Fertilität und Macht: Die Reproduktionspflicht mittelalterlicher Herrscherinnen und Herrscher
(2021)
Fertility was a key theme of medieval rulership. To conceive and give birth to sons – and thus to ensure the succession to the throne – was one of the foremost duties of medieval kings and queens. But what happened when a male child died in infancy, no male child was born, or no pregnancy ever came about? Barrenness could have dramatic consequences in the Middle Ages, for example expulsion, divorce or conflicts over royal succession. Against this historical background, it seems logical that the fate of the childless ruler would be recounted in the form of a ‘Passion narrative’. In the German literature of the Middle Ages, however, there are also interpretative models of a contrary vein to be found. In the year 1220, for instance, Ebernand of Erfurt construed the wedded life of the imperial couple Henry and Kunigunde as a tale of resistance against the royal obligation to reproduce. In his versified legend, composed in the vernacular, the couple secretly agrees not to fulfil society’s expectations, but to lead a chaste marriage. Yet above and beyond legend, childless rulers were also subject to the impact of multifarious legal, religious, medical, narrative and discursive factors. Taking Michel Foucault as a point of departure, this contribution shows how fertility became a field of power on which hierarchies between rulers and subjects, men and women were negotiated, while also shedding light on how religious and secular ideals clashed in the assessment of infertility.
Propagule pressure and an invasion syndrome determine invasion success in a plant community model
(2021)
The success of species invasions depends on multiple factors, including propagule pressure, disturbance, productivity, and the traits of native and non-native species. While the importance of many of these determinants has already been investigated in relative isolation, they are rarely studied in combination. Here, we address this shortcoming by exploring the effect of the above-listed factors on the success of invasions using an individual-based mechanistic model. This approach enables us to explicitly control environmental factors (temperature as surrogate for productivity, disturbance, and propagule pressure) as well as to monitor whole-community trait distributions of environmental adaptation, mass, and dispersal abilities. We simulated introductions of plant individuals to an oceanic island to assess which factors and species traits contribute to invasion success. We found that the most influential factors were higher propagule pressure and a particular set of traits. This invasion trait syndrome was characterized by a relative similarity in functional traits of invasive to native species, while invasive species had on average higher environmental adaptation, higher body mass, and increased dispersal distances, that is, had greater competitive and dispersive abilities. Our results highlight the importance in management practice of reducing the import of alien species, especially those that display this trait syndrome and come from similar habitats as those being managed.
Aspergillus fumigatus causes life-threatening opportunistic infections in immunocompromised patients. As therapeutic outcomes of invasive aspergillosis (IA) are often unsatisfactory, the development of targeted immunotherapy remains an important goal. Linking the innate and adaptive immune system, dendritic cells are pivotal in anti-Aspergillus defense and have generated interest as a potential immunotherapeutic approach in IA. While monocyte-derived dendritic cells (moDCs) require ex vivo differentiation, antigen-pulsed primary myeloid dendritic cells (mDCs) may present a more immediate platform for immunotherapy. To that end, we compared the response patterns and cellular interactions of human primary mDCs and moDCs pulsed with an A. fumigatus lysate and two A. fumigatus proteins (CcpA and Shm2) in a serum-free, GMP-compliant medium. CcpA and Shm2 triggered significant upregulation of maturation markers in mDCs and, to a lesser extent, moDCs. Furthermore, both A. fumigatus proteins elicited the release of an array of key pro-inflammatory cytokines including TNF-α, IL-1β, IL-6, IL-8, and CCL3 from both DC populations. Compared to moDCs, CcpA- and Shm2-pulsed mDCs exhibited greater expression of MHC class II antigens and stimulated stronger proliferation and IFN-γ secretion from autologous CD4\(^+\) and CD8\(^+\) T-cells. Moreover, supernatants of CcpA- and Shm2-pulsed mDCs significantly enhanced the oxidative burst in allogeneic neutrophils co-cultured with A. fumigatus germ tubes. Taken together, our in vitro data suggest that ex vivo CcpA- and Shm2-pulsed primary mDCs have the potential to be developed into an immunotherapeutic approach to tackle IA.
Noctes Neolatinae 38, Hildesheim/Zürich/New York 2020.“
Objective: Substantia nigra hyperechogenicity is found in children with attention- deficit hyperactivity disorder (ADHD). Research with transcranial sonography (TCS) in adults suggests that echogenic alterations are linked to subclinical behavioral deficits and that brain iron homeostasis is involved in the signal genesis. The purpose of this study was to explore substantia nigra echogenicity in healthy children, to assess age-related changes and to investigate whether echogenic signals relate to subclinical alterations in behavior. Furthermore, associations of central nigral neuromelanin measures and peripheral serum iron parameters to echogenic signals of the substantia nigra were evaluated. Methods: In a multimodal study design, neuroimaging of the substantia nigra was conducted with TCS and neuromelanin-sensitive magnetic resonance imaging (MRI) in 28 healthy children (8 − 12 years). Correlations and multiple regression analyses determined associations between the neuroimaging methods, behavioral data from Strength and Difficulties Questionnaire (SDQ) and serum iron-related parameters. Results: Substantia nigra echogenicity correlated inversely with hyperactivity ratings in healthy, non-ADHD children (r = −.602, p = .001). Echogenic sizes did not change as a function of age. Neuromelanin-sensitive MRI measures of the substantia nigra and peripheral serum iron parameters were not associated with nigral TCS signals. Conclusion: In healthy children behavioral differences in hyperactive tendencies are associated with differences in substantia nigra echogenicity. This could help to identify those children who are at risk of subclinical ADHD.
The IE languages developed different strategies for the encoding of the passive function. In some language branches, the middle voice extended to the passive function to varying extents. In addition, dedicated derivational formations arose in a number of languages, such as the Greek -ē-/-thē- aorist and the Indo-Aryan -ya-presents. Periphrastic formations involving a verbal adjective or a participle are also widely attested, and played an important role in the building of the passive paradigm in e.g. Romance and Germanic languages. As the periphrastic passive is also attested in Hittite alongside passive use of the middle, both strategies seem to be equally ancient. Some minor strategies include lexical passives and the extensive lability of verbs. A survey of possible strategies provides evidence for the rise of a disparate number of morphemes and constructions, and for their ongoing incorporation into the inflectional paradigms (paradigmaticization) of given languages, thus adding to our knowledge about cross-linguistic sources of passive morphology and grammaticalization processes involved.
The Immune system exerts its response against invading pathogens via a cumulative, sequential cooperation of immune cells coordinated by their secreted products. Immune cells, such as macrophages and dendritic cells (DCs), express toll-like receptors (TLRs) to sense the presence of pathogens through pathogen-associated molecular patterns (PAMPs). The interaction of PAMPs with TLRs elicits a cytosolic signaling cascade that enhances the expression of genes to stimulate inflammation. Interleukin 1 receptor-associated kinase 2 (IRAK2) is a component of the TLR signaling pathway. IRAK2 transduces the TLR signal via a direct interaction with TNF receptor-associated factor 6 (TRAF6) and subsequent enhancement of its ubiquitination.
During my PhD thesis, I determined that a 55-amino acid long stretch at the C-terminal end of IRAK2 is important for TLR signaling. Overexpression of C-terminal truncated IRAK2 (IRAK2Δ55) in the murine macrophage cell line RAW 264.7 led to impaired CD40 expression after TLR4 stimulation by Lipopolysaccharide (LPS). I observed attenuated competency of IRAK2Δ55 in restoring a full TLR signaling response i.e. IL-6 secretion, NO production and CD40 expression in IRAK2-deficient RAW cells generated via CRISPR-Cas9 approach. Additionally, diminished TLR4 induced activation of nuclear factor κB (NF-κB) and extracellular signal related kinase (ERK) was observed with IRAK2Δ55 reconstituted RAW cells as compared to cell reconstituted with wildtype IRAK2.
IRAK2Δ55 reconstituted RAW cells also exhibited reduced TLR4-induced cell death and phosphorylation of receptor interacting protein kinase 3 (RIP3). Co-immunoprecipitation experiments in HEK 293T cells showed that IRAK2Δ55 was still able to bind to TRAF6 alike IRAK2 but failed to induce ubiquitination of TRAF6. In conclusion, the results suggest that the IRAK2 TRAF6 interaction is not sufficient to sustain full TLR signaling. An C-terminus-dependent unknown molecular mechanism is also involved.
Through my PhD work, I also analyzed a B cell lineage-specific HECTD1 knock-out mice. HECTD1 is an E3 ubiquitin ligase for various substrate proteins, such as heat shock protein 90 (HSP90), adenomatous polyposis coli and phosphatidylinositol phosphate kinase type 1 γ. Hsp90 regulates a variety of signaling molecules in NF-κB activation pathways which are essential for an optimal B cell response.
HECTD1-deficient pro-B cells developed normally into mature B cells. However, TLR4 stimulated HECTD1-deficient B cells displayed reduced immunoglobulin (Ig) production in in vitro cultures. In addition, mice with HECTD1-deficient B cells showed a diminished Ig response after nitrophenylacetyl-keyhole limpet hemocyanin immunization. Thus, HECTD1 is necessary for efficient Ig secretion.

Within the relatively new area of research on Third Language (L3) Acquisition, the subfield of phonology is growing, but still relatively understudied. Testing the current L3 models adopted from research on L3 syntax (see Rothman 2010, Bardel & Falk 2012, Flynn et al. 2004), the studies conducted in the area have mostly focused on the source and directionality of language transfer – both into the L3 and into the respective background languages – with some recent excursions into the role of extra-linguistic factors for multilingual learners (e.g., Wrembel 2015). The findings so far (mostly on production, with perception lagging behind) have been very diverse and, depending on the concrete study, can often be taken to give evidence for any of the prevalent models. This can be attributed to the wide range of different speaker and learner biographies as well as their language combinations and state of acquisition, but crucially the dilemma seems to be inherent in the (phonological) system in and of itself since viewing phonological interlanguage transfer as a one-dimensional and immediately transparent process based on direct correspondences between language systems does not seem to capture the complex nature of the phenomenon.
In this doctoral thesis I investigate the acquisition of an additional phonological system by child and adult German heritage speakers of Turkish. Specifically, I explore how the learners deal with diverse phonological contrasts that promote positive contra negative transfer from their HL (Turkish) and their L2 (German), and how their perception and production is modulated by cognitive and affective variables. Moreover, I test contrasts that can be found neither in the HL nor in the L2 phonological system.
The studies will shed light both on the question of how a new language is shaped and affected by different existing systems and on how two or more phonological grammars co-exist and/or interact in a speaker’s mind. I will argue that, rather than being regarded as simple full projection of language-specific property sets onto the target language, phonological transfer in multilinguals needs to be considered as a process of complex interactions and layers that are established on the level of individual phonological properties and abstract (typological) associations.
Organoboron compounds are important building blocks in organic synthesis, materials science, and drug discovery. The development of practical and convenient ways to synthesize boronate esters attracted significant interest. Photoinduced borylations originated with stoichiometric reactions of arenes and alkanes with well-defined metal-boryl complexes. Now photoredox-initiated borylations, catalyzed either by transition-metal or organic photocatalysts, and photochemical borylations with high efficiency have become a burgeoning area of research. In this chapter, we summarize research in the field of photocatalytic C-X borylation, especially emphasizing recent developments and trends, based on transition-metal catalysis, metal-free organocatalysis and direct photochemical activation. We focus on reaction mechanisms involving single electron transfer (SET), triplet energy transfer (TET), and other radical processes.
We developed a highly selective photocatalytic C-F borylation method that employs a rhodium biphenyl complex as a triplet sensitizer and the nickel catalyst [Ni(IMes)2] (IMes = 1,3-dimesitylimidazolin-2-ylidene) for the C-F bond activation and defluoroborylation process. This tandem catalyst system operates with visible (400 nm) light and achieves borylation of a wide range of fluoroarenes with B2pin2 at room temperature in excellent yields and with high selectivity. Direct irradiation of the intermediary C-F bond oxidative addition product trans-[NiF(ArF)(IMes)2] leads to fast decomposition when B2pin2 is present. This destructive pathway can be bypassed by indirect excitation of the triplet states of the nickel(II) complex via the photoexcited rhodium biphenyl complex. Mechanistic studies suggest that the exceptionally long-lived triplet excited state of the Rh biphenyl complex used as the photosensitizer allows for efficient triplet energy transfer to trans-[NiF(ArF)(IMes)2], which leads to dissociation of one of the NHC ligands. This contrasts with the majority of current photocatalytic transformations, which employ transition metals as excited state single electron transfer agents. We have previously reported that C(arene)-F bond activation with [Ni(IMes)2] is facile at room temperature, but that the transmetalation step with B2pin2 is associated with a high energy barrier. Thus, this triplet energy transfer ultimately leads to a greatly enhanced rate constant for the transmetalation step and thus for the whole borylation process. While addition of a fluoride source such as CsF enhances the yield, it is not absolutely required. We attribute this yield-enhancing effect to (i) formation of an anionic adduct of B2pin2, i.e. FB2pin2-, as an efficient, much more nucleophilic {Bpin-} transfer reagent for the borylation/transmetalation process, and/or (ii) trapping of the Lewis acidic side product FBpin by formation of [F2Bpin]- to avoid the formation of a significant amount of NHC-FBpin and consequently of decomposition of {Ni(NHC)2} species in the reaction mixture.
We reported a highly selective and general photo-induced C-Cl borylation protocol that employs [Ni(IMes)2] (IMes = 1,3-dimesitylimidazoline-2-ylidene) for the radical borylation of chloroarenes. This photo-induced system operates with visible light (400 nm) and achieves borylation of a wide range of chloroarenes with B2pin2 at room temperature in excellent yields and with high selectivity, thereby demonstrating its broad utility and functional group tolerance. Mechanistic investigations suggest that the borylation reactions proceed via a radical process. EPR studies demonstrate that [Ni(IMes)2] undergoes very fast chlorine atom abstraction from aryl chlorides to give [NiI(IMes)2Cl] and aryl radicals. Control experiments indicate that light promotes the reaction of [NiI(IMes)2Cl] with aryl chlorides generating additional aryl radicals and [NiII(IMes)2Cl2]. The aryl radicals react with an anionic sp2-sp3 diborane [B2pin2(OMe)]- formed from B2pin2 and KOMe to yield the corresponding borylation product and the [Bpin(OMe)]•- radical anion, which reduces [NiII(IMes)2Cl2] under irradiation to regenerate [NiI(IMes)2Cl] and [Ni(IMes)2] for the next catalytic cycle.
A highly efficient and general protocol for traceless, directed C3-selective C-H borylation of indoles with [Ni(IMes)2] as the catalyst was achieved. Activation and borylation of N-H bonds by [Ni(IMes)2] is essential to install a Bpin moiety at the N-position as a traceless directing group, which enables the C3-selective borylation of C-H bonds. The N-Bpin group which is formed is easily converted in situ back to an N-H group by the oxidiative addition product of [Ni(IMes)2] and in situ-generated HBpin. The catalytic reactions are operationally simple, allowing borylation of of a variety of substituted indoles with B2pin2 in excellent yields and with high selectivity. The C-H borylation can be followed by Suzuki-Miyaura cross-coupling of the C-borylated indoles in an overall two-step, one-pot process providing an efficient method for synthesizing C3-functionalized heteroarenes.
