Die vorliegende Arbeit untersuchte die Veränderungen der wichtigsten hämostatischen Komponenten der plasmatischen Gerinnung in Thrombozyten- und Plasmakonzentraten während der achttägigen Lagerung bei Raumtemperatur. Es wurden keine signifikanten und klinisch relevanten Veränderungen der plasmatischen hämostatischen Kapazität des Plasmas für Fibrinogen, Gerinnungsfaktor XI, XII und XIII, Protein S und C (quantitative Messung), D-Dimere, von Willebrand-Faktor, Kollagenbindungs-aktivität und Ristocetin-Cofaktor nachgewiesen. Es kam jedoch zu einer zeitabhängigen Veränderung während der Lagerung bei PTT, Quick, Thrombinzeit, Gerinnungsfaktoren II, V, VII, VIII, IX, und X, Antithrombin III, Protein S und C (funktionelle Messung), Prothrombinfragmente F1+F2 und APC-Resistenz. Diese Veränderungen fanden sich zudem insbesondere in den gelagerten Thrombozytenkonzentraten. Die wenigen, in der Literatur verfügbaren Untersuchungen zeigen Ergebnisse, die den unseren vergleichbar sind. Erstmal wurden mit dieser Dissertationsarbeit jedoch umfassend alle diese Parameter in einem Ansatz verglichen. Vorausgesetzt, dass hämostaseologische Faktoren auch unter ungünstigen Lagerungsbedingungen weitgehend ihre hämostatische Kapazität behalten, kann bei Infusionen von Früh- oder Neugeborenen praktische Bedeutung erlangen, da die plasmatische hämostatische Kapazität in Thrombozytenkonzentraten nun in den Gesamtbedarf einbezogen werden kann. Ebenso können vereinfachte Lagerbedingungen (Raumtemperatur versus Tiefkühlung) in Krisensituationen enorme logistische Vorteile mit sich bringen. Weitere, insbesondere klinische Transfusionsstudien müssen nun zeigen, ob die ex vivo gewonnenen Erkenntnisse zur Haltbarkeit hämostatischer Gerinnungsfaktoren sich in den klinisch-praktischen Einsatz übertragen lassen. Mit unseren Versuchen wollten wir auch dazu beitragen, zukünftig möglichst schnelle und praktikable Therapievorschläge für Notfälle bereitzustellen. Eine bedeutende Rolle kann das bei Raumtemperatur gelagerten Plasma in der Transfusionsmedizin, Notfallmedizin, bei Operationen und in Kriseneinsätzen spielen. Es bleiben noch viele Fragen zu diesem Thema offen. Diese Arbeit wird zu weiterer Forschung anregen.
Several studies have linked overexpression of the LIM and SH3 domain protein 1 (LASP1) to progression of breast, colon, liver, and bladder cancer. However, its expression pattern and role in human prostate cancer (PCa) remained largely undefined.
Analysis of published microarray data revealed a significant overexpression of LASP1 in PCa metastases compared to parental primary tumors and normal prostate epithelial cells. Subsequent gene-set enrichment analysis comparing LASP1-high and -low PCa identified an association of LASP1 with genes involved in locomotory behavior and chemokine signaling. These bioinformatic predictions were confirmed in vitro as the inducible short hairpin RNA-mediated LASP1 knockdown impaired migration and proliferation in LNCaP prostate cancer cells.
By immunohistochemical staining and semi-quantitative image analysis of whole tissue sections we found an enhanced expression of LASP1 in primary PCa and lymph node metastases over benign prostatic hyperplasia. Strong cytosolic and nuclear LASP1 immunoreactivity correlated with PSA progression. Conversely, qRT-PCR analyses for mir-203, which is a known translational suppressor of LASP1 in matched RNA samples revealed an inverse correlation of LASP1 protein and mir-203 expression. Collectively, our results suggest that loss of mir-203 expression and thus uncontrolled LASP1 overexpression might drive progression of PCa.
Brustkrebs ist, verbunden mit einem Lebenszeitrisiko von 12.5 % daran zu erkranken, die häufigste maligne Erkrankung der Frau in der westlichen Welt. Das LIM- und SH3-Domänen Protein LASP-1 wurde erstmalig 1995 als spezifisch in fokalen Kontakten akkumulierendes Protein in humanen Brustkrebszellen beschrieben. Eine eigene Patientenstudie (n = 83) ergab, dass LASP-1 in 55.4 % der untersuchten invasiven Mammakarzinome eindeutig überexprimiert wird und die LASP-1-Expressionshöhe signifikant positiv mit der Tumorgröße (p < 0.05) und der Wahrscheinlichkeit für Nodalpositivität (p < 0.01) korreliert. Als Actin-bindendes Protein mit ubiquitärer Expression interagiert LASP-1 mittels seiner SH3-Domäne auch mit Zyxin. Zyxin ist ein Signalprotein, das zwischen fokalen Kontakten und dem Zellkern pendelt und an der Regulation von Transcription, Zellzyklus und Zellbewegung beteiligt ist. In dieser Doktorarbeit untersuchte ich die Funktion von LASP-1 in verschiedenen humanen Brustkrebszelllinien und einer humanen Ovarkarzinomzelllinie unter Verwendung der RNA-Interferenz zum sequenzspezifischen knock-down von LASP-1. Die Transfektion der Zellen mit LASP-1-spezifischer small-interfering-RNA (siRNA) senkte die Proteinexpression um ca. 50 - 58 %, was zu einer Arretierung des Zellzyklus in der G2-Phase führte und die Proliferation der Brust- und Eierstockkrebszellen um 30 - 50 % reduzierte. Apoptotische und nekrotische Prozesse wurden dabei durch Caspase-3 Western-blots und Trypan-Blau-Färbungen ausgeschlossen. Außerdem verringerte der knock-down von LASP-1 die Migration der Krebszellen, wohingegen die Überexpression von LASP-1 in PTK2- (Potorous tridactylis kidney) Zellen, welche kein endogenes LASP-1 exprimieren, in einer signifikanten Steigerung der Migration resultierte. Immunfluoreszenzfärbungen zeigten, dass der knock-down von LASP-1 von einer verringerten Konzentration seines Interaktionspartners Zyxin an fokalen Kontakten begleitet wird, ohne dabei das Actin-Zytoskelett oder die Morphologie der fokalen Kontakte zu verändern. Die Transfektion der Brust- und Eierstockkrebszellen mit einer Kontroll-siRNA (scrambled Neuropilin1) beeinflusste hingegen keinen der genannten Parameter. Diese Daten unterstreichen die wichtige Rolle von LASP-1 für die Tumorzellproliferation und -migration. LASP-1 könnte daher in Interaktion mit Zyxin an der molekularen Karzinogenese von Brust- und Eierstockkrebs beteiligt sein und in der Zukunft als viel versprechender Prognosemarker dienen.
LASP-1, das LIM und SH3 Protein 1, ist ein Aktin-bindendes Gerüstprotein, das in verschiedenen Tumorentitäten überexprimiert ist. Dabei scheint LASP-1 eine wichtig Rolle sowohl bei der Proliferation und Migration von Zellen als auch bei der Tumorgenese und Metastasierung zu spielen.
Ziel dieser Arbeit war es, die Expression von LASP-1 im Urothelkarzinom der Harnblase zu untersuchen und eine daraus abzuleitende klinische Relevanz für die Diagnostik zu evaluieren. Dazu wurden histologische Blasenschnitte immunhistochemisch nach LASP-1 gefärbt und Western Blot-Analysen von Urinproben durchgeführt.
Die Auswertung der immunhistochemisch gefärbten Blasenschnitte ergab, dass Urothelkarzinome signifikant mehr LASP-1 auf Proteinebene exprimieren als gesundes Blasengewebe. Allerdings konnte keine Korrelation zwischen der Stärke der LASP-1-Expression und verschiedener klinisch-pathologischer Parameter nachgewiesen werden.
Mittels Western Blot-Analysen gelang es, LASP-1 eindeutig im Urin und statistisch signifikant häufiger bei Blasenkarzinompatienten zu detektieren. Ohne Berücksichtigung einer Kontamination mit LASP-1-positiven Blut- und Entzündungszellen ist der LASP-1-Nachweis im Western Blot mit einer Gesamtsensitivität von 84,2% derzeit sensitiver als die meisten erhältlichen Tumormarker. Darüber hinaus ergab der Vergleich von Spontanurin und von Harnblasenspülflüssigkeit, dass Spontanurinproben sogar geeigneter zur Diagnostik zu sein scheinen.
Abschließend kann zusammengefasst werden, dass LASP-1 aufgrund der einfachen, nicht invasiven und kostengünstigen Probengewinnung zusammen mit den hohen Werten für Sensitivität und Spezifität als Urin-basierter Tumormarker für das Urothelkarzinom der Harnblase vielversprechend zu sein scheint.
A central question to biology is how pathogenic bacteria initiate acute or chronic infections. Here we describe a genetic program for cell-fate decision in the opportunistic human pathogen Staphylococcus aureus, which generates the phenotypic bifurcation of the cells into two genetically identical but different cell types during the course of an infection. Whereas one cell type promotes the formation of biofilms that contribute to chronic infections, the second type is planktonic and produces the toxins that contribute to acute bacteremia. We identified a bimodal switch in the agr quorum sensing system that antagonistically regulates the differentiation of these two physiologically distinct cell types. We found that extracellular signals affect the behavior of the agr bimodal switch and modify the size of the specialized subpopulations in specific colonization niches. For instance, magnesium-enriched colonization niches causes magnesium binding to S. aureusteichoic acids and increases bacterial cell wall rigidity. This signal triggers a genetic program that ultimately downregulates the agr bimodal switch. Colonization niches with different magnesium concentrations influence the bimodal system activity, which defines a distinct ratio between these subpopulations; this in turn leads to distinct infection outcomes in vitro and in an in vivo murine infection model. Cell differentiation generates physiological heterogeneity in clonal bacterial infections and helps to determine the distinct infection types.
The mammalian Vasodilator Stimulated Phosphoprotein (VASP) is a founding member of the Ena/VASP family of proteins that includes Drosophila Enabled (ena), the mammalian Ena homologue (Mena) and the Ena-VASP-like protein (Evl). VASP was initially discovered and characterized as a substrate for cGMP- and cAMP-dependent protein kinases (cGKs and cAKs). Ena/VASP proteins are involved in Actin-filament formation, plasma membrane protrusion, acceleration of Actin-based motility of Listeria and the establishment of cell-cell adhesion. Moreover, Ena/VASP proteins have been implicated as inhibitory factors in repulsive axon guidance and inhibition of plasma membrane activity and random motility in fibroblast. In order to study the physiological function of VASP, VASP-deficient mice had been generated in the laboratory by homologous recombination. VASP-/- mice showed hyperplasia of megakaryocytes in the bone marrow and spleen and a two-fold increase in thrombin- and collagen-induced platelet activation. To further investigate the cellular function of VASP, I established cardiac fibroblast cell lines derived from both wild type and VASP-/- mice. Both cell lines presented similar growth rates and normal contact dependent-growth inhibition but showed differences in morphology, migration and adhesion. Adherent VASP-/- cells, despite normal Mena and Evl expression levels, were highly spread. VASP-/- cells covered about twice the substrate surface area as wild type cells, while the cell volumes were unchanged. This shape difference suggests that VASP is involved in the regulation of spreading. Since the small GTPases Rac and Cdc 42 and their effector p21-activated kinase (Pak) are key regulators of lamellipodia formation and cell spreading, I analyzed this signalling pathway in VASP-/- cells stimulated with Platelet Derived Growth Factor-BB (PDGF-BB) or fetal calf serum. In wild type cells Rac and Pak were rapidly and transiently activated by PDGF or serum; however, in the absence of VASP both Rac and Pak activation was dramatically prolonged. The Rac/Pak pathway is known to play an essential role in cell motility. VASP deficient cells showed compromised migration and reorientation in a wound healing assay, probably due to enhanced Rac activity. The spreading phenotype, compromised migration and the effect observed on the Rac and Pak activities were reverted in VASP-/- cells stably transfected with full lenght human VASP, indicating a VASP dependent modulation of the Rac/Pak pathway and Rac/Pak regulated processes. Moreover, adhesion and detachment of VASP-deficient cells were significantly slower when compared to wild type cells. Preincubation of VASP+/+ cells with a cGMP analog accelerated adhesion. This acceleration did not take place in the VASP-/- cells, suggesting a VASP dependent effect. The second part of this work focused on VASP function in platelets. On the one hand I investigated the possibility of VASP-dependent Rac regulation in mouse platelets. Murine platelets are a good model for studying Rac regulation since they express high levels of VASP but not Mena/Evl and since VASP-deficient platelets show an increased platelet activation. Rac was activated by platelet agonists which was inhibited by preincubation with cGMP and cAMP analogs. Initial results which need to be extended showed that the cGMPcaused inhibition of Rac activation was VASP-dependent. Finally, in vivo platelet adhesion (platelet-vessel wall interactions) was studied using VASP-deficient mice. These studies demonstrated in-vivo that VASP down regulates platelet adhesion to the vascular wall under both physiological and pathophysiological conditions.
Background
Direct interaction between Red blood cells (RBCs) and platelets is known for a long time. The bleeding time is prolonged in anemic patients independent of their platelet count and could be corrected by transfusion of RBCs, which indicates that RBCs play an important role in hemostasis and platelet activation. However, in the last few years, opposing mechanisms of platelet inhibition by RBCs derived nitric oxide (NO) were proposed. The aim of our study was to identify whether RBCs could produce NO and activate soluble guanylate cyclase (sGC) in platelets.
Methods
To test whether RBCs could activate sGC under different conditions (whole blood, under hypoxia, or even loaded with NO), we used our well-established and highly sensitive models of NO-dependent sGC activation in platelets and activation of purified sGC. The activation of sGC was monitored by detecting the phosphorylation of Vasodilator Stimulated Phosphoprotein (VASPS239) by flow cytometry and Western blot. ANOVA followed by Bonferroni’s test and Student’s t-test were used as appropriate.
Results
We show that in the whole blood, RBCs prevent NO-mediated inhibition of ADP and TRAP6-induced platelet activation. Likewise, coincubation of RBCs with platelets results in strong inhibition of NO-induced sGC activation. Under hypoxic conditions, incubation of RBCs with NO donor leads to Hb-NO formation which inhibits sGC activation in platelets. Similarly, RBCs inhibit activation of purified sGC, even under conditions optimal for RBC-mediated generation of NO from nitrite.
Conclusions
All our experiments demonstrate that RBCs act as strong NO scavengers and prevent NO-mediated inhibition of activated platelets. In all tested conditions, RBCs were not able to activate platelet or purified sGC.
Das Vasodilatator stimulierte Phosphoprotein (VASP) ist ein Zytoskelett-assoziiertes Protein der Ena (Enabled)/VASP-Proteinfamilie. Seine Funktionen bezüglich Aktin- Polymerisation, Thrombozyten-Aggregation, Wachstumskegel-Führungsprozessen und Motilität sowohl von Zellen als auch von Listerien sind bisher nur unvollständig charakterisiert. In dieser Arbeit konnte ich zeigen, wie die VASP-F-Aktin Interaktion in vitro durch die Phosphorylierung zweier Aminosäuren von VASP reguliert wird. Transfektions-Experimente mit VASP und RhoA deuten eine mögliche Beteiligung von VASP im Signalweg von RhoA an. Zudem führen Überexpression und Deletion von VASP in Zellen zu demselben Stressfaser-Phänotyp, der unabhängig vom stimulierenden Einfluss von Serum ist. In VASPdefizienten Fibroblasten ist außerdem die Membranrigidität und die Phosphorylierung der leichten Kette des Myosins erhöht, was auf ein stabileres und stärker kontrahiertes Aktin- Zytoskelett in diesen Zellen schließen lässt. Die Regulation und Organisation des Aktin- Zytoskeletts beeinflusst auch die zelluläre Adhäsion, die in VASP-defizienten Zellen verändert ist. VASP-defiziente Zellen adhärieren signifikant stärker an Fibronektinbeschichtete Perlen als Wildtyp-Zellen. Der Widerstand dieser Perlen gegenüber mechanischen Kräften ist in VASP (-/-) Zellen signifikant erhöht. Dieser Unterschied beruht in erster Linie auf dem verstärkten Aktin-Zytoskelett in diesen Zellen und ist unabhängig von Mikrotubuli. Messungen mit rekonstituierten Zelllinien zeigen zudem eine VASPAbhängigkeit dieses Effekts. Der GTPase Rap1 kommt eine wichtige Bedeutung bei der Integrin-abhängigen Adhäsion von Zellen zu. Aktivierung von Epac, einem Guanin-Nukleotid- Austauschfaktor von Rap1, führt in Wildtyp-Zellen zur Verstärkung des Widerstandes gegenüber mechanischen Kräften, die auf Fibronektin-beschichtete Perlen wirken, ohne dabei die Membranrigidität zu verändern. Diese Kraft-Verstärkung wird weder vom Aktin- noch vom Mikrotubuli-Zytoskelett beeinflusst. Es exisitiert daher ein Mechanismus, bei dem die Länge von elastischen Membranfortsätzen unabhängig von der Membranfestigkeit und dem Aktin-Zytoskelett reguliert wird. Diese Experimente zeigen, dass VASP eine wichtige Rolle bei der Stabilität und Kontraktilität des Aktin-Zytoskeletts, der Membranrigidität sowie bei der zellulären Adhäsion spielt. Die vorliegenden Ergebnisse weisen darauf hin, dass hierbei weniger die direkte Interaktion von VASP mit dem Aktin-Zytoskelett von Bedeutung ist, sondern viel mehr seine mögliche Funktion als Gerüstprotein (Scaffold), das eine geregelte Signaltransduktion organisiert.
In der vorliegenden Arbeit ging es darum, die inhibierende Wirkung von extrazellulär zugefügten GMP-Derivaten auf die Thrombozytenaktivierung nachzuweisen. Anhand verschiedener thrombozytärer Aktivierungsmarker wie ERK, der Expression von P-Selektin und intrazellulärem Kalziumeinstrom zeigte sich eine signifikante Inhibition der Thrombozytenaktivierung durch zyklische GMP-Analoga. Neu ist, dass auch nicht-zyklische GMP-Derivate eindeutig einen hemmenden Effekt aufweisen. Guanosin-Derivate alleine führen dagegen weder zu einer Hemmung noch zu einer vermehrten Stimulation der Plättchenaktivierung. Die Wirkung der GMP-Analoga scheint von der negativen Phosphatgruppe abhängig zu sein. Der frühe Zeitpunkt der Inhibition und die Tatsache, dass auch Hemmer der PKG einen inhibierenden Effekt auf die Thrombozytenaktierung aufweisen, führen zu der Hypothese, dass es sich um PGK-unabhängige Effekte handelt. Der Thrombinrezeptor als Wirkort der GMP-Analoga konnte in Biacore-Messungen ausgeschlossen werden. Es gilt nun, speziell den Thromboxanrezeptor auf mögliche Interaktionen mit GMP-Derivaten hin zu untersuchen.
Chronic myeloid leukemia (CML) is characterized by a genomic translocation generating a permanently active BCR-ABL oncogene with a complex pattern of atypically tyrosine-phosphorylated proteins that drive the malignant phenotype of CML. Recently, the LIM and SH3 domain protein 1 (LASP1) was identified as a component of a six gene signature that is strongly predictive for disease progression and relapse in CML patients. However, the underlying mechanisms why LASP1 expression correlates with dismal outcome remained unresolved.
Here, we identified LASP1 as a novel and overexpressed direct substrate of BCR-ABL in CML. We demonstrate that LASP1 is specifically phosphorylated by BCR-ABL at tyrosine-171 in CML patients, which is abolished by tyrosine kinase inhibitor therapy. Further studies revealed that LASP1 phosphorylation results in an association with CRKL - another specific BCR-ABL substrate and bona fide biomarker for BCR-ABL activity. pLASP1-Y171 binds to non-phosphorylated CRKL at its SH2 domain. Accordingly, the BCR-ABL-mediated pathophysiological hyper-phosphorylation of LASP1 in CML disrupts normal regulation of CRKL and LASP1, which likely has implications on downstream BCR-ABL signaling. Collectively, our results suggest that LASP1 phosphorylation might serve as an additional candidate biomarker for assessment of BCR-ABL activity and provide a first step toward a molecular understanding of LASP1 function in CML.
