Epichloë endophytes associated with cool-season grass species can protect their hosts from herbivory and can suppress mycorrhizal colonization of the hosts’ roots. However, little is known about whether or not Epichloë endophyte infection can also change the foliar fungal assemblages of the host. We tested 52 grassland study sites along a land-use intensity gradient in three study regions over two seasons (spring vs. summer) to determine whether Epichloë infection of the host grass Lolium perenne changes the fungal community structure in leaves. Foliar fungal communities were assessed by Next Generation Sequencing of the ITS rRNA gene region. Fungal community structure was strongly affected by study region and season in our study, while land-use intensity and infection with Epichloë endophytes had no significant effects. We conclude that effects on non-systemic endophytes resulting from land use practices and Epichloë infection reported in other studies were masked by local and seasonal variability in this study’s grassland sites.
Fertilität und Macht: Die Reproduktionspflicht mittelalterlicher Herrscherinnen und Herrscher

Fertility was a key theme of medieval rulership. To conceive and give birth to sons – and thus to ensure the succession to the throne – was one of the foremost duties of medieval kings and queens. But what happened when a male child died in infancy, no male child was born, or no pregnancy ever came about? Barrenness could have dramatic consequences in the Middle Ages, for example expulsion, divorce or conflicts over royal succession. Against this historical background, it seems logical that the fate of the childless ruler would be recounted in the form of a ‘Passion narrative’. In the German literature of the Middle Ages, however, there are also interpretative models of a contrary vein to be found. In the year 1220, for instance, Ebernand of Erfurt construed the wedded life of the imperial couple Henry and Kunigunde as a tale of resistance against the royal obligation to reproduce. In his versified legend, composed in the vernacular, the couple secretly agrees not to fulfil society’s expectations, but to lead a chaste marriage. Yet above and beyond legend, childless rulers were also subject to the impact of multifarious legal, religious, medical, narrative and discursive factors. Taking Michel Foucault as a point of departure, this contribution shows how fertility became a field of power on which hierarchies between rulers and subjects, men and women were negotiated, while also shedding light on how religious and secular ideals clashed in the assessment of infertility.
The lipids phosphatidylserine (PtdSer) and phosphatidylethanolamine (PtdEth) are normally asymmetrically localized to the cytosolic face of membrane bilayers, but can both be externalized during diverse biological processes, including cell division, cell fusion, and cell death. Externalized lipids in the plasma membrane are recognized by lipid-binding proteins to regulate the clearance of cell corpses and other cell debris. However, it is unclear whether PtdSer and PtdEth contribute in similar or distinct ways to these processes. We discovered that disruption of the lipid flippases that maintain PtdSer or PtdEth asymmetry in the plasma membrane have opposite effects on phagocytosis in Caenorhabditis elegans embryos. Constitutive PtdSer externalization caused by disruption of the major PtdSer flippase TAT-1 led to increased phagocytosis of cell debris, sometimes leading to two cells engulfing the same debris. In contrast, PtdEth externalization caused by depletion of the major PtdEth flippase TAT-5 or its activator PAD-1 disrupted phagocytosis. These data suggest that PtdSer and PtdEth externalization have opposite effects on phagocytosis. Furthermore, externalizing PtdEth is associated with increased extracellular vesicle release, and we present evidence that the extent of extracellular vesicle accumulation correlates with the extent of phagocytic defects. Thus, a general loss of lipid asymmetry can have opposing impacts through different lipid subtypes simultaneously exerting disparate effects.
Plant-associated fungi can affect the plants‘ interaction with herbivores and
other microorganisms. For example, many common forage grasses are infected
with Epichloë endophytes. The endophytes systemically colonize the aerial
parts of the plants. They produce bioprotective alkaloids that can negatively
affect insects and livestock feeding on the grasses, and interact with other
fungal species which living from the plants‘ nutrients. Environmental conditions
strongly influence Epichloë endophytes. Endophyte-mediated effects
on herbivores are more pronounced under increased temperatures and the
endophytes may benefit from land use in managed grasslands. Under the
framework of the large-scale German project “Biodiversity Exploratories”, I
investigated whether infection rates and alkaloid concentrations of Epichloë
festucae var. lolii in Lolium perenne (Chapter I) and Epichloë endophytes (E.
uncinata, E. siegelii) in Festuca pratensis (Chapter II) depend on land use and
season. Further I analysed, whether foliar fungal assemblages of L. perenne
are affected by the presence of Epichloë endophytes (Chapter IV).
Psychotropic drugs are frequently prescribed ‘off-label’ to children and adolescents and carry the risk of serious adverse drug reactions (sADR). We examined the frequency of sADRs of psychotropic drugs in pediatric inpatients and explored their potential preventability through following the recommendations of a web-based pediatric drug information system (PDIS). The potential socio-economic impacts of using this online system is also addressed. Routine clinical data from all inpatients treated in a child and adolescent psychiatry department between January 2017 and December 2018 were retrospectively examined for the occurrence of sADRs as defined by the European Medicines Agency. The preventability of the sADRs was assessed based on the information of the PDIS. Furthermore, the expected prolongation of the hospital stay due to sADRs was calculated as well as the associated treatment costs. The study was supported by the Innovation Fund of the Joint Federal Committee, grant number 01NVF16021. In total, 1036 patients were screened of whom 658 (63.5%) received psychopharmacological treatment. In 53 (8.1%) of these patients 54 sADRs were documented, of which 37 sADRs were identified as potentially preventable through PDIS. Mitigating sADR through PDIS would likely have prevented prolonged hospital stays and conferred considerable savings for health insurance companies. PDIS provides systematic and evidence-based information about pediatric psychopharmacotherapy and helps to prevent prescribing errors. Therefore, PDIS is a useful tool to increase drug therapy safety in child and adolescent psychiatry. Further prospective studies are needed to confirm the results.
Das Ziel dieser Dissertation besteht darin, die für sein Künstlerleben relevanten Aspekte der Biografie Sándor (Alexander)von Wagners darzustellen. Dabei bewegen wir uns auf einem noch kaum erforschten Gebiet und bieten gleichsam die nötigen Anhaltspunkte für weitere Forschungsbeiträge. Des Weiteren sind die Werke Wagners zu betrachten. Insbesondere die anhand der Biographie belegbaren Einflüsse auf seine Werke und die von seinen Werken ausgehende Wirkung sind dabei zu berücksichtigen, um anhand der Darstellung von Wagners Position, peripher auch diejenige osteuropäischer Künstler, im gesamteuropäischen Kontext aufzudecken.
Im Mittelpunkt der vorliegenden Dissertation steht der künstlerische Werdegang Sándor (Alexander) von Wagners. Er stammte aus Ungarn, ließ sich später in München als professioneller Kunstmaler nieder und arbeitete von 1866 bis 1869 als Hilfslehrer und von 1869 bis 1910 als ordentlicher Professor für Maltechnik an der Münchner Kunstakademie. In dieser Abhandlung werden die für die künstlerische Entwicklung wesentlichen Aspekte seines Lebenslaufes dargestellt:
- die künstlerische Laufbahn Wagners unter besonderer Betrachtung seiner künstlerischen Vorbereitung und Studien sowie seiner Karriere in München
- Grundzüge seiner Werke und Einflüsse, die Wagners Lehrer und Professoren auf sein künstlerisches Schaffen ausübten
- Wagners kunstpädagogische Tätigkeit an der Königlichen Kunstakademie in München
- sein Privatleben in München
Daneben liefert diese Untersuchung eine Liste mit Wagners Studenten und legt ein vorläufiges Werkverzeichnis vor. Vollständigkeit war auf Grund der prekären Forschungslage im Rahmen einer einzelnen Dissertation nicht anzustreben, sondern es galt, eine erste grundlegende Basis zu schaffen, deren Ergänzung weiteren Forschungsbemühungen vorbehalten bleiben muss. Diese Arbeit will damit als Anregung zu weiteren Untersuchungen dienen.

Für diese retrospektive Studie wurden 95 konsekutive Patienten herausgesucht, die zwischen Januar 1997 und Dezember 1999 im König-Ludwig-Haus im Rahmen einer Rotatorenmanschettenrekonstruktion zusätzlich eine LBS-Tenodese erhielten. Diese Patienten wurden anhand der Defektlokalisation, d. h. isolierter SSP-, isolierter SSC-, kombinierter SSP+ISP-, kombinierter SSP+SSC-Defekt und kombinierter SSP+SSC+ISP-Defekt in fünf Gruppen eingeteilt. Für jede Gruppe wurden Matched-Pairs gebildet. Hierzu wurden Patienten gleicher Defektlokalisation und Rotatorenmanschettenrekonstruktion, jedoch ohne Versorgung mit einer LBS-Tenodese ausgewählt, die in Alter und Geschlecht den Patienten „mit Tenodese“ entsprachen. Mit 132 Personen (67 „mit Tenodese“ und 65 „ohne Tenodese“) konnten Matched-Pairs gebildet werden. Für die 17 Patienten mit SSP+SSC-Defekt war dies nicht möglich, da bei einer derartigen Defektsituation stets eine Tenodese der LBS durchgeführt worden war. Somit wurde 149 Patienten ein Fragebogen zugeschickt, der den Constant-Murley Score, den Simple Shoulder Test und spezielle Schulterfunktionsfragen enthielt. 111 Patienten (63 „mit Tenodese“ und 48 „ohne Tenodese“) füllten die Fragebögen aus und sendeten sie zurück. Die Fragebögen wurden statistisch ausgewertet und die Ergebnisse der Patienten „ohne Tenodese“ denen „mit Tenodese“ gegenübergestellt - sowohl in den Matched-Pairs, als auch bei der Betrachtung aller Patienten unabhängig von der Lokalisation des Rotatorenmanschettendefektes. Zusätzlich wurden bei der Auswertung noch die Gesamtergebnisse des Constant-Scores von zehn Patienten, die aus einer vorherigen Untersuchung bekannt waren, berücksichtigt. Bei der Bewertung der Operationszufriedenheit und beim Simple Shoulder Test zeigten sich keine statistisch signifikanten Unterschiede zwischen den beiden Gruppen. Beim Gesamtvergleich aller Patienten unabhängig von der Defektlokalisation, erzielten die Patienten „ohne Tenodese“ (n=53) einen durchschnittlichen Constant-Score von 68,7 Punkten und bei jedem Parameter höhere Ergebnisse als die Patienten „mit Tenodese“ (n=54), deren durchschnittlicher Constant-Score 56,7 Punkte betrug. Abgesehen von den Parametern Arbeitshöhe und Innenrotation waren diese Unterschiede bei allen vier Sektionen des Constant-Scores statistisch signifikant. Beim Matched-Pair der Patienten mit isoliertem SSP-Defekt lag bei keinem Parameter ein statistisch signifikanter Unterschied zwischen den 26 Patienten mit und den 24 Patienten ohne Tenodese vor, doch erzielten die Patienten „ohne Tenodese“ durchschnittlich höhere Werte. Im Durchschnitt betrug der Constant-Score der Patienten „ohne Tenodese“ 68,7 Punkte gegenüber 62 Punkten bei den Patienten mit Tenodese. Für das Matched-Pair der Patienten mit isoliertem SSC-Defekt wurde aufgrund der geringen Patientenzahl keine Signifikanzprüfung durchgeführt, doch zeigte sich auch hier, dass die vier Patienten „mit Tenodese“ ein niedrigeres Ergebnis im Constant-Score (63,5 Punkte) erreichten als die fünf Patienten ohne LBS-Tenodese (69 Punkte). Die Patienten des Matched-Pairs SSP+ISP-Defekt „ohne Tenodese“ (n=8) erreichten, abgesehen von der Innenrotation, bei allen Parametern ein signifikant höheres Ergebnis als Patienten „mit Tenodese“(n=9) und erzielten das höchste Ergebnis im Constant-Score (85,1 Punkte) von allen Gruppen. Bei einem vorliegendem Drei-Sehnen-Defekt konnte zwischen den beiden Gruppen nur bei der Außenrotation ein statistisch signifikanter Unterschied festgestellt werden, doch waren auch hier die einzelnen Ergebnisse bei Patienten „ohne Tenodese“ (n=16) stets höher als bei der Gruppe „mit Tenodese“ (n=15), die hier das niedrigste Gesamtergebnis (47,6 Punkte) von allen Gruppen erreichte. Obwohl nicht bei jedem Parameter statistisch signifikante Unterschiede herausgefunden werden konnten, kann man dennoch beobachten, dass die Patienten, bei denen keine LBS-Tenodese durchgeführt werden musste, tendenziell bessere Ergebnisse erzielen konnten als die Patienten „mit Tenodese“. Inwieweit sich eine vorbestehende Schädigung der LBS auf das Ergebnis einer Rotatorenmanschettenrekonstruktion auswirkt ist mit dieser vorliegenden retrospektiven Studie nicht vollständig geklärt, weitere Untersuchungen sind daher notwendig.
Endophytes live in partial symbiosis inside a plant and have been detected in all tested plants. They belong to the group of fungi or bacteria and their ecological function is mostly unknown. The fungal endophytes of the genus Epichloë belong to a special group of endophytes. Epichloë endophytes live symbiotically inside cool season grass species and some of them are able to produce alkaloids toxic to vertebrates and insects. Their symbiosis is seen as mutualistic for the following reasons: the fungus provides the plant herbivore resistance by producing alkaloids, and it increases the plant’s drought tolerance as well as its biomass production. In return, the grass provides the fungus shelter, nutrients and dispersal. Epichloë endophytes are host specific and the ability to produce alkaloids differs between species. In order to estimate intoxication risks in grasslands, it is necessary to detect infection rates of different grass species with Epichloë endophytes, and to determine the genotypes and chemotypes of the Epichloë species as well as the produced alkaloid concentrations. Factors like land-use intensity or season may have an influence on infection rates and alkaloid concentrations. Also, different methodological approaches may lead to different results. In this doctoral thesis my general aim was to evaluate intoxication risks in German grasslands caused by Epichloë endophytes. For that I investigated infection rates of different grass species and the genotypes and chemotypes of their Epichloë endophytes in German grasslands (Chapter II). Furthermore, I compared alkaloid concentrations detected with dry and fresh plant weight and different analytical methods. I also detected possible changes on the influence of season or land-use intensity (Chapter III). Additionally, I examined infections with Epichloë endophytes and alkaloid concentrations in commercially available grass seed mixtures and determined how that influences the intoxication risk of grazing animals in Europe (Chapter IV).
It is of agricultural interest to estimate intoxication risks for grazing livestock on German grasslands due to Epichloë infected grass species. Therefore, it is important to investigate which grasses are infected with the Epichloë endophyte, if the endophytes have the ability to produce vertebrate and invertebrate toxic alkaloids and if the alkaloids are indeed produced. I showed that Epichloë festucae var. lolii infecting agriculturally important Lolium perenne lacked the starting gene for ergovaline biosynthesis. Hence, vertebrate toxic ergovaline was not detected in the majority of the collected L. perenne plants. The detection of alkaloid concentrations is an important tool to estimate intoxication risk for vertebrates, but also invertebrates. My studies showed that the usage of dry plant material is crucial to quantify the correct alkaloid concentrations, and that alkaloid concentrations can vary depending on the detection method. Hence, the usage of validated, similar detection methods is important to be able to compare alkaloid concentrations from different studies. Nevertheless, the trends of seasonal changes and the influence of land-use intensity stayed the same, regardless if dry or fresh plant weight was used. Also, alkaloid concentrations were below toxicity thresholds on population level, regardless of the method used. Two commercially available forage grass and two commercially available turf grass seed mixtures were infected with Epichloë endopyhtes and alkaloids were detected. This might contribute to the spreading of Epichloë endopyhtes in Germany, therefore seed mixtures should be tested for Epichloë infections. My results indicate that the intoxication risk is generally low in Germany at the moment, although that might change due to climate change, an increase of monocultural land-use, or the seeding of Epichloë infected grass seeds.
Diese Dissertation liefert einen kulturhistorischen Überblick über die Darstellung der Frauenfiguren Gertrude und Ophelia in Hamlet-Verfilmungen des 20. Jahrhunderts. Die beiden weiblichen Gestalten werden dabei im Hinblick auf ihr äußeres Erscheinungsbild wie auch ihre Charakterzeichnung vor dem historischen, kulturellen, politischen und gesellschaftlichen Entstehungshintergrund der Filme beleuchtet. Die Analysen konzentrieren sich ausschließlich auf Kinofilme, da diese ein wesentlich breiteres Publikum erreichen als etwa die diversen Bühnen- oder Textfassungen des Stücks. Die erste der insgesamt zwanzig untersuchten Leinwandfassungen entstand im Jahre 1900, die letzte kam 2000 in die Kinos – die vorliegende Arbeit behandelt also eine genau 100-jährige Tradition der Hamlet-Verfilmungen.
Background
In individuals suffering from a rare disease the diagnostic process and the confirmation of a final diagnosis often extends over many years. Factors contributing to delayed diagnosis include health care professionals' limited knowledge of rare diseases and frequent (co-)occurrence of mental disorders that may complicate and delay the diagnostic process. The ZSE-DUO study aims to assess the benefits of a combination of a physician focusing on somatic aspects with a mental health expert working side by side as a tandem in the diagnostic process.
Study design
This multi-center, prospective controlled study has a two-phase cohort design.
Methods
Two cohorts of 682 patients each are sequentially recruited from 11 university-based German Centers for Rare Diseases (CRD): the standard care cohort (control, somatic expertise only) and the innovative care cohort (experimental, combined somatic and mental health expertise). Individuals aged 12 years and older presenting with symptoms and signs which are not explained by current diagnoses will be included. Data will be collected prior to the first visit to the CRD’s outpatient clinic (T0), at the first visit (T1) and 12 months thereafter (T2).
Outcomes
Primary outcome is the percentage of patients with one or more confirmed diagnoses covering the symptomatic spectrum presented. Sample size is calculated to detect a 10 percent increase from 30% in standard care to 40% in the innovative dual expert cohort. Secondary outcomes are (a) time to diagnosis/diagnoses explaining the symptomatology; (b) proportion of patients successfully referred from CRD to standard care; (c) costs of diagnosis including incremental cost effectiveness ratios; (d) predictive value of screening instruments administered at T0 to identify patients with mental disorders; (e) patients’ quality of life and evaluation of care; and f) physicians’ satisfaction with the innovative care approach.
Conclusions
This is the first multi-center study to investigate the effects of a mental health specialist working in tandem with a somatic expert physician in CRDs. If this innovative approach proves successful, it will be made available on a larger scale nationally and promoted internationally. In the best case, ZSE-DUO can significantly shorten the time to diagnosis for a suspected rare disease.

Obwohl der klinische Stellenwert der MRT in zahlreichen Studien für verschiedene Entitäten (AC-Komplex, coracoidale Pathologien, Rotatorenmanschette, Pathologien des Kapsel-Band-Apparates und der Bicepssehne sowie primär und reaktiv bedingte entzündliche Veränderungen) belegt ist, überrascht es, dass in der Literatur keine Arbeiten darüber existieren, inwieweit das Ergebnis der MRT-Untersuchung tatsächlich das therapeutische Procedere beeinflusst. Vorliegende Arbeit untersuchte daher den diagnostischen Impact der MRT im Setting einer hochspezialisierten Schultersprechstunde.

Pseudodistomin E : Versuche zur Totalsynthese über das Konzept der Tandem Wittig-[3+2]-Cycloaddition

Die Einleitung gibt einen kurzen Überblick über die Bedeutung von Piperidinalkaloiden und im speziellen wird kurz auf das pharmakologische Potential mariner Piperidinalkaloide eingegangen. Anschließend wird die Substanzklasse der „Pseudodistomine“ vorgestellt, gefolgt von einer Übersicht bereits literaturbekannter Synthesemöglichkeiten. Das primäre Ziel der vorliegenden Arbeit war die Entwicklung eines neuen stereoselektiven Zugangs zum all cis substituierten Grundkörper der Pseudodistomine C und E über die Kaskade einer Tandem Wittig-[3+2]-Cycloaddition. Weiterhin sollten Möglichkeiten ausgelotet werden, um hieran die Seitenkette des Pseudodistomins E aufbauen zu können, um erstmals eine Totalsynthese dessen zu ermöglichen.
The topological classification of electronic band structures is based on symmetry properties of Bloch eigenstates of single-particle Hamiltonians. In parallel, topological field theory has opened the doors to the formulation and characterization of non-trivial phases of matter driven by strong electron-electron interaction. Even though important examples of topological Mott insulators have been constructed, the relevance of the underlying non-interacting band topology to the physics of the Mott phase has remained unexplored. Here, we show that the momentum structure of the Green’s function zeros defining the “Luttinger surface" provides a topological characterization of the Mott phase related, in the simplest description, to the one of the single-particle electronic dispersion. Considerations on the zeros lead to the prediction of new phenomena: a topological Mott insulator with an inverted gap for the bulk zeros must possess gapless zeros at the boundary, which behave as a form of “topological antimatter” annihilating conventional edge states. Placing band and Mott topological insulators in contact produces distinctive observable signatures at the interface, revealing the otherwise spectroscopically elusive Green’s function zeros.

In der vorliegenden Studie wurde die Remineralisationswirkung von Fruchtgummiprodukten, die mit einer neuartigen calciumhaltigen Salzhydratschmelze angereichert waren, vergleichend in-vitro und in-situ getestet. Als Vergleich diente ein handelsübliches Fruchtgummiprodukt ohne Zusatz der zu testenden Salzhydratschmelze. Die remineralisierende Wirkung der calciumreichen Salzhydratschmelze beruhte auf dem Prinzip der forcierten dynamischen Remineralisation. Um die Mineralisationseffekte messbar zu machen, wurden künstliche, standardisiert hergestellte Schmelzplättchen den unterschiedlichen Fruchtgummiprodukten ausgesetzt und die Mineralisationseffekte in regelmäßigen Abständen gravimetrisch und radiographisch gemessen, sowie abschließend einer Härtemessung unterzogen. Dabei ergaben sich zusammengefasst folgende Resultate, die die Quantität an eingebautem Mineral betreffen: 1.Der Effekt aller salzhydratschmelzehaltigen Fruchtgummis ist signifikant größer als der der Plazebopräparate 2.Die schmelzehaltigen Fruchtgummis bewirken einen signifikant größeren Mineraleinbau als herkömmliche Fluoridpräparate in Zahnpasten, wie aus früheren gleichgearteten Untersuchungen abgeleitet werden kann. Aufgrund der vorliegenden Ergebnisse kann davon ausgegangen werden, dass trotz des Verzehrs zuckerhaltiger Fruchtgummiprodukte das Gleichgewicht zwischen Demineralisation und Remineralisation an der Zahnoberfläche nicht nur statiönär gehalten wird, sondern sogar zugunsten der Remineralisation verschoben wird, sofern die Fruchtgummiprodukte mit einer calciumhaltigen Salzhydratschmelze angereichert sind.
Der Beitrag untersucht, wie Gendervorstellungen, Geschlechtsidentitäten und Körperkonzepte in dem altfranzösischen Roman performativ konstruiert werden. Anknüpfend an die Gender- und Queertheorie Judith Butlers wird gezeigt, dass das vermeintlich natürliche Geschlecht der Protagonistin nicht essentialistisch, sondern als kulturelle Kategorie zu verstehen ist.
Indirekte Charakterisierung trägt zu einem großen Teil zur Gesamtcharakteristik von Figuren bei, ist bisher aber dennoch vergleichsweise wenig erforscht. Diese Arbeit nähert sich dem Phänomen der indirekten Figurencharakterisierung durch eine eigene Explikation theoretisch an, nennt verschiedene Repräsentationsformen von indirekter Charakterisierung und führt die Theorie dann in Bezug auf Redewiedergabe anhand zahlreicher Beispiele aus. Der Untersuchung wird ein Korpus realistischer Erzähltexte zugrunde gelegt und darin manuell Formen der Redewiedergabe annotiert, um in einem späteren Schritt computergestützt einige Experimente mit den Annotationen durchzuführen. In ihrem Aufbau entspricht diese Arbeit dadurch einem Mixed-Methods-Design von qualitativer und quantitativer Literaturwissenschaft.
„Musik auf dem Theater“ bildet eine lange Tradition, die Schauspiel und Oper großteils gemeinsam haben. Die Bezeichnung „Inzidenzmusik“ verweist auf die enge Bindung an die Handlung, in die sie „einfällt“. Richard Wagner, der alle seine musikdramatischen Werke mit Bühnenmusik versieht, knüpft an diese Tradition an. Dabei können drei Traditionsstränge unterschieden werden: Die inzidierende Musik, die sich dadurch auszeichnet, dass sie nach der Konvention feststehender Instrumenten-Emblematik im gesprochenen wie im gesungenen Drama eingesetzt wird, findet bei Wagner regelmäßig Anwendung. Die banda sul palco, die dagegen als Opernspezifikum zu betrachten ist, setzt er nur in seinen ersten Opern ein. Relativ selten treten gemischte, meist nicht sichtbar hinter der Bühne positionierte Instrumentengruppen auf, die mit transzendierender Musik eine jenseitige Welt repräsentieren sollen. Wagner weist der Bühnenmusik, neben ihren konventionellen Funktionen, weitere, neue Aufgaben zu. Die Musik auf dem Theater, die oft eher im Zusammenhang aufführungsbedingter Praxis denn als Teil auktorialer Originalität gesehen wird, erhält dadurch einen entscheidenen Platz in der Gesamtkonzeption und kann nicht beliebig ausgetauscht oder weggelassen werden. Ihre substantielle Rolle zeigt sich bereits im Kompositionsprozeß: Die Skizzen zur Bühnenmusik entstehen sehr früh, häufig weisen sie sogar in die Zeit der ersten Inspiration. Ihr motivisches Material hat Einfluß auf die Anlage des Werkganzen, formuliert aber auch in Form höchstverdichteter musikalischer Chiffren die wesentliche Aussagen des Gesamtwerkes. Am „fertigen“ Musikdrama zeigt sich, dass sie häufig Einsatz an den dramaturgisch wichtigen Stellen der Aktanfänge und -zäsuren findet. Als eine „ursprüngliche Sprache“ repräsentiert sie außerdem (ästhetisch gesehen) eine Sphäre, die sich von der Musik aus dem Orchestergraben deutlich unterscheidet. Obwohl Wagner die Bühnenmusik in seinen theoretischen Schriften kaum thematisiert, vertraut er ihr in allen Phasen seines Schaffens wesentliche Aufgaben an. Die herausgehobene Stellung der Musik auf dem Theater stellt eine bedeutende Konstante im Werk Richard Wagners dar.
Der Erste Weltkrieg und die Nachkriegszeit: Ein Thema, zu dem man scheinbar schon alles gehört und gelesen hat. Dass dem nicht so ist, zeigen Studierende der Philosophischen Fakultät der Universität Würzburg mit ihrer Ausstellung „Krieg │ Frieden. Der Erste Weltkrieg und die Nachkriegszeit“. Ihre Beiträge behandeln Politik, Kunst und Literatur in den am Krieg beteiligten Ländern.
Nach einer historischen Einführung, die auch die Nachkriegssituation in Würzburg themati-siert, vermitteln Texte von Gottfried Benn einen distanzierten Blick auf das Kampfgeschehen: Der Dichter wirkte während des Kriegs als Arzt hinter der Front und verarbeitete seine Erlebnisse u.a. in den „Rönne-Novellen“. Anhand literarischer Portraits von Schützen aus dem Senegal lässt sich wiederum nachvollziehen, wie sich der Blick auf die aus den Kolonien verpflichteten Soldaten in der französischen Literatur widerspiegelt. Unterschiedliche Reaktionen auf den Ersten Weltkrieg und die Nachkriegszeit werden demgegenüber in der italienischen und deutschen Kunst sichtbar, während die hierzulande aus propagandistischen Gründen eröffneten Kriegsmuseen in der Nachkriegszeit schlossen oder sich der Friedensarbeit widmeten. Stimmen namhafter italienischer Politiker, Journalisten, Romanciers und Dichter bringen zum Ausdruck, wie der „Große Krieg / la Grande Guerra“ Italiens kollektives Gedächtnis prägte. Auch in Großbritannien ist das Gedenken daran viel stärker als hierzulande: So prägen die von dem Gedicht „In Flanders Fields“ inspirierten Mohnblumen als „Remembrance Poppies“ die britische Erinnerungskultur bis in unsere Gegenwart.
Individuell gestaltete Text- und Bildtafeln, eine Hörstation mit Leseecke sowie eine Soundinstallation und Bildprojektion am Eröffnungsabend machen den Ausstellungsbesuch informativ und abwechslungsreich. Die interdisziplinäre Ausstellung wurde von rund 90 Studierenden der Fächer Anglistik, Amerikanistik, Germanistik, Geschichtswissenschaft, Kunstgeschichte, Museologie (Ausstellungsmanagement), Romanistik und Slavistik im Wintersemester 2018/19.
Growth and Differentiation Factor 5 (GDF5) is a secreted growth factor that belongs to the Bone Morphogenetic Protein (BMP) family and plays a pivotal role during limb development. GDF5 is a susceptibility gene for osteoarthritis (OA) and mutations in GDF5 are associated with a wide variety of skeletal malformations ranging from complex syndromes such as acromesomelic chondrodysplasias to isolated forms of brachydactylies or multiple synostoses syndrome 2 (SYNS2). Here, we report on a family with an autosomal dominant inherited combination of SYNS2 and additional brachydactyly type A1 (BDA1) caused by a single point mutation in GDF5 (p.W414R). Functional studies, including chondrogenesis assays with primary mesenchymal cells, luciferase reporter gene assays and Surface Plasmon Resonance analysis, of the GDF5 W-414R variant in comparison to other GDF5 mutations associated with isolated BDA1 (p.R399C) or SYNS2 (p.E491K) revealed a dual pathomechanism characterized by a gain-and loss-of-function at the same time. On the one hand insensitivity to the main GDF5 antagonist NOGGIN (NOG) leads to a GDF5 gain of function and subsequent SYNS2 phenotype. Whereas on the other hand, a reduced signaling activity, specifically via the BMP receptor type IA (BMPR1A), is likely responsible for the BDA1 phenotype. These results demonstrate that one mutation in the overlapping interface of antagonist and receptor binding site in GDF5 can lead to a GDF5 variant with pathophysiological relevance for both, BDA1 and SYNS2 development. Consequently, our study assembles another part of the molecular puzzle of how loss and gain of function mutations in GDF5 affect bone development in hands and feet resulting in specific types of brachydactyly and SYNS2. These novel insights into the biology of GDF5 might also provide further clues on the pathophysiology of OA.
Die Pseudodistomine gehören zu den ersten Piperidinalkaloiden marinen Ursprungs, die 1987 von Ishibashi et al. aus der Tunikate (Ascidie) Pseudodistoma kanoko isoliert wurden. Aus der gleichen Tunikate wurde 1995 das Pseudodistomin C isoliert. Die amphiphilen Piperidinalkaloide zeigen eine Antitumor-Aktivität gegen bestimmte Mäuseleukämiezellen, wobei Pseudodistomin C auch eine Cytotoxizität gegen menschliche HeLa-abgeleitete Krebszellen KB aufweist. In der Einleitung wird ausführlich auf Vorkommen, Struktur, Biogenese, pharmakologische Perspektiven und literaturbekannten Synthesen dieser amphiphilen Piperidin-Alkaloide eingegangen. Im Hauptteil wird zunächst eine gescheiterte Synthese ausgehend von D-Ribose über das Konzept einer Tandem Wittig-[3+2]-Cycloaddition beschrieben. Daraufhin wird ein völlig neuer Syntheseweg vorgestellt, welcher den formalen Aufbau des Pseudodistomin C über einen bekannten Piperidin-Grundkörper ermöglich. Des weiteren konnte das vollständig geschützte Pseudodistomin E synthetisiert werden.
Pierre Corneille (1606-1684) gehört neben Racine (1639-1699) und Molière (1622-1673) zu den drei großen Autoren des klassischen französischen Theaters. Seine erste Tragödie "Médée" wurde von Publikum und Kritikern zurückgewiesen. Corneille hatte seinen typischen Stil noch nicht entwickelt: Das Drama oszilliert zwischen Komödie und Tragödie, ist einerseits dem Handlungstheater des Barock verpflichtet und wahrt andererseits die Einheiten der "doctrine classique". Der französische Dramatiker rezipiert in seinem Stück den griechischen Tragiker Euripides (480-406 v. Chr.) und den römischen Autor Seneca (1-65 n. Chr.). Gerade den Stil Senecas ahmt er nach. Dieses Buch soll einen Beitrag zum besseren Verständnis der französischen Klassik leisten: Zwar spielt die direkte Rezeption der senecanischen Tragödie eine wichtige Rolle für das klassische französische Drama, doch Form und dramatische Technik resultierten vor allem aus der Beschäftigung mit antiken Poetiken.
The treatment landscape in multiple myeloma (MM) is shifting from genotoxic drugs to immunotherapies. Monoclonal antibodies, immunoconjugates, T-cell engaging antibodies and CART cells have been incorporated into routine treatment algorithms, resulting in improved response rates. Nevertheless, patients continue to relapse and the underlying mechanisms of resistance remain poorly understood. While Impaired death receptor signaling has been reported to mediate resistance to CART in acute lymphoblastic leukemia, this mechanism yet remains to be elucidated in context of novel immunotherapies for MM. Here, we describe impaired death receptor signaling as a novel mechanism of resistance to T-cell mediated immunotherapies in MM. This resistance seems exclusive to novel immunotherapies while sensitivity to conventional anti-tumor therapies being preserved in vitro. As a proof of concept, we present a confirmatory clinical case indicating that the FADD/BID axis is required for meaningful responses to novel immunotherapies thus we report impaired death receptor signaling as a novel resistance mechanism to T-cell mediated immunotherapy in MM.
Georg Sticker war Arzt, Hochschullehrer, Hygieniker und Medizinhistoriker. Das Themenspektrum seiner Arbeiten ist sehr umfangreich. Der Schwerpunkt dieser Arbeit liegt auf seinen Hauptforschungsgebieten, der Medizingeschichte und hier vor allem auf der Seuchengeschichte zu Pest, Lepra und Syphilis und der Bedeutung der Geschichte der Epidemien für die Epidemiologie; sowie seinen großen ärztlichen Vorbildern Hippokrates und Paracelsus. Auch über Naturheilkunst und Gesundheit und Erziehung berichtet er mit großem Engagement.Nach seiner Berufung an die Universität Würzburg beendet er seine ärztliche Tätigkeit, gründet hier im Jahr 1921 das Institut für Geschichte der Medizin und widmet sich ganz der Medizingeschichte

Staphylococcus (S.) aureus is a leading cause of bacterial infection world-wide, and currently no vaccine is available for humans. Vaccine development relies heavily on clinically relevant infection models. However, the suitability of mice for S. aureus infection models has often been questioned, because experimental infection of mice with human-adapted S. aureus requires very high infection doses. Moreover, mice were not considered to be natural hosts of S. aureus. The latter has been disproven by our recent findings, showing that both laboratory mice, as well as wild small mammals including mice, voles, and shrews, are naturally colonized with S. aureus. Here, we investigated whether mouse-and vole-derived S. aureus strains show an enhanced virulence in mice as compared to the human-adapted strain Newman. Using a step-wise approach based on the bacterial genotype and in vitro assays for host adaptation, we selected the most promising candidates for murine infection models out of a total of 254 S. aureus isolates from laboratory mice as well as wild rodents and shrews. Four strains representing the clonal complexes (CC) 8, 49, and 88 (n = 2) were selected and compared to the human-adapted S. aureus strain Newman (CC8) in murine pneumonia and bacteremia models. Notably, a bank vole-derived CC49 strain, named DIP, was highly virulent in BALB/c mice in pneumonia and bacteremia models, whereas the other murine and vole strains showed virulence similar to or lower than that of Newman. At one tenth of the standard infection dose DIP induced disease severity, bacterial load and host cytokine and chemokine responses in the murine bacteremia model similar to that of Newman. In the pneumonia model, DIP was also more virulent than Newman but the effect was less pronounced. Whole genome sequencing data analysis identified a pore-forming toxin gene, lukF-PV(P83)/lukM, in DIP but not in the other tested S. aureus isolates. To conclude, the mouse-adapted S. aureus strain DIP allows a significant reduction of the inoculation dose in mice and is hence a promising tool to develop clinically more relevant infection models.
Introduction:
Evidence from a number of open-label, uncontrolled studies has suggested that rituximab may benefit patients with autoimmune diseases who are refractory to standard-of-care. The objective of this study was to evaluate the safety and clinical outcomes of rituximab in several standard-of-care-refractory autoimmune diseases (within rheumatology, nephrology, dermatology and neurology) other than rheumatoid arthritis or non-Hodgkin’s lymphoma in a real-life clinical setting.
Methods:
Patients who received rituximab having shown an inadequate response to standard-of-care had their safety and clinical outcomes data retrospectively analysed as part of the German Registry of Autoimmune Diseases. The main outcome measures were safety and clinical response, as judged at the discretion of the investigators.
Results:
A total of 370 patients (299 patient-years) with various autoimmune diseases (23.0% with systemic lupus erythematosus, 15.7% antineutrophil cytoplasmic antibody-associated granulomatous vasculitides, 15.1% multiple sclerosis and 10.0% pemphigus) from 42 centres received a mean dose of 2,440 mg of rituximab over a median (range) of 194 (180 to 1,407) days. The overall rate of serious infections was 5.3 per 100 patient-years during rituximab therapy. Opportunistic infections were infrequent across the whole study population, and mostly occurred in patients with systemic lupus erythematosus. There were 11 deaths (3.0% of patients) after rituximab treatment (mean 11.6 months after first infusion, range 0.8 to 31.3 months), with most of the deaths caused by infections. Overall (n = 293), 13.3% of patients showed no response, 45.1% showed a partial response and 41.6% showed a complete response. Responses were also reflected by reduced use of glucocorticoids and various immunosuppressives during rituximab therapy and follow-up compared with before rituximab. Rituximab generally had a positive effect on patient well-being (physician’s visual analogue scale; mean improvement from baseline of 12.1 mm)
Mycotoxins in agriculturally used plants can cause intoxication in animals and can lead to severe financial losses for farmers. The endophytic fungus Epichloë festucae var. lolii living symbiotically within the cool season grass species Lolium perenne can produce vertebrate and invertebrate toxic alkaloids. Hence, an exact quantitation of alkaloid concentrations is essential to determine intoxication risk for animals. Many studies use different methods to detect alkaloid concentrations, which complicates the comparability. In this study, we showed that alkaloid concentrations of individual plants exceeded toxicity thresholds on real world grasslands in Germany, but not on the population level. Alkaloid concentrations on five German grasslands with high alkaloid levels peaked in summer but were also below toxicity thresholds on population level. Furthermore, we showed that alkaloid concentrations follow the same seasonal trend, regardless of whether plant fresh or dry weight was used, in the field and in a common garden study. However, alkaloid concentrations were around three times higher when detected with dry weight. Finally, we showed that alkaloid concentrations can additionally be biased to different alkaloid detection methods. We highlight that toxicity risks should be analyzed using plant dry weight, but concentration trends of fresh weight are reliable.
Propagule pressure and an invasion syndrome determine invasion success in a plant community model

The success of species invasions depends on multiple factors, including propagule pressure, disturbance, productivity, and the traits of native and non-native species. While the importance of many of these determinants has already been investigated in relative isolation, they are rarely studied in combination. Here, we address this shortcoming by exploring the effect of the above-listed factors on the success of invasions using an individual-based mechanistic model. This approach enables us to explicitly control environmental factors (temperature as surrogate for productivity, disturbance, and propagule pressure) as well as to monitor whole-community trait distributions of environmental adaptation, mass, and dispersal abilities. We simulated introductions of plant individuals to an oceanic island to assess which factors and species traits contribute to invasion success. We found that the most influential factors were higher propagule pressure and a particular set of traits. This invasion trait syndrome was characterized by a relative similarity in functional traits of invasive to native species, while invasive species had on average higher environmental adaptation, higher body mass, and increased dispersal distances, that is, had greater competitive and dispersive abilities. Our results highlight the importance in management practice of reducing the import of alien species, especially those that display this trait syndrome and come from similar habitats as those being managed.

Schafe leben seit einigen Jahrtausenden mit Menschen zusammen und mit ihnen durchwandert ihre Wolle unsere Kulturen und Gesellschaften. Ausgangspunkt der vorliegenden Arbeit ist der gegenwärtig geringe monetäre Wert von Schafwolle, der in den letzten dreißig Jahren in Deutschland stark gesunken ist.
Basierend auf ethnografischen Methoden der Feldforschung, teilnehmende Beobachtung und Interviews, verfolgt die vorliegende Arbeit Spuren von Schafwolle und fragt dabei nach den Berührungs- und Kontaktpunkten, die im Alltag zwischen Schafen, Menschen und Wolle entstehen. Menschen, die mit Schafen leben, werfen verschiedene Perspektiven auf die sogenannte Nutztierhaltung sowie auf ländliches Wirtschaften und Leben.
Schafe und Schafwolle werden hier als Miterzählende und Mitgestaltende im Prozess der Entstehung von Schaf-Woll-Geschichten betrachtet. Besonders die Perspektiven auf Schafwolle regen an, tierliche Materialitäten im Allgemeinen neu zu denken und zu reflektieren. So lädt die vorliegende Arbeit dazu ein, Menschen, Tiere und Materialitäten stärker in den Wissensgenerierungsprozess zu integrieren sowie nach dem besonderen Wissen zu suchen, das eben jene mehr-als-menschliche Akteur*innen mit uns teilen.
The field of photopharmacology has attracted considerable attention due to applying the spatial and temporal precision of light to pharmacological systems. Photoswitchable biologically active compounds have proven useful in the field of G protein-coupled receptors (GPCRs), which are of tremendous therapeutic relevance. Generally, the pharmacology of GPCRs is complex, perhaps even more complex than originally thought. Suitable tools are required to dissect the different signalling pathways and mechanisms and to unravel how they are connected in a holistic image. This is reflected in the enormous scientific interest in CB2R, as the neuroprotective and immunomodulatory effects attributed to CB2R agonists have not yet translated into effective therapeutics. This work focused on the development of a novel photoswitchable scaffold based on the privileged structure of benzimidazole and its application in photoswitchable CB2R ligands as photopharmacological tools for studying the CB2R.
The visible-light photoswitchable ligand 10d enables the investigation of CB2R activation with regard to βarr2 bias, exhibiting a unique pharmacological profile as a “cis-on” affinity switch at receptor level and as a “trans-on” efficacy-switch in βarr2-mediated receptor internalization. The novel photoswitchable scaffold developed in this work further serves as a guide for the development of novel photoswitchable GPCR ligands based on the privileged structure of benzimidazole. To obtain a different tool compound for studying CB2R activation and signalling mechanisms, a previously reported putatively dualsteric CB2R ligand was rendered photoswitchable, by linking the orthosteric agonist to a CB2R-selective PAM via photoswitchable azobenzene. Compound 27-para exhibits a desirable “cis-on” behaviour across all investigated assays with >10-fold higher potency compared to its trans-isomer and can be used as an efficacy-switch employing specific concentrations.
Long-term sequelae in hospitalized Coronavirus Disease 2019 (COVID-19) patients may result in limited quality of life. The current study aimed to determine health-related quality of life (HRQoL) after COVID-19 hospitalization in non-intensive care unit (ICU) and ICU patients. This is a single-center study at the University Hospital of Wuerzburg, Germany. Patients eligible were hospitalized with COVID-19 between March 2020 and December 2020. Patients were interviewed 3 and 12 months after hospital discharge. Questionnaires included the European Quality of Life 5 Dimensions 5 Level (EQ-5D-5L), patient health questionnaire-9 (PHQ-9), the generalized anxiety disorder 7 scale (GAD-7), FACIT fatigue scale, perceived stress scale (PSS-10) and posttraumatic symptom scale 10 (PTSS-10). 85 patients were included in the study. The EQ5D-5L-Index significantly differed between non-ICU (0.78 ± 0.33 and 0.84 ± 0.23) and ICU (0.71 ± 0.27; 0.74 ± 0.2) patients after 3- and 12-months. Of non-ICU 87% and 80% of ICU survivors lived at home without support after 12 months. One-third of ICU and half of the non-ICU patients returned to work. A higher percentage of ICU patients was limited in their activities of daily living compared to non-ICU patients. Depression and fatigue were present in one fifth of the ICU patients. Stress levels remained high with only 24% of non-ICU and 3% of ICU patients (p = 0.0186) having low perceived stress. Posttraumatic symptoms were present in 5% of non-ICU and 10% of ICU patients. HRQoL is limited in COVID-19 ICU patients 3- and 12-months post COVID-19 hospitalization, with significantly less improvement at 12-months compared to non-ICU patients. Mental disorders were common highlighting the complexity of post-COVID-19 symptoms as well as the necessity to educate patients and primary care providers about monitoring mental well-being post COVID-19.

Das Ziel dieser Arbeit war es, eine im Arbeitskreis entwickelte Methode zur Darstellung von unterschiedlich substituierten -Aminopiperidinen zu etablieren und zu verifizieren, indem unser Konzept einer Tandemreaktion zum Einsatz kommen sollte. Diese Reaktionssequenz sollte anschließend zur Totalsynthese von Microconin, einem aus Microcos paniculata isoliertem Alkaloid, genutzt werden. Den ersten Schritt in Richtung -Aminopiperidinderivate machte die Veresterung von L-Pyroglutaminsäure. Nach der Reduktion des Methylesters erfolgte die Aktivierung der Hydroxyfunktion des Alkohols in Form des Tosylats. Die Azideinführung resultierte aus einer nucleophilen Substitutionsreaktion, anschließend daran wurde der Lactam-Stickstoff mit Boc2O und einer katalytischen Menge DMAP geschützt. Das Lactam ist durch den Elektronenakzeptor aktiviert, so dass die Ringöffnung zum Methylester unter sehr milden Bedingungen und ohne weiteren Reinigungsschritt erfolgen konnte. Die Aminofunktion musste mit einer zweiten Schutzgruppe blockiert werden, die anschließende selektive Reduktion mit DiBAl-H in trockenem Ether verlief problemlos und lieferte mit dem Aldehyd das Edukt für Olefinierungen mittels verschiedener Wittig-Reaktionen. Dafür wurden stabilisierte Phosphonate hergestellt und in einer HWE-Reaktion mit dem Aldehyd umgesetzt. Die resultierenden elektronenarmen Olefine gingen dann die intramolekulare Cycloaddition mit dem Azidsubstituenten ein und bauten so den Grundkörper der -Aminopiperidinderivate in einer Reaktionssequenz auf, die wir als Tandem Wittig-[3+2]-Cycloaddition bezeichnen. Die Bildung der primären Triazoline erfolgte stereoselektiv, die Geschwindigkeit der Cycloaddition hing sowohl vom konjugierten Olefinsubstituenten als auch vom vicinalen Substituenten der Azidfunktion ab. Die Konfigurationsbestimmung erfolgte mittels NMR-Spektroskopie durch Analyse der Kopplungskonstanten und NOE-Messungen. Die asymmetrische Induktion der Cycloaddition konnte direkt für die Stereochemie am Piperidinring genutzt werden, indem, nach basischer Triazolin/Diazoamin Isomerisierung, gleich das Diazoamin hydriert wurde. Die Hydrierung der vinylogen Urethane, den Produkten aus der rhodiumkatalysierten Stickstoffextrusion, lieferte ein Diastereomerengemisch, wobei das Verhältnis der Diastereomere hauptsächlich vom Substitutionsgrad des exocyclischen Amins abhängig war. Überraschenderweise fand beim Sulfontriazolin keine Isomerisierung zum Diazoamin statt, daher musste für die Darstellung der Sulfonylmethyl--aminopiperidine eine alternative Route über ein Ketosulfon beschritten werden. Die Synthese von Microconin begann mit der Desoxygenierung von L-Rhamnose durch die sog. Fischer-Zach-Reaktion. Das Rhamnal wurde in einer drei Stufen Eintopfreaktion erhalten und mittels Perlinhydrolyse in den offenkettigen Aldehyd umgewandelt. Die Aktivierung der Hydroxyfunktion als Mesylat resultierte in einer äußerst empfindlichen Verbindung, die nur durch Verwendung des Lindlar-Katalysators mit zufrieden stellenden Ergebnis zum aliphatischen Aldehyd reduziert werden konnte. Eine bimolekulare nucleophile Substitutionsreaktion lieferte bei der Azideinführung zur Schlüsselverbindung sowohl die benötigte funktionelle Gruppe als auch die benötigte Inversion der Konfiguration. Die Tandem HWE-[3+2]-Cycloadditions-Reaktion führte auch bei dem Sulfontriazolin in eine Sackgasse, weshalb wieder eine alternative Syntheseroute eingeschlagen werden musste. Ausgehend von derselben Schlüsselverbindung gelang dies durch eine zinnkatalysierte Umsetzung mit stabilisierten Diazomethan zum Ketosulfon. Der Aufbau des Piperidin-Heterocyclus konnte dann wieder über eine intramolekulare Imin-Bildung des intermediären Amins mit dem Keton erzielt werden. Die diastereoselektive Hydrierung verlief unter Wasserstoffaddition von der sterisch weniger gehinderten -Seite und Ausbildung des all cis Substitutionsmusters. Nach dem erfolgreichen Aufbau des Heterocyclus mussten noch die beiden Heteroatome methyliert werden. Dabei wurden die besten Ergebnisse am Ringstickstoff mit der reduktiven Aminierung erzielt. Bei der anschließenden Abspaltung der Acetylgruppe zeigte sich erstmals, dass das Substitutionsmuster am Piperidinring nicht konfigurationsstabil war, da neben dem erwarteten Alkohol auch das Diastereomer isoliert wurde. Die genaue Ursache für die Epimerisierung nach der N-Methylierung konnte nicht geklärt werden. Die Einführung des Methoxy-Substituenten am Grundkörper erfolgte über eine Williamsonschen Ethersynthese. Bei den Versuchen zur Kupplung des Grundkörpers mit der Seitenkette 2,4-Nonadienal erwies sich der Zusatz von HMPT als förderlich. Weiterhin konnte die Ausbeute an -Hydroxysulfonen durch das Erwärmen der Reaktionsmischung gesteigert werden. In den Folgeschritten der Julia-Olefinierung blieben die Versuche zur Ausbildung der dreifach ungesättigten Struktureinheit in der Seitenkette des isolierten Naturstoffs jedoch erfolglos.
Current data show that resilience is an important factor in cancer patients’ well-being. We aim to explore the resilience of patients with lower grade glioma (LGG) and the potentially influencing factors. We performed a cross-sectional assessment of adult patients with LGG who were enrolled in the LoG-Glio registry. By phone interview, we administered the following measures: Resilience Scale (RS-13), distress thermometer, Montreal Cognitive Assessment Test for visually impaired patients (MoCA-Blind), internalized stigmatization by brain tumor (ISBI), Eastern Cooperative Oncological Group performance status (ECOG), patients’ perspective questionnaire (PPQ) and typical clinical parameters. We calculated correlations and multivariate regression models. Of 74 patients who were assessed, 38% of those showed a low level of resilience. Our results revealed significant correlations of resilience with distress (p < 0.001, −0.49), MOCA (p = 0.003, 0.342), ECOG (p < 0.001, −0.602), stigmatization (p < 0.001, −0.558), pain (p < 0.001, −0.524), and occupation (p = 0.007, 0.329). In multivariate analyses, resilience was negatively associated with elevated ECOG (p = 0.020, β = −0.383) and stigmatization levels (p = 0.008, β = −0.350). Occupation showed a tendency towards a significant association with resilience (p = 0.088, β = −0.254). Overall, low resilience affected more than one third of our cohort. Low functional status is a specific risk factor for low resilience. The relevant influence of stigmatization on resilience is a novel finding for patients suffering from a glioma and should be routinely identified and targeted in clinical routine.

Brain metastases are the most severe tumorous spread during breast cancer disease. They are associated with a limited quality of life and a very poor overall survival. A subtype of extracellular vesicles, exosomes, are sequestered by all kinds of cells, including tumor cells, and play a role in cell-cell communication. Exosomes contain, among others, microRNAs (miRs). Exosomes can be taken up by other cells in the body, and their active molecules can affect the cellular process in target cells. Tumor-secreted exosomes can affect the integrity of the blood-brain barrier (BBB) and have an impact on brain metastases forming. Serum samples from healthy donors, breast cancer patients with primary tumors, or with brain, bone, or visceral metastases were used to isolate exosomes and exosomal miRs. Exosomes expressed exosomal markers CD63 and CD9, and their amount did not vary significantly between groups, as shown by Western blot and ELISA. The selected 48 miRs were detected using real-time PCR. Area under the receiver-operating characteristic curve (AUC) was used to evaluate the diagnostic accuracy. We identified two miRs with the potential to serve as prognostic markers for brain metastases. Hsa-miR-576-3p was significantly upregulated, and hsa-miR-130a-3p was significantly downregulated in exosomes from breast cancer patients with cerebral metastases with AUC: 0.705 and 0.699, respectively. Furthermore, correlation of miR levels with tumor markers revealed that hsa-miR-340-5p levels were significantly correlated with the percentage of Ki67-positive tumor cells, while hsa-miR-342-3p levels were inversely correlated with tumor staging. Analysis of the expression levels of miRs in serum exosomes from breast cancer patients has the potential to identify new, non-invasive, blood-borne prognostic molecular markers to predict the potential for brain metastasis in breast cancer. Additional functional analyzes and careful validation of the identified markers are required before their potential future diagnostic use.
Background: The clinical management of teeth with complex dens invaginatus (DI) malformations and apical periodontitis may be challenging due to the lack of routine. The aim of this case report is to describe the endodontic treatment of an immature tooth with DI and to discuss strategies for preclinical training for teeth with such malformations. Case report: A 9-year-old male presented with an immature maxillary incisor with DI (Oehlers Type II) and apical periodontitis which was diagnosed by cone beam computed tomography (CBCT). Revitalization was initially attempted but then abandoned after failure to generate a stable blood clot. Nevertheless, considerable increase in both root length and thickness could be detected after medication with calcium hydroxide followed by root canal filling with MTA as an apical plug. Conclusions: The endodontic management of teeth with DI requires thorough treatment planning. In immature teeth, under certain conditions, root maturation may occur even with conventional apexification procedures. From an educational perspective, different strategies including CBCT and 3D-printed transparent tooth models for visualization of the complex internal morphology and redesigned 3D-printed replica with various degrees of difficulty for endodontic training, can be used to overcome the challenges associated with endodontic treatment of such teeth.
Objectives
Magnesium phosphate-based cements begin to catch more attention as bone substitute materials and especially as alternatives for the more commonly used calcium phosphates. In bone substitutes for augmentation purposes, atraumatic materials with good biocompatibility and resorbability are favorable. In the current study, we describe the in vivo testing of novel bone augmentation materials in form of spherical granules based on a calcium-doped magnesium phosphate (CaMgP) cement.
Materials and Methods
Granules with diameters between 500 and 710 μm were fabricated via the emulsification of CaMgP cement pastes in a lipophilic liquid. As basic material, two different CaMgP formulations were used. The obtained granules were implanted into drill hole defects at the distal femoral condyle of 27 New Zealand white rabbits for 6 and 12 weeks. After explantation, the femora were examined via X-ray diffraction analysis, histological staining, radiological examination, and EDX measurement.
Results
Both granule types display excellent biocompatibility without any signs of inflammation and allow for proper bone healing without the interposition of connective tissue. CaMgP granules show a fast and continuous degradation and enable fully adequate bone regeneration.
Conclusions
Due to their biocompatibility, their degradation behavior, and their completely spherical morphology, these CaMgP granules present a promising bone substitute material for bone augmentation procedures, especially in sensitive areas.
Clinical Relevance
The mostly insufficient local bone supply after tooth extractions complicates prosthetic dental restoration or makes it even impossible. Therefore, bone augmentation procedures are oftentimes inevitable. Spherical CaMgP granules may represent a valuable bone replacement material in many situations.
Aspergillus fumigatus causes life-threatening opportunistic infections in immunocompromised patients. As therapeutic outcomes of invasive aspergillosis (IA) are often unsatisfactory, the development of targeted immunotherapy remains an important goal. Linking the innate and adaptive immune system, dendritic cells are pivotal in anti-Aspergillus defense and have generated interest as a potential immunotherapeutic approach in IA. While monocyte-derived dendritic cells (moDCs) require ex vivo differentiation, antigen-pulsed primary myeloid dendritic cells (mDCs) may present a more immediate platform for immunotherapy. To that end, we compared the response patterns and cellular interactions of human primary mDCs and moDCs pulsed with an A. fumigatus lysate and two A. fumigatus proteins (CcpA and Shm2) in a serum-free, GMP-compliant medium. CcpA and Shm2 triggered significant upregulation of maturation markers in mDCs and, to a lesser extent, moDCs. Furthermore, both A. fumigatus proteins elicited the release of an array of key pro-inflammatory cytokines including TNF-α, IL-1β, IL-6, IL-8, and CCL3 from both DC populations. Compared to moDCs, CcpA- and Shm2-pulsed mDCs exhibited greater expression of MHC class II antigens and stimulated stronger proliferation and IFN-γ secretion from autologous CD4\(^+\) and CD8\(^+\) T-cells. Moreover, supernatants of CcpA- and Shm2-pulsed mDCs significantly enhanced the oxidative burst in allogeneic neutrophils co-cultured with A. fumigatus germ tubes. Taken together, our in vitro data suggest that ex vivo CcpA- and Shm2-pulsed primary mDCs have the potential to be developed into an immunotherapeutic approach to tackle IA.
Digital platforms, such as Amazon, represent the major beneficiaries of the Covid‐19 crisis. This study examines the role of digital platforms and their engagement in digitalisation initiatives targeting (small) brick‐and‐mortar retailers in Germany, thereby contributing to a better understanding of how digital platforms augment, substitute or reorganise physical retail spaces. This study applies a mixed‐method approach based on qualitative interviews, participant observation as well as media analysis. First, the study illustrates the controversial role of digital platforms by positioning themselves as supporting partners of the (offline) retailers, while simultaneously shifting power towards the platforms themselves. Second, digital platforms have established themselves not only as infrastructure providers but also as actors within these infrastructures, framing digital as well as physical retail spaces, inter alia due to their role as publicly legitimised retail advisers. Third, while institutions want to help retailers to survive, they simultaneously enhance retailers' dependency on digital platforms.

Das vorliegende Buch erscheint 2023 und damit 75 Jahre nach der "Allgemeinen Erklärung der Menschenrechte" der Vereinten Nationen. Menschenrechte sind eine gegen viele Widerstände und mit großen Opfern durchgesetzte Errungenschaft der Menschheit. Heute werden sie erneut zu einem verletzten und verletzbaren Gut.
Daher analysiert das Buch die ambivalente Macht der Vulnerabilität an signifi kanten Diskursorten der Gegenwart; es legt die Vulneranz der katholischen Kirche bezüglich Missbrauch, Vertuschung und Gendertrouble frei; und es fragt nach der offenbarenden Kraft an jener Schwelle, wo die Menschenrechte in Gefahr sind.
