Growth and Differentiation Factor 5 (GDF5) is a secreted growth factor that belongs to the Bone Morphogenetic Protein (BMP) family and plays a pivotal role during limb development. GDF5 is a susceptibility gene for osteoarthritis (OA) and mutations in GDF5 are associated with a wide variety of skeletal malformations ranging from complex syndromes such as acromesomelic chondrodysplasias to isolated forms of brachydactylies or multiple synostoses syndrome 2 (SYNS2). Here, we report on a family with an autosomal dominant inherited combination of SYNS2 and additional brachydactyly type A1 (BDA1) caused by a single point mutation in GDF5 (p.W414R). Functional studies, including chondrogenesis assays with primary mesenchymal cells, luciferase reporter gene assays and Surface Plasmon Resonance analysis, of the GDF5 W-414R variant in comparison to other GDF5 mutations associated with isolated BDA1 (p.R399C) or SYNS2 (p.E491K) revealed a dual pathomechanism characterized by a gain-and loss-of-function at the same time. On the one hand insensitivity to the main GDF5 antagonist NOGGIN (NOG) leads to a GDF5 gain of function and subsequent SYNS2 phenotype. Whereas on the other hand, a reduced signaling activity, specifically via the BMP receptor type IA (BMPR1A), is likely responsible for the BDA1 phenotype. These results demonstrate that one mutation in the overlapping interface of antagonist and receptor binding site in GDF5 can lead to a GDF5 variant with pathophysiological relevance for both, BDA1 and SYNS2 development. Consequently, our study assembles another part of the molecular puzzle of how loss and gain of function mutations in GDF5 affect bone development in hands and feet resulting in specific types of brachydactyly and SYNS2. These novel insights into the biology of GDF5 might also provide further clues on the pathophysiology of OA.
Die Pseudodistomine gehören zu den ersten Piperidinalkaloiden marinen Ursprungs, die 1987 von Ishibashi et al. aus der Tunikate (Ascidie) Pseudodistoma kanoko isoliert wurden. Aus der gleichen Tunikate wurde 1995 das Pseudodistomin C isoliert. Die amphiphilen Piperidinalkaloide zeigen eine Antitumor-Aktivität gegen bestimmte Mäuseleukämiezellen, wobei Pseudodistomin C auch eine Cytotoxizität gegen menschliche HeLa-abgeleitete Krebszellen KB aufweist. In der Einleitung wird ausführlich auf Vorkommen, Struktur, Biogenese, pharmakologische Perspektiven und literaturbekannten Synthesen dieser amphiphilen Piperidin-Alkaloide eingegangen. Im Hauptteil wird zunächst eine gescheiterte Synthese ausgehend von D-Ribose über das Konzept einer Tandem Wittig-[3+2]-Cycloaddition beschrieben. Daraufhin wird ein völlig neuer Syntheseweg vorgestellt, welcher den formalen Aufbau des Pseudodistomin C über einen bekannten Piperidin-Grundkörper ermöglich. Des weiteren konnte das vollständig geschützte Pseudodistomin E synthetisiert werden.
Pierre Corneille (1606-1684) gehört neben Racine (1639-1699) und Molière (1622-1673) zu den drei großen Autoren des klassischen französischen Theaters. Seine erste Tragödie "Médée" wurde von Publikum und Kritikern zurückgewiesen. Corneille hatte seinen typischen Stil noch nicht entwickelt: Das Drama oszilliert zwischen Komödie und Tragödie, ist einerseits dem Handlungstheater des Barock verpflichtet und wahrt andererseits die Einheiten der "doctrine classique". Der französische Dramatiker rezipiert in seinem Stück den griechischen Tragiker Euripides (480-406 v. Chr.) und den römischen Autor Seneca (1-65 n. Chr.). Gerade den Stil Senecas ahmt er nach. Dieses Buch soll einen Beitrag zum besseren Verständnis der französischen Klassik leisten: Zwar spielt die direkte Rezeption der senecanischen Tragödie eine wichtige Rolle für das klassische französische Drama, doch Form und dramatische Technik resultierten vor allem aus der Beschäftigung mit antiken Poetiken.
The treatment landscape in multiple myeloma (MM) is shifting from genotoxic drugs to immunotherapies. Monoclonal antibodies, immunoconjugates, T-cell engaging antibodies and CART cells have been incorporated into routine treatment algorithms, resulting in improved response rates. Nevertheless, patients continue to relapse and the underlying mechanisms of resistance remain poorly understood. While Impaired death receptor signaling has been reported to mediate resistance to CART in acute lymphoblastic leukemia, this mechanism yet remains to be elucidated in context of novel immunotherapies for MM. Here, we describe impaired death receptor signaling as a novel mechanism of resistance to T-cell mediated immunotherapies in MM. This resistance seems exclusive to novel immunotherapies while sensitivity to conventional anti-tumor therapies being preserved in vitro. As a proof of concept, we present a confirmatory clinical case indicating that the FADD/BID axis is required for meaningful responses to novel immunotherapies thus we report impaired death receptor signaling as a novel resistance mechanism to T-cell mediated immunotherapy in MM.
Georg Sticker war Arzt, Hochschullehrer, Hygieniker und Medizinhistoriker. Das Themenspektrum seiner Arbeiten ist sehr umfangreich. Der Schwerpunkt dieser Arbeit liegt auf seinen Hauptforschungsgebieten, der Medizingeschichte und hier vor allem auf der Seuchengeschichte zu Pest, Lepra und Syphilis und der Bedeutung der Geschichte der Epidemien für die Epidemiologie; sowie seinen großen ärztlichen Vorbildern Hippokrates und Paracelsus. Auch über Naturheilkunst und Gesundheit und Erziehung berichtet er mit großem Engagement.Nach seiner Berufung an die Universität Würzburg beendet er seine ärztliche Tätigkeit, gründet hier im Jahr 1921 das Institut für Geschichte der Medizin und widmet sich ganz der Medizingeschichte

Introduction:
Evidence from a number of open-label, uncontrolled studies has suggested that rituximab may benefit patients with autoimmune diseases who are refractory to standard-of-care. The objective of this study was to evaluate the safety and clinical outcomes of rituximab in several standard-of-care-refractory autoimmune diseases (within rheumatology, nephrology, dermatology and neurology) other than rheumatoid arthritis or non-Hodgkin’s lymphoma in a real-life clinical setting.
Methods:
Patients who received rituximab having shown an inadequate response to standard-of-care had their safety and clinical outcomes data retrospectively analysed as part of the German Registry of Autoimmune Diseases. The main outcome measures were safety and clinical response, as judged at the discretion of the investigators.
Results:
A total of 370 patients (299 patient-years) with various autoimmune diseases (23.0% with systemic lupus erythematosus, 15.7% antineutrophil cytoplasmic antibody-associated granulomatous vasculitides, 15.1% multiple sclerosis and 10.0% pemphigus) from 42 centres received a mean dose of 2,440 mg of rituximab over a median (range) of 194 (180 to 1,407) days. The overall rate of serious infections was 5.3 per 100 patient-years during rituximab therapy. Opportunistic infections were infrequent across the whole study population, and mostly occurred in patients with systemic lupus erythematosus. There were 11 deaths (3.0% of patients) after rituximab treatment (mean 11.6 months after first infusion, range 0.8 to 31.3 months), with most of the deaths caused by infections. Overall (n = 293), 13.3% of patients showed no response, 45.1% showed a partial response and 41.6% showed a complete response. Responses were also reflected by reduced use of glucocorticoids and various immunosuppressives during rituximab therapy and follow-up compared with before rituximab. Rituximab generally had a positive effect on patient well-being (physician’s visual analogue scale; mean improvement from baseline of 12.1 mm)
Mycotoxins in agriculturally used plants can cause intoxication in animals and can lead to severe financial losses for farmers. The endophytic fungus Epichloë festucae var. lolii living symbiotically within the cool season grass species Lolium perenne can produce vertebrate and invertebrate toxic alkaloids. Hence, an exact quantitation of alkaloid concentrations is essential to determine intoxication risk for animals. Many studies use different methods to detect alkaloid concentrations, which complicates the comparability. In this study, we showed that alkaloid concentrations of individual plants exceeded toxicity thresholds on real world grasslands in Germany, but not on the population level. Alkaloid concentrations on five German grasslands with high alkaloid levels peaked in summer but were also below toxicity thresholds on population level. Furthermore, we showed that alkaloid concentrations follow the same seasonal trend, regardless of whether plant fresh or dry weight was used, in the field and in a common garden study. However, alkaloid concentrations were around three times higher when detected with dry weight. Finally, we showed that alkaloid concentrations can additionally be biased to different alkaloid detection methods. We highlight that toxicity risks should be analyzed using plant dry weight, but concentration trends of fresh weight are reliable.
Propagule pressure and an invasion syndrome determine invasion success in a plant community model

The success of species invasions depends on multiple factors, including propagule pressure, disturbance, productivity, and the traits of native and non-native species. While the importance of many of these determinants has already been investigated in relative isolation, they are rarely studied in combination. Here, we address this shortcoming by exploring the effect of the above-listed factors on the success of invasions using an individual-based mechanistic model. This approach enables us to explicitly control environmental factors (temperature as surrogate for productivity, disturbance, and propagule pressure) as well as to monitor whole-community trait distributions of environmental adaptation, mass, and dispersal abilities. We simulated introductions of plant individuals to an oceanic island to assess which factors and species traits contribute to invasion success. We found that the most influential factors were higher propagule pressure and a particular set of traits. This invasion trait syndrome was characterized by a relative similarity in functional traits of invasive to native species, while invasive species had on average higher environmental adaptation, higher body mass, and increased dispersal distances, that is, had greater competitive and dispersive abilities. Our results highlight the importance in management practice of reducing the import of alien species, especially those that display this trait syndrome and come from similar habitats as those being managed.

Ein besonderer Fokus liegt dabei auf den zahlreichen Formen von Intertextualität.

Schafe leben seit einigen Jahrtausenden mit Menschen zusammen und mit ihnen durchwandert ihre Wolle unsere Kulturen und Gesellschaften. Ausgangspunkt der vorliegenden Arbeit ist der gegenwärtig geringe monetäre Wert von Schafwolle, der in den letzten dreißig Jahren in Deutschland stark gesunken ist.
Basierend auf ethnografischen Methoden der Feldforschung, teilnehmende Beobachtung und Interviews, verfolgt die vorliegende Arbeit Spuren von Schafwolle und fragt dabei nach den Berührungs- und Kontaktpunkten, die im Alltag zwischen Schafen, Menschen und Wolle entstehen. Menschen, die mit Schafen leben, werfen verschiedene Perspektiven auf die sogenannte Nutztierhaltung sowie auf ländliches Wirtschaften und Leben.
Schafe und Schafwolle werden hier als Miterzählende und Mitgestaltende im Prozess der Entstehung von Schaf-Woll-Geschichten betrachtet. Besonders die Perspektiven auf Schafwolle regen an, tierliche Materialitäten im Allgemeinen neu zu denken und zu reflektieren. So lädt die vorliegende Arbeit dazu ein, Menschen, Tiere und Materialitäten stärker in den Wissensgenerierungsprozess zu integrieren sowie nach dem besonderen Wissen zu suchen, das eben jene mehr-als-menschliche Akteur*innen mit uns teilen.
Long-term sequelae in hospitalized Coronavirus Disease 2019 (COVID-19) patients may result in limited quality of life. The current study aimed to determine health-related quality of life (HRQoL) after COVID-19 hospitalization in non-intensive care unit (ICU) and ICU patients. This is a single-center study at the University Hospital of Wuerzburg, Germany. Patients eligible were hospitalized with COVID-19 between March 2020 and December 2020. Patients were interviewed 3 and 12 months after hospital discharge. Questionnaires included the European Quality of Life 5 Dimensions 5 Level (EQ-5D-5L), patient health questionnaire-9 (PHQ-9), the generalized anxiety disorder 7 scale (GAD-7), FACIT fatigue scale, perceived stress scale (PSS-10) and posttraumatic symptom scale 10 (PTSS-10). 85 patients were included in the study. The EQ5D-5L-Index significantly differed between non-ICU (0.78 ± 0.33 and 0.84 ± 0.23) and ICU (0.71 ± 0.27; 0.74 ± 0.2) patients after 3- and 12-months. Of non-ICU 87% and 80% of ICU survivors lived at home without support after 12 months. One-third of ICU and half of the non-ICU patients returned to work. A higher percentage of ICU patients was limited in their activities of daily living compared to non-ICU patients. Depression and fatigue were present in one fifth of the ICU patients. Stress levels remained high with only 24% of non-ICU and 3% of ICU patients (p = 0.0186) having low perceived stress. Posttraumatic symptoms were present in 5% of non-ICU and 10% of ICU patients. HRQoL is limited in COVID-19 ICU patients 3- and 12-months post COVID-19 hospitalization, with significantly less improvement at 12-months compared to non-ICU patients. Mental disorders were common highlighting the complexity of post-COVID-19 symptoms as well as the necessity to educate patients and primary care providers about monitoring mental well-being post COVID-19.

Das Ziel dieser Arbeit war es, eine im Arbeitskreis entwickelte Methode zur Darstellung von unterschiedlich substituierten -Aminopiperidinen zu etablieren und zu verifizieren, indem unser Konzept einer Tandemreaktion zum Einsatz kommen sollte. Diese Reaktionssequenz sollte anschließend zur Totalsynthese von Microconin, einem aus Microcos paniculata isoliertem Alkaloid, genutzt werden. Den ersten Schritt in Richtung -Aminopiperidinderivate machte die Veresterung von L-Pyroglutaminsäure. Nach der Reduktion des Methylesters erfolgte die Aktivierung der Hydroxyfunktion des Alkohols in Form des Tosylats. Die Azideinführung resultierte aus einer nucleophilen Substitutionsreaktion, anschließend daran wurde der Lactam-Stickstoff mit Boc2O und einer katalytischen Menge DMAP geschützt. Das Lactam ist durch den Elektronenakzeptor aktiviert, so dass die Ringöffnung zum Methylester unter sehr milden Bedingungen und ohne weiteren Reinigungsschritt erfolgen konnte. Die Aminofunktion musste mit einer zweiten Schutzgruppe blockiert werden, die anschließende selektive Reduktion mit DiBAl-H in trockenem Ether verlief problemlos und lieferte mit dem Aldehyd das Edukt für Olefinierungen mittels verschiedener Wittig-Reaktionen. Dafür wurden stabilisierte Phosphonate hergestellt und in einer HWE-Reaktion mit dem Aldehyd umgesetzt. Die resultierenden elektronenarmen Olefine gingen dann die intramolekulare Cycloaddition mit dem Azidsubstituenten ein und bauten so den Grundkörper der -Aminopiperidinderivate in einer Reaktionssequenz auf, die wir als Tandem Wittig-[3+2]-Cycloaddition bezeichnen. Die Bildung der primären Triazoline erfolgte stereoselektiv, die Geschwindigkeit der Cycloaddition hing sowohl vom konjugierten Olefinsubstituenten als auch vom vicinalen Substituenten der Azidfunktion ab. Die Konfigurationsbestimmung erfolgte mittels NMR-Spektroskopie durch Analyse der Kopplungskonstanten und NOE-Messungen. Die asymmetrische Induktion der Cycloaddition konnte direkt für die Stereochemie am Piperidinring genutzt werden, indem, nach basischer Triazolin/Diazoamin Isomerisierung, gleich das Diazoamin hydriert wurde. Die Hydrierung der vinylogen Urethane, den Produkten aus der rhodiumkatalysierten Stickstoffextrusion, lieferte ein Diastereomerengemisch, wobei das Verhältnis der Diastereomere hauptsächlich vom Substitutionsgrad des exocyclischen Amins abhängig war. Überraschenderweise fand beim Sulfontriazolin keine Isomerisierung zum Diazoamin statt, daher musste für die Darstellung der Sulfonylmethyl--aminopiperidine eine alternative Route über ein Ketosulfon beschritten werden. Die Synthese von Microconin begann mit der Desoxygenierung von L-Rhamnose durch die sog. Fischer-Zach-Reaktion. Das Rhamnal wurde in einer drei Stufen Eintopfreaktion erhalten und mittels Perlinhydrolyse in den offenkettigen Aldehyd umgewandelt. Die Aktivierung der Hydroxyfunktion als Mesylat resultierte in einer äußerst empfindlichen Verbindung, die nur durch Verwendung des Lindlar-Katalysators mit zufrieden stellenden Ergebnis zum aliphatischen Aldehyd reduziert werden konnte. Eine bimolekulare nucleophile Substitutionsreaktion lieferte bei der Azideinführung zur Schlüsselverbindung sowohl die benötigte funktionelle Gruppe als auch die benötigte Inversion der Konfiguration. Die Tandem HWE-[3+2]-Cycloadditions-Reaktion führte auch bei dem Sulfontriazolin in eine Sackgasse, weshalb wieder eine alternative Syntheseroute eingeschlagen werden musste. Ausgehend von derselben Schlüsselverbindung gelang dies durch eine zinnkatalysierte Umsetzung mit stabilisierten Diazomethan zum Ketosulfon. Der Aufbau des Piperidin-Heterocyclus konnte dann wieder über eine intramolekulare Imin-Bildung des intermediären Amins mit dem Keton erzielt werden. Die diastereoselektive Hydrierung verlief unter Wasserstoffaddition von der sterisch weniger gehinderten -Seite und Ausbildung des all cis Substitutionsmusters. Nach dem erfolgreichen Aufbau des Heterocyclus mussten noch die beiden Heteroatome methyliert werden. Dabei wurden die besten Ergebnisse am Ringstickstoff mit der reduktiven Aminierung erzielt. Bei der anschließenden Abspaltung der Acetylgruppe zeigte sich erstmals, dass das Substitutionsmuster am Piperidinring nicht konfigurationsstabil war, da neben dem erwarteten Alkohol auch das Diastereomer isoliert wurde. Die genaue Ursache für die Epimerisierung nach der N-Methylierung konnte nicht geklärt werden. Die Einführung des Methoxy-Substituenten am Grundkörper erfolgte über eine Williamsonschen Ethersynthese. Bei den Versuchen zur Kupplung des Grundkörpers mit der Seitenkette 2,4-Nonadienal erwies sich der Zusatz von HMPT als förderlich. Weiterhin konnte die Ausbeute an -Hydroxysulfonen durch das Erwärmen der Reaktionsmischung gesteigert werden. In den Folgeschritten der Julia-Olefinierung blieben die Versuche zur Ausbildung der dreifach ungesättigten Struktureinheit in der Seitenkette des isolierten Naturstoffs jedoch erfolglos.

Current data show that resilience is an important factor in cancer patients’ well-being. We aim to explore the resilience of patients with lower grade glioma (LGG) and the potentially influencing factors. We performed a cross-sectional assessment of adult patients with LGG who were enrolled in the LoG-Glio registry. By phone interview, we administered the following measures: Resilience Scale (RS-13), distress thermometer, Montreal Cognitive Assessment Test for visually impaired patients (MoCA-Blind), internalized stigmatization by brain tumor (ISBI), Eastern Cooperative Oncological Group performance status (ECOG), patients’ perspective questionnaire (PPQ) and typical clinical parameters. We calculated correlations and multivariate regression models. Of 74 patients who were assessed, 38% of those showed a low level of resilience. Our results revealed significant correlations of resilience with distress (p < 0.001, −0.49), MOCA (p = 0.003, 0.342), ECOG (p < 0.001, −0.602), stigmatization (p < 0.001, −0.558), pain (p < 0.001, −0.524), and occupation (p = 0.007, 0.329). In multivariate analyses, resilience was negatively associated with elevated ECOG (p = 0.020, β = −0.383) and stigmatization levels (p = 0.008, β = −0.350). Occupation showed a tendency towards a significant association with resilience (p = 0.088, β = −0.254). Overall, low resilience affected more than one third of our cohort. Low functional status is a specific risk factor for low resilience. The relevant influence of stigmatization on resilience is a novel finding for patients suffering from a glioma and should be routinely identified and targeted in clinical routine.
Brain metastases are the most severe tumorous spread during breast cancer disease. They are associated with a limited quality of life and a very poor overall survival. A subtype of extracellular vesicles, exosomes, are sequestered by all kinds of cells, including tumor cells, and play a role in cell-cell communication. Exosomes contain, among others, microRNAs (miRs). Exosomes can be taken up by other cells in the body, and their active molecules can affect the cellular process in target cells. Tumor-secreted exosomes can affect the integrity of the blood-brain barrier (BBB) and have an impact on brain metastases forming. Serum samples from healthy donors, breast cancer patients with primary tumors, or with brain, bone, or visceral metastases were used to isolate exosomes and exosomal miRs. Exosomes expressed exosomal markers CD63 and CD9, and their amount did not vary significantly between groups, as shown by Western blot and ELISA. The selected 48 miRs were detected using real-time PCR. Area under the receiver-operating characteristic curve (AUC) was used to evaluate the diagnostic accuracy. We identified two miRs with the potential to serve as prognostic markers for brain metastases. Hsa-miR-576-3p was significantly upregulated, and hsa-miR-130a-3p was significantly downregulated in exosomes from breast cancer patients with cerebral metastases with AUC: 0.705 and 0.699, respectively. Furthermore, correlation of miR levels with tumor markers revealed that hsa-miR-340-5p levels were significantly correlated with the percentage of Ki67-positive tumor cells, while hsa-miR-342-3p levels were inversely correlated with tumor staging. Analysis of the expression levels of miRs in serum exosomes from breast cancer patients has the potential to identify new, non-invasive, blood-borne prognostic molecular markers to predict the potential for brain metastasis in breast cancer. Additional functional analyzes and careful validation of the identified markers are required before their potential future diagnostic use.
Background: The clinical management of teeth with complex dens invaginatus (DI) malformations and apical periodontitis may be challenging due to the lack of routine. The aim of this case report is to describe the endodontic treatment of an immature tooth with DI and to discuss strategies for preclinical training for teeth with such malformations. Case report: A 9-year-old male presented with an immature maxillary incisor with DI (Oehlers Type II) and apical periodontitis which was diagnosed by cone beam computed tomography (CBCT). Revitalization was initially attempted but then abandoned after failure to generate a stable blood clot. Nevertheless, considerable increase in both root length and thickness could be detected after medication with calcium hydroxide followed by root canal filling with MTA as an apical plug. Conclusions: The endodontic management of teeth with DI requires thorough treatment planning. In immature teeth, under certain conditions, root maturation may occur even with conventional apexification procedures. From an educational perspective, different strategies including CBCT and 3D-printed transparent tooth models for visualization of the complex internal morphology and redesigned 3D-printed replica with various degrees of difficulty for endodontic training, can be used to overcome the challenges associated with endodontic treatment of such teeth.
Objectives
Magnesium phosphate-based cements begin to catch more attention as bone substitute materials and especially as alternatives for the more commonly used calcium phosphates. In bone substitutes for augmentation purposes, atraumatic materials with good biocompatibility and resorbability are favorable. In the current study, we describe the in vivo testing of novel bone augmentation materials in form of spherical granules based on a calcium-doped magnesium phosphate (CaMgP) cement.
Materials and Methods
Granules with diameters between 500 and 710 μm were fabricated via the emulsification of CaMgP cement pastes in a lipophilic liquid. As basic material, two different CaMgP formulations were used. The obtained granules were implanted into drill hole defects at the distal femoral condyle of 27 New Zealand white rabbits for 6 and 12 weeks. After explantation, the femora were examined via X-ray diffraction analysis, histological staining, radiological examination, and EDX measurement.
Results
Both granule types display excellent biocompatibility without any signs of inflammation and allow for proper bone healing without the interposition of connective tissue. CaMgP granules show a fast and continuous degradation and enable fully adequate bone regeneration.
Conclusions
Due to their biocompatibility, their degradation behavior, and their completely spherical morphology, these CaMgP granules present a promising bone substitute material for bone augmentation procedures, especially in sensitive areas.
Clinical Relevance
The mostly insufficient local bone supply after tooth extractions complicates prosthetic dental restoration or makes it even impossible. Therefore, bone augmentation procedures are oftentimes inevitable. Spherical CaMgP granules may represent a valuable bone replacement material in many situations.
Aspergillus fumigatus causes life-threatening opportunistic infections in immunocompromised patients. As therapeutic outcomes of invasive aspergillosis (IA) are often unsatisfactory, the development of targeted immunotherapy remains an important goal. Linking the innate and adaptive immune system, dendritic cells are pivotal in anti-Aspergillus defense and have generated interest as a potential immunotherapeutic approach in IA. While monocyte-derived dendritic cells (moDCs) require ex vivo differentiation, antigen-pulsed primary myeloid dendritic cells (mDCs) may present a more immediate platform for immunotherapy. To that end, we compared the response patterns and cellular interactions of human primary mDCs and moDCs pulsed with an A. fumigatus lysate and two A. fumigatus proteins (CcpA and Shm2) in a serum-free, GMP-compliant medium. CcpA and Shm2 triggered significant upregulation of maturation markers in mDCs and, to a lesser extent, moDCs. Furthermore, both A. fumigatus proteins elicited the release of an array of key pro-inflammatory cytokines including TNF-α, IL-1β, IL-6, IL-8, and CCL3 from both DC populations. Compared to moDCs, CcpA- and Shm2-pulsed mDCs exhibited greater expression of MHC class II antigens and stimulated stronger proliferation and IFN-γ secretion from autologous CD4\(^+\) and CD8\(^+\) T-cells. Moreover, supernatants of CcpA- and Shm2-pulsed mDCs significantly enhanced the oxidative burst in allogeneic neutrophils co-cultured with A. fumigatus germ tubes. Taken together, our in vitro data suggest that ex vivo CcpA- and Shm2-pulsed primary mDCs have the potential to be developed into an immunotherapeutic approach to tackle IA.
