Key elements of sacred landscapes of the Nile Delta were lakes, canals and artificial basins connected to temples, which were built on elevated terrain. In the case of temples of goddesses of an ambivalent, even dangerous, nature, i.e. lioness goddesses and all female deities who could appear as such, the purpose of sacred lakes and canals exceeded their function as a water resource for basic practical and religious needs. Their pleasing coolness was believed to calm the goddess' fiery nature, and during important religious festivals, the barques of the goddesses were rowed on those waters. As archaeological evidence was very rare in the past, the study of those sacred waters was mainly confined to textual sources. Recently applied geoarchaeological methods, however, have changed this situation dramatically: they allow in-depth investigations and reconstructions of these deltaic sacred landscapes. Exploring these newly available data, the paper presented here focuses on the sites of Buto, Sais and Bubastis, by investigating the characteristics of their sacred lakes, canals and marshes with respect to their hydrogeographical and geomorphological context and to their role in ancient Egyptian religion and mythology as well.
Having control over the landscape played an important role in the geography and economy of Egypt from the predynastic period onwards. Especially from the beginning of the Old Kingdom, we have evidence that kings created new places (funerary domains) called (centers) and (Ezbah) for the equipment of the building projects of the royal tomb and the funerary cult of the king, as well as to ensure the eternal life of both kings and individuals. Kings used these localities in order to do so, and they oftentimes expanded the border of an existing nome and created new establishments. Consequently, these establishments were united or divided into new nomes. The paper discusses the geography of Lower Egypt and the associated royal domains in the early Fifth Dynasty based on the new discoveries from the causeway of Sahura at Abusir.
Geoarchaeological information presented here pertains to a subsidiary Nile channel that once flowed west of the main Sebennitic distributary and discharged its water and sediments at Egypt’s then north-central deltaic coast. Periodical paleoclimatic episodes during the later Middle and Upper Holocene included decreased rainfall and increased aridity that reduced the Nile’s flow levels and thus likely disrupted nautical transport and anthropogenic activity along this channel. Such changes in this deltaic sector, positioned adjacent to the Levantine Basin in the Eastern Mediterranean, can be attributed to climatic shifts triggered as far as the North Atlantic to the west, and African highland source areas of the Egyptian Nile to the south. Of special interest in a study core recovered along the channel are several sediment sequences without anthropogenic material that are interbedded between strata comprising numerous potsherds. The former are interpreted here as markers of increased regional aridity and reduced Nile flow which could have periodically disrupted the regional distribution of goods and nautical activities. Such times occurred ~5000 years B.P., ~4200–4000 years B.P., ~3200–2800 years B.P., ~2300–2200 years B.P., and more recently. Periods comparable to these are also identified by altered proportions of pollen, isotopic and compositional components in different radiocarbon-dated Holocene cores recovered elsewhere in the Nile delta, the Levantine region to the east and north of Egypt, and in the Faiyum depression south of the delta.

Cancer is one of the major causes of mortality in developed countries. In 2020, there were more than 19.3 million new cases of tumor malignancies worldwide, with more than 10 million deaths. The high rates of cancer cases and mortality necessitate extensive research and the development of novel cancer treatments and antitumor agents. In most cases, conventional treatment strategies for tumor therapy are based on chemotherapeutic treatment, which is supplemented with radiotherapy and/or surgical resection of solid tumors [1]. The use of chemotherapy for the treatment of cancer has significant side effects, the most dangerous of which is toxicity [2] [3].
Modern methods of treating tumors focus on specific drug delivery to the tumor site, actively targeting the tumor cells, as well as the reduction of side effects. One of the most promising current approaches is based on oncolytic viruses. Antitumor properties of viruses were documented at the beginning of the 20th century when some cancer patients recovered after acute viral infections, particularly influenza [4]. Vaccinia virus (VACV) is a member of the Poxviridae family, has natural antitumor properties, and provides a good basis for generating efficient recombinant oncolytic strains. Furthermore, VACV has never been shown to integrate into the host genome [5]. VACV is likely one of the safest and well-studied viruses due to extensive research being done in molecular biology and pathophysiology to investigate its potential as a vaccine for smallpox eradication programs. It has been administered to over 200 million people worldwide. VACV antitumor therapeutic effectiveness has been established in xenograft models with a variety of tumor types for human and canine cancers. Furthermore, recombinant oncolytic VACVs expressing genes encoding light-emitting proteins are a big improvement in a treatment strategy that combines tumor-specific therapies and diagnostics.
Oncolytic virus treatments are effective in xenograft cancer models in mice, however, the significant improvements found in mice do not always translate to human cancer patients. These therapies should be tested in dogs with spontaneous cancer not only to offer well translatable information regarding the possible efficiency of viral therapy for human cancers but also to improve the health of our household pets as well. Spontaneous canine tumors are starting to be regarded as an essential model of human cancers that can reproduce the tumor microenvironment and immune response of cancer patients [6]. Just as data obtained in dog experiments can improve cancer therapy for human patients, these findings can also be used to improve treatment protocols in canine patients.
Hundreds of studies and dozens of reviews have been published regarding the antitumor effects of various recombinants of VACV, but information on the anticancer features of initial, genetically-unmodified “naïve” VACV is still limited. In the first studies, we compared different wild-type, non-modified strains of VACV and tested their oncolytic properties on a panel of various cancer cells derived from different organs. In addition, we also tested a protection system based on the “Trojan horse” concept - using a combination of human Adipose tissue-derived Stem Cells (hADSC) and three different wild-type single plaque purified Vaccinia virus strains: W1, L1, and T1. We showed that all tested human cell lines (FaDu, MDA MB 231, HNT-13, HNT-35, and PC-3) are permissive to L0, W0, T0, L1, W1, and L1 infection. Furthermore, we tested the cytotoxicity of VACV in different cancer cell lines (A549, PC-3, MDA-MB 231, FaDu, HNT-13, HNT-25, and HNT-35). All strains lysed the cells, which was most visible at 96 hpi. We also showed that all tested strains could efficiently infect and multiply in hADSC at a high level. In our in vivo study, we tested the therapeutic efficacy of the wild-type Vaccinia viruses L1, W1, and T1 alone or in combination with hADSC. Wild-type VACV strains were tested for their oncolytic efficiency in human lung adenocarcinoma (A549) in a xenograft model. Treatment of A549 tumors with different doses of L1 and W1 as well as with a L1/ADSC or W1/ADSC combination led to significant tumor regression compared to the PBS control. Additionally, the treatment with L1 and W1 and the combination of L1/ADSC and W1/ADSC was well tolerated by the animals. In the case of the wild-type Tian Tan strain, results were not obtained due to the high cytotoxicity of this strain. Therefore, it should be attenuated for further studies.
In the second part of the current study, we investigated the oncolytic effect of C1-opt1, W1 opt1, and L3-opt1 strains based on the wild-type Copenhagen, Wyeth, and Lister vaccines with additional expression of turboFP635. Replication and cytotoxicity assays demonstrated that all 3 viruses were able to infect, replicate in and kill canine tumor cell lines STSA-1 and CT1258 in a virus dose- and time- dependent fashion. Cytotoxicity and replication assays were also performed on cultured canine Adipose-derived Mesenchymal Stem Cells (cAdMSC). The results showed that the cells were lysed much slower than the tumor cells. It suggests that these cells can harbour the virus for a long-term period, allowing the virus to spread into the body and there is enough time to reach the primary tumor or metastases before the cell carrier is destroyed. The viral replication in cAdMSC in our study was lower than in canine cancer cells (STSA-1 and CT1258) at the same MOI. After being studied in cell culture, C1 opt1 and their combination with cAdMSC (C1-opt1/cAdMSC) were used in canine STSA 1 tumor bearing nude mice. We tested the oncolytic effect of the C1-opt1 virus alone and in combination with cAdMSC in the canine STSA-1 xenograft mouse model. Altogether, our findings have shown that both C1-opt1 and cAdMSC/C1-opt1 significantly reduced tumor size or eliminated the tumor. There was no significant difference between C1-opt1 alone and cAdMSC/C1-opt1. The virus particles were mostly found within the tumor after 24 dpi, some amount of virus particles were found in the lungs of mice injected with a combination of cAdMSC/C1-opt1 but not in the group injected with virus alone (cAdMSC might get stuck in the lungs and cause virus propagation there).
Taken together, this study provided a proof-of-concept that hADSC/cAdMSC can be used as a carrier system for the “Trojan horse” concept. However, it should be confirmed in another experimental model system, such as canine patients. Moreover, these findings suggest that wild-type, non-modified strains of Vaccinia virus isolates can be considered promising candidates for oncolytic virotherapy, especially in combination with mesenchymal stem cells.
Um den jahrtausendealten Weg der Menschheit vom Papyrus über Buchdruck und siliziumbasierte Halbleiter in Richtung noch leistungsfähigerer Technologien zu gehen und weiterhin Heureka-Momente zu schaffen, bieten Kohlenstoffnanoröhren ein weites Forschungsfeld. Besonders die halbleitenden Charakteristika von SWNTs sowie die Manipulation dieser durch Dotierung bergen viele Möglichkeiten für zukünftige Anwendungen in moderner Elektrotechnologie. Der Weg zu einer industriellen Implementierung von SWNTs in neuartigen optoelektronischen Bauteilen ließe sich durch eine Ausweitung des Wissens bezüglich SWNTs und der dotierungsbasierten Anpassung ihrer Eigenschaften ebnen.
Mit dieser Erkenntniserweiterung als Zielsetzung wurden im Rahmen dieser Dissertation halbleitende, einwandige (6,5)-Kohlenstoffnanoröhren als chiralitätsreine, polymerstabilisierte Proben untersucht. Die ultrakurzzeitaufgelöste Spektroskopie der SWNTs erfolgte an organischen Suspensionen wie auch Dünnschichtfilmen, die je mittels eines gewissen Quantums an Gold(III)-chlorid dotiert worden waren. So konnten die ablaufenden Dynamiken auf einer ps-Zeitskala untersucht werden.
In Kapitel 4 konnte mittels transienter Absorptionsexperimente an redoxchemisch p-dotierter SWNT-Suspensionen zunächst gezeigt werden, dass sich die bei optischer Anregung gebildeten Trionen nicht analog zu Exzitonen diffusiv entlang der Nanoröhre bewegen, sondern lokalisiert vorliegen. Die längere trionischen Zerfallsdauer nach X$_1$- verglichen mit X$_1^+$-resonanter Anregung zeugt außerdem davon, dass das Trion aus dem Exziton gespeist wird. Der Einfluss der Dotierung auf die Zerfallsdynamiken von X$_1$ und X$_1^+$ wurde an SWNT-Dünnschichtfilmen untersucht. Das Photobleichsignal des Exzitons verschiebt hypsochrom und zerfällt schneller mit zunehmender Ladungsträgerdichte durch höherer Gold(III)-chloridkonzentrationen. Dies resultiert aus dem verringerten Abstand zwischen den Ladungsträgern, welche als nichtstrahlende Löschstellen fungieren. Für das X$_1^+$-PB ist ein ähnliches Verhalten zu beobachten. Dabei wird dieses Signal mit weiter steigender Dotierung von einer der H-Bande zuzuordnenden Photoabsorption überlagert. Diese lässt sich in einer starken Sättigung der Dotierung wie auch einer hohen Bandkantenverschiebung begründen.
In Kapitel 5 wurde die Größe der Exzitonen und Trionen in dotierten SWNT-Dünnschichtfilmen mittels des Phasenraumfüllmodells bestimmt. Dabei lag besonderes Augenmerk auf der Kompensation des PB/PA-Überlapps, dem schnellen Zerfall, einem Ausgleich von Differenzen zwischen Anrege- und Absorptionsspektrum sowie dem Anteil intrinsischer/dotierter Nanorohrsegmente, um korrigierte Größen $\xi_\mathrm{k}$ zu erhalten. Für die Trionengröße wurde zusätzlich der Überlapp der Absorptionsbanden einbezogen, um korrigierte Werte $\xi_{\mathrm{T,k}}$ zu bestimmen. $\xi_\mathrm{k}$ beträgt in der intrinsischen Form 6$\pm$2\,nm und bleibt bis zu einer Ladungsträgerdichte $n_{\mathrm{LT}}<0.10$\,nm$^{-1}$ etwa gleich, anschließend ist ein Absinken bis auf etwa 4\,nm bei $n_{\mathrm{LT}}\approx0.20$\,nm$^{-1}$ zu beobachten. Für diesen Trend ist die Überlagerung von Exziton- und H-Bande verantwortlich, da so der Faktor zur Bestimmung des Anteils intrinsischer Nanorohrsegmente an der SWNT verfälscht wird. Die Abweichung der intrinsischen Größe von den in der Literatur berichteten 13$\pm$3\,nm ist möglicherweise auf Unterschiede in der Probenpräparation zurückzuführen. Für die Trionengröße ergibt sich bei steigender Dotierung ein ähnliches Verhalten: Sie beträgt für $n_{\mathrm{LT}}<0.20$\,nm$^{-1}$ 1.83$\pm$0.47\,nm, was in der Größenordnung in guter Übereinstimmung mit der Literatur ist. Für höhere Dotierungen sinkt $\xi_{\mathrm{T,k}}$ bis auf 0.92$\pm$0.26nm ab. Dies erklärt sich dadurch, dass bei höherer $n_{\mathrm{LT}}$ die H-Bande das Spektrum dominiert, sodass der Einfluss der Absorptionsbandenüberlagerung nicht mehr vollständig durch den entsprechenden Korrekturfaktor kompensiert werden kann.
Kapitel 6 beschäftigte sich anstelle redoxchemischer Dotierung der nanoskaligen Halbleiter mit der (spektro-)elektrochemischen Untersuchung von Vorläufern molekularer Radikale. SWV-Messungen weisen dabei darauf hin, dass die Pyrene Pyr1-Pyr3 entsprechend der Anzahl ihrer Substituenten bei Reduktion Mono-, Bi- beziehungsweise Tetraradikale bilden. Die strukturelle Ähnlichkeit der Moleküle äußert sich in gleichen Reduktionspotentialen wie auch ähnlichen potentialabhängigen Absorptionsspektren. Während nur marginale Unterschiede in den PL-Spektren der neutralen und reduzierten Spezies festgestellt werden konnte, lieferte das zeitkorrelierte Einzelphotonenzählen aufschlussreichere Ergebnisse: So wird die Fluoreszenzlebensdauer stark von der Polarität der Umgegbung beeinflusst - bereits die Zugabe des Leitsalzes führt hier zu Änderungen. Die durchschnittliche Fluoreszenzlebensdauer $\tau_{\mathrm{av}}$ sinkt außerdem mit Reduktion und Radikalbildung; für höhere Emissionswellenlängen ist $\tau_{\mathrm{av}}$ außerdem höher. Insgesamt verdeutlichten die Experimente die gute Abschirmung zwischen Pyrenkern und Naphthalimidsubstituenten der Moleküle sowie die Sensibilität gegenüber dem Medium durch TICT, das Vorhandensein von Bi- und Tetraradikalen kann allerdings nicht vollständig belegt werden, wofür EPR-Messugen notwendig wären.
The Amduat is one of the most important Netherworld Books which was recorded in various kinds of Ancient Egyptian sources since the beginning of the 18th dynasty, especially the walls of the royal tombs. The main theme of the Amduat is the journey of the sun god through the underworld where the solar bark and its crew is the central scene of the journey. The study focuses on finding the reasons of choosing the crew’s members who manage the sun bark’s journey in the Amduat. It also aims at illustrating the functions and responsibilities of each crew member. Following a historical approach, the study analyzes the Pyramid Texts and Coffin Texts as the most important documents before the New Kingdom, and proceeding to the inscriptions and writings of the monuments which contain portrayals and inscriptions of the Amduat in the New Kingdom. Furthermore, it sheds some light on the solar cycle’s main features and primary aspects, and tries to scrutinize the date, meaning, and symbolisms of the Amduat and its indications in the earlier sources.

Thermoplastic polymers have a history of decades of safe and effective use in the clinic as implantable medical devices. In recent years additive manufacturing (AM) saw increased clinical interest for the fabrication of customizable and implantable medical devices and training models using the patients’ own radiological data. However, approval from the various regulatory bodies remains a significant hurdle. A possible solution is to fabricate the AM scaffolds using materials and techniques with a clinical safety record, e.g. melt processing of polymers. Melt Electrowriting (MEW) is a novel, high resolution AM technique which uses thermoplastic polymers. MEW produces scaffolds with microscale fibers and precise fiber placement, allowing the control of the scaffold microarchitecture. Additionally, MEW can process medical-grade thermoplastic polymers, without the use of solvents paving the way for the production of medical devices for clinical applications. This pathway is investigated in this thesis, where the layout is designed to resemble the journey of a medical device produced via MEW from conception to early in vivo experiments. To do so, first, a brief history of the development of medical implants and the regenerative capability of the human body is given in Chapter 1. In Chapter 2, a review of the use of thermoplastic polymers in medicine, with a focus on poly(ε-caprolactone) (PCL), is illustrated, as this is the polymer used in the rest of the thesis. This review is followed by a comparison of the state of the art, regarding in vivo and clinical experiments, of three polymer melt AM technologies: melt-extrusion, selective laser sintering and MEW. The first two techniques already saw successful translation to the bedside, producing patient-specific, regulatory-approved AM implants. To follow in the footsteps of these two technologies, the MEW device parameters need to be optimized. The MEW process parameters and their interplay are further discussed in Chapter 3 focusing on the importance of a steady mass flow rate of the polymer during printing. MEW reaches a balance between polymer flow, the stabilizing electric field and moving collector to produce reproducible, high-resolution scaffolds. An imbalance creates phenomena like fiber pulsing or arcing which result in defective scaffolds and potential printer damage. Chapter 4 shows the use of X-ray microtomography (µCT) as a non-destructive method to characterize the pore-related features: total porosity and the pore size distribution. MEW scaffolds are three-dimensional (3D) constructs but have long been treated in the literature as two-dimensional (2D) ones and characterized mainly by microscopy, including stereo- and scanning electron microscopy, where pore size was simply reported as the distance between the fibers in a single layer. These methods, together with the trend of producing scaffolds with symmetrical pores in the 0/90° and 0/60/120° laydown patterns, disregarded the lateral connections between pores and the potential of MEW to be used for more complex 3D structures, mimicking the extracellular matrix. Here we characterized scaffolds in the aforementioned symmetrical laydown patterns, along with the more complex 0/45/90/135° and 0/30/60/90/120/150° ones. A 2D pore size estimation was done first using stereomicroscopy, followed by and compared to µCT scanning. The scaffolds with symmetrical laydown patterns resulted in the predominance of one pore size, while those with more complex patterns had a broader distribution, which could be better shown by µCT scans. Moreover, in the symmetrical scaffolds, the size of 3D pores was not able to reach the value of the fiber spacing due to a flattening effect of the scaffold, where the thickness of the scaffold was less than the fiber spacing, further restricting the pore size distribution in such scaffolds. This method could be used for quality assurance of fabricated scaffolds prior to use in in vitro or in vivo experiments and would be important for a clinical translation. Chapter 5 illustrates a proof of principle subcutaneous implantation in vivo experiment. MEW scaffolds were already featured in small animal in vivo experiments, but to date, no analysis of the foreign body reaction (FBR) to such implants was performed. FBR is an immune reaction to implanted foreign materials, including medical devices, aimed at protecting the host from potential adverse effects and can interfere with the function of some medical implants. Medical-grade PCL was used to melt electrowrite scaffolds with 50 and 60 µm fiber spacing for the 0/90° and 0/60/120° laydown patterns, respectively. These implants were implanted subcutaneously in immunocompetent, outbred mice, with appropriate controls, and explanted after 2, 4, 7 and 14 days. A thorough characterization of the scaffolds before implantation was done, followed by a full histopathological analysis of the FBR to the implants after excision. The scaffolds, irrespective of their pore geometry, induced an extensive FBR in the form of accumulation of foreign body giant cells around the fiber walls, in a manner that almost occluded available pore spaces with little to no neovascularization. This reaction was not induced by the material itself, as the same reaction failed to develop in the PCL solid film controls. A discussion of the results was given with special regard to the literature available on flat surgical meshes, as well as other hydrogel-based porous scaffolds with similar pore sizes. Finally, a general summary of the thesis in Chapter 6 recapitulates the most important points with a focus on future directions for MEW.
Cancer is one of the leading causes of death worldwide, with currently assessed chances to develop at least one cancer in a lifetime for about 20%. High cases rates and mortality require the development of new anticancer therapies and treatment strategies. Another important concern is toxicity normally associated with conventional therapy methods, such as chemo- and radiotherapy. Among many proposed antitumoral agents, oncolytic viruses are still one of the promising and fast-developing fields of research with almost a hundred studies published data on over 3000 patients since the beginning of the new millennia.
Among all oncolytic viruses, the Vaccinia virus is arguably one of the safest, with an extremely long and prominent history of use, since it was the one and only vaccine used in the Smallpox Eradication Program in the 1970s. Interestingly enough, it was the first oncolytic virus proven to have tumor tropism in vitro and in vivo in laboratory settings, and this year we can celebrate an unofficial 100th anniversary since the publication of the fact. While being highly immunogenic, Vaccinia virus DNA replication takes place in the cytoplasm of the infected cell, and virus genes never integrate into the host genome. Another advantage of using Vaccinia as an oncolytic agent is its high genome capacity, which allows inserting up to 25 kbps of exogenous genes, thus allowing to additionally arm the virus against the tumor.
Oncolytic virus action consists of two major parts: direct oncolysis and immune activation against the tumor, with the latter being the key to successful treatment. To this moment, preclinical research data are mostly generated in immunocompromised xenograft models, which have hurdles to be properly translated for clinical use. In the first part of the current study, fourteen different recombinant Vaccinia virus strains were tested in two different murine tumor cell lines and corresponding immunocompetent animal models. We found, that Copenhagen backbone Vaccinia viruses while being extremely effective in cell culture, do not show significant oncolytic efficacy in animals. In contrast, several of the LIVP backbone viruses tested (specifically, IL-2 expressing ones) have little replication ability when compared to the Copenhagen strain, but are able to significantly delay tumor growth and prolong survival of the treated animals. We have also noted cytokine related toxicity of the animals to be mouse strain specific.
We have also tested the virus with the highest therapeutic benefit in combination with romidepsin and cyclophosphamide. While the combination with histone deacetylase inhibitor romidepsin did not result in therapeutic benefit in our settings, the addition of cyclophosphamide significantly improved the efficacy of the treatment, at the same time reducing cytokine-associated toxicity of the IL-2 expressing virus.
In the second part of the work, we analyzed the ability of adipose-derived mesenchymal stem cells to serve as a carrier for the oncolytic Vaccinia virus. We showed for the first time that the cells can be infected with the virus and can generate virus progeny. They are also able to survive for a substantially long time and, when injected into the bloodstream of tumor-bearing animals, produce the virus that is colonizing the tumor. Analysis of the systemic distribution of the cells after injection revealed that infected and uninfected cells are not distributed in the same manner, possibly suggesting that infected cells are getting recognized and cleared by an impaired immune system of athymic mice faster than non-infected cells. Despite this, injection of virus-loaded adipose-derived mesenchymal stem cells to human A549 tumor-bearing xenograft mice resulted in rapid tumor regression and reduced virus-related side effects of the treatment when compared to injection of the naked virus.
In conclusion, we have tested two different approaches to augmenting oncolytic Vaccinia virus therapy. First, the combination of recombinant Vaccinia virus expressing IL-2 and cyclophosphamide showed promising results in a syngeneic mouse model, despite the low permissivity of murine cells to the virus. Second, we loaded the oncolytic Vaccinia virus into mesenchymal stem cells and have proven that they can potentially serve as a vehicle for the virus.
In dieser Arbeit wurden Bewohner/-innen der Würzburger Gemeinschaftsunterkünfte für Geflüchtete auf das Vorliegen einer Schistosomiasis gescreent. Lag eine behandlungsdürftige Infektion vor, wurden die Teilnehmenden mit Praziquantel behandelt, um im nachfolgenden Verlauf freiwillig an der Erstellung einer Schistosomen-DNA-Kinetik mitzuwirken. Eine Besonderheit der Studie lag dabei in der fehlenden Möglichkeit einer Reinfektion, da sich die Betroffenen während des Follow-ups in einem Endemie-freien Gebiet aufhielten. Für das Screening kamen ein CCA-Urin-Schnelltest sowie ein ICT zum Einsatz. Die Diagnosesicherung wurde durch die Mikroskopie oder die qPCR angestrebt. Es zeigte sich, dass die Kombination von CCA-Test und ICT einen positiven prädiktiven Wert von 80 % für das tatsächliche Vorliegen einer Schistosomen-Infektion liefert. Die Schistosomiasis-Prävalenz der hier untersuchten, in einem Nicht-Endemiegebiet lebenden Risikopopulation, wurde auf 3,9 % bestimmt und ist im Vergleich zu bisherigen Veröffentlichungen als niedrig anzusehen. Dabei ist zu beachten, dass die Prävalenz zum Teil deutlich überschätzt werden kann, sofern der CCA-Urin-Schnelltest als alleiniges Diagnosekriterium eingesetzt wird (PrävalenzCCA = 27,6 %). Die Erstellung der DNA-Kinetik mittels qPCR zeigte, dass die Behandlung mit Praziquantel einen nach 3 Tagen messbaren, signifikanten (p < 0,05) Anstieg der DNA-Konzentration im Serum zur Folge hatte, welcher im weiteren Verlauf kontinuierlich abfiel. Im Mittel wurde nach 48 Tagen der Schwellenwert der DNA-Konzentration unterschritten, der ohne vorausgegangene Behandlung als positiv und therapiebedürftig gewertet worden wäre. Durch Inter- und Extrapolation der gewonnen Daten, konnte eine Funktion errechnet werden, die den zeitlichen Verlauf des Zerfalls der Schistosomen-DNA beschreibt und somit zur Ermittlung weiterer Therapie- und Kontrollmöglichkeit der Schistosomiasis beitragen kann.
Alzheimer´s disease (AD) is a neurodegenerative disease and the most common form of dementia with still no preventive or curative treatment. Besides several risk factors, age is one of the major risks for AD and with an aging society, there is an urgent need for disease modifying agents. The strategy to address only one target within the intertwined network of AD failed so far.
Natural products especially the phytochemical flavonoids, which are poly-phenolic natural products, have shown great potential as disease modifying agents against neurodegenerative disorders like Alzheimer´s disease (AD) with activities even in vivo. Flavonoids are produced by many plants and the native Californian plant Eriodictyon californicum is particularly rich in flavonoids. One of the major flavonoids of E. californicum is sterubin, a very potent agent against oxidative stress and inflammation, two hallmarks and drivers of AD and neurodegeneration. Herein, racemic sterubin was synthesized and separated into its pure (R)- and (S)-enantiomer by chiral HPLC. The pure enantiomers showed comparable neuroprotection in vitro with no significant differences. The stereoisomers were configurationally stable in methanol, but fast racemization was observed in culture medium. Moreover, the activity of sterubin was investigated in vivo, in an AD mouse model. Sterubin showed a significant positive impact on short- and long-term memory at low dosages.
A promising concept for the increase of activity of single flavonoids is hybridization with aromatic acids like cinnamic or ferulic acids. Hybridization of the natural products taxifolin and silibinin with cinnamic acid led to an overadditive effect of these compounds in phenotypic screening assays related to neurodegeneration and AD. Because there are more potent agents as taxifolin or silibinin, the hybrids were further developed, and different flavonoid cinnamic acid hybrids were synthesized. The connection between flavonoids and cinnamic acid was achieved by an amide instead of a labile ester to improve the stability towards hydrolysis to gain better “druggability” of the compounds. To investigate the oxidation state of the C-ring of the flavonoid part, the dehydro analogues of the respective hybrids were also synthesized. The compounds show neuroprotection against oxytosis, ferroptosis and ATP-depletion in the murine hippocampal cell line HT22. While no overall trend within the flavanones compared to the flavones could be assigned, the taxifolin and the quercetin derivative were the most active compounds in course of all assays. The quercetin derivate even shows greater activity than the taxifolin derivate in every assay. As desired no hydrolysis product was found in cellular uptake experiments after 4h, whereas different metabolites were found. The last part of this work focused on synthetic bioisoteres of the natural product curcumin. Due to the drawbacks of curcumin and flavonoids arising from poor pharmacokinetics, rapid metabolism and sometimes instability in aqueous medium, we have examined the biological activity of azobenzene compounds designed as bioisoteres of curcumin, carrying the pharmacophoric catechol group of flavonoids. These bioisosteres exceeded their parent compounds in counteracting intracellular oxidative stress, neuroinflammation and amyloid-beta aggregation. By incorporating an azobenzene moiety and the isosteric behaviour to the natural parent compounds, these compounds may act as molecular tools for further investigation towards the molecular mode of action of natural products.
The present thesis adress the synthesis and characterization of novel COFs that contain dye molecules as integral components of the organic backbone. These chromophore-containing frameworks open new research lines in the field and call for the exploration of applications such as catalysis, sensing, or in optoelectronic devices. Initially, the fabrication of organic-inorganic composites by the growth of DPP TAPP COF around functionalized iron oxide nanoparticles is reported. By varying the ratio between inorganic nanoparticles and organic COFs, optoelectronic properties of the materials are adjusted. The document also reports the synthesis of a novel boron dipyrromethene-containing (BODIPY) COF. Synthesis, full characterization and the scope of potential applications with a focus on environmental remediation are discussed in detail. Last, a novel diketopyrrolopyrrole-containing (DPP) DPP-Py-COF based on the combination of DDP and pyrene building blocks is presented. The very low bandgap of these materials and initial investigations on the photosensitizing properties are discussed.

Die Darstellung wird hier in zweiter, korrigierter und ergänzter Auflage vorgelegt.

Die Erforschung viraler Proteine ist wichtig, um virale Infektionen besser verstehen und
damit therapieren zu können. Die Aufklärung der DUB-Funktion auf dem viralen
Herpesprotein pUL36 ermöglicht ein besseres Verständnis des Infektionshergangs und
könnte zur Entwicklung eines Enzyminhibitors führen, der nur an diesem Enzym ansetzt,
nachdem es sich von den zellulären DUBs unterscheidet (Kattenhorn et al., 2005). In
dieser Arbeit konnten die vorherigen Daten, die eine stärkere Hemmung der DUB-
Mutante unter Interferoneinfluss zeigten, in unterschiedlichen Assay-Designs bestätigt
werden. Auch Versuche mit einem anderen Herpes simplex Virus Strang, bestätigten die
vorherigen Daten. Die Ergebnisse zeigen, dass die DUB-Funktion für HSV-1 wichtig ist für
die virale Evasion der zellulären Immunantwort. Die genaue Funktion der DUB in der
Infektion ist jedoch unklar. Aufgrund der vorbestehenden Datenlage erschien am
wahrscheinlichsten, dass die DUB-Funktion vor Eindringen des Herpes Simplex Virus in
den Zellkern zum Tragen kommt, womit es nach Abnahme des Interferons nicht zu einer
viralen Reaktivierung käme. Deshalb wurden Untersuchungen unternommen, um eine
mögliche Reaktivierung nach Abnahme des Interferons näher zu untersuchen. Hierfür
wurden zwei verschiedene Experimente entwickelt. Einmal wurde das Interferon direkt
nach Infektion und einmal 3 Tage nach Infektion (3dpi) abgenommen. Die Ergebnisse
zeigten beide eine stärkere Hemmung der DUB-HSV-1-Mutante unter Interferoneinfluss.
Bei Abnahme des Interferons direkt nach Infektion lag bei Wildtyp und Mutante ein
leichter Anstieg der Plaquezahlen vor, wobei dieser Effekt von der Dosis des Interferons
abhängig war. Eine hohe Interferondosis begünstigte bei beiden eine stärkere Hemmung,
allerdings bei beiden auch eine leichte Erhöhung der Plaquezahl nach Abnahme. Bei
einer niedrigen Dosis konnte nur eine stärkere Hemmung der DUB-Mutante, jedoch
keine Reaktivierung bei Wildtyp und Mutante nach Abnahme des Interferons gezeigt
werden. Bei Abnahme drei Tage nach Infektion zeigte sich sowohl bei dem Wildtyp-Virus
als auch der DUB- Mutante kein Anstieg in den Plaquezahlen. Es sind, nachdem
Deubiquitinierung nicht nur eine Rolle in der Verhinderung des proteosomalen Abbaus
von in die Zelle eingedrungenem Virus spielt, sondern auch der Zellregulation, mehrere
Szenarien denkbar, die diesen Phänotyp erklären könnten. Die DUB-Funktion könnte
zwar den proteosomalen Abbau durch Deubiqutinierung und damit Verhinderung der
Markierung des Virus zum zellulären Abbau verhindern. Allerdings könnten sich durch
einen langsameren Transport aus der Zelle oder in den Nucleus auch weniger Plaques
bei der Mutante als wie beim Wildtyp unter Interferoneinfluss bilden, nachdem das Virus
dann leichter Ziel antiviraler Proteine werden könnte. Oder die DUB-Funktion spielt eine
Rolle beim Eintritt in den Kern durch Modifikationen anderer Proteine. Virengenome
könnten auch durch eine fehlende DUB-Funktion reprimiert werden oder die Zelle durch
Apoptose absterben. Interessanterweise konnte keine Hemmung der DUB-Mutante in
Interferon behandelten U-2 OS Zellen gezeigt werden, von denen ein Defekt im STING-
vermittelten Signalweg bekannt ist. Vielleicht zeigt dies, dass das STING-Protein an dem
gezeigten DUB-Phänotyp beteiligt ist. Nachgewiesen ist außerdem bereits eine Funktion
des Enzyms bei der zweiten Umhüllung der Kapside bei Pseudorabiesvirus (Möhl, 2011).
Weitere Untersuchungen unter Einsatz bspw. von Immunfluoreszenz,
Proteasominhibitoren oder weiteren Zelllinien wie Saos-2, sind nötig, um die genaue
Funktion zu klären.
The current study presents a new a group of Demotic ostraca in the belongings of the Cairo Museum. A large part of this group stem from Medinet Habu in the western bank of modern Luxor in Upper Egypt and was discovered in the beginning of the thirties of the last century by the Chicago Oriental Institute (recently renamed as Institute for the Study of Ancient Cultures ‘ISAC’). A small portion of the collection under consideration come from other Upper Egyptian provenances including Gebelein, Edfu, Kom Ombo, and possibly elsewhere in Thebes. The main goal of the present dissertation is to decipher, translate, and provide a philological, paleographical, and cultural analysis of the group of texts in question. The results of this study are spread over two main parts, the first of which is dedicated to the main and largest part of the collection, i.e. ostraca from Medinet Habu, while the second is concerned with ostraca from other places. The first part comprises of five sections beginning with receipts of money and in-kind payments including some receipts for the payments of the different capitation charges in the Ptolemaic and Roman Periods, a few for land-related payments, as well as others related to different Ptolemaic monopolies or trades such as a receipt for the price of oil, one for the linen tax, in addition to a unique receipt for the rarely attested fish tax. The second section includes accounts and lists of different kinds be it monetary, in-kind, agriculture, or any other type of lists or accounts that record different everyday transactions. The following section presents a relatively different type of lists, namely lists of personal names. The fourth section incorporates a variety of texts of different concerns, e.g. texts of religious nature, letters, temples oaths, or other private documents. Unidentified texts occupy the fifth and final section of the first part. The second part of the study, which comprises texts that originate from different Upper Egyptian localities, includes three sections, i.e. receipts, accounts, and lists of names.
