation of 19 carrion inscet species (adults and some of their larvae) was investigated at rabbit carcasses in North Bavaria (FRG) referring to 4 niche dimensions. In the (1.) macrohabitats (forest - clearing) the distribution of saprophageous beetle larvae was mainly considered, for (2.) seasonality the differential abundance of blow flies (Colliphoridae). (3.) The stages of decay were correlated with the temperature dependent development of blow fly maggots affecting the abundance of competing saprophageous beetles and of carnivors preying upon maggots of different size classes. By using ( 4.) microhabitats (spatial subdivision of a carcass) as further niche dimension, the dustering of speries using similar food resources was domonstrated in a niche overlap dendrogram.
2. The quantitative effect of predators on blow fly maggots was investigated both in field and laboratory experiments. Predation upon maggots rcduces their scramble competition, resulting in a higher pupal weight. Thus, the reproductive succcss of the blow flies seems to be buffered by the developmental flexibility of the calliphorids. The numerical effect of predators and parasitoids on the blow fly pupae was also quantified.
3. In a case study on the staphylinid beetle, Aleochara curtula, we investigated the diffenntial abundance of sexes. The ratio at which the males and females arrive at the carcass is balanced. Here the beetles feed and copulated. Females depart into the vicinity of the carrion much earlier than males, thus shifting the sex ratio to a maale bias. In the surroundings the females deposit their eggs, and the parasitoid first instar larvae search for scattered blow fly pupae. The temporal and spatial distribution of both sexes of A. curtula is thus not only affected by the food allocation of the adults, but a.lso by limiting resources of mating end egg laying sites as well as larval hosts.
Characterization of the env gene and of two novel coding regions of the human spumaretrovirus

Recombinant clones harboring retroviral DNA were established. The nucleotide sequence of the central and 3' region of the genome of the human spumaretrovirus was determined. The 5' end of the deduced protein sequence was homologaus to the endonuclease domain of retroviral reverse transcriptases. A small intergenic region is followed by a lang open reading frame of 985 aminoacid residues that according to its genomic location and structural features is a typical retroviral env gene. Surprisingly, the postenv region contains two open reading frames that encodes two novel retroviral genes, termed bel-l and bel-2. The 3' LTR is 963 nucleotides lang and contains the signal sequences characteristic for transcriptional regulation of retrovirus genomes.
Tbe alkylating potency of unstable N-nitrosamino acids and N-nitrosopeptides was investigated in vitro using 4-(para-nitrobenzyl)pyridine (NBP) as nucleophile. Of the amino acids, Met and those with an aromatic side chain were the most potent. The relative overall alkylating potency was 23:10:5:4:2:1: for Trp, Met, His, 1)rr, Phe and Gly, respectively. The homo-dipeptides were much more potent than the amino acids, with relative potencies of 400:110:100:8:3:1, for Trp-Trp, l)T-'I)T, Met-Met, Asp-Asp, Phe-Phe and Gly, respectively. In the one-phase reaction system (in which NBP is already present durlog the nitrosation reaction at acidic pH), all amino acids tested showed a second-order reaction for nitrite. In the two-phase system (in which NBP is added only after bringing the nitrosation reaction mixture to neutrality), all amino acids tested except one again showed a second-order reaction for nitrite (Phe, His, Asp and the dipeptide artiticial sweetener aspartame); only Met under these conditions bad a reaction order of one for nitrite. This could mean that nitrosation of the side chain of Metproduces a second N-nitroso product which is relatively stable in acid but reacts with NBP under neutral conditions. In the human stomach, this side-chain nitrosation might become more important than the reactions at the primary amino group, firstly because of the greater stability of the product(s) in acid and secondly because of the tirst-order reaction rate for nitrite. A decrease in nitrite concentration from the millimolar concentrations ofthe in-vitro assay to the micromolar concentrations in the stomach reduces the reaction rate by a factor of 1000 for the side-chain nitrosation, whereas a million-fold reduction will be observed for nitrosation of the amino group.
Known mutagens and carcinogens in the dict were compiled and the risk of cancer was estimated on the basis of average exposure Ievels in Switzerland and carcinogenic potencies from rodent bioassays. The analysis showed that, except for a1cohol, the sum of all known dietary carcinogens could only explain a few percent of the cancer deaths attributed by epidemiologists to dietary factors. The discrepancy was explained by a "carcinogenicity" of excess macronutrients. This hypothesis was based on an evaluation of dietary restriction experiments in rats and mice, where a dramatic reducing effect on spontaneaus tumour formation was seen. From these experiments, a "carcinogenic potency" was deduced for food in excess (TD50 approximately 16 g/kg per day). Ovemutrition in Switzerland was converted into excess food intake and the cancer risk estimated on the basis ofthe TD50 value. The resulting risk of60,000 cases per one million lives wou1d aJlow to explain by overnutrition almost all "diet-related" cancer deaths in humans.
An improved 32P-postlabelling assay for detection and quantitation of styrene 7,8-oxide-DNA adducts

Using DNA modified with [7-3H]styrene 7,8-oxide (SO) in vitro we have standardized the 32P-postlabelling assay for detecting SO-DNA adducts. Nuclease P 1-enriched adducts were 32P-labelled and purified by high-salt ( 4.0 M ammonium formate, pH 6.1} C1s reverse-phase TLC. After elution from the layer with 2-butoxyethanol:H20 (4:6), adducts were separated by two-dimensional PEI cellulose TLC in non-urea solvents (2.0 M ammonium formate, pH 3.5, and 2.7 M sodium phosphate, pH 5.6). One major, three minor and several trace adducts were detected. The efficiency of the kinase reaction depended on the ATP concentration. Use of standard labelling conditions (['Y· 32P]ATP, <3000 Ci/mmol; <2 Mikromol) resulted in poor ( 4-7%) adduct recovery. An ATP concentration of 40 Mikromol, however, increased the labeJling efficiency by a factor of 5-8 (35-55% based on 3H-SO labelied DNA). The results indicate that the new separation technique is suitable for the relatively polar SO-DNA adducts and that high labelling efficiency can be achieved.
Investigations were carried out on the adhesion of cloned S-fimbriated E. coli, labelled with fluoresceinisothiocyanate (FITC) to human buccal epithelial cells. Fluorescence microscopic analysis revealed binding of bacteria to 75-95% of epithelial cells. Inhibition experiments with fetuin, a 1-acid glycoprotein and N-acetyl neuraminic acid confirmed the specificity of bacterial binding to sialoglycoproteins. Further studies using saliva as an inhibitor resulted in a 4-5 times stronger binding inhibition by newborn saliva in comparison to adult saliva coinciding with a 4-5 times higher content of total N-acetyl neuraminic acid in samples of newborn saliva. In Western blot analysis sialoglycoprotein bands with a molecular weight >200 kD reacting with wheat germ agglutinin (WGA), were only identified in samples of newborn saliva. These bands are classified as mucins on account of molecular weight and staining. These data suggest that saliva mucins could represent a major defense mechanism against bacterial infections at a stage of ontogeny where the secretory IgAsystem is not yet developed.
The sattering characteristics ot the n-VI semiconductors were analyzed by a method which combines the second-order finite-element method with the rigorous mode matching procedure. The method avolds the difficulty of solving the complex transcendental equation introduced in the multimode network method and calculates all the eigenvalues and eigenfunctions simultaneously which are needed for the mode matching treatment in the longitudinal direction. As a result, the whole solution procedure is significantly simplified. A comparison is given between the experimental data and the calculated results obtained with this analysis and tbe network method. Very good agreement has been achieved, the accuracy and efficiency of the present method are thus verified.
Polymorphonuclear leukocyte (PMNL) infiltration is an important characteristic in psoriatic lesions. Elevated concentrations of the chemoattractant eicosanoid leukotriene B4 (L TB4) are present in psoriatic skin. Its chemotactic activity is mediated via high affinity receptors on PMNL. The goal of our work was to ascertain whether PMNL infiltration in psoriasis can be accounted for by functional abnormalities of the circulating PMNL due to alterations in the LTB4 receptor density or affinity (or both). No significant difference was found between patients with psoriasis, healthy controls and patients with another inflammatory dermatosis (atopic eczema) with regard to the binding parameters of LTB4 receptors on PMNL. Our findings suggest that PMNL accumulation in psoriatic skin may be the result of an excess of cutaneous hemoattractant rather than the increased readiness of psoriatic PMNL to migrate towards L TB4 due to altered LTB4 receptor density or affinity.
